The World Health Organization (WHO) has identified antimicrobial resistance (AMR) as one of the top three global dangers to public health. One of the most vital factors contributing to the high prevalence of AMR is the misuse/overuse of antibiotics for treatment and/or as a growth promoter in the food industry. AMR can be transmitted to humans via food, the environment, or other channels through horizontal gene transfer. Therefore, efficient methods are urgently needed to determine whether bacteria are resistant to antibiotics. This work provides a review of the advances in machine learning (ML) techniques for predicting and identifying AMR in foodborne pathogens. We also emphasize the groundbreaking potential of whole genome sequencing (WGS) and spectroscopy technologies combined with ML in the context of AMR detection. These offer enormous potential because of their unique characteristics, which can overcome inherent limits in existing detection approaches.

Antibiotics are commonly used to address animal health issues and enhance overall productivity in animal agriculture. However, overuse and misuse of antibiotics has become a concern in the field of agriculture and pose potential food safety risks. Efficiently controlling antibiotic resistance in animal agriculture is of vital importance. Various technologies have been developed and employed to prevent and control antibiotic-resistant bacteria in this field, such as ozone sterilization technology, among others. This review provides a comprehensive overview of current sterilization technologies in animal agriculture, including chemical sanitizers, plasma oxidation, bacteriophage, ozone, ultraviolet, ultrasonic irradiation, and photodynamic inactivation. Furthermore, we present novel insights and future perspectives on the prevention and control of antibiotic resistance in animal agriculture.

Salmonella is one of the leading foodborne pathogens that cause gastroenteritis. It is imperative to monitor Salmonella contamination within poultry farms. This study focuses on the prevalence of Salmonella in composting manure at poultry farms a potential and persistent contamination sources. A batch of manure samples (n = 10) were collected from a local poultry farm, and half of the manure samples were detected to be Salmonella-positive. Serotyping results showed four serotypes among different isolated strains, including Typhimurium, Typhi, Newport, and Dublin. Isolates from manure generally demonstrated a high ciprofloxacin resistance and biofilm formation capacity. Multi-drug resistance was found in isolates from organic manure that had been fermented using high-temperature aerobic technology. Our study indicated that composting manure harboured Salmonella and might facilitate the transmission of Salmonella and the development of antimicrobial resistance. Proper composting strategies are needed to reduce the transmission of antimicrobial resistance and Salmonella.

Background: Although food-grade disinfectants are extensively used worldwide, it has been reported that the long-term exposure of bacteria to these compounds may represent a selective force inducing evolution including the emergence of antibiotic resistance. However, the mechanism underlying this correlation has not been elucidated. This study aims to investigate the genomic evolution caused by long-term disinfectant exposure in terms of antibiotic resistance in Salmonella enterica Typhimurium. Methods: S. Typhimurium isolates were exposed to increasing concentrations of benzalkonium chloride (BAC) and variations of their antibiotic susceptibilities were monitored. Strains that survived BAC exposure were analyzed at whole genome perspective using comparative genomics, and Sanger sequencing-confirmed mutations in ramR gene were identified. Next, the efflux activity in ramR-mutated strains shown as bisbenzimide accumulation and expression of genes involved in AcrAB-TolC efflux pump using quantitative reverse transcriptase PCR were determined. Results: Mutation rates of evolved strains varied from 5.82 × 10−9 to 5.56 × 10-8, with fold increase from 18.55 to 1.20 when compared with strains evolved without BAC. Mutations in ramR gene were found in evolved strains. Upregulated expression and increased activity of AcrAB-TolC was observed in evolved strains, which may contribute to their increased resistance to clinically relevant antibiotics. In addition, several indels and point mutations in ramR were identified, including L158P, A37V, G42E, F45L, and R46H which have not yet been linked to antimicrobial resistance. Resistance and mutations were stable after seven consecutive cultivations without BAC exposure. These results suggest that strains with sequence type (ST) ST34 were the most prone to mutations in ramR among the three STs tested (ST34, ST19, ST36). Conclusions: This work demonstrated that disinfectants, specifically BAC forces S. Typhimurium to enter a specific evolutionary trajectory towards antibiotic resistance illustrating the side effects of long-term exposure to BAC and probably also to other disinfectants. Most significantly, this study provides new insights in understanding the emergence of antibiotic resistance in modern society.

Abstract Background Disinfectants are important in the food industry to prevent the transmission of pathogens. Excessive use of disinfectants may increase the probability of bacteria experiencing long-term exposure and consequently resistance and cross-resistance to antibiotics. This study aims to investigate the cross-resistance of multidrug-resistant, drug-resistant, and drug-susceptible isolates of Salmonella enterica serovar Typhimurium (S. Typhimurium) with different sequence types (STs) to a group of antibiotics after exposure to different food-grade disinfectants. Methods A panel of 27 S. Typhimurium strains with different antibiograms and STs were exposed to increasing concentrations of five food-grade disinfectants, including hydrogen peroxide (H2O2), benzalkonium chloride (BAC), chlorine dioxide (ClO2), sodium hypochlorite (NaClO), and ethanol. Recovered evolved strains were analyzed using genomic tools and phenotypic tests. Genetic mutations were screened using breseq pipeline and changes in resistance to antibiotics and to the same disinfectant were determined. The relative fitness of evolved strains was also determined. Results Following exposure to disinfectants, 22 out of 135 evolved strains increased their resistance to antibiotics from a group of 14 clinically important antibiotics. The results also showed that 9 out of 135 evolved strains had decreased resistance to some antibiotics. Genetic mutations were found in evolved strains. A total of 77.78% of ST34, 58.33% of ST19, and 66.67% of the other STs strains exhibited changes in antibiotic resistance. BAC was the disinfectant that induced the highest number of strains to cross-resistance to antibiotics. Besides, H2O2 induced the highest number of strains with decreased resistance to antibiotics. Conclusions These findings provide a basis for understanding the effect of disinfectants on the antibiotic resistance of S. Typhimurium. This work highlights the link between long-term exposure to disinfectants and the evolution of resistance to antibiotics and provides evidence to promote the regulated use of disinfectants.

Recent studies have predominantly spotlighted bacterial diversity within coral microbiomes, leaving coral-associated fungi in the shadows of scientific inquiry. This study endeavors to fill this knowledge gap by delving into the biodiversity, distribution and functional differences of fungi associated with soft corals Cladiella krempfi and Sarcophyton tortuosum, gorgonian coral Dichotella gemmacea and stony coral Favia speciosa from the South China Sea. Leveraging high-throughput sequencing of fungal internal transcribed spacer-1 (ITS1) region of the rRNA gene, a total of 431 fungal amplicon sequence variants (ASVs) were identified in this study, which indicated that a large number of fungal communities were harbored in the South China Sea corals. Noteworthy among our findings is that 10 fungal genera are reported for the first time in corals, with Candolleomyces, Exophiala, Fomitopsis, Inaequalispora, Kneiffiella, Paraphaeosphaeria, and Yamadazyma belonging to the Ascomycota, and Cystobasidium, Psathyrella, and Solicoccozyma to the Basidiomycota. Moreover, significant differences (p < 0.05) of fungal communities were observed among the various coral species. In particular, the gorgonian coral D. gemmacea emerged as a veritable haven for fungal diversity, boasting 307 unique ASVs. Contrastingly, soft corals S. tortuosum and C. krempfi exhibited modest fungal diversity, with 36 and 21 unique ASVs, respectively, while the stony coral F. speciosa hosted a comparatively sparse fungal community, with merely 10 unique ASVs in total. These findings not only provide basic data on fungal diversity and function in the South China Sea corals, but also underscore the imperative of nuanced conservation and management strategies for coral reef ecosystems worldwide.

This study aims to investigate the response of Salmonella Newport to plasma-activated water (PAW), a novel disinfectant that attracts attention due to its broad-spectrum antimicrobial efficacy and eco-friendliness. In this work, we demonstrated that S. Newport of different sequence types (STs) could be induced into the viable but nonculturable (VBNC) state by PAW treatment. Notably, a remarkable 99.96% of S. Newport ST45 strain entered the VBNC state after a 12-min PAW treatment, which was the fastest observed among the five S. Newport STs (ST31, ST45, ST46, ST166, ST2364). Secretion of outer membrane vesicles was observed in ST45, suggesting a potential strategy against PAW treatment. Genes related to oxidative stress (sodA, katE, trxA), outer membrane proteins (ompA, ompC, ompD, ompF) and virulence (pagC, sipC, sopE2) were upregulated in the PAW-treated S. Newport, especially in ST45. A reduction of 38–65% in intracellular ATP level after PAW treatment was observed, indicating a contributor to the formation of the VBNC state. In addition, a rapid method for detecting the proportion of VBNC cells in food products based on pagC was established. This study contributes to understanding the formation mechanism of the VBNC state in S. Newport under PAW stress and offers insights for controlling microbial risks in the food industry.

Abstract As hypothesized by Bem (1981)’s Gender Schema Theory, individuals regulate themselves and their expectations towards others according to the gender norms in a community. The current study examines children’s gender schema regarding the language styles in compliments addressed to both the gendered self and others. Two types of oral discourse completion tasks were designed for the purpose, where twenty-five Mandarin-speaking children were instructed to pay compliments in a normal-speaking style and an imitated style of the opposite gender. Machine learning algorithms were implemented to analyze the variations of language features at lexical, discourse-pragmatic, and discourse-semantic levels. The results show that, compared to lexical features such as lexical richness and word choices, discourse-pragmatic features are more prone to gender ideologies and exhibit style-shifting in children’s imitation of the opposite sex when addressing compliments. At the discourse-semantic level, a significantly low probability of positivity was demonstrated in girls’ imitated compliments, according to the results of the logistic regression. In general, the findings support the presence of gender-differentiated language styles among pre-adolescent children. In particular, girls at this age have developed the stereotype that boys tend to use language with a less prosocial sentiment for the manifestation of their “maleness”. Directions for improving the experimental design and uncovering the possible confounding mechanisms were discussed to illuminate the multidimensional complexity of the cross-gender variations in the more nuanced speech traits, such as the use of intensifiers.

BackgroundChild sexual abuse is a major public health problem with adverse consequences for victims’ physical, mental, and reproductive health. This cross-sectional study aimed to determine the prevalence of child sexual abuse and its associated factors among 15- to 17-year-old adolescents in mainland China.MethodsFrom September 8, 2019 to January 17, 2020, a total of 48,660 participants were recruited by 58 colleges and universities across the whole country to complete the self-administered, structured, online questionnaire. This analysis was restricted to 3,215 adolescents aged between 15 and 17 years in mainland China. Chi-square tests and multivariate Logistic regression analyses were performed to identify individual, relationship, and community factors associated with child sexual abuse.ResultsThe overall prevalence of child sexual abuse was 12.0%. More specifically, 13.0% of girls and 10.6% of boys reported that they were sexually abused prior to 18 years of age. At the individual level, being female, sexual minority identity, younger age, and higher levels of knowledge, skills and self-efficacy regarding condom use were significantly related to increased odds of reporting sexual abuse. At the relationship and community level, adolescents from disrupted families and those entering into a marriage, having casual sexual partners, and having first intercourse at a younger age were more likely to report sexual abuse. On the contrary, those who had never discussed sex-related topics with their family members at home and were offered school-based sexuality education later (vs. earlier) were less likely to report sexual abuse.ConclusionMultilevel prevention programs and strategies, including targeting adolescents with high-risk characteristics, educating young children and their parents about child sexual abuse prevention and optimizing the involvement of parents, school, community, society and government in comprehensive sexuality education, should be taken to reduce child sexual abuse among 15- to 17-year-old adolescents.

Since the discovery of penicillin, β-lactam antibiotics have commonly been used to treat bacterial infections. Unfortunately, at the same time, pathogens can develop resistance to β-lactam antibiotics such as penicillins, cephalosporins, monobactams, and carbapenems by producing β-lactamases. Therefore, a combination of β-lactam antibiotics with β-lactamase inhibitors has been a promising approach to controlling β-lactam-resistant bacteria. The discovery of novel β-lactamase inhibitors (BLIs) is essential for effectively treating antibiotic-resistant bacterial infections. Therefore, this review discusses the development of innovative inhibitors meant to enhance the activity of β-lactam antibiotics. Specifically, this review describes the classification and characteristics of different classes of β-lactamases and the synergistic mechanisms of β-lactams and BLIs. In addition, we introduce potential sources of compounds for use as novel BLIs. This provides insights into overcoming current challenges in β-lactamase-producing bacteria and designing effective treatment options in combination with BLIs.

Plain language summary In this study, we aimed to determine the prevalence and factors associated with UIP among sexually active undergraduates in mainland China. Between September 8, 2019 and January 17, 2020, a total of 48,660 participants were recruited from the Chinese mainland to complete the self-administered, structured, online questionnaire. This analysis was restricted to 6347 sexually experienced, never-married 15–26 year old undergraduates. Based on a social-ecological theoretical framework, we ran separate multivariate Logistic regression models for men and women to identify sociodemographic, familial and individual variables associated with UIP. Our findings indicate that the overall prevalence of UIP was 17.7%. More specifically, 19.5% of male college students reported they had unintentionally gotten a partner pregnant, while 14.9% of female college students became unintentionally pregnant. Students who experienced UIP were more likely to belong to the older age group, live with only one parent or live without parents at home, report that their family members approve of premarital sex, initiate sexual activity younger than 14 years old and have casual sex partners. Furthermore, females with multiple partners and males who came from low-income households, experienced sexual abuse, perceived difficulties in acquiring condoms and did not know how to use condoms correctly were also at higher risk of having an unintended pregnancy. In order to prevent UIP, a comprehensive intervention measure should be taken to target older students and those engaging in risky sexual behaviors, work with young male students to improve condom use skills, improve the availability of free condoms, optimize the involvement of parents and other family members in their children’s sex education.

Drinking water disinfection is an essential process to assure public health all over the world. In this study, silver nanoparticles (AgNPs) on UiO-66 (Zr) Metal-Organic Frameworks (Ag@UiO-66) is proposed as a potential water disinfection strategy. AgNPs are synthesized using polyvinyl pyrrolidone (PVP) as stabilizing agent, and sodium borohydride as reducing agent are subsequently embedded on UiO-66, a high-stability organometallic framework. The effect of premixing time, reaction time and reactant concentration on the loading rate of AgNPs on UiO-66 was investigated. The maximum load rate of AgNPs on UiO-66 could reach 13% when the premixing time is 3 h, the reaction time is 45 min and the concentration of AgNO3 is 10 μg/mL. The formation of AgNPs loaded on UiO-66 was observed and confirmed with ultraviolet and visible spectrophotometry (UV-Vis), scanning electron microscopy (SEM), infrared emission spectroscopy (IES) and X-ray diffraction (XRD) analysis. Ag@UiO-66 exhibited strong antibacterial activity against both Gram-negative Escherichia coli and Gram-positive Staphylococcus aureus, with minimum inhibitory concentrations (MIC) of 64 and 128 μg/mL, respectively. The germicidal efficacy of Ag@UiO-66 enhanced significantly as the temperature rose from 4 °C to 37 °C. The results indicate that Ag@UiO-66 is potential candidate as a feasible water disinfection material.

Steel surface defect detection is crucial for ensuring steel quality. The traditional detection algorithm has low detection probability. This paper proposes an improved algorithm based on the YOLOv5 model to enhance detection probability. Firstly, deformable convolution is introduced in the backbone network, and a traditional convolution module is replaced by deformable convolution; secondly, the CBAM attention mechanism is added to the backbone network; then, Focal EIOU is used instead of the CIOU loss function in YOLOv5; lastly, the K-means algorithm is used to cluster the Anchor box, and the Anchor box parameters that are more suitable for this paper are obtained. The experimental results show that using deformable convolution instead of traditional convolution can get more feature information, which is more conducive to the learning of the network. This paper uses the CBAM attention mechanism, and the heat map of the attention mechanism shows that the CBAM attention mechanism is beneficial for feature extraction. Focal EIOU is optimized in high and wide loss compared with the CIOU loss function, which accelerates the convergence of the model. The Anchor box is more favorable for feature extraction. The improved algorithm achieved a detection probability of 78.8% in the NEU-DET dataset, which is 4.3% better than the original YOLOv5 network, and the inference time of each image is only increased by 1 ms; therefore, the optimized algorithm proposed in this paper is effective.

Although intestinal microbiota play crucial roles in fish digestion and health, little is known about intestinal fungi in fish. This study investigated the intestinal fungal diversity of three coral reef fish (Lates calcarifer, Trachinotus blochii, and Lutjanus argentimaculatus) from the South China Sea using a culturable method. A total of 387 isolates were recovered and identified by sequencing their internal transcribed spacer sequences, belonging to 29 known fungal species. The similarity of fungal communities in the intestines of the three fish verified that the fungal colonization might be influenced by their surrounding environments. Furthermore, the fungal communities in different intestines of some fish were significantly different, and the number of yeasts in the hindgut was less than that in fore- and mid-intestines, suggesting that the distribution of fungi in fishes’ intestines may be related to the physiological functions of various intestinal segments. In addition, 51.4% of tested fungal isolates exhibited antimicrobial activity against at least one marine pathogenic microorganism. Notably, isolate Aureobasidium pullulans SCAU243 exhibited strong antifungal activity against Aspergillus versicolor, and isolate Schizophyllum commune SCAU255 displayed extensive antimicrobial activity against four marine pathogenic microorganisms. This study contributed to our understanding of intestinal fungi in coral reef fish and further increased the library of fungi available for natural bioactive product screening.

As one core of the Guangdong–Hong Kong–Macao Greater Bay Area (GBA), Guangdong is facing some serious coastal environmental problems. Fungi are more vulnerable to changes in coastal environments than bacteria and archaea. This study investigated the fungal diversity and composition by high-throughput sequencing and detected basic parameters of seven environmental factors (temperature, dissolved oxygen, pH, salinity, total organic carbon, total nitrogen, and total phosphorus) at 11 sites. A total of 2056 fungal operational taxonomic units (OTUs) belonging to 147 genera in 6 phyla were recovered; Archaeorhizomyces (17.5%) and Aspergillus (14.19%) were the most dominant genera. Interestingly, a total of 14 genera represented the first reports of coastal fungi in this study. Furthermore, there were nine genera of fungi that were significantly correlated with environmental factors. FUNGuild analysis indicated that saprotrophs and pathogens were the two trophic types with the highest proportions. Saprotrophs were significantly correlated with total organic carbon (TOC), total nitrogen (TN), and total phosphorus (TP), while pathogens were significantly correlated with pH. This study provides new scientific data for the study of the diversity and composition of fungal communities in coastal ecosystems.

Milk is easy to be contaminated by microorganisms due to its abundant nutrients. In this study, a 360-degree radiation thermosonication (TS) system was developed and utilized for the inactivation of Staphylococcus aureus in milk. The 360-degree radiation TS system-induced inactivation kinetics of S. aureus was fitted best by the Weibull model compared with biphasic and linear models. The treatment time, the exposure temperature, and the applied ultrasound power was found to affect the bactericidal efficacy of the 360-degree radiation TS system. Additionally, the TS condition of 200 W and 63°C for 7.5 min was successfully applied to achieve complete microbial inactivation (under the limit of detection value) in raw milk. The treatment of 360-degree radiation TS can enhance the zeta potential and decrease the average particle size of milk. It also exhibited better retainment of the proteins in milk compared with the ultrahigh temperature and conventional pasteurization processing. Therefore, the 360-degree radiation TS system developed in this study can be used as an alternative technology to assure the microbiological safety and retain the quality of milk, and the Weibull model could be applied for the prediction of the inactivation levels after exposure to this technology.

In order to reduce the elemental species produced in the recycling and melting of aluminum scrap and to improve the quality of pure aluminum and aluminum alloys, it is necessary to classify the different grades of aluminum scrap before melting. For the problem of classifying different grades of aluminum scrap, most existing studies are conducted using laser-induced breakdown spectroscopy for identification and classification, which requires a clean and flat metal surface and enormous equipment costs. In this study, we propose a new classification and identification method for different grades of aluminum scrap based on the ResNet18 network model, which improves the identification efficiency and reduces the equipment cost. The objects of this research are three grades of aluminum scrap: 1060, 5052, and 6061. The surface features of the three grades were compared using a machine vision algorithm; three different datasets, using RGB, HSV, and LBP, were built for comparison to find the best training dataset for subsequent datasets, and the hyperparameters of learning rate and batch size were tuned for the ResNet18 model. The results show that there was a differentiation threshold between different grades through the comparison of surface features; the ResNet18 network model trained the three datasets, and the results showed that RGB was the best dataset. With hyperparameter optimization of the ResNet18 model, the accuracy of final classification and recognition could reach 100% and effectively achieve the classification of different grades of aluminum scrap.

Ultrasound, is thought to a potential non-thermal sterilization technology in food industry. However, the exact mechanisms underlying microbial inactivation by ultrasound still remain obscure. In this study, the action modes of ultrasound on both Gram-negative and Gram-positive microorganisms were estimated. From colony results, ultrasound acted as an irreversible effect on both Eshcerichia coli and Staphylococcus aureus without sublethal injury. The result in this study also showed that a proportion of bacteria subpopulation suffered from serious damage of intracellular components (e.g. DNA and enzymes) but with intact cell envelopes. We speculated that the inactivated effects of ultrasound on microbes might more than simply completed disruption of cell exteriors. Those microbial cells who had not enter the valid area of ultrasonic cavitation might be injected with free radicals produced by ultrasound and experienced interior injury with intact exterior structure, and others who were in close proximity to the ultrasonic wave field would be immediately and completely disrupted into debris by high power mechanic forces. These findings here try to provide extension for the inactivation mechanisms of ultrasound on microorganisms. Ultrasound has attracted great interest of both industry and scientific communities for its potential use as a physical processing and preservation tool. In this study, Escherichia coli O157:H7 was selected as the model microbe to investigate the ultrasound-induced cell death. Slight variations in membrane potential and ion exchanges across membrane induced by low-intensity ultrasound increased the membrane permeability of E. coli O157:H7, and this reversible sublethal effect can preserve the viability of E. coli O157:H7 and meanwhile be beneficial for bioprocessing application. In comparison, high-intensity ultrasound resulted in irreversible lethal effect on E. coli O157:H7, which can be applied in the field of microbial inactivation. In addition, both low- and high-intensity ultrasound induced either physical destruction or trigger genetically encoded apoptosis of E. coli O157:H7. Accumulation of reactive oxygen species and decrease of adenosine tri-phosphate might be related to the physiological and biochemical hallmarks of apoptosis, including exposed phosphatidylserine and activated caspases in E. coli O157:H7. The result provides novel insight into the mechanisms of non-thermal physical treatment on the inactivation of bacteria and lays foundation for the further research on the cell signaling and metabolic pathway in apoptotic bacteria.
