Abstract Background S100β is a biomarker of astroglial damage, the level of which is significantly increased following brain injury. However, the characteristics of S100β and its association with prognosis in patients with acute ischemic stroke following intravenous thrombolysis (IVT) remain unclear. Methods Patients in this multicenter prospective cohort study were prospectively and consecutively recruited from 16 centers. Serum S100β levels were measured 24 h after IVT. National Institutes of Health Stroke Scale (NIHSS) and hemorrhagic transformation (HT) were measured simultaneously. NIHSS at 7 days after stroke, final infarct volume, and modified Rankin Scale (mRS) scores at 90 days were also collected. An mRS score ≥ 2 at 90 days was defined as an unfavorable outcome. Results A total of 1072 patients were included in the analysis. The highest S100β levels (> 0.20 ng/mL) correlated independently with HT and higher NIHSS at 24 h, higher NIHSS at 7 days, larger final infarct volume, and unfavorable outcome at 3 months. The patients were divided into two groups based on dominant and non-dominant stroke hemispheres. ObjectivesRe-epithelialization is an important physiological process for repairing skin barrier function during wound healing. It is primarily mediated by coordinated migration, proliferation, and differentiation of keratinocytes. Long noncoding RNAs (lncRNAs) are essential components of the noncoding genome and participate in various biological processes; however, their expression profiles and function in re-epithelialization during wound healing have not been established.MethodsWe investigated the distribution of lncRNAs during wound re-epithelialization by comparing the genomic profiles of uninjured skin and acute wound (AW) from healthy donors. We performed functional screening of differentially expressed lncRNAs to identify the important lncRNAs for re-epithelialization.ResultsThe expression of multiple lncRNAs is changed during human wound re-epithelialization process. We identified VIM-AS1, SMAD5-AS1, and LINC02581 as critical regulators involved in keratinocyte migration, proliferation, and differentiation, respectively.ConclusionLncRNAs play crucial regulatory roles in wound re-epithelialization. We established lncRNA expression profile in human acute wounds compared with intact skin, offering valuable insights into the physiological mechanisms underlying wound healing and potential therapeutic targets.

IntroductionThe photosynthetic electron transport chain (ETC) is the bridge that links energy harvesting during the photophysical reactions at one end and energy consumption during the biochemical reactions at the other. Its functioning is thus fundamental for the proper balance between energy supply and demand in photosynthesis. Currently, there is a lack of understanding regarding how the structural properties of the ETC are affected by nutrient availability and plant developmental stages, which is a major roadblock to comprehensive modeling of photosynthesis.MethodsRedox parameters reflect the structural controls of ETC on the photochemical reactions and electron transport. We conducted joint measurements of chlorophyll fluorescence (ChlF) and gas exchange under systematically varying environmental conditions and growth stages of maize and sampled foliar nutrient contents. We utilized the recently developed steady-state photochemical model to infer redox parameters of electron transport from these measurements.Results and discussionWe found that the inferred values of these photochemical redox parameters varied with leaf macronutrient content. These variations may be caused either directly by these nutrients being components of protein complexes on the ETC or indirectly by their impacts on the structural integrity of the thylakoid and feedback from the biochemical reactions. Also, the redox parameters varied with plant morphology and developmental stage, reflecting seasonal changes in the structural properties of the ETC. Our findings will facilitate the parameterization and simulation of complete models of photosynthesis.

Sepsis remains a leading cause of death for humans and currently has no pathogenesis-specific therapy. Hampered progress is partly due to a lack of insight into deep mechanistic processes. In the past decade, deciphering the functions of small noncoding miRNAs in sepsis pathogenesis became a dynamic research topic. To screen for new miRNA targets for sepsis therapeutics, we used samples for miRNA array analysis of PBMCs from patients with sepsis and control individuals, blood samples from 2 cohorts of patients with sepsis, and multiple animal models: mouse cecum ligation puncture–induced (CLP-induced) sepsis, mouse viral miRNA challenge, and baboon Gram+ and Gram– sepsis models. miR-93-5p met the criteria for a therapeutic target, as it was overexpressed in baboons that died early after induction of sepsis, was downregulated in patients who survived after sepsis, and correlated with negative clinical prognosticators for sepsis. Therapeutically, inhibition of miR-93-5p prolonged the overall survival of mice with CLP-induced sepsis, with a stronger effect in older mice. Mechanistically, anti–miR-93-5p therapy reduced inflammatory monocytes and increased circulating effector memory T cells, especially the CD4+ subset. AGO2 IP in miR-93–KO T cells identified important regulatory receptors, such as CD28, as direct miR-93-5p target genes. In conclusion, miR-93-5p is a potential therapeutic target in sepsis through the regulation of both innate and adaptive immunity, with possibly a greater benefit for elderly patients than for young patients.

Perinatal autopsies are essential to establish the cause of stillbirth or neonatal death and improve clinical practice. Limited studies have provided detailed major missed diagnoses of perinatal deaths in current clinical practice. In this retrospective audit of 177 perinatal autopsies including 99 stillbirths and 78 neonatal deaths with complete pathologic evaluation, 66 cases (21 Class I and 45 Class II diagnostic errors) were revealed as major discrepancies (37.3%), with complete agreements in 80 cases (45.2%). IntroductionTo document the successional processes of shrub-herb communities after large-scale human disturbance, and understand how changing environmental conditions affect species replacement in semi-humid hilly areas.MethodsUtilizing the established permanent plots in the hilly area of Taihang Mountain, we evaluated temporal patterns of vegetation and soil following grass-to-shrub succession.Results and DiscussionAlong secondary succession, Vitex negundo var. heterophylla gradually dominated in dry sunny slope and shared the dominance with Leptodermis oblonga in shaded slope. Herbaceous dominant species in shrub-herb communities switched from Themeda japonica, Bothriochloa ischaemum, Artemisia sacrorum, and Cleistogenes chinensis in 1986 census to B. ischaemum and A. sacrorum in 2008 census, but herb was no longer dominant in 2020 census. As succession progresses, species dominance increased while richness decreased generally, and herb cover and aboveground biomass decreased, whereas shrub height, cover, and aboveground biomass increased significantly. Soil organic matter (SOM), total nitrogen (TN), total phosphorus (TP), and total potassium (TK) in topsoil increased significantly while pH declined by 1.04 units over the past three decades. Plant communities transitioned from perennial herbs to shrub-herb and then shrub communities, and V. negundo var. heterophylla dominated in the succession of shrub-herb communities. Climate and soil properties, combined with plant attributes, together drive post-disturbance secondary succession. From a management perspective, the tight coupling between vegetation and soil under local climatic conditions should be considered to improve the fragile ecosystem in the hilly area of Taihang Mountain.

Background: Tuberculosis (TB) continues to be a common disease in developing countries, among which middle ear TB is rare. Furthermore, it is relatively difficult to make an early diagnosis and provide follow-up treatment for middle ear TB. So, it is necessary to report this case for reference and further discussion. Case presentation: We reported 1 case of multidrug-resistant tuberculosis otitis media. TB otitis media is rare in tuberculosis; multidrug-resistant TB otitis media is even more rare. Our paper analyzes the possible causes, imaging, molecular biology, pathology, and clinical manifestations of multidrug-resistant TB otitis media. Conclusion: PCR and DNA molecular biology techniques are highly recommended for the early diagnosis of multidrug-resistant TB otitis media. Early, effective anti-tuberculosis treatment is the guarantee for further recovery for patients with multidrug-resistant TB otitis media.

Microbial residue nitrogen can indicate soil quality and is crucial for soil nitrogen retention and supply. However, it is still unclear how the dynamic changes in soil microbial residue nitrogen affect crop nitrogen uptake in agricultural practice. Based on a long-term wheat-maize rotation experiment with different nitrogen application rates (150 kg N ha−1, 200 kg N ha−1, and 300 kg N ha−1), 15N-labeled nitrogen fertilizer was applied during the wheat season to track the dynamics of soil microbial residue nitrogen and its mediated fertilizer nitrogen. The results showed that nitrogen addition was beneficial to the accumulation of microbial residue nitrogen (mainly fungal microbial residue nitrogen). Its contribution rate to soil total nitrogen is 54.87–56.55%, and the fertilizer nitrogen allocated to it accounts for 27.10–47.50% of the remaining fertilizer nitrogen in the soil. Ultimately, 6.77–10.24% of the nitrogen fertilizer applied remained in the soil as microbial residue nitrogen. In addition, microbial residue nitrogen is mainly mineralized during the jointing and filling stages of wheat. In a word, the accumulation and mineralization of soil microbial residue nitrogen during the application of 200 kg N ha−1 better guaranteed the uptake of nitrogen by wheat, which provided a reliable basis for guiding farmland fertility improvement and nitrogen fertilizer reduction in the North China Plain.

IntroductionUnderstanding microbial gradual shifts along species replacement can help elucidate the mechanisms driving secondary succession, and predict microbial responses to changing environments. However, how climate-induced species replacement alters microbial processes, and whether microbial shifts follow predictable assembly trajectories remain unclear.MethodsUsing space-for-time substitution approach, we studied shifts in bacterial and fungal communities in the succession from Leptodermis oblonga to Vitex negundo var. heterophylla shrubland in Taihang Mountain.Results and DiscussionSpecies replacement, induced by climate related environmental change, significantly increased the above-ground biomass of shrublands, and TP and TK contents in topsoil. The succession from L. oblonga to V. negundo var. heterophylla communities resulted in the gradually replacement of cold-tolerant microbes with warm-affinity ones, and alterations of microbial communities involved in soil biogeochemical processes. Soil and plant variables, such as above-ground biomass, soil pH, total phosphorus, and total potassium, well explained the variations in microbial communities, indicating that the coordinated changes in plant communities and soil properties during secondary succession caused accompanied shifts in microbial diversity and composition.

Abstract Background Alteration of DNA methylation is an important event in pathogenesis and progression of hepatocellular carcinoma (HCC). DNA methyltransferase (DNMT) 1, the foremost contributor in DNA methylation machinery, was revealed elevated in HCC and significantly correlates with poor prognosis. However, the transcriptional regulation of DNMT1 in HCC remains unknown. Methods Real‐time PCR and immunohistochemistry were performed to detect DNMT1 and zinc finger transcription factor 191 (ZNF191) expressions in HCCs. Transcription activity of DNMT1promoter was analyzed with Luciferase reporter activity assay. The binding capacity of ZNF191 protein to DNMT1 promoter was examined with chromatin immunoprecipitation‐qPCR (ChIP‐qPCR) and electrophoretic mobility shift assay (EMSA). DNA methylation level of hepatoma cells was detected with Methylation array. Results ZNF191 can regulate DNMT1 mRNA and protein expression positively, and increase the transcription activity of the DNMT1 promoter. ChIP‐qPCR and EMSA revealed that ZNF191 protein directly binds to the DNMT1 promoter at nt‐240 AT(TCAT)3TC. Moreover, DNMT1 and ZNF191 expression correlate positively in human HCCs. With methylation array, DNA methylation alteration was observed in hepatoma cells with ZNF191 knockdown, and the differential methylation sites are enriched in the PI3K‐AKT pathway. Furthermore, we proved DNMT1 contributes the effect of ZNF191 on hepatoma cell growth via the PI3K‐AKT pathway. Conclusion ZNF191 is a novel transcription regulator for DNMT1, and the pro‐proliferation effect of ZNF191/DNMT1/p‐AKT axis in hepatoma cells implies that ZNF191 status in HCCs may affect the therapeutic effect of DNMTs inhibitors and PI3K inhibitors for precise treatment of the disease.

Pelagic bacterioplankton exhibit biogeographical patterns linked with exporting organic carbon and energy fluxes into the deep ocean. However, knowledge of the mechanisms shaping deep-sea bacterial communities remains largely elusive. In this study, we used high throughput sequencing of the 16S rRNA gene to reveal significant annual bacterioplankton community dynamics in the South China Sea during three summer cruises (2016–2018). As we expected, the epipelagic–bathypelagic connective amplicon sequence variants (ASVs, mostly belonging to Actinobacteriota, Firmicutes, and Cyanobacteria) suggested that they not only affect the community structure but also influence the carbon cycling functions of bathypelagic bacterioplankton in different years. However, the microbial source tracking (MST) analysis indicated that the directly linked proportions between the bathypelagic and epipelagic samples were minimal. Thus, the epipelagic bacteria communities may form “seeds” rather than directly sinking into the deep ocean to influence bathypelagic bacteria. This study provides a new perspective on the mechanisms shaping the deep ocean bacterioplankton communities.

Appropriate deep application of fertilizer is the key basis for improving nitrogen use efficiency (NUE). However, the effects of different deep application methods and fertilizer types on nutrient migration, NUE and biomass in wheat season are unclear. Therefore, in this study, a barrel planting test with multilayer fertilization (15N labeled urea (U) and coated urea (CU)) was conducted in a long-term positioning trial of winter wheat in the North China Plain (NCP). We quantified the migration of fertilizer N (Ndff) in soil–plant–atmosphere and its effects on wheat biomass and NUE based on surface (Usur, CUsur), layered-strip (Ustr, CUstr) and layered-mix fertilization (Umix, CUmix) of U and CU. Compared with surface fertilization, the concentration of mineral N in root zone (0–40 cm) was increased by Ustr and Umix (8.6–50.3%), and the concentration of ammonium N was decreased by CUstr and CUmix (49.6–76.0%), but there was no change in the nitrate N. The biomass and total N absorption of wheat tissues (straw and root) were increased by 12.3–38.9% under Ustr and CUstr. Meanwhile, the distribution of Ndff in the 0–10 cm soil was decreased under Ustr and CUstr, but it was increased in the 10–30 cm soil, thereby promoting the absorption of Ndff in wheat tissues by 12.3–28.7%. The rates of absorption and loss of Ndff were the highest (57.6–58.5%) and the lowest (4.5%) under Ustr and CUstr, respectively, compared with other treatments. Consequently, layered-strip fertilization optimized the migration and utilization of Ndff within the soil–plant–atmosphere system. This approach equalized distribution, enhanced absorption and minimized losses of Ndff, resulting in an increase in NUE by 9.6–16.7%. Under the same treatment, CU was more suitable for crop nutrient requirements than U, which was more conducive to the improvement of NUE. Our findings will provide a scientific basis for the precise directional fertilization of winter wheat in the NCP.

Hypertension is a complex disease which is mainly influenced by genetic factors. Recently, genome-wide association study (GWAS) found three novel endothelial dysfunction-related sites: Vascular endothelial growth factor A (VEGFA) rs9472135, Faciogenital dysplasia 5 (FGD5) rs11128722, Zinc Finger C3HC-type Containing 1 (ZC3HC1) rs11556924. Endothelial dysfunction is one of the early events in pathophysiology of essential hypertension. To investigate the association of endothelial dysfunction-related genes with essential hypertension, we conducted a case-control study of 431 patients with hypertension and 345 controls. The polymorphisms were detected using Taqman Probe. The alleles and genotypes of ZC3HC1 rs11556924 and VEGFA rs9472135 were not statistically different between the two groups, while the allele of FGD5 rs11128722 was different [P = 0.045, OR = 1.265, 95% CI = (1.009–1.586)], especially in the male [P = 0.035, OR = 1.496, 95% CI = (1.037–2.158)]. Analyzing the different of genotype distribution of 3 SNPs in the two groups under different genetic models, the genotypes of FGD5 rs11128722 showed difference in male under dominant model [P = 0.049, OR = 1.610, 95% CI = (1.018–2.544)]. The polymorphism of FGD5 rs11128722 had a significant difference in Body Mass Index (BMI) among different genotypes; In the additive genetic model, BMI of GA genotype was higher than that of GG (P = 0.038); GA + AA was higher than GG in the dominant genetic model (P = 0.011). In our study, we found that the polymorphisms of VEGFA rs9472135 and ZC3HC1 rs11556924 may not significantly associated with the risk of essential hypertension, and FGD5 rs11128722 may increase the risk of it, especially in elderly men.

Poly(ADP-ribose) polymerase inhibitors (PARP inhibitors) have had an increasing role in the treatment of ovarian and breast cancers. PARP inhibitors are selectively active in cells with homologous recombination DNA repair deficiency caused by mutations in BRCA1/2 and other DNA repair pathway genes. Cancers with homologous recombination DNA repair proficiency respond poorly to PARP inhibitors. Cancers that initially respond to PARP inhibitors eventually develop drug resistance. We have identified salt-inducible kinase 2 (SIK2) inhibitors, ARN3236 and ARN3261, which decreased DNA double-strand break (DSB) repair functions and produced synthetic lethality with multiple PARP inhibitors in both homologous recombination DNA repair deficiency and proficiency cancer cells. SIK2 is required for centrosome splitting and PI3K activation and regulates cancer cell proliferation, metastasis, and sensitivity to chemotherapy. Here, we showed that SIK2 inhibitors sensitized ovarian and triple-negative breast cancer (TNBC) cells and xenografts to PARP inhibitors. SIK2 inhibitors decreased PARP enzyme activity and phosphorylation of class-IIa histone deacetylases (HDAC4/5/7). Furthermore, SIK2 inhibitors abolished class-IIa HDAC4/5/7–associated transcriptional activity of myocyte enhancer factor-2D (MEF2D), decreasing MEF2D binding to regulatory regions with high chromatin accessibility in FANCD2, EXO1, and XRCC4 genes, resulting in repression of their functions in the DNA DSB repair pathway. The combination of PARP inhibitors and SIK2 inhibitors provides a therapeutic strategy to enhance PARP inhibitor sensitivity for ovarian cancer and TNBC.

Long non‐coding RNAs (lncRNAs) are emerging as key molecules in various cancers, yet their potential roles in the pathogenesis of breast cancer are not fully understood. Herein, using microarray analysis, we revealed that the lncRNA RACGAP1P, the pseudogene of Rac GTPase activating protein 1 (RACGAP1), was up‐regulated in breast cancer tissues. Its high expression was confirmed in 25 pairs of breast cancer tissues and 8 breast cell lines by qRT‐PCR. Subsequently, we found that RACGAP1P expression was positively correlated with lymph node metastasis, distant metastasis, TNM stage, and shorter survival time in 102 breast cancer patients. Then, in vitro and in vivo experiments were designed to investigate the biological function and regulatory mechanism of RACGAP1P in breast cancer cell lines. Overexpression of RACGAP1P in MDA‐MB‐231 and MCF7 breast cell lines increased their invasive ability and enhanced their mitochondrial fission. Conversely, inhibition of mitochondrial fission by Mdivi‐1 could reduce the invasive ability of RACGAP1P‐overexpressing cell lines. Furthermore, the promotion of mitochondrial fission by RACGAP1P depended on its competitive binding with miR‐345‐5p against its parental gene RACGAP1, leading to the activation of dynamin‐related protein 1 (Drp1). In conclusion, lncRNA RACGAP1P promotes breast cancer invasion and metastasis via miR‐345‐5p/RACGAP1 pathway‐mediated mitochondrial fission.

Abstract Robot deburring is an effective method for improving the surface quality of the high‐voltage copper contact. The first step of robot deburring is to acquire the burr images. We propose a new burr mathematical model and build a real burr image dataset for burr image denoising. In order to improve burr image denoising effects of the high‐voltage copper contact, this study proposes an online burr image denoising algorithm, that is, block cosparsity overcomplete learning transform algorithm (BCOLTA). The penalty term and the condition number are affected by the burr parameter. The clustering and transform alternate minimisation algorithms are adopted to achieve lower computational cost and better denoising effect. In addition, BCOLTA also has a good adaptibility to inherent noise images, especially in Gaussian noise. Compared with other traditional and deep learning algorithms by no reference and full reference image quality assessment methods, BCOLTA has state‐of‐the‐art denoising effects and computational complexity on dealing with burr images. This research will play an important role in the intelligent manufacturing field.

Media supplementation has proven to be an effective technique for improving byproduct yield during microbial fermentation. This study explored the impact of different concentrations of bioactive compounds, namely alpha-tocopherol, mannitol, melatonin, sesamol, ascorbic acid, and biotin, on the Aurantiochytrium sp. TWZ-97 culture. Our investigation revealed that alpha-tocopherol was the most effective compound in reducing the reactive oxygen species (ROS) burden, both directly and indirectly. Adding 0.7 g/L of alpha-tocopherol led to an 18% improvement in biomass, from 6.29 g/L to 7.42 g/L. Moreover, the squalene concentration increased from 129.8 mg/L to 240.2 mg/L, indicating an 85% improvement, while the squalene yield increased by 63.2%, from 19.82 mg/g to 32.4 mg/g. Additionally, our comparative transcriptomics analysis suggested that several genes involved in glycolysis, pentose phosphate pathway, TCA cycle, and MVA pathway were overexpressed following alpha-tocopherol supplementation. The alpha-tocopherol supplementation also lowered ROS levels by binding directly to ROS generated in the fermentation medium and indirectly by stimulating genes that encode antioxidative enzymes, thereby decreasing the ROS burden. Our findings suggest that alpha-tocopherol supplementation can be an effective method for improving squalene production in Aurantiochytrium sp. TWZ-97 culture.

Remote sensing images often have limited resolution, which can hinder their effectiveness in various applications. Super-resolution techniques can enhance the resolution of remote sensing images, and arbitrary resolution super-resolution techniques provide additional flexibility in choosing appropriate image resolutions for different tasks. However, for subsequent processing, such as detection and classification, the resolution of the input image may vary greatly for different methods. In this paper, we propose a method for continuous remote sensing image super-resolution using feature-enhanced implicit neural representation (SR-FEINR). Continuous remote sensing image super-resolution means users can scale a low-resolution image into an image with arbitrary resolution. Our algorithm is composed of three main components: a low-resolution image feature extraction module, a positional encoding module, and a feature-enhanced multi-layer perceptron module. We are the first to apply implicit neural representation in a continuous remote sensing image super-resolution task. Through extensive experiments on two popular remote sensing image datasets, we have shown that our SR-FEINR outperforms the state-of-the-art algorithms in terms of accuracy. Our algorithm showed an average improvement of 0.05 dB over the existing method on ×30 across three datasets.

The consequences of climate change may directly or indirectly impact the marine biosphere. Although ocean stratification has been recognized as one of the crucial consequences of ocean warming, its impacts on several critical aspects of marine microbes remain largely unknown in the Indian Ocean. Here, we investigate the effects of water stratification, in both surface and subsurface layers, on hydrogeographic parameters and bacterioplankton diversity within the equatorial eastern Indian Ocean (EIO). Strong stratification in the upper 200 m of equatorial EIO was detected with evidential low primary productivity. The vertical bacterioplankton diversity of the whole water columns displayed noticeable variation, with lower diversity occurring in the surface layer than in the subsurface layers. Horizontal heterogeneity of bacterioplankton communities was also in the well-mixed layer among different stations. SAR11 and Prochlorococcus displayed uncharacteristic low abundance in the surface water. Some amplicon sequence variants (ASVs) were identified as potential biomarkers for their specific depths in strongly-stratified water columns. Thus, barriers resulting from stratification are proposed to function as an ‘ASV filter’ to regulate the vertical bacterioplankton community diversity along the water columns. Overall, our results suggest that the effects of stratification on the structure and diversity of bacterioplankton can extend up to the bathypelagic zone in the strongly-stratified waters of the equatorial EIO. This study provides the first insight into the effect of stratification on the subsurface microbial communities in the equatorial eastern Indian Ocean.

The study of heterotrophic protists in the ocean is still in its early stages, compared to other microorganisms such as bacteria, archaea, and photoautotrophic protists. Labyrinthulomycetes protists (LP) are a type of unicellular protists that are widely distributed in global waters and have the potential to produce high-value products. In this study, the abundance, diversity, and community structure of LP in the coastal zone of Hainan Island in the South China Sea were investigated through quantitative PCR and high-throughput sequencing. The results showed that LP abundance varied by location and depth, with the highest levels (37.3 × 103 copies/L) found in the middle layer offshore and the lowest (0.386 × 103 copies/L) in the bottom layer offshore. The middle layer (chlorophyll maximum layer) had higher LP abundance both inshore and offshore than the surface and bottom layers. Interestingly, the highest LP richness and diversity was found in the inshore bottom. There was a significant difference in LP abundance between the offshore surface and bottom layers. The LP community was dominated by the genus Aplanochytrium, and four different ecotypes were identified. Additionally, the genus Aurantiochytrium had different cooperative and competitive strategies with bacteria in different habitats. This study sheds light on the abundance and community structure of LP in the coastal zone of Hainan Island, explores the potential interactions between LP and bacterial populations, and raises questions about the potential differentiation of LP ecotypes.

Human disturbance and climate change can easily lead to the shift of dryland ecological quality. However, how to comprehensively assess the shift of dryland ecological quality remains challenging. The ecological quality of specific dryland ecosystem in Northern China is less investigated following the launch of long-term large-scale national ecological restoration projects. This study established a dryland ecological quality assessment framework by the “One-out, all-out (1OAO)” principle. The ecological quality dynamics of Otindag sandy land in Northern China was evaluated by choosing 16 years (2000–2015) ecological quality as a baseline and comparing it to the report period (2005–2020). Three indicators, normalized difference vegetation index (NDVI), soil organic carbon (SOC) and dust sandstorm (DSS) was used to represent vegetation, soil and atmosphere quality, respectively. Our results showed that the vegetation quality kept stable in most areas, improved area of vegetation quality accounted for 30% of Otindag sandy land. The soil quality improved and degraded area accounted for 10% and 83%, respectively. The atmosphere quality increased in 98% area of Otindag sandy land. Overall, the ecological quality of Otindag sandy land improved in one fifth but degraded in four fifth following the launch of the national ecology restoration projects. Both degradation area is persistent since 2000 and no recent degradation found compared baseline to report period. The improvement was mainly in the southeastern part and the degradation was in western part of Otindag sandy land. This study provides practical basis for the scientific evaluation of dryland ecological quality dynamics and will facilitate the regional ecosystem management.

Normal estimation is an essential task for scanned point clouds in various CAD/CAM applications. The method (GLSRNE) based on guided least squares representation (GLSR) balances speed with quality well among state-of-the-art methods. First, it segments each neighborhood into multiple sub-neighborhoods. For some neighborhoods, the segmentation is obtained by GLSR which is an efficient subspace segmentation model and widely applied in other applications. The segmentation of the rest neighborhoods is inferred via the subspace structure propagation (SSP) algorithm. Then, each sub-neighborhood is fitted by a plane. The plane achieving the minimum distance with the current point is selected for the final normal estimation. We make improvements for effectiveness and efficiency in the following three aspects. First, to improve the speed of GLSR, we propose a novel iterative algorithm to reduce the computation complexity from $O(n^{3})$ to $O(n^{2})$ with its convergence guaranteed theoretically, where $n$ represents the number of the data points. Moreover, this proposed algorithm will also be useful for other applications. Second, we add a normal constraint for SSP to improve accuracy. Third, when selecting one plane to estimate the final normal, we consider the match between the plane and all neighbors, whereas GLSRNE only considers the match between the plane and the current point. The experiments exhibit that our method is faster than GLSRNE and more effective than GLSRNE and other state-of-the-art methods.

One major drawback of 3D printing technologies is the low printing efficiency. In general, it takes 8-12 hours to print a normal 3D model. 3D printing technologies manufacture objects layer by layer where each layer is composed of one or more closed 2D polygons (named as slices in this work). As a result, the number of slices will directly affects the printing time. In addition, most 3D printing technologies need extra supporting structures to support overhang regions during printing, such as Fused Deposition Modeling (FDM), which will largely increase manufacturing time. We observe that both the slicing number and the amount of supporting structures are affected by printing direction. In this work, a novel printing direction optimization algorithm is proposed based on the amount of slices and overhang areas. In the proposed method, Genetic Algorithm is used to obtain optimal printing direction where the fitness function is designed as the weighted slice number and overhang areas. Experimental results show that the proposed algorithm is able to obtain an optimal printing direction to reduce the number of slices and overhang areas.

While recent approaches based on offline learning perform well in balancing the accuracy and speed of tracking, it is still non-trivial to accommodate a pre-trained model to an unseen target. In fact, online learning, which requires to capture specified target characteristics from one shot, is a unique attribute of single object tracking. How to favorably bridge the gap between the offline and online learning is of importance for tracking any unseen targets. In this work, in order to obtain sequence-specific information, we propose an online lightweight network consisting of feature adapting layer and ridge regression layer. Its key innovation is to interpret the ridge regression as one layer of the network. Furthermore, we integrate cross-similarity into the Siamese network and train it offline in an end-to-end manner to acquire the fine-grained local pattern of the target object. Through our effective fusion scheme for the offline and online procedures, our method can achieve considerable improvements on prevalent benchmarks.

Person re-identification (Re-ID) has greatly benefited from utilizing features of different levels. Most methods draw support from elegant network designs and elaborate fusion modules. In this paper, different from employing multi-level feature fusion like mainstream skip connection methods, we propose a Deep Classification Consistency(DCC) layer which imposes classification consistency on deep features from different stages in CNNs. DCC regularizes the training process of networks. It profoundly changes the distribution of learned features, making them more discriminative and transferable. Extensive experiments on Market-1501, DukeMTMC-reID, and CUHK03 datasets prove that the proposed method has obtained competitive performances when compared with state-of-the-art approaches, especially those advanced methods merely based on metric learning.

Automatic human gender recognition is an important and classical problem in artificial intelligence. Most of the previous gender recognition works are based on vision appearance and biometric characteristics. However, there are fewer gender recognition approaches for 3D human shapes. In this article, we propose a novel deep neural network learning method for gender recognition of 3D human shapes. Firstly, we introduce effective descriptors to distinguish male and female of 3D human shapes via probability distributions of biharmonic distances among points. Secondly, the above distances-based low-level descriptors are fed into a fully connected neural network for gender recognition. Furthermore, we construct a larger 3D human shape dataset for evaluation of the proposed gender recognition method by collecting and labeling human shape models. Compared with previous works, our method obtains higher recognition accuracy and has more advantages, such as posture invariant, robust to noises, and no need of landmarks or pre-alignment process.

PurposeSuperior limbic keratoconjunctivitis (SLK) is a bilateral, chronic inflammatory disease that recurs for up to several years; however, the fundamental processes involved in its pathogenic mechanisms remain unknown. We aimed to investigate the metabolomic alterations in the tear fluids of patients with superior limbic keratoconjunctivitis (SLK) compared with those of healthy volunteers (Ctrl group).MethodsWe performed a cross-sectional study involving 42 subjects. Tear fluid was taken from one eye of 24 SLK patients (40.13 ± 14.55 years, 83.33% female) and 18 healthy volunteers (Ctrl, 39.89 ± 9.2 years, 72.22% female) using Schirmer strips. After the liquid extraction of tear metabolites, samples were infused into the QE HFX Orbitrap mass spectrometer in both positive and negative ion mode. Metabolites were quantitatively analyzed and matched with entries in the HMDB database. Metabolic differences between the SLK group and the control group were identified based on multivariate statistical analysis. Open database sources, including SMPDB and MetaboAnalyst, were used to identify metabolic pathways.ResultsAmong 179 metabolites retained for annotation, 133 metabolites were finally identified, among which 50 were found to be significantly changed in SLK patients. Of these 50 metabolites, 31 metabolites significantly increased and 19 metabolites decreased in SLK patients. The altered metabolites are mainly involved in α linolenic acid and linoleic acid metabolism, ketone body metabolism, butyrate metabolism, mitochondrial electron transport chain, carnitine synthesis, and so on. The most significantly changed pathway was linoleic acid metabolism. To explore the utility of tear biomarkers, a model combining 9 metabolites (phenol, ethyl glucuronide, eicosapentaenoic acid, 12-keto-leukotriene B4, linoleic acid, hypoxanthine, triethanolamine, 1-nitrohexane, and terephthalic acid) was selected as a candidate biomarker.ConclusionThe results reveal that SLK has a specific metabolomic profile, of which some key elements can serve as potential biomarkers of SLK for diagnostic and prognostic purposes. The findings of this study are novel and provide a basis for further investigations of the mechanism of SLK.

The spatial pattern of color patches plays a crucial role in affecting the visual quality of peri-urban forests dominated by Cotinus coggygria var. cinerea Engl. in autumn. The impact mechanism has been studied to facilitate algorithm-based automatic visual quality estimation. The color patterns of 120 photographs were calculated after color quantization and automatic color substitution. The scenic beauty of the forest was estimated by 698 respondents. Multiple correlations between visual quality and color pattern metrics were explored with stepwise regression. Principal component analysis (PCA) was also employed to investigate the impact mechanism of color patterns on visual quality. Number of patches (NP), largest patch index (LPI), mean patch area (AREA_MN), patch size standard deviation (AREA_SD), and Shannon’s evenness index (SHEI) were the main factors affecting the visual quality of the Cotinus coggygria forest. AREA_MN correlated positively with visual quality, while NP, LPI, AREA_SD, and SHEI correlated negatively. Moreover, AREA_SD had the most significant impact on the visual quality of the landscape, while SHEI, LPI, and AREA_MN had the second-highest impact. The evenness and the size of color patches significantly affected the visual quality of the forest landscapes. Balancing the diversity and evenness of color patches plays a decisive role in creating a forest landscape with high visual quality.

Surface defect inspection is a key technique in industrial product assessments. Compared with other visual applications, industrial defect inspection suffers from a small sample problem and a lack of labeled data. Therefore, conventional deep-learning methods depending on huge supervised samples cannot be directly generalized to this task. To deal with the lack of labeled data, unsupervised subspace learning provides more clues for the task of defect inspection. However, conventional subspace learning methods focus on studying the linear subspace structure. In order to explore the nonlinear manifold structure, a novel neural subspace learning algorithm is proposed by substituting linear operators with nonlinear neural networks. The low-rank property of the latent space is approximated by limiting the dimensions of the encoded feature, and the sparse coding property is simulated by quantized autoencoding. To overcome the small sample problem, a novel data augmentation strategy called thin-plate-spline deformation is proposed. Compared with the rigid transformation methods used in previous literature, our strategy could generate more reliable training samples. Experiments on real-world datasets demonstrate that our method achieves state-of-the-art performance compared with unsupervised methods. More importantly, the proposed method is competitive and has a better generalization capability compared with supervised methods based on deep learning techniques.

Ammonia (NH3) is one of the main precursors of secondary inorganic aerosols. In 2018, the NH3 emissions of China’s cereal production (rice, wheat and maize) were estimated to be 3.3 Mt NH3-N. Numerous NH3 mitigation strategies have been developed in agriculture to reduce the emissions and improve air quality. However, due to the cost and unfeasibility of some developed techniques, the application of these mitigation measures is relatively slow in cropland. Therefore, developing low-cost, easy-operation, and feasible mitigation measures is an important breakthrough to solve the pollution of ammonia emissions in grain fields. The one-time deep application of nitrogen fertilizer in crop growing season, referred to as one-time application, is a promising ammonia mitigation measure for grain fields. It is a low-cost mode of fertilizer application suitable for grain fields as it saves labor and reduces the input of agricultural machinery. Therefore, incentive policies should be formulated to promote it for wide-range application in the whole country, especially in the areas with serious ammonia pollution, in order to achieve the goal of green and sustainable agricultural production.

Upwelling, a common oceanographic event, can make great contributions, directly or indirectly, to deep ocean carbon sequestration. However, the extent of its contribution indirectly depends on the composition of microbial communities and the interactions between bacterioplankton and other microorganisms. This study provides insights into the influence of upwelling on bacterioplankton at the whole community level and predicts their potential functional profiles. The α diversity of the bacterial community exhibited no significant differences between the upwelling area and non-upwelling area, while the community composition varied clearly in different habitats. Proteobacteria, Cyanobacteria, Bacteroidota, Firmicutes, and Actinobacteria were the five dominant phyla in all of the habitats. The proportions of members of Firmicutes were increased whereas Cyanobacteria were reduced in upwelling water. However, the percentage of Cyanobacteria was enhanced in the upwelling deep water. Functional genes that are involved in signal transductions, which belong to environmental information processing, were more active in upwelling surface water than in the other habitats. Closer and more complex relationships between bacterioplankton and microbial eukaryotes were found in the upwelling area, which altered with the variation of the external environmental conditions. Cyanobacteria showed a positive correlation with microbial eukaryotes in upwelling deep water. Combined with the high proportions of Cyanobacteria in upwelling deep water, this might be strong evidence that Cyanobacteria contribute to a deep ocean carbon sink. Overall, our study reveals the impacts of upwelling on the bacterial community composition, metabolic functions, and microbial interactions, which are significant to further understanding the carbon sink effects of upwelling.

Enhancer RNAs (eRNAs), a class of non-coding RNAs (ncRNAs) transcribed from enhancer regions, serve as a type of critical regulatory element in gene expression. There is increasing evidence demonstrating that the aberrant expression of eRNAs can be broadly detected in various human diseases. Some studies also revealed the potential clinical utility of eRNAs in these diseases. In this review, we summarized the recent studies regarding the pathological mechanisms of eRNAs as well as their potential utility across human diseases, including cancers, neurodegenerative disorders, cardiovascular diseases and metabolic diseases. It could help us to understand how eRNAs are engaged in the processes of diseases and to obtain better insight of eRNAs in diagnosis, prognosis or therapy. The studies we reviewed here indicate the enormous therapeutic potency of eRNAs across human diseases.

The burrs on high‐voltage copper contact leads to point discharge and device damage. Since the high‐voltage copper contact has different machining batch and the contour various which the machine tool remove is not economic and robot usually is used to remove the burrs of high‐voltage copper contact. The first step for robot deburring is to identify burrs. In order to improve the performance of copper contact burr video denoising, this article presents online burr video denoising sparsifying transforms algorithm, which defined two alternative values between the optimal sparse signal and transform learning dictionary, simultaneously, calculated the mean of peak signal‐to‐noise ratio, the mean of execution time, the STD (STandard Deviation), and the VAR (VARiance), accordingly presented an burr video denoising algorithm and compared with state‐of‐the‐art video denoising algorithms. The experiment results show that compared with traditional methods, the burr video denoising algorithm has higher denoising precision, faster denoising speed, and stronger high‐noise‐level processing capacity, and so on. The numeric experiments show that the proposed approach has higher peak signal‐to‐noise ratio and less computation complexity than the existing video denoising methods.

The real emission mitigation by the ecological restoration projects depends upon the integrated effect of all greenhouse gas (GHG) budgets rather than the carbon sequestration alone. However, a comprehensive and robust methodology for estimating the relevant GHG budgets and net mitigation of China's ecological restoration projects is still urgently to await development. Based on the methods from IPCC and statistical data of the management practices under the projects, we constructed a methodology for carbon accounting and determining net mitigation for ecological restoration projects in China (CANM-EP). GHG emissions generated from different processes and practices of the projects were included in the CANM-EP, and by this methodology, carbon sequestration, GHG balance changes induced by ecological response, on-site and off-site GHG emissions could be estimated. To investigate the adsorption behavior of Cu (I)-MOF material for chromium (VI) in water, the parameters of influencing adsorption were optimized and found as follows: the optimal pH was 6 for the adsorption of Cr (VI) by the Cu (I)-MOF, the optimal amount of adsorbent was 0.45 g·L−1, and the adsorption saturation time was within 180 min. Subsequently, the kinetics results were fitted by four models such as pseudo-first-order, pseudo-second-order, Elovich, and intraparticle diffusion models. Among them, the adsorption of chromium (VI) was more inclined to the pseudo-first-order model (Radj2 = 0.9230). Then, the isotherm data were fitted by Langmuir and Freundlich models. The results indicated that Langmuir isotherm was the excellent match model (Radj2 = 0.9827). It belongs to a monolayer adsorption, and the maximum adsorption capacity was 95.92 mg·g−1. Subsequently, the thermodynamic parameters of the adsorption were calculated as follows: enthalpy change (ΔHθ) was −8.583 kJ·mol−1, entropy change (ΔSθ) was −8.243 J·mol−1 K−1, and the Gibbs function change (ΔGθ) was less than zero in the temperature range of 288–328 K, indicating that the reaction was spontaneous. Finally, both the spectra of infrared and XPS supported the adsorption mechanism that belonged the ion exchange. The spectra of XRD and SEM images shown that the structure of Cu (I)-MOF remained stable for at least 3 cycles. In conclusion, Cu (I)-MOF material has a high adsorption capacity, good water stability, low cost, and easy to prepare in large quantities in practical application. It will be a promising adsorbent for the removal of Cr (VI) from water.

Corticofugal modulation of auditory responses in subcortical nuclei has been extensively studied whereas corticofugal synaptic transmission must still be characterized. This study examined postsynaptic potentials of the corticocollicular system, i.e., the projections from the primary auditory cortex (AI) to the central nucleus of the inferior colliculus (ICc) of the midbrain, in anesthetized C57 mice. We used focal electrical stimulation at the microampere level to activate the AI (ESAI) and in vivo whole-cell current-clamp to record the membrane potentials of ICc neurons. Following the whole-cell patch-clamp recording of 88 ICc neurons, 42 ICc neurons showed ESAI-evoked changes in the membrane potentials. We found that the ESAI induced inhibitory postsynaptic potentials in 6 out of 42 ICc neurons but only when the stimulus current was 96 μA or higher. In the remaining 36 ICc neurons, excitatory postsynaptic potentials (EPSPs) were induced at a much lower stimulus current. The 36 ICc neurons exhibiting EPSPs were categorized into physiologically matched neurons (n = 12) when the characteristic frequencies of the stimulated AI and recorded ICc neurons were similar (≤1 kHz) and unmatched neurons (n = 24) when they were different (>1 kHz). Compared to unmatched neurons, matched neurons exhibited a significantly lower threshold of evoking noticeable EPSP, greater EPSP amplitude, and shorter EPSP latency. Our data allow us to propose that corticocollicular synaptic transmission is primarily excitatory and that synaptic efficacy is dependent on the relationship of the frequency tunings between AI and ICc neurons.

To assess contamination levels and ecological risks of heavy metals in agricultural soil from Shanxi Province of China, a total of 33 samples in the surface soil were collected from 11 cities in Shanxi. The soil samples were digested by a mixed acid of nitric acid and hydrofluoric acid on a microwave digestion system, then the levels of eight heavy metals were analysed using an inductively coupled plasma mass spectrometer. The pollution levels of soil heavy metals were evaluated using a geo-accumulation index and their ecological risks were assessed using risk index calculated by Hakanson's method. As a result, the average concentrations of the heavy metals As, Cd, Cr, Cu, Hg, Ni, Pb and Zn were 12.9 ± 4.8, 0.35 ± 0.23, 43 ± 14, 27 ± 19, 0.25 ± 0.14, 21.7 ± 5.7, 17 ± 13 and 89 ± 53 mg kg−1, respectively. By comparison to the Chinese soil environmental quality (GB15618-2018), only 9% of Cd samples and 3% of Cu samples exceeded their corresponding screening criteria. Subsequently, the results of geo-accumulation indices suggested that Shanxi's soil suffered from moderate to heavy contamination posed by Cd and Hg, and risk indices exhibited a similar trend that Cd and Hg were the main contributors for considerable to very high ecological risk. Finally, the analysis of variance indicated that the mean levels of Cd significantly occurred at Yuncheng areas among the 11 cities (n = 3, p < 0.05), but Hg concentrations did not have significantly statistical differences. This study demonstrated that metals Cd and Hg had higher levels and ecological risks for agricultural soil in Shanxi, especially, Yuncheng City suffered from heavy Cd contamination. The findings of the present study will provide basic information on management and control of the agricultural soil contamination in Shanxi Province, China.

The neural mechanisms underlying forward suppression in the auditory cortex remain a puzzle. Little attention is paid to thalamic contribution despite the important fact that the thalamus gates upstreaming information to the auditory cortex. This study compared the time courses of forward suppression in the auditory thalamus, thalamocortical inputs and cortex using the two-tone stimulus paradigm. The preceding and succeeding tones were 20-ms long. Their frequency and amplitude were set at the characteristic frequency and 20 dB above the minimum threshold of given neurons, respectively. In the ventral division of the medial geniculate body of the thalamus, we found that the duration of complete forward suppression was about 75 ms and the duration of partial suppression was from 75 ms to about 300 ms after the onset of the preceding tone. We also found that during the partial suppression period, the responses to the succeeding tone were further suppressed in the primary auditory cortex. The forward suppression of thalamocortical field excitatory postsynaptic potentials was between those of thalamic and cortical neurons but much closer to that of thalamic ones. Our results indicate that early suppression in the cortex could result from complete suppression in the thalamus whereas later suppression may involve thalamocortical and intracortical circuitry. This suggests that the complete suppression that occurs in the thalamus provides the cortex with a "silence" window that could potentially benefit cortical processing and/or perception of the information carried by the preceding sound.

In the field of visual tracking, the methods of Discriminative Correlation Filters (DCF) have showed excellent performance, which rely heavily on the choice of feature descriptors. The Continuous Convolution Operator Tracker (C-COT) is a novel correlation filter to track the target position in the continuous domain, which achieved significant effects. However, as for various visual scenes, different feature descriptors are suitable to different environments. If each feature channel is given the same confidence during the tracking phase, it would limit the performance of some good features. To address this problem, this paper proposes an improved C-COT algorithm that can adaptively perform feature channel weighting. The Average Peak Correlation Energy (APCE) is used to evaluate the corresponding response map of each feature channel, guiding the target appearance model to give different weights to different features. Then, we can obtain the final weighted feature response map whose peak value is applied to locate the target. In addition, the C-COT updates the appearance model rigorously in every frame, which may lead to over-fitting and increase computional complexity. Therefore, in order to reduce the redundancy of the online training sample and avoid similar background interference, we adopts the method of Peak Side Lobe Ratio (PSLR) to update the model. We perform comprehensive experiments on OTB50 and OTB100. The results show that the improved tracker achieves better accuracy, especially in some specific video scenes. In addition, speed has also improved.

In this paper, inspired by an inherent characteristic of human visual system capable of recognizing salient regions from a complicated scene, we treat a defective region as a salient region in fabric images. A novel fabric defect detection method, which is based on saliency metric for color dissimilarity and positional aggregation, is proposed. In the method, the RGB color space of a given fabric image is first converted into the CIE L*a*b color space for feature representation. Then, the color dissimilarity and the positional distance between similar patches are jointly used to measure the defective values. To improve the contrast between the defective region and the non-defective region, a multi-scale analysis scheme performed on the pyramid images of the input fabric image is applied to calculate the defective values. Finally, the obtained defect map image is further enhanced by a certain threshold regarding to the obtained defective values. Thorough experimental results on several types of fabric images indicate the effectiveness of the newly proposed method.

Object tracking from LiDAR point clouds, which are always incomplete, sparse, and unstructured, plays a crucial role in urban navigation. Some existing methods utilize a learned similarity network for locating the target, immensely limiting the advancements in tracking accuracy. In this study, we leveraged a powerful target discriminator and an accurate state estimator to robustly track target objects in challenging point cloud scenarios. Considering the complex nature of estimating the state, we extended the traditional Lucas and Kanade (LK) algorithm to 3D point cloud tracking. Specifically, we propose a state estimation subnetwork that aims to learn the incremental warp for updating the coarse target state. Moreover, to obtain a coarse state, we present a simple yet efficient discrimination subnetwork. It can project 3D shapes into a more discriminatory latent space by integrating the global feature into each point-wise feature. Experiments on KITTI and PandaSet datasets showed that compared with the most advanced of other methods, our proposed method can achieve significant improvements—in particular, up to 13.68% on KITTI.

Exposure to ambient fine particulate matter (fine PM) pollution has been previously associated with ocular surface diseases. But, to the best of our knowledge, the in vivo long-term effects of fine PM on the ocular surface have not been investigated. We aimed to evaluate the effects of fine PM on cultured human corneal epithelial (HCE) cells and on the ocular surfaces of mice, with standard reference material of fine PM(SRM 2786). We applied fine PM suspension to the eyes of C57BL/6 mice for up to 6 months. In vivo examinations, including tear secretion, tear film break-up time (TBUT) and corneal fluorescein staining, were performed in the 3rd and 6th month. At the end of the in vivo study, the corneal histological changes and conjunctival goblet cells were examined by staining, and cytokines in tissue were also detected. In addition, HCE cells were treated with fine PM for 12 h and 24 h. Then, cell apoptosis and reactive oxygen species (ROS) formation was detected. We found that fine PM damages the mouse eye in a dose- and time-dependent manner. In mice, the tear secretion and tear film break-up time were significantly reduced, along with the development of corneal epithelial damage, apoptosis of conjunctival epithelial cells and hypoplasia of conjunctival goblet cells. In addition, IL-18, IL-22, IL-23 and MCP-1 were increased in both conjunctiva and cornea of the fine PM-treated animals. Furthermore, increased apoptosis and ROS production were observed in time- and dose-dependent manner in HCE cells after fine PM exposure for 12 h and 24 h. Our results indicate that fine PM is cytotoxic to both HCE cells and the ocular surface. Long-term topical application of fine PM suspension in mice results in ocular surface changes that are similar to those observed with dry eye.

Physiological studies documented highly specific corticofugal modulations making subcortical centers focus processing on sounds that the auditory cortex (AC) has experienced to be important. Here, we show the effects of focal conditioning (FC) of the primary auditory cortex (FCAI) on auditory brainstem response (ABR) amplitudes and latencies in house mice. FCAI significantly increased ABR peak amplitudes (peaks I–V), decreased thresholds, and shortened peak latencies in responses to the frequency tuned by conditioned cortical neurons. The amounts of peak amplitude increases and latency decreases were specific for each processing level up to the auditory midbrain. The data provide new insights into possible corticofugal modulation of inner hair cell synapses and new corticofugal effects as neuronal enhancement of processing in the superior olivary complex (SOC) and lateral lemniscus (LL). Thus, our comprehensive ABR approach confirms the role of the AC as instructor of lower auditory levels and extends this role specifically to the cochlea, SOC, and LL. The whole pathway from the cochlea to the inferior colliculus appears, in a common mode, instructed in a very similar way.

Canonical extrinsic representations for non-rigid shapes with different poses are preferable in many computer graphics applications, such as shape correspondence and retrieval. The main reason for this is that they give a pose invariant signature for those jobs, which significantly decreases the difficulty caused by various poses. Existing methods based on multidimentional scaling (MDS) always result in significant geometric distortions. In this paper, we present a novel shape unfolding algorithm, which deforms any given 3D shape into a canonical pose that is invariant to non-rigid transformations. The proposed method can effectively preserve the local structure of a given 3D model with the regularization of local rigid transform energy based on the shape deformation technique, and largely reduce geometric distortion. Our algorithm is quite simple and only needs to solve two linear systems during alternate iteration processes. The computational efficiency of our method can be improved with parallel computation and the robustness is guaranteed with a cascade strategy. Experimental results demonstrate the enhanced efficacy of our algorithm compared with the state-of-the-art methods on 3D shape unfolding.

Free and glycosylated sesquiterpene lactones (SLs), which are abundant in leafy vegetables including Brussels/witloof chicory, possess health-promoting effects in vivo. However, the pharmacokinetics of dietary source of SLs remain largely unknown. In this open-label and single-dose trial, sixteen healthy volunteers consumed 150 g of Brussels/witloof chicory juice containing 48.77 μmol SLs in 5 min. Blood, urine, and fecal samples were collected before and after chicory consumption in 24 h. No SLs were detected in the serum, urine, and fecal samples before chicory consumption in all of the participants. Chicory consumption increased lactucin, 11β,13-dihydrolactucin, and their glucuronide/sulfate conjugates, rather than lactucopicrin and 11β,13-dihydrolactucopicrin, as well as glycosylated SLs in biological samples. The peak concentration of total SLs in serum reached 284.46 nmol/L at 1 h, while, in urine, this peak was 220.3 nmol between 2 and 6 h. The recovery of total SLs in blood, urine, and feces was 7.03%, 1.13%, and 43.76% of the ingested dose, respectively. Human fecal suspensions with intestinal microbiota degraded glycosylated SLs in chicory, and converted lactucopicrin and 11β,13-dihydrolactucopicrin to lactucin and 11β,13-dihydrolactucin, respectively. Collectively, Brussels/witloof chicory SLs are poorly bioavailable and they undergo partial gut microbial and phase II metabolism in humans.

Summary: Renal cell carcinoma (RCC) is not a single disease, but several histologically defined cancers with different genetic drivers, clinical courses, and therapeutic responses. The current study evaluated 843 RCC from the three major histologic subtypes, including 488 clear cell RCC, 274 papillary RCC, and 81 chromophobe RCC. Comprehensive genomic and phenotypic analysis of the RCC subtypes reveals distinctive features of each subtype that provide the foundation for the development of subtype-specific therapeutic and management strategies for patients affected with these cancers. Somatic alteration of BAP1, PBRM1, and PTEN and altered metabolic pathways correlated with subtype-specific decreased survival, while CDKN2A alteration, increased DNA hypermethylation, and increases in the immune-related Th2 gene expression signature correlated with decreased survival within all major histologic subtypes. CIMP-RCC demonstrated an increased immune signature, and a uniform and distinct metabolic expression pattern identified a subset of metabolically divergent (MD) ChRCC that associated with extremely poor survival. : Ricketts et al. find distinctive features of each RCC subtype, providing the foundation for development of subtype-specific therapeutic and management strategies. Summary: Gene fusions represent an important class of somatic alterations in cancer. We systematically investigated fusions in 9,624 tumors across 33 cancer types using multiple fusion calling tools. We identified a total of 25,664 fusions, with a 63% validation rate. Integration of gene expression, copy number, and fusion annotation data revealed that fusions involving oncogenes tend to exhibit increased expression, whereas fusions involving tumor suppressors have the opposite effect. For fusions involving kinases, we found 1,275 with an intact kinase domain, the proportion of which varied significantly across cancer types. Our study suggests that fusions drive the development of 16.5% of cancer cases and function as the sole driver in more than 1% of them. Finally, we identified druggable fusions involving genes such as TMPRSS2, RET, FGFR3, ALK, and ESR1 in 6.0% of cases, and we predicted immunogenic peptides, suggesting that fusions may provide leads for targeted drug and immune therapy. : Gao et al. analyze a 9,624 sample TCGA cohort with 33 cancer types to detect gene fusion events.
