Triterpenoids widely exist in nature, displaying a variety of pharmacological activities. Determining triterpenoids in different matrices, especially in biological samples holds great significance. High-performance liquid chromatography (HPLC) has become the predominant method for triterpenoids analysis due to its exceptional analytical performance. However, due to the structural similarities among botanical samples, achieving effective separation of each triterpenoid proves challenging, necessitating significant improvements in analytical methods. Additionally, triterpenoids are characterized by a lack of ultraviolet (UV) absorption groups and chromophores, along with low ionization efficiency in mass spectrometry. Consequently, routine HPLC analysis suffers from poor sensitivity. Chemical derivatization emerges as an indispensable technique in HPLC analysis to enhance its performance. Considering the structural characteristics of triterpenoids, various derivatization reagents such as acid chlorides, rhodamines, isocyanates, sulfonic esters, and amines have been employed for the derivatization analysis of triterpenoids. This review comprehensively summarized the research progress made in derivatization strategies for HPLC detection of triterpenoids. Moreover, the limitations and challenges encountered in previous studies are discussed, and future research directions are proposed to develop more effective derivatization methods.

Key Clinical Message Maxillary canines are often impacted, which can result in tooth disorders and adversely affect occlusal and facial development. The case report describes complete bilateral impaction of maxillary canines and significant root resorption of a central incisor. The multidisciplinary approach is the optimal strategy for addressing impacted maxillary canines.

Lysosomes-associated membrane proteins (LAMPs), a family of glycosylated proteins and major constituents of the lysosomal membranes, play a dominant role in various cellular processes, including phagocytosis, autophagy and immunity in mammals. However, their roles in aquatic species remain poorly known. In the present study, three lamp genes were cloned and characterized from Micropterus salmoides. Subsequently, their transcriptional levels in response to different nutritional status were investigated. The full-length coding sequences of lamp1, lamp2 and lamp3 were 1251bp, 1224bp and 771bp, encoding 416, 407 and 256 amino acids, respectively. Multiple sequence alignment showed that LAMP1-3 were highly conserved among the different fish species, respectively. 3-D structure prediction, genomic survey, and phylogenetic analysis were further confirmed that these genes are widely existed in vertebrates. The mRNA expression of the three genes was ubiquitously expressed in all selected tissues, including liver, brain, gill, heart, muscle, spleen, kidney, stomach, adipose and intestine, lamp1 shows highly transcript levels in brain and muscle, lamp2 displays highly expression level in heart, muscle and spleen, but lamp3 shows highly transcript level in spleen, liver and kidney. To analyze the function of the three genes under starvation stress in largemouth bass, three experimental treatment groups (fasted group and refeeding group, control group) were established in the current study. The results indicated that the expression of lamp1 was significant induced after starvation, and then returned to normal levels after refeeding in the liver. The expression of lamp2 and lamp3 exhibited the same trend in the liver. In addition, in the spleen and the kidney, the transcript level of lamp1 and lamp2 was remarkably increased in the fasted treatment group and slightly decreased in the refed treatment group, respectively. Collectively, our findings suggest that three lamp genes may have differential function in the immune and energetic organism in largemouth bass, which is helpful in understanding roles of lamps in aquatic species.

Abstract Objective Accurate quantification of the root surface area (RSA) plays a decisive role in the advancement of periodontal, orthodontic, and restorative treatment modalities. In this study, we aimed to develop a dynamic threshold-based computer-aided system for segmentation and calculation of the RSA of isolated teeth on cone-beam computed tomography (CBCT) and to assess the accuracy of the measured data. Method We selected 24 teeth to be extracted, including single-rooted and multi-rooted teeth, from 22 patients who required tooth extraction. In the experimental group, we scanned 24 isolated teeth using CBCT with a voxel size of 0.3 mm. We designed a computer-aided system based on a personalized dynamic threshold algorithm to automatically segment the roots of 24 isolated teeth in CBCT images and calculate the RSA. In the control group, we employed digital intraoral scanner devices to perform optical scanning on 24 isolated teeth and subsequently manually segmented the roots using 3-matic software to calculate the RSA. We used the paired t-test (P 0.05). As per the Bland-Altman plot, the results were evenly distributed within the region of ± 1.96 standard deviations of the mean, with no increasing or decreasing trends and good consistency. Conclusion In this study, we designed a computer-aided root segmentation system based on a personalized dynamic threshold algorithm to automatically segment the roots of isolated teeth in CBCT images with a voxel size of 0.3 mm. We found that the RSA calculated using this approach was highly accurate, and a voxel of 0.3 mm in size could accurately display the surface area data in CBCT images. Overall, our findings in this study provide a foundation for future work on accurate automatic segmentation of tooth roots in full-mouth CBCT images and the computation of RSA.

Herbal medicine has been widely valued because of its remarkable efficacy and minimal side effects. The quantitative analysis of herbal medicines is essential to ensure their safety and efficacy. The simultaneous detection of multiple quality markers (Q-markers) has emerged as an important approach and trend in herbal medicine quality control. In recent years, non-targeted screening has become an effective strategy for the discovery and identification of unknown compounds. This study developed a non-targeted screening and quantitative analysis strategy to discover, identify and quantify the multiple components that truly represent the efficacy of Wuling capsule. Within this strategy, 18 types of flavonoids were tentatively discovered and identified from Wuling capsule by analyzing mass cleavage pathways, the precise molecular weights of compounds, and comparing the data with a database. Ten types of flavonoids were determined after the comparison of the standards. Additionally, following the evaluation of the regression equation, linear range, limit of detection (LOD), limit of quantitation (LOQ), precision, repeatability, and recovery of the proposed quantitative method, six flavonoids were quantified. This method successfully screened, identified, and quantified the potential active components in Wuling capsule, providing insights for improving the quality control standards in other herbal medicines.

ObjectivePrimary blepharospasm (BSP) is a clinically heterogeneous disease that manifests not only as spasmodic closure of the eyelids but also sometimes with apraxia of eyelid opening (AEO). This cross-sectional study aimed to investigate differences in the neural mechanisms of isolated BSP and BSP-associated AEO subtypes, which may reveal the pathophysiology underlying different phenotypes.MethodsA total of 29 patients manifested as isolated BSP, 17 patients manifested as BSP associated with AEO, and 28 healthy controls underwent resting-state functional near-infrared spectroscopy (fNIRS). We assessed functional connectivity (FC) between regions of interest (ROIs) in the fronto-parietal control network (PFCN) and sensorimotor network (SMN). We also examined the relationship between altered FC and behavioral data.ResultsIn the FPCN, ROI- analyses showed decreased FC between the left premotor cortex and supramarginal gyrus in the BSP with AEO group compared to the isolated BSP group. In the SMN, both subgroups showed hypoconnectivity of the left premotor cortex with the right primary motor cortex, primary sensory cortex, and somatosensory association cortex. This hypoconnectivity was positively correlated with the total number of botulinum toxin A treatments, which suggests that long-term botulinum toxin A treatment may modulate motor sequence planning and coordination.ConclusionThese findings showed different connectivity alterations in neural networks associated with motor and cognitive control among different behavioral phenotypes of BSP. The identification of specific alterations in various networks that correspond to clinical heterogeneity may inform the identification of potential biomarkers for early diagnosis and personalized neuromodulation targets for treating different BSP subphenotypes.

Abstract Objective To investigate the safety and effectiveness of implanting temporary pacemakers using ultrasound‐guidance at the bedside for rescuing patients in case of cardiac emergencies. Methods We enrolled 194 patients with cardiac emergencies requiring temporary pacemakers in this study, and randomly assigned them to either a bedside ultrasound‐guided installation group or an electrocardiogram‐guided installation group. There were 105 cases in the bedside ultrasound‐guided installation group, aged approximately 66.3 ± 10.2 years, and 89 cases in the electrocardiogram‐guided installation group, aged approximately 65.8 ± 9.5 years old, and disease composition was similar between the two groups. We then compared the duration of the procedure, success rates, and occurrence of adverse events between the two groups. Results The two groups showed similar clinical characteristics. The success rates of venipuncture and temporary pacemaker electrode placement were both 100% in the bedside ultrasound‐guided installation group, compared to 87.8% and 96.7% respectively, in the electrocardiogram‐guided installation group, with a statistically significant difference between the two groups. The duration of puncture was significantly shorter in the bedside ultrasound‐guided installation group than in the electrocardiogram‐guided installation group, with statistically significant differences. Moreover, no adverse events such as hematoma, pneumothorax and electrode dislodgement occurred in the bedside ultrasound‐guided installation group, while 13 cases in the electrocardiogram‐guided installation group experienced adverse events, and the difference was statistically significant. Conclusions The bedside installation of temporary pacemakers using ultrasound guidance is a simple, safe, effective, and cost‐efficient procedure that boasts a high success rate, does not involve radiation, and enables accurate placement of the electrode catheter.

The previously reported begonias in a limestone forest of Guangxi mainly belong to Begonia sect. Coelocentrum Irmscher. In this article, we described and illustrated a new species in sect. Platycentrum (Klotzsch) A.DC., Begonia parvibracteata X.X.Feng, R.K.Li & Z.X.Liu, which was discovered in a karst forest of south-western Guangxi. The begonia shows high morphological similarity to B. subhowii S.H. Huang and B. psilophylla Irmscher, but differs from the latter two in its narrower oblique-ovate asymmetric leaf blade, 4 (occasionally 6) tepals of pistillate flower and smaller membranous inflorescence bracts. Molecular phylogenetic analysis, based on ITS sequence data, supports the new species as monophyletic and distinct from B. subhowii and B. psilophylla. Considering its narrow distribution and the disturbance of human activities, the conservation status of new taxon is evaluated as “Vulnerable” (VU B1, B2 ab (i, iv, v), D2) according to the IUCN Red List Categories and Criteria.

Urine glucose detection is an important diagnostic tool for screening and early diagnosis of diabetes mellitus. Detection of urine glucose has many advantages over blood glucose, such as non-invasive, easy-to-detect, simple sampling and being well accepted by patients. Therefore, it is commonly used to monitor diabetes progression, assist in therapeutic intervention as well as in point-of-care testing (POCT). In recent years, with the development of material science, electrochemistry and miniaturization technology, novel applications of natural enzymes, nanozymes as well as nanomaterials, such as metal (Au, Pt, Ni, Co, etc.), alloy, grapheme, in the analysis of urine glucose level have been increasing sharply. In particular, different types of nanozymes-based biosensors, inspired by natural enzymes, have been developed with improved characteristics of being low-cost, stable, and mass-produced. On the other hand, making use of portable devices, such as smartphones and microfluidic paper-based analytical devices, has facilitated on site accurate urine glucose monitoring in real time. All these rapid advancements in nanotechnologies and devices have contributed greatly to the development of cost effective, highly sensitive, user friendly urine glucose biosensors. This review summarizes the most recent improvements in two major types of urine glucose biosensors: the optical- and electrochemical-biosensors. We also discuss the limitations, challenges and perspectives of these biosensors. Finally, we propose future research directions, development trends and potential clinical applications of nanomaterial-based biosensors developed for urine glucose detection.

Abstract Organic–inorganic halide perovskites are emerging materials for photovoltaic applications with certified power conversion efficiencies (PCEs) over 25%. Generally, the microstructures of the perovskite materials are critical to the performances of PCEs. However, the role of the nanometer-sized grain boundaries (GBs) that universally existing in polycrystalline perovskite films could be benign or detrimental to solar cell performance, still remains controversial. Thus, nanometer-resolved quantification of charge carrier distribution to elucidate the role of GBs is highly desirable. Here, we employ correlative infrared-spectroscopic nanoimaging by the scattering-type scanning near-field optical microscopy with 20 nm spatial resolution and Kelvin probe force microscopy to quantify the density of electrons accumulated at the GBs in perovskite polycrystalline thin films. It is found that the electron accumulations are enhanced at the GBs and the electron density is increased from 6 × 1019 cm−3 in the dark to 8 × 1019 cm−3 under 10 min illumination with 532 nm light. Our results reveal that the electron accumulations are enhanced at the GBs especially under light illumination, featuring downward band bending toward the GBs, which would assist in electron-hole separation and thus be benign to the solar cell performance. Correlative infrared-spectroscopic nanoimaging by the scattering-type scanning near-field optical microscopy and Kelvin probe force microscopy quantitatively reveal the accumulated electrons at GBs in perovskite polycrystalline thin films.

Background: Histological assessment of resected margins has some drawbacks. We therefore aimed to identify a panel of metabolic markers for evaluating the surgical margins of oral squamous cell carcinoma during surgery. Methods: A total of 28 case of OSCC samples were enrolled in the study. Gas chromatography-mass spectrometry based untargeted metabolic analysis was employed to acquire the metabolic perturbation of the distance-related surgical margins in the development group. The acquired MS data were then subjected to univariate and multivariate analysis by MetaboAnalyst. Ultra–high performance liquid chromatography–tandem mass spectrometerbased targeted metabolomics for quantitative analysis of the validation group was performed to verify the results of the development group. Another 60 OSCC patients with dysplastic surgical margins were used to further validate the results of the development group by immunohistochemical examination of key enzyme expression, and correlate them with clinicopathological parameters and clinical outcomes. Findings: We finally identified 4 amino acids as negative margin markers, and 6 amino acids as dysplastic margin markers. IHC analysis showed that asparagine synthetase positive expression in dysplastic surgical margins and its higher expression was correlated with tumor recurrence and local relapse-free survival. Interpretations: We developed a panel of metabolic molecular markers to supplement the evaluation of negative and dysplastic margins. The content and kind of trace elements in magnesium alloys have important effects on their as-cast and semi-solid microstructures. In this research work, effects of trace Cr on as-cast and semi-solid microstructures of ZC61 magnesium alloy were investigated by metal mold casting and semi-solid isothermal heat treatment. The results show that the addition of Cr can refine the α-Mg phase without generating a new phase, noticeably change the eutectic phase, and decrease the average size of solid particles at the same isothermal heat treatment conditions. Non-dendritic microstructures of all alloys are constituted of α1-Mg phases, α2-Mg phases and eutectic phases after water quenching. With isothermal temperature increased or holding time prolonged, the eutectic microstructure (α-Mg+MgZn2+CuMgZn) at the grain boundaries in as-cast alloy is melted preferentially and then turned into semi-solid non-dendritic microstructure by processes of initial coarsening, microstructure separation, spheroidizing and final coarsening. Especially when the ZC61-0.1Cr alloy was treated at 585 °C for 30 min, the ideal non-dendritic microstructure can be obtained, and the corresponding solid particle size and shape factor were 37.5 μm and 1.33, respectively. The coarsening process of solid α-Mg phase at higher temperature or longer time, which is affected by both combining growth and Ostwald ripening mechanism, is refrained when Cr is added to the ZC61 alloy.

Objective: To investigate the roles of serum uric acid (UA), bilirubin (BIL), albumin (ALB), and creatinine (CRE) as major intravascular antioxidants, in benign paroxysmal positional vertigo (BPPV).Methods: The serum levels of UA, BIL, ALB, and CRE were retrospectively analyzed in 70 patients with new-onset idiopathic BPPV and 140 age- and sex-matched healthy controls (HCs).Results: Serum UA, BIL, ALB, and CRE levels were significantly lower in the BPPV group than the HC group. Furthermore, serum levels of BIL and ALB were significantly lower in the BPPV group when compared by sex. Multiple stepwise logistic regression revealed that a reduction in serum ALB was independently related to BPPV (odds ratio = 0.688; 95% confidence interval = 0.607– 0.780). Receiver operating characteristic analyses revealed a cut-off value of 45.15 g/L for ALB with a sensitivity of 74.29% (62.97– 83.07%) and specificity of 73.57% (65.71– 80.18%).Conclusions: Serum levels of UA, BIL, ALB, and CRE were lower in BPPV patients, indicating a lower antioxidant status. Furthermore, a reduction in serum ALB was independently associated with BPPV. These results provide insights into the possible roles of oxidative stress in the pathogenesis of BPPV.

Odorant-binding proteins (OBPs) are important for the perception of chemical signals by insects. Effective pest management strategies can be developed by understanding the host location mechanism and the physiological functions of OBPs in olfactory detection. In this study, we cloned two OBPs from Monochamus alternatus, where MaltOBP9 was highly expressed in multiple insect tissues and MaltOBP10 was highly expressed in the female antenna according to the results of qRT-PCR. The recombinant proteins were successfully purified in vitro. Immunocytochemistry indicated the high expression of MaltOBP9 and MaltOBP10 in the sensillum lymph of sensilla basiconica, sensilla trichodea, sensilla auricillica, and sensilla chaetica, thereby demonstrating their broad participation in semiochemical detection. Both proteins were localized in the inner cavity of mechanoreceptors and they exhibited broad binding abilities with volatiles from pine bark according to fluorescence competitive binding assays. Due to its broad binding ability and distribution, MaltOBP9 may be involved in various physiological processes as well as olfactory detection. MaltOBP10 appears to play a role in the fundamental olfactory recognition process of female adults according to its broad binding ability. These findings suggest that OBPs may have various physiological functions in insects, thereby providing novel insights into the olfactory receptive mechanism.

A new rare earth magnesium alloy (Mg-6Zn-4Sm-0.4Zr, wt.%) was prepared by permanent mould casting. The microstructure and mechanical properties of the alloy sample in as-cast and various heat treatment situations were characterized with an optical microscope (OM), X-ray diffractometer (XRD), scanning electron microscope (SEM) equipped with energy dispersive spectroscope (EDS), transmission electron microscope (TEM) and mechanical tests at room temperature, respectively. The experimental results show that the as-cast alloy mainly consists of α-Mg, eutectic Mg2Zn3, MgZnSm and Mg41Sm5. These eutectic phases with continuous or semicontinuous morphology principally distribute along grain boundaries. Almost all the eutectic compounds dissolve in α-Mg and the grains have no obvious growth trend after optimum solution treatment at 490 °C for 18 h. Meanwhile, the ultimate tensile strength (UTS) of 229 MPa and elongation (EL) to rupture of 9.78% can be achieved through the optimal solution treatment, which increase by 37 MPa and 57.74%, respectively, compared with that of the as-cast alloy. Further aging treatments at 200 °C for different durations lead to the conspicuous increment of mechanical properties and prominent age-hardening response. Peak-aged alloy (treated at 200 °C for 12 h) reveals better mechanical properties (UTS 258 MPa, EL 9.42%, hardness 73.4 HV) compared with the same alloy treated in other aging conditions, which is mainly ascribed to precipitated Mg2Zn3 and MgZn2 phases. Fracture analysis demonstrates that the as-cast alloy belongs to inter-granular and cleavage fracture patterns, while the solutionized alloy (treated at 490 °C for 18 h) reveals trans-granular and quasi-cleavage fracture modes. For the peak-aged alloy, the fracture pattern obeys the mixture of trans-granular and cleavage modes.

The application of Mg-Zn binary alloys is restricted due to their developed dendritic microstructure and poor mechanical properties. In this study, an alloying method was used to improve the mechanical properties of Mg-Zn alloy. The Mg-6Zn magnesium alloys microalloyed with varying Cu content (0, 0.8, 1.5, 2.0 and 2.5wt.%) were fabricated by permanent mould casting, and the effects of Cu content on the microstructure and mechanical properties of as-cast Mg-6Zn alloys were studied using OM, SEM, XRD and tensile tests at room temperature. The obtained results show that the addition of Cu not only can refine the grains effectively, but also can modify the eutectic morphology and improve the mechanical properties of the alloys. The main phases of the studied alloys include α-Mg, MgZn2, Mg2Cu and CuMgZn. When the content of Cu exceeds 0.8wt.%, Mg2Cu phase appears. Meanwhile, the eutectic morphology is modified into dendritic shape or lamellar structure, which has an adverse effect on the tensile properties. Furthermore, among the investigated alloys, the alloy containing 0.8% Cu shows an optimal ultimate tensile strength of 196 MPa, while the alloy with 1.5wt.% Cu obtains an excellent elongation of 7.22%. The experimental alloys under different Cu contents show distinguishing fracture behaviors: the fracture of the alloy with 0.8wt.% Cu reveals a mixed mode of inter-granular and quasi-cleavage, while in other investigated alloys, the fracture behaviors are dominated by cleavage fracture.

The application of segmental semi-solid thixoforming of magnesium alloys is confined due to the dimensional distinction existing in solid particles of the alloy billet from edge to center zones. In the present study, the effects of Sm addition on the microstructural evolution of Mg-6Zn-0.4Zr and Mg-6Zn-4Sm-0.4Zr alloys by semi-solid isothermal heat treatment were investigated, to obtain optimum semi-solid microstructures for the subsequently thixoforming. The results indicate that the grains of the Sm-bearing alloy are evidently refined and gradually evolve from dendritic to globular and elliptic particles. In addition, the distinctly dimensional effect of the Mg-6Zn-0.4Zr alloy is eliminated with 4% Sm addition; the particle sizes in all zones from center to the edge of the billet are almost identical. With the increment of isothermal heat treatment temperature, the dendritic microstructures completely disappear, and meanwhile, the irregular and globular particles gradually form. The size, morphology and the distribution of solid particles mainly depend on the formation and permeation of the liquid phase in the process of isothermal heat treatment. As the isothermal temperature increases from 570 °C to 610 °C, the average size and shape factor of solid particles of both the alloys with and without Sm addition gradually decrease while the liquid fraction gradually increases.

Semisolid slurry of A356 aluminum alloy was prepared by self-inoculation method, and the microstructure and solidification behavior during rheo-diecasting process were investigated. The results indicate that the semisolid slurry of A356 aluminum alloy can be prepared by self-inoculation method at 600 °C. Primary α-Al particles with fine and spherical morphologies are uniformly distributed when the isothermal holding time of slurry is 3 min. Liquid phase segregation occurs during rheo-diecasting process of semisolid slurry and the primary particles (α1) show obvious plastic deformation in the area of high stress and low cooling rate. A small amount of dendrites resulting from the relatively low temperature of the shot chamber at the initial stage of secondary solidification are fragmented as they pass through the in-gate during the mould filling process. The amount of dendrite fragments decreases with the increase of filling distance. During the solidification process of the remaining liquid, the nucleation rate of secondary particles (α2) increases with the increase of cooling rate, and the content of Si in secondary particles (α2) are larger than primary particles (α1). With the increase of cooling rate, the content of Si in secondary particles (α2) gradually increases. The morphologies of eutectic Si in different parts of die casting are noticeably different. The low cooling rate in the first filling positions leads to coarse eutectic structures, while the high cooling rate in the post filling positions promotes small and compact eutectic structures.

(1) Background—The aim of the present study was to evaluate the correlation between the temporomandibular joint (TMJ) osseous morphology of normal skeletal pattern individuals with different dental malocclusions by using cone-beam computed tomography (CBCT). (2) Methods—The CBCT images of bilateral TMJs in 67 subjects with skeletal class I and average mandibular angle (26 males and 41 females, age range 20–49 years) were evaluated in this study. The subjects were divided into class I, class II division 1, and class II division 2 according to the molar relationship and retroclination of the maxillary incisors. Angular and linear measurements of TMJ were evaluated and the differences between the groups were statistically analyzed. (3) Results—Intragroup comparisons showed statistical differences for articular eminence inclination, the width of the glenoid fossa, the ratio of the width of the glenoid fossa to the depth of the glenoid fossa, the condylar angle, and the intercondylar angle between the malocclusion groups. The measurements of the glenoid fossa shape showed no significant difference between the left and right sides. Females showed more differences in the morphological parameters of TMJ between the three malocclusion groups than the males. (4) Conclusion—The present study revealed differences in the TMJ osseous morphology between dental class I and class II malocclusions in the normal skeletal pattern.

(1) Background: In order to determine the correlation between the inclination of articular eminence (AEI) and the development of temporomandibular disorders (TMDs), a systematic review was performed. (2) Methods: A systematic literature research was conducted between 1946 and January 2020, based on the following electronic databases: PubMed, Cochrane Library, Embase, Medline, Scope, SciELO, and Lilacs. Observational studies, analytical case-control studies, and cohort studies written in English were identified. The articles were selected and analyzed by two authors independently. The PICO format was used to analyze the studies and the Newcastle-Ottawa Scale (NOS) was used to verify the quality of the evidence. (3) Results: Sixteen articles were included in this review, ten case-control studies and six cohort studies. Eight articles (50%) established a positive relation between AEI and TMDs and eight (50%) did not. The scientific quality was medium-low, mainly influenced by the exposure to the risk of bias and the lack of clinical methods with adequate consistency and sensitivity on the diagnosis of TMDs. (4) Conclusions: It is controversial to establish a causal relationship between the TMDs and the AEI in the field of stomatology, due to limited and inconclusive evidence. However, it is suggested that the AEI defined by some specific methods may be associated with some special pathological stages of TMDs. High-quality prospective studies are required to draw any definitive conclusions.

Flavonoids represent a large group of polyphenols that have been recognized to exert a wide range of biological effects, such as anticancer, inflammation inhibition, anti-aging, and neuroprotective activities. In the forest industry, tree twigs have been treated as residues; however, tree twigs could be a rich source of high-value added compounds, which have been relatively unexplored. In this study, an investigation into the chemical constituent of extractives from S. japonica twigs resulted in the isolation of a new (Z)-caffeoyl flavonol glycoside that was elucidated as myricetin 3-O-(4′′-(Z)-caffeoyl)-α-rhamnopyranoside (IV). The structure of the new compound was established mostly on the basis of extensive spectroscopic techniques and other physiochemical evidences. Among the three known flavonoids extracted in this work, including isoquercitrin (I), isorhamnetin 3′-O-β-D-glucopyranoside (II), and myricitrin (III), II and III have never been previously reported in the Sophora genus.

Background/Aims: Nucleotide binding oligomerization domain 1 (NOD1) signal pathway and human ß defensins (hBDs) play crucial roles in innate immune. Cigarette smoke has been confirmed to dampen innate immune in some human tissues, such as oral mucosa. The aim of this study was to evaluate potential effects of smoking on NOD1 signaling and hBDs expression in oral mucosa. Methods: Tissue specimens of normal oral mucosa were collected from donors undergoing routine surgical treatment. All 20 participants were classified equally as two groups: non-smokers and smokers. By using Western blotting and immunohistochemistry, we investigated differential expression of crucial molecules in NOD1 signal pathway, hBD-1, -2, and -3 in oral mucosa tissues between non-smokers and smokers. Immortalized human oral mucosal epithelial (Leuk-1) cells were treated with various concentrations of cigarette smoke extract (CSE) for 24h. Western blotting and immunofluorescence assays were performed to study CSE-induced alteration of protein expression. Leuk-1 cells were treated with 4% CSE, iE-DAP (NOD1 agonist), CSE + iE-DAP, BAY 11-7082 (NF-κB inhibitor), 4% CSE + BAY 11-7082, respectively. Real-time PCR and ELISA were performed to detect the mRNA levels and secretion of hBD-1, -2, and -3, respectively. Results: The levels of NOD1, NF-κB, hBD-1 and hBD-3 significantly reduced in oral mucosa tissues of smokers compared with non-smokers. The levels of RIP2 (receptor-interacting protein 2), phospho-NF-κB (P-NF-κB) and hBD-2 remarkably enhanced in oral mucosal tissues of smokers. CSE treatment suppressed NOD1 and NF-κB expression and activated RIP2 and P-NF-κB expression in Leuk-1 cells. The mRNA and secretory levels of hBD-1 and -3 were down-regulated by CSE, while the mRNA and secretory level of hBD-2 were up-regulated by CSE. The iE-DAP or BAY 11-7082 treatment reversed the regulatory effects of CSE on levels of hBDs. Conclusion: The present study indicated that cigarette smoke could potentially modulate the expression of crucial molecules of NOD1 signal pathway and hBDs in human oral mucosal epithelium. NOD1 signal pathway could play an important role in the regulatory effects of CSE on hBDs levels in oral mucosal epithelial cells.

Primary blepharospasm (BPS) is a focal dystonia characterized by involuntary blinking and eyelid spasms. The pathophysiology of BPS remains unclear. Several neuroimaging studies have suggested dysfunction of sensory processing and sensorimotor integration, but the results have been inconsistent. This study aimed to determine whether patients with BPS exhibit altered functional brain connectivity and to explore possible correlations between these networks and clinical variables. Twenty-five patients with BPS and 25 healthy controls were enrolled. We found that the patient group exhibited decreased connectivity within the sensory-motor network (SMN), which involved regions of the bilateral primary sensorimotor cortex, supplementary motor area (SMA), right premotor cortex, bilateral precuneus and left superior parietal cortex. Within the right fronto-parietal network, decreased connections were observed in the middle frontal gyrus, dorsal lateral prefrontal cortex and inferior frontal gyrus. Regarding the salience network (SN), increased connectivity was observed in the left superior frontal gyrus and middle frontal gyrus. These findings suggest the involvement of multiple neural networks in primary BPS.

The phytochemical investigation, including fractionation and purification of 70% acetone extracts of Juglans sigillata seed husks, an agricultural residue, led to the isolation of five low-molecular weight galloyltannins. The structures of the extractives were elucidated as 1,2,6-tri-O-galloyl-β-D-glucose (1), 3,4,6-tri-O-galloyl-β-D-glucose (2), 2,3,4,6-tetra-O-galloyl-β-D-glucose (3), 1,2,3,4,6-penta-O-galloyl-β-D-glucose (4), and tannic acid (5), primarily based on their spectral (NMR and MS) and chemical evidence. Galloyltannins 1-5 showed strong inhibitory activity against mushroom tyrosinase, with IC50 values ranging from 35.27 to 76.37 μM; kojic acid, which was used as a positive control, had an IC50 value of 342.14 μM. It was further found that 1-5 inhibited melanin production and exhibited intracellular tyrosinase activity, as well as down-regulated mRNA and protein expression levels of tyrosinase, in B16F10 mouse melanoma cells. Therefore, the isolated extractives from seed husks of J. sigillata may serve as potential candidates for hyperpigmentation remediation and as skin-whitening agents in the cosmetics industry.

Cigarette smoke exposure is associated with increased risk of various diseases. Epithelial cells-mediated innate immune responses to infectious pathogens are compromised by cigarette smoke. Although many studies have established that cigarette smoke exposure affects the expression of Toll-liked receptor (TLR), it remains unknown whether the nucleotide-binding oligomerization domain-containing protein 1 (NOD1) expression is affected by cigarette smoke exposure. In the study, we investigated effects of cigarette smoke extract (CSE) on NOD1 signaling in an immortalized human oral mucosal epithelial (Leuk-1) cell line. We first found that CSE inhibited NOD1 expression in a dose-dependent manner. Moreover, CSE modulated the expression of other crucial molecules in NOD1 signaling and human β defensin (hBD) 1, 2 and 3. We found that RNA interference-induced Caspase-12 silencing increased NOD1 and phospho-NF-κB (p-NF-κB) expression and down-regulated RIP2 expression. The inhibitory effects of CSE on NOD1 signaling can be attenuated partially through Caspase-12 silencing. Intriguingly, Caspase-12 silencing abrogated inhibitory effects of CSE on hBD1, 3 expression and augmented induced effect of CSE on hBD2 expression. Caspase-12 could play a vital role in the inhibitory effects of cigarette smoke on NOD1 signaling and hBDs expression in oral mucosal epithelial cells.

BACKGROUND: To elucidate further from the biomechanical aspect whether microgravity-induced cerebral vascular mal-adaptation might be a contributing factor to postflight orthostatic intolerance and the underlying mechanism accounting for the potential effectiveness of intermittent artificial gravity (IAG) in preventing this adverse effect. METHODOLOGY/PRINCIPAL FINDINGS: Middle cerebral arteries (MCAs) were isolated from 28-day SUS (tail-suspended, head-down tilt rats to simulate microgravity effect), S+D (SUS plus 1-h/d -Gx gravitation by normal standing to simulate IAG), and CON (control) rats. Vascular myogenic reactivity and circumferential stress-strain and axial force-pressure relationships and overall stiffness were examined using pressure arteriography and calculated. Acellular matrix components were quantified by electron microscopy. The results demonstrate that myogenic reactivity is susceptible to previous pressure-induced, serial constrictions. During the first-run of pressure increments, active MCAs from SUS rats can strongly stiffen their wall and maintain the vessels at very low strains, which can be prevented by the simulated IAG countermeasure. The strains are 0.03 and 0.14 respectively for SUS and S+D, while circumferential stress being kept at 0.5 (106 dyn/cm2). During the second-run pressure steps, both the myogenic reactivity and active stiffness of the three groups declined. The distensibility of passive MCAs from S+D is significantly higher than CON and SUS, which may help to attenuate the vasodilatation impairment at low levels of pressure. Collagen and elastin percentages were increased and decreased, respectively, in MCAs from SUS and S+D as compared with CON; however, elastin was higher in S+D than SUS rats. CONCLUSIONS: Susceptibility to previous myogenic constrictions seems to be a self-limiting protective mechanism in cerebral small resistance arteries to prevent undue cerebral vasoconstriction during orthostasis at 1-G environment. Alleviating of active stiffening and increasing of distensibility of cerebral resistance arteries may underlie the countermeasure effectiveness of IAG.

Previous research has shown that composite shape-stabilized phase change material (CPCM) has a remarkable capacity for thermal storage and stabilization, and it can be directly applied to highway construction without leakage. However, recent studies on temperature changing behaviors of CPCM and asphalt mixture cannot intuitively reflect the thermoregulation mechanism and efficiency of CPCM on asphalt mixture. The objective of this paper is to determine the specific heat capacity of CPCM and asphalt mixtures mixed with CPCM using the heat exchange system and the data acquisition system. Studies have shown that the temperature-rise curve of 5 °C CPCM has an obvious temperature plateau, while an asphalt mixture mixed with 5 °C CPCM does not; with increasing temperature, the specific heat capacities of both 5 °C CPCM and asphalt mixture first increase and then decrease, while the variation rate of 5 °C CPCM is larger than that of the asphalt mixture, and the maximum specific heat capacity of 5 °C CPCM appears around the initial phase change temperature. It is concluded that the temperature intervals of 5 °C CPCM are −18 °C–7 °C, 7 °C–25 °C and 25 °C–44 °C, respectively, and that of the asphalt mixture are −18 °C~10 °C, −10 °C~5 °C and 5 °C~28 °C. A low dosage of 5 °C CPCM has little influence on the specific heat capacity of asphalt mixture. Finally, the functions of specific heat capacities and temperature for CPCM and asphalt mixture mixed with CPCM were recommended by the sectional regression method.

The 16th Universiade, an international multi-sport event, was hosted in Shenzhen, China from 12 to 23 August 2011. During this time, officials instituted the Pearl River Delta action plan in order to enhance the air quality of Shenzhen. To determine the effect of these controls, the current study examined the trace elements, water-soluble ions, and stable lead isotopic ratios in atmospheric particulate matter (PM) collected during the controlled (when the restrictions were in place) and uncontrolled periods. Fine particles (PM2.5) were collected at two sampling sites in Shenzhen: “LG”—a residential building in the Longgang District, with significant point sources around it and “PU”—Peking University Shenzhen Graduate School in the Nanshan District, with no significant point sources. Results from this study showed a significant increase in the concentrations of elements during the uncontrolled periods. For instance, samples at the LG site showed (controlled to uncontrolled periods) concentrations (in ng·m−3) of: Fe (152 to 290), As (3.65 to 8.38), Pb (9.52 to 70.8), and Zn (98.6 to 286). Similarly, samples at the PU site showed elemental concentrations (in ng·m−3) of: Fe (114 to 301), As (0.634 to 8.36), Pb (4.86 to 58.1), and Zn (29.5 to 259). Soluble Fe ranged from 7%–15% for the total measured Fe, indicating an urban source of Fe. Ambient PM2.5 collected at the PU site has an average 206Pb/204Pb ratio of 18.257 and 18.260 during controlled and uncontrolled periods, respectively. The LG site has an average 206Pb/204Pb ratio of 18.183 and 18.030 during controlled and uncontrolled periods, respectively. The 206Pb/204Pb ratios at the PU and the LG sites during the controlled and uncontrolled periods were similar, indicating a common Pb source. To characterize the sources of trace elements, principal component analysis was applied to the elements and ions. Although the relative importance of each component varied, the major sources for both sites were identified as residual oil combustion, secondary inorganic aerosols, sea spray, and combustion. The PM2.5 levels were severely decreased during the controlled period, but it is unclear if this was a result of the controls or change in meteorology.

Objective: To evaluate the diagnostic ability and clinical imaging features in maxillofacial soft tissue hypervascular tumours by 64-slice multidetector spiral computed tomography (64- MDCT) contrast-enhanced scanning. Methods: In a retrospective study of 21 cases of hypervascular tumours, the degree of blood supply and indexes were assessed, and the pathological results were used as the diagnostic gold standard to evaluate the sensitivity and specificity of 64-MDCT plain scan and enhanced CT in the diagnosis of oral and maxillofacial soft tissue hypervascular tumours, using the receiver operating characteristic curve to analyse and evaluate the efficacy. Results: Among 21 patients, the diagnostic accuracy of 64-MDCT contrast-enhanced scan was 90.48%, the area under the curve of venous phase CT value was 0.80, the sensitivity was 83.30% and the specificity was 72.73%. Conclusion: 64-MDCT contrast-enhanced scan can be used to evaluate the blood supply of maxillofacial soft tissue hypervascular tumours before an operation. The CT value in the venous phase of tumours has the highest diagnostic effectiveness, which can reduce the risk of blood loss during surgery for maxillofacial hypervascular tumours.
