Triphenylamine (TPA) derivatives have been widely used as useful building blocks for diverse functional materials because of their excellent redox activity. Most of the molecular structures of TPA-based organic functional materials contain 4-anisyl groups, which on one hand could reduce their oxidation potential and on the other hand significantly delocalize the spin density of the resultant TPA radical cation species and enhance their stability. However, molecular-level investigation of the redox behavior of triphenylamines consisting of 4-anisyl group and the electronic structures of their radical cation species has not been reported in the literature. Herein, we design a series of triphenylamines consisting of one, two, or three 3,5-di-tert-butyl-4-anisyl groups and investigate their redox behaviors and corresponding radical cation species. We disclose that the resonance hybrid and steric protection could both contribute to the stability of triphenylamine radical cations. Moreover, further oxidation leads to an unexpected oxidative demethylation. The findings in this work may reveal new insights for the understanding of the unique redox properties of 4-anisyl substituted triphenylamines.

Sequence isomerism plays fundamental roles in the encoding of biological information and in tuning material performance, but engineering sequences and elucidating their effects remain a major and largely unmet challenge in synthetic chemistry. Herein, We develop a novel wheel assembling approach aiming at the precise sequence control of hetero[3]rotaxanes, a typical family of mechanically interlocked molecules. Based on a key [2]rotaxane precursor with exchangeable pentafluorophenyl ester stoppers, We successfully synthesized both sequence isomers of diverse hetero[3]rotaxanes. Moreover, taking advantage of the chirality retention along with the wheel assembling process, corresponding sequence isomers of chiral functionalized hetero[3]rotaxanes were further precisely synthesized. Impressively, these hetero[3]rotaxanes revealed remarkable sequence-dependent chemical reactivity and circularly polarized luminescence, highlighting fantastic sequence effects. This proof-of-concept study lays the foundation for the investigations on the structure-property relationships of heterorotaxanes, which would further direct the rational design and precise synthesis of sequence-defined heterorotaxanes with desired properties for practical use.

This study analyzed the main chemical components of Zhuru Decoction via ultra-performance liquid chromatography coupled with quadrupole time-of-flight tandem mass spectrometry(UPLC-Q-TOF-MS/MS), and then predicted the mechanism of Zhuru Decoction in clearing heat, resolving phlegm, detoxifying, and treating vomiting and alcohol-related vomiting caused by heat in stomach based on network pharmacology. The gradient elution was conducted in Agilent ZORBAX extend-C_(18) column(2.1 mm×100 mm, 1.8 μm) with the mobile phase of 0.1% formic acid aqueous solution(A)-acetonitrile(B) at a flow rate of 0.3 mL·min~(-1) and the column temperature of 35 ℃. The MS adopted the positive and negative ion mode of electrospray ionization(ESI), and the data were collected in the scanning range of m/z 100-1 500. A total of 98 compounds in Zhuru Decoction were identified via BATMAN, SYMMAP, TCMSP, and relevant literature, including 36 flavonoids, 7 triterpenoids, 8 gingerols, 20 organic acids, 5 amino acids, and 22 other compounds. On the basis of the available studies, 9 components were selected as index components, and the protein-protein interaction(PPI) network of the common targets was established with STRING 11.0. Finally, 10 core targets associated with the pharmacodynamic effect were screened out. This study established the UPLC-Q-TOF-MS/MS method for identifying the chemical components in the classic prescription Zhuru Decoction, and employed network pharmacology to explore the core targets of its efficacy, which provided a refe-rence for the quality control and the research of the pharmacodynamic substances of Zhuru Decoction.

The design and construction of highly effective circularly polarized luminescence (CPL)-active materials has aroused considerable attention due to their widespread applications in sensors, optical devices, and asymmetric synthesis. However, the exploration of novel CPL-active materials with high luminescence dissymmetry factor (glum) values is still a challenge. Herein, we describe a new approach for the preparation of supramolecular metallacycles with amplified CPL promoted by hierarchical self-assembly involving Pt···Pt interactions. Notably, the resultant metallacycles exhibited strong CPL signals with high glum values, while their corresponding precursors were CPL silent. The CPL amplification mechanism was comprehensively validated by ultraviolet-visible absorption, emission spectroscopy, nuclear magnetic resonance spectroscopy, scanning electron microscopy, transmission electron microscopy, atomic force microscopy, coarse-grained molecular dynamics simulations, and time-dependent density functional theory calculations. This work thus provides the first example of preparing highly effective CPL-active materials based on hierarchical self-assembly involving Pt···Pt interactions.

We report herein the asymmetric total synthesis of norzoanthamine using radical reactions as key steps for rapid access to the congested carbocyclic core, which is the major synthetic challenge for most zoanthamine alkaloids. (1) The Ueno-Stork radical cyclization was applied to construct the adjacent quaternary centers at the C-9 and C-22 positions; (2) a Co-catalyzed HAT radical reaction was successfully applied to construct the quaternary center at C-12 via Csp3 -Csp2 bond formation; (3) a Mn-catalyzed HAT radical reaction was used to stereospecifically reduce the tetra-substituted olefin (C13=C18) and install the contiguous stereocenters in proximity to the quaternary center. A one-pot bio-inspired cyclization step was finally applied to forge the unstable bis-amino acetal skeleton. Our approach can precisely control the stereochemistry of seven vicinal stereocenters and effectively construct the highly congested heptacyclic skeleton.

Artificial tubular molecular pockets bearing polar functionalities on their inner surface are useful model systems for understanding the mechanisms of protein-ligand interactions in living systems. We herein report a pillar[5]arene-derived molecular tube, [P4-(OH)BPO], whose endo conformational isomer endo-[P4-(OH)BPO] possesses an inwardly pointing hydrogen-bond (H-bond) donor (OH) in its deep cavity and a strong H-bond acceptor (C═O) on its predominantly hydrophobic inner surface, rendering it a perfect protein binding pocket mimetic. A fragment-based drug design model was established using endo-[P4-(OH)BPO] and a library of various shape-complementary fragment ligands (1-38). On the basis of the binding affinity data for "fragment-pocket" complexes G⊂endo-[P4-(OH)BPO] (G = 1-38), two rationally designed "lead molecules" (39 and 40) were identified as being able to enhance binding affinity significantly by forming H-bonds with both the donor and acceptor of endo-[P4-(OH)BPO]. The described work opens new avenues for developing pillar[n]arene-derived protein binding pocket-mimetic systems for studies of protein-ligand interactions and mechanisms of enzymatic reactions.

A series of triphenylamine (TPA) derivatives with various substituent groups were prepared and showed different absorption and fluorescence characteristics due to the substituent effect. On account of the existence of pyridine units, these TPA derivatives exhibited acid-induced tunable multicolor fluorescence emission including white light emission. In addition, acid-induced fluorescence regulation of these compounds has been also realized in the solid state, which enable them to be successfully constructed the stimuli-responsive fluorescent films and fluorescent inks for inkjet printing.

The reaction conditions and scope of the excited-state Nazarov reaction of dicyclicvinyl ketones were studied. The stereochemistry of this electrocyclization is consistent with the mechanism of the pericyclic reaction and Woodward-Hoffmann rule. UV-light-promoted excited-state Nazarov reactions gave hydrofluorenones bearing a syn-cis configuration via a disrotatory cyclization. The core tricyclic hydrofluorenones of pyrrocidines and wortmannines were constructed via the excited-state Nazarov reactions, which demonstrated their synthetic potential in complex natural product total synthesis.

This investigation was aimed at determining the concentration levels of five toxic heavy metals (lead, cadmium, chromium, arsenic, and mercury) in dried Porphyra and Laminaria samples from coastal city of China. Inductively coupled plasma-mass spectrometry (ICP-MS) was used for determination of lead, cadmium and total arsenic. Atomic fluorescence spectrometry was used for mercury, and liquid chromatography coupled with ICP-MS was used for arsenic speciation. The mean concentrations of lead, cadmium, chromium, total arsenic, and mercury were 0.96 ± 0.03 mg/kg, 2.62 ± 0.07 mg/kg, 1.64 ± 0.08 mg/kg, 36.67 ± 0.53 mg/kg, and 7.56 ± 0.42 μg/kg for Porphyra samples and 0.61 ± 0.03 mg/kg, 0.48 ± 0.02 mg/kg, 3.78 ± 0.56 mg/kg, 43.85 ± 1.42 mg/kg, and 46.61 ± 2.02 μg/kg for Laminaria samples. The results were comparable with previous similar research. The potential health risk assessment was conducted by comparing the calculated weekly intakes of toxic metals from Porphyra and Laminaria with provisional tolerable weekly intake. Consumption of these seaweeds does not seem to pose a risk for the consumers' health regarding their content of heavy metals. However, the potential health risk of cadmium should not be overlooked for consumers with high intake of Porphyra.

The design and fabrication of highly efficient artificial light-harvesting systems (LHSs) are of great importance in supramolecular chemistry and materials science. Benefitting from the convenient construction process of supramolecular self-assembly, herein, a dual-donor artificial LHS was efficiently constructed through hierarchical self-assembly approaches involving metal–ligand coordination interactions, host–guest interactions and hydrophobic interactions. Notably, compared to the model single-donor artificial LHS with only one donor chromophore, the prepared dual-donor artificial LHS exhibited higher energy transfer efficiency and antenna effects. This research provides an efficient strategy for the construction of artificial LHSs with high energy transfer efficiency and antenna effects.

The photocatalytic gas-solid reaction has been regarded as a powerful mean to solve the environmental pollution and energy crisis. However, problems such as catalyst poisoning, low efficiency of reaction, and uncontrollable selectivity are still unsolved restricting the development of photocatalysis. Therefore, it is of significant importance to reveal and elucidate the reaction mechanisms of gas-solid photocatalysis. Herein, five major types of gas-solid photocatalytic reactions including oxidation of NOx, volatile organic compounds (VOCs), NH3 and reduction of CO2, N2 are discussed and summarized. It involves the composition and basic properties of gas molecules, the process of adsorption and activation, and various reaction pathways. Finally, some prospective viewpoints for future development on gas-solid photocatalysis are put forward for further improvements.

A series of fully conjugated macrocycles Mn (n = 4–7) consisting of N-(3,5-di-tert-butyl-4-methoxyphenyl) substituted carbazole (Cz-Ar) were successfully synthesized. The aryl carbazole and macrocycle M4 can be readily oxidized and the corresponding radical cation species were found to be highly stable. Moreover, macrocycle M5 was found to form 1 : 1 stoichiometric complexes with fullerenes C60 and C70 with association constants as high as (8.38 ± 0.33) × 104 M−1 and (7.64 ± 0.26) × 104 M−1, respectively.

Myeloperoxidase (MPO) is a heme peroxidase that plays an important role in innate immunity for host defense against invading microorganisms by catalyzing hydrogen peroxide (H2O2)‐mediated reactions. Although many reports indicate MPO exerts beneficial or detrimental effects on a variety of inflammatory diseases, little is known with regard to its functional role in bone homeostasis in vivo. Here, our work demonstrates that MPO was transcriptionally downregulated in response to osteoclastogenic stimuli and that exogenous alteration of MPO expression negatively regulated osteoclast (OC) differentiation in vitro. Genetic ablation of Mpo resulted in osteoporotic phenotypes and potentiated bone‐resorptive capacity in mice. Mechanistically, accumulation of intracellular H2O2 and reactive oxygen species (ROS) were observed in MPO deficiency, and MPO overexpression suppressed ROS production in mouse OC precursors. Moreover, a ROS scavenger Tempol inhibited the effect of MPO deficiency on OC formation and function as well as on receptor activator of nuclear factor‐κB ligand (RANKL)‐initiated transduction signal activation including NF‐κB, mitogen‐activated protein kinases (MAPKs), and Akt, indicating the increased ROS caused by MPO deficiency contributes to osteoclastogenesis. Taken together, our data demonstrate that MPO has a protective role in bone turnover by limiting osteoclastogenesis and bone resorption physiologically through modulating intracellular H2O2 level. © 2020 The Authors. TANK-binding kinase 1 (TBK1) belongs to the noncanonical IκB kinase (IKK) family. The ubiquitously expressed protein is well known to play a pivotal role in innate immune response and inflammation. Although excessive inflammatory activities have been shown to affect osteoclast (OC) differentiation and function, direct relevance of TBK1 in bone turnover is not known. In this work, we specifically altered the TBK1 protein level by knocking down or overexpressing it without affecting its homologous protein IKKε expression, and demonstrated the effect of TBK1 on OC differentiation in bone marrow macrophages (BMMs) and RAW264.7 cells upon induction by receptor activator of nuclear factor-κB (NF-κB) ligand (RANKL). TBK1 knockdown was found to markedly inhibit the OC differentiation and function, while TBK1 overexpression enhanced OC formation. Downregulation of TBK1 greatly suppressed RANKL-induced gene expression of Mmp9, Atp6v0d2, Acp5, Ctsk andNfatc1 involved in the regulation of OC formation and function in both BMM and RAW264.7 cells. Mechanistic studies indicated that TBK1 affected the NF-κB signaling pathway as well as mitogen-activated protein kinases (MAPKs) and protein kinase B (Akt) activation during OC differentiation. Moreover, the protein level of TNF receptor-associated factor 6 (TRAF6) was increased, and the interaction of TRAF6 with TBK1 was potentiated, by RANKL. Collectively, we provide direct evidence showing that TBK1 effectively mediates OC differentiation and function by regulating NF-κB, MAPKs and Akt signals. A TBK1-targeted therapeutic strategy may be useful for the treatment of bone-related disorders.

An unexpected bistricyclic aromatic ene AF was synthesized in a tin(II) chloride-mediated reductive aromatization reaction. The obtained AF showed a highly overcrowded structural conformation as revealed by X-ray crystallography. Interestingly, AF exhibited reversible high-contrast mechanochromism and thermochromism between pale and red color. The obvious chromism is likely ascribed to the conformation transformation and trace amount of diradical species formation upon stimulus.

OBJECTIVE: To study the value of Silverman-Anderson score versus Downes score in predicting respiratory failure in full-term neonates. METHODS: The convenience sampling method was used to select the full-term neonates with lung diseases who were hospitalized in the neonatal intensive care unit from July 2020 to July 2021. According to the diagnostic criteria for neonatal respiratory failure, they were divided into a respiratory failure group (65 neonates) and a non-respiratory failure group (363 neonates). Silverman-Anderson score and Downes score were used for evaluation. The receiver operating characteristic analysis was used to compare the value of the two noninvasive scores in predicting respiratory failure in full-term neonates. RESULTS: Among the 428 full-term neonates, 65 (15.2%) had respiratory failure. In this study, we present the de novo design of a new open-shell nitrogen-centered diradicaloid which can reversibly respond to multiple stimuli and display the tunable diradical character and chemo-physical properties. 1a was successfully synthesized through a simple and high-yielding two-step synthetic strategy. Both experimental and calculated results indicated that 1a displayed an open-shell singlet ground state with small singlet-triplet energy gap (ΔES−T = -2.311 kcal mol− 1) and a modest diradical character (y0 = 0.55). Interestingly, 1a was demonstrated to undergo reversible Lewis acid-base reaction with both Lewis acid and Brønsted acid to form acid-base adducts due to its intrinsic Lewis basicity. The formation of Lewis acid-base adduct was proven to effectively tune the ground-state electronic structures of 1a as well as its diradical character and spin density distributions. Based on this, we succeeded in devising a photoresponsive system based on 1a and a commercially available photoacid merocyanine (MEH). We believe that our studies including the entirely new molecular design methodology and the newly found stimuli-responsive organic diradicaloid system will open up a new way to develop new diradicaloids with tunable properties and even intelligent-responsive diradicaloid-based materials.

Herein we successfully developed a ring-fusion approach to extend the conjugation length of phenothiazines and synthesized a series of novel extended phenothiazines 1-5. The intriguing π-conjugation length-dependent photophysical and redox properties of 1-5, and their photocatalytic performance towards visible-light-driven oxidative coupling reactions of amines were systematically investigated. The results indicated that this series of extended phenothiazines exhibited continuous red shifts of light absorption with increasing numbers of fused rings. As compared with the conventional phenothiazine (PTZ), all the extended phenothiazines displayed reversible redox behavior and maintained a strong excited-state reduction potential as well. Consequently, 3, 4 and 5 with longer effective conjugation lengths could efficiently catalyze the oxidative coupling of amines to imines under visible-light irradiation; by comparison, the shorter 1, 2 and PTZ could only catalyze such reactions in the presence of UV light. Moreover, 3 showed superior catalytic performance which can result in better yields within a shorter reaction time, and in a broad substrate scope. Finally, a direct and efficient conversion of amines to imines under sunlight in an air atmosphere was successfully realized. We believe that our study including the new phenothiazine modification methodology and the newly developed extended phenothiazine-based photocatalysts will open up a new way to develop novel phenothiazine-based materials for optoelectronic and catalytic applications.

A total of 18 batches of Zhuru Decoction samples were prepared. Chromatographic fingerprints were established for Zhuru Decoction and single decoction pieces, the content of which was then determined. The extraction rate ranges, content, and transfer rate ranges of puerarin, liquiritin, and glycyrrhizic acid, together with the common peaks and the similarity range of the fingerprints, were determined to clarify key quality attributes of Zhuru Decoction. The 18 batches of Zhuru Decoction samples had 25 common peaks and the fingerprint similarity higher than 0.95. Puerariae Lobatae Radix, Glycyrrhizae Radix et Rhizoma, and Zingiberis Rhizoma Recens had 21, 3, and 1 characteristic peaks, respectively. The 18 batches of samples showed the extraction rates within the range of 18.45%-25.29%. Puerarin had the content of 2.20%-3.07% and the transfer rate of 38.5%-45.9%; liquiritin had the content of 0.24%-0.85% and the transfer rate of 15.9%-37.5%; glycyrrhizic acid had the content of 0.39%-1.87% and the transfer rate of 16.2%-32.8%. In this paper, the quality value transmitting of substance benchmarks of Zhuru Decoction was analyzed based on chromatographic fingerprints, extraction rate, and the content of index components. A scientific and stable method was preliminarily established, which provided a scientific basis for the quality control and formulation development of Zhuru Decoction.

Following the preparation of substance benchmarks in Huanglian Decoction from 18 batches, the method for detecting their characteristic spectra was established to identify the similarity range and peak attribution. The content and transfer rate ranges of the index components coptisine, palmatine, berberine, liquiritin, glycyrrhizic acid, 6-gingerol, and cinnamaldehyde and the extraction amount were combined for analyzing the quality value transfer from the Chinese medicinal pieces to substance benchmarks and clarifying the key quality attributes of substance benchmarks in Huanglian Decoction. The results showed that the substance benchmarks in Huang-lian Decoction of 18 batches exhibited good similarity in characteristic spectra(all greater than 0.98). There were 17 characteristic peaks identified in the substance benchmarks of Huanglian Decoction, including 10 from Coptidis Rhizoma, 3 from Glycyrrhizae Radix Et Rhizoma Praeparata Cum Melle(processed with water), 1 from Zingiberis Rhizoma, and 3 from Cinnamomi Ramulus. The contents and average transfer rates of the index components were listed as follows: coptisine 2.20-6.46 mg·g~(-1) and 18.50%±2.93%; palmatine 3.03-8.13 mg·g~(-1) and 26.56%±4.69%; berberine 7.71-22.29 mg·g~(-1) and 17.34%±3.00%; liquiritin 0.88-2.18 mg·g~(-1) and 9.88%±4.88%; glycyrrhizic acid 1.83-4.44 mg·g~(-1) and 8.50%±3.72%; 6-gingerol 0.56-1.43 mg·g~(-1) and 11.36%±2.37%; cinnamaldehyde 1.55-3.48 mg·g~(-1) and 19.02%±4.36%. The extraction amount of the substance benchmarks from the 18 batches was controlled at 10.65%-13.88%. In this paper, the quality value transfer of substance benchmarks in Huanglian Decoction was analyzed based on the characteristic spectra, the index component contents and the extraction amount, which has provided a basis for the subsequent development of Huanglian Decoction and the quality control of its related preparations.

To investigate the clinical characteristics of the patients with chronic myeloid leukemia (CML) discontinued tyrosine kinase inhibitors (TKI) therapy and the outcome of the patients.35 cases of CML patients experienced initiative discontinuation of TKI therapy in our hospital from June 1st 2015 to December 31th 2019 were retrospectively analyzed. The TFR of the patients and the factors affecting it were analyzed.The median duration of TKI administration was 72 (range 35-173) months in the 35 patients. Among these patients, 8 had experienced TKI dose reduction or suspension. All the enrolled patients have achieved at least MMR. The median time for these patients achieving MMR was 15 (range 3-75) months after administration of TKI, and for MMR maintenance before TKI suspension was 55 (range 13-164) months. After TKI withdrawal the median follow up time was 20.3 (range 3-57.9) months, 22 out of 35 patients kept TFR, among them, 2 （5.71%） patients restarted TKI after 12 month suspension, and maintained MMR during suspension. 13 （37.1%）patients lost MMR, among them, 9 patients restarted TKI treatment, and 5 of them achieved MR4.0 after the median duration of 3(2-5) month. No patients were found to have disease progression. The estimated TFR rate was 57.8% and 51.8% at 12 and 24 months after discontinuation, respectively. Other clinical characteristic related to relapse were also analyzed, including the cumulative TKI administration duration, cumulative MMR duration, time to achieve MMR, median age at diagnosis, risk stratification by Sokal score, TKI dose reduction and discontinuation history, and second-generation TKI administration before stopping TKI, however, no statistical difference was found.TKI discontinuation is practical for CML patients in our center.慢性髓细胞白血病患者酪氨酸激酶抑制剂的主动停药临床结局的回顾性分析.研究单中心慢性髓细胞白血病（CML）酪氨酸激酶抑制剂（TKI）治疗后自动停药患者的临床特征及转归情况.回顾性分析2015年6月1日至2019年12月31日在本院长期随访并主动停用TKI的35例CML患者的临床资料，分析患者主动停药后无治疗缓解（TFR）的获得情况及其影响因素.35例患者累积服用TKI中位时间为72（35-173）个月，其中8例患者有减停药史。全部患者在停药前获得主要分子学缓解（MMR）及以上疗效。患者达到MMR中位时间为开始服用TKI后15（3-75）个月，停药前MMR维持时间为55（13-164）个月。停药后中位随访20.3（3-57.9）个月，22例（62.8%）患者获得TFR，其中2例（5.71%）患者因生育停用TKI 12个月以上后重启治疗，停药期间维持MMR。13例（37.1%）患者失去MMR，9例停药后失去MMR的患者再启动药物治疗，其中5例重启后已获得MR 4.0以上疗效，中位时间为 3（2-5）个月。随访全部患者，未发现疾病进展。本组患者经统计学分析显示，12个月TFR率为57.8%，24个月TFR率为51.8%。本研究比较了中位累积TKI治疗时间、中位累积MMR时间、达到MMR时间、中位诊断年龄、Sokal积分、是否具有TKI减停史、停药前使用二代TKI，未发现与分子学复发相关具有统计学差异的临床参数.本中心达停药标准的CML患者能够安全停用 TKI.

Microplastics have been found in many environmental systems, such as oceans, terrestrial soils, sediments, and bodies of freshwater. Microplastic pollution in soils has received extensive international attention; however, there is currently no unified standard extraction method. To identify appropriate extraction and component identification methods for microplastics in typical soils in China, samples were identified and their polymer component properties examined using ATR-FTIR(Thermo Nicolet IS 20). The effects of three treatments of water+oil(T1), a saturated NaCl solution+oil(T2), and a saturated NaCl solution(T3) on the detection of four types of microplastics[polyethylene terephthalate(PET), polypropylene(PP), polystyrene(PS), and polyethylene(PE)]in four typical soils(loess, black soil, red soil, and purple soil) were examined. The results showed that:① The extraction rates of the T3 treatment decreased with microplastic density. For PP, PE, and PS, the extraction rates exceeded 86.67%, and the extraction rates of PET were 0%-13.30%. ② The extraction rates of the four kinds of microplastics were 86.67%-100.00% in loess, black soil, and purple soil using the T1 and T2 solutions; however, the residual oil on the surface of microplastics influenced the identification using FTIR. Furthermore, the cleaning effect of PE and PS was poor after wiping with anhydrous ethanol. ③ In red soil, the extraction rates of PET were 56.60% and 50.00% in T1 and T2, respectively; in the T3 treatment, the extraction rates of PET, PE, and PP were 3.33%, 10.00%, and 56.67%, respectively. These results show that the extraction rates of four types microplastics are highest for loess, black soil, and purple soil using T1 and T2, although these two methods need to be combined with anhydrous ethanol cleaning.
