In order to investigate how the addition of two common ingredients in chocolate, sugar and milk powder, affects the mechanical properties of solid chocolate, uniaxial compression tests and wedge fracture tests were carried out using four ratios of chocolate as the experimental material. Mechanical properties such as Young's modulus and fracture toughness were directly correlated with textural properties such as hardness, elasticity, and brittleness. The results show that adding sugar increases Young's modulus and fracture toughness of chocolate, while milk powder is the opposite. In equal amounts of both, sugar played a more substantial role. In combination with the properties exhibited by chocolate in the above tests, data from creep tests were collected to improve the classical Bingham model and develop a new constitutive model for predicting the mechanical behaviour of solid chocolate with different ratios of added sugar and milk powder. The new four-component constitutive model allows for a more accurate fit to the creep test data and this work provides some suggestions for making different tasting chocolates by adjusting the addition of ingredients.

Mid-infrared (mid-IR) variability in young stellar objects (YSOs) is driven by several physical mechanisms, which produce a variety of amplitudes and light curve shapes. One of these mechanisms, variable disk accretion is predicted by models of episodic accretion to drive secular variability, including in the mid-IR. Because the largest accretion bursts are rare, adding new objects to the YSO eruptive variable class aids our understanding of the episodic accretion phenomenon and its possible impact on stellar and planetary formation. A previous analysis of 6.5 yr of NeoWISE light curves (3-5 $μ$m) of ~7000 nearby YSOs found an increase in the fraction of variability and variability amplitude for objects at younger stages of evolution. To help interpret these light curves, we have obtained low- and high-resolution near-IR spectra of 78 objects from this sample of YSOs. Background Pituitary stalk interruption syndrome (PSIS) in female patients is mainly characterized by short stature, primary amenorrhea, absent or incomplete sexual maturation, and infertility. Successful pregnancies among these patients are still extremely rare. This study was designed to describe a successful pregnancy and delivery in a Chinese female with PSIS following in vitro fertilization and embryo transfer. Methods Contrast-enhanced magnetic resonance imaging (MRI) scanning of the hypothalamus-pituitary region and genetic analysis of PSIS-associated genes was performed via whole-exome sequencing to identify the potential genetic causes of this disorder. We additionally explored the feasibility to overcome the infertility by controlled ovarian hyperstimulation and frozen-thawed embryo transfer under multiple pituitary hormone supplementation. Results We found that the 28-year-old Chinese woman with PSIS exhibited characteristic symptoms including multiple pituitary hormone deficiency, typical triad signs in MRI scanning, undetectable serum gonadotropins and estradiol levels, and invisible antral follicles in both ovaries. While no pathogenic/possible pathogenic variants that could or partly explain the typical clinical phenotype of PSIS were found following whole-exome sequencing. At the first attempted controlled ovarian hyperstimulation cycle, 14 oocytes were retrieved, and 6 embryos were acquired. Artificial endometrial preparation and frozen-thawed embryo transfer were performed one month after oocyte retrieval, and one day-5 blastocyst was transferred, resulting in a clinical pregnancy. Under close monitoring during pregnancy and multiple hormones dosage modulation, she delivered a healthy boy by elective cesarean section and the newborn developed normally under 1-year follow-up. Conclusions This is the first report of a successful pregnancy achieved in a woman with PSIS following in vitro fertilization and frozen-thawed embryo transfer. Under continuous hormonal supplementation and pregnancy monitoring, in vitro fertilization and embryo transfer might serve as a safe and effective treatment for infertility among PSIS women.

Among all present quantum key distribution protocols, the reference-frame-independent measurement-device-independent quantum key distribution (RFI-MDI-QKD) is standing out due to its high security and practicability, since it can not only remove all detector-side-channel attacks, but also relax the requirement of reference-frame calibrations. However, till today, the performance degradation of RFI-MDI-QKD caused by finite-size effects is one of the biggest impediment to its practical implementations. Here, we propose an enhanced RFI-MDI-QKD protocol which is highly resistant to the finite-size effect and immensely boosts both the key rate and transmission distance. As a result, it can even surpass the most advanced MDI-QKD protocol under larger rotation angles of reference frames. Furthermore, with the state-of-the-art RFI-MDI-QKD setup, we successfully carry out a proof-of-principle experimental demonstration at a record distance of 300km. The experiments under non-ideal condition verify the advantage and robustness of our present work.

Measurement-device-independent quantum key distribution can remove all possible detector side channels, and is robust against state preparation flaws when further combined with the loss-tolerant method. However, the secure key rate in this scenario is relatively low, thus hindering its practical application. Here, we first present a four-intensity decoy-state protocol where the signal intensity is modulated only in Z basis for key generation while the decoy intensities are modulated in both Z and X bases for parameter estimation. Moreover, we adopt collective constraint and joint-study strategy in statistical fluctuation analysis. We have also experimentally demonstrated this protocol and the result indicates high performance and good security for practical applications.

Quantum key distribution (QKD) in principle can provide unconditional secure communication between distant parts. However, when finite-key length is taken into account, the security can only be ensured within certain security level. In this paper, we adopt the Chernoff bound analysis method to deal with finite-key-size effects, carrying out corresponding investigations on the relationship between the key generation rate and security parameters for different protocols, including BB84, measurement-device-independent and twin-field QKD protocols. Simulation results show that there exists a fundamental limit between the key rate and the security parameters. Therefore, this study can provide valuable references for practical application of QKD, getting a nice balance between the key generation rate and the security level.

In the present study, we focused on characterizing the proteome in granulosa cells in patients with biochemical premature ovarian insufficiency (bPOI) in order to identify differential proteins and investigate the fundamental mechanisms of POI. A total of 2688 proteins were identified based on the data-independent acquisition method, and 70 differentially expressed proteins were significant. Bioinformatic analyses, including gene expression pattern analysis, gene ontology enrichment analysis, Kyoto Encyclopedia of Genes and Genomes pathway analysis, and Search Tool for the Retrieval of Interacting Genes/Proteins analysis, revealed discrete modules and the underlying molecular mechanisms in bPOI. Importantly, we observed that Ras-related C3 botulinum toxin substrate 1 (RAC1) was downregulated in the granulosa cells of bPOI. Low expression of RAC1 may affect the development process of POI by affecting the proliferation, apoptosis, and hormone synthesis of granulosa cells. Downregulation of RAC1 expression in the KGN and COV434 cells inhibited cell proliferation, blocked cells in the G1/G0 phase, and promoted apoptosis. Western blot results showed that β-catenin and cyclin D1 in the KGN and COV434 cells transfected with RAC1-siRNA were downregulated, while P21 and Bax were upregulated. In most of the realistic measurement device-independent quantum key distribution (MDI-QKD) systems, efficient, real-time feedback controls are required to maintain system stability when facing disturbance from either external environment or imperfect internal components. Traditionally, people either use a “scanning-and-transmitting” program or insert an extra device to make a phase reference frame calibration for a stable high-visibility interference, resulting in higher system complexity and lower transmission efficiency. In this work, we build a machine learning-assisted MDI-QKD system, where a machine learning model—the long short-term memory (LSTM) network—is for the first time to apply onto the MDI-QKD system for reference frame calibrations. In this machine learning-assisted MDI-QKD system, one can predict out the phase drift between the two users in advance, and actively perform real-time phase compensations, dramatically increasing the key transmission efficiency. Furthermore, we carry out corresponding experimental demonstration over 100 km and 250 km commercial standard single-mode fibers, verifying the effectiveness of the approach.

Premature ovarian insufficiency (POI) is a highly heterogeneous ovarian disorder. Genetic factors account for the cause of POI. We aimed to analyze the genetic alterations in two affected sisters diagnosed with POI and their parents from a highly consanguineous Chinese Han family. Whole-exome sequencing was performed, and bioinformatics analysis was used to determine the potential genetic cause of POI in this family. A SYCE1 deletion was verified by Sanger sequencing. A homozygous deletion in SYCE1 was harbored by the proband and her affected sister, whereas both parents had heterozygous deletions. There were distinct differences in the amino acid sequences between wild-type and SYCE1 deletion. Domain analysis and 3D structural analysis of the SYCE1 deletion was also performed to evaluate the potential impact and pathogenicity of POI. The SYCE1 domain structure was truncated. Additionally, the 3D structure showed that the SYCE1 deletion changed the shape of the protein compared with that of wild-type SYCE1. This study revealed a novel SYCE1 deletion. This SYCE1 deletion may be the cause of POI. Genetic counseling for POI is helpful for researchers and clinicians to identify the mode of genetic inheritance for SYCE1 deletion in POI pathology.

This study aims to obtain satisfactory short-term traffic flow forecasting results in various traffic flow modes. Clustering analysis of traffic flow data is carried out using the Kohonen neural network. Then, different BP neural networks are trained using different classified traffic flow data. Different neural network models are combined with UKF to form multiple traffic estimators to realize the estimation function. Finally, the interactive method is used to fuse the prediction results of each estimator, and comprehensive traffic flow forecasting results are obtained. With the simulation example, a number of single estimators are obtained, and a joint estimator based on this method is constructed. The section flow of one given road is predicted to verify the performance of the estimator. Results show that the joint estimator has a higher prediction accuracy than the single estimator and has adaptive characteristics when traffic flow characteristics change.

This observational study included 21 patients at remarkably high risk of ovarian hyperstimulation syndrome (OHSS), characterized by more than 30 follicles measuring ≥11 mm in diameter on trigger day and/or pre-trigger peak estradiol exceeding 10 000 pg/mL, which was also the feature of women with established severe early OHSS followed by gonadotrophin-releasing hormone agonist (GnRHa) trigger and freeze-all policy that previously have been reported. All patients received a second dose of GnRHa 12 h after the first GnRHa trigger combined with administration of GnRH antagonist at 0.25 mg/day for a period of 3 days from the day of oocyte retrieval onwards. The in vitro fertilization (IVF) outcomes may be preferable compared with a bolus of GnRHa trigger and none of the included patients developed moderate-to-severe OHSS. Moreover, patients' symptoms, reproductive hormone levels and ultrasound findings were improved significantly. This new strategy seems to be efficacious and could be a further supplement of GnRHa trigger with or without applying freeze-all strategy to completely prevent early-onset moderate to severe OHSS, especially for the patients characterized by ≤30 follicles measuring ≥11 mm in diameter on trigger day and/or pre-trigger peak estradiol exceeding 10 000 pg/mL. Further studies should be performed to compare this regimen with conventional methods of OHSS prevention.

Identifying the young optically visible population in a star-forming region is essential for fully understanding the star formation event. In this paper, We identify 211 candidate members of the Perseus molecular cloud based on Gaia astronomy. We use LAMOST spectra to confirm that 51 of these candidates are new members, bringing the total census of known members to 856. The newly confirmed members are less extincted than previously known members. Two new stellar aggregates are identified in our updated census. With the updated member list, we obtain a statistically significant distance gradient of $\rm 4.84\;pc\;deg^{-1}$ from west to east. Distances and extinction corrected color-magnitude diagrams indicate that NGC 1333 is significantly younger than IC 348 and the remaining cloud regions. The disk fraction in NGC 1333 is higher than elsewhere, consistent with its youngest age. Measurement-device-independent quantum key distribution (MDI-QKD) can eliminate all detector side-channel loopholes and has shown excellent performance in long-distance secret keys sharing. Conventional security proofs, however, require additional assumptions on sources and that can be compromised through uncharacterized side channels in practice. Here, we present a general formalism based on reference technique to prove the security of MDI-QKD against any possible sources imperfection and/or side channels. With this formalism, we investigate the asymptotic performance of single-photon sources without any extra assumptions on the state preparations. Reference-frame-independent (RFI) measurement-device-independent (MDI) quantum key distribution (QKD) is immune to all detector side-channel attacks and is insensitive to slow drifts of reference frames; thus, it is a promising candidate for the realization of long-distance quantum secure communications. However, its practical performance is predominantly limited by the finite-data-size effect compared with standard MDI QKD. By combining the collective constraint and joint-study strategy with our parameter estimation processes for the RFI-MDI-QKD protocol, we reduce the influence of the finite-data-size effect and substantially improve the protocol's performance under realistic experimental conditions. Furthermore, a proof-of-principle experimental demonstration is conducted, and a transmission distance of up to 200 km is achieved. Therefore, this study represents a further step toward long-distance quantum secure communications.

Polycystic ovary syndrome (PCOS) is a common endocrine and metabolic disorder in reproductive-aged women, and its pathogenesis is still under debate. Recent studies suggest crucial roles for microRNAs (miRNAs) in PCOS development. The let-7 family miRNAs constitute the most abundant miRNAs in human granulosa cells (GCs), and plays an important role in follicular development. However, research on the let-7e implications of the non-hyperandrogenic (non-HA) phenotype remains unclear. This study aimed at determining the role of let-7e in the progression of PCOS. We performed quantitative real-time PCR to examine the levels of let-7e in fifty-two non-HA PCOS patients and fifty-two controls. A receiver operating characteristic (ROC) curve were used to reveal the diagnostic value of let-7e in non-HA PCOS. Using an immortalized human granulosa cell line, KGN, we investigated the influence of let-7e on cell proliferation and autophagy. Our data substantiated the expression of let-7e was significantly increased in non-HA PCOS group, and associated with an increased antral follicle count. The ROC curve indicated a major separation between non-HA PCOS group and the control group. Let-7e knockdown suppressed cell proliferation and enhanced cell autophagy by activating p21 pathway. Conversely, let-7e overexpression promoted cell proliferation and inhibited cell autophagy by suppressing p21 pathway. Our results indicate that increased let-7e levels in non-HA PCOS GCs may contribute to excessive follicular activation and growth, thereby involving in the pathogenesis of PCOS. Let-7e may thus be a potential therapeutic target in non-HA PCOS.

Reference-frame-independent measurement-device-independent quantum key distribution (RFI-MDI-QKD) is immune to both detector side channel attacks and slow drifts of reference frames, and thus possesses a high level of security. In the traditional protocol of RFI-MDI-QKD, each legitimate user, Alice or Bob, has to prepare six encoding states in three orthogonal bases ($X, Y$, and $Z$), which are sent to the untrusted third party, Charlie. Then all the successful counting events are utilized for parameter estimations, which suffer from a severe finite-size effect. In this paper, we propose a scheme on RFI-MDI-QKD with fewer states and demonstrate that it can be secured and made useful by utilizing semidefinite programming. Numerical simulation results reveal the equivalence between those groups of different monitoring states from Alice and Bob. In addition, when considering practical experimental conditions and taking finite-size effects into account, our proposal can achieve a better performance than the traditional six-state RFI-MDI scheme, which provides a resource-saving and simpler state-preparation approach for practical implementations without sacrificing key rates.

Blind quantum computation (BQC) is one of the most promising alternatives to solve the shortage of quantum resources in the implementation of quantum computations. However, in most reported studies on decoy-state BQCs, the source intensities are actively modulated, which, unfortunately, results in side-channel leakage and undermines system security. In this paper, we present a passive decoy-state BQC scheme with a heralded single-photon source that circumvents this problem skillfully, improving system security. Moreover, simulation results show that our passive scheme can even outperform other active schemes in the generation efficiency of blind states when using state-of-the-art equipment.

The mechanism underlying the role of oxidative stress and advanced oxidation protein products (AOPPs) in the aetiology of premature ovarian insufficiency (POI) is poorly understood. Here, we investigated the plasma AOPP level in POI patients and the effects of AOPPs on granulosa cells both in vitro and in vivo. KGN cells were treated with different AOPP doses, and cell cycle distribution, intracellular reactive oxygen species (ROS), and protein expression levels were measured. Sprague–Dawley (SD) rats were treated daily with PBS, rat serum albumin, AOPP, or AOPP+ N-acetylcysteine (NAC) for 12 weeks to explore the effect of AOPPs on ovarian function. Plasma AOPP concentrations were significantly higher in both POI and biochemical POI patients than in controls and negatively correlated with anti-Müllerian hormone and the antral follicle count. KGN cells treated with AOPP exhibited G1/G0-phase arrest. AOPP induced G1/G0-phase arrest in KGN cells by activating the ROS-c-Jun N-terminal kinase (JNK)/p38 mitogen-activated protein kinase (MAPK)-p21 pathway. Pretreatment with NAC, SP600125, SB203580, and si-p21 blocked AOPP-induced G1/G0-phase arrest. In SD rats, AOPP treatment increased the proportion of atretic follicles, and NAC attenuated the adverse effects of AOPPs in the ovary. In conclusion, we provide mechanistic evidence that AOPPs may induce cell cycle arrest in granulosa cells via the ROS-JNK/p38 MAPK-p21 pathway and thus may be a novel biomarker of POI.

Considering imperfect experimental conditions, here we give a practical proposal on the decoy-state quantum random number generator with weak coherent sources. In the proposal, three different intensities of light sources are used to estimate the contribution of single-photon pulses. System randomness is characterized and simulated through the minimum entropy. Numerical simulation results show that our new scheme can extract out randomness close to the asymptotic case where the fraction of single-photon pulses are exactly known. Furthermore, a proof-of-principle experimental demonstration is presented, showing excellent agreement between experiment and theory.

Digital signature is a key technique in information security, especially for identity authentications. Compared with classical correspondence, quantum digital signatures (QDSs) provide a considerably higher level of security, i.e., information-theoretic security. At present, its performance is limited by key generation protocols (e.g., BB84 or measurement-device-independent protocols), which are fundamentally limited in terms of channel capacity. Fortunately, the recently proposed twin-field quantum key distribution can overcome this limit. This paper presents a twin-field QDS protocol and details a corresponding security analysis. In its distribution stage, a specific key generation protocol, the sending-or-not-sending twin-field protocol, has been adopted and full parameter optimization method has been implemented. Numerical simulation results show that the new protocol exhibits outstanding security and practicality compared with all other existing protocols. Aim: To identify the expression profiles and potential functions of circular RNAs (circRNAs) in granulosa cells (GCs) from women with biochemical premature ovarian insufficiency (bPOI). Patients & methods: CircRNAs microarray analysis was performed to GCs from 8 patients with bPOI and 8 control women, followed by qRT-PCR in 15 paired samples. CircRNA–miRNA networks and the prediction of their enriched signaling pathways were conducted by bioinformatics analysis. Results: A total of 133 upregulated and 424 downregulated circRNAs was identified in women with bPOI. We constructed circRNA–miRNA networks and found that the most predominantly enriched signaling pathways were the FoxO signaling pathway and cellular senescence. Conclusion: CircRNAs are differentially expressed in bPOI, which might contribute to the pathogenesis of bPOI.

The measurement-device-independent quantum key distribution (MDI-QKD) protocol plays an important role in quantum communications due to its high level of security and practicability. It can be immune to all side-channel attacks directed on the detecting devices. However, the protocol still contains strict requirements during state preparation in most existing MDI-QKD schemes, e.g., perfect state preparation or perfectly characterized sources, which are very hard to realize in practice. In this letter, we investigate uncharacterized MDI-QKD by utilizing a three-state method, greatly reducing the finite-size effect. The only requirement for state preparation is that the state are prepared in a bidimensional Hilbert space. Furthermore, a proof-of-principle demonstration over a 170 km transmission distance is achieved, representing the longest transmission distance under the same security level on record., 5 pages, 2 figures

STUDY QUESTION Is there a specific mechanism underlying the association between lung adenocarcinoma transcript 1 (MALAT1) and endometriosis-related infertility? SUMMARY ANSWER The down-regulation of MALAT1 in endometriosis granulosa cells (GCs) may have an adverse effect on the growth and development of oocytes by inhibiting GC proliferation, due to cell cycle-dependent mechanisms that enhance P21 expression through activation of the extracellular signal-regulated kinase (ERK)/mitogen-activated protein kinase (MAPK) pathway. WHAT IS KNOWN ALREADY The association between endometriosis and infertility is well supported throughout the literature, and endometriosis per se and its surgical treatment have an adverse effect on the ovarian reserve and on oocyte development. MALAT1, one of the most extensively expressed and evolutionarily conserved transcripts, has been implicated to play a role in human development and many diseases. However, little is known about the role of MALAT1 long non-coding RNA (lncRNA) in endometriosis and its associated infertility. STUDY DESIGN, SIZE, DURATION We measured MALAT1 lncRNA expression levels in GCs from 52 endometriosis patients and 52 controls. Also, MALAT1 was knocked down in a human GC tumor-derived cell line, KGN, to investigate the role of MALAT1 and its molecular mechanism in cell proliferation. PARTICIPANTS/MATERIALS, SETTING, METHODS GCs were collected from women with or without endometriosis undergoing IVF or ICSI treatment. All endometriosis patients were diagnosed by laparoscopy or laparotomy, and control patients were limited to male factor or tubal disease and had a normal ovarian reserve. Quantitative real-time PCR (qRT-PCR) was used to measure the differential expression levels of MALAT1 lncRNA between endometriosis patients and controls. The receiver operating characteristic (ROC) curve was drawn to evaluate the diagnostic values of MALAT1 in endometriosis. In the KGN cell line, MALAT1 was knocked down with locked nucleic acid GapmeRs. Cell counting kit-8 assays, ethynyl-2-deoxyuridine assays and flow cytometry were used to study the role of MALAT1 in cell proliferation and cell-cycle progression, and western blotting was performed to detect the potential underlying mechanism. MAIN RESULTS AND THE ROLE OF CHANCE We first found that MALAT1 lncRNA was significantly down-regulated in endometriosis GCs and was associated with the antral follicle count (R = 0.376, P < 0.001 versus control). In addition, MALAT1 lncRNA levels were significantly lower in the GCs of infertile women with advanced stages of endometriosis (P = 0.01 versus control). The ROC curves illustrated strong separation between all the endometriosis patients and the control group (AUC: 0.705; 95% CI: 0.606-0.804; P < 0.001), Stage I-II and control group (AUC: 0.651; 95% CI: 0.536-0.767; P = 0.016), and Stage III-IV and control group (AUC: 0.827; 95% CI: 0.718-0.936; P < 0.001). MALAT1 lncRNA was primarily localized in the nuclei of GCs. We found a negative correlation between MALAT1 lncRNA and P21 mRNA in the GCs from patients (R = -0.628; P < 0.001). MALAT1 knockdown in KGN cells inhibited cell proliferation and cell-cycle progression. In addition, MALAT1 knockdown induced an increase in both the mRNA and protein levels of P21, and of P53, phosphorylated ERK1/2 (p-ERK1/2) and phosphorylated c-Jun N-terminal protein kinase (p-JNK) protein levels, as well as causing a decrease in cyclin dependent kinase 2 (CDK2), cyclin D1 and p-P38 MAPK protein levels. Furthermore, inhibition of the ERK/MAPK pathway with U0126, the up-regulation of p-ERK1/2, P21 and P53, and the down-regulation of CDK2 and cyclin D1 by the knockdown of MALAT1 were all attenuated by MALAT1 knockdown. Therefore, MALAT1 may regulate GC proliferation through P21/P53-dependent control of the cell cycle, and the ERK/MAPK pathway participates in this process. LARGE SCALE DATA None. LIMITATIONS, REASONS FOR CAUTION The hormonal treatment used in IVF and surgical removal of endometriotic lesions may have altered MALAT1 expression in GCs. The ovarian granulosa-like tumor cell line, KGN, was used for further functional and mechanistic studies due to the difficulties in obtaining human GCs in sizable amounts and maintaining primary cultures. WIDER IMPLICATIONS OF THE FINDINGS Our finding represents the first example of an lncRNA-based mechanism in endometriosis GCs. Women with endometriosis show altered MALAT1 expression levels in GCs that may impair fertility by regulating the function of GCs. Therefore, analysis of MALAT1 and its molecular mechanisms of action provide new insights into the pathogenesis of endometriosis and its associated infertility. In quantum key distribution (QKD), the passive decoy-state method can simplify the intensity modulation and reduce some of side-channel information leakage and modulation errors. It is usually implemented with a heralded single-photon source. In Wang et al 2016 (Phys. Rev. A 96 032312), a novel passive decoy-state method is proposed by Wang et al, which uses two local detectors to generate more detection events for tightly estimating channel parameters. However, in the original scheme, the two local detectors are assumed to be identical, including the same detection efficiency and dark count rate, which is often not satisfied in the realistic experiment. In this paper, we construct a model for this passive decoy-state QKD scheme with two mismatched detectors and explore the effect on QKD performance with certain parameters. We also take the finite-size effect into consideration, showing the performance with statistical fluctuations. The results show that the efficiencies of local detectors affect the key rate more obviously than dark count rates.

Currently, there is a growing concern regarding the safety of assisted reproductive technology (ART) due to increased risk of spontaneous abortion (SA) and imprinting disorders in ART-conceived offspring. Early investigations suggested that aberrant genetic imprinting may be related to pregnancy loss; however, few studies have used human tissue specimens. Here the DNA methylation patterns of 3 imprinted genes, including maternally inherited GRB10 and the paternally inherited IGF2 and PEG3 genes, were evaluated in human chorionic villus samples by pyrosequencing and bisulfite sequencing polymerase chain reaction. The samples were divided into 4 groups: (1) SA of natural conception (NC; n = 84), (2) induced abortion of NC (n = 94), (3) SA after ART (n = 73), and (4) fetal reduction after ART (n = 86). The methylation levels and the percentages of abnormal methylation of the IGF2, GRB10, and PEG3 genes between the ART group and the NC group showed no significant difference. Both IGF2 and GRB10 genes showed higher methylation levels in the SA group compared to the non-SA group. Additionally, determining the single-nucleotide polymorphisms of 4 loci, including IGF2 rs3741205, rs3741206, rs3741211, and GRB10 rs2237457, showed that the TC+CC genotype of IGF2 rs3741211 had a 1.91-fold increased risk of SA after ART. However, there was no association between the mutant genotype of IGF2 rs3741211 and the methylation levels of IGF2 and H19, and ART might not affect the distribution of the abovementioned genotypes. It provides support for the opinion that genetic imprinting defects may be associated with SA, which might not be due to ART treatments.

Quantum digital signature (QDS) guarantee the unforgeability, nonrepudiation and transferability of signature messages with information-theoretical security, and hence has attracted much attention recently. However, most previous implementations of QDS showed relatively low signature rates or/and short transmission distance. In this paper, we report a proof-of-principle phase-encoding QDS demonstration using only one decoy state. Firstly, such method avoids the modulation of vacuum state, thus reducing experimental complexity and random number consumption. Moreover, incorporating with low-loss asymmetric Mach-Zehnder interferometers and real-time polarization calibration technique, we have successfully achieved higher signature rate, e.g., 0.98 bit/s at 103 km, and to date a record-breaking transmission distance over 280-km installed fibers. The present work describes the in vitro antibacterial evaluation of some new pyrimidine derivatives. Twenty-two target compounds were designed, synthesized and preliminarily explored for their antimicrobial activities. The antimicrobial assay revealed that some target compounds exhibited significantly inhibitory efficiencies toward bacteria and fungal including drug-resistant pathogens. Compound 7c presented the most potent inhibitory activities against Gram-positive bacteria (e.g., Staphylococcus aureus 4220), Gram-negative bacteria (e.g., Escherichia coli 1924) and the fungus Candida albicans 7535, with an MIC of 2.4 μmol/L. Compound 7c was also the most potent, with MICs of 2.4 or 4.8 μmol/L against four multidrug-resistant, Gram-positive bacterial strains. The toxicity evaluation of the compounds 7c, 10a, 19d and 26b was assessed in human normal liver cells (L02 cells). Molecular docking simulation and analysis suggested that compound 7c has a good interaction with the active cavities of dihydrofolate reductase (DHFR). In vitro enzyme study implied that compound 7c also displayed DHFR inhibition.

Background Premature ovarian insufficiency (POI) leads to early loss of ovarian function in women aged A) associated with POI was identified by whole-exome sequencing. This mutation was heterozygous in the affected family members and was absent in the unaffected family members. In silico analysis predicted that the mutation was potentially pathogenic. Bioinformatic splice prediction tools revealed that the mutation was very likely to have a strong impact on splice site function. Results of the minigene assay revealed that the mutation changed the mRNA splicing repertory. Conclusions The missense mutation of the HFM1 gene (c.3470G > A) may be a cause of POI. The mutation altered mRNA splicing in cells. This study can provide geneticists with deeper insight into the pathogenesis of POI and aid clinicians in making early diagnoses in affected women.

We generalize BB84 quantum key distribution (QKD) to the scenario where the receiver adopts a heralded quantum memory (QM). With the heralded QM, the valid dark count rate of the receiver’s single photon detectors can be mitigated obviously, which will lower the quantum bit error rate, and thus improve the performance of decoy-state BB84 QKD systems in long distance range. Simulation results show that, with practical experimental system parameters, decoy-state BB84 QKD with QM can exhibit performance comparable to that of without QM in short distance range, and exhibit performance better than that without QM in long distance range.

The aim of this study was to explore the association of the DNA-methyltransferase (DNMT)-3A and DNMT3B promoter polymorphisms with the risk of human spontaneous abortion after assisted reproduction techniques (ARTs) and natural conception. We collected tissues from women who underwent abortion procedures: (a) chorionic villus samples (CVS) and muscle samples (MS) from spontaneous abortions conceived by ART and natural cycle (study group), n = 152; and (b) CVS and MS from normal early pregnancy and second trimester (control group), n = 155. The single-nucleotide polymorphism (SNP) −448A > G in the DNMT3A promoter region and −149C/T polymorphism of DNMT3B were determined by polymerase chain reaction–restriction fragment length polymorphism (PCR-RFLP) and confirmed by sequencing. The allele frequency of −448A among pregnancy loss group and control group was 34.2 % vs. 16.5 %, respectively. Compared with GG carriers, the DNMT3A −448AA homozygotes had an about 16-fold increased risk of spontaneous abortion [odds ratio (OR) = 16.130, 95 % confidence interval (CI), 3.665–70.984], and AG heterozygotes had an OR of 2.027 (95 % CI, 1.247–3.293). However, the distribution of −448A > G in individuals derived from ART pregnancies was not statistically significantly compared with those derived from spontaneous pregnancies (P = 0.661). For DNMT3B, we observed genotype frequencies of 100 % (TT) in the study group and the control group. The DNMT3A −448A > G polymorphism may be a novel functional SNP and contribute to its genetic susceptibility to spontaneous abortion in Chinese women, and ART may not affect the distribution of −448A > G in pregnancy loss and normal pregnancy. The observed TT genotype of DMNT3B suggests that this is the predominant genotype of this population. The findings provide new insights into the etiology of human spontaneous abortion.
