Sanggenon C is a flavonoid extracted from the root bark of white mulberry, which is a traditional Chinese medicine with anti-inflammatory, antioxidative, and antitumor pharmacological effects. In this study, sanggenon C was found to inhibit human gastric cancer (GC) cell proliferation and colony formation, induce GC cell cycle arrest in the G0-G1 phase, and promote GC cell apoptosis. Moreover, sanggenon C was found to decrease the level of mitochondrial membrane potential in GC cells and inhibit mitochondrial fission. Mechanistically, RNA sequencing, bioinformatics analysis, and a series of functional analyses confirmed that sanggenon C inhibited mitochondrial fission to induce apoptosis by blocking the extracellular regulated protein kinases (ERK) signaling pathway, and constitutive activation of ERK significantly abrogated these effects. Finally, sanggenon C was found to suppress the growth of tumor xenografts in nude mice without obvious side effects to the vital organs of animals. This study reveals that sanggenon C could be a novel therapeutic strategy for GC treatment.

We investigated the influence of DNA fragmentation index (DFI) on in vitro fertilization (IVF), embryo transfer (ET), and intracytoplasmic sperm injection (ICSI). We analyzed the semen parameters of 61 cycles in infertile couples undergoing IVF-ET and ICSI and determined DFI by sperm chromatin dispersion testing. Based on DFI, the patients were differentiated into a control group (DFI < 25%, n = 35) and a test group (DFI ≥ 25%, n = 26). Flow cytometry and immunofluorescence were used to investigate the extent of sperm reactive oxygen species (ROS) and apoptosis. We also investigated the effect of DFI on pregnancy outcomes of IVF-ET/ICSI. DFI was negatively related to sperm motility and positively correlated with ROS and apoptosis (P < 0.05). Abnormally elevated DFI reduced the rate of transplantable, high-quality embryos, implantation, clinical pregnancy, delivery, and live birth after IVF-ET, and increased the chance of early abortion per transfer cycle (P < 0.05). However, there was no significant correlation between DFI and fertilization rate, cleavage rate, transplantable rate, high-quality embryo rate, implantation rate, clinical pregnancy rate, early abortion rate, delivery rate and live birth rate when assisted by ICSI (P > 0.05). Sperm DNA integrity is crucial for fertilization and the development of healthy offspring. ROS may increase the level of DFI by inducing apoptosis in sperm.

Solid solution ceramics of (1-x)Sr0.75Ba0.25Nb2O6-xPbZr0.52Ti0.48O3 (x = 0−0.4) were prepared and investigated. The phase evolves from tungsten bronze for x = 0, coexisted tungsten bronze and pyrochlore at x = 0.3, to pyrochlore for x> 0.3. As the results, the ceramics show dramatically composition dependent microstructure and electrical property. The grain shape changes from anisotropic pillar-like with maximum length-diameter ratio of 3.3 at x = 0.2 to isotropic cubic-like for x> 0.2. At the same time, the maximum dielectric constant temperature tends to decrease with an exceptional increase at x = 0.3 due to the coexisted phases. Interestingly, the x = 0.3 ceramic endures ultrahigh breakdown strength of 340 kV/cm, which, together with enhanced polarization, results in maximum recoverable energy storage density of 3.0 J/cm3and energy storage efficiency of 81.5 % at room temperature.

Originally considered to be a plant pathogen, reports of phaeohyphomycosis due to Curvularia lunata (C. lunata) in animals and humans are increasing. However, studies on the pathogenesis, virulence, and epidemiology of C. lunata have rarely been discussed. In the present study, BALB/c mice were experimentally inoculated with C. lunata suspension by different routes and the course of infection was evaluated. In addition, the in vitro antifungal susceptibility of C. lunata against six commonly used antifungals was evaluated using the microdilution method. Inoculation resulted in skin lesions in animals inoculated intraperitonially and subcutaneously. Infection was confirmed by both mycological and histopathologic examination. C. lunata spores and hyphae were detected in the histopathologic sections stained with hexamine silver staining. In addition, voriconazole (VRC) demonstrated greater activity against C. lunata when compared to the other antifungals, whereas fluconazole (FLC) was the least active antifungal with a minimum inhibitory concentration (MIC) range of 8–16 μg/mL. Further studies are necessary to understand the pathogenicity of C. lunata and uncover the mystery of this fungus.

(1− x)Sr0.75Ba0.25Nb2O6– xNaNbO3 ( x = 0–0.8) ceramics were prepared by the traditional solid-state reaction method. The x = 0–0.4 ceramics preserve the tungsten bronze phase with a gradually unfilled–filled structure evolution accompanying a relaxor-normal ferroelectric transition. This transition process is accompanied by an increase in the maximum dielectric constant temperature or Curie temperature ( Tc), enhancement of room temperature ferroelectric and piezoelectric properties, and appearance of the stripe domain. On the other hand, ceramics with x > 0.4 consist of coexisting tungsten bronze and perovskite phases, for which Tc slightly fluctuates, and the ferroelectric and piezoelectric properties decrease with increasing x. This work provides a feasible way to manipulate the relaxor-normal ferroelectric transition and the electrical properties of the tungsten bronze family.

To observe the effect of norcantharidin (NCTD) on collagen-induced arthritis (CIA) rats. Sixty Sprague-Dawley(SD) rats were randomly divided into 6 groups (n=10): normal group, CIA model group(model group), NCTD low-dose group [1.35 mg/(kg•d)], NCTD middle-dose group [2.7 mg/(kg•d)], NCTD high-dose group [5.4 mg/(kg•d)] and methotrexate (MTX) group [1.8 mg/(kg/w)]. Anesthetized rats were sacrificed by luxation of cervical vertebra after 4 weeks of administration. The arthritis scores were evaluated twice a week. The pathological changes in the ankle joints of rats were observed by hematoxylin-eosin (H&E) staining. The serum levels of interleukin (IL) 1β, IL-6, tumor necrosis factor (TNF)-α, vascular endothelial growth factor (VEGF), IL-17 and transform growth factor (TGF) β were detected by enzyme linked immunosorbent assay (ELISA). The mRNA expression of retinoid-related orphan nuclear receptorγt (RORγt) and forkhead box P3 (Foxp3) in peripheral blood lymphocytes were confirmed by real-time polymerase chain reaction. The oriental fruit moth (OFM), Grapholita molesta (Lepidoptera: Tortricidae), is a serious pest of stone and pome fruit trees worldwide. G. molesta can switch hosts and produce multiple generations per year. Both the primary and secondary host plants of G. molesta emit aldehydes. Here, we tested whether these aldehydes are helpful for host switching of G. molesta. SPME (Solid Phase Microextraction)-GC–MS analysis identified four aldehydes (octanal, nonanal, decanal, and benzaldehyde) from volatiles of late-season yellow peach fruits. Of the four aldehydes, eletroantennogram analysis showed that nonanal produced the strongest response from the antennae of virgin and mated females. In Y-tube bioassays, only nonanal was attractive to mated G. molesta females amongst the four aldehydes, suggesting that nonanal is important for host recognition in G. molesta. In a field trial, each of the four aldehydes was tested as an attractant for both sexes of G. molesta. The traps baited with nonanal captured more female G. molesta, as compared to traps with other aldehydes and control; the traps baited with benzaldehyde captured most male G. molesta. Our research investigations provide theoretical foundation for biological control of G. molesta females. Future G. molesta lure designs should take into account sex-specific differences in the response to individual aldehydes.

The aim of this paper was to study the influence of triptolide in the immune response pathways of acquired immune deficiency syndrome( AIDS). Target proteins of triptolide and related genes of AIDS were searched in PubChem and Gene databases on line. Molecular networks and canonical pathways comparison analyses were performed by bioinformatics software( IPA). There were 15 targets proteins of triptolide and 258 related genes of AIDS. Close biological relationships of molecules of triptolide and AIDS were established by networks analysis. There were 21 common immune response pathways of triptolide and AIDS,including neuroinflammation signaling pathway,Th1 and Th2 activation pathway and role of pattern recognition receptors in recognition of bacteria and viruses. Triptolide stimulated immune response pathways by the main molecules of IFNγ,JAK2,NOD1,PTGS2,RORC. IFNγ is the focus nodes of triptolide and AIDS,and regulates genes of AIDS directly or indirectly. Triptolide may against AIDS by regulating molecules IFNγ in immune response pathways.

Rosai–Dorfman disease (RDD) with isolated central nervous system (CNS) involvement is an extremely rare disease. Most RDD of the CNS present as dural-based mass mimicking meningioma and other common lesions, which makes preoperative accurate diagnosis of great difficulty. We searched the pathology database in our hospital and 3 cases of RDD with isolated CNS involvement were finally included in our study. Radiological and clinical findings of these three cases were retrospectively analyzed. The lesions of 2 cases were dura-based against the cerebral convexity, presenting as a sheet-shaped thickened dura mater, another case was located just across the cerebral falx, the dural display in the center was intact. The 3 cases showed low signal intensity on T2-weighted image, obviously enhanced, significantly surrounding edema and finger-like protuberance but no invasion of the brain parenchyma or no sign of hyperplasia or sclerosis of the surrounding cranial bones. In conclusion, when we come across a disease that mimicking meningioma, especially when it manifests as the above radiological features, we should considered it might be a kind of proliferative disease of the meninges, such as RDD.

Constructing composite structure is an effective way to improve dielectric energy storage properties. However, ionic diffusion between composite members is generally inevitable, which mixes the effects of solid solution with composite structure on property to some extent. To exclude the ionic diffusion effect as much as possible, antiferroelectric (Pb0.97La0.02)(Zr0.5Sn0.4Ti0.1)O3 (PLZST) was sintered at a relatively high temperature to precipitate SnO2 particles, then more SnO2 were introduced to from (1-x)PLZST:xSnO2 (x = 0–0.25) composite. The results showed that composite structure had significant effects on microstructures and properties with changed lattice parameter, decreased grain size, increased Curie temperature, suppressed antiferroelectric property, and enhanced electric breakdown strength. The optimal composition of x = 0.2 showed high energy storage density of 2.38 J/cm3. This work provides a robust platform to investigate the effect of composite microstructure on electrical properties, which is helpful for further developing high performance composite materials.

Fine particulate matter (PM2.5) is a significant environmental pollutant responsible for a number of human diseases. Ginsenoside Rg1 (Rg1) is likely to have the potential to relieve PM2.5‑induced cell injury. The present study is designed to preliminarily observe the harmful effect of PM2.5 and the protective effect of Rg1 against PM2.5 on human A549 lung epithelial cells in vitro. The cytotoxic effects of the PM2.5 or Rg1 on A549 cells were measured by means of cell viability, and then exposure concentration of PM2.5 and pretreatment concentration of Rg1 used in the following assays were established. The A549 cells were pretreated with Rg1 for 1 h and then exposed to PM2.5 for 24 h. The levels of lactate dehydrogenase (LDH) in the cell culture supernatant and malondialdehyde (MDA) within the cells were assayed. The present results revealed that 200‑1,200 µg/ml of PM2.5 decreased the viability of A549 cells significantly in a concentration‑dependent manner; however, 50‑400 µg/ml of Rg1 had no significant effect. Pretreatment with 100, 200 or 400 µg/ml Rg1 significantly diminished the 200 µg/ml PM2.5‑induced A549 cell viability and decreased LDH leakage and MDA generation in a concentration‑dependent manner. These results indicated that PM2.5 induced cell injury and Rg1, antagonized PM2.5‑induced cell injury to a certain extent.

Background Diazotrophic (nitrogen-fixing) Gram-positive and endospore-formed Paenibacillus spp. have potential uses as a bacterial fertilizer in agriculture. The transcriptional analysis of nitrogen fixation in Paenibacillus is lacking, although regulation mechanisms of nitrogen fixation have been well studied in Gram-negative diazotrophs. Results Here we report a global transcriptional profiling analysis of nitrogen fixation in Paenibacillus sp. WLY78 cultured under N2-fixing condition (without O2 and NH4+) and non-N2-fixing condition (air and 100 mM NH4+). The nif (nitrogen fixation) gene operon composed of 9 genes (nifBHDKENXhesAnifV) in this bacterium was significantly up-regulated in N2-fixing condition compared to non-N2-fixing condition, indicating that nif gene transcription is strictly controlled by NH4+ and O2. qRT-PCR confirmed that these nif genes were differently expressed. Non-nif genes specifically required in nitrogen fixation, such as mod, feoAB and cys encoding transporters of Mo, Fe and S atoms, were coordinately transcribed with nif genes in N2-fixing condition. The transcript abundance of suf operon specific for synthesis of Fe-S cluster was up-regulated in N2-fixing condition, suggesting that Sul system, which takes place of nifS and nifU, plays important role in the synthesis of nitrogenase. We discover potential specific electron transporters which might provide electron from Fe protein to MoFe protein of nitrogenase. The glnR whose predicted protein might mediate nif transcription regulation by NH4+ is significantly up-regulated in N2-fixing condition. The transcription levels of nitrogen metabolism and anaerobic respiration were also analyzed. Conclusions The nif gene operon (nifBHDKENXhesAnifV) in Paenibacillus sp. WLY78 is significantly up-regulated in N2-fixing condition compared to non-N2-fixing condition. Non-nif genes specifically required in nitrogen fixation were also significantly up-regulated in N2-fixing condition. Fur and Fnr which are involved in anaerobic regulation and GlnR which might mediate nif gene transcription regulation by NH4+ were significantly up-regulated in N2-fixing condition. This study provides valuable insights into nitrogen fixation process and regulation in Gram-positive firmicutes. The dose calculation parameters of 103Pd radioactive seed need to be determined strictly before clinical application for tumor implanted treatment. The radial dose function and the anisotropy factor are data analyzed and nonlinear fitted based on the value of dose parameters in American Association of Physicists in Medicine (AAPM) TG-43 and TG-43U1 by Cftool and SFTool toolbox in Matlab. Result was selected according to the optimum principle of fitting accuracy that is the coefficient of determination is close to 1 and gets the minimum value of the sum of square of deviations in fitting process. The calculation of the radial dose function and the anisotropy factor result are obtained in an ideal situation, and then the dose calculation of 103Pd radioactive seed is determined. The calculation of 103Pd radioactive seed provides a theoretical basis for 103Pd implanted treatment of tumor in clinical.

Traditional medical extracts are commonly used as complex mixtures, which may contain naturally occurring contact sensitizers. In this investigation, the mice local lymph node assay (LLNA) was performed to evaluate the dermal sensitization potential of Myrrh, Borneolum, Olibanum, Moschus and Cassia Bark, which are widely used in topical traditional medication. In the radioactive LLNA, the stimulation index (SI) values were calculated for each medical extract. Myrrh, Borneolum, Olibanum and Moschus induced dose-dependent cell proliferation and SI was more than 3. Cassia Bark showed no positive response over the range of test concentrations. In the flow cytometry analysis, the total number of CD3+, CD4+, and CD8+ cells in local lymph nodes was increased in Moschus-, Olibanum-, Myrrh- and Borneolum-treated mice. The ratio of the B220+/CD3+ (B/T cell ratio) and the percentage of I-Ak+ cells that was also positive for the CD69 marker (I-Ak+/ CD69+) were increased in the Moschus-, Olibanum- and Myrrh-treated mice. However, no ofbvious change was observed in Borneolum-treated mice. Cassia Bark did not induce changes in the lymphocyte subpopulations. These results indicate that Moschus, Olibanum and Myrrh can be regarded as sensitizers, and Borneolum regarded as an irritant. Cassia Bark is neither a sensitizer nor an irritant. The combination of radioactive and flow cytometric LLNA can be used for the prediction of sensitizing potential of medical extracts which lead to allergic contact dermatitis in humans.

Purpose: Chemokine receptors are implicated in metastasis of several malignant tumors. This study was done to evaluate the contribution of chemokine receptors CXCR4 and CCR7 to metastasis of human nasopharyngeal carcinoma. Experimental Design: Reverse transcription-PCR, immunohistochemistry, and flow cytometry were used to evaluate mRNA and protein expression of CXCR4 and CCR7 in nasopharyngeal carcinoma tumor tissues and cell lines. Chemotaxis assays were used to evaluate the function of CXCR4 in nasopharyngeal carcinoma cells. Antisense CXCR4 was used to inhibit receptor expression and to block metastasis of human nasopharyngeal carcinoma cells in vivo in athymic mice. Results: CXCR4 protein was detected in tumor cells in 31 of 40 primary human nasopharyngeal carcinoma and in 13 of 15 lymph node metastases. CXCR4 transcripts were detected in eight CXCR4 protein–positive primary nasopharyngeal carcinoma tissues and seven nasopharyngeal carcinoma cell lines tested. On the other hand, the transcripts for CCR7 were detected only in four primary nasopharyngeal carcinoma tissues and in none of the nasopharyngeal carcinoma cell lines. In functional experiments, metastatic nasopharyngeal carcinoma cell lines that expressed high levels of CXCR4 were found to migrate in response to the CXCR4 ligand SDF-1α. Transfection of antisense CXCR4 in metastatic nasopharyngeal carcinoma cells inhibited the expression of CXCR4 and SDF-1α-induced cell migration in vitro and reduced the capacity of the tumor cells to form metastasis in the lungs and lymph nodes when injected in athymic mice. Conclusion: The expression of functional CXCR4 but not CCR7 is correlated with the metastatic potential of human nasopharyngeal carcinoma cells. Therefore, CXCR4 may be considered as a potential target for the prevention of nasopharyngeal carcinoma metastasis.

Controlled clinical trials of integrative therapies available to patients with rheumatoid arthritis (RA) improved dramatically in the past 20 years, largely because of the growing need and the methodologies improvement. Tripterygium wilfordii Hook. F., a typical example of popular use herb, has been extensively studied in trials. However, clear and convincing evidence of integrative therapy, effectiveness and safety, remains insufficient to make decision. Many research efforts are hampered by standing problems with ‘syndrome’ recruitment failure. In addition, the outcome multiplicity induces the findings inefficiency to generalize to RA patients at large. Development of validated syndrome outcomes and methodologies has also been critical. Current efforts to enhance the understanding of integrative treatment options for patients with RA include the development of drug-specific rather than disease-specific strategies, studies in predictive biomarkers, and development of peer-review trial protocol for regular clinical trials.

One of effective measures for controlling toxic reactions is to use toxic herbs according to corresponding indication syndrome. It is important to develop toxicity theory of Chinese medicine in a sound and international way using modern language to elucidate its scientific connotation. We expect to explain scientific connotation of controlling toxic reaction while toxic herbs are used to the indication syndrome by using holistic research ideas and methods capable of reflecting governing exterior to infer interior, establish appropriate corresponding syndrome animal models by cutting into dose-effect/toxicity of toxic Chinese herbs, construct and analyze multi-layer molecular network using theories and technologies of metabonomics, network biology, and bioinformatics.

A laboratory-scale intermittently aerated membrane bioreactor was applied to treat synthetic household wastewater characterized with low organic matter and high nitrogen concentration. Experimental results showed that at the condition of HRT 12 h, aeration/non-aeration cycle 30 min/60 min and a very long SRT, more than 90% of COD and close 100% ammonia nitrogen could be removed, and the average removal efficiency of TN was more than 80%, but this treatment process seemed to be not available in phosphorus removal. Nitrification was accomplished quickly and completely in the system. The denitrification appeared to be the rate-limiting step for TN removal in the system. Furthermore, a positive linear correlation between the fouling rate and the MLSS concentration was founded, and the higher the MLSS concentration was, the higher fouling rate was.

To obtain the NPC-associated antigens, a powerful new method, SEREX (serological identification of antigen by recombinant cDNA expression library), was used for identifying the antigens eliciting humoral immune response. Before performing serological analysis, a high quality cDNA library derived from human nasopharyngeal carcinoma (NPC) tissue was constructed. The primary library consisted of 3.64 x 10(6) recombinants and the recombinant rate was 94%. For better preserving the cDNA library, it was amplified. As a result, the titer of the amplified cDNA library was 3.8 x 10(9) pfu/mL. With SEREX method, immunoscreening for the detection of reactive clones in the human NPC tissue cDNA library was performed with autologous serum. As a result, 23 positive clones encoding antigenic genes were obtained after immunoscreening, and the nucleotide sequences of cDNA inserts were determined and analyzed with BLAST software in GenBank. Results showed that the 23 reactive clones were derived from 16 different genes. 10 of 16 genes had high homologous to the genes known in GenBank, such as RPL31, S100 A2, MT2A, etc. However, there were also 6 genes with low homology to the genes known in GenBank. Furthermore, 3 of 6 genes may be novel genes. The associations of these genes to NPC and the roles that they played in the occurrence and development of NPC should be revealed by further research.

Mind evolutionary computation (MEC) is a new approach of evolutionary computation. In this paper, it is adopted to train the weights of artificial neural network (ANN) to solve premature convergence problem of BP algorithm and genetic algorithm. The coding method of taking individual weights as the center of normal distribution is proposed, and information of network weights is used. Dynamic searching method is used, and weights are trained successfully. The simulation result shows that the new method is better than the common BP algorithm and genetic algorithm.

Mind evolutionary computation (MEC) is a novel stochastic algorithm that derived from man's swarm intelligence. Based on the swarm theory, the social behavior analysis about MEC is made and the searching mechanisms of its operators are studied. After that, a parameter analysis is provided for the similar axis operator, and a cooperation-based dissimilation operator (CDO) is developed. Finally, a series of experiments have been done to make a parameter choice and an evaluation for MEC. The results illustrate MEC with CDO is a viable global optimization method owning robust ability.

Low antigenicity or development of tolerance is believed to be a major contributor to the escape of malignant tumors from immune surveillance of the host. However, anti-tumor responses can be elicited by concomitant immunization of poorly antigenic tumor cells with homologous xenogeneic proteins as 'altered self' proteins. In our study, anti-tumor, but not anti-xenogeneic antigen, immune responses were generated after transduction of the gene coding for a G-protein coupled human formyl peptide receptor like-1 (FPRL1) into a mouse C26 colon cancer cell line. C26 cells transfected with FPRL1 gene exhibited markedly reduced tumorigenicity in syngeneic mice, in association with the appearance of high levels of antibody activity reacting with both FPRL1 containing and wild type C26 cells. The anti-tumor responses required the participation of CD4+ T lymphocytes, since no tumor rejection was observed in nude mice or in syngeneic mice depleted of CD4+ T cells. Furthermore, mice primed with FPRL1 transfected C26 cells were resistant to subsequent challenge by wild type C26 cells. These results indicate that the presence of human FPRL1 is capable of triggering specific anti-tumor host immune responses against poorly antigenic mouse tumor cells.

To construct phage human anti-idiotypic antibody library.Peripheral blood mononuclear cells(PBMCs) of patients with nasopharyngeal cancer(NPC) were sensitized in-vitro and transformed by Epstein-Barr virus(EBV). V(H) and V(L) genes were re-amplified by PCR and combined to single-chain fragment of variable region(ScFv) genes. ScFv genes were cloned into vector fUSE5 and transformed into MC1061 by electroporation to construct the ScFv-displaying phage library.Detection of Sandwich ELISA showed that of 10 NPC patients,8 patients' B cells transformed by EBV could produce anti-idiotypic antibodies to NPC. 5 types of V(H) genes and 7 types of V(L) genes were obtained by PCR re-amplification and then connected with (Gly(4)Ser)(3) linker to form 14 types of ScFv genes. ScFv genes digested with Sfi I were cloned into vector fUSE5 and transformed into MC1061 via electroporation. Phage anti-idiotypic antibody library with sink size being 1.1x10(7) was obtained through tetracycline-resistant secreening. The percentage of full-length ScFv gene inserted into phage DNA was 70%.A strategy for preparing human single chain anti-idiotypic antibody by means of phage antibody library technique in combination with EBV transformation technique is feasible.

The potential of anti-idiotypic antibody as a surrogate of tumor antigen for cancer therapy has been demonstrated in clinical investigations. But at present, many anti-idiotypic antibodies are mouse-original antibodies, which can cause human anti-mouse antibody (HAMA) response and decrease the curative effect. The objective of this study was to construct phage human anti-idiotypic antibody library and select beta type anti-idiotypic single chain antibodies bearing the internal image of the nasopharyngeal carcinoma (NPC) associated antigen to overcome human anti- mouse antibody response caused by application of mouse-original anti-idiotypic antibody.Peripheral blood mononuclear cells (PBMCs) of patients with NPC were immunized in vitro by anti-NPC monoclonal antibody FC2 and transformed by Epstein-Barr virus (EBV). V(H) and V(L) genes were amplified by RT-PCR and combined to single chain fragments of variable region (scFv) genes. ScFv genes were cloned into vector fUSE5 and transformed into E.coli MC1061 to construct the scFv-displaying phage library. After four rounds of panning with monoclonal antibody (mAb) FC2,the beta type Ab2 scFv were selected by Sandwich ELISA and binding inhibition test.Of 10 NPC patients, 8 patients showed their B cells immunized by FC2 and transformed by EBV produced anti-idiotypic antibodies to NPC. Five types of VH genes and 7 types of V(L) genes were obtained by RT-PCR amplification and then connected to form 14 scFv genes. ScFv genes were transducted into E.coli MC1061. The library capacity was 1.5x10(8) clones. After panning, 270 phage clones were selected randomly and 91 FC2-positive clones were obtained by Sandwich ELISA, the positive ratio was 33.7%. Five clones,which might display beta type Ab2 scFv, were selected by binding inhibition test.The strategy for preparing phage anti-idiotypic antibody library and selecting beta type Ab2 scFv by immunization in vitro, EBV transformation, and phage display technique is feasible, which provide a way for preparing cancer vaccine using beta type Ab2 scFv.

This paper presents a constraint multi-objective artificial physics optimisation (CMOAPO) algorithm by introducing a novel optimisation paradigm called artificial physics optimisation (APO) into constraint multi-objective domain. Combining with characteristics of constraint multi-objective optimisation problems, a method of virtual force decreasing is incorporated into CMOAPO to decrease the probability of individuals moving from feasible region into infeasible region. Furthermore, the convergence of CMOAPO is analysed in terms of theory with related knowledge of probability. The performance of CMOAPO algorithm is tested using several benchmark functions. The results obtained show that the proposed approach is effective.
