Abstract Anti-citrullinated protein autoantibodies (ACPA) are diagnostic for rheumatoid arthritis (RA). The antigens recognized by these autoantibodies are produced by protein arginine deiminases (PADs), particularly PAD4. However, it remains unknown why and how PAD4 causes this aberrant citrullination in RA. Here, we report that poly-perforin pores are present on freshly isolated neutrophils from RA patients, but not on healthy donor neutrophils. Neutrophils with perforin pores also contained intracellular citrullinated proteins in the region adjacent to the pores. This response was replicated in vitro by treating neutrophils with purified perforin, which generated intense dots of anti-perforin immunofluorescence, calcium influx, and intracellular citrullination. Extensive neutrophil killing in Felty’s syndrome, an aggressive form of RA, correlated with particularly high ACPA, and PAD4 autoantibodies. In contrast, other forms of death, including NETosis, apoptosis, and pyroptosis, produced minimal citrullination. We conclude that neutrophil targeting by perforin leading to intracellular citrullination takes place in patients with RA.

Abstract Background Systemic lupus erythematosus (SLE) is a chronic autoimmune disease with an unpredictable course of recurrent exacerbations alternating with more stable disease. SLE is characterized by broad immune activation and autoantibodies against double-stranded DNA and numerous proteins that exist in cells as aggregates with nucleic acids, such as Ro60, MOV10, and the L1 retrotransposon-encoded ORF1p. Results Here we report that these 3 proteins are co-expressed and co-localized in a subset of SLE granulocytes and are concentrated in cytosolic dots that also contain DNA: RNA heteroduplexes and the DNA sensor ZBP1, but not cGAS. The DNA: RNA heteroduplexes vanished from the neutrophils when they were treated with a selective inhibitor of the L1 reverse transcriptase. We also report that ORF1p granules escape neutrophils during the extrusion of neutrophil extracellular traps (NETs) and, to a lesser degree, from neutrophils dying by pyroptosis, but not apoptosis. Conclusions These results bring new insights into the composition of ORF1p granules in SLE neutrophils and may explain, in part, why proteins in these granules become targeted by autoantibodies in this disease.

Foxtail millet [Setaria italica] plays a crucial role as a multigrain crop in agricultural production. However, due to future extreme weather conditions, drought remains the main abiotic stress that limits foxtail millet yield, it is highly significant to screen for drought-tolerant varieties throughout the entire growth period and identify the regulatory genes associated with drought resistance in foxtail millet breeding. We identified 217 foxtail millet seed resources for drought resistance during the maturity stage in the field, and subsequently categorized them into different levels of drought resistance. Two cultivars with extremely strong drought resistance during the maturity stage in the field, JKH4 (Chi 5422) and JKH6 (Chigu 26), as well as two cultivars with extremely weak drought resistance during the maturity stage in the field, JRK3 (17M1309) and JRK6 (Canggu 9), were selected for physiological comparison and transcriptome sequencing before and after drought treatment. Transcriptome analysis at the seedling stage revealed that JRK3 and JRK6 cultivar primarily regulated phenylpropanoid biosynthesis, MAPK signaling pathogen-plant, and plant hormone signal transduction pathway in response to drought stress. On the other hand, the fatty acid elongation pathway of JKH4 and JKH6 variety was found to be more significant. Furthermore, 22 drought resistance related genes were screened through transcriptome analysis of four foxtail millet varieties. These findings could offer valuable theoretical guidance for breeding foxtail millet with enhanced drought resistance and potentially facilitate the development of genetically engineered drought-resistant foxtail millet varieties.

Abstract These analyses characterized tofacitinib pharmacokinetics (PKs) in children and adolescents with juvenile idiopathic arthritis (JIA). Data were pooled from phase I (NCT01513902), phase III (NCT02592434), and open‐label, long‐term extension (NCT01500551) studies of tofacitinib tablet/solution (weight‐based doses administered twice daily [b.i.d.]) in patients with JIA aged 2 to less than 18 years. Population PK modeling used a nonlinear mixed‐effects approach, with covariates identified using stepwise forward‐inclusion backward‐deletion procedures. Simulations were performed to derive dosing recommendations for children and adolescents with JIA. Two hundred forty‐six pediatric patients were included in the population PK model. A one‐compartment model with first‐order elimination and absorption with body weight as a covariate for oral clearance and apparent volume of distribution sufficiently described the data. Oral solution was associated with comparable average concentration (Cavg) and slightly higher (113.9%) maximum concentration (Cmax) versus tablet, which was confirmed by a subsequent randomized, open‐label, bioavailability study conducted in healthy adult participants (n = 12) by demonstrating adjusted geometric mean ratios (90% confidence interval) between oral solution and tablet of 1.04 (1.00–1.09) and 1.10 (1.00–1.21) for area under the curve extrapolated to infinity and Cmax, respectively (NCT04111614). A dosing regimen of 3.2 mg b.i.d. solution in patients 10 to less than 20 kg, 4 mg b.i.d. solution in patients 20 to less than 40 kg, and 5 mg b.i.d. tablet/solution in patients greater than or equal to 40 kg, irrespective of age, was proposed to achieve constant Cavg across weight groups. In summary, population PK characterization informed a simplified tofacitinib dosing regimen that has been implemented in pediatric patients with JIA.

Polyoxymethylene dimethyl ethers (DMMx, CH3O(CH2O)xCH3, x = 1–8) are appealing oxygenated chemicals with potential applications mainly used as sustainable fuels and solvents. The growth of longer C-O chain for direct oxidation of dimethyl ether (DME) to DMMx is especially important, but the construction of the acid sites with different strengths and matched redox sites faces great difficulties. Herein, the gradient acidity and enhanced oxidizability have been successfully created by regulating coordination structure of titanium sulfate (Ti(SO4)x). The formation of a higher proportion of weak-medium acid sites and appropriate redox sites over a Ti(SO4)x/activated carbon (AC) catalyst with nitrogen treatment significantly promotes the synthesis of larger molecules, with DMM2–3 selectivity up to 42.2%. Combined with the characterizations, designed experiments and theoretical calculations, the configurations of Ti(SO4)x and their local chemical environment are revealed. The different distortions of Ti(SO4)x structure in nitrogen atmosphere result in the various coordination unsaturated Ti atoms located at the center of polyhedral, which is the main reason for the gradient acidity distribution and enhanced oxidative property.

Objective The purpose of this study was to investigate the correlation between different subtypes of acute kidney injury (AKI) and clinical outcomes following lung transplantation (LTx) and to identify a reliable indicator for predicting poor prognosis in the LTx population.Methods We retrospectively analyzed the clinical data of 279 LTx patients from August 2016 to March 2023. The AKI subtypes included AKI, persistent AKI on Day 7 (P7-AKI) and Day 14 (P14-AKI) after LTx, and AKI stages. The correlations of these factors with respiratory outcomes, mortality at 90 days, mortality at 1 year and data finalization were assessed, and the risk factors for the selected AKI subtypes were evaluated.Results AKI occurred in 215 patients (77.1%), with 129 (46.2%) experiencing P7-AKI and 95 (34.1%) experiencing P14-AKI. P7-AKI was associated with more respiratory and mortality outcomes than were AKI and AKI stages, and P7-AKI surpassed P14-AKI in terms of a shorter diagnostic time. After adjusting for age, sex, BMI, type of transplant, transplant diagnosis and comorbidities, P7-AKI independently correlated with increased mortality risk at 90 days [HR 12.312 (95% CI: 2.839–53.402)], 1 year [HR 3.847 (95% CI: 1.840–8.044)], and data finalization [HR 2.010 (95% CI: 1.331–3.033)]. Five variables were identified as independent predictors for P7-AKI, including preoperative body mass index, prothrombin activity, hemoglobin and serum creatinine, and intraoperative colloid administration.Conclusion P7-AKI has been identified as a reliable indicator for predicting adverse outcomes in LTx patients, which may assist healthcare professionals in identifying high-risk individuals.

Broomcorn millet (Panicum miliaceum L.) is a crop with a good ability to adapt to the environment. Over 8800 accessions have been collected in the national gene bank of China. The huge quantity of germplasms made it difficult for analysis and evaluation. Although a broomcorn millet core collection (CC) comprising 780 accessions was established, the number is still too large for researchers to explore in depth. In this study, the genetic diversity of 634 broomcorn millet accessions from the core collection was analyzed based on SSR markers. A mini-core collection (MC) containing 256 accessions was extracted. The mini-core collection accounted for less than half of the original core collection and only about 2.8% of the total resources but still provided a good representation. In addition, the results of this study validated that Shanxi is the origin of broomcorn millet, and accessions from the South region may contain novel genes. In conclusion, this study provides a comprehensive characterization of the genetic diversities of broomcorn millet core collection in China. Moreover, an MC may aid in reasonably and efficiently selecting materials for broomcorn millet breeding as researchers could screen for aimed genetic characters within a smaller scope.

Objectives To illuminate the poorly understood aetiology of SLE by comparing the gene expression profile of SLE neutrophils with that of neutrophils from patients infected by SARS-CoV-2, a disease (COVID-19) with well-defined antigens and a similar type I interferon response.Methods RNA sequencing of neutrophils from patients with SLE (n=15) and healthy controls (n=12) was analysed for differential gene expression and modulated pathways. The same analyses were performed on a similar neutrophil dataset from patients with SARS-CoV-2 infection (n=30) and healthy controls (n=8). Data on the viral rebound and safety of nirmatrelvir/ritonavir in lung transplant (LTx) recipients are limited. The study prospectively followed four LTx recipients. Clinical characteristics, viral RNA dynamic in throat swabs, and tacrolimus blood concentration were monitored regularly. All four LTx recipients, aged 35–74 years, were not vaccinated against severe acute respiratory syndrome coronavirus 2 (SARS-CoV-2). They got coronavirus disease 2019 (COVID-19) after more than one week of admission during the era of Omicron. All cases received nirmatrelvir/ritonavir (NM/r) within two days of infection, and the relative viral RNA copies dropped quickly. Viral load rebound was observed in all four cases after discontinuation of the first five days of NM/r treatment. Three of them received another 5-days antiviral therapy with NM/r. The duration of positive viral PCR testing was 25-28 days. None of them progressed into severe or critical COVID-19. Tacrolimus was stopped 12 h before NM/r and held during the 5-day course of antiviral therapy. Blood concentration of tacrolimus were maintained at a baseline level during these five days. Tacrolimus was re‐initiated at its baseline daily dose 3-4 days after NM/r therapy. However, during the second round of antiviral therapy with NM/r, the concentration of tacrolimus fluctuated wildly. In conclusion, the 5-day course of NM/r treatment was not sufficient for LTx recipients and the viral rebound was common. More data are needed to clarify whether LTx recipients with SARS-CoV-2 viral rebound could benefit from additional treatment with NM/r.

Abstract Neurodegenerative diseases, such as Alzheimer’s disease, Parkinson’s disease, amyotrophic lateral sclerosis, and Huntington’s disease, affect millions of people worldwide. Tremendous efforts have been put into disease-related research, but few breakthroughs have been made in diagnostic and therapeutic approaches. Extracellular vesicles (EVs) are heterogeneous cell-derived membrane structures that arise from the endosomal system or are directly separated from the plasma membrane. EVs contain many biomolecules, including proteins, nucleic acids, and lipids, which can be transferred between different cells, tissues, or organs, thereby regulating cross-organ communication between cells during normal and pathological processes. Recently, EVs have been shown to participate in various aspects of neurodegenerative diseases. Abnormal secretion and levels of EVs are closely related to the pathogenesis of neurodegenerative diseases and contribute to disease progression. Numerous studies have proposed EVs as therapeutic targets or biomarkers for neurodegenerative diseases. In this review, we summarize and discuss the advanced research progress on EVs in the pathological processes of several neurodegenerative diseases. Moreover, we outline the latest research on the roles of EVs in neurodegenerative diseases and their therapeutic potential for the diseases.

Abstract The purpose of this study is to evaluate the efficacy of prophylactic use amphotericin B in patients with hematologic disorders complicated by neutropenia. We searched the PubMed, EMBASE, The Cochrane Library, CBM, CNKI, VIP and WanFang Data database and the China Clinical Trials Registry ( www.chictr.org.cn ) to collect randomized controlled trials (RCTs) of amphotericin B for patients with hematologic disorders complicated by neutropenia from inception to May 2023. The Cochrane risk-of-bias tool for RCTs was used to assess the bias risk of the included studies. The meta-analysis was performed using RevMan 5.3 software. A total of 6 studies with a total of 1019 patients were included. Abstract Background Patients with systemic lupus erythematosus (SLE) have autoantibodies against the L1-encoded open-reading frame 1 protein (ORF1p). Here, we report (i) which immune cells ORF1p emanates from, (ii) which L1 loci are transcriptionally active, (iii) whether the cells express L1-dependent interferon and interferon-stimulated genes, and (iv) the effect of inhibition of L1 ORF2p by reverse transcriptase inhibitors. Results L1 ORF1p was detected by flow cytometry primarily in SLE CD66b+CD15+ regular and low-density granulocytes, but much less in other immune cell lineages. Paxlovid (nirmatrelvir/ritonavir) is an antiviral drug used to treat COVID-19, nirmatrelvir, a SARS-CoV-2 main protease inhibitor, works by inhibiting viral replication in the early stages, and ritonavir is a strong cytochrome P450 (CYP) 3A inhibitor that helps the nirmatrelvir reach and maintain the therapeutic concentrations. Paxlovid has a potential risk of drug interaction by elevating the plasma concentration of other drugs metabolized by CYP3A, like tacrolimus. This report examines the case of a 57-year-old female lung transplant patient self-administered Paxlovid for 5 days without discontinuing tacrolimus. She presented to the hospital with symptoms of headache, dizziness, palpitations, abdominal distension, nausea, vomiting, and diarrhea. The patient presented with tacrolimus toxicity and the blood concentration of tacrolimus was measured at 106 ng/mL. Urgent medical intervention was initiated, and Rifampin was administered to induce enzyme activity and rapidly decrease the concentration of tacrolimus. By adjusting the tacrolimus dosage, the final concentration was brought within the appropriate range. Clinical pharmacists should prioritize medication education for transplant patients to prevent severe drug interactions and minimize the impact on the patient’s overall well-being.

The use of antibiotics in the livestock and poultry industries has raised significant concern about environmental and health problems. In light of this, accurate knowledge of antibiotic residues in livestock and poultry manure is important for pollution management and strategic decision-making at the national level. This study aims to provide a comprehensive report on antibiotic residues in livestock and poultry manure in China using the published data of 3751 livestock and poultry feces in 29 provincial-level units over the past 20 years. In this study, the overall status of antibiotic residues in livestock and poultry feces was analyzed by mathematical statistics. Moreover, the spatio-temporal variation characteristics were analyzed by spatial statistics, and the differences among livestock and poultry species were evaluated by subgroup analysis. The finding indicated that tetracyclines (TCs), quinolones (QLs), sulfonamides (SAs), and macrolides (MLs) were the most abundant residues in livestock and poultry manure. The spatial and temporal variation revealed that the overall trend of antibiotic residues decreased gradually, and the spatial distribution was primarily concentrated in the southeast of Hu Line, exhibiting a “northeast-southwest” distribution. The distribution range also decreased slightly, with the residues of tetracyclines (TCs), quinolones (QLs), sulfonamides (SAs), and pleuromutilins (PMs) showing a significant spatial hot spot. The center of gravity of antibiotic residue shifted to the southwest between 2003 and 2021. In comparison to cow and sheep manure, the tetracyclines (TCs), sulfonamides (SAs), and macrolides (MLs) in pig and chicken manure were higher. The results can serve as a reference for the control and reduction of antibiotic pollution in livestock and poultry manure, as well as the wise utilization of those resources and achieving goals for clean agriculture.

A series of vanadium doped cryptomelane-type manganese oxide (V-OMS-2) catalysts were prepared by a simple, low-cost reflux method, and investigated for one-pot imine synthesis from oxidative coupling of benzyl alcohol and aniline with air. The physicochemical properties of the V-OMS-2 catalysts were characterized by various techniques including XRD, BET, SEM, TEM, XPS, H2-TPR and NH3-TPD. It was found that the surface area, Lewis acid sites, the amount of Mn3+ component and active surface oxygen species were much improved with vanadium doping. Consequently, the activity of V-OMS-2 catalyst for oxidative coupling of benzyl alcohol and aniline to imine was enhanced. The highest conversion and the imine yield were obtained over the 3 mol% V-OMS-2 catalyst, being ∼99% and 92%, respectively. Higher vanadium doping (≥ 6 mol%), however, hindered the preservation of OMS-2 crystal structure, leading to a drop in the catalytic performance. The high specific surface area was suggested to be the key contributor to the high catalytic activity of 3% V-OMS-2(1) catalyst. Among the vanadium precursors studied, the catalyst prepared with vanadium pentoxide exhibited a much higher catalytic activity, which can be attributed to its larger surface area, unique mesoporous structure, increased Lewis acid sites and more readily available surface oxygen species. In addition, the stability and recyclability of the catalyst were also studied, and the reaction mechanism was discussed.

Abstract Background Stable transfection systems have been described in many protozoan parasites, including Plasmodium falciparum, Cryptosporidium parvum, Babesia bovis, Babesia ovata, and Babesia gibsoni. For Babesia sp. Xinjiang (Bxj), which is the causative pathogen of ovine babesiosis and mainly prevails across China, the platform of those techniques remains absent. Genetic manipulation techniques are powerful tools to enhance our knowledge on parasite biology, which may provide potential drug targets and diagnostic markers. Methods We evaluated the inhibition efficiency of blasticidin (BSD) and WR99210 to Bxj. Then, a plasmid was constructed bearing selectable marker BSD, green fluorescent protein (GFP) gene, and rhoptry-associated protein-1 3′ terminator region (rap 3′ TR). The plasmid was integrated into the elongation factor-1 alpha (ef-1α) site of Bxj genome by cross-over homologous recombination technique. Twenty μg of plasmid was transfected into Bxj merozoites. Subsequently, drug selection was performed 24 h after transfection to generate transfected parasites. Results Transfected parasite lines, Bxj-c1, Bxj-c2, and Bxj-c3, were successfully obtained after transfection, drug selection, and colonization. Exogenous genes were integrated into the Bxj genome, which were confirmed by PCR amplification and sequencing. In addition, results of western blot (WB) and indirect immunofluorescence assay (IFA) revealed that GFP-BSD had expressed for 11 months. Conclusions In our present study, stable transfection system for Bxj was successfully developed. We anticipate that this platform will greatly facilitate basic research of Bxj. Graphical abstract

Breast cancer (BC) is the most common malignancy and the leading cause of cancer-related deaths in women worldwide. Currently, patients’ survival remains a challenge in BC due to the lack of effective targeted therapies and the difficult condition of patients with higher aggressiveness, metastasis and drug resistance. Small extracellular vesicles (sEVs), which are nanoscale vesicles with lipid bilayer envelopes released by various cell types in physiological and pathological conditions, play an important role in biological information transfer between cells. There is growing evidence that BC cell-derived sEVs may contribute to the establishment of a favorable microenvironment that supports cancer cells proliferation, invasion and metastasis. Moreover, sEVs provide a versatile platform not only for the diagnosis but also as a delivery vehicle for drugs. This review provides an overview of current new developments regarding the involvement of sEVs in BC pathogenesis, including tumor proliferation, invasion, metastasis, immune evasion, and drug resistance. In addition, sEVs act as messenger carriers carrying a variety of biomolecules such as proteins, nucleic acids, lipids and metabolites, making them as potential liquid biopsy biomarkers for BC diagnosis and prognosis. We also described the clinical applications of BC derived sEVs associated MiRs in the diagnosis and treatment of BC along with ongoing clinical trials which will assist future scientific endeavors in a more organized direction.

The pore structure is an important factor affecting reservoir capacity and shale gas production. The shale reservoir of the Longmaxi Formation in the Changning area, Southern Sichuan Basin, is highly heterogeneous and has a complex pore structure. To quantitatively characterize the shale’s pore structure and influencing factors, based on whole rock X-ray diffraction, argon ion polishing electron microscopy observations, and low-temperature nitrogen adsorption-desorption experiments, the characteristics of the shale pore structure are studied by using the Frenkel-Halsey-Hill (FHH) model. The research reveals the following: 1) The pores of the Longmaxi Formation shale mainly include organic pores, intergranular pores, dissolution pores and microfractures. The pore size is mainly micro-mesoporous. Both ink bottle-type pores and semiclosed slit-type pores with good openness exist, but mainly ink bottle-type pores are observed. 2) The pore structure of the Longmaxi Formation shale has self-similarity, conforms to the fractal law, and shows double fractal characteristics. Taking the relative pressure of 0.45 (P/P0 = 0.45) as the boundary, the surface fractal dimension Dsf and the structural fractal dimension Dst are defined. Dsf is between 2.3215 and 2.6117, and the structural fractal dimension Dst is between 2.8424 and 2.9016. The pore structure of micropores and mesopores is more complex. 3) The mineral components and organic matter have obvious control over the fractal dimension of shale, and samples from different wells show certain differences. The fractal dimension has a good positive correlation with the quartz content but an obvious negative correlation with clay minerals. The higher the total organic carbon content is, the higher the degree of thermal evolution, the more complex the pore structure of shale, and the larger the fractal dimension. The results have guiding significance for the characterization of pore structure of tight rocks.

Exosomes are a heterogeneous subset of extracellular vesicles (EVs) that biogenesis from endosomes. Besides, exosomes contain a variety of molecular cargoes including proteins, lipids and nucleic acids, which play a key role in the mechanism of exosome formation. Meanwhile, exosomes are involved with physiological and pathological conditions. The molecular profile of exosomes reflects the type and pathophysiological status of the originating cells so could potentially be exploited for diagnostic of cancer. This review aims to describe important molecular cargoes involved in exosome biogenesis. In addition, we highlight exogenous factors, especially autophagy, hypoxia and pharmacology, that regulate the release of exosomes and their corresponding cargoes. Particularly, we also emphasize exosome molecular cargoes as potential biomarkers in liquid biopsy for diagnosis of cancer.

Abstract Background Most patients with systemic lupus erythematosus (SLE) have IgG autoantibodies against the RNA-binding p40 (ORF1p) protein encoded by the L1 retroelement. This study tested if these autoantibodies are also present in children with pediatric SLE (pSLE) and if the p40 protein itself could be detected in immune cells. Methods Autoantibodies in the plasma of pSLE patients (n = 30), healthy children (n = 37), and disease controls juvenile idiopathic arthritis (JIA) (n = 32) and juvenile dermatomyositis (JDM) (n = 60), were measured by ELISA. Expression of p40 in immune cells was assessed by flow cytometry. Markers of neutrophil activation and death were quantitated by ELISA. Results IgG and IgA autoantibodies reactive with p40 were detected in the pSLE patients, but were low in healthy controls and in JIA or JDM. pSLE patients with active disease (13 of them newly diagnosed) had higher titers than the same patients after effective therapy (p = 0.0003). IgG titers correlated with SLEDAI (r = 0.65, p = 0.0001), ESR (r = 0.43, p = 0.02), and anti-dsDNA antibodies (r = 0.49, p < 0.03), and inversely with complement C3 (r = -0.55, p = 0.002) and C4 (r = -0.51, p = 0.006). p40 protein was detected in a subpopulation of CD66b+ granulocytes in pSLE, as well as in adult SLE patients. Myeloperoxidase and neutrophil elastase complexed with DNA and the neutrophil-derived S100A8/A9 were elevated in plasma from pSLE patients with active disease and correlated with anti-p40 autoantibodies and disease activity. Conclusions Children with active SLE have elevated IgG and IgA autoantibodies against L1 p40, and this protein can be detected in circulating granulocytes in both pediatric and adult SLE patients. P40 expression and autoantibody levels correlate with disease activity. Markers of neutrophil activation and death also correlate with these autoantibodies and with disease activity, suggesting that neutrophils express L1 and are a source of p40.

Many patients suffering from autoimmune diseases have autoantibodies against proteins encoded by genomic retroelements, suggesting that normal epigenetic silencing is insufficient to prevent the production of the encoded proteins for which immune tolerance appears to be limited. One such protein is the transmembrane envelope (Env) protein encoded by human endogenous retrovirus K (HERV-K). We reported recently that patients with rheumatoid arthritis (RA) have IgG autoantibodies that recognize Env. Here, we use RNA sequencing of RA neutrophils to analyze HERV-K expression and find that only two loci with an intact open-reading frame for Env, HERV-K102, and K108 are expressed, but only the former is increased in RA. In contrast, other immune cells express more K108 than K102. Patient autoantibodies recognized endogenously expressed Env in breast cancer cells and in RA neutrophils but not healthy controls. A monoclonal anti-Env antibody also detected Env on the surface of RA neutrophils but very little on the surface of other immune cells. We conclude that HERV-K102 is the locus that produces Env detectable on the surface of neutrophils in RA. The low levels of HERV-K108 transcripts may contribute only marginally to cell surface Env on neutrophils or other immune cells in some patients.

This paper introduces the mixed incremental $H_\infty $ and incremental passivity control problem for Markov switched stochastic (MSS) nonlinear systems. The multiple incremental Lyapunov functions approach and the structure of Markov framework are utilized to establish some sufficient conditions for the MSS nonlinear systems, which will be used for the incrementally globally asymptotically stable in the mean(IGASiM) and performance index analysis. It is proved that under the proposed non-IGASiM subsystems the underlying MSS nonlinear systems are IGASiM and possess the mixed incremental $H_\infty $ and incremental passivity performance metric in the presence of specified conditions. The mathematical induction method is selected to guarantee the robust incremental stability of MSS systems with IGASiM and non-IGASiM subsystems and the performance index can be exhibited a prescribed decay rate. The effectiveness of the proposed results is demonstrated by two simulation examples.

A new kind of epoxy resin/foam nickel–iron plate connector is introduced. This kind of interpenetrating composite connector has approximate thermal insulation performance of the resin, good mechanical properties, and easy manufacture and production. This paper studied the shear performance of the connector at the elastic–plastic stage, which was the basis important performance of the connector to be widely used. Three groups of specimens made of single-layer foam nickel–iron plate and three groups of specimens made of double-layer foam nickel–iron plate were tested and analyzed by finite element simulation. The results showed that the plate connector had better shear strength. The failure mode of plate specimens was the typical bending shear failure, and finally, the web buckling occurred in the compression zone. The bearing capacity of the specimen was positively correlated with the thickness of the specimen and the material properties of the matrix phase.

Interpenetrating phase composites (IPCs) are a new type of materials with designable mechanical properties. In this work, to explore a simple, applicable, and accurate method for property designability of interpenetrated phase composites, the interaction between the matrix phase and the reinforcing phase of IPC under compressive stress is discussed, and a microstructured representative volume element is proposed for interpenetrating metal/polymer composites under compressive loading by considering the interaction of constituent phases. The stress–strain relationship of the representative volume element is established based on the iso-stress and iso-strain assumptions. Compared with the experimental results with those of interpenetrated metal/polymer composites, interpenetrated metal/metal composites, and interpenetrated metal/ceramic composites, it is shown that the analytical solutions can accurately predict the elasto-plastic compressive stress–strain curves.

Abstract Background Feline and canine babesiosis is an important tick-borne disease caused by parasites of the genus Babesia. The disease has a worldwide distribution and causes serious health problems in domestic and wild canidae and felidae. Results Genomic DNA was isolated from blood samples, which were randomly collected from pet dogs (n = 115) and cats (n = 25) in Changsha city of Hunan Province, China. Results of nested PCR assay targeting 18S rRNA gene and partial gene sequencing revealed that seven animals were infected with Babesia species, five dogs (5/115, 4.3%) and two cats (2/25, 8.0%). Sequence analysis showed that four dogs (3.5%) were positive for Babesia canis, and the other one for Babesia vogeli (0.87%). The two cats were infected by Babesia hongkongensis. Conclusions The findings of this study will expand knowledge of the distribution of Babesia species and provide important epidemiological information for the control of animal babesiosis in China.

Abstract Background Babesiosis, a tick-borne disease caused by protozoans of the genus Babesia, is widespread in subtropical and tropical countries. Mitochondria are essential organelles that are responsible for energy transduction and metabolism, calcium homeostasis and cell signaling. Mitochondrial genomes could provide new insights to help elucidate and investigate the biological features, genetic evolution and classification of the protozoans. Nevertheless, there are limited data on the mitochondrial genomes of ovine Babesia spp. in China. Methods Herein, we sequenced, assembled and annotated the mitochondrial genomes of six ovine Babesia isolates; analyzed the genome size, gene content, genome structure and cytochrome b (cytb) amino acid sequences and performed comparative mitochondrial genomics and phylogenomic analyses among apicomplexan parasites. Results The mitochondrial genomes range from 5767 to 5946 bp in length with a linear form and contain three protein-encoding genes, cytochrome c oxidase subunit 1 (cox1), cytochrome c oxidase subunit 3 (cox3) and cytb, six large subunit rRNA genes (LSU) and two terminal inverted repeats (TIR) on both ends. The cytb gene sequence analysis indicated the binding site of anti-Babesia drugs that targeted the cytochrome bc1 complex. Babesia microti and Babesia rodhaini have a dual flip-flop inversion of 184–1082 bp, whereas other Babesia spp. and Theileria spp. have one pair of TIRs, 25–1563 bp. Phylogenetic analysis indicated that the six ovine Babesia isolates were divided into two clades, Babesia sp. and Babesia motasi. Babesia motasi isolates were further separated into two small clades (B. motasi Hebei/Ningxian and B. motasi Tianzhu/Lintan). Conclusions The data provided new insights into the taxonomic relationships and drug targets of apicomplexan parasites.

Colorectal cancer (CRC) is a malignancy that seriously threatens human health, and metastasis from CRC is a major cause of death and poor prognosis for patients. Studying the potential mechanisms of small extracellular vesicles (sEVs) in tumor development may provide new options for early and effective diagnosis and treatment of CRC metastasis. In this review, we systematically describe how sEVs mediate epithelial mesenchymal transition (EMT), reconfigure the tumor microenvironment (TME), modulate the immune system, and alter vascular permeability and angiogenesis to promote CRC metastasis. We also discuss the current difficulties in studying sEVs and propose new ideas.

Previously, we reported a strong Fe-Cu synergy in CO2 hydrogenation to olefin-rich C2+ hydrocarbons over the γ-Al2O3 supported bimetallic Fe-Cu catalysts. In this work, we aimed to clarify such a synergy by investigating the catalyst structure, Fe-Cu interaction, and catalyst surface properties through a series of characterizations. H2-TPR results showed that the addition of Cu made both Fe and Cu easier to reduce via the strong interaction between Fe and Cu. It was further confirmed by X-ray absorption spectroscopy (XAS) and TEM, which showed the presence of metallic Fe and Fe-Cu alloy phases in the reduced Fe-Cu(0.17) catalyst induced by Cu addition. By correlating TPD results with the reaction performance, we found that the addition of Cu enhanced both the moderately and strongly adsorbed H2 and CO2 species, consequently enhanced CO2 conversion and C2+ selectivity. Adding K increased the adsorbed-CO2/adsorbed-H2 ratio by greatly enhancing the moderately and strongly adsorbed CO2 and slightly suppressing the moderately and strongly adsorbed H2, resulting in a significantly increased O/P ratio in the produced hydrocarbons. The product distribution analysis and in situ DRIFTS suggested that CO2 hydrogenation over the Fe-Cu catalyst involved both an indirect route with CO as the primary product and a direct route to higher hydrocarbons.

Neural architecture search (NAS) is a popular branch of automatic machine learning (AutoML), which aims to search for efficient network structures. Many prior works have explored a wide range of search algorithms for classification tasks, and have achieved better performance than manually designed network architectures. However, few works have explored NAS for object detection tasks due to the difficulty to train convolution neural networks from scratch. In this paper, we propose a framework, named as FastDARTSDet, to directly search on a larger-scale object detection dataset (MS-COCO). Specifically, we propose to apply differentiable architecture search method (DARTS) to jointly search backbone and feature pyramid network (FPN) architectures for object detection task. Extensive experimental results on MS-COCO show the efficient and efficacy of our method. Specifically, our method achieves 40.0% mean average precision (mAP) on the test set, outperforming many recent NAS methods.

Abstract Background Babesiosis is an economically important disease caused by tick-borne apicomplexan protists of the genus Babesia. Most apicomplexan parasites, including Babesia, have a plastid-derived organelle termed an apicoplast, which is involved in critical metabolic pathways such as fatty acid, iron-sulphur, haem and isoprenoid biosynthesis. Apicoplast genomic data can provide significant information for understanding and exploring the biological features, taxonomic and evolutionary relationships of apicomplexan parasites, and identify targets for anti-parasitic drugs. However, there are limited data on the apicoplast genomes of Babesia species infective to small ruminants. Methods PCR primers were designed based on the previously reported apicoplast genome sequences of Babesia motasi Lintan and Babesia sp. Xinjiang using Illumina technology. The overlapped apicoplast genomic fragments of six ovine Babesia isolates were amplified and sequenced using the Sanger dideoxy chain-termination method. The full-length sequences of the apicoplast genomes were assembled and annotated using bioinformatics software. The gene contents and order of apicoplast genomes obtained in this study were defined and compared with those of other apicomplexan parasites. Phylogenetic trees were constructed on the concatenated amino acid sequences of 13 gene products using MEGA v.6.06. Results The results showed that the six ovine Babesia apicoplast genomes consisted of circular DNA. The genome sizes were 29,916–30,846 bp with 78.7–81.0% A + T content, 29–31 open reading frames (ORF) and 23–24 transport RNAs. The ORFs encoded four DNA-directed RNA polymerase subunits (rpoB, rpoCl, rpoC2a and rpoC2b), 13 ribosomal proteins, one elongation factor TU (tufA), two ATP-dependent Clp proteases (ClpC) and 7–11 hypothetical proteins. Babesia sp. has three more genes than Babesia motasi (rpl5, rps8 and rpoB). Phylogenetic analysis showed that Babesia sp. is located in a separate clade. Babesia motasi Lintan/Tianzhu and B. motasi Ningxian/Hebei were divided into two subclades. Conclusions To our knowledge, this study is the first to elucidate the whole apicoplast genomic structural features of six Babesia isolates infective to small ruminants in China using Sanger sequencing. The data provide useful information confirming the taxonomic relationships of these parasites and identifying targets for anti-apicomplexan parasite drugs.

Small extracellular vesicles are membrane-bound vesicles secreted into extracellular spaces by virtually all types of cells. These carry a large number of membrane proteins on their surface that are incorporated during their biogenesis in cells. The composition of the membrane proteins hence bears the signature of the cells from which they originate. Recent studies have suggested that the proteins on these small extracellular vesicles can serve as biomarkers and target proteins for the diagnosis and treatment of diseases. This article classifies small extracellular vesicle membrane proteins and summarizes their pathophysiological functions in the diagnosis and treatment of diseases.

Climate change has become a worldwide concern with the rapid rise of the atmospheric Co2 concentration. To mitigate Co2 emissions, the research and development efforts in Co2 capture and separation both from the stationary sources with high Co2 concentrations (e.g., coal-fired power plant flue gas) and directly from the atmosphere have grown significantly. Much progress has been achieved, especially within the last twenty years. In this perspective, we first briefly review the current status of carbon capture technologies including absorption, adsorption, membrane, biological capture, and cryogenic separation, and compare their advantages and disadvantages. Then, we focus mainly on the recent advances in the absorption, adsorption, and membrane technologies. Even though numerous optimizations in materials and processes have been pursued, implementing a single separation process is still quite energy-intensive or costly. To address the challenges, we provide our perspectives on future directions of Co2 capture research and development, that is, the combination of flue gas recycling and hybrid capture system, and one-step integrated Co2 capture and conversion system, as they have the potential to overcome the technical bottlenecks of single capture technologies, offering significant improvement in energy efficiency and cost-effectiveness.

The Co3O4 electrode is a very promising material owing to its ultrahigh capacitance. Nevertheless, the electrochemical performance of Co3O4-based supercapacitors is practically confined by the limited active sites and poor conductivity of Co3O4. Herein, we provide a facile synthetic strategy of tightly anchoring Co3O4 nanosheets to a carbon fiber conductive cloth (Co3O4@C) using the zeolitic imidazolate framework-67 (ZIF-67) sacrificial template via in situ impregnation and the pyrolysis method. Benefiting from the enhancement of conductivity and the increase in active sites, the binder-free porous Co3O4@C supercapacitor electrodes possess typical pseudocapacitance characteristics, with an acceptable specific capacitance of ~251 F/g at 1 A/g and long-term cycling stability (90% after cycling 5000 times at 3 A/g). Moreover, the asymmetric and flexible supercapacitor composed of Co3O4@C and activated carbon is further assembled, and it can drive the red LED for 6 min.

CRISPR-Cas9 has been utilized to screen the genome in loss-of-function studies to identify genetic interactions. Here the authors compare catalytically active and dead Cas9 and observe different off-target effects in a screen for essential genes.

Abstract Background Hemotropic mycoplasmas (hemoplasmas) are emerging zoonotic pathogens with a worldwide distribution that can cause mild to severe hemolytic anemia, icterus, ill-thrift, infertility, and poor weight gain. However, understanding of the molecular epidemiology of hemoplasmas (Mycoplasma ovis and ‘Candidatus Mycoplasma haemovis’) is limited in sheep and goats, and the hemoplasma strain/species/variant ‘Candidatus M. haemovis’ was poorly studied throughout the world and had never been detected in China until now. Thus, the aim of the present study was to determine the molecular prevalence of hemoplasmas, including M. ovis and ‘Candidatus M. haemovis’ in sheep and goats from seven provinces and one autonomous region of China. Methods A total of 1364 blood samples were collected from sheep and goats in seven provinces and one autonomous region of China. All blood samples were tested for hemoplasmas (M. ovis and ‘Candidatus M. haemovis’) by nested PCR amplification based on 16S rRNA gene. Positive specimens underwent nucleotide sequencing and phylogenetic analysis. Results Overall, 610 specimens (44.7%, 610/1364) were shown to be hemoplasmas (M. ovis and ‘Candidatus M. haemovis’) -positive by nested PCR amplification based on 16S rRNA gene. The prevalence in goats was 44.1% (379/860), and 45.8% (231/504) in sheep, while that in grazing small ruminants was 54.4% (396/728) and 33.6% (214/636) in house feeding small ruminants. Sequencing of the nearly complete 16S rRNA gene was successful for the 103 randomly selected positive specimens from different farms in different sampling sites of China. Among them, analysis of the 16S rRNA gene sequences identified M. ovis (n = 56) and ‘Candidatus M. haemovis’ (n = 47). Two (KU983740 and KU983746) of the four novel genotypes obtained in this study were closely related to M. ovis, while the other two genotypes (KU983748 and KU983749) had high identity with ‘Candidatus M. haemovis’. Remarkably, the genotype (KU983740) of M. ovis in sheep and goats in this study fell in a clade with two human hemoplasmas from USA (KF313922 and GU230144) and shared 99.8%–99.9% with them. Conclusions In this study, ‘Candidatus M. haemovis’ was first detected in Chinese sheep and goats and hemoplasmas (M. ovis and ‘Candidatus M. haemovis’) are highly prevalent, and widely distributed in China.

The apicomplexan parasite Babesia bovis is a tick-borne intracellular hemoprotozoan parasite that is widespread across China. Genetic diversity is an important strategy used by parasites to escape the immune responses of their hosts. In our present study, 575 blood samples, collected from cattle in 10 provinces, were initially screened using a nested PCR (polymerase chain reaction) for detection of B. bovis infection. To perform genetic diversity analyses, positive samples were further amplified to obtain sequences of three B. bovis merozoite surface antigen genes (MSA-1, MSA-2b, MSA-2c). The results of the nested PCR approach showed that an average of 8.9% (51/575) of cattle were positive for B. bovis infection. Phylogenetic analyses of the predicted amino acid sequences revealed that unique antigen variants were formed only by Chinese isolates. Our findings provide vital information for understanding the genetic diversity of B. bovis in China.

Shenfu decoction (SFD) can be used to treat patients with sign of Yangqi decline or Yang exhaustion related to chronic heart failure (CHF). We conducted a gas chromatography with time-of-flight mass spectrometer (GC/TOF–MS)-based metabolomic study to increase the understanding of CHF and assess the efficacies and mechanisms of SFD in treating CHF induced by coronary artery ligation in rats. Based on unsupervised principal component analysis, there was a clear separation between the CHF and sham surgery group, which revealed that CHF disturbed the metabolism of endogenous substances and significantly altered the urine metabolite fingerprints. After SFD treatment, the metabolomics profile found in CHF was significantly reversed, shifting much closer to normal controls and sham surgery group, indicating that SFD has therapeutic effects in CHF, which is in accordance with the hemodynamic assay results. Metabolomic pathway analysis demonstrated that several pathways including fatty acid biosynthesis, fatty acid elongation, steroid biosynthesis, galactose metabolism, and amino acid metabolism were significantly altered in CHF rats. Therefore, we may infer that SFD shows therapeutic efficacy in CHF by restoring these disturbed metabolic pathways, especially those related to energy metabolism. This study offers new methodologies for increasing the understanding of CHF and systematically characterizing the efficacies and mechanisms of SFD in treating CHF.

Abdominal aortic aneurysm (AAA) is an inflammatory vascular disorder with high mortality. Accumulating evidence shows that toll-like receptor 2 (TLR2) plays a critical role in the regulation of wound-repairing process after tissue injury. We wondered if TLR2 signaling contributed to the pathogenesis of AAA and that targeting TLR2 would attenuate AAA development and progression. In this study, enhanced expression of TLR2 and its ligands were observed in human AAA tissue. Neutralization of TLR2 protected against AAA development and caused established AAA to regress in mouse models of AAA. In addition, TLR2-deficient mice also failed to develop AAA. The prophylactic and therapeutic effects of blocking TLR2 were accompanied by a significant resolution of inflammation and vascular remodeling, as indicated by the decreased expression or activity of MMP-2/9, α-SMA, inflammatory cytokines, and transcription factors NF-κB, AP-1 and STAT1/3 in AAA tissue. Mechanistically, blocking TLR2 decreased the expression and interaction of TLR2 and several endogenous ligands, which diminished chronic inflammation and vascular remodeling in the vascular tissue of AAA. Our studies indicate that the interactions between TLR2 and its endogenous ligands contribute to the pathogenesis of AAA and that targeting TLR2 offers great potential toward the development of therapeutic agents against AAA.

Simian Foamy Virus (SFV) can be transmitted from non-human primates (NHP) to humans. However, there are no documented cases of human to human transmission, and significant differences exist between infection in NHP and human hosts. The mechanism for these between-host differences is not completely understood. In this paper we develop a new Bayesian approach to the detection of APOBEC3-mediated hypermutation, and use it to compare SFV sequences from human and NHP hosts living in close proximity in Bangladesh. We find that human APOBEC3G can induce genetic changes that may prevent SFV replication in infected humans in vivo.
