Abstract Background The prognosis of non-small cell lung cancer (NSCLC) is substantially affected by lymph node metastasis (LNM), but there are no noninvasive, inexpensive methods of relatively high accuracy available to predict LNM in NSCLC patients. Methods Clinical data on NSCLC patients were obtained from the Surveillance, Epidemiology, and End Results (SEER) database. Risk factors for LNM were recognized LASSO and multivariate logistic regression. Six predictive models were constructed with machine learning based on risk factors. The area under the receiver operating characteristic curve (AUC) was used to assess the performance of the model. Subgroup analysis with different T-stages was performed on an optimal model. A webpage LNM risk calculator for optimal model was built using the Shinyapps.io platform. Results We enrolled 64,012 NSCLC patients, of whom 26,611 (41.57%) had LNM. Using multivariate logistic regression, we finally identified 10 independent risk factors for LNM: age, sex, race, histology, primary site, grade, T stage, M stage, tumor size, and bone metastases. GLM is the optimal model among all six machine learning models in both the training and validation cohorts. Subgroup analyses revealed that GLM has good predictability for populations with different T staging. A webpage LNM risk calculator based on GLM was posted on the shinyapps.io platform ( https://wubopredict.shinyapps.io/dynnomapp/ ). Conclusion The predictive model based on GLM can be used to precisely predict the probability of LNM in NSCLC patients, which was proven effective in all subgroup analyses according to T staging.

ABSTRACTRefractories subject to loading and unloading repeatedly because of physical effects and thermal stress attack in the service process, which is an important issue shortening the service life of the thermal equipment. Therefore, researches of predicting the service life of refractories under cyclic loading have the guiding significance. The present work investigated the synthesis of ZnAl2O4 and discussed its effects on the alumina castables. The results show that the optimized generation temperature of ZnAl2O4 phase is 1400°C and the hysteresis loop in the curves of cyclic loading reveals a general trend of first sparse and then dense. The cycle times of the specimen with 3 wt.% ZnAl2O4 is 50 times, and the time from starting to loop to fracture is about 4300 s. The fatigue resistance of the specimen with 3 wt.% ZnAl2O4 is superior to ZA-0 and ZA-1.5. The new method of cyclic loading was innovatively adopted in refractories, which provide data and theoretical support for the thermal shock evaluation methods.

The mechanical strength of rice culm, an essential factor for lodging resistance and yield maintenance, is influenced by the composition and structure of the cell wall. In this study, we characterized a rice brittle culm mutant 22 (bc22), derived from LR005 through ethyl methanesulfonate (EMS) mutagenesis. The bc22 culm exhibited increased fragility and reduced mechanical strength compared to LR005. The mutant displayed pleiotropic effects, including a shorter plant height and panicle length, a smaller grain size, and the absence of the glume hairs. Scanning electron microscopy revealed a decrease in cell density and a looser structure in the bc22 culms. Biochemical analysis demonstrated a significant increase in hemicellulose content and a marked reduction in lignin content in the culm of bc22. Genetic analysis indicated that the brittle culm trait was governed by a single recessive gene. After employing bulked segregant analysis (BSA), whole-genome resequencing, and MutMap methods, LOC_Os02g25230 was identified as the candidate gene responsible for bc22. In bc22, a point mutation from proline (Pro) to leucine (Leu) in its coding region led to the pleiotropic phenotype. A complementation test further confirmed that the missense mutation causing the proline to leucine amino acid substitution in LOC_Os02g25230 was causative of the observed bc22 phenotype. Additionally, gene expression analysis showed that BC22 had higher expression levels in the culms, leaves, and spikelets compared to the roots. Taken together, our findings indicate that BC22 is a pleiotropic gene, and the influence of BC22 on brittleness may be associated with cell wall biosynthesis in rice culm.

ObjectiveTo analyze the prognostic impact of neoadjuvant chemoradiotherapy (NCRT) on early-stage (cT1b-cT2N0M0) esophageal cancer (ESCA) and construct a prognostic nomogram for these patients.MethodsWe extracted the clinical data about patients diagnosed with early-stage esophageal cancer from the 2004–2015 period of the Surveillance, Epidemiology, and End Results (SEER) database. We applied the independent risk factors affecting the prognosis of patients with early-stage esophageal cancer obtained after screening by univariate and multifactorial COX regression analyses to establish the nomogram and performed model calibration using bootstrapping resamples. The optimal cut-off point for continuous variables is determined by applying X-tile software. After balancing the confounding factors by propensity score matching (PSM) and inverse probability of treatment weighting (IPTW) method, Kaplan-Meier(K-M) curve, and log-rank test were applied to evaluate the prognostic impact of NCRT on early-stage ESCA patients.ResultsAmong patients who met the inclusion criteria, patients in the NCRT plus esophagectomy (ES) group had a poorer prognosis for overall survival (OS) and esophageal cancer-specific survival (ECSS) than patients in the ES alone group (p

Background: Esophageal carcinoma (ESCA), a common malignant tumor of the digestive tract with insidious onset, is a serious threat to human health. Despite multiple treatment modalities for patients with ESCA, the overall prognosis remains poor. Apolipoprotein C1 (APOC1) is involved in tumorigenesis as an inflammation-related molecule, and its role in esophageal cancer is still unknown.Methods: We downloaded documents and clinical data using The Cancer Genome Atlas (TCGA)and Gene Expression Omnibus (GEO) databases. We also conducted bioinformatics studies on the diagnostic value, prognostic value, and correlation between APOC1 and immune infiltrating cells in ESCA through STRING (https://cn.string-db.org/), the TISIDB (http://cis.hku.hk/TISIDB/) website, and various other analysis tools.Results: In patients with ESCA, APOC1 was significantly more highly expressed in tumor tissues than in normal tissues (p < 0.001). APOC1 could diagnose ESCA more accurately and determine the TNM stage and disease classification with high accuracy (area under the curve, AUC≥0.807). The results of the Kaplan–Meier curve analysis showed that APOC1 has prognostic value for esophageal squamous carcinoma (ESCC) (p = 0.043). Univariate analysis showed that high APOC1 expression in ESCC was significantly associated with worse overall survival (OS) (p = 0.043), and multivariate analysis shows that high APOC1 expression was an independent risk factor for the OS of patients with ESCC (p = 0.030). In addition, the GO (gene ontology)/KEGG (Kyoto encyclopedia of genes and genomes) analysis showed a concentration of gene enrichment in the regulation of T-cell activation, cornification, cytolysis, external side of the plasma membrane, MHC protein complex, MHC class II protein complex, serine-type peptidase activity, serine-type endopeptidase activity, Staphylococcus aureus infection, antigen processing and presentation, and graft-versus-host disease (all p < 0.001). GSEA (gene set enrichment analysis) showed that enrichment pathways such as immunoregulatory-interactions between a lymphoid and non-lymphoid cell (NES = 1.493, p. adj = 0.023, FDR = 0.017) and FCERI-mediated NF-KB activation (NES = 1.437, p. adj = 0.023, FDR = 0.017) were significantly enriched in APOC1-related phenotypes. In addition, APOC1 was significantly associated with tumor immune infiltrating cells and immune chemokines.Conclusion: APOC1 can be used as a prognostic biomarker for esophageal cancer. Furthermore, as a novel prognostic marker for patients with ESCC, it may have potential value for further investigation regarding the diagnosis and treatment of this group of patients.

The rheological behaviour of Al2O3-SiC-SiO2-C shotcreting castables with several types of additives as candidate flocculants or accelerators was investigated using the newly developed castable rheometer. The device can indicate the torque changes of the castables during mixing as a function of time and rotating speed, by which it can be understood whether the castables can coagulate and work as the materials for shotcreting as well as whether the flocculants can take effect. The results show that a CaCl2 solution and a Na2Al2O4 solution take a marked effect on flocculation taking place around 20 s, and the flocculation by the former is not sensitive to the added amount.
