Sperm motility is vital to human reproduction, and malformed sperm flagella can cause male infertility. Individuals with multiple morphological abnormalities of the flagella mostly have absent, short, coiled, bent, and/or irregular-caliber flagella. In this study, a patient with male infertility underwent a physical examination along with his wife. Genetic testing was performed by whole-exome sequencing of the couple, and Sanger sequencing was performed for validation. Novel biallelic variations in the DNAH1: (NM_015512.4) gene consisting of c.1336G>C (p.E446Q) and c.2912G>A (p.R971H) were identified. In silico structural analysis revealed that the amino acid residues affected by the variation were evolutionarily conserved, and the variant p.R971H influenced the stability of the DNAH1 protein. Morphological studies of the patient's sperm showed defects in its flagella. Results of Papanicolaou staining and scanning electron microscopy demonstrated coiled and short flagella with multiple anomalies. Transmission electron microscopy of the sperm flagella showed that the inner dynein arm and radial spoke were absent, and the dense fiber and microtubule doublets were displaced. Quantitative PCR of the mRNA of the patient's sperm showed that the expression of DNALI1 was dramatically reduced. Collectively, these findings elucidated the genetic cause of the family's infertility and provided insight into the functioning of the DNAH1 gene.

Objective: To explore the clinical value of X-ray digital tomosynthesis（DTS） in the diagnosis of knee joint fractures. Methods: A total of 28 cases of thoracic trauma, X-ray film cannot be clearly diagnosed or can confirm the diagnosis but the need for further identification of forensic diagnosis of cases of DTS scan and three-dimensional reconstruction in order to control the study. Results: 1. The reconstructed images after DTS scanning showed that the knee joint fractures were clearly diagnosed, and the detection rate of 28 knee joint fractures was 92.86%. 2. DTS scanning could clearly detect the fresh knee joint fracture’s fracture line, corresponding line and broken bones. For the old fracture, DTS scanning could clearly show the condition of the fracture end’s healing. Conclusions: The technique is of great value in the diagnosis of knee joint fracture, especially in the examination of complex structure, thick body and review of internal fixation after fracture.

Location-Based Services (LBSs) have become one of the most popular activities in our daily life. With the rapid advance of LBSs, there are more threats to users’ privacy. For this reason, while enjoying the convenience provided by LBSs, we have to protect our location privacy. In this paper, we propose two all-dummy-based location privacy algorithms to achieve k-anonymity for privacy-area aware users in LBSs. Different from previous work, on the client side, our method can prevent the center attack and border attack through transforming the actual location to an anchor and the request we send to the LBSs doesn’t contain the actual location of the user. On the server side, we use a rough query before precise query to reduce the processing time and transmission bandwidth. It only returns the results that the client needs. Evaluation results show that our methods can provide more effective privacy protection and lower computation and communication cost.

Objective Microvesicles (MV) released from blood cells play an important role in the progress of diseases. The purpose of this study is to detect circulating MV level of patients suffering from severe fever with thrombocytopenia syndrome (SFTS). Methods The plasma samples of SFTS patients and healthy controls were collected. And MV from samples were isolated and MV levels were detected quantitatively. Results The results showed that the level of the circulating MV in SFTS patients was obviously higher than that of the healthy control, with statistically significant difference. Further analysis revealed that MV level was relevant to the severity of SFTS patients, namely the higher the concentration of MV, the more severity of the disease. Linear correlation analysis showed that the circulating MV level in SFTS was correlated positively with leukocyte count (r = 0.243, P < 0.05), but no correlation with the platelet count (r = 0.193, P >0.05). Conclusions We demonstrated that there existed high level of circulating MV in SFTS patients, and the level of circulating MV had a close relationship with the severity of the disease and leukocyte count. Therefore, monitoring the level of circulating MV may provide a new indicator for predicting the prognosis of SFTS patients.

An emerging infectious disease was identified as severe fever with thrombocytopenia syndrome (SFTS) in central China since late March 2009. We found the patients with SFTS had severe clinical symptoms, and progressed rapidly to multiple organ dysfunction syndrome (MODS) with high fatality rate of 25%-30%. The aim of this study was to assess the significance of risk factors predicting the development of MODS and death in SFTS patients. Consecutive SFTS admissions between May 2009 and September 2011 were analyzed for parameters of organ function during hospitalization using Marshall scoring system for MODS, and platelet counts were recorded on admission and at 24, 48, 72 h and one week after admission. We investigated the kinetics of organ failures and analyzed the association between age, platelet count and development of MODS or death. A total of 92 SFTS patients were enrolled in this study. Among them, 32 patients with dysfunction of over 4 organs were identified, 45% of them died within 72 h, 72% died within 5 days, and 76% died within 7 days after admission. To analyze the relevant documents of hospital infection administration of acupuncture manipulation with filiform needles and acupuncture aseptic technique with filiform needles. The current situation is that acupuncturists have understanding insufficiency in hospital infection management, lack the sterile concepts and consciousness of disinfection and isolation. Aseptic technic principles aren't strictly followed; disinfection and isolation systems are unsound; sanitary condition of hand of medical staff is unsatisfied; and there is shortness in traditional long filiform needle manipulation. In future, we should explore the new model of hospital infection administration of acupuncture manipulation with filiform needles from implementations of relevant rules of hospital infection administration, establishment and supervision of sound corresponding system, further research of manipulation of filiform needles and formulation of septic technic criterion of filiform needles.
