Abstract Objectives Innate immunity plays a significant role in the pathogenesis of temporomandibular joint osteoarthritis (TMJOA), which is characterized by synovial inflammation and condylar cartilage degradation. We are urged to investigate the impact of Resatorvid, a preventative drug that inhibits Toll-like receptor 4 (TLR4), on experimental inflammatory TMJOA pathology. Methods An intra-articular injection of complete Freund’s adjuvant (CFA) was used to induce an experimental inflammatory mouse TMJOA model, and TLR4 expression was identified by immunofluorescent labeling. Intraperitoneal injections of Resatorvid were administered to CFA-induced TMJOA mice, and the pathology of TMJOA animals with and without Resatorvid treatment was examined by H&E, Safranin-O/Fast Green, and TRAP staining, as well as micro-CT, immunohistochemistry, and immunofluorescence. The impact of Resatorvid on chondrocyte pyroptosis and macrophage inflammation was further investigated using ATDC5 chondrocytes and RAW264.7 macrophages pretreated with relevant antagonists. Results CFA-induced TMJOA mice revealed remarkable synovial inflammation, together with a time course of cartilage degradation and bone destruction, with TLR4 elevated in the synovium and condylar cartilage. Prophylactic treatment with Resatorvid mitigated synovial inflammation, cartilage degeneration, and bone destruction in CFA-induced TMJOA mice and downregulated MyD88/NF-κB expression. Ex vivo studies demonstrated that Resatorvid treatment alleviated NOD-like receptor protein 3 (NLRP3)-mediated chondrocyte pyroptosis and degeneration and relieved macrophage inflammation by preventing reactive oxygen species (ROS) production through NLRP3 signaling. Conclusion Prophylactic treatment with Resatorvid alleviates TMJOA pathology by inhibiting chondrocyte pyroptosis and degeneration, as well as ROS-induced macrophage inflammation, through TLR4/MyD88/NF-κB/NLRP3.

To address the issues of the low usage efficiency and illogical structure in rural regions, this study builds a rural integrated energy system (RIES) that incorporates the complementary use of biogas, wind, and light. For resolving the RIES optimum-low-carbon-economic-dispatch problem, a source–load-cooperative optimal-dispatch strategy is proposed. Firstly, a multi-energy integrated demand response (IDR) model based on time-of-use tariffs and time-varying biogas costs is established on the demand side. Secondly, power-to-gas devices are added on the supply side to optimize the system’s electricity–gas-coupling relationship and increase the wind power output space. Thirdly, an RIES-oriented carbon-trading model is constructed by considering the actual carbon emissions of gas loads and the stepped-carbon-trading mechanism. Finally, an optimal-dispatch model is built with the objective function of reducing the total energy cost, wind abandonment cost, IDR cost, and carbon emission cost, while the problem is transformed into a mixed-integer linear problem and solved using CPLEX 12.9. By setting up four scenarios for example analysis, the results show that on typical days in spring, summer, autumn, and winter, the total operating costs of the stepped-carbon-trading system (Scenario 1), taking into account the source-side power-to-gas (P2G) device and the load-side IDR, are reduced by 12.25%, 11.25%, 12.42%, and 11.56%, respectively, compared to the system without the introduction of the IDR (Scenario 3). In contrast to the system that lacks a P2G device at the source end (Scenario 2), the overall costs are decreased by 4.97%, 3.07%, 5.02%, and 5.36%, but the wind power consumption rates are increased by 11.63%, 7.93%, 11.54%, and 11.65%, respectively. Stepped emission trading (Scenario 1) reduces the total operating costs by 5.12%, 3.15%, 5.21%, and 6.84%, respectively, while reducing the biogas costs by 9.75%, 7.74%, 9.67%, and 9.57%, respectively, in comparison to traditional emission trading (Scenario 4). The example results demonstrate the economics, effectiveness, and reliability of a stepped-carbon-trading system with an integrated P2G load-side energy demand response.

Conclusion: This study suggests that sensitization of small- to medium-sized Dil+ TG neurons and CGRP- and IB4-positive Dil+ TG neurons might contribute to the development of TMJOA chronic pain in rats. This will provide valuable information for more efficient control of TMJOA chronic pain.

Photoacoustic imaging (PAI) uniquely combines optics and ultrasound, presenting a promising role in biomedical imaging as a non-invasive and label-free imaging technology. As the traditional opaque ultrasound (US) transducers could hinder the transportation of the excitation light and limit the performance of PAI system, piezoelectric transparent ultrasonic transducers (TUTs) with indium tin oxide (ITO) electrodes have been developed to allow light transmission through the transducer and illuminate the sample directly. Nevertheless, without having transparent matching materials with appropriate properties, the bandwidth of those TUTs was generally narrow. In this work, we propose to employ polymethyl methacrylate (PMMA) as the matching layer material to improve the bandwidth of lithium niobate (LN)-based TUTs. The effects of PMMA matching layer on the performance of TUTs have been systematically studied. With the optimized PMMA matching layer, the very wide bandwidth of > 50 % could be achieved for the TUTs even with different transducer frequencies, leading to the great enhancement of axial resolution when compared to the similar reported work. In addition, the imaging performance of the developed TUT prototype has been evaluated in a PAI system and demonstrated by both phantom and in vivo small animal imaging.

Osteoarthritis (OA) and rheumatoid arthritis (RA) are chronic and progressive inflammatory joint diseases that affect a large population worldwide. Intra-articular administration of various therapeutics is applied to alleviate pain, prevent further progression, and promote cartilage regeneration and bone remodeling in both OA and RA. However, the effectiveness of intra-articular injection with traditional drugs is uncertain and controversial due to issues such as rapid drug clearance and the barrier afforded by the dense structure of cartilage. Nanoparticles can improve the efficacy of intra-articular injection by facilitating controlled drug release, prolonged retention time, and enhanced penetration into joint tissue. This review systematically summarizes nanoparticle-based therapies for OA and RA management. Firstly, we explore the interaction between nanoparticles and joints, including articular fluids and cells. This is followed by a comprehensive analysis of current nanoparticles designed for OA/RA, divided into two categories based on therapeutic mechanisms: direct therapeutic nanoparticles and nanoparticles-based drug delivery systems. We highlight nanoparticle design for tissue/cell targeting and controlled drug release before discussing challenges of nanoparticle-based therapies for efficient OA and RA treatment and their future clinical translation. We anticipate that rationally designed local injection of nanoparticles will be more effective, convenient, and safer than the current therapeutic approach.

Abstract Subchondral bone degeneration is the main pathological change during temporomandibular joint (TMJ) osteoarthritis (OA) development. Netrin-1, an axon-guiding factor, might play roles in OA development and pain. The purpose of this study was to investigate the expression of Netrin-1 in TMJ OA and its possible role in the progression of TMJ OA and pain. The synovial fluids of temporomandibular joint disorders (TMDs) patients were collected for Netrin-1 by enzyme linked immunosorbent assay (ELISA). TMJ OA model was built by MIA joint injection, and then the von Frey test, hematoxylin & eosin (H&E) staining, toluidine blue (TB) staining, immunohistochemical (IHC) staining and micro-CT were performed. After induction of osteoclast differentiation of raw264.7 cells, immunofluorescence (IF) was used to detect the Netrin-1 and its receptors on osteoclast membrane. The concentration of Netrin-1 increased in the synovial fluid of TMJ OA patients. After MIA injection to TMJ, the head withdrawal threshold (HWT) was significantly decreased. Microscopically, the structural disorder of subchondral bone was the most obvious at the 2nd week after MIA injection. In addition, Netrin-1 expression increased in the subchondral bone at the 2nd week after MIA injection. In vitro, the expressions of Netrin-1 and its receptor Unc5B were upregulated on the osteoclast membrane. Netrin-1 might be an important regulator during bone degeneration and pain in the process of TMJ OA.

The use of fertilizer is closely related to crop growth and environmental protection in agricultural production. It is of great significance to develop environmentally friendly and biodegradable bio-based slow-release fertilizers. In this work, porous hemicellulose-based hydrogels were created, which had excellent mechanical properties, water retention properties (the water retention ratio in soil was 93.8% after 5 d), antioxidant properties (76.76%), and UV resistance (92.2%). This improves the efficiency and potential of its application in soil. In addition, electrostatic interaction and coating with sodium alginate produced a stable core–shell structure. The slow release of urea was realized. The cumulative release ratio of urea after 12 h was 27.42% and 11.38%, and the release kinetic constants were 0.0973 and 0.0288, in aqueous solution and soil, respectively. The sustained release results demonstrated that urea diffusion in aqueous solution followed the Korsmeyer–Peppas model, indicating the Fick diffusion mechanism, whereas diffusion in soil adhered to the Higuchi model. The outcomes show that urea release ratio may be successfully slowed down by hemicellulose hydrogels with high water retention ability. This provides a new method for the application of lignocellulosic biomass in agricultural slow-release fertilizer.

HMGB1 is a highly conserved nuclear protein that is rapidly released into the extracellular environment during infection or tissue damage. In osteoarthritis, HMGB1 acts as a pro-inflammatory cytokine inducing a positive feedback loop for synovial inflammation and cartilage degradation. The aim of this study was to explore the role of HMGB1 in inflammation and catabolism of temporomandibular joint osteoarthritis (TMJOA) and whether inhibition of HMGB1 affects TMJOA. Human synovial fibroblasts were incubated with HMGB1, the expression of pro-inflammatory cytokines and catabolic mediators were measured by Western blot and ELISA. NF-κB signaling pathway involvement was studied by the NF-κB inhibitor and detected by Western blotting and immunofluorescence staining. TMJOA was induced by an injection of Complete Freund’s adjuvant (CFA) into anterosuperior compartment of rat’s joint. An anti-HMGB1 antibody was used to assess the effect to HMGB1 in the synovium and cartilage of the CFA-induced TMJOA rats by H&E, Safranin O, Masson trichrome staining, immunohistochemistry and immunofluorescence. HMGB1 markedly increased the production of MMP13, ADAMTS5, IL-1β and IL-6 through activating NF-κB signaling pathway in human synovial fibroblasts. In vivo, application of the HMGB1 neutralizing antibody effectively ameliorated the detrimental extent of TMJOA. Furthermore, the HMGB1 neutralizing antibody reduced the expression of NF-κB, pro-inflammatory cytokines and catabolic mediators in the synovium and cartilage of CFA-induced TMJOA rats. HMGB1 inhibition alleviates TMJOA by reducing synovial inflammation and cartilage catabolism possibly through suppressing the NF-κB signaling pathway and may become a therapeutic method against TMJOA.

Abstract The slotless tubular permanent magnet linear motor (STPMLM) offers obvious advantages when it is applied to the punching system, such as space saving and controllable impact strength. Herein, the mechanical and electromagnetic coupled differential equations were established to present a comprehensive analysis of the relationship between mover mass and motor performance in the punching system. The travel time of the punching movement was expressed analytically under the condition of the mover obtaining the same impact kinetic energy, and the relationship between the mover mass and the resistive energy consumption in one punching process was also deduced under the same condition. Then the analytical results were validated by a numerical model. The studies show that a lighter mover mass can contribute to less resistive energy consumption and faster dynamic response in the punching applications. Based on that, a STPMLM with special‐shaped yoke was designed for lighter mover mass. Moreover, prototype experiments including static thrust characteristic and dynamic performance were conducted. The experimental results proved that the proposed permanent magnet linear motor with special‐shaped yoke can improve the dynamic performance and reduce resistive energy consumption while ensuring that the electromagnetic performance is not affected.
