BackgroundThe association of Remnant cholesterol (RC) with renal function and its progression in patients with Type 2 diabetes (T2DM) related chronic kidney disease (CKD) remains unclear.Methods8,678 patients with T2DM-related CKD were included in cross-sectional analysis, and 6,165 patients were enrolled in longitudinal analysis and followed up for a median of 36.0 months. The outcomes were renal composite endpoint event and rapid progression of renal function.Results24.54% developed a renal composite endpoint event, and 27.64% rapid progression of renal function. RC levels above 0.56 mmol/L independently increased the risk of both renal composite endpoint (HR, 1.17; 95% CIs, 1.03-1.33) and rapid progression of renal function (OR, 1.17; 95% CIs, 1.01- 1.37). TG levels above 1.65 mmol/L only increased the risk of renal composite endpoint (HR, 1.16; 95% CIs, 1.02 -1.32). TC levels above 5.21 mmol/L increased the risk of renal composite endpoint (HR, 1.14; 95% CIs, 1.01-1.29) only in patients with proteinuria≥0.5g/d. Conversely, HDL-C levels below 1.20 mmol/L or above 1.84 mmol/L increased the risk of rapid progression of renal function (OR, 0.88; 95% CIs, 0.70 -0.99) in patients with proteinuria

Objective:The prognostic utility of inflammatory markers in survival has been suggested in patients with cancer; however, evidence on their prognostic value in severely ill patients is very limited. We aimed to explore the prognostic value of cholinesterase (ChE), C-reactive protein (CRP), interleukin-6 (IL-6), and procalcitonin (PCT) in predicting mortality in patients from the intensive care unit (ICU).Methods:Serum levels of ChE, CRP, IL-6 and PCT were measured in ICU patients from December 13th, 2019 to June 28th, 2022. We assessed the predictive power of ChE, CRP, IL-6, and PCT using the receiver operating characteristic (ROC) curves. Furthermore, we evaluated their diagnostic accuracy by comparing the areas under the ROC curve (AUCs) along with their corresponding 95% confidence intervals (CIs). The cut-off values were determined to dichotomise these biomarkers, which were then included in multivariable logistic regression models to examine their relationship with ICU mortality.Results:Among 253 ICU patients included in the study, 66 (26%) died during the ICU stay. The AUCs to predict ICU mortality were 0.643 (95% CI, 0.566-0.719), 0.648 (95% CI, 0.633-0.735), 0.643 (95% CI, 0.563-0.723) and 0.735 (95% CI, 0.664-0.807) for ChE, CRP, IL-6 and PCT, respectively. After adjusting for age, sex and disease severity, lower ChE level (10.546 mg dL-1), IL-6 (>986.245 pg mL-1) and PCT (>0.505 μg L-1) were associated with higher mortality risk, with odd ratios of 2.70 (95% CI, 1.32-5.54), 4.99 (95% CI, 2.41-10.38), 3.24 (95% CI, 1.54-6.78) and 3.67 (95% CI, 1.45-9.95), respectively.Conclusion:ChE, CRP, IL-6 and PCT were independent ICU mortality risk factors in severely ill patients. Elevated PCT levels exhibited better predictive value than the other three biomarkers that were evaluated.

The development of temperature-sensitive sensors upgraded by poly(N-isopropylacrylamide) (PNIPAM) represents a significant stride in enhancing performance and tailoring thermoresponsiveness. In this study, an array of temperature-responsive electrochemical sensors modified with different PNIPAM-based copolymer films were fabricated via a “coating and grafting” two-step film-forming technique on screen-printed platinum electrodes (SPPEs). Chemical composition, grafting density, equilibrium swelling, surface wettability, surface morphology, amperometric response, cyclic voltammograms, and other properties were evaluated for the modified SPPEs, successively. The modified SPPEs exhibited significant changes in their properties depending on the preparation concentrations, but all the resulting sensors showed excellent stability and repeatability. The modified sensors demonstrated favorable sensitivity to hydrogen peroxide and L-ascorbic acid. Furthermore, notable temperature-induced variations in electrical signals were observed as the electrodes were subjected to temperature fluctuations above and below the lower critical solution temperature (LCST). The ability to reversibly respond to temperature variations, coupled with the tunability of PNIPAM’s thermoresponsive properties, opens up new possibilities for the design of sensors that can adapt to changing environments and optimize their performance accordingly.

The Ni61Fe10Cr10Al17Mo2 high-entropy alloys (HEAs) with low density have been developed with great potentials for applications. The forging process is one of the important methods for fabricating components. Accordingly, the study of the deformation behavior of HEAs is the essence in the processing and molding of this material for using in industry. Moreover, there are multiple phases with hierarchical structures in the as-cast Ni61Fe10Cr10Al17Mo2 alloys, with the face-centered cubic (FCC) phase as the matrix phase accompanied with hard body-centered cubic (BCC) B2 phase. Here, the hot deformation behaviors of the homogenized Ni61Fe10Cr10Al17Mo2 alloy were investigated at strain rates ranging from 0.001 to 1.0 s−1 and temperatures ranging from 1000 to 1200 °C via isothermal compression tests. The true stress-strain curves were used to reveal the relationship between the flow stress and the deformation temperature and strain rate. The constitutive equations describing the correlations between the flow stress and hot compression parameters were constructed. At the same time, the dynamic material model and instability criterion were used to plot the hot processing map. Furthermore, the deformed microstructures and the dynamic recrystallization of alloy were investigated. Combined with the processing map, the optimal processing route is obtained.

Abstract The positive and negative effects of the participation of digital platform companies in governance are a major issue of the times. Starting from the three subfields of internal governance, external governance, and co-governance, we construct an analytical framework for digital platform governance to understand the four-sided relationship among digital platform companies, the state, the market, and society. From the perspective of enterprise autonomy, we should view the effectiveness of platform company participation in social governance dialectically. The key of promoting good platform governance is to improve the external structural pressure from the state, promote the reform of the endogenous governance of enterprises, and build a sustainable architecture for co-governance. The mission of sociologists includes not only explanation but also intervention.

In order to study the multi-mode damage and fracture mechanisms of thin-walled tubular parts with cross inner ribs (longitudinal and transverse inner ribs, LTIRs), the Gurson–Tvergaard–Needleman (GTN) model was modified with a newly proposed stress state function. Thus, tension damage and shear damage were unified by the new stress state function, which was asymmetric with respect to stress triaxiality. Tension damage dominated the modification, which coupled with the shear damage variable, ensured the optimal prediction of fractures of thin-walled tubular parts with LTIRs by the modified GTN model. This included fractures occurring at the non-rib zone (NRZ), the longitudinal rib (LIR) and the interface between the transverse rib (TIR) and the NRZ. Among them, the stripping of material from the outer surface of the tubular part was mainly caused by the shearing of built-up material in front of the rollers under a large wall thickness reduction (ΔT). Shear and tension deformation were the causes of fractures occurring at the NRZ, while axial tension under a large TIR interval (l) mainly resulted in fractures on LIRs. Fractures at the interface between the TIR and NRZ were due to the shearing applied by rib grooves and radial tension during the formation of ribs. This study can provide guidance for the manufacturing of high-performance aluminum alloy thin-walled tubular components with complex inner ribs.

Abstract Triglyceride–glucose (TyG) index has been proposed to be a simple, economical, and reliable marker of insulin resistance. We aimed to investigate whether TyG is an independent predictor of hyperuricemia in diabetic kidney disease (DKD) populations by conducting a cross-sectional and longitudinal study. A total of 6,471 patients were enrolled in cross-sectional analysis, and 3,634 patients without hyperuricemia at the baseline were included in longitudinal analysis and were followed up for a median of 23.0 months. Hyperuricemia was categorized as a serum uric acid level ≥ 420 umol/L (7 mg/dL). In this study, 19.58% of participants had hyperuricemia. In the cross-sectional analysis, multivariate logistics regression analysis showed that the ORs (95% CI) for hyperuricemia in the second, third, and fourth TyG quartiles were 1.40 (95% CI 0.73–2.65), 1.69 (95% CI 0.90–3.18), and 4.53 (95% CI 2.39–8.57), respectively, compared with the first quartile. Longitudinally, the Kaplan–Meier survival analysis showed that higher TyG levels predicted higher incidence of hyperuricemia. Multivariate Cox regression model revealed that the hazard ratios for hyperuricemia in the upper quartiles of the TyG index were 1.69 (95% CI 0.97–2.93), 2.23 (95% CI 1.33–3.75), and 2.50 (95% CI 1.46–4.27), respectively, compared with the first quartile. Moreover, the subgroup analyses revealed that the relationship between TyG levels and hyperuricemia was robust in DKD patients. Our findings indicate a significant independent correlation between the TyG index and the risk of hyperuricemia in DKD patients.

Abstract Background 5-hydroxytryptophan (5-HTP), the direct biosynthetic precursor of the neurotransmitter 5-hydroxytryptamine, has been shown to have unique efficacy in the treatment of a variety of disorders, including depression, insomnia, and chronic headaches, and is one of the most commercially valuable amino acid derivatives. However, microbial fermentation for 5-HTP production continues to face many challenges, including low titer/yield and the presence of the intermediate L-tryptophan (L-Trp), owing to the complexity and low activity of heterologous expression in prokaryotes. Therefore, there is a need to construct an efficient microbial cell factory for 5-HTP production. Results We describe the systematic modular engineering of wild-type Escherichia coli for the efficient fermentation of 5-HTP from glucose. First, a xylose-induced T7 RNA polymerase-P T7 promoter system was constructed to ensure the efficient expression of each key heterologous pathway in E. coli. Next, a new tryptophan hydroxylase mutant was used to construct an efficient tryptophan hydroxylation module, and the cofactor tetrahydrobiopterin synthesis and regeneration pathway was expressed in combination. The L-Trp synthesis module was constructed by modifying the key metabolic nodes of tryptophan biosynthesis, and the heterologous synthesis of 5-HTP was achieved. Finally, the NAD(P)H regeneration module was constructed by the moderate expression of the heterologous GDH esi pathway, which successfully reduced the surplus of the intermediate L-Trp. The final engineered strain HTP11 was able to produce 8.58 g/L 5-HTP in a 5-L bioreactor with a yield of 0.095 g/g glucose and a maximum real-time productivity of 0.48 g/L/h, the highest values reported by microbial fermentation. Conclusion In this study, we demonstrate the successful design of a cell factory for high-level 5-HTP production, combined with simple processes that have potential for use in industrial applications in the future. Thus, this study provides a reference for the production of high-value amino acid derivatives using a systematic modular engineering strategy and a basis for an efficient engineered strain development of 5-HTP high-value derivatives.

In order to explore the association criteria of landslide multi-field monitoring data, we have adopted the two-step clustering method and Apriori algorithm, which belong to the classical data mining method, and we have also proposed the process of landslide monitoring data mining. Based on the Baishuihe landslide in the Three Gorges Reservoir Area, we analyzed the monitoring data of ZG93 from June 2003 to June 2016. The main inducing factors of the landslide displacement were selected, and the two-step clustering method was used to pre-cluster and cluster the different influence factors. We used Apriori algorithm to deal with the classified variables to generate frequent item sets that satisfy the minimum support degree. The association rules between the precipitating factors and the landslide deformation are established under the multi-field coupling mode of Baishuihe landslide. The results show that the correlation criterion is of great significance to the deformation analysis of landslide hazards and the data mining technology can be applied to the displacement prediction of geological hazards in the Three Gorges Reservoir Area.

Abstract This article documents and conceptualizes a mode of reproduction of elites in a society in transition from state domination to market orientation. By focusing on China’ marketization, we explore how parents’ advantageous backgrounds have influenced the chance of their children’s attainment of certain elite positions (administrative, technocratic, or market) and whether these patterns have varied across three periods (1978–1992, 1993–2002, and 2003–2010). Using data obtained from the 2011 China Social Survey, we find that although parents’ advantageous status has a persistent effect on children’s status attainment, the reproduction of the state elite and market elite still follows two separate tracks: the children of cadres do not show significant advantage in the process of becoming entrepreneurs and managerial elites, and the children of entrepreneurial and managerial elites are less likely to join cadres. We also find that the effects of the reproduction model are still enhanced and shaped by state power in different periods. These findings demonstrate the important interplay between family background and contextual inequality and give a deeper understanding of the different trajectories of elites in contemporary China.

Deformation instability is a macroscopic and microscopic phenomenon of non-uniformity and unstable deformation of materials under stress loading conditions, and it is affected by the intrinsic characteristics of materials, the structural geometry of materials, stress state and environmental conditions. Whether deformation instability is positive and constructive or negative and destructive, it objectively affects daily life at all times and the deformation instability based on metal-bearing analysis in engineering design has always been the focus of attention. Currently, the literature on deformation instability in review papers mainly focuses on the theoretical analysis of deformation instability (instability criteria). However, there are a limited number of papers that comprehensively classify and review the subject from the perspectives of material characteristic response, geometric structure response, analysis method and engineering application. Therefore, this paper aims to provide a comprehensive review of the existing literature on metal deformation instability, covering its fundamental principles, analytical methods, and engineering practices. The phenomenon and definition of deformation instability, the principle and viewpoint of deformation instability, the theoretical analysis, experimental research and simulation calculation of deformation instability, and the engineering application and prospect of deformation instability are described. This will provide a reference for metal bearing analysis and deformation instability design according to material deformation instability, structural deformation instability and localization conditions of deformation instability, etc. From the perspective of practical engineering applications, regarding the key problems in researching deformation instability, using reverse thinking to deduce and analyze the characteristics of deformation instability is the main trend of future research.

Critical heat flux (CHF) determines the safety and application of pool boiling heat transfer in many industrial fields. The influence of saturation pressure and surface roughness on saturated pool boiling CHF in hydrofluoroether HFE-7100 was experimentally studied in this investigation. Visualization and heat transfer measurements were conducted at the critical and transition boiling state, and further, the accuracy of CHF prediction models and enhancement mechanism had been analyzed. The polished boiling surfaces had various surface roughness values ranging from 0.019 to 0.587 μm and their contact angles varied from 7° to 10°, while the experimental saturation pressure changed from 0.7 to 2.0 bar. The visual images showed that the pool boiling phenomenon at a critical state was composed of different-sized bubbles, vapor column, and large mushroom vapor, whereas the unsteady blanket of vapor continually injected bubbles at a transition state. The saturation pressure and surface roughness had an obvious improvement on pool boiling CHF, which might be ascribed to the effects of bubble momentum owing to evaporation, distribution and recovery period of a heat transfer boundary layer, capillary action of the working liquid, as well as ratio of vapor jets’ area. Compared with the well-known correlations reported in the literature, CHF correlation of Bailey et al. (2006) predicted the current results more accurately. To further improve the prediction accuracy, a new empirical correlation for CHF dimensionless K considering the effects of saturation pressure and surface roughness was developed, and the predicted values were in better agreement with the experimental data.

A temperature-sensitive mutant of Corynebacterium glutamicum is widely used as the fermentation strain in L-glutamate industrial production. Rapid synthesis of L-glutamate in this type of strain can be triggered by raising the culture temperature. However, the molecular mechanism of the temperature-sensitivity trait and L-glutamate excretion has not been clarified. In this study, we compared the genome sequence of an industrial L-glutamate-producing strain C. glutamicum TCCC11822 with the wild-type strain C. glutamicum ATCC13032. Seven mutant genes related to cell membrane synthesis were screened and separately integrated into the genome of C. glutamticum ATCC13032 to replace the corresponding genes. All the recombinant strains showed temperature-sensitivity and increased L-glutamate excretion. Electron microscopy showed that there was no significant change in cell morphology of recombinant strains at different temperatures. Genotypes found in this article can be applied to the construction of temperature-sensitive engineering strains and the regulation of cell permeability.

Abstract Fitting multivariate models to the China Private Enterprises Survey (2006) data, this article reveals the differentiation of state-business relationship as a function of a private enterprise’s economic capital, thereby to a certain extend challenges the dominant paradigm of clientelism. When resolving disputes with the government, private enterprises with more economic capital are more likely to directly contact the government agency, displaying greater political privilege. On the other hand, private enterprises with middle-range economic capital show higher preference to take advantage of business associations or particularistic relationships. However, instead of economic capital, it is the Communist Party membership of private entrepreneurs that influences their contentious strategies. There is also regional variation in the selection of strategies. This article casts doubt on the existence of a “capitalist class” in China. Under the specific context of the Chinese society, characteristics of the private economy influence the path by which economic power transforms to political power.

Clear cell renal cell carcinoma (ccRCC) is one of the most frequent pathological subtypes of kidney cancer, accounting for ~70–75%, and the major cause of mortality is metastatic disease. The difference in gene expression profiles between primary ccRCC tumors and metastatic tumors has not been determined. Thus, we report integrated genomic and transcriptomic analysis for identifying differentially expressed genes (DEGs) between primary and metastatic ccRCC tumors to understand the molecular mechanisms underlying the development of metastases. The microarray datasets GSE105261 and GSE85258 were obtained from the Gene Expression Omnibus (GEO) database, and the R package limma was used for DEG analyses. In summary, the results described herein provide important molecular evidence that metastatic ccRCC tumors are different from primary tumors. Enrichment analysis indicated that the DEGs were mainly enriched in ECM–receptor interaction, platelet activation, protein digestion, absorption, focal adhesion, and the PI3K–Akt signaling pathway. Moreover, we found that DEGs associated with a higher level of tumor immune infiltrates and tumor mutation burden were more susceptible to poor prognosis of ccRCC. Specifically, our study indicates that seven core genes, namely the collagen family (COL1A2, COL1A1, COL6A3, and COL5A1), DCN, FBLN1, and POSTN, were significantly upregulated in metastatic tumors compared with those in primary tumors and, thus, potentially offer insight into novel therapeutic and early diagnostic biomarkers of ccRCC.

Objective To explore possible risk factors for poor outcomes following percutaneous vertebroplasty (PV) for painful osteoporotic compression fractures of thoracolumbar vertebra. L-theanine is the most abundant free amino acid in tea that offers various favorable physiological and pharmacological effects. Bacterial enzyme of γ-glutamylmethylamide synthetase (GMAS) can catalyze the synthesis of theanine from glutamate, ethylamine and ATP, but the manufacturing cost is uncompetitive due to the expensive substrates and complex processes. In this study, we described pathway engineering of wild-type Escherichia coli for one-step fermentative production of theanine from sugars and ethylamine. First, the synthetic pathway of theanine was conducted by heterologous introduction of a novel GMAS from Paracoccus aminovorans. A xylose-induced T7 RNA polymerase-PT7 promoter system was used to enhance and control gmas gene expression. Next, the precursor glutamate pool was increased by overexpression of native citrate synthase and introduction of glutamate dehydrogenase from Corynebacterium glutamicum. Then, in order to push more carbon flux towards theanine synthesis, the tricarboxylic acid cycle was interrupted and pyruvate carboxylase from C. glutamicum was introduced as a bypath supplying oxaloacetate from pyruvate. Finally, an energy-conserving phosphoenolpyruvate carboxykinase from Mannheimia succiniciproducens was introduced to increase ATP yield for theanine synthesis. After optimizing the addition time and concentration of ethylamine hydrochloride in the fed-batch fermentation, the recombinant strain TH11 produced 70.6 ​g/L theanine in a 5-L bioreactor with a yield and productivity of 0.42 ​g/g glucose and 2.72 ​g/L/h, respectively. To our knowledge, this is the first report regarding the pathway engineering of E. coli for fermentative production of theanine. The high production capacity of recombinant strain, combined with the easy processes, will hold attractive industrial application potential for the future.

The treatment of diabetes lies in developing novel functional carriers, which are expected to have the unique capability of monitoring blood glucose levels continuously and dispensing insulin correctly and timely. Hence, this study is proposing to create a smart self-regulated insulin delivery system according to changes in glucose concentration. Temperature and glucose dual responsive copolymer microcapsules bearing N-isopropylacrylamide and 3-acrylamidophenylboronic acid as main components were developed by bottom-spray coating technology and template method. The insulinoma β-TC6 cells were trapped in the copolymer microcapsules by use of temperature sensitivity, and then growth, proliferation, and glucose-responsive insulin secretion of microencapsulated cells were successively monitored. The copolymer microcapsules showed favorable structural stability and good biocompatibility against β-TC6 cells. Compared with free cells, the biomicrocapsules presented a more effective and safer glucose-dependent insulin release behavior. The bioactivity of secreted and released insulin did not differ between free and encapsulated β-TC6 cells. The results demonstrated that the copolymer microcapsules had a positive effect on real-time sensing of glucose and precise controlled release of insulin. The intelligent drug delivery system is supposed to mimic insulin secretion in a physiological manner, and further provide new perspectives and technical support for the development of artificial pancreas.

Abstract Inhibitor of nuclear factor kappa‐B kinase subunit epsilon (IKBKE) plays critical roles in the proliferation, invasion, and epithelial‐mesenchymal transition (EMT) of glioblastoma (GBM). However, as an immune response factor, few studies have focused on the role of IKBKE in the glioblastoma‐mediated immunosuppressive microenvironment. Here, we found a higher IKBKE expression level in gliomas corresponding to higher malignancy of the tumor. The highest level of IKBKE expression was examined in the core region of GBM tissues as well as the mesenchymal subtype, which are featured with necrosis, immunocyte infiltration, and immunosuppression. Further in silico analysis demonstrated that the JAK/STAT as the signaling pathway most associated with IKBKE in mesenchymal GBM. The co‐expression patterns of IKBKE, pSTAT3, and PD‐L1 were detected within GBM tissues. Mechanistically, IKBKE could interact with STAT3 and thus enhancing the phosphorylation level of STAT3 and its nuclear translocation. In addition, pSTAT3 could transcriptionally regulate the expression of PD‐L1 through binding to its promoter. In vivo results further confirmed the inhibitory effect of the IKBKE downregulation on tumor growth. Collectively, our findings suggest IKBKE as the central node in the crosstalk between NF‐κB and STAT3 signaling within mesenchymal GBM. Targeting GBM through inhibiting IKBKE could restrain tumor growth and tumor‐mediated immunosuppressive environment.

The category and morphology of precipitates are essential factors in determining the mechanical behaviors of aluminum alloys. It is a great challenge to synthetically modulate multiple precipitates to simultaneously improve strength and ductility. In the present work, by optimizing the precipitations of the GP zone, θ’-approximant and θ’ phase for an Al-Cu-Mn alloy, a high tensile strength of 585 MPa with large elongation of 12.35% was achieved through pre-deformation and aging. The microstructure evolution pattern was revealed by detailed characterizations of scanning electron microscopy and transmission electron microscopy. It was found that such high tensile strength of the samples was due to a combination of strengthening by the high density of dispersive fine precipitates and dislocations, and the high elongation to failure was primarily attributed to the multimodal precipitates and elimination of precipitation-free zones along the grain boundaries. The strategy proposed here is a promising way of preparing ultra-strong Al-Cu-Mn alloys.

The tremendous scale of protected vegetable cultivation incidentally produces considerable vegetable residue, which refers to the remaining parts of plants after the final harvest. The low use rate of vegetable residue results in nutrient waste and environmental pressure in China. In this study, we put forward vegetable residue directly returned to the soil and investigated its feasibility. Residue return was steadily conducted 5 times in a Chinese solar greenhouse with the cucumber–tomato rotation pattern. Results showed that residue return increased the soil alkali-hydrolysed nitrogen and available potassium contents by 4.97–26.22% and 9.31–21.92%, respectively, along with slightly reduced soil pH and bulk density by 1.00–5.39% and 6.72–11.81%, respectively. Gemmatimonadetes, Firmicutes, Acidobacteria, Basidiomycota, and Mortierellomycota were the major phyla with noticeable changes when residue return was conducted 5 times. Fruit yield began to obtain remarkable increase by 5.81–9.26 t·ha−1 after residue return was conducted 3 times, bringing about additional profits of 5382.0–8519.2 USD·ha−1. Residue return could cut down the disposal expense of vegetable residues by 480.89 USD·ha−1. Moreover, residue return could supplement nutrients to soil, potentially contributing to reducing chemical fertilizer inputs. In conclusion, in situ vegetable residue return could be considered to be a feasible and sustainable use technique for vegetable residues in the Chinese solar greenhouse.

Systems metabolic engineering is a multidisciplinary area that integrates systems biology, synthetic biology and evolutionary engineering. It is an efficient approach for strain improvement and process optimization, and has been successfully applied in the microbial production of various chemicals including amino acids. In this review, systems metabolic engineering strategies including pathway-focused approaches, systems biology-based approaches, evolutionary approaches and their applications in two major amino acid producing microorganisms: Corynebacterium glutamicum and Escherichia coli, are summarized.

Poly(N-isopropylacrylamide) (PNIPAAm) is a typical thermoresponsive polymer used widely and studied deeply in smart materials, which is attractive and valuable owing to its reversible and remote “on−off” behavior adjusted by temperature variation. PNIPAAm usually exhibits opposite solubility or wettability across lower critical solution temperature (LCST), and it is readily functionalized making it available in extensive applications. Cell culture is one of the most prospective and representative applications. Active attachment and spontaneous detachment of targeted cells are easily tunable by surface wettability changes and volume phase transitions of PNIPAAm modified substrates with respect to ambient temperature. The thermoresponsive culture platforms and matching thermal-liftoff method can effectively substitute for the traditional cell harvesting ways like enzymatic hydrolysis and mechanical scraping, and will improve the stable and high quality of recovered cells. Therefore, the establishment and detection on PNIPAAm based culture systems are of particular importance. This review covers the important developments and recommendations for future work of the preparation and characterization of temperature-responsive substrates based on PNIPAAm and analogues for cell culture applications.

In the present study, a novel breeding strategy of atmospheric and room temperature plasma (ARTP) mutagenesis was used to improve the uridine production of engineered Bacillus subtilis TD12np. A high-throughput screening method was established using both resistant plates and 96-well microplates to select the ideal mutants with diverse phenotypes. Mutant F126 accumulated 5.7 and 30.3 g/L uridine after 30 h in shake-flask and 48 h in fed-batch fermentation, respectively, which represented a 4.4- and 8.7-fold increase over the parent strain. Sequence analysis of the pyrimidine nucleotide biosynthetic operon in the representative mutants showed that proline 1016 and glutamate 949 in the large subunit of B. subtilis carbamoyl phosphate synthetase were of importance for the allosteric regulation caused by uridine 5'-monophosphate. The proposed mutation method with efficient high-throughput screening assay was proved to be an appropriate strategy to obtain uridine-overproducing strain.

Large size (>10000 μm2) precipitate-free zones in the absence of microsegregation were observed in the near-β Ti-55531 titanium alloy after furnace cooling from high temperature and longtime annealing in the single-β phase field. To reveal the formation mechanism of the large size precipitate-free zone, continuous cooling and isothermal heat treatment were carried out to investigate the β-α phase transformation process. It was found that the large size precipitate free zone is attributed to the heterogeneous nucleation of α phase. The nucleation site evolves in three different modes: I-random nucleation inside the β grain, II-network nucleation inside the β grain and, III-heterogeneous nucleation on the precipitated α phase. Modes I and II lead to homogeneous transformed structure while Mode III results in the large size precipitate-free zone. Both modes II and III are promoted at high annealing temperature, rapid cooling above 600 °C or slow cooling below 600 °C. Mode II is common as it can minimize the strain energy in phase transformation. As a result, the formation of the large size precipitate-free zone is not deterministic.

Continuing efforts to develop novel smart materials are anticipated to upgrade the quality of life of humans. Thermo-responsive poly(N-isopropylacrylamide) and glucose-responsive phenylboronic acid—typical representatives—are often integrated as multi-stimuli-sensitive materials, but few are available for side-by-side comparisons with their properties. In this study, both copolymers bearing N-isopropylacrylamide (NIPAAm), with or without 3-acrylamidophenylboronic acid (AAPBA), were synthesized by free radical polymerization, and characterized by Fourier transform infrared spectrometry, nuclear magnetic resonance hydrogen spectroscopy and gel permeation chromatography. Dynamic light scattering was used to analyze and compare the responsive behaviors of the copolymers in different aqueous solutions. Atomic force microscopy was also employed to investigate the apparent morphology changes with particle sizes. The results demonstrated that the introduction of NIPAAm endowed the composite materials with thermosensitivity, whereas the addition of AAPBA lowered the molecular weight of the copolymers, intensified the intermolecular aggregation of the nanoparticles, reduced the lower critical solution temperature (LCST) of the composites, and accordingly allowed the copolymers to respond to glucose. It was also concluded that the responding of smart copolymers to operating parameters can be activated only under special conditions, and copolymer dimension and conformation were affected by inter/intramolecular interactions.

The effect of initial structure on phase evolution in continuous heating of a near-α TA15 titanium alloy (Ti-6Al-2Zr-1Mo-1V) was experimentally investigated. To this end; three microstructures were obtained by multiple heat treatment: I-bimodal structure with 50% equaixed α, II-bimodal structure with 15% equiaxed α, III-trimodal structure with 18% equiaxed α and 25% lamellar α. Differential scanning calorimetry (DSC), dilatometry and quantitative metallography were carried out on specimens with the three initial structures at heating rates from 5 to 40 °C/min. The transformation kinetics was modeled with the Johnson–Mehl–Avrami (JMA) approach under non-isothermal condition. It was found that there exists a four-stage transformation for microstructures I and III. The secondary and third stages overlap for microstructure II. The four stages of phase transformation overlap with increasing heating rate. In the presence of α laths, the phase transformation kinetics is affected by the composition difference between lamellar α and primary equiaxed α. Phase transformation is controlled by the growth of existing large β phase.

The first episode of peritonitis affects survival of the peritoneal membrane as a medium for dialysis as well as survival of patients. The aim of this study is to investigate risk factors associated with the first episode of peritonitis in Southern Chinese continuous ambulatory peritoneal dialysis (CAPD) patients.This is a single-center, retrospective, cohort study. All incident CAPD patients from 1 January 2006 to 31 December 2010 were recruited, and followed up until their first episode of peritonitis or 31 December, 2012. Baseline demographic, socioeconomic, clinical and laboratory data were collected. Cox proportional model was used to determine the factors associated with the first episode of peritonitis.In a cumulative 30756.5 patient-months follow-up (the median vintage 26.1 months) of 1117 CAPD patients, 309(27.7%) patients presented the first episodes of peritonitis. The cumulative peritonitis-free survival was 86.2%, 78.1%, 71.4% and 57.8% at 1, 2, 3 and 5 year, respectively. The multivariate analysis showed that factors associated with risk for the first episode of peritonitis were elderly patients (>65 years) [hazard ratio (HR) = 1.427, 95% confidence interval (CI) = 1.051 to 1.938, P = 0.023], male(HR = 1.315, 95% CI = 1.028 to 1.684, P = 0.030), lower education level (HR = 1.446, 95% CI: 1.127 to 1.855, P = 0.004) and albumin

To study the role of invariant Natural Killer T cell (iNKT) cells in autoimmune thyroiditis, we derived two iNKT cell lines from the spleens of NOD·H2h4 mice, a strain that develops spontaneous autoimmune thyroiditis exacerbated by excess dietary iodine. The two lines were CD1d-restricted and expressed CD4+, DX5+, and the Vα4Jα281 gene segment, of the T-cell receptor α locus. Upon stimulation with α-galactosyl-ceramide (α-GalCer), both lines rapidly produced IL-2, IL-4, IFN-γ, IL-10, and TNF-α. Strikingly, a similar cytokine response was also induced by thyroglobulin, one of the most abundant protein in the thyroid gland and a major autoantigen in human autoimmune thyroiditis. Transfer of the iNKT cell lines to syngeneic hosts enhanced autoimmune thyroiditis. Intraperitoneal injections of α-GalCer in iodine primed mice also induced thyroid disease. This paper reports for the first time that iNKT cells respond to thyroglobulin and enhance autoimmune thyroiditis in iodine fed NOD·H2h4 mice.
