Abstract Background Nitrogen (N) is a crucial element for increasing photosynthesis and crop yields. The study aims to evaluate the photosynthetic regulation and yield formation mechanisms of different nodulating peanut varieties with N fertilizer application. Method The present work explored the effect of N fertilizer application rates (N0, N45, N105, and N165) on the photosynthetic characteristics, chlorophyll fluorescence characteristics, dry matter, N accumulation, and yield of four peanut varieties. Results The results showed that N application increased the photosynthetic capacity, dry matter, N accumulation, and yield of peanuts. The measurement of chlorophyll a fluorescence revealed that the K-phase, J-phase, and I-phase from the OJIP curve decreased under N105 treatment compared with N0, and WOI, ET0/CSM, RE0/CSM, ET0/RC, RE0/RC, φPo, φEo, φRo, and Ψ0 increased, whereas VJ, VI, WK, ABS/RC, TR0/RC, DI0/RC, and φDo decreased. Meanwhile, the photosystem activity and electron transfer efficiency of nodulating peanut varieties decreased with an increase in N (N165). However, the photosynthetic capacity and yield of the non-nodulating peanut variety, which highly depended on N fertilizer, increased with an increase in N. Conclusion Optimized N application (N105) increased the activity of the photosystem II (PSII) reaction center, improved the electron and energy transfer performance in the photosynthetic electron transport chain, and reduced the energy dissipation of leaves in nodulating peanut varieties, which is conducive to improving the yield. Nevertheless, high N (N165) had a positive effect on the photosystem and yield of non-nodulating peanut. The results provide highly valuable guidance for optimizing peanut N management and cultivation measures.

BackgroundNon-alcoholic Fatty Liver Disease (NAFLD) is a prevalent condition characterized by the accumulation of fat in the liver, often linked with increased risk for multi-systemic diseases. This study aims to investigate the relationship between NAFLD and mortality, particularly all-cause and cardiovascular mortality, among United States adults.MethodsData from the National Health and Nutrition Examination Survey (NHANES) were utilized, encompassing 80,312 participants from 2003–2004 to 2017–2018. After exclusions for incomplete data, potential other liver diseases, and significant alcohol consumption, the final analytical cohort included 32,698 participants. The Fatty Liver Index (FLI), a non-invasive diagnostic tool, was used to identify NAFLD. Covariates included demographic characteristics, lifestyle factors, and biochemical parameters. Survival analysis was conducted using a weighted Cox proportional hazards regression model to quantify the impact of NAFLD on mortality.ResultsThe study revealed that NAFLD was significantly associated with increased risks of all-cause and cardiovascular disease (CVD) mortality. The hazard ratios (HRs) from the survival analysis consistently indicated a higher risk among participants with NAFLD compared to those without. Subgroup analyses further confirmed the association, with notable exceptions in certain subgroups such as those with high school education and diabetes. Additionally, a nonlinear relationship between serum uric acid (SUA) levels and mortality risk was identified among NAFLD participants.ConclusionNon-alcoholic Fatty Liver Disease is a significant risk factor for all-cause and CVD mortality in US adults. The findings underscore the importance of early detection and intervention for NAFLD to mitigate its impact on public health. Further research is needed to explore the complex interactions between NAFLD, SUA levels, and mortality, particularly in high-risk subgroups.

Traumatic Choledochal Leak and Stenosis in Children is a relatively rare bile leakage, and there is no report of endoscopic retrograde cholangiopancreatography [ERCP] bile duct stent treatment in 3 years old, which avoids procedure and biliary tract modifications after the failure of conservative treatment of bile leakage, and solves the previous need for procedure in a minimally invasive or even non-invasive way. At the same time, it can be concluded that identifying the signs of conservative treatment failure is important, and ERCP is superior to CT and MR in diagnosing traumatic biliary leak in young children. Traumatic bile leakage common bile duct inflammatory stenosis, brittle tissue, not suitable for expansion, the choice of 5 Fr to 10 Fr plastic stent sequential treatment is an effective regimen. At the same time, it is necessary to closely monitor the biliary patency after ERCP to understand the long-term postoperative efficacy. Comprehensive evaluation before ERCP and detailed post ERCP monitoring require the participation of pediatricians.

IntroductionIn maize and peanut intercropping, shading emerges as a critical factor for restricting peanut growth, yield, and quality.MethodsThis study investigated the impact of 30% shade on shade-tolerant [Huayu 22 (HY22) and Fuhua 12 (FH12)] and shade-sensitive [Nonghua 11 (NH11) and Nonghua 5(NH5)] peanut varieties, with non-shaded condition as the control (CK). The effects of shade stress on plant morphology, photosynthetic characteristics, dry-matter accumulation, chloroplast ultra-microstructure, yield, and quality of different shade-tolerant peanut varieties were examined.ResultsCompared to that in the control, shade stress led to an elongation of the main stem, shortening of the lateral branches, and reduction in the leaf area. However, these changes were less significant in the shade-tolerant than in the shade-sensitive peanut varieties, with minimal effect on the elongation of the main stem height and shortening of the lateral branches. Differences in leaf area became significant during the later stages of shade stress, particularly pronounced in the shade-sensitive peanut varieties. To enhance light capture by leaves, the shade-tolerant peanut varieties exhibited increased chlorophyll content and chloroplast grain-layer numbers. The decrease in the chlorophyll a/b ratio was more pronounced in the shade-tolerant than in the shade-sensitive peanut varieties, with significant differences. However, reduced activities of ribulose 1,5-biphosphate (RuBP) carboxylase/oxygenase and fructose 1,6-biphosphate aldolase (FBA) resulted in decreased net photosynthetic rates, particularly evident in the shade-sensitive peanut varieties during the late shade period. Shade stress led to decreased dry-matter accumulation, reduced weight of 100 fruits and kernels, and a significant decline in yield in the shade-sensitive cultivars. Shading also affected peanut-kernel quality. Compared with that in the control, the protein content increased and amino-acid (except cysteine) content decreased in the shade-tolerant cultivars.DiscussionUnder shade stress, shade tolerant peanut varieties have increased the yield by improving the photosynthetic efficiency, which provided a reference for rational selection of shade tolerant peanut varieties in maize and peanut intercropping system.

During the longitudinal motion of a supercavitating vehicle, the stability control problem is complicated because of the nonlinear planing force on the tail part. The dynamic model of a supercavitating vehicle in longitude plane is nonlinear, simultaneously, the control instructions of a supercavitating vehicle may exceed the physical limits of an actuator. Therefore, designing a longitudinal stability control system for a supercavitating vehicle, not only the treatment of nonlinear planing force, but also the physical constraints of the actuator should be considered. For the longitudinal motion model of supercavitating vehicle, a cascade model is proposed, which decomposes the longitudinal motion of supercavitating vehicle into two subsystems. Sliding mode control based on RBF neural network compensation is adopted in the controller design process, and RBF neural network is exploited to approach the deviation caused by actuator saturation. The proposed control method can effectively compensate the performance degradation caused by control variable saturation, and has strong robustness.

We described insulin glargine (originator) and insulin glargine-yfgn (biosimilar) treatment patterns, assessed effectiveness and safety outcomes, and identified reasons for switching back to the originator product from the biosimilar. This retrospective study included 328,463 Veterans 18 years of age and older who received one or more outpatient prescriptions for insulin glargine and/or insulin glargine-yfgn between 1 June 2021 and 31 December 2022. Patients were assigned to subgroups based on the initial prescription during the study period, prevalent versus incident use for originator insulin glargine, and prior versus no prior use of the originator before the biosimilar (i.e., prevalent originator non-switcher (n = 189,734), originator switch to biosimilar (n = 81,010), incident originator non-switcher (n = 49,401), and incident biosimilar (n = 8318)). There were no differences in the outcome of mean HbA1c (7.9% for all subgroups). There were also no differences in the unadjusted rates of hospitalization and/or emergency room visits for hyper- and hypoglycemia between the prevalent originator non-switcher and originator switched to biosimilar subgroups (p = 0.09 and 0.38, respectively) or the incident originator non-switcher and incident biosimilar subgroups (p = 0.054 and 0.61, respectively). Finally, none of the HbA1c or hyperglycemia outcomes adjusted for baseline characteristics were statistically different. Adjusted analyses for rates of hospitalization and/or emergency room visits for hypoglycemia could not be performed due to the low number of events. Overall, patients who received insulin glargine-yfgn had similar effectiveness and safety outcomes as patients who received the originator.

BackgroundBorder row effects impact the ecosystem functions of intercropping systems, with high direct interactions between neighboring row crops in light, water, and nutrients. However, previous studies have mostly focused on aboveground, whereas the effects of intercropping on the spatial distribution of the root system are poorly understood. Field experiments and planting box experiments were combined to explore the yield, dry matter accumulation, and spatial distribution of root morphological indexes, such as root length density (RLD), root surface area density (RSAD), specific root length (SRL), and root diameter (RD), of maize and peanut and interspecific interactions at different soil depths in an intercropping system.ResultsIn the field experiments, the yield of intercropped maize significantly increased by 33.45%; however, the yield of intercropped peanut significantly decreased by 13.40%. The land equivalent ratio (LER) of the maize–peanut intercropping system was greater than 1, and the advantage of intercropping was significant. Maize was highly competitive (A = 0.94, CR=1.54), and the yield advantage is mainly attributed to maize. Intercropped maize had higher RLD, RSAD, and SRL than sole maize, and intercropped peanut had lower RLD, RSAD, and SRL than sole peanut. In the interspecific interaction zone, the increase in RLD, RSAD, SRL, and RD of intercropped maize was greater than that of intercropped peanut, and maize showed greater root morphological plasticity than peanut. A random forest model determined that RSAD significantly impacted yield at 15–60 cm, while SRL had a significant impact at 30–60 cm. Structural equation modeling revealed that root morphology indicators had a greater effect on yield at 30–45 cm, with interactions between indicators being more pronounced at this depth.ConclusionThese results show that border-row effects mediate the plasticity of root morphology, which could enhance resource use and increase productivity. Therefore, selecting optimal intercropping species and developing sustainable intercropping production systems is of great significance.

Photovoltaic inverters are widely utilized in microgrid systems working as the key equipment for converting solar energy into usable electricity. This paper presents a fuzzy sliding mode control (FSMC) method for the photovoltaic inverter in a microgrid. The inverter module uses voltage control to achieve stable AC output voltage. Moreover, to deal with system uncertainty and non-linearity in the photovoltaic inverter, a fuzzy controller was designed to realize real-time adaptation of the gains of the constant term and the reaching term in the sliding mode control law, which serves as a compensating controller for traditional sliding mode control. The gains of the exponential reaching law can be adjusted according to the system state instead of fixed gains, which can effectively reduce the chattering phenomenon and improve the robustness of the photovoltaic microgrid. Finally, the Lyapunov stability theory was used to ensure the stability of the entire control system, achieving a high-performance independent power supply for loads in a microgrid. Simulation results show that the designed control system is more robust to load disturbances and has superior dynamic performance.

Abstract Background Pod size is an important yield target trait for peanut breeding. However, the molecular mechanism underlying the determination of peanut pod size still remains unclear. Results In this study, two peanut varieties with contrasting pod sizes were used for comparison of differences on the transcriptomic and endogenous hormonal levels. Developing peanut pods were sampled at 10, 15, 20, 25 and 30 days after pegging (DAP). Our results showed that the process of peanut pod-expansion could be divided into three stages: the gradual-growth stage, the rapid-growth stage and the slow-growth stage. Cytological analysis confirmed that the faster increase of cell-number during the rapid-growth stage was the main reason for the formation of larger pod size in Lps. Transcriptomic analyses showed that the expression of key genes related to the auxin, the cytokinin (CK) and the gibberellin (GA) were mostly up-regulated during the rapid-growth stage. Meanwhile, the cell division-related differentially expressed genes (DEGs) were mostly up-regulated at 10DAP which was consistent with the cytological-observation. Additionally, the absolute quantification of phytohormones were carried out by liquid-chromatography coupled with the tandem-mass-spectrometry (LC–MS/MS), and results supported the findings from comparative transcriptomic studies. Conclusions It was speculated that the differential expression levels of TAA1 and ARF (auxin-related), IPT and B-ARR (CK-related), KAO, GA20ox and GA3ox (GA-related), and certain cell division-related genes (gene-LOC112747313 and gene-LOC112754661) were important participating factors of the determination-mechanism of peanut pod sizes. These results were informative for the elucidation of the underlying regulatory network in peanut pod-growth and would facilitate further identification of valuable target genes.

Solar active regions (ARs) provide the required magnetic energy and the topology configuration for flares. Apart from conventional static magnetic parameters, the evolution of AR magnetic flux systems should have nonnegligible effects on magnetic energy store and the trigger mechanism of eruptions, which would promote the prediction for the flare using photospheric observations conveniently. Here we investigate 322 large (M- and X-class) flares from 2010 to 2019, almost the whole solar cycle 24. The flare occurrence rate is obviously higher in the developing phase, which should be due to the stronger shearing and complex configurations caused by affluent magnetic emergences. However, the probability of flare eruptions in decaying phases of ARs is obviously higher than that in the developing phase. The confined flares were in nearly equal counts to eruptive flares in developing phases, whereas the eruptive flares were half over confined flares in decaying phases. Yearly looking at flare eruption rates demonstrates the same conclusion. The relationship between sunspot group areas and confined/erupted flares also suggested that the strong field make constraints on the mass ejection, though it can contribute to flare productions. The flare indexes also show a similar trend. It is worth mentioning that all the X-class flares in the decaying phase were erupted, without the strong field constraint. The decaying of magnetic flux systems had facilitation effects on flare eruptions, which may be consequent on the splitting of magnetic flux systems.

BackgroundHuman milk contains a complex mixture of triacylglycerols (TAG), making it challenging to recreate using common ingredients.ObjectiveThe study aimed to develop an innovative fermentation technique to produce essential human milk TAG, effectively tackling a significant hurdle in infant nutrition.MethodAn in-depth analysis of the literature has been conducted to identify the specific TAG to be targeted. We used a microalgal oil production platform and a two-step procedure to modify its fatty acid and TAG composition. The palmitic acid (16:0) content has been increased by classical strain improvement techniques, followed by a step involving the expression of a lysophosphatidic acid acyltransferase (LPAAT) sequence capable of esterifying 16:0 specifically at the internal position (sn-2 palmitate) of TAG. Once the strain was stabilized, the fermentation was scaled up in a 50-L reactor to yield several kilograms of biomass. Subsequently, the oil was extracted and refined using standard oil processing conditions. Liquid chromatography-mass spectrometry was employed to monitor the TAG profile and the region specificity of 16:0 at the internal position (sn-2 palmitate) of TAG.ResultsThe initial strain had a 16:0 level of 25% of total fatty acids, which was increased to 30% by classical strain improvement. Simultaneously, the oleic acid level decreased from 61% to 57% of total fatty acids. Upon expression of an exogenous LPAAT gene, the level of the 16:0 esterified in the internal position of the TAG (sn-2 palmitate) increased by a factor of 10, to reach 73% of total palmitic acid. Consequently, the concentration of oleic acid in the internal position decreased from 81% to 22% of total fatty acids, with TAG analysis confirming that the primary TAG species in the oil was 1,3-dioleoyl-2-palmitoyl-glycerol (OPO). The 50-L-scale fermentation trial confirmed the strain's ability to produce oil with a yield of >150 g of oil per liter of fermentation broth in a timeframe of 5 days, rendering the process scalable for larger-scale industrialization.ConclusionWe have demonstrated the feasibility of producing a suitable TAG composition that can be effectively integrated into the formulations of infant nutrition in combination with other fats and oils to meet the infant feeding requirements.

IntroductionTrehalose is vital for plant metabolism, growth, and stress resilience, relying on Trehalose-6-phosphate synthase (TPS) and Trehalose-6-phosphate phosphatase (TPP) genes. Research on these genes in cultivated peanuts (Arachis hypogaea) is limited.MethodsThis study employed bioinformatics to identify and analyze AhTPS and AhTPP genes in cultivated peanuts, with subsequent experimental validation of AhTPS9’s role in cold tolerance.ResultsIn the cultivated peanut genome, a total of 16 AhTPS and 17 AhTPP genes were identified. AhTPS and AhTPP genes were observed in phylogenetic analysis, closely related to wild diploid peanuts, respectively. The evolutionary patterns of AhTPS and AhTPP genes were predominantly characterized by gene segmental duplication events and robust purifying selection. A variety of hormone-responsive and stress-related cis-elements were unveiled in our analysis of cis-regulatory elements. Distinct expression patterns of AhTPS and AhTPP genes across different peanut tissues, developmental stages, and treatments were revealed, suggesting potential roles in growth, development, and stress responses. Under low-temperature stress, qPCR results showcased upregulation in AhTPS genes (AhTPS2-5, AhTPS9-12, AhTPS14, AhTPS15) and AhTPP genes (AhTPP1, AhTPP6, AhTPP11, AhTPP13). Furthermore, AhTPS9, exhibiting the most significant expression difference under cold stress, was obviously induced by cold stress in cultivated peanut, and AhTPS9-overexpression improved the cold tolerance of Arabidopsis by protect the photosynthetic system of plants, and regulates sugar-related metabolites and genes.DiscussionThis comprehensive study lays the groundwork for understanding the roles of AhTPS and AhTPP gene families in trehalose regulation within cultivated peanuts and provides valuable insights into the mechanisms related to cold stress tolerance.

Screening for nitrogen (N)-efficient germplasm to achieve high yield and high N efficiency is an important strategy to enhance the sustainability of modern agriculture. In this study, 127 peanut (Arachis hypogaea L.) germplasm resources were comprehensively evaluated by seedling hydroponics and field. At the seedling stage, with the range of low-nitrogen screening concentrations gradually narrowed through a comprehensive membership function analysis, standard normal distribution test, and variance analysis, we found that 0.15 mM N for 24 days could be the optimal condition for evaluating the N efficiency of peanuts. Through principal component analysis and correlation analysis, dry matter weight, root/shoot ratio, N content, N accumulation, N-use efficiency, and N use index were considered to be the N efficiency parameters, and a regression mathematical model was established accordingly. In the field, peanut genotypes that differ in resistance to low-nitrogen stress were evaluated by a yield nitrogen efficiency index under normal nitrogen and no nitrogen applications to verify the results at the seedling stage. Based on the multiple phenotypic analysis, N-efficient and N-inefficient peanut genotypes among germplasm were screened, and a comprehensive evaluation system was established to provide the theoretical basis for peanut breeding and cultivation techniques.

IntroductionThe TGA transcription factors, plays a crucial role in regulating gene expression. In cultivated peanut (Arachis hypogaea), which faces abiotic stress challenges, understanding the role of TGAs is important.MethodsIn this study, we conducted a comprehensive in analysis of the TGA gene family in peanut to elucidate their regulatory mechanisms and expression patterns under abiotic stress and hormone treatments. Furthermore, functional studies on the representative AhTGA gene in peanut cultivars were conducted using transgenic Arabidopsis and soybean hair roots.ResultsThe genome-wide analysis revealed that a total of 20 AhTGA genes were identified and classified into five subfamilies. Collinearity analysis revealed that AhTGA genes lack tandem duplication, and their amplification in the cultivated peanut genome primarily relies on the whole-genome duplication of the diploid wild peanut to form tetraploid cultivated peanut, as well as segment duplication between the A and B subgenomes. Promoter and Protein-protein interaction analysis identified a wide range of cis-acting elements and potential interacting proteins associated with growth and development, hormones, and stress responses. Expression patterns of AhTGA genes in different tissues, under abiotic stress conditions for low temperature and drought, and in response to hormonal stimuli revealed that seven AhTGA genes from groups I (AhTGA04, AhTGA14 and AhTGA20) and II (AhTGA07, AhTGA11, AhTGA16 and AhTGA18) are involved in the response to abiotic stress and hormonal stimuli. The hormone treatment results indicate that these AhTGA genes primarily respond to the regulation of jasmonic acid and salicylic acid. Overexpressing AhTGA11 in Arabidopsis enhances resistance to cold and drought stress by increasing antioxidant activities and altering endogenous hormone levels, particularly ABA, SA and JA.DiscussionThe AhTGA genes plays a crucial role in hormone regulation and stress response during peanut growth and development. The findings provide insights into peanut's abiotic stress tolerance mechanisms and pave the way for future functional studies.

Abstract Background Segmentation models such as latent class analysis are an increasingly popular approach to inform group-tailored interventions for high-risk complex patients. Multiple studies have identified clinically meaningful high-risk segments, but few have evaluated change in groupings over time. Objectives To describe population-level and individual change over time in latent comorbidity groups among Veterans at high-risk of hospitalization in the Veterans Health Administration (VA). Research design Using a repeated cross-sectional design, we conducted a latent class analysis of chronic condition diagnoses. We compared latent class composition, patient high-risk status, and patient class assignment in 2018 to 2020. Subjects Two cohorts of eligible patients were selected: those active in VA primary care and in the top decile of predicted one-year hospitalization risk in 2018 (n = 951,771) or 2020 (n = 978,771). Measures Medical record data were observed from January 2016–December 2020. Latent classes were modeled using indicators for 26 chronic health conditions measured with a 2-year lookback period from study entry. Results Five groups were identified in both years, labeled based on high prevalence conditions: Cardiometabolic (23% in 2018), Mental Health (18%), Substance Use Disorders (16%), Low Diagnosis (25%), and High Complexity (10%). The remaining 8% of 2018 patients were not assigned to a group due to low predicted probability. Condition prevalence overall and within groups was stable between years. However, among the 563,725 patients identified as high risk in both years, 40.8% (n = 230,185) had a different group assignment in 2018 versus 2020. Conclusions In a repeated latent class analysis of nearly 1 million Veterans at high-risk for hospitalization, population-level groups were stable over two years, but individuals often moved between groups. Interventions tailored to latent groups need to account for change in patient status and group assignment over time.

Changes in the canopy microclimate in intercropping systems, particularly in the light environment, have important effects on the physiological characteristics of photosynthesis and yield of crops. Although different row ratio configurations and strip widths of dwarf crops in intercropping systems have important effects on canopy microclimate, little information is available on the effects of intercropping on chlorophyll synthesis and photosynthetic physiological properties of dwarf crops. A 2-year field experiment was conducted in 2019 and 2020, with five treatments: sole maize (SM), sole peanut (SP), four rows of maize intercropping with eight rows of peanut (M4P8), four rows of maize intercropping with four rows of peanut (M4P4), and four rows of maize intercropping with two rows of peanut (M4P2). The results showed that the light transmittance [photosynthetically active radiation (PAR)], photosynthetic rate (Pn), transpiration rate (Tr), and stomatal conductance (Gs) of intercropped peanut canopy were reduced, while the intercellular carbon dioxide concentration (Ci) was increased, compared with SP. In particular, the M4P8 pattern Pn (2-year mean) was reduced by 5.68%, 5.33%, and 5.30%; Tr was reduced by 7.41%, 5.45%, and 5.95%; and Gs was reduced by 8.20%, 6.88%, and 6.46%; and Ci increased by 11.95%, 8.06%, and 9.61% compared to SP, at the flowering needle stage, pod stage, and maturity, respectively. M4P8 improves the content of chlorophyll synthesis precursor and conversion efficiency, which promotes the utilization efficiency of light energy. However, it was significantly reduced in M4P2 and M4P4 treatment. The dry matter accumulation and pod yield of peanut in M4P8 treatment decreased, but the proportion of dry matter distribution in the late growth period was more transferred to pods. The full pod number decreases as the peanut row ratio decreases and increases with year, but there is no significant difference between years. M4P8 has the highest yield and land use efficiency and can be used as a reference row ratio configuration for maize–peanut intercropping to obtain relatively high yield benefits.

Salt stress is a limiting factor affecting the growth, development, and yield of peanuts. Breeding improvement is a possible solution to overcome salt stress. The salt tolerance of 57 peanut cultivars in Northeast China was identified using indoor simulation at the germination stage, the seedling stage, and field natural identification. At the germination stage, 75 mM NaCl was the most suitable screening concentration, and the seed vitality index of 57 cultivars was analyzed using the membership function and cluster analysis. Among these cultivars, 11 were identified as salt-tolerant and 19 were salt-sensitive during germination. In the seedling stage, six salt tolerance coefficients (STCs) showed significant correlation. A gray relational analysis was used in combination with evaluation grading, resulting in the identification of 14 salt-tolerant cultivars and 12 salt-sensitive cultivars. In the field screening, a comprehensive analysis was conducted using a principal component analysis of nine indices, including agronomic characteristics, yield characteristics, and SPAD. This analysis led to the determination of three comprehensive indices. The weighted membership function was used for comprehensive evaluation. Finally, three salt-tolerant cultivars and four salt-sensitive cultivars suitable for planting in Northeast China were screened out to provide an excellent germplasm for researching the salt-tolerant mechanism of peanuts.

Coronal mass ejection (CME) is a powerful solar phenomenon that can lead to severe space weather events. Forecasting whether and when the corresponding interplanetary coronal mass ejection (ICME) will reach the Earth is very important in space weather study and forecast. At present, many different kinds of models use the near-Sun CME observations as model inputs to predict its propagation with similar prediction accuracies for large sample events. Among a series of physics-based models, the best-performing version of the shock propagation model (SPM) for large sample events, i.e., SPM3, had achieved a good forecast effect for the 23rd Solar Cycle events (1997.02–2006.12). To further evaluate SPM3, we collected CME events from 2013 January to 2023 July from the Community Coordinated Modeling Center (CCMC) CME scoreboard as a new data set. SPM3 achieved a total prediction success rate of 57% for these new events with a mean absolute error of 8.93 hr and a rms error of 10.86 hr for the shock's arrival time. Interestingly, SPM3 provided better predictions for the CME/shock events during high solar activity years than low solar activity years. We also analyzed the influence of input parameters on CME propagation and found that the larger the angular width of the CME event, the higher the probability of the corresponding IP shock's reaching the Earth. Source latitude had little effect on the arrival probability of the corresponding shock, while source longitude did. The CMEs originating from around W15° had the largest probability of hitting the Earth.

The formation mechanism of light bridges (LBs) is strongly related to the dynamic evolution of solar active regions (ARs). To study the relationship between LB formation and AR evolution phases, we employ 109 LB samples from 69 ARs in 2014 using observational data from the Helioseismic and Magnetic Imager on board the Solar Dynamics Observatory. LBs are well matched with the weak field lanes (WFLs), except those aligned on the polarity inversion line of δ sunspots. For penumbral intrusion (type-A) and umbral-dot emergence (type-C) LBs, the WFLs represent the splitting of magnetic flux systems. The sunspots tend to decay and split into several parts after type-A and type-C LBs are formed. For sunspot/umbra-merging (type-B) LBs, the declining WFLs are caused by collisions of flux systems. The sunspots merged and remained stable after type-B LBs formed. We conclude that type-B LBs are formed by collisions of flux systems, while type-A and type-C LBs are generated by splits. The time differences ( δ T ) between LBs appearing and ARs peaking have an average value of 1.06, −1.60, and 1.82 days for type-A, B, and C LBs, with the standard deviations of 3.27, 2.17, and 1.89, respectively. A positive value of δ T means that the LB appears after the AR peaks, whereas a negative δ T means it appears before the peak. Type-A LBs tend to form in the decaying phase or around the peak time. Type-B LBs are more likely to be formed in the developing phase. Type-C LBs mostly take shape in the decaying phase of ARs.

Since fast head-on coronal mass ejections and their associated shocks represent potential hazards to the space environment of the Earth and even other planets, forecasting the arrival time of the corresponding interplanetary shock is a priority in space weather research and prediction. Based on the radio spectrum observations of the 16-element array of the Daocheng Solar Radio Telescope (DSRT), the flagship instrument of the Meridian Project of China, during its construction, this study determines the initial shock speed of a type II solar radio burst on 2022 April 17 from its drifting speed in the spectrum. Assuming that the shock travels at a steady speed during the piston-driven phase (determined from the X-ray flux of the associated flare) and then propagates through interplanetary space as a blast wave, we estimate the propagation and arrival time of the corresponding shock at the orbit of the Solar Terrestrial Relations Observatory-A (STEREO-A). The prediction shows that the shock will reach STEREO-A at 14:31:57 UT on 2022 April 19. The STEREO-A satellite detected an interplanetary shock at 13:52:12 UT on the same day. The discrepancy between the predicted and observed arrival time of the shock is only 0.66 hr. The purpose of this paper is to establish a general method for predicting the shock’s propagation and arrival time from this example, which will be utilized to predict more events in the future based on the observations of ground-based solar radio spectrometers or telescopes like DSRT.

Abstract Background Drought stress has negative effects on plant growth and productivity. In this study, a comprehensive analysis of physiological responses and gene expression was performed. The responses and expressions were compared between drought-tolerant (DT) and drought-sensitive (DS) peanut varieties to investigate the regulatory mechanisms and hub genes involved in the impact of drought stress on culture. Results The drought-tolerant variety had robust antioxidative capacities with higher total antioxidant capacity and flavonoid contents, and it enhanced osmotic adjustment substance accumulation to adapt to drought conditions. KEGG analysis of differentially expressed genes demonstrated that photosynthesis was strongly affected by drought stress, especially in the drought-sensitive variety, which was consistent with the more severe suppression of photosynthesis. The hub genes in the key modules related to the drought response, including genes encoding protein kinase, E3 ubiquitin-protein ligase, potassium transporter, pentatricopeptide repeat-containing protein, and aspartic proteinase, were identified through a comprehensive combined analysis of genes and physiological traits using weighted gene co-expression network analysis. There were notably differentially expressed genes between the two varieties, suggesting the positive roles of these genes in peanut drought tolerance. Conclusion A comprehensive analysis of physiological traits and relevant genes was conducted on peanuts with different drought tolerances. The findings revealed diverse drought-response mechanisms and identified candidate genes for further research.

Background: Real-world data on use of PCSK9 inhibitors (PCSK9-Is), with or without statins and/or ezetimibe, and associated outcomes, can inform more effective prescribing. The objective was to evaluate clinical effectiveness and safety of PCSK9-Is within the Veterans Health Administration (VHA). Methods: In this retrospective cohort study, we included Veterans who had at least one outpatient prescription for alirocumab and/or evolocumab filled within VHA between August 21, 2015, and September 30, 2020. Analyses included 4 mutually exclusive subgroups: PCSK9-I alone, PCSK9-I+statin, PCSK9-I+ezetimibe, and PCSK9-I+statin+ezetimibe subgroups. Primary outcomes included medication possession ratio, persistence, and low-density lipoprotein (LDL). Results: Among Veterans in the analytical cohort (n = 2428), 36.2% were on PCSK9-I monotherapy; 24.0% received a PCSK9-I+statin; 27.4% were on a PCSK9-I+ezetimibe; and 12.4% received triple therapy, that is, PCSK9-I+statin+ezetimibe. The mean medication possession ratio (standard deviation [SD]) for PCSK9-I monotherapy was 83.8% (13.3) compared to 84.3% (11.2) with PCSK9-I+statin therapy, 87.1% (10.1) with PCSK9-I+ezetimibe therapy, and 85.8% (11.7) with triple therapy. The percentage of patients who discontinued PCSK9-I in the monotherapy subgroup was 12.3% vs 9.5%, 6.6%, and 7.4% in the concomitant statin, ezetimibe, and triple-therapy subgroups, respectively (p = .002 among the groups). Mean LDL level was greater in the PCSK9-I monotherapy subgroup (85.6 mg/dL) compared with the concomitant statin (66.5 mg/dL), ezetimibe (65.7 mg/dL), and triple-therapy subgroups (68.1 mg/dL). Conclusions: Veterans showed good adherence and/or persistence with PCSK9-I regimens. On average, those receiving concomitant therapy with a statin and/or ezetimibe achieved significantly lower LDL levels.

Abstract Background Intercropping, a diversified planting pattern, increases land use efficiency and farmland ecological diversity. We explored the changes in soil physicochemical properties, nutrient uptake and utilization, and microbial community composition in wide-strip intercropping of maize and peanut. Results The results from three treatments, sole maize, sole peanut and intercropping of maize and peanut, showed that intercropped maize had a marginal advantage and that the nutrient content of roots, stems and grains in side-row maize was better than that in the middle row of intercropped maize and sole maize. The yield of intercropped maize was higher than that of sole cropping. The interaction between crops significantly increased soil peroxidase activity, and significantly decreased protease and dehydrogenase activities in intercropped maize and intercropped peanut. The diversity and richness of bacteria and fungi decreased in intercropped maize rhizosphere soil, whereas the richness of fungi increased intercropped peanut. RB41, Candidatus-udaeobacter, Stropharia, Fusarium and Penicillium were positively correlated with soil peroxidase activity, and negatively correlated with soil protease and dehydrogenase activities. In addition, intercropping enriched the functional diversity of the bacterial community and reduced pathogenic fungi. Conclusion Intercropping changed the composition and diversity of the bacterial and fungal communities in rhizosphere soil, enriched beneficial microbes, increased the nitrogen content of intercropped maize and provided a scientific basis for promoting intercropping in northeastern China.

The predictions of plasma parameters in the interplanetary medium are the core of space weather forecasts, and the magnetohydrodynamics (MHD) numerical simulation is an important tool in the prediction of plasma parameters. Operational space weather forecasts are commonly produced by a heliosphere model whose inner boundary is set at 18 Rs or beyond. Such predictions typically use empirical/physics-based inner boundary conditions to solve the MHD equations for numerical simulation. In recent years, significant progress has been made in the numerical modeling of the inner heliosphere. In this paper, the numerical modeling of solar wind and coronal mass ejection in the inner heliosphere is reviewed. In particular, different inner boundary conditions used in the simulation are investigated since the MHD solutions are predetermined by the treatment of the inner boundary conditions to a large extent. Discussion is made on further development of the heliosphere model.

Although foxtail millet, as small Panicoid crop, is of drought resilient, drought stress has a significant effect on panicle of foxtail millet at the yield formation stage. In this study, the changes of panicle morphology, photosynthesis, antioxidant protective enzyme system, reactive oxygen species (ROS) system, and osmotic regulatory substance and RNA-seq of functional leaves under light drought stress (LD), heavy drought stress (HD), light drought control (LDCK) and heavy drought control (HDCK) were studied to get a snap-shot of specific panicle morphological changes, physiological responses and related molecular mechanisms. The results showed that the length and weight of panicle had decreased, but with increased empty abortive rate, and then yield dropped off 14.9% and 36.9%, respectively. The photosynthesis of millet was significantly decreased, like net photosynthesis rate, stomatal conductance and transpiration rate, especially under HD treatment with reluctant recovery from rehydration. Under LD and HD treatment, the peroxidase (POD) was increased by 34% and 14% and the same as H2O2 by 34.7% and 17.2% compared with LDCK and HDCK. The ability to produce and inhibit O2- free radicals under LD treatment was higher than HD. The content of soluble sugar was higher under LD treatment but the proline was higher under HD treatment. Through RNA-seq analysis, there were 2,393 and 3,078 different genes expressed under LD and HD treatment. According to the correlation analysis between weighted gene coexpression network analysis (WGCNA) and physiological traits, the co-expression network of several modules with high correlation was constructed, and some hub genes of millet in response to drought stress were found. The expression changes relating to carbon fixation, sucrose and starch synthesis, lignin synthesis, gibberellin synthesis, and proline synthesis of millet were specifically analyzed. These findings provide a full perspective on how drought affects the yield formation of foxtail millet by constructing one work model thereby providing theoretical foundation for hub genes exploration and drought resistance breeding of foxtail millet.

Abiotic stresses such as cold, drought and salinity are the key environmental factors that limit the yield and quality of oil crop peanut. Phospholipase Ds (PLDs) are crucial hydrolyzing enzymes involved in lipid mediated signaling and have valuable functions in plant growth, development and stress tolerance. Here, 22, 22 and 46 PLD genes were identified in Arachis duranensis, Arachis ipaensis and Arachis hypogaea, respectively, and divided into α, β, γ, δ, ε, ζ and φ isoforms. Phylogenetic relationships, structural domains and molecular evolution proved the conservation of PLDs between allotetraploid peanut and its diploid progenitors. Almost each A. hypogaea PLD except for AhPLDα6B had a corresponding homolog in A. duranensis and A. ipaensis genomes. The expansion of Arachis PLD gene families were mainly attributed to segmental and tandem duplications under strong purifying selection. Functionally, the most proteins interacting with AhPLDs were crucial components of lipid metabolic pathways, in which ahy-miR3510, ahy-miR3513-3p and ahy-miR3516 might be hub regulators. Furthermore, plenty of cis-regulatory elements involved in plant growth and development, hormones and stress responses were identified. The tissue-specific transcription profiling revealed the broad and unique expression patterns of AhPLDs in various developmental stages. The qRT-PCR analysis indicated that most AhPLDs could be induced by specific or multiple abiotic stresses. Especially, AhPLDα3A, AhPLDα5A, AhPLDβ1A, AhPLDβ2A and AhPLDδ4A were highly up-regulated under all three abiotic stresses, whereas AhPLDα9A was neither expressed in 22 peanut tissues nor induced by any abiotic stresses. This genome-wide study provides a systematic analysis of the Arachis PLD gene families and valuable information for further functional study of candidate AhPLDs in peanut growth and abiotic stress responses.

This study compared rates of empirical-therapy use and negative patient outcomes between complicated and recurrent urinary tract infection (r/cUTI) cases diagnosed with a multiplex polymerase chain reaction or pooled antibiotic susceptibility testing (M-PCR/P-AST) vs. standard urine culture (SUC). Subjects were 577 symptomatic adults (n = 207 males and n = 370 females) presenting to urology/urogynecology clinics between 03/30/2022 and 05/24/2023. Treatment and outcomes were recorded by the clinician and patient surveys. The M-PCR/P-AST (n = 252) and SUC (n = 146) arms were compared after patient matching for confounding factors. The chi-square and Fisher’s exact tests were used to analyze demographics and clinical outcomes between study arms. Reduced empirical-treatment use (28.7% vs. 66.7%), lower composite negative events (34.5% vs. 46.6%, p = 0.018), and fewer individual negative outcomes of UTI-related medical provider visits and UTI-related visits for hospitalization/an urgent care center/an emergency room (p < 0.05) were observed in the M-PCR/P-AST arm compared with the SUC arm. A reduction in UTI symptom recurrence in patients ≥ 60 years old was observed in the M-PCR/P-AST arm (p < 0.05). Study results indicate that use of the M-PCR/P-AST test reduces empirical antibiotic treatment and negative patient outcomes in r/cUTI cases.

Highlights 1. Gene transcript profiles in the elderly AML patients are different from those in young patients. 2. Statistical analysis reveals the down-regulation of protein processing in ER and pathways related to immunity in elderly patients. 3. GEO and GSEA analyses demonstrate the genes of cancer pathway and mitogen activated protein kinase (MAPK) pathway, involved in natural immunity and metabolism, are largely down-regulated in elderly patients. 4. ATF4 is a biomarker indicating different therapeutic strategies for elderly patients.

Based on historical temperature and salinity (T–S) profiles, the correlation between the sea surface temperature (SST) anomaly, sea surface dynamic height (SSH) anomaly, and temperature profile anomaly is constructed by regression analysis. A three-dimensional temperature field is reconstructed in the northern South China Sea by satellite SST and SSH, with daily temporal and 0.25°×0.25° spatial resolutions. The three-dimensional salinity field is also reconstructed based on the correlation between salinity and temperature. Compared with the observational T–S profiles, the reconstructed T–S field reflects the characteristics and structure and accurately describes the mesoscale variability of the ocean temperature field. The new expanded T–S field can be used as the initial field in numerical models and be assimilated into numerical reanalysis and prediction systems, improving their output.

Belowground interactions mediated by root exudates are critical for the productivity and efficiency of intercropping systems. Herein, we investigated the process of microbial community assembly in maize, peanuts, and shared rhizosphere soil as well as their regulatory mechanisms on root exudates under different planting patterns by combining metabolomic and metagenomic analyses. The results showed that the yield of intercropped maize increased significantly by 21.05% (2020) and 52.81% (2021), while the yield of intercropped peanut significantly decreased by 39.51% (2020) and 32.58% (2021). The nitrogen accumulation was significantly higher in the roots of the intercropped maize than in those of sole maize at 120 days after sowing, it increased by 129.16% (2020) and 151.93% (2021), respectively. The stems and leaves of intercropped peanut significantly decreased by 5.13 and 22.23% (2020) and 14.45 and 24.54% (2021), respectively. The root interaction had a significant effect on the content of ammonium nitrogen (NH4+-N) as well as the activities of urease (UE), nitrate reductase (NR), protease (Pro), and dehydrogenase (DHO) in the rhizosphere soil. A combined network analysis showed that the content of NH4+-N as well as the enzyme activities of UE, NR and Pro increased in the rhizosphere soil, resulting in cyanidin 3-sambubioside 5-glucoside and cyanidin 3-O-(6-Op-coumaroyl) glucoside-5-O-glucoside; shisonin were significantly up-regulated in the shared soil of intercropped maize and peanut, reshaped the bacterial community composition, and increased the relative abundance of Bradyrhizobium. These results indicate that interspecific root interactions improved the soil microenvironment, regulated the absorption and utilization of nitrogen nutrients, and provided a theoretical basis for high yield and sustainable development in the intercropping of maize and peanut.

Rotational strip intercropping (RSI) of cereals and legumes has been developed and widely carried out to alleviate continuous cropping obstacles, to control erosion and to improve field use efficiency. In this study, a four-year fixed-field experiment was carried out in northeast China with three treatments: continuous cropping of maize, continuous cropping of peanuts and rotational strip intercropping of maize and peanut. The results show that crop rotation improved the main-stem height, branch number, lateral branch length, and yield and quality of peanuts; the yield was the highest in 2018, when it was increased by 39.5%. RSI improved the contents of total N, available N, total P, available P, total K and available K; the content of available N was the highest in 2018, with an increase of 70%. Rhizosphere soil urease and catalase activities were significantly increased and were the highest in 2017, reaching 183.13% and 91.21%, respectively. According to a high-throughput sequencing analysis, the rhizosphere soil bacterial richness and specific OTUs decreased in peanut rhizosphere soil, while the fungal increased. There were differences in the bacterial and fungal community structures; specifically, the abundance of Acidobacteria and Planctomycetes increased among bacteria and the abundance of beneficial microorganisms such as Ascomycota increased among fungi. In conclusion, rotational strip intercropping of maize and peanut increased the yield and quality of peanuts and conducive to alleviating the obstacles facing the continuous cropping of peanuts. Among then, soil physicochemical properties, enzyme activity and microbial diversity were significantly affected the yield of peanut.

To analyze the rigid–flexible coupling effects on the dynamic performance of a robot system, a dynamic model of a parallel robot with flexible spatial links is derived in detail using a floating frame of reference (FFR) formulation. Compared to the previous rigid–flexible coupling model where the kinematic chains are all flexible links or where the joints are all flexible components, the inertia matrix and the stiffness matrix are not constant matrix which leading to the differences in respect of dynamic performance in model. To verify the correctness of the derived dynamics equations, the dynamics solutions of the spatial parallel robot from an ideal rigid–body model and the FFR model containing rigid and flexible coordinates were established by an FFR formulation. Furthermore, a finite element analysis (FEA) model, which included rigid links and flexible spatial links, was constructed for comparison. The comparison of the three models showed that the trajectory trends were the same, but the motion trajectories of the end-effector obtained by the FFR and FEA models varied within a certain range, and the maximum variations occurred at the peaks of the trajectories. However, since the FFR model considered the coupling effects of rigid and flexible links and the micro-displacement of the end-effector, the amount of deformation was the largest.

Using N2O4 donor symmetric ligand H2L and dca co-ligand, two new isostructural dinuclear CuII–LnIII complexes [Cu(Cl)(L)Ln(NO3)(CH3OH)(H2O)(dca)] [Ln=Ho (1CuHo), Gd (2CuGd)] [H2L = 6,6′-((1E,1′E)-(ethane-1,2-diylbis(azaneylylidene))bis(methaneylylidene))bis(2-methoxyphenol); dca=dicyanamide] were designed, synthesized and studied. In the two isostructural compounds, the geometric environment around the nine-coordinated Ln(III) ions is muffin, whereas the geometry of the penta-coordinated Cu(II) ions is square pyramid. The magnetic properties of both complexes were also studied. Direct current magnetic susceptibility measurements indicate ferromagnetic interactions between the Cu(II) ion and Gd(III) ion in complex 2CuGd. Alternating current (ac) magnetic measurements indicate that complex 1CuHo displays slow magnetic relaxation behaviour.

This paper presents a study of a 2021 October 9 coronal mass ejection (CME) with multipoint imaging and in situ observations. We also simulate this CME from the Sun to Earth with a passive tracer to tag the CME’s motion. The coronagraphic images show that the CME is observed as a full halo by SOHO and as a partial halo by STEREO-A. The heliospheric images reveal that the propagation speed of the CME approaches about 1° hr ^−1 , suggesting a slow CME. With simulated results matching these observation results, the simulation discloses that as the CME ejects from the Sun out to interplanetary space, its global morphology is distorted from an ellipsoid to a concave shape owing to interactions with the bimodal solar wind. The cross section of the CME’s flux rope structure transforms from a circular shape into a flat one. As a result of the deflection, the propagation direction of the CME is far away from the Sun–Earth line. This means that the CME flank (or the ICME leg) likely arrives at both Solar Orbiter and the L1 point. From the CME’s eruption to 1 au, its volume and mass increase by about two orders and one order of magnitude, respectively. Its kinetic energy is about 100 times larger than its magnetic energy at 1 au. These results have important implications for our understanding of CMEs’ morphology, as well as their space weather impacts.

Potassium is one of the most important elements for crop growth and development. However, potassium deficiencies are common in the cultivated land of oil crops in the world, which limits the increase in their yields. The photosynthesis, fluorescence, and physiological indexes of peanut plants were affected by low K stress to varying degrees, and finally the yield decreased. However, the effect of low K stress on the photosynthetic physiological mechanism of peanut plants remains unclear. In this study, in order to explore the response mechanism of peanuts to low K stress, NH18 (tolerance to low K) and HY20 (intolerance to low K) were used to explore the effects of different peanut varieties under low K stress on the dry matter accumulation, protective enzyme activities, osmotic regulatory substance accumulation capacity, fluorescence characteristics, nutrient content, grain quality, and yield. The results of the 2-year experiment showed that under the stress of low K, the content of malondialdehyde in peanut leaves increased and the activities of NH18 superoxide dismutase (SOD) and peroxidase (POD) in the low-K-tolerant variety were higher than those in the low-K-intolerant variety HY20. The decrease in osmotic regulation caused by low K was compensated for by an increase in the soluble protein content. As a result, the chlorophyll content decreased significantly, F0 increased under dark adaptation, and Fm, Fv, and Fv/Fm decreased. The photosynthetic and fluorescence physiology of low-K-tolerant NH18 was less affected by low K stress. Furthermore, under low K stress, the dry matter accumulation of NH18 was reduced less, so that the final yield was less affected by low K stress than that of HY20. Under low K stress, the potassium content in the roots, stems, leaves, and fruit needles decreased significantly, and the decreasing range of stems and leaves gradually increased with the growth period, while that of the pod gradually decreased with the growth period. Under low K stress, the sodium content in the root system significantly increased and was significantly higher than that in the stem, leaf, pod, and fruit needle, indicating that the peanut plants actively absorbed more Na+ to replace the K+ function. This study clarified the mechanism of photosynthesis and the physiology of peanut plants under low K stress, which is of great significance for the breeding and cultivation of peanut resistance.

With the development of measurement technology, the Flexible Measuring Arm (FMA) is widely used in quality test of automobile processing and industrial production. FMA is a kind of nonlinear system with many parameters. Low cost and efficient calibration method have become the focuses of attention. This article presents a fast calibration method for FMA based on an adaptive Genetic Algorithm (GA) just with several standard balls and a ball plate. It can greatly reduce the calibration cost than common external calibration method which needs high precision instruments and sensors. Firstly, the kinematic model of FMA is established by RPY theory. Secondly, the common GA is optimized and improved, and an adaptive mechanism is added to the algorithms which can realize the automatic adjustment of crossover and mutation operators. A Normalized Genetic Algorithm (NGA) with adaptive mechanism is proposed to complete the optimization calculation. It can improve the numbers of optimal individuals and the convergence speed. So, the search efficiency will be enhanced greatly. Finally, the Least square method (LSM), the General Genetic Algorithm (GGA), and the proposed NGA are respectively used to finish the calibration work. The compensation accuracy and the search efficiency with the above three different algorithms have been systematically analyzed. Experiment indicates that the performance of NGA is much better than LSM and GGA. The data also has proved that the LSM is suitable to complete optimization calculation for linear system. Its convergence stability is much poorer than NGA and GGA because of the ill-condition Jacobin matrix. GGA is easy to fall into local optimization because of the fixed operators. The proposed NGA obviously owns fast convergence speed, high accuracy and better stability than GGA. The position error is reduced from 3.17 to 0.5 mm after compensation with the proposed NGA. Its convergence rate is almost two time of GGA which applies constant genetic factors. The effectiveness and feasibility of proposed method are verified by experiment.

Abstract Background The objective of this study is to describe the use of targeted therapies for the treatment of advanced renal cell carcinoma (RCC) and overall survival (OS) among patients in clinical practice in the Veterans Health Administration (VHA). Methods A retrospective cohort of 286 patients from 24 VHA Medical Centers diagnosed with advanced clear cell RCC between Fiscal Year (FY) 2010 and FY2014 was followed through September 30, 2016. Among patients who received targeted therapy, we described the medications taken, duration of therapy, and overall survival. We also assessed the effect of the first therapy received on overall survival using Cox Proportional Hazards models. Results There were 66 patients who did not receive therapy for their advanced RCC. Of the 220 treated patients, the mean (sd) number of medications received was 1.9 (1.1). The medications most commonly used first were sunitinib (61.8%), pazopanib (17.3%), and temsirolimus (10.9%). The median duration of first‐line therapy was 86 days (interquartile range [IQR] 42, 210). Median total duration of therapy was 159 days (IQR 58, 397). 62.3% of patients had ≥ 1 dose of therapy held or reduced, mainly due to an adverse drug event (ADE). Median survival from the start of treatment to death was 1.08 years (IQR 0.80, 1.31). Finally, receipt of temsirolimus vs sunitinib (HR 1.95 [95%CI 1.09,3.47]) as the first targeted therapy was independently associated with an increased hazard of death. Conclusion Our analysis of targeted therapies for advanced RCC in VHA suggests duration of treatment is shorter in a real‐world setting than in clinical trials, and dose reductions and ADEs are more common.

Abstract Background and Aims To explore the inhibition mechanism of Saccharomyces boulardii (S. boulardii) on ulcerative colitis (UC) carcinogenesis. Methods C57BL/6 mice were treated with azoxymethane and dextran sulfate sodium (AOM/DSS) to develop a UC carcinogenesis model. The treatment group was lavaged with S. boulardii (5 × 107 CFU/d) for 12 weeks. The mice were sacrificed and the tumor load in the treatment group was compared with that of a control group. The levels of TNF-α and IL-6 in colon tissue were measured by enzyme-linked immunosorbent assays. The influence of S. boulardii on TNF-α and IL-6 regulation was also investigated using different colon cell lines. Differences in intestinal microbiota in both stool and intestinal mucosa samples were assessed using 16S rDNA sequencing. Results S. boulardii treatment reduced AOM/DSS-induced UC carcinogenesis in mice, as indicated by the reduced tumor load and reduced TNF-α and IL-6 levels in vivo, as well its effects on TNF-α and IL-6 activities in vitro. Significant changes in both fecal and mucosal microbiota were observed among the control, the AOM/DSS treated, and AOM/DSS plus S. boulardii treated groups. For fecal microbiota, the AOM/DSS treated group was lower in Lactobacillus, but higher in Oscillibacter and Lachnoclostridium than the control group. After intervention with S. boulardii, the percentage of Bacillus and Lactococcus increased, but Lachnoclostridium, Oscillibacter, Bacteroides, and Pseudomonas decreased. For the intestinal mucosal microbiota, the AOM/DSS treated group was lower in Bifidobacterium and Ruminococcaceae_UCG-014 and higher in Alloprevotella than the control group. After S. boulardii exposure, the percentage contributions of Lachnoclostridium and Lachnospiraceae_NK4A136 increased. Conclusions S. boulardii effectively reduced UC carcinogenesis in an AOM/DSS induced mice model. This positive result can likely be attributed to the reduction of TNF-α and IL-6 levels or the blockade of their function combined with alterations to the intestinal microbiota.

Abstract Background Appropriate plant architecture can improve the amount of cotton boll opening and allow increased planting density, thus increasing the level of cotton mechanical harvesting and cotton yields. The internodes of cotton fruiting branches are an important part of cotton plant architecture. Thus, studying the molecular mechanism of internode elongation in cotton fruiting branches is highly important. Results In this study, we selected internodes of cotton fruiting branches at three different stages from two cultivars whose internode lengths differed significantly. A total of 76,331 genes were detected by transcriptome sequencing. By KEGG pathway analysis, we found that DEGs were significantly enriched in the plant hormone signal transduction pathway. The transcriptional data and qRT-PCR results showed that members of the GH3 gene family, which are involved in auxin signal transduction, and CKX enzymes, which can reduce the level of CKs, were highly expressed in the cultivar XLZ77, which has relatively short internodes. Genes related to ethylene synthase (ACS), EIN2/3 and ERF in the ethylene signal transduction pathway and genes related to JAR1, COI1 and MYC2 in the JA signal transduction pathway were also highly expressed in XLZ77. Plant hormone determination results showed that the IAA and CK contents significantly decreased in cultivar XLZ77 compared with those in cultivar L28, while the ACC (the precursor of ethylene) and JA contents significantly increased. GO enrichment analysis revealed that the GO categories associated with promoting cell elongation, such as cell division, the cell cycle process and cell wall organization, were significantly enriched, and related genes were highly expressed in L28. However, genes related to the sphingolipid metabolic process and lignin biosynthetic process, whose expression can affect cell elongation, were highly expressed in XLZ77. In addition, 2067 TFs were differentially expressed. The WRKY, ERF and bHLH TF families were the top three largest families whose members were active in the two varieties, and the expression levels of most of the genes encoding these TFs were upregulated in XLZ77. Conclusions Auxin and CK are positive regulators of internode elongation in cotton branches. In contrast, ethylene and JA may act as negative regulators of internode elongation in cotton branches. Furthermore, the WRKY, ERF and bHLH TFs were identified as important inhibitors of internode elongation in cotton. In XLZ77(a short-internode variety), the mass synthesis of ethylene and amino acid conjugation of auxin led to the inhibition of plant cell elongation, while an increase in JA content and degradation of CKs led to a slow rate of cell division, which eventually resulted in a phenotype that presented relatively short internodes on the fruiting branches. The results of this study not only provide gene resources for the genetic improvement of cotton plant architecture but also lay a foundation for improved understanding of the molecular mechanism of the internode elongation of cotton branches.

Soil microorganisms play important roles in crop production and sustainable agricultural management. However, soil conditions and crop selection are key determining factors for soil microbial communities. This study investigated the effect of plant types and soil salinity on the microbial community of interspecific interaction zone (II) based on the sorghum/peanut intercropping system. Microbial community diversity and composition were determined through PacBio single molecule, real-time sequencing of 16S rDNA and internal transcribed spacer (ITS) genes. Results showed Proteobacteria, Bacteroidota, and Acidobacteriota to be the dominant bacterial phyla in IP, II, and IS, whereas Ascomycota, Basidiomycota, and Mucoromycota were the dominant fungal phyla. Under salt-treated soil conditions, the plants-specific response altered the composition of the microbial community (diversity and abundance). Additionally, the interspecific interactions were also helpful for maintaining the stability and ecological functions of microbial communities by restructuring the otherwise stable core microbiome. The phylogenetic structure of the bacterial community was greatly similar between IP and II while that of the fungal community was greatly similar between IP and IS; however, the phylogenetic distance between IP and IS increased remarkably upon salinity stress. Overall, salinity was a dominant factor shaping the microbial community structure, although plants could also shape the rhizosphere microenvironment by host specificity when subjected to environmental stresses. In particular, peanut still exerted a greater influence on the microbial community of the interaction zone than sorghum.
