In modern football matches, athlete trajectory analysis and real-time feedback play a crucial role in tactical planning and game management. Traditional trajectory prediction methods struggle to provide accurate real-time feedback due to an insufficient capture of dynamic changes. This study constructs a model for football trajectory prediction and real-time feedback based on temporal convolutional networks (TCN) and spatial channel attention. This model can effectively capture the complex motion trajectories of players and balls in time and space representation, and generate real-time feedback information to assist coach decision-making through comparison with historical data. In addition, by introducing joint trajectory prediction, we aim to address the issue of poor handling of long-term dependencies in traditional methods, and improve the accuracy and real-time performance of trajectory prediction. In the experiment, we validated the method using actual game data, and our method successfully predicted the trajectory of football in actual games. The experimental results showed that the method has high accuracy in predicting the trajectory of players and balls, with PPA and TPA reaching 65.85% and 87.12% improved by 1.73% and 1.6%, respectively, which is better than existing baseline models and can provide accurate tactical feedback.

ABSTRACT Severe acute respiratory syndrome coronavirus 2 (SARS-CoV-2) targets epithelial cells in the respiratory tract, triggering an acute proinflammatory response and chronic lung inflammation. Probiotic supplementation has shown promise in reducing the nasopharyngeal SARS-CoV-2 viral load, diminishing symptom frequency and duration, and mitigating inflammation; thus, it is a potential strategy for treating coronavirus disease 2019 (COVID-19), which is caused by SARS-CoV-2 infection. In this study, we evaluated the effects of the oral administration of the Lactobacillus plantarum GUANKE strain, a gram-positive bacterium originally isolated from a healthy individual, on SARS-CoV-2 infection in a human ACE2 transgenic mouse model. We found that GUANKE significantly reduced inflammatory cell infiltration and pulmonary interstitial exudation in mice. The transcription of CCL2, TNFA, IL1B, IL6, and IL17C in the lungs was reduced. The protein levels of TNF-α, IL-1β, IL-6, and IL-17 in the lungs were significantly lower in GUANKE-treated mice than in control mice. The viral load in GUANKE-treated mice was lower than that in saline-treated mice, although this difference did not reach statistical significance. L. plantarum GUANKE can decrease SARS-CoV-2-induced lung inflammation in mice, suggesting its potential for use as an agent for treating SARS-CoV-2 infection.IMPORTANCEMost otherwise healthy individuals develop only mild or moderate symptoms of coronavirus disease 2019 (COVID-19) caused by current strains of severe acute respiratory syndrome coronavirus 2 (SARS-CoV-2), and virus replication is mainly confined to the upper respiratory tract; however, the virus can infect the lower respiratory tract and promote inflammation. Probiotic supplementation has been shown to reduce nasopharyngeal SARS-CoV-2 viral load, reduce the overall number and duration of symptoms, and attenuate inflammation in clinical trials. We showed that a novel L. plantarum GUANKE strain alleviated SARS-CoV-2-induced pneumonia in mice. The transcription and production of inflammatory cytokines were suppressed, and GUANKE moderately reduced the viral load. L. plantarum GUANKE has the potential to become a candidate drug for the treatment of COVID-19 or other viral respiratory infections.

Terrestrial vegetation emits substantial amounts of highly reactive isoprene, significantly impacting atmospheric chemistry and climate change. Both atmospheric carbon dioxide (CO2) concentration and temperature can influence plant isoprene emissions; however, whether these factors have a synergistic effect remains unclear, particularly for tropical/subtropical plants. In this study, we conducted in-situ controlled experiments on Eucalyptus urophylla, a representative tropical/subtropical species, to investigate the seasonal variation in the response of isoprene emissions to CO2 concentrations (ISOP-CO2 response) and to identify potential controlling factors. The results showed that high CO2 exerts a nearly linear inhibitory effect on isoprene emissions, as indicated by the slope of the ISOP-CO2 response curve. This inhibitory effect exhibited evident seasonal changes, with stronger suppression during cooler seasons and weaker suppression during warmer seasons. This finding contrasts with the default ISOP-CO2 response in the MEGAN model, which ignored seasonal variation. Further analysis showed a significant correlation between the slope of the ISOP-CO2 response curve and growth temperature from the past 10 days, indicating that these metrics are effective indicators for predicting seasonal changes. Our findings reveal a synergistic mechanism between temperature and CO2 concentration effects on isoprene emissions. By coupling the effects of growth temperature with the ISOP-CO2 response, this mechanism can be integrated into models to provide more accurate predictions of future isoprene emissions, reducing prediction biases, especially during cooler seasons.

Abstract Background Community-acquired pneumonia (CAP) is a common and serious condition that can be caused by a variety of pathogens. However, much remains unknown about how these pathogens interact with the lower respiratory commensals, and whether any correlation exists between the dysbiosis of the lower respiratory microbiota and disease severity and prognosis. Methods We conducted a retrospective cohort study to investigate the composition and dynamics of sputum microbiota in patients diagnosed with CAP. In total, 917 sputum specimens were collected consecutively from 350 CAP inpatients enrolled in six hospitals following admission. The V3-V4 region of the 16 S rRNA gene was then sequenced. Results The sputum microbiota in 71% of the samples were predominately composed of respiratory commensals. Conversely, 15% of the samples demonstrated dominance by five opportunistic pathogens. Additionally, 5% of the samples exhibited sterility, resembling the composition of negative controls. Compared to non-severe CAP patients, severe cases exhibited a more disrupted sputum microbiota, characterized by the highly dominant presence of potential pathogens, greater deviation from a healthy state, more significant alterations during hospitalization, and sparser bacterial interactions. The sputum microbiota on admission demonstrated a moderate prediction of disease severity (AUC = 0.74). Furthermore, different pathogenic infections were associated with specific microbiota alterations. Acinetobacter and Pseudomonas were more abundant in influenza A infections, with Acinetobacter was also enriched in Klebsiella pneumoniae infections. Conclusion Collectively, our study demonstrated that pneumonia may not consistently correlate with severe dysbiosis of the respiratory microbiota. Instead, the degree of microbiota dysbiosis was correlated with disease severity in CAP patients.

The environmental stability of infectious viruses in the laboratory setting is crucial to the transmission potential of human respiratory viruses. Different experimental techniques or conditions used in studies over the past decades have led to diverse understandings and predictions for the stability of viral infectivity in the atmospheric environment. In this paper, we review the current knowledge on the effect of simulated atmospheric conditions on the infectivity of respiratory viruses, mainly focusing on influenza viruses and coronaviruses, including severe acute respiratory syndrome coronavirus 2 and Middle East respiratory syndrome coronavirus. First, we summarize the impact of the experimental conditions on viral stability; these involve the methods of viral aerosol generation, storage during aging and collection, the virus types and strains, the suspension matrixes, the initial inoculum volumes and concentrations, and the drying process. Second, we summarize and discuss the detection methods of viral infectivity and their disadvantages. Finally, we integrate the results from the reviewed studies to obtain an overall understanding of the effects of atmospheric environmental conditions on the decay of infectious viruses, especially aerosolized viruses. Overall, this review highlights the knowledge gaps in predicting the ability of viruses to maintain infectivity during airborne transmission.

Polyimide (PI) faces new challenges in meeting the requirements of low coefficient of thermal expansion (CTE), low dielectric content/loss, and high thermal stability to be utilized effectively as a substrate for flexible electronic materials. At present research, the CTE of PI films can be reduced to around 5 ppm/K, with Tg > 420 °C. However, it is difficult to balance low CTE and high Tg at the same time, and reducing dielectric performance is even more difficult. This study focused on synthesizing four types of PI films (using BPDA as the dianhydride) by employing biphenyl structure diamines (PDA, BZD, DPT, DMP) with an increasing number of benzene rings. The improvement in CTE, dielectric properties, and thermal stability was observed with an increase in the number of benzene rings and a decrease in the number of imine rings in the PI horizontal structure. The Tetraphenylene PI-DMP exhibited values of ε and tanθ as 3.31 and 4.73 ‰ respectively under 10 GHz. Additionally, the biphenyl PI films demonstrated commendable thermal stability (Tg = 442°C), outstanding mechanical properties (elastic modulus > 10 GPa), and a low CTE (2 ppm/K within the temperature range of 50–300 °C, and close to the 3.6 ppm/K CTE of monocrystalline silicon). The low CTE, high thermal stability, and low dielectric properties that 5G flexible substrate materials need to be qualified are simultaneously reflected in this study. MS theoretical calculations were used to analyze the results. The excellent consistency with experimental results can promote the feasibility of MS theory in PI dielectric properties at high-frequency (10 Ghz). This innovative approach is anticipated to provide foldable chip fields with intrinsic biphenyl PI materials possessing low dielectric, low CTE, and high thermal stability.

BackgroundFerroptosis has been proven to contribute to the progression of myocardial ischemia/reperfusion (I/R) injury and can be inhibited or promoted by ATF3. Short-chain fatty acids (SCFAs) have shown benefits in various cardiovascular diseases with anti-inflammatory and antioxidant effects. However, the impact of SCFAs on ferroptosis in ischemic-stimulated cardiomyocytes remains unknown. This study aimed to investigate the effect of SCFAs on cardiomyocyte ferroptosis, the expression of ATF3, and its potential upstream regulators.Methods and resultsThe expression of ATF3, ferroptosis pathway geneset (FPG), and geneset of potential regulators for ATF3 (GPRA, predicted by the PROMO database) was explored in the public human myocardial infarction single-cell RNA-seq (sma) dataset. Cardiomyocyte data was extracted from the dataset and re-clustered to explore the FPG, ATF3, and GPRA expression patterns in cardiomyocyte subclusters. A dose-dependent toxic experiment was run to detect the suitable dose for SCFA treatment. The erastin-induced ferroptosis model and hypoxia-reoxygenation (H/R) model (10 h of hypoxia followed by 6 h of reoxygenation) were adopted to assess the effect of SCFAs via the CCK8 assay. Gene expression was examined via RT-PCR and western blot. Ferroptosis markers, including lipid peroxides and Fe2+, were detected using the liperfluo and ferroOrange probes, respectively. In the sma dataset, upregulated ferroptosis pathway genes were mainly found in the infarction-stimulated cardiac cells (border zone and fibrotic zone), particularly the cardiomyocytes and adipocytes. The ATF3 and some of its potential transcription factors (VDR, EGR3, PAX5, and SP1) can be regulated by SCFA. SCFA can attenuate erastin-induced lipid peroxidation in cardiomyocytes. SCFA treatment can also reverse erastin-induced Fe2+ increase but may strengthen the Fe2+ in the H/R model. We also precisely defined a ferroptosis subcluster of cardiomyocytes (CM09) that highly expressed FPG, ATF3, and GPRA.ConclusionThe ATF3 and the ferroptosis pathway are elevated in cardiomyocytes of injury-related cardiac regions (border zone, ischemic zone, and fibrotic zone). SCFA can attenuate cardiomyocyte ferroptosis and regulate the expression of ATF3. Our study offers novel insights into the potential targets of SCFAs in the cardiovascular system.

Abstract Nitrogen is a significant element that constitutes ambient organic aerosol. Individual N-containing organic molecules are known to have both natural and anthropogenic sources and implicated in a wide-ranging health and environmental effects. Yet, unlike carbon (C), the total quantity of aerosol organic nitrogen (ON) remains largely unquantified, hindering a quantitative understanding of their major sources and impacts. Here, aerosol ON is quantitatively surveyed in hundreds of aerosol filter samples collected from sites of varying urban influence in China using our recently developed method that permits simple, and yet sensitive, simultaneous detection of inorganic and organic nitrogen. Annual average ON concentration was in the range of 0.4–1.4 μg N m−3, representing 17–31% of aerosol total nitrogen. Monte Carlo simulations constrained by paired ON and OC measurements suggest N-containing organic molecules contributed typically 37–50%, with a 95% confidence interval of [12%, 94%], to ambient organic aerosols. Source apportionment analysis reveals that biomass burning and secondary formation are dominant ON sources, accounting for 21–24% and ~ 30% of ON, respectively. Primary biological aerosol is also a significant source of ON (7–18%), with its contribution more prominent in non-urban atmospheres. The results provide the quantitative data for the extent of presence of organic nitrogenous aerosol and the origin of their major sources. Such data, we anticipate, would bring forth a breakthrough in our ability to describe and model organic aerosols and to assess their environmental impacts, such as atmospheric nitrogen nutrient inputs to ecosystems.

Among persons born in China before 1980 and tested for vaccinia virus Tiantan strain (VVT), 28.7% (137/478) had neutralizing antibodies, 71.4% (25/35) had memory B-cell responses, and 65.7% (23/35) had memory T-cell responses to VVT. Because of cross-immunity between the viruses, these findings can help guide mpox vaccination strategies in China.

Gastrodiae Rhizoma (GE), a popular food in China, is stored and consumed after steaming, which can lead to the degradation of active substances and a decrease in its quality. Therefore, this study explored the potential application of ultra-high-pressure (UHP)-assisted hot air drying in improving the quality of GE. The results indicated that UHP pre-treatment could preserve the original cross-sectional color of GE and increase the degrees of freedom of water in GE samples. Compared with traditional steaming pre-treatment (18 h), UHP pre-treatment at 500 MPa significantly shorted the time (10 h) required for the GE samples to reach drying equilibrium. Meanwhile, the UHP-assisted hot air drying method (60 °C) could reduce the activity of β-D-glucosidase and avoid the degradation of active substances. Finally, UHP pre-treatment improved the neuroprotective activity in vivo. Overall, UHP-assisted hot air drying could improve the quality of GE samples. This study provides a simple method for improving the quality of GE samples and offers a reference for subsequent research on the influence of UHP on GE.

Carbonyl compounds are important oxygenated volatile organic compounds (VOCs) that play significant roles in the formation of ozone (O3) and atmospheric chemistry. This study presents comprehensive field observations of carbonyl compounds during an unusual wintertime ozone pollution event at a suburban site in Guangzhou, South China, from 19 to 28 December 2020. The aim was to investigate the characteristics and sources of carbonyls, as well as their contributions to O3 formation. Formaldehyde, acetone, and acetaldehyde were the most abundant carbonyls detected, with average concentrations of 7.11 ± 1.80, 5.21 ± 1.13, and 3.00 ± 0.94 ppbv, respectively, on pollution days, significantly higher than those of 2.57 ± 1.12, 2.73 ± 0.88, and 1.10 ± 0.48 ppbv, respectively, on nonpollution days. The Frame for 0-D Atmospheric Modeling (F0AM) box model simulations revealed that local production accounted for 62–88% of observed O3 concentrations during the pollution days. The calculated ozone formation potentials (OFPs) for various precursors (carbonyls and VOCs) indicated that carbonyl compounds contributed 32.87% of the total OFPs on nonpollution days and 36.71% on pollution days, respectively. Formaldehyde, acetaldehyde, and methylglyoxal were identified as the most reactive carbonyls, and formaldehyde ranked top in OFPs, and it alone contributed 15.92% of total OFPs on nonpollution days and 18.10% of total OFPs on pollution days, respectively. The calculation of relative incremental reactivity (RIR) indicates that ozone sensitivity was a VOC-limited regime, and carbonyls showed greater RIRs than other groups of VOCs. The model simulation showed that secondary formation has a significant impact on formaldehyde production, which is primarily controlled by alkenes and biogenic VOCs. The characteristic ratios and backward trajectory analysis also indicated the indispensable impacts of local primary sources (like industrial emissions and vehicle emissions) and regional sources (like biomass burning) through transportation. This study highlights the important roles of carbonyls, particularly formaldehyde, in forming ozone pollution in megacities like the Pearl River Delta region.

Abstract Accurately measuring emission factors (Es) of biogenic volatile organic compounds (BVOCs) with consideration of intraspecific variability is vital but often overlooked. This study presents in‐situ measurements of BVOC emissions from 114 Eucalyptus urophylla individuals using the LI‐6800 portable photosynthesis system. We observed intraspecific variability exceeding an order of magnitude in BVOC Es. Despite this variability, our approach yielded statistically representative Es for E. urophylla, yet challenging the feasibility of extensive field measurements. By quickly screening net photosynthesis rate (Pn) across a broad set of individuals and selecting those within a specific Pn range, such as mean ± 0.1 × SD (standard deviation) of Pn for all screened individuals, for detailed BVOC emission measurements, we achieved comparable mean Es with approximately 10% of the original sampling effort. This offers a practical solution for efficient and accurate field measurement of representative BVOC Es, significantly reducing required sample size while effectively addressing intraspecific variability.

Abstract Background Coronavirus disease 19 (COVID-19) is a major public health problem that cannot be ignored. As a widely used drug in the treatment of COVID-19, whether glucocorticoids may accelerate the clearance of COVID-19 is still not clear, and the glucocorticoids may improve the prognosis of patients is also controversial. Therefore, to explore the relationship between COVID-19 viral load and the use of glucocorticoids we designed this study. Methods Patients with COVID-19 infection who were admitted to the emergency department of Peking Union Medical College Hospital from the end of 2022 to early 2023 were enrolled in this study. Characteristics of baseline, clinical and laboratory evaluation especially immunological indicator and daily viral load were carefully collected. Kolmogorov–Smirnov test, Student’s t test, Mann–Whitney U test and proportional-hazards model (Cox model) were chosen as appropriate for comparison of variables. Results By comparing the daily COVID-19 viral load and prognosis of patients with and without glucocorticoid therapy, we found that glucocorticoids did not statistically enhance the clearance or replication of COVID-19, nor did it change the 28-days and in-hospital mortality. However, glucocorticoid therapy may be a favorable factor for COVID-19 negative conversion in Cox model. The inflammatory factors in patients with glucocorticoid therapy were significantly decreased. Conclusions We believe that the real effect of glucocorticoids may be to improve the destruction of host immune system caused by inflammatory storm through host immune regulation and then achieve the improvement of clinical symptoms.

Abstract Type 1 conventional dendritic cells (cDC1) are the most efficient cross-presenting cells that induce protective cytotoxic T cell response. However, the regulation of their homeostasis and function is incompletely understood. Here we observe a selective reduction of splenic cDC1 accompanied by excessive cell death in mice with Zeb1 deficiency in dendritic cells, rendering the mice more resistant to Listeria infection. Additionally, cDC1 from other sources of Zeb1-deficient mice display impaired cross-presentation of exogenous antigens, compromising antitumor CD8+ T cell responses. Mechanistically, Zeb1 represses the expression of microRNA-96/182 that target Cybb mRNA of NADPH oxidase Nox2, and consequently facilitates reactive-oxygen-species-dependent rupture of phagosomal membrane to allow antigen export to the cytosol. Cybb re-expression in Zeb1-deficient cDC1 fully restores the defective cross-presentation while microRNA-96/182 overexpression in Zeb1-sufficient cDC1 inhibits cross-presentation. Therefore, our results identify a Zeb1-microRNA-96/182-Cybb pathway that controls cross-presentation in cDC1 and uncover an essential role of Zeb1 in cDC1 homeostasis.

To investigate the characteristics and sources of volatile organic compounds (VOCs) as well as their impacts on secondary organic aerosols (SOAs) formation during high-incidence periods of PM2.5 pollution, a field measurement was conducted in December 2019 in Hefei, a typical city of the Yangtze River Delta (YRD). During the whole process, the mixing ratios of VOCs were averaged as 21.1 ± 15.9 ppb, with alkanes, alkenes, alkyne, and aromatics accounting for 59.9%, 15.3%, 15.0%, and 9.8% of the total VOCs, respectively. It is worth noting that the contributions of alkenes and alkyne increased significantly during PM2.5 pollution periods. Based on source apportionment via the positive matrix factorization (PMF) model, vehicle emissions, liquefied petroleum gas/natural gas (LPG/NG), and biomass/coal burning were the main sources of VOCs during the research in Hefei. During pollution periods, however, the contribution of biomass/coal burning to VOCs increased significantly, reaching as much as 47.6%. The calculated SOA formation potential (SOAFP) of VOCs was 0.38 ± 1.04 µg m−3 (range: 0.04–7.30 µg m−3), and aromatics were the dominant contributors, with a percentage of 96.8%. The source contributions showed that industrial emissions (49.1%) and vehicle emissions (28.3%) contributed the most to SOAFP during non-pollution periods, whereas the contribution of biomass/coal burning to SOA formation increased significantly (32.8%) during PM2.5 pollution periods. These findings suggest that reducing VOCs emissions from biomass/coal burning, vehicle, and industrial sources is a crucial approach for the effective control of SOA formation in Hefei, which provides a scientific basis for controlling PM2.5 pollution and improving air quality in the YRD region.

Abstract Heterogeneous reaction of gas phase NO2 with atmospheric humic-like substances (HULIS) is potentially an important source of volatile organic compounds (VOCs) including nitrogen (N)-containing compounds, a class of brown carbon of emerging importance. However, the role of ubiquitous water-soluble aerosol components in this multiphase chemistry, namely nitrate and iron ions, remains largely unexplored. Here, we used secondary electrospray ionization ultrahigh-resolution mass spectrometry for real-time measurements of VOCs formed during the heterogeneous reaction of gas phase NO2 with a solution containing gallic acid (GA) as a proxy of HULIS at pH 5 relevant for moderately acidic aerosol particles. Results showed that the number of detected N-containing organic compounds largely increased from 4 during the NO2 reaction with GA in the absence of nitrate and iron ions to 55 in the presence of nitrate and iron ions. The N-containing compounds have reduced nitrogen functional groups, namely amines, imines and imides. These results suggest that the number of N-containing compounds is significantly higher in deliquescent aerosol particles due to the influence of relatively higher ionic strength from nitrate ions and complexation/redox reactivity of iron cations compared to that in the dilute aqueous phase representative of cloud, fog, and rain water.

Abstract A pediatric patient suffered from renal and hepatic abscesses (through hepatorenal space) and recovered by antibiotics and other therapies. By clinical analysis, the multi-organ abscesses might be caused by bloodstream-disseminated infection. In order to identify the pathogen, we collected kidney biopsy tissue, swabs, and plasma samples, and used metagenomics next-generation sequencing (mNGS) and some traditional methods. The results revealed that polymicrobial especially anaerobic bacteremia (Bacteroides fragilis, et al.) contributed to the abscess formation. What is more, systematic human adenovirus C (HAdV-C) infection was shown, and the virus was isolated. The titer of HAdV-2 neutralizing antibodies was 1/4 in the plasma after symptoms onset. Although the exact mechanism of HAdV-2 infection in multiple abscess formation has not been clarified, the case of multi-organ abscesses in the context of polymicrobial especially anaerobic bacteremia and HAdV infection in healthy children is infrequent.

The effect of configuration entropy (ΔSconf) on thermal properties and magnetocaloric effects (MCE) of metallic glasses (MGs) remains vague. In this work, Tb55Co17.5Al27.5 low-, (Gd0.5Tb0.5)55Co17.5Al27.5 medium-, and (Gd1/3Tb1/3Dy1/3)55Co17.5Al27.5 high-entropy (LE, ME, and HE) MGs with critical diameters more than 5 mm were successfully fabricated. Systematic investigations into their thermodynamic behaviors suggest that high ΔSconf might deteriorate glass forming ability (GFA) along with reduced viscosity. Simultaneously, an improved thermal stability was observed in ME and HE MGs for their high activation energies for glass transition, crystal nucleation and growth, while the notable ordered structure in HE MG could accelerate glass transition and crystal growth by reducing activation energies. Studies of MCE indicate that the distinguished spin glass behaviors and higher structural order in HE MGs comprehensively contribute to an enhanced relative cooling power and a broader magnetic-transition temperature range, whereas the change of Curie temperature and maximum magnetic entropy more depends on the de Gennes factor rather than ΔSconf. All three designed MGs manifesting the competitive GFA and MCE could be candidates for magnetic refrigeration. Our studies show that entropy tailoring seems to play a dominant role in determining thermodynamic behaviors, which could help to design more stable MCE MGs.

The gut microbiota undergoes substantial changes in COVID-19 patients; yet, the utility of these alterations as prognostic biomarkers at the time of hospital admission, and its correlation with immunological and hematological parameters, remains unclear. The objective of this study is to investigate the gut microbiota's dynamic change in critically ill patients with COVID-19 and evaluate its predictive capability for clinical outcomes alongside immunological and hematological parameters. In this study, anal swabs were consecutively collected from 192 COVID-19 patients (583 samples) upon hospital admission for metagenome sequencing. Simultaneously, blood samples were obtained to measure the concentrations of 27 cytokines and chemokines, along with hematological and biochemical indicators. Our findings indicate a significant correlation between the composition and dynamics of gut microbiota with disease severity and mortality in COVID-19 patients. Recovered patients exhibited a higher abundance of Veillonella and denser interactions among gut commensal bacteria compared to deceased patients. Furthermore, the abundance of gut commensal bacteria exhibited a negative correlation with the concentration of proinflammatory cytokines and organ damage markers. The gut microbiota upon admission showed moderate prognostic prediction ability with an AUC of 0.78, which was less effective compared to predictions based on immunological and hematological parameters (AUC 0.80 and 0.88, respectively). Noteworthy, the integration of these three datasets yielded a higher predictive accuracy (AUC 0.93). Our findings suggest the gut microbiota as an informative biomarker for COVID-19 prognosis, augmenting existing immune and hematological indicators.

Abstract Background Community-acquired pneumonia (CAP) is a major public health challenge worldwide. However, the aetiological and disease severity-related pathogens associated with CAP in adults in China are not well established based on the detection of both viral and bacterial agents. Methods A multicentre, prospective study was conducted involving 10 hospitals located in nine geographical regions in China from 2014 to 2019. Sputum or bronchoalveolar lavage fluid (BALF) samples were collected from each recruited CAP patient. Multiplex real-time PCR and bacteria culture methods were used to detect respiratory pathogens. The association between detected pathogens and CAP severity was evaluated. Results Among the 3,403 recruited eligible patients, 462 (13.58%) had severe CAP, and the in-hospital mortality rate was 1.94% (66/3,403). At least one pathogen was detected in 2,054 (60.36%) patients, with two or more pathogens were co-detected in 725 patients. The ten major pathogens detected were Mycoplasma pneumoniae (11.05%), Haemophilus influenzae (10.67%), Klebsiella pneumoniae (10.43%), influenza A virus (9.49%), human rhinovirus (9.02%), Streptococcus pneumoniae (7.43%), Staphylococcus aureus (4.50%), adenovirus (2.94%), respiratory syncytial viruses (2.35%), and Legionella pneumophila (1.03%), which accounted for 76.06–92.52% of all positive detection results across sampling sites. Abstract Myocardial infarction (MI) is a major cause of mortality worldwide. The major limitation of regenerative therapy for MI is poor cardiac retention of therapeutics, which results from an inefficient vascular network and poor targeting ability. In this study, a two‐layer intrinsically magnetic epicardial patch (MagPatch) prepared by 3D printing with biocompatible materials like poly (glycerol sebacate) (PGS) is designed, poly (ε‐caprolactone) (PCL), and NdFeB. The two‐layer structure ensured that the MagPatch multifariously utilized the magnetic force for rapid vascular reconstruction and targeted drug delivery. MagPatch accumulates superparamagnetic iron oxide (SPION)‐labelled endothelial cells, instantly forming a ready‐implanted organization, and rapidly reconstructs a vascular network anastomosed with the host. In addition, the prefabricated vascular network within the MagPatch allowed for the efficient accumulation of SPION‐labelled therapeutics, amplifying the therapeutic effects of cardiac repair. This study defined an extendable therapeutic platform for vascularization‐based targeted drug delivery that is expected to assist in the progress of regenerative therapies in clinical applications.

ABSTRACTThe adaptive immunity against SARS-CoV-2 prototype strain and Omicron sublineages induced by BA.1 breakthrough infection in vaccinees of inactivated COVID-19 vaccines have not been well characterized. Here, we report that BA.1 breakthrough infection induced mucosal sIgA and resulted in higher IgG titers against prototype strain and Omicron sublineages in vaccinees than in vaccine naïve-infected individuals. BA.1 breakthrough infection boosted antibody-dependent cellular cytotoxicity and antibody-dependent cellular phagocytosis to prototype strain and BA.1, BA.1.1, BA.2, BA.2.12.1, and BA.2.75 but not BA.4/5 and induced neutralization against prototype strain and BA.1, BA.1.1, BA.2, BA.2.12.1, BA.2.75, and BA.4/5 but not BF.7, BQ.1, and XBB. In total, BA.1 breakthrough infection individuals produced less extensive sIgA, plasma IgG and NAb responses against Omicron sublineages compared with those against prototype strain. Further, BA.1 breakthrough infection induced recall B cell response to prototype strain and Omicron variant, primarily targeting memory B cells producing conserved epitopes. Memory T cell responses against Omicron is largely preserved. Individuals with vaccine booster did not induce more beneficial immune responses to Omicron sublineages upon BA.1 breakthrough infection than those with primary vaccine dose only. The breakthrough infection individuals produced stronger adaptive immunity than those of inactivated vaccine-healthy individuals. These data have important implications for understanding the vaccine effectiveness and adaptive immunity to breakthrough infection in individuals fully immunized with inactivated vaccines. Omicron sublineages, especially for those emerged after BA.4/5 strain, evade NAb responses induced by BA.1 breakthrough infection. It is urgent to optimize the vaccine immunogen design and formulations to SARS-CoV-2 variants.

Abstract The profile of antibodies against antigenic epitopes of severe acute respiratory syndrome coronavirus 2 (SARS‐CoV‐2) during neutralizing antibody (NAb) decay has not been clarified. Using a SARS‐CoV‐2 proteome microarray that contained viral antigenic peptides, we analyzed the characteristics of the humoral response in patients with coronavirus disease 19 (COVID‐19) in a longitudinal study. A total of 89 patients were recruited, and 226 plasma samples were serially collected in 2020. In the antigenic peptide microarray, the level of immunoglobulin G (IgG) antibodies against peptides within the S2 subunit (S‐82) and a conserved gene region in variants of interest, open reading frame protein 10 (ORF10‐3), were closely associated with the presence of SARS‐CoV‐2 NAbs. In an independent evaluation cohort of 232 plasma samples collected from 116 COVID‐19 cases in 2020, S82‐IgG titers were higher in NAbs‐positive samples (p = 0.002) than in NAbs‐negative samples using enzyme‐linked immunosorbent assay. We further collected 66 plasma samples from another cohort infected by Omicron BA.1 virus in 2022. Compared with the samples with lower S82‐IgG titers, NAb titers were significantly higher in the samples with higher S82‐IgG titers (p = 0.04). Our findings provide insights into the understanding of the decay‐associated signatures of SARS‐CoV‐2 NAbs.

The exchange of isoprenoids, which includes isoprene, monoterpenes, and sesquiterpenes, between ecosystem soils and the atmosphere plays a significant role in soil ecology and atmospheric chemistry. However, research on flux exchange rates in subtropical ecosystems has been limited, as previous studies have mainly focused on temperate and boreal environments. In this study, we aimed to quantify the exchange of isoprenoids between the soil (with or without surface litter) and the atmosphere in a subtropical Eucalyptus urophylla plantation forest during the daytime in the wet season of subtropical China. Additionally, we investigated the influence of soil and litter variables on the fluxes of isoprenoids. Our results unveiled the exchange of isoprene and 17 terpenoid compounds, comprising 11 monoterpenes and 6 sesquiterpenes, between the studied soils and the atmosphere. Interestingly, regardless of the presence of surface litter, the studied soils acted as net sinks for isoprenoids, with isoprene being the most absorbed compound (−71.84 ± 8.26 μg m−2 h−1). The removal of surface litter had a significant impact on the exchange rates of two monoterpenes (α-pinene and β-pinene), resulting in decreased fluxes. Furthermore, the exchange rates of isoprene were positively correlated with litter dry weight and negatively correlated with soil temperature. The higher exchange rates of monoterpenes and sesquiterpenes were associated with increased levels of soil respiration and the abundance of leaf litter. These findings suggest that, in the context of projected global warming scenarios, the capacity of subtropical soils to act as sinks for isoprenoids is expected to increase in subtropical China. These changes in sink capacity may have implications for regional-scale atmospheric chemistry and ecosystem functioning.
