Polyaluminum chloride modified clay (PAC-MC) is a safe and efficient red tide control agent that has been studied and applied worldwide. Although it is well known that the distribution of hydrolytic aluminum species in PAC affects its flocculation, little is known about the influence of particulars aluminum species on the microalgae removal efficiency of PAC-MC; this lack of knowledge creates a bottleneck in the development of more efficient MCs based on aluminum salts. The ferron method was used in this study to quantitatively analyze the distributions of and variations in different hydrolytic aluminum species during the process of microalgae removal by PAC-MC. The results showed that Ala, which made up 5%–20% of the total aluminum, and Alp, which made up 15%–55% of the total aluminum, significantly affected microalgae removal, with Pearson's correlation coefficients of 0.83 and 0.89, respectively. Most of the aluminum in the PAC-MC sank rapidly into the sediments, but the rate and velocity of settlement were affected by the dose of modified clay. The optimal dose of PAC-MC for precipitating microalgae was determined based on its aluminum profile. These results provide guidance for the precise application of PAC-MC in the control of harmful algal blooms.

Electrophysiology studies the electrical properties of cells and tissues including bioelectrical signals and membrane ion channel activities. As an important means to reveal ion channel related physiological functions and the underlying mechanisms, electrophysiological techniques have been widely used in studies of animals, higher plants and algae that are closely related to higher plants. However, few electrophysiological studies have been carried out in red tide organisms, especially in dinoflagellates, which is mainly due to the complex surface structure of dinoflagellate amphiesma. In this study, the surface amphiesma of Alexandrium pacificum, a typical red tide species, was removed by centrifugation, low-temperature treatment and enzymatic treatment. In all three treatments, low-temperature treatment with 4 °C for 2 h had high ecdysis rate and high fixation rate, and the treated cells were easy to puncture, so low-temperature treatment was used as a preprocessing treatment for subsequent current recording. The abalone industry has suffered immense economic losses due to the occurrence of harmful algal blooms (HABs). Among the methods for mitigating HABs, modified clay is considered the most promising strategy and has been successfully used for field applications in many countries, and its environmental effects have become a subject of global concern. The effects of modified clay on the survival, growth, nutritional quality, and oxidative stress indicators of abalone were studied based on both laboratory and field experiments. The results showed that modified clay at 3-10 times the concentrations used for HAB treatment did not affect the survival of abalone. During the laboratory experiments, the increases in abalone shell length and weight nonsignificantly decreased with increasing concentrations of modified clay at 1-15 d, whereas the weight of abalone in the experimental groups increased rapidly during the recovery period at 16-30 d. The growth and nutrition qualities of abalone in field experiments showed negligible differences between the control and experimental groups. Catalase (CAT) activity in the hepatopancreas and superoxide dismutase (SOD) activity in the gills were significantly affected by certain concentrations of modified clay at individual time points, whereas the malondialdehyde (MDA) content decreased in all experimental groups within 96 h. The removal of bacteria and the mitigation of water quality decline were among the effects of modified clay that contributed to the decrease in MDA content. The present study showed that modified clay had no obvious adverse effects on the survival, growth, quality, or oxidative stress indicators of abalone at the experimental concentrations, thus providing a reference for the field application of modified clay in typical aquaculture areas. Phaeocystis globosa is a harmful algal bloom (HAB) species worldwide. Using modified clay (MC) to control HABs and to mitigate their adverse effects is currently a commonly used method in China. In this paper, the effects of oxidized composite modified clay (OXI-MC) on P. globosa were studied from different perspectives. The results show that the OXI-MC could effectively remove P. globosa and inhibit both the growth of residual algal cells and the formation of new colonies. The P. globosa algal biomass removal efficiencies after 3 h reached 90% at a dose of 0.1 g/L, and the number of colonies with different particle sizes was greatly reduced. Compared with those of the control, the superoxide dismutase (SOD) activity, catalase (CAT) activity, and malondialdehyde (MDA) content of the residual algae significantly increased, indicating that OXI-MC caused oxidative stress in the algal cells. In addition, we evaluated the effects of OXI-MC on the photosynthesis of residual microalgae and found that the maximal photochemical efficiency of photosystem II (PSII) under dark adaptation (Fv/Fm) and actual photochemical efficiency of PSII (ΦPSII) decreased, severely damaging the photosynthesis efficiency, implying that OXI-MC effected the photosynthesis system of P. globosa. The results of this study clarify that OXI-MC could remove the most of algal cells and break up the colonies of P. globosa by collision, flocculation, and releasing active substances, as well as inhibit effectively the growth and colony formation of residual P. globosa by causing oxidative stress, reducing photosynthesis activity, accelerating the degradation of polysaccharides, and inhibiting the formation of colonies.

Microscopic propagules of Ulva species (UMPs) spread on Subei Shoal are believed to play a significant role in the formation of the Yellow Sea green tide. Previous laboratory and ship-based studies indicated that modified clay (MC) can effectively remove UMPs from the water column and restrict their germination. To evaluate the effectiveness of this method under natural conditions, a mesocosm experiment was conducted on the Zhugensha sandbank on Subei Shoal. The results show that the suspended particles on the sandbank would partially disturb the flocculation process between the MC and UMPs. However, the MC at a proper dose could effectively remove UMPs from the water column and decrease the adhesion and germination of UMPs on the nylon ropes that comprised the mariculture rafts by > 75%. This method was proven a potential strategy to restrict the initial biomass accumulation of green algae on Subei Shoal. However, field trials in larger time-space scales are still needed to assess the efficiency of this method when used in the open sea.

The composition and stability of microbial communities in aquaculture water are crucial for the healthy growth of shrimp and present considerable risk to aquatic ecosystems. The modified clay (MC) method has been proposed as an efficient and safe solution for the mitigation of harmful algal blooms (HABs). Currently, the effects of MC on microbial communities in aquaculture water remain unknown. Here, we adopted the MC method to regulate shrimp-culture water quality and evaluated the effects of MC on the composition and stability of phytoplankton together with bacteria communities through high-throughput sequencing. On the one hand, a prominent change in the composition of microbial community was observed, with green algae becoming the most abundant genera and pathogens being infrequent in the MC-treated pond, which was more conducive to the growth of shrimp than that in the control pond. Moreover, MC could increase the diversity and stability of the microbial community structure in the water column, which had a higher anti-interference ability, as demonstrated by the analysis of the diversity and molecular ecological network. Taken together, MC could reduce the possibility for the occurrence of HABs and maintain a stable microbial community, which is beneficial for the health and high yield of shrimp.

On the basis of field experience, a bloom does not continue after treatment with modified clay (MC), even though the residual harmful algal bloom (HAB) biomass accounts for 20–30% of the initial cells. This interesting phenomenon indicates that, in addition to causing flocculation, MC can inhibit the growth of residual cells. Here, from a cell morphology perspective, Aureococcus anophagefferens was used as a model organism to explore this scientific issue and clarify the mechanism by which MC mitigates harmful algal blooms (HABs). The results showed that, at an ~70% removal efficiency, neutral clay (NC) could not effectively inhibit the growth of residual cells, although it caused various forms of damage to residual cells, such as cell deformation, cell breakage, decreased extracellular polysaccharides (EPS), increased cell membrane permeability, and increased cytoplasmic granularity, due to physical collisions. After modification, some physical and chemical properties of the clay particle surface were changed, for example, the surface electrical properties changed from negative to positive, lamellar spacing increased, hardness decreased, adhesion chains increased, adhesion improved, and the number of absorption sites increased, enhancing the occurrence of chemical and electrochemical effects and physical collisions with residual cells, leading to severe cell deformation and chemical cell breakage. Thus, MC effectively inhibited the growth of residual cells and controlled HABs.

Phaeocystis globosa is a globally distributed harmful algal blooms (HABs) species dominated by the colonial morphotype, which presents dramatic environmental hazards and poses a threat to human health. Modified clay (MC) can effectively flocculate HAB organisms and prevent their subsequent growth, but the effects of MC on colony-dominated P. globosa blooms remain uncertain. In this paper, a series of removal and incubation experiments were conducted to investigate the growth, colony formation and colony development of P. globosa cells after treatment with MC. The results show that the density of colonies was higher at MC concentrations below 0.2 g/L compared to those in the control, indicating the role of P. globosa colonies in resistance to environmental stress. Concentrations of MC greater than 0.2 g/L could reduce the density of solitary cells and colonies, and the colony diameter and extracellular polysaccharide (EPS) content were also decreased. The adsorption of MC to dissolved inorganic phosphorus (DIP) and the cell damage caused by collision may be the main mechanisms underlying this phenomenon. These results elucidate that the treatment with an appropriate concentration of MC may provide an effective mitigation strategy for P. globosa blooms by preventing their growth and colony formation.

The harmful algal bloom (HAB) species Phaeocystis globosa is commonly observed in global temperate and tropical oceans, and colonies of P. globosa exhibit a dominant morphotype during blooms. The use of polyaluminium chloride modified clay (PAC-MC) is an effective mitigation strategy for P. globosa blooms. Although previous studies have found that PAC-MC can stimulate P. globosa colony formation at low concentrations and inhibit it at higher concentrations, the underlying mechanisms of these effects are poorly understood. Here, we comprehensively compared the physiochemical indices and transcriptomic response of residual P. globosa cells after treatment with two concentrations of PAC-MC. The results showed that PAC-MC induced oxidative stress, photosynthetic inhibition, and DNA damage in residual cells. Moreover, it could activate antioxidant responses and enhance the repair of photosynthetic structure and DNA damage in cells. The biosynthesis of polysaccharides was enhanced and genes associated with cell motility were down-regulated after treatment with PAC-MC, resulting in the accumulation of colonial matrixes. After treatment with a low concentration of PAC-MC (0.1 g/L), the residual cells were slightly stressed, including physical damage, oxidative stress and other damage, and polysaccharide synthesis was enhanced to promote colony formation to alleviate environmental stress. Moreover, the damage to residual cells was slight; thus, normal cell function provided abundant energy and matter for colony formation. After treatment with a high concentration of PAC-MC (0.5 g/L), the residual cells suffered severe damage, which disrupted normal physiological processes and inhibited cell proliferation and colony formation. The present study elucidated the concentration-dependent mechanism of PAC-MC affecting the formation of P. globosa colonies and provided a reference for the application of PAC-MC to control P. globosa blooms.

Vibrio alginolyticus is a globally distributed opportunistic pathogen that causes different degrees of disease in various marine organisms, such as fish, shrimp and shellfish. At present, vibriosis caused by V. alginolyticus has a wide epidemic range and causes frequent outbreaks, resulting in substantial losses in aquaculture. According to previous studies, modified clay (MC) could effectively flocculate and reduce the density of Vibrio in water, but it is still unknown whether MC inhibits growth and how it affects virulence in bottom flocs. Here, we studied the response mechanism of V. alginolyticus in flocs treated with MC at the transcriptome level and verified the transcriptomic data combined with relevant physiological experiments and reverse transcription quantitative real-time PCR (RT-qPCR) for the first time. It was found that the morphology of Vibrio in the MC flocs changed, the membrane function was damaged, the antioxidant system was activated, and the material and energy metabolism also changed. In addition, MC could inhibit the expression of virulence factors of V. alginolyticus; for example, flagella, pilus, siderophores, quorum sensing, and other related genes were significantly downregulated. In general, MC effectively inhibited the growth of Vibrio and reduced its virulence potential in flocs, which could provide theoretical support for a new model of healthy aquaculture.

In this study, Karenia brevis 165 (K. brevis 165), a Chinese strain, was used to research brevetoxin (BTX) metabolites. The sample pretreatment method for the enrichment of BTX metabolites in an algal culture medium was improved here. The method for screening and identifying intracellular and extracellular BTX metabolites was established based on liquid chromatography-time-of-flight mass spectrometry (LC-ToF-MS) and liquid chromatography triple quadrupole tandem mass spectrometry (LC-QqQ-MS/MS). The results show that the recovery rates for BTX toxins enriched by a hydrophilic–lipophilic balance (HLB) extraction column were higher than those with a C18 extraction column. This method was used to analyze the profiles of extracellular and intracellular BTX metabolites at different growth stages of K. brevis 165. This is the first time a Chinese strain of K. brevis has been reported that can produce toxic BTX metabolites. Five and eight kinds of BTX toxin metabolites were detected in the cell and culture media of K. brevis 165, respectively. Brevenal, a toxic BTX metabolite antagonist, was found for the first time in the culture media. The toxic BTX metabolites and brevenal in the K. brevis 165 cell and culture media were found to be fully proven in terms of the necessity of establishing a method for screening and identifying toxic BTX metabolites. The results found by qualitatively and quantitatively analyzing BTX metabolites produced by K. brevis 165 at different growth stages show that the total toxic BTX metabolite contents in single cells ranged between 6.78 and 21.53 pg/cell, and the total toxin concentration in culture media ranged between 10.27 and 449.11 μg/L. There were significant differences in the types and contents of toxic BTX metabolites with varying growth stages. Therefore, when harmful algal blooms occur, the accurate determination of BTX metabolite types and concentrations will be helpful to assess the ecological disaster risk in order to avoid hazards and provide appropriate disaster warnings.

For many years, the dispersal of modified clay (MC) has been used to control harmful algal blooms (HABs) in coastal waters of China. MC flocculation efficiency can be influenced by many factors in variable and complex natural environments, including high concentrations of dissolved organic matter (DOM) in the water to be treated. Since many HABs occur in nearshore waters where DOM concentrations are high, this is a significant problem that requires urgent attention. This study involved the use of humic acid as a representative form of DOM to study the influence of organic matter on the MC flocculation process. At high concentrations, humic acid was adsorbed onto MC particles, resulting in a decrease in surface potential and an increase in electrostatic repulsion between the clay particles; this decreased the MC settling rate and increased the water clarification time. Flocs were characterized by their relatively small particle size, high particle concentration, and low collision efficiency, which together resulted in slow clarification of the water after MC spraying. Based on the mechanism of the DOM-MC interaction and combined with the Derjaguin-Landau-Verwey-Overbeek theory and theoretical considerations of clay surface modification, the “ionic atmosphere compression” method was used to improve MC flocculation efficiency in high-organic water. This method increased the ionic strength of the clay stock solution by adding salt, thereby compressing the ionic atmosphere of MC particles and lowering the potential barrier, allowing the MC particles in the treated water to flocculate rapidly and form large flocs, followed by further floc growth and rapid settling via differential sedimentation. The settling rate of MCs improved by a factor of two and the removal efficiency of the HAB cells increased by 7–28%. This study provides important baseline information that will extend the application of MC to HAB control in water bodies with high organic loadings.

The cosmopolitan alga Phaeocystis globosa forms massive colonies during blooms. In addition to producing haemolytic toxins that affect the ecosystem, centimetre-sized colonies can block nuclear power plant cooling systems. Nitrogen plays a key role in P. globosa blooms; however, the preferred form of inorganic nitrogen taken up by colonies remains unknown. Therefore, we set up four groups with different nitrogen types and levels of enrichment. The culture medium and intracolonial fluid were sampled at different times for the determination of nutrient concentrations to elucidate the nutrient uptake strategy that presumably favours colonies. Although $${{\text{NH}}_{4}}^{ + } {\text{ {-} N}}$$ and $${{\text{NO}}_{2}}^{ - } {\text{{-}N}}$$ were absorbed by the colonies, $${{\text{NO}}_{3}}^{ - } {\text{{-}N}}$$ was the favoured nitrogen, and the average uptake rate of $${{\text{NO}}_{3}}^{ - } {\text{{-}N}}$$ was significantly higher than that of $${{\text{NH}}_{4}}^{ + } {\text{{-}N}}$$ and $${{\text{NO}}_{2}}^{ - } {\text{{-}N}}$$ in each group (P

Using modified clay is one of the most promising methods for the mitigation of harmful algal blooms (HABs). However, the environmental impact of modified clay has become a subject of global concern. In this study, turbot (Scophthalmus maximus L.) embryos were used as a model to assess the effect of modified clay on this sensitive stage of fish development. Results show that the 24 and 48h LC50 (median lethal concentrations) of the modified clay were 1.70 and 1.65 g/L, and the safe concentration was 0.47 g/L, which is much higher than the concentration used to treat HAB. The modified clay did not affect significantly the hatchability of turbot embryos but when the concentration exceeded 0.50 g/L, the deformity rate of newly hatched larvae increased significantly. The total length, specific growth rate (SGR) and yolk sac absorption rate of larvae reached their peaks at 0.50 g/L and then gradually decreased as the concentration of modified clay increased. Therefore, a moderate amount of modified clay does not harm the survival and hatching of turbot embryos, or the growth and development of newly hatched larvae.

Shellfish aquaculture is an important marine industry that has been threatened by harmful algal blooms (HABs) and, in recent years, toxic HABs in particular. The modified clay (MC) treatment is the most promising method to mitigate HABs and has been frequently applied in the field. To determine how the application of MC affects the accumulation, transformation, and detoxification of paralytic shellfish toxins (PSTs) in the bay scallop Argopecten irradians and surrounding sediments, a series of experiments over 16 days in a laboratory simulated Alexandrium tamarense bloom were conducted. After 3 h of exposure to A. tamarense at 2070 (±120) cells mL−1, the toxicity level in the tissues of the scallops increased to 105.9 (±16.6) μg STX eq. 100 g−1 in the control group (group A, with no addition of MC), while the toxicity levels in the scallops of groups B and C (0.1 g L−1 and 0.5 g L−1 MC added, respectively) remained at relatively low levels. During the 3 h, the scallops in group A had incorporated 73.2 mol% of the toxin contained in the initial A. tamarense algal culture, the scallops in group B had incorporated 13.5 mol% of the toxin, and the scallops in group C showed almost no elevation in tissue toxin content. Meanwhile, 56.8 mol% and 107.1 mol% of the PSTs contained in the initial algal culture were settled in the sediments along with the MC-algal cells combination in group B and C, respectively. The results indicated that the application of MC can effectively prevent filter-feeding bivalves from ingesting toxic algal cells and decrease the accumulation of toxins by settling the toxic algal cells in the sediment. It was also indicated that the toxins accumulated within the sediments were more rapidly detoxified than that in the scallop tissues. Toxins accumulated by the scallops persisted in the tissue for approximately 16 days, while in the sediments, all of the PSTs contents were below the HPLC detection limits 4 days after the addition of MC.

Increasing numbers of approaches to assess eutrophication, such as estuarine trophic status, the Oslo-Paris Commission Common Procedure, and the Water Framework Directive, focus on the symptoms of eutrophication. In China, however, nutrient index methods dominate the assessment of coastal waters. In this study, an integrated method that includes both water quality and ecological response was compared with the Northwest Pacific Action Plan (NOWPAP) Common Procedure. Observation data from Jiaozhou Bay, Shandong, China, were used in a comparison of the two methods in a trophic status study. Overall, both clearly revealed a high level of nutrient enrichment in the bay, indicated by high nutrient concentrations. Though the two methods differed in their methodological design in the assessment of the ecological effects of nutrient enrichment, they have acquired similar results: the integrated method suggested that the status was good, and the NOWPAP Common Procedure suggested that the status was low (indicating that the bay had no serious eutrophication problem). The introduction of filter feeders (shellfish aquaculture) into the bay on a reasonable scale may have been effective in mitigating eutrophic conditions, and perhaps explains the low ecological impacts there. Our results will be useful to ecosystem-based eutrophication management in the bay and in similar areas.

In order to depict the distribution of diatom fossils in surface sediments and to establish a reliable reference data for further paleoenvironmental study in the Changjiang (Yangtze) River estuary and its adjacent waters, the diatom fossils from 34 surface sediment samples and their relationship with environmental variables were analyzed by principal component analysis and redundancy correspondence analysis. The diversity and abundance of diatom fossils were analyzed. Some annual average parameters of the overlying water (salinity, temperature, turbidity, dissolved oxygen, depth, dissolved inorganic nitrogen, dissolved inorganic phosphate and dissolved inorganic silicate) were measured at each sampling site. A total of 113 diatom taxa and one silicoflagellate species were identified in the investigation area. Diatom fossils were better preserved in fine sediments. The absolute abundance of diatom fossils did not significantly differ between inshore and offshore areas, the species diversity decreased from inshore to offshore. This may be because high nutrients and low salinity promoted the growth of more brackish species in coastal waters. The diatom taxa were divided into three groups, on the basis of their response and indication to environmental changes. For example, Actinocyclus ehrenbergii and Cyclotella stylorum were dominant in coastal waters (Group 1 and Group 3) with high nutrients and low salinity; the relative abundances of Paralia sulcata and Podosira stelliger were significantly higher in offshore sites (Group 2, average 39.5%), which were characterized by high salinity and deep water. Four environmental variables (salinity, dissolved inorganic nitrogen, temperature and water depth) explained the composition and distribution of diatom taxa independently ( P< 0.05), this finding can be applied in further paleoenvironmental reconstruction research in this area.

The application of modified clay (MC) to mitigate harmful algal blooms (HABs) is becoming a widespread practice in China because of its low cost, high efficiency and environmental friendliness. Due to its success in China, this technology has also gained international recognition in recent years. Nevertheless, the eco-environmental impacts of this technology still remain to be a concern to many customers and government agencies, which has motivated scientists in both China and many other countries to evaluate its potential effects on nontarget aquatic organisms, water quality and the benthic environment. This paper reviews the results from these studies both in the laboratory and in the field, which showed that MC does not harm nontarget aquatic organisms and has no negative impact on water quality or the benthic environment. Additionally, findings show that MC can alter nutrient cycling and reduce algal toxins in water bodies. Furthermore, researchers also found that MC affects cyst formation and germination in sedimentary environments. This review is expected to provide scientific guidance for mitigating HABs in China and worldwide using clay or MC.

Modified clay (MC), an effective material used for the emergency elimination of algal blooms, can rapidly reduce the biomass of harmful algal blooms (HABs) via flocculation. After that, MC can still control bloom population through indirect effects such as oxidative stress, which was initially proposed to be related to programmed cell death (PCD) at molecular level. To further study the MC induced cell death in residual bloom organisms, especially identifying PCD process, we studied the physiological state of the residual Prorocentrum donghaiense. The experimental results showed that flocculation changed the physiological state of the residual cells, as evidenced by growth inhibition and increased reactive oxygen species production. Moreover, this research provides biochemical and ultrastructural evidence showing that MC induces PCD in P. donghaiense. Nuclear changes were observed, and increased caspase-like activity, externalization of phosphatidylserine and DNA fragmentation were detected in MC-treated groups and quantified. And the mitochondrial apoptosis pathway was activated in both MC-treated groups. Besides, the features of MC-induced PCD in a unicellular organism were summarized and its concentration dependent manner was proved. All our preliminary results elucidate the mechanism through which MC can further control HABs by inducing PCD and suggest a promising application of PCD in bloom control.

Hypoxia off the Changjiang Estuary (CE) and its adjacent waters is purported to be the most severe in China, attracting considerable concern from both the scientific community and the general public. Currently, continuous observations of dissolved oxygen (DO) levels covering hypoxia from its appearance to disappearance are lacking. In this study, twelve consecutive monthly cruises (from February 2015 to January 2016) were conducted. The consecutive spatiotemporal variations in hypoxia throughout the annual cycle were elucidated in detail, and the responses of annual variations in hypoxia to the different influential factors were explored. Overall, hypoxia experienced a consecutive process of expanding from south to north, then disappearing from north to south. The annual variations in hypoxia were mainly contingent on stratification variations. Among different stages, there was significant heterogeneity in the dominant factors. Specifically, low-DO waters initially appeared from the intrusion of nearshore Kuroshio branch current (NKBC), as NKBC intrusion provided a low-DO background and triggered stratification. Thereafter, stratification was enhanced and gradually expanded northward, which promoted the extension of low-DO areas. The formation of hypoxia was regionally selective, and more intense organic matter decomposition at local regions facilitated the occurrence and discontinuous distribution of hypoxia. Hypoxic zones were observed at the Changjiang bank and Zhejiang coastal region from August (most extensively at 14,800 km2) to October. Thereafter, increased vertical mixing facilitated the dissipation of hypoxia from north to south.

The marine iodine cycle has significant impacts on air quality and atmospheric chemistry. Specifically, the reaction of iodide with ozone in the top few micrometres of the surface ocean is an important sink for tropospheric ozone (a pollutant gas) and the dominant source of reactive iodine to the atmosphere.
