Abstract Spatial Kramers-Kronig (KK) media offer a possible route to obtain omnidirectional light absorption within a thin layer of material. However, the experimental realizations are typically limited to a specific polarization, i.e., either transverse electric (TE) or transverse magnetic (TM), hence lacking specific implementations for the absorption of unpolarized light. In this work, we propose theoretically and demonstrate experimentally a polarization-independent KK medium which performs omnidirectional reflectionless absorption for both TE and TM polarized waves. Our design makes use of a special matryoshka metamaterial, whose electric and magnetic responses can be independently controlled with minimized crosstalk. To extend the absorption spectrum, the inner truncation boundary of the KK medium is set at a position far away from the spatial Lorentz resonance, where the constitutive parameter of the metamaterial remains unitary over a broad frequency band. A mini anechoic chamber, 6.83-wavelength in diameter, is constructed using the designed annulus-shaped KK medium. The measured fields for both TE and TM polarizations confirm the polarization-independent omnidirectional and nearly reflectionless absorption in a broadband frequency range.

Abstract Background Delayed graft function (DGF) is an important complication after kidney transplantation surgery. The present study aimed to develop and validate a nomogram for preoperative prediction of DGF on the basis of clinical and histological risk factors. Methods The prediction model was constructed in a development cohort comprising 492 kidney transplant recipients from May 2018 to December 2019. Data regarding donor and recipient characteristics, pre-transplantation biopsy results, and machine perfusion parameters were collected, and univariate analysis was performed. The least absolute shrinkage and selection operator regression model was used for variable selection. The prediction model was developed by multivariate logistic regression analysis and presented as a nomogram. An external validation cohort comprising 105 transplantation cases from January 2020 to April 2020 was included in the analysis. Results 266 donors were included in the development cohort, 458 kidneys (93.1%) were preserved by hypothermic machine perfusion (HMP), 96 (19.51%) of 492 recipients developed DGF. Twenty-eight variables measured before transplantation surgery were included in the LASSO regression model. The nomogram consisted of 12 variables from donor characteristics, pre-transplantation biopsy results and machine perfusion parameters. Internal and external validation showed good discrimination and calibration of the nomogram, with Area Under Curve (AUC) 0.83 (95%CI, 0.78–0.88) and 0.87 (95%CI, 0.80–0.94). Decision curve analysis demonstrated that the nomogram was clinically useful. Conclusion A DGF predicting nomogram was developed that incorporated donor characteristics, pre-transplantation biopsy results, and machine perfusion parameters. This nomogram can be conveniently used for preoperative individualized prediction of DGF in kidney transplant recipients.

Highlights Challenges in developing practical all-solid-state lithium–sulfur batteries (ASSLSBs) and recently devised concepts to address those critical challenges have been discussed. Recent developments in comprehending solid-state electrolytes, cathodes, and highperformance anodes, including key challenges associated with ion transport, electrochemical properties, and processing methods, have been discussed. Prospects of ASSLSBs for commercial use and guiding forthcoming research and development efforts in this area have been presented.

This study aimed to investigate the metallographic structure and the impact of the heat treatment process on the MAR-M247 superalloy, a high-temperature nickel-based superalloy commonly used in turbine blades. The heat treatment process can potentially influence the mechanical properties of the MAR-M247 superalloy at different temperatures. A strength simulation analysis of gas turbine blades should include the variations in the mechanical properties of the material. The effect of heat treatment on grain size was investigated by metallographic experiments, and numerical calculations of material mechanical properties were conducted. The mechanical property parameters necessary for finite element analysis of turbine blades were determined. Finally, a finite element simulation model of the blade was established based on these mechanical property parameters, and strength analysis was performed. The simulation results provided the stress distribution and the strength of the turbine blade.

Background Patients access relevant information concerning their orthopaedic surgery resources through multiple information channels before presenting for clinical treatment. Recently, artificial intelligence (AI)-powered chatbots have become another source of information for patients. The currently developed AI chat technology ChatGPT (OpenAI LP) is an application for such purposes and it has been rapidly gaining popularity, including for patient education. This study sought to evaluate whether ChatGPT can correctly answer frequently asked questions (FAQ) regarding periprosthetic joint infection (PJI). Methods Twelve FAQs about PJI after hip and knee arthroplasty were identified from the websites of fifteen international clinical expert centres. ChatGPT was confronted with these questions and its responses were analysed for their accuracy using an evidence-based approach by a multidisciplinary team. Responses were categorised in four groups: (1) Excellent response that did not require additional improvement; (2) Satisfactory responses that required a small amount of improvement; (3) Satisfactory responses that required moderate improvement; and (4) Unsatisfactory responses that required a large amount of improvement. Results From the analysis of the responses given by the chatbot, no reply received an ‘unsatisfactory’ rating; one did not require any correction; and the majority of the responses required low (7 out of 12) or moderate (4 out of 12) clarification. Although a few responses required minimal clarification, the chatbot responses were generally unbiased and evidence-based, even when asked controversial questions. Conclusions The AI-chatbot ChatGPT was able to effectively answer the FAQs of patients seeking information around PJI diagnosis and treatment. The given information was also written in a manner that can be assumed to be understandable by patients. The chatbot could be a valuable clinical tool for patient education and understanding around PJI treatment in the future. Further studies should evaluate its use and acceptance by patients with PJI.

Abstract Transformation optics (TO) provides a powerful tool to manipulate electromagnetic waves, enabling the design of invisibility cloaks, which can render objects invisible. Despite many years of research, however, invisibility cloaks experimentally realized thus far can only operate at a single frequency. The narrow bandwidth significantly restricts the practical applications of invisibility cloaks and other TO devices. Here, a general design strategy is proposed to realize a multiband anisotropic metamaterial characterized by two principal permittivity components, i.e., one infinite and the other spatially gradient. Through a proper transformation and combination of such metamaterials, an omnidirectional invisibility cloak is experimentally implemented, which is impedance‐matched to free space at multiple frequencies. Both far‐field numerical simulations and near‐field experimental mappings confirm that this cloak can successfully suppress scattering from multiple large‐scale objects simultaneously at 5 and 10 GHz. The design strategy and corresponding practical realization bring multiband transformation optical devices one step closer to reality.

Abstract Background Familial hypercholesterolemia (FH) is an autosomal semi-dominant disease, characterized by markedly elevated levels of low-density lipoprotein cholesterol (LDL-c) from conception and accelerated atherosclerotic cardiovascular disease, often resulting in early death. The aim of this study was to evaluate the prevalence of clinically defined FH in Chinese Han patients with acute coronary syndrome (ACS) and compare the long-term prognosis of ACS patients with and without FH receiving lipid-lowering therapy containing statins after a coronary event. Methods All ACS patients were screened at the Second Affiliated Hospital of Xi’an Jiaotong University between Jan 2019 and Sep 2020, and 531 participants were enrolled. All were examined for FH under the Dutch Lipid Clinical Network (DLCN) criteria, and those patients were divided into definite/probable FH, possible FH and unlikely FH. The severity of coronary artery disease was evaluated by the Gensini scoring system. Plasma levels of total cholesterol (TC), triacylglycerol (TG), HDL-cholesterol (HDL-c), LDL-cholesterol (LDL-c), very low-density lipoproteins-cholesterol (VLDL-c), apolipoprotein A1 (apoA1), apolipoprotein B (apoB) and lipoprotein (a) (Lp(a)) were determined centrally at baseline and the last follow-up visit in the fasting state. The non-high-density lipoprotein cholesterol (non-HDL-c) concentration, the TC/HDL-c and apoB/apoA1 ratios were calculated. After FH patients received lipid-lowering treatment containing statin, the target LDL-c levels recommended by the guidelines (LDL-c 50% from baseline) were evaluated, and the occurrence of major adverse cardiovascular and cerebrovascular events (MACCE) during the 12-month follow-up was recorded. Results The prevalence of clinically definite or probable FH was 4.3%, and the prevalence of possible FH was 10.6%. Compared with the unlikely FH patients with ACS, the FH patients had higher levels of TC, LDL-c, apoB, Lp(a), non-HDL-c, TC/HDL-c and apoB/apoA1 ratio, more severe coronary artery diseases and greater prevalence of left main and triple or multiple vessel lesions. After lipid-lowering therapy containing statins, a minority of FH patients reached the target LDL-c levels defined by the guidelines (χ2 = 33.527, P 0.05). Conclusions FH was an independent risk factor for MACCE in adult patients after a coronary event during long-term follow-up. However, there was inadequate high-intensity statins prescriptions for high-risk patients in this current study. It is important for FH patients to optimize lipid-lowering treatment strategies to reach the target LDL-c level to improve the long-term prognosis of clinical outcomes.

The silicon vacancy (SiV) color centers in diamond have significant potential in the fields of quantum information, nanophotonics, bioimaging and sensing, due to its good structural stability at room temperature, strong photostability, short luminescence lifetime, narrow zero-phonon line bandwidth, and avoidance of biological autofluorescence. They have attracted extensive attention and in-depth research from researchers. The applications of SiV color centers in the above areas require addressing key issues such as control over the number and charge state of SiV color centers in diamond, as well as improving the luminous efficiency. Therefore, this review summarizes some recent relevant research progress, including common methods for preparing SiV color centers, control over their luminescence intensity, preparation of SiV single-photon sources, and research on the control of SiV color center charge states. Based on these, an outlook is provided for the preparation and application of SiV color centers in diamond.

We investigated the correlation between microstructures and optical properties of boron ion implanted microcrystalline diamond (B-MCD) thin films annealed at different temperatures. Ellipsometry characterization of B-MCD films was performed in the wavelength range of 380–1000 nm and the disperse parameters of the films such as single oscillator energy and dispersive energy were uniquely determined by using a Gaussian model. The results show that the refractive index increases from 2.18 to 2.21 to 2.28–2.29 with annealing temperature increasing from 700 to 800 °C and then it slightly decreases to 2.25–2.28 with annealing temperature further increasing to 900 °C, implying that the annealing temperature has a significant impact on the optical properties of B-MCD films. Raman spectra reveal that the full width at half maximum of diamond peak significantly decreases after 800 °C annealing, indicating a better quality of diamond crystalline. X-ray photoelectron spectroscopy results show that this sample contains higher sp3 carbon content, lower sp2 and C–O contents on the film surface after 800 °C annealing, implying that fewer defects exist. The results are of great significance for the preparation of excellent boron ion implanted microcrystalline diamond for waveguide based Mach-Zehnder Interferometer liquid sensing.

Experimental results showed that for the diamond film prepared by hot filament chemical vapor deposition (HFCVD) using Ta filament, TaC existed between diamond and the silicon substrate, and diamond grew directly on TaC, while the inherent mechanism was not clear. Here, a special coherent interface Diamond(111)//TaC(111) is observed using high resolution transmission electron microscopy, and then we explore the effects of the TaC with different lattice planes on the diamond formation by first-principle calculations. The results show that C tends to adsorb on the TaC(111) C-terminated surface. The strong covalent bond between C from diamond and Ta from TaC is formed in the Diamond(111)//TaC(111) interface, while only C–C covalent bonds are formed at the Graphite(002)/TaC(111). This makes diamond thermodynamically more stable than graphite on the TaC surfaces. Our investigations provide critical information to understand the complex diamond formation mechanism, especially with the presence of TaC.

Abstract Background Detection of exotic plant pathogens and preventing their entry and establishment are critical for the protection of agricultural systems while securing the global trading of agricultural commodities. High-throughput sequencing (HTS) has been applied successfully for plant pathogen discovery, leading to its current application in routine pathogen detection. However, the analysis of massive amounts of HTS data has become one of the major challenges for the use of HTS more broadly as a rapid diagnostics tool. Several bioinformatics pipelines have been developed to handle HTS data with a focus on plant virus and viroid detection. However, there is a need for an integrative tool that can simultaneously detect a wider range of other plant pathogens in HTS data, such as bacteria (including phytoplasmas), fungi, and oomycetes, and this tool should also be capable of generating a comprehensive report on the phytosanitary status of the diagnosed specimen. Results We have developed an open-source bioinformatics pipeline called PhytoPipe (Phytosanitary Pipeline) to provide the plant pathology diagnostician community with a user-friendly tool that integrates analysis and visualization of HTS RNA-seq data. PhytoPipe includes quality control of reads, read classification, assembly-based annotation, and reference-based mapping. The final product of the analysis is a comprehensive report for easy interpretation of not only viruses and viroids but also bacteria (including phytoplasma), fungi, and oomycetes. PhytoPipe is implemented in Snakemake workflow with Python 3 and bash scripts in a Linux environment. The source code for PhytoPipe is freely available and distributed under a BSD-3 license. Conclusions PhytoPipe provides an integrative bioinformatics pipeline that can be used for the analysis of HTS RNA-seq data. PhytoPipe is easily installed on a Linux or Mac system and can be conveniently used with a Docker image, which includes all dependent packages and software related to analyses. Diaporthe longicolla (syn. Phomopsis longicolla) is an important seed-borne fungal pathogen and the primary cause of Phomopsis seed decay (PSD) in soybean. PSD is one of the most devastating seed diseases, reducing soybean seed quality and yield worldwide. As part of a genome sequencing project on the fungal Diaporthe–Phomopsis complex, draft genomes of eight D. longicolla isolates were sequenced and assembled. Sequences of mitochondrial genomes were extracted and analyzed. The circular mitochondrial genomes ranged from 52,534 bp to 58,280 bp long, with a mean GC content of 34%. A total of 14 core protein-coding genes, 23 tRNA, and 2 rRNA genes were identified. Introns were detected in the genes of atp6, cob, cox1, cox2, cox3, nad1, nad2, nad5, and rnl. Three isolates (PL7, PL10, and PL185E) had more introns than other isolates. Approximately 6.4% of the mitochondrial genomes consist of repetitive elements. Moreover, 48 single-nucleotide polymorphisms (SNPs) and were identified. The mitochondrial genome sequences of D. longicolla will be useful to further study the molecular basis of seed-borne pathogens causing seed diseases, investigate genetic variation among isolates, and develop improved control strategies for Phomopsis seed decay of soybean.

With the development of urban rail transportation, the environmental vibration problem caused by the running of metro vehicles has received attention. In order to reduce ground vibration near buildings caused by metro vehicles running on viaducts, this paper establishes the train–track–viaduct rigid–flexible coupling dynamics model and pier–soil–building finite element model and carries out the simulation calculation and analysis of ground vibration. The influence of train speed and fastener stiffness on ground vibration is explored, and the vibration reduction effect of the track vibration reduction pad and continuous support vibration reduction structure is studied. The results show that the ground vibration near the building caused by the train running on the viaduct decreases with the reduction in speed, when the speed is reduced to 40 km/h, the vibration attenuation is slower as the speed continues to be reduced; the reduction in the vertical stiffness of fasteners can reduce ground vibration; the arrangement of the vibration damping pad can effectively reduce ground vibration, and after installing a vibration damping pad, 0–23 Hz and 50–80 Hz ground vibration speeds are effectively suppressed. In order to meet the environmental requirements for ground vibration, the vehicle speed can be reduced to less than 35 km/h or vibration damping mats can be installed.

Abstract High throughput sequencing (HTS) can supplement and may replace diagnostic tests for plant pathogens. However, the methodology and processing of HTS data must first be optimized and standardized to ensure the sensitivity and repeatability of the results. Importation of sugarcane into the United States is highly regulated, and sugarcane plants are subjected to strict quarantine measures and diagnostic testing, especially for the presence of certain viruses of regulatory concern. Here, we tested whether HTS could reliably detect four RNA and three DNA sugarcane viruses over three seasons (fall, winter, and spring) and in three tissue types (root, stem, and leaves). Using HTS on ribosomal depleted total RNA samples, we reliably detected RNA viruses in all tissue types and across all seasons, but we failed to confidently detect DNA viruses in some samples. We recommend that future optimization be employed to ensure the robust and reliable detection of all regulated sugarcane viruses by HTS.

In order to broaden the application of coconut oil, virgin coconut oil, gelatin and pectin were used to prepare complex gel fudge. The optimal formulation of coconut oil complex gel fudge was obtained via single-factor test and orthogonal tests with sensory score as evaluation index, and the effect of storage time on the texture of gel fudge was also investigated. The results showed that the optimal technological parameters of coconut oil complex gel fudge were as follows: Coconut oil 9.44 g, total sugar 34.00 g, total gum 35.89 g, among which the mass ratio of maltose, xylitol and brown sugar was 24:5:5, the mass ratio of total sugar and total gum was 36:38, and the mass ratio of coconut oil and sugar gum was 10:74. The complex gel fudge prepared under this formula had rich coconut flavor, smooth and non-sticky, proper sweetness, and the sensory score was 84.22±1.17. Results of texture analysis showed that the hardness, gumminess and chewiness of the coconut oil complex gel fudge increased extremely significantly with the extension of storage time (P0.05). This study provides references for coconut oil utilization.

Understanding the degradation kinetics and mechanisms of trace organic contaminants (TrOCs) by UV-based advanced oxidation processes (UV-AOPs) are pivotal in realizing their efficient application in water treatment. However, the relevant knowledge in practical flow-through reactors remains a void, compared with that of commonly used batch reactors. To fill the knowledge gaps, the current work investigated the degradation of atrazine (ATZ) in flow-through UV-AOP systems with different light sources and chlorine additions. The results showed that UV/Cl2 in the reactors (with a diameter of 50 mm) was not very efficient in ATZ degradation while the pseudo-first order degradation rate constant was elevated by over 2.7 times with vacuum UV (VUV)/UV. In contrast to observations in the batch reactors, the addition of chlorine to the flow-through VUV/UV system unexpectedly decreased the rate constant by about 39%. The analysis of the relative contributions of different degradation pathways revealed that the inhibitory effect of the chlorine addition arose from the transformation of HO• to reactive chlorine species (e.g., ClO•) which had low reaction rate constants with ATZ. The baffle implementation promoted the ATZ degradation by 12–58%, mainly due to an enhanced mixing that facilitated the radical oxidation. The energy costs of the UV-AOPs in ATZ removal ranged within 0.40–1.11 kWh m−3 order−1. The findings of this work are helpful in guiding efficient VUV/UV and VUV/UV/Cl2 processes in drinking water treatment.

Partitioning experiments and the chemistry of iron meteorites indicate that the light element nitrogen could be sequestered into the metallic core of rocky planets during core–mantle differentiation. The thermal conductivity and the mineralogy of the Fe–N system under core conditions could therefore influence the planetary cooling, core crystallization, and evolution of the intrinsic magnetic field of rocky planets. Limited experiments have been conducted to study the thermal properties and phase relations of Fe–N components under planetary core conditions, such as those found in the Moon, Mercury, and Ganymede. In this study, we report results from high-pressure experiments involving electrical resistivity measurements of Fe–N phases at a pressure of 5 GPa and temperatures up to 1400 K. Four Fe–N compositions, including Fe–10%N, Fe–6.4%N, Fe–2%N, and Fe–1%N (by weight percent), were prepared and subjected to recovery experiments at 5 GPa and 1273 K. These experiments show that Fe–10%N and Fe–6.4%N form a single hexagonal close-packed phase (ɛ-nitrides), while Fe–2%N and Fe–1%N exhibit a face-centered cubic structure (γ-Fe). In separate experiments, the resistivity data were collected during the cooling after compressing the starting materials to 5 GPa and heating to ~1400 K. The resistivity of all compositions, similar to the pure γ-Fe, exhibits weak temperature dependence. We found that N has a strong effect on the resistivity of metallic Fe under rocky planetary core conditions compared to other potential light elements such as Si. The temperature-dependence of the resistivity also revealed high-pressure phase transition points in the Fe–N system. A congruent reaction, ε ⇌ γ’, occurs at ~673 K in Fe–6.4%N, which is ~280 K lower than that at ambient pressure. Furthermore, the resistivity data provided constraints on the high-pressure phase boundary of the polymorphic transition, γ ⇌ α, and an eutectoid equilibrium of γ’ ⇌ α + ε. The data, along with the recently reported phase equilibrium experiments at high pressures, enable construction of a phase diagram of the Fe–N binary system at 5 GPa.

To explore the effects of ultra-high pressure treatment (UHP) on the quality properties of shrimp slides, the color, water holding capacity, cooking yield, gel strength, texture properties and sensory characteristics of shrimp slides were investigated when treated by different pressure (200~400 MPa) and holding time (5~20 min). The results illustrated that the brightness (L*) and whiteness (W) of shrimp slides were not significantly affected by the different pressures and holding times, whereas the redness (a*) decreased significantly and the yellowness (b*) increased compared to the control group. Meanwhile, the water holding capacity and cooking yield of shrimp slides were affected by the different pressures and holding times, and the maximum values of both properties reached to 84.17% and 92.80% when treated for 10 min under 200 MPa pressure, respectively. Compared to the control group, the gel strength, hardness, chewiness, and springiness of the shrimp slides decreased when pressure and holding time increased, while the cohesiveness did not change significantly. The results of sensory evaluation showed that the UHP treatment had no negative impact on the sensory properties of shrimp slides, and the overall acceptability of shrimp slides held at 200 MPa for 10 min was closest to that of the control group. Therefore, the cooking yield and texture characteristics of shrimp slips products could be improved upon UHP treatment at 200 MPa for 10 min. It is expected that this study will provide a theoretical basis for the development of shrimp slippery products.

Small incision lenticule extraction (SMILE) has become one of the mainstream refractive surgeries in recent years, with satisfactory efficacy, safety, and predictability. SMILE-derived refractive lenticule, the byproduct of the surgery, holds great potential in clinical practice given its easy access and good biocompatibility. Numerous studies have been published to describe its applications in refractive correction, corneal ectasia diseases, and corneal defects. The feasibility and safety were validated in both animal models and clinical studies. Moreover, the preservation method is also crucial for its further promotion and application. Novel techniques are also evaluated and applied in lenticule preservation. We covered the recent advances in the preservation of corneal stromal lenticules and their clinical reuse in this review.

Abstract Aims Abnormalities in potassium homeostasis are frequently seen in hospitalized patients. A poor outcome in heart failure (HF) has been linked to both hypokalaemia and hyperkalaemia. The studies on the connection between variations in potassium levels and all‐cause mortality remain scarce. We delineated trajectories of potassium levels and investigated the association of these trajectories with all‐cause mortality of critically ill patients with HF. Methods and results A retrospective analysis of blood potassium levels (9 times) in patients with HF after being admitted to the intensive care unit (ICU). Potassium levels were divided into three groups according to the first serum potassium level in ICU and thereafter categorized as follows: hypokalaemia group (n = 336) (5.0 mmol/L). According to the group‐based trajectory modelling (GBTM), the hyperkalaemia group and the normal blood potassium‐level group can be divided into three trajectory groups: the low‐level stable group, the medium‐level stable group, and the high‐level decline group. The hypokalaemia group can be divided into two trajectory groups: the low‐level rise group and the high‐level rise group. A total of 4053 HF patients were included (mean age 71.81 ± 13.12 years, 54.90% males, 45.10% females). After adjusting for possible confounding variables, in the hyperkalaemia group, the low‐level stable group had lower 28 day [high‐level decline group vs. low‐level stable group hazard ratio (HR), 95% confidence interval (CI): 2.917, 1.555–5.473; P

High-speed direct-drive permanent magnet synchronous motors (PMSMs), supported by elastic foil gas bearings, have broad applications, such as in microcompressors. However, some problems remain to be solved for the electrical performance analysis of PMSMs. For example, there is presently no related analytical model that can be used in rotor dynamics expression for this type of PMSM. This study aimed to establish theoretical models for electromagnetic force density and torque. The process involved both theoretical and experimental research. The analytic models of air gap magnetic density, electromagnetic force density, and electromagnetic performance were established for a PMSM with a parallel magnetized cylindrical permanent magnet. The analytic calculation was conducted, and the results of the analytic model were obtained. The analytical model of the electromagnetic torque and force can be applied in theoretical research on rotor dynamics. The model provides a theoretical basis and method for studying the influence of the electromagnetic load on rotor dynamics. A finite element simulation analysis of the electrical performance of the PMSM was carried out. An electrical performance experiment was conducted. The deviation between the experimental result and the theoretical value was less than 4%. This result indicated that the analytic models could be used in a dynamics analysis of compressors that are directly driven by a PMSM for application in engineering and industrial contexts.

Based on the concept of component assembly, a novel star polygon-coupled honeycomb metamaterial, which achieves a collaborative improvement in load-bearing capacity and vibration suppression performance, is proposed based on a common polygonal structure. The compression simulation and experiment results show that the load-bearing capacity of the proposed metamaterial is three times more than that of the initial metamaterial. Additionally, metal pins are attached and particle damping is applied to the metamaterial to regulate its bandgap properties; the influence of configuration parameters, including the size, number, position, and material of the metal pins, on bandgaps is also investigated. The results show that the bandgap of the proposed metamaterial can be conveniently and effectively regulated by adjusting the parameters and can effectively suppress vibrations in the corresponding frequency band. Particle damping can be used to continuously adjust the frequency of the bandgap and further enhance the vibration suppression capacity of the metamaterial in other frequency bands. This paper provides a reference for the design and optimization of metamaterials.

Summary: Budd-Chiari syndrome (BCS) is characterized by hepatic venous outflow obstruction, posing life-threatening risks in severe cases. Reported risk factors include inherited and acquired hypercoagulable states or other predisposing factors. However, many patients have no identifiable etiology, and causes of BCS differ between the West and East. This study recruited 500 BCS patients and 696 normal individuals for whole-exome sequencing and developed a polygenic risk scoring (PRS) model using PLINK, LASSOSUM, BLUP, and BayesA methods. Risk factors for venous thromboembolism and vascular malformations were also assessed for BCS risk prediction. Ultimately, we discovered potential BCS risk mutations, such as rs1042331, and the optimal BayesA-generated PRS model presented an AUC >0.9 in the external replication cohort. This model provides particular insights into genetic risk differences between China and the West and suggests shared genetic risks among BCS, venous thromboembolism, and vascular malformations, offering different perspectives on BCS pathogenesis.

Sorafenib (SFN) is a multi-kinase inhibitor drug for the treatment of advanced hepatocellular carcinoma (HCC), but its limited efficacy is a major obstacle to the clinical outcomes of patients with HCC. We aimed to explore a novel molecular mechanism underlying the chemosensitivity of HCC to SFN, and to identify a promising therapeutic target for HCC treatment. In this study, bioinformatic analysis revealed that DDX24 was associated with poor survival in HCC cases, and significantly related to the pathways modulating tumor development. DDX24 regulated HCC cell proliferation and migration potentials. Moreover, reduction of DDX24 promoted the sorafenib-mediated inhibition of HCC cell growth and migration, the elevation of sorafenib-induced HCC cell apoptosis. DDX24 overexpression suppressed the inhibitory effect of SFN on cell proliferation and migration and reduced the apoptosis induced by SFN. Further, DDX24, combined with SFN treatment, presented a synergistic enhancement of the sensitivity of SFN to the growth and migration of HCC cells via AKT/ERK and the epithelial-mesenchymal transition (EMT) pathways, and that it modulated apoptosis via the caspase/PARP pathway. Mechanistically, SNORA18 served as a target gene for DDX24, regulating the chemosensitivity of sorafenib-treated HCC cells. Furthermore, SNORA18 knockdown or overexpression could partially reverse the inhibition or elevation of cell viability, colony formation and migration induced by DDX24 in sorafenib-treated HCC cells, respectively. Collectively, our results suggest that DDX24 regulates the chemosensitivity of HCC to SFN by mediating the expression of SNORA18, which may act as an effective therapeutic target for improving SFN efficiency in HCC treatment.

Microorganisms promoted corrosion has caused significant loss to marine engineering and the antibacterial coatings have served as a solution that has gained attention. In this study, the chemical vapour deposition technique has been employed to grow three different types of diamond coatings, namely, ultrananocrystalline diamond (UNCD), nanocrystalline diamond (NCD), and microcrystalline diamond (MCD) coatings. The evolution of associated surface morphology and the surface functional groups of the grown coatings have demonstrated antibacterial activity in seawater environments. It is found that different ratio of sp3/sp2 carbon bonds on the diamond coatings influences their surface property (hydrophobic/hydrophilic), which changes the anti-adhesion behaviour of diamond coatings against bacteria. This plays a critical role in determining the antibacterial property of the developed coatings. The results show that the diamond coatings arising from the deposition process kill the bacteria via a combination of the mechanical effects and the functional groups on the surface of UNCD, NCD, and MCD coatings, respectively. These antibacterial coatings are effective to both Gram-negative bacteria (E. coli) and Gram-positive bacteria (B. subtilis) for 1–6 h of incubation time. When the contact duration is prolonged to 6 h or over, the MCD coatings begin to reduce the bacteria colonies drastically and enhance the bacteriostatic rate for both E. coli and B. subtilis.

Background and aim: Percutaneous transhepatic biliary drainage (PTBD) does not always lead to a reduction in serum total bilirubin (TBil) level in patients with hepatocellular carcinoma and obstructive jaundice. We aimed to develop a model for pre-PTBD prediction of post-procedural TBil decrease in these patients. Materials and methods: Retrospective database searches were conducted at four teaching hospitals (reference period: January 2010 to December 2018), and baseline characteristics of eligible patients were extracted. Any decrease in TBil after PTBD and the lowest level of TBil post-PTBD

Co-based catalyst has excellent chlorine resistance, so it is an effective catalyst to degrade VC at present. A series of high-performance CoxBa1 catalysts were prepared by doping Ba into Co3O4 and changing the Ba/Co mol ratio. Under the condition of space velocity of 15,000 ml·g−1·h−1, the conversion rate of VC reached 90% at 199 °C in Co19Ba1, the temperature is much lower than T90 of pure Co3O4 (309 °C), and the catalysis activity of Co19Ba1 was higher than that of the other alkali metal-doped Co19M1 (M = Sr, Ca). Ba species in CoxBa1 mainly exist in the form of BaCO3. With the addition of BaCO3, the particle size of CoxBa1 decreases, resulting in the increase of active site of CoxBa1. BaCO3 can also make CoxBa1 obtain better low-temperature reducibility and more active components. In addition, the increase of BaCO3 content will also increase the basic sites of CoxBa1, and more basic sites can effectively protect Co3O4 from Cl poisoning. When Ba/Co is 1/19, the content of BaCO3 is the highest, so Co19Ba1 also shows the best activity.

Tire cavity resonance noise (TCRN) is one of main contributors to vehicle interior noise, which has long been a concern in the automotive industry and academia. As suspension is crucial for the propagation of TCRN energy into the vehicle, the propagation characteristics of energy in automobile suspension is studied in this research. Firstly, the finite element model of a McPherson suspension system connected to an aluminum alloy wheel with a Y-shaped spoke was established. Then, a modal analysis and response calculations of the McPherson suspension system connected to the aluminum alloy wheel with a Y-shaped spoke were carried out. Finally, the propagation characteristics of TCRN in the McPherson suspension system connected to the aluminum alloy wheel with a Y-shaped spoke were studied and analyzed by the power flow method under different working conditions. The power flow output via the lower arm front bushing was the largest, while the output via the rear bushing was the smallest in the Y-spoke aluminum alloy wheel and suspension system. The areas in the suspension system with high stress are located at the steering knuckle, lower swing arm, and shock absorber. Therefore, study of the propagation characteristics can provide a basis for a McPherson suspension system design.

To investigate the differences of aroma characteristics between Jingui green tea (JGT) and Jingui flowers, Jingui (JG) and Dangui (DG) flowers were analyzed the aroma quality and volatile compounds by the method of sense evaluation and GC-MS, respectively. JG flowers were selected to scent green tea and the aroma quality and chemical characteristics of Jingui green tea were further researched. The results showed that JG showed a more excellent aroma quality than that of DG, and its total content of volatile components was 22.7% higher than DG, so JG flowers was used to make flower tea and acted as a control like green tea (GT). Comparing the aroma characteristics of JGT, JG and GT, the fragrance and floral characteristics of JGT were slightly inferior to those of JG and higher than that of GT. JGT absorbed 8 ingredients such as linalool, α-ylangene, megastigma-4,6(E),8(E)-triene, cis-pyranoid linalool oxide 1, α-ionone, 2,6,6-tetramethyl-1-cyclohexene-1-propanol, γ-decalactone and 4-hydroxy-β-ionone from JG and 6 new components such as 3,4-dimethylaniline, trans-Z-α-bisabolene epoxide, geranyl isovalerate, 1,2,3,4-tetrahydro-1,1,6-trimethyl-naphthalene, dihydro-beta-ionone and phytol acetate formed during green tea scenting, and kept 10 common volatile components with green tea (control), but its volatile components was still 60.4% lower than that of JG. In summary, JGT obtained the parts of aroma components from JG and GT, and formed its unique components by scenting technology, so it was beneficial for technology optimization and quality improvement of flower tea.

Defensive behaviors are critical for animal’s survival. Both the paraventricular nucleus of the hypothalamus (PVN) and the parabrachial nucleus (PBN) have been shown to be involved in defensive behaviors. However, whether there are direct connections between them to mediate defensive behaviors remains unclear. Here, by retrograde and anterograde tracing, we uncover that cholecystokinin (CCK)-expressing neurons in the lateral PBN (LPBCCK) directly project to the PVN. By in vivo fiber photometry recording, we find that LPBCCK neurons actively respond to various threat stimuli. Selective photoactivation of LPBCCK neurons promotes aversion and defensive behaviors. Conversely, photoinhibition of LPBCCK neurons attenuates rat or looming stimuli-induced flight responses. Optogenetic activation of LPBCCK axon terminals within the PVN or PVN glutamatergic neurons promotes defensive behaviors. Whereas chemogenetic and pharmacological inhibition of local PVN neurons prevent LPBCCK-PVN pathway activation-driven flight responses. These data suggest that LPBCCK neurons recruit downstream PVN neurons to actively engage in flight responses. Our study identifies a previously unrecognized role for the LPBCCK-PVN pathway in controlling defensive behaviors.

Wheat stripe rust is a destructive disease in many cool and temperate regions around the world. Exploiting novel sources of resistance can provide wheat cultivars with robust and durable resistance to stripe rust. The wheat–Thinopyrum intermedium addition line TAI-14 was proven to carry a stripe rust resistance gene (named as YrT14) on the alien Th. intermedium chromosome. In order to transfer the resistance gene to wheat, wheat–Th. intermedium translocation lines were created by irradiating the pollen of the line TAI-14. We totally obtained 153 wheat–Th. intermedium translocation lines, among which the long alien segmental translocation line Zhongke 78 and the intercalary translocation line Zhongke 15 not only showed good integrated agronomic traits but also were identified as highly resistant to stripe rust in both seedling and adult plant stages. The alien chromatin in Zhongke 15 was identified as an insertion into the satellite of chromosome 6B, a type of translocation never reported before in chromosome engineering. By screening Simple Sequence Repeat (SSR) and Expressed Sequence Tag (EST) markers as well as the markers developed from RNA-sequencing (RNA-Seq) data, 14 markers were identified specific for the alien chromosome and a physical map was constructed. Both Zhongke 78 and Zhongke 15 could be used as a novel source of stripe rust resistance for wheat breeding, and the linked marker T14K50 can be used for molecular marker–assisted breeding. Finally, based on the karyotype, reaction to stripe rust, and genome resequencing data of different wheat–Th. intermedium translocation lines, the stripe rust resistance gene YrT14 was located to an 88.1 Mb interval from 636.7 to 724.8 Mb on Th. intermedium chromosome 19 corresponding to 7J or 7Js.

Tetrabromobisphenol A (TBBPA) is one of the most prevalently used brominated flame retardants. Due to its persistence, it is predominantly found in environmental matrices and has the potential to generate multi-generational toxicity. However, knowledge of its adaptive response or long-term residual effect in multi-generations, and molecular mechanisms remain understudied. In the current study, the model animal nematode Caenorhabditis elegans (C. elegans) was exposed to TBBPA at environmentally realistic concentrations (0.1–1000 μg L−1) for four consecutive generations (G0 to G3). Degenerative age-related multiple endpoints including lifespan, locomotion behaviors, growth, reproduction, oxidative stress-related biochemical responses, cell apoptosis, and stress related gene expressions were assessed in the continuous exposure generations (G0 and G3) and the discontinuously exposed generations (T3 and T′3). The results showed that changes in degenerative age-related response monitored four generations varied in direction and magnitude depending on the TBBPA concentrations, and the response intensify ranked as G0 > T′3/G3 > T3. TBBPA at 1 μg L−1 dosage was detected as the lowest observed effect concentration in multi-biomarkers. The underlying mechanism of aging phenotypes was that reactive oxygen species accumulation led to cell apoptosis regulated by gene ape-1, and confirmed catalase enzyme and superoxide dismutase activity played a crucial role in the detoxification process of TBBPA at the molecular level. This study provided insights into the underlying mechanism of TBBPA-interfered longevity and its environmental multi-generational potential risks.

Herein, the mid-infrared (7.7–13.7 μm) diamond-based phase shifter was designed and optimized by finite-element analysis. The ridge-shaped diamond waveguide is designed and doped to form the internal p–n structure, and the internal carrier distribution is changed by applying forward and reverse voltages to change the effective refractive index to achieve the effect of π-phase shift. The results show that when p-doping concentration is 4 × 1017 cm−3 and n doping concentration is 1 × 1018 cm−3, upon the reverse voltage (8 V) is applied, the change of the real part of effective refractive index (ΔR) is 1.6 × 10−5, and the length of the phase shifter (L) required to realize the π-phase shift is 241 mm; upon the forward voltage (–8 V) is applied, ΔR increases to 3.2 × 10−4, and the length of the phase shifter required is shortened to 12.03 mm. Such a short length is relatively easy in industrial production. In order to make the refractive index distribution more uniform, the carrier concentration has been optimized as 1 × 1017 cm−3 for p-type and 4 × 1017 cm−3 for n-type, respectively.

Abstract In this study, a novel coupling structure based on half‐mode substrate‐integrated waveguide (HMSIW) is proposed with a small size and broad bandwidth. The HMSIW coupler is realised by connecting their equivalent magnetic walls with metal strips, which avoids exposing the semi‐enclosed magnetic walls outside. Hence, it has greater anti‐interference capability than the existing HMSIW couplers. The directional coupling principle is introduced by analysing the proposed single connecting strip coupler. In order to lower return loss and higher isolation, a 3 dB directional coupler working from 10.8 to 16.8 GHz is designed with two connecting strips. Finally, a six‐port circuit is achieved by utilising this compact 3 dB directional coupler. Furthermore, a broadband HMSIW power divider is optimised to satisfy the performance of this six‐port circuit. The proposed coupler with metal strips helps to increase bandwidth and reduce size, which makes it more applicable to demanding systems with complex requirements such as detecting and transceiver systems. The 3 dB coupler and six‐port circuit are fabricated and measured. The measured results are in accordance with the simulated data.

Tire acoustic cavity resonance noise (TACRN) contributes significantly to the interior noise of electric cars and passenger cars with lower powertrain noise, which affects the comfort of the ride. To suppress TACRN effectively, it is crucial to clarify the characteristics of TACRN. In previous studies, the acoustic wave in the tire acoustic cavity is straightforwardly assumed to be the plane wave for convenience. In fact, there exist strict conditions for the acoustic wave propagating in the pipeline to act as a plane wave. The aim of this paper is to make the characteristics and evolution of acoustic waves in tire acoustic cavities clear. To do so, a simplified model of the tire cavity is established, and the sound field distribution and the acoustic wave propagation characteristics in the tire cavity are analyzed based on the theory of acoustic waveguide. Then, the existence ranges of higher-order waves (non-plane waves), the conditions of an acoustic wave evolving into a plane wave, and the frequency range of a plane wave are investigated. Finally, the characteristics and evolution law of an acoustic wave in a tire acoustic cavity are obtained. The work in this paper may deepen the understanding of the characteristics and mechanism of acoustic waves in the tire cavity and be helpful and meaningful for analyzing and suppressing TACRN. Therefore, it is of practical significance to reduce TACRN transmitted to the vehicle and improve the sound quality inside the vehicle.

Summary: A high point of Tibetan Plateau (TP) civilization, the expansive Tubo Empire (618–842 AD) wielded great influence across ancient western China. However, whether the Tubo expansion was cultural or demic remains unclear due to sparse ancient DNA sampling. Here, we reported ten ancient genomes at 0.017- to 0.867-fold coverages from the Dulan site with typical Tubo archaeological culture dating to 1308–1130 BP. Nine individuals from three different grave types have close relationship with previously reported ancient highlanders from the southwestern Himalayas and modern core-Tibetan populations. A Dulan-related Tubo ancestry contributed overwhelmingly (95%–100%) to the formation of modern Tibetans. A genetic outlier with dominant Eurasian steppe-related ancestry suggesting a potential population movement into the Tubo-controlled regions from Central Asia. Together with archeological evidence from burial styles and customs, our study suggested the impact of the Tubo empire on the northeast edge of the TP involved both cultural and demic diffusion.

A comprehensive diagnostic method of known plant viruses and viroids is necessary to provide an accurate phytosanitary status of fruit trees. However, most widely used detection methods have a small limit on either the number of targeted viruses/viroids or the number of samples to be evaluated at a time, hampering the ability to rapidly scale up the test capacity. Here we report that by combining the power of high multiplexing PCR (499 primer pairs) of small amplicons (120-135bp), targeting 27 viruses and 7 viroids of fruit trees, followed by a single high-throughput sequencing (HTS) run, we accurately diagnosed the viruses and viroids on as many as 123 pome and stone fruit tree samples. We compared the accuracy, sensitivity, and reproducibility of this approach and contrast it with other detection methods including HTS of total RNA (RNA-Seq) and individual RT-qPCR for every fruit tree virus or viroid under the study. We argue that this robust and high-throughput cost-effective diagnostic tool will enhance the viral/viroid knowledge of fruit trees while increasing the capacity for large scale diagnostics. This approach can also be adopted for the detection of multiple viruses and viroids in other crops.

In this paper, some new versions of Bohr-type inequalities for bounded analytic functions of Schwarz functions are established. Most of these inequalities are sharp. Some previous inequalities are generalized.

The high morbidity of patients with coronavirus disease 2019 (COVID-19) brings on a panic around the world. COVID-19 is associated with sex bias, immune system, and preexisting chronic diseases. We analyzed the gene expression in patients with COVID-19 and in their microbiota in order to identify potential biomarkers to aid in disease management. A total of 129 RNA samples from nasopharyngeal, oropharyngeal, and anal swabs were collected and sequenced in a high-throughput manner. Several microbial strains differed in abundance between patients with mild or severe COVID-19. Microbial genera were more abundant in oropharyngeal swabs than in nasopharyngeal or anal swabs. Oropharyngeal swabs allowed more sensitive detection of the causative SARS-CoV-2. Microbial and human transcriptomes in swabs from patients with mild disease showed enrichment of genes involved in amino acid metabolism, or protein modification via small protein removal, and antibacterial defense responses, respectively, whereas swabs from patients with severe disease showed enrichment of genes involved in drug metabolism, or negative regulation of apoptosis execution, spermatogenesis, and immune system, respectively. Microbial abundance and diversity did not differ significantly between males and females. The expression of several host genes on the X chromosome correlated negatively with disease severity. In this way, our analyses identify host genes whose differential expression could aid in the diagnosis of COVID-19 and prediction of its severity via non-invasive assay.

Potassium ion batteries (KIBs) have received increasing popularity owing to their distinct advantages. We discover a hitherto unknown C4S nanosheet, a novel carbon-based material with carbon and sulfur consisting of pentagons and hexagons rings. The proposed C4S nanosheet is highly stable dynamically, thermodynamically, mechanically, and chemically, according to first-principles calculations. Moreover, the graphene-like C4S nanosheet is a prospective KIBs anode material, which has a metallic band structure, a relatively low diffusion barrier (0.07 eV), a large capacity (1340 mA h g−1), and an acceptable average voltage (0.44 V). Finally, we demonstrate good cycling stability of the C4S nanosheet. Our findings indicate that the proposed C4S nanosheet is a potentially favorable KIBs anode material.

High-throughput sequencing (HTS), more specifically RNA sequencing of plant tissues, has become an indispensable tool for plant virologists to detect and identify plant viruses. During the data analysis step, plant virologists typically compare the obtained sequences to reference virus databases. In this way, they are neglecting sequences without homologies to viruses, which usually represent the majority of sequencing reads. We hypothesized that traces of other pathogens might be detected in this unused sequence data. In the present study, our goal was to investigate whether total RNA-seq data, as generated for plant virus detection, is also suitable for the detection of other plant pathogens and pests. As proof of concept, we first analyzed RNA-seq datasets of plant materials with confirmed infections by cellular pathogens in order to check whether these non-viral pathogens could be easily detected in the data. Next, we set up a community effort to re-analyze existing Illumina RNA-seq datasets used for virus detection to check for the potential presence of non-viral pathogens or pests. In total, 101 datasets from 15 participants derived from 51 different plant species were re-analyzed, of which 37 were selected for subsequent in-depth analyses. In 29 of the 37 selected samples (78%), we found convincing traces of non-viral plant pathogens or pests. The organisms most frequently detected in this way were fungi (15/37 datasets), followed by insects (13/37) and mites (9/37). The presence of some of the detected pathogens was confirmed by independent (q)PCRs analyses. After communicating the results, 6 out of the 15 participants indicated that they were unaware of the possible presence of these pathogens in their sample(s). All participants indicated that they would broaden the scope of their bioinformatic analyses in future studies and thus check for the presence of non-viral pathogens. In conclusion, we show that it is possible to detect non-viral pathogens or pests from total RNA-seq datasets, in this case primarily fungi, insects, and mites. With this study, we hope to raise awareness among plant virologists that their data might be useful for fellow plant pathologists in other disciplines (mycology, entomology, bacteriology) as well.

Multiorgan venous and lymphatic defect (MOVLD) syndrome is a unique visceral vascular malformations with complex etiologies. In this study, primary skin fibroblasts were obtained from three MOVLD patients and reprogrammed into iPSCs by Yamanaka’s classical strategy. The MOVLD- iPSCs carrying the DDX24 p.Glu271Lys mutation were confirmed by Sanger sequencing. The pluripotency of MOVLD-iPSCs was verified by the specific molecular markers and gene expression, trilineage differentiation potential. The establishment of the MOVLD-iPSCs will provide a useful model for understanding the mechanisms involved the MOVLD and promoting the development of medical treatment.

Nano-crystalline diamond has been extensively researched and applied in the fields of tribology, optics, quantum information and biomedicine. In virtue of its hardness, the highest in natural materials, diamond outperforms the other materials in terms of wear resistance. Compared to traditional single-crystalline and poly-crystalline diamonds, nano-crystalline diamond consists of disordered grains and thus possesses good toughness and self-sharpening. These merits render nano-crystalline diamonds to have great potential in tribology. Moreover, the re-nucleation of nano-crystalline diamond during preparation is beneficial to decreasing surface roughness due to its ultrafine grain size. Nano-crystalline diamond coatings can have a friction coefficient as low as single-crystal diamonds. This article briefly introduces the approaches to preparing nano-crystalline diamond materials and summarizes their applications in the field of tribology. Firstly, nano-crystalline diamond powders can be used as additives in both oil- and water-based lubricants to significantly enhance their anti-wear property. Nano-crystalline diamond coatings can also act as self-lubricating films when they are deposited on different substrates, exhibiting excellent performance in friction reduction and wear resistance. In addition, the research works related to the tribological applications of nano-crystalline diamond composites have also been reviewed in this paper.

Histopathologic grade of hepatocellular carcinoma (HCC) is an important predictor of early recurrence and poor prognosis after curative treatments. This study aims to develop a radiomics model based on preoperative gadoxetic acid-enhanced MRI for predicting HCC histopathologic grade and to validate its predictive performance in an independent external cohort. Clinical and imaging data of 403 consecutive HCC patients were retrospectively collected from two hospitals (265 and 138, respectively). Patients were categorized into poorly differentiated HCC and non-poorly differentiated HCC groups. A total of 851 radiomics features were extracted from the segmented tumor at the hepatobiliary phase images. Three classifiers, logistic regression (LR), support vector machine, and Adaboost were adopted for modeling. The areas under the curve of the three models were 0.70, 0.67, and 0.61, respectively, in the external test cohort. Alpha-fetoprotein (AFP) was the only significant clinicopathological variable associated with HCC grading (odds ratio: 2.75). When combining AFP, the LR+AFP model showed the best performance, with an AUC of 0.71 (95%CI: 0.59–0.82) in the external test cohort. A radiomics model based on gadoxetic acid-enhanced MRI was constructed in this study to discriminate HCC with different histopathologic grades. Its good performance indicates a promise in the preoperative prediction of HCC differentiation levels.

Abstract A low‐loss Ka‐band coupled‐resonator bandpass filter based on the printed ridge gap waveguide (PRGW) technology is presented for millimetre‐wave communication applications. The PRGW is employed to allow electromagnetic‐wave propagation in the air gap in order to avoid dielectric loss and improve transmission performance which is suitable for millimetre‐wave applications. The bandpass filter is implemented with two transmission zeros which are easily obtained by adjusting the stub length of two T‐shape stub‐loaded resonators (SLR). From its simulation frequency response, the filter has an insertion loss of 0.8 dB, a return loss less than −20 dB during the passband and high selectivity in cut‐off frequency. The prototype of the proposed filter is fabricated and tested and a good agreement can be observed between the measured and simulated results.

In this paper, we develop a simple design for simultaneous detection of melamine (MEL) and cyromazine (CYR) by surface-enhanced Raman scattering (SERS) with the formation of well-defined MEL or CYR-aptamer-AgNPs. This detection will take only 10 min, and SERS signals generated by MEL-aptamer-AgNPs exhibits a linear dependence on melamine (R2 = 0.998) concentration ranging from 0.1 to 0.5 ppm on melamine. The detection limit is sensitive down to 43.5 ppb. SERS signals generated by CYR-aptamer-AgNPs exhibits a linear dependence on cyromazine (R2 = 0.997) concentration ranging from 0.1 to 0.5 ppm on cyromazine. The detection limit is sensitive down to 23.6 ppb. This SERS biosensor was first reported for the simultaneous detection of melamine and cyromazine by using aptamer-based in situ silver nanoparticles synthesis. So we expected that the development of this biosensor could provide a good platform for the rapid detection of other pollutants in raw milk.

A major challenge in translating findings from genome-wide association studies (GWAS) to biological mechanisms is pinpointing functional variants because only a very small percentage of variants associated with a given trait actually impact the trait. We used an extensive epigenetics, transcriptomics, and genetics analysis of the TBX15/WARS2 neighbourhood to prioritize this region’s best-candidate causal variants for the genetic risk of osteoporosis (estimated bone density, eBMD) and obesity (waist-hip ratio or waist circumference adjusted for body mass index). TBX15 encodes a transcription factor that is important in bone development and adipose biology. Manual curation of 692 GWAS-derived variants gave eight strong candidates for causal SNPs that modulate TBX15 transcription in subcutaneous adipose tissue (SAT) or osteoblasts, which highly and specifically express this gene. None of these SNPs were prioritized by Bayesian fine-mapping. The eight regulatory causal SNPs were in enhancer or promoter chromatin seen preferentially in SAT or osteoblasts at TBX15 intron-1 or upstream. They overlap strongly predicted, allele-specific transcription factor binding sites. Our analysis suggests that these SNPs act independently of two missense SNPs in TBX15. Remarkably, five of the regulatory SNPs were associated with eBMD and obesity and had the same trait-increasing allele for both. We found that WARS2 obesity-related SNPs can be ascribed to high linkage disequilibrium with TBX15 intron-1 SNPs. Our findings from GWAS index, proxy, and imputed SNPs suggest that a few SNPs, including three in a 0.7-kb cluster, act as causal regulatory variants to fine-tune TBX15 expression and, thereby, affect both obesity and osteoporosis risk.
