We investigated the relationship between membership in an accredited Clubhouse for mental health support and psychiatric hospitalization in Canada using linked administrative data. Results show that Clubhouse members were less likely to be hospitalized after enrollment and after longer-term enrollment, and younger members diagnosed with schizophrenia and/or bipolar disorders were at increased risk of hospitalization compared to older members without such diagnoses. These findings provide evidence of the possible benefits of Clubhouses in Canada and the characteristics of members who may benefit from support.

The western Tianshan Gold Belt hosts numerous giant and large gold deposits that have been formed during the late Paleozoic amalgamation of the Tianshan orogen. However, little is known about their exhumation histories during the Mesozoic to Cenozoic intracontinental evolution of the orogen. The Carboniferous Katebasu orogenic gold deposit in northwestern China is a new gold discovery within the western Tianshan Gold Belt, and it shares many similarities with other orogenic gold deposits in the belt. In this contribution, new 40 Ar/ 39 Ar and (U–Th)/He ages were combined with previous geochronology and numerical modelling to quantify its post-Carboniferous cooling and exhumation history. The results revealed a three-phase cooling history and two phases of post-mineralization exhumation. We suggest that a large volume ( c. 0.8 km) of the mineralized roof parts of the Katebasu deposit might have been removed during uplift and erosion, whereas significant ore reserves could still exist at depth. The large erosion depth of the Katebasu gold deposit in the Nalati Range of the Chinese western Tianshan also signifies that shallow-emplaced porphyry and epithermal systems that formed prior to Permo-Triassic uplift might have been largely eroded.

Youth in special education have complex needs that are supported across multiple systems. Our research investigates the use of adult income assistance, as one structure that supports youth as they transition to adulthood. We created a cohort of youth (5-22 years old) using linked administrative data from British Columbia government ministries from 1996 to 2018. Youth were grouped by their special education funding (most to least; Level 1, Level 2, Level 3, Unfunded, and no special education). We investigated (1) youth characteristics and service use patterns, (2) which youth used income supports after the child-to-adult transition (19-22 years old), and what youth characteristics and service use patterns were associated with use, and (3) how much income support they used (CAD$). Of 174,527 youth, 254 (0.1%) were Level 1, 6020 (3.4%) were Level 2, 4409 (2.5%) were Level 3, 21,232 (12.2%) were Unfunded, and 142,612 (81.7%) were not in special education. Youth assigned higher funding levels, compared to lower levels, generally had increased service use, and in the transition to adult services were more likely to use income supports, and received more income support. An important exception was youth with serious behavioural/mental health special education funding (Level 3), who had increased service use for their level of funding, but received less income support due to a reliance on Temporary versus Disability Assistance. Youth that received an accredited diploma were less likely to use income supports. Factors related to the use of income supports are further described. This study highlights differences in access to income support when youth transition to adult services and considerations around equitable access to support.

The emergence of the plasmid-borne colistin-resistant gene (mcr-1) poses a great threat to human health. What is worse, the recent observations of the co-existence of mcr-1 with other antimicrobial resistance genes in some bacteria cause further concern. Here, we present the first report of a wild Escherichia coli strain that co-carries an mcr-1 encoding phage-like IncY plasmid (pR15_MCR-1) and a bla NDM-5 encoding IncX3 plasmid (pR15_NDM-5) from a pharmaceutical industry, wastewater treatment plant, in China. This study highlights the spreading of E. coli carrying both mcr-1 and bla NDM-5 genes in the pharmaceutical industry. Importance Escherichia coli strains that carry both mcr-1 and bla NDM-5 genes are of great health concern and are already found in humans and animals worldwide, yet there is a paucity of observations of this resistant strain in the environment. Here we present the first isolation of an E. coli strain (R15) that co-carries mcr-1 and bla NDM-5 genes from a wastewater treatment plant in China. Whole-genome sequencing indicated that R15 harbored two plasmids, pR15_MCR-1 and pR15_NDM-5, that carry mcr-1 and bla NDM-5, respectively. The observation of this wild-derived E. coli strain that carries mcr-1 and bla NDM-5 genes simultaneously calls for the urgency to improve monitoring and reducing its further spreading.

A multidrug-resistant Citrobacter freundii strain R17 was isolated from a wastewater treatment plant in China. Whole-genome sequencing of strain R17 revealed a new sequence type (ST412) chromosome (length 5,124,258 bp) and an Inc FII (Yp) group plasmid pCFR17_1 (length 206,820 bp). A total of 13 antibiotic-resistance genes (ARGs) that confer resistance to eight different antibiotic groups were encoded by strain R17 and 12 of them were carried by plasmid pCFR17_1. These data and analysis suggest that the environment-derived C. freundii strains may serve as potential sources of ARGs and highlight the need of further surveillance of this bacteria in the future.

Youth with complex service needs require extraordinary services that are often managed by different ministries (government departments) and in many cases from various service sectors. They often require education, child welfare, and justice services, and have increased exposure to poverty, mental health issues, chronic health conditions, and decreased high school graduation. While public services are designed to improve outcomes for youth with complex service need, little is known about how these youth use services during the child to adult transition period in Canada. Knowledge of service use in this transition is important as access and eligibility for services changes during this period for youth. To better understand the needs of youth with complex service need, we used cross-ministry administrative data from Alberta to identify service use pattern clusters from age 15 through 20 years. For revealed clusters, we report and discuss poverty, mental health use, high cost health service use, and high school completion covariate proportions. The data analysis revealed that those that used more services generally showed higher proportions of poverty, mental health use, high cost health care use, and high school incompletion than those that use less services. These service use clusters and covariate trajectories can be used to inform decision makers on how to allocate resources to support youth with complex service need during the child to adult transition.

The western Tianshan orogen is one of the world’s most important gold provinces. It hosts numerous world-class gold deposits along the southern and northern margins of the Middle Tianshan. Most deposits occur within large-scale fault zones and/or ductile-brittle deformation zones that were formed during the Paleozoic evolution of the Paleo-Asian Ocean, thereby indicating a correlation between orogenic tectonics and the gold mineralization. However, the correlation is often poorly constrained and remains controversially discussed. The Katebasu gold deposit in Xinjiang, China, is located on the northern margin of Middle Tianshan and is hosted by secondary fault zones to the south of the North Nalati Fault. The main period of gold mineralization is suggested to be the Late Carboniferous, which is broadly coeval with the late Paleozoic amalgamation of the western Tianshan. Therefore, the Katebasu gold deposit provides an excellent opportunity to search for possible correlations between structural processes at a deposit scale and orogenic processes at a regional scale to better understand the tectonic controls on large-scale gold mineralization in the western Tianshan. In this paper, we report detailed structural mapping and field investigations on the fault geometries and kinematics of the Katebasu gold deposit that permit inversion of the tectonic stress field. Our structural data, combined with results of previous studies of the regional tectonics, allow us to reconstruct the structural history of the Katebasu gold deposit, to look for possible correlations with the orogenic processes in the western Tianshan and to determine their control on gold mineralization. The first stage of deformation (D1) was top-to-the-north thrusting that was caused by a roughly N–S compression and is represented by ENE–WSW-striking thrust faults. D1 was likely related to the closure of the South Tianshan Ocean and the resulting collision between the Tarim craton and Middle Tianshan–Yili block. The second stage of deformation (D2) was dextral strike-slip motion along the ENE–WSW-striking faults that was caused by the WNW–ESE stress field and that corresponds to Permian dextral wrenching in the Tianshan orogen. Subsequent NE–SW compression led to the third deformation event (D3) that mainly formed NW–SE-striking reverse faults and NNE–SSW-striking dextral faults.

Mesenchymal stem cell-derived exosomes (MSC-exos) are phospholipid bimolecular vesicles containing various materials, and they mediate crosstalk among cells. MSC-exos can maintain glucose homeostasis and delay the progression of diabetes and its microvascular complications through multiple mechanisms, such as by improving β-cell viability and insulin resistance as well as through multiple signal transduction pathways. However, related knowledge has not yet been systematically summarized. Therefore, we reviewed the applications and relevant mechanisms of MSC-exos in treatments for diabetes and its microvascular complications, particularly treatments for improving islet β-cells viability, insulin resistance, diabetic nephropathy, and retinopathy.

Background Children with disabilities often face limitations that cross support sectors. Objective: Our aim was to measure cross-ministry service use, outcomes, and functional limitations faced by children who qualified for special education. Methods We used longitudinal British Columbia ministry data linked to children (0-18y) registered in K-12 education. Children were grouped by special education funding (most to least; Level 1, Level 2, Level 3, Unfunded, and no special education), and related to 1) service use patterns, 2) the age they first used disability services, and 3) functional limitations reported in health visits. We also reported how length of special education use related to disability service use. Results Of 111,274 children, 154(0.1%) were Level 1, 4427(4.0%) Level 2, 2897(2.6%) Level 3, 13472(12.1%) Unfunded, and 90324(81.2%) not in special education. Children with higher funding levels, compared to lower levels of funding, generally were more likely to experience poorer outcomes, have functional limitations, have service needs, and receive early support. One exception was children with serious behavioural/mental health special education coding, which had poorer outcomes for their level of funding. Children received child disability supports early (about half of users started by 4y), but use was mostly limited to those with many years (9+years) of funded special education (70.7% of the all users) and biased to certain special education codes (i.e., Level 1, severe intellectual disability, and autism). Conclusions This study provides evidence of the long-term, diverse needs of children in special education and may be used to inform decisions surrounding their support.

Copper mine tailings are causing great environmental concern nowadays due to their high contents of heavy metals. These hazards may release to air, water, and soil, posing great threat to the living organisms in the surroundings. In the present work, we profiled the heavy metal contents, microbiome and resistome of a mine tailing in Dexing Copper Mine, which is the largest open-pit copper mine in China. A total of 39.75 Gb clean data was generated by metagenomics sequencing and taxonomy analysis revealed Actinobacteria, Proteobacteria, Acidobacteria, Euryarchaeota, and Nitrospirae as the most abundant phylum in this tailing. In general, 76 heavy metal resistance genes (HMRGs) and 194 antimicrobial resistance genes (ARGs) were identified with merA and rpoB2 as the most abundant HMRG and ARG, respectively. We also compared the differences of heavy metal concentrations among the six sampling sites in the same tailing and found that significant differences exited in copper and zinc. Hierarchical cluster analysis showed that the samples from the six sampling sites were clustering in two groups based on heavy metal concentrations. Accordingly, clustering based on microbial composition and relative abundances of resistance genes exhibited the same clustering pattern, indicating a possible shaping influence of heavy metals on the microbiome and resistome in this tailing. Our work presented heavy metal contents, microbial composition and resistance genes in a copper mine tailing of the largest copper mine in China, and these data will of great use in the surveillance, maintenance, and remediation of this tailing.

Aim To investigate which families with young children with disabilities used disability services and when they used services to inform policy on service delivery. Method We used linked administrative data from different ministries in Alberta to describe families’ use of disability services when their children were between the ages of 3 and 8 years old. Disability was investigated on the basis of the presence of a severe special education code for children, and level of special education code. The outcome was the use of family disability services. Results Of 31 346 children, 24 761 (79.0%) had no special education code, 3982 (12.7%) had a mild special education code, and 2603 (8.3%) had a severe special education code. Level of special education code was associated with child characteristics and service use. Children with severe special education codes generally were more likely to report service use and have poor outcomes than those with less severe codes. Of note, 26% of children with severe special education codes used family disability services. In addition, among children with severe special education codes, many years of severe coding (compared with fewer years) had the strongest association with family disability service use (prevalence ratio 5.50; 95% confidence interval 4.10–7.37). Associations with family disability service use were seen with mental health, health care, and educational achievement. Interactions between child characteristics and service use were observed. Interpretation This study provides evidence that families were more likely to use disability services when they were involved with other services, and that use interacts with various factors. Background and objectives: Wastewater treatment plants (WWTPs) are one of the major reservoirs for antimicrobial resistant bacteria (ARB) and antimicrobial resistance genes (ARGs) in the environment. Thus, the investigation on ARB and ARGs from WWTPs has attracted increasing attention in recent years. In order to uncover the resistome in a WWTP treating effluents from a pharmaceutical industry in China, the extended-spectrum beta-lactamase (ESBL)-producing Escherichia coli strains were isolated and their whole genome sequences were obtained and analyzed. Moreover, metagenomic sequencing was applied to give a comprehensive view of antibiotic resistance in this WWTP. Methods: 18 ESBL-producing E. coli strains were isolated from a WWTP located in Taizhou, China on April, 2017. All strains were sequenced using Illumina HiSeq 2000 sequencer. The whole genome sequences were assembled using SPAdes software and annotated with RAST server. Sequence types (STs), plasmids, ARGs and virulence genes were predicted from the genomes using MLST, Plasmid Finder, ResFinder and Virulence Finder, respectively. Metagenomic DNA of the same sample was extracted and sequenced using Illumina Hiseq X Ten platform. Metagenomic sequences were assembled using SOAPdenovo software. Results: All 18 ESBL-producing E. coli strains were resistant to ampicillin, cefazolin and ceftriaxone. Analysis of their genomes revealed that all strains carried beta-lactamase encoding genes and the most prevalent type was blaCTX-M. Various virulence genes and ARGs confronting resistance to other types of antimicrobial agents were also predicted. Further investigation on the metagenomics data indicated 11 ARGs with high amino acid identities to the known ARGs. Five of these ARGs, aadA1, aac(6’)-lb-cr, flo(R), sul2 and sul1, were also present in the genomes of the ESBL-producing E.coli isolated from the same sample. Conclusions: Our study revealed the resistome of a pharmaceutical WWTP by both culture-dependent and metegenomic methods. The existence of ESBL-producing E. coli strains, indicating that pharmaceutical WWTP can play a significant role in the emergence of ARB. The occurrence of ARGs annotated from the metagenomic data suggests that pharmaceutical WWTP can play a significant role in the emergence of ARGs. Our findings highlight the need for strengthening the active surveillance of ARB and ARGs from pharmaceutical industry.

Metallo-β-lactamases (MBLs) are a group of enzymes that can inactivate most commonly used β-lactam-based antibiotics. Among MBLs, New Delhi metallo-β-lactamase-1 (NDM-1) constitutes an urgent threat to public health as evidenced by its success in rapidly disseminating worldwide since its first discovery. Here we report the biochemical and genetic characteristics of a novel MBL, ElBla2, from the marine bacterium Erythrobacter litoralis HTCC 2594. This enzyme has a higher amino acid sequence similarity to NDM-1 (56%) than any previously reported MBL. Enzymatic assays and secondary structure alignment also confirmed the high similarity between these two enzymes. Whole genome comparison of four Erythrobacter species showed that genes located upstream and downstream of elbla2 were highly conserved, which may indicate that elbla2 was lost during evolution. Furthermore, we predicted two prophages, 13 genomic islands and 25 open reading frames related to insertion sequences in the genome of E. litoralis HTCC 2594. However, unlike NDM-1, the chromosome encoded ElBla2 did not locate in or near these mobile genetic elements, indicating that it cannot transfer between strains. Finally, following our phylogenetic analysis, we suggest a reclassification of E. litoralis HTCC 2594 as a novel species: Erythrobacter sp. HTCC 2594.

IntroductionInnovative data platforms (e.g. biobanks, repositories) continually emerge to facilitate data sharing. Extant and emerging data platforms must navigate myriad tensions for successful data sharing and re-use. Two Alberta data platforms navigated such processes and factors regarding administrative, research and nonprofit data: the Child & Youth Data Laboratory (CYDL) and Secondary Analysis to Generate Evidence (SAGE). ObjectivesTo clarify the social and policy factors that influenced CYDL and SAGE establishment and implementation, and the relationships, if any, between these factors and data type. MethodsThis paper involves a qualitative secondary analysis of two developmental evaluations on CYDL and SAGE establishment. Six-years post-implementation, the CYDL evaluation entailed document review; website user analysis; informant interviews (n=30); online stakeholder survey (n=260); and an environmental scan. One-year post implementation, the SAGE evaluation included 15 interviews and document review. We used thematic analysis and comparisons with the literature to identify key factors. ResultsThree (not mutually exclusive) categories of social and policy factors influenced the navigation towards CYDL and SAGE realization: trusting relationships; sustainability amidst readiness; and privacy within social context. For these platforms to be able to manage, link or share data, trust had to be fostered and maintained across multiple, dynamic and intersecting relationships between primary data producers, data subjects, secondary users and institutions. Platform sustainability and readiness required capacity building and innovation. Privacy and information sharing evolved culturally and correspondingly for these data platforms, which required constant flexibility and awareness. ConclusionsThis commentary calls for more empirical research on the value of data re-use or the detriment in not re-using data. While the culture of information sharing is progressing towards greater openness and capacity for data sharing and re-use, successful data platforms must advocate, facilitate and mobilize analysis and innovation using data re-use while being cognizant of social and policy influences.

Background Mandates abound to share publicly-funded research data for reuse, while data platforms continue to emerge to facilitate such reuse. Birth cohorts (BC) involve longitudinal designs, significant sample sizes and rich and deep datasets. Data sharing benefits include more analyses, greater research complexity, increased opportunities for collaboration, amplification of public contributions, and reduced respondent burdens. Sharing BC data involves significant challenges including consent, privacy, access policies, communication, and vulnerability of the child. Research on these issues is available for biological data, but these findings may not extend to BC data. We lack consensus on how best to approach these challenges in consent, privacy, communication and autonomy when sharing BC data. We require more stakeholder engagement to understand perspectives and generate consensus. Methods Parents participating in longitudinal birth cohorts completed a web-based survey investigating consent preferences for sharing their, and their child’s, non-biological research data. Results from a previous qualitative inquiry informed survey development, and cognitive interviewing methods (n = 9) were used to improve the question quality and comprehension. Recruitment was via personalized email, with email and phone reminders during the 14-day window for survey completion. Results Three hundred and forty-six of 569 parents completed the survey in September 2014 (60.8%). Participants preferred consent processes for data sharing in future independent research that were less-active (i.e. no consent or opt-out). Parents’ consent preferences are associated with their communication preferences. Twenty percent (20.2%) of parents generally agreed that their child should provide consent to continue participating in research at age 12, while 25.6% felt decision-making on sharing non-biological research data should begin at age 18. Conclusions These finding reflect the parenting population’s preference for less project-specific permission when research data is non-biological and de-identified and when governance practices are highly detailed and rigourous. Parents recognize that children should become involved in consent for secondary data use, but there is variability regarding when and how involvement occurs. These findings emphasize governance processes and participant notification rather than project-specific consent for secondary use of de-identified, non-biological data. Ultimately, parents prefer general consent processes for sharing de-identified, non-biological research data with ultimate involvement of the child. IntroductionLinked datasets are important resources for research, but linkage errors can lead to incorrect results. For data security and privacy concerns, when linkage of personal identifiers is performed anonymously, it is difficult to assess the quality of the linked dataset. We describe the method used to perform linkage quality. Objectives and ApproachWe explored how to check the quality of linkages while preserving the privacy of individuals. We also adopted an approach that minimized time and burden on data providers involved in physical verification using randomly-generated appropriate sample sizes. To validate these linkages, data providers were given random samples of 50 unique records from both linked and unlinked individuals across two other Government programs. Data providers were asked to look at the records associated with those individuals in their original datasets. Three types of linkage results were validated: cross-program linkages, cross-program non-linkages, and within-program linkages. Proportions of false-matches and missed-matches were estimated. ResultsTwenty data providers checked their samples with two other programs which gave us a sample of 2000 individuals. The linkage process, based on anonymized personal identifiers, resulted in high true positive and high true negative rates. Agreement between human judges and the linkage software was strong. Results of this exercise and other linkage validation examinations provided confidence in the accuracy of the linkage process. With false matches occurring approximately only 3% of the time and virtually no missed-matches occurring, no adjustments were deemed necessary. Although linkage rates were reassuring, the sample sizes used for comparison were small, so it is expected that there would be significant variation associated with this 3% estimate; caution is advised in its use. Conclusion/ImplicationsProportions of false-matches and missed-matches determine linkage quality which is the base for research when linkages are performed anonymously. A low proportion of false-matches and an absence of missed-matches was an indication of robust linkages.

IntroductionYouth with complex-needs are vulnerable as a consequence of exposure to social adversity and/or chronic health conditions, and are at a high risk of school failure and justice involvement. Information about the patterns of service use across government sectors that influence the life outcomes of complex-needs youth is unknown. Objectives and ApproachYouth with complex needs often engage with multiple services across multiple government sectors for extended periods of time. Understanding the patterns and trajectory of their service use may inform programs, decision makers and government in the optimal allocation of resources to increase their life outcomes. It may reveal where and when interventions would be most effective to improve the life course for vulnerable youth. In this study, through a unique approach to link over 20 administrative longitudinal datasets and a novel trajectory clustering technique, the patterns of service use among complex-needs young Albertans is revealed and visualized. ResultsA trajectory clustering technique was applied to reveal patterns of service use among complex-needs individuals. Compared to the general population, higher proportions of youth with complex needs lived in low socio-economic neighborhoods, suffered from mental health issues, were high cost health service users, and had lower rates of high school completion. Furthermore, youth having complex needs for a longer period of time and who required multiple complex services in a given year had the poorest outcomes, in terms of high school completion, mental health issues, and other health problems. The majority of complex-needs youth came in contact with services via the education system, followed by child services/welfare. Conclusion/ImplicationsThe trajectories of service use among complex-needs youth reveals that these individuals are primarily identified through education. Consequently, educational supports would best address the development of effective programs including mental health supports and other needs.

IntroductionData collected by myriad not-for-profit (NFPs) agencies while providing social services represent a significant untapped source of intelligence. While much research has been conducted on the governance and capacity to share administrative and research data, little work has been done in the area of NFP data sources. Objectives and ApproachSix NFP agencies in Calgary participated in a data sharing project to measure various aspects of poverty. This collaboration required understanding the legal, ethical, policy and data challenges to working with data collected in the NFP sector. This case study will expose (a) the legal analysis on legal and governance issues for, and obligations of, NFPs when aiming to share and re-use information beyond service delivery in Alberta, Canada; and (b) the practical and policy challenges faced by the NFPs and repository. The legal analysis involved a systematic search of academic and grey literature; relevant legislation and case-law (especially privacy law). ResultsThe legal analysis demonstrated three key things. First, in many circumstances an NFP is not held to any information-handling legal obligations. This does not absolve NFPs from social privacy expectations, especially involving vulnerable populations. Second, privacy best practices are available to NFPs from other more-regulated sectors and from NFP sector standards. The trifecta of purpose, reasonableness and minimal extent are critical. Third, adhering to privacy best practices would not be overly costly, more efficient, and less risky of negative publicity and lost public trust from privacy complaints. These findings informed the NFP data sharing project. The six participant NFPs varied greatly in their approach to consent, governance and information-handling, which affected their data-readiness for cross-organizational data linkage and analysis. Conclusion/ImplicationsUnlike government, most NFPs have limited experience with data linkage. This paper informs ethical approaches to include NFP data in broad cross-sector data linkage. Appropriate data governance is imperative for data sharing amongst NFPs, researchers and public organizations, which promotes community service delivery, evaluation and improvements to benefit vulnerable families.

IntroductionLinking administrative data provides valuable information about individuals using government services and can be very useful for policy-makers in improving and developing services and policies. The Child and Youth Data Laboratory (CYDL) links and analyses administrative data from Alberta Government ministries to provide evidence for policy and program development. Objectives and ApproachData from 20 programs of six Government of Alberta ministries (Advanced Education, Education, Health, Children’s Services, Community and Social Services, and Justice and Solicitor General) were linked anonymously. The data spans six years from 2005/06 to 2010/11 and consists of almost 50 million records corresponding to over 2 million unique Albertans aged 0 to 25 years. A data visualization tool called the Program Overlap Matrix summarises the overlap rates among the programs. It is comprised of a matrix of squares, where each cell represents the overlap between two programs. ResultsThe Program Overlap Matrix is publically available at https://visualization.policywise.com/P2matrix/. It consists of overlap rates between programs in any study year (2005/06 to 2010/11), individual years, the first year vs. future years, and the last year vs. previous years which can be used to answer many policy-related questions such as: other service use (e.g., what other services do ESL students use?), over-represented programs (e.g., in what programs are Child Care Subsidy clients over-represented?), resilience (e.g., what is the proportion of Child Intervention clients in post-secondary institutions?), transitions (e.g., what types of services do students with special needs receive as they transition to adulthood?), and time trend (e.g., what types of services did Income Support clients receive in the past?) Conclusion/ImplicationsThe program overlap matrix is a powerful tool to discover relationships between programs. It is a useful instrument to inform public and policy-makers about the overlap rates between government programs. It can be used to answer a variety of policy-related questions.

Information is increasingly digital, creating opportunities to respond to pressing issues about human populations in near real time using linked datasets that are large, complex, and diverse. The potential social and individual benefits that can come from data-intensive science are large, but raise challenges of balancing individual privacy and the public good, building appropriate socio-technical systems to support data-intensive science, and determining whether defining a new field of inquiry might help move those collective interests and activities forward. A combination of expert engagement, literature review, and iterative conversations led to our conclusion that defining the field of Population Data Science (challenge 3) will help address the other two challenges as well. We define Population Data Science succinctly as the science of data about people and note that it is related to but distinct from the fields of data science and informatics. A broader definition names four characteristics of: data use for positive impact on citizens and society; bringing together and analyzing data from multiple sources; finding population-level insights; and developing safe, privacy-sensitive and ethical infrastructure to support research. One implication of these characteristics is that few people possess all of the requisite knowledge and skills of Population Data Science, so this is by nature a multi-disciplinary field. Other implications include the need to advance various aspects of science, such as data linkage technology, various forms of analytics, and methods of public engagement. These implications are the beginnings of a research agenda for Population Data Science, which if approached as a collective field, can catalyze significant advances in our understanding of trends in society, health, and human behavior.

As one of the key systems of the marine power plant diesel engine, the turbocharger directly affects whether the diesel engine can continuously and stably provide the power required for the ship. Owing to a number of uncontrollable factors, such as harsh working conditions and complex structures, the turbocharger may have various failures, causing it to lose its intended function. At present, the fault diagnosis of the marine turbocharger has not been paid enough attention yet and in most cases, the method of ‘ex post diagnosis’ is still adopted. When analysing the nonlinear correspondence between the failure symptoms and failure causes, it is difficult for the existing theories to meet the actual diagnostic requirements. This paper introduces the concept of gas-path diagnosis into the condition monitoring for a marine turbocharger for the first time and proposes the flow capacity index which characterizes the flow capacity of the component and the isentropic efficiency index which characterizes the operating efficiency of the component as two dimensionless evaluation indicators for turbocharger health status. Moreover, the nonlinear mapping relationship between these two health parameters and the gas-path measurable parameters of the turbocharger is studied, and a novel performance degradation evaluation method for a turbocharger is established. The proposed method has been tested in three test cases where the degradation of a model turbocharger has been analysed. These case studies have illustrated that the proposed method can accurately isolate the degraded components and further quantify the degradation of the components.

Research data repositories (RDRs) are data storage entities where data can be submitted, stored, and subsequently accessed for purposes beyond the original intent. There is little information relating to non-biological RDRs, nor considerations regarding pediatric data storage and re-use. We examined parent perspectives on pediatric, non-biological RDRs. Qualitative, descriptive methods including both interviews and focus groups were used. Purposive sampling of adult participants in two provincial birth cohorts yielded 19 interviewees and 18 focus group participants (4 groups). Transcripts were analyzed by thematic content analysis. Parent research participants strongly supported the sharing of their own, and their child’s, non-biological research data. Four themes emerged: that altruism has limits, that participants have ongoing privacy concerns, that some participants need the assurance of congruent values between themselves and researchers/research questions, and that opinions diverge for some governance issues. The establishment of RDRs is important and maximizes participants’, researchers’, and funders’ investments. Participants as data donors have concerns relating to privacy, relationships, and governance that must be considered in RDR development.

Infections with Staphylococcus aureus, a common inducer of mastitis, often result in mammary gland damage and death of various cell types. Although S. aureus was suggested to induce apoptosis in a bovine mammary epithelial cell (BMEC) line, MAC-T, it is unknown whether primary BMECs (pBMECs) apoptosis is triggered by S. aureus and the associated underlying molecular mechanisms have not been determined. Here, we demonstrated that S. aureus induced apoptosis in pBMECs in a time- and dose-dependent manner. Further, S. aureus-induced apoptosis in pBMECs was associated with activation of caspase-3 and caspase-8, but caspase-9 was not. In addition, pBMECs apoptosis was mitigated by caspase-3 and caspase-8 inhibitors, suggesting that apoptosis is initiated via caspase-8 activation. Moreover, S. aureus infection significantly increased expressions of Fas and Fas-associated death domain (FADD) of pBMECs. Taken together, our results demonstrated that S. aureus induced apoptosis in pBMECs via the Fas-FADD death receptor and subsequently triggered the caspase-8-dependent signaling.

ObjectivesUsing existing data for research can generate new knowledge and evidence for policy with relatively little cost. Privacy concerns are paramount in such secondary usage of data collected on human subjects. Information privacy protection and data security are critical considerations in reuse and repurposing of data especially linked data, longitudinal data, and large amounts of data. Data sharing and privacy protection are both in the public interest and we need to assess the risk of “doing” (sharing) as well as the risk of “not doing” (not sharing or not protecting). ApproachThe Alberta Centre for Child Family and Community Research (the Centre) establishes the Child Youth Data Lab that links and analyzes administrative data from multiple provincial ministries and the Child Data Centre of Alberta that repurposes research data and manages its access for reuse. The Centre partners with provincial Office of the Information Privacy Commissioner, Research Ethics Boards and leaders in the research communities and technology industry to design and develop measures to enable secondary use while safeguarding the data, and to explore and adopt best practices on data sharing processes, governance, and technologies. ResultsIn principle current privacy laws and regulations provide good guidance in collection, use, and disclosure of data, however there is a lack of consistency in the interpretation of these laws at the operational level with regard to secondary data use. The experiences of establishing different data sharing models at the Centre through multiple initiatives are discussed. Cross-sectoral broad partnership brings understanding and builds trusting relationships, which are crucial to establishing data sharing processes. The recognition of the significance of secondary data use to provide direction for policy and program development at the executive level provides commitment for data sharing initiatives. Strong governance structure consists multi-level ministry and multiple stakeholder involvement ensures ongoing support and engagement. The highest data security standards and anonymous solution for data linkage enables the sharing of data with good privacy protection. ConclusionSecondary use of data to improve system performance and contributing to scientific discovery has been broadly recognized. A balance between utility and privacy can be realized through broad partnership in building proper governance, technology, processes and policies.

As the main cause of failure and damage to rotating machinery, rolling bearing failure can result in huge economic losses. As the rolling bearing vibration signal is nonlinear and has non-stationary characteristics, the health status information distributed in the rolling bearing vibration signal is complex. Using common time-domain or frequency-domain approaches cannot easily enable an accurate assessment of rolling bearing health. In this paper, a novel rolling bearing fault diagnostic method based on multi-dimensional characteristics was developed to meet the requirements for accurate diagnosis of different fault types and severities with real-time computational performance. First, a multi-dimensional feature extraction algorithm based on entropy characteristics, Holder coefficient characteristics and improved generalized fractal box-counting dimension characteristics was performed to extract the health status feature vectors from the bearing vibration signals. Second, a grey relation algorithm was employed to achieve bearing fault pattern recognition intelligently using the extracted multi-dimensional feature vector. This experimental study has illustrated that the proposed method can effectively recognize different fault types and severities after integration of the improved fractal box-counting dimension into the multi-dimensional characteristics, in comparison with existing pattern recognition methods.

Schistosomiasis japonica is an endemic, zoonotic disease of major public health importance in China. Vaccination is needed as a complementary approach to the ongoing control programs. In the present study, we determined if the efficacies of DNA vaccine encoding the SjGST and Sj32 asparaginyl endopeptidase protein could be enhanced by boosting with SjGST-32 protein vaccines. Mice were inoculated with a VR1012-SjGST-32 DNA vaccine followed by boosting with rSjGST-32 at 0, 14 and 28 d. Two weeks after the final boost, mice were challenged percutaneously with cercariae. On day 45 following the challenge, all mice were sacrificed and the numbers of recovered worms and hepatic eggs were counted. Moreover, we analyzed the immune response among various vaccination groups. The results showed that DNA vaccine efficacy was enhanced when mice were boosted with protein vaccine. Adult worm and liver egg burdens were reduced 42.3% and 59.6%, respectively. We further found that DNA vaccine followed by boosting with protein significantly increased the IgG titer and T cell proliferation over those seen in mice vaccinated solely with DNA vaccines. Furthermore, the higher level of IFN-gamma expression in the splenetic CD4+ T cell showed that DNA prime-Protein boosting vaccine induced CD4+ Th1-type responses. Thus, DNA vaccine efficacy was significantly enhanced via boosting protein vaccine which might provide a basis for rational application of the Schistosoma vaccine.

SUMMARY Estimates of national population were studied in two experiments. In Experiment 1, Canadian and Chinese undergraduates rated their knowledge of 112 countries and then estimated the population of each. In Experiment 2, Canadians rated their knowledge of 52 countries and then provided population estimates for these primed countries and for a comparable set of 52 unprimed countries. In Experiment 1, participants from both nations produced estimates that resembled those obtained from Americans in prior studies (Brown and Siegler, 1992, 1993, 1996, 2001). However, there were several reliable cross-national differences in performance which appear to reflect cross-cultural differences in task-relevant naive domain knowledge. In addition, both experiments produced findings consistent with the claim that availability-based intuitions play an important role in this task. In Experiment 1, cross-national differences in rated knowledge predicted cross-national differences in estimated population; in Experiment 2, primed country names elicited larger population estimates than unprimed country names. We conclude by arguing for the general utility of this hybrid approach to real-world estimation. Copyright # 2002 John Wiley & Sons, Ltd.
