Our study endeavors to investigate the causal association between BC and CVD, with a focus on identifying potential metabolic mediators and elucidating their mediation effects in this causality. Methods: In this study, we conducted two-sample Mendelian randomization (MR) to estimate the causal effect of BC (overall BC, ER+ BC, ER- BC) from the Breast Cancer Association Consortium (BCAC) on CVD including coronary heart disease (CHD), hypertensive heart disease (HHD), ischaemic heart disease (IHD), and heart failure (HF) from the FinnGen consortium. Then, we used two-step MR to evaluate 18 metabolic mediators of the association and calculate the mediated proportions. Results: Genetically predicted ER+ BC was causally associated with an increased risk of CVD including CHD (OR = 1.034, 95% CI: 1.004– 1.065, p = 0.026), HHD (OR = 1.061, 95% CI: 1.002– 1.124, p = 0.041), IHD (OR = 1.034, 95% CI: 1.007– 1.062, p=0.013), and HF (OR = 1.055, 95% CI: 1.013– 1.099, p = 0.010), while no causality was observed for overall BC and ER- BC. Furthermore, high-density lipoprotein cholesterol (HDL-C) was identified as a mediator of the association between ER+BC and CVD, including CHD (with 15.2% proportion)) and IHD (with 15.5% proportion), respectively. Conclusion: This study elucidates the potential causal impact of ER+ BC on subsequent risk of CVD, including CHD, HHD, IHD, and HF. We also outline the metabolic mediator HDL-C as a priority target for preventive measures to reduce excessive risk of CVD among patients diagnosed with ER+BC. [ABSTRACT FROM AUTHOR]

Abstract

Cancer vaccines represent a promising form of immunotherapy employed in the treatment of cancer. However, their efficiency in eliciting immune responses is limited, and satisfactory results have yet to be achieved. Optimizing adjuvants and antigens is an important approach to promoting the anti‐tumor efficacy of cancer vaccines. Here, a novel nanoadjuvant (LNP/siRNA) designed to silence T‐cell immunoglobulin and mucin‐domain containing‐3 (TIM3) and activate Toll‐like receptors (TLRs) is presented. The LNP/siRNA demonstrates significant potential in promoting dendritic cell (DC) maturation and enhancing the anti‐tumor response. Furthermore, desialylated cancer cell membrane is utilized as antigens, providing a variety of tumor antigens for DCs and enhancing their function. Additionally, they are integrated to create a core‐shell structured nanovaccine (dClip‐LNP/siRNA) through coextrusion, which collectively enhances the cross‐presentation ability of DCs, thus achieving a dual enhancement strategy. The dClip‐LNP/siRNA significantly silences TIM3 expression in DCs and promotes antigen presentation by DCs. Besides, dClip‐LNP/siRNA significantly promotes the activation of T cells in lymph nodes and induces robust and durable anti‐tumor immunity in tumor sites to eliminate established B16‐OVA tumors, prevent tumor occurrence, and suppress tumor lung metastasis. The dClip‐LNP/siRNA is also suitable for combination with adoptive OT‐I cell therapy to enhance cancer immunotherapy. Abstract

The red blood cell membrane (RBCm) provides tight protection, lowers the immunogenicity, and prolongs the circulation time of drugs in vivo when acting as the coating of drug delivery systems. However, the cellular uptake and release of drugs are hindered by RBCm. Docetaxel (DTX) is the first‐line medicine for treating triple‐negative breast cancer (TNBC), but it induces tumor metastasis. To solve these dilemmas, in this study, the photosensitizer 1,1‐dioctadecyl‐3,3,3,3‐tetramethylindotricarbocyanine iodide (DiR)‐modified RBCm (DM) is prepared, which is coated onto a hybrid micelle consisting of the prodrugs of DTX and the anti‐metastasis agent calcitriol (CTL), obtaining a nanoparticle, named HDC‐DM. In a 4T1 tumor‐bearing mouse model, after injecting HDC‐DM, the intratumoral DTX and CTL concentrations are increased by 1.7 and 2.5 times compared with the free drug groups. After irradiating tumors with near‐infrared laser, DiR elicits the photothermal effect, triggering the rupture of RBCm and drug release, promoting drug penetration in tumors, and inducing immunogenic cell death. The tumor growth inhibition rate is 77%, and the formation of lung metastases is reduced by 82%, with good biocompatibility. It is suggested that the combination of phototherapy, chemotherapy, and anti‐metastatic therapy using HDC‐DM is expected to be a powerful strategy for treating TNBC. [ABSTRACT FROM AUTHOR]

Abstract

The first mitochondrion-anchoring photosensitizer that specifically generates singlet oxygen (1O2) in mitochondria under white light irradiation that can serve as a highly effective radiosensitizer is reported here, significantly sensitizing cancer cells to ionizing radiation. An aggregation-induced emission luminogen (AIEgen), namely DPA-SCP, is rationally designed with α-cyanostilbene as a simple building block to reveal AIE, diphenylamino (DPA) group as a strong electron donating group to benefit red emission and efficient light-controlled 1O2 generation, as well as a pyridinium salt as the targeting moiety to ensure specific mitochondrial localization. The AIE signature endows DPA-SCP with the capacity to visualize mitochondria in a fluorescence turn-on mode. It is found that under optimized experimental condition, DPA-SCP with white light does not lead to apoptosis/death of cancer cells, whereas provides an elevated 1O2 environment in the mitochondria. More importantly, increasing intracellular level of 1O2 originated from mitochondria is demonstrated to be a generic method to enhance the radiosensitivity of cancer cells with a supra-additive synergistic effect of “0 + 1 > 1.” Noteworthy is that “DPA-SCP + white light” achieves a high SER10 value of 1.62, which is much larger than that of the most popularly used radiosensitizers, gold nanoparticles (1.19), and paclitaxel (1.32). © 2017 WILEY-VCH Verlag GmbH & Co. Recently, resveratrol (Res), herbal medicine extracted from grape seeds, has been demonstrated for its potential use in chemosensitization. In the current study, both these drugs were loaded simultaneously into nanoparticles with methoxy poly(ethylene glycol)-poly epsilon caprolactone (mPEG-PCL) as drug carriers in order to achieve better antitumor efficiency. Tem/Res-coloaded mPEG-PCL nanoparticles were constructed, characterized, and tested for antitumor effect on glioma cells by using in vitro and xenograft model system. The nanoparticle constructs were satisfactory with drug loading content (Res =~12.4%; Tem =~9.3%) and encapsulation capacity of >85% for both the drugs. In addition, the coencapsulation led to better in vitro stability of the nanoparticles than Tem-loaded nanoparticles. An in vitro uptake study demonstrated a high uptake efficiency of the nanoparticles by glioma cells.
