Fly ash grouting is a common method for mine fire prevention and control. Aiming at the problems of fly ash slurry which is prone to water-loss cracking, poor stability and low fire prevention efficiency, fly ash colloids were prepared by adding high molecular polymer xanthan gum (XG) and hydroxypropyl methyl cellulose (HPMC) to fly ash slurry, and the gel formation time, water-retaining property and thermal stability of fly ash colloids with different water-cement ratios were tested. The effect of colloid on the inhibition of coal spontaneous combustion was studied by programmed temperature. The results showed that when the water-cement ratio of fly ash slurry was 10∶1, the glue formation time was 9.52 min, and the water-retaining property and static stability were good. The fly ash colloid exhibited the shear dilution phenomenon of non-Newtonian fluid, and the energy storage modulus G′ was higher than the loss modulus G″. It was a pseudo-plastic fluid conforming to the power law model, and exhibited the properties of elastic solid. The weight loss process of fly ash colloidal heating can be divided into three stages (30-100 ℃, 218-380 ℃ and 380-800 ℃), and the total mass loss was about 6.17%. Fly ash colloid had the function of high moisture content, oxygen isolation and fissure sealing. The inhibition rate of coal sample after inhibition was 75.86%, and the corresponding temperature point of CO and C2H4 gas generation were 30 ℃ and 20 ℃ later than that of raw coal. The spontaneous combustion process of coal had been effectively inhibited.

IntroductionThe thickening of radish taproots is primarily determined by secondary growth driven by the vascular cambium and is a highly intricate process regulated by plant hormones, transcription factors, and many metabolic pathways. Gibberellin (GA), a plant hormone associated with cell elongation, is essential in secondary growth. However, the mechanism through which exogenous GA3 regulates secondary taproot growth in radishes remains unclear.MethodsIntegrated morphological, anatomical, hormonal, and transcriptomic analyses of taproots in radishes treated with GA3 and its biosynthesis inhibitor paclobutrazol (PBZ) were performed to explore their effects on taproot secondary growth and key regulatory pathways.ResultsGA3 significantly hindered taproot thickening by inhibiting the formation and maintenance of the vascular cambium, and PBZ promoted root development by increasing root length rather than root diameter. Transcriptome analysis revealed 2,014, 948, and 1,831 differentially expressed genes identified from the control vs. GA3, control vs. PBZ, and GA3 vs. PBZ comparisons, respectively. Kyoto Encyclopedia of Genes and Genome pathway enrichment analysis revealed that differentially expressed genes were primarily involved in the biosyntheses of secondary metabolites and metabolic pathways. GA3 significantly increased the levels of endogenous indole-acetic acid and the expression of auxin synthesis and signal transduction genes.DiscussionExogenous GA3 significantly inhibited the expression of genes involved in the maintenance and differentiation of vascular cambium, including WOX14, ER/ERL1, and XCP2. Exogenous GA3 affects root thickening in radishes primarily by regulating hormone signal transduction pathways, vascular cambium activity, and substance and energy metabolisms. Our findings provide insights into the mechanisms underlying taproot thickening in radishes and provide a valuable gene database for future studies.

In recent years, the advancement of photovoltaic power generation technology has led to a surge in the construction of photovoltaic power stations in desert gravel areas. However, traditional equal cross-section photovoltaic bracket pile foundations require improvements to adapt to the unique challenges of these environments. This paper introduces a new type of photovoltaic bracket pile foundation named the “serpentine pile foundation” based on the principle of biomimicry. Utilizing experimental data, numerical simulation technology was employed to comprehensively investigate the pullout resistance, compressive resistance, and horizontal bearing performance of the serpentine pile foundation. Comparative analysis with traditional square and circular pile foundations revealed the serpentine pile foundation’s significant advantages in all performance indexes. The serpentine pile exhibits a significantly higher ultimate uplift bearing capacity of 70.25 kN, which is 8.56 times that of the square pile and 10.94 times that of the circular pile. This study not only offers valuable technical support for the construction of photovoltaic power plants in desert gravel areas but also holds great significance in advancing the sustainable development of the global photovoltaic industry.

Impacts of wheat flour substituted with varying levels of vine tea (Ampelopsis grossedentata) powder (from 0% to 5%) on the sensory quality, texture properties, internal structure and antioxidant properties of bread were investigated. Compared with white bread, the chewiness and springness of bread with 1% berry tea did not change significantly (P>0.05), and its quality was great. However, the quality of berry tea bread decreased significantly as the proportion of vine tea powder increased. Scanning electron microscope (SEM) results indicated that the internal structure of bread was intact and the edge of pores was smooth when the addition amount was 1%, but with the increase of the addition amount, the internal structure of vine tea bread was destroyed and the quality of bread decreased. Objectives: We describe a new method, FlowSpot, to assess CMV-specific T-cell response by quantification of interferon-gamma (IFN-γ). CMV-specific, T-cell-released IFN-γ was captured by flow beads and measured via flow cytometry. In the present study, we used FlowSpot to assess CMV-specific T-cell response in healthy individuals. The FlowSpot results were compared with those of serological analysis and enzyme-linked immunospot (ELISpot) assay. Methods: Experimental results and parameter analysis were investigated by using serological, ELISpot, and FlowSpot assays. Results: The levels of IFN-γ, which is released from CMV-specific T-cells, were measured, and the results and parameter analysis showed a good correlation between FlowSpot and ELISpot. However, FlowSpot was more sensitive and better reflected the strength of IFN-γ secretion than did ELISpot. Conclusions: Compared to ELISpot, FlowSpot has a high sensitivity and is cost and time effective. Thus, this method can be used in wider clinical and scientific applications.

Deep eutectic solvent (DES) and hot-water extraction (HWE) methods were utilized to extract polysaccharides from Polygonatum sibiricum, referred to as DPsP and WPsP, respectively. The extracted polysaccharides were purified using the Superdex-200 dextran gel purification system, resulting in three components for each type of polysaccharide. The structures of these components were characterized. The molecular weight analysis revealed that DPsP components had slightly larger molecular weights compared with WPsP, with DPsP-A showing a slightly higher dispersity index and broader molecular weight distribution. The main monosaccharide components of both DPsP and WPsP were mannose and glucose, while DPsP exhibited a slightly greater variety of sugar components compared with WPsP. FTIR analysis demonstrated characteristic polysaccharide absorption peaks in all six PSP components, with a predominance of acidic pyranose sugars. NMR analysis revealed the presence of pyranose sugars, including rhamnose and sugar aldehyde acids, in both DPsP-B and WPsP-A. DPsP-B primarily exhibited β-type glycosidic linkages, while WPsP-A predominantly displayed α-type glycosidic linkages, with a smaller fraction being β-type. These findings indicated differences in monosaccharide composition and structure between PSPs extracted using different methods. Overall, this study provided experimental evidence for future research on the structure–function relationship of PSPs.

Mapping potential landslides is crucial to mitigating and preventing landslide disasters and understanding mountain landscape evolution. However, the existing methods to map and demonstrate potential landslides in mountainous regions are challenging to use and inefficient. Therefore, herein, we propose a method using hot spot analysis and convolutional neural networks to map potential landslides in mountainous areas at a regional scale based on ground deformation detection using multitemporal interferometry synthetic aperture radar. Ground deformations were detected by processing 76 images acquired from the descending and ascending orbits of the Sentinel-1A satellite. In total, 606 slopes with large ground deformations were automatically detected using hot spot analysis in the study area, and the extraction accuracy rate and the missing rate are 71.02% and 7.89%, respectively. Subsequently, based on the high-deformation areas and potential landslide conditioning factors, we compared the performance of convolutional neural networks with the random forest algorithm and constructed a classification model with the area under the curve (AUC), accuracy, recall, and precision for testing being 0.75, 0.75, 0.82, and 0.75, respectively. Our approach underpins the ability of interferometric synthetic aperture radar (InSAR) to map potential landslides regionally and provide a scientific foundation for landslide risk management. It also enables an accurate and efficient identification of potential landslides within a short period and under extremely hazardous conditions.

Abstract Camellia is a genus of flowering plants in the family Theaceae, and several species in this genus have economic importance. Although a great deal of molecular makers has been developed for molecular assisted breeding in genus Camellia in the past decade, the number of simple sequence repeats (SSRs) publicly available for plants in this genus is insufficient. In this study, a total of 28,854 potential SSRs were identified with a frequency of 4.63 kb. A total of 172 primer pairs were synthesized and preliminarily screened in 10 C. japonica accessions, and of these primer pairs, 111 were found to be polymorphic. Fifty-one polymorphic SSR markers were randomly selected to perform further analysis of the genetic relationships of 89 accessions across the genus Camellia. Cluster analysis revealed major clusters corresponding to those based on taxonomic classification and geographic origin. Furthermore, all the genotypes of C. japonica separated and consistently grouped well in the genetic structure analysis. The results of the present study provide high-quality SSR resources for molecular genetic breeding studies in camellia plants.

Abstract Background Access to kidney transplantation by uremic children is very limited due to the lack of donors in many countries. We sought to explore small pediatric kidney donors as a strategy to provide transplant opportunities for uremic children. Methods A total of 56 cases of single pediatric kidney transplantation and 26 cases of en bloc kidney transplantation from pediatric donors with body weight (BW) less than 10 kg were performed in two transplant centers in China and the transplant outcomes were retrospectively analyzed. Results The 1-year and 2-year death-censored graft survival in the en bloc kidney transplantation (KTx) group was inferior to that in the single KTx group. Subgroup analysis of the single KTx group found that the 1-year and 2-year death-censored graft survival in the group where the donor BW was between 5 and 10 kg was 97.7 and 90.0%, respectively. ObjectivesB cell-activating factor (BAFF), which is critical in the activation and differentiation of B cells, is a candidate diagnostic and predictive biomarker for antibody-mediated rejection (ABMR). We aimed to investigate the value of serum soluble BAFF (sBAFF) for the diagnosis and risk stratification of ABMR after kidney transplantation.MethodsIn the diagnostic study, sBAFF level among ABMR (n = 25), T cell-mediated rejection (TCMR) (n = 14), 4 other pathological lesions (n = 21), and stable allograft function group (n = 15) were compared. In the nested case-control study, kidney allograft recipients with de novo donor-specific antibody (DSA) or ABMR (n = 16) vs. stable allograft function (n = 7) were enrolled, and sBAFF was measured preoperatively, at D7, M1, M3, M6, M9, M12, M18 posttransplant and at allograft biopsy.ResultsThere was no significant difference in sBAFF level at biopsy between ABMR and non-ABMR groups. Longitudinal study showed that the sBAFF levels decreased dramatically at D7 in both groups. The sBAFF level in the DSA group started to increase within M1, while in the stable group, it maintained a low level until M3 and M6. The sBAFF levels of the DSA group were significantly higher than that of the stable group at M1 [1,013.23 (633.97, 1,277.38) pg/ml vs. 462.69 (438.77, 586.48) pg/ml, P = 0.005], M3 [1,472.07 (912.79, 1,922.08) pg/ml vs. 561.63 (489.77, 630.00) pg/ml, P = 0.002], and M6 [1,217.95 (965.25, 1,321.43) pg/ml vs. 726.93 (604.77, 924.60) pg/ml, P = 0.027]. sBAFF levels at M3 had the best predictive value for the DSA/ABMR with the area under the receiver operating characteristic (AUROC) curve value of 0.908. The predictive performance of the maximum (max) change rate from D7 to the peak within M3 was also excellent (AUROC 0.949, P = 0.580).ConclusionWe clarified by a diagnostic study that sBAFF is not a diagnostic biomarker for ABMR in kidney transplantation and revealed by a nested case-control study that sBAFF values at M3 posttransplant and dynamic changes in sBAFF within M3 posttransplant have a good predictive value for the DSA/ABMR. It provides a useful tool for early screening of low-risk patients with negative preoperative DSA for the risk of developing postoperative DSA in kidney allograft recipients.

Background: Donor-derived cell-free DNA (ddcfDNA) has been suggested as an indicator of allograft injury in adult and pediatric kidney transplantation (KTx). However, the dynamics of ddcfDNA in pediatric KTx have not been investigated. In addition, it has not been demonstrated whether donor-recipient (D/R) size mismatch affect ddcfDNA level.Methods: Pediatric KTx recipients with a single donor kidney were enrolled and followed up for 1 year. ddcfDNA, calculated as a fraction (%) in the recipient plasma, was examined longitudinally within 3 months post-transplant. D/R size mismatch degree was described as D/R height ratio. The 33rd percentile of D/R height ratio (0.70) was used as the cut-off to divide the patients into low donor-recipient height ratio group (30% from stable value with a peak value >1% during elevation.Results: Twenty-one clinically stable recipients were enrolled. The median D/R height ratio was 0.83 (0.62–0.88). It took a median of 8 days for ddcfDNA to drop from day 1 and reach a stable value of 0.67% (0.46–0.73%). Nevertheless, 61.5% patients presented ddcfDNA>1% at day 30. Besides, 81.0% (17/21) of patients experienced elevated ddcfDNA and 47.6% (10/21) met the standard of ddcfDNA flare-up. (1) Calculated panel-reactive antibody (CPRA) is a measure of sensitization based on unacceptable antigens (UAs). Determination of UAs based on single-antigen bead assays at allele or antigen levels may be inappropriate. We aimed to introduce eplets for better assessment of sensitization; (2) 900 recipients and 1427 donors were enrolled for candidate or donor pools, respectively. Eplets were from the HLA Epitope Registry. UAs were determined by anti-HLA antibodies identified using LIFECODES Single Antigen (LSA) kits. CPRA values were calculated using a simplified method of donor filtering; (3) HLA antigens containing all eplets of an HLA antigen in LSA kits (LSA antigen) were defined as eplet-predicted (EP) antigens, the reactivity of which could be predicted by that LSA antigen. High reactivity concordance was found between LSA and EP antigens. More HLA antigens were covered by EP antigens in the population than LSA antigens. CPRA values at the EP level were higher than at the allele level and lower than at the antigen level. The EP antigens facilitated UA determination for non-LSA antigens and avoided acute rejection; (4) UA determination using EP antigens can lead to more accurate assessment of sensitization, enabling a high probability of compatible organs and a low risk of adverse outcomes.

(1) Background: The need to elucidate the microbial patterns in preservation fluid and explore their relationship with early infection-related events post kidney transplant and investigate antimicrobial resistance and the effects of preemptive antibiotic therapy. (2) Methods: This retrospective study analyzed the clinical data of 514 kidney transplant donors and 808 recipients from April 2015 to October 2020. Clinical data of donor and recipient characteristics, preservation fluid microbes, early infections (≤30 days), probable donor-derived infections (P-DDIs), antimicrobial resistance and preemptive antibiotic therapy was collected. (3) Results: The incidence of bloodstream (10.3% versus 5.2%, p = 0.006) and graft-site infections (9.7% versus 4.6%, p = 0.004) was significantly higher in recipients with culture-positive preservation fluid. In addition, recipients with ESKAPE pathogens or Candida species had a notably higher rate of bloodstream infections (14.1% versus 6.9%, p = 0.033) and graft-site infections (16.7% versus 3.5%, p < 0.01) than those with other positive pathogens. Preemptive antibiotic therapy decreased the bloodstream infection rate (11.8% versus 35.7%, p = 0.047) when preservation fluid was positive for ESKAPE pathogens. (4) Conclusions: Culture-positive preservation fluid has potential implications for kidney transplant recipients. ESKAPE pathogens or Candida species in preservation fluid as well as their antimicrobial resistance properties and non-preemptive antibiotic therapy could pose a risk of early infection-related events.

The Hunza Valley, in the northwestern Karakoram Mountains, North Pakistan, is a typical region with many towns and villages, and a dense population and is prone to landslides. The present study completed landslide identification, updating a comprehensive landslide inventory and analysis. First, the ground surface deformation was detected in the Hunza Valley by SBAS-InSAR from ascending and descending datasets, respectively. Then, the locations and boundaries were interpreted and delineated, and a comprehensive inventory of 118 landslides, including the 53 most recent InSAR identified active landslides and 65 landslides cited from the literature, was completed. This study firstly named all 118 landslides, considering the demand for globally intensive research and hazard mitigation. Finally, the deformation, spatial–topographic development, and distribution characteristics in the Hunza Valley scale and three large significant landslides were analyzed. Information on 72 reported landslides was used to construct an empirical power law relationship linking landslide area (AL) to volume (VL) (VL = 0.067 × AL1.351), and this formula predicted the volume of 118 landslides in this study. We discovered that the landslides from the literature, which were interpreted from optical images, had lower levels of velocity, area, elevation, and height. The SBAS-InSAR-detected active landslide was characterized by higher velocity, larger area, higher elevation, larger slope gradient, larger NDVI (normalized difference vegetation index), and greater height. The melting glacier water and rainfall infiltration from cracks on the landslide’s upper part may promote the action of a push from gravity on the upper part. Simultaneously, the coupling of actions from river erosion and active tectonics could have an impact on the stability of the slope toe. The up-to-date comprehensive identification and understanding of the characteristics and mechanism of landslide development in this study provide a reference for the next step in landslide disaster prevention and risk assessment.

Epilepsy is one of the most common neurological disorders in pediatric patients with other underlying neurological defects. Identifying the underlying etiology is crucial for better management of the disorder. We performed trio-whole exome sequencing in 221 pediatric patients with epilepsy. Probands were divided into seizures with developmental delay/intellectual disability (DD/ID) and seizures without DD/ID groups. Pathogenic (P) or likely pathogenic (LP) variants were identified in 71/110 (64.5%) patients in the seizures with DD/ID group and 21/111 (18.9%) patients in the seizures without DD/ID group (P < 0.001). Eighty-seven distinct P/LP single nucleotide variants (SNVs)/insertion deletions (Indels) were detected, with 55.2% (48/87) of them being novel. All aneuploidy and P/LP copy number variants (CNVs) larger than 100 Kb were identifiable by both whole-exome sequencing and copy number variation sequencing (CNVseq) in 123 of individuals (41 pedigrees). Ten of P/LP CNVs in nine patients and one aneuploidy variant in one patient (Patient #56, #47, XXY) were identified by CNVseq. Herein, we identified seven genes (NCL, SEPHS2, PA2G4, SLC35G2, MYO1C, GPR158, and POU3F1) with de novo variants but unknown pathogenicity that were not previously associated with epilepsy. Potential effective treatment options were available for 32 patients with a P/LP variant, based on the molecular diagnosis. Genetic testing may help identify the molecular etiology of early onset epilepsy and DD/ID and further aid to choose the appropriate treatment strategy for patients.

ObjectiveTo investigate the efficacy and safety of bone marrow-derived mesenchymal stem cells (BM-MSCs) on chronic active antibody-mediated rejection (cABMR) in the kidney allograft.MethodsKidney recipients with biopsy-proven cABMR were treated with allogeneic third-party BM-MSCs in this open-label, single-arm, single-center, two-dosing-regimen phase I/II clinical trial. In Regimen 1 (n=8), BM-MSCs were administered intravenously at a dose of 1.0×106 cells/kg monthly for four consecutive months, while in Regimen 2 (n=15), the BM-MSCs dose was 1.0×106 cells/kg weekly during four consecutive weeks. The primary endpoints were the absolute change of estimated glomerular filtration rate (eGFR) from baseline (delta eGFR) and the incidence of adverse events associated with BM-MSCs administration 24 months after the treatment. Contemporaneous cABMR patients who did not receive BM-MSCs were retrospectively analyzed as the control group (n =30).ResultsTwenty-three recipients with cABMR received BM-MSCs. The median delta eGFR of the total BM-MSCs treated patients was -4.3 ml/min per 1.73m2 (interquartile range, IQR -11.2 to 1.2) 2 years after BM-MSCs treatment (P=0.0233). The median delta maximum donor-specific antibody (maxDSA) was -4310 (IQR -9187 to 1129) at 2 years (P=0.0040). The median delta eGFR of the control group was -12.7 ml/min per 1.73 m2 (IQR -22.2 to -3.5) 2 years after the diagnosis, which was greater than that of the BM-MSCs treated group (P=0.0342). The incidence of hepatic enzyme elevation, BK polyomaviruses (BKV) infection, cytomegalovirus (CMV) infection was 17.4%, 17.4%, 8.7%, respectively. There was no fever, anaphylaxis, phlebitis or venous thrombosis, cardiovascular complications, or malignancy after BM-MSCs administration. Flow cytometry analysis showed a significant decreasing trend of CD27-IgD- double negative B cells subsets and trend towards the increase of CD3+CD4+PD-1+/lymphocyte population after MSCs therapy. Multiplex analysis found TNF-α, CXCL10, CCL4, CCL11 and RANTES decreased after MSCs treatment.ConclusionKidney allograft recipients with cABMR are tolerable to BM-MSCs. Immunosuppressive drugs combined with intravenous BM-MSCs can delay the deterioration of allograft function, probably by decreasing DSA level and reducing DSA-induced injury. The underlying mechanism may involve immunomodulatory effect of MSCs on peripheral B and T cells subsets.

Enzymatic browning is a major issue that reduces the commercial value of Luffa cylindrica during storage, processing, and transportation. Our results showed that 1% CaCl2 treatment was optimal for reducing the surface browning of fresh-cut luffa. After storage at 25 °C for four days, the treated luffa had a significantly higher total phenolic (TP) content than the untreated luffa. At the end of the storage period, the calcium treatment showed low malondialdehyde (MDA) and hydrogen peroxide (H2O2) accumulation in the luffa. The treated luffa maintained higher superoxide dismutase (SOD), catalase (CAT), and phenylalanine ammonia lyase (PAL) activities and lower polyphenol oxidase (PPO) activity as compared to the untreated luffa. Furthermore, the genes regulating SOD (e.g., SOD1, SOD2, and SOD3), CAT (e.g., LcCAT1 and CAT2), and PAL (e.g., PAL1 and PAL2) in calcium-treated luffa were upregulated to varying degrees, suggesting that Ca2+ inhibited the browning of fresh-cut tissue by regulating the activities of those enzymes. Ultrastructure images showed that the treated luffa could maintain the relative integrity of the cell membrane and organelles. Therefore, Ca2+ might act as a second messenger to reduce ROS oxidative damage and maintain the cell membrane integrity. This study provides new insights into the breeding of new luffa varieties that are resistant to browning and post-harvest treatments to reduce the browning of luffa tissue.

Landslide risk assessment is important for risk management and loss–damage reduction. Herein, we assessed landslide susceptibility, hazard, and risk in the urban area of Yan’an City, which is located on the Loess Plateau of China and affected by many loess landslides. Based on 1841 slope units mapped in the study area, a random forest machine learning classifier and eight environmental factors influencing landslides were used for a landslide susceptibility assessment. In addition, differential synthetic aperture radar interferometry (DInSAR) technology was used for a hazard assessment. The accuracy of the random forest is 0.903 and the area under the receiver operating characteristics (ROC) curve is 0.96. The results show that 16% and 22% of the slope units were classified as being at very high and high-susceptibility levels for landslides, respectively, whereas 16% and 24% of the slope units were at very high and high-hazard levels for landslides, respectively. The landslide risk was obtained based on the susceptibility map and hazard map of landslides. The results show that only 26% of the slope units were located at very high and high-risk levels for landslides and these are mainly concentrated in urban centers. Such risk zones should be taken seriously and their dynamics must be monitored. Our landslide risk map is expected to provide information for planners to help them choose appropriate locations for development schemes and improve integrated geohazard mitigation in Yan’an City.

In order to explore the processing and application potential of Chinese yam starch, nine kinds of Chinese yam starch (GY11, GY5, GY2, GXPY, LCY, SFY, MPY, SYPY, ASY) from South China were collected and characterized. The chemical composition, rheological properties, thermal properties, and in vitro starch digestion were compared, and the correlation between the structure and processing properties of these yam starches was analyzed using Pearson correlation. The results show that GY2 had the highest amylose content of 28.70%. All the yam starches were similarly elliptical, and all the yam starch gels showed pseudoplastic behavior. Yam starches showed similar pasting temperatures and resistant starch content, but SYPY showed the largest particle size (28.4 μm), SFY showed the highest setback (2712.33 cp), and LCY showed the highest peak viscosity (6145.67 cp) and breakdown (2672.33 cp). In addition, these yam starches also showed different crystal types (A-type, B-type, C-type), relative crystallinity (26.54–31.48%), the ratios of 1045/1022 cm−1 (0.836–1.213), pasting properties, and rheological properties, so the yam starches have different application potentials. The rheological and pasting properties were related to the structural properties of starch, such as DI, Mw, and particle size, and were also closely related to the thermodynamic properties. The appropriate processing methods and purposes of the processed products of these yam starches can be selected according to their characteristics.

To clarify the mechanisms underlying the growth-promoting effects of yam polysaccharide on Streptococcus thermophilus (S. thermophilus), the yam polysaccharide was extracted using a deep eutectic solvents (DESs) method and separated into four fractions by DEAE-cellulose 52. These fractions were used as the alternative carbon source to substitute lactose to compare their growth-promoting effects on S. thermophilus. Furthermore, their molecular weight, monosaccharide and functional groups’ composition, microscopic forms and other basic structure characterizations were analyzed. The results showed that all the fractions could significantly promote S. thermophilus growth, and fractions exhibited significantly different growth-promoting effects, whose viable count increased by 6.14, 6.03, 11.48 and 11.29%, respectively, relative to those in the M17 broth medium. Structure-activity relationship analysis revealed that the high growth-promoting activity of yam polysaccharide might be more dependent on the higher molecular weight, the higher galacturonic acid content and its complex spatial configuration, and the existence of β-glycosides would make the yam polysaccharide have a better growth-promoting effect on S. thermophilus.

1,8-diiodooctane (DIO) additive is an important method for optimizing the morphology and device performance of polythieno[3,4-b]-thiophene-co-benzodithiophene (PTB7)-based polymer solar cells. However, the effect of DIO additive on charge photogeneration dynamics of PTB7-based polymer solar cells is still poorly understood. In this work, the effect of DIO additive on the carrier photogeneration dynamics, as well as device performance of PTB7: [6,6]-phenyl-C71-butyric acid methyl ester (PC71BM) solar cells was studied. Bias-dependent photoluminescence (PL) experiments of a neat PTB7 device show that the exciton cannot be dissociated by the electric field in the device within the operating voltage range, but it can be effectively dissociated by the high electric field. PL and time-resolved PL studies show that DIO additive reduces the phase size of PTB7 in the blend film, resulting in an increased exciton dissociation efficiency. The carrier recombination processes were studied by transient absorption, which shows geminate carrier recombination was suppressed in the DIO-treated PTB7:PC71BM device in ultrafast time scale. The increased exciton dissociation efficiency and suppressed carrier recombination in ultrafast time scale play an important role for DIO-treated PTB7:PC71BM solar cells to attain a higher power conversion efficiency.

The China–Pakistan Karakoram Highway is an important land route from China to South Asia and the Middle East via Pakistan. Due to the extremely hazardous geological environment around the highway, landslides, debris flows, collapses, and subsidence are frequent. Among them, debris flows are one of the most serious geological hazards on the Karakoram Highway, and they often cause interruptions to traffic and casualties. Therefore, the development of debris flow susceptibility mapping along the highway can potentially facilitate its safe operation. In this study, we used remote sensing, GIS, and machine learning techniques to map debris flow susceptibility along the Karakoram Highway in areas where observation data are scarce and difficult to obtain by field survey. First, the distribution of 544 catchments which are prone to debris flow were identified through visual interpretation of remote sensing images. The factors influencing debris flow susceptibility were then analyzed, and a total of 17 parameters related to geomorphology, soil materials, and triggering conditions were selected. Model training was based on multiple common machine learning methods, including Ensemble Methods, Gaussian Processes, Generalized Linear models, Navies Bayes, Nearest Neighbors, Support Vector Machines, Trees, Discriminant Analysis, and eXtreme Gradient Boosting. Support Vector Classification (SVC) was chosen as the final model after evaluation; its accuracy (ACC) was 0.91, and the area under the ROC curve (AUC) was 0.96. Among the factors involved in SVC, the Melton Ratio (MR) was the most important, followed by drainage density (DD), Hypsometric Integral (HI), and average slope (AS), indicating that geomorphic conditions play an important role in predicting debris flow susceptibility in the study area. SVC was used to map debris flow susceptibility in the study area, and the results will potentially facilitate the safe operation of the highway.

From a geological standpoint, northern Pakistan is one of the most active and unstable areas in the world. As a consequence, many massive landslides have occurred in the area in historical times that have destroyed infrastructure, blocked the Hunza River, and damaged the Karakoram Highway repeatedly. However, despite the high frequency of large magnitude landslide events, and the consequent damages, the entire area is largely understudied, mainly due to the difficult logistics and the large distances involved. This work is aimed at applying the potential use of Interferometric Synthetic Aperture Radar (InSAR) for landslide identification and investigation for the Hunza-Nagar Region. Sentinel-1 images covering a period of more than two years (February 2017–August 2019) were used and processed by adopting the small baseline subset (SBAS) method. The obtained deformation rate measured along the line of sight (VLOS) varies from −114 to 20 mm/year. The downslope velocity deformation rates (Vslope) range from 0 to −300 mm/year. The Vslope stability threshold for our study area was calculated to be −14 mm/year from the Vslope standard deviation. Four active landslides with Vslope exceeding 14 mm/year were recognizable and have been confirmed by field inspection. The identified landslides listed from the most active to least active are the Humarri, Mayoon, Khai, and Ghulmet landslides, respectively. VLOS exceeding 114 mm/year was observed in the Humarri landslide, which posed a threat of damming a lake on the Hispar River and was also a risk to the Humarri Village located below the landslide. The maximum mean deformation detected in the Ghulmet, and Mayoon landslide was in the order of 30 mm/year and 20 mm/year, respectively. More importantly, it was found that in places, the slope deformation time series showed a patchy correlation with precipitation and seismic events in the area. This may indicate a complex, and possibly uncoupled, relationship between the two controlling agents promoting the deformation. However, the collective impact of the two factors is evident in the form of a continuously descending deformation curve and clearly indicates the ground distortion. The results indicate a potentially critical situation related to the high deformation rates measured at the Humarri landslide. On this specific slope, conditions leading to a possible catastrophic failure cannot be ruled out and should be a priority for the application of mitigation measures.

Geological conditions along the Karakorum Highway (KKH) promote the occurrence of frequent natural disasters, which pose a serious threat to its normal operation. Landslide susceptibility mapping (LSM) provides a basis for analyzing and evaluating the degree of landslide susceptibility of an area. However, there has been limited analysis of actual landslide activity processes in real-time. The SBAS-InSAR (Small Baseline Subsets-Interferometric Synthetic Aperture Radar) method can fully consider the current landslide susceptibility situation and, thus, it can be used to optimize the results of LSM. In this study, we compared the results of LSM using logistic regression and Random Forest models along the KKH. Both approaches produced a classification in terms of very low, low, moderate, high, and very high landslide susceptibility. The evaluation results of the two models revealed a high susceptibility of land sliding in the Gaizi Valley and the Tashkurgan Valley. The Receiver Operating Characteristic (ROC) curve and historical landslide verification points were used to compare the evaluation accuracy of the two models. The Area under Curve (AUC) value of the Random Forest model was 0.981, and 98.79% of the historical landslide points in the verification points fell within the range of high and very high landslide susceptibility degrees. The Random Forest evaluation results were found to be superior to those of the logistic regression and they were combined with the SBAS-InSAR results to conduct a new LSM. The results showed an increase in the landslide susceptibility degree for 2808 cells. We conclude that this optimized landslide susceptibility mapping can provide valuable decision support for disaster prevention and it also provides theoretical guidance for the maintenance and normal operation of KKH.

Background Dual kidney transplantation (DKT) from small pediatric donors, either en-bloc or split dual kidney transplantation, contributes to mitigating organ scarcity. This study investigates the prognosis of DKT from pediatric deceased donors, and influencing factors.Method A retrospective study included recipients who underwent DKT from pediatric donors between 2012 and 2022. Recipients were categorized into low mismatch (BWLM, n = 30) and high mismatch (BWHM, n = 10) groups based on donor-recipient weight ratio of 1:10. Outcome encompassed recipient and graft survival, renal function, and adverse events.Result Forty recipients were included. The average follow-up period was 54.6 months. The 1, 3, and 5-year patient survival were 97.4%, with no significant differences between en-bloc and split dual kidney transplantation or between BWLM and BWHM groups. The graft survival at 1, 3, and 5 years were 89.9%, the graft survival of BWHM group was lower than BWLM group (70% vs 96.7%, p = 0.039). The average eGFR at 1, 3, and 5 years postoperatively were (78.93 ± 25.23), (83.82 ± 32.4), and (85.92 ± 37.08) mL/min/1.73 m2, respectively. The BWHM group also experienced higher rates of graft-related surgical complications (p = 0.006) and urinary tract surgical complications (p = 0.042).Conclusion DKT from pediatric donors yields favorable outcomes, with similar graft survival and complication rates across surgical subgroups. However, significant donor-recipient weight mismatch, particularly when the ratio is less than 1:10, may contribute to increased surgical complications and poorer graft survival. Efforts to minimize extreme weight mismatch are recommended to optimize outcomes.

The phytohormone abscisic acid (ABA) and the lipoxygenases (LOXs) pathway play important roles in seed germination and seedling growth and development. Here, we reported on the functional characterization of Arabidopsis CPR5 in the ABA signaling and LOX pathways. The cpr5 mutant was hypersensitive to ABA in the seed germination, cotyledon greening and root growth, whereas transgenic plants overexpressing CPR5 were insensitive. Genetic analysis demonstrated that CPR5 gene may be located downstream of the ABI1 in the ABA signaling pathway. However, the cpr5 mutant showed an ABA independent drought-resistant phenotype. It was also found that the cpr5 mutant was hypersensitive to NDGA and NDGA treatment aggravated the ABA-induced delay in the seed germination and cotyledon greening. Taken together, these results suggest that the CPR5 plays a regulatory role in the regulation of seed germination and early seedling growth through ABA and LOX pathways independently.

Abstract With the accelerated urbanization and economic development in Northwest China, the efficiency of urban wastewater treatment and the importance of water quality management have become increasingly significant. This work aims to explore urban wastewater treatment and carbon reduction mechanisms in Northwest China to alleviate water resource pressure. By utilizing online monitoring data from pilot systems, it conducts an in-depth analysis of the impacts of different wastewater treatment processes on water quality parameters. This work pays particular attention to their impact on key indicators such as Chemical Oxygen Demand (COD), NH4 +-N, Total Phosphorus (TP), and Total Nitrogen (TN), and the application of predictive models. The work first establishes a Random Forest Regression (RFR) model. The RFR algorithm integrates Bagging ensemble learning and random subspace theory to construct multiple decision trees and aggregate their predictions, thereby enhancing the model’s prediction accuracy and stability. Using bootstrap sampling, the RFR model generates multiple training subsets from the original data and randomly selects subsets of variables to construct regression trees. Its performance in predicting various water quality indicators is then evaluated. The results show that the RFR model exhibits excellent performance, achieving high levels of prediction accuracy and stability for all indicators. For example, the R2 for COD prediction is 0.99954, while the R2 values for NH4 +-N, TP, and TN predictions reach 0.99989. Compared to five other models, the RFR model demonstrates the best performance across all water quality indicator predictions. This work provides critical support for optimizing wastewater treatment technologies and developing water resource management policies. These findings also offer essential theoretical and empirical insights for the future improvement of urban wastewater treatment technologies and water resource management decision-making.

Abstract Friction stir processing (FSP) is successfully employed to alleviate their hook defects of friction stir lap welding (FSLW) of aluminum alloys. The mechanical properties and microstructural characteristics are compared and analyzed between the FSLW&FSP joint fabricated by FSLW and FSP and the FSLW joint. The microstructural analysis shows that the hook defect zone at the advancing side of the FSLW joint is changed into the overlap zone (OZ) of the FSLW&FSP joint due to microstructure reconstruction caused by performing the FSP. The heterogeneous and coarse grains at the hook defect zone of the FSLW joint are transformed into refined and equiaxed grains at the OZ of the FSLW&FSP joint as a result of dynamic recrystallization. The results of tensile tests show that tensile strength and fracture toughness of the FSLW&FSP joint are 85.7% and 220% higher compared to those of the FSLW joint, respectively. The cross sections of broken lap joints reveal that their failure location is shifted from the hook defect at AS of the FSLW joint to the thermo-mechanically affected zone of the FSP zone of FSLW&FSP joint. The combined action of effective sheet thickness increasement, microstructure uniformity, grain refinement, and local stress concentration reduction act as the strengthening mechanism of the lap joint of aluminum alloys.
