Abstract Erectile dysfunction (ED) is prevalent among males experiencing fertility challenges, yet attitudes towards actively treating ED in this group are under-researched. From a cohort of 1256 men with reproductive needs, 303 were identified with ED. The survey encompassed 296 respondents who correctly completed the second questionnaire, revealing that 50.3% sought ED treatment, with higher ED severity increasing the likelihood of seeking treatment. Infertile men were more likely to seek treatment than those with pregnancy loss (OR 3.18, 95% CI 1.74–5.83). Men with normal semen parameters were more open to ED therapy (OR 3.02, 95% CI 1.69–5.36), whereas those undergoing Assisted Reproduction Treatment were less inclined (OR 0.32, 95% CI 0.18–0.58). PDE-5 inhibitors (PDE-5Is) were preferred by 51.0% of those seeking treatment, with 29.7% of men with pregnancy loss and 60.1% of infertile men choosing PDE-5Is as their first option. Concerns included potential adverse effects of PDE-5Is on fetal health (78.7% of men with pregnancy loss) and on sperm quality (44.2% of infertile men). In conclusion, the different fertility requirements, semen parameters, and whether received ART are significant factors influencing the acceptance of treatment, PDE-5Is utilization among individuals in men with couple pregnancy loss is notably limited.

In recent years, exploration breakthroughs have been successively achieved in the strata deeper than the fifth member of the Lower Ordovician Majiagou Formation in the Ordos Basin, revealing its tremendous exploration potential. With the deployment of high-precision 3D seismic data and the deepening of oil and gas exploration, the control exerted by faults on the gas reservoirs in the deeper layers of the fifth member has become increasingly prominent. Based on high-precision 3D seismic data, this study systematically characterized the geometric features of faults in the Gaojiabao area, dissected typical fault structural patterns, established fault formation and evolution models, and, in conjunction with drilling and testing data, discussed the control of faults on the differential enrichment of gas reservoirs in the deeper layers of the Majiagou Formation. The study yielded the following insights: Faults in the Lower Paleozoic of the Gaojiabao area in the Ordos Basin exhibit distinct stratified deformation characteristics, with the two sets of gypsum-salt rocks in the third and fifth members being major contributors to fault stratification. The Lower Paleozoic in the Gaojiabao area mainly develops strike-slip faults with compressional and torsional properties and thrust faults. Through studying the structural morphology and fault patterns of the Gaojiabao area, it is inferred that the Yanshan tectonic movement was the most significant phase of reformation affecting the faults formed during the Caledonian tectonic movement. The coupling of the structural uplift period with the oil-gas charging period in the Gaojiabao area means that faults play multiple roles, including improving reservoirs, connecting source rocks, and controlling traps.

Under the global climate challenges, accurate assessment of forest variables is becoming increasingly important for effective forest management and monitoring. While long-wavelength satellite radar data (P-, L-band) has been widely used for this purpose, the potential of airborne Ku-band microwave radar remains underexplored. In this paper, an airborne Ku-band frequency modulated continuous wave (FMCW) profiling radar data was used to predict forest variables including plot-level basal-area weighted mean canopy height (Hg), mean diameter at breast height (DBHg), stem volume (V) and above ground biomass (AGB) using the random forest method. In addition, the maximum canopy heights were computed from the radar-derived results. The estimated results were compared with 55 manually-measured and accurately matched field plots consisting of thousands of trees. The comparison results show that the estimation accuracy is at a good level, with the relative Root Mean Square Errors (RMSE%) and the coefficient of determination (R2) of the four variables were Hg (10.04 %, 0.75), DBHg (17.25 %, 0.63), V (21.11 %, 0.67) and AGB (19.75 %, 0.63), respectively. The estimation of plot-level maximum heights resulted in a relative RMSE of 7.95 % and R2 of 0.89. Through comparing with previous similar studies, the accuracies of estimating the four variables from airborne Ku-band data are comparable to those of high-precision satellite microwave radar data. The study results indicate that the canopy gaps and adequate penetration of airborne Ku-band signals into the canopy enable the characterization of the canopy layer and the ground floor, allowing for accurate estimation of forest variables in boreal forests.

This study investigated the Maillard reaction in Baijiu and the effects of extended aging in the presence of Maillard reaction intermediates (MRIs) on aromatic compounds, particularly focusing on heterocyclic changes. MRIs with different aroma types in Baijiu aged 1–18 years and force-aged for 6 weeks were determined. Results revealed that MRIs in soy sauce aroma–type Baijiu were significantly more abundant than those in other types of Baijiu. Changes in MRIs were observed and compared in aging and forced-aging Baijiu. Additionally, the distribution and variation of heterocycles in Baijiu were examined, which revealed an increase in N-heterocycle levels but a decrease in S- and O-heterocycle levels to a certain extent. The results of this study demonstrate that the Maillard reaction during the aging of Baijiu influences heterocycle concentrations, thereby improving flavor of aged Baijiu. Research into heterocycles and the Maillard reaction may help elucidate the aromatic evolution of Baijiu with aging and provide guidance for Baijiu storage.

Abstract Background Frogs are critical economic animals essential to agricultural ecosystem equilibrium. However, Meningitis-like Infectious Disease (MID) often affects them in agricultural settings. While frog-associated microbiota contribute to elemental cycling and immunity, the effects of frog sex and health on gut bacteria remain understudied, and the relationship between frog habitat and soil microbes is unclear. We aimed to determine how frog sex, health status and habitat influence symbiotic bacteria and community assembly mechanism to provide guidance for sustainable frog farming and conservation. Results We employed 16S rRNA sequencing to investigate gut microbiota differences in relation to frog sex and health status. We also compared symbiotic communities in frog-aggregation, native and soybean soil on the farm. Results showed that gut bacterial β-diversity and taxonomy were markedly influenced by frog sex and health. Healthy frogs had more robust gut bacterial metabolism than frogs infected with MID. Cooccurrence network analysis revealed that healthy female frogs had more complex microbial network structure than males; however, diseased males showed the greatest network complexity. The assembly mechanism of gut bacteria in male frogs was dominated by deterministic processes, whereas in female frogs it was dominated by stochastic processes. Among symbiotic bacteria in frog habitat soils, deterministic processes predominantly shaped the community assembly of soybean soil. In particular, soybean soil was enriched in pathogens and nitrogen functions, whereas frog-aggregation soil was markedly increased in sulphur respiration and hydrocarbon degradation. Conclusion Our study reveals that sex mainly alters the interaction network and assembly mechanism of frog intestinal bacteria; MID infection significantly inhibits the metabolic functions of intestinal bacteria. Furthermore, diverse frog habitat soils could shape more symbiotic bacteria to benefit frog farming. Our findings provide new horizons for symbiotic bacteria among frogs, which could contribute to sustainable agriculture and ecological balance.

Abstract Background and objective Calcaneal Sanders type II or III fractures are highly disabling with significant burden. Surgical treatment modalities include open reduction and internal fixation (ORIF) techniques and a variety of minimally invasive surgical (MIS) approaches. ORIF techniques are associated with complications and traditional MIS techniques need extensive intraoperative fluoroscopic procedures. The present study aims to investigate the effects of three different minimally invasive internal fixation (MIIF) techniques used to treat Sanders type II intra-articular calcaneal fractures using finite element analyses. Methods A 64-row spiral computed tomography scan was used to observe the calcaneus of a healthy adult. The scanning data were imported into Mimics in a DICOM format. Using a new model of a Sanders type II-B intra-articular calcaneal fracture, three minimally invasive techniques were simulated. Technique A involved fixation using an isolated minimally invasive locking plate; Technique B used a minimally invasive locking plate with one medial support screw; and Technique C simulated a screw fixation technique using four 4.0-mm screws. After simulating a 640-N load on the subtalar facet, the maximum displacement and von Mises stress of fragments and implants were recorded to evaluate the biomechanical stability of different fixation techniques using finite element analyses. Results After stress loading, the maximum displacements of the fragments and implants were located at the sustentaculum tali and the tip of sustentaculum tali screw, respectively, in the three techniques; however, among the three techniques, Technique B had better results for displacement of both. The quality of apple picking affects the sales of apples, and the grasping force of the end effector of an apple picking robot is very important for apple picking. It is easy to cause apple damage due to excessive contact force, or when the contact force is too small to grasp the apple. However, the current research lacks an analysis of the minimum stable grasping force of apples. Therefore, in order to realize the stable grasping of apples by the end-effector of a picking robot and reduce fruit damage, this study first analyzes the grasping stability of the end-effector based on the force closure theory, and comprehensively considers the force closure constraints, nonlinear friction cone constraints and the introduction of torque constraints. Next, the constraint conditions are processed using an obstacle function, and a penalty factor is introduced to construct an optimization model of the contact force distribution of the end-effector. Then, the improved Newton method is used to grasp and solve the contact force distribution model. Under the premise of selecting the penalty factor, the optimal contact force of grasping an apple is determined using a method of numerical example simulation analysis, and the validity of the solution is verified. In order to verify the reliability of the contact force distribution optimization model, the practical significance of the method for apple grasping is verified in an actual grasping experiment. The actual experiment shows that the method can provide the minimum stable grasping force to the end-effector to achieve stable grasping.

At the dawn of of Artificial General Intelligence (AGI), the emergence of large language models such as ChatGPT show promise in revolutionizing healthcare by improving patient care, expanding medical access, and optimizing clinical processes. However, their integration into healthcare systems requires careful consideration of potential risks, such as inaccurate medical advice, patient privacy violations, the creation of falsified documents or images, overreliance on AGI in medical education, and the perpetuation of biases. It is crucial to implement proper oversight and regulation to address these risks, ensuring the safe and effective incorporation of AGI technologies into healthcare systems. By acknowledging and mitigating these challenges, AGI can be harnessed to enhance patient care, medical knowledge, and healthcare processes, ultimately benefiting society as a whole.

The impact of various fatty acid types on adaptive immunity remains uncertain, and their roles remain unelucidated. Stearoyl-CoA desaturase (Scd) is a Δ-9 desaturase, which is a key rate-limiting enzyme for the conversion of saturated fatty acids (SFA) to monounsaturated fatty acids (MUFA) in the fatty acid de novo synthesis. Scd-1 converts stearic acid (SA) and palmitic acid (PA) to oleic acid (OA) and palmitoleic acid (PO), respectively. In this study, through a series of experiments, we showed that Scd-1 and its resulting compound, OA, have a substantial impact on the transformation of CD8+ naïve T cells into effector T cells. Inactivation of Scd-1 triggers the specialization of CD8+ T cells into the Teff subset, enhancing the effector function and mitochondrial metabolism of Teff cells, and OA can partially counteract this. A deeper understanding of lipid metabolism in immune cells and its impact on cell function can lead to new therapeutic approaches for controlling the immune response and improving prognosis.

Purpose: To explore stable and sensitive indicators for clinical diagnosis of acute angle closure (AAC) secondary to lens subluxation (LS) through quantitative analysis of CASIA 2 imaging.Design: A prospective cross-sectional study. Methods: Setting: Clinical practice.Participants: 23 patients with unilateral acute angle closure secondary to lens subluxation and 23 cataract patients without lens subluxation were recruited. Lens subluxation was confirmed by ultrasound biomicroscope diagnosis. The contralateral eyes without LS served as fellow control group. The cataract eyes without LS were enrolled in blank control group.Intervention: Participants underwent ophthalmologic examinations including slit-lamp biomicroscope, best corrected visual acuity, intraocular pressure, central corneal thickness measurement, axial length, gonioscopy, ultrasound biomicroscope and 360-degree anterior chamber and crystalline lens scan protocols of CASIA 2 system.Main outcome measures: Automated circumferential anterior segment and lens morphological parameters under anterior segment optical coherence tomography were analyzed via three-dimensional analysis. Results: Significant differences were found in the front and back radius of the lens, the front and back radius of steep curvature of the lens, lens thickness, lens decentration, lens diameter, iris-trabecular contact (ITC) index, ITC area, anterior chamber depth (ACD), lens vault (LV), and iris volume between LS and controls. Among these parameters, LV, the anterior radius of steep curvature of the lens and ACD demonstrated the highest prediction power (AUC = 0.87, 0.89, and 0.86, respectively). The prediction power of tilt/axis was much higher in the Gaussian Naive Bayes model (AUCs = 0.90) than in the logistic model (AUCs = 0.74). Combination of LV_mean, LV_std, tilt and tilt axis in Gaussian Naive Bayes model presented as most stable and excellent diagnostic markers for AAC secondary to LS (AUCs = 0.98). Conclusions: The combination of markers including lens tilt and lens vault in the mathematic model facilitate clinical work as it not only provides novel diagnostic indications and possible prompt treatment for AAC secondary to lens subluxations, but also enhances our understanding of the pathogenic role of zonulopathy in angle closure glaucoma.

A hybrid fiber reinforced cementitious composite (HFRCC) was produced by mixing steel fiber (SF) and polyethylene fiber (PE) together in the cement matrix with an appropriate ratio. Under the same PE volume percentage, the effect of various SF aspect ratios (30, 50, 65, 83.3, and 108.3) and volume percentages (0%, 0.3%, 0.6%, and 0.9%) on the compressive behavior of HFRCC was experimentally studied. The results indicated that the aspect ratio and volume percentage of SF had little effect on the elastic modulus of HFRCC. With the addition of SF, the maximum increase of compressive strength and peak strain reached 33.82% and 24.29%, respectively. And the HFRCC presented maximum compressive strength and peak strain when the aspect ratio of SF was 108.3 and the volume percentage was 0.6%. In order to reveal the compressive constitutive relationship of HFRCC, a piecewise function was adopted based on the comparison of existing theory and furthermore in which the fiber reinforcing index, compressive strength, peak strain, and elastic modulus had been introduced. It is concluded that the proposed compressive constitutive model has more agreement with test data.

Abstract Background Adding acid protease to feed can enhance protein digestibility, boost feed utilization, and stimulate the growth of animals in breading industry. In order to obtain an acid protease with high hydrolysis efficiency to plant protein, in this study, an aspartic protease from Aspergillus niger was heterologous expressed in Pichia pastoris (P. pastoris). The enzymatic properties and application in soybean protein degradation were also studied. Results In our investigation, the high aspartic protease (Apa1) activity level of 1500 U/mL was achieved in 3 L bioreactor. After dialysis and anion exchange chromatography, the total enzyme activity and specific enzyme activity were 9412 U and 4852 U/mg, respectively. The molecular weight of the purified protease was 50 kDa, while the optimal pH and temperature were 3.0 and 50 °C, respectively. It was stable at pH 2.0–5.0 and 30–60 °C. Apa1 was used to hydrolyze soybean isolate protein (SPI) at 40 °C and pH 3.0, and a high hydrolysis degree (DH) of 61.65% was achieved. In addition, the molecular weight distribution of SPI hydrolysis products was studied, the result showed that the hydrolysis products were primarily oligopeptides with molecular weights of 189 Da or below. Conclusions In this study, Apa1 was successfully expressed in P. pastoris and high expression level was obtained. In addition, the highest protein hydrolysis rate to SPI degradation so far was achieved. The acid protease in this study provides a new protease that is suitable for the feed industry, which will be very helpful to improve the feed utilization and promote the development of the breeding industry.

The gold deposit offshore of Northern Sanshandao is an ultra-large-scale gold deposit discovered in the Jiaodong ore area in recent years. This deposit is a fractured-zone altered-rock-type gold deposit; however, its ore genesis and precise mineralization processes are still highly controversial. Based on petrographical observation, the trace elements, sulfur isotopes, and rubidium–strontium isotopes of the gold-bearing pyrite were analyzed using LA-MC-ICP-MS to obtain the source of the ore-forming fluids and ore genesis. The results show that Au has a good positive correlation with Ag, As, and Cu. It is speculated that the As in the pyrite of the gold deposit offshore of Northern Sanshandao is in the form of As−, replacing S− and entering the pyrite, causing its lattice defects, and thus promoting the entry of Au+ into the gold-bearing pyrite. The Co/Ni ratios mainly range between 0.1 and 10, indicating that the mineralization process has experienced different forms of hydrothermal evolution and the mixing of different fluids. The results of the in-situ sulfur isotope analysis show that pyrite δ34S in the mineralization period is characterized by a high sulfur value. The authors of this study believe that the initial sulfur isotope composition has mantle-derived components. The large-scale, deep cutting, and high degree of fragmentation in the Sanshandao fault zone are conducive to the interaction between fluids and rocks, as well as the mixing and addition of seawater, resulting in the characteristic high δ34S value. The Sr isotopic compositions indicate a crust–mantle mixing attribute of the mineralized material source. The Rb–Sr isochron age of the pyrite is 118.5 ± 0.65 Ma, which represents the age of gold mineralization. According to the characteristics of the trace elements and sulfur isotopes, it is inferred that the gold deposit minerals offshore of Northern Sanshandao originated from deep magmatic-hydrothermal reservoirs, and the mixing of seawater and Au–As-rich hydrothermal fluids was the formation mechanism of huge amounts of gold precipitation.

Land use and cover change are major factors driving global change and greatly impact terrestrial organisms, especially soil microbial diversity. Little is known, however, about bacterial diversity, distribution patterns and assembly processes across different land use types. In this study, therefore, we conducted a large-scale field survey of 48 sampling sites, encompassing different land use types in Xuancheng city, China, with different degrees of soil disturbance and different soil horizons. The distance-decay relationships (DDRs), assembly processes and the spatial patterns of soil bacterial communities were investigated based on high-throughput sequencing data. We found that the DDRs might be weakened by anthropogenic disturbances, which were not observed in tilled soils, while a decreasing trend was observed along the soil horizon in untilled soils. The relative importance of environmental factors and geographic distance varied with soil tillage. Specifically, bacterial communities in tilled soils were driven by non-spatially autocorrelated environmental factors, while untilled soils were more susceptible to geographic distance. In addition, the heterogeneity of soil properties, as well as the differences in soil bacterial niche width and niche overlap, determined the assembly processes of the bacterial community, resulting in opposite trends along the soil layers in tilled and untilled soils. These findings expand the current understanding of the biogeography of soil bacterial communities across different land use types.

Abstract Background Erectile dysfunction is a common problem in males of couples experiencing pregnancy loss. Erectile dysfunction in males with couple infertile has been extensively investigated and found to be closely linked with semen quality impairment and psychological distress, but it is less clear if this relation exists in males of couples experiencing pregnancy loss. Method A cross-sectional analysis of 437 men who attended our outpatient clinic between June 2021 and October 2021 for couple pregnancy loss. All subjects underwent a complete physical examination, palpation, inspection of the male genitalia, and semen analysis. Validated assessment tools for erectile dysfunction (the International Index of Sexual Function5 -IIEF-5) and anxiety (the seven-item Generalized Anxiety Disorder Scale- GAD-7) were used. Results Among 437 men of couples with pregnancy loss, we found several relevant sperm parameters confirmed a significant correlation between IIEF-5 scores and sperm parameters, including: sperm progressive motility (r = 0.1627, p = 0.001), sperm normal morphology (r = 0.1373, p = 0.004) and sperm DNA fragmentation (r =—0.1248, p = 0.009). Males with an IIEF-5 scores range between 5–11 presented the worst results in terms of sperm progressive motility (p = 0.002), normal morphology (p = 0.001), and SDF levels (p = 0.003). GAD-7 score, as well as anxiety level, was significantly higher in those males with an IIEF-5 score between 5 and 11 (p = 0.000). Conclusion Although current evidence does not demonstrate the importance of spermatozoa in the etiology of pregnancy loss, significant correlations have been observed between impaired sperm quality and low IIEF-5 scores. Also, anxiety is more likely to occur in males with sexual dysfunction.

Abstract Men in couples that have experienced pregnancy loss have a higher risk of sexual dysfunction. Semen quality impairment is common in men of couples with pregnancy loss. The objective of this article is to evaluate the differences in the incidence of male sexual dysfunction in a cohort of pregnancy loss couples with different types of semen quality impairment. A cross-sectional analysis of 426 men who attended our outpatient clinic for couples’ pregnancy loss, those without genetic abnormalities were included in the final analysis covering June 2021 to October 2021. The patients were divided into 5 groups according to type of semen quality impairment: normozoospermia group (group normal; N = 161), high sperm DNA fragmentation group (group high-SDF; N = 87), isolated asthenozoospermia group (group iAstheno; N = 45), isolated teratozoopermia group (group iTerato; N = 44), and ≥ 2 abnormal sperm parameters group (group multiple; N = 89). All subjects underwent a complete physical inspection, including palpation of the male genitalia and semen analysis. Validated assessment tools for erectile dysfunction (the International Index of Erectile Function -IIEF-5) and anxiety (the seven-item Generalized Anxiety Disorder Scale- GAD-7) were also used. Men with high sperm DNA fragmentation and isolated teratozoopermia were associated with increased erectile dysfunction risk when compared with normozoospermic men, with an OR of 2.75 [1.49–5.09; p = 0.001] and 2.44 [1.22–5.31; p = 0.024], respectively. It is interesting to note that there was no difference in prevalence of erectile dysfunction between Group iAstheno and Group normal (20.0% vs. 18.0%; OR = 1.24 [0.52–2.97]; P = 0.625). China has proposed the ambitious target of peaking the carbon emissions by 2030 and realizing the carbon neutrality by 2060 (hereafter referred to as “double carbon” goal). Central China is supposed to be one of the most essential aspect for this target. However, renewable resources in the central China region encounter the issues of inferior capacity factor and restricted resource potential. A resource analysis for local onshore wind and solar PV is conducted here, incorporating inversion analysis data with high spatial and temporal resolutions (include wind field, surface radial, local elevation, slope and landscape), to propose the (1) spatial distribution and seasonal fluctuation of local onshore wind and solar PV; (2) supplied curves for all the provincial regions in Central China at every renewable investment level; (3) optimal provincial allocation and overall portfolio for local renewables in the Central China. This research is supposed to provide data and model support for the ongoing deployment of renewables in Central China, and the fulfillment of the “double carbon” goal in the entire country.

Abstract Background Infertile men with higher sexual dysfunction risk and increased psychological burden, were also associated with more inclined to timed intercourse. Decreased semen quality may have adverse effects on male sexual function. However, it is also likely that many of these sequences do not play a direct role, those negative consequences may depend mainly on the later failed attempting pregnancy. Research is limited in this area. Methods This cross-sectional study was based on a group of 509 men who were assessed for couple’s infertility at the First Hospital of Jilin University between June 2021 and October 2021. All the men completed a comprehensive questionnaire, and then were divided in two groups. Group A included patients who either never received a routine infertility work-up or done so recently within the last 6 months. Group B included patients who previously received a sperm quality assessment at least 6 months or more prior. Patients were further categorized into three subgroups according to the severity of the decreases in their sperm parameters: severe, mild-moderate, and normozoospermic. Results The prevalence of erectile dysfunction was higher in Group B Mild-Moderate and Group B Severe in comparison to Group A (OR=1.86 [1.07–3.24], P = 0.027; OR=5.312 [2.69–10.49], P < 0.001, respectively). No significant differences were found between Group A and Group B-normozoospermic. Similar results were observed in the prevalence of premature ejaculation between the groups. Timed intercourse was seen in 11.8% (20/170) of men in Group A and 16.2% (19/117) in Group B-normozoospermic. It was more commonly practiced among infertile men in Group B-Mild-Moderate and Group B Severe, as 28.2% (44/156) and 25.7% (17/66) of these couples had attempted to conceive through timed intercourse (P < 0.001). Conclusions Our findings indicate that the severity of sperm quality impairment was negatively associated with sexual dysfunction only in infertile men who with known impairment of sperm quality for a long period. Timed intercourse was more common among these couples. For those individuals had never test their sperm quality, although more than half of these patients showed a decrease in sperm quality, the incidence of sexual dysfunction is relatively low and were comparable to those men examined previously known as normozoospermic.

The automatic classification of apple tree organs is of great significance for automatic pruning of apple trees, automatic picking of apple fruits, and estimation of fruit yield. However, there are some problems of dense foliage, partial occlusion and clustering of apple fruits. All of the problems above would contribute to the difficulties of organs classification and yield estimation of the apple trees. In this paper a method based on Color and Shape Multi-features Fusion and Support Vector Machine (SVM) for 3D apple tree organs classification and yield estimation was proposed. The method was designed for dwarf and densely planted apple trees at the early and late maturity stages. 196-dimensional feature vectors composed with Red Green Blue (RGB), Hue Saturation Value (HSV), Curvatures, Fast Point Feature Histogram (FPFH), and Spin Image were extracted firstly. And then the SVM based on linear kernel function was trained, after that the trained SVM was used for apple tree organs classification. Then the position weighted smoothing algorithm was used for classified apple tree organs smoothing. Then the agglomerative hierarchical clustering algorithm was used to recognize single apple fruit for yield estimation. On the same training and test set the experimental results showed that the SVM based on linear kernel function outperformed the KNN algorithm and Ensemble algorithm. The Recall, Precision and F1 score of the proposed method for yield estimation were 93.75%, 96.15% and 94.93% respectively. In summary, to solve the problems of apple tree organs classification and yield estimation in natural apple orchard, a novelty method based on multi-features fusion and SVM was proposed and achieve good performance. Moreover, the proposed method could provide technical support for automatic apple picking, automatic pruning of fruit trees, and automatic information acquisition and management in orchards.

We showed that a mobile handheld laser scanner (HHLS) provides useful features concerning the wood quality-influencing external structures of trees. When linked with wood properties measured at a sawmill utilizing state-of-the-art X-ray scanners, these data enable the training of various wood quality models for use in targeting and planning future wood procurement. A total of 457 Norway spruce sample trees (Picea abies (L.) H. Karst.) from 13 spruce-dominated stands in southeastern Finland were used in the study. All test sites were recorded with a ZEB Horizon HHLS, and the sample trees were tracked to a sawmill and subjected to X-rays. Two branch extraction techniques were applied to the HHLS point clouds: 1) a method developed in this study that was based on the density-based spatial clustering of applications with noise (DBSCAN) and 2) segmentation-based quantitative structure model (treeQSM). On average, the treeQSM method detected 46% more branches per tree than the DBSCAN did. However, compared with the X-rayed references, some of the branches detected by the treeQSM may either be false positives or so small in size that the X-rays are unable to detect them as knots, as the method overestimated the whorl count by 19% when compared with the X-rays. On the other hand, the DBSCAN method only detected larger branches and showed a −11% bias in whorl count. Overall, the DBSCAN underestimated knot volumes within trees by 6%, while the treeQSM overestimated them by 25%. When we input the HHLS features into a Random Forest model, the knottiness variables measured at the sawmill were predicted with R2s of 0.47–0.64. The results were comparable with previous results derived with the static terrestrial laser scanners. The obtained stem branching data are relevant for predicting wood quality attributes but do not provide data that are directly comparable with the X-ray features. Future work should combine terrestrial point clouds with dense above-canopy point clouds to overcome the limitations related to vertical coverage.

Tree species characterise biodiversity, health, economic potential, and resilience of an ecosystem, for example. Tree species classification based on remote sensing data, however, is known to be a challenging task. In this paper, we study for the first time the feasibility of tree species classification using high-density point clouds collected with an airborne close-range multispectral laser scanning system – a technique that has previously proved to be capable of providing stem curve and volume accurately and rapidly for standing trees. To this end, we carried out laser scanning measurements from a helicopter on 53 forest sample plots, each with a size of 32 m × 32 m. The plots covered approximately 5500 trees in total, including Scots pine (Pinus sylvestris L.), Norway spruce (Picea abies (L.) H.Karst.), and deciduous trees such as Downy birch (Betula pubescens Ehrh.) and Silverbirch (Betula pendula Roth). The multispectral laser scanning system consisted of integrated Riegl VUX-1HA, miniVUX-3UAV, and VQ-840-G scanners (Riegl GmbH, Austria) operating at wavelengths of 1550 nm, 905 nm, and 532 nm, respectively. A new approach, layer-by-layer segmentation, was developed for individual tree detection and segmentation from the dense point cloud data. After individual tree segmentation, 249 features were computed for tree species classification, which was tested with approximately 3000 trees. The features described the point cloud geometry as well as single-channel and multi-channel reflectance metrics. Both feature selection and the tree species classification were conducted using the random forest method. Using the layer-by-layer segmentation algorithm, trees in the dominant and co-dominant categories were found with detection rates of 89.5% and 77.9%, respectively, whereas suppressed trees were detected with a success rate of 15.2%–42.3%, clearly improving upon the standard watershed segmentation. The overall accuracy of the tree species classification was 73.1% when using geometric features from the 1550 nm scanner data and 86.6% when combining the geometric features with reflectance information of the 1550 nm data. The use of multispectral reflectance and geometric features improved the overall classification accuracy up to 90.8%. Classification accuracies were as high as 92.7% and 93.7% for dominant and co-dominant trees, respectively.

The plum rain season is a special climatic phenomenon in east Asia, which is characterized by persistent rainfall, a high temperature, and humidity, providing suitable environmental conditions for certain pathogenic bacteria, thus increasing the incidence of respiratory, gastrointestinal, and urinary diseases. However, studies on human opportunistic pathogenic bacteria communities during the plum rain season are still limited. In this study, the characteristics of human opportunistic pathogenic bacterial communities on daily necessities during the non-plum and plum rain seasons were investigated using high-throughput sequencing technology. The results revealed that the relative abundance of human opportunistic pathogenic bacteria was higher in the plum rain season (cotton cloth: 2.469%, electric bicycles: 0.724%, rice: 3.737%, and washbasins: 5.005%) than in the non-plum rain season (cotton cloth: 1.425%, electric bicycles: 0.601%, rice: 2.426%, and washbasins: 4.801%). Both temperature and relative humidity affected human opportunistic pathogenic bacterial communities. Stochastic processes dominated the assembly process of human opportunistic pathogenic bacterial communities, and undominated processes prevailed. The stability of the co-occurrence network was higher in the non-plum rain season than that in the plum rain season. In addition, the proportion of deterministic processes showed the same trend as the complexity of the co-occurrence network.

During the picking process of the apple harvesting robot, the attitude of the end effector holding the apple and the movement method of separating the apple directly affect the success rate of picking. In order to improve the stability of the picking process, reduce the gripping force, and avoid apple dislodgement and damage, this work studies the new apple-picking pattern of the flexible three-fingered end-effector based on the analysis of the existing apple-picking pattern. First, two new three-finger grasping postures for wrapping the apple horizontally and vertically on the inside of the fingers are proposed, and a new method of separating the stem with a circular-pull-down motion of the end-effector picking the apple is designed. Then, the pressure on the apple under different picking patterns was analyzed, and a branch–stem–apple simulation model was established. Combining the constraint conditions such as the angle between the apple stem and the vertical direction, the movement speed, the root impulse, and so on, the optimal angle of apple circular movement and the force required to realize the movement are obtained through dynamic simulation experiments. Finally, the experiments of apple picking patterns were carried out with the flexible three-fingered end-effector. The experiment shows that the best angle for apple picking is 15°~20° using the circular-pull-down movement separation method. In terms of average grasping force peaks and pressures, the combination of the vertical holding posture of the inner finger and the circular-pull-down movement separation method is the best picking pattern. In this pattern, the average peak exerts force on the inner side of a single finger is about 8.52 N, and the pressure is about 20.9 KPa.

Abstract The link between sexual dysfunction and male infertility has been well established. In addition to male infertility, male patients with couple pregnancy loss and preconception care are the most frequent reasons for the treatment of andrology outpatients. However, there is a paucity of information simultaneously investigating male sexual dysfunction in these males with different reproduction situations. A cross-sectional study was performed in consecutive series of 1256 participants, including 509 men with infertility, 437 couples with pregnancy loss, and 310 men for preconception care. All men completed a questionnaire on baseline demographic information, sexual behavior characteristics and validated research tools, including Premature Ejaculation Diagnostic Tool, seven-item Generalized Anxiety Disorder Scale, and International Index of Erectile Function. ABSTRACT: β-Galactosidase (lacA) from Aspergillus oryzae is widely used in the dairy industry. Its acidic pH optimum and severe product inhibition limit its application for lactose hydrolysis in milk. In the present study, structure-based sequence alignment was conducted to determine the candidate mutations to shift the pH optimum of lacA to the neutral range. The Y138F and Y364F mutants shifted the pH optimum of lacA from 4.5 to 5.5 and 6.0, respectively. The acid dissociation constant (pKa) values of catalytic acid/base residues with upwards shift were consistent with the increased pH optimum. All variants in the present study also alleviated galactose inhibition to various extents. Molecular dynamics demonstrated that the less rigid tertiary structures and lower galactose-binding free energy of Y138F and Y364F might facilitate the release of the end product. Both Y138F and Y364F mutants exhibited better hydrolytic ability than lacA in milk lactose hydrolysis. The higher pH optimum and lower galactose inhibition of Y138F and Y364F may explain their superiority over lacA. The Y138F and Y364F mutants in the present study showed potential in producing low-lactose milk, and our studies provide a novel strategy for engineering the pH optimum of glycoside hydrolase.

BackgroundAmbroxol is a widely used mucoactive drug in sputum clearance of respiratory diseases taken orally and by injection. However, there is a paucity of evidence for inhaled ambroxol in sputum clearance.MethodsThis study performed a multicenter, randomized, double-blind, placebo-controlled, phase 3 trial at 19 centers in China. Hospitalized adult patients with mucopurulent sputum and expectoration difficulty were recruited. Patients were randomized by 1:1 to receive inhalation of either ambroxol hydrochloride solution 3 mL (22.5 mg) + 0.9% sodium chloride 3 mL or 0.9% sodium chloride 6 mL twice daily for 5 days, with an interval of more than 6 h. The primary efficacy endpoint was the absolute change in the sputum property score after treatment compared to the baseline in the intention-to-treat population.ResultsBetween 10 April 2018 and 23 November 2020, 316 patients were recruited and assessed for eligibility, of whom 138 who received inhaled ambroxol and 134 who received a placebo were included. Patients who received inhaled ambroxol had a significantly greater decrease in the sputum property score compared with patients who received inhalation of placebo (difference: −0.29; 95% CI: −0.53 to −0.05; p = 0.0215). Compared with the placebo, inhaled ambroxol also significantly reduced more expectoration volume in 24 h (difference: −0.18; 95% CI: −0.34 to −0.03; p = 0.0166). Introductionβ-Mannanases can hydrolyze mannans, which are widely available in nature. However, the optimum temperature of most β-mannanases is too low to be directly utilized in industry.MethodsTo further improve the thermostability of Anman (mannanase from Aspergillus niger CBS513.88), B-factor and Gibbs unfolding free energy change were used to modify the flexible of Anman, and then combined with multiple sequence alignment and consensus mutation to generate an excellent mutant. At last, we analyzed the intermolecular forces between Anman and the mutant by molecular dynamics simulation.ResultsThe thermostability of combined mutant mut5 (E15C/S65P/A84P/A195P/T298P) was increased by 70% than the wild-type Amman at 70°C, and the melting temperature (Tm) and half-life (t1/2) values were increased by 2°C and 7.8-folds, respectively. Molecular dynamics simulation showed reduced flexibility and additional chemical bonds in the region near the mutation site.DiscussionThese results indicate that we obtained a Anman mutant that is more suitable for industrial application, and they also confirm that a combination of rational and semi-rational techniques is helpful for screening mutant sites.

BackgroundInfected bone nonunion is the toughest problem in fracture-related infection, leading to high disability and recurrence. The aim of this study was to evaluate the effectiveness of the Masquelet technique with radical debridement and alternative fixation in the management of infected bone nonunion.Patients and MethodsA retrospective study of prospectively collected data in two trauma centers was performed from 2016 to 2020. Patients diagnosed as infected bone nonunion were included in this study. The initial implant was removed and all patients received a two-stage Masquelet procedure with radical debridement and alternative fixation. The disappearance of inflammatory manifestations and regression of infection indicators (such as interleukin-6 (IL-6), C-reactive protein, white blood cell count) to the normal range were regarded as radical debridement. The alternative fixation depended on local soft tissue conditions. Results were evaluated according to clinical and radiographic assessment and patient satisfaction.ResultsA total of 23 patients were included in our study. Six of them received internal fixation, while the other 17 received external fixation. Of the 23 cases, 21 were successfully reconstructed without infection recurrence, except 2 reinfected cases. Mean full weight bearing time was 6.6 months follow-up post last surgery. Out of the 23, 20 cases had satisfactory functional outcomes without additional bone or soft tissue comorbidities. Discrepancies in leg length and joint stiffness were observed in three cases and marked as unsatisfied results.ConclusionsInfected bone nonunion can be successfully managed using the Masquelet technique under radical debridement combined with an alternative fixation method.

BackgroundInflammatory markers are being increasingly used to predict the prognosis of cancer patients. We hereby conducted the first meta-analysis assessing the association between systemic inflammation score (SIS) and prognosis of gastric cancer patients undergoing surgical intervention.MethodsA literature search was carried out on PubMed, CENTRAL, Scopus, and Embase up to 3rd June 2022 for relevant studies. Adjusted data reported as hazard ratios (HR) was combined in a random-effects model.ResultsA total of seven studies with 5,338 patients could be included. All studies were from either China or Japan and published in the last four years. Meta-analysis showed that higher SIS scores (1 or 2) were significant predictors of poor overall survival (OS) in gastric cancer patients (HR: 1.25 95% CI: 1.05, 1.49, I2 = 11%). Similarly, the meta-analysis demonstrated that an SIS score of 2 was associated with poor OS as compared to scores of 0/1 (HR: 2.53 95% CI: 1.30, 4.89, I2 = 45%). Data on disease-free survival (DFS) was scarce to draw conclusions.ConclusionThe SIS score can be a simple and useful tool to predict OS in gastric cancer patients undergoing surgery. Data on DFS is scarce and conflicting. Many techniques have been proposed to harden programs with protection mechanisms to defend against vulnerability exploits. Unfortunately the vast majority of them cannot be applied to closed source software because they require access to program source code. This paper presents our work on automatically hardening binary code with security workarounds, a protection mechanism that prevents vulnerabilities from being triggered by disabling vulnerable code. By working solely with binary code, our approach is applicable to closed source software. To automatically synthesize security workarounds, we develop binary program analysis techniques to identify existing error handling code in binary code, synthesize security workarounds in the form of binary code, and instrument security workarounds into binary programs. We designed and implemented a prototype or our approach for Windows and Linux binary programs. Our evaluation shows that our approach can apply security workarounds to an average of 69.3% of program code and the security workarounds successfully prevents exploits to trigger real-world vulnerabilities.

ABSTRACT Little is unknown about the regulatory mechanisms underlying the pathogenesis of osteomyelitis induced by Staphylococcus aureus. Hypoxia-inducible factor-1α (HIF-1α) and transforming growth factor β1 (TGF-β1) were both upregulated in S. aureus-infected MC3T3-E1 cells and osteomyelitis patients. HIF-1α directly targets the hypoxia-responsive elements (HREs) of TGF-β1 mRNA to induce its expression. Silencing HIF-1α and TGF-β1, as well as treatment of hypoxia inhibitor IDF-11774, consistently elevated OPN and RUNX2 expression and alizarin Red S (ARS) and alkaline phosphatase (ALP) staining levels in MC3T3-E1 cells with S. aureus infection. S. aureus infection increased HIF-1α expression and serum TGF-β1 concentration in a mouse model of osteomyelitis. Hypoxia inhibitor IDF-11774 treatment reduced serum levels of interleukin (IL)-6, IL-1β, and C-reactive protein. Upon S. aureus infection, hypoxia was activated to trigger TGF-β1 upregulation through direct targeting of HRE on TGF-β1 mRNA by HIF-1α, eventually leading to osteomyelitis symptoms in terms of osteogenesis and mineralization deficiencies as well as elevated inflammation. This study hereby suggests a novel signaling cascade involving hypoxia/HIF-1α/TGF-β1 in osteomyelitis pathogenesis, which could potentially serve as a target for therapeutic measures. IMPORTANCE The pathogenesis of osteomyelitis induced by Staphylococcus aureus remains unclear. To develop therapeutic approaches for osteomyelitis, it is important to understand the molecular mechanisms of its pathogenesis. Our results suggests that hypoxia/HIF-1α/TGF-β1 signaling is involved in osteomyelitis pathogenesis. Thus, these findings highlight the potential of this signaling components as therapeutic targets for the treatment of osteomyelitis.

Background: The complexity of the spatial dynamic flexion axis (DFA) of the elbow joint makes the elbow prosthesis design and humeral component alignment challenging. This study aimed to 1) investigate the variations of the spatial DFA during elbow flexion and 2) investigate the relationship between the distal humeral trochlear geometry and the in vivo spatial variation of the DFA.Methods: Ten healthy subjects participated in this study. Each subject performed a full elbow extension to maximum flexion with hand supination under dual fluoroscopic imaging system (DFIS) surveillance. The 2D fluoroscopic images and the 3D bone models were registered to analyze the in vivo elbow kinematics and DFAs. The spatial DFA positions were defined as inclination with the medial and lateral epicondyle axes (MLA) in the transverse and coronal planes. The range of the DFA positions was also investigated during different flexion phases. The Spearman correlation method was used to analyze the relationship between the distal humeral trochlear’s morphological parameters and the position of DFAs during different flexion phases.Results: The pathway of the DFAs showed an irregular pattern and presented individual features. The medial trochlear depth (MTD) (r = 0.68, p = 0.03) was positively correlated with the range of the DFA position (2.8° ± 1.9°) in the coronal plane from full extension to 30° of flexion. Lateral trochlear height (LTH) (r = −0.64, p = 0.04) was negatively correlated with the DFA position (−1.4° ± 3.3°) in the transverse plane from 30° to 60° of flexion. A significant correlation was found between LTH with the DFA position in the coronal (r = −0.77, p = 0.01) and transverse planes (r = −0.76, p = 0.01) from 60° to 90° of flexion.Conclusion: This study showed that the pathway of the dynamic flexion axis has an individual pattern. The medial and lateral trochlear sizes were the key parameters that might affect the elbow joint flexion function. When recovering complex distal humeral fractures or considering the implant design of total elbow arthroplasty, surgeons should pay more attention to the medial and lateral trochlea’s geometry, which may help restore normal elbow kinematics.

Today, high-quality reference tree measurements, including the position, diameter, height and volume, are cumbersome and slow to carry out, but highly needed for forest inventories based on airborne laser scanning. Mobile laser scanning technologies hold the promise for collecting reference data for forest inventories with an extremely high efficiency. Perhaps, the most efficient approach for reference data collection would be to mount a high-resolution laser scanning system on board an airborne vehicle flying at a low altitude above the forest canopy since this would allow recording reference samples of individual trees with the speed of flight. To demonstrate the potential of this technology, we mounted an in-house developed HeliALS-DW laser scanning system on board a helicopter and collected point cloud data in a boreal forest on three test sites containing a total of 1469 trees. The obtained point clouds incorporated sufficiently many high-quality stem hits for estimating the stem curves and stem volumes of individual trees since the point clouds had a relatively high point density of 2200–3800 echoes/m2, and the scanner had been tilted by 15° from the nadir to increase the possibility of recording stem hits. To automatically estimate the diameters at breast height (DBH) and stem curves of individual trees, we used algorithms designed to tolerate moderate drifts in the trajectory of the laser scanner. Furthermore, the stem volumes of individual trees were computed by using the estimated stem curves and tree heights without any allometric models. Using the proposed methods, we were able to estimate the stem curves with a root-mean-square error (RMSE) of 1.7–2.6 cm (6–9%) while detecting 42–71% of the trees. The RMSE of stem volume estimates was 0.1–0.15 m3 (12–21%). We also showed that the tree detection rate could be improved up to 87–96% for trees with a DBH exceeding 20 cm if slightly larger average errors for the stem attributes were allowed. Our results pave the way for using high-resolution airborne laser scanning for field reference data collection by conducting direct measurements of tree stems with a high efficiency.

ABSTRACT While circulating cell-free DNA (cfDNA) is becoming a powerful marker for noninvasive identification of infectious pathogens in liquid biopsy specimens, a microbial cfDNA baseline in healthy individuals is urgently needed for the proper interpretation of microbial cfDNA sequencing results in clinical metagenomics. Because noninvasive prenatal testing (NIPT) shares many similarities with the sequencing protocol of metagenomics, we utilized the standard low-pass whole-genome-sequencing-based NIPT to establish a microbial cfDNA baseline in healthy people. Sequencing data from a total of 107,763 peripheral blood samples of healthy pregnant women undergoing NIPT screening were retrospectively collected and reanalyzed for microbiome DNA screening. It was found that more than 95% of exogenous cfDNA was from bacteria, 3% from eukaryotes, and 0.4% from viruses, indicating the gut/environment origins of many microorganisms. Overall and regional abundance patterns were well illustrated, with huge regional diversity and complexity, and unique interspecies and symbiotic relationships were observed for TORCH organisms (Toxoplasma gondii, others [Treponema pallidum {causing syphilis}, hepatitis B virus {HBV}, and human parvovirus B19 {HPV-B19}], rubella virus, cytomegalovirus [CMV], and herpes simplex virus [HSV]) and another common virus, Epstein-Barr virus (EBV). To sum up, our study revealed the complexity of the baseline circulating microbial cfDNA and showed that microbial cfDNA sequencing results need to be interpreted in a more comprehensive manner. Purpose: To retrospectively report a case of Weill-Marchesani syndrome 4 (WMS4) with compound heterozygous variants of ADAMTS17 gene. Observations: The patient was a 7-year-old boy with progressively worsening eyesight and intermittent elevated intraocular pressure (IOP) for two years. His IOPs were temporarily controlled using anti-glaucoma drugs. At presentation he had a shallow anterior chamber, lens subluxation, spherophakia and extensive synechial angle closure with high myopia in both eyes. Ultrasound biomicroscopy (UBM) identified thickened zonule fibers and anteriorly rotated, flat and slender ciliary processes, both of which worsened and were accompanied by obvious iris bombe after miosis. Gene testing showed compound heterozygosity of a maternal submicroscopic deletion on chromosome 15q26.3 (0.774 Mb) affecting the sequences of ADAMTS17, LYSMD4 and CERS3 as well as a paternal nonsense variant (c.1051_1053delAAGinsTAA, P.K351X) in the ADAMTS17 gene in the proband. The diagnosis of WMS4 was confirmed by genetic testing. Phacoemulsification (Phaco), intraocular lens (IOL) implantation, and irido-zonulo-hyaloid-vitrectomy (IZHV) combined with Ahmed Glaucoma Valve (AGV) implantation as a staged or one-stage surgery effectively lowered IOP, deepened ACD, improved visual acuity, and resolved the configuration of the ciliary processes in both eyes. Conclusion and Importance: Recessive ADAMTS17 variants are associated with WMS4. We report here compound heterozygous variants in ADAMTS17 causing WMS4, and anatomically highlighted the possible pathophysiology for its clinical phenotype. A modified surgical approach with Phaco, IOL implantation, and IZHV combined with AGV implantation could be used to treat these complicated cases.

Abstract Background Osteoarthritis (OA) is a commonly known prevalent joint disease, with limited therapeutic methods. This study aimed to investigate the functions of miRNA-132 (miR-132) in the modulation of PTEN/PI3K/AKT signaling pathway in the development and progression of osteoarthritis. Methods Eight male osteoarthritic patients and eight healthy males were recruited. Male Sprague Dawley (SD) rats were used for cellular experiments. QRT-PCR was performed to detect the expression levels of miR-132, PTEN, PI3K and AKT. MTT assay and apoptosis assay were carried out to measure the cell proliferation rate and cell apoptosis rate, respectively. Western blotting was employed to detect the protein expression of related RNAs and inflammatory factors. Results In osteoarthritic patients, the expression level of miR-132 was decreased, compared with that in the normal group. Over-expression of miR-132 elevated cell proliferation and decreased apoptosis of chondrocytes. Down-regulation of miR-132 decreased cell proliferation and induced apoptosis in chondrocytes. In addition, down-regulation of miR-132 promoted the expression of Bax protein and activated caspase-3/9, increased inflammation divisors. PTEN inhibitor antagonized the destructive effect of the miR-132 inhibitor on cell proliferation of chondrocytes. PI3K inhibitor increased the destructive effect of the miR-132 inhibitor on osteoarthritis. Conclusion In conclusion, miR-132 is an important regulator of osteoarthritis in chondrocytes through the PTEN/PI3K/AKT signaling pathway.

The continuous flow of autonomous vehicle-based data could revolutionize current map updating procedures and allow completely new types of mapping applications. Therefore, in this article, we demonstrate the feasibility of using perception data of autonomous vehicles to replace traditionally conducted mobile mapping surveys with a case study focusing on updating a register of roadside city trees. In our experiment, we drove along a 1.3-km-long road in Helsinki to collect laser scanner data using our autonomous car platform ARVO, which is based on a Ford Mondeo hybrid passenger vehicle equipped with a Velodyne VLS-128 Alpha Prime scanner and other high-grade sensors for autonomous perception. For comparison, laser scanner data from the same region were also collected with a specially-planned high-grade mobile mapping laser scanning system. Based on our results, the diameter at breast height, one of the key parameters of city tree registers, could be estimated with a lower root-mean-square error from the perception data of the autonomous car than from the specially-planned mobile laser scanning survey, provided that time-based filtering was included in the post-processing of the autonomous perception data to mitigate distortions in the obtained point cloud. Therefore, appropriately performed post-processing of the autonomous perception data can be regarded as a viable option for keeping maps updated in road environments. However, point cloud-processing algorithms may need to be adapted for the post-processing of autonomous perception data due to the differences in the sensors and their arrangements compared to designated mobile mapping systems. We also emphasize that time-based filtering may be required in the post-processing of autonomous perception data due to point cloud distortions around objects seen at multiple times. This highlights the importance of saving the time stamp for each data point in the autonomous perception data or saving the temporal order of the data points.

Abstract Background Current automated forest investigation is facing a dilemma over how to achieve high tree- and plot-level completeness while maintaining a high cost and labor efficiency. This study tackles the challenge by exploring a new concept that enables an efficient fusion of aerial and terrestrial perspectives for digitizing and characterizing individual trees in forests through an Unmanned Aerial Vehicle (UAV) that flies above and under canopies in a single operation. The advantage of such concept is that the aerial perspective from the above-canopy UAV and the terrestrial perspective from the under-canopy UAV can be seamlessly integrated in one flight, thus grants the access to simultaneous high completeness, high efficiency, and low cost. Results In the experiment, an approximately 0.5 ha forest was covered in ca. 10 min from takeoff to landing. The GNSS-IMU based positioning supports a geometric accuracy of the produced point cloud that is equivalent to that of the mobile mapping systems, which leads to a 2–4 cm RMSE of the diameter at the breast height estimates, and a 4–7 cm RMSE of the stem curve estimates. Conclusions Results of the experiment suggested that the integrated flight is capable of combining the high completeness of upper canopies from the above-canopy perspective and the high completeness of stems from the terrestrial perspective. Thus, it is a solution to combine the advantages of the terrestrial static, the mobile, and the above-canopy UAV observations, which is a promising step forward to achieve a fully autonomous in situ forest inventory. Future studies should be aimed to further improve the platform positioning, and to automatize the UAV operation.

Conservation tillage is an advanced agricultural technology that seeks to minimize soil disturbance by reducing, or even eliminating tillage. Straw or stubble mulching in conservation tillage systems help to increase crop yield, maintain biodiversity and increase levels of exogenous nutrients, all of which may influence the structure of fungal communities in the soil. Currently, however, the assembly processes and co-occurrence patterns of fungal sub-communities remain unknown. In this paper, we investigated the effects of no-tillage and straw mulching on the composition, assembly process, and co-occurrence patterns of soil fungal sub-communities in a long-term experimental plot (15 years). The results revealed that combine straw mulching with no-tillage significantly increased the richness of fungi but not their diversity. Differential abundance analysis and principal component analysis (PCA) indicated that tillage management had a greater effect on the fungal communities of abundant and intermediate taxa than on the rare taxa. Available phosphorus (AP) and total nitrogen (TN) were the major determinants of fungal sub-communities in NT treatment. The abundant fungal sub-communities were assembled by deterministic processes under medium strength selection, while strong conservation tillage strength shifts the abundant sub-community assembly process from deterministic to stochastic. Overall, the investigation of the ecological network indicated that no-tillage and straw mulching practices decreased the complexity of the abundant and intermediate fungal networks, while not significantly influencing rare fungal networks. These findings refine our knowledge of the response of fungal sub-communities to conservation tillage management techniques and provide new insights into understanding fungal sub-community assembly.

Diabetes mellitus is a prevalent disease result in several complications, including bone problems. Previous studies have shown that microRNA (miR)-26a regulates glucose metabolism and plays a protective role in diabetes. However, whether miR-26a also affects bone quality in diabetes remains unknown. In the present study, we evaluated the potential effects of miR-26a on bone in diabetic mice. We administrated miR-26a in streptozotocin-induced diabetic mice. The metabolic parameters, bone quality, osteoblast and osteoclast markers, and insulin signaling activation were measured. miR-26a ameliorated insulin resistance and glucose tolerance, improved bone microarchitecture and quality, increased osteoblasts and bone formation, decreased osteoclasts, and promoted the insulin signaling pathway in diabetic mice. These effects were abolished in insulin receptor-compromised Col1a1-Insr+/– mice. In conclusion, miR-26a could ameliorate bone-specific insulin resistance and bone quality in diabetic mice, which depended on the insulin receptors on osteoblasts. Our findings highlight the potential of miR-26a as a therapeutic target for diabetes mellitus-related bone metabolism and diseases.

Abstract Background Whether IL-15 is involved in the development of steroid-induced osteonecrosis of the femoral head (ONFH) is investigated. Methods C57BL/6 J and l15−/−mice were injected with methylprednisolone to induce wide type osteonecrosis (WT ON) and IL-15 deficiency osteonecrosis (IL-15−/− ON). Hematoxylin-Eosin (H&E) staining and micro-computed tomography (micro-CT) scanning was used to detect the microstructure. The differentiation and formation of osteoclasts were determined with colony-forming unit-granulocyte macrophages (CFU-GM), colony-forming unit-macrophage/mononuclear (CFU-M) per tibia, and tartrate-resistant acid phosphatase (TRACP or TRAP) positive cells. Serum interleukin (IL)-15, osteocalcin, bone alkaline phosphatase (BAP), bone Gla protein (BGP), and TRACP were assayed with enzyme-linked immunosorbent assay (ELISA). The receptor activator of nuclear factor-κB (RANK), RANK ligand (RANKL), and osteoprotegerin (OPG) in the femoral heads were detected by Western blot. CD34 staining was performed to detect microvascular density. Results IL-15 secretion was increased in the femoral heads and the serum of steroid-induced ONFH mice. IL-15 deficiency may lead to up-regulated vessel remodeling, improved microstructure, and up-regulated serum osteocalcin, BAP, and BGP secretion. Both the expression of RANKL/RANK/OPG and osteoclast differentiation and formation can be down-regulated by IL-15 deficiency. Conclusion IL-15 deficiency alleviates steroid-induced ONFH by impact osteoclasts via RANKL-RANK-OPG system.

Objective(s): The purpose of this study is to investigate the indication function of the calcium circulation-related factors on the damage to muscle strength and contraction function after nerve injury. The target factors include ryanodine receptor (RyR), inositol-1,4,5-triphosphate receptor (IP3R), phospholamban (PLN), cryptocalcitonin (CASQ), ATPase and troponin C (TNNC).Materials and Methods: Sprague-Dawley (SD) rats were randomly divided into sham-operated group (SO), sciatic nerve injury group (SNI) and sciatic nerve disconnection group (SNT). Sciatic nerve function index and stretching test were used to examine the changes to muscle strength; bilateral gastrocnemius muscles were extracted after execution for gastrocnemius wet weight ratio test. HE staining slides and average cross-sectional area of muscle fibers were acquired to analyze the muscle atrophy. The transcription level of the factors was also measured.Results: Sciatic nerve damage in SNI group was significantly higher than that in SO group in the 6 weeks, but there was no significant difference between SNT and SO groups fallowing sciatic nerve damage. Sciatic nerve function in SNT group was worse than that in SNI group. The average cross-sectional area of gastrocnemius muscle fibers in SNI and SNT groups was significantly reduced compared to that in SO group. The transcriptional levels of RyR, PLN, CASQ, ATPase and TNNC in SNI and SNT groups were significantly different from those in SO group.Conclusion: Calcium circulation-related factors could be used as potential indicators for assessment of damages to muscle strength.

Abstract Background The antisense cerebellar degenerative-related protein-1 (CDR1as) has been identified as a sponge for several microRNAs. MiR-641 has been shown to be downregulated in osteoarthritic human chondrocytes, but its regulation and function in osteoarthritis (OA) has not been reported. Methods OA cartilage samples were obtained from the knee joints of 12 patients (8 males and 4 females at age of 57–73 years old) who underwent total knee arthroplasty. Normal articular cartilage samples were obtained from the knee joints of 10 trauma patients at age of 29–65 years old (6 males and 4 females). The levels of circRNA-CDR1as mRNA and miR-641 were examined by qRT-PCR and the contents of type II collagen (Col II), IL-6, MMP13 and GAPDH in chondrocytes were examined by Western blot. Results In this study, we found that circRNA-CDR1as level was significantly upregulated in OA chondrocytes, and negatively related with that of miR-641. RNA pull down assay confirmed that circRNA-CDR1as directly targets to miR-641. Furthermore, downregulation of circRNA-CDR1as increased type II collagen level but reduced MMP13 and IL-6 contents, while these effects were partly reversed by down-regulation of miR-641. Conclusion Overall, our results indicate that circRNA-CDR1as plays a crucial role in regulating OA progression via modulating extracellular matrix metabolism and inflammation via sponging miR-641 and provide a novel regulatory role of circRNA-CDR1as in OA.

Forest is one of the most challenging environments to be recorded in a three-dimensional (3D) digitized geometrical representation, because of the size and the complexity of the environment and the data-acquisition constraints brought by on-site conditions. Previous studies have indicated that the data-acquisition pattern can have more influence on the registration results than other factors. In practice, the ideal short-baseline observations, i.e., the dense collection mode, is rarely feasible, considering the low accessibility in forest environments and the commonly limited labor and time resources. The wide-baseline observations that cover a forest site using a few folds less observations than short-baseline observations, are therefore more preferable and commonly applied. Nevertheless, the wide-baseline approach is more challenging for data registration since it typically lacks the required sufficient overlaps between datasets. Until now, a robust automated registration solution that is independent of special hardware requirements has still been missing. That is, the registration accuracy is still far from the required level, and the information extractable from the merged point cloud using automated registration could not match that from the merged point cloud using manual registration. This paper proposes a discrete overlap search (DOS) method to find correspondences in the point clouds to solve the low-overlap problem in the wide-baseline point clouds. The proposed automatic method uses potential correspondences from both original data and selected feature points to reconstruct rough observation geometries without external knowledge and to retrieve precise registration parameters at data-level. An extensive experiment was carried out with 24 forest datasets of different conditions categorized in three difficulty levels. The performance of the proposed method was evaluated using various accuracy criteria, as well as based on data acquired from different hardware, platforms, viewing perspectives, and at different points of time. The proposed method achieved a 3D registration accuracy at a 0.50-cm level in all difficulty categories using static terrestrial acquisitions. In the terrestrial-aerial registration, data sets were collected from different sensors and at different points of time with scene changes, and a registration accuracy at the raw data geometric accuracy level was achieved. These results represent the highest automated registration accuracy and the strictest evaluation so far. The proposed method is applicable in multiple scenarios, such as 1) the global positioning of individual under-canopy observations, which is one of the main challenges in applying terrestrial observations lacking a global context, 2) the fusion of point clouds acquired from terrestrial and aerial perspectives, which is required in order to achieve a complete forest observation, 3) mobile mapping using a new stop-and-go approach, which solves the problems of lacking mobility and slow data collection in static terrestrial measurements as well as the data-quality issue in the continuous mobile approach. Furthermore, this work proposes a new error estimate that units all parameter-level errors into a single quantity and compensates for the downsides of the widely used parameter- and object-level error estimates; it also proposes a new deterministic point sets registration method as an alternative to the popular sampling methods.

Oxidative damage can lead to severe ovarian dysfunctions and even premature ovarian failure. Nrf2, a significant transcription factor that regulates the oxidative stress response of cells, declines with age. Daphnetin, as a kind of natural Chinese herbal medicine, can activate Nrf2 and further promote the antioxidant defense of cells. However, whether Daphnetin treatment can protect ovary from premature ovarian failure and the specific mechanism involved are not understood. This study aimed to investigate the protective function of Daphnetin against the ovarian aging induced by D-galactose in wild-type and Nrf2−/− mice. Female C57BL/6 mice with Wild-type and Nrf2−/− were divided into five groups separately and the premature ovarian failure model were established by D-galactose and then Daphnetin and VE were given for treatment. After 42 days, ovaries tissue and serum were collected for biochemical determination, H&E staining, Immunohistochemical staining and western blot analysis. In the WT-POF group, ovarian function was broke, and the expression of the ovarian senescence-associated protein P16 and the level of oxidative stress were significantly increased, while the expression of the anti-senescence protein klotho was significantly decreased. In addition, the expression of Nrf2 and the antioxidases GCLC, HO-1 and NQO1 were decreased, but TXNIP and NLRP3 were significantly increased. Furthermore, the characteristics of premature ovarian failure were more significant in Nrf2 knockout mice than in wild-type mice, especially the expression of NLRP3 and TXNIP. Moreover, daphnetin, an Nrf2 activator, rescued d-gal-induced POF in a dose-dependent manner, while the protective effect was weakened or even lost in Nrf2 knockout mice. Our results suggested that daphnetin is likely to be a candidate drug for premature ovarian failure treatment and it is mostly possible referred to the molecular mechanism of increasing Nrf2 expression and inhibiting NLRP3 activation in the ovarian aging process.

In this paper, we present a simple, efficient, and robust algorithm for 2D coarse registration of two point clouds. In the proposed algorithm, the locations of some distinct objects are detected from the point cloud data, and a rotation- and translation-invariant feature descriptor vector is computed for each of the detected objects based on the relative locations of the neighboring objects. Subsequently, the feature descriptors obtained for the different point clouds are compared against one another by using the Euclidean distance in the feature space as the similarity criterion. By using the nearest neighbor distance ratio, the most promising matching object pairs are found and further used to fit the optimal Euclidean transformation between the two point clouds. Importantly, the time complexity of the proposed algorithm scales quadratically in the number of objects detected from the point clouds. We demonstrate the proposed algorithm in the context of forest inventory by performing coarse registration between terrestrial and airborne point clouds. To this end, we use trees as the objects and perform the coarse registration by using no other information than the locations of the detected trees. We evaluate the performance of the algorithm using both simulations and three test sites located in a boreal forest. We show that the algorithm is fast and performs well for a large range of stem densities and for test sites with up to 10 ​000 trees. Additionally, we show that the algorithm works reliably even in the case of moderate errors in the tree locations, commission and omission errors in the tree detection, and partial overlap of the data sets. We also demonstrate that additional tree attributes can be incorporated into the proposed feature descriptor to improve the robustness of the registration algorithm provided that reliable information of these additional tree attributes is available. Furthermore, we show that the registration accuracy between the terrestrial and airborne point clouds can be significantly improved if stem positions estimated from the terrestrial data are matched to stem positions obtained from the airborne data instead of matching them to tree top positions estimated from the airborne data. Even though the 2D coarse registration algorithm is demonstrated in the context of forestry, the algorithm is not restricted to forest data and it may potentially be utilized in other applications, in which efficient 2D point set registration is needed.

Background: The present study aims to investigate the roles of microRNA-155 in collagen-induced arthritis (CIA) and its underlying mechanisms. Methods: CIA mouse model was established and miR155 inhibitor was intravenously injected. In in vitro studies, bone marrow-derived macrophages (BMDMs) were induced into M1 macrophages followed by the treatment of miR155 inhibitor. Quantitative reverse transcription PCR (RT-qPCR) was applied to determine the mRNA expressions. Flow cytometry was applied to determine the frequency of M1 or M2 macrophages. Western blotting was determined to detect protein expressions. Enzyme-linked immunosorbent assay (ELISA) was applied to determine the levels of inflammatory cytokines and anti-collagen antibody. Results: The levels of miR155 were increased in macrophages from rheumatoid arthritis (RA) patients and M1 macrophages. The treatment of miR155 inhibitor decreased inflammatory cytokines in M1 macrophages. Besides, treatment of miR155 inhibitors promoted the differentiation of M0 macrophages into M2 macrophages. In vivo studies demonstrated that the treatment of miR155 inhibitors ameliorated the RA symptoms by decreasing inflammatory cytokines in the CIA mouse model. Treatment of miR155 also resulted in a decrease of M1 macrophage biomarker and an increase of M2 macrophage biomarker. Conclusion: microRNA-155 inhibitor ameliorates RA symptoms in part by regulating macrophage phenotypes.

Reviewing forest carbon sinks is of the utmost importance in efforts to control climate change. This study focuses on reporting the 20-year boreal forest growth values acquired with airborne laser scanning (ALS). The growth was examined on the Kalkkinen research site in southern Finland as a continuation of several earlier growth studies performed in the same area. The data for the study were gathered with three totally different airborne laser scanning systems, namely using Toposys-I Falcon in June 2000 and Riegl VUX-1HA and miniVUX-3UAV in June 2021 with approximate point densities of 11, 1360, and 460 points/m2, respectively. The ALS point cloud was preprocessed to identify individual trees, from each of which different features were extracted either for direct or indirect growth measurement. In the direct method, the growth value is predicted based on differences of features, whereas in the indirect method, the growth value is obtained by subtracting the results of two independent predictions of different years. The growth in individual tree attributes, such as growth in height, diameter at breast height (DBH), and stem volume, were calculated for direct estimation. Field reference campaigns were performed in the summer of 2001 and in November 2021 to validate the obtained growth values. The study showed that long-term series growth of height, DBH, and stem volume are possible to record with a high-to-moderate coefficient of determination (R2) of 0.90, 0.48, and 0.45 in the best-case scenarios. The respective root-mean-squared errors (RMSE) values were 0.98 m, 0.02 m, and 0.17 m3, and the biases were −0.06 m, 0.00 m, and 0.17 m3. The direct method produced better metrics in terms of RMSE-% and bias, but the indirect method produced better best-fit lines. Additionally, the mean growth values for height, diameter, and stem volume intervals were compared, and they are presumed to be usable even for forest modelling.
