Calculating the parameters of the ground shock induced by an underground explosion is a complex energy coupling problem. It is difficult to establish a unified ground shock coupling law from limited test data. This paper summarizes the research results obtained at home and abroad and systematically analyzes the coupling mechanism of craters formed by an underground explosion and the ground shock. The differences between the concepts of “closed-explosion critical depth” and “equivalent closed-explosion critical depth” are clearly explained. The spreading of the ground shock energy is attributed to the explosive expansion of the air cavity, revealing a linear relationship between the volume of the cavity region (or the volume of the crack region) and the ground shock energy associated with the underground explosion. The proportionality factor is related to the mechanical properties of the medium and is independent of the magnitude of the explosion equivalent. Based on this, a theoretical calculation formula and conversion method for the ground shock coupling coefficient were established. Explosion tests were conducted in clay and Plexiglass under varying burial depths. The test results were consistent with the theoretically calculated results. Our study provides a theoretical basis for the design of explosion-resistant structures in underground engineering.

Abstract Background Ubiquitination plays an important role in proliferating and invasive characteristic of glioblastoma (GBM), similar to many other cancers. Tripartite motif 25 (TRIM25) is a member of the TRIM family of proteins, which are involved in tumorigenesis through substrate ubiquitination. Methods Difference in TRIM25 expression levels between nonneoplastic brain tissue samples and primary glioma samples was demonstrated using publicly available glioblastoma database, immunohistochemistry, and western blotting. TRIM25 knockdown GBM cell lines (LN229 and U251) and patient derived GBM stem-like cells (GSCs) GBM#021 were used to investigate the function of TRIM25 in vivo and in vitro. Co-immunoprecipitation (Co-IP) and mass spectrometry analysis were performed to identify NONO as a protein that interacts with TRIM25. The molecular mechanisms underlying the promotion of GBM development by TRIM25 through NONO were investigated by RNA-seq and validated by qRT-PCR and western blotting. Results We observed upregulation of TRIM25 in GBM, correlating with enhanced glioblastoma cell growth and invasion, both in vitro and in vivo. Subsequently, we screened a panel of proteins interacting with TRIM25; mass spectrometry and co-immunoprecipitation revealed that NONO was a potential substrate of TRIM25. TRIM25 knockdown reduced the K63-linked ubiquitination of NONO, thereby suppressing the splicing function of NONO. Dysfunctional NONO resulted in the retention of the second intron in the pre-mRNA of PRMT1, inhibiting the activation of the PRMT1/c-MYC pathway. Conclusions Our study demonstrates that TRIM25 promotes glioblastoma cell growth and invasion by regulating the PRMT1/c-MYC pathway through mediation of the splicing factor NONO. Targeting the E3 ligase activity of TRIM25 or the complex interactions between TRIM25 and NONO may prove beneficial in the treatment of GBM.

Abstract Accumulating evidence suggests that changes in the tumor microenvironment caused by radiotherapy are closely related to the recurrence of glioma. However, the mechanisms by which such radiation‐induced changes are involved in tumor regrowth have not yet been fully investigated. In the present study, how cranial irradiation‐induced senescence in non‐neoplastic brain cells contributes to glioma progression is explored. It is observed that senescent brain cells facilitated tumor regrowth by enhancing the peripheral recruitment of myeloid inflammatory cells in glioblastoma. Further, it is identified that astrocytes are one of the most susceptible senescent populations and that they promoted chemokine secretion in glioma cells via the senescence‐associated secretory phenotype. By using senolytic agents after radiotherapy to eliminate these senescent cells substantially prolonged survival time in preclinical models. The findings suggest the tumor‐promoting role of senescent astrocytes in the irradiated glioma microenvironment and emphasize the translational relevance of senolytic agents for enhancing the efficacy of radiotherapy in gliomas.

Abstract Objective Risk stratification plays a critical role in patients with asymptomatic carotid atherosclerotic stenosis. Heavy macrophage infiltration (HMC) is an important factor of plaque destabilization. However, in vivo imaging technologies and screening criteria for HMC remain limited. We aimed to (i) introduce algorithms for in vivo detection of macrophage infiltrations using optical coherence tomography (OCT) and (ii) to investigate the threshold of HMC and its association with plaque vulnerability. Methods Ex vivo OCT images were co‐registered with histopathology in 282 cross‐sectional pairs from 19 carotid endarterectomy specimens. Of these, 197 randomly selected pairs were employed to define the parameters, and the remaining 85 pairs were used to evaluate the accuracy of the OCT‐based algorithm in detecting macrophage infiltrations. Clinical analysis included 93 patients receiving carotid OCT evaluation. The prevalence and burden of macrophage infiltration were analyzed. Multivariable and subgroup analysis were performed to investigate the association between HMC and plaque rupture. Results The sensitivity and specificity of algorithm for detecting macrophage infiltration were 88.0% and 74.9%, respectively. Purpose – The purpose of this study is to examine the impact of managerial short-termism on green innovation of firms and the moderating role of digital transformation of enterprises in the association between managerial short-termism and green innovation. Design/methodology/approach – This study uses data from Chinese A-share listed companies from 2001 to 2021 and employ panel fixed model and moderating effect model to examine the impact of managerial short-termism on green innovation of firms and the moderating role of digital transformation of enterprises in the association between managerial short-termism and green innovation. Findings – The findings of this study reveal that managerial short-termism exerts negative influence on green innovation. Digital transformation enables firms to reduce the adverse effect of managerial short-termism on green innovation because digital transformation enhances information processing ability and then improves internal corporate governance and analyst coverage. Moreover, the moderating role of digital transformation is more prominent for firms with lower internal corporate governance, for firms with less analyst coverage and for non-state-owned enterprises. Originality/value – This paper intends to address the following two questions: what is the impact of managerial short-termism on green innovation and what is the role of digital transformation in the two variables’ association? By using data of Chinese A-share listed companies from 2001 to 2021 and developing two individual indexes to measure managerial short-termism and digital transformation, the authors empirically test these above two questions. The results of this study indicate that: First, drawn on time-oriented theory and upper echelon theory, managerial short-termism has an adverse effect on firms’ green innovation. Second, digital transformation enables firms to reduce the negative effect of managerial short-termism on green innovation. Furthermore, the moderating mechanism tests show that the corporate governance effects of digital transformation play a supervisory role that impels managers to reduce short-term investments and promote firms’ green R&D investments, which helps to reduce the negative effect of managerial short-termism on green innovation. Additionally, the heterogeneity checks show that the moderating role of digital transformation in the relation between managerial short-termism and green innovation is more prominent for firms with lower internal corporate governance, with less analyst coverage and for non-state-owned enterprises.

ABSTRACT: Phage therapy is gaining momentum as an alternative to antibiotics in the treatment of salmonellosis caused by Salmonella. In this study, a novel Salmonella phage, vB_SalS_JNS02, was isolated successfully from poultry farms in Shandong, China. The biological characteristics of vB_SalS_JNS02 were analysed, which revealed a short latent period of approximately 10 min and a burst size of 110 PFU/cell. Moreover, vB_SalS_JNS02 exhibited remarkable stability across a wide pH range (pH 3–12) and temperatures ranging from 30 to 80°C. Genome sequencing analysis provided valuable insights into the genetic composition of vB_SalS_JNS02, which consists of a double-stranded DNA genome that spans 42,450 base pairs and has a G + C content of 49.4%. Of significant importance, the genomic sequence of vB_SalS_JNS02 did not contain any genes related to lysogenicity, virulence, or antibiotic resistance. The phage's efficacy was evaluated in a larval challenge study. Treatment with the phage resulted in increased survival of Galleria mellonella larvae (100, 70, and 85%) (MOI 0.1) in the prophylactic treatment, co-infection treatment, and remedial treatment experiments, respectively. Another in vivo experiment investigated the potential application of the phage in broiler chickens and revealed that a single oral dose of vB_SalS_JNS02 (108 PFU/mL, 100 µL/chick) administered 3 h after S. enteritidis oral administration provided effective protection. The introduction of bacteriophage not only enhances the production of secretory immunoglobulin A (sIgA), but also induces alterations in the composition of the gut microbial community. Phage therapy increases the relative abundance of beneficial bacteria, which helps to maintain intestinal barrier homeostasis. However, it is unable to fully restore the disrupted intestinal microbiome caused by S. enteritidis infection. Importantly, no significant adverse effects were observed in the animal subjects following oral administration of the phage, and our findings highlight vB_SalS_JNS02 is a hopeful candidate as a promising tool to target Salmonella infections in poultry.

The emergence of triploid oysters has freed the oyster market from the problem of interruptions in the breeding season, allowing for year-round availability. However, according to recent statistics, the phenomenon of fertility in triploid oysters has been increasing yearly, perhaps eventually leading to the loss of the sterility characteristic. This has in turn affected the oyster farming industry. Therefore, we attempted to mine key genes through transcriptomics to find the causes of triploid fertility. During our surveys and sampling, we found that most of the fertile triploid Crassostrea gigas were females, with a much smaller percentage of males. Therefore, the main objective of this study was to analyze the differences between fertile and sterile triploid female C. gigas. We performed transcriptome analysis of fertile female triploid C. gigas and sterile female triploid C. gigas and identified 2361 up-regulated differentially expressed genes (DEGs) and 1541 down-regulated DEGs, respectively. Enrichment analysis of these DEGs revealed that most GO terms and KEGG pathways were associated with mitosis. We then created a protein–protein interaction (PPI) network using genes enriched in mitosis-associated KEGG pathways. Finally, we screened and validated 26 key genes by quantitative RT-PCR and all were found to be downregulated in expression in sterile female triploids. The main cause of sterility is the suppression of the expression of genes that control the process of germ cell proliferation, the failure of the mitotic cycle to proceed smoothly, and the inability to produce an oogonium. These results can be used to explore ways to reduce the fertility of triploid C. gigas and to promote the breeding of higher quality triploid oysters.

Many societies around the world are rapidly aging and this has implications for social and economic development. We collect data on NEEQ-listed enterprises from 2010 to 2021 in China and empirically test the effect of population aging on the digital transformation of small and medium-sized enterprises (SMEs). The findings show that population aging has a significant positive impact on SME digital transformation, and private enterprises and enterprises in eastern regions of China tend to benefit more than other regions. The mechanism studies find that population aging positively impacts SME digital transformation by increasing labor costs, facilitating human capital accumulation, and raising savings rates. Furthermore, the threshold effect analyses find that the marginal promotion effect of population aging will weaken with greater aging and will strengthen with a higher level of marketization. Finally, we provide policy recommendations for promoting digital transformation in SMEs against the background of population aging.

BackgroundExposure to heavy metals (cadmium, mercury, and lead) has been linked with adverse health outcomes, especially their nephrotoxic effects at high levels of exposure. We conducted a replication study to examine the association of low-level heavy metal exposure and chronic kidney disease (CKD) using a larger NHANES data set compared to previous studies.MethodsThe large cross-sectional study comprised 5,175 CKD cases out of 55677 participants aged 20-85 years from the 1999-2020 National Health and Nutrition Examination Survey [NHANES]. Logistic regression analysis was applied to estimate the associations between CKD and heavy metals [Cd, Pb, Hg] measured as categorical variables after adjusting with age, race, gender, socioeconomic status, hypertension, diabetes mellitus and blood cotinine level as smoking status.ResultsCompared to the lowest quartile of blood Cd, exposures to the 2nd, 3rd and 4th quartiles of blood Cd were statistically significantly associated with higher odds of CKD after adjustment for blood Pb and Hg, with OR = 1.79, [95% CI; 1.55-2.07, pConclusionsOur findings showed that low blood levels of Cd and Pb were associated with higher odds of CKD while low blood levels of Hg were associated with lower odds of CKD in the US adult population. However, temporal association cannot be determined as it is a cross sectional study.

BackgroundSarcopenia can lead to significant personal, social, and economic burdens. The diagnosis of sarcopenia heavily relies on the identification of Low Skeletal Muscle Mass (LSMM), which is an independent predictor of frailty, disability, and increased risk of death among seniors. Women have physiologically lower levels of skeletal muscle mass than men, and female sarcopenia appears to be more influenced by menopause. They also tend to have higher body fat levels than man, which increases the risk of sarcopenia obesity. On another front, it’s also recognized that humans are largely prone to Helicobacter pylori (H. pylori) infection, with global prevalence rates often surpassing 50%. Nevertheless, the interconnection between H. pylori infection and LSMM remains relatively unexplored. Hence, our study specifically targeted women as the research population and sought to explore several risk factors for LSMM. Additionally, we delved into the potential correlation between LSMM and H. pylori infection in women, hoping to gain insights into potential preventative measures or treatment options that may enhance the quality of life for women affected by sarcopenia.MethodsWe conducted a cross-sectional study among women aged over 18 years undergoing physical examination. We performed 13C-urea breath test (UBT) for diagnosis of H. pylori infection and Bioelectrical impedance analysis (BIA) for the assessment of LSMM. Logistic regression models were used to analyze the associations of H. pylori infection with LSMM.ResultsThis study enrolled 1984 Chinese women who were undergoing health check-ups. A univariate logistic regression analysis did not reveal a direct correlation between H. pylori infection and LSMM among this female population (OR=1.149, 95% CI 0.904-1.459, p=0.257). Yet, upon dividing the participants into age-based subgroups, an evident link was observed between H. pylori infection and LSMM in women aged 40 or above (OR=1.381, 95%CI 1.032-1.848, p= 0.030). After adjusting for variables including Age, BMI, TP, ALK, Cre, this relationship remained statistically relevant (OR=1.514, 95%CI 1.085-2.113, p= 0.015).ConclusionsWomen who are over 40 years old and currently infected with H. pylori have an increased risk of developing LSMM. Therefore, timely treatment for H. pylori eradication is recommended for this group of women to reduce the occurrence of LSMM.

Abstract One of the main characteristics of optical imaging systems is spatial resolution, which is restricted by the diffraction limit to approximately half the wavelength of the incident light. Along with the recently developed classical super-resolution techniques, which aim at breaking the diffraction limit in classical systems, there is a class of quantum super-resolution techniques which leverage the non-classical nature of the optical signals radiated by quantum emitters, the so-called antibunching super-resolution microscopy. This approach can ensure a factor of $$\sqrt{n}$$ n improvement in the spatial resolution by measuring the n -th order autocorrelation function. The main bottleneck of the antibunching super-resolution microscopy is the time-consuming acquisition of multi-photon event histograms. We present a machine learning-assisted approach for the realization of rapid antibunching super-resolution imaging and demonstrate 12 times speed-up compared to conventional, fitting-based autocorrelation measurements. The developed framework paves the way to the practical realization of scalable quantum super-resolution imaging devices that can be compatible with various types of quantum emitters.

Extracellular vesicles (EVs) derived from mesenchymal stem cells (MSCs) have emerged as a new mode of intercellular crosstalk and are responsible for many of the therapeutic effects of MSCs. To promote the application of MSC-EVs, recent studies have focused on the manipulation of MSCs to improve the production of EVs and EV-mediated activities. The current paper details an optimization method using non-invasive low-intensity pulsed ultrasound (LIPUS) as the stimulation for improving oral MSC-EV production and effectiveness. Stem cells from apical papilla (SCAP), a type of oral mesenchymal stem cell, displayed intensity-dependent pro-osteogenic and anti-inflammatory responses to LIPUS without significant cytotoxicity or apoptosis. The stimuli increased the secretion of EVs by promoting the expression of neutral sphingomyelinases in SCAP. In addition, EVs from LIPUS-induced SCAP exhibited stronger efficacy in promoting the osteogenic differentiation and anti-inflammation of periodontal ligament cells in vitro and alleviating oral inflammatory bone loss in vivo. In addition, LIPUS stimulation affected the physical characteristics and miRNA cargo of SCAP-EVs. Further investigations indicated that miR-935 is an important mediator of the pro-osteogenic and anti-inflammatory capabilities of LIPUS-induced SCAP-EVs. Taken together, these findings demonstrate that LIPUS is a simple and effective physical method to optimize SCAP-EV production and efficacy.

The bonding force of hardened magnesium phosphate cement (MPC) based materials comes from the ionic and covalent bonds with high bond energy and strength between cementitious phosphate hydrates, resulting in high brittleness under stress. Fiber reinforced MPC matrix composites can consume energy through the various effects of fibers, thereby improving the brittleness of the MPC matrix. This study uses glass fiber three-dimensional spacing fabric with good fiber directional reinforcement ability and interlayer shear resistance as the reinforcement material, and potassium magnesium phosphate cement (MPKC) mortar was used as the matrix to prepare glass fiber reinforced MKPC based composite materials. Design and optimize the surface treatment method of three-dimensional spacing fabrics through uniaxial tensile testing of fiber reinforcement and its pull-out test in MKPC matrix. The mechanical properties of MKPC based composite material specimens reinforced with fabric were studied through compressive and four point bending tests. The results are as follows: the axial tensile force of the glass fibre reinforcement treated with water glass impregnation containing an appropriate amount of magnesium oxide powder is 2.5 times that of the untreated fibre reinforcement (the reference sample), and its tensile force in the MKPC matrix can reach 2 times that of the reference sample. The three-dimensional spacing fabric can improve the compressive strength of MKPC-based material specimens, and the larger the spacing between the fabric sandwich layers, the better the reinforcement effect. The reinforcement effect of the three-dimensional fabric impregnated with water glass containing magnesium oxide powder is better, and its compressive strength can be increased by 14.3% in comparison to the reference specimens without fabric fibers. The MKPC thin plate containing three-dimensional spacing fabric can withstand larger loads after initial cracking, and multiple cracks develop simultaneously. The initial cracking strength and initial cracking displacement slightly increase, and the ultimate strength and ultimate displacement significantly increase. The toughening effect of spacing fabrics with low-core columns on MKPC-based thin plate specimens is better than that of spacing fabrics with high-core columns. Impregnating three-dimensional spacer fabrics with water glass containing magnesium oxide powder can significantly improve the initial crack strength, deformation, ultimate strength, and displacement of MKPC-based thin plate specimens. In comparison to the reference sample without fabric fibers, the surface treated fabric reinforced MKPC matrix composite material specimen M1–1 exhibited a significant increase in cracking load and strain (26.01% and 33.96%), and its ultimate load and displacement were significantly increased (113.26% and 1125.13%). The above research content provides a theoretical basis for further research and engineering applications of this type of composite material.

Background: Echocardiographic data investigating the association between left ventricular (LV) function and gout is still limited.Purpose: To analyze the association of echocardiographic parameters based on two-dimentional speckle tracking analysis with clinically related indicators in patients with gout, and to provide a clinical basis for the early diagnosis and treatment of cardiovascular disease in patients with gout.Methods: This study collected gout patients who visited the outpatient and inpatient departments of the first affiliated hospital of chengdu medical college from November 2019 to December 2020. Spearman correlation test was performed to analyze the correlation coefficients between the laboratorial indicators with echocardiographic parameters. And the logistic regression analysis was performed to evaluate the independent effects.Results: The results of multivariate logistic regression showed that fasting plasma glucose (FPG) was a risk factor for the decrease in absolute value of global longitudinal strain [GLS (OR = 2.34; 95% CI, 1.01–5.39; p = 0.04)], Urea was a risk factor for absolute reduction in GCS (OR = 1.40; 95% CI, 1.07–1.85; p = 0.02), age (OR = 1.09, 95% CI, 1.04–1.16; p = 0.001), and hypertension (OR = 8.35; 95% CI, 1.83–38.02; p = 0.006) were risk factors for increased E/Em. High urea levels were significantly related with high risks of LVH (OR = 1.59, 95% CI, 1.04–2.43; p = 0.03) and enlargement of LAVI (OR = 1.68, 95% CI, 1.01–2.80; p = 0.04).Conclusion: Our study found that elevated urea and FPG were risk factors for subclinical LV myocardial dysfunction in patients with gout, which might provide a theoretical basis for the early diagnosis and treatment of heart disease in clinical practice.

Background: Studies suggest associations between long-term ambient air pollution exposure and outcomes related to Alzheimer’s disease (AD). Whether a link exists between pollutants and brain amyloid accumulation, a biomarker of AD, is unclear. We assessed whether long-term air pollutant exposures are associated with late-life brain amyloid deposition in Atherosclerosis Risk in Communities (ARIC) study participants. Methods: We used a chemical transport model with data fusion to estimate ambient concentrations of PM2.5 and its components, NO2, NOx, O3 (24-hour and 8-hour), CO, and airborne trace metals. We linked concentrations to geocoded participant addresses and calculated 10-year mean exposures (2002 to 2011). Brain amyloid deposition was measured using florbetapir amyloid positron emission tomography (PET) scans in 346 participants without dementia in 2012–2014, and we defined amyloid positivity as a global cortical standardized uptake value ratio ≥ the sample median of 1.2. We used logistic regression models to quantify the association between amyloid positivity and each air pollutant, adjusting for putative confounders. In sensitivity analyses, we considered whether use of alternate air pollution estimation approaches impacted findings for PM2.5, NO2, NOx, and 24-hour O3. Results: At PET imaging, eligible participants (N = 318) had a mean age of 78 years, 56% were female, 43% were Black, and 27% had mild cognitive impairment. We did not find evidence of associations between long-term exposure to any pollutant and brain amyloid positivity in adjusted models. Findings were materially unchanged in sensitivity analyses using alternate air pollution estimation approaches for PM2.5, NO2, NOx, and 24-hour O3. Conclusions: Air pollution may impact cognition and dementia independent of amyloid accumulation, though whether air pollution influences AD pathogenesis later in the disease course or at higher exposure levels deserves further consideration.

Lung injury and pulmonary fibrosis contribute to morbidity and mortality, and, in particular, are characterized as leading cause on confirmed COVID-19 death. To date, efficient therapeutic approach for such lung diseases is lacking. N-Acetylglucosamine (NAG), an acetylated derivative of glucosamine, has been proposed as a potential protector of lung function in several types of lung diseases. The mechanism by which NAG protects against lung injury, however, remains unclear. Here, we show that NAG treatment improves pulmonary function in bleomycin (BLM)-induced lung injury model measured by flexiVent system. At early phase of lung injury, NAG treatment results in silenced immune response by targeting ARG1+ macrophages activation, and, consequently, blocks KRT8+ transitional stem cell in the alveolar region to stimulate PDGF Rβ+ fibroblasts hyperproliferation, thereby attenuating the pulmonary fibrosis. This combinational depression of immune response and extracellular matrix deposition within the lung mitigates lung injury and pulmonary fibrosis induced by BLM. Our findings provide novel insight into the protective role of NAG in lung injury.

This paper discusses a type of mixed-delay quaternion-valued neural networks (QVNNs) under impulsive and stochastic disturbances. The considered QVNNs model are treated as a whole, rather than as complex-valued neural networks (NNs) or four real-valued NNs. Using the vector Lyapunov function method, some criteria are provided for securing the mean-square exponential stability of the mixed-delay QVNNs under impulsive and stochastic disturbances. Furthermore, a type of chaotic QVNNs under stochastic and impulsive disturbances is considered using a previously established stability analysis method. After the completion of designing the linear feedback control law, some sufficient conditions are obtained using the vector Lyapunov function method for determining the mean-square exponential synchronization of drive–response systems. Finally, two examples are provided to demonstrate the correctness and feasibility of the main findings and one example is provided to validate the use of QVNNs for image associative memory.

Abstract Liver hepatocellular carcinoma (LIHC) is one of the most common malignancies and places a heavy burden on patients worldwide. HAUS augmin-like complex subunit 5 (HAUS5) is involved in the occurrence and development of various cancers. However, the functional role and significance of HAUS5 in LIHC remain unclear. The Cancer Genome Atlas (TCGA), Genotype-Tissue Expression (GTEx), Cancer Cell Line Encyclopedia (CCLE) and Gene Expression Omnibus (GEO) databases were used to analyze the mRNA expression of HAUS5. The value of HAUS5 in predicting LIHC prognosis and the relationship between HAUS5 and clinicopathological features were assessed by the Kaplan–Meier plotter and UALCAN databases. Functional enrichment analyses and nomogram prediction model construction were performed with the R packages. The LinkedOmics database was searched to reveal co-expressed genes associated with HAUS5. The relationship between HAUS5 expression and immune infiltration was explored by searching the TISIDB database and single-sample gene set enrichment analysis (ssGSEA). The Clinical Proteomic Tumor Analysis Consortium (CPTAC) and the Human Protein Atlas (HPA) databases were used to evaluate HAUS5 protein expression. Finally, the effect of HAUS5 on the proliferation of hepatoma cells was verified by CCK-8, colony formation and EdU assays. HAUS5 is aberrantly expressed and associated with a poor prognosis in most tumors, including LIHC. The expression of HAUS5 is significantly correlated with clinicopathological indicators in patients with LIHC. Functional enrichment analysis showed that HAUS5 was closely related to DNA replication, cell cycle and p53 signaling pathway. HAUS5 may serve as an independent risk factor for LIHC prognosis. The nomogram based on HAUS5 had area under the curve (AUC) values of 0.74 and 0.77 for predicting the 3-year and 5-year overall survival (OS) of LIHC patients. Immune correlation analysis showed that HAUS5 was significantly associated with immune infiltration. Finally, the results of in vitro experiments showed that when HAUS5 was knocked down, the proliferation of hepatoma cells was significantly decreased. The pan-oncogene HAUS5 is a positive regulator of LIHC progression and is closely associated with a poor prognosis in LIHC. Moreover, HAUS5 is involved in immune infiltration in LIHC. HAUS5 may be a new prognostic marker and therapeutic target for LIHC patients.

Autism spectrum disorder (ASD)—a congenital mental disorder accompanied by social dysfunction and stereotyped behaviors—has attracted a great deal of attention worldwide. A combination of genetic and environmental factors may determine the pathogenesis of ASD. Recent research of multiple ASD models indicates that microRNAs (miRNAs) play a central role at the onset and progression of ASD by repressing the translation of key mRNAs in neural development and functions. As such, miRNAs show great potential to serve as biomarkers for ASD diagnosis or prognosis and therapeutic targets for the treatment of ASD. In this review, we discuss the regulatory mechanisms by which miRNAs influence ASD phenotypes through various in vivo and in vitro models, including necropsy specimens, animal models, cellular models, and, in particular, induced pluripotent stem cells derived from patients with ASD. We then discuss the potential of miRNA-based therapeutic strategies for ASD currently being evaluated in preclinical studies.

Feathers are important economic products. Anser cygnoide (AC) is superior in reproduction, growth and feather quality, while Anser anser (AA) has an advantage in feather yield. Understanding the development of feather follicles facilitates deliberate crossbreeding. Here, we compared feather follicles morphogenesis between AC and AA. Feathers and dorsal skin were sampled at multiple embryonic days to investigate feather phenotypes, feather follicle morphology and the expression of hub molecules (FZD4, β-catenin, LEF1, TCF4 and c-Myc) in the Wnt signalling pathway. We found a marked difference in plumage colour between the two breeds. Feather colour in AA has undergone a ‘black dorsum’ stage and cavity formation in AC was earlier. Furthermore, different trends in molecular hub expression helped explain the phenotypical and morphological differences: the upregulated Wnt signals in AC at E12 and/or E15, as well as in AA at E18, were associated with early epidermal placode formation and cavity formation in AC, also, related to the development of the secondary feather follicles and ‘black dorsum’ in AA. Summarily, breed-specific expression patterns of Wnt signalling pathway between AC and AA might participate in the shaping of breed-specific feather follicles morphogenesis and feather phenotypes. This work provides references for feather follicles development in geese.

Sepia esculenta is an economically important mollusk distributed in the coastal waters of China. Juveniles are more susceptible to stimulation by the external environment than mature individuals. The ocean salinity fluctuates due to environmental changes. However, there is a lack of research on the salinity adaptations of S. esculenta. Therefore, in this study, we investigated the differential expression of genes in S. esculenta larvae after stimulation by low salinity. RNA samples were sequenced and 1039 differentially expressed genes (DEGs) were identified. Then, enrichment analysis was performed using the Gene Ontology (GO) and Kyoto Encyclopedia of Genes and Genomes (KEGG) databases. Finally, a protein–protein interaction network (PPI) was constructed, and the functions of key genes in S. esculenta larvae after low-salinity stimulation were explored. We suggest that low salinity leads to an excess proliferation of cells in S. esculenta larvae that, in turn, affects normal physiological activities. The results of this study can aid in the artificial incubation of S. esculenta and reduce the mortality of larvae.

Sepia esculenta is a cephalopod widely distributed in the Western Pacific Ocean, and there has been growing research interest due to its high economic and nutritional value. The limited anti-stress capacity of larvae renders challenges for their adaptation to high ambient temperatures. Exposure to high temperatures produces intense stress responses, thereby affecting survival, metabolism, immunity, and other life activities. Notably, the molecular mechanisms by which larval cuttlefish cope with high temperatures are not well understood. As such, in the present study, transcriptome sequencing of S. esculenta larvae was performed and 1,927 differentially expressed genes (DEGs) were identified. DEGs were subjected to functional enrichment analyses using the Gene Ontology (GO) and Kyoto Encyclopedia of Genes and Genomes (KEGG) databases. The top 20 terms of biological processes in GO and 20 high-temperature stress-related pathways in KEGG functional enrichment analysis were identified. A protein-protein interaction network was constructed to investigate the interaction between temperature stress-related genes. A total of 30 key genes with a high degree of participation in KEGG signaling pathways or protein-protein interactions were identified and subsequently validated using quantitative RT-PCR. Through a comprehensive analysis of the protein-protein interaction network and KEGG signaling pathway, the functions of three hub genes (HSP90AA1, PSMD6, and PSMA5), which belong to the heat shock protein family and proteasome, were explored. The present results can facilitate further understanding of the mechanism of high temperature resistance in invertebrates and provide a reference for the S. esculenta industry in the context of global warming.

The impacts of hydraulic fracturing (HF) on birth defects have been suggested by previous studies but remain largely inconclusive. In this study, we assessed whether pregnant women who lived in areas with high HF activities had increased risks of giving birth to offspring with overall or specific birth defects, including atrial septal defect (ASD), ventricular septal defect (VSD), patent ductus arteriosus (PDA), microcephaly (MIC), and hydrocephaly without spina bifida (HSB). All live births between 1999 and 2014 among the residents in the four core counties of Denton, Johnson, Tarrant, and Wise in the Barnett Shale region, Texas, were analyzed. Standardized Morbidity Ratio (SMR) and Poisson regressions were applied for statistical analysis. Compared to the statewide risk, the risks of ASD, VSD, and PDA in four selected counties with high HF activities were significantly higher. The Annual Natural Gas Production from HF was significantly correlated with risks of ASD, PDA, MIC, and total birth defect after adjusting for counties and years. No significant associations of HF activities were found with VSD and HSB. This ecological study suggested that hydraulic fracturing might be associated with the increased risk of some birth defects in the Barnett Shale Region, TX, which warrants further investigations due to the limitation of an ecological study design.

Sea cucumber Apostichopus japonicus, an important marine benthic animal, is particularly sensitive to environmental temperature stress, which affected the growth, metabolism, and reproduction even survival. In this study, we used whole genome bisulfite sequencing (WGBS) to perform a comprehensive comparative analysis of the DNA methylation profiles of A. japonicus at three temperatures: normal (20 °C), aestivation (26 °C), and lethal (32 °C). The results showed that the total methylation level was highest in group T26 and lowest in group T32. Most of the DNA methylation occurred on the C bases of the CG type, and the degree of DNA methylation on the CG-type C bases was 13.32–14.99%, reflecting a moderate level of DNA methylation. Comparison of differentially methylated regions (DMRs) in the CG sequence context, we identified 17,174 DMRs in group T26_vs_T20 and 16,216 DMRs in group T32_vs_T26. There were more hypermethylated regions than hypomethylated regions, and this difference was more obvious in group T32_vs_T26. DMRs analysis of the promoter regions and the gene body regions identified more DMR-related genes in the gene body region than in the promoter region in both comparison groups (T26_vs_T20 and T32_vs_T26). GO and KEGG analysis showed that pathways such as protein processing in the endoplasmic reticulum, ubiquitin mediated proteolysis, and endocytosis, etc., were highly enriched in the DMR-related genes in the comparison groups. Some of these pathways that were significantly enriched in the hypermethylated genes in group T26_vs_T20 but were enriched in the hypomethylated genes in group T32_vs_T26. Enrichment analysis of genes both differentially methylated and differentially expressed showed that downregulated genes in group T32_vs_T26 that were also hypomethylated in the gene body region were significantly enriched in the metabolic and the antioxidant-related glutathione metabolism pathway. Our results suggested that DNA methylation develop a lot of variation in the regulation of the response of A. japonicus to two levels of heat stress: at aestivation temperature and at lethal temperature, and there are many differences between the two. The results of this study provide important insights into the epigenetic mechanisms underlying the response of A. japonicus to the pressure of environmental temperature.

Sperm storage is an important reproductive strategy in Amphioctopus fangsiao. Sperm can be stored in the oviducal glands up to eight months before fertilization. However, studies on the underlying storage mechanisms remain limited. In this study, the morphological structures of the oviductal glands and the changes adapted for sperm storage and fertilization were investigated by Hematoxylin eosin (HE) staining, transmission electron microscopy (TEM), and Alcian blue periodic acid Schiff (AB-PAS) staining. Results revealed that the nucleo-cytoplasmic ratio of the peripheral gland cells decreased and the eosinophilic secretory granules (glycoproteins) appeared in the cytoplasm and lumen during sperm storage. The cytoplasm of the central gland appeared vacuolated and AB pH 2.5 (+) secretions (acid mucins) were dispersed in the cells. These secretions may pass through the central cavity into the oviducts and be involved in zygote packaging. The spermathecae are composed of columnar epithelial ciliated cells with secretory functions. PAS (+) glycoproteins were found in the lumen, providing a microenvironment conducive to sperm survival. Cilia in the oviducts, central cavity, and spermathecae may help with sperm delivery, storage, and release, and egg transport. This study enhances our understanding of the biological reproduction process in A. fangsiao.

Using data on Chinese A-share listed firms from 2008 to 2017, we explore how corporate social responsibility (CSR) performance is affected by managerial short-termism and what factors influence the association between the two. First, by employing text analysis in conjunction with machine learning, we construct a new managerial short-termism indicator. Using panel fixed models, we find that managerial short-termism has an adverse impact on CSR performance, and the results are consistent in a series of robustness checks. The heterogeneous test results show that the negative effect is significant only for firms with lower internal corporate governance, for firms in less competitive industries, for firms with less analyst attention, and for state-owned enterprises (SOEs). Additionally, a better institutional environment weakens the negative impact of managerial short-termism on CSR performance. The findings shed light on policy implications for emerging countries.

The virtual inertia technology of DGs (Distributed Generations) can provide inertia and damping support for the power system by imitating the traditional synchronous machine. However, the inherent delay problems around the communication and process links of the virtual inertia control will make an impact on the fast support effect. In this paper, the virtual inertia based hierarchical control scheme considering communication delay for distributed generations integration systems is proposed. The hierarchical control architecture including the PQ primary control and the virtual inertia based secondary control method which can realize the optimal power utilization and certain frequency support under the situation of communication signal delay at the same time. Firstly, the basic hierarchical control scheme for distributed generations and corresponding small signal modeling considering communication delay are presented. Then to enhance the inertia support ability of the distributed generation integration system, an improved hierarchical control strategy considering communication delay is designed based on the robust passivity method to compensate the equivalent delay disturbance. Finally, the effectiveness verification of the proposed control is carried out with the simulation cases in PSCAD platform.

As many schools remain closed during the COVID-19 pandemic, various courses have to be migrated online. Previous studies have showed that students’ satisfaction of the courses could reflect the quality of online learning, which is determined by students’ perception of online courses. Although the community of inquiry (CoI) framework provided an effective tool for measuring students’ perception in online learning, the mechanisms between the CoI and satisfaction, especially the role of academic emotion and self-regulation, still need to be investigated in the online context. The present study aimed to (1) explore the relationships among three elements of the CoI framework; and (2) explore the relationships between the CoI and satisfaction, as well as the mediating role of academic emotion and self-regulation. The data was collected from 461 university students who were taking online courses in China. The results of Structural Equation Modeling showed that teaching presence significantly and positively predicted social presence and cognitive presence; both positive and negative academic emotions played the mediating roles between teaching presence, social presence and satisfaction; self-regulation played the mediating role between teaching presence, cognitive presence and satisfaction. The present study provided empirical evidence for the dynamics among the CoI framework as well as mechanisms between CoI and satisfaction in the online education environment.

As a biological sound source widely distributed in temperate and tropical coastal waters, snapping shrimp produce strong pulses which can serve as honest signals to indicate habitat-specific soundscape. The past decade has witnessed the growing interest in investigating the acoustical activity of snapping shrimp across many shallow waters including the coastal line of the west Pacific Ocean. It was extended to the Southeast China coastal area where snapping sounds and the associated soundscape were recorded at four sites. Customized codes incorporating bandwidth and amplitude threshold operations were developed to detect snaps from the ambient noise to estimate snap rate and extract snaps individually. The subsequent analysis suggested that snaps recorded at different sites were unanimously stronger than background noise. Sound pressure level of the snaps ranged from 150 dB to 190 dB (re 1 μPa). The characteristics of snaps, including sound pressure level, duration, peak frequency, -3dB bandwidth from different sites are examined to evaluate the variability across the sites. Though snapping pulses had peak frequencies and the -3 dB bandwidth consistently below 10 kHz, snaps had considerable energy extending to the high frequency range over 200 kHz. The analysis of the acoustic data received for 7 consecutive days at one site indicated that the snap rate corresponded to tidal level periodicity. A high tide was accompanied with a local high snap rate regardless of light but this local snap rate peak was much higher at night. The mean rate fluctuated between 2000 and 4000 snaps per minute and more snaps were recorded after sunset suggesting that snapping shrimp living in the area snapped in response to light. These data may indicate that snaps are important communication means in light-limited condition and deepen our understanding on the correlation of snapping behavior and ecological environments.

IntroductionNumerous studies have reported the modification of particulate matters (PMs) on the association between cold temperature and health. However, it remains uncertain whether the modification effect may vary by size of PMs, especially in Shandong Province, China where the disease burdens associated with cold temperature and PMs are both substantial. This study aimed to examine various interactive effects of cold exposure and ambient PMs with diameters ≤1/2.5 μm (PM1 and PM2.5) on premature deaths in Shandong Province, China.MethodsIn the 2013-2018 cold seasons, data on daily mortality, PM1 and PM2.5, and weather conditions were collected from the 1822 sub-districts of Shandong Province. A time-stratified case-crossover study design was performed to quantify the cumulative association between ambient cold and mortality over lag 0-12 days, with a linear interactive term between temperature and PM1 and PM2.5 additionally added into the model.ResultsThe mortality risk increased with temperature decline, with the cumulative OR of extreme cold (−16.9°C, the 1st percentile of temperature range) being 1.83 (95% CI: 1.66, 2.02), compared with the minimum mortality temperature. The cold-related mortality risk was 2.20 (95%CI: 1.83, 2.64) and 2.24 (95%CI: 1.78, 2.81) on high PM1 and PM2.5 days, which dropped to 1.60 (95%CI: 1.39, 1.84) and 1.60 (95%CI: 1.37, 1.88) on low PM1 and PM2.5 days. PM1 showed greater modification effect for per unit concentration increase than PM2.5. For example, for each 10?g/m3 increase in PM1 and PM2.5, the mortality risk associated with extreme cold temperature increased by 7.6% (95% CI: 1.3%, 14.2%) and 2.6% (95% CI: −0.7%, 5.9%), respectively.DiscussionThe increment of smaller PMs' modification effect varied by population subgroups, which was particularly strong in the elderly aged over 75 years and individuals with middle school education and below. Specific health promotion strategies should be developed towards the greater modification effect of smaller PMs on cold effect.

Traditional Chinese medicine (TCM) is still considered a global complementary or alternative medical system, but exogenous hazardous contaminants remain in TCM even after decocting. Besides, it is time-consuming to conduct a risk assessment of trace elements in TCMs with a non-automatic approach due to the wide variety of TCMs. Here, we present MRTCM, a cloud-computing infrastructure for automating the probabilistic risk assessment of metals and metalloids in TCM. MRTCM includes a consumption database and a pollutant database involving forty million rows of consumption data and fourteen types of TCM potentially toxic elements concentrations. The algorithm of probabilistic risk assessment was also packaged in MRTCM to assess the risks of eight elements with Monte Carlo simulation. The results demonstrated that 96.64% and 99.46% had no non-carcinogenic risk (hazard indices (HI) were 10-4 for TCM consumers, indicating that they are at potential risk for carcinogenicity. Sensitivity analysis revealed that annual consumption and concentration were the main variables affecting the assessment results. Ultimately, a priority management list of TCMs was also generated, indicating that more attention should be paid to the non-carcinogenic risks of As, Mn, and Hg and the carcinogenic risks of As and Cr in Pheretima and Cr in Arcae Conch. In general, MRTCM could significantly enhance the efficiency of risk assessment in TCM and provide reasonable guidance for policymakers to optimize risk management.

In this study, diploid, triploid, and tetraploid Crassostrea gigas samples were subjected to gas chromatography and ion mobility (GC-IMS) to identify and analyze volatile compounds and flavor fingerprints under conditions of high-temperature incubation. The GC-IMS technology identified a total of 54 volatile components in C. gigas. The contents of 1-octen-3-ol, butyl pentanoate, p-methyl anisole, and 2-methyl-2-hepten-6-one in male oysters were significantly higher than in females, while the contents of phenylacetaldehyde, benzaldehyde, 2-ethyl-3-methylpyrazine, 2-ethylfuran, and 2,4-hexadienal in female oysters were significantly higher than in males. The contents of non-3-en-2-one-M and 1-pentanol in diploids were significantly higher than in triploids and tetraploids, while the content of 2,4-hexadienal in tetraploids was significantly higher than in diploids and tetraploids. The contents of ethyl acetate, ethyl-2-butenoate, and butanal in tetraploids were significantly higher than those in diploids and triploids. The results of a principal components analysis showed that different samples were relatively independently clustered, allowing the ability to distinguish different oyster samples. The chemical fingerprints of volatile compounds of C. gigas with different ploidy and gender under high-temperature incubation were established, and the volatile substance contours of C. gigas were visualized. The results provide a reference for distinguishing the ploidy and gender of C. gigas under conditions of high-temperature incubation.

As antimicrobial resistance continues to escalate, the exploration of alternative approaches to safeguard food safety becomes more crucial than ever. Phage endolysins are enzymes derived from phages that possess the ability to break down bacterial cell walls. They have emerged as promising antibacterial agents suitable for integration into food processing systems. Their application as food preservatives can effectively regulate pathogens, thus contributing to an overall improvement in food safety. This review summarizes the latest techniques considering endolysins’ potential for food safety. These techniques include native and engineered endolysins for controlling bacterial contamination at different points within the food production chain. However, we find that characterizing endolysins through in vitro methods proves to be time consuming and resource intensive. Alternatively, the emergence of advanced high-throughput sequencing technology necessitates the creation of a robust computational framework to efficiently characterize recently identified endolysins, paving the way for future research. Machine learning encompasses potent tools capable of analyzing intricate datasets and pattern recognition. This study briefly reviewed the use of these industry 4.0 technologies for advancing the research in food industry. We aimed to provide current status of endolysins in food industry and new insights by implementing these industry 4.0 strategies revolutionizes endolysin development. It will enhance food safety, customization, efficiency, transparency, and collaboration while reducing regulatory hurdles and ensuring timely product availability.

Background: Emotional mimicry, a phenomenon frequently observed in our everyday interactions, is the act of replicating another individual’s facial expression. The Emotion Mimicry in Context View and the Correction Hypothesis underscore the critical role of context and intention within emotional mimicry. Methods: In two distinct studies, participants were presented with facial expressions of models (happiness and anger) within various contexts (affiliative, distancing, and neutral). Concurrently, we recorded electromyography (EMG) to index emotional mimicry, while participants explicitly rated the models’ intentions. Results: We observed context swiftly influences emotional mimicry within 500 ms, notably when the intentions of contexts are opposing to the intentions of facial expressions, leading to weakened muscle responses and diminished perceived intention. Furthermore, a notable correlation was discovered in the mimicry of angry faces; the more distancing the context, the stronger the corrugator supercilii (CS) muscle activity after context processing. Conclusions: First, emotional mimicry should not be simply viewed as an output corresponding to the expresser’s facial expressions but the dynamic process involving the active participation of the observer. Second, intention serves as a pivotal anchor, effectively integrating facial and contextual information. As such, we provided empirical support for the Correction Hypothesis.

Plastic products are widely used due to their lightness, durability, low cost, and malleability, and their threat to the environment is becoming increasingly apparent. However, the detailed mechanism of such an effect is unclear. Due to improper treatment, microplastic contamination is very much studied in the areas around sewage outfalls or some plastic manufacturers, and its content is high. To investigate the ecological impact of microplastics in this particular area, we evaluated the toxic effect of polystyrene microplastics (PS-MPs) on the microalgae Scenedesmus obliquus (S. obliquus). In particular, the cell growth, photosynthesis, antioxidant enzyme activity, membrane permeability, and morphology of the microalgae in the presence of different PS-MP concentrations were monitored. These new data revealed an increase in microalgae cell growth as an early response to low-dose PS-MP exposure with a maximum inhabitation rate of −3.33%. In contrast, a higher concentration of PS-MP solution leads to a significant inhibition within the test concentration, with a maximum inhabitation rate of 43.62%. Meanwhile, a decreased photosynthesis activity of S. obliquus was observed during the exposure. Moreover, PS-MPs induced oxidative damage to the algae cells, exhibiting decreased antioxidant activities and enhanced lipid peroxidation reactions, as evidenced by the reduction in the superoxide dismutase (SOD) level and increased malondialdehyde (MDA) content, as well as damaged cell membrane and soluble protein. Collectively, our study systematically investigates the toxicity of microplastics on microalgae, providing a deep insight into the possible influences of plastic pollution on the whole food web in the aquatic ecosystem.

Objective: Females are more vulnerable to renal impairment (RI) in people living with HIV (PLWH), but few studies have examined sex disparity in the association of HIV serostatus with RI. Methods: In total, 2,101 PLWH on antiretroviral treatment (ART) and 4,202 HIV-negative people were selected and frequency matched in 1:2 ratio by sex and age categories. RI was defined as estimated glomerular filtration rate (eGFR)

Abstract This randomized, double-blind, placebo-controlled, multiple ascending–dose study evaluated safety, tolerability, pharmacokinetics, and pharmacodynamics of multiple doses of milvexian, an oral small-molecule FXIa inhibitor, in healthy Japanese participants. Participants received oral milvexian daily under fasted (50 mg and 200 mg) or fed conditions (500 mg) or placebo over 14 days; 24 participants (8/cohort: 6 milvexian; 2 placebo) were planned. Due to an unblinding event, participants in one cohort (200 mg daily) were discontinued, and a second cohort enrolled; 32 participants were included in safety and pharmacodynamic analyses, and 24/32 in pharmacokinetic analyses. Milvexian up to 500 mg daily for 14 days was generally well tolerated, with no deaths, serious adverse events, or discontinuations due to adverse events. Milvexian exposure increased between 50-mg and 200-mg doses. Median Tmax was similar with 50-mg and 200-mg doses (2.5–3.0 h) and delayed under fed conditions (500 mg, 7.0–8.0 h). Median T1/2 was similar across doses (8.9–11.9 h). Multiple oral milvexian administrations resulted in concentration-related prolongation of aPTT and decreased FXI clotting activity. Milvexian was generally safe and well tolerated. The pharmacokinetic and pharmacodynamic profile of milvexian demonstrates suitability for further clinical development in Japanese participants.

Abstract Background OsWRKY62 and OsWRKY76, two close members of WRKY transcription factors, function together as transcriptional repressors. OsWRKY62 is predominantly localized in the cytosol. What are the regulatory factors for OsWRKY62 nuclear translocation? Results In this study, we characterized the interaction of OsWRKY62 and OsWRKY76 with rice importin, OsIMα1a and OsIMα1b, for nuclear translocation. Chimeric OsWRKY62.1-GFP, which is predominantly localized in the cytoplasm, was translocated to the nucleus of Nicotiana benthamiana leaf cells in the presence of OsIMα1a or OsIMαΔIBB1a lacking the auto-inhibitory importin β-binding domain. OsIMαΔIBB1a interacted with the WRKY domain of OsWRKY62.1, which has specific bipartite positively charged concatenated amino acids functioning as a nuclear localization signal (NLS). Similarly, we found that OsIMαΔIBB1a interacted with the AvrPib effector of rice blast fungus Magnaporthe oryzae, which contains a scattered distribution of positively charged amino acids. Furthermore, we identified a nuclear export signal (NES) in OsWRKY62.1 that inhibited nuclear transportation. Overexpression of OsIMα1a or OsIMα1b enhanced resistance to M. oryzae, whereas knockout mutants decreased resistance to the pathogen. However, overexpressing both OsIMα1a and OsWRKY62.1 were slightly more susceptible to M. oryzae than OsWRKY62.1 alone. Ectopic overexpression of OsWRKY62.1-NES fused gene compromised the enhanced susceptibility of OsWRKY62.1 to M. oryzae. Conclusion These results revealed the existence of NLS and NES in OsWRKY62. OsWRKY62, OsWRKY76, and AvrPib effector translocate to nucleus in association with importin α1s through new types of nuclear localization signals for negatively regulating defense responses.

Semicarbazide (SMC), also known as carbamoyl hydrazide, is a key intermediate for the organic synthesis of drugs, pesticides, and a panoply of other applications. It is also regarded as a landmark metabolite of nitrofurazone, a banned veterinary drug. SMC produced in different ways will eventually enter the ocean and become an emerging marine pollutant, affecting the physiological metabolism, behavioral activities, and even survival of aquatic organisms. Sea cucumbers are sediment-feeding organisms, and their risk of exposure to pollutants has attracted increasing attention. In this study, an integrated proteomic and metabolomic approach was used to investigate the responses of Apostichopus japonicus treated with SMC (3.72 g/L) for 72 h. After SMC treatment, the proteins and metabolites of A. japonicus intestine changed significantly. The results showed that 342 differentially expressed proteins were identified, of which 174 were upregulated, 168 were downregulated, and 74 differentially expressed metabolites, of which 62 were upregulated and 12 were downregulated. These differential proteins and metabolites were primarily involved in energy metabolism, lipid metabolism, signal transduction, immune regulation, autophagy, and apoptosis. On the basis of a combination of proteomic and metabolomic data, a hypothetical network of proteins, metabolites, and pathways in sea cucumbers was also described; the resulting network indicated several significant biological activities in response to SMC. This work offers a thorough analysis of the intricate mechanisms by which sea cucumbers respond to SMC stress and indicates numerous possible indicators for further research on creatures exposed to SMC. Further, our results provide scientific guidance for pollution control of Apostichopus japonicus culture to ensure healthy breeding.

To ensure that the potassium magnesium phosphate cement (MKPC) paste is both easily pourable and compact when repairing narrow cracks, the paste must be capable of self-compacting. Herein, the effects of fly ash (FA) on the workability, rheological properties, and early strength of the MKPC paste were tested, and the physical phase composition and microstructure of the paste were analyzed by X-ray diffraction (XRD) and scanning electron microscopy (SEM). The results suggested that the working properties of the resulting MKPC paste satisfy the index requirements of the European Design Code for Self-Compacting Concrete of European Federation of National Associations Representing for Concrete when 10 % and 20 % FA are used instead of magnesium oxide powder. The early plastic viscosity of the slurry mixed with 10 % and 20 % (M1, M2) increased by 0.114 and 0.174 Pa s, respectively, in relation to that of the MKPC paste without FA (M0). This result indicated that the FA admixture increases the plastic viscosity of the MKPC paste because FA can promote the early hydration of the MKPC paste and consume more free water. The static yield stress of M2 pastes blended with 20 % FA and the thixotropic ring area decreased by 31.89 Pa and 971.12 Pa s−1, respectively, in relation to those of M0. The addition of an appropriate amount of FA reduces the early static yield stress and improves the thixotropy of the MKPC pastes owing to the dispersing effect of spherical particles in FA and the tumbling effect. Owing to heterogeneous core-forming, active, and microaggregate effects of FA, the structure of the hydrated 1 d M2 sample was significantly denser than that of M0, which subsequently improved the 1 d strength of the hardened body.

Periodontitis is among most common human inflammatory diseases and characterized by destruction of tooth-supporting tissues that will eventually lead to tooth loss. Diabetes mellitus (DM) is a group of metabolic disorders characterized by chronic hyperglycemia which results from defects in insulin secretion and/or insulin resistance. Numerous studies have provided evidence for the inter-relationship between DM and periodontitis that has been considered as the sixth most frequent complication of DM. However, the mechanisms are not fully understood yet. The impact of DM on periodontitis through hyperglycemia and inflammatory pathways is well described, but the effects of DM on oral microbiota remain controversial according to previous studies. Recent studies using next-generation sequencing technology indicate that DM can alter the biodiversity and composition of oral microbiome especially subgingival microbiome. This may be another mechanism by which DM risks or aggravates periodontitis. Thus, to understand the role of oral microbiome in periodontitis of diabetics and the mechanism of shifts of oral microbiome under DM would be valuable for making specific therapeutic regimens for treating periodontitis patients with DM or preventing diabetic patients from periodontitis. This article reviews the role of oral microbiome in periodontal health (symbiosis) and disease (dysbiosis), highlights the oral microbial shifts under DM and summarizes the mechanism of the shifts.

Abstract This pilot study aimed to determine if a biophilic Green Therapy or Virtual Reality environment can decrease an oncology patient’s pain and distress while receiving chemotherapy. A case-crossover pilot study was conducted in a comprehensive cancer infusion center. 33 participants with breast, gynecologic, gastrointestinal, pancreatic and prostate cancers were all included in three rooms in a random order at different cycles: control room, Green Therapy room, and Virtual Reality room to receive chemotherapy, respectively. Participants’ pain, distress, heart rate, blood pressure, and saliva cortisol were measured before and after infusion in each room. No statistical significance differences were shown in the changes of heart rate, systolic, or diastolic blood pressure, saliva cortisol, pain, or distress before and after infusion between the control, Green Therapy, and Virtual Reality rooms. However, more patients reported the experience as “fun” and “enjoyable” when they were in Green Therapy or Virtual reality room as compared to in the control room. Additionally, since participating in the study, 14 patients reported spending at least 30 min or more outside in nature. In this study, we found that patients’ heart rate, blood pressure, and self-reported distress levels were reduced after each biophilic intervention although results are not statistically significant. The study also suggested that biophilic interventions are safe and feasible and may complement the standard of care for oncology patients.

Artificial modification of Bacillus thuringiensis (Bt) proteins can effectively improve their resistance to target pests, but the effect of such modification on the diversity of rhizosphere microorganisms remains unclear. Transgenic maize 2A-7 contains two artificially modified Bt proteins, mCry1Ab and mCry2Ab. These proteins can enter soil and pose a potential threat to soil microbial diversity. To assess their impacts on rhizosphere bacteria communities, the contents of the two Bt proteins and changes in bacterial community diversity in the rhizosphere soils of transgenic maize 2A-7 and its control variety were analyzed at different growth stages in 2020. The results showed that the two Bt proteins were detected at low levels in the rhizosphere soils of 2A-7 plants. No significant differences in soil bacterial diversity were detected between 2A-7 and its control variety at any of the growth stages. Bioinformatics analysis indicated that the growth stage, rather than the cultivar, was the main factor causing changes in bacterial communities. This research provides valuable data for understanding the impact of Bt crops on the soil microbiome, and establishes a theoretical basis for evaluation of their safety.

Pancreatic cancer is the third leading cause of cancer-related death in the United States. Pancreatic ductal adenocarcinoma (PDAC) is the major form of pancreatic cancer with the worst outcomes. Early detection is key to improving the overall survival rate of PDAC patients. Recent studies have demonstrated that microRNA (miRNA) signatures in plasma small extracellular vesicles (EVs) are potential biomarkers for the early detection of PDAC. However, published results are inconsistent due to the heterogeneity of plasma small EVs and the methods used for small EV isolation. We have recently refined the process of plasma small EV isolation using double filtration and ultracentrifugation. In the present study, we applied this protocol and analyzed plasma small EV miRNA signatures by small RNA sequencing and quantitative RT-PCR in a pilot cohort, consisting of patients with early-stage PDAC, and age- and gender-matched healthy subjects (n = 20). We found, via small RNA sequencing, that there are several miRNAs enriched in plasma small EVs of PDAC patients, and the levels of miR-18a and miR-106a were confirmed by quantitative RT-PCR to be significantly elevated in patients with early-stage PDAC compared with age- and gender-matched healthy subjects. Furthermore, using an immunoaffinity-based plasma small EV isolation approach, we confirmed that the levels of miR-18a and miR-106a in plasma small EVs were significantly higher in PDAC patients versus the healthy subjects. We thus conclude that the levels of miR-18a and miR-106a in plasma small EVs are promising biomarkers for the early detection of PDAC.

The leakage prediction calculation method for dynamic seal rings in underground equipment is presented in this paper. The framework of the method is given. The leakage prediction model is built. The non-Newtonian fluid interface element is brought in. The leakage prediction calculation method was developed based on the thermal–structural coupled method and the fluid–structural coupled method. A test is performed to validate the proposed method. It is proved that the film thickness of an O-ring made of nitrile rubber in pulling-in travel is thicker than that in pushing-out travel. The leakage of an O-ring made of fluororubber is larger than that of an O-ring made of nitrile rubber in the same environmental condition. The presented method is useful for predicting the sealing ability of dynamic seal rings in underground equipment. Evaluation costs will be reduced with the given leakage prediction calculation method.

Abstract Perforated wells in ultra‐deep sandstone reservoirs are characterized by high reservoir temperature, high formation pressure, and high production, and sand production will cause severe safety hazards to these wells. Based on Drucker‐Prager (DP) rock failure criterion, a practical analytical model for predicting critical drawdown pressure (CDP) of sanding onset for perforated wells is established by comprehensively considering the influence of drag stress, formation water production, reservoir pressure depletion, temperature difference in the perforated wellbore. The proposed model is verified by field sanding onset monitoring data. The calculation results show that CDP changes with well inclination, and it decreases with the increase in wellbore temperature difference, reservoir pressure depletion, and water saturation. However, the CDP increases with the number of perforations and the ratio of diameter to length. The optimization of perforation parameters has an important influence on sand control. This study can effectively guide the prediction of CDP and provides basis for efficient and safe development of similar ultra‐deep and high yield sandstone reservoirs.
