Abstract Objective Clinically, it has been found that patients undergoing knee replacement have a high incidence of concomitant hallux valgus. In this study, we analyzed whether patients with osteoarthritis who underwent surgery and those patient who did not have surgery had an increased risk of hallux valgus by Mendelian randomization and performed reverse causal analysis. Design Genomewide association study (GWAS) data for osteoarthritis, categorized by knee arthritis with joint replacement, knee arthritis without joint replacement, hip arthritis with joint replacement, and hip arthritis without joint replacement.And acquired hallux valgus were downloaded for Mendelian randomized studies. MR analysis was performed using inverse variance-weighted (IVW), weighted median, and MR-Egger methods. MR-egger regression, MR pleiotropic residuals and outliers (MR-presso), and Cochran's Q statistical methods were used to evaluate heterogeneity and pleiotropy. Results The IVW results indicate that, compared to healthy individuals, patients who meet the criteria for knee osteoarthritis joint replacement surgery have a significantly higher risk of acquired hallux valgus. There were no significant causal relationships found for the remaining results. No significant heterogeneity or multiplicity was observed in all the Mr analyses. Conclusion Our study supports the increased risk of acquired hallux valgus in patients eligible for knee replacement. There is necessary for clinicians to be concerned about the hallux valgus status of patients undergoing knee arthroplasty.

Abstract Introduction Dual eligible beneficiaries are a vulnerable population who often experience inferior access to care and outcomes compared to non‐dual eligible beneficiaries. The Oncology Care Model (OCM) is an alternative payment model that aims to improve coordination and quality of care in beneficiaries receiving chemotherapy and thus may improve care for dual eligible beneficiaries with cancer. Methods We used 100% Medicare claims data from 2014 through 2019 and included beneficiaries with bladder, breast, esophageal, colorectal, kidney, lung, pancreatic, or prostate cancer receiving chemotherapy. We constructed multivariable difference‐in‐differences regression models to evaluate the effect of OCM participation on healthcare utilization and quality of care at the end‐of‐life among dual eligible beneficiaries. We also compared healthcare utilization and quality of care outcomes to non‐dual eligible beneficiaries. Results We identified 3,043,944 episodes of care among 1,260,892 unique Medicare beneficiaries. Ten percent of all beneficiaries (n = 126,758) were dual eligible and 64,087 (22%) of episodes among dual eligible patients were in an OCM participating practice. We noted no effect of OCM participation on healthcare utilization or end‐of‐life quality of care for dual eligible beneficiaries. However, we observed higher rates of hospitalization, emergency department visits, intensive care unit stays, and a lower number of office visits among dual eligible beneficiaries compared to non‐dual eligible beneficiaries. Conclusions Participation in OCM was not associated with improvements in quality of care or healthcare utilization for dual eligible beneficiaries. Dual eligible beneficiaries experience lower quality of care across several measures compared to non‐dual eligible beneficiaries. Focused policies and incentives may be necessary to address disparities within emerging health reforms.

Objective Glucocorticoid (GC) overuse is strongly associated with steroid‐induced osteonecrosis of the femoral head (SINFH). However, the underlying mechanism of SINFH remains unclear. This study aims to investigate the effect of dexamethasone (Dex)‐induced oxidative stress on osteocyte apoptosis and the underlying mechanisms. Methods Ten patients with SINFH and 10 patients with developmental dysplasia of the hips (DDH) were enrolled in our study. Sixty rats were randomly assigned to the Control, Dex, Dex + N‐Acetyl‐L‐cysteine (NAC), Dex + Dibenziodolium chloride (DPI), NAC, and DPI groups. Magnetic resonance imaging (MRI) was used to examine edema in the femoral head of rats. Histopathological staining was performed to assess osteonecrosis. Immunofluorescence staining with TUNEL and 8‐OHdG was conducted to evaluate osteocyte apoptosis and oxidative damage. Immunohistochemical staining was carried out to detect the expression of NOX1, NOX2, and NOX4. Viability and apoptosis of MLO‐Y4 cells were measured using the CCK‐8 assay and TUNEL staining. 8‐OHdG staining was conducted to detect oxidative stress. 2′,7′‐Dichlorodihydrofluorescein diacetate (DCFH‐DA) staining was performed to measure reactive oxygen species (ROS). The expression of NOX1, NOX2, and NOX4 in MLO‐Y4 cells was analyzed by Western blotting. Multiple comparisons were performed using one‐way analysis of variance (ANOVA). Results In patients and the rat model, hematoxylin–eosin (HE) staining revealed a significantly higher rate of empty lacunae in the SINFH group than in the DDH group. Immunofluorescence staining indicated a significant increase in TUNEL‐positive cells and 8‐OHdG‐positive cells in the SINFH group compared to the DDH group. Immunohistochemical staining demonstrated a significant increase in the expression of NOX1, NOX2, and NOX4 proteins in SINFH patients compared to DDH patients. Moreover, immunohistochemical staining showed a significant increase in the proportion of NOX2‐positive cells compared to the Control group in the femoral head of rats. In vitro, Dex significantly inhibited the viability of osteocyte cells and induced apoptosis. After Dex treatment, the intracellular ROS level increased. However, Dex treatment did not alter the expression of NOX proteins in vitro. Additionally, NAC and DPI inhibited the generation of intracellular ROS and partially alleviated osteocyte apoptosis in vivo and in vitro. Conclusion This study demonstrates that GC promotes apoptosis of osteocyte cells through ROS‐induced oxidative stress. Furthermore, we found that the increased expression of NOXs induced by GC serves as an important source of ROS generation.

To improve the prediction accuracy of monocular depth estimation networks and address issues such as edge blurring and excessive artifacts in the generated depth maps, this paper proposes a deep network architecture based on a global-local feature fusion module and a residual pooling module. The encoder utilizes a Hierarchical Transformer, while the decoder incorporates a U-Net structure model that combines multi-dimensional attention features aggregation and residual pooling. The residual pooling module facilitates better extraction of background contextual information from the feature maps to obtain more precise scene depth information. The global-local feature fusion module enables the network to learn features that encompass both global and local information. Experimental evaluations conducted on the NYU Depth V2 and KITTI datasets demonstrate that the proposed method achieves a $\delta 1$ of 0.916 on the NYU Depth V2 dataset, along with enhanced generalization ability and robustness. Furthermore, the effectiveness of each module is validated through ablation studies on the NYU Depth V2 dataset.

AbstractPurpose This retrospective, single-center, case-control study evaluated the safety and efficacy of Computed tomography (CT)-guided microwave ablation (MWA) for pulmonary nodules located in the right middle lobe (RML), a challenging location associated with a high frequency of complications.Methods Between May 2020 and April 2022, 71 patients with 71 RML pulmonary nodules underwent 71 MWA sessions. To comparison, 142 patients with 142 pulmonary nodules in non-RML were selected using propensity score matching. The technical success, technique efficacy, complications, and associated factors were analyzed. The duration of the procedure and post-ablation hospital stay were also recorded.Results Technical success was achieved in 100% of all patients. ObjectiveAmong adolescents with depression, the occurrence of non-suicidal self-injury (NSSI) behavior is prevalent, constituting a high-risk factor for suicide. However, there has been limited research on the neuroimaging mechanisms underlying adolescent depression and NSSI behavior, and the potential association between the two remains unclear. Therefore, this study aims to investigate the alterations in functional connectivity (FC) of the regions in the prefrontal cortex with the whole brain, and elucidates the relationship between these alterations and NSSI behavior in adolescents with depression.MethodsA total of 68 participants were included in this study, including 35 adolescents with depression and 33 healthy controls. All participants underwent assessments using the 17-item Hamilton Depression Rating Scale (17-HAMD) and the Ottawa Self-Harm Inventory. In addition, functional magnetic resonance imaging (fMRI) data of the participants’ brains were collected. Subsequently, the FCs of the regions in the prefrontal cortex with the whole brain was calculated. The FCs showing significant differences were then subjected to correlation analyses with 17-HAMD scores and NSSI behavior scores.ResultCompared to the healthy control group, the adolescent depression group exhibited decreased FCs in several regions, including the right frontal eye field, left dorsolateral prefrontal cortex, right orbitofrontal cortex, left insula and right anterior cingulate coetex. The 17-HAMD score was positively correlated with the frequency of NSSI behavior within 1 year (rs = 0.461, p = 0.005). The FC between the right anterior cingulate cortex and the right precuneus showed a negative correlation with the 17-HAMD scores (rs = −0.401, p = 0.023). Additionally, the FC between the right orbitofrontal cortex and the right insula, demonstrated a negative correlation with the frequency of NSSI behavior within 1 year (rs = −0.438, p = 0.012, respectively).ConclusionAdolescents with depression showed decreased FCs of the prefrontal cortex with multiple brain regions, and some of these FCs were associated with the NSSI frequency within 1 year. This study provided neuroimaging evidence for the neurophysiological mechanisms underlying adolescent depression and its comorbidity with NSSI behavior.

Abstract Background The relationship between aging and osteoporosis is well established. However, the relationship between the body's physiological age, i.e. epigenetic age, and osteoporosis is not known. Our goal is to analyze the bidirectional causal relationship between epigenetic clocks and osteoporosis using a bidirectional Mendelian randomization study. Methods We used SNPs closely associated with GrimAge, Hannum, PhenoAge, and HorvathAge in epigenetic age and SNPs closely associated with femoral neck bone mineral density, lumbar spine bone mineral density, and forearm bone mineral density as instrumental variables, respectively, using the inverse variance weighting method and several other MR methods to assess the bidirectional causal relationship between epigenetic age and osteoporosis. Result There was no evidence of a clear causal relationship of epigenetic age (GrimAge, Hannum, PhenoAge, and HorvathAge) on femoral neck bone mineral density, lumbar spine bone mineral density, and forearm bone mineral density. In reverse Mendelian randomization analysis showed a significant causal effect of lumbar spine bone mineral density on GrimAge: odds ratio (OR) = 0.692, 95% confidence interval (CI) = (0.538–0.890), p = 0.004. The results suggest that a decrease in lumbar spine bone mineral density promotes an acceleration of GrimAge. Conclusion There was no significant bidirectional causal relationship between epigenetic age and osteoporosis A decrease in lumbar spine bone density may lead to an acceleration of the epigenetic clock "GrimAge". Our study provides partial evidence for a bidirectional causal effect between epigenetic age and Osteoporosis.

Foggy and hazy weather conditions can significantly reduce the clarity of images captured by cameras, making it difficult for object detection algorithms to accurately recognize targets. This degradation can cause failures in autonomous or assisted driving systems, posing severe safety threats to both drivers and passengers. To address the issue of decreased detection accuracy in foggy weather, we propose an object detection algorithm specifically designed for such environments, named AODs-CLYOLO. To effectively handle images affected by fog, we introduce an image dehazing model, AODs, which is more suitable for detection tasks. This model incorporates a Channel–Pixel (CP) attention mechanism and a new Contrastive Regularization (CR), enhancing the dehazing effect while preserving the integrity of image information. For the detection network component, we propose a learnable Cross-Stage Partial Connection Module (CSPCM++), which is used before the detection head. Alongside this, we integrate the LSKNet selective attention mechanism to improve the extraction of effective features from large objects. Additionally, we apply the FocalGIoU loss function to enhance the model’s performance in scenarios characterized by sample imbalance or a high proportion of difficult samples. Experimental results demonstrate that the AODs-CLYOLO detection algorithm achieves up to a 10.1% improvement in the mAP (0.5:0.95) metric compared to the baseline model YOLOv5s.

Objectives. To compare four DNA extraction methods for recovering bacterial DNA from mammalian faecal samples. Methods and Results. Three commercial kits from QIAamp, TIANamp, and MAGEN, together with the classic cetyltrimethylammonium bromide (CTAB) method, were evaluated for their performance in extracting bacterial DNA from the gut microbiota of five mammals, including humans (Homo sapiens), macaques (Macaca mulatta), dogs (Canis lupus familiaris), golden hamsters (Mesocricetus auratus), and mice (Mus musculus). First, we assessed the efficiency of the four methods based on DNA yield, purity, and integrity. Then, we investigated the impact of these methods on microbial composition and diversity and examined the relative abundance of dominant phyla bacteria based on 16S rRNA gene sequencing and real-time quantitative PCR. Our results showed that the CTAB method yielded relatively larger amounts of DNA, while the MAGEN kit yielded more Firmicutes DNA and reflected the true status of the microbiota more accurately. Conclusions. Of the four methods tested, the traditional CTAB method and commercial MAGEN kit accomplished the best performance in terms of DNA concentration and Firmicutes abundance across most of the tested species. As the CTAB method and MAGEN kit exhibited different advantages, we further tested and compared their DNA extraction performance on a defined microbiota comprising six strains from four dominant phyla to see which one better reflected the true status of the microbiota. In conclusion, the MAGEN kit was found to be superior to the CTAB method, as the testing results were closer to that of the defined microbiota.

Single crystals of the intermetallic phase with composition Ti4Ni2C were serendipitously obtained by high-pressure sintering of a mixture with initial chemical composition Ti2Ni. The Ti4Ni2C phase crystallizes in the Fd\overline{3}m space group and can be considered as a partially filled Ti2Ni structure with the C atom occupying an octahedral void. Ti4Ni2C is isotypic with Ti4Ni2O, Nb4Ni2C and Ta4Ni2C, all of which were studied previously by means of powder diffraction.

Background Despite the health effects of basal metabolic rate (BMR), the causal effect of BMR on cardiovascular diseases (CVDs) remains undetermined. To elucidate the causal nature, Mendelian randomization (MR) analyses were performed. Methods and Results Summary genome‐wide association statistics regarding BMR and 5 CVDs were obtained from European databases. A 2‐sample bidirectional MR was performed to assess the causal association between BMR and CVDs. The causal effects were estimated using inverse variance weighting. Simultaneously, multiple sensitivity analyses were performed to validate the robustness and reliability of the results. This study utilized a plant-wearable sap flow sensor developed by a multidisciplinary team at Zhejiang University to monitor water distribution patterns in watermelon fruit stalks throughout their developmental stages. The dynamic rules of sap flow at different stages of fruit development were discovered: (1) In the first stage, sap flow into the fruit gradually halts after sunrise due to increased leaf transpiration, followed by a rapid increase post-noon until the next morning, correlating with fruit expansion. (2) In the second stage, the time of inflow sap from noon to night is significantly shortened, while the outflow sap from fruit is observed with the enhancement of leaf transpiration after sunrise, which is consistent with the slow fruit growth at this stage. (3) In the third stage, the sap flow maintains the diurnal pattern. However, the sap flow that inputs the fruit at night is basically equal to the sap flow that outputs the fruit during the day; the fruit phenotype does not change anymore. In addition, a strong correlation between the daily mass growth in fruit and the daily sap flow amount in fruit stalk was identified, validating the sensor’s utility for fruit growth monitoring and yield prediction.

The structure and function of ecological spaces play a crucial role in resisting risk interference and providing ecological services. Conducting resilience assessments of ecological spaces is of great significance for maintaining ecological security. Taking the Baiyangdian Basin as the research object, based on the evaluation results of ecosystem service function importance and ecological environmental sensitivity, the ecological space importance level of the Baiyangdian Basin is divided. Based on the three basic characteristics of “resistance, adaptability, recoverability”, the resilience of ecological space model was constructed by selecting ecosystem service value, landscape structure stability, ecological resilience model and coefficient. The research results show that: (1) The overall importance index of ecological space in the Baiyangdian Basin is relatively low. The non-critical area accounted for 43.00% of the total area, the general area accounted for 30.78%, and the core area accounted for 26.22%, which mainly distributed in the northern part of the study area and at the confluence of rivers. (2) The proportion of areas with extremely important ecosystem service functions in Baiyangdian Basin is 31.73%, concentrated in the central and southwestern parts of the basin. The highly sensitive ecological environment areas accounted for 21.96%, mainly distributed in high-altitude areas in the northern part of the basin and river convergence areas. (3) On the whole, the overall resilience level of the study area is not high and there is a significant difference in resilience between the north and south, with 68.60% of the regions in the low level of resilience. The area with high resilience level accounted for only 1.51%, and the highest resilience level was found at river confluence. The research results provide a theoretical basis for maintaining and improving the ecological environment of the basin, and provide a basis for implementing precise policies to optimize the ecological space of the basin.

Renal interstitial fibrosis (RIF), a progressive process affecting the kidneys in chronic kidney disease (CKD), currently lacks an effective therapeutic intervention. Traditional Chinese medicine (TCM) has shown promise in reducing RIF and slowing CKD progression. In this study, we demonstrated the dose-dependent attenuation of RIF by Ootheca mantidis (SPX), a commonly prescribed TCM for CKD, in a mouse model of unilateral ureteral obstruction (UUO). RNA-sequencing analysis suggested that SPX treatment prominently downregulated apoptosis and inflammation-associated pathways, thereby inhibiting the fibrogenic signaling in the kidney. We further found that transplantation of fecal microbiota from SPX-treated mice conferred protection against renal injury and fibrosis through suppressing apoptosis in UUO mice, indicating that SPX ameliorated RIF via remodeling the gut microbiota and reducing apoptosis in the kidneys. Further functional exploration of the gut microbiota combined with fecal metabolomics revealed increased levels of some probiotics, including Akkermansia muciniphila (A. muciniphila), and modulations in glutamine-related amino acid metabolism in UUO mice treated with SPX. Subsequent colonization of A. muciniphila and supplementation with glutamine effectively mitigated cell apoptosis and RIF in UUO mice. Collectively, these findings unveil a functionally A. muciniphila- and glutamine-involved gut–renal axis that contributes to the action of SPX, and provide important clue for the therapeutic potential of SPX, A. muciniphila, and glutamine in combatting RIF.

This study aimed to explore the mechanism through which Tibetan medicine Liuwei Muxiang (LWMX) pills acts against colorectal cancer (CRC). We firstly retrieved the active ingredients and the correlated targets of LWMX pills from public databases. The CRC-related targets were determined through bioinformatic analysis of a public CRC dataset. By computing the intersection of the drug-specific and disease-related targets, LWMX pill–CRC interaction networks were constructed using the protein–protein interaction (PPI) method and functional enrichment analysis. Subsequently, we determined the hub genes using machine learning tools and further verified their critical roles in CRC treatment via immune infiltration analysis and molecular docking studies. We identified 81 active ingredients in LWMX pills with 614 correlated targets, 1877 differentially expressed genes, and 9534 coexpression module genes related to CRC. A total of 5 target hub genes were identified among the 108 intersecting genes using machine learning algorithms. The immune infiltration analysis results suggested that LWMX pills could affect the CRC immune infiltration microenvironment by regulating the expression of the target hub genes. Finally, the molecular docking outcomes revealed stable binding affinity between all target hub proteins and the primary active ingredients of LWMX pills. Our findings illustrate the anti-CRC potential and the mechanism of action of LWMX pills and provide novel insights into multitarget medication for CRC treatment.

Abstract Objective High intake of caffeoylquinic acid (CQA)-rich dietary supplements, such as green coffee bean extracts, offers health-promoting effects on maintaining metabolic homeostasis. Similar to many active herbal ingredients with high pharmacological activities but low bioavailability, CQA has been reported as a promising thermogenic agent with anti-obesity properties, which contrasts with its poor oral absorption. Intestinal tract is the first site of CQA exposure and gut microbes might react quickly to CQA. Thus, it is of interest to explore the role of gut microbiome and microbial metabolites in the beneficial effects of CQA on obesity-related disorders. Results Oral CQA supplementation effectively enhanced energy expenditure by activating browning of adipose and thus ameliorated obesity-related metabolic dysfunctions in high fat diet-induced obese (DIO) mice. Here, 16S rRNA gene amplicon sequencing revealed that CQA treatment remodeled the gut microbiota to promote its anti-obesity actions, as confirmed by antibiotic treatment and fecal microbiota transplantation. CQA enriched the gut commensal species Limosilactobacillus reuteri (L. reuteri) and stimulated the production of short-chain fatty acids, especially propionate. Mono-colonization of L. reuteri or low-dose CQA treatment did not reduce adiposity in DIO mice, while their combination elicited an enhanced thermogenic response, indicating the synergistic effects of CQA and L. reuteri on obesity. Exogenous propionate supplementation mimicked the anti-obesity effects of CQA alone or when combined with L. reuteri, which was ablated by the monocarboxylate transporter (MCT) inhibitor 7ACC1 or MCT1 disruption in inguinal white adipose tissues to block propionate transport. Conclusions Our data demonstrate a functional axis among L. reuteri, propionate, and beige fat tissue in the anti-obesity action of CQA through the regulation of thermogenesis. These findings provide mechanistic insights into the therapeutic use of herbal ingredients with poor bioavailability via their interaction with the gut microbiota. Video Abstract

IntroductionThe causal relationship between Coronavirus disease 2019 (COVID-19) and osteoporosis (OP) remains uncertain. We aimed to assess the effect of COVID-19 severity (severe acute respiratory syndrome coronavirus 2 (SARS-CoV-2) infection, COVID-19 hospitalization, and severe COVID-19) on OP by a two-sample Mendelian randomization (MR) study.MethodsWe conducted a two-sample MR analysis using publicly available genome-wide association study (GWAS) data. Inverse variance weighting (IVW) was used as the main analysis method. Four complementary methods were used for our MR analysis, which included the MR–Egger regression method, the weighted median method, the simple mode method, and the weighted mode method. We utilized the MR-Egger intercept test and MR pleiotropy residual sum and outlier (MR-PRESSO) global test to identify the presence of horizontal pleiotropy. Cochran’s Q statistics were employed to assess the existence of instrument heterogeneity. We conducted a sensitivity analysis using the leave-one-out method.ResultsThe primary results of IVW showed that COVID-19 severity was not statistically related to OP (SARS-CoV-2 infection: OR (95% CI) = 0.998 (0.995 ~ 1.001), p = 0.201403; COVID-19 hospitalization: OR (95% CI) =1.001 (0.999 ~ 1.003), p = 0.504735; severe COVID-19: OR (95% CI) = 1.000 (0.998 ~ 1.001), p = 0.965383). In addition, the MR-Egger regression, weighted median, simple mode and weighted mode methods showed consistent results. The results were robust under all sensitivity analyses.ConclusionThe results of the MR analysis provide preliminary evidence that a genetic causal link between the severity of COVID-19 and OP may be absent.

BackgroundRecent studies have shown that major depressive disorder (MDD) is associated with altered intrinsic functional connectivity (FC) of the thalamus; however, investigations of these alterations at a finer time scale and the level of thalamic subregions are still lacking.MethodsWe collected resting-state functional MRI data from 100 treatment-naïve, first-episode MDD patients and 99 age-, gender- and education-matched healthy controls (HCs). Seed-based whole-brain sliding window-based dFC analyses were performed for 16 thalamic subregions. Between-group differences in the mean and variance of dFC were determined using threshold-free cluster enhancement algorithm. For significant alterations, there relationships with clinical and neuropsychological variables were further examined via bivariate and multivariate correlation analyses.ResultsOf all thalamic subregions, only the left sensory thalamus (Stha) showed altered variance of dFC in the patients characterized by increases with the left inferior parietal lobule, left superior frontal gyrus, left inferior temporal gyrus, and left precuneus, and decreases with multiple frontal, temporal, parietal, and subcortical regions. These alterations accounted for, to a great extent, clinical, and neuropsychological characteristics of the patients as revealed by the multivariate correlation analysis. In addition, the bivariate correlation analysis revealed a positive correlation between the variance of dFC between the left Stha and right inferior temporal gurus/fusiform and childhood trauma questionnaires scores (r = 0.562, P < 0.001).ConclusionThese findings suggest that the left Stha is the most vulnerable thalamic subregion to MDD, whose dFC alterations may serve as potential biomarkers for the diagnosis of the disease.

Fish head cutting is one of the most important processes during fish pre-processing. At present, the identification of cutting positions mainly depends on manual experience, which cannot meet the requirements of large-scale production lines. In this paper, a fast and contactless identification method of cutting position was carried out by using a constructed line laser data acquisition system. The fish surface data were collected by a linear laser scanning sensor, and Principal Component Analysis (PCA) was used to reduce the dimensions of the dorsal and abdominal boundary data. Based on the dimension data, Least Squares Support Vector Machines (LS-SVMs), Particle Swarm Optimization-Back Propagation (PSO-BP) networks, and Long and Short Term Memory (LSTM) neural networks were applied for fish head cutting position identification model establishment. According to the results, the LSTM model was considered to be the best prediction model with a determination coefficient (R2) value, root mean square error (RMSE), mean absolute error (MAE), and residual predictive deviation (RPD) of 0.9480, 0.2957, 0.1933, and 3.1426, respectively. This study demonstrated the reliability of combining line laser scanning techniques with machine learning using LSTM to identify the fish head cutting position accurately and quickly. It can provide a theoretical reference for the development of intelligent processing and intelligent cutting equipment for fish.

In order to study the strength change of concrete specimens under different loading conditions, based on the principle of minimum energy dissipation, the damage energy per unit area of concrete was studied. By using finite element numerical simulation software for concrete specimens with different failure modes of tension, pressure, bending and torsion, a double-broken line damage constitutive model is adopted. The failure forms of concrete specimens under different loading conditions, as well as the failure area and failure energy of each specimen during loading, are simulated and analyzed. The failure energy per unit area under different failure modes was quantitively calculated, the relationship between the failure area and failure energy consumption under different failure modes was analyzed. The results show that, under different failure modes, the failure area of concrete specimens is different, the energy consumed during failure is different, and the strength is different. However, no matter how the failure mode changes during the failure process, the failure energy W per unit area remains constant and fluctuates in the range of 2.0~6.0 mJ/cm2, which is related to the physical properties of concrete itself.

Abstract Background Grass carp are an important farmed fish in China that are infected by many pathogens, especially grass carp reovirus (GCRV). Notably, grass carp showed age-dependent susceptibility to GCRV; that is, grass carp not older than one year were sensitive to GCRV, while those over three years old were resistant to this virus. However, the underlying mechanism remains unclear. Herein, whole genome-wide DNA methylation and gene expression variations between susceptible five-month-old (FMO) and resistant three-year-old (TYO) grass carp were investigated aiming to uncover potential epigenetic mechanisms. Results Colorimetric quantification revealed that the global methylation level in TYO fish was higher than that in FMO fish. Whole-genome bisulfite sequencing (WGBS) of the two groups revealed 6214 differentially methylated regions (DMRs) and 4052 differentially methylated genes (DMGs), with most DMRs and DMGs showing hypermethylation patterns in TYO fish. Correlation analysis revealed that DNA hypomethylation in promoter regions and DNA hypermethylation in gene body regions were associated with gene expression. Enrichment analysis revealed that promoter hypo-DMGs in TYO fish were significantly enriched in typical immune response pathways, whereas gene body hyper-DMGs in TYO fish were significantly enriched in terms related to RNA transcription, biosynthesis, and energy production. RNA-seq analysis of the corresponding samples indicated that most of the genes in the above terms were upregulated in TYO fish. Moreover, gene function analysis revealed that the two genes involved in energy metabolism displayed antiviral effects. Conclusions Collectively, these results revealed genome-wide variations in DNA methylation between grass carp of different ages. DNA methylation and gene expression variations in genes involved in immune response, biosynthesis, and energy production may contribute to age-dependent susceptibility to GCRV in grass carp. Our results provide important information for disease-resistant breeding programs for grass carp and may also benefit research on age-dependent diseases in humans.

Background and objectiveIntracranial fusiform aneurysms (IFAs) are considered to have a complex pathophysiology process and poor natural history. The purpose of this study was to investigate the pathophysiological mechanisms of IFAs based on the characteristics of aneurysm wall enhancement (AWE), hemodynamics, and morphology.MethodsA total of 21 patients with 21 IFAs (seven fusiform types, seven dolichoectatic types, and seven transitional types) were included in this study. Morphological parameters of IFAs were measured from the vascular model, including the maximum diameter (Dmax), maximum length (Lmax), and centerline curvature and torsion of fusiform aneurysms. The three-dimensional (3D) distribution of AWE in IFAs was obtained based on high-resolution magnetic resonance imaging (HR-MRI). Hemodynamic parameters including time-averaged wall shear stress (TAWSS), oscillatory shear index (OSI), gradient oscillatory number (GON), and relative residence time (RRT) were extracted by computational fluid dynamics (CFD) analysis of the vascular model, and the relationship between these parameters and AWE was investigated.ResultsThe results showed that Dmax (p = 0.007), Lmax (p = 0.022), enhancement area (p = 0.002), and proportion of enhancement area (p = 0.006) were significantly different among three IFA types, and the transitional type had the largest Dmax, Lmax, and enhancement area. The instability of rock fractures has been extensively studied in domestic and international research. This paper investigates two coal types from the Jiaozuo area, which were subjected to liquid nitrogen cyclic treatment using Brazil’s indirect tensile testing (BITT). The study analyzes the characteristics of acoustic emission (AE) and crack formation during the coal cracking process and evaluates the impact of freeze-thaw treatment with liquid nitrogen cycles on the mechanical properties of coals. The results demonstrate that liquid nitrogen has a significant impact on the formation and development of coal fractures under the Brazil splitting test conditions. As the number of cyclic treatments increases, the maximum load-bearing capacity of coal decreases, indicating a gradual reduction in the effective stress on coal samples. Furthermore, the acoustic emission activity intensity of coal mass during phased loading increases as the number of freeze-thaw cycles of liquid nitrogen increases. Finally, during phased loading of coal specimens, the primary factor is the germination, extension, and failure of tensile cracks, while the secondary factor is the development, extension, and failure of shear cracks. Overall, this study provides valuable insights into the behavior of coals under freeze-thaw cycles of liquid nitrogen treatment and highlights the importance of understanding the characteristics of coal fracture instability.

The ongoing development of renewable energy and microgrid technologies has gradually transformed the conventional energy infrastructure and upgraded it into a modernized system with more distributed generation and localized energy storage options. Compared with power grids utilizing synchronous generation, inverter-based networks cannot physically provide large amounts of inertia, which means that more advanced and extensive studies regarding stability considerations are required for such systems. Therefore, appropriate analytical methods are needed for the voltage stability analysis of renewable-dominated power systems, which incorporate a large number of inverters and distributed energy sources. This paper provides a comprehensive literature review of voltage stability analyses of power systems with high levels of renewable energy penetration. A series of generalized evaluation schemes and improvement methods relating to the voltage stability of power systems integrated with various distributed energy resources are discussed. The existing voltage stability analysis methods and corresponding simulation verification models for microgrids are also reviewed in a systematic manner. The traditional and improved voltage stability analysis methods are reviewed according to the microgrid operation mode, the types of distributed generators, and the microgrid configurations. Moreover, the voltage stability indices, which play a crucial role in voltage stability assessments, are critically evaluated in terms of the applicable conditions. The associated modeling and simulation techniques are also presented and discussed. This contribution presents guidelines for voltage stability analysis and instability mitigation methods for modern renewable-rich power systems.

In this paper, we propose a novel method for point cloud complementation called PADPNet. Our approach uses a combination of global and local information to infer missing elements in the point cloud. We achieve this by dividing the input point cloud into uniform local regions, called perceptual fields, which are abstractly understood as special convolution kernels. The set of point clouds in each local region is represented as a feature vector and transformed into N uniform perceptual fields as the input to our transformer model. We also designed a geometric density-aware block to better exploit the inductive bias of the point cloud’s 3D geometric structure. Our method preserves sharp edges and detailed structures that are often lost in voxel-based or point-based approaches. Experimental results demonstrate that our approach outperforms other methods in reducing the ambiguity of output results. Our proposed method has important applications in 3D computer vision and can efficiently recover complete 3D object shapes from missing point clouds.

ABSTRACT Grass carp is an important commercial fish in China that is plagued by various diseases, especially the hemorrhagic disease induced by grass carp reovirus (GCRV). Nevertheless, the mechanism by which GCRV hijacks the host metabolism to complete its life cycle is unclear. In this study, we performed lipidomic analysis of grass carp liver samples collected before and after GCRV infection. GCRV infection altered host lipid metabolism and increased de novo fatty acid synthesis. Increased de novo fatty acid synthesis induced accumulation of lipid droplets (LDs). LDs are associated with GCRV viroplasms, as well as viral proteins and double-stranded RNA. Pharmacological inhibition of LD formation led to the disappearance of viroplasms, accompanied by decreased viral replication capacity. Moreover, transmission electron microscopy revealed LDs in close association with the viroplasms and mounted GCRV particles. Collectively, these data suggest that LDs are essential for viroplasm formation and are sites for GCRV replication and assembly. Our results revealed the detailed molecular events of GCRV hijacking host lipid metabolism to benefit its replication and assembly, which may provide new perspective for the prevention and control of GCRV. IMPORTANCE Grass carp reovirus (GCRV) is the most virulent pathogen in the genus Aquareovirus, which belongs to the family Reoviridae. GCRV-induced hemorrhagic disease is a major threat to the grass carp aquaculture industry. Viruses are obligate intracellular parasites that require host cell machinery to complete their life cycle; the mechanism by which GCRV hijacks the host metabolism to benefit viral replication and assembly remains unclear. Our study demonstrated that GCRV infection alters host lipid metabolism and increases de novo fatty acid synthesis. The increased de novo fatty acid synthesis induced accumulation of LDs, which act as sites or scaffolds for GCRV replication and assembly. Our findings illustrate a typical example of how the virus hijacks cellular organelles for replication and assembly and hence may provide new insights for the prevention and control of GCRV.

In view of the difficulties in optimal dispatch caused by diverse energy and various types of equipment in the regional integrated energy system(RIES), this paper established a generalized energy storage model by extracting the flexibility characteristics of cooling, heat, and electrical load based on abstract equivalence. Instead of reducing the accuracy of regulation, this modeling method can greatly simplify the information interaction between the energy dispatching center and energy consumers, thus cutting down the time of collaborative analysis calculations. With the scheduling characteristics of multiple types of flexible loads in day-ahead and intra-day time scales considered comprehensively, a multi-time-scale optimization scheduling model of RIES is established, which takes into account the economy of system operation and the demand response. The simulation results show that the proposed optimization model can fully tap the load-side scheduling potential, along with effectively reducing the computational complexity. Through the coordination of resources with different characteristics, the pressure of balancing energy supply and demand can be relieved, and the operating cost of the system can be minimized.

In order to study the permeability characteristics of cohesive soil in the consolidation process more accurately, in this work, the pores occupied by the combined water were defined as invalid pores, and the effective pores and ineffective pores were decoupled to study the relationship between effective pores and permeability coefficient. The widely used empirical formulas of the permeability coefficient were modified, and the empirical formulas of the permeability coefficient suitable for the consolidation process of cohesive soil were obtained. Combined with the e−lgσ relationships of normally consolidated soil, underconsolidated soil, and overconsolidated soil, the formulas of permeability coefficient, which considering the influence of initial consolidation state and consolidation stress, in the consolidation process of cohesive soil, were proposed. Finally, the rationality of the new formulas proposed in this paper was analyzed by indoor consolidation-permeability test and existing research. The results showed that the permeability coefficient of cohesive soil in the consolidation process predicted by the new formulas were more consistent with the measured average value. This study possesses practical engineering significance to solve the problem of foundation consolidation settlement.

Objective To evaluate the efficacy and safety of cilostazol, pentoxifylline, beraprost for intermittent claudication due to lower extremity arterial occlusive disease. Methods Randomized controlled clinical trials were identified from PubMed, Scopus, EMbase, Cochrane Library, Web of Science, China National Knowledge Infrastructure, SinoMed, Wanfang and Chongqing VIP databases, from the database inception to 31/12/2021. The outcome measures were walking distance measured by treadmill (maximum and pain-free walking distance), ankle-brachial index and adverse events. The quality of included studies was assessed by the Cochrane bias risk assessment tool. A network meta-analysis was carried out with Stata 16.0 software. Results There were 29 RCTs included in the study, covering total 5352 patients. Cilostazol was ranked first for both maximum and pain-free walking distance, followed by beraprost and pentoxifylline. For cilostazol, pentoxifylline and beraprost, maximum walking distance increased by 62.93 95%CI(44.06, 81.79), 32.72 95%CI(13.51, 55.79) and 43.90 95%CI(2.10, 85.71) meters, respectively relative to placebo, and pain-free walking distance increased by 23.92 95%CI(11.24, 36.61), 15.16 95%CI(2.33, 27.99) and 19.78 95%CI(-3.07, 42.62) meters. For cilostazol, pentoxifylline, beraprost and cilostazol combined with beraprost, ankle-brachial index increased by 0.06 95%CI(0.04, 0.07), -0.01 95%CI(-0.08, 0.05), 0.18 95%CI(0.12, 0.23) and 0.23 95%CI(0.18, 0.27), respectively relative to placebo. The pentoxifylline and cilostazol was associated with a lower ratio of adverse events than beraprost and cilostazol combined with beraprost. Conclusion Cilostazol, pentoxifylline and beraprost were all effective treatments for intermittent claudication; cilostazol with good tolerance was likely to be the most effective in walking distance, while beraprost and cilostazol combined with beraprost were more prominent in the ankle-brachial index.

Integrated electrical and heating systems (IEHSs) are promising for increasing the flexibility of power systems by exploiting the heat energy storage of pipelines. With the recent development of advanced communication technology, distributed optimization is employed in the coordination of IEHSs to meet the practical requirement of information privacy between different system operators. Existing studies on distributed optimization algorithms for IEHSs have seldom addressed packet loss during the process of information exchange. In this paper, a distributed paradigm is proposed for coordinating the operation of an IEHS considering communication packet loss. The relaxed alternating direction method of multipliers (R-ADMM) is derived by applying Peaceman-Rachford splitting to the Lagrangian dual of the primal problem. The proposed method is tested using several test systems in a lossy communication and transmission environment. Simulation results indicate the effectiveness and robustness of the proposed R-ADMM algorithm.

In order to obtain an accurate state estimation of the operation in the combined heat and power system, it is necessary to carry out state estimation. Due to the limited information sharing among various energy systems, it is practical to perform state estimation in a decentralized manner. However, the possible communication packet loss is seldomly considered among various energy systems. This paper bridges this gap by proposing a relaxed alternating direction method of multiplier algorithm. It can also improve the computation efficiency compared with the conventional alternating direction of the multiplier algorithm. Case studies of two test systems are carried out to show the validity and superiority of the proposed algorithm.

Background: Major depressive disorder (MDD) patients face an increased risk of developing cognitive impairments. One of the prominent cognitive impairments in MDD patients is verbal fluency deficit. Nonetheless, it is not clear which vulnerable brain region in MDD is interactively linked to verbal fluency deficit. It is important to gain an improved understanding for verbal fluency deficit in MDD.Methods: Thirty-four MDD patients and 34 normal controls (NCs) completed resting-state fMRI (rs-fMRI) scan and a set of verbal fluency tests (semantic VFT and phonemic VFT). Fourteen brain regions from five brain networks/systems (central executive network, default mode network, salience network, limbic system, cerebellum) based on their vital role in MDD neuropathology were selected as seeds for functional connectivity (FC) analyses with the voxels in the whole brain. Finally, correlations between the z-score of the FCs from clusters showing significant between-group difference and z-score of the VFTs were calculated using Pearson correlation analyses.Results: Increased FCs in MDD patients vs. NCs were identified between the bilateral posterior cingulate cortex (PCC) and the right inferior frontal gyrus (triangular part), in which the increased FC between the right PCC and the right inferior frontal gyrus (triangular part) was positively correlated with the z score of phonemic VFT in the MDD patients. Moreover, decreased FCs were identified between the right hippocampal gyrus and PCC, as well as left cerebellum Crus II and right parahippocampal gyrus in MDD patients vs. NCs.Conclusions: The MDD patients have altered FCs among key brain regions in the default mode network, the central executive network, the limbic system, and the cerebellum. The increased FC between the right PCC and the right inferior frontal gyrus (triangular part) may be useful to better characterize pathophysiology of MDD and functional correlates of the phonemic verbal fluency deficit in MDD.

Peroxiredoxins are a family of antioxidant proteins that protect cells from oxidative damage caused by reactive oxygen species (ROS). Herein, the peroxiredoxin 3 gene from grass carp (Ctenopharyngodon idellus), named CiPrx3, was cloned and analyzed. The full-length cDNA of CiPrx3 is 1068 bp long, with a 753 bp open reading frame (ORF) that contains a thioredoxin-2 domain, two peroxiredoxin signature motifs, and two highly conserved cysteine residues. CiPrx3 was ubiquitously expressed in all the tested tissues, while its expression level was altered significantly after exposure to grass carp reovirus (GCRV) and pathogen-associated molecular patterns (PAMPs). CiPrx3 was localized in the mitochondria of transfected cells and concentrated in the nucleus after poly (I:C) treatment. Transformation of CiPrx3 into Escherichia coli enhanced host resistance to H2O2 and heavy metals. Purified recombinant CiPrx3 proteins could protect DNA against oxidative damage. Overexpression of CiPrx3 in fish cells reduced intracellular ROS, increased cell viability, and decreased cell apoptosis caused by H2O2 stimulation and GCRV infection. Further study indicated that CiPrx3 induced autophagy to inhibit GCRV replication in fish cells. Collectively, these results imply that grass carp Prx3 elevates host antioxidant activity and induces autophagy to inhibit GCRV replication.

In the data acquisition and processing of Unmanned Aerial Vehicle(UAV) oblique photography monomer modeling, it is hard to balance the model quality and the production efficiency. This research applied the Analytic Hierarchy Process (AHP) evaluation method in the field of systems engineering to the management and decision of UAV oblique photography monomer modeling. Firstly, the AHP model is constructed by the expert survey method, and the relative weights of 6 evaluation indicators in the first hierarchies and the comprehensive ones of 3 in the second hierarchies are calculated. Then, each index data of different photo modeling schemes were collected and processed through experiments, and the AHP model was used to systematically evaluate each modeling scheme. Finally, the quadratic function is constructed with two variables, the number of photos and the AHP system score, meanwhile obtaining the optimal scheme by calculation. This is a useful attempt to apply the scientific evaluation method in the field of systems engineering to the production management of UAV aerial surveys. In this way, the efficiency of internal and external data collection and processing can be maximally improved while guaranteeing modeling accuracy.

Objective: To retrospectively investigate the comparative efficacy, safety and cost-benefits of three nutritional treatment schemes including short peptide jejunal nutrition (SPJN), whole protein jejunal nutrition (WPJN), and partial parenteral nutrition (PPN) in patients underwent esophagectomy for esophageal cancer in our hospital. This study was carried out in accordance with the conceptual framework of nutritional therapy in fast-track rehabilitation surgery.Methods: We retrospectively reviewed 305 patients who were assigned to receive esophagectomy for esophageal cancer. Eligible patients was naturally divided into SPJN group [n = 98 (32.1%)], WPJN group [n = 95 (31.1%)], and PPN group [n = 112 (36.7%)] according to the type of nutritional scheme which was actually prescribed to patients by the attendingphysician in clinical practice. The differences of the serum total protein (TP), albumin (ALB), pre-albumin (PA), hemoglobin (HGB), white blood cells (WBCs), red blood cells (RBCs) and neutrophils were compared among 3 nutritional schemes groups. We also investigated the relationship of the fluid intake, urine output, gastric juice drainage volume and thoracic drainage volume among 3 nutritional groups at 3 days after surgery. Moreover, the differences of cost-benefit indexes, complications, length of hospitalization and hospital expenditure were also compared.Results: The serum TP, ALB, and PA in the SPJN group were all higher than those in the WPJN and PPN groups (p < 0.05). The gastric juice volume of gastrointestinal decompression drainage and fluid volume of thoracic drainage in the SPJN group were all less than that in the WPJN group (p < 0.05). The overall hospital stay and post-operative hospital stay in the SPJN group were all shorter than that in WPJN group (p < 0.05). Moreover, the incidence of post-operative complications including anastomotic leakage, infection, and gastrointestinal reaction was remarkably lower in the SPJN group compared to the WPJN group (p < 0.05). Interesting, hospital expenditure in the PPN group was less than that in the SPJN and the WPJN groups (p < 0.001).Conclusion: Patients may obtain benefits in improving protein level after receiving SPJN scheme at the early stage after esophagectomy. Meanwhile, patients may obtain benefits in improving post-operative complications and hospital stay after receiving SPJN or PPN compared to WPJN protocol. However, the difference between SPJN and PPN requires further study because no difference was detected in terms of clinical outcomes including complications and the length of hospitalization although PPN may achieve a possible decrease of medical expenditure.

Grass carp (Ctenopharyngodon idellus) is an important aquaculture species in China that is affected by serious diseases, especially hemorrhagic disease caused by grass carp reovirus (GCRV). Grass carp have previously shown age-dependent susceptibility to GCRV, however, the mechanism by which this occurs remains poorly understood. Therefore, we performed transcriptome and metabolome sequencing on five-month-old (FMO) and three-year-old (TYO) grass carp to identify the potential mechanism. Viral challenge experiments showed that FMO fish were susceptible, whereas TYO fish were resistant to GCRV. RNA-seq showed that the genes involved in immune response, antigen presentation, and phagocytosis were significantly upregulated in TYO fish before the GCRV infection and at the early stage of infection. Metabolome sequencing showed that most metabolites were upregulated in TYO fish and downregulated in FMO fish after virus infection. Intragroup analysis showed that arachidonic acid metabolism was the most significantly upregulated pathway in TYO fish, whereas choline metabolism in cancer and glycerophospholispid metabolism were significantly downregulated in FMO fish after virus infection. Intergroup comparison revealed that metabolites from carbohydrate, amino acid, glycerophospholipid, and nucleotide metabolism were upregulated in TYO fish when compared with FMO fish. Moreover, the significantly differentially expressed metabolites showed antiviral effects both in vivo and in vitro. Based on these results, we concluded that the immune system and host biosynthesis and metabolism, can explain the age-dependent viral susceptibility in grass carp.

The left and right hemispheres of the human brain are two connected but relatively independent functional modules; they show multidimensional asymmetries ranging from particular local brain unit properties to entire hemispheric connectome topology. To date, however, it remains largely unknown whether and how hemispheric functional hierarchical structures differ between hemispheres. In the present study, we adopted a newly developed resting-state (rs) functional connectivity (FC)-based gradient approach to evaluate hemispheric functional hierarchical structures and their asymmetries in right-handed healthy young adults. Our results showed an overall mirrored principal functional gradient between hemispheres, with the sensory cortex and the default-mode network (DMN) anchored at the two opposite ends of the gradient. Interestingly, the left hemisphere showed a significantly larger full range of the principal gradient in both males and females, with males exhibiting greater leftward asymmetry. Similarly, the principal gradient component scores of two regions around the middle temporal gyrus and posterior orbitofrontal cortex exhibited similar hemisphere × sex interaction effects: a greater degree of leftward asymmetry in males than in females. Moreover, we observed significant main hemisphere and sex effects in distributed regions across the entire hemisphere. All these results are reproducible and robust between test-retest rs-fMRI sessions. Our findings provide evidence of functional gradients that enhance the present understanding of human brain asymmetries in functional organization and highlight the impact of sex on hemispheric functional gradients and their asymmetries.

The major barrier to curing HIV-1 infection is a small pool of latently infected cells that harbor replication-competent viruses, which are widely considered the origin of viral rebound when antiretroviral therapy (ART) is interrupted. The difficulty in distinguishing latently infected cells from the vast majority of uninfected cells has represented a significant bottleneck precluding comprehensive understandings of HIV-1 latency. Here we reported and validated a newly designed dual fluorescent reporter virus, DFV-B, infection with which primary CD4+ T cells can directly label latently infected cells and generate a latency model that was highly physiological relevant. Applying DFV-B infection in Jurkat T cells, we generated a stable cell line model of HIV-1 latency with diverse viral integration sites. High-throughput compound screening with this model identified ACY-1215 as a potent latency reversing agent, which could be verified in other cell models and in primary CD4+ T cells from ART-suppressed individuals ex vivo. In summary, we have generated a meaningful and feasible model to directly study latently infected cells, which could open up new avenues to explore the critical events of HIV-1 latency and become a valuable tool for the research of AIDS functional cure.

Abstract The white coat colour of Yorkshire and Landrace pig breeds is caused by the dominant white I allele of KIT, associated with 450-kb duplications and a splice mutation (G > A) at the first base in intron 17. To test whether genome editing can be employed to correct this structural mutation, and to investigate the role of KIT in the control of porcine coat colour, we designed sgRNAs targeting either intron 16 or intron 17 of KIT, and transfected Cas9/sgRNA co-expression plasmids into the kidney cells of Yorkshire pigs. The copy number of KIT was reduced by about 13%, suggesting the possibility of obtaining cells with corrected structural mutations of the KIT locus. Using single cell cloning, from 24 successfully expanded single cell clones derived from cells transfected with sgRNA targeting at intron 17, we obtained 3 clones with a single copy of KIT without the splice mutation. Taken together, the 12.5% (3/24) efficiency of correction of structural mutations of 450 kb fragments is highly efficient, providing a solid basis for the generation of genome edited Yorkshire pigs with a normal KIT locus. This provides an insight into the underlying genetic mechanisms of porcine coat colour.

Abstract Objective This study was designed to verify the association between dementia and mortality in the elderly undergoing hip fracture surgery, and assessed the mortality of patients with dementia after hip fracture surgery. Material and methods PubMed, Embase, and Web of Science were searched until April, 2018 without language restrictions. Two reviewers selected related studies, assessed study quality, and extracted data independently. Risk ratios (RRs) with 95% confidence intervals (CI) were derived using random-effects model throughout all analyses. The endpoints included 30-day, 6-month, 1-year, and more than 1-year mortality. Major depressive disorder (MDD) represents a grand challenge to human health and society, but the underlying pathophysiological mechanisms remain elusive. Previous neuroimaging studies have suggested that MDD is associated with abnormal interactions and dynamics in two major neural systems including the default mode - salience (DMN-SAL) network and the executive - limbic (EXE-LIM) network, but it is not clear which network plays a central role and which network plays a subordinate role in MDD pathophysiology. To address this question, we refined a newly developed Multiscale Neural Model Inversion (MNMI) framework and applied it to test whether MDD is more affected by impaired circuit interactions in the DMN-SAL network or the EXE-LIM network. The model estimates the directed connection strengths between different neural populations both within and between brain regions based on resting-state fMRI data collected from normal healthy subjects and patients with MDD. Results show that MDD is primarily characterized by abnormal circuit interactions in the EXE-LIM network rather than the DMN-SAL network. Specifically, we observe reduced frontoparietal effective connectivity that potentially contributes to hypoactivity in the dorsolateral prefrontal cortex (dlPFC), and decreased intrinsic inhibition combined with increased excitation from the superior parietal cortex (SPC) that potentially lead to amygdala hyperactivity, together resulting in activation imbalance in the PFC-amygdala circuit that pervades in MDD. Moreover, the model reveals reduced PFC-to-hippocampus excitation but decreased SPC-to-thalamus inhibition in MDD population that potentially lead to hypoactivity in the hippocampus and hyperactivity in the thalamus, consistent with previous experimental data. Overall, our findings provide strong support for the long-standing limbic-cortical dysregulation model in major depression but also offer novel insights into the multiscale pathophysiology of this debilitating disease.

Functional integration among neural units is one of the fundamental principles in brain organization that could be examined using resting-state functional connectivity (rs-FC). Interhemispheric functional integration plays a critical role in human cognition. Homotopic functional connectivity (HoFC) under resting state provide an avenue to investigate functional integration between the two brain hemispheres, which can improve the present understanding of how interhemispheric interactions affect cognitive processing. In this review, we summarize the progress of HoFC studies under resting state and highlight how these findings have enhanced our understanding of interhemispheric functional organization of the human brain. Future studies are encouraged to address particular methodological issues and to further ascertain behavioral correlates, brain disease’s modulation, task influence, and genetic basis of HoFC.

Background: Major depressive disorder (MDD) is one of the most common and costly psychiatric disorders. In addition to significant changes in mood, MDD patients face an increased risk of developing cognitive dysfunction. It is important to gain an improved understanding of cognitive impairments and find a biomarker for cognitive impairment diagnosis in MDD.Methods: One hundred MDD patients and 100 normal controls (NCs) completed resting-state fMRI (rs-fMRI) scan, in which 34 MDD patients and 34 NCs had scores in multiple cognitive domains (executive function, verbal fluency, and processing speed). Twenty-seven regions of interest from the default mode network (DMN), central executive network (CEN), salience network (SN), and limbic system (LS) were selected as seeds for functional connectivity (FC) analyses with the voxels in the whole brain. Finally, partial correlations were conducted for cognitive domain scores and FCs with significant differences between the MDD and NC groups.Results: Significant FC differences between groups were identified among the seeds and clusters in the DMN, CEN, LS, visual network, somatomotor network, ventral attention network, and dorsal attention network. In the MDD patients, the magnitude of the Stroop interference effect was positively correlated with the illness duration, and the illness duration was negatively correlated with the FC between the right ventral hippocampal gyrus and the left inferior frontal gyrus. However, the correlation between the Stroop interference effect and the FC of the right anterior prefrontal cortex with the left cerebellum_4_5 was disrupted in these patients.Conclusions: The MDD patients have altered FCs among multiple brain networks and a disrupted correlation between the FC of prefrontal cortex and executive function. The disrupted correlation could present before the symptoms develop and may be the core process in the development of executive function impairment.

The dominant white phenotype in pigs is thought to be mainly due to a structural mutation in the KIT gene, a splice mutation (G > A) at the first base in intron 17 which leads to the deletion of exon 17 in the mature KIT mRNA. However, this hypothesis has not yet been validated by functional studies. Here, we created two mouse models, KIT D17/+ to mimic the splice mutation, and KIT Dup/+ to partially mimic the duplication mutation of KIT gene in dominant white pigs using CRISPR/Cas9 technology. We found that the splice mutation homozygote is lethal and the heterozygous mice have a piebald coat. Slightly increased expression of KIT in KIT Dup/+mice did not confer the patched phenotype and had no obvious impact on coat color. Interestingly, the combination of these two mutations reduced the phosphorylation of PI3K and MAPK pathway associated proteins, which may be related to the impaired migration of melanoblasts observed during embryonic development that eventually leads to the dominant white phenotype.

In a wireless ad hoc network, the size of the virtual backbone (VB) is an important factor for measuring the quality of the VB. The smaller the VB is, the less the overhead caused by the VB. Since ball graphs (BGs) have been used to model 3-dimensional wireless ad hoc networks and since a connected dominating set can be used to represent a VB undertaking routing-related tasks, the problem of finding the smallest VB is transformed into the problem of finding a minimum connected dominating set (MCDS). Many research results on the MCDS problem have been obtained for unit disk graphs and unit ball graphs, in which the transmission ranges of all nodes are identical. In some situations, the node powers can vary. One can model such a network as a graph with different transmission ranges for different nodes. In this paper, we focus on the problem of minimum strongly connected dominating and absorbing sets (MSCDASs) in a strongly connected directed ball graph with different transmission ranges, which is also NP-hard. We design an algorithm considering the construction of a strongly connected dominating and absorbing set (SCDAS), whose size does not exceed 319/15k3+116/5k2+29/5kopt+29/3k3+116/5k2+87/5k+13/15, where opt is the size of an MCDAS and k denotes the ratio of rmax to rmin in the ad hoc network with transmission range rmin,rmax. Our simulations show the feasibility of the algorithm proposed in this paper.

This paper studies and introduces the successful case of gob-side entry retaining technology and the typical mining pressure law in Luan mining area, which is the main mining coal seam in Qinshui coalfield. Qinshui coalfield has an estimated coal reserve of 300 billion tons, accounting for 9.58% of the total national coal reserve in China, especially anthracite, chemical coal, and coking coal. The methods of field investigation, theoretical analysis, physical experiment, and industrial test are adopted. Through the field investigation, theoretical analysis, physical experiments, and industrial test, the following conclusions have been drawn in this study: (1) A thorough engineering geological investigation was conducted on the entry retaining along the gob side on noncoal pillar mining working face, which covers multiple periods of mining process including the roadway excavation period, primary mining period, primary mining stability period, and secondary mining influence period. A series of analysis and tests were conducted such as core sampling, rock mechanics property testing, borehole detection, and flexible formwork support evaluation, which laid a foundation for identifying the mining pressure law of gob-side entry retaining by using noncoal pillar mining. (2) The mining pressure law was studied through the collection of the field measurements taken from the entry retaining along the gob side on noncoal pillar mining working face. The keys to achieve the roadway surrounding rock stability through noncoal pillar mining are obtained. According to the study, the stability control of retained roadway surrounding rock mainly depends on the stability of top coal, coal side, and shoulder angle coal. (3) In this study, a roadway reinforcement scheme is proposed to improve the surrounding rock control technology for gob-side entry retaining by noncoal pillar mining, whose effectiveness has been verified by a series of industrial test. Therefore, the wide adoption of the noncoal pillar mining method in Number #3 coal mine can significantly relieve the predicament of coal pressing under a large number of buildings in Luan mining area, which provides insightful guidance to the coal-free pillar mining in the whole Luan mining area.

Abstract Background Platelet glycoprotein Ibα (GPIbα) extracellular domain, which is part of the receptor complex GPIb-IX-V, plays an important role in tumor metastasis. However, the mechanism through which GPIbα participates in the metastatic process remains unclear. In addition, potential bleeding complication remains an obstacle for the clinical use of anti-platelet agents in cancer therapy. Methods We established a series of screening models and obtained rat anti-mouse GPIbα monoclonal antibodies (mAb) 1D12 and 2B4 that demonstrated potential value in suppressing cancer metastasis. To validate our findings, we further obtained mouse anti-human GPIbα monoclonal antibody YQ3 through the same approach. Results 1D12 and 2B4 affected the von Willebrand factor (vWF)-GPIbα interaction via binding to GPIbα aa 41-50 and aa 277-290 respectively, which markedly inhibited the interaction among platelets, tumor cells, and endothelial cells in vitro, and reduced the mean number of surface nodules in the experimental and spontaneous metastasis models in vivo. As expected, YQ3 inhibited lung cancer adhesion and demonstrated similar value in metastasis. More importantly, for all three mAbs in our study, none of their Fabs induced thrombocytopenia. Conclusion Our results therefore supported the hypothesis that GPIbα contributes to tumor metastasis and suggested potential value of using anti-GPIbα mAb to suppress cancer metastasis.
