We sought to define the landscape of alternative pre-mRNA splicing in prostate cancers and the relationship of exon choice to known cancer driver alterations. To do so, we compiled a metadataset composed of 876 RNA-sequencing (RNA-Seq) samples from five publicly available sources representing a range of prostate phenotypes from normal tissue to drug-resistant metastases. We subjected these samples to exon-level analysis with rMATS-turbo, purpose-built software designed for large-scale analyses of splicing, and identified 13,149 high-confidence cassette exon events with variable incorporation across samples. We then developed a computational framework, pathway enrichment-guided activity study of alternative splicing (PEGASAS), to correlate transcriptional signatures of 50 different cancer driver pathways with these alternative splicing events. We discovered that Myc signaling was correlated with incorporation of a set of 1,039 cassette exons enriched in genes encoding RNA binding proteins. Using a human prostate epithelial transformation assay, we confirmed the Myc regulation of 147 of these exons, many of which introduced frameshifts or encoded premature stop codons. Our results connect changes in alternative pre-mRNA splicing to oncogenic alterations common in prostate and many other cancers. We also establish a role for Myc in regulating RNA splicing by controlling the incorporation of nonsense-mediated decay-determinant exons in genes encoding RNA binding proteins.

The current research examined the effect of group potential-characterized by upward trend-on individuals' preference for groups. In three experiments, participants (Total N = 189) assessed group members' impressions, anticipated future achievements, and willingness to join high-potential moderate-achievement and moderate-potential high-achievement groups. As predicted, anticipated future achievements led to willingness to join in the high-potential groups. Specifically, compared with high-achievement groups, participants formed better impressions of high-potential groups, anticipated higher future achievements of them, and were more willing to join them. Importantly, anticipated future achievements totally mediated the effect of group potential on willingness to join. A major limitation of RNA sequencing (RNA-seq) analysis of alternative splicing is its reliance on high sequencing coverage. We report DARTS (https://github.com/Xinglab/DARTS), a computational framework that integrates deep-learning-based predictions with empirical RNA-seq evidence to infer differential alternative splicing between biological samples. DARTS leverages public RNA-seq big data to provide a knowledge base of splicing regulation via deep learning, thereby helping researchers better characterize alternative splicing using RNA-seq datasets even with modest coverage.

Electrically controlled droplet manipulation is attractive for lots of applications ranging from material synthesis and drug delivery. However, the present techniques using bulky and expensive power supplies to generate high‐voltage electrical signals are easy to cause electrode electrolysis, bubble formation in buffer solution, and are incapable for point‐of‐care testing. Herein, a novel and portable electrical method for self‐powered emulsion droplet core coalescence and release based on triboelectric nanogenerator (TENG) is presented. The electrical signal with a 2.25 kV open‐voltage and a 35 µA short‐circuit current can be conveniently generated via frictional electrification effect, and the harmful electrochemical reaction in solutions can be avoided. The high transient pulse voltage and stable sinusoidal signal between two electrodes in microfluidic device can generate a strong electric field, which causes apparent Maxwell electrical stresses at droplet interfaces and subsequently promotes the core coalescence and release of double‐emulsions. By adjusting the voltage magnitude, the core coalescence and release behaviors of double‐emulsion droplets can be controllably achieved, followed by the synthesis copper‐based metal–organic framework (Cu‐MOF) nanoparticles.
