After undergoing 75% hot deformation and 40% cold deformation, a variant of TWIP (Twin Induced Plasticity) steel with a composition of Fe–22Mn−2.7Al−0.95C−0.75Si (wt.%) was subjected to annealing at various temperatures (850 °C, 650 °C, 550 °C) for 20min. The recovery annealed C550 sample exhibited a yield strength of 1289.20 MPa, a tensile strength of 1442.29 MPa and an elongation of 30.78%. Characterization and comparison of the samples were conducted using XRD, SEM, EBSD, TEM and uniaxial tensile tests. The yield strength of the sample was assessed through different strengthening mechanisms to analysis the exceptional properties of the recovery annealed sample. Experimental findings indicate that the high-density dislocation and the refinement of slip bands in the recovery annealed C550 samples contribute to their superior performance compared to the partially and fully recrystallized samples.

Abstract Background Early prediction of pathological complete response (pCR) is important for deciding appropriate treatment strategies for patients. In this study, we aimed to quantify the dynamic characteristics of dynamic contrast-enhanced magnetic resonance images (DCE-MRI) and investigate its value to improve pCR prediction as well as its association with tumor heterogeneity in breast cancer patients. Methods The DCE-MRI, clinicopathologic record, and full transcriptomic data of 785 breast cancer patients receiving neoadjuvant chemotherapy were retrospectively included from a public dataset. Dynamic features of DCE-MRI were computed from extracted phase-varying radiomic feature series using 22 CAnonical Time-sereis CHaracteristics. Dynamic model and radiomic model were developed by logistic regression using dynamic features and traditional radiomic features respectively. Various combined models with clinical factors were also developed to find the optimal combination and the significance of each components was evaluated. All the models were evaluated in independent test set in terms of area under receiver operating characteristic curve (AUC). To explore the potential underlying biological mechanisms, radiogenomic analysis was implemented on patient subgroups stratified by dynamic model to identify differentially expressed genes (DEGs) and enriched pathways. Results A 10-feature dynamic model and a 4-feature radiomic model were developed (AUC = 0.688, 95%CI: 0.635–0.741 and AUC = 0.650, 95%CI: 0.595–0.705) and tested (AUC = 0.686, 95%CI: 0.594–0.778 and AUC = 0.626, 95%CI: 0.529–0.722), with the dynamic model showing slightly higher AUC (train p = 0.181, test p = 0.222). The combined model of clinical, radiomic, and dynamic achieved the highest AUC in pCR prediction (train: 0.769, 95%CI: 0.722–0.816 and test: 0.762, 95%CI: 0.679–0.845). As advanced silicon manufacturing processes slow down in scaling, three-dimensional stacking of chips has become a future trend in microelectronic industry. High-density bonding technology is one of the core technologies used to achieve the stacking. In this work, we explored a new bonding technology that is possible to manufacture future high-density interconnects. We fabricated a series of sandwich structure solder joints with different thicknesses using SnBiIn-based nanoparticles under a low bonding temperature of 100 °C. The intermetallic compound (IMC) at the interface is proven to be η′-Cu6(Sn, In)5 nanocrystals with random grain orientations through X-ray diffraction (XRD), high-resolution transmission electron microscope (HRTEM), and atomic probe tomography (APT). The hardness and elastic modulus of η′-Cu6(Sn, In)5 were measured to be 4.093 GPa and 79.586 GPa by micropillar compression tests, respectively, decreasing by 35% and 29.1% compared to Cu6Sn5. Therefore, doping In atoms reduces the brittleness of IMCs, providing a new approach for mitigating the mechanical failures of full IMC bump. First-principles calculation also verified the change in mechanical properties. Furthermore, a full-Cux(Sn, In)y IMCs solder joint and an array of 3 μm solder joints were fabricated experimentally using the bonding technology. This technology successfully demonstrates a pioneer interconnection bonding methodology for future ultra-high-density chip stacking using low melting point solder nanoparticles.

To investigate the coupling strengthening mechanism of precipitate and Cu solute atoms in Al-Mg-Si alloy, the molecular dynamics model of edge dislocation motion interacted with precipitate and Cu solute atoms was established. The dislocation length is increased in the dislocation-precipitate interaction due to the bending and bowing of the edge dislocation around the precipitate. No coupling between precipitate and solute can be found in material strengthening in cases with lower solute concentrations. The exponential effect of the coupling effects between precipitate and solute on material strengthening is found when the solute concentration is increased above 0.3.at% at room temperature. The change of the edge dislocation caused by solute atoms around the precipitate is the key reason for the strengthening coupling effect. Furthermore, the effects of temperature variations on the coupling effect are more pronounced than the effects of shear strain rate variations. When the temperature is increased from 200K to 500K, the critical ratio of solute and precipitate strengthening is decreased from 1.5 to 0.4.

Abstract Phase-to-height mapping is one of the important processes in three dimensional phase measurement profilometry. But, in traditional phase-to-height mapping method, the measurement accuracy is affected by device attitude, so it needs saving a large amount of mapping equations to achieve high-quality phase-to-height mapping. In order to improve that, this paper proposes an improved phase-to-height mapping method combine with device attitude. Firstly, we get the unwrapped phase of the target. Then, using generalized regression neural network is used to reduce the offset of phase information at the same height due to the randomness of device attitude. Last, the phase-to-height mapping is completed by substituting the unwrapped phase (the difference between having detected object and no detected object) of eliminate the offset into improved phase-to-height mapping method. Experimental results show that the proposed method could achieve high-quality phase-to-height mapping with less mapping equation and less memory space. Compared with the nonlinear phase-to-height mapping method (probabilistic neural network to eliminate phase offset), its accuracy is improved by 44.30%. Compared with the nonlinear phase-to-height mapping method (radial basis function neural network to eliminate phase offset), the accuracy is improved by 39.58%.

Abstract The phenomenon of semantic satiation, which refers to the loss of meaning of a word or phrase after being repeated many times, is a well-known psychological phenomenon. However, the microscopic neural computational principles responsible for these mechanisms remain unknown. In this study, we use a deep learning model of continuous coupled neural networks to investigate the mechanism underlying semantic satiation and precisely describe this process with neuronal components. Our results suggest that, from a mesoscopic perspective, semantic satiation may be a bottom-up process. Unlike existing macroscopic psychological studies that suggest that semantic satiation is a top-down process, our simulations use a similar experimental paradigm as classical psychology experiments and observe similar results. Satiation of semantic objectives, similar to the learning process of our network model used for object recognition, relies on continuous learning and switching between objects. The underlying neural coupling strengthens or weakens satiation. Taken together, both neural and network mechanisms play a role in controlling semantic satiation.

Abstract As a critical component of space-air-ground integrated IoT, the aerial network provides highly reliable, low-latency and ubiquitous information services to ground users by virtue of their high mobility, easy deployment and low cost. However, the current computation and resource management model of air-ground integrated networks are insufficient to meet the latency demanding of emerging intelligent services such as autonomous systems, extended reality and haptic feedback. To tackle these challenges, we propose a computation offloading and optimization method based on potential game. First, we construct an cloud-edge collaborative computing model. Secondly, we construct Offloading Decision Objective Functions (ODOF) with the objective of minimum task processing latency and energy consumption. ODOF is proved to be a Mixed Inferior Nonlinear Programming (MINLP) problem, which is hard to solve. ODOF is converted to be a full potential game, and the Nash equilibrium solution exists. Then, a computational resource allocation algorithm based on Karush–Kuhn–Tucker (KKT) conditions is proposed to solve resource allocation problem. On this basis, a distributed game-based computational offloading algorithm is proposed to minimize the offloading cost. Extensive simulation results demonstrate that the convergence performance of the proposed algorithm is reduced by 50%, the convergence time is reduced by 13.3% and the average task processing delay is reduced by 10%.

Abstract Background Melanoma is the most lethal skin cancer characterized by its high metastatic potential. In the past decade, targeted and immunotherapy have brought revolutionary survival benefits to patients with advanced and metastatic melanoma, but these treatment responses are also heterogeneous and/or do not achieve durable responses. Therefore, novel therapeutic strategies for improving outcomes remain an unmet clinical need. The aim of this study was to evaluate the therapeutic potential and underlying molecular mechanisms of RC48, a novel HER2-target antibody drug conjugate, either alone or in combination with dabrafenib, a V600-mutant BRAF inhibitor, for the treatment of advanced BRAF-mutant cutaneous melanoma. Methods We evaluated the therapeutic efficacy of RC48, alone or in combination with dabrafenib, in BRAF-mutant cutaneous melanoma cell lines and cell-derived xenograft (CDX) models. We also conducted signaling pathways analysis and global mRNA sequencing to explore mechanisms underlying the synergistic effect of the combination therapy. Results Our results revealed the expression of membrane-localized HER2 in melanoma cells. RC48 effectively targeted and inhibited the growth of HER2-positive human melanoma cell lines and corresponding CDX models. When used RC48 and dabrafenib synergically induced tumor regression together in human BRAF-mutant melanoma cell lines and CDX models. Mechanically, our results demonstrated that the combination therapy induced apoptosis and cell cycle arrest while suppressing cell motility in vitro. Furthermore, global RNA sequencing analysis demonstrated that the combination treatment led to the downregulation of several key signaling pathways, including the PI3K-AKT pathway, MAPK pathway, AMPK pathway, and FOXO pathway. Conclusion These findings establish a preclinical foundation for the combined use of an anti-HER2 drug conjugate and a BRAF inhibitor in the treatment of BRAF-mutant cutaneous melanoma.

Abstract Background Laryngeal squamous cell carcinoma (LSCC) is one of the most common malignant tumors of the head and neck. Vasculogenic mimicry (VM) is crucial for tumor growth and metastasis and refers to the formation of fluid channels by invasive tumor cells rather than endothelial cells. However, the regulatory mechanisms underlying VM during the malignant progression of LSCC remain largely unknown. Methods Gene expression and clinical data for LSCC were obtained from the TCGA and Gene GEO (GSE27020) databases. A risk prediction model associated with VM was established using LASSO and Cox regression analyses. Based on their risk scores, patients with LSCC were categorized into high- and low-risk groups. The disparities in immune infiltration, tumor mutational burden (TMB), and functional enrichment between these two groups were examined. The core genes in LSCC were identified using the machine learning (SVM-RFE) and WGCNA algorithms. Subsequently, the involvement of bone morphogenetic protein 2 (BMP2) in VM and metastasis was investigated both in vitro and in vivo. To elucidate the downstream signaling pathways regulated by BMP2, western blotting was performed. Additionally, ChIP experiments were employed to identify the key transcription factors responsible for modulating the expression of BMP2. Results We established a new precise prognostic model for LSCC related to VM based on three genes: BMP2, EPO, and AGPS. The ROC curves from both TCGA and GSE27020 validation cohorts demonstrated precision survival prediction capabilities, with the nomogram showing some net clinical benefit. Multiple algorithm analyses indicated BMP2 as a potential core gene. Further experiments suggested that BMP2 promotes VM and metastasis in LSCC. The malignant progression of LSCC is promoted by BMP2 via the activation of the PI3K-AKT signaling pathway, with the high expression of BMP2 in LSCC resulting from its transcriptional activation by runt-related transcription factor 1 (RUNX1). Conclusion BMP2 predicts poor prognosis in LSCC, promotes LSCC VM and metastasis through the PI3K-AKT signaling pathway, and is transcriptionally regulated by RUNX1. BMP2 may be a novel, precise, diagnostic, and therapeutic biomarker of LSCC.

Abstract Background Despite the rapid advances in modern medical technology, kidney renal clear cell carcinoma (KIRC) remains a challenging clinical problem in urology. Researchers urgently search for useful markers to break through the therapeutic conundrum due to its high lethality. Therefore, the study explores the value of ADH5 on overall survival (OS) and the immunology of KIRC. Methods The gene expression matrix and clinical information on ADH5 in the TCGA database were validated using external databases and qRT-PCR. To confirm the correlation between ADH5 and KIRC prognosis, univariate/multivariate Cox regression analysis was used. We also explored the signaling pathways associated with ADH5 in KIRC and investigated its association with immunity. Results The mRNA and protein levels showed an apparent downregulation of ADH5 in KIRC. Abstract The structural characteristics of plant communities in urban green spaces have a significant impact on their carbon sequestration function. In this study, comprehensive data were collected from 106 plant communities (each 20 m × 20 m) in Zhengzhou Green Expo Park. We assessed aboveground and soil carbon storage, alongside maintenance carbon emissions, to quantify carbon dynamics. Our primary objective was to establish a statistical model that correlates the structural attributes of plant communities with their total annual carbon sequestration. This model aims to provide a quantitative framework for optimizing community structures to maximize carbon sequestration in urban green spaces. The results showed that density and coverage were significantly and positively correlated with aboveground and soil carbon stocks. Density and mean height were significantly and positively correlated with maintenance carbon emissions. Density played a key structural role in regulating the total carbon sequestration of the plant communities, being 27.24 times more effective than coverage. The total annual carbon sequestration of the plant community reached an optimal value of 327.67 kg CO2-eq/y−1 at a density and cover of 0.15 and 1, respectively. This study provides valuable data for increasing the carbon sink ability of urban green spaces through plant structure regulation and supporting low-carbon development strategies in urban management.

Abstract It is a long-standing scientific controversy to achieve anti-Kasha-type multiple emissions by tuning the structures at a molecular level. Although it is known that some conjugated structures have excitation-dependent multiple emissions, no all-benzenoid molecules have yet been reported, the emissions of which originate from different excited states. Herein, we report the design of two symmetry-breaking heterogeneous carbon bisnanohoops that in solution become multiple fluorescent emitters with unusual anti-Kasha characteristics. This phenomenon can be spectroscopically and theoretically explained and will find applications in a wide range of sensing and imaging technologies.

Abstract In this study, the corrosion behavior of AlMn lightweight weathering steel (LWS) in the simulated marine atmosphere was investigated by means of the dry/wet corrosion cycle test. The results showed that Al was present as FeAl2O4 and enriched in the inner layer, which significantly optimizes the rust layer in terms of compactness, elemental distribution, phase constitution, and electrochemical properties. The Mn oxides promoted the formation of FeAl2O4 and enhanced the anti–rupture ability of the LWS’s rust layer.

Abstract Craniofacial malformations, often associated with syndromes, are prevalent birth defects. Emerging evidence underscores the importance of m6A modifications in various bioprocesses such as stem cell differentiation, tissue development, and tumorigenesis. Here, in vivo, experiments with zebrafish models revealed that mettl3-knockdown embryos at 144 h postfertilization exhibited aberrant craniofacial features, including altered mouth opening, jaw dimensions, ethmoid plate, tooth formation and hypoactive behavior. Similarly, low METTL3 expression inhibited the proliferation and migration of BMSCs, HEPM cells, and DPSCs. Loss of METTL3 led to reduced mRNA m6A methylation and PSEN1 expression, impacting craniofacial phenotypes. Co-injection of mettl3 or psen1 mRNA rescued the level of Sox10 fusion protein, promoted voluntary movement, and mitigated abnormal craniofacial phenotypes induced by mettl3 knockdown in zebrafish. Mechanistically, YTHDF1 enhanced the mRNA stability of m6A-modified PSEN1, while decreased METTL3-mediated m6A methylation hindered β-catenin binding to PSEN1, suppressing Wnt/β-catenin signaling. Pharmacological activation of the Wnt/β-catenin pathway partially alleviated the phenotypes of mettl3 morphant and reversed the decreases in cell proliferation and migration induced by METTL3 silencing. This study elucidates the pivotal role of METTL3 in craniofacial development via the METTL3/YTHDF1/PSEN1/β-catenin signaling axis.

Abstract Whether in the realms of computer vision, robotics, or environmental monitoring, the ability to monitor and follow specific targets amidst intricate surroundings is essential for numerous applications. However, achieving rapid and efficient target tracking remains a challenge. Here we propose an optical implementation for rapid tracking with negligible digital post-processing, leveraging an all-optical information processing. This work combines a diffractive-based optical nerual network with a layered liquid crystal electrical addressing architecture, synergizing the parallel processing capabilities inherent in light propagation with liquid crystal dynamic adaptation mechanism. Through a one-time effort training, the trained network enable accurate prediction of the desired arrangement of liquid crystal molecules as confirmed through numerical blind testing. Then we establish an experimental camera architecture that synergistically combines an electrically-tuned functioned liquid crystal layer with materialized optical neural network. With integrating the architecture into optical imaging path of a detector plane, this optical computing camera offers a data-driven diffractive guidance, enabling the identification of target within complex backgrounds, highlighting its high-level vision task implementation and problem-solving capabilities.

China is one of the countries with the fastest growing prevalence of diabetes in the world, and also has the largest number of people with diabetes. The "Health China 2030" plan clearly proposes to further prevent and control major diseases, implement diabetes prevention and control, and guide patients with diabetes to strengthen health management and delay or prevent the development of diabetes. In the past 20 years, value co-creation theory has been well developed in the field of health services, and different countries (regions) have explored programs to prevent and control diabetes at low cost and high effectiveness, but it is currently lacking in the field of chronic disease management in China. Based on the perspective of value co-creation, this paper further summarizes the domestic and international experience of chronic disease management taking community as the research and analysis point, proposes the prospect from four aspects of active health, organizational guidance, digital technology, and incentive assessment, which provides references for optimizing and improving the service system of diabetes management at primary health care, establishing a model of collaborative participation of multiple subjects in community chronic disease management based on value co-creation, and realizing co-creation, co-management and sharing of chronic disease management.

ObjectiveThis study investigates the interactive collision avoidance （CA） problem of autonomous and manned ships in mixed navigation scenarios in order to achieve the efficient collision avoidance and safe navigation of ships. To this end, a multi-agent interactive ship dynamic game collision avoidance decision-making method is proposed. MethodsAccording to sailing practices, the ship collision avoidance problem in mixed navigation environments is analyzed and understood, and the ship encounter situation and collision risk are quantified on the basis of the International Regulations for Preventing Collisions at Sea (COLREGs). Dynamic game theory is introduced to model individual ships with collision risk as participants with independent thinking in the game, and the course changes of ships are taken as the strategy for determining the optimal action sequence under the constraints of ship safety and social and economic benefits. Different ship maneuvering modes are introduced to the simulation experiment, and the effectiveness of the collision avoidance decision-making method is verified using Python. ResultsThe results show that this method can realize the safe encounter of multiple ships in a mixed navigation environment of autonomous and manned ships, and each ship can adjust its behavior strategy to achieve safe avoidance when facing target ships with different sailing styles. ConclusionThis study can provide valuable references for the collision avoidance decision-making of autonomous and manned ships.

Abstract B-cell subsets in peripheral blood (PB) and tumor microenvironment (TME) were evaluated to determine myasthenia gravis (MG) severity in patients with thymoma-associated MG (TMG) and the distribution of B cells in type B TMG. The distribution of mature B cells, including Bm1–Bm5, CD19+ and CD20+ B cells and non-switched (NSMBCs) and switched (SMBCs) memory B cells, were determined in 79 patients with thymoma or TMG. Quantitative relationships between the T and TMG groups and the TMG-low and TMG-high subgroups were determined. NSMBCs and SMBCs were compared in TME and PB. Type B thymoma was more likely to develop into MG, with types B2 and B3 being especially associated with MG worsening. The percentage of CD19+ B cells in PB gradually increased, whereas the percentage of CD20+ B cells and the CD19/CD20 ratio were not altered. The (Bm2 + Bm2′)/(eBm5 + Bm5) index was significantly higher in the TMG-high than in thymoma group. The difference between SMBC/CD19+ and NSMBC/CD19+ B cell ratios was significantly lower in the thymoma than TMG group. NSMBCs assembled around tertiary lymphoid tissue in thymomas of patients with TMG. Few NSMBCs were observed in patients with thymoma alone, with these cells being diffusely distributed. MG severity in patients with TMG can be determined by measuring CD19+ B cells and Bm1-Bm5 in PB. The CD19/CD20 ratio is a marker of disease severity in TMG patients. Differences between NSMBCs and SMBCs in PB and TME of thymomas can synergistically determine MG severity in patients with TMG.

Abstract Diabetic nephropathy (DN), one of the more prevalent microvascular complications in patients diagnosed with diabetes mellitus, is attributed as the main cause of end-stage renal disease (ESRD). Lipotoxicity in podocytes caused by hyperglycemia has been recognised as a significant pathology change, resulting in the deterioration of the glomerular filtration barrier. Research has demonstrated how dapagliflozin, a kind of SGLT2i, exhibits a multifaceted and powerful protective effect in DN, entirely independent of the hypoglycemic effect, with the specific mechanism verified. In this present study, we found that dapagliflozin has the potential to alleviate apoptosis and restore cytoskeleton triggered by high glucose (HG) in vivo and in vitro. We also discovered that dapagliflozin could mitigate podocyte cholesterol accumulation by restoring the expression of ABCA1, which is the key pathway for cholesterol outflows. This research also mechanistically demonstrates that the protective effect of dapagliflozin can be mediated by KLF-5, which is the upstream transcription factor of ABCA1. Taken together, our data suggest that dapagliflozin offers significant potential in alleviating podocyte injury and cholesterol accumulation triggered by high glucose. In terms of the mechanism, we herein reveal that dapagliflozin could accelerate cholesterol efflux by restoring the expression of ABCA1, which is directly regulated by KLF-5.

Abstract Background Ovarian cancer (OC) is a prevalent malignancy in the female reproductive system, and developing effective targeted therapies for this disease remains challenging. The aim of this study was to use clinically-relevant OC models to evaluate the therapeutic effectiveness of RC48, an antibody-drug conjugate (ADC) targeting HER2, either alone or in combination with the VEGFR inhibitor Cediranib Maleate (CM), for the treatment of advanced OC. Methods OC tumor specimens and cell lines were analyzed to determine HER2 and VEGFR expression by Western blot, immunocytochemistry and immunofluorescence. Moreover, the OC cell lines, cell-derived xenograft (CDX) and patient-derived xenograft (PDX) models were treated with RC48 and/or CM and then subjected to cell proliferation, viability, apoptosis, and tumor growth analyses to evaluate the feasibility of combination therapy for OC both in vitro and in vivo. Additionally, RNA-Seq was performed to investigate the critical mechanism underlying the combination therapy of RC48 and CM. Results Our results demonstrated that RC48 alone effectively targeted and inhibited the growth of HER2-positive OC tumors in both cell lines and PDX models. Furthermore, the combination of RC48 and CM synergistically induced tumor regression in human OC cell lines, as well as CDX and PDX models. Mechanistically, we observed that the combination treatment inhibited the growth of OC cells involved inducing apoptosis and suppressing cell motility. RNA-seq analysis provided further mechanistic insights and revealed that co-administration of RC48 and CM downregulated multiple cancer-related pathways, including the AKT/mTOR pathway, cell cycle, and cell proliferation. Notably, our data further confirmed that the PI3K-AKT pathway played a key role in the inhibition of proliferation triggered by combinational treatment of RC48 and CM in OC cells. Conclusions These findings provide a preclinical framework supporting the potential of dual targeting HER2 and VEGFR as a promising therapeutic strategy to improve outcomes in patients with OC.

Abstract Background This study explores the effectiveness of Photobiomodulation Therapy (PBMT) in enhancing orthodontic tooth movement (OTM), osteogenesis, and angiogenesis through a comprehensive series of in vitro and in vivo investigations. The in vitro experiments involved co-culturing MC3T3-E1 and HUVEC cells to assess PBMT’s impact on cell proliferation, osteogenesis, angiogenesis, and associated gene expression. Simultaneously, an in vivo experiment utilized an OTM rat model subjected to laser irradiation at specific energy densities. Methods In vitro experiments involved co-culturing MC3T3-E1 and HUVEC cells treated with PBMT, enabling a comprehensive assessment of cell proliferation, osteogenesis, angiogenesis, and gene expression. In vivo, an OTM rat model was subjected to laser irradiation at specified energy densities. Statistical analyses were performed to evaluate the significance of observed differences. Results The results revealed a significant increase in blood vessel formation and new bone generation within the PBMT-treated group compared to the control group. In vitro, PBMT demonstrated positive effects on cell proliferation, osteogenesis, angiogenesis, and gene expression in the co-culture model. In vivo, laser irradiation at specific energy densities significantly enhanced OTM, angiogenesis, and osteogenesis. Conclusions This study highlights the substantial potential of PBMT in improving post-orthodontic bone quality. The observed enhancements in angiogenesis and osteogenesis suggest a pivotal role for PBMT in optimizing treatment outcomes in orthodontic practices. The findings position PBMT as a promising therapeutic intervention that could be seamlessly integrated into orthodontic protocols, offering a novel dimension to enhance overall treatment efficacy. Beyond the laboratory, these results suggest practical significance for PBMT in clinical scenarios, emphasizing its potential to contribute to the advancement of orthodontic treatments. Further exploration of PBMT in orthodontic practices is warranted to unlock its full therapeutic potential.

Background: Aggrephagy is a lysosome-dependent process that degrades misfolded protein condensates to maintain cancer cell homeostasis. Despite its importance in cellular protein quality control, the role of aggrephagy in glioma remains poorly understood. Objective: To investigate the expression of aggrephagy-related genes (ARGs) in glioma and in different cell types of gliomas and to develop an ARGs-based prognostic signature to predict the prognosis, tumor microenvironment, and immunotherapy response of gliomas. Methods: ARGs were identified by searching the Reactome database. We developed the ARGs-based prognostic signature (ARPS) using data from the Cancer Genome Atlas (TCGA, n = 669) by Lasso-Cox regression. We validated the robustness of the signature in clinical subgroups and CGGA cohorts (n = 970). Gene set enrichment analysis (GSEA) was used to identify the pathways enriched in ARPS subgroups. The correlations between ARGs and macrophages were also investigated at single cell level. Results: A total of 44 ARGs showed heterogeneous expression among different cell types of gliomas. Five ARGs (HSF1, DYNC1H1, DYNLL2, TUBB6, TUBA1C) were identified to develop ARPS, an independent prognostic factor. GSEA showed gene sets of patients with high-ARPS were mostly enriched in cell cycle, DNA replication, and immune-related pathways. High-ARPS subgroup had higher immune cell infiltration states, particularly macrophages, Treg cells, and neutrophils. APRS had positive association with tumor mutation burden (TMB) and immunotherapy response predictors. At the single cell level, we found ARGs correlated with macrophage development and identified ARGs-mediated macrophage subtypes with distinct communication characteristics with tumor cells. VIM+ macrophages were identified as pro-inflammatory and had higher interactions with malignant cells. Conclusion: We identified a novel signature based on ARGs for predicting glioma prognosis, tumor microenvironment, and immunotherapy response. We highlight the ARGs-mediated macrophages in glioma exhibit classical features.

Objective: To evaluate the effectiveness and safety of modified Xiaoyao powder for postpartum depression (PPD) by conducting a systematic review of randomized controlled trials (RCTs). Methods: The Chinese National Knowledge Infrastructure Databases (CNKI), the Chinese Scientific Journals Database (VIP), Wanfang, Google Scholar, the SinoMed, Embase, Cochrane Library, and PubMed databases were searched from their inception to April 25, 2023. The Cochrane Risk of Bias tool was used to assess the quality of the trials. We applied the risk ratio to present dichotomous data and the mean difference to present continuous data. Data with similar characteristics were pooled for meta-analysis and heterogeneity was assessed using I2. Results: This review included 35 trials involving 2848 participants. The quality of the included studies was low (unclear randomization processes and insufficient reporting of blinding). Abstract Background Immunological nonresponders (INRs) living with HIV are at increased risk of co-infection and multiple tumors, with no effective strategy currently available to restore their T-cell immune response. This study aimed to explore the safety and efficacy of thymosin α1 in reconstituting the immune response in INRs. Methods INRs with CD4 + T cell counts between 100 and 350 cells/μL were enrolled and received two-staged 1.6 mg thymosin α1 subcutaneous injections for 24 weeks (daily in the first 2 weeks and biweekly in the subsequent 22 weeks) while continuing antiretroviral therapy. T cell counts and subsets, the expression of PD-1 and TIM-3 on T cells, and signal joint T cell receptor excision circles (sjTREC) at week 24 were evaluated as endpoints. Results Twenty three INRs were screened for eligibility, and 20 received treatment.
