Abstract Hypervirulent Klebsiella pneumoniae (HvKP) is an emerging bacterial pathogen causing invasive infection in immune-competent humans. The hypervirulence is strongly linked to the overproduction of hypermucoviscous capsule, but the underlying regulatory mechanisms of hypermucoviscosity (HMV) have been elusive, especially at the post-transcriptional level mediated by small noncoding RNAs (sRNAs). Using a recently developed RNA interactome profiling approach iRIL-seq, we interrogate the Hfq-associated sRNA regulatory network and establish an intracellular RNA-RNA interactome in HvKP. Our data reveal numerous interactions between sRNAs and HMV-related mRNAs, and identify a plethora of sRNAs that repress or promote HMV. One of the strongest HMV repressors is ArcZ, which is activated by the catabolite regulator CRP and targets many HMV-related genes including mlaA and fbp. We discover that MlaA and its function in phospholipid transport is crucial for capsule retention and HMV, inactivation of which abolishes Klebsiella virulence in mice. ArcZ overexpression drastically reduces bacterial burden in mice and reduces HMV in multiple hypervirulent and carbapenem-resistant clinical isolates, indicating ArcZ is a potent RNA inhibitor of bacterial pneumonia with therapeutic potential. Our work unravels a novel CRP-ArcZ-MlaA regulatory circuit of HMV and provides mechanistic insights into the posttranscriptional virulence control in a superbug of global concern.

IntroductionThe role of individual nutrients including vitamins and minerals in cognitive function gained increasing attention in recent years. With regard to the association between dietary minerals and cognitive function, the results of human studies are inconclusive. The objective of this study was to explore the association between mineral intake and cognitive function using the data from Shanghai Health and Nutrition Survey (SHNS) in 2018.MethodsIn total, 835 adults were included in a crosssectional study, and completed a three-day dietary record to estimate their average daily intake of minerals. Mini-Mental State Examination (MMSE) was used for the assessment of cognitive function, and logistic regression analyses were performed on participants to examine the association between dietary mineral intake and cognitive performance. The participants were divided into tertiles according to their mineral intake.ResultsParticipants in the second and third tertile of the dietary copper intake had lower rates of low MMSE scores compared to those in the lowest tertile. We found the adjusted OR and 95%CI values were 0.44 (0.21–0.89) and 0.40 (0.16–0.94), respectively. Participants in the second tertile of dietary magnesium intake showed a trend of lower rates of low MMSE score compared to those in the lowest tertile (p = 0.06). The adjusted OR and 95%CI values were 0.35 (0.16–0.72). No significant association were observed between any of the other minerals including iron, zinc, selenium and manganese and cognitive function.DiscussionOur findings suggest that dietary intake of copper and magnesium may have a protective effect on cognitive performance in elderly over 60 years old. To prevent cognitive decline, elderly should get recommended amounts of copper and magnesium from diet or supplements.

Abstract Direct in situ imaging of nucleic acids on biological samples is advantageous for rapid analysis without DNA extraction. However, traditional nucleic acid amplification in aqueous solutions tends to lose spatial information because of the high mobility of molecules. Similar to a cellular matrix, hydrogels with biomimetic 3D nanoconfined spaces can limit the free diffusion of nucleic acids, thereby allowing for ultrafast in situ enzymatic reactions. In this study, hydrogel‐based in situ space‐confined interfacial amplification (iSCIA) is developed for direct imaging of single nucleic acid and single pathogen on biological samples without formaldehyde fixation. With a polyethylene glycol hydrogel coating, nucleic acids on the sample are nanoconfined with restricted movement, while in situ amplification can be successfully performed. As a result, the nucleic acids are lighted‐up on the large‐scale surface in 20 min, with a detection limit as low as 1 copy/10 cm2. Multiplex imaging with a deep learning model is also established to automatically analyze multiple targets. Furthermore, the iSCIA imaging of pathogens on plant leaves and food is successfully used to monitor plant health and food safety. The proposed technique, a rapid and flexible system for in situ imaging, has great potential for food, environmental, and clinical applications.

Abstract Photoelectrocatalysis has attracted significant attention for water splitting and contaminant degradation. However, the lifetime of photoelectrocatalysis devices is hampered by the severe instability and photocorrosion of the photo‐active nanomaterial on the photoelectrode, which is a key limitation to realizing industrialization. Typically, the conventional protection strategy of photoelectrodes usually suffers from the trade‐off between the photoelectrocatalytic activity and stability. Inspired by biological cell membrane with water channels, here a highly permeable and ultrathin silica coating with ultrasmall straight nanochannels is in situ grown that stabilizes the photoelectrode. These ultrasmall channels boost photoelectrocatalysis by accelerating water transport and reducing the reaction energy within the confined nanochannels. Specifically, the ultrathin coating imparts significant mechanical and structural stability to the photo‐active nanomaterial, thereby preventing its detachment, dissolution, and crystal damage without compromising performance. As a result, the protected photoelectrode exhibits enhanced water splitting activity and excellent stability over 120 h, whereas the photocurrent of the unprotected photoelectrode degrades rapidly. Meanwhile, the coated photoelectrode also exhibits superior photoelectrocatalytic degradation efficiency (>97%), even after the 10th cycle. This strategy is facile and universal and can be extended to construct other stable and high‐performance electrodes for promoting photoelectrocatalysis in practical applications.

Abstract Transcription is crucial for the expression of genetic information and its efficient and accurate termination is required for all living organisms. Rho-dependent termination could rapidly terminate unwanted premature RNAs and play important roles in bacterial adaptation to changing environments. Although Rho has been discovered for about five decades, the regulation mechanisms of Rho-dependent termination are still not fully elucidated. Here we report that Rof is a conserved antiterminator and determine the cryogenic electron microscopy structure of Rho-Rof antitermination complex. Rof binds to the open-ring Rho hexamer and inhibits the initiation of Rho-dependent termination. Rof’s N-terminal α-helix undergoes conformational changes upon binding with Rho, and is key in facilitating Rof-Rho interactions. Rof binds to Rho’s primary binding site (PBS) and excludes Rho from binding with PBS ligand RNA at the initiation step. Further in vivo analyses in Salmonella Typhimurium show that Rof is required for virulence gene expression and host cell invasion, unveiling a physiological function of Rof and transcription termination in bacterial pathogenesis.

Abstract Recent development of new immune checkpoint inhibitors has been particularly successfully in cancer treatment, but still the majority patients fail to benefit. Converting resistant tumors to immunotherapy sensitive will provide a significant improvement in patient outcome. Here we identify Mi-2β as a key melanoma-intrinsic effector regulating the adaptive anti-tumor immune response. Studies in genetically engineered mouse melanoma models indicate that loss of Mi-2β rescues the immune response to immunotherapy in vivo. Mechanistically, ATAC-seq analysis shows that Mi-2β controls the accessibility of IFN-γ-stimulated genes (ISGs). Mi-2β binds to EZH2 and promotes K510 methylation of EZH2, subsequently activating the trimethylation of H3K27 to inhibit the transcription of ISGs. Finally, we develop an Mi-2β-targeted inhibitor, Z36-MP5, which reduces Mi-2β ATPase activity and reactivates ISG transcription. Consequently, Z36-MP5 induces a response to immune checkpoint inhibitors in otherwise resistant melanoma models. Our work provides a potential therapeutic strategy to convert immunotherapy resistant melanomas to sensitive ones.

In recent years, extensive research had centered on the application of 1-methylcyclopropene (1-MCP) in fruit preservation, with a predominant focus on climacteric fruits. However, the development of efficient sustained-release products remained insufficient, and the effects of 1-MCP on the quality of non-climacteric fruits such as strawberry in postharvest were not fully understood. This research investigated the creation of a 1-MCP hydrogel-based controlled release product for strawberry preservation. The hydrogel system used in the preservative was synthesized using hydroxyethyl acrylate (HEA) and acrylic acid (AA) as monomers, with polyethylene glycol diacrylate (PEGDA) serving as a cross-linking agent. 1-MCP sustained-release hydrogel, which was referred to as 1-MCP hydrogel, was prepared by wrapping 1-MCP powder in a porous water gel system in an aqueous solvent system and initiated by diphenyl phosphorus oxide (TPO). The 1-MCP hydrogel was applied to the preservation of strawberry stored at room temperature. The results showed that the 1-MCP hydrogel had excellent 1-MCP sustained release performance, and could release 1-MCP evenly and slowly. Its release rate was 7% of the 1-MCP powder group and 3% of the 1-MCP powder solution group, respectively, at 12 h, achieving a long-term effective release. Fruit preservation experiments showed that the 1-MCP hydrogel group maintained a lower weight loss rate and decay rate, which were 20% and 21% lower than the control group on day 6, respectively. In addition, 1-MCP hydrogel maintained better fruit color, higher hardness, soluble solids content (TSS), and titratable acid content (TA). On day 2, the a* value, b* value and L* value of the experimental group were 11%, 11%, and 6% higher than the control group, respectively. The hardness, TSS, and TA on day 6 were 21%, 15%, and 18% higher than the control group, respectively. To sum up, 1-MCP hydrogel has multiple functions, such as the sustained release of 1-MCP, improving fruit quality. It is also convenient to prepare and can be used in the postharvest preservation of strawberries and potentially other fruit.

‘Fengtang‘ plums soften quickly and lose flavor after harvest. This study comprehensively evaluated the effect of exogenous melatonin on the fruit quality of ‘Fengtang’ plums. According to our findings, exogenous melatonin prevented plum fruit from losing water, delayed the decline in firmness, and preserved a high TSS/TA level. Additionally, exogenous melatonin also enhanced the activity of antioxidant enzymes and increased the non-enzymatic antioxidants, thereby further increasing the antioxidant capacity of plum fruit. Notably, exogenous melatonin delayed the degradation of covalent soluble pectin (CSP), cellulose, and hemicellulose, as well as the rise in water-soluble pectin (WSP) concentration and the activity of cell wall degrading enzymes. Further investigation using atomic force microscopy (AFM) revealed that the chain-like structure of ionic-soluble pectin (ISP) and the self-assembly network structures of CSP were depolymerized, and melatonin treatment retarded the depolymerization of pectin structures. Our results showed that exogenous melatonin preserved the postharvest quality of plum fruits by controlling fruit softness and antioxidant capacity during storage.

Summary: Background: Some locally advanced (IIIA/IIIB) non-small cell lung cancers (NSCLCs) might have surgical options available. However, information regarding the effectiveness of neoadjuvant immunotherapy for potentially resectable IIIA/IIIB NSCLC is limited. The intent of this investigation was to offer a more favourable alternative to the standard approach of chemoradiotherapy (concurrent or sequential chemoradiotherapy) followed by immunotherapy for potentially resectable stage III NSCLC. Methods: This prospective, single-arm, phase 2 clinical trial (NCT04326153) enrolled treatment-naïve patients with ‘potentially resectable’ IIIA/IIIB NSCLC who were deemed unsuitable for complete (R0) resection upon initial diagnosis. The study period was between March 20, 2020, and August 20, 2021. Patients underwent neoadjuvant chemoimmunotherapy (sintilimab combined with nab-paclitaxel and carboplatin) for two to three cycles prior to surgical resection of the lung carcinoma and systematic nodal dissection within 30–45 days. The primary endpoint was the 2-year disease-free survival (DFS) rate, with secondary endpoints encompassing major pathological response (MPR) rate, pathological complete response (pCR) rate, overall survival, objective response rate (ORR), downstaging rate, and adverse events (AEs). Tumour immune cell infiltrates, identified via immunohistochemistry, were assessed as biomarkers at baseline and after surgery. Findings: Among 30 patients who received neoadjuvant chemoimmunotherapy, 20 underwent complete resection. The disease control rate was 96.7% (95% CI: 90.3%–99.99%), with an ORR of 55% (95% CI: 37.2%–72.8%) and a downstaging rate of 80% (95% CI: 65.7%–94.3%). In the subgroup of 20 patients who underwent surgery, the MPR rate was 65% (95% CI: 43.3%–82.9%), and the pCR rate was 40% (95% CI: 21.2%–46.3%). The 2-year DFS rate in the surgical group was 75% (95% CI 56%–94%). Notably, the MPR group demonstrated significantly prolonged DFS compared with the non-MPR group (p = 0.00024). A significant increase in pretreatment CD8 expression correlated with improved DFS (p = 0.00019). Three patients (10%) experienced grade 3 or higher immune-related AEs—one case of grade 3 elevated myocardial enzymes, one case of grade 3 interstitial pneumonia, and one case of grade 5 bronchopleural fistula. Interpretation: Neoadjuvant immunotherapy markedly enhanced the rate of pathological response and 2-year DFS in patients with potentially resectable IIIA/IIIB NSCLC. Overexpression of CD8 before treatment (H score≥3) may serve as a potential predictive biomarker for DFS. Consequently, the treatment landscape for potentially resectable IIIA/IIIB NSCLC could undergo changes. Funding: This study did not receive any financial support.

In this study, we sequenced two complete mitogenomes from Deroceras laeve and Ambigolimax valentianus. The mitogenome of Ambigolimax valentianus represented the first such data from the family Limacidae. The lengths of the mitogenomes of Deroceras laeve and Ambigolimax valentianus were 14,773 bp and 15,195 bp, respectively. The entire set of 37 mitochondrial genes were identified for both mitogenomes. Compared with the mitogenome of Achatina fulica, the trnP_trnA tRNA cluster was rearranged in both Deroceras laeve and Ambigolimax valentianus. The secondary structures of tRNA and rRNA genes for the two species were predicted. Phylogenetic analyses based on amino acid sequences supported (1) monophyly of Stylommatophora, (2) division of Stylommatophora into the ‘achatinoid’ clade (i.e., the suborder Achatinina) and the ‘non-achatinoid’ clade (i.e., the suborder Helicina), (3) placement of the Orthurethra in the ‘non-achatinoid’ clade, and (4) monophyly of each of the superfamilies Helicoidea, Urocoptoidea, Succineoidea, Arionoidea, Pupilloidea and Limacoidea. The exemplars of Helicidae, Philomycidae and Achatinellidae displayed many more mitochondrial gene rearrangements than other species of Stylommatophora.

Abstract Toxoplasma gondii (T. gondii) is an obligate intracellular parasite with a wide range of hosts, including humans and many warm-blooded animals. The parasite exists in two interconvertible forms, namely tachyzoites and bradyzoites in intermediate hosts that are responsible for acute and chronic infections respectively. Mature bradyzoites accumulate large amounts of amylopectin granules but their roles have not been fully characterized. In this study, the predicted key enzymes involved in amylopectin synthesis (UDP-sugar pyrophospharylase, USP) and degradation (alpha-glucan water dikinase, GWD) of ME49 strain were individually knocked out, and then bradyzoite-related phenotyping experiments in vitro and in vivo were performed to dissect their roles during parasite growth and development. Deletion of the usp or gwd gene in the type II strain ME49 reduced the replication rates of tachyzoites in vitro and parasite virulence in vivo, suggesting that amylopectin metabolism is important for optimal tachyzoite growth. Interestingly, the Δusp mutant grew slightly faster than the parental strain under stress conditions that induced bradyzoite transition, which was likely due to the decreased efficiency of bradyzoite formation of the Δusp mutant. Although the Δgwd mutant could convert to bradyzoite robustly in vitro, it was significantly impaired in establishing chronic infection in vivo. Both the Δusp and Δgwd mutants showed a dramatic reduction in the reactivation of chronic infection in an in vitro model. Together, these results suggest that USP and GWD, which are involved in amylopectin synthesis and degradation have important roles in tachyzoite growth, as well as in the formation and reactivation of bradyzoites in T. gondii.

The family Elateridae, known as click beetles, is a mega-diverse lineage of Coleoptera. Wireworms are the larval stage of click beetles, which are generalist herbivores and which are recognized as economically important pests of crops. To more effectively control and monitor wireworms, it is crucial to understand the genetics, taxonomy and phylogenetics of Elateridae. Here, we sequenced and characterized three complete mitochondrial genomes (mitogenomes) from the subfamily Elaterinae using a next-generation sequencing approach. In addition, we provided the annotated mitogenomes of the newly sequenced species, namely Parasilesis musculus (Candèze, 1873), Melanotus cribricollis Candèze, 1860 and Glyphonyx sp., and compared their arrangement with other closely related species. The secondary structures of tRNA genes and rRNA genes were predicted. Combined with the published mitogenomes of elaterid species, we reconstructed the phylogenetic framework for Elateridae under maximum likelihood and Bayesian inference methods using nucleotide and amino acid sequence datasets separately. The results from the Bayesian analysis based on the nucleotide dataset PCGRNA including all 37 mitochondrial genes were congruent with previous studies. Within the monophyletic Elateridae, two main clades were recovered. The first clade included Elaterinae and Melanotus. The second clade consisted of the remaining subfamilies. Physodactylinae and Cardiophorinae formed a sister group. Agrypninae was monophyletic. A subclade comprised Negastriinae and Dendrometrinae.

The effect of the increase of inclined length of fully mechanized mining face on roof breaking and stress evolution is complex, which leads to regional differences in the support characteristics of support in mining face. In order to deeply understand the interaction between support and surrounding rock in super-long working face, taking the 450 m super-long working face in Xiaobaodang Coal Mine as the engineering background, the mechanical model of elastic foundation rock beam is constructed, and the mathematical statistics and homogenization cycle analysis method are used to analyze the end-cycle resistance, working resistance distribution frequency and increasing resistance characteristics of support in working face. Clarify the characteristics of roof movement and support working resistance distribution in the inclined direction of working face with different face lengths, and explore the resistance increasing characteristics of support in the inclined zone of super-long working face. The results show that with the increase of the inclination length of the working face, the support of the coal body in the two roadways to the central roof is weakened, the peak area of the middle working face moves closer to the two roadways, and the distribution trend of working resistance of support in super-long working face is M-type three-peak distribution. When the support in the upper, middle and lower parts of the super-long working face shows a logarithmic resistance increase, the homogenization curve is still a logarithmic function, which shows that the resistance increases rapidly at first and then tends to smooth. The resistance increase of the support reflects that the roof of the working face gradually changes from rapid subsidence to slow subsidence during the pressure period. In the case of logarithmic-exponential drag increase, the resistance increase homogenization curve in the middle and upper region is a logarithmic function, and the middle and lower homogenization curve is a logarithmic-exponential compound function, which shows the characteristics that the resistance increases rapidly at first, then tends to smooth and finally increases rapidly. When the working cycle time of the support exceeds a certain value, the logarithmic-exponential compound resistance increasing support will rapidly increase the resistance, which is not conducive to the roof control. By optimizing the coal cutting speed of the working face and reducing the working cycle time of the support, the resistance increase of the support can be reduced, and the instantaneous resistance increase of the support in the state of high resistance can be avoided and the supporting effect of the support in the working face can be improved.

The study explored the effect of exogenous chlorine dioxide (ClO2) treatment on retaining freshness of post-harvest fresh ginger. The fresh tender ginger were treated with 80 mg/L ClO2 at room temperature for 10 min, and the rest were soaked in distilled water for 10 min as control, then stored at 4 ℃ for up to 28 days. The results showed that ClO2 treatment suppressed the respiration rate, reduced decay rate, delayed the decrease of soluble solids (TSS) and decreased the content of hydrogen peroxide (H2O2) and malondialdehyde (MDA) during the entire storage time. Compared with the control, ClO2 treatment significantly enhanced the enzymes activities of superoxide dismutase (SOD) and peroxidase (POD). The research indicated that ClO2 maintained the storage quality of fresh tender ginger by promoting the antioxidant capacity.

Sugars and organic acids are the main factors determining the flavor of citrus fruit. The WRKY transcription factor family plays a vital role in plant growth and development. However, there are still few studies about the regulation of citrus WRKY transcription factors (CsWRKYs) on sugars and organic acids in citrus fruit. In this work, a genome-wide analysis of CsWRKYs was carried out in the citrus genome, and a total of 81 CsWRKYs were identified, which contained conserved WRKY motifs. Cis-regulatory element analysis revealed that most of the CsWRKY promoters contained several kinds of hormone-responsive and abiotic-responsive cis-elements. Furthermore, gene expression analysis and fruit quality determination showed that multiple CsWRKYs were closely linked to fruit sugars and organic acids with the development of citrus fruit. Notably, transcriptome co-expression network analysis further indicated that three CsWRKYs, namely, CsWRKY3, CsWRKY47, and CsWRKY46, co-expressed with multiple genes involved in various pathways, such as Pyruvate metabolism and Citrate cycle. These CsWRKYs may participate in the metabolism of fruit sugars and organic acids by regulating carbohydrate metabolism genes in citrus fruit. These findings provide comprehensive knowledge of the CsWRKY family on the regulation of fruit quality.

Integration of sensors with engineering thermoplastics allows to track their health and surrounding stimuli. As one of vital backbones to construct sensor systems, copper (Cu) is highly conductive and cost-effective, yet tends to easily oxidize during and after processing. Herein, an in-situ integrated sensor system on engineering thermoplastics via hybrid laser direct writing is proposed, which primarily consists of laser-passivated functional Cu interconnects and laser-induced carbon-based sensors. Through a one-step photothermal treatment, the resulting functional Cu interconnects after reductive sintering and passivation are capable of resisting long-term oxidation failure at high temperatures (up to 170 °C) without additional encapsulations. Interfacing with signal processing units, such an all-in-one system is applied for long-term and real-time temperature monitoring. This integrated sensor system with facile laser manufacturing strategies holds potentials for health monitoring and fault diagnosis of advanced equipment such as aircrafts, automobiles, high-speed trains, and medical devices.

The rapid development of the electric vehicle industry produces large amounts of retired power lithium-ion batteries, thus resulting in the echelon utilization technology of such retired batteries becoming a research hotspot in the field of renewable energy. The relationship between the cycle times and capacity decline of retired batteries performs as a fundamental guideline to determine the echelon utilization. The cycle conditions can influence the characteristics of the degradation of battery capacity; especially neglection of the SOC ranges of batteries leads to a large error in estimating the capacity degradation. Practically, the limited cycle test data of the SOC ranges of the retired battery cannot support a model to comprehensively describe the characteristics of the capacity decline. In this background, based on the limited cycle test data of SOC ranges, this paper studies and establishes a capacity degradation model of retired batteries that considers the factors affecting the battery cycle more comprehensively. In detail, based on the data-driven method and combined with the empirical model of retired battery capacity degradation, three semi-empirical modeling methods of retired battery capacity degradation based on limited test data of SOC ranges are proposed. The feasibility and accuracy of these methods are verified through the experimental data of retired battery cycling, and the conclusions are drawn to illustrate their respective scenarios of applicability.

Convenient, and sensitive detection of pesticides and their metabolites in environmental or food samples is critical for assessing potential environmental and health risks. Here, a three-dimensional (3D) electrochemical sensing platform is proposed based on the integration of nanochannel array on pre-activated 3D graphene (p-3DG) electrodes with no need of additional adhesive layers, which enables sensitive detection of prochloraz and 2,4,6-trichlorophenol (TCP) in environmental and food samples. Through two-step electrochemical polarization, organic phase anodic oxidation, and aqueous phase cathodic reduction, p-3DG electrodes with high active area and excellent electrocatalytic performance were obtained. Vertically-ordered mesoporous silica-nanochannel film (VMSF) can be rapidly grown on the surface of p-3DG by an electrochemical-assisted self-assembly (EASA) method. Taking advantage of the high electrocatalytic activity of p-3DG and the ability of nanochannels to enrich TCP through hydrogen bonding, the VMSF/p-3DG sensor can sensitively detect TCP in the range of 10 nM to 0.1 μM and 0.1–15 μM with a low limit of detection (LOD) of 2.4 nM. Compared with p-3DG and VMSF-modified 2D electrodes, the fabricated sensor has a wide detection linear range and low LOD. The coexistence of model interferents such as protein, surfactant, and humic acid did not affect the electrochemical response of TCP, confirming the high anti-fouling ability of the VMSF/p-3DG sensor. In addition, prochloraz in vegetable and fruit samples was indirectly determined because TCP was the metabolite of prochloraz.

Foodborne pathogenic microorganisms have become major threats that endanger human life and health. The current technology cannot perform rapid screening of foodborne pathogenic bacteria, and fail to timely control food safety risks. Here, we develop a novel microfluidic sensor for real-time and label-free bacteria classification at the single-cell level. Concretely, a low-aspect-ratio SiN micropore with PDMS coating was fabricated, which could significantly reduce the noise of the sensing system, and makes the microfluidic pore sensor sensitive to bacteria discrimination. The prepared SiN micropore equipped with the high temporal-spatial resolution was applied to observe bacterial translocation “events” and the current pulse signals could be obtained, which depend on the size, charge, and morphology of the target bacteria. According to the variation of the current pulse signals produced by different bacteria across the micropore, three common foodborne pathogens such as Salmonella enteric, Listeria monocytogenes, and Escherichia coli were identified. Due to convenience, rapidity, and accuracy, the label-free method we report here has great potential for the identification of diverse foodborne microorganisms at single-cell sensitivity.

Herein we report a highly specific electrochemical aptasenseor for AFB1 determination based on AFB1-controlled diffusion of redox probe (Ru(NH3)63+) through nanochannels of AFB1-specific aptamer functionalized VMSF. A high density of silanol groups on the inner surface confers VMSF with cationic permselectivity, enabling electrostatic preconcentration of Ru(NH3)63+ and producing amplified electrochemical signals. Upon the addition of AFB1, the specific interaction between the aptamer and AFB1 occurs and generates steric hindrance effect on the access of Ru(NH3)63+, finally resulting in the reduced electrochemical responses and allowing the quantitative determination of AFB1. The proposed electrochemical aptasensor shows excellent detection performance in the range of 3 pg/mL to 3 μg/mL with a low detection limit of 2.3 pg/mL for AFB1 detection. Practical analysis of AFB1 in peanut and corn samples is also accomplished with satisfactory results by our fabricated electrochemical aptasensor.

Sterol regulatory element binding protein-1 (SREBP-1), a transcription factor with a basic helix–loop–helix leucine zipper, has two isoforms, SREBP-1a and SREBP-1c, derived from the same gene for regulating the genes of lipogenesis, including acetyl-CoA carboxylase, fatty acid synthase, and stearoyl-CoA desaturase. Importantly, SREBP-1 participates in metabolic reprogramming of various cancers and has been a biomarker for the prognosis or drug efficacy for the patients with cancer. In this review, we first introduced the structure, activation, and key upstream signaling pathway of SREBP-1. Then, the potential targets and molecular mechanisms of SREBP-1-regulated lipogenesis in various types of cancer, such as colorectal, prostate, breast, and hepatocellular cancer, were summarized. We also discussed potential therapies targeting the SREBP-1-regulated pathway by small molecules, natural products, or the extracts of herbs against tumor progression. This review could provide new insights in understanding advanced findings about SREBP-1-mediated lipogenesis in cancer and its potential as a target for cancer therapeutics.

In this study, we newly sequenced the complete mitochondrial genomes (mitogenomes) of two phytophagous scarab beetles, and investigated the deep level relationships within Scarabaeidae combined with other published beetle mitogenome sequences. The complete mitogenomes of Dicronocephalus adamsi Pascoe (Cetoniinae) and Amphimallon sp. (Melolonthinae) are 15,563 bp and 17,433 bp in size, respectively. Both mitogenomes have the typical set of 37 genes (13 protein-coding genes, 22 transfer RNA genes, two ribosomal RNA genes) and an A+T-rich region, with the same gene arrangement found in the majority of beetles. The secondary structures for ribosomal RNA genes (rrnL and rrnS) were inferred by comparative analysis method. Results from phylogenetic analyses provide support for major lineages and current classification of Scarabaeidae. Amino acid data recovered Scarabaeidae as monophyletic. The Scarabaeidae was split into two clades. One clade contained the subfamilies Scarabaeinae and Aphodiinae. The other major clade contained the subfamilies Dynastinae, Rutelinae, Cetoniinae, Melolonthinae and Sericini. The monophyly of Scarabaeinae, Aphodiinae, Dynastinae, Cetoniinae and Sericini were strongly supported. The Scarabaeinae was the sister group of Aphodiinae. The Cetoniinae was sister to the Dynastinae + Rutelinae clade. The Melolonthinae was a non-monophyletic group. The removal of fast-evolving sites from nucleotide dataset using a pattern sorting method (OV-sorting) supported the family Scarabaeidae as a monophyletic group. At the tribe level, the Onthophagini was non-monophyletic with respect to Oniticellini. Ateuchini was sister to a large clade comprising the tribes Onthophagini, Oniticellini and Onitini. Eurysternini was a sister group of the Phanaeini + Ateuchini clade.

The diversity and high adaptability of insects are heavily associated with their symbiotic microbes, which include bacteria, fungi, viruses, protozoa, and archaea. These microbes play important roles in many aspects of the biology and physiology of insects, such as helping the host insects with food digestion, nutrition absorption, strengthening immunity and confronting plant defenses. To maintain normal development and population reproduction, herbivorous insects have developed strategies to detoxify the substances to which they may be exposed in the living habitat, such as the detoxifying enzymes carboxylesterase, glutathione-S-transferases (GSTs), and cytochrome P450 monooxygenases (CYP450s). Additionally, insect symbiotic bacteria can act as an important factor to modulate the adaptability of insects to the exposed detrimental substances. This review summarizes the current research progress on the role of insect symbiotic bacteria in metabolizing phytochemicals and agrochemicals (insecticides and herbicides). Given the importance of insect microbiota, more functional symbiotic bacteria that modulate the adaptability of insects to the detrimental substances to which they are exposed should be identified, and the underlying mechanisms should also be further studied, facilitating the development of microbial-resource-based pest control approaches or protective methods for beneficial insects.

Some innovative works have been done to probabilistic power flow (PPF) analysis for hybrid HVAC and LCC-VSC HVDC system in this paper. Firstly, a unified method considering precise model of converters is proposed to solve a general deterministic power flow (DPF) calculation including hybrid LCC (Line Current Converter) and VSC (Voltage Source Converter), the pure VSC-MTDC (Voltage Source Converter-Multiple Terminal Direct Current) and pure LCC system. Meanwhile, with a large amount of renewable energy sources integrated to the main grid through DC grids, it will impose a stochastic impact on the secure operation of such hybrid AC/DC grids. Therefore, it becomes necessary to model the probabilistic uncertainties and analyze their effects on the operation of hybrid AC/DC systems under different control modes. Nevertheless, most power flow analysis methods for hybrid AC/DC system are still deterministic in nature. Therefore, a probabilistic method based on the combination of Nataf transformation and Latin hypercube sampling (LHS) is developed and proposed to solve this complex PPF problem in an efficient manner, which considers correlated various probabilistic uncertainties, e.g. wind speeds, solar radiations and loads following different types of probability distribution. Finally, the effectiveness of the unified DPF method is validated in a modified IEEE 14-bus system, while the proposed PPF is verified in a modified IEEE 118-bus system and the effects of uncertainties on the diverse operation modes of hybrid AC/DC grids are discussed as well.

In this article, the finite element model considering the casing eccentricity of high-pressure, high-temperature wellbore integrity is established, where the cohesive zone element is introduced to evaluate the mechanical behavior of the casing–cement sheath interface. In this analysis, the bilinear traction–separation law is used to estimate the interfacial failure, and a damage factor is simultaneously defined to describe the damage evolution of the cohesive element. In addition, to achieve the interfacial separation, the pressure fluctuation inside the casing is applied. Using the finite element approach, we investigate the effect of mechanical parameters including Young’s modulus, Poisson’s ratio, thermal conductivity of the cement sheath, and pressure fluctuation inside the casing on the interfacial failure of the casing–cement sheath. The simulation results show that the damage factor increases with the increase in the mechanical parameters including Young’s modulus and Poisson’s ratio of the cement sheath and the casing pressure fluctuation. We also observe that the maximum damage factor under higher thermal conductivity is higher than that under lower thermal conductivity for the casing concentricity, while it is lower than that under lower thermal conductivity for the casing eccentricity. This indicates that the casing eccentricity has an important impact on the damage factor. What’s more, the casing eccentricity can change the location where the maximum damage factor occurs.

Modified atmosphere packaging (MAP) has been widely known to delay the postharvest fruit senescence; nevertheless, its effect on antioxidant activity and cell wall metabolism of wolfberry fruit is largely unknown. The present study investigated the impact of elevated CO2 on the quality attributes and cell wall degradation of wolfberry fruit during storage. The results showed that 10% CO2 better maintained the physiological quality and conferred the reduction in weight loss, decay index, and color change. Higher 2,2′-azino-bis (3-ethylbenzothiazoline-6-sulfonic acid) (ABTS) and 1,1-diphenyl-1-picrylhydrazil (DPPH) radical scavenging activity, total phenol and flavonoid content, and superoxide dismutase (SOD) and catalase (CAT) activity of wolfberry were detected at elevated CO2 concentrations. Elevated CO2 atmosphere contributed to the maintenance of the cell integrity, the decrease of cell wall degradation (polygalacturonase, pectate lyase, cellulase, and β-glucosidase), and the increase of cellulose and proto pectin content. Overall, we revealed the potential mechanism of elevated CO2 on the antioxidant activity enhancement and cell wall homeostasis of fresh berry fruit.

An InGaN laser diode with InGaN–GaN–InGaN delta barriers was designed and investigated numerically. The laser power–current–voltage performance curves, carrier concentrations, current distributions, energy band structures, and non-radiative and stimulated recombination rates in the quantum wells were characterized. The simulations indicate that an InGaN laser diode with InGaN–GaN–InGaN delta barriers has a lower turn-on current, a higher laser power, and a higher slope efficiency than those with InGaN or conventional GaN barriers. These improvements originate from modified energy bands of the laser diodes with InGaN–GaN–InGaN delta barriers, which can suppress electron leakage out of, and enhance hole injection into, the active region.

This paper proposes a new probabilistic power flow method for the hybrid AC/VSC-MTDC (Voltage Source Control-Multiple Terminal Direct Current) grids, which is based on the combination of ninth-order polynomial normal transformation (NPNT) and inherited Latin hypercube sampling (ILHS) techniques. NPNT is utilized to directly handle historical records of uncertain sources to build the accurate probability model of random inputs, and ILHS is adopted to propagate the randomness from inputs to target outputs. Regardless of whether the underlying probability distribution is known or unknown, the proposed method has the ability to adaptively evaluate the sample size according to a specific operational scenario of the power systems, thus achieving a good balance between computational accuracy and speed. Meanwhile, the frequency histograms, probability distributions, and some more statistics of the results can be accurately and efficiently estimated as well. The modified IEEE 118-bus system, together with the realistic data of wind speeds and diverse consumer behaviors following irregular distributions, is used to demonstrate the effectiveness and superiority of the proposed method.
