Abstract Liver injury is a core pathological process in the majority of liver diseases, yet the genetic factors predisposing individuals to its initiation and progression remain poorly understood. Here we show that asialoglycoprotein receptor 1 (ASGR1), a lectin specifically expressed in the liver, is downregulated in patients with liver fibrosis or cirrhosis and male mice with liver injury. ASGR1 deficiency exacerbates while its overexpression mitigates acetaminophen-induced acute and CCl4-induced chronic liver injuries in male mice. Mechanistically, ASGR1 binds to an endoplasmic reticulum stress mediator GP73 and facilitates its lysosomal degradation. ASGR1 depletion increases circulating GP73 levels and promotes the interaction between GP73 and BIP to activate endoplasmic reticulum stress, leading to liver injury. Neutralization of GP73 not only attenuates ASGR1 deficiency-induced liver injuries but also improves survival in mice received a lethal dose of acetaminophen. Collectively, these findings identify ASGR1 as a potential genetic determinant of susceptibility to liver injury and propose it as a therapeutic target for the treatment of liver injury.

Distribution of shrubs expanding in grasslands – shrubification – is ongoing worldwide in grasslands and is common on the Qinghai–Tibetan Plateau (QTP). But the consequences of shrubification for plant carbon (C) input and fate in soil are unclear. We used 13C pulse labelling in a meadow on the QTP to compare photosynthetic capacity and photosynthate distribution in shoots, roots, soil and microbial functional groups between herbaceous plants (herbs) and shrubby Potentilla fruticosa. During 3 h of labeling in 13CO2 atmosphere, the 13C amount assimilated by shrubs (0.81 g C/m−2) was only 38 % of that by herbs. Over 8 days after labeling, 13C amount respired jointly by roots and soil microorganisms under shrubs (0.049 g m−2) was less than half of that under herbs. The mean residence time of 13C for respiration jointly by roots and rhizosphere microorganisms was longer under shrubs (0.61 day) than under herbs (0.44 day). Within 22 days after labelling, 13C amounts in roots, soil, and microorganisms were consistently smaller under shrubs than those under herbs. Consequently, shrub P. fruticosa had not only smaller photosynthetic potential, but also allocated less photosynthate belowground and slowed down C cycling in soil compared with herbs. The distribution of total 13C in microbial functional groups indicated by PLFA analysis was similar between herbs and shrubs. Averaged over vegetation patch types, gram negative bacteria and AMF accounted for 22 % and 4 % of the total microbial PLFAs in the 0–20 cm, respectively, but these two functional groups took up 51 % and 23 % of the total 13C absorbed by microorganisms, respectively. This indicates that gram negative bacteria and AMF are major consumers of rhizodeposits. Concluding, shrubification leads to smaller C allocation belowground and slows down C cycling in the soil.

Abstract Background Though bictegravir/emtricitabine/tenofovir (BIC/FTC/TAF) have been regulatory approved and included in the National Reimbursement Drug List in China, due to the affordability concern, generic version of efavirenz + lamivudine + tenofovir (EFV + 3TC + TDF) is still recommended as the first-line therapy in the clinical guideline and widely used in clinical practice. The aim of the study is to assess the persistence with first-line BIC/TAF/TAF and EFV + 3TC + TDF in newly treated HIV-1 patients in the real-world setting in Hunan Province in China. Methods A retrospective analysis of the medical records of HIV patients initiating first-line antiretroviral therapy in the First Hospital of Changsha in January 1st, 2021-July 31st, 2022 was conducted. Persistence was assessed as the number of days on the therapy from the index until treatment discontinuation or end of data availability. Kaplan-Meier Curves and Cox Proportional Hazard models were used to evaluate the discontinuation rates. Subgroup analysis was performed excluding BIC/FTC/TAF patients with treatment discontinuation due to economic reason, and EFV + 3TC + TDF patients with a viral load > 500,000 copies/mL. Results A total of 310 eligible patients were included in the study, with 244 and 66 patients in the BIC/FTC/TAF group and EFV + 3TC + TDF group, respectively. Compared with EFV + 3TC + TDF patients, BIC/FTC/TAF patients were older, more living in the capital city currently, and had significantly higher total cholesterol and low-density level (all p 500,000 copies/mL, the analysis showed similar results (HR = 10.1, 95% CI = 1.2–84.1). 79.4% of the EFV + 3TC + TDF patients discontinued treatment due to clinical reasons, while 83.3% of the BIC/FTC/TAF patients discontinued treatment due to economic reasons. Conclusions Compared with BIC/FTC/TAF, EFV + TDF + 3TC patients were significantly more likely to discontinue the first-line treatment in Hunan Province in China.

Abstract Background/aims To investigate the clinical situation, treatment methods, and clinical predictors of surgical intervention in children with magnetic foreign bodies in the digestive tract. Materials and methods From January 2019 to June 2022, we retrospectively analyzed the clinical data of 72 children who ingested magnetic foreign bodies inadvertently in our hospital, including their general information, admissions, clinical manifestations, and treatment methods, as well as pertinent literature and statistical data. Following software processing, univariate and multivariate logistic regression analyses were conducted to determine the independent risk factors of this study. Results In this study, 16 patients (22.2%) were discharged smoothly following conservative treatment and 19 patients (26.4%) were cured by gastroscopy. The remaining 37 patients (51.4%) were underwent surgery, in which 26 cases developed gastrointestinal perforation. ABSTRACT Colletotrichum spp. are economically important phytopathogenic fungi that cause anthracnose in a variety of plant species worldwide. Hypovirulence-associated mycoviruses provide new options for the biological control of plant fungal diseases. Here, we found a novel partitivirus from Colletotrichum alienum and named it Colletotrichum alienum partitivirus 1 (CaPV1). CaPV1 contained two dsRNA segments encoding an RNA-dependent RNA polymerase and a capsid protein and was classified under the genus Gammapartitivirus of the family Partitiviridae. CaPV1 significantly decreased host virulence, mycelial growth, appressorial development, and appressorium turgor but increased conidial production with abnormal morphology. In addition, CaPV1 could be successfully transfected into other Colletotrichum species, including C. fructicola, C. spaethianum, and C. gloeosporioides, and caused hypovirulence, indicating the broad application potential of this virus. CaPV1 caused significant transcriptional rewiring of the host fungus C. alienum. Notably, some genes related to vesicle transport in the CaPV1-infected strain were downregulated, consistent with the impaired endocytosis pathway in this fungus. When the Rab gene CaRab7, which is associated with endocytosis in vesicle transport, was knocked out, the virulence of the mutants was reduced. Overall, our findings demonstrated that CaPV1 has the potential to control anthracnose caused by Colletotrichum, and the mechanism by which Colletotrichum induces hypovirulence is caused by affecting vesicle transport.IMPORTANCEColletotrichum is a kind of economically important phytopathogenic fungi that cause anthracnose disease in a variety of plant species worldwide. We found a novel mycovirus of the Gammapartitivirus genus and Partitiviridae family from the phytopathogenic fungus Colletotrichum alienum and named it CaPV1. This study revealed that CaPV1 infection significantly decreased host virulence and fitness by affecting mycelial growth, appressorial development, and appressorium turgor. In addition, CaPV1 could also infect other Colletotrichum species, including C. fructicola, C. spaethianum, and C. gloeosporioides, by viral particle transfection and resulting in hypovirulence of these Colletotrichum species. Transcriptomic analysis showed that CaPV1 caused significant transcriptional rewiring of the host fungus C. alienum, especially the genes involved in vesicle transport. Moreover, endocytosis and gene knockout assays demonstrated that the mechanism underlying CaPV1-induced hypovirulence is, at least in part, caused by affecting the vesicle transport of the host fungus. This study provided insights into the mechanisms underlying the pathogenesis of Colletotrichum species and mycovirus-fungus interactions, linking the role of mycovirus and fungus vesicle transport systems in shaping fungal pathogenicity.

In order to solve the problem that the high-voltage DC power supply system of the communication base station of the intelligent fire protection system in the open-pit coal mine area has the risk of personal electric shock when the insulation of any branch decreases, an on-line insulation protection of the high-voltage DC power supply system cable based on the signal injection method is proposed. We discuss the principle and method of HVDC power supply cable insulation on-line detection technology, and design a leakage fault detection system for HVDC power supply system. The leakage simulation experiment of HVDC power supply system cable is carried out through eight lines of different lengths and different degrees of insulation drop successively. The research results show that when the leakage of eight power cables of different lengths occurs in the HVDC power supply system, the on-line insulation detection system can accurately select the line of leakage fault and measure the degree of insulation decline, and the error is within the range of ±5%. The leakage protection device provides a guarantee for personal electric shock safety.

The design or simulation analysis of special equipment products must follow the national standards, and hence it may be necessary to repeatedly consult the contents of the standards in the design process. However, it is difficult for the traditional question answering system based on keyword retrieval to give accurate answers to technical questions. Therefore, we use natural language processing techniques to design a question answering system for the decision-making process in pressure vessel design. To solve the problem of insufficient training data for the technology question answering system, we propose a method to generate questions according to a declarative sentence from several different dimensions so that multiple question-answer pairs can be obtained from a declarative sentence. In addition, we designed an interactive attention model based on a bidirectional long short-term memory (BiLSTM) network to improve the performance of the similarity comparison of two question sentences. Finally, the performance of the question answering system was tested on public and technical domain datasets.

BackgroundEarly gastric cancer (EGC) has a high survival rate, but it is difficult to diagnosis. Recently, artificial intelligence (AI) based on deep convolutional neural network (DCNN) has made significant progress in the field of gastroenterology. The purpose of this study was to establish a DCNN assist system to improve the detection of EGC.Methods3400 EGC and 8600 benign images were collected to train the DCNN to detect EGC. Subsequently, its diagnostic ability was compared to that of endoscopists using an independent internal test set (ITS, including 1289 images) and an external test set (ETS, including 542 images) come from three digestive center.ResultsThe diagnostic time of DCNN and endoscopists were 0.028s, 8.05 ± 0.21s, 7.69 ± 0.25s in ITS, and 0.028s, 7.98 ± 0.19s, 7.50 ± 0.23s in ETS, respectively. In ITS, the diagnostic sensitivity and accuracy of DCNN are 88.08%(95% confidence interval,95%CI,85.24%-90.44%), 88.60% (95%CI,86.74%-90.22%), respectively. In ETS, the diagnostic sensitivity and accuracy are 92.08% (95%CI, 87.91%- 94.94%),92.07%(95%CI, 89.46%-94.08%),respectively. DCNN outperformed all endoscopists in ETS, and had a significantly higher sensitivity than the junior endoscopists(JE)(by18.54% (95%CI, 15.64%-21.84%) in ITS, also higher than JE (by21.67%,95%CI, 16.90%-27.32%) and senior endoscopists (SE) (by2.08%, 95%CI, 0.75%-4.92%)in ETS. The accuracy of DCNN model was higher (by10.47%,95%CI, 8.91%-12.27%) than that of JE in ITS, and also higher (by14.58%,95%CI, 11.84%-17.81%; by 1.94%,95%CI,1.25%-2.96%, respectively) than JE and SE in ETS.ConclusionThe DCNN can detected more EGC images in a shorter time than the endoscopists. It will become an effective tool to assist in the detection of EGC in the near future.

Alternaria dianthicola is a pathogenic fungus that causes serious leaf or flower blight on some medicinal plants worldwide. In this study, multiple dsRNA bands in the range of 1.2-10 kbp were found in a Alternaria dianthus strain HNSZ-1, and eleven full-length cDNA sequences of these dsRNA were obtained by high-throughput sequencing, RT-PCR detection and conventional Sanger sequencing. Homology search and phylogenetic analyses indicated that the strain HNSZ-1 was infected by at least nine mycoviruses. Among the nine, five viruses were confirmed to represent novel viruses in the families Hypoviridae, Totiviridae, Mymonaviridae and a provisional family Ambiguiviridae. Virus elimination and horizontal transmission indicated that the (-) ssRNA virus, AdNSRV1, might be associated with the slow growth and irregular colony phenotype of the host fungus. As far as we know, this is the first report for virome characterization of A. dianthus, which might provide important insights for screening of mycovirus for biological control and for studying of the interactions between viruses or viruses and their host.

Currently, the practical application of SiOx still has a huge hindrance in the area of lithium ion battery, because it is unable to achieve an effective contact with surrounding conducting materials, resulting in failure to form lithium ion migration tunnels. In this work, we presented a facile method to synthesize the B-doped SiOx composite by adhering SiOx particles with MWCNT (multi-walled carbon nanotube) under the assistance of lithium metaborate (LiBO2). LiBO2, as a sintering aid, not only can react with SiOx to form a compacted framework, but also build a three-dimensional (3D) conductive network for ions transportation. Furthermore, B-SiOx@CNT@LBO anode delivers a remarkable lithium storage performance in terms of long cycles and high rate capability. A full cell coupled with NCM622 cathode achieves a high energy density of 429.5 Wh kg−1 based on the total mass of cathode.

In this paper, the numerical simulation of supercritical carbon dioxide (S−CO2) flowing in large diameter inclined circular tubes with different heat flux and inlet temperature is studied. According to the experimental results of S−CO2 flow in a horizontal tube, the SSTk−ω model was found to be the most accurate predictor of heat transfer. Based on the phase transition temperature of the pseudo-boiling phenomenon under subcritical conditions, a gas-like film radius model was established. The results show that when heat flux is too large, the wall temperature rises quickly, which makes the density gradient in the cross-section, buoyancy and gas-like film radius increase constantly, resulting in the gathering of a large number of low-density and poor heat transfer performances at the top generatrix, causing a sudden increase in wall temperature and the heat transfer coefficient decrease significantly. As the inlet temperature increases, buoyancy and relative secondary flow energy decreases and the fluid temperature rises dramatically, eventually leading to a rapid decrease in thermal conductivity and heat transfer capacity.

Our current research aims to examine whether protocatechuic acid (PCA) can be used as a therapeutic agent for the development of cerebral aneurysm (CA) and to elucidate the mechanisms behind this. We assessed the effects of PCA at 50 and 100 mg/kg on the activation of signaling pathways for tissue necrosis factor (TNF)-α/nuclear factor (NF)-κB/nuclear factor erythroid 2 (Nrf-2) on progression and development in an elastase-induced CA model, accompanied by a high-salt diet to induce hypertension. The expression of inflammatory cytokines, chemokines, tumor necrosis factor-α, interleukins (IL)-8, IL-17, IL-6, IL-1β, and matrix metalloproteinase (MMP)-2 and MMP-9 was analyzed by ELISA, western blot, and reverse transcriptase quantative polymerase chain reaction. The expression levels of antioxidant enzymes and translocation of Nrf-2 were also determined. The group treated with PCA demonstrated a significant (P < 0.05) decrease in the aneurysmal size in rats compared to the CA-induced group. We found that PCA treatment suppressed the invasion of macrophage and activation of TNF-α/NF-κB/Nrf-2 signaling pathways. There was a significant decrease (P < 0.05) in pro-inflammatory cytokine and chemokine levels in a dose-dependent manner. We found that PCA treatment exerts protective effects by suppressing the development and progression of CA through the inhibition of inflammatory responses in macrophages via TNF-α/NF-κB/Nrf-2 signaling pathways, thus demonstrating that PCA can act as a treatment for CA.

[Introduction] The paper aims to sort out the contents of the design of solar field of the solar tower plant systematically and to evaluate the impact of solar field design on the economy of the plant. The solar field design is summarized and introduced. The factors affecting solar field efficiency is analyzed and the economic research is conducted. [Method] The design of the collector field was divided into three parts: design of the position and height of the receiver, design of the heliostat layout and design of the road in solar field. The method of decomposing the sub efficiencies of solar field efficiency was adopted and the research was focused on the two main design factors that affect the economy of the solar field. [Result] In this paper, several factors that need to be considered in the design of solar field are summarized and the design conclusions of various factors to improve the efficiency of solar field are listed. Under the current engineering conditions, the larger solar field results in a better the economy of the plant. The influence of the density of the heliostat layout on the economy is obtained. [Conclusion] The conclusions are of great guiding significance and can be used for reference in the design of solar tower plant.

We discuss the modes B→K(⁎)νν¯ in the context of non-standard neutrino interactions that add incoherently to the SM rates. We consider two scenarios: an additional light neutrino; and neutrino lepton flavour violation. We find that an additional light neutrino that interacts with SM fields via a non-universal Z′ can increase RK(⁎)ν by up to a factor of two without conflicting with Bs−B¯s mixing. This model then predicts rates for Bs→τ+τ− up to six times larger than the SM. In the context of neutrino lepton flavour violation mediated by leptoquarks we find that the current experimental upper bounds on RK(⁎)ν are already more constraining than direct bounds from Bs→τℓ and B→K(⁎)τℓ modes for ℓ=e,μ.

Objective The aim of this study was to compare the treatment effects of high-intensity focused ultrasound (HIFU) and laparoscopic excision (LE) in patients with adenomyosis and infertility. Materials and methods A total of 93 patients with adenomyosis and infertility who were treated with HIFU (50 patients) or LE (43 patients) from January 2012 to January 2017 at the Third Xiangya Hospital of Central South University were retrospectively analyzed. Clinical characteristics including dysmenorrhea severity pain score, menorrhagia severity scores, reproductive outcomes, complications during pregnancy and delivery, adverse effects, surgical complications, and other clinical variables were compared between the HIFU and LE groups. Results Of the total 93 patients with adenomyosis and infertility, 50 were treated with HIFU and 43 underwent LE. Both HIFU and LE treatments achieved significant relief of dysmenorrhea and menorrhagia. The total hospital stay was shorter in patients treated with HIFU than in those who underwent LE surgery. Neither HIFU nor LE treatment led to severe complications after treatment. Most importantly, patients treated with HIFU showed significantly higher pregnancy rates and natural conception rates than those who underwent LE surgery. Notably, in the HIFU treatment group, those with diffuse adenomyotic lesions had significantly lower postoperative pregnancy rates than those with focal adenomyosis. Conclusion HIFU showed a safe and effective profile as a therapeutic management option for patients with adenomyosis. In comparison with LE, HIFU treatment achieved better postoperative reproductive outcomes. HIFU treatment should be encouraged and implemented in clinical practice.

Ethylene-Vinyl Acetate (EVA) polymer foam was prepared using a mold for this investigation. The effect of the cell structure parameters and pore filling gas type on the sound absorption performance of the EVA foam materials were analyzed in terms of their sound absorption capability. The results show that the cell size, cell wall thickness and pore filling gas type have significant influence on the sound absorption performance. When the cell size is 71.3 μm, at low frequency (1000 Hz) the absorption coefficient was the largest at 0.487 and the absorption effect was good. With the increase of the cell wall thickness, the peak value of the sound absorption coefficient first increased and then decreased, and gradually transferred to the low frequency area. When the cell wall thicknesses were 14.3 μm, the foam material had optimal sound absorption properties. The cell filling gas was hydrogen, the EVA foam material had optimal sound absorption effect, and the sound absorption peak value was 0.553 at 800 Hz in the low frequency range (Ⅰ), the sound absorption performance of the foamed materials was ideal.

Here, we report a novel double-stranded RNA virus designated Colletotrichum liriopes partitivirus 1 (ClPV1) from the plant pathogenic fungus C. liriopes. ClPV1 genome has two double stranded RNAs (dsRNAs), named as dsRNA 1 and dsRNA 2, which in the lengths of 1,807 and 1,706 bp, respectively. The dsRNA 1 and dsRNA 2 encoded proteins showing significant amino acid (aa) sequence identity to the RNA-dependent RNA polymerase (RdRp) and coat protein (CP) of partitiviruses, respectively. Phylogenetic analysis using the aa sequences of RdRp and CP indicated that ClPV1 was grouped to members of the putative Epsilonpartitivirus genus in the Partitiviridae family. Spherical viral particles in approximately 35 nm in diameter and packaging the ClPV1 genome were isolated. Virus elimination and virus transfection with purified viral particles, and biological comparison revealed that ClPV1 could reduce the virulence and conidia production of C. liriopes. To the best of our knowledge, this is the first report of mycovirus in C. liriopes fungus.

Patchiness is an important feature in grassland ecosystems, but the distribution pattern of plant species in small patches on the Qinghai-Tibetan Plateau (QTP) is largely unknown. We questioned: (1) whether the power law can be applied to determine the spatial pattern of plant species in small patches; and (2) how species frequency (dominant, companion and rare) affects the spatial heterogeneity of small patches on the QTP? The power law was used to examine the spatial pattern of plant species and examine the relationship between the plant species presence frequency and spatial pattern in three types of small patches (Cyperaceae, Polygonaceae and Compositae), each with three spatial extents according to the size of the area (scale 1: 0.6–0.9 m2, scale 2: 3.0–3.8 m2 and scale 3: 6.5–8.8 m2) on the QTP. Plant species in patches showed an aggregated distribution and the spatial heterogeneity index differed among species. The spatial heterogeneities of Cyperaceae and Polygonaceae patches were influenced strongly by dominant and companion plant species, while the Compositae patch was influenced by dominant and rare species. The Compositae patches exhibited a lower species diversity and higher spatial heterogeneity index than the other patches, and the extensive distribution of Compositae community can be linked to degradation processes in the alpine meadow. We concluded that the spatial pattern of plant species and the heterogeneity index for small patches could be determined by the species presence frequency on the QTP. Abstract Potential intensity (PI), an upper thermodynamic limit of tropical cyclone (TC) intensity, is a useful index in TC statistical forecasting. This study evaluated the relationship between the current intensity of TCs and four PI indexes calculated with different sea surface indicators over the Western North Pacific (WNP) from 2009 to 2016. To mirror the degree of sea cooling caused by TCs, three sea surface temperature (SST) indicators (pre‐SST, 80 m depth mean sea temperature, and dynamic mixed mean sea temperature) were compared with real‐time SST obtained from HYbrid Coordinate Ocean Model (HYCOM) data. The results showed that four versions of PI all overestimated the current intensity of developing TCs. In contrast, when a TC at its strongest or in its weakening stages in a favorable atmospheric environment, pre‐sea conditions were strongly correlated with TC intensity. For strong TCs, the ocean‐coupled PI index was best able to describe TC intensity.
