The interplay between G4s and R-loops are emerging in regulating DNA repair, replication, and transcription. A comprehensive picture of native co-localized G4s and R-loops in living cells is currently lacking. Here, we describe the development of HepG4-seq and an optimized HBD-seq methods, which robustly capture native G4s and R-loops, respectively, in living cells. We successfully employed these methods to establish comprehensive maps of native co-localized G4s and R-loops in human HEK293 cells and mouse embryonic stem cells (mESCs). We discovered that co-localized G4s and R-loops are dynamically altered in a cell type-dependent manner and are largely localized at active promoters and enhancers of transcriptional active genes. We further demonstrated the helicase Dhx9 as a direct and major regulator that modulates the formation and resolution of co-localized G4s and R-loops. Depletion of Dhx9 impaired the self-renewal and differentiation capacities of mESCs by altering the transcription of co-localized G4s and R-loops -associated genes. Taken together, our work established that the endogenous co-localized G4s and R-loops are prevalently persisted in the regulatory regions of active genes and are involved in the transcriptional regulation of their linked genes, opening the door for exploring broader roles of co-localized G4s and R-loops in development and disease.

Abstract Background This study aims to compare the clinical outcomes and safety of a novel hand-held retractor system-assisted Wiltse TLIF with that P-TLIF and assess whether this hand-held retractor system assisted Wiltse TLIF can yield less paraspinal muscle injury. Methods 56 patients (P-TLIF: 26, Wiltse TLIF: 30) were included in this one year prospective controlled study. The operation time, intraoperative blood loss, postoperative drainage, mobilization time, and discharge time were recorded. The clinical outcomes were evaluated by ODI, VAS, JOA, and SF-36 scores (7 days, 3, 6, and 12 months after surgery). Paraspinal muscle injury was assessed by postoperative MRI (6 months after surgery). CK and C-reaction protein were measured pre and postoperatively, and CT or X-ray (one year postoperatively) was used to assess bony union/non-union. Results The Wiltse (study) group was associated with significantly less estimated blood loss (79.67 ± 28.59 ml vs 192.31 ± 59.48 ml, P = 0.000*), postoperative drainage (43.33 ± 27.89 ml vs 285.57 ± 123.05 ml, P = 0.000*), and shorter mobilization (4.1 ± 1.2 d vs. 3.0 ± 0.9 d, P

We aimed to assess the effects of standard resin preparation models with five different thicknesses of occlusal surface on the fracture strengths of zirconia (ZrO2) and lithium disilicate glass ceramics. The specimens of 10 first maxillary molars collected between January 2019 and January 2020 were selected. Standard mathematical models were formed after scanning the resin matrices using software. The full crowns with five different thicknesses of occlusal surface were established, among which the molar specimens prepared by ZrO2 glass ceramic composites alone were assigned into ZrO2 group (n=5, 40 specimens) while those prepared using ZrO2-lithium disilicate glass ceramic composites were allocated into ZTCLDC group (n=5, 40 specimens). When the thickness of glass-ceramic full crowns was 0.5, 0.8, 1.0, 1.2 and 1.5 mm, the fracture load of the specimens in ZTCLDC group was not significantly different from that in ZrO2 group, and there was no significant difference in the three-point flexural strength between ZTCLDC group and ZrO2 group (P>0.05). The fracture toughness was not significantly different between the two groups in the case of the thickness of glass-ceramic full crown at 0.5, 0.8, 1.0, 1.2 and 1.5 mm (P>0.05). For the purpose of preparing anti-fenitrothion nanobodies with high-purity efficiently, the expression conditions of a recombinant anti-fenitrothion nanobody with low expression level in E. coli was optimized. The IPTG concentration and inducing temperature were selected as the independent variables and the nanobody expression level was used as the dependent variable for single-factor experiments, and the purification strategy was also investigated. The results showed that the highest expression level of 6 mg/L anti-fenitrothion nanobody was achieved at 37 ℃ without IPTG. Moreover, an interactive effect of IPTG dosage and inducing temperature was found. After a two-step purification of Ni affinity chromatography and gel filtration chromatography, the anti-fenitrothion nanobodies was finally yielded with more than 98% purity, indicating that the expression and purification strategies in this study can obtain anti-fenitrothion nanobodies efficiently. An ic-ELISA assay was then established based on the obtained nanobody, with an IC50 of 5.81 ng/mL and an LOD of 0.25 ng/mL, and a detection range of 0.78~43.07 ng/mL. The assay was applied to determine fenitrothion in Chinese cabbage and lettuce samples. The recovery rate of addition was between 93.3%~111.7%, and the coefficient of variation (CV) was between 2.3%~18.2%. The proposed assay based on anti-fenitrothion nanobody has high sensitivity, which can meet the requirements for monitoring of fenitrothion under the national standard, thus can be used for rapid screening of fenitrothion. This study laid a foundation for the development of efficient and rapid immunoassays for fenitrothion.

In direction-of-arrival (DOA) estimation with sensor arrays, the background noise is usually modeled to be uncorrelated uniform white noise, such that the related algorithms can be greatly simplified by making use of the property of the noise covariance matrix being a diagonal matrix with identical diagonal entries. However, this model can be easily violated by the nonuniformity of sensor noise and the presence of outliers that may arise from unexpected impulsive noise. To tackle this problem, we first introduce an exploratory factor analysis (EFA) model for DOA estimation in nonuniform noise. Then, to deal with the outliers, a generalized extreme Studentized deviate (ESD) test is applied for outlier detection and trimming. Based on the trimmed data matrix, a modified EFA model, which belongs to weighted least-squares (WLS) fitting problems, is presented. Furthermore, a monotonic convergent iterative reweighted least-squares (IRLS) algorithm, called the iterative majorization approach, is introduced to solve the WLS problem. Simulation results show that the proposed algorithm offers improved robustness against nonuniform noise and observation outliers over traditional algorithms.

Morphing wingtips have the potential to improve aircraft performance. By connecting the wingtips and the wings with a compliant structure, a continuous aerodynamic surface can be achieved for a better aerodynamic performance. However, how to maintain the shape-changing capability while keeping a high stiffness to carry aerodynamic loads is a key problem. In this paper, based on asymmetric stiffness, a type of single-row corrugated panel is designed to satisfy the limited space around the wingtip. A finite element model of the single-row corrugated panels is established, and parameter analysis is performed to investigate the impact of the thickness characteristics of the corrugated panel on the folding angle. The corrugated panel is then optimised to find the maximum folding angle. Based on the optimisation results, corrugated panels with asymmetric and symmetric stiffness are fabricated and tested. The results demonstrate that the asymmetric stiffness corrugated panels have the capability to increase the wingtip folding angle.

Snow Lotus Seed (SLS), esteemed for its nutritional and market value, faces challenges of authentication due to the absence of appropriate testing standards and methods. This results in frequent adulteration of SLS sourced from Gleditsia sinensis (G. sinensis) with other plant seeds endosperm. Traditional chloroplast DNA barcoding methods are inadequate for species identification due to the absence of chloroplasts in G. sinensis seeds endosperm. In this study, the homology of 11 ITS genes among 6 common Gleditsia species was analyzed. Universal primers suitable for these species were designed and screened. A DNA barcoding method for distinguishing SLS species was developed using Sanger sequencing technology, leveraging existing GenBank and Barcode of Life Data System (BOLD) databases. Optimized sample pretreatment facilitated effective DNA extraction from phytopolysaccharide-rich SLS. Through testing of commercial SLS products, the species origin has been successfully identified. Additionally, a novel instance of food fraud was uncovered, where the Caesalpinia spinosa endosperm was used to counterfeit SLS for the first time. The study established that the developed DNA barcoding method is effective for authenticating SLS species. It is of great significance for combating food fraud related to SLS, ensuring food safety, and promoting the healthy development of the SLS industry.

During the water entry process of a trans-domain morphing aircraft, significant impact forces are generated when the aircraft hits the water surface, which will potentially cause the deformation of the cabin structure and might damage the structure or onboard devices. Thus, it is necessary to investigate the water entry process of the cabin structure. This paper analyses changes in fluid loads and the corresponding structural responses during the water entry process. Firstly, the numerical model is established for the water entry process and the modeling method is validated by comparing the results to the experimental data. An empirical formula is developed to correlate the impact loads with the water entry velocities. Then, fluid–structure interaction analysis of the water entry process is performed using a two-way coupling approach. The relationship between structural deformation and the water entry process is then investigated. The results are compared with those without considering the structural deformation. The empirical formula is then modified to reflect the effects of the deformation. The results show that structural deformation will disperse the impact load, which represents different responses compared to the rigid cabin structure.

The adulteration of goat milk powder occurs frequently; cattle-derived and soybean-derived ingredients are common adulterants in goat milk powder. However, simultaneously and rapidly detecting cattle-derived and soybean-derived components is still a challenge. An efficient, high-throughput screening method for adulteration detection is needed. In this study, a rapid method was developed to detect the adulteration of common cattle-derived and soybean-derived components simultaneously in goat milk powder by combining the CRISPR/Cas12a system with recombinant polymerase amplification (RPA). A dual DNA extraction method was employed. Primers and crRNA for dual detection were designed and screened, and a series of condition optimizations were carried out in this experiment. The optimized assay rapidly detected cattle-derived and soybean-derived components in 40 min. The detection limits of both cattle-derived and soybean-derived components were 1% (w/w) for the mixed adulteration models. The established method was applied to a blind survey of 55 commercially available goat milk powder products. The results revealed that 36.36% of the samples contained cattle-derived or soybean-derived ingredients, which revealed the noticeable adulteration situation in the goat milk powder market. This study realized a fast flow of dual extraction, dual amplification, and dual detection of cattle-derived and soybean-derived components in goat milk powder for the first time. The method developed can be used for high-throughput and high-efficiency on-site primary screening of goat milk powder adulterants, and provides a technical reference for combating food adulteration.

Ionizing radiation (IR)-induced hematopoietic injury has become a global concern in the past decade. The underlying cause of this condition is a compromised hematopoietic reserve, and this kind of hematopoietic injury could result in infection or bleeding, in addition to lethal mishaps. Therefore, developing an effective treatment for this condition is imperative. Fluacrypyrim (FAPM) is a recognized effective inhibitor of STAT3, which exhibits anti-inflammation and anti-tumor effects in hematopoietic disorders. In this context, the present study aimed to determine whether FAPM could serve as a curative agent in hematopoietic-acute radiation syndrome (H-ARS) after total body irradiation (TBI). The results revealed that the peritoneally injection of FAPM could effectively promote mice survival after lethal dose irradiation. In addition, promising recovery of peripheral blood, bone marrow (BM) cell counts, hematopoietic stem cell (HSC) cellularity, BM colony-forming ability, and HSC reconstituting ability upon FAPM treatment after sublethal dose irradiation was noted. Furthermore, FAPM could reduce IR-induced apoptosis in hematopoietic stem and progenitor cells (HSPCs) both in vitro and in vivo. Specifically, FAPM could downregulate the expressions of p53-PUMA pathway target genes, such as Puma, Bax, and Noxa. These results suggested that FAPM played a protective role in IR-induced hematopoietic damage and that the possible underlying mechanism was the modulation of apoptotic activities in HSCs.

Propisochlor is a chloroacetamide herbicide causing liver toxicity and suppressing immunity in human and animal. Although the herbicide has been used for years, the effects of propisochlor on intestinal health remain poorly understood. Hence, the impacts of propisochlor in intestinal health and gut microbiota were analyzed by using molecular approach and bacterial 16S rRNA sequencing. The result showed that the intake of propisochlor in mice impaired gut morphology, reduced expression of tight junction proteins, decreased thickness of mucus layer and activated pyroptosis signaling. Moreover, the exposure of propisochlor in mice led to significant alterations in gut microbial diversity and composition, including an increase of Bacteroidetes and a decrease of Firmicutes. The gut microbiota, such as Parabacteroides, Parasutterella, and Bacteroides, demonstrated a strong negative correlation with the intestinal health. These findings suggested that gut microbiota could play a critical role in the propisochlor-induced pyroptosis.

This paper based on the first-principles simulations sheds light on the chalcogenide-defected behavior of the Janus SnSSe monolayer and the sensing property of the defected-SnSSe monolayer upon three typical thermal runaway fault gases, namely H2, CO and C2H2. Results indicate that the Se-vacancy is more likely to be formed in a SnSSe monolayer, and the Se–SnSSe monolayer performs physisorption, with adsorption energies of −0.14, −0.61 and −0.37 eV for three systems, and electron-donating property, contributing 0.047, 0.087 and 0.070 e to such three gases. The band structure and work function analyses reveal the desirable potential of Se–SnSSe monolayer as a resistance-type or WF-based sensor for CO detection. Also, the recovery property is analyzed to consider the reusability of such sensing material. Besides, the applied biaxial strains reveal that Se–SnSSe monolayer performs high and tunable sensitivity for these gas species, especially for H2 and C2H2 detections. This work paves the way to explore SnSSe-based material for gas sensing application in many typical fields involving H2, CO and C2H2 detection, which is also guidable to explore the sensing potential of many other Janus 2D materials.

Abstract Background IgA vasculitis (IgAV) is a common type of vasculitis seen in children. IgAV recurrence can result in chronic kidney disease. We aimed to explore the association between common laboratory indices and IgAV recurrence in children, and to establish a prediction model. Methods This retrospective study included children with diagnosed with IgAV hospitalized in Bazhong Central Hospital, Sichuan, from January 2014 to December 2019. Children were assigned to two groups based on IgAV recurrence, and baseline clinical data were collected for comparison. A logistic regression model to predict IgAV recurrence was established. The receiver operating characteristic curve was plotted. The area under the curve (AUC) was used to detect performance of the predictive model. Results This study included 193 children (39 [20.2%], recurrence group; 154 [79.8%], non-recurrence group). Based on multivariate regression analysis, the duration of illness and joint involvement were independent predictors of IgAV recurrence in children (P < 0.05). No significant differences were observed in common laboratory indices (P > 0.05). The AUC of the prediction model was 0.766 (P < 0.001) with sensitivity of 74.4% and specificity of 68.8%. Conclusion Common laboratory indices were not associated with recurrence of IgAV in children.

AimTo investigate the clinical characteristics and health resource costs among children hospitalised for injuries in southern Sichuan, China, and to provide guidance for prevention and treatment.MethodsWe collected clinical data concerning children aged from 29 days to 18 years hospitalised for injuries from January 1, 2017, to December 31, 2021, retrospectively analysing the basic characteristics, evolution of injury characteristics over time, risk factors for events with adverse outcomes, and health resource costs.ResultsAmong 5,826 hospitalised children with injuries, males (63.6%), those in rural areas (40.3%), and adolescents (33.5%) were most commonly injured. Most injuries occurred at home (52.6%), and during summer. The most common injury types were falls, burns, road traffic injuries, poisoning, and foreign body injuries (32.0%, 17.9%, 13.6%, 8.8%, and 7.9%, respectively). After 2019, the proportion of intentional injuries among adolescent girls was significantly higher. Road traffic injuries most commonly led to poor clinical outcomes (95%CI: 5.39–31.51), followed by falls (95%CI: 2.20–10.67). Adolescents were at higher risk of poor prognosis. Injuries occurring in rural areas, adolescents, road traffic injuries, and falls cost high health resource.ConclusionInjuries among children remain serious, with males and adolescents from villages predominantly affected. Attention should be paid to intentional injuries among adolescent females also. Targeted prevention and control measures for road traffic injuries and falls should be strengthened.

Post-fermented tea (PFT) is one of the most commonly consumed beverages worldwide. Rapid microbial growth and significant changes in the microbial composition of PFT during processing and storage pose a potential risk of contamination with mycotoxins such as zearalenone (ZEN). Screening for ZEN contamination in a simple, rapid, and inexpensive manner is required to ensure that PFT is safe for consumption. To monitor ZEN in PFT, ZEN was conjugated with bovine serum albumin to prepare egg yolk immunoglobulins (IgY). A specific indirect competitive enzyme-linked immunosorbent assay (ic-ELISA) based on IgY was developed and validated. ZEN was extracted with acetonitrile and water (50:50, v/v) containing 5% acetic acid and purified using a mixture of primary and secondary amines and graphitized carbon black to remove matrix interference from the PFT samples. Under optimal conditions, the linear range of this assay was 13.8−508.9 ng mL−1, the limit of detection was 9.3 ng mL−1, and the half-maximal inhibitory concentration was 83.8 ng mL−1. Cross-reactivity was negligible, and the assay was specific for ZEN-related molecules. The recovery rate of ZEN in the control blanks of PFT samples spiked with a defined concentration of ZEN of 89.5% to 98.0%. The recovery and accuracy of the method were qualified for PFT matrices. No significant differences were evident between the results of the actual PFT samples analyzed by high-performance liquid chromatography and ic-ELISA. The collective data indicate that the developed ic-ELISA can be used for the rapid and simple detection of ZEN in PFT products.

Flywheels play a critical role as core components in satellite attitude control systems. However, their high-speed rotation inevitably generates vibrations that have a detrimental impact on the in-orbit imaging capabilities of high-precision remote sensing payloads. This study focuses on the passive vibration isolation design of satellite flywheels. The flywheel-mounted structural plate and flywheel vibration isolation platform are considered as a whole system (termed a plate-isolator system). In this system, the structural plate is treated as an elastomer. By simplifying the plate-isolator system as a 2-degree-of-freedom vibration system, it becomes evident that obtaining an ideal vibration isolation effect through the optimization of the flywheel vibration isolation platform (FVIP) alone is difficult. In order to enhance the passive vibration isolation effect for satellite flywheels, this study introduces the concept of an elastic boundary applied to the flywheel-mounted structural plate, thus treating the elastic boundary as a design factor. Consequently, the plate-isolator system can be simplified as a 3-stage vibration isolation system. The optimization of the elastic boundary condition of the structural plate is performed using the kinetic model of the simplified 3-stage system. The vibration isolation effect of the plate-isolator system with an elastic boundary is further confirmed through finite element simulation. The calculation results demonstrate that, after establishing a reasonable elastic boundary for the satellite structural plate, the overall vibration/force transmission rate of the plate-isolator system becomes similar to that of a single-degree-of-freedom dynamic system. Finally, the proposed concept is validated through kinetic response analysis of a cube satellite. The results reveal that the vibration amplitude of the satellite’s top and side structural plates can be effectively lowered if the elastic boundary condition is set for the flywheel-mounted bottom structural plate.

The driving operations of the subway system are of great significance in ensuring the safety of trains. There are several hand actions defined in the driving instructions that the driver must strictly execute while operating the train. The actions directly indicate whether equipment is normally operating. Therefore, it is important to automatically sense the region of the driver and detect the actions of the driver from surveillance cameras to determine whether they are carrying out the corresponding actions correctly or not. In this paper, a lightweight two-stage model for subway driver action sensoring and detection is proposed, consisting of a driver detection network to sense the region of the driver and an action recognition network to recognize the category of an action. The driver detection network adopts the pretrained MobileNetV2-SSDLite. The action recognition network employs an improved ShuffleNetV2, which incorporates a spatial enhanced module (SEM), improved shuffle units (ISUs), and shuffle attention modules (SAMs). SEM is used to enhance the feature maps after convolutional downsampling. ISU introduces a new branch to expand the receptive field of the network. SAM enables the model to focus on important channels and key spatial locations. Experimental results show that the proposed model outperforms 3D MobileNetV1, 3D MobileNetV3, SlowFast, SlowOnly, and SE-STAD models. Furthermore, a subway driver action sensoring and detection system based on a surveillance camera is built, which is composed of a video-reading module, main operation module, and result-displaying module. The system can perform action sensoring and detection from surveillance cameras directly. According to the runtime analysis, the system meets the requirements for real-time detection.

Food Nutriology is one of the main course for students of Food Science in South China Agricultural University. The teaching group is devoted to reforming and innovation of teaching theory and practice, constantly improving the teaching resource, and applying the technical means of intelligent and information to teaching. The “learning-thinking-distinguishing-applying combination” teaching model is gradually formed. The course is awarded the national top-class undergraduate courses by the Ministry of Education in November 2020. The construction and development process of course, course positioning and objectives, construction of teaching group, course content and resources construction, teaching mode and characteristic innovation are introduced in this paper, to provide some useful references for advancing the construction and teaching reform of food science courses.

High-capacity Li-rich oxide materials have received extensive attention due to their unique anion–cation charge compensation involvement. However, the high operating voltage, poor cycling performance, unsafe oxygen evolution, and voltage decay limit their industrial application. The emergence and development of solid-state batteries offer a great opportunity to solve these issues by replacing flammable and unstable liquid electrolytes with solid electrolytes. Meanwhile, utilization of high-capacity Li-rich oxide cathodes enables to establish high-energy-density solid-state batteries with wide voltage ranges, light weight, and high mechanical properties. This review summarizes the recent progress of Li-rich oxide materials and solid electrolytes, emphasizing their major advantages, interface challenges, and modification approaches in the development of Li-rich solid-state batteries. We also propose possible characterization strategies for effective interfacial observation and analyses. It is hoped that this review should inspire the rational design and development of better solid-state batteries for application in portable devices, electric vehicles, as well as power grids.

Background: The association between serum bilirubin level and heart failure (HF) was controversial in previous observational studies and the causal effects of bilirubin on HF have not been investigated. Here, we conducted a Mendelian randomization (MR) study to investigate the associations between genetically determined bilirubin level and HF.Methods: Summary data on the association of single nucleotide polymorphisms (SNPs) with serum bilirubin levels were obtained from genome-wide association study (GWAS) for individuals of European descent and East Asian descent separately. Statistical data for gene-HF associations were extracted from three databases: the HERMES Consortium (47,309 cases and 930,014 controls), FinnGen study (30,098 cases and 229,612 controls) for European population and Biobank Japan (2,820 HF cases and 192,383 controls) for East Asian population. We applied a two-sample Mendelian randomization framework to investigate the causal association between serum bilirubin and HF.Results: Findings from our MR analyses showed that genetically determined serum bilirubin levels were not causally associated with HF risk in either European or East Asian population (odds ratio [OR] = 1.01 and 95% confidence interval [CI] = .97–1.05 for HERMES Consortium; OR = 1.01 and 95% CI = .98–1.04 for FinnGen Study; OR = .82, 95% CI: .61–1.10 for Biobank Japan). These results remained unchanged using different Mendelian randomization methods and in sensitivity analyses.Conclusion: Our study did not find any evidence to support a causal association between serum bilirubin and HF.

The advantages of genetic modification and preferable physicochemical qualities make nanobody (Nb) easy to develop a sensitive and stable immunosensor platform. Herein, an indirect competitive chemiluminescence enzyme immunoassay (ic-CLEIA) based on biotinylated Nb was established for the quantification of diazinon (DAZ). The anti-DAZ Nb, named Nb-EQ1, with good sensitivity and specificity, was obtained from an immunized library via a phage display technique, where the molecular docking results indicated that the hydrogen bond and hydrophobic interactions between DAZ and complementarity-determining region 3 and framework region 2 in Nb-EQ1 played a critical role in the Nb-DAZ affinity processes. Subsequently, the Nb-EQ1 was further biotinylated to generate a bi-functional Nb-biotin, and then an ic-CLEIA was developed for DAZ determination via signal amplification of the biotin–streptavidin platform. The results showed that the proposed method based on Nb-biotin had a high specificity and sensitivity to DAZ, with a relative broader linear range of 0.12–25.96 ng/mL. After being 2-folds dilution of the vegetable samples matrix, the average recoveries were 85.7–113.9% with a coefficient of variation of 4.2–19.2%. Moreover, the results for the analysis of real samples by the developed ic-CLEIA correlated well with that obtained by reference method GC-MS (R2 ≥ 0.97). In summary, the ic-CLEIA based on biotinylated Nb-EQ1 and streptavidin recognition demonstrated itself to be a convenient tool for the quantification of DAZ in vegetables.

Abstract Background The high quality of antibody (Ab) is critical for an immunoassay; usually, an Ab with low affinity is often regarded as a “bad” one in the immunoassay development. How to use a “bad” Ab to develop a highly sensitive immunoassay is still a huge challenge. Methods In this study, a heterologous immunoassay strategy was designed to enhance the sensitivity for the detection of banned dye, rhodamine B (RB), in fraudulent food. The RB Ab could not recognize RB by pairing with homologous coating antigen (Ag). However, the Ab showed unexpected high specificity and sensitivity recognition after being replaced by heterologous coating Ag. Indirect competitive enzyme-linked immunosorbent assay (icELISA) was developed based on the heterologous strategy. Results The detection limit of icELISA for chilli powder, Chinese prickly ash, hot-pot seasoning, and chilli sauce was 0.002 μg/kg, and the recoveries of the four samples ranged from 76.0 to 102.0%, with the coefficient of variation between 3.9 and 18.8%. Parallel experiment for 20 market samples with high-performance liquid chromatography (HPLC) was performed on to confirm the performance of the practical application of the developed icELISA, and the results of the two methods had good correlation. Molecular modeling inferred that the carboxyl group of hapten and its exposure level played an important role in the hapten-Ab recognition. Conclusions The proposed icELISA can be used for the surveillance screening of RB in a range of seasoning foods, and the heterologous strategy is an effective approach to enhance the sensitivity in an immunoassay.

Pathogenic variants of Burkholderia gladioli pose a serious threat to human health and food safety, but there is a lack of rapid and sensitive field detection methods for Burkholderia gladioli. In this study, the CRISPR/Cas12a system combined with recombinant enzyme polymerase amplification (RPA) was used to detect Burkholderia gladioli in food. The optimized RPA-CRISPR/Cas12a assay was able to specifically and stably detect Burkholderia gladioli at a constant 37 °C without the assistance of large equipment. The detection limit of the method was evaluated at two aspects, the genomic DNA (gDNA) level and bacterial quantity, of which there were 10−3 ng/μL and 101 CFU/mL, respectively. Three kinds of real food samples were tested. The detection limit for rice noodles, fresh white noodles, and glutinous rice flour samples was 101 CFU/mL, 102 CFU/mL, and 102 CFU/mL, respectively, without any enrichment steps. The whole detection process, including sample pretreatment and DNA extraction, did not exceed one hour. Compared with the qPCR method, the established RPA-CRISPR /Cas12a method was simpler and even more sensitive. Using this method, a visual detection of Burkholderia gladioli that is suitable for field detection can be achieved quickly and easily.

Single-atom catalysts (SACs) have been synthesized using a variety of methods in recent years, and they have shown excellent catalytic activities. However, metal atoms show a high tendency to agglomerate in liquid media, making the single atom synthesis more difficult in liquid media. The synthesis of such metal single-atom catalysts that do not have strong ligand coordination is rarely reported in the literature. Herein, we report the facile synthesis of monodispersed Au atoms (Au1) through the reduction in HAuCl4 in 15-crown-5. The complete reduction in HAuCl4 was confirmed through UV-Vis spectroscopy. In addition, the Au was found in a zero valence state after reduction, which was confirmed through XPS and XANES results. Moreover, the dispersion of Au was confirmed as a single atom (Au1) through transmission electron microscopy and spherical aberration electron microscopy. The possible structure of this catalyst was proposed by matching the EXAFS results with the structure of Au1@15-crown-5 as -(OC2H4O)-AuCl2H2. The Au1@15-crown-5 showed high activity (TOF as high as 22,075) in the reduction in nitrophenol and nitroaniline to aminophenol and phenylenediamine by sodium borohydride. Because of the monodispersion of Au atoms, its performance is much better than noble nanoparticles and non-precious metal catalysts.

Because of the serious adulteration of goat milk, the rapid on-site detection of goat milk powder adulteration is needed. In this study, the CRISPR/Cas12a detection system combined with recombinase polymerase amplification (RPA) was employed to qualitatively detect the adulteration of goat milk powder with cattle-derived components. Specific primers and crRNA were designed and screened. After the optimization of RPA and the Cas system, the RPA-CRISPR/Cas12a detection method was established. The detection can complete the rapid identification of cattle-derived components in 45 min, without the assistant of large equipment. The absolute detectability of the RPA-CRISPR/Cas12a assay could reach 10−2 ng/μL for cattle genomic DNA, and 1% (w/w) for cattle milk powder, which is suitable to meet the testing requirements for on-site detection. In total, 55 commercial goat milk powder products were collected for blind testing. The results showed that 27.3% of the samples were adulterated with cattle ingredients, revealing a serious adulteration situation in goat milk powder market. The RPA-CRISPR/Cas12a assay established in this research exhibited its potential for practical use of on-site detection to detect cow milk powder in goat milk powder and can provide reliable technical reference for combating food fraud of adulteration of goat milk products.

Corrugated panels are promising in the field of morphing structures due to their high anisotropy. A low stiffness in the morphing direction allows for a reduced actuation force, which provides the benefit of reducing the system weight. However, a low stiffness also leads to reduced critical buckling loads, which makes it easier for corrugated panels to buckle in the morphing process. The conflict between the actuation force and the critical buckling load requires an efficient method to predict the critical buckling load, which can then be applied in a trade-off study to find the optimal design. In the current study, a simplified method to predict the critical buckling load is proposed based on the derivation of equivalent properties. The method is verified using the finite element analysis and applied for the optimisation of corrugated panels. A conceptual design to reinforce corrugated panels is also proposed, which shows a significant increase in the critical buckling load of flexible corrugated panels.

Exposure to medium and high doses of ionizing radiation (IR) can induce long-term bone marrow (BM) suppression. We previously showed that recombinant human thrombopoietin (rhTPO) significantly promotes recovery from hematopoietic-acute radiation syndrome, but its effect on long-term BM suppression remains unknown. C57BL/6 mice were exposed to 6.5 Gy γ-rays of total body irradiation (TBI) at a dose-rate of 63.01 cGy per minute, and the mice were treated with rhTPO (100 μg; intramuscular injection) or vehicle at 2 h after TBI. All mice were killed one or two months after TBI for analysis of peripheral blood cell counts, long-term hematopoietic stem cell (HSC) frequency, and BM-derived clonogenic activity. The HSC self-renewal capacity was analyzed by BM transplantation. The levels of reactive oxygen species (ROS) production and ratios of γH2AX+ and p16, p53, and p21 mRNA in HSCs were measured by flow cytometry and real-time polymerase chain reaction, respectively. Treatment with rhTPO reduced long-term myelosuppression by improving long-term hematopoietic reconstitution (p < 0.05) after transplantation and resting state maintenance of HSCs (p < 0.05). Moreover, rhTPO treatment was associated with a sustained reduction in long-term ROS production, reduction of long-term DNA damage, diminished p53/p21 mRNA expression, and prevention of senescence after TBI. This study suggests rhTPO is an effective agent for treating IR-induced long-term BM injury because it regulates hematopoietic remodeling and HSC cycle disorder through the ROS/p53/p21/p16 pathway long term after IR.

Abstract Background General practitioners (GPs) are the foundation of any primary healthcare system. Their quality and quantity are directly associated with the effectiveness and quality of the health services of a nation. GPs’ shortage and turnover have become an important issue in developed and developing countries. An accurate estimate of turnover intention prevalence among GPs would have important health policy implications, but the overall prevalence is unknown. We aimed to summarize the global prevalence of turnover intention and associated factors among GPs. Methods We systematically reviewed the PubMed, Embase, Web of Science and China National Knowledge Infrastructure (CNKI) databases from their inception up to May 2020, as well as the reference lists of all included studies. We included observational studies that reported data on turnover intention or their prevalence rate among GPs could be calculated based on the information provided. The prevalence rate of the turnover intentions was estimated using a random-effects meta-analysis. The heterogeneity was evaluated using I 2 statistic. Differences by study level characteristics were estimated via subgroup analysis and meta-regression. Results A total of 25 cross-sectional studies were included (a total of 27,285 participants). The prevalence of turnover intention was 0.47 (95% CI: 0.39–0.55). Those having a lower level of salary (OR = 1.38, 95% CI: 1.13–1.63) and job satisfaction (OR = 1.35, 95% CI: 1.12–1.70) or having lower level of morale (OR = 2.68, 95% CI: 1.56–3.80) had a higher intention. In contrast, GPs with a lower level of professional title had a lower turnover intention (OR = 0.81, 95% CI: 0.65–0.98). Conclusions In this systematic review, approximately half of the GPs had the intention to leave their current posts worldwide. The factors associated with turnover intention were higher professional title, lower income level, lower job satisfaction and lower morale.

Na-ion batteries (NIBs) have received significant interest as potential candidates for large-scale energy storage owing to the widespread distribution of sodium and superior low-temperature performance. However, their commercial application is usually hindered by the high production cost and inadequate performance for electrode materials, particularly for cathodes. Na3(VOPO4)2F (NVOPF) has been recognized as one of the most promising cathodes for high-energy NIBs owing to the high working voltage and energy density. Here, we report a facile highly efficient room-temperature solution protocol for large-scale synthesis of NVOPF cathode for NIBs. By simply regulating pH, NVOPF can be obtained, which delivered a discharge capacity of 120.2 mAh g-1 at 0.1 C and 72% capacity retention over 8000 cycles at 25 C. Besides, the kilogram-level NVOPF products have been synthesized, and 26650 cylindrical cells were fabricated, which exhibit excellent cycling stabilities, remarkable low-temperature performance with comparable safety features. We hope our findings could provide insights on the industrial application of NVOPF in NIBs.

Low Phosphate Root (LPR) encodes a protein localized to the endoplasmic reticulum (ER) and cell wall. This gene plays a key role in responding to phosphate (Pi) deprivation, especially in remodeling the root system architecture (RSA). An identification and expression analysis of the OsLPR family in rice (Oryza sativa) has been previously reported, and OsLPR5, functioning in Pi uptake and translocation, is required for the normal growth and development of rice. However, the role of OsLPR3, one of the five members of this family in rice, in response to Pi deficiency and/or in the regulation of plant growth and development is unknown. Therefore, in this study, the roles of OsLPR3 in these processes were investigated, and some functions were found to differ between OsLPR3 and OsLPR5. OsLPR3 was found to be induced in the leaf blades, leaf sheaths, and roots under Pi deprivation. OsLPR3 overexpression strongly inhibited the growth and development of the rice but did not affect the Pi homeostasis of the plant. However, oslpr3 mutants improved RSA and Pi utilization, and they exhibited a higher tolerance to low Pi stress in rice. The agronomic traits of the oslpr3 mutants, such as 1000-grain weight and seed length, were stimulated under Pi-sufficient conditions, indicating that OsLPR3 plays roles different from those of OsLPR5 during plant growth and development, as well as in the maintenance of the Pi status of rice.

In this study, UPLC-MS/MS was used to study the effects of smoking duration and temperature on the formation of heterocyclic amines (HAs) in smoke-processed meat patties. Four kinds of free HAs—including F-7,8-DiMeIQx; F-MeAαC; F-Harman and F-Norharman—and six kinds of protein-bound HAs—including B-AαC; B-7,8-DiMeIQx; B-Glu-p-1; B-MeAαC; B-Harman and B-Norharman—were detected and quantified. Among the free HAs, we observed a 23-fold content increase (p < 0.05), from 0–4 h (at 0 h and 4 h they were 4.24 ng·g−1 and 98.33 ng·g−1, respectively), and the content of the free HAs decreased to 78.80 ng·g−1, at 5 h. At the same time, the free HAs content increased from 53.52 ng·g−1, at 50 °C, to 127.16 ng·g−1, at 60 °C, and then decreased continuously. The content of the free HAs was the highest at 60 °C. For the protein-bound HAs, their content was found to generally decrease with the increase in smoking duration and temperature. However, at 5 h, the content of protein-bound HAs slightly increased to 984.2 ng·g−1. Meanwhile, at 90 °C, it increased to 1643.53 ng·g−1. Additionally, a total of 16 volatile organic compounds (VOCs) were found in all of the meat samples, of which 10 VOCs (one acid, three aldehydes and seven phenols) were significantly related to the formation of free HAs. These findings showed that all the different types of HAs were produced under low-temperature processing, which provided scientific insights into the potential generation of HAs during meat smoking processes and could be used as a reference to minimize the risks of cancer related to the consumption of smoked meat products.

The identification of damage based on Lamb waves can hardly avoid obtaining the reference signal under healthy conditions. A non-reference damage localization (NRDL) method is proposed in this paper. The NRDL method is established by the improved two-arrival-time difference method (2/ATDM) and BFGS method. The layout principles of the piezoelectric ceramic lead zirconate titanate (PZT) transducer array in the specimen are proposed. In contrast to existing methods, the damage outside the array in the specimen is identified based on the NRDL. The full-area damage location in the specimen is realized. Furthermore, the optimization of the excitation signal center frequency and transducer array layout is carried out. The damage location accuracy is greatly improved.

Sulfonylureas, a family of anti-diabetic drugs widely used in the clinical treatment of type 2 diabetes, have recently emerged as an illegal adulterant in functional foods, to enhance the claimed anti-diabetic activity. To establish a screening assay method against their adulteration, with the aid of molecular simulation of hapten, two antibodies were raised and complementarily used to enhance the broad-specificity of an enzyme-linked immunosorbent assay (ELISA), which demonstrated simultaneous detection capability to 6 sulfonylureas; the detection limits ranged from 0.02 to 1.0 ng/mL, and recoveries were between 78.3% to 104.5%. Liquid chromatography with tandem mass spectrometry (LC-MS/MS) confirmed the reliability of the proposed ELISA, based on real samples. These results suggest that the proposed ELISA could be an ideal method for screening to monitor for illicit adulteration of sulfonylureas in functional pill products.

In this study, dehydroepiandrosterone (DHEA) hapten and high sensitivity polyclonal antibody against DHEA were prepared, a competitive indirect enzyme-linked immunosorbent assay (ciELISA) and lateral flow immunochromatography (LFA) for the determination of DHEA in slimming products were developed for the first time. The sample pretreatment was very simple and fast, the limit of detection (LOD) of the ciELISA for DHEA in slimming products was 9.6 μg/kg. The average recoveries of slimming tea, slimming tablets and slimming capsules ranged from 93.2% to 99.4%, 86.8% to 100.0%, and 80.5% to 115.4%, respectively, with the coefficient of variations (CVs) between 7.7% and 14.1%, 2.7% and 11.0%, and 2.2% and 8.6%, respectively. The pAb had negligible cross-reactivity to the structural and functional analogs of DHEA. Simultaneously, the LFA showed a cut-off value of 500 μg/kg in slimming products, and the results could be attained within 3 min. A parallel HPLC analysis in 30 commercial slimming products validated a good agreement with the proposed ciELISA and LFA.

The demands for carbon fiber reinforced composites (CFRCs) are growing in the aviation industry for fuel consumption savings, despite the increasing risk of electromagnetic interference (EMI). In this work, polyacrylonitrile (PAN) sheets were prepared by electrospinning. Carbon nanofiber (CNF) sheets were obtained by the carbonization of PAN sheets. The laminate structures of the CF reinforced bismaleimide (BMI)-based composites were specially designed by introducing two thin CNF sheets in the upper and bottom plies, according to EMI shielding theory. The results showed that the introduction of CNF sheets led to a substantial increase in the EMI shielding effectiveness (SE) by 35.0% compared with CFRCs free of CNF sheets. The dominant EMI shielding mechanism was reflection. Noticeably, the introduction of CNF sheets did not impact the interlaminar shear strength (ILSS) of CFRCs, indicating that the strategy provided in this work was feasible for fabricating CFRCs with a high EMI shielding performance without sacrificing their mechanical properties. Therefore, the satisfactory EMI shielding and ILSS properties, coupled with a high service temperature, made BMI-based composites a promising candidate in some specific fields, such as high-speed aircrafts and missiles.

This work concentrated on the interlaminar mechanical properties and toughening mechanism of carbon fiber-reinforced bismaleimide resin (CF/BMI) composites modified by polyacrylonitrile (PAN) nanofiber films. The PAN nanofiber films were prepared by electrospinning. End-notched flexure (ENF) and short-beam strength tests were conducted to assess the mode II fracture toughness (GIIc) and interlaminar shear strength (ILSS). The results showed that the GIIc and ILSS of PAN-modified specimens are 1900.4 J/m2 and 93.1 MPa, which was 21.4% and 5.4% higher than that of the virgin specimens (1565.5 J/m2 and 88.3 MPa), respectively. The scanning electron microscopy (SEM) images of the fracture surface revealed that the PAN nanofiber films toughen the composite on two scales. On the mesoscopic scale, the composite laminates modified by PAN formed a resin-rich layer with high strength and toughness, which made the crack propagate across the layers. At the microscopic scale, the crack propagation between two-dimensional nanofiber films led to constant pull-out and breakage of the nanofibers. As a result, the interlaminar fracture toughness of the composite laminates improved.

Friction is one of the important factors in sheet metal forming. It greatly affects dynamic behaviors of metal sheets and stress and strain distributions in the metal sheets. In this study, deformation characteristics, stress–strain distribution, and change law of symmetrical parts in the process of deep drawing are analyzed using a new theoretical model based on the plastic flow law and partitioning the forming area. In the model, the least-square method is used to linearize the friction coefficient in nonlinear problems and reverse the calculation of friction coefficients to interpret the friction coefficient. To evaluate the model, the friction coefficient in sheet metal drawing of axis-symmetric deep drawing parts under various friction conditions was measured using a self-developed measuring system. The comparison between the experimental results and the calculation using the model shows a good agreement. The results show that the drawing force increases with the increase in punch depth; the friction coefficient decreases with the rise in punch depth. The friction coefficient obtained by fitting is relatively stable, and the average error is less than 3%. Using the friction coefficient model in finite element simulation analysis, it shows that the thickness and blank shape errors are less than 5%. The novel method studied in this paper shows great significance in support for theoretical research, numerical simulation research, and sheet metal stamping performance evaluation.

In wind tunnel tests, the cantilever sting is usually used to support aircraft models because of its simple structure and low aerodynamic interference. However, in some special conditions, big-amplitude and low-frequency vibration would occur easily on the model not only in the pitch direction but also in the yaw direction, resulting in inaccurate data and even damage of the supporting structure. In this paper, aiming at suppressing the vibration in pitch and yaw plane, a multidimensional system identification and active vibration control system on the basis of piezoelectric actuators is established. A vibration monitoring method based on the strain-displacement transformation (SDT) matrix is proposed, which can transform strain signals into vibration displacements. The system identification based on chirp-Z transform (CZT) is applied to improve the adaptability and precision of the building process for the system model. After that, the hardware platform as well as the software control system based on the classical proportional-derivative (PD) algorithm is built. A series of experiments are carried out, and the results show the exactness of the vibration monitoring method. The system identification process is completed, and the controller is designed. Vibration control experiments verify the effectiveness of the controller, and the results indicate that vibrations in pitch and yaw directions are attenuated apparently. The spectrum power is reduced over 14.8 dB/Hz, which prove that the multidimensional identification and active vibration control system has the capability to decline vibration from different directions.

Propiconazole (PCZ) is a fungicide popularly used to prevent and control wheat and rice bakanae disease, etc. However, it was recently found to be illegally employed as a plant regulator to induce thick stems and dark green leaves of Brassica campestris, a famous vegetable in Guangdong, South China. Due to a lack of available recognition molecules to the target analyte, it is still a big challenge to establish a rapid surveillance screening method. In this study, a novel chiral hapten was rationally designed, and an artificial immunogen was then prepared for the generation of a specific antibody against propiconazole for the first time. Using the obtained antibody, a highly sensitive time-resolved fluorescence microspheres lateral flow immunochromatographic assay (TRFMs-LFIA) was established with a visual limit of detection of 100 ng/mL and a quantitative limit of detection of 1.92 ng/mL for propiconazole. TRFMs-LFIA also exhibited good recoveries ranging from 78.6% to 110.7% with coefficients of variation below 16%. The analysis of blind real-life samples showed a good agreement with results obtained using HPLC-MS/MS. Therefore, the proposed method could be used as an ideal screening surveillance tool for the detection of propiconazole in vegetables.

In order to alleviate the problems of complex structure and low reliability of traditional Shape Memory Alloy (SMA) rotary actuator, a planar vortex actuator (PVA) based on SMA material was proposed to directly output torque and angular displacement. Based on the calculation method of PVA and the constitutive model of the phase transition equation of SMA, the mechanical model is established, and the pre-tightening torque, temperature, output torque, and rotation angle are obtained. The relationship expression between the tests has verified the mechanical model. The results show that the relationship between the excitation temperature and the output torque, the coefficient of determination between the calculated value and the tested value, is 0.938, the minimum error is 0.46%, and the maximum error is 49.8%. In the relationship between angular displacement and torque, the coefficient of determination between the calculated value and the test value is 0.939, the maximum error is 58.5%, and the minimum error is 28.0%. The test results show that the calculated values of mechanical model and experimental data have similar representation form.

In recent years, furosemide has been found to be abused in slimming health foods. There is an urgent need for a simpler, faster method for detecting furosemide in slimming health foods. In this study, a rapid, convenient and sensitive lateral flow immunochromatography (LFIA) based on Au nanoparticles (AuNPs) was established for the first time. Under optimal conditions, the qualitative limit of detection (LOD) of the AuNPs-based LFIA was 1.0~1.2 μg/g in slimming health foods with different substrates. AuNPs-LFIA could specifically detect furosemide within 12 min (including sample pretreatment) and be read by the naked eye. The developed AuNPs-LFIA showed high consistency with liquid chromatography with tandem mass spectrometry (LC-MS/MS), and no false positive or false negative results were found in spiked slimming health foods, proving that the AuNPs-LFIA should be accurate and reliable. The AuNPs-LFIA reported here provides a serviceable analytical tool for the on-site detection and rapid initial screening of furosemide for the first time.

Aflatoxin B1 (AFB1), a mycotoxin, is hepatotoxic, carcinogenic, and nephrotoxic in humans and animals, and contaminate a wide range of maize. In this study, an immunochromatographic assay (ICA) based on polystyrene microspheres (PMs) was developed for sensitive and quantitative detection of AFB1 in maize. The amounts of PMs, the condition for activating carboxyl groups of PMs, the amount of monoclonal antibody (mAb), and the volume of the immune probe were optimized to enhance the performance PMs-ICA for point-of-care testing of AFB1 in maize. The PMs-ICA showed the cut-off value of 1 ng/mL in phosphate buffer (PB) and 6 µg/kg in maize samples, respectively. The quantitative limit of detection (qLOD) was 0.27 and 1.43 µg/kg in PB and maize samples, respectively. The accuracy and precision of the PMs-ICA were evaluated by analysis of spiked maize samples with recoveries of 96.0% to 107.6% with coefficients of variation below 10%. In addition, the reliability of PMs-ICA was confirmed by the liquid chromatography-tandem mass spectrometry method. The results indicated that the PMs-ICA could be used as a sensitive, simple, rapid point-of-care testing of AFB1 in maize.

Abstract Background Bone marrow derived stem cells (BMSCs) have the potential to differentiate into cardiomyocytes, but the rate of differentiation is low and the mechanism of differentiation is unclear completely. Here, we aimed to investigate the role of miR1-2 in differentiation of mouse BMSCs into cardiomyocyte-like cells and reveal the involved signaling pathways in the procedure. Methods Mouse BMSCs were treated with miR1-2 and 5-azacytine (5-aza). The expression of cardiac cell markers: NKx2.5, cTnI and GATA4 in BMSCs were examined by qPCR. The apoptosis rate was detected by flow cytometry and the activity of the Wnt/β-catenin signaling pathway was evaluated by measuring the upstream protein of this signaling pathway. Results After over-expression of miR1-2 in mouse BMSCs, the apoptosis rate was significantly lower than the 5-aza group, while the expressions of cardiac-specific genes: such as Nkx2.5, cTnI and GATA4 were significantly increased compared to the control group and the 5-aza group. Meanwhile, over-expression of miR1-2 in mouse BMSCs enhanced the expression of wnt11, JNK, β-catenin and TCF in the Wnt/β-catenin signaling pathway. Use of LGK-974, an inhibitor of Wnt/β-catenin signaling pathway, significantly reduced the expression of cardiac-specific genes and partially blocked the role of the miR1-2. Conclusion Over-expression of miR1-2 in mouse BMSCs can induce them toward promoted cardiomyocyte differentiation via the activation of the Wnt/β-catenin signaling pathway. Compared to 5-aza, miR1-2 can induce differentiation of BMSCs into cardiomyocytes more effectively with a less cytotoxicity.

Carbon dynamics in forests and in particular in soils are of primary importance in the context of climate change. A better understanding of the drivers controlling carbon storage is needed to improve climate mitigation strategies. Carbon storage is the result of a balance between inputs and outputs. Carbon inputs in the soil come from plant residues and root exudates, which are further transformed by microorganisms and stored in the long term. Here, we measured litter and fine-root production in three mixed forests dominated by Pinus koraiensis along a latitudinal gradient and performed a litterbag experiment to better understand the driving factors of decomposition. We found that over the three sites litter production was controlled by climatic factors, soil properties, and forest stand characteristics, whereas decay rates were mainly controlled by microbial community structure and soil stoichiometry. For fine roots, production differed among sites, and higher production was consistently observed in the top soil layers compared to deep soil, although the root distribution along the soil profile differed among sites. Fine-root decay rates were mainly controlled by fine-root stoichiometric characteristics. This article emphasizes the complexity of fine-root dynamics even for a single species. Environmental drivers impact on both production and decay, and we suggest performing manipulative field experiments to better identify the mechanisms involved in soil carbon cycling.

In this paper, a thermo-responsive shape memory effect in a polyvinyl chloride thermoset foam is characterized. Excellent shape recovery performance is observed in foam samples programmed both at room temperature and above their transition temperature. The conversion of porous structures in the foam from closed-cell to open-cell after a shape memory effect cycle is revealed via a series of specially designed oil-dripping experiments and optical images of the micro pores. Programming the strain higher than 20% results in an apparent increase in open-cell level, whereas programming temperatures have almost no influence.

In wind tunnel tests, cantilever stings are often used as model-mount in order to reduce flow interference on experimental data. In this case, however, large-amplitude vibration and low-frequency vibration are easily produced on the system, which indicates the potential hazards of gaining inaccurate data and even damaging the structure. This paper details three algorithms, respectively, Classical PD Algorithm, Artificial Neural Network PID (NNPID), and Linear Quadratic Regulator (LQR) Optimal Control Algorithm, which can realize active vibration control of sting used in wind tunnel. The hardware platform of the first-order vibration damping system based on piezoelectric structure is set up and the real-time control software is designed to verify the feasibility and practicability of the algorithms. While the PD algorithm is the most common method in engineering, the results show that all the algorithms can achieve the purpose of over 80% reduction, and the last two algorithms perform even better. Besides, self-tuning is realized in NNPID, and with the help of the Observer/Kalman Filter Identification (OKID), LQR optimal control algorithm can make the control effort as small as possible. The paper proves the superiority of NNPID and LQR algorithms and can be an available reference for vibration control of wind tunnel system.

Pain hypersensitivity is the most common category of chronic pain and is difficult to cure. Oxidative stress and certain cells apoptosis, such as dorsal root ganglion (DRG) neurons, play an essential role in the induction and development of pain hypersensitivity. The focus of this study is at a more specific molecular level. We investigated the role of advanced oxidative protein products (AOPPs) in inducing hypersensitivity and the cellular mechanism underlying the proapoptotic effect of AOPPs. Normal rats were injected by AOPPs-Rat serum albumin (AOPPs–RSA) to cause pain hypersensitivity. Primary cultured DRG neurons were treated with increasing concentrations of AOPPs–RSA or for increasing time durations. The MTT, flow cytometry and western blot analyses were performed in the DRG neurons. A loss of mitochondrial membrane potential (MMP) and an increase in intracellular reactive oxygen species (ROS) were observed. We found that AOPPs triggered DRG neurons apoptosis and MMP loss. After AOPPs treatment, intracellular ROS generation increased in a time- and dose-dependent manner, whereas, N-acetyl-L-cysteine (NAC), a specific ROS scavenger could inhibit the ROS generation. Proapoptotic proteins, such as Bax, caspase 9/caspase 3, and PARP-1 were activated, whereas anti-apoptotic Bcl-2 protein was down-regulated. AOPPs also increased Nox4 and JNK expression. Taken together, these findings suggest that AOPPs cause pain hypersensitivity in rats, and extracellular AOPPs accumulation triggered Nox4-dependent ROS production, which activated JNK, and induced DRG neurons apoptosis by activating caspase 3 and PARP-1.

The seeds of terrestrial flowering plants often start their life cycle in subterranean darkness. To protect the fragile apical meristematic tissues and cotyledons from mechanical injuries during soil penetration, dicotyledonous seedlings form an elegant apical hook at the top of the hypocotyl. The apical hook has been considered as an adaption structure to the subterranean environment. However, the role of the apical hook in seedling emergence and the molecular mechanism of apical hook formation under real-life conditions remain highly speculative. Here, we find that HOOKLESS 1 (HLS1), a critical gene in apical hook formation in Arabidopsis thaliana, is required for seedling emergence from the soil. When grown under soil, hls1 mutant exhibits severe emergence defects. By contrast, HLS1 overexpression in the hls1 background fully restores emergence defects and displays better emergence capacity than that of WT. Our results indicate that HLS1 transcription is stimulated in response to the mechanical stress of soil cover, which is dependent on the function of the transcription factors ETHYLENE INSENTIVE 3 (EIN3) and EIN3-LIKE 1 (EIL1). Soil-conferred mechanical stress activates the ethylene signaling pathway to stabilize EIN3 by repressing the activity of the F-box proteins EBF1 and EBF2. These combined results reveal a signaling pathway in which plant seedlings transduce the mechanical pressure of soil covering to regulate apical hook formation accordingly for emerging from the soil.
