Edible bird’s nest (EBN), a most highly priced and valuable foodstuff, contains high percentage of proteins and carbohydrates. However, proteins adhering to these carbohydrates make the EBN hard and tough, which need to be boiled as the bird’s nest soup to make the Chinese cuisine. To overcome the hard and tough texture of EBN and improve the digestion degrees, the present study screened and identified a probiotic strain Bacillus amyloliquefaciens YZW02 from 5-year stored EBN sample completely solubilizing EBN for the first time. The 24-h B. amyloliquefaciens fermented EBN contained 20.30–21.48 mg/mL of the soluble protein contents with a recovery rate of 98–100%, DPPH radical scavenging rate of 84.76% and ABTS radical scavenging capacity of 41.05%. The mixed fermentation of B. amyloliquefaciens YZW02 and Bacillus natto BN1 were further applied to improve the low-MW peptide percentages and antioxidant activities. The mixed-fermentation of B. natto BN1 with 4-h cultured B. amyloliquefaciens YZW02 had the lowest percentage (82.23%) of >12-kDa proteins/peptides and highest percentages of 3–12 kDa, 1–3 kDa and 0.1–1 kDa peptides of 8.6% ± 0.08, 7.57% ± 0.09, 1.77% ± 0.05 and 0.73% ± 0.05, with the highest DPPH, ABTS and •OH scavenging capacity of 90.23%, 46.45% and 49.12%, respectively. Cotton (Gossypium spp.) is a vital global source of renewable fiber and ranks among the world's most important cash crops. While extensive nuclear genomic data of Gossypium has been explored, the organellar genomic resources of allotetraploid cotton, remain largely untapped at the population level. The plastid genome (plastome) is well suited for studying plant species relationships and diversity due to its nonrecombinant uniparental inheritance. Here, we conducted de novo assembly of 336 Gossypium plastomes, mainly from domesticated cultivars, and generated a pan‐plastome level resource for population structure and genetic diversity analyses. The assembled plastomes exhibited a typical quadripartite structure and varied in length from 160 103 to 160 597 bp. At the species level, seven allotetraploid species were resolved into three clades, where Gossypium tomentosum and Gossypium mustelinum formed an early diverging clade rooted by diploids, followed by splitting two sister clades of Gossypium darwinii–Gossypium barbadense and Gossypium hirsutum–Gossypium ekmanianum–Gossypium stephensii. Within the G. hirsutum clade the resolution of cultivated accessions was less polyphyletic with landrace and wild accessions than in G. barbadense suggesting some selection on plastome in the domestication of this adaptable species of cotton. The nucleotide diversity of G. hirsutum was higher than that of G. barbadense. We specifically compared the plastomes of G. hirsutum and G. barbadense to find mutational hotspots within each species as potential molecular markers. These findings contribute a valuable resource for exploring cotton evolution as well as in the breeding of new cotton cultivars and the preservation of wild and cultivated germplasm. The prevention and treatment of gastric cancer has been the focus and difficulty of medical research. We aimed to explore the mechanism of inhibiting migration and invasion of gastric cancer cells by methionine restriction (MR). The human gastric cancer cell lines AGS and MKN45 cultured with complete medium (CM) or medium without methionine were used for in vitro experiments. MKN45 cells were injected tail vein into BALB/c nude mice and then fed with normal diet or methionine diet for in vivo experiments. MR treatment decreased cell migration and invasion, increased E-cadherin expression, decreased N-cadherin and p-p65 expressions, and inhibited nuclear p65 translocation of AGS and MKN45 cells when compared with CM group. MR treatment increased IκBα protein expression and protein stability, and decreased IκBα protein ubiquitination level and TRIM47 expression. TRIM47 interacted with IκBα protein, and overexpression of TRIM47 reversed the regulatory effects of MR. TRIM47 promoted lung metastasis formation and partially attenuated the effect of MR on metastasis formation in vivo compared to normal diet group mice. MR reduces TRIM47 expression, leads to the degradation of IκBα, and then inhibits the translocation of nuclear p65 and the migration and invasion of gastric cancer cells. [ABSTRACT FROM AUTHOR]

Head injuries induced by tennis ball impacts are computationally studied. The impact of a two-piece tennis ball on a human head is simulated by using an established full body model and a newly constructed tennis ball model. The new tennis ball model is validated against existing experimental data. The frontal impact of a tennis ball on a human head at a velocity of 25 m/s is first studied as the baseline case. The effects of the impact location, velocity, and angle as well as the ball spinning are then examined. It is revealed that the lateral impact results in a higher risk of head injury than the frontal and crown impacts. In addition, it is found that the impact force and von Mises stress in the skull, the intracranial pressure and first principal strain in the brain, and the translational and rotational accelerations at the center of gravity of the head all increase with the increase of the impact velocity. Moreover, the normal (90-deg) impact has the highest risk of head injury, which is followed by the 60-deg, 45-deg and 30-deg impacts. Further, it is observed that the spinning of the tennis ball has insignificant effects on the head response. The simulation results show that there will be no skull fracture or mild brain injury in the baseline case. However, traumatic brain injuries may occur after the impact velocity exceeds 40 m/s. The propensity to metastasize is the most important prognostic indicator for solid cancers. New insights into the mechanisms of early carcinogenesis have revealed micrometastases are generated far earlier than previously thought. Evidence supports a synergistic relationship between vascular and lymphatic seeding which can occur before there is clinical evidence of a primary tumour. Early vascular seeding prepares distal sites for colonisation while regional lymphatics are co‐opted to promote facilitative cancer cell mutations. In response, the host mounts a global inflammatory and immunomodulatory response towards these cells supporting the concept that cancer is a systemic disease. Cancer staging systems should be refined to better reflect cancer cell loads in various tissue compartments while clinical perspectives should be broadened to encompass this view when approaching high‐risk cancers. Measured adjunctive therapies implemented earlier for low‐volume, in‐transit cancer offers the prospect of preventing advanced disease and the need for heroic therapeutic interventions. This review seeks to re‐appraise how we view the metastatic process for solid cancers. It will explore in‐transit metastasis in the context of high‐risk skin cancer and how it dictates disease progression. Unintended end-of-process depression (EOPD) commonly occurs in laser powder bed fusion (LPBF), leading to poor surface quality and lower fatigue strength, especially for many implants. In this study, a high-fidelity multi-physics meso-scale simulation model is developed to uncover the forming mechanism of this defect. A defect-process map of the EOPD phenomenon is obtained using this simulation model. It is found that the EOPD formation mechanisms are different under distinct regions of process parameters. At low scanning speeds in keyhole mode, the long-lasting recoil pressure and the large temperature gradient easily induce EOPD. While at high scanning speeds in keyhole mode, the shallow molten pool morphology and the large solidification rate allow the keyhole to evolve into an EOPD quickly. Nevertheless, in the conduction mode, the Marangoni effects along with a faster solidification rate induce EOPD. Finally, a 'step' variable power strategy is proposed to optimise the EOPD defects for the case with high volumetric energy density at low scanning speeds. Purpose: Dynamic PET is an essential tool in oncology due to its ability to visualize and quantify radiotracer uptake, which has the potential to improve imaging quality. However, image noise caused by a low photon count in dynamic PET is more significant than in static PET. This study aims to develop a novel denoising method, namely the Guided Block Matching and 4-D Transform Domain Filter (GBM4D) projection, to enhance dynamic PET image reconstruction. Methods: The sinogram was first transformed using the Anscombe method, then denoised using a combination of hard thresholding and Wiener filtering. Each denoising step involved guided block matching and grouping, collaborative filtering, and weighted averaging. The guided block matching was performed on accumulated PET sinograms to prevent mismatching due to low photon counts. The performance of the proposed denoising method (GBM4D) was compared to other methods such as wavelet, total variation, non-local means, and BM3D using computer simulations on the Shepp–Logan and digital brain phantoms. The denoising methods were also applied to real patient data for evaluation. Results: In all phantom studies, GBM4D outperformed other denoising methods in all time frames based on the structural similarity and peak signal-to-noise ratio. Moreover, GBM4D yielded the lowest root mean square error in the time-activity curve of all tissues and produced the highest image quality when applied to real patient data. Conclusion: GBM4D demonstrates excellent denoising and edge-preserving capabilities, as validated through qualitative and quantitative assessments of both temporal and spatial denoising performance. Proactively programming materials toward target nonlinear mechanical behaviors is crucial to realize customizable functions for advanced devices and systems, which arouses persistent explorations for rapid and efficient inverse design strategies. Herein, we propose a "mechanical Fourier transform" strategy to program mechanical behaviors of materials by mimicking the concept of Fourier transform. In this strategy, an arbitrary target force-displacement curve is decomposed into multiple cosine curves and a constant curve, each of which is realized by a rationally designed multistable module in an array-structured metamaterial. Various target curves with distinct shapes can be rapidly programmed and reprogrammed through only amplitude modulation on the modules. Two exemplary metamaterials are demonstrated to validate the strategy with a macroscale prototype based on magnet lattice and a microscale prototype based on an etched silicon wafer. Diverse β-carboline (βC) alkaloids are produced by microbes, plants, and animals with myriad bioactivities and drug potentials. However, the biosynthetic mechanism of βCs remains largely elusive, especially regarding the hydroxyl and glucosyl modifications of βCs. Here, we report the presence of the bacterial-like Pictet–Spenglerase gene Fcs1 in the entomopathogenic Beauveria fungi that can catalyze the biosynthesis of the βC skeleton. The overexpression of Fcs1 in Beauveria bassiana led to the identification of six βC methyl glycosides, termed bassicarbosides (BCSs) A–F. We verified that the cytochrome P450 (CYP) genes adjacent to Fcs1 cannot oxidize βCs. Alternatively, the separated CYP684B2 family gene Fcs2 was identified to catalyze βC hydroxylation together with its cofactor gene Fcs3. The functional homologue of Fcs2 is only present in the Fcs1-containing fungi and highly similar to the Fcs1-connected yet nonfunctional CYP. Both evolved quicker than those from fungi without Fcs1 homologues. Finally, the paired methyl/glucosyl transferase genes were verified to mediate the production of BCSs from hydroxy-βCs. All these functionally verified genes are located on different chromosomes of Beauveria, which is in contrast to the typical content-clustered feature of fungal biosynthetic gene clusters (BGCs). We also found that the production of BCSs selectively contributed to fungal infection of different insect species. Our findings shed light on the biosynthetic mechanism of βC glycosides, including the identification of a βC hydroxylase. An extended Hill's lemma is provided for non-Cauchy continua satisfying the modified couple stress theory in the bulk and the surface elasticity theory in the surface layer. Based on the Hill–Mandel condition, four sets of boundary conditions (BCs) are identified. It is shown that each of these four sets of BCs satisfies the admissibility and average field requirements. In addition, the set of kinetic BCs meets the force and moment balance conditions, and the set of modified kinematic BCs satisfies the displacement and micro-rotation compatibility constraint. By applying the newly extended Hill's lemma, a homogenization analysis of a two-phase composite is performed, in which the average strain energy of the composite is computed using a finite element model constructed using COMSOL. In order to study the variation law of temperature and stress of overlying rock under the thermal solid coupling condition of underground coal gasification (UCG), the coal measure sandstone samples were heated in one direction, the variation law of temperature and thermal stress was analyzed, and the experiment was analyzed using the numerical simulation software COMSOL. The temperature field, stress field, and volume strain distribution of rock mass are obtained. The results show that in the heating stage of the heating plate, the temperature curve of each measuring point changes rapidly and the temperature rises rapidly, and due to the decrease of the heating rate of the heating plate, the change rate of thermal stress tends to decrease. In the circumferential region of the lower end face, stress stretching occurs and the radial strain is the largest. Under the constraint of unidirectional heating, the temperature distribution of rock along the height direction is obvious. The principal stress is mainly concentrated near the lower end face of the specimen. The research results are helpful to study the thermal damage and fracture characteristics of overlying sandstone of UCG from the perspective of thermal solid coupling. [ABSTRACT FROM AUTHOR]

Hybrid rocket engines suffer from the restricted mechanical properties and low regression rates of current polymeric fuel grains. We propose a three-dimensional printed aluminium (Al) nested composite fuel grain with millimetre-scale lattice pores (referred to as Al-L). In this study, breathable Al blades with micrometer-scale interconnected pores (Al-B) and blades combining millimetre-scale and micrometer-scale pores (Al-B&L) are designed. The formation mechanisms, characteristics, and effects of the breathable blades are analysed in simulations, microcomputed tomography, and cyclic compression tests. The mechanical properties of the composite fuel grains are investigated numerically and in compression tests. Al-B has the highest Young's modulus at more than 15 times that of a paraffin-based fuel grain and Al-B&L has the highest yield stress at 4 times that of the paraffin-based fuel grain. Referring to combustion properties, the regression rates of the Al-B and Al-B&L grains are respectively 63.3% and 58.2% greater than the regression rate of the paraffin-based fuel grain. Multifunctional materials are an attractive research area. Organic‐inorganic hybrid perovskites are widely used in the design of these materials due to their rich properties and flexible composition. It is easy to obtain more photoelectric properties by introducing chiral groups as ligands. In this work, we synthesized chiral one‐dimensional organic‐inorganic hybrid perovskites, namely (R/S‐3‐HP)PbBr3 (1R/1S) (3‐HP=3‐hydroxy‐piperidine). The enantiomer compounds undergo reversible phase transition at 349/336 K. Under the excitation light of 339 nm, 1R and 1S have a wide emission peak at 635 nm, showing orange light. In addition, the indirect bandgap is 3.29 eV and the SHG intensity is comparable to that of KDP. This work provides a way to design multifunctional chiral perovskite materials. Objective: Bi-level positive airway pressure (BiPAP) and synchronized intermittent mandatory ventilation (SIMV) can be used to achieve peak inspiratory pressure and positive end-expiratory pressure to avoid alveolar collapse and improve oxygenation in preterm infants during the treatment of respiratory distress syndrome (RDS), and there is an urgent demand for evaluating the effects and prognoses of these two ventilation modes. Study design: We conducted a retrospective study on preterm infants (≤32 weeks and <2500 g) from March 2015 to March 2020 with BiPAP (n = 63) and SIMV (n = 63). The primary outcomes were successful treatment and weaning within 72 h, the demand for a second pulmonary surfactant supply and the need for a second respiratory support. The secondary outcome was the incidence of complications. Results: There were no significant differences (p > .05) in the primary outcomes or the incidence of complications (pneumonia, apnea, respiratory failure, air leak syndrome, persistence of patent ductus arteriosus, neonatal sepsis, necrotizing enterocolitis, retinopathy of prematurity, and intraventricular hemorrhage). There were significant differences (p < .05) in the incidence of pulmonary hemorrhage, bronchopulmonary dysplasia and IVH (grade II). Conclusions: Although both BiPAP and SIMV achieved good early treatment outcomes of RDS in preterm infants, BiPAP support is recommended for reducing the incidence of pulmonary hemorrhage, bronchopulmonary dysplasia and IVH (grade II) if infants are tolerant. Background: Both TP53 mutation and MYC amplification indicate poor outcomes in breast cancer (BC), but the clinical values of concurrent TP53 and MYC alterations have not been well-characterized. Methods: A total of 494 BC patients diagnosed at Guangdong Provincial People's Hospital (GDPH) were retrospectively analyzed. Genomic alterations were determined using next-generation sequencing. Survival analysis was applied to assess the effects of genetic alterations on relapse-free survival. The prognosis was verified based on 1405 patients from METABRIC cohort. Additionally, we used logistic regression to identify the factors associated with pathological complete response (pCR) after neoadjuvant chemotherapy. Results: In GDPH cohort, patients with TP53/MYC co-alteration exhibited higher grade and stage, more positive HER2 status and higher Ki67 levels, but less luminal A subtypes. They also had more mutations in genes involved in ERBB and TGF-β signaling pathways, as well as exclusive FANCG/CDKN2B/QKI copy number amplifications and SUFU/HIST3H3/ERCC4/JUN/BCR mutations. Concurrent TP53 and MYC alterations independently increased hazards of relapse (HR, 5.425; 95% CI: 2.019--14.579; p < 0.001). They maintained independent significance for relapse-free (HR, 1.310; 95% CI: 1.012--1.697; = 0.041) and overall survival (HR, 1.373; 95% CI: 1.093--1.725; p = 0.006) in METABRIC cohort. Among the 81 patients receiving chemotherapy, TP53 mutation (OR, 5.750; 95% CI: 1.553--25.776; p = 0.013) and earlier stage (OR, 0.275; 95% CI 0.088--0.788; p = 0.020) were associated with pCR, while the co-alter- ation did not serve as an independent predictor (p = 0.199). Conclusions: TP53/MYC co-alteration was associated with distinct clinicopathologi- cal and genomic features. They also conferred unfavorable prognosis in BC patients, and did not improve pCR after neoadjuvant chemotherapy. [ABSTRACT FROM AUTHOR]

Kapok fiber (KF), with the characteristics of a natural hollow structure, light weight, and low density, can be used as acoustic and thermal insulation, buoyancy, adsorption, filling, and composite material. The flame-retardant treatment can expand the functionality and application of KF. In this work, the phosphorylation of KF using phytic acid (PA) in the presence of urea at a high temperature was used to enhance its flame retardancy. The phosphorylation reaction conditions were discussed, and the surface topography, thermal degradation, heat release, and combustion properties of phosphorylated KF were studied. The Fourier transform infrared spectroscopy and 31P solid-state nuclear magnetic resonance spectroscopy analyses confirmed the grafting of PA on cellulose by the formation of phosphate ester bonds. Due to the covalent binding of PA, phosphorylated KF exhibited good washing durability. The surface topography, Raman spectroscopy, thermogravimetric (TG), and microcalorimetry analyses revealed the excellent charring ability of phosphorylated KF. In the TG test in nitrogen, the char residue increased to 42.6% of phosphorylated KF from 8.3% of raw KF at 700 °C. In the vertical combustion, raw KF sheet was almost completely burned out within 30 s, while phosphorylated KF was very difficult to catch fire. In the microcalorimetry analysis, the heat release capacity and total heat release of phosphorylated KF decreased to 67 J/g∙K and 3.9 kJ/g, respectively from 237 J/g∙K and 18.1 kJ/g of raw KF. Critical velocities and middle-surface displacements of anisotropic axisymmetric cylindrical shells (tubes) under a uniform internal pressure moving at a constant velocity are derived in closed-form expressions by using the Love–Kirchhoff thin shell theory incorporating the rotary inertia and material anisotropy. The formulation is based on the general three-dimensional constitutive relations for orthotropic elastic materials and provides a unified treatment of orthotropic, transversely isotropic, cubic and isotropic tubes, which can represent various composite and metallic tubes. Closed-form formulas are first obtained for the general case with both the rotary inertia and radial stress effects, which are then reduced to the special cases without the rotary inertia effect and/or radial stress effect. It is shown that when the rotary inertia effect is suppressed and the radial normal stress is neglected, the newly derived formulas for the critical velocities of orthotropic and isotropic tubes recover the two existing ones for thin tubes as special cases. An example for an isotropic tube is provided to illustrate the new formulas, which give the values of the critical velocity and dynamic amplification factor that agree well with those obtained experimentally and computationally by others. Glutamate acts at eight metabotropic glutamate (mGlu) receptor subtypes expressed in a partially overlapping fashion in distinct brain circuits. Recent evidence indicates that specific mGlu receptor protomers can heterodimerize and that these heterodimers can exhibit different pharmacology when compared to their homodimeric counterparts. Group III mGlu agonist-induced suppression of evoked excitatory potentials and induction of long-term potentiation at Schaffer collateral-CA1 (SC-CA1) synapses in the rodent hippocampus can be blocked by the selective mGlu7 negative allosteric modulator (NAM), ADX71743. Curiously, a different mGlu7 NAM, 6-(4-methoxyphenyl)-5-methyl-3-pyridin-4-ylisoxazo nolo[4,5-c]pyridin-4(5H)-one, failed to block these responses in brain slices despite its robust activity at mGlu7 homodimers in vitro. We hypothesized that this might result from heterodimerization of mGlu7 with another mGlu receptor protomer and focused on mGlu8 as a candidate given the reported effects of mGlu8-targeted compounds in the hippocampus. Here, we used complemented donor acceptor-resonance energy transfer to study mGlu7/8 heterodimer activation in vitro and observed that ADX71743 blocked responses of both mGlu7/7 homodimers and mGlu7/8 heterodimers, whereas 6-(4-methoxyphenyl)-5-methyl-3-pyridin-4-ylisoxazonolo[4,5-c]pyridin-4(5H)-one only antagonized responses of mGlu7/7 homodimers. Taken together with our electrophysiology observations, these results suggest that a receptor with pharmacology consistent with an mGlu7/8 heterodimer modulates the activity of SC-CA1 synapses. Building on this hypothesis, we identified two additional structurally related mGlu7 NAMs that also differ in their activity at mGlu7/8 heterodimers, in a manner consistent with their ability to inhibit synaptic transmission and plasticity at SC-CA1. The study of topology in elastic media has been primarily focused on achieving non-trivial topological states in discrete elastic lattices through active or chiral microscopic interactions. Realization of such topological states in continuous elastic media remains largely unexplored. In this study, a new continuum theory of micropolar gyroelasticity is developed and applied to attain non-trivial topological boundary states in elastic continua. According to the new theory, an elastic continuum is composed of elastically interacting micro-volume elements that can translate and rotate and are connected at their mass centers to gyroscopes, which contribute to the linear and orbital angular momenta but not to the spin angular momentum of the continuum. By applying this micropolar gyroelasticity theory to elastic media with both periodic and finite domains, the emergence of topological boundary states in 2D micropolar gyroelastic continua is demonstrated. Through using the Floquet–Bloch method for periodic domains, the bulk-boundary correspondence is analytically established, and the emergence of non-trivial topological bulk states characterized by Mexican-hat band structures is observed. In addition, by employing an asymptotic analytical model based on the extended Bloch theorem and performing numerical analyses of micropolar gyroelastic continua with finite domains of different geometries, it is shown that the non-trivial Mexican-hat band structure is associated with and provides protection for topological boundary states confined at the boundaries. Finally, the application of the newly developed micropolar gyroelasticity theory to Zinc-blende structured materials (including ZnTe, GaP, InP and ZnS) reveals that the emergence of the topological boundary states in an elastic continuum is not triggered solely by the gyroscopic effect but also depends on the material properties of the micropolar continuum. Grain quality has become one of the foci in rice breeding worldwide since it is crucial for farmers, millers and consumers. Previous studies have revealed that rice gain quality is mainly determined with several major characteristics including milling, physical appearance, eating and cooking, sensory and nutritional value. In genetics, rice grain quality is a complex trait controlled by polygene/QTL. Better understanding the mechanism underlying the grain quality is fundamental for developing new breeding strategies. Therefore, abundant studies had been conducted to discover the factors that control the quality traits, and led to dozens of QTL identified and several major genes cloned. This has enabled development of the functional markers to facilitate selection of the complex traits. But breeding for rice varieties with desired grain quality needs to pyramid multiple and beneficial loci or alleles, some of which are still untapped. In this chapter, we first summarize the literatures on rice grain quality traits and the relevant phenotyping approaches, and then review the progress in genetic dissection of these traits. This review also presented the breeding attempts using marker-assisted selection, genomics-assisted breeding approaches, as well as genome editing technologies, and finally provided innovative insights into the future of rice grain quality breeding by using the modern technology with integration of machine learning and genomic selection. RNA-binding protein RBM28 (RBM28), as a nucleolar component of spliceosomal small nuclear ribonucleoproteins, is involved in the nucleolar stress response. Whether and how RBM28 regulates tumor progression remains unclear. Here, we report that RBM28 is frequently overexpressed in various types of cancer and that its upregulation is associated with a poor prognosis. Functional and mechanistic assays revealed that RBM28 promotes the survival and growth of cancer cells by interacting with the DNA-binding domain of tumor suppressor p53 to inhibit p53 transcriptional activity. Upon treatment with chemotherapeutic drugs (e.g., adriamycin), RBM28 is trans-located from the nucleolus to the nucleoplasm, which is likely mediated via phosphorylation of RBM28 at Ser122 by DNA checkpoint kinases 1 and 2 (Chk1/2), indicating that RBM28 may act as a nucleolar stress sensor in response to DNA damage stress. Our findings not only reveal RBM28 as a potential biomarker and therapeutic target for cancers but also provide mechanistic insights into how cancer cells convert stress signals into a cellular response linking the nucleolus to regulation of the tumor suppressor p53. Gastric cancer metastasis is a major cause of poor prognosis. Our previous research showed that methionine restriction (MR) lowers the invasiveness and motility of gastric carcinoma. In this study, we investigated the particular mechanisms of MR on gastric carcinoma metastasis. In vitro, gastric carcinoma cells (AGS, SNU-5, MKN7, KATO III, SNU-1, and MKN45) were grown in an MR medium for 24 h. In vivo, BALB/c mice were given a methionine-free (Met−) diet. Transwell assays were used to investigate cell invasion and migration. The amounts of Krüppel like factor 10 (KLF10) and cystathionine β-synthase (CBS) were determined using quantitative real-time PCR and Western blot. To determine the relationship between KLF10 and CBS, chromatin immunoprecipitation and a dual-luciferase reporter experiment were used. Hematoxylin–eosin staining was used to detect lung metastasis. Liquid chromatography–mass spectrometry was used to determine cystathionine content. MR therapy had varying effects on the invasion and migration of gastric carcinoma cells AGS, SNU-5, MKN7, KATO III, SNU-1, and MKN45. KLF10 was highly expressed in AGS cells but poorly expressed in KATO III cells. KLF10 improved MR's ability to prevent gastric carcinoma cell invasion and migration. In addition, KLF10 may interact with CBS, facilitating transcription. Further detection revealed that inhibiting the KLF10/CBS-mediated trans -sulfur pathway lowered Met−'s inhibitory effect on lung metastasis development. In this research, five new indolequinazoline alkaloids (1 – 5), along with six known indolequinazoline alkaloids (6 – 11) were obtained from the fruits of Tetradium ruticarpum. Their structures were elucidated through comprehensive spectroscopic data of 1D and 2D NMR, HRESIMS and ECD spectra. Additionally, all isolates were assayed for their SIRT1 inhibitory activities in vitro and compounds 2 , 7 , 10 and 11 exhibited activities with IC 50 values ranged from 43.16 to 118.35 μM. Objective: Immune inflammatory cells and cytokines play an important role in the occurrence and development of diabetic nephropathy (DN). Acteoside has been reported to regulate the inflammation and immune response. The study aims to investigate the effects of acteoside on the expressions of MCP-1 and TGF-β1 on nephropathy in diabetic mice. Methods: C57BL/6J mice in the model group were given a single intraperitoneal injection of STZ (150 mg/kg). Model mice were divided randomly into two groups: 5 without treatment, 5 treated with acteoside. After continuous administration for 8 weeks, serum, urine, and kidney tissue were collected, then, ralated biochemical parameters, pathological characteristics and MCP-1 and TGF-β1 mRNA or protein were detected. The NRK-52E cells were divided into three groups as follows: the normal control group (NC group), the high glucose group (HS group), the high glucose+acteoside group (HS+ACT group). The expressions of MCP-1 and TGF-β1 in the mRNA and protein levels were assessed with RT-PCR, western blot and ELISA. Results: The expressions of MCP-1 and TGF-β1 were significantly enhanced in DN group and dramatically reduced after acteoside treatment. Compared with those in NC group, the expressions of MCP-1 and TGF-β1 in NRK-52E cell of HS group were sizgnificantly enhanced, while both were significantly decreased in HS+ACT group compared with HS group. This study aimed to investigate whether the exosomal lncRNA HCG18 regulated the M2 macrophage polarization in GC and the possible mechanism. The isolated GC cells (GCCs)-Exos were identified using transmission electron microscopy, Nanoparticle Tracking Analysis and Western blot. The GCCs-Exos function was verified by enzyme-linked immunosorbent assay and flow cytometry. Meanwhile, the exosomal lncRNA HCG18 function was determined using the in vitro assays. Furthermore, the underlying mechanism of the exosomal lncRNA HCG18 that regulated M2 macrophage polarization in GC was investigated using dual-luciferase reporter gene assay and RNA pull-down. After the validation of GCCs-Exos, the GCCs-Exos facilitated the M2 macrophage polarization. The in vitro assays confirmed that the exosomal lncRNA HCG18 positively regulated the M2 macrophage polarization. Mechanistically, lncRNA HCG18 bound to miR-875-3p, miR-875-3p bound to KLF4. In order to better explore the spontaneous combustion characteristics of lignite, a low-temperature heating experimental device was used to study the changes in the physical and chemical structure of lignite in an air atmosphere of 50–350°C. Through FTIR, XRD and SEM, the coal sample's chemical structure and surface microstructure change at low temperature are correlated and analysed, and the physical and chemical fabric change law of the coal sample at low temperature is obtained. The results show that in the water loss stage, the hydroxyl content in the coal sample increases, and the surface pores of the coal sample become larger. As the oxidation temperature increases, the pores further expand, secondary cracks appear, and gas escapes. In the decarboxylation stage, the coal sample is further coked, the methyl group increases, the oxygen-containing functional group increases, and the coal sample crystallisation degree increases, which accelerates the spontaneous combustion of the coal sample. Recent years have witnessed the emergence of worldwide megalopolises and the accompanying public safety events, making urban safety a top priority in modern urban management. Among various threats, dangerous goods such as gas and hazardous chemicals transported through cities have bred repeated tragedies and become the deadly "bomb" we sleep with every day. While tremendous research efforts have been devoted to dealing with dangerous goods transportation (DGT) issues, further study is still in great need to quantify this problem and explore its intrinsic dynamics from a big data perspective. In this article, we present a novel system called DGeye, to feature a fusion between DGT trajectory data and residential population data for dangers perception and prediction. Specifically, DGeye first develops a probabilistic graphical model-based approach to mine spatio-temporally adjacent risk patterns from population-aware risk trajectories. Then, DGeye builds the novel causality network among risk patterns for risk pain-point identification, risk source attribution, and online risky state prediction. Experiments on both Beijing and Tianjin cities demonstrate the effectiveness of DGeye in real-life DGT risk management. Large temperature gradient and local hot spot in solar receiver seriously impact the operation safety and thermal performance. To improve the temperature uniformity of porous foam absorber in a volumetric solar receiver, this work presents a receiver with an annular inner window to reshape the fluid field in the receiver. To examine the improvement effect of the proposed receiver, one numerical model is developed to analyze the coupled heat transfer in the receiver. The model takes the propagation of concentrated solar irradiation and thermal radiation in the consideration by using the Monte-Carlo ray tracing method. The effects of the inner window on the fluid flow and temperature distribution of the absorber as well as the efficiency of the receiver are analyzed. The results show that the standard difference of temperature in absorber of the proposed receiver is 35–158.6 K lower than that of the single windowed one, and the thermal efficiency is increased by 2.86%–7.18%, within the inner radius of the annular window range of 15–45 mm and flow rate of 4–7 g/s. Purpose: The aim of the study was to determine the feasibility of interstitial brachytherapy under non-anesthetist-administered moderate sedation, to identify factors influencing the insertion, and the total procedural time. Material and methods: A total of 47 insertions with hybrid intracavitary and interstitial applicators were performed in 23 patients from March 2017 to March 2020. Moderate sedation was achieved with intravenous midazolam and fentanyl administered by non-anesthetist. Insertion time and procedural time was recorded. Univariate and multivariate analysis were performed to evaluate the impact of different factors on insertion and procedural time. Results: A total of 238 needles (range, 2-8 per insertion) were implanted, with an average insertion depth of 30 mm (range, 20-40 mm). The mean doses for midazolam and fentanyl were 3 mg (standard deviation [SD] = 1) and 53.3 mcg (SD = 23.9) per insertion, respectively. The median insertion time was 30 minutes (interquartile range [IQR] = 22-40), and the median total procedural time was 4.3 hours (IQR = 3.6-5.2). First time insertion, insertions performed before 2019, and higher midazolam dose were associated with significantly longer insertion time, whereas longer insertion time, MRI-based planning, and insertions performed before 2019 were associated with significantly longer total procedural time. Conclusions: Outpatient interstitial brachytherapy with non-anesthetist-administered sedation is achievable and well-tolerated. Background/Aims: Targeted drug delivery vehicles with low immunogenicity and toxicity are needed for cancer therapy. Here, we prepare an active targeting drug carrier of low immunogenicity and toxicity for targeted therapy. Methods: Immature dendritic cells (imDCs) from BALB/c mice were used as donor cells of exosomes (Exos) that were transfected with the plasmids expressing fusion proteins of a tumor-targeting peptide known as internalizing RGD (iRGD) to construct a type of tumor-targeting iRGD-Exos and observe the interaction between these iRGD-Exos. Also, recombinant methioninase (rMETase) was loaded into the iRGD-Exos by electroporation to construct iRGD-Exos-rMETase and to assess the tumor-targeting function of the iRGD-Exos-rMETase. Finally, 30 BALB/c were randomly divided into five groups (n = 6), to observe tumor growth in vivo. Results: The iRGD-Exos-rMETase was 99.58 nm in diameter and presented a unique "goblet" structure under transmission electron microscopy (TEM), with the encapsulation efficiency (EE) of 19.05%. iRGD-Exos-rMETase group has the strongest tumor suppressive effect. Compared to the iRGD-Exos-rMETase group, rMETase group and the blank-Exos-rMETase group were less effective, while the PBS group and the iRGD-Exos group showed no inhibitory effect on tumor growth. After treatment, the iRGD-Exos-rMETase group had gastric tumors significantly smaller and lighter than the other groups (P < 0.05). Conclusion: The iRGD-Exos-rMETase is an effective antitumor therapy that delivers rMETase to tumor tissue using the iRGD-Exos. Dark tea is a post-fermented tea with unique organoleptic characteristics. A unique microbial fermentation procedure is involved in dark tea processing that is considered a major factor to manage the quality of dark tea. Dark tea is gaining increasing attention recently because it contains various bioactive compounds with diverse biological functions. This review summarizes the microbial fermentation process, main bioactive compounds, and important biological functions or health benefits of dark tea, emphasizing its protective effects against different diseases and relevant molecular mechanisms, and proposes the perspectives in research and application of dark tea. Functional core microorganism-mediated bioconversion is crucial for the formation of dark tea's main quality components during the fermentation period. Dark tea contains a variety of bioactive compounds, including alkaloids, free amino acids, peptides, polyphenols, pigments, polysaccharides, and volatile compounds. Consumption of dark teas has been linked to multiple health benefits, such as antioxidant, anti-obesity, anti-diabetic, anti-cancer, cardiovascular protective, gastrointestinal protective, hepatoprotective, neuroprotective, and photoprotective activities. Objective(s): Acute renal ischemia may cause acute renal dysfunction due to lack of blood supply; the manifestations are renal tubular cell apoptosis, infiltration of macrophages, and microvascular destruction. Many studies have shown that erythropoietin (EPO) and vitamin D3 (VD3) can be used to prevent or treat renal ischemia-reperfusion (I/R) injury, and VD3 may interact with EPO. In the present study, the effects of the combination of VD3 and EPO in I/R acute kidney injury were studied. Materials and Methods: Rats were divided into 5 groups: sham-operated (SHAM), AKI without treatment (AKI-control), AKI treatment with VD3(AKI+VD3), AKI treatment with EPO(AKI+EPO), AKI treatment with VD3 and EPO(AKI+VD3+EPO). The effects of the combination of VD3 and EPO on AKI were assessed by histologic, inflammation, and apoptosis studies. Results: The degree of damage in renal tissue was significantly reduced in VD3, EPO, and combined groups. Combination therapy with VD3 and EPO markedly improved Creatinine clearance rate (CCr). The combined treatment group showed the lowest F4/80+ and CD68+ expressions. The expression of Bcl-2 in the combined treatment group was higher than those in VD3 group and the EPO group, while Bax's expression goes in the opposite direction. Conclusion: This provides further evidence that VD3 and EPO have beneficial effects in I/R injury via anti-inflammatory and anti-apoptosis pathways. A multispectral radiometry method was developed to simultaneously determine normal spectral emissivity of continuum spectra and temperature of materials. Linear dependence assumption would be a constraint to associate the spectral emissivity to the temperature when spectral radiance for various spectrum was measured within small temperature differences. The methodology was evaluated for different parameter constraints, and validated by seven emissivity hypothesis models. To verify the reliability of the experimental device and the multispectral radiometry method, the spectral emissivities of graphite at high temperatures were verified that the deviations between the official experimental data and the results in this work are all less than 5 %. Besides, the spectral emissivity and temperature of the graphite-carbon fiber composite in the spectral range of 0.3–2.5 μm at 1473–1873 K were measured. The temperature uniformity of the sample front surface was evaluated, and the relative error of the temperature distribution is less than 2 %. The uncertainties of the spectral emissivity and temperature in the studied spectrum are less than5.0 % and 6.0 %, respectively. In order to solve the problem of large errors in the high temperature measurement of these materials, this paper constructs an inversion model for the radiative and conductive coupled heat transfer through wavelet decomposition coefficients, and compares it with traditional methods. The radiative and conductive coupled heat transfer model is solved by finite volume method and Monte Carlo method. Daubechies nine wavelet is used for signal wavelet analysis. It has been discovered that the wavelet decomposition parameters of the temperature field are mainly concentrated in the 4–8 wavelet frequency band of wavelet, it can separate from the random error that is mainly concentrated in the 1–3 frequency band of wavelet. Most of physical properties like the heating power, convective heat transfer coefficient, material thermal conductivity, extinction coefficient has a great impact on the waveform and amplitude of wavelet detail coefficient and approximation coefficient, and the change trend of approximation coefficient and detail coefficient amplitude is consistent with the trend of temperature field. However, the effect of scattering albedo on wavelet parameters is tiny. Feeding and oviposition deterrents help phytophagous insects to identify host plants. The taste organs of phytophagous insects contain bitter gustatory receptors (GRs). To explore their function, the GRs in Plutella xylostella were analyzed. Through RNA sequencing and qPCR, we detected abundant PxylGr34 transcripts in the larval head and adult antennae. Functional analyses using the Xenopus oocyte expression system and 24 diverse phytochemicals showed that PxylGr34 is tuned to the canonical plant hormones brassinolide (BL) and 24-epibrassinolide (EBL). Electrophysiological analyses revealed that the medial sensilla styloconica of 4th instar larvae are responsive to BL and EBL. Dual-choice bioassays demonstrated that BL inhibits larval feeding and female oviposition. Knock-down of PxylGr34 by RNAi attenuates the taste responses to BL, and abolishes BL-induced feeding inhibition. These results increase our understanding of how herbivorous insects detect compounds that deter feeding and oviposition, and may be useful for designing plant hormone-based pest management strategies. The pseudo-analog wireless video transmission technology can improve the effectiveness, reliability, and robustness of the conventional digital system in video broadcast scenarios. Although some prototypes of IEEE 802.11 series have been developed for researchers to do simulations and experiments, they are usually expensive and provide very limited access to the physical layer. More importantly, these prototypes cannot be used to verify the correctness of the new proposed pseudo-analog wireless video transmission algorithms directly due to limited modulation modes they can support. In this paper, we present a novel design of software radio platform (SDR)-based pseudo-analog wireless video transceiver which is completely transparent and allows users to learn all the implementation details. Firstly, we prove that the analog method can also achieve the optimal performance as the digital method from the perspective of the rate-distortion theory. Then, we describe the two hardware implementation difficulties existed in the designing process including the data format modification and the non-linear distortion. Next, we introduce the implementation details of the designed transceiver. Finally, we analyze the performance of the designed transceiver. Summary: An independent correlation between pre-RDW and 1-year mortality after surgery in elderly hip fracture can be used to predict mortality in elderly hip fracture patients and has predictive significance in anemia patients. With further research, a treatment algorithm can be developed to potentially identify patients at high risk of preoperative mortality. Introduction: Red blood cell distribution width (RDW) is an independent predictor of various disease states in elderly individuals, but its association with the prognosis of elderly hip fracture patients is controversial. This study aimed to evaluate the prognostic value of RDW in such patients, construct a prediction model containing RDW using random survival forest (RSF) and Cox regression analysis, and compare RDW in patients with and without anemia. Methods: We retrospectively analyzed the data of elderly patients who underwent hip fracture surgery, selected the best variables using RSF, stratified the independent variables by Cox regression analysis, constructed a 1-year mortality prediction model of elderly hip fracture with RDW, and conducted internal validation and external validation. Results: Two thousand one hundred six patients were included in this study. The RSF algorithm selects 12 important influencing factors, and Cox regression analysis showed that eight variables including preoperative RDW (pre-RDW) were independent risk factors for death within 1-year after hip fracture surgery in elderly patients. Stratified analysis showed that pre-RDW was still independently associated with 1-year mortality in the non-anemia group and not in the anemia group. The nomogram prediction model had high differentiation and fit, and the prediction model constructed by the total cohort of patients was also used for validation of patients in the anemia patients and obtained good clinical benefits. This work presents a numerical study on transmission performance of ellipsoidal windows used in receivers under concentrated solar irradiation. The Monte Carlo ray tracing method is used to analyze the transmission of concentrated solar radiation through an ellipsoidal window. The effects of concentrator shape error (1–3 mrad) and geometry parameters of the window, including window height (30–60 mm), radius of concave region (20–55 mm), and shape factor (0–100 mm), on the transmittance are examined. It is found that the transmittance increases with the increase of shape factor and window height and the decrease of the slope error of the concentrator. The dependence of the transmittance on the radius of the concave region is nonmonotonic. To maximize window transmittance, a single-variable nonlinear optimizer is used to optimize the concave region radius. The results indicate that the transmittance can be increased by 1.7% at most by increasing the shape factor, within the parameter range. Only if the window height is greater than a certain critical value, could the transmittance of the ellipsoidal window be significantly higher than that of plane window. The critical value increases with the increasing slope error and decreasing shape factor, varying from 20 to 40 mm. The transmittance of the ellipsoidal window with optimal parameter can range up to 98.9%, which is 7.82% higher than that of flat window. To investigate the changes of organic and inorganic substances produced by coal heat pyrolysis in the dry distillation channel in coal underground gasification, lignite coal was used as the object of this study. The pyrolysis reaction in the range of room temperature to 600 °C was carried out using coal pyrolysis reactor of high-temperature tubular heating furnace. SEM, FTIR and x-ray diffractometer analyzed the sample. The results show: The increase in pyrolysis temperature leads to a decrease in tar yield and an increase in gas and char coke yield. The surface of the coal sample shows molten state and gradually harden. The expansion of porosity accelerates the transportation of gas in coal seams. Coal pyrolysis is prone to hydrogen bond breakage at 400 °C. In addition, the content of unsaturated halogenated hydrocarbons increases under pyrolysis. • The most abundant functional groups in coal are oxygen-containing functional groups. • The pores of coal samples are cracked by high temperature. • The high temperature leads to reduction of alicyclic structure and lipid chain in coal. Recently, pseudo analog transmission has gained increasing attentions due to its ability to alleviate the cliff effect in video multicast scenarios. The existing pseudo analog systems are optimized under the minimum mean squared error criterion. However, their power allocation strategies do not take the perceptual video quality into consideration. In this article, we propose a human-perception-based pseudo analog video transmission system named ROIC-Cast, which aims to intelligently enhance the transmission quality of the region-of-interest (ROI) parts. Firstly, the classic deep learning based saliency detection algorithm is adopted to decompose the continuous video sequences into ROI and non-ROI blocks. Secondly, an effective compression method is used to reduce the data amount of side information generated by the ROI extraction module. Then, the power allocation scheme is formulated as a convex problem, and the optimal transmission power for both ROI and non-ROI blocks is derived in a closed form. Finally, the simulations are conducted to validate the proposed system by comparing with a few of existing systems, e.g., KMV-Cast, SoftCast, and DAC-RAN. The proposed ROIC-Cast can achieve over 4.1 dB peak signal-to-noise ratio gains of ROI compared with other systems, given the channel signal-to-noise ratio as −5 dB, 0 dB, 5 dB, and 10 dB, respectively. The amplitude of the atmospheric CO2 seasonal cycle has increased by 30 to 50% in the Northern Hemisphere (NH) since the 1960s, suggesting widespread ecological changes in the northern extratropics. However, substantial uncertainty remains in the continental and regional drivers of this prominent amplitude increase. Here we present a quantitative regional attribution of CO2 seasonal amplification over the past 4 decades, using a tagged atmospheric transport model prescribed with observationally constrained fluxes. We find that seasonal flux changes in Siberian and temperate ecosystems together shape the observed amplitude increases in the NH. At the surface of northern high latitudes, enhanced seasonal carbon exchange in Siberia is the dominant contributor (followed by temperate ecosystems). Arctic-boreal North America shows much smaller changes in flux seasonality and has only localized impacts. These continental contrasts, based on an atmospheric approach, corroborate heterogeneous vegetation greening and browning trends from field and remote-sensing observations, providing independent evidence for regionally divergent ecological responses and carbon dynamics to global change drivers. Over surface midlatitudes and throughout the midtroposphere, increased seasonal carbon exchange in temperate ecosystems is the dominant contributor to CO2 amplification, albeit with considerable contributions from Siberia. Representing the mechanisms that control the high-latitude asymmetry in flux amplification found in this study should be an important goal for mechanistic land surface models moving forward. Epigallocatechin gallate (EGCG) is a natural phenolic compound found in many plants, especially in green tea, which is a popular and restorative beverage with many claimed health benefits such as antioxidant, anti-cancer, anti-microbial, anti-diabetic, and anti-obesity activities. Despite its great curative potential, the poor bioavailability of EGCG restricts its clinical applcation. However, nanoformulations of EGCG are emerging as new alternatives to traditional formulations. This review focuses on the nanochemopreventive applications of various EGCG nanoparticles such as lipid-based, polymer-based, carbohydrate-based, protein-based, and metal-based nanoparticles. EGCG hybridized with these nanocarriers is capable of achieving advanced functions such as targeted release, active targeting, and enhanced penetration, ultimately increasing the bioavailability of EGCG. Cancer stem cells are undifferentiated cancer cells that have self‐renewal ability, a high tumorigenic activity, and a multilineage differentiation potential. MicroRNAs play a critical role in regulating gene expression during carcinogenesis. Here, we investigated the role of miR‐7 and the mechanism by which it is dysregulated in gastric cancer stem cells (GCSCs). The stem cell marker, CD44, was used to sort GCSCs by fluorescence‐activated cell sorting. We found that CD44 (+) cells have higher invasiveness and form more number of sphere colonies than CD44 (−) cells. Quantitative real‐time polymerase chain reaction (PCR) revealed that the miR‐7‐5p expression was remarkably downregulated in GCSCs but was significantly increased in the methionine‐deprived medium. The downregulation of miR‐7‐5p results from the increased DNA methylation in the promoter region using the methylation‐specific PCR. Overexpression of miR‐7‐5p reduced the formation of colony and decreased the invasion of GCSCs through targeting Smo and Hes1 and subsequent repressing Notch and Hedgehog signaling pathways in vitro. Notably, upregulating miR‐7‐5p inhibited the growth of tumor in the xenograft model. Hence, these data demonstrated that miR‐7‐5p represses GCSC invasion through inhibition of Smo and Hes1, which provides a potential therapeutic target of gastric cancer treatment. Dilated cardiomyopathy (DCM) is one of the leading causes of heart failure. A large proportion of genetic cause remains unexplained, especially in idiopathic DCM. We performed target next-generation sequencing of 102 genes which were known causes or candidate genes for cardiomyopathies and channelpathies in 118 prospectively recruited Han Chinese patients with idiopathic DCM. 41 of the 118 patients carried 40 pathogenic or likely pathogenic variants, providing a molecular diagnosis in 34.7% of patients. 32 of these variants were novel. TTN truncating variants were predominant, with a frequency of 31.0%, followed by variants of LMNA (14.3%), RBM20 (4.8%), and NEXN (4.8%). These 4 genes accounted for over half variants identified. No significant difference in clinical characteristics or rates of reaching the composite end point (cardiac transplantation and death from cardiac causes) between pathogenic or likely pathogenic variant carriers and noncarriers (hazard ratio 1.11, 95% CI: 0.41 to 3.00), or between patients with TTN truncating variants or without (hazard ratio 0.49, 95% CI: 0.36 to 6.10). In our prospective study, we first determined the overall genetic profiles and genotype-phenotype correlations in Han Chinese idiopathic DCM patients, which could provide insight for genetic diagnosis of DCM in this population. The mechanical properties of poly(arylene sulfide sulfone) (PASS) electrospun membrane were significantly enhanced by co-electrospinning with semi-aromatic nylon poly(m-xylene adipamide) (MXD6), another engineering plastic with high thermal stability and good mechanical properties. The tensile strength of PASS membrane increased with increased incorporation of MXD6, and was tripled when 20% MXD6 was incorporated. The mechanism of the mechanical property improvement is the existence of hydrogen bonding interaction between PASS and MXD6 and between adjacent fibers at the intersections. Thermal properties of the PASS/MXD6 membranes were evaluated by differential scanning calorimetry (DSC) and thermogravimetric analysis (TGA), which showed that the membranes could be stably utilized up to 180 °C without any change in appearance and without decomposition. Contact angle measurements of all the membranes showed hydrophobic character. To demonstrate the potential applications of PASS/MXD6 blend membranes, their oil absorption capacities were evaluated with three oils of different viscosities, which proved that the PASS/MXD6 membranes are better absorbents than commercial non-woven polypropylene fibers. Primary Objective: This study characterized the acute and chronic effects of tau reduction in traumatic brain injury (TBI). Research Design: A fluid percussion injury (FPI) or a sham-injury was administered to wild type (WT) or tau knockout (Tau-/-) mice. Mice were assigned to a one-week or twelve-week recovery period before behavioral testing and analysis of brain tissue. Methods and Procedures: Mice were tested on the elevated-plus maze, the Y-maze, and rotarod. The twelve-week recovery mice underwent in vivo MRI. Phosphorylated tau in brain tissue was analyzed post-mortem using western blots. Main Outcomes and Results: FPI mice, regardless of genotype, had abnormalities on the elevated-plus maze (a task to assess anxiety-like behavior) at one-week post-injury. However, after twelve-weeks recovery, the Tau-/- mice that were given an FPI were less anxious and had improved motor function compared to their WT counterparts. MRI analysis found that while all FPI mice had brain damage, the Tau-/- mice had larger hippocampal volumes. The WT+FPI mice also had increased phosphorylated tau compared to WT+sham mice at both the one-week and twelve-week recovery times. Conclusion: These findings suggest that tau may play an important role in some of the consequences of TBI, particularly the long-term functional deficits. [ABSTRACT FROM AUTHOR]
