Abstract: The Water Cherenkov Detector Array (WCDA) is one of the components of Large High Altitude Air Shower Observatory (LHAASO) and can monitor any sources over two-thirds of the sky for up to 7 hours per day with >98\% duty cycle. In this work, we report the detection of two outbursts of the Fanaroff-Riley I radio galaxy NGC 1275 that were detected by LHAASO-WCDA between November 2022 and January 2023 with statistical significance of 5.2~$\sigma$ and 8.3~$\sigma$. The observed spectral energy distribution in the range from 500 GeV to 3 TeV is fitted by a power-law with a best-fit spectral index of $\alpha=-3.37\pm0.52$ and $-3.35\pm0.29$, respectively. The outburst flux above 0.5~TeV was ($4.55\pm 4.21)\times~10^{-11}~\rm cm^{-2}~s^{-1}$ and ($3.45\pm 1.78)\times~10^{-11}~\rm cm^{-2}~s^{-1}$, corresponding to 60\%, 45\% of Crab Nebula flux. Variation analysis reveals the variability time-scale of days at the TeV energy band. A simple test by one-zone synchrotron self-Compton model reproduces the data in the gamma-ray band well., Comment: 11 pages, 8 figures, 3 tables

Abstract: We report the detection of an extended very-high-energy (VHE) gamma-ray source coincident with the locations of middle-aged (62.4~\rm kyr) pulsar PSR J0248+6021, by using the LHAASO-WCDA data of live 796 days and LHAASO-KM2A data of live 1216 days. A significant excess of \gray induced showers is observed both by WCDA in energy bands of 1-25~\rm TeV and KM2A in energy bands of $>$ 25~\rm TeV with 7.3 $\sigma$ and 13.5 $\sigma$, respectively. The best-fit position derived through WCDA data is R.A. = 42.06$^\circ \pm$ 0.12$^\circ$ and Dec. = 60.24$^\circ \pm $ 0.13$^\circ$ with an extension of 0.69$^\circ\pm$0.15$^\circ$ and that of the KM2A data is R.A.= 42.29$^\circ \pm $ 0.13$^\circ$ and Dec. = 60.38$^\circ \pm$ 0.07$^\circ$ with an extension of 0.37$^\circ\pm$0.07$^\circ$. No clear extended multiwavelength counterpart of this LHAASO source has been found from the radio band to the GeV band. The most plausible explanation of the VHE \gray emission is the inverse Compton process of highly relativistic electrons and positrons injected by the pulsar. These electrons/positrons are hypothesized to be either confined within the pulsar wind nebula or to have already escaped into the interstellar medium, forming a pulsar halo., Comment: 12 pages, 10 figures, Accepted by Sci. However, the mechanism by which circ-OGDH regulates ESCC progression is unclear.Methods: Expression levels of circ-OGDH, microRNA (miR)-615-5p, and PDX1 (pancreatic and duodenal homeobox 1) mRNA were evaluated with quantitative real-time polymerase chain reaction (qRT-PCR). The proliferation, apoptosis, migration, invasion, and cell cycle progression of ESCC cells were analyzed by MTT (3-(4,5-Dimethylthiazol-2-yl)-2,5-Diphenyltetrazolium Bromide), colony formation, flow cytometry, and transwell assays. Measurement of glutamine consumption, α-KG (α-ketoglutarate) production, and ATP (Adenosine Triphosphate) content using corresponding kits. Protein levels were analyzed by Western blotting. The targeting relationship between circ-OGDH or PDX1 and miR-615-5p was verified by dual-luciferase reporter and RNA immunoprecipitation (RIP) assays. The function of circ-OGDH in ESCC was confirmed by animal experiments.Results: Circ-OGDH was upregulated in ESCC. Circ-OGDH inhibition reduced ESCC growth in vivo and accelerated cell apoptosis, cell cycle arrest, repressed cell proliferation, migration, invasion, and reduced cell glutamine metabolism in ESCC cells in vitro. MiR-615-5p was downregulated in ESCC, while PDX1 had an opposite result. Circ-OGDH sponged miR-615-5p to regulate PDX1 expression. MiR-615-5p inhibitor neutralized the repressive effect of circ-OGDH knockdown on malignancy and glutamine metabolism of ESCC cells. PDX1 overexpression counteracted the inhibitory impact of miR-615-5p mimic on malignancy and glutamine metabolism of ESCC cells.Conclusion: Circ-OGDH sponged miR-615-5p to elevate PDX1 expression, thus elevating glutamine metabolism and promoting tumor growth in ESCC. Abstract: In this work we try to search for signals generated by ultra-heavy dark matter at the Large High Altitude Air Shower Observatory (LHAASO) data. We look for possible gamma-ray by dark matter annihilation or decay from 16 dwarf spheroidal galaxies in the field of view of LHAASO. Dwarf spheroidal galaxies are among the most promising targets for indirect detection of dark matter which have low fluxes of astrophysical $\gamma$-ray background while large amount of dark matter. By analyzing more than 700 days observational data at LHAASO, no significant dark matter signal from 1 TeV to 1 EeV is detected. Accordingly we derive the most stringent constraints on the ultra-heavy dark matter annihilation cross-section up to EeV. The anti.-metastasis has been considered as a feasible strategy for clinical cancer management. It is well known that diosgenin could inhibit tumor metastasis and doxorubicin (DOX) could induce tumor apoptosis. However, their efficient delivery remains challenging.Purpose: To address these issues, a novel pH-sensitive polymer-prodrug based on diosgenin nanoparticles (NPs) platform was developed to enhance the efficiency of DOX delivery (DOX/NPs) for synergistic therapy of cutaneous melanoma, the most lethal form of skin cancer with high malignancy, early metastasis and high mortality.Methods and Results: The inhibitory effect of DOX/NPs on tumor proliferation and migration was superior to that of NPs or free DOX. What is more, DOX/NPs could combine mitochondria-associated metastasis and apoptosis with unique internalization pathway of carrier to fight tumors. In addition, biodistribution experiments proved that DOX/NPs could efficiently accumulate in tumor sites through enhancing permeation and retention (EPR) effect compared with free DOX. Importantly, the data from in vivo experiment revealed that DOX/NPs without heart toxicity significantly inhibited tumor metastasis by exerting synergistic therapeutic effect, and reduced tumor volume and weight by inducing apoptosis.Conclusion: The nanocarrier DOX/NPs with satisfying pharmaceutical characteristics based on the establishment of two different functional agents is a promising strategy for synergistically enhancing effects of cancer therapy.Keywords: anti-metastasis, antitumor, self-assembly, codelivery, doxorubicin

Platelets and blood coagulation system are highly associated with tumor hematogenous metastasis. Therefore, the purpose of this study was to evaluate whether dihydrodiosgenin (dydio) mediated-platelet inhibition or coagulation factor level modulation is involved in hepatocellular carcinoma cell (HCC) metastasis.Methods: Cell viability was examined by MTT and colony formation assays. Platelet aggregation text and morphology were used to assess dydio’s role on tumor cell-induced platelet activation (TCIPA). Scratch assay, adhesion assay and Western blot were used to evaluate dydio’s role on platelet-mediated metastasis. Western blot and fluorescence detection were performed to clarify dydio’s role on endothelial cell (EC) function. The mice lung metastasis model was constructed to investigated dydio’s function on coagulation factor and platelet-mediated metastasis.Results: This study found that pretreatment with dydio caused a significant inhibition of TCIPA. Platelets exposed to dydio significantly inhibited their adhesion to tumor cells, meanwhile, releasates of platelets that pretreated with dydio led to diminished cancer cell proliferation and migration along with the increase of epithelial markers E-cadherin and loss of mesenchymal phenotype. Additionally, ECs pretreated with dydio suppressed factor VIII (FVIII) level which in turn restrained the activation of platelets and the adhesion of cancer cells or platelets to ECs. Interestingly, our study demonstrated that FVIII could promote HCC proliferation. In vivo study revealed that mice intragastrical (i.g.) administration with dydio significantly inhibited the lung metastasis of hepal-6 cells which is highly correlated with the altered platelet function and coagulation level.Conclusion: Taken together, these results demonstrated that dydio altered platelet function and coagulation FVIII level, resulting in decreased metastatic potential of HCC. Abstract: The first source catalog of Large High Altitude Air Shower Observatory reported the detection of a very-high-energy gamma ray source, 1LHAASO J1219+2915. In this paper a further detailed study of the spectral and temporal behavior of this point-like source have been carried. The best-fit position of the TeV source ($\rm{RA}=185.05^{\circ}\pm0.04^{\circ}$, $\rm{Dec}=29.25^{\circ}\pm0.03^{\circ}$) is compatible with NGC 4278 within $\sim0.03$ degree. Variation analysis shows an indication of the variability at a few months level in the TeV band, which is consistent with low frequency observations. Based on these observations, we report the detection of TeV $\gamma$-ray emissions from this low-luminosity AGN NGC 4278. The observations by LHAASO-WCDA during active period has a significance level of 8.8\,$\sigma$ with best-fit photon spectral index $\varGamma=2.56\pm0.14$ and a flux $f_{1-10\,\rm{TeV}}=(7.0\pm1.1_{\rm{sta}}\pm0.35_{\rm{syst}})\times10^{-13}\,\rm{photons\,cm^{-2}\,s^{-1}}$, or approximately $5\%$ of the Crab Nebula. The discovery of VHE from NGC 4278 indicates that the compact, weak radio jet can efficiently accelerate particles and emit TeV photons., Comment: 11 pages, 5 figures

Abstract: KM2A is one of the main sub-arrays of LHAASO, working on gamma ray astronomy and cosmic ray physics at energies above 10 TeV. Detector simulation is the important foundation for estimating detector performance and data analysis. It is a big challenge to simulate the KM2A detector in the framework of Geant4 due to the need to track numerous photons from a large number of detector units (>6000) with large altitude difference (30 m) and huge coverage (1.3 km^2). In this paper, the design of the KM2A simulation code G4KM2A based on Geant4 is introduced. The process of G4KM2A is optimized mainly in memory consumption to avoid memory overffow. Some simpliffcations are used to signiffcantly speed up the execution of G4KM2A. The running time is reduced by at least 30 times compared to full detector simulation. The particle distributions and the core/angle resolution comparison between simulation and experimental data of the full KM2A array are also presented, which show good agreement.

Abstract: We present the measurements of all-particle energy spectrum and mean logarithmic mass of cosmic rays in the energy range of 0.3-30 PeV using data collected from LHAASO-KM2A between September 2021 and December 2022, which is based on a nearly composition-independent energy reconstruction method, achieving unprecedented accuracy. Our analysis reveals the position of the knee at $3.67 \pm 0.05 \pm 0.15$ PeV. Below the knee, the spectral index is found to be -$2.7413 \pm 0.0004 \pm 0.0050$, while above the knee, it is -$3.128 \pm 0.005 \pm 0.027$, with the sharpness of the transition measured with a statistical error of 2%. The mean logarithmic mass of cosmic rays is almost heavier than helium in the whole measured energy range. It decreases from 1.7 at 0.3 PeV to 1.3 at 3 PeV, representing a 24% decline following a power law with an index of -$0.1200 \pm 0.0003 \pm 0.0341$. This is equivalent to an increase in abundance of light components. Above the knee, the mean logarithmic mass exhibits a power law trend towards heavier components, which is reversal to the behavior observed in the all-particle energy spectrum. Additionally, the knee position and the change in power-law index are approximately the same. These findings suggest that the knee observed in the all-particle spectrum corresponds to the knee of the light component, rather than the medium-heavy components., Comment: 8 pages, 3 figures

Abstract: For decades, supernova remnants (SNRs) have been considered the prime sources of Galactic Cosmic rays (CRs). But whether SNRs can accelerate CR protons to PeV energies and thus dominate CR flux up to the knee is currently under intensive theoretical and phenomenological debate. The direct test of the ability of SNRs to operate as CR PeVatrons can be provided by ultrahigh-energy (UHE; $E_\gamma \geq 100$~TeV) $\gamma$-rays. In this context, the historical SNR Cassiopeia A (Cas A) is considered one of the most promising target for UHE observations. This paper presents the observation of Cas A and its vicinity by the LHAASO KM2A detector. The exceptional sensitivity of LHAASO KM2A in the UHE band, combined with the young age of Cas A, enabled us to derive stringent model-independent limits on the energy budget of UHE protons and nuclei accelerated by Cas A at any epoch after the explosion. Abstract: The highest energy gamma-rays from gamma-ray bursts (GRBs) have important implications for their radiation mechanism. Here we report for the first time the detection of gamma-rays up to 13 TeV from the brightest GRB 221009A by the Large High Altitude Air-shower Observatory (LHAASO). The LHAASO-KM2A detector registered more than 140 gamma-rays with energies above 3 TeV during 230$-$900s after the trigger. The intrinsic energy spectrum of gamma-rays can be described by a power-law after correcting for extragalactic background light (EBL) absorption. Such a hard spectrum challenges the synchrotron self-Compton (SSC) scenario of relativistic electrons for the afterglow emission above several TeV. Observations of gamma-rays up to 13 TeV from a source with a measured redshift of z=0.151 hints more transparency in intergalactic space than previously expected. Alternatively, one may invoke new physics such as Lorentz Invariance Violation (LIV) or an axion origin of very high energy (VHE) signals., Comment: 49pages, 11figures

Abstract: We present the first catalog of very-high energy and ultra-high energy gamma-ray sources detected by the Large High Altitude Air Shower Observatory (LHAASO). The catalog was compiled using 508 days of data collected by the Water Cherenkov Detector Array (WCDA) from March 2021 to September 2022 and 933 days of data recorded by the Kilometer Squared Array (KM2A) from January 2020 to September 2022. This catalog represents the main result from the most sensitive large coverage gamma-ray survey of the sky above 1 TeV, covering declination from $-$20$^{\circ}$ to 80$^{\circ}$. In total, the catalog contains 90 sources with an extended size smaller than $2^\circ$ and a significance of detection at $> 5\sigma$. Based on our source association criteria, 32 new TeV sources are proposed in this study. Among the 90 sources, 43 sources are detected with ultra-high energy ($E > 100$ TeV) emission at $> 4\sigma$ significance level. Abstract: The diffuse Galactic $\gamma$-ray emission, mainly produced via interactions between cosmic rays and the interstellar medium and/or radiation field, is a very important probe of the distribution, propagation, and interaction of cosmic rays in the Milky Way. In this work we report the measurements of diffuse $\gamma$-rays from the Galactic plane between 10 TeV and 1 PeV energies, with the square kilometer array of the Large High Altitude Air Shower Observatory (LHAASO). Diffuse emissions from the inner ($15^{\circ}10$~TeV). The energy spectrum in the inner Galaxy regions can be described by a power-law function with an index of $-2.99\pm0.04$, which is different from the curved spectrum as expected from hadronic interactions between locally measured cosmic rays and the line-of-sight integrated gas content. Furthermore, the measured flux is higher by a factor of $\sim3$ than the prediction. A similar spectrum with an index of $-2.99\pm0.07$ is found in the outer Galaxy region, and the absolute flux for $10\lesssim E\lesssim60$ TeV is again higher than the prediction for hadronic cosmic ray interactions. The latitude distributions of the diffuse emission are consistent with the gas distribution, while the longitude distributions show clear deviation from the gas distribution. Abstract: We study ionization of atoms in strong orthogonal two-color ($\omega,2\omega$) (OTC) laser fields numerically and analytically. The calculated photoelectron momentum distribution shows two typical structures: a rectangular-like structure and a shoulder-like structure, the positions of which depend on the laser parameters. Using a strong-field model which allows us to quantitatively evaluate the Coulomb effect, we show that these two structures arise from attosecond response of electron inside an atom to light in OTC-induced photoemission. 15. Response time of electron inside a molecule to light in strong-field ionization

Abstract: We study ionization of aligned H$_2^+$ in strong elliptically-polarized laser fields numerically and analytically. The calculated offset angle in photoelectron momentum distribution is several degrees larger for the molecule than a model atom with similar ionization potential at diverse laser parameters. Using a strong-field model that considers the properties of multi-center and single-center Coulomb potentials, we are able to quantitatively reproduce this angle difference between the molecule and the atom. Further analyses based on this model show that the response time of electron to light which is encoded in the offset angle and is manifested as the time spent in tunneling ionization, is about 15 attoseconds longer for the molecule than the atom. Abstract: We study ionization of atoms in strong elliptically-polarized laser fields. We focus on the physical origin of the offset angle in the photoelectron momentum distribution and its possible relation to a specific time. By developing a model which is based on strong-field approximation and considers the classical Coulomb scattering, we are able to quantitatively explain recent attoclock experiments in a wide region of laser and atomic parameters. The offset angle can be understood as arising from the scattering of the electron by the ionic potential when the electron exits the laser-Coulomb-formed barrier through tunneling. 18. Single-shot Foothold Selection and Constraint Evaluation for Quadruped Locomotion

Abstract: In this paper, we propose a method for selecting the optimal footholds for legged systems. The goal of the proposed method is to find the best foothold for the swing leg on a local elevation map. We apply the Convolutional Neural Network to learn the relationship between the local elevation map and the quality of potential footholds. The proposed network evaluates the geometrical characteristics of each cell on the elevation map, checks kinematic constraints and collisions. During execution time, the controller obtains the qualitative measurement of each potential foothold from the neural model. This method allows to evaluate hundreds of potential footholds and check multiple constraints in a single step which takes 10~ms on a standard computer without GPGPU. The experiments were carried out on a quadruped robot walking over rough terrain in both simulation and real robotic platforms.

Abstract: The Large High Altitude Air Shower Observatory (LHAASO) has three sub-arrays, KM2A, WCDA and WFCTA. The flux variations of cosmic ray air showers were studied by analyzing the KM2A data during the thunderstorm on 10 June 2021. The number of shower events that meet the trigger conditions increases significantly in atmospheric electric fields, with maximum fractional increase of 20%. The variations of trigger rates (increases or decreases) are found to be strongly dependent on the primary zenith angle. The flux of secondary particles increases significantly, following a similar trend with that of the shower events. To better understand the observed behavior, Monte Carlo simulations are performed with CORSIKA and G4KM2A (a code based on GEANT4). We find that the experimental data (in saturated negative fields) are in good agreement with simulations, assuming the presence of a uniform upward electric field of 700 V/cm with a thickness of 1500 m in the atmosphere above the observation level. Due to the acceleration/deceleration and deflection by the atmospheric electric field, the number of secondary particles with energy above the detector threshold is modified, resulting in the changes in shower detection rate., Comment: 18 pages, 11 figures

Abstract: The response time of the electron to light in photoemission is difficult to define and measure. Tunneling ionization of atoms, a strong-laser-induced photoemission process, provides a semiclassical case for visiting the problem. Here, we show that the response time can be determined at the boundary between quantum and classic. Specifically, tunneling is instantaneous but a finite response time (about 100 attoseconds) is needed for the state of the tunneling electron to evolve into the ionized state around tunnel exit. This time can be well described with a compact expression related to some basic laser and atomic parameters. Moreover, it can be directly mapped to and easily decoded from photoelectron momentum with a simple mapping, allowing an unambiguous measurement. Abstract: The Crab pulsar and the surrounding nebula powered by the pulsar's rotational energy through the formation and termination of a relativistic electron-positron wind is a bright source of gamma-rays carrying crucial information about this complex conglomerate. We report the detection of $\gamma$-rays with a spectrum showing gradual steepening over three energy decades, from $5\times 10^{-4}$ to $1.1$ petaelectronvolt (PeV). The ultra-high-energy photons exhibit the presence of a PeV electron accelerator (a pevatron) with an acceleration rate exceeding 15% of the absolute theoretical limit. Assuming that unpulsed $\gamma$-rays are produced at the termination of the pulsar's wind, we constrain the pevatron's size, between $0.025$ and $0.1$ pc, and the magnetic field $\approx 110 \mu$G. The production rate of PeV electrons, $2.5 \times 10^{36}$ erg $\rm s^{-1}$, constitutes 0.5% of the pulsar's spin-down luminosity, although we do not exclude a non-negligible contribution of PeV protons to the production of the highest energy $\gamma$-rays., Comment: 43 pages, 13 figures, 2 tables; Published in Science

Abstract: The Wide Field-of-View Cherenkov Telescope Array (WFCTA) and the Water Cherenkov Detector Arrays (WCDA) of LHAASO are designed to work in combination for measuring the energy spectra of various cosmic ray species over a very wide energy range from a few TeV to 10 PeV. The energy calibration of WCDA can be achieved with a proven technique of measuring the westward shift of the Moon shadow of galactic cosmic rays due to the geomagnetic field. This deflection angle $\Delta$ is inversely proportional to the energy of the cosmic rays. The precise measurements of the shifts by WCDA allows us to calibrate its energy scale for energies as high as 35 TeV. The energy scale measured by WCDA can be used to cross calibrate the energy reconstructed by WFCTA, which spans the whole energy range up to 10 PeV. In this work, we will demonstrate the feasibility of the method using the data collected from April 2019 to January 2020 by the WFCTA array and WCDA-1 detector, the first of the three water Cherenkov ponds, already commissioned at LHAASO site.

Abstract: Electrons in atoms and molecules can not react immediately to the action of intense laser field. A time lag (about 100 attoseconds) between instants of the field maximum and the ionization-rate maximum emerges. This lag characterizes the response time of the electronic wave function to the strong-field ionization event and has important effects on subsequent ultrafast dynamics of the ionized electron. The absolute time lag is not accessible in experiments. Here, a calibrated attoclock procedure, which is related to a simple Coulomb-induced temporal correction to electron trajectories, is proposed to measure the relative lag of two different ionization events. Using this procedure,the difference (i.e., the relative lag) between the ionization time lags of polar molecules in two consecutive half laser cycles can be probed with a high accuracy., Comment: 5 pages, 4 figures

Abstract: The focal plane camera is the core component of the Wide Field-of-view Cherenkov/fluorescence Telescope Array (WFCTA) of the Large High-Altitude Air Shower Observatory (LHAASO). Because of the capability of working under moonlight without aging, silicon photomultipliers (SiPM) have been proven to be not only an alternative but also an improvement to conventional photomultiplier tubes (PMT) in this application. Eighteen SiPM-based cameras with square light funnels have been built for WFCTA. The telescopes have collected more than 100 million cosmic ray events and preliminary results indicate that these cameras are capable of working under moonlight. The characteristics of the light funnels and SiPMs pose challenges (e.g. dynamic range, dark count rate, assembly techniques). In this paper, we present the design features, manufacturing techniques and performances of these cameras. Abstract: As a sub-array of the Large High Altitude Air Shower Observatory (LHAASO), KM2A is mainly designed to cover a large fraction of the northern sky to hunt for gamma-ray sources at energies above 10 TeV. Even though the detector construction is still underway, a half of the KM2A array has been operating stably since the end of 2019. In this paper, we present the pipeline of KM2A data analysis and the first observation on the Crab Nebula, a standard candle in very high energy gamma-ray astronomy. We detect gamma-ray signals from the Crab Nebula in both energy ranges of 10$-$100 TeV and $>$100 TeV with high significance, by analyzing the KM2A data of 136 live days between December 2019 and May 2020. With the observations, we test the detector performance including angular resolution, pointing accuracy and cosmic ray background rejection power. The energy spectrum of the Crab Nebula in the energy range 10-250 TeV fits well with a single power-law function dN/dE =(1.13$\pm$0.05$_{stat}$$\pm$0.08$_{sys}$)$\times$10$^{-14}$$\cdot$(E/20TeV)$^{-3.09\pm0.06_{stat}\pm0.02_{sys}}$ cm$^{-2}$ s$^{-1}$ TeV$^{-1}$. It is consistent with previous measurements by other experiments. 27. Strong-field double ionization dynamics of vibrating HeH$^+$ versus HeT$^+$

Abstract: We study double ionization (DI) dynamics of vibrating HeH$^+$ versus its isotopic variant HeT$^+$ in strong laser fields numerically. Our simulations show that for both cases, these two electrons in DI prefer to release together along the H(T) side. At the same time, however, the single ionization (SI) is preferred when the first electron escapes along the He side. This potential mechanism is attributed to the interplay of the rescattering of the first electron and the Coulomb induced large ionization time lag. On the other hand, the nuclear motion increases the contributions of these two electrons releasing together along the He side. 28. Tracing origins of asymmetric momentum distribution for polar molecules in strong linearly-polarized laser fields

Abstract: We study the ionization dynamics of oriented HeH$^+$ in strong linearly-polarized laser fields by numerically solving the time-dependent Schr\"{o}dinger equation. The calculated photoelectron momentum distributions for parallel orientation show a striking asymmetric structure. With a developed model pertinent to polar molecules, we trace the electron motion in real time. We show that this asymmetric structure arises from the interplay of the Coulomb effect and the permanent dipole in strong laser fields. Abstract: After electrons tunnel out of a laser-Coulomb-formed barrier, %formed by the strong laser field and the atomic Coulomb potential, the movement of the tunneling electron can be affected by the Coulomb tail. We show that this Coulomb effect induces a large time difference (longer than a hundred attoseconds) between the exiting time at which the electron exits the barrier and the ionization time at which the electron is free. Abstract: Our main result here is that the specialization at $t=1/q$ of the $Q_{km,kn}$ operators studied in [4] may be given a very simple plethystic form. This discovery yields elementary and direct derivations of several identities relating these operators at $t=1/q$ to the Rational Compositional Shuffle conjecture of [3]. In particular we show that if $m,n $ and $k$ are positive integers and $(m,n)$ is a coprime pair then $$ q^{(km-1)(kn-1)+k-1\over 2} Q_{km,kn}(-1)^{kn}\Big|_{t=1/q} \,=\, \textstyle{[k]_q\over [km]_q} e_{km}\big[ X[km]_q\big] $$ where as customarily, for any integer $s \geq 0$ and indeterminate $u$ we set $[s]_u=1+u+\cdots +u^{s-1}$. We also show that the symmetric polynomial on the right hand side is always Schur positive. Moreover, using the Rational Compositional Shuffle conjecture, we derive a precise formula expressing this polynomial in terms of Parking functions in the $km\times kn$ lattice rectangle.

35. Some new applications of the Stanley-Macdonald Pieri Rules

Abstract: In a seminal paper Richard Stanley derived Pieri rules for the Jack symmetric function basis. These rules were extended by Macdonald to his now famous symmetric function basis. The original form of these rules had a forbidding complexity that made them difficult to use in explicit calculations. In the early 90's it was discovered that, due to massive cancellations, the dual rule, which expresses skewing by $e_1$ the modified Macdonald polynomial ${\tilde H}_\mu[X;q,t]$, can be given a very simple combinatorial form in terms of corner weights of the Ferrers' diagram of $\mu$. A similar formula was later obtained by the last named author for the multiplication of ${\tilde H}_\mu[X;q,t]$ by $e_1$, but never published. In the years that followed we have seen some truly remarkable uses of these two Pieri rules in establishing highly non trivial combinatorial results in the Theory of Macdonald polynomials. This theory has recently been spectacularly enriched by various Algebraic Geometrical results in the works of Hikita, Schiffmann, Schiffmann-Vasserot, A. Negut and Gorsky-Negut. This development opens up the challenging task of deriving their results by purely Algebraic Combinatorial methods. Abstract: Abstract Recent studies have uncovered that TIPE2 is involved in the development of cancer. However, less research has been conducted on the role of TIPE2 in melanoma. Our study aims to elucidate the mechanism of action of TIPE2 in the development of melanoma. We examined TIPE2 expression in paracarcinoma tissue and melanoma tissues and found that TIPE2 expression was downregulated in melanoma tissue compared with paracarcinoma tissue. Overexpression of TIPE2 significantly inhibited the proliferation of melanoma cells in vitro and even inhibited tumor formation in vivo. The CCK8 assay results indicated that TIPE2 overexpression suppressed the proliferation of melanoma cells. The colony-forming ability and wound healing ability of TIPE2-overexpressing melanoma cells were significantly reduced compared with those of control cells. Moreover, immunohistochemistry experiments using a nude mouse tumor model showed consistent results. TIPE2 inhibited the phosphorylation of MEK and ERK. In summary, TIPE2 suppresses the proliferation and migration of melanoma cells by affecting proliferation-related factors and possibly by regulating the MEK/ERK pathway. TIPE2 could be used to inhibit melanoma growth and is a potential drug target for future drug development.

However, the presence of consistent evidence remains elusive. Our study aimed to conduct a systematic review and meta-analysis to identify differences in small-molecule metabolites between BPD and non-BPD preterm infants. Through meticulous screening of numerous samples, we identified promising candidates, providing valuable insights for future research. We searched PubMed, the Cochrane Library, Embase, Web of Science, China National Knowledge Internet, Wan-fang database, Chinese Science and Technique Journal Database and Chinese Biomedical Literature Database from inception until January 16, 2024. Studies were comprehensively reviewed against inclusion criteria. We included case-control studies and adhered to Preferred Reporting Items for Systematic Reviews and Meta-Analysis guidelines. Study quality was assessed with the Newcastle-Ottawa scale. We compared the changes in metabolite levels between the BPD and non-BPD preterm infants. A meta-analysis was conducted on targeted metabolomics research data based on the strategy of standardized mean differences (MD) and 95% confidence intervals (CI).Fifteen studies (1357 participants) were included. 40. On the spatial double peak of the 2023–2024 El Niño event

Abstract: Abstract The 2023–2024 El Niño has raised widespread concern in scientific and public communities. Here, using latest observational reanalysis, we show that this event matured with two distinct spatial peaks of sea surface temperature anomaly in the equatorial central and eastern Pacific, respectively. Like other double-peaked El Niños, the central Pacific sea surface temperature peaked in winter due to zonal advective and thermocline feedbacks. However, the eastern counterpart matured asynchronously in autumn and then stopped growing. This peculiar behavior results primarily from a cooling zonal advection by the anomalous westward current in the eastern Pacific during the autumn of 2023, which is associated with the local wind-driven sea surface height meridional concavity. We further propose that the relatively stronger and more eastward-displaced autumn precipitation in the Pacific intertropical convergence zone in 2023, compared to other double-peaked El Niños, is the primary cause of this distinct wind and sea surface height pattern.

41. TDFPS-Designer: an efficient toolkit for barcode design and selection in nanopore sequencing

Abstract: Abstract Oxford Nanopore Technologies (ONT) offers ultrahigh-throughput multi-sample sequencing but only provides barcode kits that enable up to 96-sample multiplexing. We present TDFPS-Designer, a new toolkit for nanopore sequencing barcode design, which creates significantly more barcodes: 137 with a length of 20 base pairs, 410 at 24 bp, and 1779 at 30 bp, far surpassing ONT’s offerings. It includes GPU-based acceleration for ultra-fast demultiplexing and designs robust barcodes suitable for high-error ONT data. TDFPS-Designer outperforms current methods, improving the demultiplexing recall rate by 20% relative to Guppy, without a reduction in precision.

42. The gut microbial metabolite indole-3-aldehyde alleviates impaired intestinal development by promoting intestinal stem cell expansion in weaned piglets

Abstract: Abstract Background Weaning stress-induced diarrhea is widely recognized as being associated with gut microbiota dysbiosis. However, it has been challenging to clarify which specific intestinal microbiota and their metabolites play a crucial role in the antidiarrhea process of weaned piglets. Results In this study, we first observed that piglets with diarrhea exhibited a lower average daily gain and higher diarrhea score, and elevated levels of lipopolysaccharide (LPS) and D-lactate (D-LA) compared to healthy piglets. Subsequently, we analyzed the differences in intestinal microbial composition and metabolite levels between healthy and diarrheal weaned piglets. Diarrheal piglets demonstrated intestinal microbiota dysbiosis, characterized primarily by a higher Firmicutes to Bacteroidota ratio, a deficiency of Lactobacillus amylovorus and Lactobacillus reuteri, and an increased abundance of Bacteroides sp.HF-5287 and Bacteroides thetaiotaomicron. Functional profiling of the gut microbiota based on Kyoto Encyclopedia of Genes and Genomes (KEGG) data was performed, and the results showed that tryptophan metabolism was the most significantly inhibited pathway in piglets with diarrhea. Most tryptophan metabolites were detected at lower concentrations in diarrheal piglets than in healthy piglets. Furthermore, we explored the effects of dietary indole-3-aldehyde (IAld), a key tryptophan metabolite, on intestinal development and gut barrier function in weaned piglets. Supplementation with 100 mg/kg IAld in the diet increased the small intestine index and improved intestinal barrier function by promoting intestinal stem cell (ISC) expansion in piglets. The promotion of ISC expansion by IAld was also confirmed in porcine intestinal organoids. Conclusions These findings revealed that intestinal microbial tryptophan metabolite IAld alleviates impaired intestinal development by promoting ISC expansion in weaned piglets.

Abstract: Abstract Extracellular vesicles (EVs) carry a variety of bioactive molecules and are becoming a promising alternative to cell therapy. Scale-up EV isolation is necessary for their functional studies and biological applications, while the traditional methods are challenged by low throughput, low yield, and potential damage. Herein, we developed an ion osmolarity-driven sequential concentration-enrichment strategy (IOSCE) for the EV isolation. IOSCE is composed of a novel superabsorbent polymers (SAPs) for EV concentration and a charged polymer for EV enrichment. Based on the driving force of ionic osmotic pressure, IOSCE can isolate EVs on a large scale from cell culture medium. The saturated water absorption capacity of IOSCE is 13.62 times higher than that of commercial SAPs. Compared with the ultracentrifugation method, IOSCE exhibited a 2.64 times higher yield (6.33 × 108 particles/mL). Moreover, the mesenchymal stem cell-derived EVs isolated using IOSCE demonstrate strong biological activity and can reduce neuroinflammation by affecting RNA metabolism and translation processes. IOSCE provides a cost-effective, high-throughput, and low-damage method for the scale up EV isolation, which is promising for disease diagnosis and treatment. Graphical Abstract

Abstract: Abstract Long non-coding RNAs (lncRNAs) are emerging as crucial regulators of gene expression through diverse mechanisms, including regulation of protein localization, sequestration of miRNAs, recruitment of chromatin modifiers, and modulation of signaling pathways. Accumulating evidence highlights their pivotal roles in tumor initiation, progression, and the development of therapeutic resistance. In this review, we comprehensively summarized the existing literature to identify lncRNAs associated with treatment responses in non-small cell lung cancer (NSCLC). Specifically, we categorized these lncRNAs based on their mechanisms of action in mediating resistance to chemotherapy, targeted therapy, and radiotherapy. Our analysis revealed that aberrant expression of various lncRNAs contributes to the development, metastasis, and therapeutic resistance in NSCLC, ultimately leading to poor clinical outcomes. By elucidating the intricate mechanisms through which lncRNAs modulate therapeutic responses, this review aims to provide mechanistic insights into the heterogeneous treatment outcomes observed in NSCLC patients and unveil potential therapeutic targets for overcoming drug resistance.

Abstract: The AlN particles reinforced Mg-10Gd-3Y–1Zn (GWZ) composites were prepared by mixing AlN/Al particles using ball milling followed by stirring casting method. The influence of AlN/Al particles on the microstructural evolution and high-temperature mechanical properties of GWZ alloys were revealed and discussed. With AlN/Al particles added into the as-cast alloys, the Mg3RE phase was transformed into lamellar LPSO phase, while the content of Al2RE and LPSO phase increased. Since the Al2RE and AlN were effective nucleation sites for α-Mg, the grain size of AlN/Al-GWZ composites was reduced obviously from 126.1 to 39.8 μm. After homogenization, due to the coherent lattice matching relationships between 18R-LPSO phase and Al2RE or AlN particles, the 18R-LPSO phase nucleated on the Al2RE and AlN particles. After ageing treatment, the 2AlN/Al-GWZ composite showed the optimum mechanical properties at 250 °C, with the yield strength, ultimate tensile strength and elongation of 247.1 MPa, 265.5 MPa and 4.0%, respectively. The strengthening effect was primarily originated from the AlN particles, Al2RE and LPSO phases in the 2AlN/Al-GWZ composite. Besides, the composites can achieve the enhancement of high-temperature strength without the sacrifice of plasticity due to the coordinated deformation of AlN particles as well as the refinement of grains.

46. Longitudinal changes of serum metabolomic profile after laparoscopic sleeve gastrectomy in obesity

Abstract: Background: Bariatric surgery induces significant weight loss, increases insulin sensitivity, and improves dyslipidemia. As one of the most widely performed bariatric surgeries, laparoscopic sleeve gastrectomy (LSG) is thought to improve the metabolic profile along with weight loss. The objective of this study was to evaluate longitudinal changes in the serum metabolite levels after LSG and elucidate the underlying mechanisms of metabolic improvement. Methods: Clinical metabolic parameters and serum samples were collected preoperatively and at 1, 3, and 6 months postoperatively from nine patients with obesity undergoing LSG. Serum metabolites were measured using a non-targeted metabolic liquid chromatography–mass spectrometry method. Results: During the 1, 3, and 6 months postoperative follow-up, the body mass index, HOMA-IR, and liver fat content showed a gradual descending trend. A total of 328 serum metabolites were detected, and 38 were differentially expressed. The up-regulated metabolites were mainly enriched in ketone body metabolism, alpha-linolenic acid and linoleic acid metabolism, pantothenate and CoA biosynthesis, glycerolipid metabolism, and fructose and mannose degradation, while the down-regulated metabolites were closely related to caffeine metabolism, oxidation of branched-chain fatty acids, glutamate metabolism, and homocysteine degradation. Notably, nine metabolites (oxoglutarate, 2-ketobutyric acid, succinic acid semialdehyde, phthalic acid, pantetheine, eicosapentaenoate, 3-hydroxybutanoate, oxamic acid, and dihydroxyfumarate) showed persistent differential expression at 1, 3, and 6 months follow-up. Some were found to be significantly associated with weight loss, insulin resistance improvement, and liver fat content reduction. Conclusions: This finding may provide a new perspective for revealing novel biomarkers and mechanisms of metabolic improvement in obesity and related comorbidities.

Abstract: Abstract Using the Kreuzer-Skarke database of 4-dimensional reflexive polytopes, we systematically constructed a new database of orientifold Calabi-Yau threefolds with h 1,1(X) ≤ 12. Our approach involved non-trivial ℤ2 involutions, incorporating both divisor exchanges and multi-divisor reflections acting on the Calabi-Yau threefolds. Each proper involution results in an orientifold Calabi-Yau threefolds and we constructed 320, 386, 067 such examples. We developed a novel algorithm that significantly reduces the complexity of determining all the fixed loci under the involutions, and clarifies the types of O-planes. Our results show that under proper involutions, the majority of cases end up with O3/O7-plane systems, and most of these further admit a naive Type IIB string vacua. Additionally, a new type of free action was determined. We also computed the smoothness and the splitting of Hodge numbers in the ℤ2-orbifold limit for these orientifold Calabi-Yau threefolds.

Abstract: Abstract Pantropical climate interactions across ocean basins operate on a wide range of timescales and can improve the accuracy of climate predictions. Here, we show in observations that Central Pacific (CP) El Niño-like sea surface temperature (SST) anomalies have coevolved with tropical South Atlantic SST anomalies on a quasi-decadal (~10-year) timescale over the past seven decades. During the austral autumn–winter season, decadal warm SSTs in the tropical CP effectively induce tropical SST cooling in the South Atlantic, mainly by strengthening the South Atlantic subtropical anticyclone via an extratropical atmospheric wave teleconnection in the southern hemisphere. Partially coupled pacemaker simulations corroborate the observational findings, indicating that tropical CP decadal SSTs play a primary pacing role, while Atlantic feedback is of secondary importance throughout the study period. Our results suggest that the tropical CP could be an important source of decadal predictability for tropical South Atlantic SST and the surrounding climate.

49. Modulation of Autophagy–Lysosome Axis by African Swine Fever Virus and Its Encoded Protein pEP153R

Abstract: The autophagy–lysosome axis is an evolutionarily conserved intracellular degradation pathway which constitutes an important component of host innate immunity against microbial infections. Here, we show that African swine fever virus (ASFV), one of most devastating pathogens to the worldwide swine industry, can reshape the autophagy–lysosome axis by recruiting the critical lysosome membrane proteins (LAMP1 and LAMP2) to viral factories while inhibiting autophagic induction in macrophages. The screening of viral membrane proteins led to the identification of several ASFV membrane proteins, exemplified by viral protein pEP153R, that could significantly alter the subcellular localization of LAMP1/2 when expressed alone in transfected cells. Further analysis showed that pEP153R was also a component of viral factories and could induce endoplasmic reticulum (ER) retention of LAMP1/2, leading to the inhibition of the fusion of autophagosomes with lysosomes. Interestingly, the ASFV mutant lacking EP153R could still actively recruit LAMP into viral factories (VFs) and inhibit autophagic flux, indicating the existence of a functional redundancy of other viral proteins in the absence of pEP153R and highlighting the complexity of ASFV replication biology. Taken together, our results reveal novel information about the interplay of ASFV with the autophagy–lysosome axis and a previously unrecognized function of ASFV protein pEP153R in regulating the cellular autophagic process.

50. Osteopetrosis-like disorders induced by osteoblast-specific retinoic acid signaling inhibition in mice

Abstract: Abstract Osteopetrosis is an inherited metabolic disease, characterized by increased bone density and narrow marrow cavity. Patients with severe osteopetrosis exhibit abnormal bone brittleness, anemia, and infection complications, which commonly cause death within the first decade of life. Pathologically, osteopetrosis impairs not only the skeletal system, but also the hemopoietic and immune systems during development, while the underlying osteoimmunological mechanisms remain unclear. Osteoclastic mutations are regarded as the major causes of osteopetrosis, while osteoclast non-autonomous theories have been proposed in recent years with unclear underlying mechanisms. Retinoic acid (RA), the metabolite of Vitamin A, is an essential requirement for skeletal and hematopoietic development, through the activation of retinoic acid signaling. RA can relieve osteopetrosis symptoms in some animal models, while its effect on bone health is still controversial and the underlying mechanisms remain unclear. In this study, we constructed an osteoblast-specific inhibitory retinoic acid signaling mouse model and surprisingly found it mimicked the symptoms of osteopetrosis found in clinical cases: dwarfism, increased imperfectly-formed trabecular bone deposition with a reduced marrow cavity, thin cortical bone with a brittle skeleton, and hematopoietic and immune dysfunction. Micro-CT, the three-point bending test, and histological analysis drew a landscape of poor bone quality. Single-cell RNA sequencing (scRNA-seq) of the femur and RNA-seq of osteoblasts uncovered an atlas of pathological skeletal metabolism dysfunction in the mutant mice showing that osteogenesis was impaired in a cell-autonomous manner and osteoclastogenesis was impaired via osteoblast-osteoclast crosstalk. Moreover, scRNA-seq of bone marrow and flow cytometry of peripheral blood, spleen, and bone marrow uncovered pathology in the hematopoietic and immune systems in the mutant mice, mimicking human osteopetrosis. Results showed that hematopoietic progenitors and B lymphocyte differentiation were affected and the osteoblast-dominated cell crosstalk was impaired, which may result from transcriptional impairment of the ligands Pdgfd and Sema4d. In summary, we uncovered previously unreported pathogenesis of osteopetrosis-like disorder in mice with skeletal, hematopoietic, and immune system dysfunction, which was induced by the inhibition of retinoic acid signaling in osteoblasts, and sheds new insights into a potential treatment for osteopetrosis.
