Objective Percutaneous repair is an alternative to open surgical repair of the Achilles tendon with comparable, functional results and low re‐rupture and infection rates; however, sural nerve injury is a known complication. The purpose of this study is to design a new surgical procedure, the minimally invasive peritendinous submembrane access technique (MIS‐PSAT). It offers optimal results, with excellent functional outcomes, and with minimal soft tissue complications and sural nerve injury. Methods This retrospective study included 249 patients with acute closed Achilles tendon ruptures treated at our institution between 2009 and 2019. All patients underwent MIS‐PSAT at our institution and were followed up for 8–48 months. Functional evaluation was based on the Achilles tendon total rupture score (ATRS) and the American Orthopedic Foot and Ankle Society Ankle‐Hindfoot Scale (AOFAS‐AHS), associated with local complications and isokinetic tests. Results None of the patients had infection, necrosis, or sural nerve injury. Re‐rupture occurred in two cases. The average times to return to work and sports was 10.4 and 31.6 weeks, respectively. The average ATRS and AOFAS‐AHS scores were 90.2 and 95.7, respectively, with an excellent rate of 99.5%. Isokinetic tests showed that ankle function on the affected side was comparable with that on the healthy side (p > 0.05). Conclusion The MIS‐PSAT for acute Achilles tendon rupture is easy to perform with few complications. Importantly, the surgical technique reduces the risk of sural nerve injuries. Patients have high postoperative satisfaction, low re‐rupture rates, and muscle strength, and endurance can be restored to levels similar to those on the healthy side.

Background and aims: The serum metabolites changes in patients with hepatitis B virus (HBV)-related cirrhosis as progression. Peroxisome proliferator-activated receptor gamma (PPARγ) is closely related to lipid metabolism in cirrhotic liver. However, the relationship between fatty acids and the expression of hepatic PPARγ during cirrhosis regression remains unknown. In this study, we explored the serum metabolic characteristics and expression of PPARγ in patients with histological response to treatment with entecavir.Methods: Sixty patients with HBV-related cirrhosis were selected as the training cohort with thirty patients each in the regression (R) group and non-regression (NR) group based on their pathological changes after 48-week treatment with entecavir. Another 72 patients with HBV-related cirrhosis and treated with entecavir were collected as the validation cohort. All of the serum samples were tested using ultra-performance liquid chromatography coupled to tandem mass spectrometry. Data were processed through principal component analysis and orthogonal partial least square discriminant analysis. Hepatic PPARγ expression was observed using immunohistochemistry. The relationship between serum fatty acids and PPARγ was calculated using Pearson’s or Spearman’s correlation analysis.Results: A total of 189 metabolites were identified and 13 differential metabolites were screened. Compared to the non-regression group, the serum level of fatty acids was higher in the R group. At baseline, the expression of PPARγ in hepatic stellate cells was positively correlated with adrenic acid (r2 = 0.451, p = 0.046). The expression of PPARγ in both groups increased after treatment, and the expression of PPARγ in the R group was restored in HSCs much more than that in the NR group (p = 0.042). The adrenic acid and arachidonic acid (AA) in the R group also upgraded more than the NR group after treatment (p = 0.037 and 0.014).Conclusion: Baseline serum differential metabolites, especially fatty acids, were identified in patients with HBV-related cirrhosis patients who achieved cirrhosis regression. Upregulation of adrenic acid and arachidonic acid in serum and re-expression of PPARγ in HSCs may play a crucial role in liver fibrosis improvement.

Soil flame disinfestation (SFD) is a form of physical disinfestation that can be used both in greenhouses and on field crops. Its use for soil disinfestation in different crop growing conditions makes it increasingly attractive for controlling soil-borne pathogens and weeds. But little is known about the effect on weeds and soilbrone diseases. This study reports on greenhouses and field crops in China that determined the efficacy of SFD to control weeds, nematodes and fungi. It also determined the impact of SFD on the soil physical and chemical properties (water content, bulk density, NO3−-N content, NH4+-N content, conductivity and organic matter) in three field trials. A second generation SFD machine was used in these trials. SFD treatment significantly reduced weeds (>87.8%) and root-knot nematodes (Meloidogyne incognita) (>98.1%). Plant height and crop yield was significantly increased with SFD treatment. NO3−-N and NH4+-N increased after the SFD treatment, and there was also an increase in soil conductivity. Water content, bulk density and organic matter decreased significantly in the soil after the SFD treatment compared to the control. Soil flame disinfestation is a potential technique for controlling weeds and diseases in greenhouses or in fields. SFD is a non-chemical, safe, environmentally-friendly soil disinfection method.

The highly-damaging root-knot nematode (Meloidogyne spp., RKN) cannot be reliably controlled using only a nematicide such as fosthiazate because of increasing pest resistance. In laboratory and greenhouse trials, we showed that chloropicrin (CP) or dazomet (DZ) synergized the efficacy of fosthiazate against RKN. The combination significantly extended the degradation half-life of fosthiazate by an average of about 1.25 times. CP or DZ with fosthiazate reduced the time for fosthiazate to penetrate the RKN cuticle compared to fosthiazate alone. CP or DZ combined with low or medium rate of fosthiazate increased the total cucumber yield, compared to the use of each product alone. A low-dose fosthiazate with DZ improved total yield more than a low dose fosthiazate with CP. Extending the half-life of fosthiazate and reducing the time for fosthiazate or fumigant to penetrate the RKN cuticle were the two features that gave the fumigant-fosthiazate combination its synergistic advantage over these products used singularly. This synergy provides the opportunity for farmers to use a low dose of fosthiazate which lowers the risk of RKN resistance. Farmers could combine DZ at 30 g m−2 with fosthiazate at a low rate of 0.375 g m−2 to control RKN and adequately control two major soil-borne diseases in cucumber greenhouses.

Exogenous toxicants (e.g. arsenic, cigarette smoke), trigger oxidative stress, and initiate the pathogenesis of many human diseases. Induction of nuclear factor E2-related factor 2 (Nrf2) counteracts exogenous toxicant-induced oxidative insults. Using a high-throughput screen, we firstly identified a citrus polymethoxyflavone, (2S)-5,6,7,3′,4′-pentamethoxyflavanone (PMF), to be an Nrf2 activator, and an investigation targeting Nrf2 pathway was performed. Our study indicated that: (i) PMF activated Nrf2 and its downstream genes, NQO1 and γ-GCS, and enhanced the stabilization of Nrf2 through inhibiting Nrf2 ubiquitination. (ii) Inhibition of PI3K, p38 MAPK, PKC and PERK by specific kinase inhibitors suppressed PMF-induced activation of Nrf2. (iii) PMF conferred Nrf2-dependent protection against sodium arsenite- and cigarette smoke extract- induced cytotoxicity in lung epithelial cells. Collectively, our data demonstrate that PMF is a novel Nrf2 activator with potential prevention against oxidative insults. The diverse pharmacological functions of citrus polymethoxyflavone might be related to their ability of activating Nrf2 pathway.

Abstract Because of the structural novelty and interesting biological profiles, the synthesis of higher-carbon sugars has been highly pursued. In this work, we first synthesized a series of structurally novel bis-uracil containing tricyclic higher-carbon sugar nucleosides (4a–e) using D-xylose as the starting material and the classical Vorbruggen glycosylation as the key synthetic step. The yields of the target compound were good. Unfortunately, despite the presence of pharmaceutically relevant uracil fragment, compounds 4a–e were inactive against the proliferation of several cancer cell lines (EC109, EC9706, PC-3, and MGC-803). Whether and how 4a–e functioned as anticancer agents would be further studied in our laboratory.

Salvianolic acid B (Sal B) is one of the main active ingredients of Salvia miltiorrhiza, with strong antioxidant effects. Recent findings have shown that Sal B has anti-inflammatory, anti-apoptotic, anti-fibrotic effects and can promote stem cell proliferation and differentiation, and has a beneficial effect on cardiovascular and cerebrovascular diseases, aging, and liver fibrosis. Reactive oxygen species (ROS) include oxygen free radicals and oxygen-containing non-free radicals. ROS can regulate cell proliferation, survival, death and differentiation to regulate inflammation, and immunity, while Sal B can scavenge oxygen free radicals by providing hydrogen atoms and reduce the production of oxygen free radicals and oxygen-containing non-radicals by regulating the expression of antioxidant enzymes. The many pharmacological effects of Sal B may be closely related to its elimination and inhibition of ROS generation, and Nuclear factor E2-related factor 2/Kelch-like ECH-related protein 1 may be the core link in its regulation of the expression of antioxidant enzyme to exert its antioxidant effect. What is confusing and interesting is that Sal B exhibits the opposite mechanisms in tumors. To clarify the specific target of Sal B and the correlation between its regulation of oxidative stress and energy metabolism homeostasis will help to further understand its role in different pathological conditions, and provide a scientific basis for its further clinical application and new drug development. Although Sal B has broad prospects in clinical application due to its extensive pharmacological effects, the low bioavailability is a serious obstacle to further improving its efficacy in vivo and promoting clinical application. Therefore, how to improve the availability of Sal B in vivo requires the joint efforts of many interdisciplinary subjects.

Currently, it is still a challenge to study the degradation mechanisms of the space traveling wave tube amplifier (TWTA) with no failure and small sample tests. Given that the Copula functions are used to describe the correlation of multiple performance characteristics, this paper develops a bivariate hybrid stochastic degradation model to evaluate the in-orbit reliability of TWTA. Firstly, based on the impact analysis of the life of TWTA, helix current and anode voltage are selected as the performance degradation parameters. Secondly, stochastic processes with random effects based on the one-dimensional Wiener process and Gamma process are applied to describe the degradation of TWTA’s helix current and anode voltage, respectively, and the corresponding marginal distribution function is obtained. Then, the Copula function is utilized to describe the correlation between two different performance parameters of TWTA. Meanwhile, this paper also proposed a two-step method to estimate the reliability level of TWTA based on its in-orbit telemetry data through a two-step method, which contains a Markov Chain Monte Carlo (MCMC) algorithm and a maximum likelihood estimation (MLE) algorithm. Besides, the Bayes-Bootstrap sampling method is also used to improve the evaluation accuracy to overcome the defect of an in-orbit small sample of TWTA. Finally, a TWTA degradation case with a set of telemetry data is carried out, and the results show that the method proposed in this paper is more applicable and more accurate than other methods.

Oxidative stress is closely associated to the onset and progression of many human diseases. Activation of the nuclear factor erythroid 2-related factor 2 (Nrf2)/antioxidant response element (ARE) signaling pathway using naturally-derived molecules is an efficient strategy for alleviating the intracellular oxidative insults, and thus blocking the pathogenesis of oxidative stress-induced diseases. In the present study, a naturally-derived isopimarane-type diterpenoid sphaeropsidin C (SC) was identified to be an activator of Nrf2/ARE signaling pathway. Our data indicated that SC was able to stimulate Nrf2-mediated defensive system through promoting Nrf2 translocation, inhibiting Nrf2 ubiquitination, and enhancing Nrf2 stability in normal human lung epithelial Beas-2B cells. Furthermore, SC-induced Nrf2 activation required the involvement of protein kinases, exemplified by protein kinase C (PKC), protein kinase R-like endoplasmic reticulum kinase (PERK), and phosphatidylinositol-4,5-bisphosphate 3-kinase (PI3K). It alleviated sodium arsenite [As(III)]-induced intracellular oxidative stress in an Nrf2-dependent manner. These results suggested that SC displayed potential application for the prevention and therapy against oxidative stress-induced diseases. Moreover, isopimarane-type diterpenoid represents a promising skeleton for developing Nrf2 activators.

Human lung tissue, directly exposed to the environmental oxidants and toxicants, is apt to be harmed to bring about acute or chronic oxidative insults. The nuclear factor erythroid 2-related factor 2 (Nrf2) represents a central cellular defense mechanism, and is a target for developing agents against oxidative insult-induced human lung diseases. Our previous study found that the EtOH extract of Cinnamomum chartophyllum protected human bronchial epithelial cells against oxidative insults via Nrf2 activation. In this study, a systemic phytochemical investigation of the aerial parts of C. chartophyllum led to the isolation of thirty chemical constituents, which were further evaluated for their Nrf2 inducing potential using NAD(P)H: quinone reductase (QR) assay. Among these purified constituents, a sesquiterpenoid bearing α, β-unsaturated ketone group, 3S-(+)-9-oxonerolidol (NLD), and a diphenyl sharing phenolic groups, 3, 3â², 4, 4â²-tetrahydroxydiphenyl (THD) significantly activated Nrf2 and its downstream genes, NAD(P)H quinone oxidoreductase 1 (NQO-1), and γ-glutamyl cysteine synthetase (γ-GCS), and enhanced the nuclear translocation and stabilization of Nrf2 in human lung epithelial cells. Importantly, NLD and THD had no toxicities under the Nrf2 inducing doses. THD also demonstrated a potential of interrupting Nrf2-Keap1 proteinâprotein interaction (PPI). Furthermore, NLD and THD protected human lung epithelial cells against sodium arsenite [As(III)]-induced cytotoxicity. Taken together, we conclude that NLD and THD are two novel Nrf2 activators with potential application of preventing acute and chronic oxidative insults in human lung tissue. Abstract Background China is one of 22 countries with a high tuberculosis (TB) burden in the world. Healthcare workers (HCWs) have a high risk of contracting Mycobacterium tuberculosis infection due to insufficient infection control practices. We conducted a cross-sectional study to explore the prevalence of TB and its associated risk factors among HCWs in Chinese TB facilities. Methods Two hundred and forty-one TB facilities employing a total of 9663 HCWs were selected from 12 provinces in China to represent healthcare settings at the provincial, prefectural, and county levels. Structured questionnaires were used to collect information on TB infection control practices and HCWs in those facilities. Data was double entered into EpiData 3.1; TB prevalence and associated risk factors were analyzed using SPSS 21.0 with bivariate and multivariate regression models. Results The results showed that 71 HCWs had been diagnosed with TB, accounting for a prevalence of 760/100 000. The multivariate analysis showed that associated risk factors included belonging to the age group of 51 years and above (aOR: 6.17, 95% CI: 1.35–28.28), being a nurse (aOR = 3.09, 95% CI: 1.15–8.32), implementation of 0–9 items of management measures (aOR = 2.57, 95% CI: 1.37–4.80), and implementation of 0–1 items of ventilation measures (aOR = 2.42, 95% CI: 1.31–4.47). Conclusion This was the first national large sampling survey on TB prevalence among HCWs in China. It was found that the implementation of TB infection control practices in some facilities was poor. The TB prevalence in HCWs was higher than that in the general population. Therefore, TB infection control practices in Chinese medical facilities should be strengthened.

Background: Excessive reactive oxygen species (ROS) may lead to a number of reproductive diseases such as polycystic ovary syndrome. This study aimed to establish an animal model of ovarian oxidative stress and to assess the protective effect of curcumin against oxidative injury. Methods: Ovarian oxidative stress was induced in female Kunming mice (n = 40) with intraperitoneal injection of 8 mg/kg sodium arsenite (As) once every other day for 16 days; meanwhile, they were, respectively, treated by intragastric administration of 0, 100, 150, or 200 mg/kg (n = 10/group) curcumin once per day for 21 days. Ten normal mice were used as control. Then, the mice were injected intraperitoneally with BrdU and sacrificed; the right ovaries were collected for hematoxylin and eosin (HE) staining and BrdU immunohistochemistry, and the left ovaries for enzyme-linked immunosorbent assay (ELISA) and Western blotting analyses. Results: The ELISA results showed that ROS (11.74 ± 0.65 IU/mg in 8 mg/kg AS + 0 mg/kg curcumin group vs. 10.71 ± 0.91 IU/mg in control group, P= 0.021) and malondialdehyde (MDA) (0.32 ± 0.02 nmol/g in 8 mg/kg AS + 0 mg/kg curcumin group vs. 0.27 ± 0.02 nmol/g in control group, P= 0.048) increased while superoxide dismutase (SOD) (3.96 ± 0.36 U/mg in 8 mg/kg AS + 0 mg/kg curcumin group vs. 4.51 ± 0.70 U/mg in control group, P= 0.012) and glutathione peroxidase (17.36 ± 1.63 U/g in 8 mg/kg AS + 0 mg/kg curcumin group vs. 18.92 ± 1.80 U/g in control group, P= 0.045) decreased in the ovary after injection of As, indicating successful modeling of oxidative stress. Curcumin treatment could considerably increase SOD (4.57 ± 0.68, 4.49 ± 0.27, and 4.56 ± 0.25 U/mg in 100 mg/kg, 150 mg/kg, and 200 mg/kg curcumin group, respectively, allP < 0.05) while significantly reduce ROS (10.64 ± 1.38, 10.73 ± 0.71, and 10.67 ± 1.38 IU/mg in 100 mg/kg, 150 mg/kg, and 200 mg/kg curcumin group, respectively, allP < 0.05) and MDA (0.28 ± 0.02, 0.25 ± 0.03, and 0.27 ± 0.04 nmol/g in 100 mg/kg, 150 mg/kg, and 200 mg/kg curcumin group, respectively; bothP < 0.05) in the ovary. HE staining and BrdU immunohistochemistry of the ovarian tissues indicated the increased amount of atretic follicles (5.67 ± 0.81, 5.84 ± 0.98, and 5.72 ± 0.84 in 100 mg/kg, 150 mg/kg, and 200 mg/kg curcumin group, respectively, all P < 0.05), and the inhibited proliferation of granular cells under oxidative stress would be reversed by curcumin. Furthermore, the Western blotting of ovarian tissues showed that the p66Shc expression upregulated under oxidative stress would be lowered by curcumin. Conclusion: Curcumin could alleviate arsenic-induced ovarian oxidative injury to a certain extent.

Fibrosis is the remodeling and repair processes of chronic injuries. There are few effective therapies. Chinese medicine formula, the main pattern of traditional Chinese medicine (TCM) in clinic, applies a multi-component, multi-target and complicated approach in the treatment of diseases, and certainly shows good comprehensive therapeutic effects on fibrosis. In this review, the clinical study, effects and mechanism of action of Fuzheng Huayu recipe in the treatment of liver, pulmonary, and renal fibrosis was analyzed and evaluated. Meanwhile, based on the understanding of TCM pathogenesis for liver cirrhosis, this review will also briefly introduce the research of different classical Chinese medicine formulae with various functions in the treatment of experimental liver fibrosis induced by different methods, including comparing the efficacy, analyzing the action characteristics and mechanism of effective formulae, exploring pathological and biological basis of TCM diagnostic and therapeutic pattern, which will contribute to the research of TCM in the treatment of organ fibrosis.

Salmonella typhimurium (S. typhimurium) is a common foodborne pathogen that not only causes diseases and contaminates food, but also causes considerable economic losses. Therefore, it is necessary to find effective and feasible methods to control S. typhimurium. In this study, changes in S. typhimurium after treatment with benzyl isothiocyanate (BITC) were detected by transcriptomics to explore the antibacterial effect of BITC at subinhibitory concentration. The results showed that, in contrast to the control group (SC), the BITC-treated group (SQ_BITC) had 197 differentially expressed genes (DEGs), of which 115 were downregulated and 82 were upregulated. We screened out eight significantly downregulated virulence-related genes and verified gene expression by quantitative Real-time Polymerase Chain Reaction (qRT-PCR). We also selected motility and biofilm formation to observe the effects of BITC on the other virulence related factors of S. typhimurium. The results showed that both swimming and swarming were significantly inhibited. BITC also had a significant inhibitory effect on biofilm formation, and showed an effect on bacterial morphology. These results will be helpful for understanding the mechanism of the antibacterial action of BITC against S. typhimurium and other foodborne pathogens.

Biological functional traits help to understand specific stressors that are ignored in taxonomic data analysis. A combination of biological functional traits and taxonomic data is helpful in determining specific stressors which are of significance for fish conservation and river basin management. In the current study, the Taizi River was used as a case study to understand the relationships between the taxonomic and functional structure of fish and land use and water quality, in addition to determining the thresholds of these stressors. The results showed that taxonomic structure was significantly affected by the proportion of urban land and specific conductivity levels, while functional metrics were influenced by the proportions of farmland and forest. Threshold indicator taxa analysis found that Phoxinus lagowskii, Barbatula barbatula nuda, Odontobutis obscura, and Cobitis granoei had negative threshold responses along the gradients of urban developments and specific conductivity. There was a significant change in fish taxonomic composition when the proportion of urban land exceeded a threshold of 2.6–3.1%, or specific conductivity exceeded a threshold of 369.5–484.5 μS/cm. Three functional features—habitat preference, tolerance to disturbances, and spawning traits—showed threshold responses to the proportion of farmland and forest. The abundance of sensitive species should be monitored as part of watershed management, as sensitive species exhibit an earlier and stronger response to stressors than other functional metrics. Sensitive species had a positive threshold response to the proportion of forest at 80.1%. These species exhibited a negative threshold response to the proportion of farmland at 13.3%. The results of the current study suggest that the taxonomic and functional structure of fish assemblages are affected by land use and water quality. These parameters should be integrated into routine monitoring for fish conservation and river basin management in the Taizi River. In addition, corresponding measures for improving river habitat and water quality should be implemented according to the thresholds of these parameters.

For evaluating the classification model of an information system, a proper measure is usually needed to determine if the model is appropriate for dealing with the specific domain task. Though many performance measures have been proposed, few measures were specially defined for multi-class problems, which tend to be more complicated than two-class problems, especially in addressing the issue of class discrimination power. Confusion entropy was proposed for evaluating classifiers in the multi-class case. Nevertheless, it makes no use of the probabilities of samples classified into different classes. In this paper, we propose to calculate confusion entropy based on a probabilistic confusion matrix. Besides inheriting the merit of measuring if a classifier can classify with high accuracy and class discrimination power, probabilistic confusion entropy also tends to measure if samples are classified into true classes and separated from others with high probabilities. Analysis and experimental comparisons show the feasibility of the simply improved measure and demonstrate that the measure does not stand or fall over the classifiers on different datasets in comparison with the compared measures.

Fish species tolerance used as a component of fish-index of biological integrity (F-IBI) can be problematic as it is usually classified using the historical data, data from literature or expert judgments. In this study, fish assemblages, water quality parameters and physical habitat factors from 206 sampling sites in the Huntai River Basin were analyzed to develop tolerance indicator values (TIVs) of fish based on a (Fb-TIVs) and the weighted averaging (WA) method (FW-TIVs). The two quantitative methods for fish tolerance were then compared. The FW-TIVs and Fb-TIVs of fish species were calculated separately using a WA inference model based on ten water quality parameters (WT, pH, DO, SC, TDS, NH3, NO2−, NO3−, TP, Cl−, and SO42−), and six biological traits (lithophilic spawning, benthic invertivores, cold water species, equilibrium or periodic life history strategies, families of Cottidae, and species distribution range). Fish species were then classified into biological traits approach three categories (tolerant species, moderately tolerant species, and sensitive species). The results indicated that only 30.3% fish species have the same classification based on FW-TIVs and Fb-TIVs. However, the proportion of tolerant species based on two methods had a similar response to environmental stress, and these tolerant species were correlated with PCA axes 1 site scores obtained by (FW-TIVs, p < 0.05, R2 = 0.434; Fb-TIVs, p < 0.05, R2 = 0.334) and not correlated with PCA axis 2 site scores (FW-TIVs, p > 0.05, R2 = 0.001; Fb-TIVs, p > 0.05, R2 = 0.012) and PCA axis 3 site scores (FW-TIVs, p > 0.05, R2 = 0.000; Fb-TIVs, p > 0.05, R2 = 0.013). The results of linear regression analyses indicated that Fb-TIVs can be used for the study of fish tolerance. Fish tolerance assessments based on FW-TIVs requires long-term monitoring of fish assemblages and water quality parameters to provide sufficient data for quantitative studies. The Fb-TIV method relies on the accurate identification of fish traits by an ichthyologist. The two methods used in this study can provide methodological references for quantitative studies of fish tolerance in other regions, and are of great significance for the development of biological assessment tools.

Cell-in-cell structures refer to a unique phenomenon that one living cell enters into another living cell intactly, occurring between homotypic tumor cells or tumor (or other tissue cells) and immune cells (named as heterotypic cell-in-cell structure). In the present study, through a large scale of survey we observed that heterotypic cell-in-cell structure formation occurred commonly in vitro with host cells derived from different human carcinomas as well as xenotypic mouse tumor cell lines. Most of the lineages of human immune cells, including T, B, NK cells, monocytes as well as in vitro activated LAK cells, were able to invade tumor cell lines. Poorly differentiated stem cells were capable of internalizing immune cells as well. More significantly, heterotypic tumor/immune cell-in-cell structures were observed in a higher frequency in tumor-derived tissues than those in adjacent tissues. In mouse hepatitis models, heterotypic immune cell/hepatocyte cell-in-cell structures were also formed in a higher frequency than in normal controls. After in vitro culture, different forms of internalized immune cells in heterotypic cell-in-cell structures were observed, with one or multiple immune cells inside host cells undergoing resting, degradation or mitosis. More strikingly, some internalized immune cells penetrated directly into the nucleus of target cells. Multinuclear cells with aneuploid nucleus were formed in target tumor cells after internalizing immune cells as well as in situ tumor regions. Therefore, with the prevalence of heterotypic cell-in-cell structures observed, we suggest that shielding of immune cells inside tumor or inflammatory tissue cells implies the formation of aneuploidy with the increased multinucleation as well as fine-tuning of microenvironment under pathological status, which may define distinct mechanisms to influence the etiology and progress of tumors.

The title compound, C10H11I2NO, was prepared by the reaction of 3,5-diiodosalicylaldehyde with propylamine in ethanol. The molecule adopts an E conformation with respect to the C=N bond and the aromatic ring. The aromatic ring and the imino unit are close to being coplanar, with a dihedral angle of 2.6 (3)° between their planes. This planarity is assisted by the formation of an intramolecular O—H...O hydrogen bond.

The asymmetric unit of the title compound, C20H16ClN3S, contains two independent molecules, A and B. In molecule A, the dihedral angles between the central benzene ring and the pendant chlorobenzene and phenyl rings are 6.37 (15) and 64.79 (15)°, respectively. The corresponding values in molecule B are 28.21 (14) and 82.11 (16)°, respectively. Each molecule features an intramolecular N—H...N hydrogen bond, which generates an S(5) ring. In the crystal, molecules A and B form dimers, being linked by two N—H...S hydrogen bonds with graph-set notation R22(8).

Nasopharyngeal carcinoma (NPC) is an Epstein-Barr virus (EBV) associated malignancy with high prevalence in Southern Chinese. In order to assess whether defects of EBV-specific immunity may contribute to the tumor, the phenotype and function of circulating T-cells and tumor infiltrating lymphocytes (TILs) were investigated in untreated NPC patients. Circulating naïve CD3+CD45RA+ and CD4+CD25- cells were decreased, while activated CD4+CD25+ T-cells and CD3-CD16+ NK-cells were increased in patients compared to healthy donors. The frequency of T-cells recognizing seven HLA-A2 restricted epitopes in LMP1 and LMP2 was lower in the patients and remained low after stimulation with autologous EBV-carrying cells. TILs expanded in low doses of IL-2 exhibited an increase of CD3+CD4+, CD3+CD45RO+ and CD4+CD25+ cells and 2 to 5 fold higher frequency of LMP1 and LMP2 tetramer positive cells compared to peripheral blood. EBV-specific cytotoxicity could be reactivated from the blood of most patients, whereas the TILs lacked cytotoxic activity and failed to produce IFNgamma upon specific stimulation. Thus, EBV-specific rejection responses appear to be functionally inactivated at the tumor site in NPC.

In China, university sports are mainly conducted through physical education (PE) classes and amateur training, which are basically in a disorganized state. Due to the highly open nature of the modern sports system, changes in the way decisions are made in this information-based society have had a profound impact on the decision-making process in the field of sports. However, current school data processing is still at a rudimentary stage of data backup, querying and simple statistics, and does not provide an effective means to help teachers make decisions about school processes. Data mining (DM) is an in-depth approach to data analysis by mining useful information from large amounts of data, and this technique is now being used in an increasing number of fields. In this paper, we present the design of a real-time data analysis system for sports training based on DM technology, and use the corresponding mining tools of DM technology to discover relevant patterns or laws hidden in the data. Therefore, using the real-time data analysis system for sports training based on DM technology, useful information and patterns for improving examination performance can be obtained, which can improve targeted teaching methods and help students overcome learning difficulties, providing rational teaching. Background and aims: The serum metabolites changes in patients with hepatitis B virus (HBV)-related cirrhosis as progression. Peroxisome proliferator-activated receptor gamma (PPARγ) is closely related to lipid metabolism in cirrhotic liver. However, the relationship between fatty acids and the expression of hepatic PPARγ during cirrhosis regression remains unknown. In this study, we explored the serum metabolic characteristics and expression of PPARγ in patients with histological response to treatment with entecavir. Methods: Sixty patients with HBV-related cirrhosis were selected as the training cohort with thirty patients each in the regression (R) group and non-regression (NR) group based on their pathological changes after 48-week treatment with entecavir. Another 72 patients with HBV-related cirrhosis and treated with entecavir were collected as the validation cohort. All of the serum samples were tested using ultra-performance liquid chromatography coupled to tandem mass spectrometry. Data were processed through principal component analysis and orthogonal partial least square discriminant analysis. Hepatic PPARγ expression was observed using immunohistochemistry. The relationship between serum fatty acids and PPARγ was calculated using Pearson’s or Spearman’s correlation analysis. Results: A total of 189 metabolites were identified and 13 differential metabolites were screened. Compared to the non-regression group, the serum level of fatty acids was higher in the R group. At baseline, the expression of PPARγ in hepatic stellate cells was positively correlated with adrenic acid (r² = 0.451, p = 0.046). The expression of PPARγ in both groups increased after treatment, and the expression of PPARγ in the R group was restored in HSCs much more than that in the NR group (p = 0.042). The adrenic acid and arachidonic acid (AA) in the R group also upgraded more than the NR group after treatment (p = 0.037 and 0.014). Conclusion: Baseline serum differential metabolites, especially fatty acids, were identified in patients with HBV-related cirrhosis patients who achieved cirrhosis regression. Upregulation of adrenic acid and arachidonic acid in serum and reexpression of PPARγ in HSCs may play a crucial role in liver fibrosis improvement. [ABSTRACT FROM AUTHOR]
