Perfluorononanoic acid (PFNA), an acknowledged environmental endocrine disruptor, is increasingly utilized as a substitute for perfluorooctanesulfonic acid (PFOS) and perfluorooctanoic acid (PFOA). Despite its growing use, limited research has been conducted to investigate its potential impact on tumorigenesis and progression, and the potential molecular mechanisms. Earlier studies linked perfluoroalkyl and polyfluoroalkyl substances (PFAS) exposure to breast and gynecological cancer progression in humans, lacking a clear understanding of the underlying mechanisms, notably in ovarian cancer. Our investigation into PFNA's effects at environmental concentrations (0.25–2 mM) showed no significant impact on cell proliferation but a notable increase in invasion and migration of ovarian cancer cells. This led to alterations in epithelial-mesenchymal transition (EMT) markers, including Claudin1, Vimentin, and Snail. Notably, PFNA exposure activated the TGF-β/SMADs signaling pathway. Crucially, SMAD7 degradation through the ubiquitin-proteasome system emerged as PFNA's pivotal molecular target for inducing EMT, corroborated in mouse models. In summary, this study presented evidence that environmentally relevant concentrations of PFNA could induce SMAD7 degradation via the proteasome pathway, subsequently activating the TGF-β/SMADs signaling pathway, and promoting EMT in ovarian cancer. These results illuminated the association between PFNA exposure and metastasis of ovarian cancer.

Abstract Background Ovarian cancer is commonly associated with a poor prognosis due to metastasis and chemoresistance. PINK1 (PTEN-induced kinase 1) is a serine/threonine kinase that plays a crucial part in regulating various physiological and pathophysiological processes in cancer cells. Methods The ATdb database and "CuratedOvarianData" were used to evaluate the effect of kinases on ovarian cancer survival. The gene expression in ovarian cancer cells was detected by Western blot and quantitative real-time PCR. The effects of gene knockdown or overexpression in vitro were evaluated by wound healing assay, cell transwell assay, immunofluorescence staining, immunohistochemistry, and flow cytometry analysis. Mass spectrometry analysis, protein structure analysis, co-immunoprecipitation assay, nuclear-cytoplasmic separation, and in vitro kinase assay were applied to demonstrate the PINK1-PTEN (phosphatase and tensin homolog) interaction and the effect of this interaction. The metastasis experiments for ovarian cancer xenografts were performed in female BALB/c nude mice. Results PINK1 was strongly associated with a poor prognosis in ovarian cancer patients and promoted metastasis and chemoresistance in ovarian cancer cells. Although the canonical PINK1/PRKN (parkin RBR E3 ubiquitin protein ligase) pathway showed weak effects in ovarian cancer, PINK1 was identified to interact with PTEN and phosphorylate it at Serine179. Remarkably, the phosphorylation of PTEN resulted in the inactivation of the phosphatase activity, leading to an increase in AKT (AKT serine/threonine kinase) activity. Moreover, PINK1-mediated phosphorylation of PTEN impaired the nuclear import of PTEN, thereby enhancing the cancer cells’ ability to resist chemotherapy and metastasize. Conclusions PINK1 interacts with and phosphorylates PTEN at Serine179, resulting in the activation of AKT and the inhibition of PTEN nuclear import. PINK1 promotes ovarian cancer metastasis and chemotherapy resistance through the regulation of PTEN. These findings offer new potential therapeutic targets for ovarian cancer management.

Abstract Hypobaric hypoxia (HH) exposure affects appetite and serum iron levels in both humans and animals. Thus, whether appetite-regulating ghrelin is involved in iron regulation under HH needs to be elucidated. In vivo, C57BL/6J mice were placed in a hypobaric chamber to establish a 6000-m-high altitude exposure animal model. In vitro, mouse primary hepatocytes and peritoneal macrophages were exposed to hypoxia (1% O2) to examine the effects of ghrelin on iron-regulating proteins. HH obviously reduced the body weight of mice and significantly increased the levels of erythrocytes, and also significantly enhanced the levels of serum iron and plasma ghrelin. However, iron content in the liver and spleen was decreased, while ferroportin (Fpn) expression was increased. Moreover, ghrelin significantly induced Fpn and pERK expression in both hepatocytes and macrophages under hypoxia, which were reversed by pretreatment with growth hormone secretagogue receptor 1a (GHSR1a) antagonist or pERK inhibitor. Our findings indicated that HH leads to decreased appetite and insufficient dietary intake, which may negatively regulate the levels of ghrelin. Furthermore, GHSR1a/ERK signalling pathway is further activated to upregulate the expression of Fpn, and then promoting iron mobilization both in the liver/hepatocytes and spleen/macrophages in mice. Thus, these results revealed that ghrelin may be a potential iron regulatory hormone, and raised the possibility of ghrelin as a promising therapeutic target against iron disorders under HH.

Inland waterway transport is an important mode of transportation for many countries and regions. Route planning optimization can reduce navigation time, avoid traffic congestion, and improve transportation efficiency. In actual operations, many vessels determine their navigation routes based on the experience of their shipowners. When the captain fails to obtain accurate information, experience-based routes may pose significant navigation risks and may not consider the overall economic efficiency. This study proposes a comprehensive method for optimizing inland waterway vessel routes using automatic identification system (AIS) data, considering the geographical characteristics of inland waterways and navigation constraints. First, AIS data from vessels in inland waters are collected, and the multi-objective Peak Douglas–Peucker (MPDP) algorithm is applied to compress the trajectory data. Compared to the traditional DP algorithm, the MPDP algorithm reduces the average compression rate by 5.27%, decreases length loss by 0.04%, optimizes Euclidean distance by 50.16%, and improves the mean deviations in heading and speed by 23.53% and 10.86%, respectively. Next, the Ordering Points to Identify the Clustering Structure (OPTICS) algorithm is used to perform cluster analysis on the compressed route points. Compared to the traditional DBSCAN algorithm, the OPTICS algorithm identifies more clusters that are both detailed and hierarchically structured, including some critical waypoints that DBSCAN may overlook. Based on the clustering results, the A* algorithm is used to determine the connectivity between clusters. Finally, the nondominated sorting genetic algorithm II is used to select suitable route points within the connected clusters, optimizing objectives, including path length and route congestion, to form an optimized complete route. Experiments using vessel data from the waters near Shuangshan Island indicate that, when compared to three classic original routes, the proposed method achieves path length optimizations of 4.28%, 1.67%, and 0.24%, respectively, and reduces congestion by 24.15%. These improvements significantly enhance the planning efficiency of inland waterway vessel routes. These findings provide a scientific basis and technical support for inland waterway transport.

Wheat dwarf virus (WDV, genus Mastrevirus, family Geminiviridae) is one of the causal agents of wheat viral disease, which severely impacts wheat production in most wheat-growing regions in the world. Currently, there is little information about natural resistance against WDV in common wheat germplasms. CRISPR/Cas9 technology is being utilized to manufacture transgenic plants resistant to different diseases. In the present study, we used the CRISPR/Cas9 system targeting overlapping regions of coat protein (CP) and movement protein (MP) (referred to as CP/MP) or large intergenic region (LIR) in the wheat variety ‘Fielder’ to develop resistance against WDV. WDV-inoculated T1 progenies expressing Cas9 and sgRNA for CP/MP and LIR showed complete resistance against WDV and no accumulation of viral DNA compared with control plants. Mutation analysis revealed that the CP/MP and LIR targeting sites have small indels in the corresponding Cas9-positive plants. Additionally, virus inhibition and indel mutations occurred in T2 homozygous lines. Together, our work gives efficient results of the engineering of CRISPR/Cas9-mediated WDV resistance in common wheat plants, and the specific sgRNAs identified in this study can be extended to utilize the CRISPR/Cas9 system to confer resistance to WDV in other cereal crops such as barley, oats, and rye.

Objectives This study aimed to evaluate the effectiveness of the Trauma Rating Index in Age, Glasgow Coma Scale, Respiratory rate and Systolic blood pressure score (TRIAGES) in predicting 24-hour in-hospital mortality among patients aged 65 years and older with isolated traumatic brain injury (TBI).Design A retrospective, single-centre cohort study.Setting This study was conducted at a government-run tertiary comprehensive hospital.Participants This study included 982 patients aged 65 years or older with isolated TBI, who were admitted to the emergency department between 1 January 2020 and 31 December 2021.Interventions None.Primary outcome 24-hour in-hospital mortality was the primary outcome.Results Among the 982 patients, 8.75% died within 24 hours of admission. The non-survivors typically had higher TRIAGES and lower GCS scores. Logistic regression showed significant associations of both TRIAGES and GCS with mortality; the adjusted ORs were 1.98 (95% CI 1.74 to 2.25) for TRIAGES and 0.72 (95% CI 0.68 to 0.77) for GCS. Receiver operating characteristic (ROC) analysis indicated an area under the ROC curve of 0.86 for GCS and 0.88 for TRIAGES, with a significant difference (p=0.012). However, precision–recall curve (PRC) analysis revealed an area under the PRC of 0.38 for GCS and 0.47 for TRIAGES, without a significant difference (p=0.107).Conclusions The TRIAGES system is a promising tool for predicting 24-hour in-hospital mortality in older patients with TBI, demonstrating comparable or slightly superior efficacy to the GCS. Further multicentre studies are recommended for validation.

Abstract Background Clear cell renal cell carcinoma (ccRCC) is one of the most common malignancies. Recently, immunotherapy has been considered a promising treatment for metastatic ccRCC. NUF2 is a crucial component of the Ndc80 complex. NUF2 can stabilize microtubule attachment and is closely related to cell apoptosis and proliferation. This research is dedicated to investigating the role of NUF2 in ccRCC and the possible mechanisms. Methods First, analysis of NUF2 mRNA expression levels in ccRCC and normal tissues by The Cancer Genome Atlas (TCGA) database and further verified by analysis of independent multiple microarray data sets in the Gene Expression Omnibus (GEO) database. Moreover, we evaluated and identified correlations between NUF2 expression, clinicopathologic variable, and overall survival (OS) in ccRCC by various methods. We investigated the relationship between NUF2 and tumor immune infiltration and the expression of corresponding immune cell markers via the Gene Expression Profiling Interactive Analysis (GEPIA) and Tumor Immune Estimation Resource (TIMER) databases. Then, we performed functional enrichment analysis of NUF2 co-expressed genes using R software and protein-protein interactions (PPIs) using the search tool used to retrieve interacting genes/proteins (STRING) databases. Results We discovered that NUF2 mRNA expression was upregulated in ccRCC tissues and was associated with sex, grade, pathological stage, lymph node metastasis, and worse prognosis. In addition, NUF2 was positively linked to tumor immune cells in ccRCC. Moreover, NUF2 was closely related to genetic markers of different immune cells. Finally, functional enrichment and protein–protein interaction (PPI) analysis suggested that NUF2 and its closely related genes may be involved in the regulation of the cell cycle and mitosis. Our results suggested that NUF2 is correlated with a poor prognosis and immune infiltration in ccRCC.

To systematically evaluate the differences in therapeutic response to chemotherapy or immunotherapy between different molecular subtypes of bladder cancer (BC). A comprehensive literature search was performed up to December 2021. Consensus clusters 1 (CC1), CC2 and CC3 molecular subtypes were used to perform meta-analysis. Pooled odds ratios (ORs) with 95% confidence intervals (CIs) were used to assess the therapeutic response by fix-effect modeling. Eight studies involving 1,463 patients were included. The automatic identification system (AIS) provides a massive database for ocean science. The original AIS data are redundant. Direct use will cause a waste of data storage space and computation costs; hence, data compression must be performed. The Douglas–Peucker algorithm (DP) is an effective trajectory compression algorithm that can well preserve the spatial characteristics of a trajectory but has the following shortcomings: first, it has poor track recovery when compressing multi-turn routes; second, it does not consider the ship speed and heading; and third, it may have the wrong result of the compressed trajectory crossing the obstacle. To address these situations, this study proposes a multi-objective peak DP algorithm (MPDP) that adopts a peak sampling strategy, considers three optimization objectives (spatial characteristics, heading and speed) of trajectory and adds an obstacle detection mechanism to realize a compression algorithm more suitable for curved trajectories. The classical DP algorithm is compared with the MPDP algorithm by simulating trajectory and real trajectory experiments. The results show that the MPDP algorithm optimizes the length loss rate, simultaneous Euclidean distance, and average deviations of the speed and the heading while maintaining a high compression rate similar to that of the DP algorithm. Moreover, it can also successfully avoid obstacles. The optimization effect is most obvious for the multi-turn or hovering trajectory. The optimization rate of length loss, synchronous Euclidean distance, and average deviation of the heading can reach 40%.

Automatic Identification System (AIS) can obtain a large amount of data on ship trajectories and movement characteristics, which provides data support for ship route planning, navigation safety, and maritime traffic control. Therefore, it is of great importance to analyze and cluster the AIS data to obtain an understanding of the movement behavior of ships. The Density-Based Spatial Clustering of Applications with Noise (DBSCAN) algorithm is capable of discovering arbitrarily shaped clusters and is suitable for clustering ship trajectories. However, the traditional DBSCAN algorithm suffers from the following shortcomings. First, the DBSCAN algorithm requires the input of a similarity matrix between the trajectories, and the similarity matrix obtained from the traditional Hausdorff distances does not identify the trajectory directions. Second, the DBSCAN algorithm is sensitive to the input parameters, and the clustering results obtained from different datasets with the same parameters vary widely. Finally, the DBSCAN algorithm is poor at clustering multi-density distribution datasets. To address these shortcomings, this study proposes a multi-density adaptive trajectory clustering (MDA-Traclus) algorithm, which considers the multi-density distribution trajectory dataset, adaptively determines the input parameters, and adds a trajectory direction identification mechanism to realize the clustering of trajectory clusters with different directions and different densities. Here, the classical DBSCAN algorithm, KANN-DBSCAN algorithm, and MDA-Traclus algorithm are compared and analyzed using four manually labeled trajectory datasets and a real trajectory dataset. The results show that the MDA-Traclus algorithm and KANN-DBSCAN algorithm are able to automatically determine the input parameters when clustering trajectory datasets with uniformly distributed densities and achieve the same clustering effect as the DBSCAN algorithm. When the trajectory density distribution is not uniform and the directions of the trajectory clusters are not consistent, the MDA-Traclus algorithm can effectively identify the low-density regions, differentiate the different directions of trajectories, and improve the clustering of the trajectories.

Per- and polyfluoroalkyl substances (PFAS), the versatile anthropogenic chemicals, are popular with the markets and manufactured in large quantities yearly. Accumulation of PFAS has various adverse health effects on human. Albeit certain members of PFAS were found to have genotoxicity in previous studies, the mechanisms underlying their effects on DNA damage repair remain unclear. Here, we investigated the effects of Perfluorodecanoic acid (PFDA) on DNA damage and DNA damage repair in ovarian epithelial cells through a series of in vivo and in vitro experiments. At environmentally relevant concentration, we firstly found that PFDA can cause DNA damage in primary mouse ovarian epithelial cells and IOSE-80 cells. Moreover, nuclear cGAS increased in PFDA-treated cells, which leaded to the efficiency of DNA homologous recombination (HR) decreased and DNA double-strand breaks perpetuated. In vivo experiments also verified that PFDA can induce more DNA double-strand breaks lesions and nuclear cGAS in ovarian tissue. Taken together, our results unveiled that low dose PFDA can cause deleterious effects on DNA and DNA damage repair (DDR) in ovarian epithelial cells and induce genomic instability.

In recent years, the rapid development of artificial intelligence algorithms has promoted the intelligent transformation of the ship industry; unmanned surface vessels (USVs) have become a widely used representative product. The dynamic window approach (DWA) is an effective robotic collision avoidance algorithm; however, there are deficiencies in its application to the ship field. First, the DWA algorithm does not consider International Regulations for Preventing Collisions at Sea (COLREGs), which must be met for ship collision avoidance to ensure the navigational safety of the USV and other ships. Second, the DWA algorithm does not consider the influence of wind and waves on the collision avoidance of USVs in actual navigational environments. Reasonable use of windy and wavy environments not only improves navigational safety but also saves navigational time and fuel consumption, thereby improving the economy. Therefore, this paper proposes an improvement algorithm by DWA referred to as utility DWA (UDWA) based on COLREGs considering the sailing environment. The velocity sampling area was improved by dividing the priority, and the velocity function in the objective function was enhanced to convert the effect of wind and waves on the USVs into a change in velocity. The simulation results showed that the UDWA algorithm optimized the distance to the obstacle ship by 43.25%, 31.36%, and 67.81% in a head-on situation, crossing situation, and overtaking situation, respectively, compared to the COLREGs-compliant DWA algorithm, which considers the COLREGs. The improved algorithm not only follows the COLREGs but also has better flexibility in emergency collision avoidance and can safely and economically navigate and complete collision avoidance in windy and wavy environments.

Alterations in tryptophan (Trp) metabolism facilitate the continuous modulation of tumor progression, including tumor growth, distant metastasis, and chemoresistance development. Although there is a high correlation between Trp metabolism and tumor progression, it is unknown whether and how Trp metabolism affects the development of prostate cancer. In this study, we reported that the overexpression of Trp hydroxylase 1 (TPH1) caused the upregulation of Trp hydroxylation and mediated the production of 5-hydroxytryptamine (5-HT), contributing to tumor growth and poor prognosis in patients with prostate cancer. An increase in 5-HT levels triggered the activation of the Axin 1/β-catenin signaling pathway, thus enhancing cell proliferation and migration. Consequently, β-catenin cooperated with the Krüppel-type zinc finger family transcription factor ZBP-89 to upregulate TPH1 expression, further promoting Trp hydroxylation and forming the TPH1/5-HT/β-catenin/ZBP-89/THP1 positive feedback signaling loop. Interruption of the signaling loop by the THP1 inhibitor 4-chloro-dl-phenylalanine (PCPA) significantly improved anticancer effects and suppressed lung metastasis in prostate cancer–bearing mice. Our findings revealed a mechanism by which TPH1 promotes prostate cancer growth by inducing Trp hydroxylation and identified a novel THP1 target for an innovative prostate cancer therapeutic strategy.

Based on meteorological observations, traffic flow data and information of traffic accidents caused by fog or agglomerate fog along the expressways in Jiangsu Province and Anhui Province in China from 2012 to 2021, key impact factors including meteorological conditions, road hidden dangers and traffic flow conditions are integrated to establish the prediction model for risk levels of expressway agglomerate fog-related accidents. This model takes the discrimination of the occurrence conditions of agglomerate fog as the starting term, and determines the hazard levels of agglomerate fog-related accidents by introducing the probability prediction value of meteorological conditions for fog-related accident as the disaster-causing factor. On this basis, the hourly road traffic flow and the location of road sections with a hidden danger of agglomerate fog are taken as traffic and road factors to construct the correction scheme for the hazard levels, and the final predicted risk level of agglomerate fog-related accident is obtained. The results show that for the criteria of disaster-causing factor classification threshold, 72.3% of fog-related accidents correspond to a hazard of a medium level or above, and 86.2% of the road traffic flow conditions are consistent with the levels of the traffic factor defined based on parametric indexes. For risk level prediction, six out of the seven agglomerate fog-related accidents correspond to the level of higher risk or above, which can help provide meteorological support for traffic safety under severe weather conditions. Moreover, the model takes into account the impacts of traffic flow and the road environment, which is conducive to further improving the reliability of the risk assessment results.

Based on the pavement temperature observation data of the transportation meteorological stations along the Xianyang Airport Expressway, China, as well as the datasets of precipitation and sunshine hours obtained from the nearby weather stations, the variation characteristics of local pavement temperatures are investigated for winter in this study. Results indicate that during the daytime, the pavement temperatures are always higher on sunny and cloudy days than those on rainy and snowy days, while during the nighttime, the temperatures on sunny and cloudy days are higher than those on the days with freezing rain and snow, and with the temperatures on rainy and snowy days without icing being further higher. In general, the pavement temperatures in winter features significant periodic oscillations with cycles of roughly 24 h, 12 h, 8 h, 6 h, 5 h and 4 h, which differ slightly at different times for different stations. Moreover, the nowcasting experiments on the local pavement temperatures are also carried out using a regression model via extracting the corresponding periodic features. It shows the mean absolute errors of about 0.6 °C, 1.2 °C, and 1.5 °C for lead times of 1 h, 2 h, and 3 h, respectively. The nowcasting skills are higher on rainy and snowy days, while are inferior on sunny days. For nowcasting cases initialized at nighttime (daytime), the mean absolute errors are 0.4 °C (0.7 °C) and 0.9 °C (1.4 °C) for lead times of 1 h and 2 h. Examinations suggest that the nowcasting system could be well utilized in plain areas of China, whereas it shows relatively larger biases in plateau areas with complex terrain.

Objectives Our study aimed to support evidence-informed policy-making on patient-centred care by investigating preferences for healthcare services among hypertension patients.Design We identified six attributes of healthcare services for a discrete choice experiment (DCE), and applied Bayesian-efficient design with blocking techniques to generate choice sets. After conducting the DCE, we used a mixed logit regression model to investigate patients’ preferences for each attribute and analysed the heterogeneities in preferences. Introduction: Given the ongoing coronavirus disease 2019 (COVID-19) pandemic and the consequent global healthcare crisis, there is an urgent need to better understand risk factors for symptom deterioration and mortality among patients with COVID-19. This systematic review aimed to meet the need by determining the predictive value of chronic diseases for COVID-19 severity and mortality.Methods: We searched PubMed, Embase, Web of Science, and Cumulative Index to Nursing and Allied Health Complete to identify studies published between December 1, 2019, and December 31, 2020. Two hundred and seventeen observational studies from 26 countries involving 624,986 patients were included. We assessed the risk of bias of the included studies and performed a cumulative meta-analysis.Results: We found that among COVID-19 patients, hypertension was a very common condition and was associated with higher severity, intensive care unit (ICU) admission, acute respiratory distress syndrome, and mortality. Chronic obstructive pulmonary disease was the strongest predictor for COVID-19 severity, admission to ICU, and mortality, while asthma was associated with a reduced risk of COVID-19 mortality. Patients with obesity were at a higher risk of experiencing severe symptoms of COVID-19 rather than mortality. Patients with cerebrovascular disease, chronic liver disease, chronic renal disease, or cancer were more likely to become severe COVID-19 cases and had a greater probability of mortality.Conclusions: COVID-19 patients with chronic diseases were more likely to experience severe symptoms and ICU admission and faced a higher risk of mortality. Aggressive strategies to combat the COVID-19 pandemic should target patients with chronic diseases as a priority.

Triphenyltin (TPT) is a widely used pesticide that is highly toxic to a variety of organisms, including humans, and is a potential contributor to environmental pollution. The present study was conducted to evaluate the oxidative stress and immunotoxicity induced by TPT in goldfish (Carassius auratus) and the protective effects of fructooligosaccharide (FOS). Goldfish (mean weight of 13.3 ± 0.2 g) were randomly divided into six groups with three replicates: (G1) the control group, (G2) the 10 ng/L TPT group, (G3) the 0.4% FOS group, (G4) the 10 ng/L TPT + 0.4% FOS group, (G5) the 0.8% FOS group, and (G6) the 10 ng/L TPT + 0.8% FOS group. The results showed that 10 ng/L TPT induced oxidative stress and significantly decreased the activities of antioxidant enzymes, such as superoxide dismutase (SOD), catalase (CAT) and glutathione peroxidase (GPx), in the liver and the gene expression of SOD, GPx, metallothionein (MT), and peroxiredoxin-4 (Prdx-4). The concentration of malondialdehyde (MDA) and the gene expression of cytochrome P450 (CYP) and glutathione S-transferase (GST) in the liver were significantly increased in the TPT-treated group. Exposure to 10 ng/L TPT in water induced immune suppression and significantly decreased the activities of immune enzymes, such as lysozyme, myeloperoxidase (MPO), alternative complement (ACH50), acid phosphatase (ACP) and alkaline phosphatase (AKP), in the serum. TPT could stimulate the fish to generate large amounts of proinflammatory cytokines, including increased tumor necrosis factor-α (TNF-α), interleukin-6 (IL-6), interleukin-1β (IL-1β), and nitric oxide (NO) levels and TNF-α, IL-6, IL-1β, and NF-κB mRNA expression. However, TPT-induced toxicity was significantly ameliorated in the groups treated with FOS, and FOS partly prevented alterations in the activities of antioxidant enzymes and the expression of antioxidant- and ROS scavenger-related genes. In addition, TPT-induced immune toxicity was significantly ameliorated in the groups treated with FOS. FOS markedly suppressed TNF-α, IL-6, IL-1β, and NO production and TNF-α, IL-6, and IL-1β mRNA expression in the TPT-treated groups. The study indicated that TPT-induced oxidative stress may play a critical role in inhibiting immunity. However, FOS administration attenuates TPT-induced oxidative stress and immune suppression in goldfish.

Parkinson’s disease (PD) is a neurodegenerative disorder, and the hallmarks of this disease include iron deposition and α-synuclein (α-syn) aggregation. Hepcidin could reduce iron in the central and peripheral nervous systems. Here, we hypothesized that hepcidin could further decrease α-syn accumulation via reducing iron. Therefore, rotenone or α-syn was introduced into human neuroblastoma SH-SY5Y cells to imitate the pathological progress of PD in vitro. This study investigated the clearance effects of hepcidin on α-syn induced by a relatively low concentration of rotenone exposure or α-syn overexpression to elucidate the potential clearance pathway involved in this process. We demonstrated that SH-SY5Y cell viability was impaired after rotenone treatment in a dose-dependent manner. α-syn expression and iron content increased under a low concentration rotenone (25 nM for 3 days) treatment in SH-SY5Y cells. Pre-treatment with hepcidin peptide suppressed the abovementioned effects of rotenone. However, hepcidin did not affect treatment with rotenone under high iron conditions. Hepcidin also played a role in reducing α-syn accumulation in rotenone and α-syn overexpression conditions. We identified that the probable clearance effect of hepcidin on α-syn was mediated by the autophagy pathway using pretreatment with autophagy inhibitors (3-MA and CQ) and detection of autophagy protein markers (LC3II/I and p62). In conclusion, hepcidin eliminated α-syn expression via the autophagy pathway in rotenone-treated and α-syn overexpression SH-SY5Y cells. This study highlights that hepcidin may offer a potential therapeutic perspective in α-syn accumulation diseases.

Abstract Objective Traumatic brain injury (TBI) results not only in gray matter damage, but also in severe white matter injury (WMI). Previous findings support hypoxic preconditioning (HP) could augment the efficacy of bone marrow stromal cell (BMSC) transplantation in a TBI mouse model. However, whether HP‐treated BMSCs (H‐BMSCs) could overcome remyelination failure after WMI is unclear, and the molecular mechanisms remain to be explored. Here, we focused on the therapeutic benefits of H‐BMSC transplantation for treating WMI, as well as its underlying mechanisms. Methods In vitro, BMSCs were incubated at passage 4 in the hypoxic preconditioning (1.0% oxygen) for 8 hr. In vivo, a TBI mouse model was established, and DMEM cell culture medium (control), normal cultured BMSCs (N‐BMSCs), or H‐BMSCs were transplanted to mice 24 hr afterward. Neurobehavioral function, histopathological changes, and oligodendrogenesis were assessed for up to 35 days post‐TBI. Results Compared with the control group, improvement of cognitive functions and smaller lesion volumes was observed in the two BMSC‐transplanted groups, especially the H‐BMSC group. H‐BMSC transplantation resulted in a greater number of neural/glial antigen 2 (NG2)–positive and adenomatous polyposis coli (APC)–positive cells than N‐BMSC transplantation in both the corpus callosum and the striatum. In addition, we observed that the expression levels of hypoxia‐inducible factor‐1a (HIF‐1α), phosphorylated mechanistic target of rapamycin (p‐mTOR), and vascular endothelial growth factor (VEGF) were all increased in H‐BMSC–transplanted mice. Furthermore, the mTOR pathway inhibitor rapamycin attenuated the impact of HP both in vivo and in vitro. Conclusion The results provided mechanistic evidences suggesting that HP‐treated BMSCs promoted remyelination partly by modulating the pro‐survival mTOR/HIF‐1α/VEGF signaling pathway.

The Yuanba Gas Field is a large gas field in the marine strata with the largest burial depth in China up to date. The studies performed on the mentioned gas field have achieved significant progress made possible by determining the characteristics of reef flat reservoirs in the marine strata and the main controlling factors of hydrocarbon accumulation. However, there is no consensus on the origin of natural gases stored in the reservoir in the Changxing-Feixianguan Formations in the Yuanba and its adjacent area. The study on geochemical characteristics indicates that natural gases in the reservoir in the Changxing-Feixianguan Formations in the Yuanba Gas Field are mainly composed of alkane gases which are dominated by methane with dryness coefficients generally higher than 0.995. The CO2 and H2S display an average content of 8.55% and 6.47%, respectively. The δ13C1 and δ13C2 values are from −31.2‰ to −27.9‰, and from −29.9‰ to −25.0‰, respectively, displaying the positive carbon isotopic series. The δ13CCO2 values are generally higher than −8‰, and the δD1 values are from −156‰ to −107‰. The identification of gas origin and gas–source correlation indicate that the natural gases reservoired in the Changxing-Feixianguan Formations in the Yuanba Gas Field have been altered by thermochemical sulfate reduction, and they are mainly composed of oil cracking gases derived from the secondary cracking of oil generated by the sapropelic-prone source rocks in the Permian Longtan Formation. The CO2 in the gas pools are mainly inorganic and were derived from the interaction between the acidic fluid and carbonate reservoirs.

The effects of two representative dissolved organic matters (DOMs) (derived from corrupt plants (PDOM) and chicken manure (MDOM)) on sorption characteristic of oxytetracycline to three typical sediments (first terrace (FT), river floodplain (RF), and riverbed (RB) sediments collected from the Weihe River) were investigated. Results showed that both DOMs can make the adsorption equilibrium time advance about 6 hours. The presence of DOMs changed the sorption kinetics model and the spontaneous degree of the reaction but did not change the sorption isotherm models. The adsorption of oxytetracycline (OTC) could be promoted by adding PDOM, and its maximum adsorption amount increased by 23.8% for FT, 38.0% for RB, and 28.3% for RF, respectively, whereas MDOM could inhibit the adsorption and maximum adsorption amount decreased by 23.3% for FT, 11.6% for RB, and 16.1% for RF, respectively. In addition, the DOM concentration also affected the adsorption. Overall, this study suggests that the humus-like DOM can promote the adsorption of OTC while the protein-like DOM can inhibit the adsorption of OTC to sediments, which is determined by the aromaticity, hydrophilicity, and polarity of the DOMs.

Urban agglomerations mirror the highest level of spatial organization in urban development. They are characterized by one or more mega-cities serving as central hubs within a delineated region, upheld by three or more major cities. With advanced transportation, communication, and other facilities as the foundation, these agglomerations establish close spatial and economic ties, achieving a high degree of urbanization and integration. Against the backdrop of global and anti-globalization forces leading to shifts in foreign situations, coupled with domestic economic downturns resulting from changes in pandemic policies, the imperative to form an integrated regional economic society has become increasingly prominent. This study takes the construction and development of the Yangtze River Delta Urban Agglomeration (YRDUA) as an example, examining the necessity and urgency of promoting regional economic and social integration through the proportional changes in the three major industries of each city. As the earliest and representative urban agglomeration in national planning, the YRDUA serves as an illustrative model. The findings indicate that applying the experiences of regional economic and social integration in the YRDUA to the entire country is representative and offers valuable insights.
