Precipitation change and dry‒wet trend are the most critical natural factors that determine the social development and civilization process in Xinjiang. The Aridity Index (AI), calculated from 99 homogeneous meteorological stations from 1961 to 2020,was used to analyze the variation of dry‒wet climate change in Xinjiang in the past 60 years . The results show that the annual AI in Xinjiang has shown a significant decreasing trend over the past 60 years; that is, the climate in Xinjiang, especially southern Xinjiang, has become wetter. The interdecadal variations from the 1960s-2010s show that the total station ratio of arid and extremely arid areas in Xinjiang showed a decadal decreasing trend, while the subarid areas and humid and semihumid areas showed increasing trends, especially since the beginning of the 21st century. The interdecadal spatial evolution characteristics show that the dry‒wet climate in Xinjiang reversed in the 1990s. An abrupt change in the annual aridity index occurred in 1986, after which the study region was basically in a continuous wetting process. The first empirical orthogonal function (EOF) decomposition mode is consistent in Xinjiang; that is, the climate in Xinjiang is generally dry or wet, and the intensity of this change varies among different regions. The second mode reflects the opposite spatial distribution characteristics of the dry‒wet climate in southern and northern Xinjiang with the Tianshan Mountains. Dry‒wet climate changes in Xinjiang mainly exhibit 2.5-year and 6-year oscillation periods, between which the 6-year period is more significant.

Dihydro-β-ionone is a characteristic aroma compound of Osmanthus fragrans and is widely applied in the flavorfragrance industry. However, the main focus is on chemical synthesis due to the metabolic pathways of dihydro-β-ionone is still unclear. Here, we explored the one-pot synthesis system for dihydro-β-ionone production using carotenoid cleavage dioxygenase (CCD) and enoate reductase. After screening the CCD enzyme, PhCCD1 from the Petunia hybrid was identified as the suitable enzyme for the first step of dihydro-β-ionone synthesis due to the high enzyme activity for carotenoid. The PhCCD1 was expressed in Escherichia coli and further characterized. The optimal activity of PhCCD1 was observed at pH 6.8 and 45 °C. The enzyme was stable over the pH range of 6.0-8.0 and had good thermal stability below 40 °C. Then, we optimized the coupled reaction conditions for dihydro-β-ionone production by PhCCD1 and enoate reductase AaDBR1 from Artemisia annua. Furthermore, we introduced the NADPH regeneration system with a 1.5-fold enhancement for dihydro-β-ionone production. Collectively, approximately 13.34 mg/L dihydro-β-ionone was obtained by the one-pot biosystem with a corresponding molar conversion of 85.8%. For the first time, we successfully designed and constructed a new synthesis pathway for dihydro-β-ionone production in vitro. The coupled catalysis reported herein illustrates the feasibility of producing dihydro-β-ionone from carotenoids and guides further engineering in the food industry.

Epimedin C, a major flavonoid extracted from Herba Epimedii, is a precursor of minor flavonoid icaritin that is a desired drug candidate with remarkable anti-cancer activities. For enhancing the biotransformation efficiency of icaritin, a novel α- l -rhamnosidase gene was cloned from hyperthermophiles Thermotoga petrophila DSM 13995. TpeRha displayed optimal activity at a pH of 4.5 and a temperature of 90 °C. The Km and Kcat of TpeRha for p-nitrophenyl-α- l -rhamnopyranoside were 2.99 mM and 651.73 s−1, respectively. It displayed broad catalytic ability in cleavage of the outer and inner rhamnopyranosyl moieties on the C-3 carbon of epimedin C. Further, this enzyme was utilized to improve the efficiency of the co-conversion system in transforming epimedin C into icaritin, in combined with a thermostable β-glucosidase Tpebgl1. In addition, a transformation pathway (epimedin C -icariin - icariside I - icaritin) with a high efficiency for icaritin production was screened. After a two-stage transformation under optimized conditions (90 °C, pH 4.5, 80 U/mL of TpeRha and 1.2 U/mL of Tpebgl1), 1 g/L of epimedin C was transformed into 0.4337 g/L of icaritin within 150 min, with a corresponding molar conversion rate of 96.9 %. This is the first report of enzymatic transformation on preparing icaritin from epimedin C by using thermostable glycosidase.

Natural β-ionone, a high-value flavoring agent, has been widely applied in the food, cosmetics, and perfume industry. However, attempts to overproduce β-ionone in microorganisms have been limited by the efficiency of carotenoid cleavage dioxygenases (CCDs), which catalyzes β-carotene in the biosynthesis pathway. In order to obtain CCD genes responsible for the specific cleavage of carotenoids generating β-ionone, a novel carotenoid cleavage dioxygenase 1 from Helianthus annuus was cloned and overexpressed in Escherichia coli BL21(DE3). The recombinant CCD was able to cleave a variety of carotenoids at the 9, 10 (9', 10') sites to produce C13 products in vitro , including β-ionone, pseudoionone, 3-hydroxy-4-oxo-β-ionone, 3-hydroxy-β-ionone, and 3-hydroxy-α-ionone, which vary depending on the carotenoid substrates. In comparison with lycopene and zeaxanthin, HaCCD1 also showed the high specificity for β-carotene to cleave the 9, 10 (9', 10') double bond to produce β-ionone in E.coli accumulating carotenoids. Finally, the expression of HaCCD1 in E.coli was optimized, and biochemical characterizations were further clarified. The optimal activity of HaCCD1 was at pH 8.8 and 50 °C. The V max for β-apo-8'-carotenal was 10.14 U/mg, while the K m was 0.32 mM. Collectively, our study provides a valuable enzyme for the synthesis of natural β-ionone by biotransformation and synthetic biology platform.

A novel β-xylosidase Dt-2286 from Dictyoglomus turgidum was cloned and overexpressed in Escherichia coli BL21 (DE3). Dt-2286 belonging to glycoside hydrolase (GH) family 3 encodes a polypeptide with 762 amino acid residues with a molecular weight of 85.1 kDa. By optimization of the growth and induction conditions, the activity of β-xylosidase reached 273 U/mL, which is the highest yield reported to date from E. coli in a shake-flask. The optimal activities of the purified Dt-2286 were found at pH 5.0 and 98 °C. It also shows excellent thermostable/haloduric/organic solvent-tolerance. Dt-2286 was revealed to be a multifunctional enzyme with β-xylosidase, α-arabinofuranoside, α-arabinopyranoside and β-glucosidase activities, and Kcat/Km was 5245.316 mM−1 s−1, 2077.353 mM−1 s−1, 1626.454 mM−1 s−1, and 470.432 mM−1 s−1 respectively. Dt-2286 showed significant synergistic effects on the degradation of xylans, releasing more reduced sugars (up to 15.08 fold) by simultaneous addition with endoxylanase. Moreover, this enzyme has good activity in the hydrolysis of epimedium B, demonstrating its versatility in practical applications.

A global self-labeled distribution analysis (GSLDA) for hyperspectral image (HSI) band selection is proposed in this paper, which focuses on an unsupervised method to ascertain the band discrimination. In order to generate the band labels for further analysis, the concept of the local minimum spanning forest (LMSF) is introduced into the construction of the global self-labeled band partitions based on graph theory. Meanwhile, the novel scoring strategy of triple-density indexes is applied to analyze the labeled-band distribution for determining the selected band subset with clear discrimination. The feasibility of the proposed method is evaluated on real hyperspectral data and the experiment results show a competitive good performance, which demonstrates that the selected bands hold apparent global discrimination and robust noise immunity.

With the aim of finding an extracellular biocatalyst that can efficiently remove the C-7 xylose group from 10-deacetyl-7-xylosltaxol, a Dictyoglomus turgidum β-xylosidase was cloned and expressed in Escherichia coli BL21 (DE3). The molecular mass of purified Dt-Xyl3 was approximately 84 kDa. The recombinant Dt-Xyl3 was most active at pH 5.0 and 75 °C, retaining 88% activity at 65 °C for 1 h, and displaying excellent stability over pH 4.0–7.5 for 24 h. In terms of kinetic parameters, the Km and Vmax values for pNPX were 0.8316 mM and 5.0178 μmol/mL·min, respectively. Moreover, Dt-Xyl3 was activated by Mn2+ and Ba2+ and inhibited by Cu2+, Ni+ and Al3+. In particular, it displayed high tolerance to salts with 60.8% activity in 20% (w/v) NaCl. Ethanol and methanol at 5–15% showed little effect on the enzymatic activity. Dt-Xyl3 demonstrated multifunctional activities followed by pNPX, pNPAraf and pNPG and had a high selectivity for cleaving the outer xylose moieties of 10-deacetyl-7-xylosltaxol with Kcat/Km 110.87 s−1/mM, which produced 10-deacetyl-taxol to semi-synthesize paclitaxel. Under the optimized conditions (60 °C, pH 4.5, enzyme dosage of 0.5 U/mL), 1 g of 10-deacetyl-7-xylosltaxol was transformed to its corresponding aglycone 10-deacetyl-taxol within 30 min, with a molar conversion of 98%. This is the first report that Dictyoglomus turgidum can produce extracellular GH3 β-xylosidase with highly specific activity for 10-deacetyl-7-xylosltaxol biotransformation, thus leading to the application of β-xylosidase Dt-Xyl3 as a biocatalyst in biopharmaceutics.

Conventional antibody-based targeted cancer therapy is one of the most promising avenues of successful cancer treatment, with the potential to reduce toxic side effects to healthy cells surrounding tumor cells. However, the full potential of antibodies is severely limited due to their large size, low stability, slow clearance, and high immunogenicity. Alternatively, recently discovered nanobodies, which are the smallest naturally occurring antigen-binding format, have shown great potential for addressing these limitations. Bioconjugation of nanobodies to functional groups such as toxins, enzymes, radionucleotides, and fluorophores can improve the efficacy and potency of nanobodies, enhance their in vivo pharmacokinetics, and expand the range of potential applications. Herein, we review the superior characteristics of nanobodies in comparison to conventional antibodies and provide insight into recent developments in nanobody conjugates for targeted cancer therapy and imaging.

The ideal therapy would target cancer cells while sparing normal tissue. However, in most conventional chemotherapies normal cells are damaged together with cancer cells resulting in the unfortunate side effects. The principle underlying enzyme/prodrug therapy is that a prodrug-activating enzyme is delivered or expressed in tumor tissue following which a non-toxic prodrug is administered systemically. Non-invasive imaging modalities can fill an important niche in guiding prodrug administration when the enzyme concentration is detected to be high in the tumor tissue but low in the normal tissue. Therefore, high therapeutic efficacy with minimized toxic effect can be anticipated. This review introduces the latest developments of molecular imaging in enzyme/prodrug cancer therapies. We focus on the application of imaging modalities including magnetic resonance imaging, position emission tomography and optical imaging in monitoring the enzyme delivery/expression, guiding the prodrug administration and evaluating the real-time therapeutic response in vivo.
