Abstract

The objective of the present study was to investigate the effects of different doses of dietary Eucommia ulmoides leaf extract (ELE) on juvenile red claw crayfish (Cherax quadricarinatus). A total number of 720 red claw crayfish (initial body weight of 0.24 ± 0.01 g) were randomly assigned to six groups and fed diets containing 0 (Diet 1), 0.5 (Diet 2), 1 (Diet 3), 2 (Diet 4), 4 (Diet 5) and 10 (Diet 6) g dry weight (dw) ELE kg (dw)−1 diets for eight weeks and challenged with microcystin-LR stress. The results indicated that dietary supplementation with 1–2 g dw ELE kg (dw)−1 diet could significantly improve the weight gain rate (WGR) and specific growth rate (SGR) of crayfish. Muscle crude protein contents of crayfish fed Diet 2, Diet 3, and Diet 4 were significantly higher than those of the control group. Compared with the control group, dietary ELE could increase total antioxidant capacity (T-AOC), superoxide dismutase (SOD), glutathione peroxidase (GPx), acid phosphatase (ACP), alkaline phosphatase (AKP), and phenoloxidase (PO) activities and decrease malondialdehyde (MDA) level of crayfish. Dietary ELE significantly increased the relative expression levels of SOD, thioredoxin 1 (TRX1), GPx, selenium-dependent glutathione peroxidase (Se-GPx), cytochrome P450 (CYP450), anti-lipopolysaccharide factor (ALF) and C-type lysozyme (C-LZM) mRNA of crayfish compared with the control group during the feeding experiment. When subjected to MC-LR stress for 48 h, the mRNA expression levels of SOD, GPx, Se-GPx, glutathione-s-transferase 1 (GST1), ALF, hemocyanin (HEM), and C-LZM in the hepatopancreas could be improved to varying degrees compared with the Diet 1. Supplementation of 1–2 g dw ELE kg (dw)−1 diet could improve the survival rate (SR) of crayfish under MC-LR stress. These results indicated that dietary ELE (1–2 g dw ELE kg (dw)−1 diet) could improve the growth performance, muscle protein, and non-specific immune response and increase the SR of crayfish under MC-LR stress by regulating the mRNA expression levels of the immune- and antioxidant-related genes. 27. Reconstructing mean temperature of April-July in 1809-2018 based on tree-ring of Pinus taiwanensis in the Tianmu Mountain, East China.

Abstract

We established 340-year chronologies of total ring width, early wood width, and late wood width with tree-ring samples of Pinus taiwanensis at high altitude collected from the western Tianmu Mountain in northern Zhejiang Province. According to the criterion that subsample signal strength (SSS) should be larger than 0.8, the reliable period was from 1810 to 2019. Through the correlation analysis between chronologies and climatic factors, we examined the responses of tree ring growth to climate. The results showed that radial growth of P. taiwanensis was more sensitive to temperature than to precipitation. Comprehensively considering the correlation analysis results for the raw and first-order difference series, early wood width was significantly correlated with the early growing season mean and maximum temperatures of the prior year, while late wood width with prior May and current September mean and maximum temperatures. The correlation pattern of total ring width was similar to that of early wood width, although at a low level. The optimal correlation was between early wood width and prior April-July mean temperature. Based on this relationship, April-July mean temperature of the Tianmu Mountain, East China was reconstructed for the period of 1809-2018 with an explained variance of 61.5%. Both the raw and first-order difference series passed the split sample calibration-verification test. The warm periods were 1809-1833 and 1965-2018, with a cold period in 1834-1964. Temperature had risen rapidly since the 1960s. From the standpoint of low frequency, it reached an unprecedented level since the 1980s over the past 210 years. Spatial correlation analysis showed that the reconstructed temperature series could represent temperature variations of East China, which had a good agreement with a reconstructed regional temperature series from East China. However, the efficacy of this treatment strategy is limited for treatment of triple negative breast cancer (TNBC). Gene therapy may be a more effective strategy for improving the prognosis of TNBC.Methods: A novel 25 nucleotide sense strand of miRNA was designed to treat TNBC by silencing the Slug gene, and encapsulated into DSPE-PEG2000-tLyp-1 peptide-modified functional liposomes. The efficacy of miRNA liposomes was evaluated on invasive TNBC cells and TNBC cancer-bearing nude mice. Furthermore, functional vinorelbine liposomes were constructed to investigate the anticancer effects of combined treatment.Results: The functional miRNA liposomes had a round shape and were nanosized (120 nm). Functional miRNA liposomes were effectively captured by TNBC cells in vitro and were target to mitochondria. Treatment with functional liposomes silenced the expression of Slug and Slug protein, inhibited the TGF-β1/Smad pathway, and inhibited invasiveness and growth of TNBC cells. In TNBC cancer-bearing mice, functional miRNA liposomes exerted a stronger anticancer effect than functional vinorelbine liposomes, and combination therapy with these two formulations resulted in nearly complete inhibition of tumor growth. Preliminary safety evaluations indicated that the functional miRNA liposomes did not affect body weight or cause damage to any major organs. Furthermore, the functional liposomes significantly increased the half-life of the drug in the blood of cancer-bearing nude mice, and increased drug accumulation in breast cancer tissues.Conclusion: In this study, we constructed novel functional miRNA liposomes. These liposomes silenced Slug expression and inhibited the TGF-β1/Smad pathway in TNBC cells, and enhanced anticancer efficacy in mice using combined chemotherapy. Abstract

Transcatheter arterial chemoembolization (TACE) is an effective therapy for hepatocellular carcinoma (HCC). However, acute kidney injury (AKI) may occur after TACE due to the contrast agent and cytotoxic anticancer drugs used in this procedure. Post-TACE AKI is not an unusual event, and may adversely affect patient outcome. Coexisting situations including cirrhosis, renal insufficiency, diabetes and hypertension play a role in the development of HCC, and may predispose patients to AKI after TACE. Most post-TACE are transient and reversible, while prolonged AKI may predict a decreased survival. The best strategy to manage post-TACE AKI is prevention. Patients, before undergoing TACE, should be carefully assessed. In this study, we reviewed the current literature published in English about the incidence rate, risk factors, management and prognosis of AKI in patients with HCC undergoing TACE for a better understanding of this complication.

Abstract

This study was to investigate the effects of Alpinia officinarumHance stems and leaves extract (AOE) on the growth, non-specific immunity, intestinal microflora of shrimp (Litopenaeus vannamei). A total of 540 juvenile L. vannamei(2.69 ± 0.03 g) were randomly divided into six groups, and feed experiment with diet supplement of 0 g/kg (CG), 0.1 g/kg (AO-1), 0.2 g/kg (AO-2), 0.3 g/kg (AO-3), 0.5 g/kg (AO-4) and 0.7 g/kg(AO-5) AOE was carried out for 8 weeks, respectively. Compared with the control group (CG), the WGR and SGR of the AO-1, AO-3, and AO-4 groups were significantly increased. Crude protein content in the muscle of L. vannameiin AO-2 was significantly increased, while the water content was significantly decreased. The crude lipid content of each extract group was significantly decreased. Compared with the control group, activities of GSH-Px, SOD, and T-AOC in the cell-free hemolymph of AO-3 group were significantly enhanced, and the MDA content of each extract group was significantly decreased. Activities of CAT, SOD, and T-AOC in the hepatopancreas of AO-2 group were significantly increased. Gene expression levels of lvcat, lvgsh-px,and lvmn-sodin AO-3 group were obviously higher than those in the control group, while lvpropoexpression was dramatically higher in AO-2 and AO-3 groups. Moreover, AOE addition in feed changed the composition and species abundance of intestinal microflora of L. vannameiat phylum and genus levels. Through α diversity analysis, the biodiversity of the flora was improved, while β diversity analysis changed the similarity of the flora between the control and addition groups. Therefore, addition of 0.2–0.3 g/kg AOE could improve the growth, immunity and optimize the composition of intestinal microorganisms of L. vannamei.
