Abstract Background T cell-based immunotherapies are facing great challenges in the recruitment and activation of tumor-specific T cells against solid tumors. Among which, utilizing nanobody (Nb) or nanobodies (Nbs) to construct T cell engager has emerged as a more practical potential for enhancing the anti-tumor effectiveness of T cells. Here, we designed a new Nb-guided multifunctional T cell engager (Nb-MuTE) that not only recruited effector T cells into the tumor tissues, but also efficiently activated T cells anti-tumor immunity when synergies with photothermal effect. Results The Nb-MuTE, which was constructed based on an indocyanine green (ICG)-containing liposome with surface conjugation of CD105 and CD3 Nbs, and showed excellent targetability to both tumor and T cells, following enhancement of activation, proliferation and cytokine secretion of tumor-specific T cells. Notably, the immunological anti-tumor functions of Nb-MuTE-mediated T cells were further enhanced by the ICG-induced photothermal effect in vitro and in vivo. Conclusions Such a new platform Nb-MuTE provides a practical and “all-in-one” strategy to potentiate T cell responses for the treatment of solid tumor in clinic.

Abstract The Chinese government has introduced a carbon neutral policy to cope with the rapid changes in the global climate. It is not clear what impact this policy will have on wildlife. Therefore, this study analyzed the suitable habitat distribution of China’s unique leopard subspecies in northern Shaanxi, and simulated the potential suitable habitat distribution under different carbon emission scenarios at two time points of future carbon peak and carbon neutralization. We found that in the future SSPs 126 scenario, the suitable habitat area and the number of suitable habitat patches of North China leopard will continue to increase. With the increase of carbon emissions, it is expected that the suitable habitat of North China leopard will continue to be fragmented and shifted. When the annual average temperature is lower than 8 °C, the precipitation seasonality is 80–90 mm and the precipitation of the warmest quarter is greater than 260 mm, the probability of occurrence of North China leopard is higher. The increase in carbon emissions will lead to the reduction, migration, and fragmentation of the suitable habitat distribution of the North China leopard. Carbon neutrality policies can protect suitable wild habitats. In the future, the impact of carbon neutrality policies on future wildlife habitat protection should be carried out in depth to effectively promote the construction of wildlife protection projects.

Abstract Doxorubicin’s antitumor effectiveness may be constrained with ineffective tumor penetration, systemic adverse effects, as well as drug resistance. The co-loading of immune checkpoint inhibitors and doxorubicin into liposomes can produce synergistic benefits and address problems, including quick drug clearance, toxicity, and low drug penetration efficiency. In our previous study, we modified a nanobody targeting CTLA-4 onto liposomes (LPS-Nb36) to be an extremely potent CTLA-4 signal blocker which improve the CD8+ T-cell activity against tumors under physiological conditions. In this study, we designed a drug delivery system (LPS-RGD-Nb36-DOX) based on LPS-Nb36 that realized the doxorubicin and anti-CTLA-4 Nb co-loaded and RGD modification, and was applied to antitumor therapy. We tested whether LPS-RGD-Nb36-DOX could targets the tumor by in vivo animal photography, and more importantly, promote cytotoxic T cells proliferation, pro-inflammatory cytokine production, and cytotoxicity. Our findings demonstrated that the combination of activated CD8+ T cells with doxorubicin/anti-CTLA-4 Nb co-loaded liposomes can effectively eradicate tumor cells both in vivo and in vitro. This combination therapy is anticipated to have synergistic antitumor effects. More importantly, it has the potential to reduce the dose of chemotherapeutic drugs and improve safety.

Objectives Despite the use of multimodal analgesia, patients undergoing knee arthroplasty still encounter residual moderate pain. The addition of betamethasone to local anesthetic has been shown to improve postoperative pain. However, it remains uncertain whether the positive effects of perineural or intravenous administration of betamethasone on analgesia outcomes lead to better early mobility and postoperative recovery. Methods Between June 2022 and February 2023, a total of 159 patients who were undergoing knee arthroplasty were included in this study. These patients were allocated randomly into three groups: (i) the NS group, received ropivacaine 0.375% and intravenous 3mL 0.9% normal saline; (ii) the PNB group, received ropivacaine 0.375% plus perineural betamethasone (12mg) 3mL and intravenous 3mL 0.9% normal saline; and (iii) the IVB group, received ropivacaine 0.375% and intravenous betamethasone (12mg) 3mL. Results Both perineural and intravenous administration of betamethasone led to improved median (IQR) numeric rating scale (NRS) scores on the 6‐meter walk test, with a score of 1.0 (1.0–2.0) for both groups, compared with 2.0 (1.0–2.0) for the NS group (p = 0.003). Compared to the NS group, both the PNB and IVB groups showed significant reductions in NRS scores at 24 and 36 h after surgery, along with a significant increase in ROM at 24, 36, and 48 h post‐operation. Additionally, it exhibited lower levels of cytokine IL‐1β and TNF‐α in fluid samples, as well as lower level of HS‐CRP in blood samples in the PNB and IVB groups compared to the NS group. Conclusion The administration of perineural and intravenous betamethasone demonstrated an enhanced analgesic effect following knee arthroplasty. Furthermore, it was associated with reduced levels of IL‐1β, TNF‐α, and HS‐CRP, as well as enhanced knee ROM, which is conducive to early ambulation and postoperative rehabilitation after knee arthroplasty.

Since the onset of the SARS-CoV-2 pandemic in early 2020, there has been a notable rise in sodium hypochlorite disinfectants. Sodium hypochlorite undergoes hydrolysis to generate hypochlorous acid for virus eradication. This chlorine-based disinfectant is widely utilized for public disinfection due to its effectiveness. Although sodium hypochlorite disinfection is convenient, its excessive and indiscriminate use can harm the water environment and pose a risk to human health. Hypochlorous acid, a reactive oxygen species, plays a crucial role in the troposphere, stratospheric chemistry, and oxidizing capacity. Additionally, hypochlorous acid is vital as a reactive oxygen species in biological systems, and its irregular metabolism and level is associated with several illnesses. Thus, it is crucial to identify hypochlorous acid to comprehend its environmental and biological functions precisely. Here, we constructed a new fluorescent probe, utilizing the twisted intramolecular charge transfer mechanism to quickly and accurately detect hypochlorous acid in environmental water and biosystems. The probe showed a notable increase in fluorescence when exposed to hypochlorous acid, demonstrating its excellent selectivity, fast response time (less than 10 seconds), a large Stokes shift (∼ 102 nm), and a low detection limit of 15.5 nM.

This study aimed to assess the efficiency of CNV-seq and WES in detecting genetic cause of congenital heart disease (CHDs) in prenatal diagnoses and to compare CNV detection rate between isolated and non-isolated CHD cases. We conducted a retrospective study of 118 Chinese fetuses diagnosed with CHD by prenatal ultrasound. Participants underwent CNV-seq and, if necessary, WES to detect chromosomal and single nucleotide variations. The overall detection rate for pathogenic or likely pathogenic chromosomal abnormalities was 16.9%, including 7.6% aneuploidies and 9.3% pathogenic/likely pathogenic copy number variations (CNVs), predominantly 22q11.2 deletion syndrome (54.4%). The sensitivity and specificity of CNV-Seq for detecting P/Lp CNVs were 95% and 100%, respectively. CNV-Seq offered a 6.7% improvement in detecting chromosomal abnormalities over karyotyping. WES further identified significant single nucleotide and small indel variations contributing to CHD in genes such as TMEM67, PLD1, ANKRD11, and PNKP, enhancing diagnostic yield by 14.8% in cases negative for CNVs. Non-isolated CHD cases exhibited higher rates of detectable chromosomal abnormalities compared to isolated cases (32.4% vs. 9.9%, p = 0.005), underlining the genetic complexity of these conditions. The combined use of CNV-seq and WES provides a comprehensive approach to prenatal genetic testing for CHDs, unveiling significant genetic cause that could impact clinical management and parental decision-making. This study supports the integration of these advanced genomic technologies in routine prenatal diagnostics to increase detection diagnostic yields of causal genetic variants associated with CHDs.

BackgroundFerroptosis has been proven to contribute to the progression of myocardial ischemia/reperfusion (I/R) injury and can be inhibited or promoted by ATF3. Short-chain fatty acids (SCFAs) have shown benefits in various cardiovascular diseases with anti-inflammatory and antioxidant effects. However, the impact of SCFAs on ferroptosis in ischemic-stimulated cardiomyocytes remains unknown. This study aimed to investigate the effect of SCFAs on cardiomyocyte ferroptosis, the expression of ATF3, and its potential upstream regulators.Methods and resultsThe expression of ATF3, ferroptosis pathway geneset (FPG), and geneset of potential regulators for ATF3 (GPRA, predicted by the PROMO database) was explored in the public human myocardial infarction single-cell RNA-seq (sma) dataset. Cardiomyocyte data was extracted from the dataset and re-clustered to explore the FPG, ATF3, and GPRA expression patterns in cardiomyocyte subclusters. A dose-dependent toxic experiment was run to detect the suitable dose for SCFA treatment. The erastin-induced ferroptosis model and hypoxia-reoxygenation (H/R) model (10 h of hypoxia followed by 6 h of reoxygenation) were adopted to assess the effect of SCFAs via the CCK8 assay. Gene expression was examined via RT-PCR and western blot. Ferroptosis markers, including lipid peroxides and Fe2+, were detected using the liperfluo and ferroOrange probes, respectively. In the sma dataset, upregulated ferroptosis pathway genes were mainly found in the infarction-stimulated cardiac cells (border zone and fibrotic zone), particularly the cardiomyocytes and adipocytes. The ATF3 and some of its potential transcription factors (VDR, EGR3, PAX5, and SP1) can be regulated by SCFA. SCFA can attenuate erastin-induced lipid peroxidation in cardiomyocytes. SCFA treatment can also reverse erastin-induced Fe2+ increase but may strengthen the Fe2+ in the H/R model. We also precisely defined a ferroptosis subcluster of cardiomyocytes (CM09) that highly expressed FPG, ATF3, and GPRA.ConclusionThe ATF3 and the ferroptosis pathway are elevated in cardiomyocytes of injury-related cardiac regions (border zone, ischemic zone, and fibrotic zone). SCFA can attenuate cardiomyocyte ferroptosis and regulate the expression of ATF3. Our study offers novel insights into the potential targets of SCFAs in the cardiovascular system.

Abstract Background A significant reduction in regional cerebral oxygen saturation (rSO2) is commonly observed during one-lung ventilation (OLV), while positive end-expiratory pressure (PEEP) can improve oxygenation. We compared the effects of three different PEEP levels on rSO2, pulmonary oxygenation, and hemodynamics during OLV. Methods Forty-three elderly patients who underwent thoracoscopic lobectomy were randomly assigned to one of six PEEP combinations which used a crossover design of 3 levels of PEEP—0 cmH2O, 5 cmH2O, and 10 cmH2O. The primary endpoint was rSO2 in patients receiving OLV 20 min after adjusting the PEEP. The secondary outcomes included hemodynamic and respiratory variables. Results After exclusion, thirty-six patients (36.11% female; age range: 60–76 year) were assigned to six groups (n = 6 in each group). The rSO2 was highest at OLV(0) than at OLV(10) (difference, 2.889%; [95% CI, 0.573 to 5.204%]; p = 0.008). Arterial oxygen partial pressure (PaO2) was lowest at OLV(0) compared with OLV(5) (difference, -62.639 mmHg; [95% CI, -106.170 to -19.108 mmHg]; p = 0.005) or OLV(10) (difference, -73.389 mmHg; [95% CI, -117.852 to -28.925 mmHg]; p = 0.001), while peak airway pressure (Ppeak) was lower at OLV(0) (difference, -4.222 mmHg; [95% CI, -5.140 to -3.304 mmHg]; p

Against the backdrop of near-saturation in global marine capture fisheries, current mariculture production now exceeds one-third of capture production and continues to hold significant growth potential. However, the lack of spatially detailed global offshore surface seawater mariculture (OSSM) data hampers scientific understanding and management from a spatial perspective. In this study, utilizing multi-source remote sensing time-series imagery, we achieve the world’s first spatially detailed mapping of both raft and cage mariculture types. The results show that (1) in 2020, the total global OSSM area was 20,079.97 km², comprising 85.04% raft mariculture and 14.96% cage mariculture. 95.33% of the OSSM was concentrated in Asia. (2) The interpreted national OSSM areas align well with the FAO production statistics trends. Obtaining these areas using remote sensing offers greater timeliness compared to production statistics, which is advantageous for dynamic monitoring and production estimation. (3) Through analysis of global pond aquaculture data, it was found that most coastal areas worldwide had a single-type of aquaculture. However, in parts of Asia, regions with both OSSM and pond aquaculture coexisted. The data acquired in this study, characterized by detailed spatial information, provide significant insights for constructing spatial models of mariculture, researching resource-environment effects, and informing management practices.

ABSTRACTMulti-scale landscape functions play a critical role in revealing intricate functional structures within large regions. However, previous studies on landscape functions have predominantly focused on a single macro or micro scale, impeding a holistic multi-scale understanding of the spatial distribution and heterogeneity of landscape functions. To address this gap, this study proposes a framework leveraging the power of big geodata to mine multi-scale landscape functions from parcel to entire urban agglomerations, as well as non-administrative divisions. Our study focuses on the Guangdong-Hong Kong-Macao Greater Bay Area (GBA) in China. Firstly, we integrated multi-source big geodata to derive parcel-scale landscape functions. Subsequently, we employed the Normalized Revealed Comparative Advantage index to derive landscape functions at broader scales, including towns, counties and cities. The effectiveness of our approach is validated through in-field investigations and comparisons with established policy planning positions. The outcomes not only offer distinctive planning insights at various scales but also highlight the versatility of big geodata in extracting landscape functions across scales. This study demonstrates that big geodata is adept at uncovering multi-scale landscape functions irrespective of administrative boundaries, providing valuable insights for fostering multi-scale regional coordinated development.

Pesticide residues and microplastics (MPs) in agricultural soils are two major concerns for soil health and food safety. The degradation of chlorpyrifos (CPF), an organophosphorus pesticide, releases phosphates. This process may be affected by the presence of MPs in the soil. The combination of CPF and MPs presence in the soil may thus produce interaction effects that alter the soil phosphorus (P) balance. This study explores the degradation pathways of CPF (6 mg kg−1, 12 mg kg−1 of CPF addition) in soils with different levels of polylactic acid MPs (PLA-MPs) (0.0 %, 0.1 %, 0.5 %, 1.0 % w/w), and analyzes soil P fractions and phosphatase enzyme activities to investigate soil P bioavailability under different treatments. Results show that the degradation of CPF fits to a first-order decay model, with half-lives (DT50) ranging from 11.0 to 14.8 d depending on PLA-MPs treatment. The concentration of its metabolite 3, 5, 6-trichloropyridine 2-phenol (TCP) reached a peak of 0.93–1.67 mg kg−1 within 7–14 days. Similarly, the degradation of CPF led to a significant transient increase in P bioavailability within 3–7 days (p < 0.05), with a peak range of 22.55–26.01 mg kg−1 for Olsen-P content and a peak range of 4.63–6.76 % for the proportions of available P fractions (H2O-P+NaHCO3-P+NaOH-P), before returning to prior levels (Olsen-P: 11.28–19.52 mg kg−1; available soil P fractions: 4.15–5.61 %). CPF degradation (6 mg kg−1) was significantly inhibited in soil with 1.0 % PLA-MPs addition. The effects of MPs and CPF on soil P fractions occur at different time frames, implying that their modes of action and interactions with soil microbes differ.

The study explores the impact of mine grassland restoration on plant communities and soil properties in alpine grasslands, a subject of significant interest due to the observed relationship between grassland changes, plant communities, and soil properties. While prior research has mainly focused on the consequences of grassland degradation on plant diversity and soil characteristics, the specific effects of varying restoration degrees in alpine mining grasslands at the regional scale remain poorly understood. To address this knowledge gap, we established 15 sampling plots (0.5m×0.5m) across five different restoration degrees within alpine mining grasslands in the Qilian Mountains, China. Our objective was to assess the variations in plant diversity and soil properties along these restoration gradients. We conducted comprehensive analyses, encompassing soil properties [soil water content (SWC), available nitrogen (AN), total phosphorus (TP), nitrate nitrogen (NO3-N), ammonium nitrogen (NH4-N), total nitrogen (TN), available phosphorus (AP), soil organic carbon (SOC), nitrate nitrogen, soil pH, and electrical conductivity (EC)], plant characteristics (height, density, frequency, coverage, and aboveground biomass), and plant diversity indices (Simpson, Shannon-Wiener, Margalef, Dominance, and Evenness indexes). Our findings included the identification and collection of 18 plant species from 11 families and 16 genera across the five restoration degrees: Very Low Restoration Degree (VLRD), Low Restoration Degree (LRD), Moderate Restoration Degree (MRD), High Restoration Degree (HRD), and Natural Grassland (NGL). Notably, species like Carex duriuscula, Cyperus rotundus, and Polygonum viviparum showed signs of recovery. Principal component analysis and Pearson correlation analysis revealed that soil pH, SWC, SOC, NO3-N, and AN were the primary environmental factors influencing plant communities. Specifically, soil pH and EC decreased as restoration levels increased, while SWC, AN, TP, NH4-N, TN, AP, SOC, and NO3-N exhibited a gradual increase with greater restoration efforts. Furthermore, the HRD plant community demonstrated similarities to the NGL, indicating the most effective natural recovery. In conclusion, our study provides valuable insights into the responses of plant community characteristics, plant diversity, and soil properties across varying restoration degrees to environmental factors. It also elucidates the characteristics of plant communities along recovery gradients in alpine grasslands.

Abstract Background T cell engagers (TCEs) have been established as an emerging modality for hematologic malignancies, but solid tumors remain refractory. However, the upregulation of programmed cell death 1 (PD-1) is correlated with T cell dysfunction that confer tumor-mediated immunosuppression. Developing a novel nanobody-based trispecific T cell engager (Nb-TriTE) would be a potential strategy to improve therapeutic efficacy. Methods Given the therapeutic potential of nanobodies (Nbs), we first screened Nb targeting fibroblast activation protein (FAP) and successfully generated a Nb-based bispecific T cell engager (Nb-BiTE) targeting FAP. Then, we developed a Nb-TriTE by fusing an anti-PD-1 Nb to the Nb-BiTE. The biological activity and antitumor efficacy of the Nb-TriTE were evaluated in vitro and in both cell line-derived and patient-derived xenograft mouse models. Results We had for the first time successfully selected a FAP Nb for the generation of novel Nb-BiTE and Nb-TriTE, which showed good binding ability to their targets. Nb-TriTE not only induced robust tumor antigen-specific killing, potent T cell activation and enhanced T cell function in vitro, but also suppressed tumor growth, improved survival and mediated more T cell infiltration than Nb-BiTE in mouse models of different solid tumors without toxicity. Conclusions This novel Nb-TriTE provides a promising and universal platform to overcome tumor-mediated immunosuppression and improve patient outcomes in the future.

To improve the gel properties of squid surimi products, we optimized the addition of modified corn starch acetylated distarch phosphate (ADSP) and the non-muscle proteins egg white protein (EWP) and soy protein isolate (SPI), and we evaluated the effects of the addition of ADSP, EWP and SPI on the texture, intermolecular force, protein secondary structure and microstructure of squid surimi products. Furthermore, we explored the mechanism by which modified starch and non-muscle protein improve the gel quality of squid surimi products. The results showed that the gel properties of squid surimi products with 10% ADSP, 7% EWP and 6% SPI were the best. Compared with the control group, the addition of modified starch or non-muscle protein decrease the relative contents of random coil and α-helix in proteins, and increased the contents of β-sheet and β-turn. Their combined addition was beneficial for the retention of β-sheet and β-turn structures during the gelation process, and promoted the hydrophobic interaction between proteins, resulting in the formation of a denser gel network structure, which in turn improved the gel strength and water holding capacity of squid surimi products. Correlation analysis showed that the gel strength and water holding capacity, hydrophobic interaction, and relative contents of β-turn and immobilized water of squid surimi products were significantly positively correlated with each other (P < 0.01), and significantly negatively correlated with the contents of free water and random coil (P < 0.01). Therefore, the addition of 10% ADSP, 7% EWP and 6% SPI improved the gel quality of squid surimi products by affecting protein secondary structure and water distribution.

Daucosterol, a natural saponin from Eleocharis dulcis peel, exhibited anti-hyperglycemic properties. This study investigated daucosterol's effects on glycemic control, insulin resistance, and gut microbiota in type 2 diabetic mice. The results demonstrated that daucosterol treatment reduced fasting glucose, improved glucose intolerance and pancreatic islet damage. It decreased HOMA-IR and increased ISI, indicating enhanced insulin sensitivity. Daucosterol supplementation enriched gut microbiota diversity, including reducing Firmicutes and Desulfobacterota while increasing Bacteroidia. This involved an increase in Enterococcus, Akkermansia, Alistipes, Clostridium_sensu_stricto_1, and Odoribacter, coupled with decreased Aerococcus, Desulfovibrio, and Blautia, improved gut health. Uncultured_bacterium_g_Clostridium_sensu_stricto_1 and Bifidobacterium_pseudolongum were identified as major biomarkers. PICRUSt analysis revealed daucosterol improved the pathways of glycan biosynthesis and metabolism, lipid metabolism, carbohydrate metabolism, and some organismal systems. Therefore, daucosterol act as a potential anti-hyperglycemic agent, improving insulin resistance and optimizing gut microbiota to alleviate type 2 diabetes.

Rural residents are exposed to both particulate and gaseous pesticides in the indoor-outdoor nexus in their daily routine. However, previous personal exposure assessment mostly focuses on single aspects of the exposure, such as indoor or gaseous exposure, leading to severe cognition bias to evaluate the exposure risks. In this study, residential dust and silicone wristbands (including stationary and personal wearing ones) were used to screen pesticides in different phases and unfold the hidden characteristics of personal exposure via indoor-outdoor nexus in intensive agricultural area. Mento-Carlo Simulation was performed to assess the probabilistic exposure risk by transforming adsorbed pesticides from wristbands into air concentration, which explores a new approach to integrate particulate (dust) and gaseous (silicone wristbands) pesticide exposures in indoor and outdoor environment. Microplastics (MPs) is a major threat to agroecosystems. Their accumulation and impacts should be evaluated to advance our understanding of soil function and health. Uncovering the role of cascade effects in regulating crop growth is crucial to understanding the link between MPs disturbance and environmental functions. Therefore, we aimed to assess how the cascade changes between (non-) biological factors and functional traits of maize regulate the response of maize growth to MPs in different nutrient soil environments. We found that soil dehydration induced by MPs may disrupt the balance of the physiological status of maize, negatively affect photosynthetic performance, and enhance competition among organisms for limited nutrients. However, root-responsive nutrient cues with a high degree of tectonic freedom allowed adaptive phenotypic plasticity to occur, masking the negative effects of MPs. In nutrient-rich soil environments, moderate and high intensity (>0.5 %) MPs disturbances initiated root nutrient foraging activities, and maize tended to decrease its cost of investing in root construction, i.e., increasing specific root length (SRL) to promote its own growth. The growth of maize was mainly characterized by increases in the belowground biomass (BGB, 7.11 to 20.81 g) and aboveground biomass (AGB, 61.11 to 118.26 g). Our study suggests that a cascade effect between environmental factors initiated by MPs and the functional architecture of the maize root system drives maize to regulate its growth by responding to nutrient cues. These findings will help to ensure food security, formulate environmental risk management policies and protect soil health, especially in the context of future agriculture.

In this work, the PEEK/short carbon fiber (CF) composites were prepared, a new thermodynamic coupling (preheating and impact compaction) process of the FDM method is proposed, and the warp deformation mechanism was obtained by finite element simulation analysis. Results show that a new method could improve the forming quality of an FDM sample. The porosity of FDM samples of the PEEK/CF composite gradually decreased from 10.15% to 6.83% with the increase in impact temperature and frequency. However, the interlayer bonding performance was reduced from 16.9 MPa to 8.50 MPa, which was attributed to the influence of the printing layer height change from the printhead to the forming layer. To explain the above phenomenon, a thermodynamic coupling model was established and a relevant mechanism was analyzed to better understand the interlayer mechanical and porosity properties of PEEK/CF composites. The study reported here provides a reference for improving the forming quality of fabricated PEEK/CF composites by FDM.

To understand how the soil microbial community structure responds to vegetation restoration in alpine mining areas, this study specifically examines the grassland ecosystem in the Qianmalong mining area of the Qilian Mountains after five years of artificial restoration. High-throughput sequencing methods were employed to analyze soil bacteria and fungi microbial characteristics in diverse grassland communities. Combined with modifications in vegetation diversity as well as soil physicochemical properties, the impact of vegetation restoration on soil microbiome diversity in this alpine mining area was investigated. The findings indicated that the dominant plants were Cyperus rotundus, Carex spp., and Elymus nutans. As the extent of the grassland’s restoration increased, the number of plant species, importance values, and plant community diversity showed an increasing trend. The plant functional groups were mainly dominated by Cyperaceae, followed by Poaceae. Plant height, density, plant cover, frequency, and aboveground biomass showed an increasing trend, and soil water content (SWC) increased. While soil pH and soil electrical conductivity (EC) exhibited a declining trend, available phosphorus (AP), total phosphorus (TP), total nitrogen (TN), nitrate nitrogen (NO3-N), soil organic carbon (SOC), and soil water content (SWC) showed an increasing trend. The dominant bacterial communities were Actinobacteriota, Proteobacteria, Acidobacteriota, Chloroflexi, Firmicutes, and Gemmatimonadota, while the dominant fungal communities were Ascomycota, Mortierellomycota, Basidiomycota, unclassified_k_Fungi, and Glomeromycota. Significant differences were detected within soil microbial community composition among different degrees of restoration grasslands, with bacteria generally dominating over fungi. SWC, TP, and TN were found to be the main soil physicochemical factors affecting the distribution of soil bacterial communities’ structure; however, SOC, TN, and NO3-N were the primary factors influencing the soil distribution of fungal communities. The results of this study indicate that different degrees of vegetation restoration in alpine mining areas can significantly affect soil bacterial and fungal communities, and the degree of restoration has varying effects on the soil bacteria and fungi community structure in alpine mining areas.

Multiple datasets related to pond and marine aquaculture have been published using diverse remote sensing technologies, yet a comprehensive dataset detailing spatial distribution on both land and sea sides is lacking. Firstly, a meticulous comparison of datasets which we selected related to aquaculture ponds and marine, ensuring consistency in trends. Subsequently, the datasets published by our team were edited and integrated to illustrate aquaculture activities on both sides of China’s coastal zone. Finally, a spatial differentiation of coastal aquaculture in major provinces was analyzed. This analysis also utilizes the types of coastline and statistical data, guiding coordinated resource management efforts. The results unveil a distinctive spatial distribution pattern, concentrating aquaculture in the northern regions—Bohai Sea, Jiangsu, Fujian, and Pearl River coasts in Guangdong. The provinces rich in aquaculture resources, such as Shandong, Guangdong, and Liaoning, exhibit extensive coastlines. However, remote sensing monitoring suggests an underestimation of Liaoning’s marine aquaculture compared to statistical yearbook data. Furthermore, southern provinces like Guangdong and Fujian exhibit significantly higher aquaculture output than Liaoning. Zhejiang leads in fishing output. The paper outlines the future development direction of coastal aquaculture, emphasizing a strategic, integrated land–sea approach for sustainable development.

Abstract The mid-depth ocean circulation is critically linked to actual changes in the long-term global climate system. However, in the past few decades, predictions based on ocean circulation models highlight the lack of data, knowledge, and long-term implications in climate change assessment. Here, using 842,421 observations produced by Argo floats from 2001-2020, and Lagrangian simulations, we show that only 3.8% of the mid-depth oceans, including part of the equatorial Pacific Ocean and the Antarctic Circumpolar Current, can be regarded as accurately modelled, while other regions exhibit significant underestimations in mean current velocity. Knowledge of ocean circulation is generally more complete in the low-latitude oceans but is especially poor in high latitude regions. Accordingly, we propose improvements in forecasting, model representation of stochasticity, and enhancement of observations of ocean currents. The study demonstrates that knowledge and model representations of global circulation are substantially compromised by inaccuracies of significant magnitude and direction, with important implications for modelled predictions of currents, temperature, carbon dioxide sequestration, and sea-level rise trends.

Mariculture can provide a large amount of food without occupying land resources, which provides important assistance for humans to achieve sustainable development. In this paper, China, where the output of mariculture exceeds half of the global output, is taken as the research area. Based on the spatial distribution data of three periods of mariculture along the coast of the Chinese Mainland in 2000, 2010, and 2020 obtained from remote sensing images and from the perspective of the mariculture center, the spatial development of mariculture in three periods in China is explored. Furthermore, by spatially overlaying mariculture data with concurrent water quality data, the relationship between the development and changes in the mariculture center and the seawater quality in different geographical environments was analyzed at the national scale.. We found that (1) Mariculture in China exhibits a noticeable geographical differentiation, with the scale and development speed of mariculture in northern regions surpassing that of the southern regions. (2) Good seawater quality is the prerequisite for the formation of mariculture centers, while water quality deterioration is detrimental to the development of mariculture. (3) The formation of large-scale high-density mariculture centers does not necessarily lead to deterioration of the seawater quality. Furthermore, we found that the water depth is an important factor leading to the spatial distribution pattern of mariculture in China, i.e., dense in the north and sparse in the south. The flow velocity is an important factor determining the water quality changes in mariculture centers.

BackgroundPostoperative delirium (POD) presents as a serious neuropsychiatric syndrome in patients undergoing off-pump coronary artery bypass grafting (OPCABG) surgery. This is correlated with higher mortality, cognitive decline, and increased costs. The Age-adjusted Charlson Comorbidity Index (ACCI) is recognized as an independent predictor for mortality and survival rate. The purpose of our study is to estimate the predictive value of the ACCI on the POD in patients undergoing OPCABG surgery.MethodsThis prospective cohort study enrolled patients undergoing OPCABG surgery between December 2020 and May 2021 in Qilu Hospital. Patients were divided into the low-ACCI group (score, 0–3) and the high-ACCI group (score ≥4) according to their ACCI scores. The Confusion Assessment Method for the Intensive Care Unit (CAM-ICU) and CAM were used to diagnose POD within 7 days after surgery. The general, laboratory, and clinical data of the patients were recorded and collected. The characteristic ROC curve was applied to further assess the predictive value of the ACCI for POD in patients following OPCABG surgery.ResultsA total of 89 patients were enrolled, including 45 patients in the low-ACCI group and 44 patients in the high-ACCI group. The incidence of POD was higher in the high-ACCI group than in the low-ACCI group (45.5% vs. 15.6%, P = 0.003). Multivariate logistic regression analyses showed that the ACCI (OR, 2.433; 95% CI, 1.468–4.032; P = 0.001) was an independent risk factor for POD. The ACCI accurately predicted POD in patients following OPCABG surgery with an AUC of 0.738, and the Hosmer–Lemeshow goodness of fit test yielded X2 = 5.391 (P = 0.145).ConclusionThe high-ACCI group showed a high incidence of POD. The ACCI was an independent factor associated with POD in patients following OPCABG surgery. In addition, the ACCI could accurately predict POD in patients following OPCABG surgery. Aquaculture, a crucial component of global food production, faces sustainability challenges due to increased competition for natural resources, environmental pollution, and subpar management practices. Understanding spatial–temporal changes in aquaculture distribution can offer insights into guiding its scientific and sustainable development. While Southeast Asia constitutes a key player in global aquaculture, there exists a significant gap in long-term remote sensing monitoring of aquaculture changes at a sub-continental scale. To fill this gap, this study introduces a workflow for aquaculture extraction that combines both automated and manual methods, based on water and edge frequency detection. By using time series of Landsat images and Sentinel-2 images on the GEE platform, we extracted spatial distribution data of aquaculture in Southeast Asia for five periods from 1990 to 2020. This data, boasting an accuracy exceeding 89.53% and peaking at 95.49%, laid the foundation for investigating the spatial distribution patterns, temporal-spatial changes, and area center changes of aquaculture across the study periods. The results showed that the period of aquaculture rapid expansion was concentrated in the first two decades, followed by a gradual decline in the expansion rate, and a consistent northwestward movement of the area center. The spatial distribution pattern of aquaculture in Southeast Asia gradually changed from “Indonesia and the other producers each account for 40% and 60%” to “Vietnam, Indonesia and the other producers each account for 40%, 30%, and 30%”.

In recent years, subsynchronous control interaction (SSCI) has frequently taken place in renewable-connected power systems. To counter this issue, utilities have been seeking tools for fast and accurate identification of SSCI events. The main challenges of SSCI monitoring are the time-varying nature and uncertain modes of SSCI events. Accordingly, this paper presents a simple but effective method that takes advantage of intrinsic time-scale decomposition (ITD). The main purpose is to improve the accuracy and robustness of ITD by incorporating the least-squares method. Results show that the proposed method strikes a good balance between dynamic performance and estimation accuracy. More importantly, the method does not require any prior information, and its performance is therefore not affected by the frequency constitution of the SSCI. Comprehensive comparative studies are conducted to demonstrate the usefulness of the method through synthetic signals, electromagnetic temporary program (EMTP) simulations, and field-recorded SSCI data. Finally, real-time simulation tests are conducted to show the feasibility of the method for real-time monitoring.

Abstract Nanobodies (Nbs) are the variable domain of heavy‐chain antibodies derived from the blood of camelids or sharks. Nanobodies are the smallest antibody fragment with intact antigen‐binding ability. Compared to conventional antibodies, nanobodies have unique properties such as small size, excellent stability and solubility, low immunogenicity, ability to recognize hidden epitopes, high tissue penetration, and industrialized production. More excitingly, the camelid‐specific amino acid sequences in the framework are mutated to their human heavy‐chain variable domain equivalent, which is humanized, to a wide range of applications. These superior characteristics make nanobody an ideal alternative to conventional antibody, showing excellent prospects for various applications in structural biology, molecular imaging, disease diagnosis and therapy, agricultural products, and environmental chemicals detection. With the continuous updating of theories and the rapid development of technology, the screening and expression methods of nanobodies are increasingly mature. Consequently, several technologies to identify and express nanobodies have been established, and various use cases have been described. In this review, we summarize recent advances in the discovery and production of novel nanobodies, and their use in detection and diagnosis platforms.

Although biodegradable plastic film is a promising alternative product for reducing polyethylene plastic pollution in agricultural soils, the effects of its residues on plant growth and soil properties remain unclear. In this study, we conducted an experiment to investigate root properties and soil enzyme activities in Poly (butylene adipate-co-terephthalate) microplastics (PBAT-MPs) contaminated soil (0 % (CK), 0.1 %, 0.2 %, 0.5 % and 1 % of dry soil weight) with soybean (Glycine max (Linn.) Merr.) and maize (Zea mays L.). The results show that PBAT-MP accumulation in soil negatively affects root growth, and alter soil enzyme activities, which may then constrain C/N cycling and potential yields. For soybean, the total root length, total root surface area and root biomass decreased by 34 %− 58 %, 34 %− 54 % and 25 %− 40 % at the harvesting stage compared to CK, respectively. The negative effects of PBAT-MPs on maize roots were greater than on soybean roots. Summary: The p53 tumor suppressor exerts antitumor functions through its ability to regulate the transcription of its downstream targets. Long noncoding RNAs (lncRNAs) act as oncogenes or tumor suppressors implicated in tumorigenesis and tumor progression. Here, we identify the lncRNA LINC00324 (long intergenic noncoding RNA 00324) as a direct p53 transcriptional target. Knockdown of LINC00324 expression promotes tumor growth by reducing p53 transcriptional activity, whereas ectopic LINC00324 expression demonstrates a reverse effect. Notably, LINC00324 is present in the endogenous p53 complex in tumor cells and directly binds to the C-terminal domain of p53 in vitro. Mechanistically, LINC00324 enables p53 transactivation by competitively disrupting the p53-SET interaction, resulting in an increase of p300/CBP-mediated H3K18 and H3K27 acetylation on the p53 target promoters. Lower LINC00324 expression is associated with more aggressive disease status and predicts worse overall survival of patients with cancer. Our study identifies a p53/LINC00324 positive feedback loop that suppresses tumor growth by counteracting SET-mediated transcriptional repression.

Due to the weak information about cultural targets in the complex marine environment, an omission problem exists in large-scale mariculture extraction using single-view and single-source images. To overcome this problem, we developed a mariculture extraction method that combines dense time-series Sentinel-2 and Sentinel-1 data. A high-precision Chinese mariculture distribution map for 2020 was produced with an overall accuracy of 94.00% and a kappa coefficient of 0.91. The results show that (1) the total area of mariculture was 1173249.22 ha on the national scale, which was significantly larger than the previous studies (459595.70 and 205920.28 ha, respectively), with Shandong Province (39.09%) having the largest proportion; (2) China’s mariculture presented a spatial distribution characteristic of ‘Denser North and Sparser South’, and mariculture was centralized in the coastal zones of the northern provinces (60.76%) rather than the southern provinces; (3) the official production statistics and remote sensing-derived mariculture area revealed a highly corresponding trend at the provincial level, with an R2 reaching 0.78, which is much higher than the 0.07 and 0.41 values of the comparison data. The results directly provide data reference for mariculture production estimation and site selection or ideas for mariculture extraction in other regions and globally.

Abstract Interstitial fluid (ISF) from brain drains along the basement membranes of capillaries and arteries as Intramural Periarterial Drainage (IPAD); failure of IPAD results in cerebral amyloid angiopathy (CAA). In this study, we test the hypothesis that IPAD fails after subarachnoid haemorrhage (SAH). The rat SAH model was established using endovascular perforation method. Fluorescence dyes with various molecular weights were injected into cisterna magna of rats, and the pattern of IPAD after SAH was detected using immunofluorescence staining, two-photon fluorescent microscope, transmission electron microscope and magnetic resonance imaging tracking techniques. Our results showed that fluorescence dyes entered the brain along a periarterial compartment and were cleared from brain along the basement membranes of the capillaries, with different patterns based on individual molecular weights. After SAH, there was significant impairment in the IPAD system: marked expansion of perivascular spaces, and ISF clearance rate was significantly decreased, associated with the apoptosis of endothelial cells, activation of astrocytes, over-expression of matrix metalloproteinase 9 and loss of collagen type IV. In conclusion, experimental SAH leads to a failure of IPAD, clinically significant for long term complications such as CAA, following SAH.

Abstract Non‐intrusive load monitoring (NILM) is an important technology for deeply mining consumers' internal electricity consumption information, which can improve the level of awareness of the load and significantly improve the demand‐side management capability of the smart grid. For a long time, traditional NILM faces the challenge of huge data, long training time and low accuracy. To address this issue, this paper proposes an adaptative lightweight seq2subseq model. The model adaptively determines the optimum window length of the active power segment to extract the appliance characteristics and thus reduces the redundant data to construct the appliance‐level‐seq2subseq model. The proposed model significantly improves the ability to extract and utilize information. At the same time, transfer learning is used to ban the training of appliance models with similar characteristics, further reducing model complexity and training time. The experimental results based on the REDD dataset verify the method's effectiveness. Compared with the seq2seq and seq2point models, the proposed model achieved better accuracy and F1 score, up to 98.10% and 90.29%, respectively. At the same time, the proposed method is further verified on UK‐DALE, which proves that the proposed method has good portability and robustness.

Abstract Background Recurrence and chemoresistance constitute the leading cause of death in colorectal cancer (CRC). Thus, it is of great significance to clarify the underlying mechanisms and identify predictors for tailoring adjuvant chemotherapy to improve the outcome of CRC. Methods By screening differentially expressed genes (DEGs), constructing random forest classification and ranking the importance of DEGs, we identified membrane associated guanylate kinase, WW and PDZ domain containing 3 (MAGI3) as an important gene in CRC recurrence. Immunohistochemical and western blot assays were employed to further detect MAGI3 expression in CRC tissues and cell lines. Cell counting kit-8, plate colony formation, flow cytometry, sub-cutaneous injection and azoxymethane plus dextran sulfate sodium induced mice CRC assays were employed to explore the effects of MAGI3 on proliferation, growth, cell cycle, apoptosis, xenograft formation and chemotherapy resistance of CRC. The underlying molecular mechanisms were further investigated through gene set enrichment analysis, quantitative real-time PCR, western blot, co-immunoprecipitation, ubiquitination, GST fusion protein pull-down and immunohistochemical staining assays. Results Our results showed that dysregulated low level of MAGI3 was correlated with recurrence and poor prognosis of CRC. MAGI3 was identified as a novel substrate-binding subunit of SKP1-Cullin E3 ligase to recognize c-Myc, and process c-Myc ubiquitination and degradation. Expression of MAGI3 in CRC cells inhibited cell growth, promoted apoptosis and chemosensitivity to fluoropyrimidine-based chemotherapy by suppressing activation of c-Myc in vitro and in vivo. In clinic, the stage II/III CRC patients with MAGI3-high had a significantly good recurrence-free survival (~ 80%, 5-year), and were not necessary for further adjuvant chemotherapy. The patients with MAGI3-medium had a robustly good response rate or recurrence-free survival with fluoropyrimidine-based chemotherapy, and were recommended to undergo fluoropyrimidine-based adjuvant chemotherapy. Conclusions MAGI3 is a novel E3 ubiquitin ligase by degradation of c-Myc to regulate CRC development and may act as a potential predictor of adjuvant chemotherapy for CRC patients. Background: Mining activities are known to exert significant effects on the structure and function of grassland ecosystems. However, the role of mining grasslands restoration in altering the plant community and soil quality remains poorly understood, especially in alpine regions. Here, we investigated species diversity in grasslands with dynamic changes and different restoration levels in the Tianzhu alpine mining area locating in the Qilian Mountains.Methods: The plant community structure and species composition of the grasslands with different restoration levels were analyzed by the sample method. We used five different restoration levels: very low recovered degree (VLRD), low recovered degree (LRD), medium recovered degree (MRD), and high recovered degree (HRD), and selected natural grassland (NGL, CK) as the control.Results: Plant community structure and species composition were significantly higher than those under the VLRD in the Tianzhu alpine mining area (p < 0.05), with HRD > MRD > LRD > VLRD. There were 11 families, 18 genera, and 17 species of plants, mainly in the families of Leguminosae, Asteraceae, Gramineae, Rosaceae, and Salicaceae; among them, Salicaceae and Gramineae played a decisive role in the stability of the community. The ecotype community showed that perennial herbaceous plants were the most dominant, with annual herbaceous plants being the least dominant, and no tree and shrub layers were observed; the dominance index was the highest in VLRD at 0.32, the richness index was the highest in HRD at 2.73, the diversity of HRD was higher at 1.93, soil pH and EC showed a decreasing trend, and SMC, SOC, TN, NO3-N, NH4-N, AN, TP, and AP content showed an increasing trend with the increase of grassland restoration.Conclusion: In summary, with the increase of restored grassland in the Tianzhu alpine mining area, plant diversity gradually increased and plant community structure gradually diversified, which was close to the plant diversity of NGL. The protection of partially VLRD and LRD grasslands in the mining area should be emphasized, and the mine grassland should be used rationally and scientifically restored.

Background: Remimazolam tosilate is a new ultra-short-acting benzodiazepine sedative medicine. In this study, we evaluated the effect of remimazolam tosilate on the incidence of hypoxemia during sedation in elderly patients undergoing gastrointestinal endoscopy.Methods: Patients in the remimazolam group received an initial dose of 0.1 mg/kg and a bolus dose of 2.5 mg of remimazolam tosilate, whereas patients in the propofol group received an initial dose of 1.5 mg/kg and a bolus dose of 0.5 mg/kg of propofol. Patients received ASA standard monitoring (heart-rate, non-invasive blood pressure, and pulse oxygen saturation) during the entire examination process. The primary outcome was the incidence of moderate hypoxemia (defined as 85%≤ SpO2< 90%, >15s) during the gastrointestinal endoscopy. The secondary outcomes included the incidence of mild hypoxemia (defined as SpO2 90%–94%) and severe hypoxemia (defined as SpO2< 85%, >15s), the lowest pulse oxygen saturation, airway maneuvers used to correct hypoxemia, patient’s hemodynamic as well as other adverse events.Results: 107 elderly patients (67.6 ± 5.7 years old) in the remimazolam group and 109 elderly patients (67.5 ± 4.9 years old) in the propofol group were analyzed. The incidence of moderate hypoxemia was 2.8% in the remimazolam group and 17.4% in the propofol group (relative risk [RR] = 0.161; 95% confidence interval [CI], 0.049 to 0.528; p < 0.001). The frequency of mild hypoxemia was less in the remimazolam group, but not statistically significant (9.3% vs. 14.7%; RR = 0.637; 95% CI, 0.303 to 1.339; p = 0.228). There was no significant difference in the incidence of severe hypoxemia between the two groups (4.7% vs. 5.5%; RR = 0.849; 95% CI, 0.267 to 2.698; p = 0.781). The median lowest SpO2 during the examination was 98% (IQR, 96.0%–99.0%) in patients in the remimazolam group, which was significantly higher than in patients in the propofol group (96%, IQR, 92.0%–99.0%, p < 0.001). Patients in the remimazolam group received more drug supplementation during endoscopy than patients in the propofol group (p = 0.014). There was a statistically significant difference in the incidence of hypotension between the two groups (2.8% vs. 12.8%; RR = 0.218; 95% CI, 0.065 to 0.738; p = 0.006). No significant differences were found in the incidence of adverse events such as nausea and vomiting, dizziness, and prolonged sedation.Conclusion: This study explored the safety of remimazolam compared with propofol during gastrointestinal endoscopy in elderly patients. Despite the increased supplemental doses during sedation, remimazolam improved risk of moderate hypoxemia (i.e., 85%≤ SpO2 < 90%) and hypotension in elderly patients.

In response to the significant ecological damage caused by unsustainable mining practices in the Qilian Mountains, ecological restoration projects have been undertaken in recent years. Analyzing the changes in soil microbial communities during the restoration process of mine meadows helps to reveal the mechanism of the restoration process in alpine mining areas. To explore the characteristics of soil microbial community distribution and their relationships with soil environmental factors during the restoration of alpine grasslands in the Qilian Mountains, we conducted surveys and analyses in two restoration levels low restoration (LR) and high restoration (HR) in the eastern Qilian Mountains, along with an undisturbed natural grassland control (NG). We found that as the degree of high-altitude mining area recovery increases, there were significant increases in vegetation cover, vegetation height, above-ground biomass, vegetation Shannon–Wiener index, soil organic carbon (SOC), soil water content (SWC), total nitrogen (TN), total phosphorus (TP), total potassium (TK), available phosphorus (AP), and available nitrogen (AN). Conversely, soil pH and electrical conductivity (EC) significantly decreased, with soil pH decreasing from 6.93 to 4.13. Restoration of high-altitude mining area grasslands significantly alters the distribution and composition of soil bacteria and fungi, while the impact on soil microbial community changes was not significant. Notably, with increasing recovery level, the dominant bacterial phyla are Acidobacteria and Proteobacteria, while the dominant fungal phyla are Ascomycota and Basidiomycota. These results indicate that changes in vegetation and soil properties both affect the composition of soil microbial communities, with soil properties having a greater influence. Soil fertility and nutrient levels emerge as the primary drivers influencing soil microbial composition communities and the degree of high-altitude mining area grassland recovery.

Obtaining spatial distribution information on mariculture in a low-cost, fast, and efficient manner is crucial for the sustainable development and regulatory planning of coastal zones and mariculture industries. This study, based on the Segment Anything Model (SAM) and high-resolution remote sensing imagery, rapidly extracted mariculture areas in Liaoning Province, a typical northern province in China with significant mariculture activity. Additionally, it explored the actual marine ownership data to investigate the marine use status of Liaoning Province’s mariculture. The total area of mariculture we extracted in Liaoning Province is 1052.89 km2. Among this, the area of cage mariculture is 27.1 km2, while raft mariculture covers 1025.79 km2. Through field investigations, it was determined that in the western part of Liaodong Bay, cage mariculture predominantly involves sea cucumbers. In the southern end of Dalian, the raft mariculture focuses on cultivating kelp. On the other hand, around the islands in the eastern region, the primary crop in raft mariculture is scallops, showing a significant geographical differentiation pattern. In the planned mariculture areas within Liaoning Province’s waters, the proportion of actual development and utilization is 11.2%, while the proportion approved for actual mariculture is 90.2%. This indicates a suspicion that 9.8% of mariculture is possibly in violation of sea occupation rights, which could be due to the untimely updating of marine ownership data. Based on SAM, efficient and accurate extraction of cage mariculture can be achieved. However, the extraction performance for raft mariculture is challenging and remains unsatisfactory. Manual interpretation is still required for satisfactory results in this context.

Most current research on aquaculture ponds focuses on coastal areas, leaving a gap in understanding of inland regions, such as the strategically significant Yangtze River Economic Belt in China. This study introduces an intelligent extraction method for extensive monitoring of aquaculture ponds in Yangtze River Economic Belt, using Landsat and Sentinel data from 1985 to 2020 with five-year intervals based on the Google Earth Engine (GEE) platform. Land cover change data were also analyzed to understand the impact of aquaculture-related changes. Results indicate a significant increase in aquaculture ponds in the Yangtze River Economic Belt from 3235.51 km2 to 14,207.08 km2 between 1985 and 2020. Aquaculture activity primarily shifted eastward from 1985 to 2015, then westward from 2015 to 2020. Approximately 2018.36 km2 of aquatic areas underwent conversion, mainly to water bodies or croplands, with fewer transitions to impervious surfaces, grasslands, or forests. This study highlights that inland areas can also experience significant increases in aquaculture ponds, particularly alongside large rivers, and that the environmental impacts of these changes differ from those in coastal areas, warranting specific attention.

China is one of the countries that suffers severe damage from storm surges. Assessing the vulnerability to storm surges holds great significance for promoting sustainable development and minimizing disaster losses in coastal areas. This study first developed a vulnerability index by integrating 15 indicators from three components (exposure, sensitivity, and adaptability) that provide a comprehensive portrayal of the multidimensional structure of vulnerability. Subsequently, the vulnerability of Chinese coastal areas was comprehensively evaluated from the perspective of prefecture-level cities using a weight combination strategy. Furthermore, spatial statistical techniques were utilized to analyze the spatial heterogeneity of vulnerability. The results show that 64% of coastal cities are classified as being in the very high and high vulnerability categories, with Zhanjiang, Lingao, Dalian, Yancheng, and Shanwei exhibiting the highest vulnerability levels. Among the provinces, Guangxi and Hainan Provinces demonstrate the highest vulnerability, with more than 90% of their coastal cities facing high vulnerability. Additionally, the vulnerability of Chinese coastal cities exhibits significant spatial heterogeneity. Specifically, coastal cities located in the Yangtze River Delta and the Pearl River Delta regions are identified as low–low (LL) vulnerability clusters, whereas high–high (HH) vulnerability clusters are observed in coastal cities within the Beibu Gulf region. These results provide valuable insights for the formulation of disaster reduction policies at the provincial level and the focus for action at the local level.

Background Cardiac failure is the primary cause of death in most patients with pulmonary arterial hypertension (PH). As pleiotropic cytokines, human resistin (Hresistin) and its rodent homolog, resistin‐like molecule α, are mechanistically critical to pulmonary vascular remodeling in PH. However, it is still unclear whether activation of these resistin‐like molecules can directly cause PH‐associated cardiac dysfunction and remodeling. Methods and Results In this study, we detected Hresistin protein in right ventricular (RV) tissue of patients with PH and elevated resistin‐like molecule expression in RV tissues of rodents with RV hypertrophy and failure. In a humanized mouse model, cardiac‐specific Hresistin overexpression was sufficient to cause cardiac dysfunction and remodeling. Dilated hearts exhibited reduced force development and decreased intracellular Ca2+ transients. In the RV tissues overexpressing Hresistin, the impaired contractility was associated with the suppression of protein kinase A and AMP‐activated protein kinase. Mechanistically, Hresistin activation triggered the inflammation mediated by signaling of the key damage‐associated molecular pattern molecule high‐mobility group box 1, and subsequently induced pro‐proliferative Ki67 in RV tissues of the transgenic mice. Intriguingly, an anti‐Hresistin human antibody that we generated protected the myocardium from hypertrophy and failure in the rodent PH models. Conclusions Our data indicate that Hresistin is expressed in heart tissues and plays a role in the development of RV dysfunction and maladaptive remodeling through its immunoregulatory activities. Targeting this signaling to modulate cardiac inflammation may offer a promising strategy to treat PH‐associated RV hypertrophy and failure in humans.
