Abstract Many plants associate with endophytic microbes that improve root phosphorus (P) uptake. Understanding the interactions between roots and endophytes can enable efforts to improve P utilization. Here, we characterize the interactions between lateral roots of endophytes in a core collection of 50 rapeseed (Brassica napus L.) genotypes with differing sensitivities to low P conditions. With the correlation analysis result between bacterial abundance and plant physiological indices of rapeseeds, and inoculation experiments on plates and soil, we identify one Flavobacterium strain (C2) that significantly alleviates the P deficiency phenotype of rapeseeds. The underlying mechanisms are explored by performing the weighted gene coexpression network analysis (WGCNA), and conducting genome-wide association studies (GWAS) using Flavobacterium abundance as a quantitative trait. Under P-limited conditions, C2 regulates fatty acid and lipid metabolic pathways. For example, C2 improves metabolism of linoleic acid, which mediates root suberin biosynthesis, and enhances P uptake efficiency. In addition, C2 suppresses root jasmonic acid biosynthesis, which depends on α-linolenic acid metabolism, improving C2 colonization and activating P uptake. This study demonstrates that adjusting the endophyte composition can modulate P uptake in B. napus plants, providing a basis for developing agricultural microbial agents.

Long non-coding RNA (lncRNA) H19 is an extensively studied lncRNA that is related to numerous pathological changes. Our previous findings have documented that serum lncRNA H19 levels are decreased in patients with chronic kidney disorder and lncRNA H19 reduction is closely correlated with renal tubulointerstitial fibrosis, an essential step in developing end-stage kidney disease. Nonetheless, the precise function and mechanism of lncRNA H19 in renal tubulointerstitial fibrosis are not fully comprehended. The present work utilized a mouse model of unilateral ureteral obstruction (UUO) and transforming growth factor-β1 (TGF-β1)-stimulated HK-2 cells to investigate the possible role and mechanism of lncRNA H19 in renal tubulointerstitial fibrosis were investigated. Levels of lncRNA H19 decreased in kidneys of mice with UUO and HK-2 cells stimulated with TGF-β1. Up-regulation of lncRNA H19 in mouse kidneys remarkably relieved kidney injury, fibrosis and inflammation triggered by UUO. Moreover, the increase of lncRNA H19 in HK-2 cells reduced epithelial-to-mesenchymal transition (EMT) induced by TGF-β1. Notably, up-regulation of lncRNA H19 reduced lipid accumulation and triacylglycerol content in kidneys of mice with UUO and TGF-β1-stimulated HK-2 cells, accompanied by the up-regulation of long-chain acyl-CoA synthetase 1 (ACSL1). lncRNA H19 was identified as a sponge of microRNA-130a-3p, through which lncRNA H19 modulates the expression of ACSL1. The overexpression of microRNA-130a-3p reversed the lncRNA H19-induced increases in the expression of ACSL1. The suppressive effects of lncRNA H19 overexpression on the EMT, inflammation and lipid accumulation in HK-2 cells were diminished by ACSL1 silencing or microRNA-130a-3p overexpression. Overall, the findings showed that lncRNA H19 ameliorated renal tubulointerstitial fibrosis by reducing lipid deposition via modulation of the microRNA-130a-3p/ACSL1 axis.

Abstract Osteoblasts and osteoclasts play an important role in maintaining the structural integrity of bone tissue, in which osteoclasts degrade bone structure and osteoblasts restore bone tissue. The imbalance of osteoblast and osteoclast function can lead to many bone-related diseases, such as osteoporosis and inflammatory osteolysis. The drug that can both promote bone formation and inhibit bone loss will be able to treat those diseases. In this study, it was found that LMK-235, an selective HDAC4/5 inhibitor, inhibited the differentiation and maturation of osteoclasts by regulating NF-κB and p-Smad2/3 signaling pathways via inhibition of HDAC4. At the same time, we found that LMK-235 promoted osteoblast mineralization by upregulating Runx2 expression via inhibition of HDAC4. In vivo, LMK-235 was able to alleviate lipopolysaccharide (LPS)-induced calvarial osteolysis and promote the repair of bone defects. Taken together, LMK-235 suppresses osteoclast differentiation and promotes osteoblast formation by inhibiting HDAC4. This may provide a valuable treatment for bone diseases caused by abnormal osteoclast bone resorption and osteoblast bone regeneration.

Abstract The estuarine plastisphere, a novel ecological habitat in the Anthropocene, has garnered global concerns. Recent geochemical evidence has pointed out its potential role in influencing nitrogen biogeochemistry. However, the biogeochemical significance of the plastisphere and its mechanisms regulating nitrogen cycling remain elusive. Using 15N- and 13C-labelling coupled with metagenomics and metatranscriptomics, here we unveil that the plastisphere likely acts as an underappreciated nitrifying niche in estuarine ecosystems, exhibiting a 0.9 ~ 12-fold higher activity of bacteria-mediated nitrification compared to surrounding seawater and other biofilms (stone, wood and glass biofilms). The shift of active nitrifiers from O2-sensitive nitrifiers in the seawater to nitrifiers with versatile metabolisms in the plastisphere, combined with the potential interspecific cooperation of nitrifying substrate exchange observed among the plastisphere nitrifiers, collectively results in the unique nitrifying niche. Our findings highlight the plastisphere as an emerging nitrifying niche in estuarine environment, and deepen the mechanistic understanding of its contribution to marine biogeochemistry.

IntroductionPhosphorus recovery from waste streams is a global concern due to open nutrient cycles. However, the reliability and efficiency of recycled P fertilizers are often low. Biostimulants (BS), as a potential enhancer of P availability in soil, could help to overcome current barriers using recycled P fertilizers. For this, a deeper understanding of the influence of BSs on soil P turnover and the interaction of BSs with plants is needed. MethodsWe conducted an incubation and a pot trial with maize in which we testednon-microbial (humic acids and plant extracts) and microbial BSs (microbial consortia) in combination with two recycled fertilizers for their impact on soil P turnover, plant available P, and plant growth.Results and discussionBSs could not stimulate P turnover processes (phosphatase activity, microbial biomass P) and had a minor impact on calcium acetate-lactate extractable P (CAL-P) in the incubation trial. Even though stimulation of microbial P turnover by the microbial consortium and humic acids in combination with the sewage sludge ash could be identified in the plant trial with maize, this was not reflected in the plant performance and soil P turnover processes. Concerning the recycled P fertilizers, the CAL-P content in soil was not a reliable predictor of plant performance with both products resulting in competitive plant growth and P uptake. While this study questions the reliability of BSs, it also highlights the necessity toimprove our understanding and distinguish the mechanisms of P mobilization in soil and the stimulation of plant P acquisition to optimize future usage.

Vegetables are important economic crops globally, and their production has approximately doubled over the past 20 years. Globally, vegetables account for 13% of the harvested area but consume 25% of the fertilizer, leading to serious environmental impacts. However, the quantitative evaluation of vegetable production systems in subtropical plateau lake basins and the establishment of optimal management practices to further reduce environmental risks are still lacking. Using the life cycle assessment method, this study quantified the global warming, eutrophication, acidification, and energy depletion potential of vegetable production in a subtropical plateau lake basin in China based on data from 183 farmer surveys. Our results indicated that vegetable production in the study area, the Erhai Lake Basin, was high but came at a high environmental cost, mainly due to low fertilizer efficiency and high nutrient loss. Root vegetables have relatively high environmental costs due to the significant environmental impacts of fertilizer production, transportation, and application. A comprehensive analysis showed that the vegetable production in this region exhibited low economic and net ecosystem economic benefits, with ranges of 7.88–8.91 × 103 and 7.35–8.69 × 103 $ ha−1, respectively. Scenario analysis showed that adopting strategies that comprehensively consider soil, crop, and nutrient conditions for vegetable production can reduce environmental costs (with reductions in global warming potential (GWP), eutrophication potential (EP), acidification potential (AP), and energy depletion potential (EDP) by 10.6–28.2%, 65.1–73.5%, 64.5–71.9%, 47.8–70.4%, respectively) compared with the current practices of farmers. This study highlighted the importance of optimizing nutrient management in vegetable production based on farmers’ practices, which can achieve more yield with less environmental impacts and thereby avoid the “trade-off” effect between productivity and environmental sustainability.

Despite the essential role of nitrogen fertilizers in achieving high crop yields, current application practices often exhibit low efficiency. Optimizing nitrogen (N) fertilization in agriculture is, therefore, critical for enhancing crop productivity while ensuring sustainable food production. This study investigates the effects of nitrification inhibitors (Nis) such as Dimethyl Pyrazole Phosphate (DMPP) and Dimethyl Pyrazole Fulvic Acid (DMPFA), plant growth-promoting bacteria inoculation, and phosphorus (P) application on the soil-plant-microbe system in maize. DMPFA is an organic nitrification inhibitor that combines DMP and fulvic acid for the benefits of both compounds as a chelator. A comprehensive rhizobox experiment was conducted, employing varying levels of P, inoculant types, and Nis, to analyze the influence of these factors on various soil properties, maize fitness, and phenotypic traits, including root architecture and exudate profile. Additionally, the experiment examined the effects of treatments on the bacterial and fungal communities within the rhizosphere and maize roots. Our results showed that the use of Nis improved plant nutrition and biomass. For example, the use of DMPFA as a nitrification inhibitor significantly improved phosphorus use efficiency by up to 29%, increased P content to 37%, and raised P concentration in the shoot by 26%, compared to traditional ammonium treatments. The microbial communities inhabiting maize rhizosphere and roots were also highly influenced by the different treatments. Among them, the N treatment was the major driver in shaping bacterial and fungal communities in both plant compartments. Notably, Nis reduced significantly the abundance of bacterial groups involved in the nitrification process. Moreover, we observed that each experimental treatment employed in this investigation could select, promote, or reduce specific groups of beneficial or detrimental soil microorganisms. Overall, our results highlight the intricate interplay between soil amendments, microbial communities, and plant nutrient dynamics, suggesting that Nis, particularly DMPFA, could be pivotal in bolstering agricultural sustainability by optimizing nutrient utilization.

Cover crops are one of the climate-smart agricultural practices used to increase soil organic carbon (SOC) sequestration. However, the SOC sequestration potential and underlying mechanisms under different cover crops, especially in orchard agroecosystems, have not been fully elucidated. Here, we investigated three orchards in China using SOC fractionation methods, high-throughput sequencing, and biomarker analysis. Our objectives were to determine the effect of cover crops on the physical fractions and chemical compositions of SOC, as well as on microbial properties, and to clarify why legume and non-legume cover crops sequester SOC differently. The results showed different increases in SOC between legume and non-legume cover crops (+38% vs. +16%) compared with those in the control plots without cover crops. Legume cover crops increased mineral-associated and particulate organic carbon, whereas non-legume cover crops increased mineral-associated organic carbon only. These differences were attributed to their distinct effects on microbial SOC transformation pathways. Legume cover crops positively impacted microbial pathways by increasing the availability of soil substrates and nitrogen, such as dissolved organic carbon (+84%), O-alkyl carbon (+18%), and ammonium nitrogen (+42%). These results were supported by the increases in carbon and nitrogen enzyme activities, microbial community diversity indices, the abundance of dominant fungal taxa (Sordariomycetes), microbial biomass carbon (+105%), and microbial necromass carbon (+47%). Non-legume cover crops might have induced microbial nitrogen starvation, decreasing the efficiency of microbial pathways, as evidenced by the low β-glucosidase to β-N-acetylglucosaminidase ratios (−7%) and the lack of significant changes in the bacterial Shannon index or microbial necromass carbon. In addition, redundancy analysis revealed that enzyme activity, the microbial community, and microbial necromass carbon collectively dominated the changes in the SOC physical fraction. Site-specific soil properties such as soil texture and nitrogen availability were important factors influencing SOC sequestration under cover crops. Our study provides essential insights for optimizing cover crop management to increase SOC sequestration in orchard agroecosystems.

Abstract Delayed repair of fractures seriously impacts patients’ health and significantly increases financial burdens. Consequently, there is a growing clinical demand for effective fracture treatment. While current materials used for fracture repair have partially addressed bone integrity issues, they still possess limitations. These challenges include issues associated with autologous material donor sites, intricate preparation procedures for artificial biomaterials, suboptimal biocompatibility, and extended degradation cycles, all of which are detrimental to bone regeneration. Hence, there is an urgent need to design a novel material with a straightforward preparation method that can substantially enhance bone regeneration. In this context, we developed a novel nanoparticle, mPPTMP195, to enhance the bioavailability of TMP195 for fracture treatment. Our results demonstrate that mPPTMP195 effectively promotes the differentiation of bone marrow mesenchymal stem cells into osteoblasts while inhibiting the differentiation of bone marrow mononuclear macrophages into osteoclasts. Moreover, in a mouse femur fracture model, mPPTMP195 nanoparticles exhibited superior therapeutic effects compared to free TMP195. Ultimately, our study highlights that mPPTMP195 accelerates fracture repair by preventing HDAC4 translocation from the cytoplasm to the nucleus, thereby activating the NRF2/HO-1 signaling pathway. In conclusion, our study not only proposes a new strategy for fracture treatment but also provides an efficient nano-delivery system for the widespread application of TMP195 in various other diseases.

Abstract Accurate prediction of remaining useful life (RUL) for aircraft engines is essential for proactive maintenance and safety assurance. However, existing methods such as physics-based models, classical recurrent neural networks, and convolutional neural networks face limitations in capturing long-term dependencies and modeling complex degradation patterns. In this study, we propose a novel deep-learning model based on the Transformer architecture to address these limitations. Specifically, to address the issue of insensitivity to local context in the attention mechanism employed by the Transformer encoder, we introduce a position-sensitive self-attention (PSA) unit to enhance the model's ability to incorporate local context by attending to the positional relationships of the input data at each time step. Additionally, a gated hierarchical long short-term memory network (GHLSTM) is designed to perform regression prediction at different time scales on the latent features, thereby improving the accuracy of RUL estimation for mechanical equipment. Experiments on the C-MAPSS dataset demonstrate that the proposed model outperforms existing methods in RUL prediction, showcasing its effectiveness in modeling complex degradation patterns and long-term dependencies.

C2H2 zinc finger protein is widely involved in the occurrence and development of cancer. However, the function and mechanism of most C2H2 zinc finger proteins in breast caner (BC) remains unclear. Here, we reported the expression prognosis of C2H2 type zinc finger protein ZNF775 in BC patients and its possible biological mechanism. First, multiple public databases showed that ZNF775 was significantly overexpressed in BC tissues. Interestingly, high expression of ZNF775 was significantly associated with a better prognosis. Immunohistochemistry were used for verification, and the expression of ZNF775 was consistent with the databases. Considering the large heterogeneity of different breast cancer cells, we temporarily selected MCF-7 cell line for verification. In vitro overexpression experiments showed that overexpression of ZNF775 significantly inhibited the proliferation and migration of MCF-7 BC cell. We further combined RNA-sequencing (RNA-seq) and CUT & Tag, and found that overexpression of ZNF775 can down-regulate the expression of most genes in the Wnt signaling pathway. The cBioportal database showed that ZNF775 was negatively correlated with the expression of Wnt5a, suggesting that its downstream target was likely Wnt5a. Finally, we discovered that Wnt5a could partially reverse the inhibitory effect of ZNF775 on MCF-7 BC cell migration through transwell migration experiments. In conclusion, our findings will provide new ideas for the diagnosis, treatment and prognosis assessment of BC in the future.

Abstract Pathogen genetic diversity varies in response to environmental changes. However, it remains unclear whether plant barriers to invasion could be considered a genetic bottleneck for phytopathogen populations. Here, we implement a barcoding approach to generate a pool of 90 isogenic and individually barcoded Ralstonia solanacearum strains. We used 90 of these strains to inoculate tomato plants with different degrees of physical permeability to invasion (intact roots, wounded roots and xylem inoculation) and quantify the phytopathogen population dynamics during invasion. Our results reveal that the permeability of plant roots impacts the degree of population bottleneck, genetic diversity, and composition of Ralstonia populations. We also find that selection is the main driver structuring pathogen populations when barriers to infection are less permeable, i.e., intact roots, the removal of root physical and immune barriers results in the predominance of stochasticity in population assembly. Taken together, our study suggests that plant root permeability constitutes a bottleneck for phytopathogen invasion and genetic diversity.

Abstract Gears, as indispensable components of machinery, demand accurate prediction of their Remaining Useful Life (RUL). To enhance the utilization of ordered information within time series data and elevate RUL prediction precision, this study introduces the attention-guided multi-hierarchy LSTM (AGMLSTM). This innovative approach leverages attention mechanisms to capture the intricate interplay between high and low hierarchical features of the input data, marking the first application of such a technique in gear RUL prediction. Additionally, a refined health indicator (HI) is introduced, constructed through a diffusion model, to precisely reflect the gears' health condition. The proposed RUL prediction method unfolds as follows: firstly, HIs are computed from gear vibration data. Subsequently, leveraging the known HIs, AGMLSTM predicts future HIs, and the RUL of the gear is determined upon surpassing the failure threshold. Quantitative analysis of experimental results conclusively demonstrates the superiority of the proposed RUL prediction method over existing approaches for gear RUL estimation.

IntroductionThe micronutrient deficiency of iron and boron is a common issue affecting the growth of rapeseed (Brassica napus). In this study, a non-destructive diagnosis method for iron and boron deficiency in Brassica napus (genotype: Zhongshuang 11) using hyperspectral imaging technology was established.MethodsThe recognition accuracy was compared using the Fisher Linear Discriminant Analysis (LDA) and Support Vector Machine (SVM) recognition models. Recognition results showed that Multiple Scattering Correction (MSC) could be applied for the full band hyperspectral data processing, while the LDA models presented better performance on establishing the leaf iron and boron deficiency symptom recognition than the SVM models.ResultsThe recognition accuracy of the training set reached 96.67%, and the recognition rate of the prediction set could be 91.67%. To improve the model accuracy, the Competitive Adaptive Reweighted Sampling algorithm (CARS) was added to construct the MSC-CARS-LDA model. 33 featured wavelengths were selected via CARS. The recognition accuracy of the MSC-CARS-LDA training set was 100%, while the recognition accuracy of the MSC-CARS-LDA prediction set was 95.00%.DiscussionThis study indicates that, it is capable to identify the iron and boron deficiency in rapeseed using hyperspectral imaging technology.

Abstract Due to anthropogenic activities, heavy metal (HM) pollution in soils has increased, resulting in severe ecological problems and posing a constant threat to human health. Among various remediation methods, bacterial remediation is a relatively clean, efficient, and minimally negative approach. However, bacterial agents face multiple environmental stresses, making them challenging to achieve long-lasting and stable restoration effects. To address this issue, supportive organic substances such as biochar can be added to the soil with bacteria. According to bibliometric studies, integrating biochar and bacteria is extensively researched and widely used for HM-contaminated soil remediation. By integrating biochar and bacteria, heavy metals in the soil can be remediated, and soil conditions can be improved over time. Bacteria can also better promote plant growth or contribute effectively to phytoremediation processes when assisted by biochar. However, the remediation agents integrating biochar and bacteria are still some distance away from large-scale use because of their high cost and possible environmental problems. Therefore, further discussion on the interaction between biochar and bacteria and the integration approach, along with their remediation efficiency and environmental friendliness, is needed to achieve sustainable remediation of HM-contaminated soils by integrating biochar and bacteria. This paper discusses the potential mechanisms of biochar-bacteria-metal interactions, current advancements in biochar-bacteria combinations for HM-contaminated soil treatment, and their application in sustainable remediation, analyzes the interaction between biochar and bacteria and compares the remediation effect of different ways and feedstocks to integrate biochar and bacteria. Finally, future directions of biochar-bacteria combinations are presented, along with evidence and strategies for improving their commercialization and implementation. Graphical Abstract

The combination of nanoporous materials with organic working fluids holds the promise of further enhancing the performance of refrigerants based thermodynamics cycles. In this study, the adsorption and desorption properties of several organic refrigerants, e.g., R1234yf, R134a, R32, and their mixtures in metal-organic framework materials MOF-5 and Co-MOF-74 are investigated via molecular dynamics methods. The results indicate that the adsorption capacity is inversely proportional to the temperature during the adsorption process, and the adsorption capacity of the R1234yf/R32 mixture (molar ratio 2:1) is higher than that of the corresponding pure working fluids. The desorption amount, desorption regeneration rate, and desorption heat are directly proportional to the temperature. The interactions between different molecular atoms in the mixed working fluids promote the desorption process.

Pigments derived from red pepper fruits are widely used in food and cosmetics as natural colorants. Nitrogen (N) is a key nutrient affecting plant growth and metabolism; however, its regulation of color-related metabolites in pepper fruit has not been fully elucidated. This study analyzed the effects of N supply (0, 250, and 400 kg N ha-1) on the growth, fruit skin color, and targeted and non-target secondary metabolites of field-grown pepper fruits at the mature red stage. Overall, 16 carotenoids were detected, of which capsanthin, zeaxanthin, and capsorubin were the dominant ones. N application at 250 kg ha-1 dramatically increased contents of red pigment capsanthin, yellow-orange zeaxanthin and β-carotene, with optimum fruit yield. A total of 290 secondary metabolites were detected and identified. The relative content of most flavonoids and phenolic acids was decreased with increasing N supply. Correlation analysis showed that color parameters were highly correlated with N application rates, carotenoids, flavonoids, phenolic acids, lignans, and coumarins. Collectively, N promoted carotenoid biosynthesis but downregulated phenylpropanoid and flavonoid biosynthesis, which together determined the spectrum of red color expression in pepper fruit. Our results provide a better understanding of the impact of N nutrition on pepper fruit color formation and related physiology, and identification of target metabolites for enhancement of nutritional quality and consumer appeal.

Abstract Using agricultural wastes as an alternative phosphorus (P) source has great prospects to improve soil P status. A 70-day incubation experiment was carried out to investigate the effects of superphosphate (SSP), poultry manure (PM), cattle manure (CM), maize straw (MS), and cattle bone meal (CB) with the same total P input on soil P availability and fractions in typical acidic (red soil) and alkaline (fluvo-aquic soil) soils. The results showed that in both fluvo-aquic and red soils, CM out-performed other P sources in improving soil P availability. Changes in soil Olsen-P (ΔOlsen-P) were greater in fluvo-aquic soils with SSP, PM and CM additions than in red soils. Among the different P sources used, only CM has increased the labile soil P fractions to levels similar to that with SSP. Compared with SSP, more monoester P and inositol hexakisphosphate were detected in soils amended with PM and CM. A structural equation model (SEM) analysis suggested that soil pH had a direct positive effect on the labile P fractions in the acidic red soil amended with different P sources. In summary, CM is a superior P source for increasing plant available soil P, with considerable practical implications for P recycling.

The quality of sintered ore, which serves as the primary raw material for blast furnace ironmaking, is directly influenced by the moisture in the sintering mixture. In order to improve the precision of water addition in the sintering process, this paper proposes an intelligent model for predicting water-filling volume based on Temporal Fusion Transformer (TFT), whose symmetry enables it to effectively capture long-term dependencies in time series data. Utilizing historical sintering data to develop a prediction model for the amount of mixing and water addition, the results indicate that the TFT model can achieve the R squared of 0.9881, and the root mean square error (RMSE) of 3.5951. When compared to the transformer, long short-term memory (LSTM), and particle swarm optimization–long short-term memory (PSO-LSTM), it is evident that the TFT model outperforms the other models, improving the RMSE by 8.5403, 6.9852, and 0.453, respectively. As an application, the TFT model provides an effective interval reference for moisture control in normal sintering processes, which ensures that the error is within 1 t.

Our study aims to investigate the water quality and distribution patterns of phytoplankton communities in the Jinjiang River Basin in Quanzhou, as well as their relationship with environmental factors. We integrated data from the national water quality databases of the two main tributaries of the West and East Jinjiang Rivers between 2020 and 2023, supplemented by field surveys. Redundancy analysis was used to explore the effect of environmental factors on phytoplankton communities. Our findings revealed that the West Jinjiang River experienced a significant influence from excessive fertilizer use in tea cultivation, leading to an increase in TN concentrations compared to the East Jinjiang River. The abundance of phytoplankton in the Jinjiang River Basin was 105 cells·L−1, with phytoplankton being dominated by Chlorophyta, Cyanphyta, and diatoms, accounting for an average of 50%, 20%, and 19% of the total phytoplankton abundance, respectively. Redundancy analysis indicated that temperature, pH, and nutrient concentrations were important factors influencing the phytoplankton communities. With increasing temperature and nutrients concentrations, the abundance of Chlorophyta and Dinophyta significantly increased. This study provides a solid foundation for the regular “health diagnosis” of crucial rivers and lakes in Quanzhou and supports the establishment of a health guarantee system for rivers and lakes.

Excessive fertilizer input, low nutrient use efficiency, soil quality, and environmental degradation hinder greenhouse vegetable production. Integrated agronomic strategies of soil, crop, and nutrient management are needed to sharply improve the vegetable yield and simultaneously maintain sustainable production. A three-season field experiment was conducted from 2015 to 2018, aiming to evaluate the effect of integrated soil–crop system management (ISSM) on the agronomy, environment, and economy of greenhouse vegetable systems in the Yangtze River Basin, China. Three treatments were included in the experiment: (1) farmers’ current practice (FP), based on a local farmers’ survey; (2) soil remediation treatment (SR), the application of soil conditioner and compost fertilizer instead of chicken manure; (3) ISSM, a combination of soil conditioner, reducing plant density, and using formula fertilizer as well as increasing the fertilization times. The results indicated that ISSM (47.7 Mg ha−1) improved the pepper yield by 17% relative to farmers’ current practice (FP, 40.7 Mg ha−1). Soil remediation (SR), as a single approach, mainly made a contribution to improving the yield (by 6.9%) and nutrient use efficiency while reducing apparent nitrogen (N) losses. Higher yields were mainly attributed to increasing the fruit number per plant. On average, apparent N losses were reduced by 245 kg N ha−1 per season for ISSM compared to FP. In addition, higher net profits were obtained under SR and ISSM relative to FP. Overall, both SR and ISSM have advantages for the agronomy, environment, and economy in greenhouse vegetable production, but ISSM would be the optimal choice to achieve higher yields with lower environmental impacts.

Breast cancer has become the number one cancer in the world, and intestinal flora may be closely linked to it. Geographic location also has an important impact on human intestinal flora. We conducted the first study on the intestinal flora of breast cancer patients and non-breast cancer patients in a tropical region - Hainan Province in China. At the same time, Pacbio platform based on third-generation sequencing was used for the first time to conduct 16S full-length sequencing of fecal microorganism DNA. We completed the species diversity analysis and differential species analysis of the intestinal flora between the two groups, inferred their functional genetic composition and performed functional difference analysis. There were statistically significant differences in alpha diversity between the two groups in Hainan Province. By species composition difference analysis, at the phylum level, Bacteroidales (P = 0.006) and Firmicutes (P = 0.002) was differed between the two groups, and at the genus level, 17 breast cancer-related differential species such as Bacteroides were screened. The occurrence of antibiotics in agricultural soils has raised concerns due to their potential risks to ecosystems and human health. However, a comprehensive understanding of antibiotic accumulation, distribution, and potential risks to terrestrial ecosystems on a global scale is still limited. Therefore, in this study, we evaluated the accumulation of antibiotics and their potential risks to soil microorganisms and plants, and highlighted the driving factors of antibiotic accumulation in agricultural soils based on 134 peer-reviewed studies (between 2000 and 2022). The results indicated that 56 types of antibiotics were detected at least once in agricultural soils with concentrations ranging from undetectable to over 7000 µg/kg. Doxycycline, tylosin, sulfamethoxazole, and enrofloxacin, belonging to the tetracyclines, macrolides, sulfonamides, and fluoroquinolones, respectively, were the most accumulated antibiotics in agricultural soil. The accumulation of TCs, SAs, and FQs was found to pose greater risks to soil microorganisms (average at 29.3%, 15.4%, and 21.8%) and plants (42.4%, 26.0%, and 38.7%) than other antibiotics. East China was identified as a hot spot for antibiotic contamination due to high levels of antibiotic concentration and ecological risk to soil microorganisms and plants. Antibiotic accumulation was found to be higher in vegetable fields (245.5 µg/kg) and orchards (212.4 µg/kg) compared to croplands (137.2 µg/kg). Furthermore, direct land application of manure resulted in a greater accumulation of TCs, SAs, and FQs accumulation in soils than compost fertilization. The level of antibiotics decreased with increasing soil pH and organic matter content, attributed to decreasing adsorption and enhancing degradation of antibiotics. In conclusion, this study highlights the need for further research on the impacts of antibiotics on soil ecological function in agricultural fields and their interaction mechanisms. Additionally, a whole-chain approach, consisting of antibiotic consumption reduction, manure management strategies, and remediation technology for soil contaminated with antibiotics, is needed to eliminate the potential environmental risks of antibiotics for sustainable and green agriculture.

The high loss of foliar-applied fertilizers in the wet season and other extreme weather conditions reduces fertilizer use efficiency remarkably low. Emerging nano-iron materials, owing to their highly desirable properties such as exceptionally small size, particle surface and interface attributes, strong adhesion and high iron ratio, could overcome the limitations of traditional fertilizers. However, their anti-leaching property, leaching resistance mechanism, iron nutrition enrichment capacity and cost effectiveness remain unclear. In this study, four nano-iron materials (Fe nanoparticles (Fe-NPs), Fe3O4-NPs, α-Fe2O3-NPs and γ-Fe2O3-NPs) were compared with traditional iron fertilizers (FeSO4 and Fe-EDTA). The results showed that the efficiency of traditional iron fertilizers decreased greatly under simulated rainfall conditions due to their poor leaching resistance. In contrast, nano-iron fertilizers performed exceptionally well under simulated rainfall conditions with significant increase in leaf iron concentration, photosynthesis and growth of peanut. This was mainly attributed to their decreased contact angle, increased wetting capacity and increased adhesion on plant surface, which collectively decreased fertilizer leaching remarkably. The adhesion capacity of Fe3O4-NPs (53.5 mN/m), was nearly three times that of traditional iron fertilizer (18.3 mN/m). Therefore, only a small amount of Fe3O4-NPs (4 mg/plant) was needed relative to traditional iron fertilizers (FeSO4, 59 mg/plant) to achieve comparable high leaf Fe concentration (1076.96 μg/g) under rainfall conditions. Cost evaluation suggests that the cost-competitiveness of Fe3O4-NPs will become comparable to that of FeSO4 when its price comes down from the current cost of $139/kg to $36/kg, the cost of FeSO4, which is likely considering the pace of nano-material technology development. Together, these results demonstrate the remarkable potential of nano-iron fertilizers in mitigating the escalating fertilizer cost and improving the environmental outcomes of peanut, as well as potentially other crops grown in wet season.

Article highlights A meta-analysis of biochar preparation and application methods for disease severity and plant growth is needed. Soil application of biochar from straw at 350–600 °C and at 3–5% rate was effective in reducing disease severity. Biochar application has better suppression effect on cash crop diseases caused by airborne pathogens. The results of the meta-analysis are particularly useful for maximizing the effect of biochar on plant disease control.

This paper investigates the impact of increasing maximum wind speed of tropical cyclones on the return periods of water levels in the sea area of the Yangtze River Delta. To conduct this study, a series of numerical experiments are performed using historical tropical cyclones that impacted the Yangtze River Delta from 1949 to 2019. The aim is to analyze the effects of global climate change on extreme water levels and the corresponding return periods. To obtain the historical water levels in the sea areas of the Yangtze River Delta, a storm surge model is driven by the selected tropical cyclones. The simulated astronomical tidal levels during the same period are also used. The extreme water levels of different return periods are then calculated. The maximum wind speeds of the selected tropical cyclones are increased by 11% according to the expected amount of increase under global climate change. The extreme water levels of different return periods under this scenario are calculated with the same procedure. The results of the study show that the impact of increasing maximum wind speed of tropical cyclones on the increases of extreme water levels and the decrease of return periods is more significant in the inner area of the estuaries than in the outer areas. Moreover, the responses of the extreme water levels and the corresponding return periods in the Yangtze River Estuary and the Hangzhou Bay show different characteristics. The results of this study provide significant reference value for the management of future coastal disaster prevention and mitigation in the Yangtze River Delta. Furthermore, the methodology used in this study can be applied in other estuaries to investigate the potential impacts of changes in climate and hydrology factors on extreme water levels and the corresponding return periods.

Nasopharyngeal carcinoma (NPC) is one of the most predominant cancers occurring in China with high morbidity. Lately, large quantities of long non-coding RNAs (lncRNAs) have been highlighted to regulate the biological activities in multiple tumors, including NPC. Our study centered on whether TTN-AS1 was involved in NPC and how it modulated the progression of NPC. Here, qRT-PCR data uncovered that TTN-AS1 expression was conspicuously high in NPC cells. Based on the results of functional assays, TTN-AS1 silence hampered the proliferative, migratory, and invasive abilities but stimulated the apoptotic capability of NPC cells. After a series of mechanism assays, TTN-AS1 was found to competitively bind with miR-876-5p and recruit UPF1 to enhance NETO2 expression. In addition, TTN-AS1 could be transcriptionally activated by YY1 in NPC cells. It was also found that miR-876-5p overexpression or NETO2 downregulation had inhibitory effects on cell proliferation, migration, and invasion in NPC. Moreover, NETO2 upregulation could restore the suppressive impacts of TTN-AS1 depletion on NPC cell and tumor growth. In conclusion, YY1-activated TTN-AS1 interacted with both miR-876-5p and UPF1 to upregulate NETO2, thus strengthening NPC cell malignant behaviors, which might provide more useful information for people to develop effective NPC treatments.

Researchers have previously described the response of crop productivity and greenhouse gas (GHG) emissions to fertilizer nitrogen (N) additions, but they have not determined how to maximize yields while minimizing GHG emissions. We conducted an experiment at 2293 sites with four N levels to simulate both grain yield and yield-scaled GHG emissions in response to the N addition. The yield-scaled GHG emissions decreased by 16% as the N rate increased from treatments without the N addition to the minimum yield-scaled GHG emissions, which was comparable to the values associated with the maximum grain yields. The sites with both high soil productivity and high crop productivity had the highest yield and lowest yield-scaled GHG emissions, with 43% higher yield and 38% lower yield-scaled GHG emissions than sites with low soil and low crop productivity. These findings are expected to enhance evaluations of wheat production and GHG emissions in China, and thereby contribute to addressing disparities in the global food and GHG budget.

Pepper is one of the most vitamin C enriched vegetables worldwide. Although applying nitrogen (N) fertilizer is an important practice for high fruit yield in pepper production, it is still unclear how N application regulates pepper fruit vitamin C anabolism at different maturity stage. To further the understanding, we combined physiological and metabolomic analysis to investigate the fruit vitamin C content (including ascorbic acid (AsA) and dehydroascorbic acid (DHA)), related enzyme activity and non-targeted metabolites of field-grown chili pepper produced under different N levels at mature green and red stages. The results showed that increasing N application reduced AsA content in pepper fruit at both maturity stages, but highly elevated DHA content only at mature green stage. Regardless of N application level, AsA content displayed an increasing trend while DHA content was reduced as pepper fruit maturity advanced, resulting in a higher content of total vitamin C at the mature green stage. The L-galactose pathway, D-galacturonate pathway, and myo-inositol pathway were identified for AsA biosynthesis. The involved precursor metabolites were mainly negatively regulated by increasing N application, and their accumulation increased when pepper fruit developed from green to red stage. Meanwhile, the activities of key enzymes and metabolites in relation to degradation and recycling processes of AsA and DHA were increased or did not change with increasing N application, and they were differently influenced as fruit maturing. As a result, the recommended N application level (250 kg N ha-1) could maintain relatively high total vitamin C content in pepper fruits without yield loss at both maturity stages. These findings highlight the importance of optimizing N application level to maximize vitamin C content in pepper fruits, and provide a better understanding of the maturity stage-dependent N regulation on vitamin C anabolism.

Osteoclast (OC) abnormalities lead to many osteolytic diseases, such as osteoporosis, inflammatory bone erosion, and tumor-induced osteolysis. Exploring effective strategies to remediate OCs dysregulation is essential. FTY720, also known as fingolimod, has been approved for the treatment of multiple sclerosis and has anti-inflammatory and immunosuppressive effects. Here, we found that FTY720 inhibited osteoclastogenesis and OC function by inhibiting nuclear factor kappa-B (NF-κB) signaling. Interestingly, we also found that FTY720 inhibited osteoclastogenesis by upregulating histone deacetylase 4 (HDAC4) expression levels and downregulating activating transcription factor 4 (ATF4) expression levels. In vivo, FTY720 treatment prevented lipopolysaccharide- (LPS-) induced calvarial osteolysis and significantly reduced the number of tartrate-resistant acid phosphatase- (TRAP-) positive OCs. Taken together, these results demonstrate that FTY720 can inhibit osteoclastogenesis and ameliorate inflammation-induced bone loss. Which may provide evidence of a new therapeutic target for skeletal diseases caused by OC abnormalities.

Abstract Background A variety of hematopoietic abnormalities are commonly seen in human immunodeficiency virus-1 (HIV-1) infected individuals despite antiviral therapy, but the underlying mechanism remains elusive. Nef plays an important role in HIV-1 induced T cell loss and disease progression, but it is not known whether Nef participates in other hematopoietic abnormalities associated with infection. Results In the current study we investigated the influence of HIV-1LAI Nef (LAI Nef) on the development of hematopoietic stem/progenitor cells (HSPCs) into myeloid-erythroid lineage cells, and found that nef expression in HSPCs blocked their differentiation both in vitro and in humanized mice reconstituted with nef-expressing HSPCs. Conclusions Our novel findings demonstrate LAI Nef compromised the development of myeloid-erythroid lineage cells, and therapeutics targeting Nef would be promising in correcting HIV-1 associated hematopoietic abnormalities.

Abstract Background Interferon regulatory factor 2 (IRF2) is an important transcription factor, which can regulate the IFN response and plays a role in antiviral innate immunity in teleost. Results In the present study, the full-length cDNA sequence of IRF2 (CcIRF2) was characterized in common carp (Cyprinus carpio L.), which encoded a protein containing a conserved DNA-binding domain (DBD) and an IRF-associated domain (IAD). Phylogenetic analysis showed that CcIRF2 was most closely related with IRF2 of Ctenopharyngodon idella. CcIRF2 transcripts were detectable in all examined tissues, with higher expression in the gills, spleen and brain. CcIRF2 expression was upregulated in immune-related tissues of common carp upon polyinosinic:polycytidylic acid (poly (I:C)) and Aeromonas hydrophila stimulation and induced by poly (I:C), lipopolysaccharide (LPS), peptidoglycan (PGN) and flagellin in the peripheral blood leucocytes (PBLs) and head kidney leukocytes (HKLs). In addition, overexpression of CcIRF2 decreased the expression of IFN and IFN-stimulated genes (ISGs), and a dual-luciferase reporter assay revealed that CcIRF2 could increase the activation of NF-κB. Conclusions These results indicate that CcIRF2 participates in antiviral and antibacterial immune response and negatively regulates the IFN response, which provide a new insight into the regulation of IFN system in common carp, and are helpful for the prevention and control of infectious diseases in carp farming.

Plant roots have important functions, such as acquiring nutrients and water from the surrounding soil and transporting them upwards to the shoots. Simultaneously, they must be able to exclude potentially harmful substances and prevent the entry of pathogens into the roots. The endodermis surrounds the vascular tissues and forms hydrophobic diffusion barriers including Casparian strips and suberin lamella. Suberin in cell walls can be induced by a range of environmental factors and contribute to against biotic and abiotic threats. Tremendous progress has been made in biosynthesis of suberin and its function, little is known about the effect of its plasticity and distribution on stress tolerance. In field conditions, biotic and abiotic stress can exist at the same time, and little is known about the change of suberization under that condition. This paper update the progress of research related to suberin biosynthesis and its function, and also discuss the change of suberization in plant roots and its role on biotic and abiotic stresses tolerance.

IntroductionA better understanding of the regulatory role of microorganisms on soil phosphorous (P) mobilization is critical for developing sustainable fertilization practices and reducing P resource scarcity. The phoD genes regulate soil organic P (Po) mobilization.MethodsBased on the long-term P application experiments in acid purple soil of maize system in Southwest China (started in 2010), the experiment included five P levels: 0, 16, 33, 49, and 65.5 kg P hm–2 (P0, P16, P33, P49, and P65.5, respectively). The molecular speciation of organic P in soil was determined by 31P-nuclear magnetic resonance (NMR), high-throughput sequencing technology, and real-time qPCR were used to analyze the bacterial community and abundance of phoD-harboring bacterial genes, exploring the bacterial community and abundance characteristics of phoD gene and its relationship with the forms of Po and alkaline phosphatase (ALP) activity in the soil.ResultsThe results showed that the orthophosphate monoesters (OM) were the main Po speciation and varied by P fertilization in acid purple soil. ALP activity decreased as P fertilization increased. Co-occurrence network analysis identified the overall network under five P fertilizations. The keystone taxon base on the network showed that Collimonas, Roseateles, Mesorhizobium, and Cellulomonas positively correlated with both OM and Po. The random forest showed that Cellulomonas, Roseateles, and Rhodoplanes were the key predictors for ALP activity. The keystone taxon was a more important predictor than the dominant taxon for ALP, OM, and Po. The structural equation model (SEM) showed that soil organic matter (SOM), available P (AP), and OM were the main factors influencing the ALP by reshaping phoD-harboring bacteria alpha diversity, community composition, and phoD abundance.DiscussionThe phoD-harboring bacterial community composition especially the keystone taxon rather than alpha diversity and abundance dominated the ALP activity, which could promote P utilization over an intensive agroecosystem. These findings improve the understanding of how long-term gradient fertilization influences the community composition and function of P-solubilizing microorganisms in acid purple soil.

Intensive vegetable production has been characterized by high nitrogen (N) fertilizer input in southwest China. Optimizing the N fertilizer rate is the basis for the optimal management of regional N fertilizer. A two-year field experiment with five N fertilizer rates was conducted during 2019–2021 in southwest China, and the aim of this study was to identify the effects of different N application rates on yield, dry matter biomass (DMB), N uptake, N use efficiency (NUE) and soil mineral N (Nmin) residues for Chinese cabbage (Brassica chinensis L.) and further determine the critical plant N concentration and root-zone soil Nmin residues required to reach the maximum DMB of Chinese cabbage. Five N treatments were established: control without N input (CK); optimal N fertilizer rate decreased by 30% (70% OPT, 175 kg N ha−1), optimized N fertilizer rate (OPT, 250 kg N ha−1), optimal N fertilizer rate increased by 30% (130% OPT, 325 kg N ha−1) and farmers’ N fertilizer practice (FP, 450 kg N ha−1). The N source in all treatments was conventional urea (N ≥ 46.2%). The results showed that the total yield of Chinese cabbage followed a “linear-plateau” trend with an increasing N fertilizer rate. There was no significant difference in yield between the OPT, 130% OPT and FP treatments. The aboveground plant DMB and N uptake showed a ‘slow-fast-slow’ pattern with the growth period. There was no significant difference in aboveground plant DMB and N uptake between the OPT, 130% OPT and FP treatments. Moreover, the OPT treatment significantly increased the aboveground plant DMB and N accumulation by 29.6% and 40.5%, respectively, compared with the 70% OPT treatment. The OPT treatment significantly increased the NUE by 23.8%, 31.2% and 43.1% compared with that in the 70% OPT, 130% OPT and FP treatments, respectively. The linear-plateau model provided the best fit for the relationship among aboveground DMB of Chinese cabbage, plant N concentration and root-zone soil Nmin content. The critical root-zone soil Nmin and plant N concentrations were 94.1, 63.4 and 68.3 kg ha−1 and 34.4, 33.5 and 32.9 g kg−1 during the rosette, heading and harvest periods, respectively. In summary, compared to the FP treatment, the optimized N fertilizer rate (250 kg N ha−1) could significantly reduce the N application rate, maintain yield, increase aboveground plant DMB and N uptake, and improve NUE. Moreover, the study has great significance for guiding the green utilization of vegetable N fertilizer in southwest China.

Heavy metal contamination in soils endangers humans and the biosphere by reducing agricultural yield and negatively impacting ecosystem health. In recent decades, this issue has been addressed and partially remedied through the use of “green technology,” which employs metal-tolerant plants to clean up polluted soils. Furthermore, the global climate change enhances the negative effects of climatic stressors (particularly drought, salinity, and extreme temperatures), thus reducing the growth and metal accumulation capacity of remediating plants. Plant growth-promoting bacteria (PGPB) have been widely introduced into plants to improve agricultural productivity or the efficiency of phytoremediation of metal-contaminated soils via various mechanisms, including nitrogen fixation, phosphate solubilization, phytohormone production, and biological control. The use of metal-tolerant plants, as well as PGPB inoculants, should hasten the process of moving this technology from the laboratory to the field. Hence, it is critical to understand how PGPB ameliorate environmental stress and metal toxicity while also inducing plant tolerance, as well as the mechanisms involved in such actions. This review attempts to compile the scientific evidence on this topic, with a special emphasis on the mechanism of PGPB involved in the metal bioremediation process [plant growth promotion and metal detoxification/(im)mobilization/bioaccumulation/transformation/translocation] and deciphering combined stress (metal and climatic stresses) tolerance.

Abstract Rapeseed (Brassica napus L.) production experienced a considerable increase during the past decades, and there is growing concern about how to realize the dual objectives of meeting the increasing demand for rapeseed and reducing the associated greenhouse gas (GHG) emissions. Most reported studies on crop‐sourced GHG emissions to date were aimed at optimizing crop management to reduce emission, but often without a detailed spatio‐temporal assessment of GHG emissions for layout management practices. Herein, we used a newly developed modelling approach that integrates the NUtrient Flows in food chains, Environment, and Resource use (NUFER) model with a dynamic reactive nitrogen (Nr) loss module for rapeseed and Life Cycle Assessment (LCA) approach to assess the GHG emissions from rapeseed production in China, where produces 18% of the world's rapeseed. Results showed that the GHG emissions from rapeseed production in China increased by 1.90% yearly from 2004 to 2018, largely due to increased inputs of agricultural materials, especially diesel. Further, carbon footprints of rapeseed production were found vary greatly between production regions, due to diverse climatic conditions, field management practices and the level of mechanization. Scenario analysis showed that adoption of both optimized nutrient and layout management practice is required to mitigate the current resource and environmental burdens, and to achieve edible oil security. Thus, our study provides new insights for policymakers on the formulation of management practices to reduce crop‐sourced GHG emissions, going along with an increase of food demand in the future.

Ralstonia solanacearum RSc2741 has been predicted as a gamma-glutamyl phosphate reductase ProA catalyzing the second reaction of proline formation from glutamate. Here, we experimentally demonstrated that proA mutants were proline auxotrophs that failed to grow in a minimal medium, and supplementary proline, but not glutamate, fully restored the diminished growth, confirming that ProA is responsible for the biosynthesis of proline from glutamate in R. solanacearum. ProA was previously identified as one of the candidates regulating the expression of genes for type three secretion system (T3SS), one of the essential pathogenicity determinants of R. solanacearum. Supplementary proline significantly enhanced the T3SS expression both in vitro and in planta, indicating that proline is a novel inducer of the T3SS expression. Deletion of proA substantially impaired the T3SS expression both in vitro and in planta even under proline-supplemented conditions, indicating that ProA plays additional roles apart from proline biosynthesis in promoting the expression of the T3SS genes. It was further revealed that the involvement of ProA in the T3SS expression was mediated through the pathway of PrhG-HrpB. Both the proA mutants and the wild-type strain grew in the intercellular spaces of tobacco leaves, while their ability to invade and colonize tobacco xylem vessels was substantially impaired, which was about a 1-day delay for proA mutants to successfully invade xylem vessels and was about one order of magnitude less than the wild-type strain to proliferate to the maximum densities in xylem vessels. It thus resulted in substantially impaired virulence of proA mutants toward host tobacco plants. The impaired abilities of proA mutants to invade and colonize xylem vessels were not due to possible proline insufficiency in the rhizosphere soil or inside the plants. All taken together, these results extend novel insights into the understanding of the biological function of ProA and sophisticated regulation of the T3SS and pathogenicity in R. solanacearum.

Global warming has driven the expansion of cultivated land to high-altitude areas. Intensive vegetable production, which is generally considered to be a high economic value and high environmental risk system, has expanded greatly in high-altitude mountainous areas of China. However, the environmental cost of vegetable production in these areas is poorly understood. In this study, pepper production at low (traditional pepper production area) and high (newly expanded area) altitudes were investigated in Shizhu, a typical pepper crop area. The output and environmental cost at the two altitudes were identified. the influence of resource inputs, climate, and soil properties on pepper production was evaluated. There were obvious differences in output and environmental cost between the two altitudes. High-altitude pepper production achieved a 16.2% lower yield, and had a higher fertilizer input, resulting in a 22.3% lower net ecosystem economic benefit (NEEB), 23.0% higher nitrogen (N) footprint and 24.0% higher carbon (C) footprint compared to low-altitude farming. There is potential for environmental mitigation with both high- and low-altitude pepper production; Compared to average farmers, high-yield farmers groups reduced their N and C footprints by 16.9–24.8% and 18.3–25.2%, respectively, with 30.6–34.1% higher yield. A large increase in yield could also be achieved by increasing the top-dress fertilizer rate and decreasing the plant density. Importantly, high-altitude pepper production was achieved despite less advanced technology and inferior conditions (e.g., a poor road system and uneven fields). It provides a reference for the study of the environmental cost of other high-altitude regions or other crop systems at high-altitude areas.

Background: The goal of this work was to compare the serum vitamin D level in women with uterine leiomyoma to that of controls, and to examine its association with the etiopathogenesis of this disease. Methods: This case-control study included 110 premenopausal women referred to a university hospital from January 2020 to December 2020. The study group consisted of 55 cases who underwent surgery for uterine fibroids, while the control group was comprised of 55 subjects without uterine fibroids. The number and volume of existing fibroids in each subject was assessed by transvaginal ultrasonography. Serum vitamin D status was determined by radioimmunoassay for 25-hydroxyvitamin D. Results: Average serum 25-hydroxyvitamin D levels in patients with uterine fibroids (20.52 ± 6.17 ng/mL) were significantly lower than in controls (24.18 ± 6.88 ng/mL; p = 0.004). The Yangtze River Basin (YRB) has been recognized as one of the key strategic development regions in China. Agriculture p52 roduction systems in the YRB have contributed considerably to China’s goal of food security. Realizing Agriculture Green Development (AGD) means agriculture production systems with high productivity, high resource use efficiency and low environmental costs. However, challenges and barriers still exist for realizing AGD in the YRB. Here, we summarize four main challenges for AGD in the YRB, and identify two approaches (top-down and bottom-up) including main strategies needed to achieve AGD. The four challenges include, but are not limited to, (1) low agricultural productivity and nutrient use efficiencies, (2) an uneven agricultural production structure, (3) rapid urbanization, and (4) uncoordinated targets for environmental protection and food production. We conclude that both top-down and bottom-up approaches are needed to deliver AGD in the YRB. Top-down approaches are mainly operated by government and underpinned by research, which uses spatial planning to promote the balance between agricultural production and the ecological environment, and to optimize the proportions of cereal and cash crop production with monogastric and ruminant animal production. The bottom-up approach needs strategies to close the yield gap of various cropping and livestock systems, improve resource use efficiencies to control environmental impacts. Furthermore, training and education are needed to increase awarenessand improve skills for farmers and advisers. Our review can serve as example for other global regions that are in transition from unsustainable agriculture production towards sustainable withclean environment and healthy economies.

Bioeffector (BE) application is emerging as a strategy for achieving sustainable agricultural practices worldwide. However, the effect of BE on crop growth and quality is still controversial and there is still no adequate impact assessment that determines factors on the efficiency of BE application. Therefore, we carried out a network metaanalysis on the effect of BEs using 1,791 global observations from 186 studies to summarize influencing factors and the impact of BEs on crop growth, quality, and nutrient contents. The results show that BEs did not only improve plant growth by around 25% and yield by 30%, but also enhanced crop quality, e.g., protein (55% increase) and soluble solids content (75% increase) as well as aboveground nitrogen (N) and phosphate (P) content by 28 and 40%, respectively. The comparisons among BE types demonstrated that especially non-microbial products, such as extracts and humic/amino acids, have the potential to increase biomass growth by 40–60% and aboveground P content by 54–110%. The soil pH strongly influenced the efficiency of the applied BE with the highest effects in acidic soils. Our results showed that BEs are most suitable for promoting the quality of legumes and increasing the yield of fruits, herbs, and legumes. We illustrate that it is crucial to optimize the application of BEs with respect to the right application time and technique (e.g., placement, foliar). Our results provide an important basis for future research on the mechanisms underlying crop improvement by the application of BEs and on the development of new BE products.

Abstract Background Interferon (IFN) regulatory factors (IRFs), as transcriptional regulatory factors, play important roles in regulating the expression of type I IFN and IFN- stimulated genes (ISGs) in innate immune responses. In addition, they participate in cell growth and development and regulate oncogenesis. Results In the present study, the cDNA sequence of IRF10 in common carp (Cyprinus carpio L.) was characterized (abbreviation, CcIRF10). The predicted protein sequence of CcIRF10 shared 52.7–89.2% identity with other teleost IRF10s and contained a DNA-binding domain (DBD), a nuclear localization signal (NLS) and an IRF-associated domain (IAD). Phylogenetic analysis showed that CcIRF10 had the closest relationship with IRF10 of Ctenopharyngodon idella. CcIRF10 transcripts were detectable in all examined tissues, with the highest expression in the gonad and the lowest expression in the head kidney. CcIRF10 expression was upregulated in the spleen, head kidney, foregut and hindgut upon polyinosinic:polycytidylic acid (poly I:C) and Aeromonas hydrophila stimulation and induced by poly I:C, lipopolysaccharide (LPS) and peptidoglycan (PGN) in peripheral blood leucocytes (PBLs) and head kidney leukocytes (HKLs) of C. carpio. In addition, overexpression of CcIRF10 was able to decrease the expression of the IFN and IFN-stimulated genes PKR and ISG15. Conclusions These results indicate that CcIRF10 participates in antiviral and antibacterial immunity and negatively regulates the IFN response, which provides new insights into the IFN system of C. carpio.

Our previous study manifested that lanthanum chloride (LaCl3) can enhance the anticancer ability of cisplatin (DDP) in ovarian cancer cells. Here, ovarian cancer cells SKOV3 and SKOV3/DDP were subjected to DDP and LaCl3. Cell viability, apoptosis, DNA repair, and PI3K/Akt pathway were detected. LaCl3 induced more cell death and apoptosis caused by DDP in two cell lines, accompanied by upregulation of Bax and Cleaved caspase 3 proteins, and downregulation of Bcl-2 protein. LaCl3 also could decrease RAD51 protein by inactivation of the PI3K/Akt pathway. These data indicated that LaCl3 could be a potential drug to modulate DDP resistance by inactivating of PI3K/Akt pathway and attenuating DNA repair in ovarian cancer.

Abstract Small farms are the mainstay of maize production in China. Its productivity is relatively low despite large farm inputs and the associated environmental footprints. Here, we studied public–private partnership (PPP) model for sustainable intensification of maize production to achieve co‐benefits of food security and environmental sustainability. The PPP model enabled the development of an effective partnership by bringing complementary skills, knowledge, proprietary products and technologies, and resources of public research community and private enterprises to create a new, operational maize farming system in China. We conducted on‐farm research with farmer participation in four major maize‐growing regions spanning temperate to sub‐tropical zones in China for 2 years. The PPP model achieved 78.7% of maize yield potential compared with 61.8% realized in smallholder farm (SHF) (11.0 Mg ha−1 vs. 8.6 Mg ha−1). Overall, environmental externalities of PPP were up to 32.7% lower than that of SHF, depending on the region studied. PPP significantly reduced reactive nitrogen losses by 31.3%–35.5% compared with SHF in both years. There was no significant difference between PPP and SHF for greenhouse gas emission in 2018, but it was significantly lower in PPP (19%) compared to SHF in 2019. Similarly, PPP significantly reduced soil acidification potential (by 10.1%–42.2%) and eutrophication of waterbodies (21.5%) in comparison to SHF. Overall, the net ecosystem economic budget increased 277 USD ha−1 with PPP. The PPP model provides new insights into improving food security and ecosystem and economic budget. As a logical progression to our research, future work should focus on (a) the reasons for the persistence of inter‐regional yield gap in PPP model and (b) to gain a better understanding of socioeconomic drivers critical for successful PPP in different maize‐growing regions.

Background: Increasing circular RNAs (circRNAs) are reported to participate in cancer progression. Nonetheless, the role of circRNAs in nasopharyngeal carcinoma (NPC) has not been fully clarified. This work is aimed to probe the role of circ_0000215 in NPC.Methods: Circ_0000215 expression in NPC tissues and cell lines was examined by quantitative real-time polymerase chain reaction (qRT-PCR). Cell counting kit-8 (CCK-8) assay, 5-bromo-2′-deoxyuridine (BrdU) assay, scratch healing assay and Transwell experiment were executed to investigate the regulatory function of circ_0000215 on the proliferation, migration and invasion of NPC cells. RNA immunoprecipitation (RIP), pull-down and dual-luciferase reporter experiments were utilized to determine the binding relationship between circ_0000215 and miR-512-5p, and between miR-512-5p and phosphoinositide-3-kinase regulatory subunit 1 (PIK3R1) 3′UTR. The effects of circ_0000215 on NPC growth and metastasis in vivo were examined with nude mice model. Western blot was applied to detect the regulatory effects of circ_0000215 and miR-512-5p on PIK3R1 expression.Results: Circ_0000215 was overexpressed in NPC tissues and cell lines. The functional experiments confirmed that knockdown of circ_0000215 impeded the growth and metastasis of NPC cells in vitro and in vivo. Additionally, circ_0000215 could also work as a molecular sponge to repress miR-512-5p expression. PIK3R1 was validated as a target gene of miR-512-5p, and circ_0000215 could increase the expression level of PIK3R1 in NPC cells via suppressing miR-512-5p.Conclusion: Circ_0000215 is overexpressed in NPC and exerts oncogenic effects in NPC through regulating miR-512-5p/PIK3R1 axis.

Mangroves provide many ecosystem services, including coastal protection against storm surges and waves. As an adaptive method for coastal defense, mangroves were widely restored and planted in tropical and subtropical regions, such as the coastal regions in Southeast Asia. Field surveys were conducted to quantify the nature-based coastal protection provided by a planted mangrove forest along the coasts of Shanwei, Guangdong Province, China, under typhoon influence. The resilience of mangrove trees was assessed under the impact of Typhoon Mangkhut (2018), which induced a maximum storm surge of 1.74 m with a maximum wave height of 1.16 m in the study area. The pre- and post-typhoon surveys and hydrodynamic measurements were conducted at a mudflat with planted mangroves. The wave height reduction reached 77% over 100 m wide mangrove forest. Our results suggest that a six-year-old planted mangrove forest with a ~100 m width might withstand a super typhoon impact and provide substantial protection for the fish ponds and embankments behind the mangrove forest. No uproots or deadly breakage of stems were observed in the mangrove forest, while severe defoliation was spotted for a small portion of trees in the study area, mainly along the wind path, the windward edge of the forest, and among the taller plants. Obvious sedimentation in the mangrove front and the tidal flat was observed during the typhoon Mangkhut (2018) and the entire typhoon season of 2018.

Symptoms of coronavirus disease 2019 (COVID-19) range from asymptomatic to severe pneumonia and death. A deep understanding of the variation of biological characteristics in severe COVID-19 patients is crucial for the detection of individuals at high risk of critical condition for the clinical management of the disease. Herein, by profiling the gene expression spectrum deduced from DNA coverage in regions surrounding transcriptional start site in plasma cell-free DNA (cfDNA) of COVID-19 patients, we deciphered the altered biological processes in the severe cases and demonstrated the feasibility of cfDNA in measuring the COVID-19 progression. The up- and downregulated genes in the plasma of severe patient were found to be closely related to the biological processes and functions affected by COVID-19 progression. More importantly, with the analysis of transcriptome data of blood cells and lung cells from control group and cases with severe acute respiratory syndrome-coronavirus 2 (SARS-CoV-2) infection, we revealed that the upregulated genes were predominantly involved in the viral and antiviral activity in blood cells, reflecting the intense viral replication and the active reaction of immune system in the severe patients. Pathway analysis of downregulated genes in plasma DNA and lung cells also demonstrated the diminished adenosine triphosphate synthesis function in lung cells, which was evidenced to correlate with the severe COVID-19 symptoms, such as a cytokine storm and acute respiratory distress. Overall, this study revealed tissue involvement, provided insights into the mechanism of COVID-19 progression, and highlighted the utility of cfDNA as a noninvasive biomarker for disease severity inspections.

Compression sutures are primarily used to treat atonic postpartum hemorrhage. We herein describe three cases of selective arterial ligation combined with B-Lynch or modified B-Lynch suture for the treatment of intractable postpartum hemorrhage unresponsive to available conservative interventions. Three pregnant women underwent a cesarean section for a macrosomic fetus, fetal distress, and oligohydramnios, respectively. All three women developed intractable postpartum hemorrhage due to uterine atony with no chance of embolization therapy. B-Lynch or modified B-Lynch suture and additional selective arterial ligation were performed using braided absorbable suture. The first woman developed postoperative hematometra and infection without response to drainage and antibiotic therapy. Although laparoscopic exploration was performed to loosen the suture line and drain the hematometra and pyometra, the necrosis and infection could not be controlled. Subtotal hysterectomy was therefore conducted, and the necrotic uterine adnexa was removed. The other two women developed subinvolution of the uterus resulting in prolonged menstruation and amenorrhea, although the uterus was preserved and the bleeding was controlled. Modified B-Lynch suture combined with vascular ligation is an invaluable technique for women with severe intractable postpartum hemorrhage. However, it can lead to serious complications such as uterine necrosis, infection, and subinvolution.
