Studies that focus on brazing high-nitrogen steel (HNS) to high-entropy alloys (HEAs) are currently limited. In this study, the HEA Al0·3CoCrFeNi was successfully brazed to HNS using a commercial BNi-2 filler at 1080–1140 °C. The microstructure and shear strength of the Al0·3CoCrFeNi/HNS brazed joints were observed and evaluated. The typical microstructure of the brazed joint was HNS/infiltration layer/reaction layer/Ni(s,s) (containing NiAl3)/reaction layer/infiltration layer/Al0·3CoCrFeNi. The higher brazing temperature and longer holding time promoted the diffusion of B as a brazing seam with homogeneous Ni(s,s), and more intergranular Cr-rich borides formed in the base materials. The optimized shear strength attained 549.2 MPa after the joint brazed at 1120 °C for 15 min, and ductile fracture occurred in the reaction zone. The insufficient and excessive diffusion of B resulted in residual Cr–B compounds in the brazing seam and excess borides in the HNS, respectively, which negatively affected shear strength. All the joints showed poor corrosion resistance following immersion in a 3.5 wt% NaCl solution because the excess Cr-rich compounds resulted in decreased Cr content in the surrounding matrix. Therefore, Ni-based fillers without B were more suitable for obtaining Al0·3CoCrFeNi/HNS joints with a favorable corrosion resistance.

A gradual increase in avian influenza outbreaks has been found in recent years. It is highly possible to trigger the next human pandemic due to the characteristics of antigenic drift and antigenic shift in avian influenza virus (AIV). Although great improvements in understanding influenza viruses and the associated diseases have been unraveled, our knowledge of how these viruses impact the gut microbiome of both poultry and humans, as well as the underlying mechanisms, is still improving. The “One Health” approach shows better vitality in monitoring and mitigating the risk of avian influenza, which requires a multi-sectoral effort and highlights the interconnection of human health with environmental sustainability and animal health. Therefore, monitoring the gut microbiome may serve as a sentinel for protecting the common health of the environment, animals, and humans. This review summarizes the interactions between AIV infection and the gut microbiome of poultry and humans and their potential mechanisms. With the presented suggestions, we hope to address the current major challenges in the surveillance and prevention of microbiome-related avian influenza with the “One Health” approach.

Interest point detection and matching play an extremely important role in many computer vision applications. In recent years, deep learning-based algorithms are being used for interest point detection, of which SuperPoint is the most notable algorithm. Although SuperPoint has achieved good results, its interest point detection accuracy is limited to the pixel level; additionally, the simple rounding of noninteger ground truth coordinates in this algorithm results in the loss of decimal information, introducing quantization errors. To overcome these limitations, this study introduces the subpixel module and randomized round encoding methods to reduce the prediction error and quantization error in SuperPoint, respectively. First, we propose a differentiable decoder, soft-random to achieve subpixel-level accuracy of interest point detection. Additionally, to further reduce the interest point localization error, we propose the reprojection homography adaptation of the training step based on SuperPoint’s homography adaptation that is, weincorporate the reprojection error into the algorithm’s training loss. The optimized algorithm proposed in this study, called SRSP, is tested on the HPatches dataset and Euroc dataset. The results showed that SRSP performed better than SuperPoint regarding all indicators, indicating the effectiveness of SRSP on both image matching dataset and real-world dataset.

Abstract Ni-based hydrogen oxidation reaction (HOR) electrocatalysts are promising anode materials for the anion exchange membrane fuel cells (AEMFCs), but their application is hindered by their inherent instability for practical operations. Here, we report a TiO2 supported Ni4Mo (Ni4Mo/TiO2) catalyst that can effectively catalyze HOR in alkaline electrolyte with a mass activity of 10.1 ± 0.9 A g−1 Ni and remain active even up to 1.2 V. The Ni4Mo/TiO2 anode AEMFC delivers a peak power density of 520 mW cm−2 and durability at 400 mA cm−2 for nearly 100 h. The origin for the enhanced activity and stability is attributed to the down-shifted d band center, caused by the efficient charge transfer from TiO2 to Ni. The modulated electronic structure weakens the binding strength of oxygen species, rendering a high stability. The Ni4Mo/TiO2 has achieved greatly improved stability both in half cell and single AEMFC tests, and made a step forward for feasibility of efficient and durable AEMFCs.

This study provides a spatial configuration method to improve the cooling efficiency of multiple VRV outdoor units placed on equipment layers with high floors. Relevant factors include wind parameters, the placement of multiple outdoor units, and louver. A total of 96 cases were designed. CFD simulations were used to obtain the inlet air temperature distributions of multiple outdoor units and then calculate their cooling efficiency. The results found that these factors have effects on the average cooling efficiency of outdoor units in a single row to a certain extent. The influencing degrees of these factors, from large to small, were the louver angle, wind parameters, and the placement of multiple outdoor units. When the cooling efficiency of outdoor units was maximum and the louver angle was 15°, the louvers could be oriented perpendicular to the dominant wind direction (90°) when wind speed was ≥6 m/s, and the unit spacing was 600 mm. Based on this, when the number of outdoor units was expanded in the limited space, staggered arrangements with different directions of heat exchange surfaces were a recommended optimization layout. This study provides technical support for improving the working efficiency of VRV outdoor units in an equipment layer.

This study evaluated the differences in oral processing and texture perception of breads with varying compositions. The research investigated the dynamic changes in moisture content (MC), reducing sugars (RSs), and chewiness of the bolus formed from white bread (B0) and 50% whole-wheat bread (B50) during oral processing. Hyperspectral imaging (HSI) combined with chemometric methods was used to establish quantitative prediction models for MC, RSs, and chewiness, and to create visual distribution maps of these parameters. The results showed that B0 had a higher moisture content and a faster hydration rate than B50 during the initial stages of oral processing, indicating greater hydrophilicity and ease of saliva wetting. Additionally, the uniformity of moisture distribution in the bolus of B0 was higher than that of B50. B50 exhibited significantly lower RSs content and poorer distribution uniformity compared to B0. The primary differences in chewiness between the two types of bread were observed during the early stages of oral processing, with B50 requiring more chewing effort initially. This study demonstrated that HSI technology can effectively monitor and elucidate the compositional changes in food particles during oral processing, providing new insights into bread texture perception and offering a scientific basis for improving bread processing and texture.

Mycotoxin contamination in grain has been an ongoing concern in the world. Wheat, as a staple crop in China, is particularly notable for its mycotoxin contamination. The main mycotoxins in wheat include deoxynivalenol (DON) and its derivates, zearalenone (ZEN) and aflatoxin B1 (AFB1). After harvest, drying process is an effective technique and a necessary step to ensure the long-term safe storage of wheat. In this study, the moisture content, the concentrations of total fungi and main mycotoxins in post-harvest wheat of three wheat growing areas in the North China Plain were examined, and the effect of different drying methods on wheat quality was evaluated. The results showed that 87.5% of wheat samples were simultaneously contaminated with two or more mycotoxins. Due to the pre-harvest heavy rainfall, the moisture content, the levels of total fungi and mycotoxins in wheat samples of Liaocheng city were significantly higher compared to other regions. Moreover, the effects of different drying methods on the starch gelatinization and viscosity properties of wheat were investigated. The results showed that both natural air drying and dryer drying altered the crystal structure within starch particles and affected the gelatinization and viscosity properties of wheat starch. However, there is no significant difference between the wheat samples treated with two drying methods.

IntroductionThe study assessed the correlation and concordance of 25-hydroxyvitamin D [25(OH)D] levels in capillary and venous plasma collected simultaneously after vitamin D3 supplementation in 42 healthy adults. They were randomly divided into three groups by random number table method. Group A took 1,000 IU vitamin D3 daily, group B took 10,000 IU vitamin D3 every 10 days, and group C took 30,000 IU vitamin D3 every 30 days until the end of the 12th month. Venous blood serum 25(OH)D level was detected by chemiluminescence immunoassay (CLIA) and mass spectrometry (LC-MS) at day 1, day 14, day 28, month 6, and month 12 respectively, the capillary blood serum 25(OH)D level was detected by chemiluminescence immunoassay (CLIA) at the same time. Pearson correlation analysis and linear regression analysis were employed to investigate the relationship and transformation equation between the findings of the two samples and the results obtained from different detection methods within the same sample. The Bland-Altman method, Kappa analysis, and receiver operating characteristic (ROC) curve were utilized for assessing consistency, sensitivity, and specificity.ResultsThe three groups all reached a stable peak at 6 months, and the average levels of the three groups were 49.21, 42.50 and 43.025 nmol/L, respectively. The average levels of group A were higher than those of group B and group C (P < 0.001). The mean values of serum 25(OH)D measured by LC-MS and CLIA in 42 healthy adults were 45.32 nmol/L and 49.88 nmol/L, respectively, and the mean values of 25(OH)D measured by LC-MS in capillary blood were 52.03 nmol/L, and the difference was statistically significant (P < 0.001). Pearson correlation analysis showed that the linear fitting formula of scatter data was as follows: venous 25(OH)D concentration (nmol/L) = 1.105 * capillary 25(OH)D concentration −7.532 nmol/L, R2 = 0.625. Good agreement was observed between venous and corrected capillary 25(OH)D levels in clinical diagnosis (Kappa value 0.75). The adjusted serum 25(OH)D in capillary blood had a high clinical predictive value.ConclusionsThe agreement between the two methods is good when the measured 25(OH)D level is higher. Standardized capillary blood chemiluminescence method can be used for 25(OH)D detection.

Abstract Urogenital congenital anomalies (UCAs) is defined as “any live-birth with a urinary or genital condition” and affects millions of men and women worldwide. However, sex differences and related environmental risk factors in UCAs burden on a global scale have not been assessed. Using data from the Global Burden of Diseases, Injuries, and Risk Factors Study (GBD) 2019, we estimated prevalence, incidence, mortality and disability-adjusted life years (DALYs) of UCAs from 1990 to 2019 by sex, region, and socio-demographic Index (SDI) in 204 countries and territories. The disease burden of UCAs was also estimated attributable to each risk factor were estimated according to risk exposure. In 2019, UCAs caused 10,200 all-ages deaths (95% UI 7550–13,400). The combined global incidence rate was 8.38 per 1000 (95% UI 5.88–12.0) live births. The ASIR increased slightly, while the ASDR decreased from 1990 to 2019.The UCAs burden varies greatly depending on the development level and geographical location. The UCAs burden was significantly higher in men than in women, and the sex differences showed an enlarging trend. Health risks and issues, including pollution, child and maternal malnutrition, diet habits, unsafe sanitation and water source, were detected to be positively related to UCAs burden. Albeit the age-standardised prevalence, mortality, incidence, and DALYs of UCAs have decreased, they still cause a public health challenge worldwide. The high deaths and DALYs rates in low and low-middle SDI countries highlight the urgent need for improved preventive, diagnostic, and therapeutic measures. Global strategies for enhancing water safety, reducing pollution, and healthy diets are crucial steps in reducing the burden of UCAs.

Objective: To assess the anti-aging effect and mechanism underlying the effects of ethanol extract from Clinacanthus nutans (CNEE) using Caenorhabditis elegans as the model organism. Methods: The anti-aging effects of ethanol extract from Clinacanthus nutans on nematodes were detected by experiments to assay longevity, oviposition, and exercise. The antioxidant capacity of the ethanolic extracts from Clinacanthus nutans and their mechanisms of action in delaying aging were investigated by assessing the acute oxidative stress, determining the antioxidant enzyme activity, and determining the reactive oxygen species (ROS) levels along with real-time fluorescence quantitative PCR experiments. Results: The ethanol extract from Clinacanthus nutans at 100 μg/mL could prolong the life span of N2 wild type nematode (P0.05). The extract also increased nematode longevity (P0.05). Conclusion: The ethanol extract from Clinacanthus nutans enhanced the ability of nematodes to resist aging and the underlying mechanism of action may be related to the regulation of oxidative stress by daf-16.

Abstract Epidemiological studies have confirmed salt sensitivity as a crucial risk factor for the development of hypertension. However, few studies have investigated the association between salt sensitivity of blood pressure (SSBP) and hypertension in Chinese Tibetan population. Therefore, we conducted a cross‐sectional study based on a Tibetan population to evaluate the association between SSBP and the risk of hypertension. Seven hundred and eighty‐four participants with hypertension and 645 participants without hypertension were included from five villages in Tibetan Autonomous Region of Gannan during 2013–2014. The assessment of salt sensitivity (SS) and non‐salt sensitivity (NSS) was performed according to mean arterial pressure (MAP) changes by the modified Sullivan's acute oral saline load and diuresis shrinkage test (MSAOSL‐DST). Logistic regression models and restricted cubic models were used to examine the association between SSBP and hypertension. There were 554 (70.5%) salt‐sensitive participants with hypertension and 412 (63.9%) salt‐sensitive participants without hypertension in this study. Compared with individuals with NSS, individuals with SS had a significantly increased risk of hypertension, and the multiple‐adjusted odds ratios were 2.582 with 95% confidence interval of 1.357–4.912. Furthermore, a significant linear trend was found between MAP changes and hypertension. Subgroup analyses showed significant and stronger associations between SSBP and the risk of hypertension in the older (age ≥ 55 years old), males and participants who took exercise less than 1 time per week. Our results suggest that SS is associated with an increased risk of hypertension in Tibetan population, indicating a need for clinicians dealing with SSBP to decrease the risk of hypertension.

Abstract Surfactin has the potential to be used as a food preservative. However, efficiency and action mechanism in various applications need more assessments and research. In this study, the antifungal effects and the mechanism of action of surfactin on the fungus Botrytis cinerea were investigated. The effects of applying surfactin for the removal of gray mold on the quality of winter jujube were investigated based on the changes in fruit fatty acids. The results showed that (1) surfactin significantly inhibited the growth of B. cinerea, the EC50 at 5 d was 46.42 mg/L. (2) Surfactin significantly reduced the disease incidence and diameter of gray mold-inoculated winter jujube in a concentration-dependent manner. For that treated with surfactin at the EC50, the incidence decreased by 38.89%. (3) For B. cinerea under surfactin treatment, the mycelial morphology changed, the levels of total lipids and ergosterol decreased, the reactive oxygen species levels increased, and the cell integrity was completely damaged. (4) For winter jujube inoculated by B. cinerea, the contents of saturated fatty acids decreased and unsaturated fatty acids increased. For those under the surfactin treatments, winter jujube maintained the fatty acid composition at the level of non-inoculated groups. Mechanical injury significantly changed the fatty acid composition of winter jujube; however, surfactin not only was able to inhibit the growth of gray mold but also mitigated the adverse effects from mechanical injury. The present study demonstrated the potential applications of surfactin in the preservation of postharvest fruit quality.

Inadequate invasion and excessive apoptosis of trophoblast cells are associated with the development of preeclampsia. Vitamin D deficiency in pregnant women may lead to an increased risk of preeclampsia. However, the underlying mechanisms by which vitamin D is effective in preventing preeclampsia are not fully understood. The objectives of this study were to investigate the role of lysosome-associated membrane glycoprotein 3 (LAMP3) in the pathogenesis of preeclampsia and to evaluate whether vitamin D supplementation would protect against the development of preeclampsia by regulating LAMP3 expression. Firstly, the mRNA and protein levels of LAMP3 were significantly upregulated in the placentas of preeclampsia patients compared to normal placentas, especially in trophoblast cells (a key component of the human placenta). In the hypoxia/reoxygenation (H/R)-exposed HTR-8/Svneo trophoblast cells, LAMP3 expression was also upregulated. H/R exposure repressed cell viability and invasion and increased apoptosis of trophoblast cells. siRNA-mediated knockdown of LAMP3 increased cell viability and invasion and suppressed apoptosis of H/R-exposed trophoblast cells. We further found that 1,25(OH)2D3 (the hormonally active form of vitamin D) treatment reduced LAMP3 expression in H/R exposed trophoblast cells. In addition, 1,25(OH)2D3 treatment promoted cell viability and invasion and inhibited apoptosis of H/R-exposed trophoblast cells. Notably, overexpression of LAMP3 abrogated the protective effect of 1,25(OH)2D3 on H/R-exposed trophoblast cells. Collectively, we demonstrated trophoblast cytoprotection by vitamin D, a process mediated via LAMP3.

The BBSome is an octameric protein complex that regulates ciliary transport and signaling. Mutations in BBSome subunits are closely associated with ciliary defects and lead to ciliopathies, notably Bardet-Biedl syndrome. Over the past few years, there has been significant progress in elucidating the molecular organization and functions of the BBSome complex. An improved understanding of BBSome-mediated biological events and molecular mechanisms is expected to help advance the development of diagnostic and therapeutic approaches for BBSome-related diseases. Here, we review the current literature on the structural assembly, transport regulation, and molecular functions of the BBSome, emphasizing its roles in cilium-related processes. We also provide perspectives on the pathological role of the BBSome in ciliopathies as well as how these can be exploited for therapeutic benefit.

Previous studies on the association of endocrine-disrupting chemicals (EDCs) with metabolic dysfunction-associated fatty liver disease (MAFLD) are very limited. This study analyzed the association of EDCs exposure with MAFLD among 5073 American adults from the 2017–2018 National Health and Nutrition Examination Survey. The results showed that increased exposure to 3 EDCs metabolites (namely As, DiNP and PFOA) were significantly associated with MAFLD, the odds ratio of which were 1.819 (95% CI: 1.224, 2.702), 1.959 (95% CI: 1.224, 3.136) and 2.148 (95% CI: 1.036, 4.456), respectively. Further, the bayesian kernel machine regression model also revealed that phthalates exposure was strongly connected with the MAFLD, particularly in females and the elderly over 65. Moderating effect analysis suggested that higher body mass index (BMI) and inflammatory diet habit (indicated by dietary inflammatory index) strengthened the association between EDCs and MAFLD, whereas population with higher level of insulin sensitivity showed lower risk. In conclusion, our results suggest that either single or combined exposure to EDCs metabolites is link to MAFLD. Our findings also encourage people to sustain a healthy diet, normal levels of insulin sensitivity and BMI, which may help to alleviate the association of MAFLD risk in exposure to EDCs. These results also help us to better understand the association of EDCs and MAFLD and provide effective evidences for preventing MAFLD from the EDCs exposure aspect.

BackgroundIndoor air pollution is an important risk factor affecting health of the respiratory system. Studies on indoor air pollution in China are mostly limited to the central and eastern regions, and there are few studies in the rural areas of northwest China. ObjectiveTo explore the influencing factors of lung ventilation function and its relationship with indoor air pollution in rural areas of Gansu Province based on a cross-sectional investigation. MethodsA total of 399 subjects were selected from four villages in Baiyin and Yuzhong of Gansu Province. Questionnaires were used to collect demographic information, lifestyle, disease history, fuel use, and other information, and physical and functional tests were ordered such as height, weight, and lung function. The Indoor Air Pollution (IAP) exposure index was calculated based on smoking, fuel type, and weekly ventilation. IAP > 5 was defined as a high level of indoor air pollution. Lung function indexes included forced vital capacity (FVC), forced expiratory volume in the first second (FEV1), FEV1/FVC, forced vital capacity as a percentage of predicted value (FVC%), and forced expiratory volume in the first second as a percentage of predicted value (FEV1%), which were used to determine pulmonary ventilation dysfunction. Logistic regression model was used to evaluate the relationship between indoor air pollution and pulmonary ventilation function, and subgroup analysis was further conducted according to home address and BMI , in order to identify the high-risk population of pulmonary ventilation dysfunction. ResultsThe mean age of the subjects was (56.75±7.31) years old; 155 subjects (38.85%) had normal pulmonary ventilation function, and the other 244 subjects (61.14%) had impaired pulmonary ventilation function; about 89.97% of the subjects were exposed to high level of indoor air pollution (IAP > 5). We found that IAP > 5 (OR=2.327, 95%CI: 1.089-4.974) and use of bituminous coal as the main heating fuel in winter (OR=3.467, 95%CI: 1.197-10.037) increased the risk of pulmonary ventilation dysfunction after adjusting for age, BMI, residence, gender, smoking, drinking, and cardiovascular disease. The subgroup analysis results showed that no ventilation in the living room/bedroom (OR=3.460, 95%CI: 1.116-10.268) increased the risk of pulmonary ventilation dysfunction in Baiyin. Heating with coal stoves and Chinese Kang in the bedroom (OR=2.092, 95%CI: 1.030-4.247) and cooking in the bedroom in winter ( OR =2.954, 95% CI : 1.046-8.344) also increased the risk of pulmonary ventilation dysfunction in the residents with BMI≤24 kg·m−2. IAP > 5 (OR=3.739, 95%CI: 1.147-12.182) was associated with a significantly increased risk of pulmonary ventilation dysfunction in the BMI > 24 kg·m−2 subgroup. ConclusionThe pulmonary ventilation function of rural residents in Gansu is poor, which is negatively correlated with indoor air pollution. Coal use, overweight, cooking in bedroom, and use of coal stoves and Chinese Kang for heating may increase the risk of pulmonary ventilation dysfunction, while room ventilation is a beneficial factor.

Endothelial cell apoptosis is an important pathophysiology in many cardiovascular diseases. The gasotransmitter nitric oxide (NO) is known to regulate cell survival and apoptosis. However, the mechanism underlying the effect of NO remains unclear. In this research, by targeting cytosolic copper/zinc superoxide dismutase (SOD1) monomerization, we aimed to explore how NO inhibited endothelial cell apoptosis. We showed that treatment with the NO synthase (NOS) inhibitor nomega‐nitro‐l‐arginine methyl ester hydrochloride (L‐NAME) significantly decreased the endogenous NO content of endothelial cells, facilitated the formation of SOD1 monomers, inhibited dismutase activity, and promoted reactive oxygen species (ROS) accumulation in human umbilical vein endothelial cells (HUVECs); by contrast, supplementation with the NO donor sodium nitroprusside (SNP) upregulated NO content, prevented the formation of SOD1 monomers, enhanced dismutase activity, and reduced ROS accumulation in L‐NAME‐treated HUVECs. Mechanistically, tris(2‐carboxyethyl) phosphine hydrochloride (TCEP), a specific reducer of cysteine thiol, increased SOD1 monomer formation, thus preventing the NO‐induced increase in dismutase activity and the decrease in ROS. Furthermore, SNP inhibited HUVEC apoptosis caused by the decrease in endogenous NO, whereas TCEP abolished this protective effect of SNP. In summary, our data reveal that NO protects endothelial cells against apoptosis by inhibiting cysteine‐dependent SOD1 monomerization to enhance SOD1 activity and inhibit oxidative stress.

Abstract Background Global developmental delay (GDD) has a heterogeneous clinical profile among patients, accounting for approximately 1%–3% of cases in children. An increasing number of gene defects have been demonstrated to be associated with GDD; up to now, only limited studies have reported developmental disorders driven by WDR45B. Methods Trio‐whole exome sequencing (Trio‐WES) was performed for the patient and her family. All variants with a minor allele frequency C (p. Arg226Thr)] was identified from the proband. The variant was absent in published databases such as gnomAD and Exome Aggregation Consortium (ExAC). The variant was predicted to be damaging for proteins and classified as VUS according to the ACMG guidelines. We reviewed the literature, and the development delay level in our case was less severe than the other reported cases. Conclusion We reported another case with a novel homozygous variant of WDR45B and showed the heterogeneity of clinical features.

Maturity is a crucial indicator in assessing the quality of tomatoes, and it is closely related to lycopene content. Using hyperspectral imaging, this study aimed to monitor tomato maturity and predict its lycopene content at different maturity stages. Standard normal variable (SNV) transformation was applied to preprocess the hyperspectral data. Then, using competitive adaptive reweighted sampling (CARS), the characteristic wavelengths were selected to simplify the calibration models. Based on the full and characteristic wavelengths, a support vector classifier (SVC) model was developed to determine tomato maturity qualitatively. The results demonstrated that the classification accuracy using the characteristic wavelength led to the obtention of better results with an accuracy of 95.83%. In addition, the support vector regression (SVR) and partial least squares regression (PLSR) models were utilized to predict lycopene content. With a coefficient of determination for prediction (R2P) of 0.9652 and a root mean square error for prediction (RMSEP) of 0.0166 mg/kg, the SVR model exhibited the best quantitative prediction capacity based on the characteristic wavelengths. Following this, a visual distribution map was created to evaluate the lycopene content in tomato fruit intuitively. The results demonstrated the viability of hyperspectral imaging for detecting tomato maturity and quantitatively predicting the lycopene content during storage.

Introduction: The proliferation of vascular smooth muscle cells (VSMCs) is an important physiological and pathological basis for many cardiovascular diseases. Endogenous hydrogen sulfide (H2S), the third gasotransmitter, is found to preserve vascular structure by inhibiting VSMC proliferation. However, the mechanism by which H2S suppresses VSMC proliferation has not been fully clear. Objectives: This study aimed to explore whether H2S persulfidates the transcription factor FOXO1 to inhibit VSMC proliferation. Methods: After the proliferation of VSMC A7r5 cells was induced by endothelin-1 (ET-1), FOXO1 phosphorylation and proliferating cell nuclear antigen (PCNA) expression were detected by Western blotting, the degree of FOXO1 nuclear exclusion and PCNA fluorescent signals in the nucleus were detected by immunofluorescence, and the persulfidation of FOXO1 was measured through a biotin switch assay. Results: The results showed that ET-1 stimulation increased cell proliferation, FOXO1 phosphorylation and FOXO1 nuclear exclusion to the cytoplasm in the cells. However, pretreatment with NaHS, an H2S donor, successfully abolished the ET-1-induced increases in the VSMC proliferation, FOXO1 phosphorylation, and FOXO1 nuclear exclusion to the cytoplasm. Mechanistically, H2S persulfidated the FOXO1 protein in A7r5 and 293T cells, and the thiol reductant DTT reversed this effect. Furthermore, the C457S mutation of FOXO1 abolished the H2S-induced persulfidation of FOXO1 in the cells and the subsequent inhibitory effects on FOXO1 phosphorylation at Ser256, FOXO1 nuclear exclusion to the cytoplasm and cell proliferation. Conclusion: Thus, our findings demonstrated that H2S might inhibit VSMC proliferation by persulfidating FOXO1 at Cys457 and subsequently preventing FOXO1 phosphorylation at Ser256.

Abstract Background Stem cells, including induced pluripotent stem cells (iPSCs), have tremendous potential in health care, though with several significant limitations. Each of the limitations, including immunogenicity, may block most of the therapeutic potentials. Beta2 microglobulin (B2M) and MHC II transactivator (CIITA) are critical for MHC I and II, respectively. MHCs are responsible for immunogenic recognition. Methods B2M and CIITA were knocked out from human iPSCs, either separately or simultaneously. The effects of single or dual knockout of B2M and CIITA on iPSC properties were evaluated in a xenogeneic model of human-to-monkey transplantation. Results B2M or CIITA knockout in human induced pluripotent stem cells (iPSCs) diminishes the expression of MHC I or II alleles, respectively, without changing iPSC pluripotency. Dual knockout was better than either single knockout in preserving the ability of human iPSCs to reduce infiltration of T and B lymphocytes, survive, and promote wound healing in monkey wound lesions. The knockouts did not affect the xenogeneic iPSC-induced infiltration of macrophages and natural killer cells. They, however, decreased the iPSC-promoted proliferation of allogeneic peripheral blood mononuclear cells and T lymphocytes in vitro, although not so for B lymphocytes isolated from healthy human donors. Although the dual knockout cells survived long enough for suiting therapeutic needs, the cells eventually died, possibly due to innate immune response against them, thereby eliminating long-term risks. Conclusions Having these iPSCs with diminished immunogenicity-recognizable to allogeneic recipient may provide unlimited reproducible, universal, standardized “ready-to-use” iPSCs and relevant derivatives for clinical applications.

Tunnel side ditches in the high-latitude cold regions of China experience severe freezing-induced damage. In this paper, a three-dimensional transient temperature calculation model for tunnel in cold area is proposed, and the reliability of the temperature calculation model is verified by the field test results. The characteristics of tunnel temperature distribution under the influence of many factors, especially tunnel section and tunnel length, which are ignored in the existing research, are studied. The influence of tunnel design parameters on temperature field under different railway tunnel types (single-track and double-track tunnel) was also studied. The results show that the length with a temperature below 0 °C of single-track tunnel increases with the increase of tunnel length, while that of double-track tunnel decreases. No matter what type of tunnel, increasing the train frequency and train speed can significantly increase the freezing range of the tunnel. However, the influence of piston wind on temperature distribution in single-track tunnel and double-track tunnel is very different. For single-track tunnel, the growth rate of the length with a temperature below 0 °C decreases with the increase of train speed; However, for double-track tunnel, the growth rate of the length with a temperature below 0 °C increases with train speed. The frost damage of double-track tunnel in cold area is more serious than that of single-track tunnel. Under the same conditions, the antifreezing length of double-track tunnel is much longer than that of single-track tunnel. Based on the calculation results, a prediction model for the freezing length of cold-region tunnel side ditches was proposed, thereby providing a reference for the prevention and control of freezing-induced damage in the side ditches of cold-region tunnels.

Transcranial direct current stimulation (tDCS) has been applied in training and competition, but its effects on physical performance remain largely unknown. This study aimed to observe the effect of tDCS on muscular strength and knee activation. Nineteen healthy young men were subjected to 20 min of real stimulation (2 mA) and sham stimulation (0 mA) over the primary motor cortex (M1) bilaterally on different days. The maximal voluntary contraction (MVC) of the knee extensors and flexors, and surface electromyography (sEMG) of the rectus femoris (RF) and biceps femoris (BF) were recorded before, immediately after, and 30 min after stimulation. MVC, rate of force development (RFD), and sEMG activity were analyzed before and after each condition. MVC of the non-dominant leg extensor and flexor was significantly higher immediately after real stimulation and 30 min after stimulation than before, and MVC of the non-dominant leg flexor was significantly higher 30 min after real stimulation than that after sham stimulation (P < 0.05). The RFD of the non-dominant leg extensor and flexor immediately after real stimulation was significantly higher than before stimulation, and the RFD of the non-dominant leg extensor immediately after real stimulation and 30 min after stimulation was significantly higher than that of sham stimulation (P < 0.05). EMG analysis showed the root mean square amplitude and mean power frequency (MPF) of the non-dominant BF and RF were significantly higher immediately after real stimulation and 30 min after stimulation than before stimulation, and the MPF of the non-dominant BF EMG was significantly higher 30 min after real stimulation than that after sham stimulation (P < 0.05). Bilateral tDCS of the M1 can significantly improve the muscle strength and explosive force of the non-dominant knee extensor and flexor, which might result from increased recruitment of motor units. This effect can last until 30 min after stimulation, but there is no significant effect on the dominant knee.

Potato flour is an important raw material for potato staple food products; nevertheless, the quality and flavor vary significantly due to process changes. In this study, the physicochemical features of fresh and five different dehydration temperature potato samples, including the degree of starch gelatinization (DG), pasting, structure properties and volatile components, were compared to investigate the effect of hot air drying (HAD) on potato flour. The results showed that the degree of pasting, viscosity and volatile aroma components changed significantly with differences in drying temperature. With the increase in drying temperature, the gelatinization degree and peak viscosity of potato powder increased or decreased, the breakdown viscosity of HAD-50 was higher, the setback viscosity of HAD-90 was higher, while the crystallization zone of HAD-90 was destroyed due to the high temperature. The flavor components of potato flour are increased during processing due to lipid oxidation, Maillard reaction and thermal degradation. The level of aldehydes,3,5-Octadien-2-one and E,E)-3,5-Octadien-2-one gradually reduced as the processing temperature increased, while the content of furans grew and then decreased, nonanal and 2-Penty-l-Furan increased. Overall, lower HAD temperatures are beneficial for the quality and flavor of potato flour. The information presented here will be useful for the further development of potato flour products.

Cardiac involvement, displayed as premature cardiovascular disease (CVD), is one of common clinical symptoms of patients with systemic lupus erythematosus (SLE), contributing to mortality of the disease. The precise underlying pathological mechanism(s) for the cardiac involvement in lupus remains poorly understood. Lipids and their metabolites are directly involved in atherosclerosis development, oxidative stress, and inflammation, which are closely related to the development of CVD. In the study, shotgun lipidomics was exploited to quantitatively analyze cellular lipidomes in the cardiac tissue of MRL/lpr mice at two different time points (i.e., pre-lupus and lupus state) with/without treatment with glucocorticoids (GCs). Urine protein, spleen index, and renal histopathological evaluation of the mice were also performed for assessment of SLE onset and/or outcome. Lipidomics analysis revealed that the deposition of cholesterol and the aberrant metabolism of lipids caused by the increased energy metabolism and the enhanced activation of phospholipases, both of which were originally induced by inflammation, were already present in cardiac tissues from lupus-prone mice even at pre-lupus state. These lipid alterations could further induce inflammation and autoimmune responses, accelerating the process of CVD. In addition, the present study also demonstrated that GCs therapy could not only delay the progression of SLE, but also partially corrected these alterations of lipid species in cardiac tissue due to their anti-inflammatory effect. Thus, the medications with better anti-inflammatory effect might be a useful therapeutic method for premature CVD of SLE.

Atherosclerosis is a chronic inflammatory arterial disease characterized by build-up of atheromatous plaque, which narrows the lumen of arteries. Hypercholesterolemia and excessive oxidative stress in arterial walls are among the main causative factors of atherosclerosis. Transient receptor potential channel M2 (TRPM2) is a Ca2+-permeable cation channel activated by oxidative stress. However, the role of TRPM2 in atherosclerosis in animal models is not well studied. In the present study, with the use of adeno-associated virus (AAV)-PCSK9 and TRPM2 knockout (TRPM2−/−) mice, we determined the role of TRPM2 in hypercholesterolemia-induced atherosclerosis. Our results demonstrated that TRPM2 knockout reduced atherosclerotic plaque area in analysis of En face Oil Red O staining of both whole aortas and aortic-root thin sections. Furthermore, TRPM2 knockout reduced the expression of CD68, α-SMA, and PCNA in the plaque region, suggesting a role of TRPM2 in promoting macrophage infiltration and smooth-muscle cell migration into the lesion area. Moreover, TRPM2 knockout reduced the expression of ICAM-1, MCP-1, and TNFα and decreased the ROS level in the plaque region, suggesting a role of TRPM2 in enhancing monocyte adhesion and promoting vascular inflammation. In bone-marrow-derived macrophages and primary cultured arterial endothelial cells, TRPM2 knockout reduced the production of inflammatory cytokines/factors and decreased ROS production. In addition, a TRPM2 antagonist N-(p-amylcinnamoyl) anthranilic acid (ACA) was able to inhibit atherosclerotic development in an ApoE−/− mouse model of atherosclerosis. Taken together, the findings of our study demonstrated that TRPM2 contributes to the progression of hypercholesterolemia-induced atherosclerosis. Mechanistically, TRPM2 channels may provide an essential link that can connect ROS to Ca2+ and inflammation, consequently promoting atherosclerotic progression.

Biosurfactant has potential application value in the removal of microalgal blooms, but the ecological risks require more research. In this paper, the effects of surfactin on the toxic dinoflagellate Karlodinium veneficum were studied. The coaction of surfactin and K. veneficum was also evaluated through toxicological experiments on Artemia and juvenile clams. The results showed that: (1) in the concentration range of 0–10 mg/L, surfactin significantly killed algal cells in a dose-dependent manner within 48 h; the 24 h EC50 was 3.065 mg/L; (2) K. veneficum had the ability to restore population growth after stress reduction and the restored proliferation was positively correlated with the initial surfactin concentration; (3) the ability to restore population growth was associated with protection afforded by the promotion of antioxidant enzymes, including catalase (CAT), peroxidase (POD) and superoxide dismutase (SOD), whose increase was positively correlated with the surfactin concentration; (4) the toxicity of the coculture of surfactin and K. veneficum was significantly greater than that of the K. veneficum culture or surfactin alone and was dose and time dependent. The potential ecological risks should be considered when applying biosurfactants, such as surfactin, in the removal of harmful algal blooms.

Human epididymis protein 4 (HE4) is well known to be a predictor of ovarian cancer clinically. HE4 is reported to play crucial roles in ovarian cancer progression and metastasis. The purpose of the present study was to explore its biological role and molecular mechanism in ovarian cancer. In our study, we found that expression levels of HE4 in tissues, serum and urine in ovarian cancer were upregulated compared to healthy and benign groups. HE4 expression was elevated in ovarian cancer cells. Knockdown of HE4 dampened cell proliferation and Ki67 expression, as well as enhanced apoptosis, caspase-3 activity and cleaved-caspase-3 expression. In addition, HE4 downregulation repressed invasion and migration capabilities of ovarian cancer cells. Western blot analyses showed that knockdown of HE4 reduced the levels of matrix metalloproteinases (MMP-2 and MMP-9) and inhibited epithelial to mesenchymal transition (EMT) in ovarian cancer cells. In vivo animal experiments revealed that HE4 downregulation constrained the growth of xenograft tumor. Mechanism research showed that knockdown HE4 inhibited the activity of JAK/STAT3 pathway in vitro and in vivo. In conclusion, our findings reported that knockdown of HE4 suppresses aggressive cell growth and malignant progression of ovarian cancer by inhibiting the JAK/STAT3 pathway, which provides valuable insights to contribute to develop novel HE4-targeted therapies.

Objective To investigate tumor and pregnancy outcomes after fertility-sparing surgery for cervical cancer. Methods A total of 83 patients with cervical cancer who received fertility-sparing surgery at 10 gynecologic cancer research centers in Henan Province were enrolled from January 2010 to June 2016. Clinical data and follow-up results were collected. Of them, 78 had cervical squamous carcinoma and five had cervical adenocarcinoma. International Federation of Gynecology and Obstetrics (2009) staging showed that 26 patients had stage IA1, 11 had stage IA2, and 46 had stage IBI. Seventy-two patients underwent radical trachelectomy and retroperitoneal lymphadenectomy, whereas 11 underwent subradical trachelectomy and retroperitoneal lymphadenectomy. Moreover, 17 patients received one to two courses of preoperative neoadjuvant chemotherapy and five received two to four courses of postoperative chemotherapy. Eighty-three patients were followed up postoperatively (median follow-up duration, 36.2 months). Results With regard to tumor outcomes, one (1.2%) patient showed recurrence following fertility-sparing surgery. In 69 patients with planned pregnancy after treatment, 54 had 58 pregnancies, including 42 full-term births and eight premature births. Seventy-nine patients were satisfied with their quality of life. Conclusions Radical/subradical trachelectomy is safe and effective as fertility-sparing surgery for young patients with early cervical cancer, with good pregnancy outcomes.

Sulfur dioxide (SO2) is a colorless and irritating gas. Recent studies indicate that SO2 acts as the gas signal molecule and inhibits vascular smooth muscle cell (VSMC) proliferation. Cell proliferation depends on intracellular pH (pHi). Transmembrane cystein mutation of Na+- independent Cl-/HCO3- exchanger (anion exchanger, AE) affects pHi. However, whether SO2 inhibits VSMC proliferation by reducing pHi is still unknown. Here, we investigated whether SO2 reduced pHi to inhibit the proliferation of VSMCs and explore its molecular mechanisms. Within a range of 50–200 μM, SO2 was found to lower the pHi in VSMCs. Concurrently, NH4Cl pre-perfusion showed that SO2 significantly activated AE, whereas the AE inhibitor 4,4′-diisothiocyanatostilbene- 2,20-disulfonic acid (DIDS) significantly attenuated the effect of SO2 on pHi in VSMCs. While 200 μM SO2 sulphenylated AE2, while dithiothreitol (DTT) blocked the sulphenylation of AE2 and subsequent AE activation by SO2, thereby restoring the pHi in VSMCs. Furthermore, DIDS pretreatment eliminated SO2-induced inhibition of PDGF-BB-stimulated VSMC proliferation. We report for the first time that SO2 inhibits VSMC proliferation in part by direct activation of the AE via posttranslational sulphenylation and induction of intracellular acidification.

Endogenous hydrogen sulfide (H2S) and sulfur dioxide (SO2) are regarded as important regulators to control endothelial cell function and protect endothelial cell against various injuries. In our present study, we aimed to investigate the effect of endogenous H2S on the SO2 generation in the endothelial cells and explore its significance in the endothelial inflammation in vitro and in vivo. The human umbilical vein endothelial cell (HUVEC) line (EA.hy926), primary HUVECs, primary rat pulmonary artery endothelial cells (RPAECs), and purified aspartate aminotransferase (AAT) protein from pig heart were used for in vitro experiments. A rat model of monocrotaline (MCT)-induced pulmonary vascular inflammation was used for in vivo experiments. We found that endogenous H2S deficiency caused by cystathionine-γ-lyase (CSE) knockdown increased endogenous SO2 level in endothelial cells and enhanced the enzymatic activity of AAT, a major SO2 synthesis enzyme, without affecting the expressions of AAT1 and AAT2. While H2S donor could reverse the CSE knockdown-induced increase in the endogenous SO2 level and AAT activity. Moreover, H2S donor directly inhibited the activity of purified AAT protein, which was reversed by a thiol reductant DTT. Mechanistically, H2S donor sulfhydrated the purified AAT1/2 protein and rescued the decrease in the sulfhydration of AAT1/2 protein in the CSE knockdown endothelial cells. Furthermore, an AAT inhibitor l-aspartate-β-hydroxamate (HDX), which blocked the upregulation of endogenous SO2/AAT generation induced by CSE knockdown, aggravated CSE knockdown-activated nuclear factor-κB pathway in the endothelial cells and its downstream inflammatory factors including ICAM-1, TNF-α, and IL-6. In in vivo experiment, H2S donor restored the deficiency of endogenous H2S production induced by MCT, and reversed the upregulation of endogenous SO2/AAT pathway via sulfhydrating AAT1 and AAT2. In accordance with the results of the in vitro experiment, HDX exacerbated the pulmonary vascular inflammation induced by the broken endogenous H2S production in MCT-treated rat. In conclusion, for the first time, the present study showed that H2S inhibited endogenous SO2 generation by inactivating AAT via the sulfhydration of AAT1/2; and the increased endogenous SO2 generation might play a compensatory role when H2S/CSE pathway was downregulated, thereby exerting protective effects in endothelial inflammatory responses in vitro and in vivo.

We wish to identify determinants of endothelial lineage. Murine embryonic stem cells (mESC) were fused with human endothelial cells in stable, non-dividing, heterokaryons. Using RNA-seq, it is possible to discriminate between human and mouse transcripts in these chimeric heterokaryons. We observed a temporal pattern of gene expression in the ESCs of the heterokaryons that recapitulated ontogeny, with early mesodermal factors being expressed before mature endothelial genes. A set of transcriptional factors not known to be involved in endothelial development was upregulated, one of which was POU class 3 homeobox 2 (Pou3f2). We confirmed its importance in differentiation to endothelial lineage via loss- and gain-of-function (LOF and GOF). Its role in vascular development was validated in zebrafish embryos using morpholino oligonucleotides. These studies provide a systematic and mechanistic approach for identifying key regulators in directed differentiation of pluripotent stem cells to somatic cell lineages.

The PdNi film hydrogen sensors with Wheatstone bridge structure were designed and fabricated with the micro-electro-mechanical system (MEMS) technology. The integrated sensors consisted of four PdNi alloy film resistors. The internal two were shielded with silicon nitride film and used as reference resistors, while the others were used for hydrogen sensing. The PdNi alloy films and SiN films were deposited by magnetron sputtering. The morphology and microstructure of the PdNi films were characterized with X-ray diffraction (XRD). For efficient data acquisition, the output signal was converted from resistance to voltage. Hydrogen (H2) sensing properties of PdNi film hydrogen sensors with Wheatstone bridge structure were investigated under different temperatures (30 °C, 50 °C and 70 °C) and H2 concentrations (from 10 ppm to 0.4%). The hydrogen sensor demonstrated distinct response at different hydrogen concentrations and high repeatability in cycle testing under 0.4% H2 concentration. Towards 10 ppm hydrogen, the PdNi film hydrogen sensor had evident and collectable output voltage of 600 μV.

The construction and gradual installation of turbines on wind farms has been hindered by the high cost of the energy production. An effective way to minimize energy costs is via the optimal design of wind turbines and their layout, but relevant and synthetic studies are lacking. This paper proposes a method to minimize the energy cost of offshore wind farms by simultaneously optimizing the rated wind speed, the rotor radius of wind turbines and their layout. Firstly, a new, mixed mathematical formulation of the energy cost is presented, considering the Weibull distribution for wind, the characterizing parameters of wind turbines and the distance between two turbines. Secondly, to obtain the minimum energy cost, a composite optimization algorithm was developed, which consists of an iterative method and an improved particle swarm optimization algorithm. The former was used to search the minimal energy costs that relate to the design parameters of a single wind turbine, while the latter was adopted for optimizing the layout of the wind turbines iteratively. Finally, the proposed method was applied to three case studies with variable wind speed and constant wind direction. Results of the case studies show that the reduced energy cost after optimization has a range of 0⁻0.001 $/kWh, which confirms the effectiveness of the proposed approach. Meanwhile, the layout of the wind turbines after optimization tends to locate the two wind turbines with the biggest spacing in the wind direction, which justifies the utilization of layout optimization. Furthermore, exploring the optimally designed parameters of wind turbines revealed that the wind farms with a high mean wind speed can have a wider range of turbine capacity than those with a low wind speed, which offers more freedom for the designers when constructing offshore wind farms at wind sites with rich wind resources.

OBJECTIVE:To compare the natural fertility outcomes of salpingotomy and salpingectomy among women treated for tubal pregnancy. METHODS:An online database search including PubMed, Embase, CENTRAL and Web of Science was performed to identify studies comparing salpingotomy and salpingectomy to treat women with tubal pregnancy. The search included papers published after the databases were established until May 2015. Two reviewers independently screened literature according to the inclusion and exclusion criteria and then extracted data and assessed the methodological quality of all of the included studies. The meta-analysis was conducted using RevMan 5.3 software. The registration number is CRD42015017545 in PROSPERO. RESULTS:Two randomized controlled trials (RCTs) and eight cohort studies, including a total of 1,229 patients, were znalyzed. The meta-analysis of the RCT subgroup indicated that there was no statistically significant difference in IUP rates (RR = 1.04, 95% CI = 0.89-1.21, P = 0.61) nor the repeat ectopic pregnancy (REP) rate (RR = 1.30, 95% CI = 0.72-2.38, P = 0.39) between the salpingotomy and salpingectomy group. In contrast, the cohort study subgroup analysis revealed that the IUP rate was higher in the salpingotomy group compared with the salpingectomy group (RR = 1.24, 95% CI = 1.08-1.42, P = 0.002); Salpingotomy also increased the risk of REP rate (RR = 2.27, 95% CI = 1.12-4.58, P = 0.02). The persistent ectopic pregnancy (PEP) occurred more frequently in the salpingotomy group than the salpingectomy group (RR = 11.61, 95% CI = 3.17-42.46, P = 0.0002). An IUP would be more likely to occur after salpingotomy than salpingectomy when the follow-up time was more than 36 months (RR = 1.16, 95% CI = 1.02-1.32, P = 0.03). The IUP rate (RR = 1.13, 95% CI = 1.01-1.26, P = 0.03), and the REP rate (RR = 1.62, 95% CI = 1.02-2.56, P = 0.04) was higher after salpingotomy than salpingectomy among patients from Europe compared with those from America. CONCLUSIONS:Based on the available evidence, we believe that for patients with a healthy contralateral tube operated for tubal pregnancy, the subsequent fertility after salpingectomy and salpingotomy are similar in the long term. The fertility prospects will not be improved via salpingotomy compared with salpingectomy.

As onshore wind energy has depleted, the utilization of offshore wind energy has gradually played an important role in globally meeting growing green energy demands. However, the cost of energy (COE) for offshore wind energy is very high compared to the onshore one. To minimize the COE, implementing optimal design of offshore turbines is an effective way, but the relevant studies are lacking. This study proposes a method to minimize the COE of offshore wind turbines, in which two design parameters, including the rated wind speed and rotor radius are optimally designed. Through this study, the relation among the COE and the two design parameters is explored. To this end, based on the power-coefficient power curve model, the annual energy production (AEP) model is designed as a function of the rated wind speed and the Weibull distribution parameters. On the other hand, the detailed cost model of offshore turbines developed by the National Renewable Energy Laboratory is formulated as a function of the rated wind speed and the rotor radius. Then, the COE is formulated as the ratio of the total cost and the AEP. Following that, an iterative method is proposed to search the minimal COE which corresponds to the optimal rated wind speed and rotor radius. Finally, the proposed method has been applied to the wind classes of USA, and some useful findings have been obtained.

Current sharing control is a challenge for a discontinuous conduction mode (DCM) micro-inverter based on interleaved flyback topology. To solve this problem, this study proposes a novel and systemic model-based approach. Firstly, an accurate fourth-order model is presented for the interleaved flyback circuit, which takes the two flybacks’ parameter mismatch and coupling into account. Secondly, based on the presented model, a continuous time sliding mode current controller is proposed to tackle the output imbalance caused by parameter mismatch, coupling and disturbance. The proposed controller is derived from the Lyapunov function without switching conditions. Finally, the effectiveness of the proposed model and control method is validated by simulation tests using MATLAB/SIMULINK. Simulation results show that the proposed approach improves the current sharing for the interleaved flyback micro-inverter when compared to the conventional current sharing approach.

This paper introduces a distributed secondary control algorithm for automatic generation control (AGC) and automatic voltage control (AVC), which aims at matching area generation to area load and minimizing the total generation cost in an alternating current (AC) microgrids. Firstly, the control algorithm utilizes a continuous-time distributed algorithm to generate additional control variables to achieve frequency-voltage recovery for all distributed generators (DGs). Secondary, it solves the economic dispatch problem (EDP) by a distributed economic incremental algorithm in the secondary control level, which avoids the problem caused by communication speed inconsistency between secondary and tertiary control levels. This study also utilizes a fully distributed strategy based on secondary communication network to estimate the total load demand. In addition, the proposed algorithm can be used to realize a seamless handover from the islanded mode to the grid-connected mode, run under the condition of short time communication system out of action, and help to realize the plug and play function. Lastly, the stability of the proposed control algorithm is analyzed and proved, and the effectiveness of the method is verified in some case studies.

In this work, a PdNi thin film hydrogen gas sensor with integrated Pt thin film temperature sensor was designed and fabricated using the micro-electro-mechanical system (MEMS) process. The integrated sensors consist of two resistors: the former, based on Pt film, is used as a temperature sensor, while the latter had the function of hydrogen sensing and is based on PdNi alloy film. The temperature coefficient of resistance (TCR) in both devices was measured and the output response of the PdNi film hydrogen sensor was calibrated based on the temperature acquired by the Pt temperature sensor. The SiN layer was deposited on top of Pt film to inhibit the hydrogen diffusion and reduce consequent disturbance on temperature measurement. The TCR of the PdNi film and the Pt film was about 0.00122/K and 0.00217/K, respectively. The performances of the PdNi film hydrogen sensor were investigated with hydrogen concentrations from 0.3% to 3% on different temperatures from 294.7 to 302.2 K. With the measured temperature of the Pt resistor and the TCR of the PdNi film, the impact of the temperature on the performances of the PdNi film hydrogen sensor was reduced. The output response, response time and recovery time of the PdNi film hydrogen sensors under the hydrogen concentration of 0.5%, 1.0%, 1.5% and 2.0% were measured at 313 K. The output response of the PdNi thin film hydrogen sensors increased with increasing hydrogen concentration while the response time and recovery time decreased. A cycling test between pure nitrogen and 3% hydrogen concentration was performed at 313 K and PdNi thin film hydrogen sensor demonstrated great repeatability in the cycling test.
