Objective To evaluate the association between body composition and subsequent risk of the major gynecologic malignancies. Methods This is a prospective analysis of participants from the UK Biobank. We measured baseline body composition and confirmed cancer diagnosis through linkage to cancer and death registries. We evaluated hazard ratios (HRs) and confidence interval (CIs) with COX models adjusting for potential confounders. Results We document 1430 cases of the top three gynecologic malignancies (uterine corpus cancer 847 cases, ovarian cancer 514 cases, and cervical cancer 69 cases) from 245,084 female participants (75,307 were premenopausal and 169,777 were postmenopausal). For premenopausal women, whole body fat‐free mass (WBFFM) was associated with an increased risk of uterine corpus cancer (Adjusted HR per unit increase 1.04, 95% CI 1.02–1.06). For postmenopausal women, compared with the first quartile, the fourth quartile of WBFFM and whole body fat mass(WBFM) was associated with 2.16 (95% CI 1.49–3.13) times and 1.89 (95% CI 1.31–2.72) times of increased uterine corpus cancer risk, respectively. Regarding the distribution of body fat mass (FM)/fat‐free mass (FFM), FFM distributed in the trunk was associate with increased uterine corpus cancer risk in premenopausal (HR 1.18,95% CI 1.07–1.31) and postmenopausal women (HR 1.13,95% CI 1.09–1.18). Meanwhile, FM/FFM distributed in the limbs present an U‐shaped associations with uterine corpus cancer risk. We did not observe any association between aforementioned body composition indices with ovarian or cervical cancer. Conclusion FM is associated with an increased risk of uterine corpus cancer in postmenopausal women. Meanwhile, FFM is found to be a risk factor for uterine corpus cancer in both premenopausal and postmenopausal women. No association of body composition with ovarian or cervical cancer was observed., The aim of this study is to reveal the relationship between body composition and the risk of gynecological malignancies. In addition to finding that body fat mass(FM) is a risk factor for postmenopausal uterine corpus cancer, we also find that fat‐free mass(FFM) is a stronger risk factor for uterine corpus cancer in both premenopausal and postmenopausal women. No association of body composition with ovarian or cervical cancer was observed.

Background Familial chylomicronemia syndrome (FCS) is a rare autosomal recessive lipid disorder often associated with recurrent episodes of pancreatitis. It is documented in most cases with FCS due to the mutations of key proteins in lipolysis, including LPL, APOC2, APOA5, LMF1 and GPIHBP1. Case presentation We report the successful management of a 35-year-old pregnant woman carrying a novel homozygous frameshift mutation c.48_49insGCGG (p.P17A fs*22) in the GPIHBP1 gene with previous severe episodes of acute pancreatitis triggered by pregnancy, resulting in adverse obstetrical outcomes. With careful monitoring, the patient underwent an uneventful pregnancy and delivered a baby with no anomalies. Conclusions The case report contributes to the understanding of GPIHBP1-deficient familial chylomicronemia syndrome (FCS) and highlights gestational management of FCS patient.

Satellites have shown free-space quantum-communication ability; however, they are orbit-limited from full-time all-location coverage. Meanwhile, practical quantum networks require satellite constellations, which are complicated and expensive, whereas the airborne mobile quantum communication may be a practical alternative to offering full-time all-location multi-weather coverage in a cost-effective way. Here, we demonstrate the first mobile entanglement distribution based on drones, realizing multi-weather operation including daytime and rainy nights, with a Clauser-Horne-Shimony-Holt S-parameter measured to be 2.41 ± 0.14 and 2.49 ± 0.06, respectively. Such a system shows unparalleled mobility, flexibility and reconfigurability compared to the existing satellite and fiber-based quantum communication, and reveals its potential to establish a multinode quantum network, with a scalable design using symmetrical lens diameter and single-mode-fiber coupling. All key technologies have been developed to pack quantum nodes into lightweight mobile platforms for local-area coverage, and arouse further technical improvements to establish wide-area quantum networks with high-altitude mobile communication., We demonstrate the first drone-based entanglement distribution under multi-weather conditions, which exhibits the third possible experimental strategy for quantum communication, in addition to fiber and satellite.

To observe the effect of electroacupuncture (EA) on Leptin and Leptin receptor (OB-Rb) of insulin resistant obese (OIR) rats, so as to explore its possible mechanism for obesity.Twenty-four Wistar rats were randomly divided into normal, model and EA groups, with 8 rats in each group. The OIR model was established by feeding the rats with high-fat diet for 8 weeks. EA (2 Hz, 1 mA) was applied to "Zhongwan" (CV12), "Guanyuan" (CV4), "Zusanli" (ST36) and "Fenglong" (ST40) for 30 min, once every other day for 8 weeks. At the 6th week of intervention, glucose contents of intraperitoneal glucose tolerance test (IPGTT) and intraperitoneal insulin tolerance test (IPITT) were measured. After 8 weeks' intervention, the body weight and food intake were recorded. Serum total cholesterol (TC) and total triglyceride (TG) were assayed by using an automatic biochemical analyzer. The expression levels of Leptin and OB-Rb proteins in the small intestine and hypothalamus were detected by Western blot.Following modeling, the body weight, food intake, IPGTT, IPITT, the contents of serum TC and TG, and the expressions of Leptin protein in small intestine and hypothalamus were significantly up-regulated (EA can improve insulin resis-tance by up-regulating the OB-Rb protein expression and enhancing the binding force of Leptin and OB-Rb.

Entanglement distribution has been accomplished using a flying drone, and this mobile platform can be generalized for multiple mobile nodes with optical relay among them. Here we develop the first optical relay to reshape the wave front of photons for their low diffraction loss in free-space transmission. Using two drones, where one distributes the entangled photons and the other serves as relay node, we achieve entanglement distribution with Clauser-Horne-Shimony-Holt S parameter of 2.59±0.11 at 1 km distance. Key components for entangled source, tracking, and relay are developed with high performance and are lightweight, constructing a scalable airborne system for multinode connectio and toward mobile quantum networks.

We study the effect of waveguide thickness variations on the frequency spectrum of spontaneous parametric down-conversion in the periodically-poled lithium niobate on insulator (LNOI) waveguide. We analyze several variation models and our simulation results show that thickness variations in several nanometers can induce distinct effects on the central peak of the spectrum, such as narrowing, broadening, and splitting. We also prove that the effects of positive and negative variations can be canceled and thus lead to a variation-robust feature and an ultra-broad bandwidth. Our study may promote the development of on-chip photon sources in the LNOI platform, as well as opensup a way to engineer photon frequency state.

Two core-shell structure poly methyl methacrylate-caprolactam (PMMA-CPL) and poly methyl methacrylate-caprolactam-sericin (PMMA-CPL-SS) nanoparticles were synthesized by emulsion polymerization. A latex blending process was performed on preparation of natural rubber (NR) blends, NR/PMMA-CPL and NR/PMMA-CPL-SS. Transmission electron microscopy (TEM) images showed that PMMA-CPL and PMMA-CPL-SS nanoparticles covering on the surface of NR particles homogeneously. SEM, AFM, ATR-FTIR and DSC were used to compare the difference between NR/PMMA-CPL-SS and NR/PMMA-CPL. The results showed that addition of SS improved the smoothness and reduced the amount of incompatible particles on the surface of NR/PMMA-CPL-SS film. Due to the introduction of CPL and SS, the number of escaping particles on the surface of NR was decreased during film forming process and the Tg of NR shifted to a higher temperature. The results indicated that the interfacial adhesion and compatibility of PMMA and NR were improved. Mechanical testing showed that PMMA-CPL nanoparticles could improve the mechanical property of NR. When the content of PMMA-CPL-SS was 10%, the tensile strength of NR/PMMA-CPL-SS film was increased by 19.6% compared to NR/PMMA-CPL. Moreover, the NR/PMMA-CPL-SS film exhibited a higher hydrophilic property after modification with PMMA-CPL-SS nanoparticles.

As an initial channel for users learning about a mobile game, the interactive experience of the navigation interface will directly affect the first impression of the users on the game and their subsequent behaviors and willingness to use. This study aims to investigate players&rsquo, visual attention mechanisms of various interactive levels of mobile games&rsquo, interfaces under free-browsing and task-oriented conditions. Eye-tracking glasses and a questionnaire were used to measure the interactive experience of mobile games. The results show that in the free-browsing condition, the fixation count, saccade count and average saccade amplitude can be used to reflect and predict the interactive experiences of mobile games&rsquo, navigation interface, while in the task-oriented condition, the fixation count, first fixation duration, dwell time ratio and saccade count can be used to reflect and predict the interactive experience of mobile games&rsquo, navigation interface. These findings suggest that apart from the different eye movement indicators, players&rsquo, motivations should also be considered during the process of the games&rsquo, navigation interface design.

Recent evidence indicates that long noncoding RNA colon cancer-associated transcript-1 (lncRNA CCAT1) is abundantly expressed in esophageal cancer and is closely related to the occurrence, development, invasion, metastasis, and drug resistance of this disease. However, the role and molecular mechanisms of CCAT1 in the cell proliferation and chemoresistance of esophageal cancer are largely unknown. The correlation between CCAT1 expression and drug resistance to cisplatin (CDDP) in esophageal squamous cell carcinoma (ESCC) cells was analyzed using 3-(4,5-dimethylthiazol-2-yl)-2,5-diphenyl-tetrazolium bromide (MTT) and quantitative real-time polymerase chain reaction (qRT-PCR) assays. CCAT1 knockdown and miR-143 overexpression or inhibition were used to verify the effects on proliferation and drug resistance via MTT, western blotting, flow cytometry, and immunofluorescence assays. qRT-PCR and western blotting were applied to detect the potential regulatory relationship among CCAT1, miR-143, PLK1, and BUBR1. A xenograft tumor assay was performed to validate the role of CCAT1 in vivo. The expression of CCAT1 was positively correlated with drug resistance in several ESCC cell lines. CCAT1 knockdown and miR-143 overexpression inhibited cell proliferation and CDDP drug resistance. Moreover, the downstream target of CCAT1 was found to be miR-143, which can regulate the expression of PLK1 and BUBR1. In vivo assays showed that CCAT1 knockdown suppressed tumor growth and enhanced the sensitivity of tumors to CDDP in nude mice. Taken together, we discovered a novel mechanism by which CCAT1 promotes cell proliferation and enhances drug resistance by regulating the miR-143/PLK1/BUBR1 signaling axis both in vitro and in vivo. Our findings further suggest that lncRNA CCAT1 may be a potential therapeutic target for overcoming chemoresistance in esophageal cancer.

We study the effect of dimension variation for second-harmonic generation (SHG) in lithium niobate on insulator (LNOI) waveguides. Non-trivial SHG profiles in both type-0 and type-I quasi-phase matching are observed during the wavelength tuning of the fundamental light. Theoretical modeling shows that the SHG profile and efficiency can be greatly affected by the waveguide cross-section dimension variations, especially the thickness variations. In particular, our analysis shows that a thickness variation of tens of nanometers is in good agreement with the experimental results. Such investigations could be used to evaluate fabrication performance of LNOI-based nonlinear optical devices.

Quaternized polyvinyl alcohol (QPVA) was synthesized and used as an intermedium to improve dispersion of silica (SiO2) in natural rubber (NR). QPVA/SiO2 nanoclusters reinforced NR nanocomposite was prepared by latex compounding via electrostatic interaction. TEM micrographs demonstrated QPVA/SiO2 nanoclusters were distributed around NR particles, forming shell-core structure. The mechanical properties and thermal ageing resistance of NR-QPVA/SiO2 were significantly improved compared with that of neat NR. The tensile strength of NR-QPVA/SiO2 film was improved by 60%, when the SiO2 content is 3%. SEM pictures indicated SiO2 was homogenous dispersed throughout NR matrix in the presence of QPVA. It also demonstrated that SiO2 could enhance thermal stability of NR, as NR-QPVA/SiO2 had best surface morphology after 72 hours thermal ageing at 100 °C. The thermal decomposition temperature and glass transition temperature of NR-QPVA/SiO2 film increased to a higher temperature due to strong polymer–filler interaction, which also indicated that all the ingredients were compatible and homogenous.

The present study was designed to investigate the chemical constituents of the whole herb of Dichrocephala benthamii. A new megastigmane glucoside (compound 1), together with its four known analogues (compounds 2-5), was obtained. Their structures were elucidated on the basis of spectroscopic analyses (UV, IR, MS, and 1D and 2D NMR). The absolute configuration of compound 1 was assigned on the basis of CD method and chemical evidence. In addition, their cytotoxicity against human hepatoma cells (HepG-2) was evaluated by the MTT method. Compound 5 showed weak activity against HepG-2, while the other compounds did not show remarkable inhibitory effects.

A novel organometallic chromophore with potential infrared (IR) chemical sensing property, named as N'-(2-pyridinyl)-N-phenylurea tricarbonyl chromium, was synthesized and systematically characterized by nuclear magnetic resonance (NMR), mass spectra (MS) and elemental analysis. The recognition and IR sensing behavior toward fluoride and acetate anions was investigated in chloroform. The results of IR titration indicated that, when the concentration of the two anions was greater than 10(-5) mol.L-1, a good linear relationship between the stretching vibration of metal carbonyl of N'-(2-pyridinyl)-N-phenylurea tricarbonyl chromium and the concentration of the two anions was observed, which exhibits expected application in trace analysis of fluoride and acetate with detection error less than 5%.

Activation of NF-κB pathway and co-stimulatory system CD40/CD40L promotes the inflammation, which plays a key role in the failure of islet graft. Therefore, the purpose of this study was to determine if simultaneous blockade of CD40/CD40L and IκB/NF-κB pathways could protect islet graft. Streptozocin-induced diabetic Wistar rats were transplanted intraportally with 2000 IEQ islets isolated from Sprague-Dawley rats. The rats were divided into five groups: nontreatment group, AdGFP-treated group, Ad-IκBα-treated group, Ad-sCD40LIg-treated group, and Ad-IκBα-IRES(2) -sCD40L-treated group. The islet graft mean survival time (MST), insulin expression of islet grafts, and the levels of cytokines in peripheral blood, were measured for the animals in each group. Our study confirmed that islet cells transfected with low doses of adenovirus could achieve high transfection efficiency, and would not affect the function of islet cells (P0.05). Splenocytes cultured with Ad-IκBα-IRES2-CD40L-transfected islets resulted in homospecific hyporesponsiveness. The islet graft MST (100 d) in the Ad-IκBα-IRES2-sCD40L-treated group was dramatically prolonged compared with that in the nontreatment group (7.1 ± 1.16 d). In addition, TNF-α, IL-1β, and IFN-γ were diminished in the Ad-IκBα-IRES2-sCD40L-treated group, which was commensurate with the reduced cellular infiltration (P0.01). Simultaneous blockade of the CD40/CD40L and IκB/NF-κB pathways could effectively extend the survival of islet grafts.

To establish a more convenient and effective method for isolating adult Sertoli cells and apply the method to islet transplantation.Trypsin, DNase, collagenase and hyaluronidase were used for a one-step digestion in group A. A two-step digestion was used in group B, in which testis tissues were first digested by trypsin and DNase and then were digested by collagenase and hyaluronidase. In group C, trypsin and DNase were used in the first step of digestion, hyaluronidase was used in the second step and collagenase was used in the third step. Sertoli cells were identified by morphology and immunohistochemistry and the viability and purity of Sertoli cells were detected by MTT and flow cytometry (FCM). Expression of Fas-L was detected by Western blot and the effects of the co-transplantation of islets and Sertoli cells were compared between the three groups.Typical Sertoli cells were seen after isolation using the three different methods. Sertoli cells isolated by method A and method B were large in number while those isolated by method B and method C were pure. The results of MTT showed that the viability of Sertoli cells in group B was significantly higher than that in group A and group C (P0.05) and Western blot results showed that expression of Fas-L on Sertoli cells in group B was significantly stronger than that in group A and group C (P0.05). The purity of Sertoli cells detected by FCM in group B and group C were significantly higher than that in group A (P0.05). The survival of islets co-transplanted with Sertoli cells to renal capsule of diabetic mice was significantly longer in group B compared with that in control group as well as in group A and group C (P0.05).Sertoli cells obtained by the two-step digestion method are of higher purity and viability, which significantly prolong the survival of islet graft by co-transplantation.

To construct a replication-incompetent recombinant adenovirus mediating short hairpin RNA (shRNA)-induced tissue factor gene silencing in the islet.Four pairs of complementary oligonucleotides were designed and synthesized to create double-stranded oligonucleotides (ds oligo). The ds oligos were cloned into Pentr/U6 vector to construct the shuttle plasmid pENTR/U6-shRNA, which was transduced into human islets via liposome after sequence verification. The plasmid with the best silencing effect was identified by real-time RT-PCR, followed by homologous recombination with the adenovirus backbone plasmid. The functional clone was transfected into 293A cells to amplify the adenovirus, whose silencing effect against TF expression was tested using real-time RT-PCR and Western blotting.The pENTR/U6-shRNA shuttle plasmid was constructed and verified by sequencing. The recombinant adenovirus-mediated shRNA against TF was constructed, and real-time RT-PCR and Western blotting demonstrated that the strongest silencing effect of the adenovirus against TF occurred on the 4th day following islet transfection.Replication-incompetent recombinant adenovirus-mediated shRNA against TF has been successfully constructed, which has good silencing effect against TF expression in human islet in vitro.

To study the impacts of kidney transplantation on erectile function and analyse its contributing factors.In order to evaluate the severity of erectile dysfunction (ED), a total of 250 married male kidney transplant recipients (KTR) with functioning graft were assessed with the International Index of Erectile Function (IIEF) questionnaire. Data of clinical characteristics, medical and sexual history and laboratory examination were collected. Univariate and multivariate logistic regression analyses were carried out to determine which have independent impacts on erectile function.The investigation was accomplished in 84.8% of the KTRs. There was no significant difference in ED incidence before and after renal transplantation (53.8% vs. 44.3%, P0.05). According to the IIEF score, erectile function improved in 43.9% of the KTRs, remained unchanged in 42.9%, and deteriorated in 13.2%, as compared with pre-transplantation. Logistic regression analysis showed that significant and independent influencing factors in erectile function were age, hemoglobin level, presence of DM and/or peripheral neuropathy and iterative transplantations, and their relative risks were 3.01, 2.01, 3.15, 3.89 and 2.67, respectively.ED is highly prevalent among KTRs and its pathogenesis is multifactorial. Age, presence of DM and/or peripheral neuropathy, hemoglobin level and iterative transplantations were chief contributing factors in erectile function.

To investigate the feasibility and benefits of co-culture of cryopreserved islets with small intestinal submucosa (SIS).Purified rat islets cryopreserved for one month were divided into SIS group and control group, and after culture in standard islet culture media RPMI1640 for 1 week, the morphology and function of the islets were assessed.The SIS protects the fragile islets from damage by cryopreservation, and increased the recovery from (60.6-/+3.3)% to (91.7-/+1.8) % (P0.05). Compared with the control group, incubation of the islets of the SIS group in high-glucose (16.7 mmol/L) solution resulted in significantly enhanced insulin secretion (23.7-/+1.6 vs 12.5-/+1.1 mU/L, P0.05). When the islets were incubated in high-glucose solution containing theophylline, the calculated stimulation index of SIS group was about 3-fold higher than that of the control group.Co-culture of cryopreserved rat islets with SIS can increase the recovery of islet cells and improve their function.

To investigate the role of alginate-polylysine-alginate (APA) microcapsules in protecting rat islet cells in cryopreservation.Purified rat islet cells microencapsulated with APA and free islet cells were cryopreserved for one month and then thawed for culture in RPMI 1640 overnight. The morphology of the cells was observed and their function assessed by stimulated insulin release test.APA microcapsulation protected the fragile islets from freezing damage by increasing the recovery rate of the cells from 68.6%+/-2.9% to 94.7%+/-1.4% (P0.05). After incubation with high glucose (16.7 mmol/L) solution, the insulin release from the encapsulated cells after cryopreservation significantly increased in comparison with that of the nonencapsulated cells (22.6+/-1.8 mU/L vs 11.7+/-1.5 mU/L, P0.05). In high glucose solution containing theophylline, the calculated stimulation index of the encapsulated cells was about 3 times that of the nonencapsulated cells.APA microencapsulation may significantly increase the post-thaw recovery and improve the function for cryopreserved rat islets.

To evaluate the effect of HSGJ on chronic allograft nephropathy (CAN) using standard rat model of CAN.Renal transplantation was performed with Fisher rats as donors and Lewis rats as recipients. All the recipients were randomly divided into control group and medication groups (high and low dosage of HSGJ, fed every other day). After 16 weeks of treatment, renal function and the histological alteration of CAN were measured. The expression of the TGFbeta1 mRNA in the allograft was evaluated by real-time PCR.The content of 24 h urine protein and the level of serum creatinine in the medication groups were significantly decreased (P0.01) as compared with control group, whereas the creatinine clearance was increased (P0.01). The degree of glomerular sclerosis and the Banff score of medication groups were lower than the control group respectively (P0.01), in consistent with decreased expression of the TGF 1mRNA.HSGJ can prevent the chronic allograft nephropathy and the mechanism may be related with its influence on the expression of the TGFbeta1.

The ability to maintain isolated human islet preparation in tissue culture has recently been adopted by most islet transplant centers to improve the safety and practicality of islet transplantation. However, maintaining islet viability and recovery remains a challenge in clinical setting. Extracellular matrix (ECM) is one of the most important components of islet microenvironment. The reconstruction of the cell-matrix relationship seems to be effective in improving the loss of differentiated islet structure and function. Small intestinal submucosa (SIS), a naturally occurring ECM, has been investigated to be able to promote wound healing, tissue remodeling, and cell growth. The purpose of this study was to evaluate the recovery and function of isolated rat pancreatic islets after in vitro culture with SIS.Pancreatic islets were isolated from Wistar rats by using standard surgical procurement followed by intraductal collagenase distension, mechanical dissociation, and EuroFicoll purification. Groups of purified islets were cultured in plates which were coated with multilayer SIS (SIS-treated group) or without (standard cultured group) for 7 days and 14 days in standard islet culture conditions of RPMI 1640 tissue culture media in humidified atmosphere containing 95% air and 5% CO2 at 37 degree centigrade. The mean recovery of islets after the culture period was determined by sizing duplicate counts of a known volume and their viability was assessed by static incubation with low glucose (2.7 mmol), high glucose (16.7 mmol) and high glucose solution supplemented with 50 mum 3-isobutyl-1-methylxanthine (IBMX) solution.After 7 days and 14 days of in vitro tissue culture, the SIS-treated group showed a significantly higher recovery compared with those cultured under standard conditions. The recovery in the SIS-treated group was about two times of the control group cultured in standard conditions after 14 days culture. In the SIS-treated group, there was no statistically difference between the short and long periods of culture(95.8+/-1.0% vs. 90.8+/-1.5%, P0.05). During incubation in high glucose (16.7 mmol) solution, there was a 2-3 fold increase in insulin secretion from both groups,but the SIS-treated group showed a higher increase than the standard cultured group after 14-day culture (20.7+/-1.1 mU/L vs. 11.8+/-1.1 mU/L, P0.05). When islets were placed in the high glucose solution supplemented with IBMX, the stimulated insulin response in the SIS-treated group was higher than that in the standard cultured group in spite of the duration of the culture. The stimulation index of the SIS-treated group was about 2-3 times of the standard cultured group. In addition, after a long period of culture, the stimulation index of the SIS-treated group was statistically equivalent with that of the short period of culture (9.5+/-0.2 vs. 10.2+/-1.2, P0.05).The co-culture of isolated rat islets with native sheet-like SIS can provide an excellent extracellular matrix, possible biotrophic and growth factors that promote the recovery and subsequent function of islets after in vitro tissue culture. In view of results of this study and rapid degradation of SIS in vitro, future studies will investigate the extended duration of culture and the effect of SIS on islets in vitro.

To investigate the effect of Danshen injection (DSI) on early stage of renal transplantation.One hundred and twelve patients in early stage after renal transplantation were allocated in the treated group, they were treated by conventional treatment with DSI 60 ml given additionally once a day for 10 days. And 109 patients who received conventional treatment alone after renal transplantation at the corresponding period were allocated in the control group. Indexes in the two groups, including volume of urine, serum creatinine (SCr), endogenous creatinine clearance rate, incidence of delayed graft function and acute rejection reaction, blood viscosity (BV), platelet aggregation rate (PAR) as well as the blood flow resistance in graft measured by color Doppler ultrasonography.The urinary volume and endogenous creatinine clearance rate in the treated group were significantly higher, but levels of SCr, incidence of renal function recovery retardation, BV, PAR and blood flow resistance in graft were significantly lower than those in the control group (P0.05). The difference of incidence of acute rejection reaction between the two groups was insignificant (P0.05).DSI can improve blood microcirculation, decrease the incidence of renal function recovery retardation, these effects are helpful for recovery of renal function after renal transplantation.
