This study proposed an improved bio-carbonation of reactive magnesia cement (RMC) method for dredged sludge stabilization using the urea pre-hydrolysis strategy. Based on unconfined compression strength (UCS), pickling-drainage, and scanning electron microscopy (SEM) tests, the effects of pre-hydrolysis duration (T), urease activity (UA) and curing age (CA) on the mechanical properties and microstructural characteristics of bio-carbonized samples were systematically investigated and analyzed. The results demonstrated that the proposed method could significantly enhance urea hydrolysis and RMC bio-carbonation to achieve efficient stabilization of dredged sludge with 80% high water content. A significant strength increment of up to about 1063.36 kPa was obtained for the bio-carbonized samples after just 7 d of curing, which was 2.64 times higher than that of the 28-day cured ordinary Portland cement-reinforced samples. Both elevated T and UA could notably increase urea utilization ratio and carbonate ion yield, but the resulting surge in supersaturation also affected the precipitation patterns of hydrated magnesia carbonates (HMCs), which weakened the cementation effect of HMCs on soil particles and further inhibited strength enhancement of bio-carbonized samples. The optimum formula was determined to be the case of T = 24 h and UA = 10 U/mL for dredged sludge stabilization. A 7-day CA was enough for bio-carbonized samples to obtain stable strength, albeit slightly affected by UA. The benefits of high efficiency and water stability presented the potential of this method in achieving dredged sludge stabilization and resource utilization. This investigation provides informative ideas and valuable insights on implementing advanced bio-geotechnical techniques to achieve efficient stabilization of soft soil, such as dredged sludge.

Fresh water shortage is a major problem for grain production in the low plain around Bohai Sea in the North China Plain. Relative abundance of shallow saline groundwater could serve as an alternative water resource for use during dry seasons. A continuous 8-year field study was conducted from 2015 to 2023 to assess the effects of salt content in irrigation water on soil salt accumulation and crop production. Fresh water (FW) with electrical conductivity (EC) at 1.6 dS/m and three levels of saline water (SW) with EC at 4.7 (SW1), 6.3 (SW2) and 7.8 dS/m (SW3) were used for irrigation. Results showed that a single irrigation event at the jointing stage of winter wheat increased grain production averagely by 18.6 %, 22.5 %, 12.9 % and 9.5 % compared with a rain-fed treatment (RF) under FW, SW1, SW2 and SW3, respectively. With an additional irrigation applied at flowering stage, both irrigations using FW increased the yield by 28.6 %, and both irrigations using SW2 increased the yield by 19.3 % compared with RF. Negative effects of salt on winter wheat overshadowed the positive effects of increased water supply under two irrigations both using SW. With an irrigation at maize sowing and the subsequent summer rainy season, the yield of maize following winter wheat was not affected by a one-time SW irrigation to the previous crop, but showed a 5.3 % yield reduction when two irrigations of SW were applied. There was no apparent salt accumulation in the top 1 m of the soil profile, but a slight increasing trend in the salt content in the 1–2 m layer of the soil profile under SW2 and SW3 irrigation. No apparent changes in soil physical properties were observed for continuous application of SW. It was suggested that SW with EC not exceeding 6.3 dS/m should be applied for a single irrigation during the winter wheat season. This practice could alleviate the fresh water shortage in this region and allow for the maintenance of a relatively stable yield of winter wheat and maize without the risk of salt accumulation in the soil.

Using drought-resistant cultivars is important to increase crop production in water limited regions. Cultivar performance is influenced by genetic, management and environmental factors, and selection good performance cultivars is often a challenge. Five seasons (2016–2021) of field experiments and three seedling growing durations in a greenhouse, both under three water supply levels (serious water deficit, moderate water deficit and normal water supply), were conducted to assess 10 winter wheat cultivars for their water use characteristics and drought tolerance ability based on seven drought resistance indices and agronomic traits. The performance of the cultivars varied seasonally under field growing conditions. Drought resistance screened by a single index or based on the results from a single season was inconsistent. To avoid seasonal changes in cultivar performance, principal component analyses (PCA) and hierarchical cluster analysis (HCA) were conducted based on the seven indices to classify the ten cultivars into four types of drought responses. The drought resistance classification under field growing conditions at the grain yield level was highly consistent with that obtained under greenhouse growing conditions for biomass at the seedling stage. The results showed that the biomass at the seedling growing stage of the winter wheat cultivars could be used to predict the yield performance and water use characteristics when grown in the field under different water supply conditions. Precisely controlled greenhouse pot experiments could amplify the water use characteristics of different wheat cultivars for assessment. The cultivars with strong stomatal control ability had higher drought resistance and biomass production. Under field growing conditions, lower canopy temperature, deep root growth, a high root/shoot ratio and the maintenance of stomatal conductance under water stress were associated with high drought resistance. These findings provide references for selection methods and indicators for assessing drought-resistant winter wheat cultivars under different growing conditions.

AbstractThe complete mitochondrial genomes of two spine-color individuals, red and white, of the tropical sea urchin species Tripneustes gratilla (Linnaeus, 1758) were sequenced on Illumina system platform. The red-spined species had a genome size of 15,774 bp, while the white-spined species had a genome size of 15,723 bp. Both genomes contained 13 protein-coding genes, 2 rRNA genes, and 22 tRNA genes. The GC composition in both species was above 40%. In order to investigate the phylogenetic relationships of two different spine-color individuals, a comprehensive analysis was conducted using eight complete mitochondrial genomic sequences of the genus Tripneustes on the software MEGA X. It was observed that the two spine color types of T. gratilla species showed a high similarity of 98.91%. However, different color-spined species of T. gratilla were found in separate branches of the phylogenetic tree of the same sea urchin species.

BackgroundImmune checkpoint therapy, involving the programmed cell death 1 (PD-1) monoclonal antibody, has revolutionized the treatment of cancer. Tertiary lymphatic structure (TLS) serves as an immune indicator to predict the efficacy of PD-1 antibody therapy. However, there is no clear result whether the distribution, quantity, and maturity of TLS can be effective indicators for predicting the clinical efficacy of anti-PD1 immunotherapy in patients with colorectal cancer (CRC).MethodsFifty-seven patients who underwent surgical resection and thirty-nine patients who received anti-PD-1 immunotherapy were enrolled in this retrospective study. Immunohistochemical staining and multiple fluorescence immunohistochemistry were used to evaluate the mismatch repair (MMR) subtypes and TLS distribution, quantity, and maturity, respectively.ResultsA comprehensive patient score system was built based on TLS quantity and maturity. We found that the proportion of patients with score >1 was much higher in the deficient mismatch repair(dMMR) group than in the proficient mismatch repair(pMMR) group, and this difference was mainly due to intratumoral TLS. Patient score, based on the TLS evaluation of whole tumor, peritumor, or intratumor, was used to evaluate the efficacy of anti-PD1 immunotherapy. Based only on the intratumor TLS evaluation, the proportion of patients with a score >1 was higher in the response (PR + CR) group than in the non-response (PD) group. Multivariate analysis revealed that patient scores were positively correlated with the clinical efficacy of immunotherapy. Further analysis of immune-related progression-free survival was performed in patients with CRC who received anti-PD-1 immunotherapy. Patients with score >1 based on the intratumor TLS evaluation had significantly better survival.ConclusionsThese results suggest that the patient score based on intratumor TLS evaluation may be a good immune predictive indicator for PD-1 antibody therapy in patients with CRC.

Noninvasive and nondestructive root phenotyping techniques under field conditions are needed to advance plant root science. Soil electrical parameters including capacitance and resistance hold potential to meet this need, although their specific ability to detect roots remains uncertain. In this study, we developed a two-phase root and soil permittivity model at high frequency enabling accurate and noninvasive prediction of root volume. The validation calculation showed that the two-phase model successfully predicts root volume with a minimal error of less than 1 % under the control conditions with roots placed in wet soil and water. Furthermore, we demonstrated that the high-frequency bulk soil capacitance when integrated into an empirical model, can accurately estimate root volume for summer maize (Zea mays L.) in both the field (root mean square error (RMSE) = 1.52 cm3) and in root washing-free measurement applications in the absence of soil (RMSE = 0.36 cm3). When the empirical relationship between high-frequency bulk soil capacitance and root volume is not adequate, the two-phase model can be used to calibrate the empirical model. To this end, a series of soil pot experiments with winter wheat (Triticum aestivum L.) showed that the measurement accuracy can be fundamentally improved. These results highlighted the effectiveness of the two-phase permittivity model, making high-frequency bulk soil capacitance a reliable method for in situ measurement of root volume. It is anticipated that the availability of new equipment for measuring high-frequency soil electrical parameters will lead to widespread adoption of this root phenotyping technique in the future.

BackgroundThe infiltration and activation of immune cells in the tumor microenvironment (TIME) affect the prognosis of patients with cancer. Tertiary lymphoid structure (TLS) formation favors tumour- infiltrating-lymphocyte (TIL) recruitment and is regarded as an important indicator of good prognosis associated with immunotherapy in patients with tumors. Chemotherapy is currently one of the most commonly used clinical treatment methods. However, there have been no clear report to explore the effects of different types of chemotherapy on TLS formation in the TIME. This study examined the effects of immunogenic cell death (ICD)-inducing chemotherapeutics on immune cells, high-endothelial venules (HEV), and TLSs in mouse melanomas.MethodsDoxorubicin (an ICD inducer), gemcitabine (non-ICD inducer), and a combination of the two drugs was delivered intra-peritoneally to B16F1-loaded C57BL/6 mice. The infiltration of immune cells into tumor tissues was evaluated using flow cytometry. HEV and TLS formation was assessed using immunohistochemistry and multiple fluorescent immunohistochemical staining.ResultsDoxorubicin alone, gemcitabine alone, and the two-drug combination all slowed tumor growth, with the combined treatment demonstrating a more pronounced effect. Compared with the control group, the doxorubicin group showed a higher infiltration of CD8+ T cells and tissue-resident memory T cells (TRM) and an increase in the secretion of interferon-γ, granzyme B, and perforin in CD8+ T subsets and activation of B cells and dendritic cells. Doxorubicin alone and in combination with gemcitabine decreased regulatory T cells in the TIME. Moreover, doxorubicin treatment promoted the formation of HEV and TLS. Doxorubicin treatment also upregulated the expression of programmed cell death protein (PD)-1 in CD8+ T cells and programmed cell death protein ligand (PD-L)1 in tumor cells.ConclusionsThese results indicate that doxorubicin with an ICD reaction promotes TLS formation and increases PD-1/PD-L1 expression in tumor tissues. The results demonstrate the development of a therapeutic avenue using combined immune checkpoint therapy.

Abstract Powdery mildew is a major disease in melon, primarily caused by Podosphaera xanthii (Px). Some melon varieties were resistant to powdery mildew, while others were susceptible. However, the candidate genes associated with resistance and the mechanism of resistance/susceptibility to powdery mildew in melon remain unclear. In this study, disease-resistant melon cultivar TG-1 and disease-susceptible melon cultivar TG-5 were selected for comparative transcriptome analysis. The results suggested that the numbers of differentially expressed genes (DEGs) in TG-5 was always more than that in TG-1 at each of the four time points after Px infection, indicating that their responses to Px infection may be different and that the active response of TG-5 to Px infection may be earlier than that of TG-1. Transcription factors (TFs) analysis among the DEGs revealed that the bHLH, ERF, and MYB families in TG-1 may play a vital role in the interaction between melon and powdery mildew pathogens. GO enrichment analysis of these DEGs in TG-5 showed that the SBP, HSF, and ERF gene families may play important roles in the early stage of melon development after Px infection. Finally, we speculated on the regulatory pathways of melon powdery mildew and found PTI and ABA signaling genes may be associated with the response to Px infection in melon.

The water quality of the Golmud River is essential for environmental preservation and economic growth of Golmud city and Qarhan Salt Lake in China. Thirty-four samples of surface water and sediment from seventeen places in the Golmud River and thirty-two dustfall samples in the Qaidam Basin were collected. The concentrations of heavy metals (HMs) were measured; water quality, risk assessment, and multiple source analysis were applied. Concentrations of HMs in water were Zn > Cu > Ni > As > Pb > Cd > Hg, and in sediment were Ni > Zn > Pb > As > Cu > Cd > Hg. In water, the Nemerow pollution index (NP) values indicated that most of the sampling points seemly were seriously polluted; other water quality assessment results suggested no pollution. In sediment, the concentrations of 27% HMs exceeded the background values of soil in Qinghai; 48% exceeded the Earth crust background values, which were As, Hg, and Cd. The single factor index method (Pi), geological accumulation index (Igeo), and contamination factor (CF) revealed that As pollution is serious, followed by Hg and Cd; the pollution load index (PLI) and modified pollution index (mCd) values indicated that 64% and 57% of samples were polluted. NP values are shown serious pollution. The ecological risk results demonstrated a low risk in water and a medium risk in sediment. The average total hazard quotient values in sediment and water for adults and children revealed low non-carcinogenic risks. Carcinogenic risk indicated Ni in water and sediment, and As in sediment may be involved in cancer risk. Multivariate statistics showed that the HMs mainly came from nature, and human activities will also impact them. The upper continental crust values indicated that As and Hg have high background values. The saline dust storm was one of the essential sources of HMs, especially Hg. Various provenances constituted the material cycling of HMs in the surface environment.

Improving water use efficiency (WUE) has been proven to be a prosperous way to produce more grain in drought-prone areas. Transpiration efficiency (TE) has been proposed as a criterion for screening cultivars with high WUE. This study quantifies the relations of TE to relative soil water content (RSWC) gradients using pot experiments and evaluates the capability of the relations of TE-RSWC on assessing the cultivar performance in field yield and WUE. Twelve winter wheat cultivars were grown at 6 RSWC, 12.1, 24.2, 36.3, 48.4, 60.5, and 72.6% of field capacity (FC = 24.8 g/g) for 33 days in tightly sealed pots preventing soil evaporation. The results showed that TE decreased power functionally following the increase in RSWC for all cultivars. The relationship could be described as TE = TE FC × (RSWC) b, named TE–RSWC curve. This curve could be divided into an orderly area where the rank of cultivars was stable when RSWC ≤ 12.1% or RSWC ≥ 72.6% and a disorderly area where the rank was unstable when 12.1% < RSWC < 72.6%. To assess the consistency of pot TE to field yield and WUE, the same 12 varieties were grown under rainfed and two irrigations (75 mm at the jointing and flowering stages, respectively). TE FC was found to be positively related to field yield and WUE independent of irrigation. TE measured near the wilting point was negatively related to field yield and WUE. These results indicated that TE FC could be used as a surrogate for screening high-yield and high-WUE cultivars. The consistency and inconsistency can be attributed to the orderly area and disorderly area of the TE–RSWC curves.

The Qinghai-Tibet Plateau is a unique area with water sources for approximately 40 % of the population in the world. Water resources and water quality are closely associated with ecological security and human health. Fifty-one trace elements in surface water samples (n = 40) were measured, and water quality, health and ecological risks were assessed. Trace elements showed significant variations in different surface water bodies in the study area. Concentrations of minor elements were relatively high in saline and salt lakes while those of REEs varied from 0.05 to 33.62 μg/L with an average value of 3.80 μg/L. The Nemerow pollution index (NP) values of trace elements ranged from 0.08 to 3.48, with an average value of 0.36 in rivers, fresh lakes and reservoir water samples; The heavy metal pollution index (HPI) values ranged from 3.70 to 21.18, indicating that most samples were within the critical limit; The heavy metal evaluation index (HEI) values and degree of contamination (DC) values indicated a free pollution status. The water quality index (WQI) values showed that 96 % of the samples belonged to excellent status in rivers, fresh lakes and reservoir water samples. More attention should be given to the Cr, Zn and Hg in the study area according to potential ecological risk assessment. Hazard quotients for residential children in 30 sites exceed 1.0 with maximal value of 10.97, suggesting the high non-carcinogenic risks for children in the study area. U, Zr and Cr for the ingestion pathway, Cr and U for the dermal pathway were primary contributors to the total health risk. Carcinogenic risk values of trace elements for residential and recreational receptors were in the range of 3.20 × 10−5-7.38 × 10−3 and 8.62 × 10−6-3.63 × 10−3, respectively. The carcinogenic risk values of Cr in surface water were higher than the target risk of 1 × 10−4, while the carcinogenic risk values of As were below the target risk. The results of this study provided information on trace elements for human health protection and water management in the northeastern Qinghai-Tibet Plateau.

The micromixer is a key component of the microfluidic chip analysis system. Micromixers are widely used in applications, such as DNA hybridization and protein synthesis. A high-efficiency mixer can speed up the biochemical analysis process. In order to study how to improve the mixing efficiency of the mixer, this paper designs passive micromixers with three different blocks: cylindrical, equilateral triangle, and square. The effects of them on the mixing performance and pressure drop of the mixer were studied, respectively. Through numerical simulation, the study shows that the mixing efficiency of the mixer with equilateral triangle blocks is 96% at Re = 100, and the maximum pressure drop is 18 135.8 Pa. In addition, through the analysis of three-dimensional numerical simulation, the block causes the fluid to generate a horizontal and vertical vortex flow state in the mixing unit, thereby breaking the laminar flow and greatly improving the mixing efficiency. Through structural optimization, ETOM4, which has four mixing units and a side length of 150 μm equilateral triangle blocks, is the best passive micromixer with its mixing efficiency of 99.1%.

Creativity has become prevalent in the routine work of knowledge employees in contemporary enterprises. From the perspective of ambidexterity, drawing upon expectancy theory and self-determination theory (SDT), the present study highlights the driver behind proactive and responsive creativity. Using two-stage longitudinal data collected from 373 knowledge employee-supervisor dyads in information and manufacturing companies in China, the results show that: (1) performance-contingent rewards have an inverted U-shaped influence on proactive creativity and a U-shaped influence on responsive creativity; (2) performance-contingent rewards have an inverted U-shaped influence on autonomous and controlled motivation; (3) autonomous motivation has a positive influence on proactive creativity, but controlled motivation has a negative influence on responsive creativity; (4) autonomous and controlled motivation play a partly mediating mechanism in the non-linear effects of performance-contingent rewards on proactive and responsive creativity, respectively. Managers should enhance the ambidextrous ability to deal with proactive and responsive creativity and establish an applied and dynamic policy of performance-contingent rewards intensity to drive ambidextrous creativity.

The Nangqen and Qamdo (NQ-QD) basins in China have very rich geothermal and brine resources. The origin and spatiotemporal evolutionary processes of its hot and saline springs however remain unclear. Geochemical and isotopic (18O, 2H, 3H) studies have therefore been conducted on the water from the geothermal and saline springs in the NQ-QD Basin. All saline springs in the study area are of the Na-Cl geochemical type while geothermal waters show different geochemical types. The oxygen and hydrogen isotopic compositions of the springs in the NQ-QD Basin are primarily controlled by meteoric water or ice-snow melt water and are influenced by rock-water interactions. It is found that the saline springs in the study area are derived from the dissolution of halite and sulfate that occur in the tertiary Gongjue red bed, while the hot springs in the QD Basin are greatly influenced by the dissolution of carbonatites and sulfates from the Bolila (T3b) and Huakaizuo (J2h) formations. Results from silica geothermometry and a silicon-enthalpy hybrid model indicate that the apparent reservoir temperatures and reservoir temperatures for the hot springs in the QD Basin range from 57–130°C to75–214°C, respectively. Deuterium analysis indicates that most of the hot springs are recently recharged rain water. Furthermore, the saline springs have a weaker groundwater regeneration capacity than the hot springs. Tritium data shows that the ranges of calculated residence times for springs in this study are 25 to 55 years, and that there is a likelihood that hot springs in the QD Basin originated from two different hydrothermal systems. The geochemical characteristics of the NQ-QD springs are similar to those of the Lanping-Simao Basin, indicating similar solute sources. Thus, the use of water isotope analyses coupled with hydrogeochemistry proves to be an effective tool to determine the origin and spatiotemporal evolution of the NQ-QD spring waters.

Kunteyi Salt Lake (KSL), located in the northwest of the Qaidam Basin (QB), is rich in polyhalite resources. However, there is no relevant research on the ore-formed temperature of polyhalite in nature, such as KSL. The homogenization temperature (Th) of salt mineral inclusions can directly reveal the form temperature of minerals. In view of the poor diagenesis of polyhalite in KSL, almost no polyhalite crystals are formed. Therefore, the ore-formed temperature of polyhalite in KSL is revealed by using the Th of fluid inclusions in halite associated with polyhalite as a substitute index. A total of 472 Th data from 34 halite samples and 34 maximum homogenization temperature (ThMAX) data ranged from 17.1°C to 35.5°C, among which 24 data were concentrated at 17-23°C and the average value is 22.1°C. Brine temperature of other salt lakes in QB and paleoclimate characteristics of the study area were combined. It suggests that the temperature conditions of polyhalite mineralization in the study area are generally low. However, under the overall low-temperature background, polyhalite seems to be easily enriched at relatively high temperature; for example, the content of polyhalite is generally high in the first relatively dry and hot salt-forming period, and the brine temperature at the peak stage of polyhalite at 45 m is relatively high, which indicates that the high temperature conditions promote the formation of polyhalite in KSL. As far as the overall relationship between temperature and polyhalite is concerned, polyhalite is deposited at both low temperature and relatively high temperature, which verifies the previous understanding that polyhalite is a mineral with wide temperature phase, and also shows that temperature has a limited effect on polyhalite formation under natural conditions. In addition, combined with the chemical composition of halite fluid inclusions, it is found that the concentration of Mg2+ in nature has an influence on the temperature measurement process. According to the previous experimental research, speculate that the actual temperature of ancient brine and ore-formed temperature of polyhalite in KSL are lower than the measured Th. The confirmation of the influence of Mg2+ on temperature measurement is convenient for more accurate reconstruction of the metallogenic temperature of evaporite such as polyhalite. The research on the ore-formed temperature of KSL polyhalite enriches and perfects the polyhalite mineralization theory and provides theoretical basis for the basic and applied research of polyhalite.

Agricultural droughts cause a great reduction in winter wheat productivity; therefore, timely and precise irrigation recommendations are needed to alleviate the impact. This study aims to assess drought stress in winter wheat with the use of an unmanned aerial system (UAS) with multispectral and thermal sensors. High-resolution Water Deficit Index (WDI) maps were derived to assess crop drought stress and evaluate winter wheat actual evapotranspiration rate (ETa). However, the estimation of WDI needs to be improved by using more appropriate vegetation indices as a proximate of the fraction of vegetation cover. The experiments involved six irrigation levels of winter wheat in the harvest years 2019 and 2020 at Luancheng, North China Plain on seasonal and diurnal timescales. Additionally, WDI derived from several vegetation indices (VIs) were compared: near-infrared-, red edge-, and RGB-based. The WDIs derived from different VIs were highly correlated with each other and had similar performances. The WDI had a consistently high correlation to stomatal conductance during the whole season (R2 between 0.63–0.99) and the correlation was the highest in the middle of the growing season. On the contrary, the correlation between WDI and leaf water potential increased as the season progressed with R2 up to 0.99. Additionally, WDI and ETa had a strong connection to soil water status with R2 up to 0.93 to the fraction of transpirable soil water and 0.94 to the soil water change at 2 m depth at the hourly rate. The results indicated that WDI derived from multispectral and thermal sensors was a reliable factor in assessing the water status of the crop for irrigation scheduling.

Polyhalite occurrence in the Kunteyi Playa in the Qaidam Basin has been known for many years. However, the genetic mechanism of this deposit remains unclear. In this study, a typical section in the playa depocenter is selected to study the polyhalite mineralogy combined with the homogenization temperature and composition of halite fluid inclusions in shallow evaporitic strata. The results show that 1) the main evaporite minerals in the strata are halite and polyhalite; no common gypsum is found; 2) analyses of homogenization temperatures of halite fluid inclusions indicate that a higher temperature is needed for polyhalite generation compared with other saline minerals; and 3) the fluid inclusion chemical analysis shows that they are sulfate-type minerals with a shortage of Ca. Thus, it can be concluded that the formation of polyhalite is not related to gypsum replacement, and deep oilfield brines may provide a Ca source and a higher temperature for polyhalite formation, where the mixing and interaction occurred between K- and Mg-enriched sulfate brines and deep Ca-enriched brines under the control of climate and tectonics in the study area. While most polyhalite was generated natively, some formed during secondary generation, which was potentially related to replacement with carnallites or sylvites.

The fluid inclusion composition of halite can help track chemical composition of ancient fluids and, thus, serves as a reliable index to analyze ancient brine in salt lakes. Qarhan Salt Lake (QSL) is the largest potash brine deposit in China. Although the mixing of modern river water and Ca-Cl deep water is widely accepted as potassium formation, the mixing characteristics in the time domain and driving factors of deep water are still unclear. Here, the chemical composition of fluid inclusions in primary halite samples collected from the ISL1A borehole in QSL was measured by LA-ICP-MS technology. The analysis results show that, during the formation stage of the S4 salt layer in QSL, the main potassium salt layer, the contents of Ca2+ and Sr2+ in brine increased significantly. There is evidence confirming that Ca-Cl deep water is beneficial to the enrichment of potassium and the surrounding rivers generally develop terraces. It suggests that, during the formation stage of the QSL potassium salt layer, more Ca-Cl inflow water of the northern margin supplies the salt lake, inferring that it was driven by tectonic activities. In addition, the chemical composition of halite fluid inclusions shows that there is an anomaly in geochemistry at the early stage of salt formation in QSL. By combining the time of tectonic activities, it is inferred that the anomaly is not caused by tectonic activities but maybe caused by a salt-forming event. This work indicates that deep water and tectonic movement have a huge impact on the evolution of salt lakes. Therefore, it is necessary to consider the influence of deep water and tectonic activities on the salt-forming evolution stage of salt lakes when studying the salt-forming evolution stage of salt lakes and paleoclimate by using salt lake deposition.

Image identification technology has great significance for forestry production and forestry management. Highly similar object identification tasks, such as tree species with similar leaves, are extremely challenging. Simply using typical Convolutional Neural Networks (CNNs) or simply adding more convolutional layers still performs poorly in the above tasks. In this paper, we present a novel attention mechanism to enhance the CNN for identification of tree species with highly similar leaves. This paper presents a highly discriminative network, namely attention branch based convolutional neural networks (ABCNN), to better distinguish the differences between leaves features. Firstly, we proposed a novel structure, in which an attention branch is added in all block layers of network besides the typical normal branch. Secondly, our attention branch adopts a condensation process to obtain a region of interest (ROI) from global information of input and designs a reconstruction process to amplify the features difference to focus on the ROI. Thirdly, we design a fusion process, which carefully combines the attention branch with a normal branch to improve the network performance in the training process. The proposed ABCNN is tested on special dataset of Leafsnap with highly similar tree leaves. Our approach achieved 91.43% classification accuracy, which is higher than previous methods. Furthermore, ABCNN is also tested on general data set of SVHN and obtains 98.27% classification accuracy, which is the most competitive when considering the lower computational resources for ordinary applications. Both above experiments demonstrate the discrimination and robustness of the proposed method.

There are more than 1,000 lakes within the Tibetan Plateau (TP), all of which are sensitive to changes in regional climate and local hydrology. Lacustrine sediments within these lakes preserve a good record of these changes. However, determining their precise ages is difficult due to the complex nature of lake reservoir effects (LRE), which limit our understanding of paleoenvironmental changes. Focusing on an exposed 600 cm thick lacustrine sediment profile located in western Zhari Namco, we used a combination of both radiocarbon and optically stimulated luminescence (OSL) dating methods in order to evaluate the carbon reservoirs of bulk organic matter (BOM) and aquatic plant remnants (APR), and to explore the age differences between 14C and OSL and their respective reliability. We demonstrated that (i) OSL ages were changed in stratigraphic order, and the OSL age just below the beach gravel layer was consistent with previously reported paleoshoreline ages; (ii) 14C ages were divergent between BOM and grass leaves; (iii) 14C ages of BOM were older than 14C ages of APR; and (iv) all 14C ages were older than OSL ages. This could be attributed to changing LRE in the past, causing the 14C ages to appear unstable during the deposition period. Although the 14C ages of terrestrial plant remnants (TPR) were not affected by LRE, an analyzed twig nonetheless returned a 14C age older than its respective layer’s OSL age, suggesting it may have been preserved on land prior to transportation into the lake. Our study suggests that OSL ages are more reliable than 14C ages with respect to Zhari Namco lacustrine sediments. We recommend caution when interpreting paleoenvironmental changes based on lacustrine sediment 14C ages alone.

In recent years, the tree of life has expanded substantially. Despite this, many abundant yet uncultivated microbial groups remain to be explored.

Soil management practices are important parts of sustainable agriculture. Improving tillage practice is important for alleviating soil degradation and promoting sustainable grain production. A four year field experiment was conducted to examine the effects of deep tillage (DT), incorporated into the minimum tillage (MT), on soil physical properties and crop performance. The field experiments included continuous rotary tillage (RT), continuous DT, and intermittent DT every three years, every two years, and every other year, into RT. The results showed that the introduction of DT into continuous RT reduced the soil bulk density of the 20–30 cm soil layer by 5.6% and reduced nutrient stratification rates by 20–30%, which favored more uniformly distributed soil nutrients in the top soil layer. The root growth for treatments with DT in the deep soil layers (1–1.5 m) was significantly improved, which resulted in a higher soil water depletion. Under deficit irrigation scheduling, the improved root growth and soil water uptake in the deep soil layer improved crop growth and grain production. Overall, a 10.5% increase in yield and 18.3% increase in water productivity were observed when intermittent DT was introduced during the four years. The effects of DT could be maintained for two continuous years. Therefore, it was suggested that after two or three years of RT, DT should be applied to improve soil physical properties and ensure high grain production.

The different skeletal muscle fiber types exhibit distinctively different physiological and metabolic properties, and have been linked to both human metabolic diseases and meat quality traits in livestock. Circular RNAs (circRNAs) are a new class of endogenous RNA regulating gene expression, but regulatory mechanisms of skeletal muscle fibers involved in circRNAs remain poorly understood. Here, we constructed circRNA expression profiles of three fast-twitch biceps femoris (Bf) and three slow-twitch soleus (Sol) muscles in pigs using RNA-seq and identified 16,342 distinct circRNA candidates. Notably, 242 differentially expressed (DE) circRNAs between Bf and Sol muscles were identified, including 105 upregulated and 137 downregulated circRNAs, and are thus potential candidates for the regulation of skeletal muscle fiber conversion. Moreover, Gene Ontology (GO) and Kyoto Encyclopedia of Genes and Genomes (KEGG) pathway enrichment analysis of host genes of DE circRNAs revealed that host genes were mainly involved in skeletal muscle fiber-related GO terms (e.g., muscle contraction, contractile fiber part, and Z disk) and skeletal muscle fiber-related signaling pathways (e.g., AMPK and cGMP-PKG). We also constructed co-expression networks of DE circRNA-miRNA-mRNA using previously acquired high-throughput sequencing mRNA and miRNA data, from which 112 circRNA-miRNA and 95 miRNA-mRNA interactions were identified. Multiple circRNAs essentially serve as a sponge for miR-499-5p, which is preferentially expressed in slow-twitch muscle and reduces the severity of Duchenne muscular dystrophy (DMD). Taken together, a series of novel candidate circRNAs involved in the growth and development of porcine skeletal muscle was identified. Furthermore, they provide a comprehensive circRNA resource for further in-depth research on the regulatory mechanisms of circRNA in the formation of skeletal muscle fiber, and may provide insights into human skeletal muscle diseases.

TNM stage is not enough to accurately predict the prognosis of patients with non-small cell lung cancer (NSCLC). This study aimed to establish the immunological score (IS) in lung adenocarcinoma (LUAD) and lung squamous cell carcinoma (LUSC), separately, and propose a new staging system in NSCLC. We used the multiplex fluorescent immunohistochemistry (mIHC) technology to detect 17 immune biomarkers of 304 patients with NSCLC. The LASSO-COX regression model was used to establish the ISNSCLC in the training cohorts. The ISNSCLC was then validated in the validation cohort. The constructed ISLUAD contained three immune features: CD4+CD73+core of tumor (CT), PD-L1+CT, and IDO+invasive margin (IM). ISLUSC also contained two immune features: CD8+CD39−CD73−CT, CD8+Tim-3+IM. In the training cohort, significant prognostic differences were found upon comparing low-ISNSCLC patients with high-ISNSCLC patients. The high hygroscopicity of salt aerosol particles makes the particles active in aerosol and cloud formations. Inland saline lakes are an important and dynamic source of salt aerosol. The salt particles can be mixed with mineral dust and transported over long distances. During transportation, these particles participate in atmospheric heterogeneous chemistry and further impact the climate and air quality on a global scale. Despite their importance and potential, relatively little research has been done on saline lake salt mixtures from atmospheric perspectives. In this study, we use experimental and model methods to evaluate the hygroscopic properties of saline lake brines, fresh salt aerosol particles, and aged salt aerosol particles. Both original samples and literature data are investigated. The original brine samples are collected from six salt lakes in Shanxi and Qinghai provinces in China. The ionic compositions of the brines are determined and the hygroscopicity measurements are performed on crystallized brines. The experimental results agree well with theoretical deliquescence relative humidity (DRH) values estimated by a thermodynamic model. The correlations between DRHs of different salt components and the correlations between DRHs and ionic concentrations are presented and discussed. Positive matrix factorization (PMF) analysis is performed on the ionic concentrations data and the hygroscopicity results, and the solutions are interpreted and discussed. The fresh and aged salt aerosol particles are analyzed in the same way as the brines, and the comparison shows that the aged salt aerosol particles completely alter their hygroscopic property, i.e., transferring from MgCl2− governed to NH4NO3− governed.

Precision irrigation is a promising method to mitigate the impacts of drought stress on crop production with the optimal use of water resources. However, the reliable assessment of plant water status has not been adequately demonstrated, and unmanned aerial systems (UAS) offer great potential for spatiotemporal improvements. This study utilized UAS equipped with multispectral and thermal sensors to detect and quantify drought stress in winter wheat (Triticum aestivum L.) using the Water Deficit Index (WDI). Biennial field experiments were conducted on coarse sand soil in Denmark and analyses were performed at both diurnal and seasonal timescales. The WDI was significantly correlated with leaf stomatal conductance (R2 = 0.61–0.73), and the correlation was weaker with leaf water potential (R2 = 0.39–0.56) and topsoil water status (the highest R2 of 0.68). A semi-physical model depicting the relationship between WDI and fraction of transpirable soil water (FTSW) in the root zone was derived with R2 = 0.74. Moreover, WDI estimates were improved using an energy balance model with an iterative scheme to estimate the net radiation and land surface temperature, as well as the dual crop coefficient. The diurnal variation in WDI revealed a pattern of the ratio of actual to potential evapotranspiration, being higher in the morning, decreasing at noon hours and ‘recovering’ in the afternoon. Future work should investigate the temporal upscaling of evapotranspiration, which may be used to develop methods for site-specific irrigation recommendations.

Climate changes show asymmetrical warming, and warming is typically greater at night than during the day. To understand how nocturnal warming (NW) affects the performance of maize (Zea mays L.), an open-field experiment with a free air temperature increase (FATI) facility was conducted for three seasons during 2014 to 2016 at Luancheng eco-agro-experimental station on the North China Plain (NCP). Three nocturnal warming scenarios were set up: the entire growing period (T1, from V4 to maturity), only the vegetative stages (T2, from V4 to a week presilking) and the reproductive stages (T3, from a week presilking to R6). The treatment without NW was the control. Maize lodged seriously in 2015 due to heavy rainfall combined with strong winds, and the experiment failed. The results from 2014 and 2016 were analyzed in this study. During the experimental duration, the average nocturnal temperature was increased by approximately 3.6 and 3.3 °C at 150 cm height and 2.0 and 1.7 °C at the soil surface during the vegetative stages. The corresponding increases were 2.1 and 2.5 °C and 0.7 and 1.2 °C at the soil surface during the reproductive stages in 2014 and 2016, respectively, as compared with that of the CK treatment. NW during the whole growth period significantly decreased maize yield for the two seasons. Treatment T2 had a smaller impact on maize yield than T1 and T3. The silking stage was delayed by 2 days in 2014 and 2016 under T1. As a result, presilking duration and VT-R1 interval were prolonged by 1–2 days; and the postsilking duration were shortened by 1–3 days under T1. The soil moisture in the warmed plots was slightly lower than that in the control plots in the 2014 and during the stages before the earlier grain-filling stages in 2016, but NW decreased soil water content greatly at the later grain-filling stages in 2016, which caused the fast green leaf senescence and exacerbated the negative effects of NW on maize yield. NW for the whole growth duration (T1) significantly decreased seed weight and harvest index. NW increased leaf nighttime respiration rate in both seasons. No significant effects of NW on ear leaf net photosynthesis, leaf area, and specific leaf weight at early grain-filling stage were observed, irrespective of the warming stage and season. The results suggested that reproductive stages were more sensitive to NW compared to vegetative stages under the growing conditions of NCP. The negative effects of NW were worsened in dry seasons. The reduction in maize yield with nocturnal warming was driven by the reduction in the aboveground carbon allocation from shoot to grain during postanthesis stage.

Reducing irrigation water use by improving water use efficiency (WUE) in grain production is critical for the development of sustainable agriculture in the North China Plain (NCP). This article summarizes the research progresses in WUE improvement carried out at the Luancheng station located in the Northern part of NCP for the past three decades. Progresses in four aspects of yield and WUE improvement are presented, including yield and WUE improvement associated with cultivar selection, irrigation management for improving yield and WUE under limited water supply, managing root system for efficient soil water use and reducing soil evaporation by straw mulch. The results showed that annual average increase of 0.014 kg·m-3 for winter wheat and 0.02 kg·m-3 in WUE were observed for the past three decades, and this increase was largely associated with the improvement in harvest index related to cultivar renewal and an increase in chemical fertilizer use and soil fertility. The results also indicated that deficit irrigation for winter wheat could significantly reduce the irrigation water use, whereas the seasonal yield showed a smaller reduction rate and WUE was significantly improved. Straw mulching of summer maize using the straw from winter wheat could reduce seasonal soil evaporation by 30–40 mm. With new cultivars and improved management practices it was possible to further increase grain production without much increase in water use. Future strategies to further improve WUE are also discussed.

Prox1 is involved in muscle fiber conversion, adult-onset obesity, and type 2 diabetes. However, information regarding porcine Prox1 and its relationship with meat quality traits is still unknown. In this study, we characterized the full-length cDNA and proximal promoter of two transcript variants of porcine Prox1. Moreover, Prox1 was expressed abundantly in the skeletal muscle and its expression was higher in the soleus muscle than that in the biceps femoris muscle. Its expression pattern in the high and low meat color (redness) value a* groups was similar to that of myoglobin and MyHC I, but opposed to that of MyHC IIB. Importantly, there was a significant positive correlation between Prox1 expression and meat color (redness) value a* (r = 0.3845, p = 0.0394), and a significant negative correlation between Prox1 expression and drip loss (r = −0.4204, p = 0.0232), as well as the ratio of MyHC IIB to MyHC I expression (r = −0.3871, p = 0.0380). In addition, we found that the polymorphisms of three closely linked SNPs in Prox1 promoter 1 were significantly associated with pH24h in a pig population. Taken together, our data provide valuable insights into the characteristics of porcine Prox1 and indicate that Prox1 is a promising candidate gene affecting meat quality traits.

The major wheat production region of China the North China Plain (NCP) is seriously affected by air pollution. In this study, yield of winter wheat (Triticum aestivum L.) was analyzed with respect to the potential impact of air pollution index under conditions of optimal crop management in the NCP from 2001 to 2012. Results showed that air pollution was especially serious at the early phase of winter wheat growth significantly influencing various weather factors. However, no significant correlations were found between final grain yield and the weather factors during the early growth phase. In contrast, significant correlations were found between grain yield and total solar radiation gap, sunshine hour gap, diurnal temperature range and relative humidity during the late growing phase. To disentangle the confounding effects of various weather factors, and test the isolated effect of air pollution induced changes in incoming global solar radiation on yield under ceteris paribus conditions, crop model based scenario-analysis was conducted. The simulation results of the calibrated Agricultural Production Systems Simulator (APSIM) model indicated that a reduction in radiation by 10% might cause a yield reduction by more than 10%. Increasing incident radiation by 10% would lead to yield increases of (only) 7%, with the effects being much stronger during the late growing phase compared to the early growing phase. However, there is evidence that APSIM overestimates the effect of air pollution induced changes on radiation, as it does not consider the changes in radiative properties of solar insulation, i.e. the relative increase of diffuse over direct radiation, which may partly alleviate the negative effects of reduced total radiation by air pollution. Concluding, the present study could not detect a significantly negative effect of air pollution on wheat yields in the NCP.

Plant roots play a significant role in plant growth by exploiting soil resources via the uptake of water and nutrients. Root traits such as fine root diameter, specific root length, specific root area, root angle, and root length density are considered useful traits for improving plant productivity under drought conditions. Therefore, understanding interactions between roots and their surrounding soil environment is important, which can be improved through root phenotyping. With the advancement in technologies, many tools have been developed for root phenotyping. Canopy temperature depression (CTD) has been considered a good technique for field phenotyping of crops under drought and is used to estimate crop yield as well as root traits in relation to drought tolerance. Both laboratory and field-based methods for phenotyping root traits have been developed including soil sampling, mini-rhizotron, rhizotrons, thermography and non-soil techniques. Recently, a non-invasive approach of X-ray computed tomography (CT) has provided a break-through to study the root architecture in three dimensions (3-D). This review summarizes methods for root phenotyping. On the basis of this review, it can be concluded that root traits are useful characters to be included in future breeding programs and for selecting better cultivars to increase crop yield under water-limited environments.
