The development of digital technology has brought great impact and changes to the management practice of Chinese manufacturing enterprises. Improving the digital ability of enterprises, so as to improve the internal and external environment adaptability of enterprises, and enhancing the resilience of enterprises is an urgent consideration for every enterprise. Based on the theory of Dynamic ability and Knowledge management, this paper discusses the internal action mechanism of digital ability (digital technology application ability and digital resource integration ability) on organizational resilience, and verifies the intermediary role of Ambidextrous innovation and the regulatory role of knowledge management. This paper uses a sample of 400 senior managers of Chinese manufacturing enterprises to conduct empirical research, and the results show that: number Positive relationship between digital resource integration and organizational resilience; digital capability positively affects Ambidextrous innovation; Ambidextrous innovation partially mediates digital capability and organizational resilience; and knowledge management mechanism positively regulates the relationship between digital capability and organizational resilience. BACKGROUND: Denosumab entered the Chinese market for the first time in 2020. Since it is a short period of time, there is a lack of data on its effectiveness and safety in Chinese people. The objective of this study was to evaluate the effectiveness and safety of denosumab in delaying skeletal-related events (SREs) in patients with breast cancer metastatic to bone. METHODS: The study retrospectively analyzed data from breast cancer patients with bone metastases (BM) who were treated with denosumab in the First Affiliated Hospital of Nanjing Medical University from September 2020 to January 2022. The primary endpoint was SRE incidence at 1 year after receiving denosumab treatment. The secondary endpoints included time to first on-study SRE and safety. Descriptive analysis was utilized to display clinicopathological features. The Kaplan-Meier method was used to estimate the median time to first onstudy SRE in total population and subgroups. Logistic regression analysis and χ² test were employed to determine the potential factors influencing the occurrence of SREs. RESULTS: Fifty breast cancer patients with BM were enrolled in our study, and 54.0% of the patients had 5 or more metastatic bone lesions. After a median follow-up of 17.00 months, 24% of the patients developed SREs at 1 year after receiving denosumab treatment, and the median time to first on-study SREs was not reached. Five or more metastatic bone lesions were an independent risk factor for SRE occurrence (odds ratio = 6.06, 95% CI: 1.09-33.54, P = .039). The adverse events (AEs) associated with denosumab mainly included hypocalcemia (68.0%), periodontitis (28.0%), and myalgia (14.0%). Only 3 cases of grade III/IV AEs were reported, and no serious AEs occurred. CONCLUSION: Denosumab was effective and well tolerated in Chinese breast cancer patients with BM. [ABSTRACT FROM AUTHOR]

Immune-stimulatory ligands, such as major histocompatibility complex molecules and the T-cell costimulatory ligand CD86, are central to productive immunity. Endogenous mammalian membrane-associated RING-CHs (MARCH) act on these and other targets to regulate antigen presentation and activation of adaptive immunity, whereas virus-encoded homologs target the same molecules to evade immune responses. Substrate specificity is encoded in or near the membraneembedded domains of MARCHs and the proteins they regulate, but the exact sequences that distinguish substrates from nonsubstrates are poorly understood. Here, we examined the requirements for recognition of the costimulatory ligand CD86 by two different MARCH-family proteins, human MARCH1 and Kaposi's sarcoma herpesvirus modulator of immune recognition 2 (MIR2), using deep mutational scanning. We identified a highly specific recognition surface in the hydrophobic core of the CD86 transmembrane (TM) domain (TMD) that is required for recognition by MARCH1 and prominently features a proline at position 254. In contrast, MIR2 requires no specific sequences in the CD86 TMD but relies primarily on an aspartic acid at position 244 in the CD86 extracellular juxtamembrane region. Surprisingly, MIR2 recognized CD86 with a TMD composed entirely of valine, whereas many different single amino acid substitutions in the context of the native TM sequence conferred MIR2 resistance. These results show that the human and viral proteins evolved completely different recognition modes for the same substrate. That some TM sequences are incompatible with MIR2 activity, even when no specific recognition motif is required, suggests a more complicated mechanism of immune modulation via CD86 than was previously appreciated. Objective(s): Previously we reported functional leukocyte immunoglobulin-like receptor A3 (LILRA3) leads to susceptibility and sub-phenotypes of several autoimmune diseases. LILRA3 levels in blood serum and CD14+ monocytes enhanced in systemic lupus erythematosus and resulted in disease severity. However, the mechanism of LILRA3 in the pathogenesis of autoimmunity remains elusive. This study aims to explore the potential impact of LILRA3 on the differentiation, maturation, and function of monocyte-derived DCs (MoDCs). Materials and Methods: The human monocytic cell line (THP-1) was cultured to derive MoDCs in vitro. We performed plasmid transfection to examine the impact of LILRA3 on monocyte differentiation. Surface markers on MoDCs were measured using FACS. To assess the function of mature MoDCs, IL- 12p70, IFN-γ and IL-4 levels were detected after the mixed leucocyte response by enzyme-linked immunosorbent assay. Western blot assay was employed in this study to determine the signaling pathways in MoDCs activation. Results: LILRA3 promotes MoDCs maturation, our results showed significant up-regulation of CD40, CD80, CD86, CD209, and HLA-DR and increased production of pro-inflammatory cytokine IL-12. LILRA3-treated MoDCs exhibited a robust proliferation of allogeneic CD4+ T cells and induced naive CD4+ T cell polarization into the Th1 phenotype. Furthermore, the preceding activation of MoDCs maturation and LILRA3 function might be attributed to p38 MAPK and STAT1 signaling pathway's aberrant activation. Conclusion: This is the first study to report that LILRA3 played a critical role in promoting MoDCs maturation and directing MoDCs to modulate Th1 cell differentiation, which may have a role in the pathogenesis of autoimmune diseases. In this study, the authors assess the performance of a cognitive multiple-input single-output (MISO) network, where the secondary base station schedules a secondary destination Dk* to communicate from K candidates, by employing scheduled maximal ratio transmission (MRT) scheme. A practical scenario is considered where imperfect channel state information (CSI) is taken into account. Specially, assuming Rayleigh fading channels, the exact expression for the cumulative distribution function of the received signal-to-noise ratio (SNR) at Dk* is derived, from which the outage probability (OP) at Dk* is obtained. An asymptotic expression for the OP at high SNR region is also presented, indicating the diversity gain that the scheduled MRT scheme can achieve. Furthermore, the interference OP at the primary receiver is discussed due to imperfect CSI in the link from base station to primary receiver link. Multiple cascaded hydropower reservoir systems serving the same area need to cooperate to increase overall benefits and reliability. To meet system demand while balancing among cascaded systems, both centralized and distributed five segment energy available based operating rules, making power decisions for subsystems and the whole system, are proposed for cooperation across multiple cascaded hydropower reservoirs. Unlike existing methods, the cooperative operating rules are optimized to the global objective of maximizing the minimum system power production, with constraints on local cascade minimum energy generation to confine the possible local profit earnings or losses because of cooperation, making the operating rules more acceptable for cooperative operations among local cascade system agents. The operating rule optimization models are solved using a genetic algorithm. A case study for three cascaded systems in southwest China shows that distributed and centralized cooperative operating rules can increase minimum total power by more than 3,600 MW and 4,700 MW, respectively, in the dry season with little energy changes in each cascaded system, taking advantages of reservoir storages and different timing of inflows. The centralized rule is superior to the distributed rule for increasing minimum power, although the distributed rule is more effective to control the energy for each cascaded system. Gait phase is widely used for gait trajectory generation, gait control and gait evaluation on lower-limb exoskeletons. So far, a variety of methods have been developed to identify the gait phase for lower-limb exoskeletons. Angular sensors on lower-limb exoskeletons are essential for joint closed-loop controlling; however, other types of sensors, such as plantar pressure, attitude or inertial measurement unit, are not indispensable. Therefore, to make full use of existing sensors, we propose a novel gait phase recognition method for lower-limb exoskeletons using only joint angular sensors. The method consists of two procedures. Firstly, the gait deviation distances during walking are calculated and classified by Fisher's linear discriminant method, and one gait cycle is divided into eight gait phases. The validity of the classification results is also verified based on large gait samples. Secondly, we build a gait phase recognition model based on multilayer perceptron and train it with the phase-labeled gait data. The experimental result of cross-validation shows that the model has a 94.45% average correct rate of set (CRS) and an 87.22% average correct rate of phase (CRP) on the testing set, and it can predict the gait phase accurately. Background: Streptococcus mutans is the primary etiological agent of human dental caries. It can metabolize a wide variety of carbohydrates and produce large amounts of organic acids that cause enamel demineralization. Phosphoenolpyruvate-dependent sugar phosphotransferase system (PTS) plays an important role in carbohydrates uptake of S. mutans. The ptxA and ptxB genes in S. mutans encode putative enzyme IIA and enzyme IIB of the L-ascorbate-specific PTS. The aim of this study was to analyze the function of these proteins and understand the transcriptional regulatory mechanism. Results: ptxA-, ptxB-, as well as ptxA-, ptxB- double-deletion mutants all had more extended lag phase and lower growth yield than wild-type strain UA159 when grown in the medium using L-ascorbate as the sole carbon source. Acid production and acid killing assays showed that the absence of the ptxA and ptxB genes resulted in a reduction in the capacity for acidogenesis, and all three mutant strains did not survive an acid shock. According to biofilm and extracellular polysaccharides (EPS) formation analysis, all the mutant strains formed much less prolific biofilms with small amounts of EPS than wild-type UA159 when using L-ascorbate as the sole carbon source. Moreover, PCR analysis and quantitative real-time PCR revealed that sgaT, ptxA, ptxB, SMU.273, SMU.274 and SMU.275 appear to be parts of the same operon. The transcription levels of these genes were all elevated in the presence of L-ascorbate, and the expression of ptxA gene decreased significantly once ptxB gene was knockout. Conclusions: The ptxA and ptxB genes are involved in the growth, aciduricity, acidogenesis, and formation of biofilms and EPS of S. mutans when L-ascorbate is the sole carbon source. In addition, the expression of ptxA is regulated by ptxB. ptxA, ptxB, and the upstream gene sgaT, the downstream genes SMU.273, SMU.274 and SMU.275 appear to be parts of the same operon, and L-ascorbate is a potential inducer of the operon. So far, the role of Ether à go-go 1 (Eag1) potassium channels in migration and invasion progression of cancers remains elusive. In the present study, the effects of Eag1 knockdown on osteosarcoma cell proliferation, growth, and apoptosis were examined.Then, we evaluated the effects of Eag1 silencing on osteosarcoma cell migration and invasion. In addition, we detected the expression of vascular endothelial growth factor (VEGF) and signal transducer and activator of transcription 3 (STAT3) in osteosarcoma cell treated with Eag1 small interfering RNAs (siRNAs). Finally, STAT3 siRNA was employed to determine the influence of downregulation of STAT3 on cell proliferation and migration. The results showed that knockdown of Eag1 significantly suppressed osteosarcoma cell proliferation and osteosarcoma xenografts growth. However, Eag1 silencing had little effect on cell apoptosis. Additionally, osteosarcomacell adhesion, migration, and invasionwere also potently attenuated.Notably, the expression levels ofVEGFdecreased evidently upon Eag1 siRNAs treatment, paralleled with reductions in the expression levels of STAT3. Moreover, a similar pattern was observed in osteosarcoma cell proliferation and migration suppression between STAT3 siRNA and Eag1 siRNAs groups. A multiobjective model with chance constraints is proposed to obtain hedging rules for hydropower reservoirs based on initial reservoir operating rules derived using stochastic dynamic programming. In the model, hedging rules are evaluated through simulations and the maximizations of minimum powers in simulation horizon of hydropower reservoirs are taken as objectives. The maximum-minimum objectives are substituted by aggregated functions. A genetic algorithm is used to solve the model, with a fuzzy optimal method evaluating the individuals. A case study shows that the proposed approach is feasible and efficient to produce a series of hedging rules with different reliability levels, and valuable to balance the magnitudes of generation below firm power and the chances of such events. Wireless Sensor and Actor Networks (WSANs) refer to a group of sensors and actors linked by wireless medium to probe environment and perform specific actions. Such certain actions should always be taken before a deadline when an event of interest is detected. In order to provide such services, the whole monitor area is divided into several virtual areas and nodes in the same area form a cluster. Clustering of the WSANs is often pursued to give that each actor acts as a cluster-head. The number of actors is related to the size and the deployment of WSANs cluster. In this paper, we find a method to determine the accurate number of actors which enables them to receive data and take actions in an imposed time-delay. The k-MinTE and the k-MaxTE clustering algorithm are proposed to form the minimum and maximum size of cluster, respectively. In those clustering algorithms, actors are deployed in such a way that sensors could route data to actors within k hops. Then, clusters are arranged by the regular hexagon. Mouse models of prostate cancer (PCa) are critical for understanding the biology of PCa initiation, progression, and treatment modalities. Here, we summarize recent advances in PCa mouse models that led to new insights into specific gene functions in PCa. For example, the study of transgenic mice with TMPRSS2/ERG, an androgen-regulated fusion protein, revealed its role in developing PCa precursor lesions, prostate intraepithelial neoplasia; however, it is not sufficient for PCa development. Double deficiency of Pten and Smad4 leads to a high incidence of metastatic PCa. Targeted deletion of Pten in castration-resistant Nkx3-1-expressing cells results in rapid carcinoma formation after androgen-mediated regeneration, indicating that progenitor cells with luminal characteristics can play a role in initiation of PCa. Transgenic mice with activated oncogenes, growth factors, and steroid hormone receptors or inactivated tumor suppressors continue to provide insights into disease progression from initiation to metastasis. Further development of new PCa models with spatial and temporal regulation of candidate gene expression will probably enhance our understanding of the complex events that lead to PCa initiation and progression, thereby invoking novel strategies to combat this common disease in men. Ether à go-go 1 (Eag1) channel is overexpressed in a variety of cancers but the therapeutic potential of Eag1 in osteosarcoma remains elusive. In this study, we constructed an Ad5-Eag1-shRNA vector and evaluated its efficiency for Eag1 knockdown and its effects on osteosarcoma. Our results showed that Ad5-Eag1-shRNA had high interference efficiency of Eag1 expression and suppressed osteosarcoma growth both in vitro and in vivo. To explore the molecular mechanism underlying tumor growth inhibition induced by Eag1 silencing, the intratumoral microvessel density (MVD) was assessed by CD31 staining and the expression of vascular endothelial growth factor (VEGF) was detected by Western blot analysis. We found that Eag1 silencing led to decreased angiogenesis and VEGF expression in the xenograft model of osteosarcoma. Finally, we detected a time-dependent decrease in VEGF expression and considerably reduced phosphoinositide 3-kinase (PI3K) and protein kinase B (AKT) activation in osteosarcoma cells treated by Eag1 shRNA. Taken together, our results suggest that Eag1 silencing inhibits tumor growth and angiogenesis in osteosarcoma via the down regulation of VEGF/PI3K/AKT signaling. [ABSTRACT FROM AUTHOR]

Objectives: Dendritic cell immunoreceptor (DCIR) has been implicated in development of autoimmune disorders in rodent and DCIR polymorphisms were associated with anti-citrullinated proteins antibodies (ACPA)-negative rheumatoid arthritis (RA) in Swedish Caucasians. This study was undertaken to further investigate whether DCIR polymorphisms are also risk factors for the development of RA in four Asian populations originated from China and Malaysia. Methods: We genotyped two DCIR SNPs rs2377422 and rs10840759 in Han Chinese population (1,193 cases, 1,278 controls), to assess their association with RA. Subsequently, rs2377422 was further genotyped in three independent cohorts of Malaysian-Chinese subjects (MY•Chinese, 254 cases, 206 controls), Malay subjects (MY•Malay, 515 cases, 986 controls), and Malaysian-Indian subjects (MY•Indian, 378 cases, 285 controls), to seek confirmation of association in various ethnic groups. Meta-analysis was preformed to evaluate the contribution of rs2377422 polymorphisms to the development of ACPAnegative RA in distinct ethnic groups. Finally, we carried out association analysis of rs2377422 polymorphisms with DCIR mRNA expression levels. Results: DCIR rs2377422 was found to be significantly associated with ACPA -negative RA in Han Chinese (OR 1.92, 95% CI 1.27-2.90, P = 0.0020). Meta-analysis confirms DCIR rs2377422 as a risk factor for ACPA-negative RA across distinct ethnic groups (ORoverall = 1.17, 95% CI 1.06-1.30, P = 0.003). The SNP rs2377422 polymorphism showed significant association with DCIR mRNA expression level, i.e. RA-risk CC genotype exhibit a significant increase in the expression of DCIR (P = 0.0023, Kruskal-Wallis). Conclusions: Our data provide evidence for association between DCIR rs2377422 and RA in non-Caucasian populations and confirm the influence of DCIR polymorphisms on RA susceptibility, especially on ACPA-negative RA. Background: During normal development in human and other placental mammals, the embryonic cloacal cavity separates along the axial longitudinal plane to give rise to the urethral system, ventrally, and the rectum, dorsally. Defects in cloacal development are very common and present clinically as a rectourethral fistula in about 1 in 5,000 live human births. Yet, the cellular mechanisms of cloacal septation remain poorly understood. Methodology/Principal Findings: We previously detected Bone morphogenetic protein 7 (Bmp7) expression in the urorectal mesenchyme (URM), and have shown that loss of Bmp7 function results in the arrest of cloacal septation. Here, we present evidence that cloacal partitioning is driven by Bmp7 signaling in the cloacal endoderm. We performed TUNEL and immunofluorescent analysis on cloacal sections from Bmp7 null and control littermate embryos. We found that loss of Bmp7 results in a dramatic decrease in the endoderm survival and a delay in differentiation. We used immunological methods to show that Bmp7 functions by activating the c-Jun N-terminal kinase (JNK) pathway. We carried out confocal and 3D imaging analysis of mitotic chromosome bundles to show that during normal septation cells in the cloacal endoderm divide predominantly in the apical-basal direction. Loss of Bmp7/JNK signaling results in randomization of mitotic angles in the cloacal endoderm. We also conducted immunohistochemical analysis of human fetal sections to show that BMP/phospho- SMAD and JNK pathways function in the human cloacal region similar as in the mouse. Conclusion/Significance: Our results strongly indicate that Bmp7/JNK signaling regulates remodeling of the cloacal endoderm resulting in a topological separation of the urinary and digestive systems. Electric power transmission lines in service in southwest China have fallen far behind the power sources. With rapid development of hydropower in this area, severe water spills for hydropower frequently occur during the flood season because of limited transmission capacity. Effective coordinated operation among large and small hydropower plants (LHPPs and SHPPs) in the area is a feasible approach for alleviating this problem. Using the Yunnan Power Grid (YNPG) of southwest China as an example, a long-term coordinated optimal operation model (COOM) for an integrated system of LHPPs and SHPPs is proposed in this paper. Total electricity that can be consumed or transmitted is defined as electricity absorption. Maximizing the total electricity absorption, which implies balancing high average water heads with reservoir spills, was selected as the objective. Satisfying multiple voltage transmission constraints is a critical issue. A progressive optimality algorithm (POA) was used to solve the model based on a multidimensional search in two-stage optimizations. Experimental investigations were performed on the single-phase flow and heat transfer characteristics through the silicon-based trapezoidal microchannels with a hydraulic diameter of 194.5 um using Al2O3-H2O nanofluids with particle volume fractions of 0, 0.15% and 0.26% as the working fluids. The effects of the Reynolds number, Prandtl number and nanoparticle concentration on the pressure drop and convective heat transfer were investigated. Experimental results show that the pressure drop and flow friction of the nanofluids increased slightly when compared with that of the pure water, while the Nusselt number increased considerably. At the same pumping power, using nanofluids instead of pure water caused a reduction in the thermal resistance. It was also found that the Nusselt number increased with the increase in the particle concentration, Reynolds number and Prandtl number. Based on the experimental data, the dimensionless correlations for the flow friction and heat transfer of Al2O3-H2O nanofluids through silicon microchannels were proposed for the first time. The agglomeration and deposition of nanoparticles in the silicon microchannels were also examined in this paper. It was found that the Al2O3 nanoparticles deposited on the inner wall of microchannels more easily with increasing wall temperature, and once boiling commenced, there is a severe deposition and adhesion of nanoparticles to the inner wall, which makes the boiling heat transfer of nanofluids in silicon microchannels questionable. An experimental investigation was performed on the flow friction and convective heat transfer characteristics of the ethanol-water solutions flowing through five sets of trapezoidal silicon microchannels having hydraulic diameters ranging from 141.7 um to 268.6 um. Four kinds of ethanol-water solutions with the ethanol volume concentrations ranging from 0 to 0.8 were tested under different flow and heating conditions. It was found that the cross-sectional geometric parameters had great effect on the flow friction and heat transfer, and the microchannels with a larger Wb/Wt (bottom width-to-top width ratio) and a smaller H/Wt (depth-to-top width ratio) usually had a larger friction constant and a higher Nusselt number. Entrance effects were significant for the flow friction and heat transfer in silicon microchannels, and decreased with the increase of dimensionless hydrodynamic length L and dimensionless thermal length L+h. When L > 1.0, the hydrodynamic entrance effect on the flow friction was ignorable. For the developed laminar flow in silicon microchannels, the Navier-Stokes equation was applicable. It was also found that the volume concentrations had different effects on the flow friction and heat transfer. Within the experimental range, the effect of volume concentrations on the flow friction was ignorable, and the friction constants of the ethanol-water solutions having different concentrations were the same as those of the pure water. However, volume concentrations had great effect on the convection heat transfer in silicon microchannels. With the increase of the volume concentrations, the Nusselt number of the ethanol-water solutions increased obviously, which was attributed to the combination effect of the increase in the Prantdtl number as well as the volatilization effect of the ethanol. Based on the experimental data, the dimensionless correlations for the flow friction and heat transfer of the ethanol-water solutions in the silicon microchannels in a function of the cross-sectional geometrical parameters, entrance effect and volume concentrations were proposed for the first time. In this paper, we present a novel approach for video-anomaly detection in crowded and complicated scenes. The proposed approach detects anomalies based on a hierarchical activity-pattern discovery framework, comprehensively considering both global and local spatio-temporal contexts. The discovery is a coarse-to-fine learning process with unsupervised methods for automatically constructing normal activity patterns at different levels. A unified anomaly energy function is designed based on these discovered activity patterns to identify the abnormal level of an input motion pattern. Deregulation of voltage-gated potassium channel subunit Kv1.3 has been reported in many tumors. Kv1.3 promotes tumorigenesis by enhancing cell proliferation while suppressing apoptosis. However, the expression and function of Kv1.3 in osteosarcoma are unknown. In the present study, we detected the expression of Kv1.3 in human osteosarcoma cells and tissues by RT-PCR, Western blot and immunohistochemistry. We further examined cell proliferation and apoptosis in osteosarcoma MG-63 cells and xenografts following knockdown of Kv1.3 by short hairpin RNA (shRNA). We found that Kv1.3 was upregulated in human osteosarcoma. Knockdown of Kv1.3 significantly suppressed cell proliferation and increased apoptosis as demonstrated by enhanced cleavage of poly (ADP-ribose) polymerase (PARP) and the activation of Caspase-3/7. Furthermore, adenovirus delivered shRNA targeting Kv1.3 significantly inhibited the growth of MG-63 xenografts. Plin4 is a lipid droplet protein (LDP) found predominantly in white adipose tissue (WAT). The Plin4 gene is immediately downstream of the Plin5 gene; the two genes exhibit distinct though overlapping tissue expression patterns. Plin4 is absent in brown adipose tissue (BAT) and liver and expressed at low levels in heart and skeletal muscle, whereas Plin5 is highly expressed in these oxidative tissues but at a low level in WAT. The physiological role of Plin4 remains unclear. We have generated Plin4-1- mice by gene targeting. Loss of Plin4 has no effect on body weight or composition or on adipose mass or development. However, the triacylglycerol (TAG) content in heart, but not other oxidative tissues such as BAT, soleus muscle, and liver, is markedly reduced in Plin4-1- mice. The heart of Plin4-1- mice displays reduced Plin5 mRNA and protein levels (by ~38 and 87%, respectively, vs. wild-type) but unchanged mRNA levels of other perilipin family genes (Plin2 and Plin3) or genes involved in glucose and lipid metabolism. Despite reduced cardiac TAG level, both young and aged Plin4-1- mice maintain normal heart function as wild-type mice, as measured by echocardiography. Interestingly, Plin4 deficiency prevents the lipid accumulation in the heart that normally occurs after a prolonged (48-h) fast. It also protects the heart from cardiac steatosis induced by high-fat diet or when Plin4-1- mice are bred into Lep-1- obese background. This paper presents a novel low-rank affinity based local-driven algorithm to robustly propagate the multilabels from training images to test images. A graph is constructed over the segmented local image regions. The labels for vertices from the training data are derived based on the context among different training images, and the derived vertex labels are propagated to the unlabeled vertices via the graph. The multitask low-rank affinity, which jointly seeks the sparsity-consistent low-rank affinities from multiple featurematrices, is applied to compute the edge weights between graph vertices. The inference process of multitask low-rank affinity is formulated as a constrained nuclear norm and l2,1-norm minimization problem. The optimization is conducted efficiently with the augmented Lagrange multiplier method. Based on the learned local patch labels we can predict the multilabels for the test images. The indoor environment has brought new challenges for micro Unmanned Aerial Vehicles (UAVs) in terms of their being able to execute tasks with high positioning accuracy. Conventional positioning methods based on GPS are unreliable, although certain circumstances of limited space make it possible to apply new technologies. In this paper, we propose a novel indoor self-positioning system of UAV based on a heterogeneous sensing system, which integrates data from a structured light scanner, ultra-wideband (UWB), and an inertial navigation system (INS).We made the structured light scanner, which is composed of a low-cost structured light and camera, ourselves to improve the positioning accuracy at a specified area. We applied adaptive Kalman filtering to fuse the data from the INS and UWB while the vehicle was moving, as well as Gauss filtering to fuse the data from the UWB and the structured light scanner in a hovering state. The results of our simulations and experiments demonstrate that the proposed strategy significantly improves positioning accuracy in motion and also in the hovering state, as compared to using a single sensor. For the power systems, for which few data are available for mid-term electricity market clearing price (MCP) forecasting at the early stage of market reform, a novel grey prediction model (defined as interval GM(0, N) model) is proposed in this paper. Over the traditional GM(0, N) model, three major improvements of the proposed model are: (i) the lower and upper bounds are firstly identified to give an interval estimation of the forecasting value; (ii) a novel whitenization method is then established to determine the definite forecasting value from the forecasting interval; and (iii) the model parameters are identified by an improved particle swarm optimization (PSO) instead of the least square method (LSM) for the limitation of LSM. Finally, a newly-reformed electricity market in Yunnan province of China is studied, and input variables are contrapuntally selected. The accuracy of the proposed model is validated by observed data. Compared with the multiple linear regression (MLR) model, the traditional GM(0, N) model and the artificial neural network (ANN) model, the proposed model gives a better performance and its superiority is further ensured by the use of the modified Diebold-Mariano (MDM) test, suggesting that it is suitable for mid-term electricity MCP forecasting in a data-sparse electricity market. In this paper, we propose a biomechatronic design of an anthropomorphic artificial hand that is able to mimic the natural motion of human fingers. The prosthetic hand has 5 fingers and 15 joints, which are actuated by 5 embedded motors. Each finger has three phalanges that can fulfill flexion-extension movements independently. The thumb is specially designed to move along a cone surface when grasping, and the other four fingers are well developed based on the four-bar link mechanism to imitate the motion of the human finger. To accomplish the sophisticated control schemes, the fingers are equipped with numerous torque and position sensors. The mechanical parts, sensors, and motion control systems are integrated in the hand structure, and the motion of the hand can be controlled through electromyography (EMG) signals in real-time. A new concept for the sensory feedback system based on an electrical stimulator is also taken into account. The low-cost prosthetic hand is small in size (85% of the human hand), of low weight (420 g) and has a large grasp power (10 N on the fingertips), hence it has a dexterous and humanlike appearance. Kv1.5 (also known as KCNA5) is a protein encoded by the KCNA5 gene, which belongs to the voltage-gated potassium channel, shaker-related subfamily. Recently, a number of studies have suggested that Kv1.5 is overexpressed in numerous cancers and plays crucial roles in cancer development. However, until now, the expression and functions of Kv1.5 in osteosarcoma are still unclear. To characterize the potential biological functions of Kv1.5 in osteosarcoma, herein, we examined the expression levels of Kv1.5 in osteosarcoma cells and tissues using quantitative real-time polymerase chain reaction (qRT-PCR), western blot, and immunohistochemistry assays. Four short hairpin RNAs (shRNAs) targeting Kv1.5 were designed and homologous recombination technology was used to construct pGeneSil-Kv1.5 vectors. In addition, the vectors were transfected into osteosarcoma MG63 cells and Kv1.5 mRNA level was measured by qRT-PCR and the Kv1.5 protein level was examined by western blot. We also examined the effects of Kv1.5 silencing on proliferation, cell cycle and apoptosis of the osteosarcoma cells using CCK-8, colony formation, flow cytometry and terminal deoxynucleotidyl transferase dUTP nick end labeling (TUNEL) assays. Our results showed that Kv1.5 was aberrantly expressed in osteosarcoma and that the synthesized shRNA targeting Kv1.5 reduced Kv1.5 mRNA and protein expression effectively. Silencing Kv1.5 expression in the osteosarcoma cells significantly inhibited the proliferation of osteosarcoma cells, induced cell cycle arrest at G0/G1 phase, and induced cell apoptosis through up-regulation of p21, p27, Bax, Bcl-XL and caspase-3 and down-regulation of cyclins A, cyclins D1, cyclins E, Bcl-2 and Bik. In summary, our results indicate that Kv1.5 silencing could suppress osteosarcoma progression through multiple signaling pathways and suggest that Kv1.5 may be a novel target for osteosarcoma therapeutics. Coal-fired thermal power plants, which represent the largest proportion of China's electric power system, are very sluggish in responding to power system load demands. Thus, a reasonable and feasible scheme for the medium-term optimal commitment of thermal units (MOCTU) can ensure that the generation process runs smoothly and minimizes the start-up and shut-down times of thermal units. In this paper, based on the real-world and practical demands of power dispatch centers in China, a flexible mathematical model for MOCTU that uses equal utilization hours for the installed capacity of all thermal power plants as the optimization goal and that considers the award hours for MOCTU is developed. MOCTU is a unit commitment (UC) problem with characteristics of large-scale, high dimensions and nonlinearity. For optimization, an improved progressive optimality algorithm (IPOA) offering the advantages of POA is adopted to overcome the drawback of POA of easily falling into the local optima. In the optimization process, strategies of system operating capacity equalization and single station operating peak combination are introduced to move the target solution from the boundary constraints along the target isopleths into the feasible solution's interior to guarantee the global optima. The results of a case study consisting of nine thermal power plants with 27 units show that the presented algorithm can obtain an optimal solution and is competent in solving the MOCTU with high efficiency and accuracy as well as that the developed simulation model can be applied to practical engineering needs. Real-time stereo matching in image sequences is important in video monitoring, robotic navigation and intelligent vehicle, etc. Spatiotemporal stereo and scene flow can be used to produce temporally coherent disparity of dynamic scenes. However, most methods do not use the previous disparity map sufficiently to compute the current one. Thus, the disparity range limits the speed of disparity computation for each stereo pair. This paper integrates the temporal information into the stereo computation, and presents the relationship between consecutive disparity maps, which makes the disparity prediction reasonable. The scheme can produce a sequence of temporally coherent disparity maps rapidly. The tests performed on simulated and real stereo sequences confirm the validity of our approach. Androgen receptor (AR), a steroid hormone receptor, is critical for prostate cancer growth. However, activation of AR by androgens can also lead to growth suppression and differentiation. Transcriptional cofactors play an important role in this switch between proliferative and anti-proliferative AR target gene programs. Transducin b-like-related protein 1 (TBLR1), a core component of the nuclear receptor corepressor complex, shows both corepressor and coactivator activities on nuclear receptors, but little is known about its effects on AR and prostate cancer. We characterized TBLR1 as a coactivator of AR in prostate cancer cells and determined that the activation is dependent on both phosphorylation and 19S proteosome. We showed that TBLR1 physically interacts with AR and directly occupies the androgen-response elements of the affected AR target genes in an androgen-dependent manner. TBLR1 is primarily localized in the nucleus in benign prostate cells and nuclear expression is significantly reduced in prostate cancer cells in culture. Similarly, in human tumor samples, the expression of TBLR1 in the nucleus is significantly reduced in the malignant glands compared with the surrounding benign prostatic glands (P<0.005). Stable ectopic expression of nuclear TBLR1 leads to androgen-dependent growth suppression of prostate cancer cells in vitro and in vivo by selective activation of androgenregulated genes associated with differentiation (e.g. KRT18) and growth suppression (e.g. NKX3-1), but not cell proliferation of the prostate cancer. Understanding the molecular switches involved in the transition from AR-dependent growth promotion to AR-dependent growth suppression will lead to more successful treatments for prostate cancer. Hormones, including estrogen and progesterone, and their receptors play an important role in the development and progression of ovarian carcinoma. Androgen, its receptor and coactivators have also been implicated in these processes. p44/Mep50/WDR77 was identified as a subunit of the methylosome complex and lately characterized as a steroid receptor coactivator that enhances androgen receptor as well as estrogen receptor-mediated transcriptional activity in a liganddependent manner. We previously described distinct expression and function of p44 in prostate, testis, and breast cancers. In this report, we examined the expression and function of p44 in ovarian cancer. In contrast to findings in prostate and testicular cancer and similar to breast cancer, p44 shows strong cytoplasmic localization in morphologically normal ovarian surface and fallopian tube epithelia, while nuclear p44 is observed in invasive ovarian carcinoma. We observed that p44 can serve as a coactivator of both androgen receptor (AR) and estrogen receptor (ER) in ovarian cells. Further, overexpression of nuclear-localized p44 stimulates proliferation and invasion in ovarian cancer cells in the presence of estrogen or androgen. These findings strongly suggest that p44 plays a role in mediating the effects of hormones during ovarian tumorigenesis. [ABSTRACT FROM AUTHOR]

Gender is an important cue in social activities. In this correspondence, we present a study and analysis of gender classification based on human gait. Psychological experiments were carried out. These experiments showed that humans can recognize gender based on gait information, and that contributions of different body components vary. The prior knowledge extracted from the psychological experiments can be combined with an automatic method to further improve classification accuracy. The proposed method which combines human knowledge achieves higher performance than some other methods, and is even more accurate than human observers. We also present a numerical analysis of the contributions of different human components, which shows that head and hair, back, chest and thigh are more discriminative than other components. We also did challenging cross-race experiments that used Asian gait data to classify the gender of Europeans, and vice versa. Encouraging results were obtained. All the above prove that gait-based gender classification is feasible in controlled environments. In real applications, it still suffers from many difficulties, such as view variation, clothing and shoes changes, or carrying objects.
