Abstract Real-time and accurate guidance for tumor resection has long been anticipated by surgeons. In the past decade, the flourishing material science has made impressive progress in near-infrared fluorophores that may fulfill this purpose. Fluorescence imaging-guided surgery shows great promise for clinical application and has undergone widespread evaluations, though it still requires continuous improvements to transition this technique from bench to bedside. Concurrently, the rapid progress of artificial intelligence (AI) has revolutionized medicine, aiding in the screening, diagnosis, and treatment of human doctors. Incorporating AI helps enhance fluorescence imaging and is poised to bring major innovations to surgical guidance, thereby realizing precision cancer surgery. This review provides an overview of the principles and clinical evaluations of fluorescence-guided surgery. Furthermore, recent endeavors to synergize AI with fluorescence imaging were presented, and the benefits of this interdisciplinary convergence were discussed. Finally, several implementation strategies to overcome technical hurdles were proposed to encourage and inspire future research to expedite the clinical application of these revolutionary technologies.

Abstract. Background. This study aimed to explore the patterns of treatment-related adverse events (AEs) associated with immune checkpoint inhibitor (ICI) monotherapy and in combination with chemotherapy or tyrosine kinase inhibitor (TKI) therapy and to summarize the corresponding management and nursing strategies. Materials and methods. A total of 69 patients with malignant urological tumors who received ICI treatment between June 2019 and October 2022 were retrospectively analyzed, and AEs that occurred during treatment were observed and reported. Based on the different types of treatment, the patients were divided into ICI monotherapy, ICI plus chemotherapy, and ICI plus TKI therapy groups. Subgroup analysis was performed. The incidence, distribution, and severity of AEs in the different subgroups were evaluated. Results. A total of 138 AEs occurred in 69 patients, among which grade 1 plus 2, and grade 3 plus 4 AEs accounted for 78.99% and 21.01%, respectively. The incidence of AEs per patient in the ICI-TKI therapy group was the highest (3.75 times/person), followed by the ICI-chemotherapy (2.33 times/person) and ICI monotherapy (0.82 times/person) groups. Specific AEs, such as fatigue, nausea, and myelosuppression, were much more common in the ICI-gemcitabine and cisplatin group, whereas renal injury, skin lesions, and diarrhea were most common ones in the ICI-TKI group. Conclusions. Immune checkpoint inhibitors are new treatment options for advanced urological tumors and renal cell carcinoma. Distinctive AE patterns were observed among the different treatment groups. Therefore, strict and meticulous clinical management and nursing measures are required to ensure the safety of patients receiving ICI treatment.

Summary: Examining nasal mucosa samples is crucial for nasal cavity disease research and diagnosis. Simultaneously obtaining high-quality data for single-cell transcriptomics (single-cell RNA sequencing [scRNA-seq]) and epigenomics (single-cell assay for transposase-accessible chromatin using sequencing [scATAC-seq]) of nasal mucosa tissues is challenging. Here, we present a protocol for processing human nasal mucosa samples to obtain data for both scRNA-seq and scATAC-seq. We describe steps for extracting human nasal mucosa tissue, mechanical and enzymatic dissociation, lysis of red blood cells, and a viability assay. The European sweet cherry Prunus avium (L.), a member of the Rosaceae family, is one of the most popular and economically valuable fruits. However, the rapid spread of gummosis and poor management practices have become the major obstacles to their production. To identify pathogenic microorganisms responsible for gummosis disease, we conducted observations comparing the garden of Bailuyuan, which heavily suffered from gummosis disease and horn beetle damage, with the orchard of Mayuhe, which only suffered from gummosis disease, both from Xi’an, Shaanxi, China. Samples were obtained from the healthy tissues and gummosis disease tissues that used the Illumina sequence of 16S rRNA and the internal transcribed spacer region (ITS) to identify bacterial and fungal communities in these samples. An alpha diversity analysis revealed a significantly higher fungal diversity of disease than in healthy tissue in the gummosis period. The results suggested that an imbalance in the fungal genera may be associated with gummosis disease. Species relative analyses showed some bacterial genera (Pelagibacterium, Halomonas, Azospirillum, Aquabacterium and Alistipes) and fungal genera (Penicillium, Alternaria and Rhodotorula) in the diseased tissues of gummosis. Among these, the increased relative abundance of the bacteria genes Halomonas, Pelagibacterium, Chelativorans, Pantoea, Aquabacterium, Alternaria and fungi genes Penicillium, Cystobasidium, Rhodotorula may be associated with gummosis of P. avium. The bacterial genera Methylobacterium, Psychroglaciecola, Aeromonas, Conexibacter and fungal genera Didymella, Aureobasidium, Mycosphaerella, Meyerozyma are probably antagonists of the pathogen of gummosis. These findings are an initial step in the identification of potential candidates for the biological control of the disease.

Dysfunction of the vascular angiocrine system is critically involved in regenerative defects and fibrosis of injured organs. Previous studies have identified various angiocrine factors and found that risk factors such as aging and metabolic disorders can disturb the vascular angiocrine system in fibrotic organs. One existing key gap is what sense the fibrotic risk to modulate the vascular angiocrine system in organ fibrosis. Here, using human and mouse data, we discovered that the metabolic pathway hydrogen sulfide (H2S)-AMP-activated protein kinase (AMPK) is a sensor of fibrotic stress and serves as a key mechanism upregulating the angiocrine factor plasminogen activator inhibitor-1 (PAI-1) in endothelial cells to participate in lung fibrosis. Activation of the metabolic sensor AMPK was inhibited in endothelial cells of fibrotic lungs, and AMPK inactivation was correlated with enriched fibrotic signature and reduced lung functions in humans. The inactivation of endothelial AMPK accelerated lung fibrosis in mice, while the activation of endothelial AMPK with metformin alleviated lung fibrosis. In fibrotic lungs, endothelial AMPK inactivation led to YAP activation and overexpression of the angiocrine factor PAI-1, which was positively correlated with the fibrotic signature in human fibrotic lungs and inhibition of PAI-1 with Tiplaxtinin mitigated lung fibrosis. Further study identified that the deficiency of the antioxidative gas metabolite H2S accounted for the inactivation of AMPK and activation of YAP-PAI-1 signaling in endothelial cells of fibrotic lungs. H2S deficiency was involved in human lung fibrosis and H2S supplement reversed mouse lung fibrosis in an endothelial AMPK-dependent manner. These findings provide new insight into the mechanism underlying the deregulation of the vascular angiocrine system in fibrotic organs.

PurposeTo assess tumor growth using tumor doubling rate (TDR) during active surveillance (AS) in China.MethodsBetween January 2016 and June 2020, a total of 219 patients with low-risk papillary thyroid microcarcinoma (PTMC) (aged 23-75 years) were consecutively enrolled in the AS program.ResultsFour sections of TDR, >0.5, 0.1~0.5, -0.1~0.1 and 179.5 mm3), the proportion of non-growth was 68.75%.ConclusionTDR may be a better metric for evaluating tumor growth in observational PTMCs. A certain proportion of PTMCs grow during the period of AS and tumor growth was associated with age and volume of PTMC at initial diagnosis. Therefore, how to block tumor growth during the AS period, especially for young patients and patients with early-stage PTMC (size ≤ 5 mm), will be a new challenge.

Abstract Fine-particle pollution associated with high sulfate and secondary organic aerosol (SOA) contents still occurs in winter, despite considerable reductions in precursor emissions in China. The chemical mechanisms involved are consistently acknowledged to be linked with aerosol water but remain poorly understood. Here, we present findings demonstrating that the synergistic effect of elevated aerosol water content and particles originating from fossil fuel combustion significantly enhanced the rapid formation of sulfate and SOAs under high humidity conditions during winter in northern China. The presence of high aerosol water content promoted the aging process of particles containing elemental carbon (EC) and facilitated the formation of SOAs. Evidence from single particles reveals an increased potential for SOA formation from primary organic aerosols (POAs) originating from fossil fuel combustion, in the presence of increasing aerosol water. Under conditions of relative humidity exceeding 85%, the multiphase oxidation of SO2 by in-particle nitrite/nitrous acid, generated through reactions involving aged EC-containing and iron-rich particles, as well as aqueous-phase formation of hydroxymethanesulfonate (HMS), led to a substantial increase in sulfate production. This study emphasizes the critical role of aerosol water in the aerosol chemistry concerning sulfate and SOAs, highlighting the urgent necessity to mitigate primary emissions from fossil fuel combustion.

This study investigates the distribution, morphology, and potential functions of antennal sensilla in various wasp species, including Dolichovespula flora, D. intermedia, Vespula structor, Vl. vulgaris, Provespa barthelemyi, Vespa bicolor, V. ducalis, V. mocsaryana, and V. velutina var. nigothorax. The study thoroughly analyzes the antennal structure of these species, representing all four genera of the yellow-jacket and hornet subfamily Vespinae. Using scanning electron microscopy (SEM), the study identifies a total of nineteen types of sensilla, including sensilla trichodea (ST-I, ST-II, ST-III), sensilla campaniform (SCF-I, SCF-II, SCF-III), pit organs (SCO-I, SCO-II, and SA), sensilla placodea (SP-I, SP-II), sensilla chaetica (SCH-I, SCH-II), sensilla basiconica (SB-I, SB-II), sensilla agmon (SAG-I, SAG-II), and sensilla coelocapitular (SCA). Additionally, tyloids were observed in the males of seven species, except for Vl. structor and Vl. vulgaris. The study provides insights into these sensilla types’ morphology, abundance, and distribution. It discusses the variations in sensilla morphology among different species and the presence of gender-specific sensilla. This study provides new data about the morphology and distribution patterns of sensilla and tyloid.

The phototactic behavior of insects is commonly used to manage pest populations in practical production. However, this elusive behavior is not yet fully understood. Investigating whether the opsin genes play a crucial role in phototaxis is an intriguing topic. Vespinae (Hymenoptera: Vespidae) are a common group of social wasps that are closely associated with human activities. Efficiently controlling wasp populations while maintaining ecological balance is a pressing global challenge that still has to be resolved. This research aims to explore the phototactic behavior and key opsin genes associated with Vespinae. We found significant differences in the photophilic rates of Vespula germanica and Vespa analis under 14 different light conditions, indicating that their phototactic behavior is rhythmic. The results also showed that the two species exhibited varying photophilic rates under different wavelengths of light, suggesting that light wavelength significantly affects their phototactic behavior. Additionally, the opsin genes of the most aggressive hornet, Vespa basalis, have been sequenced. There are only two opsin genes, one for UV light and the other for blue light, and Vespa basalis lacks long-wavelength visual proteins. However, they exhibit peak phototaxis for long-wavelength light and instead have the lowest phototaxis for UV light. This suggests that the visual protein genes have a complex regulatory mechanism for phototactic behavior in Vespinae. Additionally, visual protein sequences have a high degree of homology among Hymenoptera. Despite the hypotheses put forward by some scholars regarding phototaxis, a clear and complete explanation of insect phototaxis is still lacking to date. Our findings provide a strong theoretical basis for further investigation of visual expression patterns and phototactic mechanisms in Vespinae.

Abstract Vanadium nitride (VN) electrode displays high‐rate, pseudocapacitive responses in aqueous electrolytes, however, it remains largely unclear in nonaqueous, Na+‐based electrolytes. The traditional view supposes a conversion‐type mechanism for Na+ storage in VN anodes but does not explain the phenomena of their size‐dependent specific capacities and underlying causes of pseudocapacitive charge storage behaviors. Herein, we insightfully reveal the VN anode exhibits a surface‐redox pseudocapacitive mechanism in nonaqueous, Na+‐based electrolytes, as demonstrated by kinetics analysis, experimental observations, and first‐principles calculations. Through ex situ X‐ray photoelectron spectroscopy and semiquantitative analyses, the Na+ storage is characterized by redox reactions occurring with the V5+/V4+ to V3+ at the surface of VN particles, which is different from the well‐known conversion reaction mechanism. The pseudocapacitive performance is enhanced through nanoarchitecture design via oxidized vanadium states at the surface. The optimized VN‐10 nm anode delivers a sodium‐ion storage capability of 106 mAh g−1 at the high specific current of 20 A g−1, and excellent cycling performance of 5000 cycles with negligible capacity losses. This work demonstrates the emerging opportunities of utilizing pseudocapacitive charge storage for realizing high‐rate sodium‐ion storage applications.

Hepatocellular carcinoma is one of the leading cancers worldwide and is a potential consequence of fibrosis. Therefore, the identification of key cellular and molecular mechanisms involved in liver fibrosis is an important goal for the development of new strategies to control liver-related diseases. Here, single-cell RNA sequencing data (GSE136103 and GES181483) of clinical liver non-parenchymal cells were analyzed to identify cellular and molecular mechanisms of liver fibrosis. The proportion of endothelial subpopulations in cirrhotic livers was significantly higher than that in healthy livers. Gene ontology and gene set enrichment analysis of differentially expressed genes in the endothelial subgroups revealed that extracellular matrix (ECM)-related pathways were significantly enriched. Since anthrax toxin receptor 2 (ANTXR2) interacts with the ECM, the expression of ANTXR2 in the liver endothelium was analyzed. ANTXR2 expression in the liver endothelium of wild-type (WT) mice significantly decreased after a 4-time sequential injection of carbon tetrachloride (CCl4) to induce liver fibrosis. Next, conditional knockout mice selectively lacking Antxr2 in endothelial cells were generated. After endothelial-specific Antxr2 knockout mice were subjected to the CCl4 model, the degree of liver fibrosis in the knockout group was significantly more severe than that in the control group. In addition, ANTXR2 in human umbilical vein endothelial cells promoted matrix metalloproteinase 2 (MMP2) activation to degrade the ECM in vitro. Finally, endothelial-specific overexpression of Antxr2 alleviated the development of liver fibrosis following adeno-associated virus treatment. Collectively, these results suggested that endothelial ANTXR2 plays a protective role in liver fibrosis. This function of ANTXR2 may be achieved by promoting MMP2 activation to degrade the ECM.

Due to the good conductivity and adjustable mechanical properties, ionic conductive hydrogel is considered an ideal candidate for use in wearable electronic devices. However, maintaining good flexibility, conductivity, and high sensitivity of electromechanical performance, though highly desirable, remains a great challenge. In this work, a zwitterionic ionic conductive hydrogel (CMHZ: chitosan/polyacrylic acid-acrylamide/ZnSO4) was prepared with excellent mechanical properties, ionic conductivity, and high sensitivity. The CMHZ showed excellent adjusted mechanical properties (up to 1210% tensile strain under 0.15 MPa, toughness 5.42 MJ/m3) and good shape-recovery ability. Meanwhile, the free-moving metal ions in the hydrogel guaranteed the CMHZ with good conductivity (33.73 mS/cm). More importantly, the unique zwitterionic structure made the hydrogel have a wide working voltage range (0 - 2.0 V), which endowed the hydrogel with a significant improvement of sensitivity up to GF = 4.22. Based on these obvious advantages, the CMHZ hydrogel was used as a stretchable sensor demonstrated high-sensitivity physiological signal detection and motion direction recognition, suggesting wide potential in flexible electronic devices.

Abstract Cancer cell membrane (CCM) derived nanotechnology functionalizes nanoparticles (NPs) to recognize homologous cells, exhibiting translational potential in accurate tumor therapy. However, these nanoplatforms are majorly generated from fixed cell lines and are typically evaluated in cell line-derived subcutaneous-xenografts (CDX), ignoring the tumor heterogeneity and differentiation from inter- and intra- individuals and microenvironments between heterotopic- and orthotopic-tumors, limiting the therapeutic efficiency of such nanoplatforms. Herein, various biomimetic nanoplatforms (CCM-modified gold@Carbon, i.e., Au@C-CCM) were fabricated by coating CCMs of head and neck squamous cell carcinoma (HNSCC) cell lines and patient-derived cells on the surface of Au@C NP. The generated Au@C-CCMs were evaluated on corresponding CDX, tongue orthotopic xenograft (TOX), immune-competent primary and distant tumor models, and patient-derived xenograft (PDX) models. The Au@C-CCM generates a photothermal conversion efficiency up to 44.2% for primary HNSCC therapy and induced immunotherapy to inhibit metastasis via photothermal therapy-induced immunogenic cell death. The homologous CCM endowed the nanoplatforms with optimal targeting properties for the highest therapeutic efficiency, far above those with mismatched CCMs, resulting in distinct tumor ablation and tumor growth inhibition in all four models. This work reinforces the feasibility of biomimetic NPs combining modular designed CMs and functional cores for customized treatment of HNSCC, can be further extended to other malignant tumors therapy.

Flight is a complex physiological process requiring precise coordination of muscular contraction. A key protein in insect flight is flightin, which plays an integral role in the flight muscles. This research sought to evaluate the flight competence of the social wasp V. basalis by characterizing the molecular components involved. Our study focused on Vespa basalis, one of the most dangerous hornet species, utilizing PCR to obtain a partial cDNA sequence of the flightin protein. We then employed phylogenetic and sequence analysis to gain insights into this protein in flight-related adaptations. The cDNA has an 1189-base pair sequence including an open reading frame (453 bp) encoding 150 amino acids. Analyzing the deduced amino acid sequence using an online tool revealed a molecular weight of 18.05 kDa, an isoelectric point of 5.84, four functional site patterns, and no transmembrane topology. We constructed a phylogenetic tree of flightin based on 38 species. Our analysis indicated that V. basalis is most closely related to V. mandarinia; this alignment is consistent with their similar aggressive behavior, but their evolutionary relationship, based on mitochondrial sequences, presents a contrast. These initial findings on the flightin gene in V. basalis lay the groundwork for future functional studies to elucidate its specific role in flight adaptations and explore its potential as a target for pest management strategies.

Abstract Surface‐redox pseudocapacitive nanomaterials show promise for fast‐charging energy storage. However, their high surface area usually leads to low density, which is not conducive to achieving both high volumetric capacity and high‐rate capability. Herein, we demonstrate that TiO2 nanosheets (meso‐TiO2‐NSs) with densely packed mesoporous are capable of fast pseudocapacitance‐dominated sodium‐ion storage, as well as high volumetric and gravimetric capacities. Through compressing treatment, the compaction density of meso‐TiO2‐NSs is up to ~1.6 g/cm3, combined with high surface area and high porosity with mesopore channels for rapid Na+ diffusion. The compacted meso‐TiO2‐NSs electrodes achieve high pseudocapacitance (93.6% of total charge at 1 mV/s), high‐rate capability (up to 10 A/g), and long‐term cycling stability (10,000 cycles). More importantly, the space‐efficiently packed structure enables high volumetric capacity. The thick‐film meso‐TiO2‐NSs anode with the mass loading of 10 mg/cm2 delivers a gravimetric capacity of 165 mAh/g and a volumetric capacity of 223 mAh/cm3 at 5 mA/cm2, much higher than those of commercial hard carbon anode (80 mAh/g and 86 mAh/cm3). This work highlights a pathway for designing a dense nanostructure that enables fast charge kinetics for high‐density sodium‐ion storage.

Self-collection of saliva samples has attracted considerable attention in recent years, particularly during the coronavirus disease 2019 pandemic. However, studies investigating the detection of other common respiratory pathogens in saliva samples are limited. In this study, nasopharyngeal swabs (NPS), oropharyngeal swabs (OPS), and “hock-a-loogie” saliva (HLS) were collected from 469 patients to detect 13 common respiratory pathogens. Overall positivity rates for NPS (66.1 %), HLS (63.5 %), and OPS (57.8 %) were statistically different (P = 0.028), with an overall concordance of 72.7 %. Additionally, detection rates for NPS (85.9 %) and HLS (83.2 %) for all pathogens were much higher than for OPS (73.3 %). Pseudocapacitive materials with surface‐redox reactions are capable of realizing high capacities at ultrahigh rates; however, it remains a challenge in the synthesis of active components with high surface area to boost surface‐redox sodiation but restrain side reactions. Herein, a two‐step, topochemical synthesis of 2D mesoporous TiN (2D‐meso‐TiN) with high surface area and rich mesoporosities is presented. It is demonstrated that the sodium‐ion storage mechanism of TiN anode is based on the existence of surficial titanium oxides via redox reactions between Ti4+ and Ti3+. The interconnected, highly conductive 2D‐meso‐TiN with high surface area largely increases the pseudocapacitive capacities, leading to a high capacity of 160/93 mAh g−1 at 0.1/10 A g−1, which is much higher than 2D‐TiN (120/72 mAh g−1) and commercial TiN nanoparticles (57/30 mAh g−1). The surface‐redox (de)sodiation undergoes no destruction of crystalline TiN, which enables high initial coulombic efficiency and long‐term cycles. Furthermore, a novel hybrid sodium‐ion capacitor consisting of 2D‐meso‐TiN anode and Na3V2(PO4)3 cathode is assembled without any presodiation treatments. The hybrid capacitor delivers both high energy density (94 Wh kg−1 at 64 W kg−1) and high power density (38 Wh kg−1 at 4.4 kW kg−1), as well as long cycling stability.

Immunotherapy has revolutionized cancer treatment. B7-H3 is a promising target for cancer immunotherapy (CI). The present study aimed to utilize bibliometric methods to assess the current research status and explore future trends in the use of B7-H3 for CI. We collected publications related to B7-H3/CI from the Clarivate Web of Science Core Collection database. VOSviewer, Microsoft Excel, the bibliometrix R package, and an online platform were used to conduct qualitative and visualized analyses of the literature. A total of 555 papers were analyzed, revealing a significant increase in annual publications since 2018. The most productive countries were China and the USA, and the leading institutions were Soochow University and Sichuan University. Zang and Ferrone were the most popular authors. Among the journals, Frontiers in Immunology had the highest number of papers, whereas Clinical Cancer Research was the most influential. Historical citation analysis reveals the development of B7-H3/CI. Top-cited papers and keyword analyses were performed to highlight current hotspots in the domain. Using cluster analysis, we classified all keywords into four clusters: “immunotherapy,” “co-stimulatory molecule,” “B7 family,” and “PD-L1.” Finally, Trends analysis suggested that future research might focus on “chimeric antigen receptor,” “pathways,” and “targeting B7-H3.” This is the first bibliometric crosstalk analysis between B7-H3 and CI. Our study illustrates that the topic of B7-H3/CI is very popular and has great clinical implications and that the number of correlative publications will continue to increase. B7-H3-based CI may lead to new research trends.

China experienced severe haze pollution in the past decades. However, systematic characterization of atmospheric fine particles with advanced measurement techniques has been very scarce in Shanghai, which is the largest megacity in China. Herein, we present the characterization of non-refractory submicron aerosol (NR-PM1) in urban Shanghai during winter 2017 by applying an Aerosol Chemical Speciation Monitor (ACSM, Aerodyne Research Inc.). The NR-PM1 is predominated by organics (43%), followed by nitrate (29%), sulfate (18%), ammonium (7%), and chloride (3%). Regarding the organic aerosol (OA) factors, three organic aerosol factors were identified and classified as hydrocarbon-like organic aerosol (HOA), less-oxygenated organic aerosol (LO-OOA), and more-oxygenated organic aerosol (MO-OOA), which contributed 32%, 22%, and 46% to total organic aerosol respectively. The NR-PM1 composition exhibited an increase in secondary inorganic aerosol (SIA) (sulfate, nitrate and ammonium) contribution from clean episodes (49%) to pollution episodes (59%), while organic aerosol contribution decreased accordingly from 48% to 39%. The strong increase of sulfate in high-relative humidity (RH) pollution episodes indicated that aqueous-phase oxidation of SO2 could be an important formation process for sulfate during particulate air pollution period. The contribution of nitrate was elevated from 25% during clean episodes to 32% during polluted episodes, likely owing to the increase of relative humidity which facilitates the hydrolysis of N2O5 and the gas-to-particle partitioning of hydrophilic NH4NO3 and. Further analysis of atmospheric formation relevance suggested that less-oxygenated organic aerosol formation was mainly driven by aqueous-phase chemistry reactions, whereas photochemical oxidation became an important process for more-oxygenated organic aerosol formation. Meanwhile, less-oxygenated organic aerosol formation may also be influenced by atmospheric oxidative tracer (i.e., Ox), as less-oxygenated organic aerosol exhibited a distinct peak at noon under high-relative humidity condition.

Accurate quantification of terrestrial gross primary production (GPP) is integral for enhancing our understanding of the global carbon budget and climate change. The light use efficiency (LUE) model is undoubtedly the most extensively applied method for GPP estimation. However, the two-leaf (TL)-LUE model using a ‘potential’ sunlit leaf area index (LAI _su ) can separate a portion of LAI _su even when the canopy does not receive any direct radiation, leading to the underestimation of GPP under cloudy and overcast days. Here, we developed a dynamic-leaf (DL) LUE model by introducing an ‘effective’ LAI _su to improve GPP estimation, which considers the comprehensive contribution of LAI _su when the canopy does and does not receive direct radiation. In particular, the new model decreases LAI _su to zero when direct radiation reaches zero. Our evaluation at eight ChinaFLUX sites showed that (1) the DL-LUE model outperformed the most well-known BL-LUE (namely, the MOD17 GPP algorithm) and TL-LUE models in reproducing the daily in situ GPP, especially at four forest sites [reducing the root mean square error (RMSE) from 1.74 g C m ^−2 d ^−1 and 1.53 g C m ^−2 d ^−1 to 1.36 g C m ^−2 d ^−1 and increasing the coefficient of determination ( R ^2 ) from 0.74 and 0.79–0.82, respectively]. Moreover, the improvements were particularly pronounced at longer temporal scales, as indicated by the RMSE decreasing from 29.32 g C m ^−2 month ^−1 and28.11 g C m ^−2 month ^−1 to 25.81 g C m ^−2 month ^−1 at a monthly scale and from 231.82 g C m ^−2 yr ^−1 and 221.60 g C m ^−2 yr ^−1 –200.00 g C m ^−2 yr ^−1 at a yearly scale; (2) the DL-LUE model mitigated the systematic underestimation of the in situ GPP by both the TL-LUE and BL-LUE models when the clearness index (CI) was below 0.5, as indicated by the Bias reductions of 0.25 g C m ^−2 d ^−1 and 0.46 g C m ^−2 d ^−1 , respectively; and (3) the contributions of the shaded GPP to the total GPP from the DL-LUE model were higher by 0.07–0.16 than those from the TL-LUE model across the eight ChinaFLUX sites. The proposed parsimonious and effective DL-LUE model not only has great potential for improving global GPP estimations but also provides a more mechanism-based approach for partitioning the total GPP into its shaded and sunlit components.

Background: Small intestinal neuroendocrine tumors (SI-NETs) are the most common malignant tumors of the small intestine, with many patients presenting with metastases and their incidence increasing. We aimed to find effective diagnostic biomarkers for patients with primary and metastatic SI-NETs that could be applied for clinical diagnosis.Methods: We downloaded GSE65286 (training set) and GSE98894 (test set) from the GEO database and performed differential gene expression analysis to obtain differentially expressed genes (DEGs) and differentially expressed long non-coding RNAs (DElncRNAs). The functions and pathways involved in these genes were further explored by Gene Ontology (GO) and Kyoto Encyclopedia of Genes and Genomes (KEGG) enrichment analyses. In addition, a global regulatory network involving dysregulated genes in SI-NETs was constructed based on RNAInter and TRRUST v2 databases, and the diagnostic power of hub genes was identified by receiver operating characteristic curve (ROC).Results: A total of 2,969 DEGs and DElncRNAs were obtained in the training set. Enrichment analysis revealed that biological processes (BPs) and KEGG pathways were mainly associated with cancer. Based on gene set enrichment analysis (GSEA), we obtained five BPs (cytokinesis, iron ion homeostasis, mucopolysaccharide metabolic process, platelet degranulation and triglyceride metabolic process) and one KEGG pathway (ppar signaling pathway). In addition, the core set of dysregulated genes obtained included MYL9, ITGV8, FGF2, FZD7, and FLNC. The hub genes were upregulated in patients with primary SI-NETs compared to patients with metastatic SI-NETs, which is consistent with the training set. Significantly, the results of ROC analysis showed that the diagnostic power of the hub genes was strong in both the training and test sets.Conclusion: In summary, we constructed a global regulatory network in SI-NETs. In addition, we obtained the hub genes including MYL9, ITGV8, FGF2, FZD7, and FLNC, which may be useful for the diagnosis of patients with primary and metastatic SI-NETs.

Abstract Background Photodynamic therapy (PDT) is a promising antitumor strategy with fewer adverse effects and higher selectivity than conventional therapies. Recently, a series of reports have suggested that PDT induced by Cerenkov radiation (CR) (CR-PDT) has deeper tissue penetration than traditional PDT; however, the strategy of coupling radionuclides with photosensitizers may cause severe side effects. Methods We designed tumor-targeting nanoparticles (131I-EM@ALA) by loading 5-aminolevulinic acid (ALA) into an 131I-labeled exosome mimetic (EM) to achieve combined antitumor therapy. In addition to playing a radiotherapeutic role, 131I served as an internal light source for the Cerenkov radiation (CR). Results The drug-loaded nanoparticles effectively targeted tumors as confirmed by confocal imaging, flow cytometry, and small animal fluorescence imaging. In vitro and in vivo experiments demonstrated that 131I-EM@ALA produced a promising antitumor effect through the synergy of radiotherapy and CR-PDT. The nanoparticles killed tumor cells by inducing DNA damage and activating the lysosome-mitochondrial pathways. No obvious abnormalities in the hematology analyses, blood biochemistry, or histological examinations were observed during the treatment. Conclusions We successfully engineered a nanocarrier coloaded with the radionuclide 131I and a photosensitizer precursor for combined radiotherapy and PDT for the treatment of breast cancer. Soil moisture (SM) affects ecosystem homeostasis through interactions among land use type (LUT), precipitation, heat and human activity (HA). In the core areas of China Loess Plateau, we selected three sites along gradients of annual precipitation and annual accumulated temperature ≥ 0℃ and compare SM variations among three LUTs, including cropland, woodland, and rangeland, and in three precipitation years, including dry, wet, and normal year. The SM of three LUTs were sensitive to wet years in the north, dry years in the south, and both wet and dry years in the middle, respectively. From north to south, water use (WU) increased while water use efficiency (WUE) decreased. The WU for cropland was the highest and WUE was the lowest, while rangeland had the opposite tendency. The mean annual water deficiencies for cropland, rangeland and woodland were 42.41 mm, 21.38 mm, and 15.49 mm, respectively. HA for three LUTs increased as precipitation increased and the order was cropland > woodland > rangeland. The stability of SM to precipitation was as following order: rangeland > woodland > cropland. Rangeland was insensitive to precipitation, while cropland was sensitive to precipitation. Productivity of rangeland and cropland were stabilized mainly by increasing resistance to SM and the resilience to SM, respectively. To maximize the WUE of the rain fed system and enhance the stability of SM, the area ratios of cropland, woodland, and rangeland were recommended as 4:2:19, 3:2:5 and 3:2:4 in the northern, central, and southern regions, respectively.

Abstract Background Inflammation markers have an important effect on tumor proliferation, invasion, and metastasis. Oligometastatic disease (OMD) is an intermediate state between widespread metastases and locally confined disease, where curative strategies may be effective for some patients. We aimed to explore the predictive value of inflammatory markers in patients with oligometastatic colorectal cancer (OMCC) and build a nomogram to predict the prognosis of these patients. Methods Two hundred nine patients with OMCC were retrospectively collected in this study. The Kaplan-Meier survival curves and Cox regression analysis were used to estimate overall survival (OS) and progression-free survival (PFS). A multivariate Cox analysis model was utilized to establish the nomogram. The concordance index (C-index), calibration curve, and receiver operating characteristics (ROC) were established to verify the validity and accuracy of the prediction model. Results According to the multivariate analysis, decreased platelet-to-lymphocyte ratio (PLR) might independently improve OS in patients with OMCC (HR = 2.396, 95% CI 1.391–4.126, P = 0.002). Metastases of extra-regional lymph nodes indicated poor OS (HR = 2.472, 95% CI 1.247–4.903, P = 0.010). While the patients with early N stage had better OS (HR = 4.602, 95% CI 2.055–10.305, P = 0.001) and PFS (HR = 2.100, 95% CI 1.364–3.231, P = 0.007). Primary tumor resection (HR = 0.367, 95% CI 0.148–0.908, P = 0.030) and lower fibrinogen (HR = 2.254, 95% CI 1.246–4.078, P = 0.007) could significantly prolong the OS in patients with OMCC. PLR, metastases of extra-regional lymph nodes, N stage, primary tumor resection, and fibrinogen were used to make up the nomogram. The C-index and area under the curve (AUC) of the ROC in nomogram were 0.721 and 0.772 respectively for OS, showed good consistency between predictive probability of OS and actual survival. Conclusions Decreased PLR could predict a good prognosis in patients with OMCC. The nomogram including inflammatory factors and clinicopathological markers was credible and accurate to predict survivals in patients with OMCC.

Abstract This study investigates the protective effect of Erigeron breviscapus injection, a classic traditional Chinese medicine most typically used by Chinese minority to treat stroke, on cerebral ischemia–reperfusion injury and the related signaling pathways. Use network pharmacology methods to study the relationship between E. breviscapus (Vant.) Hand-Mazz. and ischemic stroke, predict the mechanism and active ingredients of E. breviscapus (Vant.) Hand-Mazz. in improving ischemic stroke disease. We study the protective effect of E. breviscapus injection on blood–brain barrier (BBB) injuries induced by cerebral ischemia in rats by regulating the ROS/RNS-MMPs-TJs signaling pathway. The rat model of focal cerebral ischemia–reperfusion injury has been prepared using the wire-suppository method. Firstly, the efficacy of E. breviscapus injection, Scutellarin and 3,5-dicaffeoylquinic acid in protecting BBB injury caused by cerebral ischemia has been evaluated. Secondly, the following two methods have been used to study the mechanism of E. breviscapus injection in regulating the ROS/RNS-MMPS-TJS signaling pathway: real-time PCR and western blot for the determination of iNOS, MMP-9, claudin-5, occludin, ZO-1 mRNA and protein expression in brain tissue. We find that PI3K-Akt signaling pathway predicted by network pharmaology affects the blood–brain barrier function, so we chose the blood–brain barrier-related MMP-9, claudin-5, iNOS, occludin and ZO-1 proteins are used for research. The results of our research show that 3 drugs can reduce the rate of cerebral infarction in rats, relieve the abnormal neuroethology of rats, reduce the degree of brain tissue lesion, increase the number of the Nissl corpuscle cells and repair the neuron ultrastructure in injured rats. At the same time, it can obviously reduce the ultrastructure damage of the BBB in rats. All three drugs significantly reduced the content of Evans blue in the ischemic brain tissue caused by cerebral ischemia in rats with BBB injury. In addition, E. breviscapus injection, Scutellarin and 3,5-dicaffeoylquinic acid can decrease the protein expression of iNOS and MMP-9 in rat ischemic brain tissue. In addition, 3,5-dicaffeoylquinic acid can increase the protein expression of claudin-5. We conclude that E. breviscapus injection, Scutellarin and 3,5-dicaffeoylquinic acid have obvious therapeutic effects on BBB and neuron injury induced by cerebral ischemia in rats. Our results from studying the mechanism of action show that E. breviscapus injection and Scutellarin inhibited the activation of MMP-9 by inhibiting the synthesis of iNOS, 3,5-dicaffeoylquinic acid inhibits the expression and activation of MMP-9 by inhibiting the activation of iNOS and reducing the generation of free radicals, thus reducing the degradation of important cytoskeleton connexin claudin-5 in the tight junction (TJ) structure by inhibiting the expression and activation of MMP-9. Finally BBB structure integrity was protected.

Vegetation gross primary production (GPP) in terrestrial ecosystems is a key element of the carbon cycle, and its estimation highly determines the accuracy of carbon budget assessments. Currently, several global datasets of vegetation production are available at coarse or moderate spatial resolutions, but globally they still have large uncertainties, which hindered the application of GPP, especially in strongly heterogeneous agriculture ecosystems and mountainous areas. Here, we used the Markov chain Monte Carlo (MCMC) approach with the footprints of FLUXNET data to optimize the parameters of the high resolution of Global LAnd Surface Satellite (Hi-GLASS) GPP algorithm (i.e., the revised EC-LUE model) using 30 m spatial resolution Landsat data as driving data. Then, we generated a new set of algorithm parameters for high resolution GPP estimates. We used the optimized parameters with integrating footprint to calculate Hi-GLASS GPP based on Landsat data and our results revealed that on average, Hi-GLASS GPP explained 76% of variance in tower GPP at the total of 78 sites across ten vegetation types. Moreover, compared with previous 500 m GPP product such as GLASS GPP and MODerate Resolution Imaging Spectroradiometer (MODIS) GPP, our optimized Hi-GLASS algorithm using Landsat data had large superiority in simulating GPP for wetlands, savannas, shrubland and C3, C4 cropland ecosystems, and had a slightly improvement for deciduous broadleaf forests and evergreen broadleaf forest ecosystem. Our study is an effort to optimize and quantify parameter uncertainty of Hi-GLASS algorithm using high spatial resolution (30 m) Landsat data and improve the high resolution GPP estimation for better understanding global ecosystem carbon dynamics and carbon-climate feedbacks.

Abstract Background There was little known on how the interaction effect between obesity and current smoking affected the incidence of hypertension. The aim of this study was to investigate how body mass index (BMI) modified the effect of current smoking on the incidence of hypertension. Methods Data were obtained from the China Health and Nutrition Survey (CHNS). According to the WHO recommendations for Chinese people, the normal weight, overweight, and obesity were defined using the BMI cutoff values 18.5 kg/m2, 23.0 kg/m2, and 27.5 kg/m, respectively. Current smokers were defined as having smoked at least 100 cigarettes or electronic cigarettes, 20 cigars, or 20 tobacco pipes and other type of tobacco in the last 30 days preceding the survey. Hypertension was defined as systolic blood pressure (SBP)/ diastolic blood pressure (DBP) ≥ 140/90 mmHg, use of anti-hypertensive medications, or a self-reported diagnosis. Results This study included 12,900 subjects. There were interaction effects between obesity and current smoking in females (P = 0.030) and the 50–59 years group (P = 0.049). Current smoking was a significant predictor of incident hypertension only in the total and female populations with normal weight (HR: 1.119 and 1.274; HR 95% CI: 1.013–1.236 and 1.143–1.415; and P = 0.027 and 0.040, respectively). Stratified by age, current smoking affected the development of hypertension only in the 50–59 years subjects with the normal weight (HR: 1.356; HR 95% CI: 1.084–1.697; and P = 0.008). Conclusions Current smoking was a significant predictor of incident hypertension only in the female and middle-age populations with normal weight but not in the overweight and obesity as well as the younger and elder populations.

Land surface temperature (LST) is an essential parameter in land–atmosphere interaction. However, it is still poorly understood about the effects of seasonal LST on the interannual LST change and the dominant driving force for the variation in LST. In this study, trends of time-series LST were analyzed by using both linear and nonlinear methods. Two indices were developed to evaluate the effects of seasonal LST trends on interannual LST change. The main driving factor of LST was identified based on each pixel. Two turning points were founded in 2007 and 2011 during the study period of 2003–2019. A significant cooling trend of LST appeared from 2007 to 2011/2012 with the rates of −0.2237 K/year (daytime) and −0.1239 K/year (nighttime). LST increased in almost all seasons except the daytime in autumn. The warming effect in spring and winter contributed 69.43% to interannual warming of daytime LST, and accounted for 59.02% of interannual warming of nighttime LST. In most regions of China, air temperature and vegetation were the dominant factors influencing the change of LST. The current research improved our understanding of changes of LST and the results can serve for mitigating and adapting to climate change.

Eddy-covariance (EC) measurements are widely used to optimize the terrestrial vegetation gross primary productivity (GPP) model because they provide standardized and high-quality flux data within their footprint areas. However, the extent of flux data taken from a tower site within the EC footprint, represented by the satellite-based grid cell between Landsat and Moderate Resolution Imaging Spectroradiometer (MODIS), and the performance of the model derived from the Normalized Difference Vegetation Index (NDVI) within the EC footprint at different spatial resolutions (e.g., Landsat and MODIS) remain unclear. Here, we first calculated the Landsat-footprint NDVI and MODIS-footprint NDVI and assessed their spatial representativeness at 78 FLUXNET sites at 30 m and 500 m scale, respectively. We then optimized the parameters of the revised Eddy Covariance-Light Use Efficiency (EC-LUE) model using NDVI within the EC-tower footprints that were calculated from the Landsat and MODIS sensor. Finally, we evaluated the performance of the optimized model at 30 m and 500 m scale. Our results showed that matching Landsat data with the flux tower footprint was able to improve the performance of the revised EC-LUE model by 18% for savannas, 14% for croplands, 9% for wetlands. The outperformance of the Landsat-footprint NDVI in driving model relied on the spatial heterogeneity of the flux sites. Our study assessed the advantages of remote sensing data with high spatial resolution in simulating GPP, especially for areas with high heterogeneity of landscapes. This could facilitate a more accurate estimation of global ecosystem carbon sink and a better understanding of plant productivity and carbon climate feedbacks.

Accurate simulations of the spatial and temporal changes in vegetation gross primary production (GPP) play an important role in ecological studies. Previous studies highlighted large uncertainties in GPP datasets based on satellite data with coarse spatial resolutions (>500 m), and implied the need to produce high-spatial-resolution datasets. However, estimating fine spatial resolution GPP is time-consuming and requires an enormous amount of computing storage space. In this study, based on the Eddy Covariance-Light Use Efficiency (EC-LUE) model, we used Google Earth Engine (GEE) to develop a web application (EC-LUE APP) to generate 30-m-spatial-resolution GPP estimates within a region of interest. We examined the accuracy of the GPP estimates produced by the APP and compared them with observed GPP at 193 global eddy covariance sites. The results showed the good performance of the EC-LUE APP in reproducing the spatial and temporal variations in the GPP. The fine-spatial-resolution GPP product (GPPL) explained 64% of the GPP variations and had fewer uncertainties (root mean square error = 2.34 g C m−2 d−1) and bias (−0.09 g C m−2 d−1) than the coarse-spatial-resolution GPP products. In particular, the GPPL significantly improved the GPP estimations for cropland and dryland ecosystems. With this APP, users can easily obtain 30-m-spatial-resolution GPP at any given location and for any given year since 1984.

The transition from outcrossing to selfing is a common evolutionary trend in flowering plants, and floral traits change significantly with the evolution of selfing. Whether or not plant traits are subjected to selection remains an open question in species with mixed mating systems. We examined phenotypic selection in two populations of Halenia elliptica with different selfing rates. We found that the pollen–ovule ratio, seed size, plant height, spur length, and pollinator visitation rate in the population with the higher selfing rate were lower than those in the population with the lower selfing rate. Selfing provides reproductive assurance for populations when pollinator service is low, and the floral traits that are associated with selfing syndrome are evident in populations with a higher selfing rate but are subjected to weak selection in each of the two populations with different selfing rates. Directional selection for an early flowering time indicated that late blooming flowers could experience a risk of seed development in alpine environments, and for large plants, selection indicated that seed production could be limited by the available resources. The floral traits that are associated with pollinator attraction and specialization could be subjected to weak selection at the plant level as selfing evolves, and the selective pressures that are independent of pollinators might not change significantly; highlighting the selective biotic and abiotic pressures that shape the morphological traits of plant species and their independence from the mating system.

Winter cereals, including wheat, rye, barley, and triticale, are important food crops, and it is crucial to identify the distribution of winter cereals for monitoring crop growth and predicting yield. The production and plating area of winter cereals in Europe both contribute 12.57% to the total global cereal production and plating area in 2020. However, the distribution maps of winter cereals with high spatial resolution are scarce in Europe. Here, we first used synthetic aperture radar (SAR) data from Sentinel-1 A/B, in the Interferometric Wide (IW) swath mode, to distinguish rapeseed and winter cereals; we then used a time-weighted dynamic time warping (TWDTW) method to discriminate winter cereals from other crops by comparing the similarity of seasonal changes in the Normalized Difference Vegetation Index (NDVI) from Landsat and Sentinel-2 images. We generated winter cereal maps for 2016–2020 that cover 32 European countries with 30 m spatial resolution. Validation using field samples obtained from the Google Earth Engine (GEE) platform show that the producer’s and user’s accuracies are 91% ± 7.8% and 89% ± 10.3%, respectively, averaged over 32 countries in Europe. The winter cereal map agrees well with agricultural census data for planted winter cereal areas at municipal and country levels, with the averaged coefficient of determination R2 as 0.77 ± 0.15 for 2016–2019. In addition, our method can identify the distribution of winter cereals two months before harvest, with an overall accuracy of 88.4%, indicating that TWDTW is an effective method for timely crop growth monitoring and identification at the continent level. The winter cereal maps in Europe are available via an open-data repository.

Extraction conditions can exert a remarkable influence on extraction efficiency. The aim of this study was to improve the extraction efficiency of carotenoids from Dunaliella parva (D. parva). Dimethyl sulfoxide (DMSO) and 95% ethanol were used as the extraction solvents. The extraction time, extraction temperature and the proportions of mixed solvent were taken as influencing factors, and the experimental scheme was determined by Central Composite Design (CCD) of Design Expert 10.0.4.0 to optimize the extraction process of carotenoids from D. parva. The absorbance values of the extract at 665 nm, 649 nm and 480 nm were determined by a microplate spectrophotometer, and the extraction efficiency of carotenoids was calculated. Analyses of the model fitting degree, variance and interaction term 3D surface were performed by response surface analysis. The optimal extraction conditions were as follows: extraction time of 20 min, extraction temperature of 40 °C, and a mixed solvent ratio (DMSO: 95% ethanol) of 3.64:1. Under the optimal conditions, the actual extraction efficiency of carotenoids was 0.0464%, which was increased by 18.19% (the initial extraction efficiency of 0.03926%) with a lower extraction temperature (i.e., lower energy consumption) compared to the standard protocol.

The climate oscillations of the quaternary periods have profoundly affected the geographic distributions of current species. Acer davidii is a deciduous forest tree species mainly distributed in East Asia and China, playing a dominant role in the local forest ecosystem. In order to study the potential changes of geographic distributions of A. davidii in climate fluctuations, we collected the relate geographical distribution data and six climatic variables, using maximum entropy modelling to determine the species distribution. The results showed that the Areas Under Curve (AUC) values of the working characteristic curves of the subjects in the five historical periods were all greater than 0.93, suggesting that the results of maximum entropy modelling were accurate. The simulation of species distribution showed that the suitable area of A. davidii was mainly concentrated in central and northern China in contemporary times. From the Last Interglacial Age (LIG) to the Last Glacial Maximum (LGM), and then to the future (2050, 2070), the distribution area of this species experienced a decrease (LGM~Current; the high adaptability areas of central China became moderate) then an increase (Current~2050, the adaptation areas expanded to South Asia, Southeast Asia, and Siberia), and finally decreased (2050~2070, the suitable areas of South Asia, Southeast Asia, and Siberia shrank returning to China at latitude 25 °N). Compared to the LGM, the area of contemporary suitable area increased. Interestingly, the area of suitable growth range under future climatic conditions (2050) increased by half than before, and the suitable distribution area moved from Midwest China to Northeast China. This study on the change of species distribution can provide a typical case for the model study on the response of plants to climate change in the north temperate and subtropical zones of East Asia. Meanwhile, it can also give a basis for planting planning, species protection, and management.

Abstract Stimuli‐responsive nanomedicines have become a recent research focus as a candidate for cancer treatment because of their effectiveness, sensibility, and minimal invasiveness. In this work, a novel nanosystem is developed based on Cu2−xS@MnS core–shell nanoparticles (CSNPs) in which the Cu2−xS core serves as a photosensitizer to generate hyperthermia and reactive oxygen species (ROS), and the MnS shell is used in H2O2‐responsive O2 production. Cu2−xS@MnS CSNPs with an independent core and shell ratio are synthesized by a controllable hot‐injection method, resulting in an optimal photothermal (PT) effect with a PT conversion efficiency of up to 47.9%. An enhanced photodynamic (PD) effect also occurs in an H2O2 environment. More significantly, in vivo experiments demonstrate that Cu2−xS@MnS CSNPs can mediate tumor shrinkage in both HeLa tumor cell line‐derived xenograft (CDX) and head and neck squamous cell carcinoma (HNSCC) patient‐derived xenograft (PDX) models, with the capability of being used as a T1‐enhanced magnetic resonance (MR) contrast agent. These results suggest the great potential of as‐prepared Cu2−xS@MnS CSNPs as photo/H2O2‐responsive therapeutic‐agents against tumors, even in a complicated and heterogeneous environment, thus promoting the clinical translation of nanomedicine.

The objective was to explore a fast, accurate, non-destructive, and less disturbance method for predicting the aboveground biomass (AGB) of the typical steppe, by using plant height and canopy diameter of the dominant species, Stipa bungeana, Artemisia capillaris, and Lespedeza davurica, data were observed from 165 quadrats during the peak plant growing season, and the product of plant height (PH) and canopy diameter (PC) were calculated for each species. AGB of population were predicted for the same species and other species through using 2/3 of the measured data, and the optimal predictive equation was linear in terms of determination coefficient. The other 1/3 of the data, which was measured from no grazing paddocks or rotational grazing paddocks, was substituted into the predictive equations for validation. Results showed that PC of one dominant species could be used to predict AGB of the same species or other species well. The predicted and measured values were significantly correlative, and most of the predictive accuracy was above 80%, and not affected by managements of grassland, including rotational grazing or no grazing. A combination of 3 to 6 representative species was used to predict AGB of the community, and the predictive equations with PC of six species as an independent variable were the most optimal because explaining 83.5% variation of AGB. The predictive methods cost 1/15, 1/9, and 1/51 of time, labor, and capital as much as the destructive sample method (quadrat sampling method), respectively, and thus improved the efficiency of field study and protecting the fragile study areas, especially the long-term study sites in grassland.

Satellite-derived vegetation indices (VIs) have been widely used to approximate or estimate gross primary productivity (GPP). However, it remains unclear how the VI-GPP relationship varies with indices, biomes, timescales, and the bidirectional reflectance distribution function (BRDF) effect. We examined the relationship between VIs and GPP for 121 FLUXNET sites across the globe and assessed how the VI-GPP relationship varied among a variety of biomes at both monthly and annual timescales. We used three widely-used VIs: normalized difference vegetation index (NDVI), enhanced vegetation index (EVI), and 2-band EVI (EVI2) as well as a new VI - NIRV and used surface reflectance both with and without BRDF correction from the moderate resolution imaging spectroradiometer (MODIS) to calculate these indices. The resulting traditional (NDVI, EVI, EVI2, and NIRV) and BRDF-corrected (NDVIBRDF, EVIBRDF, EVI2BRDF, and NIRV, BRDF) VIs were used to examine the VI-GPP relationship. At the monthly scale, all VIs were moderate or strong predictors of GPP, and the BRDF correction improved their performance. EVI2BRDF and NIRV, BRDF had similar performance in capturing the variations in tower GPP as did the MODIS GPP product. The VIs explained lower variance in tower GPP at the annual scale than at the monthly scale. The BRDF-correction of surface reflectance did not improve the VI-GPP relationship at the annual scale. The VIs had similar capability in capturing the interannual variability in tower GPP as MODIS GPP. VIs were influenced by temperature and water stresses and were more sensitive to temperature stress than to water stress. VIs in combination with environmental factors could improve the prediction of GPP than VIs alone. Our findings can help us better understand how the VI-GPP relationship varies among indices, biomes, and timescales and how the BRDF effect influences the VI-GPP relationship.

To gain insights into the impacts of emission reduction measures on the characteristics and sources of trace elements during the 2014 Asia-Pacific Economic Cooperation (APEC) summit, PM2.5 samples were simultaneously collected from an urban site and a suburban site in Beijing from September 15th to November 12th, and fifteen metallic elements were analyzed, including five crustal elements (Mg, Al, K, Ca and Fe), nine trace metals (V, Cr, Mn, Co, Cu, Zn, Ag, Cd and Pb) and As. Most of the trace metals (V, Cr, Mn, As, Cd and Pb) decreased more than 40% due to the emission regulations during APEC, while the crustal elements decreased considerably (4⁻45%). Relative to the daytime, trace metals increased during the nighttime at both sites before the APEC summit, but no significant difference was observed during the APEC summit, suggesting suppressed emissions from anthropogenic activities. Five sources (dust, traffic exhaust, industrial sources, coal and oil combustion and biomass burning) were resolved using positive matrix factorization (PMF), which were collectively decreased by 30.7% at the urban site and 14.4% at the suburban site during the APEC summit. Coal and oil combustion regulations were the most effective for reducing the trace elements concentrations (urban site: 63.1%; suburban site: 52.0%), followed by measures to reduce traffic exhaust (52.8%) at the urban site and measures to reduce biomass burning (37.7%) at the suburban site. Our results signify that future control efforts of metallic elements in megacities like Beijing should prioritize coal and oil combustion, as well as traffic emissions.

Human flap endonuclease 1 (FEN1), one of the structure-specific 5′ nucleases, is integral in replication, repair, and recombination of cellular DNA. The 5′ nucleases share significant unifying features yet cleave diverse substrates at similar positions relative to 5′ end junctions. Using single-molecule Förster resonance energy transfer, we find a multistep mechanism that verifies all substrate features before inducing the intermediary-DNA bending step that is believed to unify 5′ nuclease mechanisms. This is achieved by coordinating threading of the 5′ flap of a nick junction into the conserved capped-helical gateway, overseeing the active site, and bending by binding at the base of the junction. We propose that this sequential and multistep substrate recognition process allows different 5′ nucleases to recognize different substrates and restrict the induction of DNA bending to the last common step. Such mechanisms would also ensure the protection of DNA junctions from nonspecific bending and cleavage.

Pulmonary hypertension syndrome (PHS) has always been of significant importance related to cardiovascular diseases. The PHS is one of the major reasons for the high morbidity and mortality in broilers negatively impacting the broiler farming industry. In this study samples from lungs and blood of broilers with enlarged and normal abdomen were collected and fluorescence quantification, immunohistochemistry and histopathology of the selected organs were done to diagnose this disease in broilers. Clinically, it was observed that morbid broilers were depressed, dyspneic, right heart hypertrophied, right ventricle to total ventricle volume (RV/TV) was greater than 0.25 and abdomen was enlarged having yellowish fluid. The diagnosis of PHS was confirmed by the RV/TV and clinical symptoms whereas, the histopathological observations revealed that with age the pulmonary vascular density decreased, lumen of the pulmonary vessels was narrowed and the middle layer of pulmonary vessels was thickened. Hematoxylin and Eosin (HE) staining of the selected samples was done. It was found that age increased in greater interstitial spaces in the lungs of PHS broilers along with inflammatory cells infiltrating the lungs. Results of fluorescence quantification and immunohistochemistry revealed that all types of PDK (PDK1, PDK2, PDK3, PDK4) exhibited significant upregulation in the lungs of broilers with PHS onset and PDK4 was significantly expressed on pulmonary vessels. Therefore, it was concluded that PHS leads to remodeling of the pulmonary vasculature leading to ascites and PDK plays an important role in the progression of PHS. [ABSTRACT FROM AUTHOR]
