In order to reveal the mineral filling sequence and fluid action process in the dissolution pores of the bedded siliceous rocks in the Dongrong Formation of the Sanjiang Basin, the petrological characteristics, geochemical analysis and testing methods were comprehensively applied to research, and the databases of major amount and rare earth elements of siliceous rocks were established, and the sedimentary environment, origin and source of siliceous materials were systematically judged again. The results show that the siliceous rocks of the Nadanhada terrane exposed in the Sanjiang Basin are biogenic. On the basis of early metasomatic microbial dolomite, the layered siliceous rocks have experienced six stages (three fluid properties) of fluid filling, including magnesium rich hydrothermal fluid, siliceous hydrothermal fluid, asphalt and siliceous hydrothermal fluid. Siliceous rock cavity filling material is the product of fluid rock coupling under different temperatures, concentrations and reaction systems under the control of different tectonic movements and hydrocarbon maturation.

Through field geological survey and drilling exploration in Sanjiang Basin, combined with geochemical analysis, the tectonic thermal evolution history of intrusive rocks is studied. The results show that the Early Mesozoic siliceous rocks in Qianjin Depression of Sanjiang Basin are widely distributed. The siliceous rocks are of post diagenetic hydrothermal metasomatic origin, and the hydrocarbon source rock indicators are good, which is expected to be a new formation for oil and gas exploration in Sanjiang area. The time temperature thermal history simulation of fission track shows that there have been two rapid cooling events (late Early Cretaceous Late Cretaceous and Paleocene Eocene) in the Nadanhadaling area since the Mesozoic era, with cooling rates of 3.42~4.81 ℃/Ma and 1.43~1.83 ℃/Ma, respectively. The two rapid cooling processes were caused by the subduction of the Pacific plate. The rapid cooling during the Early Cretaceous to the Late Cretaceous was caused by the change of the subduction direction of the Pacific plate and the collision and superposition of the Okot Motsk block and the East Asian continental margin; The rapid cooling from Paleocene to Eocene was mainly caused by the subduction and retreat of the Pacific plate, which made the East Asian continental margin in an extensional environment and caused denudation and cooling in most parts of Northeast China.

Pilomatricoma, a benign skin appendage tumor, also known as calcifying epithelioma, consists of islands of epithelial cells histologically that contain anucleated cells in the center surrounded by basophilic cells and partial calcification. Sporadic pilomatricomas commonly have somatic mutations in the gene CTNNB1, but causative genes from germline and the underlying pathophysiology are unclear. In this study, we identified a germline missense variant of PLCD1 encoding PLCδ1, c.1186G>A (p.Glu396Lys), in a large Chinese family with autosomal dominant multiple pilomatricomas. Phospholipase C, a key enzyme playing critical roles in intracellular signal transduction, is essential for epidermal barrier integrity. The p.Glu396Lys variant increased the enzymatic activity of PLCδ1, leading to protein kinase C/protein kinase D/extracellular signal–regulated kinase1/2 pathway activation and TPRV6 channel closure, which not only resulted in excessive proliferation of keratinocytes in vitro and in vivo but also induced local accumulation of calcium in the pilomatricoma-like tumor that developed spontaneously in the skin of Plcd1E396K/E396K mice. Our results implicate this p.Glu396Lys variant of PLCD1 from germline leading to gain-of-function of PLCδ1 as a causative genetic defect in familial multiple pilomatricomas.

Adrenal cortical carcinoma (ACC) is a severe malignant tumor with low early diagnosis rates and high mortality. In this study, we used a variety of bioinformatic analyses to find potential prognostic markers and therapeutic targets for ACC. Gene Expression Omnibus (GEO) and The Cancer Genome Atlas (TCGA) data sets were used to perform differential expressed analysis. WebGestalt was used to perform enrichment analysis, while String was used for protein-protein analysis. Our study first detected 28 up-regulation and 462 down-regulation differential expressed genes through the GEO and TCGA databases. Then, GO functional analysis, four pathway analyses (KEGG, REACTOME, PANTHER, and BIOCYC), and protein-protein interaction network were performed to identify these genes by WebGestalt tool and KOBAS website, as well as String database, respectively, and finalize 17 hub genes. After a series of analyses from GEPIA, including gene mutations, differential expression, and prognosis, we excluded one candidate unrelated to the prognosis of ACC and put the remaining genes into pathway analysis again. We screened out CCNB1 and NDC80 genes by three algorithms of Degree, MCC, and MNC. We subsequently performed genomic analysis using the TCGA and cBioPortal databases to better understand these two hub genes. Our data also showed that the CCNB1 and NDC80 genes might become ACC biomarkers for future clinical use.

Phosphorus is a necessary component of all living organisms. This nutrient is mainly transported from the maternal blood to the fetus via the placenta, and insufficient phosphorus availability via the placenta disturbs the normal development of the fetus, especially fetal bone formation in late gestation. Key proteins (phosphate transporters and exporters) that are responsible for the maintenance of placental-fetal phosphorus homeostasis have been identified. A deficiency in the phosphate transporter Pit2 has been shown to result in placental calcification and the retardation of fetal development in mice. What roles does XPR1 (the only known phosphate exporter) play in maintaining placental-fetal phosphorus homeostasis? In this study, we found that Xpr1 expression is strong in the murine placenta and increases with age during gestation. We generated a global Xpr1 knockout mouse and found that heterozygous (Xpr1+/- ) and homozygous (Xpr1-/- ) fetuses have lower inorganic phosphate (Pi) levels in amniotic fluid and serum and a decreased skeletal mineral content. Xpr1-deficient placentas show abnormal Pi exchange during gestation. Therefore, Xpr1 deficiency in the placenta disrupts placental-fetal Pi homeostasis. We also discovered that the placentas of the Xpr1+/- and Xpr1-/- embryos are severely calcified. Mendelian inheritance statistics for offspring outcomes indicated that Xpr1-deficient embryos are significantly reduced in late gestation. In addition, Xpr1-/- mice die perinatally and a small proportion of Xpr1+/- mice die neonatally. RNA sequence (RNA-Seq) analysis of placental mRNA revealed that many of the transcripts are significantly differentially expressed due to Xpr1 deficiency and are linked to dysfunction of the placenta. This study is the first to reveal that XPR1 plays an important role in maintaining placental-fetal Pi homeostasis, disruption of which causes severe placental calcification, delays normal placental function, and restricts fetal growth. © 2019 American Society for Bone and Mineral Research.

Traditionally, moving ship detection by Synthetic Aperture Radar (SAR) image is primarily based on the ship wake feature. However, many ship wakes cannot be imaged by SAR owing to changes in imaging conditions, such as the SAR band, polarisation, incident angle, and sea state. In this study, we discovered a unique phenomenon called ‘azimuth tail’ from Radarsat-2 SAR images. Following research and analysis, we determined that the azimuth tail is not manifested as a visible disturbance on sea surface waves. Instead, it is an observation enabled by certain SAR imaging principles. Consequently, we propose a new method for extracting information on moving vessels after a preliminary analysis of the principle of the azimuth tail. The results of experimental analysis of the correctness of the method indicate that the error of the vessel’s velocity from its azimuth tail is less than 20%, and the azimuth tail can be applied to the detection of moving vessels in oceans using Radarsat-2 SAR imagery.

Objective. Testicular germ cell tumors (TGCT) are a serious malignant tumor with low early diagnosis rates and high mortality. Methods. To investigate novel biomarkers to predict the diagnosis and prognosis of this cancer, bioinformatics analysis was used as an accurate, efficient, and economical method. Results. Our study detected 39 upregulated and 589 downregulated differentially expressed genes (DEGs) using the GEO and TCGA databases. To identify the function of DEGs, GO functional analysis, three pathway analysis (KEGG, REACTOME, and PANTHER), and protein-protein interaction network were performed using the KOBAS website, as well as the String database. After a series of analyses in GEPIA and TIMER, including differential expression, we found one candidate gene related to the prognosis and diagnosis of TGCT. LAPTM5 was also associated with CD8+ T cell and PDCD1 expression, which suggests that it may affect immune infiltration. Conclusions. LAPTM5 was identified as a hub gene, which could be used as a potential biomarker for TGCT diagnosis and prognosis.

Objective. This study is aimed at using genes related to the peroxisome proliferator-activated receptor (PPAR) pathway to establish a prognostic risk model in kidney renal clear cell carcinoma (KIRC). Methods. For this study, we first found the PPAR pathway-related genes on the gene set enrichment analysis (GSEA) website and found the KIRC mRNA expression data and clinical data through TCGA database. Subsequently, we used R language and multiple R language expansion packages to analyze the expression, hazard ratio analysis, and coexpression analysis of PPAR pathway-related genes in KIRC. Afterward, using the Search Tool for the Retrieval of Interacting Genes/Proteins (STRING) website, we established the protein-protein interaction (PPI) network of genes related to the PPAR pathway. After that, we used LASSO regression curve analysis to establish a prognostic survival model in KIRC. Finally, based on the model, we conducted correlation analysis of the clinicopathological characteristics, univariate analysis, and multivariate analysis. Results. We found that most of the genes related to the PPAR pathway had different degrees of expression differences in KIRC. Among them, the high expression of 27 genes is related to low survival rate of KIRC patients, and the high expression of 13 other genes is related to their high survival rate. Most importantly, we used 13 of these genes successfully to establish a risk model that could accurately predict patients’ prognosis. There is a clear correlation between this model and metastasis, tumor, stage, grade, and fustat. Conclusions. To the best of our knowledge, this is the first study to analyze the entire PPAR pathway in KIRC in detail and successfully establish a risk model for patient prognosis. We believe that our research can provide valuable data for future researchers and clinicians.

Background To summary the clinical characteristics of common primary adrenal malignancies in adults, the survival time of the patients, and the prognostic factors associated with these rare malignancies. Methods The Surveillance, Epidemiology, and End Results database (1975–2016) was queried for all patients who were diagnosed with primary adrenal malignancies, including adrenocortical carcinoma(ACC), pheochromocytoma and paraganglioma (PPGL), lymphoma, and sarcoma (SA). The clinical characteristics, overall survival (OS) and cancer-specific survival (CSS) were analyzed, in which propensity score methodology, logistic regression modeling and Kaplan-Meier survival curves were used. The Multivariate Cox proportional hazard model was used to identify factors affecting the prognosis of patients with ACC. Results 3204 patients with primary adrenal malignancies were identified, including 2180 with ACC, 593 with PPGL, 307 with lymphoma, and 124 with SA. Generally, PPGL had a better prognosis than others. For ACC and PPGL, the non-metastatic group had better OS and CSS than the metastatic group; for SA, the non-metastatic group had better OS, however, there was no significant difference in CSS between the two groups. Furthermore, For ACC, the single metastasis group had a better OS than the multiple metastasis group. After tumors metastasis, the surgery group, surgery of primary adrenal lesions, had better OS and CSS than the non-surgery group for ACC and PPGL. For lymphoma, the chemotherapy group had better OS and CSS; for ACC and PPGL, the chemotherapy group had better CSS and worse OS; for SA, the survival showed no significant difference between the two groups. Radiotherapy had the same effect on patients' survival as chemotherapy, except for the lymphoma and the ACC. As for lymphoma, the radiotherapy group had better CSS, whereas the survival showed no significant difference in OS between the two groups. For ACC, the non- radiotherapy group had better CSS and OS. More importantly, for lymphoma, under the condition that both cohorts were the same basically, the surgical group had a better prognosis. The prognosis of ACC was related to sex, age, seer historic stage, surgery, chemotherapy. Conclusion Understanding the clinical characteristics of these tumors and factors affecting prognosis can facilitate the selection of more appropriate clinical treatment options.

Structure heterogeneity and host nucleic acids contamination are two major problems for virus-like particles (VLPs) produced by various host cells. In this study, an in vitro optimized disassembly-purification-reassembly process was developed to obtain uniform and nucleic acid free hepatitis B core (HBc) based VLPs from E. coli fermentation. The process started with ammonium sulfate precipitation of all heterogeneous HBc structures after cell disintegration. Then, dissolution and disassembly of pellets into basic subunits were carried out under the optimized disassembly condition. All contaminants, including host nucleic acids and proteins, were efficiently removed with affinity chromatography. The purified subunits reassembled into VLPs by final removal of the chaotropic agent. Two uniform and nucleic acid free HBc-based VLPs, truncated HBc149 and chimeric HBc183-MAGE3 I, were successfully prepared. It was found that disassembly degree of HBc-based VLPs had a great influence on the protein yield, nucleic acid removal and reassembly efficiency. 4 M urea was optimal because lower concentration would not disassemble the particles completely while higher concentration would further denature the subunits into disordered aggregate and could not be purified and reassembled efficiently. For removal of strong binding nucleic acids such as in the case of HBc183-MAGE3 I, benzonase nuclease was added to the disassembly buffer before affinity purification. Through the optimized downstream process, uniform and nucleic acid free HBc149 VLPs and HBc183-MAGE3 I VLPs were obtained with purities above 90% and yields of 55.2 and 43.0 mg/L, respectively. This study would be a reference for efficient preparation of other VLPs.

Background COVID-19 infection is associated with D-dimer elevations, high rates of thrombus formation, and poor clinical outcomes. We sought to determine if empiric therapeutic anticoagulation (AC) affected survival in COVID-19 patients compared to standard prophylactic AC. Methods Retrospective analysis of 402 COVID-19 patients hospitalized between March 15 and May 31, 2020 was performed. Clinical outcomes were compared between 152 patients treated with therapeutic AC to 250 patients on prophylactic AC. An elastic net logistic regression was designed to first identify the important variables affecting mortality. These variables were then included as covariates to AC in standard multivariate logistic regression models studying the effect of AC on death. Nonparametric survival analysis was conducted, and Kaplan Meier curves were constructed. Results Increased mortality was associated with therapeutic AC [OR 3.42 (2.06, 5.67)]. The log-rank test was statistically significant at p = 0.001 showing higher mortality for patients treated with therapeutic AC compared to prophylactic AC. Subset analysis of critically ill and intubated patients had similar survival curves regardless of AC dose. The log-rank test was not significant even with Prentice modification. For non-ICU patients, the log rank test favoring prophylactic AC disappeared when the analysis was stratified by D-dimer level less or greater than 3 μg/mL. Approximately 9% of patients receiving therapeutic AC experienced clinically significant bleeding or thrombocytopenia, versus 3% in those receiving prophylactic AC. Conclusions In our cohort, therapeutic anticoagulation provided no mortality benefit over thromboprophylaxis, independent of co-morbidities or disease severity. More adverse events were observed with therapeutic AC., Highlights • Anticoagulation strategies for patients with COVID-19 infections remain unclear. • Outcomes of prophylactic versus therapeutic anticoagulation were compared. • In our cohort, therapeutic anticoagulation provided no mortality benefit over thromboprophylaxis. • Therapeutic anticoagulation was associated with increased adverse events.

Plasma fractionation industry is by far the largest protein pharmaceutical provider, but there are still some plasma components in its industrial waste liquid that have not been utilized. This study aimed to develop a simple and efficient method for plasma protein recovery from Cohn fraction V supernatant (FVS), an effluent containing about 40% ethanol. A new affinity chromatography medium was synthesized with a fatty acid ligand. When the medium was applied to recovery of human serum albumin (HSA) from FVS at physiological pH7.4, the process was unsuccessful due to substantial decrease in capacity in the presence of high ethanol concentration. Nevertheless, change of pH from 7.4 to 4.2 emerged an improved adsorption capacity. The carboxyl group of the ligand began to act as cationic ion exchange role. Both HSA and α2HS-glycoprotein were adsorbed by the column, but α2HS-glycoprotein could be eluted by increasing pH from 4.2 to 7.4, while HSA was retained by the column and could only be eluted by addition of fatty acid. Therefore, the adsorption of albumin under pH 4.2 is charge-induced affinity adsorption, not simple ion exchange. The so-called dual-mode adsorption depends not only on the chromatographic medium but also on the separated object and environment. HPSEC showed that the purity of recovered HSA was greater than 98%. Circular dichroism and fluorescence spectra were consistent with that of the commercial product. Furthermore, the measurement by isothermal titration calorimetry showed that the separated HSA still maintained the binding activities with the ligands of warfarin and naproxen. It is therefore possible to directly recover high-purity and high-quality human serum albumin from the effluent of plasma fractionation industry by one-step chromatography.

The maritime administrative department employs synthetic aperture radar (SAR) satellite remote sensing technology to obtain evidence of illegal discharge of ships. If the ship is discharged during navigation, it forms a long dark wake on the SAR image due to the suppression of the Bragg wave by the oil film. This study investigates key techniques for rapid detection of long ship wakes, thereby providing law enforcement agencies with candidate ships for possible discharge. This paper presents a rapid long ship wake detection method that uses satellite imaging parameters and the axial direction of the ship in images to determine the potential detection area of the wake. Then, the threshold of long ship wake detection is determined using statistical analysis, the area is binarized, and isolated points are removed using a morphological filter operator. The method was tested with ENVISAT Synthetic Aperture Radar and GF-3 SAR data, and results showed that the method was effective, and the overall accuracy of the decision reaches 71%. We present two innovations; one is a method that draws a Doppler shift curve, and uses the SAR imaging parameters to determine the detection area of the long wake to achieve rapid detection and reduce the image detection area. The other is where a classical linear fitting method is used to quickly and accurately determine whether the detected dark area is a long ship wake and realizes the twisted long ship wake detection caused by the sea surface flow field, which is otherwise difficult to detect by the traditional Radon and Hough transform methods. This method has good suppression performance for the dark spot false alarm formed by low speed wind region or upward flow. The method is developed for maritime ship monitoring system and will promote the operational application of maritime ship monitoring system.

The siliceous shale oil and gas in Nadanhada terrane has been studied through field geological survey, geochemistry and sedimentary petrology. The results show that the area belongs to a good shale reservoir, and the siliceous rock is brittle and suitable for exploitation. The abundance of organic matter is relatively high. From west to east and from south to north, the abundance of organic matter increases and the maturity of organic matter decreases. From west to east and from south to north, the dynamic metamorphism and thermal contact metamorphism of the siliceous rocks in the Nadanhada terrane are gradually weakened, and the folds and magmatic intrusion of the strata are alleviated.

The Nadanhada terrane is basically a large tectonic sedimentary hybrid, which is an accretionary complex belt formed by the subduction of the Pacific plate to the Eurasian plate. Through field geological survey, paleomagnetic analysis, paleontological analysis and microscopic observation of core slices, its structural and petrological characteristics were studied. The results show that the accretionary complex is mainly composed of 7 parts, including undeformed sedimentary rocks, shear deformed sedimentary rocks, shear deformed volcanic rocks, turbidites, deformed siliceous rocks, limestone blocks and basic ultrabasic rocks. Shear deformation structures of deformed sedimentary rocks and volcanic rocks are developed. The sediment grain size of the Lower Cretaceous reservoirs in each basin in the northern region is relatively coarse, the sorting is moderately deviated, and the rounding is poor, mainly secondary to angular. The grain size of the lower Cretaceous sediments of each basin in the southern region is relatively fine, with coarse, medium and fine sandstones, but mainly medium fine sandstones, with medium sorting preference and sub angular to sub round roundness.

Production of natural pertactin for pharmaceutical use is limited by its low abundance. In this study, recombinant pertactin was highly expressed in the form of inclusion bodies in E. coli . However, up to 75% of the soluble turned out as aggregates when refolding by pulse-fed batch dilution. The conceivable route for aggregate formation was proposed as that the C-terminus of partially folded intermediate with a strong hydrophobic core would intertwine with that region of newly added denatured protein, resulting in aggregation between proteins with different folding states. The key factor for prevention of aggregate formation was to improve the synchronization of refolding. For this purpose, flash-batch dilution was conducted at a scale of 5 L and achieved a monomeric refolding yield of above 70%. Aggregates formed were efficiently removed along with impurities by one-step chromatography of Ni-resin. The purity of monomeric pertactin was >98%. An overall yield was 320 mg per liter fermentation liquor with a total recovery of about 59%. The purified protein was characterized by MALDI-TOF, circular dichroism, fluorescence, HPLC and DSC, and showed similar physiochemical properties compared to its natural counterpart. Animal study showed similar immunological responses and antibodies elicited demonstrated a comparable reactivity.

Human ferritin (HFn) nanocaging is becoming an appealing platform for anticancer drugs delivery. However, protein aggregation always occurs during the encapsulation process, resulting in low production efficiency. A new approach using high hydrostatic pressure (HHP) was explored in this study to overcome the problem of loading doxorubicin (DOX) in HFn. At the pressure of 500MPa and pH 5.5, DOX molecules were found to be encapsulated into HFn. Meanwhile, combining it with an additive of 20mM arginine completely inhibited precipitation and aggregation, resulting in highly monodispersed nanoparticles with almost 100% protein recovery. Furthermore, stepwise decompression and incubation of the complex in atmospheric pressure at pH 7.4 for another period could further increase the DOX encapsulation ratio. The HFn-DOX nanoparticles (NPs) showed similar morphology and structural features to the hollow cage and no notable drug leakage occurred for HFn-DOX NPs when stored at 4°C and pH 7.4 for two weeks. HFn-DOX NPs prepared through HHP also showed significant cytotoxicity in vitro and higher antitumor bioactivity in vivo than naked DOX. Moreover, This HHP encapsulation strategy could economize on DOX that was greatly wasted during the conventional preparation process simply through a desalting column. These results indicated that HHP could offer a feasible approach with high efficiency for the production of HFn-DOX NPs.

Protein refolding from inclusion bodies (IBs) often encounters a problem of low recovery at high protein concentration. In this study, we demonstrated that high hydrostatic pressure (HHP) could simultaneously achieve high refolding concentration and high refolding yield for IBs of recombinant human ciliary neurotrophic factor (rhCNTF), a potential therapeutic for neurodegenerative diseases. The use of dilution refolding obtained 18% recovery at 3 mg/mL, even in the presence of 4 M urea. In contrast, HHP refolding could efficiently increase the recovery up to almost 100% even at 4 mg/mL. It was found that in the dilution, hydrophobic aggregates were the off-path products and their amount increased with the protein concentration. However, HHP could effectively minimize the formation of hydrophobic aggregates, leading to almost complete conversion of the rhCNTF IBs to the correct configuration. The stable operation range of concentration is 0.5–4.0 mg/mL, in which the refolding yield was almost 100%. Compared with the literatures where HHP failed to increase the refolding yield beyond 90%, the reason could be attributed to the structural difference that rhCNTF has no disulfide bond and is a monomeric protein. After purification by one-step of anionic chromatography, the purity of rhCNTF reached 95% with total process recovery of 54.1%. The purified rhCNTF showed similar structure and in vitro bioactivity to the native species. The whole process featured integration of solubilization/refolding, a high refolding yield of 100%, a high concentration of 4 mg/mL, and a simple chromatography to ensure a high productivity.

Dextran-grafted Protein A affinity chromatographic medium was prepared by grafting dextran to agarose-based matrix, followed by epoxy-activation and Protein A coupling site-directed to sulfhydryl groups of cysteine molecules. An enhancement of both the binding performance and the stability was achieved for this dextran-grafted Protein A chromatographic medium. Its dynamic binding capacity was 61 mg immunoglobulin G/mL suction-dried gel, increased by 24% compared with that of the non-grafted medium. The binding capacity of dextran-grafted medium decreased about 7% after 40 cleaning-in-place cycles, much lower than that of the non-grafted medium as decreased about 15%. Confocal laser scanning microscopy results showed that immunoglobulin G was bound to both the outside and the inside of dextran-grafted medium faster than that of non-grafted one. Atomic force microscopy showed that this dextran-grafted Protein A medium had much rougher surface with a vertical coordinate range of +/- 80 nm, while that of non-grafted one was +/- 10 nm. Grafted dextran provided a more stereo surface morphology and immunoglobulin G molecules were more easily to be bound. This high-performance dextran-grafted Protein A affinity chromatographic medium has promising applications in large-scale antibody purification.

A temperature controlled low field nuclear magnetic resonance (LF-NMR) T2 relaxometry technique based on the mobility changes of water trapped in hydrogels, was successfully used for on-line determination of the sol-gel transition temperature for chitosan/β-glycerophosphate (CS/GP) hydrogels in real time. The LF-NMR results indicated that the gelation temperature decreased gradually with increasing GP concentration, and the results were supported by both thermogravimetric differential scanning calorimetry (DSC) and rheological findings; however, LF-NMR allows non-destructive monitoring of samples during continuous heating. Moreover, as the mobility of water molecules varies greatly during the sol-gel phase transition, the LF-NMR measurement was more sensitive and accurate (RSD ≤ 0.1 %, n = 5) compared with DSC (RSD: 1.2 %–3.7 %, n = 5) and rheology (RSD: 1.1 %–2.3 %, n = 5).

Background To describe the pathological distribution, imaging manifestations, and surgical managements and prognosis of large adrenal tumors (LATs) ≥ 5 cm Methods A total of 251 patients with LATs were analyzed on the basis of pathological or clinical diagnosis. Regarding surgery, open adrenalectomy was performed on 89 patients, and laparoscopic adrenalectomy was performed on 89 patients. Thirty-two patients with bilateral tumors were analyzed in terms of clinical characteristics. The survival rate was determined for 43 patients with adrenal metastases and 29 patients with primary adrenal malignancies. The CT characteristics including tumor diameter, shape, edge, heterogeneity, necrosis, calcification, pre-contrast attenuation, and contrast attenuation were analyzed for 117 patients. Results The majority of LATs were still benign, but they had a higher probability to be malignant. Benign LATs made up 68.13% of all cases, mainly adrenal cysts (19.52%), pheochromocytoma (18.73%), benign adenoma (16.73%), and myelolipoma (7.17%). Malignant LATs accounted for 28.69% of cases, mainly including adrenocortical carcinoma (8.76%) and metastases (17.13%). Laparoscopic surgery was found to involve less trauma than open surgery. It was also safer and postoperative recovery was faster, but it had drawbacks and could not completely replace open surgery. CT features had obvious specificity for the diagnosis of benign and malignant tumors. For example, benign adenomas had a smaller pre-contrast ( Conclusion LATs were more likely to be benign; however, they still had a high probability of being a malignant tumor. Understanding the clinicopathological characteristics of LATs can facilitate selection of more effective clinical treatment options.

Adjuvants are important to enhance the antigens immunogenicity, but may also alter the structures of antigens. Currently off-line methods for adjuvants induced antigen alteration suffer from incomplete release and possible structural alteration of antigens. Here we investigated the differential scanning fluorimetry (DSF) as an in-situ and high-throughput strategy to analyze the stability of inactivated foot-and-mouth disease virus (iFMDV), known as 146S, in three representative adjuvants including aluminum hydroxide (AH), oil-in-water (O/W) emulsion, and water-in-oil (W/O) emulsion. Under optimized DSF conditions, the Tm referring to 146S dissociation can be detected in all three adjuvants. Using SYBR Green II as fluorescent dye enables detection of iFMDV as low as 5 μg/mL. By comparing the Tm in different pH, three adjuvants showed different effects on 146S. Screening for excipients was successfully conducted using DSF. Sugars and glycerol increased the Tm of iFMDV in all three adjuvants, but to different degree. The stabilization by 20% (w/v) sucrose and glycerol was further verified by differential scanning calorimetry (DSC) and high performance size exclusion chromatography (HPSEC). DSF is proved also applicative for low-purity iFMDV and pre-adjuvanted iFMDV vaccines. In summary, the DSF can be a powerful tool in formulation study and vaccine quality control for inactivated virus vaccines.

Increasing evidence has indicated that circular (circ)RNAs participate in carcinogenesis; however, the specific regulatory mechanisms underlying the effects of circRNAs, microRNAs (miRNAs/miRs) and genes on the development of clear cell renal cell carcinoma (CCRCC) remain unclear. In the present study, RNA microarray data from CCRCC tissues and control samples were downloaded from the Gene Expression Omnibus and The Cancer Genome Atlas, in order to identify significantly dysregulated circRNAs, miRNAs and genes. The Cancer-Specific circRNA Database was used to explore the interactions between miRNAs and circRNAs, whereas TargetScan and miRDB were employed to predict the mRNA targets of miRNAs. Functional enrichment and prognostic analyses were conducted in R. The results revealed that 324 circRNAs were downregulated, whereas 218 circRNAs were upregulated in cancer. In addition, a circRNA-miRNA-mRNA interaction network was constructed. Gene Ontology analysis of the upregulated genes revealed that these genes were enriched in biological processes, including ‘flavonoid metabolic process’, ‘cellular glucuronidation’ and ‘T cell activation’. The downregulated genes were mainly enriched in biological processes, such as ‘nephron development’, ‘kidney development’ and ‘renal system development’. The hub genes, including membrane palmitoylated protein 7, aldehyde dehydrogenase 6 family member A1, transcription factor AP-2α, collagen type IV α 4 chain, nuclear receptor subfamily 3 group C member 2, plasminogen, Holliday junction recognition protein, claudin 10, kinesin family member 18B and thyroid hormone receptor β, and the hub miRNAs, including miR-21-3p, miR-155-3p, miR-144-3p, miR-142-5p, miR-875-3p, miR-885-3p, miR-3941, miR-224-3p, miR-584-3p and miR-138-1-3p, were significantly associated with CCRCC survival. In conclusion, these results suggested that the significantly dysregulated circRNAs, miRNAs and genes identified in this study may be considered potential biomarkers of the carcinogenesis of CCRCC and the survival of patients with this disease.

MiR-27b downregulation is significantly associated with tamoxifen resistance in breast cancer cells. However, how it is downregulated in tamoxifen resistant (TamR) breast cancer cells and its downstream regulation were not clear. By performing MSP assay and QRT-PCR analysis with the use of 5-AZA-dC, a DNA methyltransferase inhibitor, we observed that TamR MCF-7 cells had significantly higher levels of methylation in the miR-27b promoter region than tamoxifen sensitive MCF-7 (TamS) cells and demethylation restored miR-27b expression. Re-expression of miR-27b sensitized TamR MCF-7 cells to tamoxifen, inhibited invasion and reversed epithelial-mesenchymal transition (EMT)-like properties. By using bioinformatics analysis and following dual luciferase and western blot analysis, this study confirmed a direct regulation of miR-27b on HMGB3 expression by binding to the 3′UTR. In addition, this study also found that silencing of HMGB3 indeed partially phenocopied the effects of miR-27b in reducing tamoxifen resistance and cell invasion and in reversing EMT-like properties. Therefore, we infer that HMGB3 is a functional target of miR-27b in modulation of tamoxifen resistance and EMT.

A porous structure is the key factor to successful chromatography separation. Agarose gel as one of the most popular porous media has been extensively used in chromatography separation. As the cooling process in the agarose gelation procedure can directly influence the pore structure, ten kinds of 4% agarose media with different cooling rates from 0.132 to 16.7°C/min were synthesized, and the pore structure was determined accurately by using low-field NMR spectroscopy. The curves of pore structure and cooling rate can be divided into two stages with the boundary of 6°C/min. In stage I, the pore structure met a power equation with the decrease of the cooling rate, and in stage II, the process reached a plateau. Confirmatory experiments proved that, by adjusting the cooling rate, a precise control of the pore structure of agarose media can be realized, furthermore, cooling rate optimization was an effective way to control the pore size of agarose media and can further tailor the pore structure for more effective separation of different proteins.

Remote sensing (RS) monitoring of ships has important significance in both military and civilian fields. The RS ship detection aims to locate the position of the ship in the remote sensing image and extract its characteristics. Traditional ship target detection algorithms cannot meet the demands for speed and precision of SAR remote sensing and optical remote sensing data. With the development of artificial intelligence technology, the target detection technology such as deep learning algorithms has made significant progress in RS ship detection. Deep learning has become a heated topic in research. This paper has analyzed and summarized previous researches on the application of deep learning algorithms in ship detection technologies based on SAR and optical remote sensing images in recent years and has provided suggestions for future studies. In the future, deep learning-based technologies for RS ship detection will use more data, such as data from multiple sensors in multiple channels. Deep neural networks will also include more rules and specialized knowledge. Its structure will become more complicated and eventually develop into a neural network like the human brain.

The porous structure characteristics of twenty-one porous microspheres with agarose framework were investigated based on the low-field nuclear magnetic resonance (LF NMR) relaxation time (T 2) distribution measurements. The feasibility of the technique was confirmed by direct relationship between T 2 and the mean pore size of the polymer networks (Rh) of agarose hydrogel which was established using the “Fiber-Cell” model. The expected pore radius distribution curve was obtained and the reliability was validated by comparing them with the results obtained by inverse size-exclusion chromatography. Thereafter, the technique was applied to characterize the pore size distribution (PSD) of soft microspheres and the pore size transformation of microspheres with or without grafted polymers in the process of protein adsorption. All of these results strongly support LF NMR as a promising, rapid and nondestructive technique for the determination of PSDs in many kinds of soft porous microspheres.

In this study, low-cost konjac glucomannan (KGM) microspheres used for desalting were developed by an inverse dispersion method. High concentration of KGM was pretreated to reduce viscosity by acid hydrolysis method under the condition of high temperature and pressure. The selectivity of the obtained cross-linked KGM gels with different degree of crosslinking was studied by constructing calibration curves (K(av)) of standard molecular weight substances. The stability of the gels was investigated, which showed that the KGM microspheres are tolerant to a wide range of pH and stable in all commonly used aqueous buffers, and insensitive to autoclaving as well. Furthermore, protein-desalting performances of GM-1250, a cross-linked KGM microsphere, were evaluated with two proteins, bovine serum albumin and filamentous hemagglutinin, which turned out that GM-1250 is comparable to the widely used commercial product--Sephadex G25 Fine. From economic considerations, KGM gel is a prospective alternative for dextran gels in protein desalting process.
