Abstract Resveratrol has been reported to promote immunity and decrease oxidative stress, but which demonstrates biphasic effects relied on the use concentration. In this study, the effects of diet supplement with a relative high concentration of resveratrol (0.32 mg/kg) on metabolism, antioxidation and apoptosis of liver were investigated in Siberian sturgeon. The results showed that resveratrol significantly increased the lipid synthesis and the apoptosis, but did not either activate the antioxidant NRF2/KEAP1 pathway or enhance the antioxidant enzyme activity. Transcriptome analysis revealed significant changes in regulatory pathways related to glycolipid, including PPAR signaling pathway, Insulin signaling pathway, Fatty acid biosynthesis, and Glycolysis/Gluconeogenesis. In addition, resveratrol significantly increased the lipid synthesis genes (accα and fas), fatty acid transport gene (fatp 6) and gluconeogenesis gene (gck), but decreased the survival-promoting genes (gadd45β and igf 1). These findings highlight a significant effect of resveratrol on glycolipid metabolism in Siberian sturgeon. Moreover, this study also demonstrated that 0.32 mg/kg resveratrol has physiological toxicity to the liver of Siberian sturgeon, indicating that this dose is too high for Siberian sturgeon. Thus, our study provides a valuable insight for future research and application of resveratrol in fish.

Abstract Aim This study aims to explore the duration and influencing factors of care‐seeking delay among patients with heart failure (HF) in China. Methods and results A convergent mixed method containing a cross‐sectional study and two parts of qualitative studies was designed, following the STROBE and COREQ guidelines. Convenience sampling was applied to recruit patients with HF from two general hospitals from December 2021 to December 2022. Purposive sampling was used to enrol healthcare professionals from two general hospitals and two community hospitals from June to November 2022. Among the 258 patients with HF in the cross‐sectional study, the median duration of care‐seeking delay was 7.5 days. The result integration indicated that the delay duration was influenced by the dyspnoea symptom burden, the oedema symptom burden, and the depression status. The lower dyspnoea symptom burden, the higher oedema symptom burden, and the higher depression score were related to the prolonged care‐seeking delay duration. The duration was also affected by the COVID‐19 pandemic, level of support from medical system, and the symptom management abilities of the caregivers. The COVID‐19 pandemic, low level of support from medical system, and limited symptom management abilities of caregivers were related to the prolonged care‐seeking delay duration. Conclusions Care‐seeking delay among patients with HF needs attention in China. The duration of care‐seeking delay of patients with HF was influenced by the dyspnoea symptom burden, the oedema symptom burden, and depression status, as well as the COVID‐19 pandemic, level of support from medical system, and the symptom management abilities of the caregivers.

Abstract Genes associated with endoplasmic reticulum stress (ERS) and mitophagy can be conducive to predicting solid tumour prognosis. The authors aimed to develop a prognosis prediction model for these genes in lung adenocarcinoma (LUAD). Relevant gene expression and clinical information were collected from public databases including Gene Expression Omnibus (GEO) and The Cancer Genome Atlas (TCGA). A total of 265 differentially expressed genes was finally selected (71 up‐regulated and 194 downregulated) in the LUAD dataset. Among these, 15 candidate ERS and mitophagy genes (ATG12, CSNK2A1, MAP1LC3A, MAP1LC3B, MFN2, PGAM5, PINK1, RPS27A, SQSTM1, SRC, UBA52, UBB, UBC, ULK1, and VDAC1) might be critical to LUAD based on the expression analysis after crossing with the ERS and mitochondrial autophagy genes. The prediction model demonstrated the ability to effectively predict the 5‐, 3‐, and 1‐year prognoses of LUAD patients in both GEO and TCGA databases. Abstract Background In aquaculture, sturgeons are generally maintained in the confined spaces, which not only hinders sturgeon movement, but also threatens their flesh quality that seriously concerned by aquaculture industry. As a typical antioxidant, resveratrol can improve the flesh quality of livestock and poultry. However, the mechanism of resveratrol’s effect on the muscle of Siberian sturgeon is still unclear. Results In this study, the dietary resveratrol increased the myofiber diameter, the content of the amino acids, antioxidant capacity markers (CAT, LDH and SOD) levels and the expression levels of mTORC1 and MYH9 in muscle of Siberian sturgeon. Further transcriptome analysis displayed that ROS production-related pathways (“Oxidative phosphorylation” and “Chemical carcinogenes-reactive oxygen species”) were enriched in KEGG analysis, and the expression levels of genes related to the production of ROS (COX4, COX6A, ATPeF1A, etc.) in mitochondria were significantly down-regulated, while the expression levels of genes related to scavenging ROS (SOD1) were up-regulated. Conclusions In summary, this study reveals that resveratrol may promote the flesh quality of Siberian sturgeon probably by enhancing myofiber growth, nutritional value and the antioxidant capacity of muscle, which has certain reference significance for the development of a new type of feed for Siberian sturgeon.

AbstractBackground The burden of carbapenem-resistant gram-negative bacteria (CRGNB) among solid organ transplant (SOT) recipients has not been systematically explored. Here, we discern the risk factors associated with CRGNB infection and colonization in SOT recipients.Methods This study included observational studies conducted among CRGNB-infected SOT patients, which reported risk factors associated with mortality, infection or colonization. Relevant records will be searched in PubMed, Embase and Web of Science for the period from the time of database construction to 1 March 2023.Results A total of 23 studies with 13,511 participants were included, enabling the assessment of 27 potential risk factors. The pooled prevalence of 1-year mortality among SOT recipients with CRGNB was 44.5%. Prolonged mechanical ventilation, combined transplantation, reoperation and pre-transplantation CRGNB colonization are salient contributors to the occurrence of CRGNB infections in SOT recipients. Renal replacement therapy, post-LT CRGNB colonization, pre-LT liver disease and model for end-stage liver disease score increased the risk of infection. Re-transplantation, carbapenem use before transplantation and ureteral stent utilization increaesd risk of CRGNB colonization.Conclusion Our study demonstrated that SOT recipients with CRGNB infections had a higher mortality risk. Invasive procedure may be the main factor contribute to CRGNB infection.

Abstract Background Astragaloside IV (AS‐IV) is the most active monomer in the traditional Chinese herbal medicine Radix Astragali, which has a wide range of antiviral, anti‐inflammatory, and antifibrosis pharmacological effects, and shows protective effects in acute lung injury. Methods This study utilized the immunofluorescence, flow cytometry, enzyme‐linked immunosorbent assay, quantitative reverse transcription‐polymerase chain reaction, western blot, and hematoxylin and eosin staining methods to investigate the mechanism of AS‐IV in reducing viral pneumonia caused by influenza A virus in A549 cells and BALB/c mice. Results The results showed that AS‐IV suppressed reactive oxygen species production in influenza virus‐infected A549 cells in a dose‐dependent manner, and subsequently inhibited the activation of nucleotide‐binding oligomerization domain‐like receptor thermal protein domain associated protein 3 inflammasome and Caspase‐1, decreased interleukin (IL) ‐1β and IL‐18 secretion. In BALB/c mice infected with Poly (I:C), oral administration of AS‐IV can significantly reduce Poly (I:C)‐induced acute pneumonia and lung pathological injury. Conclusions AS‐IV alleviates the inflammatory response induced by influenza virus in vitro and lung flammation and structural damage caused by poly (I:C) in vivo.

Diabetes mellitus (DM)-induced endothelial dysfunction (DED) contributes to macrovascular complications of DM, resulting in high mortality. Fish oil (FO) is a dietary source of omega-3 polyunsaturated fatty acids, with little known for its action on DED. Thus, the objective of the current study was to investigate the effect of FO on aortic DED using a mouse model of DM. To this end, C57BL/6 J male mice (n = 8/each group) were intraperitoneally injected with streptozotocin (50 mg/kg/day, 5 days), following feeding with a high-fat diet containing FO for 24 weeks, with fasting blood glucose levels and glucose tolerance monitored. At the end of the procedure, aortic rings were harvested and evaluated for the contraction and relaxation in response to phenylephrine and acetylcholine respectively. Aortic pathological injuries were assessed by hematoxylin and eosin, and Masson's trichrome staining. Aortic expression of proinflammatory, oxidative stress and profibrotic markers were determined by qRT-PCR and immunohistochemical staining. The diabetic mice developed impaired glucose tolerance, abnormal endothelial dependent aortic contraction and relaxation, thickened tunica media, and increased accumulation of fibrosis in the aortas. These effects were remarkably reversed by FO supplementation. Further investigation revealed that FO inhibited aortic mRNA expression of Tnf-α, Il-6, Il-1β and Vcam-1 as markers for inflammation, Nox1, Nox2, Nox4 and Nos2 as markers for oxidative stress, as well as Acta 2, Col1a1, Col3 and Ccn2 as markers for fibrosis. Moreover, FO decreased the aortic protein expression of TNF-α, 4-HNE and COL1, all of which were increased under the diabetic condition. The present study may provide a molecular basis for the application of FO in prevention of macrovascular complications of DM.

Abstract Background Mutations in fibrillin-1 (FBN1) are known to be associated with Marfan syndrome (MFS), an autosomal dominant connective tissue disorder. Most FBN1 mutations are missense or nonsense mutations. Traditional molecular genetic testing for the FBN1 gene, like Sanger sequencing, may miss disease-causing mutations in the gene’s regulatory regions or non-coding sequences, as well as partial or complete gene deletions and duplications. Methods Next-generation sequencing, multiplex ligation-dependent probe amplification and gap PCR were conducted on two MFS patients to screen for disease-causing mutations. Results We identified two large deletions in FBN1 from two MFS patients. One patient had a 0.23 Mb deletion (NC_000015.9:g.48550506_48779360del) including 5’UTR-exon6 of FBN1. The other patient harbored a 1416 bp deletion (NC_000015.9:g.48410869_48412284del) affecting the last exon, exon 66, of the FBN1 gene. Conclusion Our results expanded the number of large FBN1 deletions and highlighted the importance of screening for large deletions in FBN1 in clinical genetic testing, especially for those with the classic MFS phenotype.

To enhance the precision of rolling bearing fault detection and lessen the likelihood of safety mishaps, this paper proposes a fault detection method grounded in improved variational mode decomposition. This technique initially employs the Northern Eagle Algorithm to determine the optimal parameter value for variational mode decomposition, subsequently decomposing the signal. This is followed by the utilization of the Spearman correlation coefficient to differentiate between the effective component and the noise-dominant component. Finally, the wavelet packet decomposition is adopted to filter noise and yield the Hilbert envelope spectrum of the de-noised signal to ascertain the bearing’s health status based on the extraction of characteristic fault frequencies and harmonics. The experimental findings illustrate that the enhanced variational mode decomposition technique not only escalates the inner ring signal’s signal-to-noise ratio from −10.844 dB to 8.4471 dB and the outer ring signal’s ratio from −4.5852 dB to 3.0997 dB but also reduces the error of outer ring fault detection from 3.14% to 0.37%, and improves the frequency of inner ring fault detection from a feature extraction inability to an error frequency of 0.45%.

The gastrointestinal tract is an important part of the human immune system. The gut microbiome, which constitutes a major component of the gastrointestinal tract, plays a crucial role in maintaining normal physiological functions and influences the development, diagnosis, and immunotherapy of colorectal cancer (CRC). Natural polysaccharides can be extracted from animals, plants, and traditional Chinese medicines. They serve as an essential energy source for the gut microbiome, promoting probiotic proliferation and regulating the intestinal microecological balance. Moreover, polysaccharides exhibit anti-tumor effects due to their immune regulatory functions and low toxicity. This review focuses on discussing these anti-tumor effects in CRC, along with improving gut microbiome dysbiosis and regulating the tumor immune microenvironment, providing evidence for effective therapeutic strategies against CRC.

TonB-dependent siderophore receptors play a critical transport role for Flavobacterium columnare virulence formation and growth, and have become valuable targets for the development of novel antimicrobial agents. Traditional Chinese medicine has demonstrated notable efficacy in the treatment of fish diseases and includes potential antibacterial agents. Herein, we performed molecular docking-based virtual screening to discover novel TonB-dependent siderophore receptor inhibitors from traditional Chinese medicine and provide information for developing novel antibacterial agents. Firstly, we efficiently obtained 11 potential inhibitors with desirable drug-like characteristics from thousands of compounds in the TCM library based on virtual screening and property prediction. The antibacterial activity of Enoxolone, along with its interaction characteristics, were determined via an MIC assay and molecular dynamic simulation. Transcriptional profiling, along with validation experiments, subsequently revealed that an insufficient uptake of iron ions by bacteria upon binding to the TonB-dependent siderophore receptors is the antibacterial mechanism of Enoxolone. Finally, Enoxolone's acceptable toxicity was illustrated through immersion experiments. In summary, we have used virtual screening techniques for the first time in the development of antimicrobial agents in aquaculture. Through this process, we have identified Enoxolone as a promising compound targeting the TonB-dependent siderophore receptor of F. columnare. In addition, our findings will provide new ideas for the advancement of innovative antimicrobial medications in aquaculture.

Abstract Introduction Hypoxia is an important characteristic of gastric mucosal diseases, and hypoxia‐inducible factor‐1α (HIF‐1α) contributes to microenvironment disturbance and metabolic spectrum abnormalities. However, the underlying mechanism of HIF‐1α and its association with mitochondrial dysfunction in gastric mucosal lesions under hypoxia have not been fully clarified. Objectives To evaluate the effects of hypoxia‐induced HIF‐1α on the development of gastric mucosal lesions. Methods Portal hypertensive gastropathy (PHG) and gastric cancer (GC) were selected as representative diseases of benign and malignant gastric lesions, respectively. Gastric tissues from patients diagnosed with the above diseases were collected. Portal hypertension (PHT)‐induced mouse models in METTL3 mutant or NLRP3‐deficient littermates were established, and nude mouse gastric graft tumour models with relevant inhibitors were generated. The mechanisms underlying hypoxic condition, mitochondrial dysfunction and metabolic alterations in gastric mucosal lesions were further analysed. Results HIF‐1α, which can mediate mitochondrial dysfunction via upregulation of METTL3/IGF2BP3‐dependent dynamin‐related protein 1 (Drp1) N6‐methyladenosine modification to increase mitochondrial reactive oxygen species (mtROS) production, was elevated under hypoxic conditions in human and mouse portal hypertensive gastric mucosa and GC tissues. While blocking HIF‐1α with PX‐478, inhibiting Drp1‐dependent mitochondrial fission via mitochondrial division inhibitor 1 (Mdivi‐1) treatment or METTL3 mutation alleviated this process. Furthermore, HIF‐1α influenced energy metabolism by enhancing glycolysis via lactate dehydrogenase A. In addition, HIF‐1α‐induced Drp1‐dependent mitochondrial fission also enhanced glycolysis. Drp1‐dependent mitochondrial fission and enhanced glycolysis were associated with alterations in antioxidant enzyme activity and dysfunction of the mitochondrial electron transport chain, resulting in massive mtROS production, which was needed for activation of NLRP3 inflammasome to aggravate the development of the PHG and GC. Conclusions Under hypoxic conditions, HIF‐1α enhances mitochondrial dysfunction via Drp1‐dependent mitochondrial fission and influences the metabolic profile by altering glycolysis to increase mtROS production, which can trigger NLRP3 inflammasome activation and mucosal microenvironment alterations to contribute to the development of benign and malignant gastric mucosal lesions.

Increasing attention has been paid to diabetes-associated cognitive dysfunction (DCD) due to its high prevalence and severe impact on life quality in the diabetic population. However, effective interventions for DCD currently are limited. In this study, we investigated the preventive effect of krill oil (KO) on DCD using a mouse model of type 2 diabetes induced by streptozotocin and high-fat diet. The diabetic mice developed cognitive impairment, as well as neuron loss, oxidative stress and inflammation in the hippocampi, the effects of which were significantly prevented by KO. Cell experiments found that KO did not offer a direct protection against advanced glycation end products-induced neuronal activation of apoptotic signaling pathway and neuron loss. In contrast, KO remarkably inactivated the M1 polarized microglia induced by lipopolysaccharide, the effect of which mitigated apoptosis of co-cultured neurons. In M1 polarized microglia, KO predominantly inhibited inflammation and oxidative stress pathways as revealed by RNA-sequencing. These effects were further confirmed in M1 polarized microglia by the KO-decreased expression of inflammatory and oxidative genes, as well as reactive oxygen species level. The current study reported that KO improved DCD through inactivation of microglia, providing a potential effective approach for the intervention of DCD.

Abstract Background Although the majority of pregnancies with preeclampsia are characterised by elevated blood pressure, preeclampsia is often associated with nephrotic syndrome with similar symptoms such as high proteinuria and bilateral lower limb oedema. In this study, we compared the maternal–foetal outcomes of pregnant women with preeclampsia in a population with nephrotic syndrome and explored the factors that contribute to the corresponding outcomes and disease development. Methods A total of 90 pregnant women were included in this study, of whom 30 had nephrotic syndrome and were diagnosed with preeclampsia during pregnancy, and 60 had nephrotic syndrome alone. Descriptive statistical analyses of baseline data were performed to analyse the effect of combined preeclampsia on maternal and foetal pregnancy outcomes using unadjusted and adjusted logistic regression models. Results In this study, the baseline data of the two study populations demonstrated no differences except for the history of caesarean section and 24-h proteinuria results, which were significantly different (P 0.05). Abstract Hepatitis B is a risk factor for the development of intrahepatic cholangiocarcinoma. The prognosis of HBV-related ICC remains to be further investigated. To investigate the clinical, pathological and imaging features of intrahepatic cholangiocarcinoma of hepatitis B virus-positive and -negative patients. Data from January 31, 2012 to December 31, 2019 of 138 patients were retrospectively analyzed. The patients were divided into hepatitis B virus-positive group (group A[n = 66]) and virus-negative group (group B[n = 72]), and the patients were divided into groups according to pathological differentiation degree and tumor size. The differences in clinical, imaging characteristics and the progression-free survival between groups were analyzed. There were significant differences in gender, age, HBc antibody, CA125 and AFP, tumor distribution site, maximum diameter, plain scan density, inferior hepatic angle, peritumoral bile duct dilatation, vascular encasement invasion, intrahepatic bile duct dilatation and lymphadenopathy between the two groups (P 0.05). The clinical and imaging features of ICC of hepatitis B virus-positive and -negative patients are different, and there is little difference in postoperative disease-free survival time.

Abstract Background Tumor cells can resist chemotherapy-induced pyroptosis through glycolytic reprogramming. Estrogen-related receptor alpha (ERRα) is a central regulator of cellular energy metabolism associated with poor cancer prognosis. Herein, we refine the oncogenic role of ERRα in the pyroptosis pathway and glycolytic metabolism. Methods The interaction between ERRα and HIF-1α was verified using co-immunoprecipitation. The transcriptional binding sites of ERRα and NLRP3 were confirmed using dual-luciferase reporter assay and cleavage under targets and tagmentation (CUT&Tag). Flow cytometry, transmission electron microscopy, scanning electron microscopy, cell mito stress test, and extracellular acidification rate analysis were performed to investigate the effects of ERRα on the pyroptosis pathway and glycolytic metabolism. The results of these experiments were further confirmed in endometrial cancer (EC)-derived organoids and nude mice. In addition, the expression of ERRα-related pyroptosis genes was analyzed using The Cancer Genome Atlas and Gene Expression Omnibus database. Results Triggered by a hypoxic microenvironment, highly expressed ERRα could bind to the promoter of NLRP3 and inhibit caspase-1/GSDMD signaling, which reduced inflammasome activation and increased pyroptosis resistance, thereby resulting in the resistance of cancer cells to cisplatin. Moreover, ERRα activated glycolytic rate-limiting enzyme to bridge glycolytic metabolism and pyroptosis in EC. This phenomenon was further confirmed in EC-derived organoids and nude mice. CUT & Tag sequencing and The Cancer Genome Atlas database analysis showed that ERRα participated in glycolysis and programmed cell death, which resulted in EC progression. Conclusions ERRα inhibits pyroptosis in an NLRP3-dependent manner and induces glycolytic metabolism, resulting in cisplatin resistance in EC cells.

Abstract Background The hepatopancreas of crustaceans serves as a significant organ for both the synthesis and secretion of digestive enzymes, as well as energy storage. In the event of food shortage, the hepatopancreas can provide energy for survival. To investigate the potential regulatory mechanisms of the hepatopancreas in response to starvation in Eriocheir Sinensis, transcriptome analysis, histological study and qRT-PCR were performed. Results The results showed that starvation caused a decrease in the hepatopancreas index of E. sinensis, which had certain effects on the tissue structure, metabolism and angiogenesis in the hepatopancreas. In addition, WGCNA and linear regression analysis showed that the genes significantly related to the hepatopancreas index were mainly enriched in the angiogenesis pathway, in which AKT signaling played an important role. Starvation may inhibit AKT signaling pathway by reducing the expression of TGFBI, HSP27, HHEX, and EsPVF1, thereby hindering angiogenesis, promoting apoptosis, and leading to hepatopancreas atrophy. Conclusion These results indicate that AKT plays an important role in the angiogenesis pathway and apoptosis of the starvation induced hepatopancreas index reduction, which is beneficial to further understand the effect of starvation stress on hepatopancreas of Chinese mitten crab.

We propose an efficient outer space branch-and-bound algorithm for minimizing linear multiplicative problems (LMP). First, by introducing auxiliary variables, LMP is transformed into an equivalent problem (ELMP), where the number of auxiliary variables is equal to the number of linear functions. Subsequently, based on the properties of exponential and logarithmic functions, further equivalent transformation of ELMP is performed. Next, a novel linear relaxation technique is used to obtain the linear relaxation problem, which provides a reliable lower bound for the global optimal value of LMP. Once more, branching operation takes place in the outer space of the linear function while embedding compression technique to remove infeasible regions to the maximum extent possible, which significantly reduces the computational cost. Therefore, an outer space branch-and-bound algorithm is proposed. In addition, we conduct convergence analysis and complexity proof for the algorithm. Finally, the computational performance of the algorithm is demonstrated based on the experimental results obtained by testing a series of problems.

Abstract This study is performed to explore the role of P2X4 in intracerebral hemorrhage (ICH) and the association between P2X4 and the NLRP1/Caspase-1 pathway. The mouse ICH model was established via collagenase injection into the right basal ganglia. P2X4 expression in brain tissues was knocked down via intracerebroventricular injection with adeno-associated virus (AAV) harboring shRNA against shP2X4. The gene expression of P2X4 and protein levels related to NLRP1 inflammasome were detected using qRT-PCR and Western blot analysis, respectively. Muramyl dipeptide (an activator of NLRP1) was used to activate NLRP1 in brain tissues. ICH induced high expression of P2X4 in mouse brain tissues. The knockdown of P2X4 alleviated short- and long-term neurological deficits of ICH mice, as well as inhibited the tissue expression and serum levels of pro-inflammatory cytokines, including TNF-α, interleukin (IL)-6, and IL-1β. Additionally, the expressions of NLRP1, ASC, and pro-Caspase-1 were down-regulated upon P2X4 silencing. Moreover, neurological impairment and the expression and secretion of cytokines after P2X4 silencing were aggravated by the additional administration of MDP. P2X4 knockdown represses neuroinflammation in brain tissues after ICH. Mechanistically, P2X4 inhibition exerts a neuroprotective effect in ICH by blocking the NLRP1/Caspase-1 pathway.

Abstract Aim To examine specific correlates that may affect retention outcomes of neural stem cell therapy trials in families screened for cerebral palsy. Design A prospective correlational study. Methods Primary caregivers completed surveys of psychological resilience, care burden and family caregiver tasks. The overall data and differences between groups were analysed and compared. Results Resilience was negatively correlated with the care ability and closely related to the monthly household income and educational level of the caregivers. Factors affecting the final retention rate included the type of disease, number of combined disorders, monthly household income, primary caregivers' education level and resilience. Conclusion Economic level, literacy and psychological status may affect trial retention. These findings can provide tips for preparing for subsequent screening, identification and intervention in stem cell clinical trials. Implication for the Profession and/or Patient Care The study results may provide nursing care tips to make recruitment more efficient, reduce trial costs, support patient‐centredness and accelerate trial progress. HtpG, a bacterial homolog of the eukaryotic 90 kDa heat-shock protein (Hsp90), represents the simplest member of the heat shock protein family. While the significance of Hsp90 in fungal and cancer drug resistance has been confirmed, the role of HtpG in bacterial antibiotic resistance remains largely unexplored. This research aims to investigate the impact of the htpG gene on antibiotic resistance in Vibrio mimicus. Through the creation of htpG gene deletion and complementation strains, we have uncovered the essential role of htpG in regulating the structural integrity of the bacterial cell envelope. Our transcriptomics analysis demonstrates that the deletion of htpG increases the sensitivity of V. mimicus to antimicrobial peptides, primarily due to upregulated lipopolysaccharide synthesis, reduced glycerophospholipid content, and weakened efflux pumps activity. Conversely, reduced sensitivity to β-lactam antibiotics in the ΔhtpG strain results from decreased peptidoglycan synthesis and dysregulated peptidoglycan recycling and regulation. Further exploration of specific pathway components is essential for a comprehensive understanding of htpG-mediated resistance mechanisms, aiding in the development of antimicrobial agents. To our knowledge, this is the first effort to explore the relationship between htpG and drug resistance in bacteria.

BackgroundThe correlation between sarcopenia and hematological malignancy prognosis is still controversial. Design: A systematic review and meta-analysis. Objectives: To explore sarcopenia’s prevalence and prognostic value in hematologic malignancies.Data sources and methodsWe searched Embase, MEDLINE, and Cochrane Library through Ovid SP using an appropriate search strategy on August 28, 2022, and updated the search results on January 9, 2023. Study quality was assessed using the Newcastle-Ottawa scale. The pooled prevalence of sarcopenia was calculated with a 95% confidence interval (CI). Relationships between sarcopenia and prognostic value were expressed as hazard ratio (HR) and 95% CI. HR means the probability of something undesirable, i.e., death or disease progression.ResultsThe search identified more than 3992 studies, and 21 (3354 patients, median or mean age ranging from 36 to 78 years) were finally included. The risk of bias in the studies was low to medium. All included studies were diagnosed based on low muscle mass (LMM). Muscle mass was assessed mainly through imaging technologies, and different cut-offs were applied to determine LMM. The prevalence of sarcopenia was 44.5%, which could fluctuate by age. Subgroup analysis showed that older people had a higher sarcopenic rate than the non-elderly group. Sarcopenia resulted in an inferior prognosis [overall survival: HR 1.821, 95% CI 1.415-2.343; progression-free survival: HR 1.703, 95% CI 1.128-2.571).ConclusionSarcopenia has a prevalence of over 30% in malignant hematologic patients and is associated with a poorer prognosis. Future studies with a standardized sarcopenia diagnostic criterion were needed to investigate sarcopenia’s prevalence and prognostic effects in hematologic malignancies.

Abstract Ternary hydrides are regarded as an important platform for exploring high-temperature superconductivity at relatively low pressures. Here, we successfully synthesized the hcp-(La,Ce)H9-10 at 113 GPa with the initial La/Ce ratio close to 3:1. The high-temperature superconductivity was strikingly observed at 176 K and 100 GPa with the extrapolated upper critical field H c2(0) reaching 235 T. We also studied the binary La-H system for comparison, which exhibited a T c of 103 K at 78 GPa. The T c and H c2(0) of the La-Ce-H are respectively enhanced by over 80 K and 100 T with respect to the binary La-H and Ce-H components. The experimental results and theoretical calculations indicate that the formation of the solid solution contributes not only to enhanced stability but also to superior superconducting properties. These results show how better superconductors can be engineered in the new hydrides by large addition of alloy-forming elements.

Link prediction is recognized as a crucial means to analyze dynamic social networks, revealing the principles of social relationship evolution. However, the complex topology and temporal evolution characteristics of dynamic social networks pose significant research challenges. This study introduces an innovative fusion framework that incorporates entropy, causality, and a GCN model, focusing specifically on link prediction in dynamic social networks. Firstly, the framework preprocesses the raw data, extracting and recording timestamp information between interactions. It then introduces the concept of “Temporal Information Entropy (TIE)”, integrating it into the Node2Vec algorithm’s random walk to generate initial feature vectors for nodes in the graph. A causality analysis model is subsequently applied for secondary processing of the generated feature vectors. Following this, an equal dataset is constructed by adjusting the ratio of positive and negative samples. Lastly, a dedicated GCN model is used for model training. Through extensive experimentation in multiple real social networks, the framework proposed in this study demonstrated a better performance than other methods in key evaluation indicators such as precision, recall, F1 score, and accuracy. This study provides a fresh perspective for understanding and predicting link dynamics in social networks and has significant practical value.

The Chinese mitten crab (Eriocheir sinensis) is one of the most commercially important crustacean species in China. The aim of this study was to characterize the toxic effects of butachlor (an herbicide of the acetanilide class) on juvenile E. sinensis crabs. The lethal effects and the acute toxicity of butachlor on juvenile E. sinensis specimens were assessed through a semi-static in vitro experiment. We determined the activities of superoxide dismutase (SOD) and catalase (CAT) as well as the levels of glutathione (GSH) and malondialdehyde (MDA) in the gills and the hepatopancreas of the juvenile crabs, at different time points over a 14-day short-term exposure to butachlor. Moreover, we measured the residual levels of butachlor in three different tissues (gills, hepatopancreas, and muscles) of the juvenile crabs over a longer period. Our findings revealed that butachlor is highly toxic for juvenile E. sinensis crabs. In fact, the median lethal concentration (LC50) values of butachlor at 24, 48, 72, and 96 h were found to be 4.22, 1.84, 0.34, and 0.14 mg/L, respectively, while the safe concentration was 0.014 mg/L. The antioxidant defense ability of the juvenile E. sinensis crabs against butachlor was induced after exposure to the herbicide at a concentration of 0.01 mg/L. After 14 days of exposure to butachlor at 0.04 and 0.16 mg/L, both SOD and CAT were found to be significantly inhibited (p < 0.05), the GSH levels were found to be significantly decreased (p < 0.05) and the MDA levels were identified as significantly increased (p < 0.05). Moreover, after 14 days of exposure to butachlor at 0.16 mg/L, the activities of SOD and CAT as well as the content of GSH in the hepatopancreas were found to be significantly decreased (p < 0.05). Our results revealed that a high concentration of butachlor was capable of inducing oxidative stress and damage in juvenile E. sinensis crabs. The maximal residual value of butachlor was obtained in the gills, with a content of 4.56 μg/kg. Butachlor was not detected after 24 days in the aforementioned three tissues of the juvenile crabs, thereby indicating that it was effectively metabolized.

The integration of graph embedding technology and collaborative filtering algorithms has shown promise in enhancing the performance of recommendation systems. However, existing integrated recommendation algorithms often suffer from feature bias and lack effectiveness in personalized user recommendation. For instance, users’ historical interactions with a certain class of items may inaccurately lead to recommendations of all items within that class, resulting in feature bias. Moreover, accommodating changes in user interests over time poses a significant challenge. This study introduces a novel recommendation model, RCKFM, which addresses these shortcomings by leveraging the CoFM model, TransR graph embedding model, backdoor tuning of causal inference, KL divergence, and the factorization machine model. RCKFM focuses on improving graph embedding technology, adjusting feature bias in embedding models, and achieving personalized recommendations. Specifically, it employs the TransR graph embedding model to handle various relationship types effectively, mitigates feature bias using causal inference techniques, and predicts changes in user interests through KL divergence, thereby enhancing the accuracy of personalized recommendations. Experimental evaluations conducted on publicly available datasets, including “MovieLens-1M” and “Douban dataset” from Kaggle, demonstrate the superior performance of the RCKFM model. The results indicate a significant improvement of between 3.17% and 6.81% in key indicators such as precision, recall, normalized discount cumulative gain, and hit rate in the top-10 recommendation tasks. These findings underscore the efficacy and potential impact of the proposed RCKFM model in advancing recommendation systems.

In recent years, the interaction of intracellular organelles such as mitochondria and lysosomal functions has attracted increasing attention. Recent evidence suggests that mitochondrion-lysosomal contact plays a key role in regulating lysosomal biogenesis and maintaining cellular homeostasis. Myocardial ischemia and reperfusion will lead to corresponding changes in the autophagy flux in cardiomyocytes, and lysosomes are a key link in the process of autophagy, and the fusion of lysosomes and autophagosomes is an essential link in the occurrence of autophagy. Therefore, the function and homeostasis of lysosomes also undergo different changes during myocardial ischemia and reperfusion. Lysosomal-related biological factors and membrane proteins also play different roles. This article will review the mechanism of lysosomes in myocardial ischemia-reperfusion injury and the research progress of lysosomal-related proteins.

In this study, 32 numerical models of CHST columns were established in the ABAQUS program to evaluate the effect of mid-span local defects on the impact resistance of circular hollow steel tube (CHST) columns. The simulation studies were conducted from three aspects: notch length, notch angle, and impact energy. The results showed that under a lateral impact load, the mid-span of the CHST column presented global bending failure patterns accompanied by local indentation deformation in the impact region and local buckling deformation at the bottom of the fixed end. Compared with the mid-span indentation displacement of the non-notch model, when the impact velocities were 30 km/h and 60 km/h, the horizontal notch model surpassed the maximum by 29.3% and 36.3%, the oblique notch model surpassed the maximum by 47.8% and 115.6%, and the vertical notch model only increased by 9.7% and 1.1%. The local damage area and impact force time-history curves of the vertical notch model agreed well with those of the non-notch model. Among the three notch angles, the impact plateau values of the vertical notch model and the global bending displacement in the mid-span were least affected by the notch length, notch location, and impact energy. The energy absorption of the CHST column was mainly due to indentation deformation in the mid-span, and the global bending deformation was auxiliary. Compared with the energy absorption ratio (EAR) of the non-notch model, with increased impact energy, the EAR of the vertical notch model increased by 20.2%, 13.5%, and 17.3% on average. The horizontal and oblique notch models decreased by 28.2%, 61.0%, 42.4%, and 29.1%, 62.7%, and 49.3%, respectively. The Rd of all notch models showed an overall upward trend as the impact energy increased, and the Rd of the horizontal notch model increased the most. According to the parametric analysis results, the dynamic flexural capacity prediction formula of the CHST columns section was obtained, considering the influence of notch length, notch angle, and impact energy within the parameter range of this study.

Microplastics (MPs) increase the effective state of heavy metals (HMs) in soil and seriously threaten the yield and quality of peanuts (Arachis Hypogea L.). Kaolinite (KL) has the potential to ameliorate MP- and HM- contaminated soils, but the mechanism of action between them is not well understood. Therefore, 60-day experiments were conducted, where KL (1 %, 2 %) and MPs (0.1 %, 1 %) were individually or jointly mixed into soils with different cadmium (Cd) concentrations (0.5, 2.5, and 5.0 mg·kg−1) to cultivate peanuts in a greenhouse. Finally, soil-bioavailable Cd, peanut dry weight, peanut Cd concentrations, the pH, cation exchange capacity (CEC), dissolved organic carbon (DOC), microbial biomass carbon (MBC) and microbial biomass nitrogen (MBN) were determined. It was shown that MPs negatively affected the peanut dry weight and increased the content of soil-bioavailable Cd and Cd concentration in peanut. In the MP- and Cd-contaminated soils, KL mitigated the negative influence of MPs by increasing the dry weight of peanuts by 8.40 %−40.59 %, decreasing the soil-bioavailable Cd by 23.70–35.74 %, and significantly decreasing peanut Cd concentrations by 9.65–30.86 %. The presence of MPs decreased soil pH (7.69–7.87) and the CEC (20.96–23.95 cmol·L−1) and increased the soil DOC (1.84–2.26 mg·kg−1). KL significantly increased soil pH (7.79–8.03) and the CEC (24.96–28.28 cmol·L−1) and mitigated the adverse influence of MPs on the pH and CEC of Cd-contaminated soils. A regression path analysis (RPA) evidenced that KL decreased Cd accumulation in plants by changing the properties of soil contaminated with MPs and Cd. The research results revealed the mechanism of KL on peanut growth and Cd absorption in MP- and Cd-contaminated soil. The results of this study provide a foundation to improve the quality of MP- and HM-contaminated soils and realize safe peanut production.

ObjectiveDuring the Omicron wave of the COVID-19 pandemic in Shanghai, Fangcang Shelter Hospital (FSH) served as the major way in patient quarantine. Many COVID patients served as volunteers in FSH providing a lot of assistance for the medical workers and other COVID patients. The aim of this study was to explore the experiences of patient volunteers in FSH. It helps health professionals better understand their motivational incentives and barriers in their volunteer work, and improves recruiting and managing volunteers in subsequent public health emergencies.MethodsThis is a qualitative study using semi-structured interviews. Thirteen patient volunteers working in an FSH in Shanghai were included. Thematic analysis was applied to data analysis.ResultsFour themes and nine subthemes were identified. The wishes to give back to society and the responsibility of politics and religion were the main reasons for the patients to serve as volunteers in FSH. The patient volunteers served as the bridge to reduce the communication barriers between other patients and healthcare professionals. They also provided support in supply distribution and psychological counseling. They viewed voluntary work as a usual task and tried to solve the barriers in their work. In turn, the voluntary work brought them benefits in mental and physical health, as well as another chance for growth.ConclusionWorking as volunteers in FSHs not only brought personal benefits to the COVID patients but also fulfilled the needs of the healthcare system during public health emergencies. The mode of mutual help between patients could be taken as an example in other public health emergencies.

BackgroundDepression and anxiety are common comorbidities in cardiovascular clinic outpatients. Timely identification and intervention of these mental and psychological disorders can contribute to correct diagnosis, better prognosis, less medical expenses, and improved quality of life. The convenience of online doctor-patient communication platforms has increasingly attracted patients to online consultations. However, online health care and offline health care are very different. Research on how to identify psychological disorders in patients who engage in an online cardiology consultation is lacking. ObjectiveThis study aimed to explore the feasibility of using a self-rating scale to assess mental illness among patients who consult with a cardiologist online and to compare the differences in anxiety and depression between online and offline patients. MethodsFrom June 2022 to July 2022, we conducted follow-up visits with 10,173 patients on the Haodf platform. We conducted detailed consultations with 286 patients who visited the same cardiologist in the outpatient department. We used the self-rated Generalized Anxiety Disorder (GAD-7) and Patient Health Questionnaire (PHQ-9) scales to assess anxiety and depression, respectively. We analyzed the influencing factors related to the degree of coordination of online patients. We also compared the prevalence of anxiety or depression between online and offline patients and analyzed the factors related to anxiety or depression. ResultsOf the 10,173 online consultation patients, only 186 (1.8%) responded effectively. The response rate of the offline consultation patients was 96.5% (276/286). Eucommia ulmoides Oliver (EuO) is a natural medicine that can improve the composition of intestinal flora in fish, but more experiments and data are needed to support whether it can effectively improve the changes of intestinal flora and intestinal damage caused by high starch. This study examined the changes in intestinal structure as well as intestinal flora before and after the addition of EuO to high-starch diets and analyzed the effects of such changes on immune and digestive functions. The results showed that EuO reduces mortality during Nocardia seriolae attack and can reduce starch-induced intestinal inflammation. Eucommia ulmoides Oliver supplementation was able to alter the changes of intestinal flora in fatty acid degradation, bacterial chemotaxis, porphyrin metabolism and flagella assembly caused by high starch. By analyzing the abundance and correlation of bacterial communities, three bacterial communities that were significantly related to the intervention effect of EuO were screened. Further analysis revealed that EuO supplementation reduced the increase in abundance of Limnochordaceae, Nitrolancea, Lysinibacillus, and Hydrogenispora induced by high starch, which were negatively correlated with levels of the immunoreactive substance LZM in fish. This study reveals the regulatory effects of EuO on the intestinal flora of Micropterus salmoides fed on high starch diets, and provides a theoretical basis for reducing starch damage to fish in production.

Nitrogen from ammonia is one of the most common pollutants toxics to aquatic species in aquatic environment. The intestinal mucosa is one of the key mucosal defenses of aquatic species, and the accumulation of ammonia nitrogen in water environment will cause irreversible damage to intestinal function. In this study, histology, immunohistochemistry, ultrastructural pathology, enzyme activity analysis and qRT-PCR were performed to reveal the toxic effect of ammonia nitrogen stress on the intestine of Pelteobagrus fulvidraco. According to histological findings, ammonia nitrogen stress caused structural damage to the intestine and reduced the number of mucous cells. Enzyme activity analysis revealed that the activity of bactericidal substances (Lysozyme, alkaline phosphatase, and ACP) had decreased. The ultrastructure revealed sparse and shortened microvilli as well as badly degraded tight junctions. Immunohistochemistry for ZO-1 demonstrated an impaired intestinal mucosal barrier. Furthermore, qRT-PCR revealed that tight junction related genes (ZO-1, Occludin, Claudin-1) were downregulated, while the pore-forming protein Claudin-2 was upregulated. Furthermore, as ammonia nitrogen concentration grew, so did the positive signal of Zap-70 (T/NK cell) and the expression of inflammation-related genes (TNF, IL-1β, IL-8, IL-10). In light of the above findings, we conclude that ammonia nitrogen stress damages intestinal mucosal barrier of Pelteobagrus fulvidraco and induces intestinal inflammation.

Lane-level road maps are crucial for urban traffic management, autonomous driving, and vehicle navigations. Optical remote sensing image suffers from trees and buildings occlusion for lane-level road mapping due to the top-down view. While street view images (SVIs) have been used for road detection, however, most of the previous articles focused on extracting road in image space. The reconstruction of lane-level road maps with measurability in geographic space remains challenging. Hence, this article proposed an operational framework for extracting and reconstructing lane-level road maps from urban open access data. First, a sample strategy was used to collect SVIs based on OpenStreetMap (OSM) road central lines. Then, a deep-learning-based method was adopted to identify lanes accurately, and road width was extracted based on design knowledge and OSM information. Finally, the lane-level road map was reconstructed by integrating the lane and its width information. The proposed framework achieves the transformation from image space to geographic space. The case study shows that 82.43% of the roadway is accurately reconstructed in lane-level. The difference between the reconstructed width of the roadway and the reference true value is within the m-level and the RMSE is 0.32 m. The proposed method is cost-effective and accurate-acceptable for acquiring lane-level road datasets in cities.

Abstract Background Maintaining osmotic equilibrium plays an important role in the survival of cold-water fishes. Heat stress has been proven to reduce the activity of Na+/K+-ATPase in the gill tissue, leading to destruction of the osmotic equilibrium. However, the mechanism of megatemperature affecting gill osmoregulation has not been fully elucidated. Results In this study, Siberian sturgeon (Acipenser baerii) was used to analyze histopathological change, plasma ion level, and transcriptome of gill tissue subjected to 20℃, 24℃and 28℃. The results showed that ROS level and damage were increased in gill tissue with the increasing of heat stress temperature. Plasma Cl− level at 28℃ was distinctly lower than that at 20℃ and 24℃, while no significant difference was found in Na+ and K+ ion levels among different groups. Transcriptome analysis displayed that osmoregulation-, DNA-repair- and apoptosis-related terms or pathways were enriched in GO and KEGG analysis. Moreover, 194 osmoregulation-related genes were identified. Amongst, the expression of genes limiting ion outflow, occluding (OCLN), and ion absorption, solute carrier family 4, member 2 (AE2) solute carrier family 9, member 3 (NHE3) chloride channel 2 (CLC-2) were increased, while Na+/K+-ATPase alpha (NKA-a) expression was decreased after heat stress. Conclusions This study reveals for the first time that the effect of heat stress on damage and osmotic regulation in gill tissue of cold-water fishes. Heat stress increases the permeability of fish’s gill tissue, and induces the gill tissue to keep ion balance through active ion absorption and passive ion outflow. Our study will contribute to research of global-warming-caused effects on cold-water fishes.

This study investigated the zooplankton community structure in Ulungur Lake, located in the arid region of Northwest China, which has been slightly polluted by exogeneous sources. A total of 44 zooplankton species were identified in Ulungur Lake, which were sorted into seven functional groups: protozoan filter feeders (PF), rotifer filter feeders (RF), rotifer carnivores (RC), small copepod and cladoceran filter feeders (SCF), mid-sized copepod and cladoceran filter feeders (MCF), mid-sized copepod and cladoceran carnivores (MCC), and large copepod and cladoceran filter feeders (LCF). The dominant zooplankton functional group varied according to seasonal change. The RF group was dominant in the spring, comprising 87.77% of the total biomass. In the summer, the dominant group was SCF, comprising 90.72% of the total biomass, while the RF group accounted for only 7.24%. In the autumn, the biomass was mainly distributed between the RC (36.38%), SCF (30.62%), and RF (30.06%) groups. This seasonal difference in zooplankton community structure is related to fish production in Ulungur Lake, as well as to the competition and predation relationship among zooplankton species. This study showed a significant positive correlation between the PF and LCF groups, and between the RC, MCC, and MCF groups. This positive correlation was found to be related to the competition and predatory relationships between these functional groups. The predation pressure within the zooplankton functional groups in Ulungur Lake was not as significant as the pressure of food competition. The zooplankton biomass showed no significant difference between spring (0.0615 mg/L) and summer (0.0621 mg/L), but the biomass in autumn (0.3051 mg/L) was significantly increased. Nitrite-nitrogen (NO2-N) and Secchi Depth (SD) had significant correlations between species and functional groups, and were the major driving factors affecting the community structure of Ulungur Lake. In this study, the seasonal variations in the biomass of zooplankton were strongly influenced by environmental factors, which played an important driving role in structuring the ecological communities of zooplankton.

Frailty is a common geriatric syndrome characterized by increased vulnerability and is associated with adverse clinical events such as falls, hospitalization, and death. Early diagnosis and early intervention can delay or reverse frailty and ensure the healthy aging of older individuals. At present, there are no gold standard biomarkers for the diagnosis of frailty, which mainly relies on the scale to assess frailty, and the scale has shortcomings such as lagging assessment, subjectivity, and poor consistency. Frailty biomarkers help in early diagnosis and intervention in frailty. The purpose of this review is to summarize the existing inflammatory markers of frailty and focus on novel inflammatory biomarkers of frailty that can be used to help identify frailty early and explore potential intervention targets.

In 2002 and 2020–2022, KH-9 HEXAGON mapping camera system (MCS) and panoramic camera system (PCS) images were made available to the public, respectively. Although great efforts have been made by the scientific community to develop applications that utilize KH-9 HEXAGON images, little attention has been paid to de-noising and contrast enhancement of these images particularly over urban landscapes. This paper focuses on developing a de-noising and contrast enhancement pipeline for KH-9 HEXAGON MCS and PCS over urban regions. The proposed approach employs first a wavelet transform trained using a suite of ‘degree of over-smoothing’ metrics (DOSM) for image de-noising. These metrics are sensitive to structure, texture, edges and local homogeneity of image objects. Then the de-noised image is subjected to the multi-resolution Top-hat to optimize the contrast. This method incorporates a range of shapes and neighborhoods at multiple scales. The method was applied to a KH-9 HEXAGON MCS image (acquired in 1975) and PCS image (acquired in 1974) representing a complex urban landscape, to support comprehensive evaluation under a range of settings. Performance was assessed against three state-of-the-art benchmark approaches: residual learning (deep learning), blind deconvolution and spatial filtering. To evaluate the performance of the proposed pipeline against the benchmarks, we employed the saturation image edge difference standard-deviation, co-occurrence metrics and the semivariogram. Additionally, the potential applications of pre-processed results were demonstrated using change detection, identification reference points and stereo images. The proposed method not only improved the quality of the KH-9 image across the different urban landscape types, but also preserved the original spatial characteristics of the image in comparison with the benchmark methods. At a time when understanding the nature of our changing planet is paramount, the proposed pipeline should be of great benefit to investigators wishing to use KH program images to extend their historical or time-series analyses further back in time.

The use of artificial substrates and biofloc technology can favor fish culture and improve water quality. The aim of this study was to evaluate whether artificial substrates and carbon source additions modify the microbial activity of water bodies. The diversity and structure of microflora in the water after adding artificial substrates and carbon sources to the ponds were analyzed using high-throughput sequencing based on the V3-V4 region of 16S rRNA genes. The results showed that there was no difference in the richness and diversity of intestinal microflora between the control and experimental groups. Principal coordinate analysis (PCoA) and nonmetric multidimensional scaling (NMDS) showed that artificial substrate and carbon source addition changed the structure of the microflora. The results of a linear discriminant analysis (LDA) effect size (LefSe) indicated 11 biomarkers in the EGˍst group. Spearman correlation heatmap analysis showed that environmental factors affected the bacterial communities, and the results of a redundancy analysis indicated that chemical oxygen demand was a critical factor in controlling the bacterial communities in the water. These results provide an understanding of the effect of artificial substrate and carbon source addition on bacterial diversity and community composition in water.

Vibrio mimicus is a serious pathogen in aquatic animals, resulting in significant economic losses. The cAMP receptor protein (CRP) often acts as a central regulator in highly pathogenic pathogens. V. mimicus SCCF01 is a highly pathogenic strain isolated from yellow catfish; the crp gene deletion strain (Δcrp) was constructed by natural transformation to determine whether this deletion affects the virulence phenotypes. Their potential molecular connections were revealed by qRT-PCR analysis. Our results showed that the absence of the crp gene resulted in bacterial and colony morphological changes alongside decreases in bacterial growth, hemolytic activity, biofilm formation, enzymatic activity, motility, and cell adhesion. A cell cytotoxicity assay and animal experiments confirmed that crp contributes to V. mimicus pathogenicity, as the LD50 of the Δcrp strain was 73.1-fold lower compared to the WT strain. Moreover, qRT-PCR analysis revealed the inhibition of type II secretion system genes, flagellum genes, adhesion genes, and metalloproteinase genes in the deletion strain. This resulted in the virulence phenotype differences described above. Together, these data demonstrate that the crp gene plays a core regulatory role in V. mimicus virulence and pathogenicity.

Determining the amount of nitrogen (N) and phosphorus (P) discharged into lakes and assessing the effectiveness of reduction measures in a basin that involve the interaction of multiple factors are still daunting challenges. In this study, the random forest (RF) model was employed to simulate the impact of controlling measures on the amount of N and P discharged in 2025 under seven specific reduction scenarios. Discharged N and P decreased in the basin by 23.38% and 31.69% from 2011 to 2020, respectively. The N and P nutrient discharge intensities were significantly higher in the western part of the basin (13.31 kg·ha−1·a−1 and 1.34 kg·ha−1·a−1) than those in the eastern region (10.24 kg·ha−1·a−1 and 0.74 kg·ha−1·a−1). Fertilizer runoff (N: 34.72%, 5934.49 t; P: 13.60%, 199.76 t), domestic sewage (N: 29.14%, 4009.27 t; P: 34.84%, 496.59 t), and livestock farming (N: 15.11%, 2657.50 t; P: 40.05%, 616.05 t) were the key sources of N and P. The RF model shows that (R2 > 0.994, p < 0.01) the multi-factor reduction effect is the best, and under this discharge reduction effect, the amount of N and P discharged in 2025 are expected to decrease by 13.79% and 19.42%, respectively, compared with those in 2020. In addition, different key sources in sub-basins might lead to regional differences in the discharge reduction effects of various measures. Ultimately, we recommend that the synergistic treatment of point and non-point sources, using treatments with multiple measures, should be implemented in different regions to reduce the amount of N and P discharged in the Chaohu Lake Basin.

Chronic heat stress caused by global warming can have serious implications for fish survival. The kidney plays a central role in many homeostatic functions, including water and electrolyte regulation. However, there is limited knowledge about the effect of heat stress on fish kidneys. In this study, water temperatures were increased from 20 °C to 24 °C and 28 °C in 8 days at a warming rate of 1 °C/d, and then maintained for 12 days. We investigated the effects of mild heat stress (24 °C) and high heat stress (28 °C) on Siberian Sturgeon (Acipenser baerii) kidneys using histological observation, flow cytometry detection, and RT-qPCR. Our histological observations revealed that heat stress caused significant infiltration of inflammatory cells in the kidney, especially at 28 °C. The flow cytometry assay demonstrated a significant increase in the number of apoptotic cells after heat stress at 28 °C compared to a control group at 20 °C (p = 0.033). The level of plasma creatinine was significantly increased in the 28 °C group compared to the control group (p = 0.001). In addition, the mRNA expression levels of heat shock protein GRP75 increased (p = 0.009). The results indicate that heat stress at 28 °C caused damage to the kidneys of A. baerii and triggered the protective response of heat shock proteins. In conclusion, this study contributes to the understanding of the coping strategies of the kidney of A. baerii for chronic heat stress.

Abstract Background The expression and biological functions of circular RNAs (circRNAs) in reproductive organs have been extensively reported. However, it is still unclear whether circRNAs are involved in sex change. To this end, RNA sequencing (RNA-seq) was performed in gonads at 5 sexual stages (ovary, early intersexual stage gonad, middle intersexual stage gonad, late intersexual stage gonad, and testis) of ricefield eel, and the expression profiles and potential functions of circRNAs were studied. Results Seven hundred twenty-one circRNAs were identified, and the expression levels of 10 circRNAs were verified by quantitative real-time PCR (qRT–PCR) and found to be in accordance with the RNA-seq data, suggesting that the RNA-seq data were reliable. Then, the sequence length, category, sequence composition and the relationship between the parent genes of the circRNAs were explored. A total of 147 circRNAs were differentially expressed in the sex change process, and GO and KEGG analyses revealed that some differentially expressed (such as novel_circ_0000659, novel_circ_0004005 and novel_circ_0005865) circRNAs were closely involved in sex change. Furthermore, expression pattern analysis demonstrated that both circSnd1 and foxl2 were downregulated in the process of sex change, which was contrary to mal-miR-135b. Finally, dual-luciferase reporter assay and RNA immunoprecipitation showed that circSnd1 and foxl2 can combine with mal-miR-135b and mal-miR-135c. These data revealed that circSnd1 regulates foxl2 expression in the sex change of ricefield eel by acting as a sponge of mal-miR-135b/c. Conclusion Our results are the first to demonstrate that circRNAs have potential effects on sex change in ricefield eel; and circSnd1 could regulate foxl2 expression in the sex change of ricefield eel by acting as a sponge of mal-miR-135b/c. These data will be useful for enhancing our understanding of sequential hermaphroditism and sex change in ricefield eel or other teleosts.

Abstract Background Genetic factors are important risk factors to develop coronary heart disease (CHD). In this study, we mainly explored whether CYP11B1 mutations influence CHD risk among Chinese Han population. Methods Six variants were genotyped using Agena MassARRAY system from 509 CHD patients and 509 healthy controls. The correlations between CYP11B1 mutations and CHD risk were assessed using odds ratio (OR) and 95% confidence interval (95% CI) by logistic regression. The haplotype analysis and were ultifactor dimensionality reduction (MDR) were conducted. Results In the overall analysis, CYP11B1 polymorphisms were not correlated with CHD susceptibility.
