The global coronavirus disease 2019 (COVID-19) pandemic caused by the severe acute respiratory syndrome coronavirus 2 (SARS-CoV-2) has become the greatest worldwide public health threat of this century, which may predispose multi-organ failure (especially the lung) and death despite numerous mild and moderate symptoms. Recent studies have unraveled the molecular and clinical characteristics of the infectivity, pathogenicity, and immune evasion of SARS-CoV-2 and thus improved the development of many different therapeutic strategies to combat COVID-19, including treatment and prevention. Previous studies have indicated that nitric oxide (NO) is an antimicrobial and anti-inflammatory molecule with key roles in pulmonary vascular function in the context of viral infections and other pulmonary disease states. This review summarized the recent advances of the pathogenesis of SARS-CoV-2, and accordingly elaborated on the potential application of NO in the management of patients with COVID-19 through antiviral activities and anti-inflammatory properties, which mitigate the propagation of this disease. Despite the increasing impact of atmospheric nitrogen (N) deposition on terrestrial greenhouse gas (GHG) budget, through driving both the net atmospheric CO2 exchange and the emission or uptake of non‐CO2 GHGs (CH4 and N2O), few studies have assessed the climatic impact of forests and grasslands under N deposition globally based on different bottom‐up approaches. Here, we quantify the effects of N deposition on biomass C increment, soil organic C (SOC), CH4 and N2O fluxes and, ultimately, the net ecosystem GHG balance of forests and grasslands using a global comprehensive dataset. We showed that N addition significantly increased plant C uptake (net primary production) in forests and grasslands, to a larger extent for the aboveground C (aboveground net primary production), whereas it only caused a small or insignificant enhancement of SOC pool in both upland systems. Nitrogen addition had no significant effect on soil heterotrophic respiration (RH) in both forests and grasslands, while a significant N‐induced increase in soil CO2 fluxes (RS, soil respiration) was observed in grasslands. Nitrogen addition significantly stimulated soil N2O fluxes in forests (76%), to a larger extent in grasslands (87%), but showed a consistent trend to decrease soil uptake of CH4, suggesting a declined sink capacity of forests and grasslands for atmospheric CH4 under N enrichment. Overall, the net GHG balance estimated by the net ecosystem production‐based method (forest, 1.28 Pg CO2‐eq year−1 vs. grassland, 0.58 Pg CO2‐eq year−1) was greater than those estimated using the SOC‐based method (forest, 0.32 Pg CO2‐eq year−1 vs. grassland, 0.18 Pg CO2‐eq year−1) caused by N addition. Our findings revealed that the enhanced soil C sequestration by N addition in global forests and grasslands could be only marginally offset (1.5%–4.8%) by the combined effects of its stimulation of N2O emissions together with the reduced soil uptake of CH4. The pandemic due to Severe Acute Respiratory Syndrome Coronavirus 2 (SARS-CoV-2) has emerged as a serious global public health issue. Besides the high transmission rate from individual to individual, indirect transmission from inanimate objects or surfaces poses a more significant threat. Since the start of the outbreak, the importance of respiratory protection, social distancing, and chemical disinfection to prevent the spread of the virus has been the prime focus for infection control. Health regulatory organizations have produced guidelines for the formulation and application of chemical disinfectants to manufacturing industries and the public. On the other hand, extensive literature on the virucidal efficacy testing of microbicides for SARS-CoV-2 has been published over the past year and a half. This review summarizes the studies on the most common chemical disinfectants and their virucidal efficacy against SARS-CoV-2, including the type and concentration of the chemical disinfectant, the formulation, the presence of excipients, the exposure time, and other critical factors that determine the effectiveness of chemical disinfectants. Modern human activity is profoundly changing our relationship with microorganisms with the startling rise in the rate of emerging infectious diseases. Nipah virus together with Ebola virus and SARS-CoV-2 are prominent examples. Since COVID-19 and the West African Ebola virus disease outbreak, different chemical disinfectants have been developed for preventing the direct spread of viruses and their efficacy has also been evaluated. However, there are currently no published efficacy studies for the chemical disinfection of Nipah virus. In this study, the virucidal efficacy of three disinfectants (Micro-Chem Plus detergent disinfectant cleaner, FWD and Medical EtOH) against Nipah virus was evaluated in quantitative suspension tests including. Our results showed that the > 4 log reduction achieved for all products in inactivating Nipah virus in 15 s. Even, 19% ethanol was able to inactivate Nipah virus when applied for at least 8 min contact time. Comparative analysis displayed virucidal efficacy of each of the evaluated disinfectants against SARS-CoV-2, Ebola virus and Nipah virus, with only minor differences in working concentrations and contact times required for complete inactivation. We expect that our study can assist in decontamination in healthcare settings and high level biosafety laboratories and can be beneficial to control for emerging enveloped viruses. How to achieve high soil organic carbon (SOC) sequestration and low greenhouse gases emissions but without compromising yields is the challenge of rice cultivation in China. This study explored the potential to simultaneously increase SOC stocks and reduce methane (CH 4) emissions by optimizing rice cropping rotation and fertilization based on a 2-year field experiment in southeast China. We tested five optimized N management practices including directly reducing chemical N fertilizer input and replacing chemical N fertilizers with organic and controlled-release fertilizers in double-rice cropping and Chinese milk vetch-rice cropping rotation systems. Results showed higher seasonal rice yields, nitrogen use efficiency (NUE) and SOC stocks for Chinese milk vetch-rice rotation systems (CR) as compared to double-rice cropping systems (DR). Seasonal rice yields were the highest in the 2019 single-rice season (2019-SR), followed by the 2018 late (2018-LR) and early rice seasons (2018-ER). SOC stocks of CR for the treatment receiving organic N fertilizer (OF) were the largest among all rice seasons and fertilization treatments. Seasonal CH 4 emissions were 8–12% greater in the OF than in the chemical N fertilizer treatments across all rice seasons, with the greatest in 2019-SR and lowest in 2018-ER. Fertilization stimulated CH 4 emissions through enhancing mcrA gene activity, but regulated by soil properties, while rice rotation models indirectly influenced CH 4 emissions through direct effects on soil properties. However, yield-scaled CH 4 emissions remained unaffected by N fertilization in the two-year rice rotation systems. The NUE was significantly enhanced in OF than in other fertilizer treatments over all seasons. Seasonal net ecosystem C budget (NECB) was 24–41% greater in OF than in chemical N fertilizer treatments across all rice seasons, with the greatest in 2019-SR and lowest in 2018-LR. The NECB was increased by 46% in CR as compared to DR. Our results revealed that the optimized rice cropping rotation by incorporation of Chinese milk vetch with organic N fertilization is beneficial for enhancing soil C sequestration and maintaining rice yields in southeast China. Convolutional neural networks (CNNs) have shown great performance in computer vision tasks, from image classification to pattern recognition. However, CNNs′${\rm CNNs}^{\prime }$ superior performance arises at the expense of high computational costs, which restricts their employment in real‐time decision‐making applications. Computationally intensive convolutions can be offloaded to optical metasurfaces, enabling sub‐picosecond latency and nearly zero energy consumption, but the currently reported approaches require additional bulk optics and can only process polarized light, which limits their practical usages in integrated lightweight systems. To solve these challenges, a novel design of the metasurface‐based optical convolutional accelerator is experimentally demonstrated, offering an ultra‐compact volume of 0.016 mm3${\rm mm}^{3}$, a low cross‐talk of ‐20 dB, polarization insensitivity, and is capable of implementing multiple convolution operations and extracting simultaneously various features from light‐encoded images. The ultra‐compact metasurface‐based optical accelerator can be compactly integrated with a digital imaging system to constitute an optical‐electronic hybrid CNN, which experimentally achieves a consistent accuracy of 96 % in arbitrarily polarized MNIST digits classification. The proposed ultra‐compact metasurface‐based optical convolutional accelerator paves the way for power‐efficient edge‐computing platforms for a range of machine vision applications. Rice paddies and rivers draining agricultural watersheds have been documented as two major sources of atmospheric methane (CH 4), while parallel flux measurements have been rarely taken on CH 4 from rice paddies against the drainage rivers in agricultural watersheds. Moreover, the drainage events during the rice growing season deliver sufficient organic and inorganic nutrients to rivers, which makes there an increasing concerned source of atmospheric CH 4. Here, we compared CH 4 emissions from rice paddies and the associated rivers draining agricultural watersheds by a parallel field experiment in southeast China. Over the whole rice–growing season (June–November), CH 4 fluxes from drainage rivers averaged 25 mg m–2 h–1, which were 82 % higher than those from rice paddies (13.7 mg m–2 h–1). Besides the dependence of riverine CH 4 fluxes on water dissolved oxygen (DO), both ecosystem CH 4 fluxes were significantly correlated with water/soil dissolved organic carbon (DOC), oxidation-reduction potential (Eh) and the microbial functional genes that drive CH 4 production or uptake [methanogens (mcrA and methanogenic archaeal 16S rRNA) and methanotrophs (pmoA)]. Our results highlighted that the CH 4 mitigation potential benefited from the drainage practice in rice paddies might have been totally offset by its induced increases in CH 4 emissions from rivers draining agricultural watersheds at the regional scale. ● Rivers draining agricultural watersheds are an important source of atmospheric CH 4. ● CH 4 fluxes from drainage rivers have been rarely related to the abundance of microbes. ● CH 4 fluxes from drainage rivers were 82 % greater than those from rice paddies. ● The increase in CH 4 emissions in rivers may offset the mitigation benefit of CH 4 in rice paddies. Porcine reproductive and respiratory syndrome virus (PRRSV) is one of the most economically important viruses affecting the swine industry worldwide. MicroRNAs (miRNAs) play vital roles in virus-host interactions by regulating the expression of viral or host gene at posttranscriptional level. Our previous research showed that PRRSV infection down-regulates the expression of heme oxygenase-1 (HO-1), a pivotal cytoprotective enzyme, and overexpression of HO-1 inhibits PRRSV replication. In this study, we demonstrate that host miRNA miR-22 can downregulate HO-1 expression by directly targeting its 3′ untranslated region. Suppression of HO-1 expression by miR-22 facilitates PRRSV replication. This work suggests that PRRSV may utilize cellular miRNA to modify antiviral host factor expression, enabling viral replication, which not only provides new insights into virus-host interactions during PRRSV infection, but also suggests potential therapies for PRRSV infection. Virus replication depends upon host-cell processes in infected cells, and this is true for porcine reproductive and respiratory syndrome virus (PRRSV), the causative agent of PRRS that is a worldwide threat to the swine industry. Heme oxygenase-1 (HO-1) is a ubiquitously expressed inducible isoform of the first and rate-limiting enzyme for heme degradation. Our previous research suggested that HO-1 may play an important role in PRRSV infection. However, the function of HO-1 in PRRSV infection is unclear. In the present study, Marc-145, PK-15 CD163 cell lines and porcine alveolar macrophages (PAMs) were used to evaluate the effects of HO-1 induction and over-expression on the replication of two different PRRSV strains. Induction of HO-1 markedly decreased the replication of PRRSV strains in the different cells. Similarly, adenoviral-mediated over-expression of HO-1 also greatly decreased the replication of PRRSV. In contrast, ablation of HO-1 using small interfering RNA concomitantly increased PRRSV replication. Therefore, the data were consistent with HO-1 acting as an antiviral factor and these findings suggested that over-expression or induction of HO-1 may provide a potential therapeutic strategy against PRRSV infection. Background: There was a large scale outbreak of the highly pathogenic porcine reproductive and respiratory syndrome (PRRS) in China and Vietnam during 2006 and 2007 that resulted in unusually high morbidity and mortality among pigs of all ages. The mechanisms underlying the molecular pathogenesis of the highly virulent PRRS virus (H-PRRSV) remains unknown. Therefore, the relationship between pulmonary gene expression profiles after H-PRRSV infection and infection pathology were analyzed in this study using high-throughput deep sequencing and histopathology. Results: H-PRRSV infection resulted in severe lung pathology. The results indicate that aberrant host innate immune responses to H-PRRSV and induction of an anti-apoptotic state could be responsible for the aggressive replication and dissemination of H-PRRSV. Prolific rapid replication of H-PRRSV could have triggered aberrant sustained expression of pro-inflammatory cytokines and chemokines leading to a markedly robust inflammatory response compounded by significant cell death and increased oxidative damage. The end result was severe tissue damage and high pathogenicity. Conclusions: The systems analysis utilized in this study provides a comprehensive basis for better understanding the pathogenesis of H-PRRSV. Given that ebolavirus causes severe and frequently lethal disease, its rapid and accurate detection using available and validated methods is essential for controlling infection. Real-time reverse-transcription PCR (RT-PCR) has proven to be an invaluable tool for ebolaviruses diagnostics. Many assays with different targets have been developed, but they have not been externally compared or validated, and limits of detection are not uniformly reported. Here we compared and evaluated the sensitivity, reproducibility and specificity of 23 in-house assays under the same conditions. Our results showed that these assays were highly gene- and species- specific when evaluated using in vitro RNA transcripts and viral RNA, and the potential limits of detection were uniformly reported ranging from 102 to 106 in vitro synthesized RNA transcripts copies perμL and 1–100 TCID50/mL. The comparison of these assays indicated that those targeting the more conservative NP gene could be the better option for EVD case definition and quantitative measurement because of its higher sensitivity for the same species. Our analysis could contribute to the standardization of ebolavirus detection and quantification assays, which can offer a better understanding of the meaning of results across laboratories and time points, as well as make them easy to implement, especially under outbreak conditions. Inland waters (rivers, reservoirs, lakes, ponds, streams) and estuaries are significant emitters of methane (CH4) and nitrous oxide (N2O) to the atmosphere, while global estimates of these emissions have been hampered due to the lack of a worldwide comprehensive data set of CH4 and N2O flux components. Here, we synthesize 2997 in‐situ flux or concentration measurements of CH4 and N2O from 277 peer‐reviewed publications to estimate global CH4 and N2O emissions from inland waters and estuaries. Inland waters including rivers, reservoirs, lakes, and streams together release 95.18 Tg CH4 year−1 (ebullition plus diffusion) and 1.48 Tg N2O year−1 (diffusion) to the atmosphere, yielding an overall CO2‐equivalent emission total of 3.06 Pg CO2 year−1. The estimate of CH4 and N2O emissions represents roughly 60% of CO2 emissions (5.13 Pg CO2 year−1) from these four inland aquatic systems, among which lakes act as the largest emitter for both CH4 and N2O. Ebullition showed as a dominant flux component of CH4, contributing up to 62%–84% of total CH4 fluxes across all inland waters. Chamber‐derived CH4 emission rates are significantly greater than those determined by diffusion model‐based methods for commonly capturing of both diffusive and ebullitive fluxes. Water dissolved oxygen (DO) showed as a dominant factor among all variables to influence both CH4 (diffusive and ebullitive) and N2O fluxes from inland waters. Our study reveals a major oversight in regional and global CH4 budgets from inland waters, caused by neglecting the dominant role of ebullition pathways in those emissions. The estimated indirect N2O EF5 values suggest that a downward refinement is required in current IPCC default EF5 values for inland waters and estuaries. Our findings further indicate that a comprehensive understanding of the magnitude and patterns of CH4 and N2O emissions from inland waters and estuaries is essential in defining the way of how these aquatic systems will shape our climate. Interferon-gamma (IFN-γ) is a critical indicator of the immune response to many infections and diseases. Identifying excellent IFN-γ recognition molecules is of great significance for the early diagnosis and treatment of diseases. Herein, anti-IFN-γ aptamers were selected based on capillary electrophoresis-systematic evolution of ligands by exponential enrichment (CE-SELEX) and their affinities were characterized by the biolayer interferometry (BLI) assay. A rational molecular docking strategy was utilized to predict precisely the binding behavior between aptamer and IFN-γ and remove redundant bases of original sequence. Therefore, aptamer IFNG-8 with the highest affinity can be efficiently identified within only three rounds and the truncated aptamer IFNG-8 T with comparable affinity is eventually obtained. With IFNG-8 T as the detection probe, a fiber-optic evanescent wave aptasensor is constructed for the detection of IFN-γ, which shows a wide linear range from 0.1 to 10000 pM and a low limit of detection (LOD) of 0.0718 pM. The developed aptasensor exhibits high selectivity and can be utilized for IFN-γ detection in human serum samples with good recoveries. In summary, the truncated aptamer can serve as a novel optional transducing element and the developed aptasensor provides an innovative methodology for detection of IFN-γ in trace amount. Microring resonators, due to their ability to enable robust strong light–matter interactions within their structures, have garnered substantial interest for their utility in sensing applications, particularly in the realm of gas detection. However, there is an inherent trade-off between a microring resonator's quality factor and confinement factor in the air, making it difficult to balance them. Here, we demonstrate a novel solution with a suspended nanomembrane silicon (SNS) microring resonator. This resonator has ultrathin sub-wavelength thicknesses (0.02–0.03λ), which breaks the trade-off, offering not only a high intrinsic quality factor of 6 × 105 but also an extraordinarily large confinement factor of ∼80% in the air at mid-infrared wavelengths. As a proof-of-concept demonstration, we applied the SNS microring resonator for CO2 gas sensing, exhibiting a sensitivity over 10 times higher than conventional silicon resonators and a large dynamic sensing range spanning from 0% to 100% with a high resolution of better than 4% and chemical specificity. By virtue of its excellent properties, the SNS microring resonator has the potential to open new possibilities for the development of unprecedented nanophotonic integrated circuits, with a broad range of applications in on-chip sensing scenarios. Gaseous reactive nitrogen (Nr) emissions from agricultural soils to the atmosphere constitute an integral part of global N cycle, directly or indirectly causing climate change impacts. The extensive use of N fertilizer in crop production will compromise our efforts to reduce agricultural Nr emissions in China. A national inventory of fertilizer N‐induced gaseous Nr emissions from croplands in China remains to be developed to reveal its role in shaping climate change. Here we present a data‐driven estimate of fertilizer N‐induced soil Nr emissions based on regional and crop‐specific emission factors (EFs) compiled from 379 manipulative studies. In China, agricultural soil Nr emissions from the use of synthetic N fertilizer and manure in 2018 are estimated to be 3.81 and 0.73 Tg N yr−1, with a combined contribution of 23%, 20% and 15% to the global agricultural emission total of ammonia (NH3), nitrous oxide (N2O) and nitric oxide (NO), respectively. Over the past three decades, NH3 volatilization from croplands has experienced a shift from a rapid increase to a decline trend, whereas N2O and NO emissions always maintain a strong growth momentum due to a robust and continuous rise of EFs. Regionally, croplands in Central south (1.51 Tg N yr−1) and East (0.99 Tg N yr−1) of China exhibit as hotspots of soil Nr emissions. In terms of crop‐specific emissions, rice, maize and vegetable show as three leading Nr emitters, together accounting for 61% of synthetic N fertilizer‐induced Nr emissions from croplands. The global warming effect derived from cropland N2O emissions in China was found to dominate over the local cooling effects of NH3 and NO emissions. Our established regional and crop‐specific EFs for gaseous Nr forms provide a new benchmark for constraining the IPCC Tier 1 default EF values. The spatio‐temporal insight into soil Nr emission data from N fertilizer application in our estimate is expected to advance our efforts towards more accurate global or regional cropland Nr emission inventories and effective mitigation strategies. Abstract Microcystin-LR (MC-LR), a toxin produced by cyanobacteria, is very toxic and poses a threat to public health when entering water treatment works. In this study, UV/chlorine process, as an advanced oxidation process (AOP), has been demonstrated for effective elimination of MC-LR levels and associated toxicity. At a chlorine dose of 3.0 mg L−1 and UV fluence of 125 mJ cm−2, MC-LR (initial concentration 1.0 μM) was reduced by 92.5%, which was much higher than 20.3% removal under UV irradiation alone and 65.1% removal during dark chlorination. Enhanced degradation was attributed by hydroxyl radicals (HO) and reactive chlorine species (RCS), mainly Cl 2 - and ClO. Increasing chlorine doses or lowering pH favored MC-LR removal. Increased bicarbonate and natural organic matter concentrations inhibited MC-LR removal, but bromide ions enhanced MC-LR removal instead. MC-LR elimination rates in natural waters were roughly two times smaller than those in ultrapure water. The reactive radicals promoted hydroxylation of both diene of Adda moiety and double bond of Mdha moiety in MC-LR. UV exposure enhanced the dechlorination of chloro-MC-LR via the cleavage of C Cl bond. The toxicity was evaluated by a protein phosphatase (PP2A) inhibition assay. At a chlorine dose of 3.0 mg L−1 and UV fluence of 125 mJ cm−2, the toxicity of the treated water was reduced by 75.0%, which was also higher than 25.7% and 46.7% removal under UV irradiation alone and during dark chlorination, respectively. These results highlight UV/chlorine is an efficient AOP for MC-LR degradation and detoxification. Rice paddies and rice-aquaculture co-culture systems are important sources of methane (CH 4) and nitrous oxide (N 2 O) to the atmosphere. Due to the growing human demand of aquaculture protein and the rapid development of green agriculture, rice-aquaculture co-culture systems have been increasingly developed from rice paddies in southeast China. However, the strength of CH 4 and N 2 O emissions from rice-aquaculture co-culture systems remains poorly quantified, in particular the contribution of these emissions from different functional cultivation areas. Here, a two-year parallel field experiment was conducted to examine the changes in CH 4 and N 2 O fluxes following the conversion from rice paddies to rice-crayfish co-culture systems in southeast China. Over the two-year period, annual CH 4 and N 2 O fluxes from rice-crayfish co-culture systems averaged 4.59 mg m-2 h-1 and 39.50 μg m -2 h -1, amounting to 391.9 kg ha−1 and 3.46 kg ha−1, respectively. The conversion from rice paddies to rice-crayfish co-culture systems significantly reduced CH 4 and N 2 O emissions by 14% and 31%, respectively. The emission factors of N 2 O were estimated to be 1.25–1.61% in the rice-crayfish co-culture system. Relative to rice planted areas, CH 4 and N 2 O fluxes from crayfish ditched culture areas were three times higher and 50% lower, which contributed 26% and 5% to the total CH 4 and N 2 O emissions from rice-crayfish co-culture systems, respectively. Our results highlight that the conversion of paddy rice monoculture to rice-crayfish co-culture systems can benefit low greenhouse gas emissions and high ecosystem economic profits as a potential option to sustain agricultural development and achieve carbon neutrality. Porcine reproductive and respiratory syndrome (PRRS) is a leading disease in pig industry worldwide and can result in serious economic losses each year. The PRRS epidemic situation in China has been very complicated since the unprecedented large-scale highly pathogenic PRRS (HP-PRRS) outbreaks in 2006. And now the HP-PRRS virus (HP-PRRSV) and classical North American type PRRSV strains have coexisted in China. Rapid differential detection of the two strains of PRRSV is very important for effective PRRS control. The real-time RT-PCR for simultaneous detection and differentiation of HP-PRRSV and PRRSV by using both SYBR Green and TaqMan probes was developed and validated. Both assays can be used for rapid detection and strain-specific identification of HP-PRRSV and PRRSV. However, the TaqMan probe method had the highest detection rate whereas the conventional RT-PCR was the lowest. The real-time RT-PCR developed based on SYBR Green and TaqMan probe could be used for simultaneous detection and differentiation of HP-PRRSV and PRRSV in China, which will benefit much the PRRS control and research. Rotavirus (RV) is a non–enveloped icosahedral virus with an 11–segment double–stranded RNA genome, belonging to the family of rotaviruses. RV is one of the pathogens causing diarrhea in infants and young animals, and it induces the production of type I interferons (IFNs), which can trigger antiviral function by inducing the production of interferon–stimulated genes (ISGs). Although IFITM3, an ISG localizing to late endosomes, can limit many viral infections, whether or not it restricts the infection of RV is still unknown. Therefore, we attempted to determine whether IFITM3 also restricts RV infection by using over–expression and knockout cell strains. It was found that IFITM3–expressing cell strains were less susceptible to RV infection, as the replication of RV in over–expressing cells was significantly less than in control group cells. Correspondingly, IFITM3–knockout cells were significantly susceptible compared to the normal cells. Furthermore, the IFN–induced antiviral effect was significantly attenuated in the absence of IFITM3, and IFITM3 delayed RV escape from endosomes in the presence of IFITM3, suggesting that endogenous IFITM3 is of great importance in type I IFN–mediated antiviral responses and may restrict infection by affecting the function of the late endosomal compartment. Porcine circovirus type 2 (PCV2) is intensely prevalent in global pig farms. The PCV2 vaccine is an important means of preventing and controlling PCV2. The quality control of PCV2 vaccines is predominantly based on detection techniques such as animal testing and neutralizing antibody titration. Measuring the content of effective proteins in vaccines to measure vaccine efficacy is an excellent alternative to traditional methods, which can greatly accelerate the development speed and testing time of vaccines. In this study, we screened a monoclonal antibody (mAb) that can effectively recognize not only the exogenous expression of PCV2 Cap protein but also PCV2 virus. The double antibody sandwich ELISA (DAS-ELISA) was developed using this mAb that specifically recognize PCV2 Cap. The minimum protein content detected by this method is 3.5 ng/mL. This method can be used for the quality control of PCV2 inactivated vaccine and subunit vaccine, and the detection results are consistent with the results of mice animal experiments. This method has the advantages of simple operation, good sensitivity, high specificity and wide application. Hybrid integration of two-dimensional (2D) materials with photonic integrated circuits can enable additional functionality in planar waveguides based on excellent optical and electrical properties of 2D materials. 2D layered palladium diselenide (PdSe2) has a narrow energy bandgap and high carrier mobility, and its stability under normal laboratory environment conditions makes it of interest for use as high-performance infrared photodetectors. In this work, we propose and experimentally demonstrate a high-speed waveguide-integrated photodetector which uses chemical vapor deposition grown PdSe2 transferred onto a silicon waveguide. At 1550 nm wavelength, the photodetector can be operated without external bias with a responsivity of 0.57 mA/W and a responsivity of 20 mA/W at 6 V bias voltage. The detector had an impulse response full-width-half-maximum pulse width of about 11 ps, corresponding to a 3-dB bandwidth of 40 GHz. [ABSTRACT FROM AUTHOR]

Wastewater-based epidemiology (WBE) has emerged as an effective environmental surveillance tool in monitoring fecal-oral pathogen infections within a community. Congruently, SARS-CoV-2, the etiologic agent of COVID-19, has been demonstrated to infect the gastrointestinal tissues, and be shed in feces. In the present study, SARS-CoV-2 RNA was concentrated from wastewater, sludge, surface water, ground water, sediment, and soil samples of municipal and hospital wastewater systems and related environments in Wuhan during the COVID-19 middle and low risk periods, and the viral RNA copies quantified using reverse transcription quantitative polymerase chain reaction (RT-qPCR). From the findings of this study, during the middle risk period, one influent sample and three secondary effluents collected from waste water treatment plant 2, as well as two samples from Jinyintan Hospital wastewater system influent were SARS-CoV-2 RNA positive. One sludge sample collected from Guanggu Branch of Tongji Hospital, which was obtained during the low risk period, was also positive for SARS-CoV-2 RNA. These study findings demonstrate the significance of WBE in continuous surveillance of SARS-CoV-2 at the community level, even when the COVID-19 prevalence is low. Overall, this study can be used as an important reference for contingency management of wastewater treatment plants and COVID-19 prevention and control departments of Wuhan. Porcine reproductive and respiratory syndrome virus is one of the main pathogens threatening the global pig industry, and there is still a lack of effective therapeutic drugs. Sanggenon C is a flavanone Diels-Alder adduct compound extracted from the root bark of the mulberry genus, which has blood pressure-reducing, anti-atherosclerotic, anti-oxidative, and anti-inflammatory effects. In our previous study, Sanggenon C was confirmed to significantly inhibit PRRSV replication in vitro. However, its antiviral potential to inhibit PRRSV infection in vivo has not been evaluated in piglets. Here, the antiviral effect of Sanggenon C was evaluated in PRRSV-challenged piglets based on assessments of rectal temperature, viral load, pathological changes of lung tissue and secretion of inflammatory cytokines. The results showed that Sanggenon C treatment relieved the clinical symptoms, reduced the viral loads in the lungs and bloods, alleviated the pathological damage of lung tissue, decreased the secretion of inflammatory cytokines, and shorten the excretion time of virus from the oral and nasal secretions and feces of piglets after PRRSV infection. Abstract: The immunoglobulin heavy chain variable region in vertebrates is composed of many genes. However, the gene number and the extent of diversity among the VH gene segments vary with the species. To gain insight into the dog VH repertoire, we analyzed its genome sequence. Eighty VH, six DH and three JH segments were mapped to a 1.28Mb region of the dog chromosome 8. Forty-one of these VH genes were potentially functional, and 39 were pseudogenes. The sequence similarity analysis suggests that the dog IgH locus harbors three VH gene families, VH1 (including 76 members), VH2 (including 3 members) and VH3 (including one member), that are classified into mammalian clan III, I and II, respectively. Subsequently, 111 cDNA positive clones from dog 26 μ, 22 γ, 23 α, 16 δ and 24 ɛ chains containing almost full-length VH, DH and JH segments were selected at random. All of the sequences, except for clone G6, which belonged to the dog VH2 family, originated from the dog VH1 family. The germline segments, VH62 (human VH3-21 homology) and VH44 (human VH3-23 homology), seemed to be preferentially used. Our results suggest that the dog VH repertoire appears to be derived from limited germline gene families, and its diversity may rely on junctional diversity and somatic hypermutation. Recombination is an important phenomenon that accelerates evolution and enriches the genetic diversity of porcine reproductive and respiratory syndrome virus (PRRSV). Recombinant PRRSV isolates sometimes have different genetic backgrounds. In this study, we report a recombinant PRRSV (SD-YL1712) isolated from a pig farm. The genome of SD-YL1712 is 15,014 nucleotides in length, and its nucleotide and amino acid sequence conservation is higher than that of PRRSV strain JXA1 except within the NSP2 region. The NSP2 region of SDYL1712 shares the highest nucleotide (85.9%) and amino acid (84.1%) sequence identity with PRRSV strain NADC30. SD-YL1712 was found to contain a characteristic 131-amino-acid deletion in the NSP2 region. Two recombination breakpoints were detected at nt 2134 and nt 3958 within the NSP2 region, which revealed that SD-YL1712 originated from a recombination event between NADC30-like and HP-PRRSV-derived MLV-like strains. Interestingly, SD-YL1712 had an additional deletion at position 586, similar to that found in strain TJnh1501. Moreover, the pathogenicity of strain SD-YL1712 was found to be similar to that of HP-PRRSV JXA1, which was higher than that of the CH1a strain. Warming can accelerate the decomposition of soil organic matter and stimulate the release of soil greenhouse gases (GHGs), but to what extent soil release of methane (CH4) and nitrous oxide (N2O) may contribute to soil C loss for driving climate change under warming remains unresolved. By synthesizing 1,845 measurements from 164 peer‐reviewed publications, we show that around 1.5°C (1.16–2.01°C) of experimental warming significantly stimulates soil respiration by 12.9%, N2O emissions by 35.2%, CH4 emissions by 23.4% from rice paddies, and by 37.5% from natural wetlands. Rising temperature increases CH4 uptake of upland soils by 13.8%. Warming‐enhanced emission of soil CH4 and N2O corresponds to an overall source strength of 1.19, 1.84, and 3.12 Pg CO2‐equivalent/year under 1°C, 1.5°C, and 2°C warming scenarios, respectively, interacting with soil C loss of 1.60 Pg CO2/year in terms of contribution to climate change. The warming‐induced rise in soil CH4 and N2O emissions (1.84 Pg CO2‐equivalent/year) could reduce mitigation potential of terrestrial net ecosystem production by 8.3% (NEP, 22.25 Pg CO2/year) under warming. Soil respiration and CH4 release are intensified following the mean warming threshold of 1.5°C scenario, as compared to soil CH4 uptake and N2O release with a reduced and less positive response, respectively. Soil C loss increases to a larger extent under soil warming than under canopy air warming. Warming‐raised emission of soil GHG increases with the intensity of temperature rise but decreases with the extension of experimental duration. This synthesis takes the lead to quantify the ecosystem C and N cycling in response to warming and advances our capacity to predict terrestrial feedback to climate change under projected warming scenarios. Abstract: To investigate the influence of extractable organic matter (EOM) on pore evolution of lacustrine shales, Soxhlet extraction, using dichloromethane, was performed on a series of Chang 7 shale samples (Ordos Basin, China) with vitrinite reflectance of 0.64% to 1.34%. Low‐pressure gas adsorption experiments were conducted on the samples before and after extraction. The pore structure parameters were calculated from the gas adsorption data. The results show complex changes to the pore volumes and surface areas after extraction. The pore development of both the initial and extracted samples is strongly controlled by total organic carbon (TOC) content. Micropores developed mainly in organic matter (OM), while mesopores and macropores predominantly developed in fractions other than OM. The influence of EOM on micropores is stronger than on mesopores and macropores. Organic solvents with a higher boiling point should be used to explore the effect of EOM on pore structure in the future. [ABSTRACT FROM AUTHOR]

Hepatitis E virus (HEV) has been detected in rabbits, a recently identified natural reservoir. In this study, anti-HEV antibodies and viral RNA were detected in rabbits sourced from a specific-pathogen-free (SPF) rabbit vendor in Shaanxi Province, China. BLAST results of partial HEV ORF2 genes cloned here indicated that two viral strains circulated in the rabbits. Sequence determination of the complete genome (7302 bp) of one strain and a partial ORF1 gene (1537 bp) of the other strain showed that they shared 90% identity with one another and 78%–94% identity with other known rabbit HEVs. In addition, inoculation with rabbit HEV from SPF rabbits studied here resulted in infection of SPF pigs; this cross-species transmission was evidenced by seroconversion, viremia and faecal virus shedding. These results suggest that to prevent spread of this zoonotic pathogen, rabbits should be tested routinely for HEV RNA in SPF vendor facilities. [ABSTRACT FROM AUTHOR]

Porcinereproductiveandrespiratorysyndromevirus (PRRSV) causes significant economic losses to the pork industry worldwide. Previously, we demonstrated that heme oxygenase-1 (HO-1) interferes with PRRSV replication. To elucidate the mechanisms involved, here we assess whether the HO-1 downstream metabolites biliverdin (BV) and/or iron mediate the HO-1 antiviral effect. We demonstrate a BV concentration-dependent suppression of PRRSV replication and show that virions are not directly inactivated by BV. Additionally, BV or N-acetyl cysteine (NAC) significantly reduced reactive oxygen species (ROS) in PRRSV-infected MARC-145 cells; however, because NAC did not reduce viral load, the BV antiviral effect is independent of decreased ROS levels. Moreover, a secondary metabolite of BV, bilirubin (BR), specifically mediates this anti-PRRSV activity via a nitric oxide (NO)-dependent cGMP/PKG signaling pathway. While increased iron via addition of FeCl 3 did not interfere with PRRSV replication, iron depletion by deferoxamine (DFO) after cobalt-protoporphyrin IX induction of HO-1 did not restore PRRSV replication. Collectively, our findings identify a HO-1-BV/BR-NO-cGMP/PKG cascade as a novel pathway underlying the host cell antiviral effect. Severe Acute Respiratory Syndrome Coronavirus 2 (SARS-CoV-2), as the pathogen of coronavirus disease 2019 (COVID-19), has infected millions of people and took hundreds of thousands of lives. Unfortunately, there is deficiency of effective medicines to prevent or treat COVID-19. 3C like protease (3CLPro) of SARS-CoV-2 is essential to the viral replication and transcription, and is an attractive target to develop anti-SARS-CoV-2 agents. Targeting on the 3CLPro, we screened our protease inhibitor library and obtained compound 10a as hit to weakly inhibit the SARS-CoV-2 3CLPro, and determined the co-crystal structure of 10a and the protease. Based on the deep understanding on the protein-ligand complexes between the hit and SARS-CoV-2 3CLPro, we designed a series of peptidomimetic inhibitors, with outstanding inhibitory activity against SARS-CoV-2 3CLPro and excellent anti-viral potency against SARS-CoV-2. The protein-ligand complexes of the other key inhibitors with SARS-CoV-2 3CLPro were explicitly described by the X-ray co-crystal study. All such results suggest these peptidomimetic inhibitors could be further applied as encouraging drug candidates. Porcine reproductive and respiratory syndrome (PRRS) is a widespread viral disease affecting the swine industry, with no cure or effective treatment. Current vaccines are inefficient mainly due to the high degree of genetic and antigenic variation within PRRS virus (PRRSV) strains. Thus, the development of novel anti-PRRSV strategies is an important area of research. The nonstructural protein 9 (Nsp9) of PRRSV is essential for viral replication, and its sequence is relatively conserved, making it a logical antiviral target for PRRSV. Camel single-domain antibodies (nanobodies) represent a promising antiviral approach because of their small size, high specificity, and solubility. However, no nanobodies against PRRSV have been reported to date. In this study, Nsp9-specific nanobodies were isolated from a phage display library of variable domains of Camellidae heavy chain-only antibodies (VHH). One of the isolated nanobodies, Nb6, was chosen for further investigation. Co-immunoprecipitation experiments indicated that Nb6 can still maintain antigen binding capabilities when expressed in the cell cytoplasm. A MARC-145 cell line stably expressing Nb6 was established to investigate its potential antiviral activity. Our results showed that intracellularly expressed Nb6 could potently suppress PRRSV replication by inhibiting viral genome replication and transcription. More importantly, Nb6 could protect MARC-145 cells from virus-induced cytopathic effect (CPE) and fully block PRRSV replication at an MOI of 0.01 or lower. To our knowledge, this is the first report of a nanobody based antiviral strategy against PRRSV, and this finding has the potential to lead to future developments of novel antiviral treatments for PRRSV infection. Cell apoptosis is common after infection with porcine reproductive and respiratory syndrome virus (PRRSV). PRRSV GP5 has been reported to induce cell apoptosis. To further understand the role of GP5 in PRRSV induced cell apoptosis, we established Marc-145 cell lines stably expressing full-length GP5, GP5 Δ84-96 (aa 84-96 deletion), and GP5 Δ97-119 (aa 97-119 deletion). Cell proliferation, cell cycle progression, cell apoptosis and virus replication in these cell lines were evaluated. Neither truncated nor full-length GP5 induced cell apoptosis in Marc-145 cells. However, GP5 Δ97-119 , but not full-length or GP5 Δ84-96 , induced a cell cycle arrest at the G2/M phase resulting in a reduction in the growth of Marc-145 cells. Additionally, GP5 Δ84-96 inhibited the replication of PRRSV in Marc-145 cells through induction of IFN-β. These findings suggest that PRRSV GP5 is not responsible for inducing cell apoptosis in Marc-145 cells under these experimental conditions; however it has other important roles in virus/host cell biology. Monoclonal anti-idiotypic antibody (Mab2-5G2) raised against idiotypic antibodies to membrane glycoprotein GP5 of porcine reproductive and respiratory syndrome virus (PRRSV). The variable regions of the heavy chain (VH) and light chain (VL) of Mab2-5G2 were cloned and connected with a (Gly 4 Ser) 3 linker. The recombinant scFv gene was cloned into the pEasy-E1 vector and expressed in E. coli as inclusion bodies. The expressed scFv-His proteins renatured in a pH and urea gradient buffer retained the same immunological properties as that of Mab2-5G2. Renatured scFv-His protein retained the same characteristics as that of Mab2-5G2 by recognizing and binding to Marc-145 cells. Furthermore, renatured scFv-His along with Mab2-5G2 were used to immunize rabbits to produce anti-anti-idiotypic antibodies (Ab3) that neutralized PRRSV infection of Marc-145 cells. These results demonstrated that the expressed scFv-His protein possessed the same characteristics of Mab2-5G2 and will be suitable for future investigations of Mab2-5G2 antibody structure and its ability to interact with potential PRRSV cellular receptor as well as immunological properties against PRRSV infection. The major neutralizing epitope of porcine reproductive and respiratory syndrome virus (PRRSV) is mainly located on virus glycoprotein 5 (GP5). Immunization with exogenous GP5 or exposure to native GP5 by means of DNA immunization can provide some degree of immune protection to PRRSV infection in pigs. However, during PRRSV infection in pigs, the production of neutralization antibodies induced by GP5 is delayed or suppressed. This suggests that the synthesis of GP5 is late than some PRRSV proteins or other PRRSV proteins interfering with the function of GP5 in inducing host responses during virus infection. Here, to exclude the impacts of the other PRRSV proteins and determine the role of GP5 in the replication of PRRSV in vitro , a Marc-145 cell line stably expressing GP5 (Marc-145-GP5 Flag ) was constructed. Cell proliferation and cell apoptosis measurements indicated that the expression of GP5 in Marc-145 cells did not disturb the cells’ viability. Following infection with different PRRSV strains PRRSV replication in Marc-145-GP5 Flag cells was inhibited significantly. Type I interferon assay results showed that beta interferon (IFN-β) in the Marc-145-GP5 Flag cells were increased at mRNA and protein levels. When siRNA was introduced into the cells to knock down IFN-β mRNA, PRRSV infectivity of these cells was recovered. These data suggest that early GP5 expression is not favorable for further infection by PRRSV, because it not only stimulates production of neutralization antibodies in pigs, but also induces IFN-β production in host cells. The lysophosphatidic acid receptor 1 (LPA1), a G-protein coupled receptor, regulates cell proliferation, migration, and cytokine release. Here, we investigate the molecular signature of LPA1 trafficking to the cell surface. The overexpressed LPA1 with a C-terminal V5 tag (LPA1-V5) is majorly expressed on the cell surface, while two deletion mutants (C320 and ∆84–87) failed to be trafficked to the cell surface. Further, site-directed mutagenesis analysis of the LPA1 revealed that Ile325, Tyr85, and Leu87 within these two fragments regulate LPA1 maturation and trafficking to the cell surface. Over-expression of Sar1, a component of coat protein complex II (COPII), enhances glycosylation of LPA1 wild type, but not these mutants. The mutants of LPA1 are majorly localized in the endoplasmic reticulum (ER) and exhibit a higher binding affinity to heat shock protein 70 (Hsp70), when compared to the LPA1 wild type. Further, we found that all these mutants failed to increase phosphorylation of Erk, and the cytokine release in response to LPA treatment. The diversity of porcine reproductive and respiratory syndrome virus (PRRSV) in China is increasing rapidly along with mutation and recombination. Recombination could occur between inter- and intra-lineage of PRRSV, which accelerated the complexity of pathogenicity and cell tropism of the recombinant strain. In the present study, a novel PRRSV strain named HN-YL1711 was isolated from a pig farm suffering from severe respiratory difficulty in Henan province, China. The whole genomic sequence analysis indicated that the genome of HN-YL1711 was 15018 nt. It shared 86%, 87.3%, 88.1%, 91.1%, 84.2%, and 84.1% nucleotide similarities with PRRSVs VR2332, CH1a, JXA1, NADC30, QYYZ, and GM2, respectively. Based on phylogenetic analysis of Nsp2, ORF5 and complete genomes, HN-YL1711 was classified into lineage 1 of PRRSV. However, seven genomic break points were detected in recombination analysis, which indicated that the HN-YL1711 originated from multiple recombination among NADC30-like (major parent, lineage 1), JXA1-like (minor parent, lineage 8), and QYYZ-like (minor parent, lineage 3) PRRSV. Porcine alveolar macrophages (PAMs), 3D4/21-CD163 and MARC-145 cells were used to explore the viral adaptation of HN-YL1711. The results indicated that it could infect the PAMs but failed to infect MARC-145 cells. Challenge experiments showed that HN-YL1711 exhibits intermediate virulence in pigs, compared with HP-PRRSV JXA1 and LP-PRRSV CH1a. Abstract: Streptococcus equi ssp. zooepidemicus (SEZ) is an important pathogen associated with a wide range of diseases in many mammalian species. The development of novel effective vaccines would be beneficial to control SEZ infection. In the present study, the importance of the SEZ capsule was examined using a newly constructed capsule-deficient mutant ΔhasB strain. Transmission electron microscopy confirmed a decrease in the abundance of extracellular capsular polysaccharide on the mutant SEZ. Compared to the parental wild-type SEZ, the ΔhasB mutant was highly attenuated in mice and provided 100% protection against lethal challenge when administered as a live vaccine. Real-time PCR analysis showed a marked increased in the levels of IL-4 and IFN-γ mRNA in immunized mice. The role that the capsule plays in SEZ pathogenicity was also explored with respect to the mechanistic design of an attenuated vaccine target. The capsule could resist complement C3 deposition on the surface of SEZ cells and aid in preventing complement-mediated opsonization and phagocytosis by cultured macrophages. These results suggest that the capsule of SEZ plays an important role in pathogenicity and may serve as a target for attenuation in vaccine development. [Copyright &y& Elsevier]

Abstract: This study was designed to investigate the single nucleotide polymorphism (SNP) in intron 1 of the gene A-FABP in 127 Junmu No. 1 white swine using PCR–SSCP. The association between the polymorphism and meat quality traits was also studied. The cloning and sequencing results indicated that the polymorphism of intron 1 was due to a point mutation in position 3481bp of A-FABP, giving 3 genotypes (CC, CD and DD). Association analysis indicated that the polymorphism had a significant effect on marbling (P<0.05). Genotype DD had higher marbling than CD and CC, but the difference between CD and CC was no significant. Polymorphism had a highly significant effect on intramuscular fat (IMF) content (P<0.01). DD was higher than CD, which was higher than CC. No significant conclusions can be drawn regarding other traits. Immunoblot analysis of A-FABP levels was carried out on 3 different genotype individuals. Expression was markedly reduced in DD compared with genotype CC. Thus A-FABP may be a candidate gene or a quantitative trait locus-linked gene associated with meat quality traits. [Copyright &y& Elsevier]

Progestin and adipoQ receptor family member VI (PAQR6) is a new member of the Progestin and adipoQ receptors (PAQRs) family that functions as an important factor in the nongenomic actions of rapid steroid response. Considering the important role of PAQR6 in these nongenomic actions, we determined porcine PAQR6 expression and regulation. We cloned the cDNA of porcine PAQR6 and analyzed its genomic structure. Subcellular localization analysis showed that PAQR6-green fluorescent protein fusion protein was distributed in cytoplasm. Spatial expression patterns analysis revealed that porcine PAQR6 was more highly expressed in small intestinal than in other tissues. To investigate the regulatory mechanism of porcine PAQR6, we cloned approximately 1.5 kb of porcine PAQR6 5′-regulatory region and generated sequential deletion constructs and evaluated their activity in a dual-luciferase reporter assay. The results suggested that the core promoter region of this gene is located in the first exon region from +90 to +241. Site-directed mutagenesis indicated that adiponectin receptor protein 1, E2F transcription factor 1, and Sp1 transcription factor might be important transcription factors for porcine PAQR6. This study is the first attempt to elucidate the transcriptional regulation of porcine PAQR6, which established a foundation for further study and contributed to the deeper investigation of the regulation and function of PAQR6. The innate immune response provides the initial defence mechanism against infection by other organisms. However, an excessive immune response will cause damage to host tissues. In an attempt to identify microRNAs (miRNAs) that regulate the innate immune response in inflammation and homeostasis, we examined the differential expression of miRNAs using microarray analysis in the spleens of mice injected intraperitoneally with lipopolysaccharide (LPS) and saline, respectively. Following challenge, we observed 19 miRNAs up-regulated (1·5-fold) in response to LPS. Among these miRNAs, miR-1224, whose expression level increased 5·7-fold 6 hr after LPS injection and 2·3-fold after 24 hr, was selected for further study. Tissue expression patterns showed that mouse miR-1224 is highly expressed in mouse spleen, kidney and lung. Transfection of miR-1224 mimics resulted in a decrease in basal tumour necrosis factor-α (TNF-α) promoter reporter gene activity and a down-regulation of LPS-induced TNF-α mRNA in RAW264.7 cells. With public databases of miRNA target prediction, miR-1224 was shown to bind to the 3′ untranslated region (UTR) of Sp1 mRNA, whose coding product controls TNF-α expression at the transcriptional level. Furthermore, we found that in HEK-293 cells, the activity of the luciferase reporter bearing Sp1 mRNA 3′ UTR was down-regulated significantly when transfected with miR-1224 mimics. After transfection of miR-1224 in RAW264.7 cells, nucleus Sp1 protein level decreased, and when endogenous miR-1224 was blocked, the decrease was abolished. Therefore, we initially speculated that miR-1224 was a negative regulator of TNF-α in an Sp1-dependent manner, which was confirmed in vivo by chromatin immunoprecipitation assay, and might be involved in regulating the LPS-mediated inflammatory responses. Abstract: The follicle-stimulating hormone (FSH) acts on the Sertoli cells in the seminiferous tubules of the testis and regulates spermatogenesis up to the secondary spermatocyte stage. This study aimed to investigate molecular genetic characteristics of the bovine FSH β-subunit gene (FSHB) and elucidate the effects of single nucleotide polymorphisms (SNPs) of FSHB on the quality of fresh and frozen semen and on fertility in bulls. We used polymerase chain reaction single-strand conformation polymorphism (PCR-SSCP) and sequencing of the FSHB gene in 56 bulls belonging to three breeds. We identified 13 substitutions and 1 insertion in the upstream regulation region and in the coding region of exon 3, which were all linked together. Bioinformatics analysis suggested that mutations of the 5′-upstream regulation region altered the binding sites for transcription factors, and radioimmunoassay demonstrated that mutations may result in alterations in the serum FSH concentrations. The least-squares analysis revealed that bulls with this genotype exhibited a significantly lower sperm concentration in fresh semen and a lower percentage of acrosome integrity in both fresh and frozen semen (P <0.05). These bulls also exhibited a significantly higher percentage of sperm deformity in fresh semen (P <0.05), which was more pronounced in frozen semen (P <0.01), and a significantly lower sperm motility in frozen semen (P <0.05). For fertility evaluation, the nonreturn rates obtained from 14,416 inseminations with the analyzed batches revealed that bulls with this genotype showed significantly lower nonreturn rates (P <0.05). In other words, bulls with this genotype exhibited lower semen quality, poor freeze resistance, and lower fertility. Porcine reproductive and respiratory syndrome (PRRS) is a serious infectious disease in the swine industry, which causes severe economic losses to current swine production worldwide. There are no effective antiviral strategies for preventing this disease. Previous studies showed that microRNAs (miRNAs) play important role in virus-host interactions. In this study, we demonstrated that the expression level of ssc-miR-124a was significantly downregulated during both high and low pathogenic PRRSV infection. Overexpression of ssc-miR-124a markedly inhibits PRRSV replication in PAMs. Luciferase reporter experiments and RISC immunoprecipitation assay were used to identify the ssc-miR-124a could directly target the 3'UTR of pig CD163 mRNA in a sequence-specific manner and that CD163 mRNA and protein levels were reduced in PAMs overexpressing ssc-miR-124a. These data not only provide new insights into virus-host interactions during PRRSV infection, but also suggest potential new antiviral strategies against PRRSV infection in the future. Porcine epidemic diarrhea virus (PEDV) causes an emerging and re-emerging coronavirus disease characterized by vomiting, acute diarrhea, dehydration, and up to 100% mortality in neonatal suckling piglets, leading to huge economic losses in the global swine industry. Vaccination remains the most promising and effective way to prevent and control PEDV. However, effective vaccines for PEDV are still under development. Understanding the genomic structure and function of PEDV and the influence of the viral components on innate immunity is essential for developing effective vaccines. Bovine viral diarrhea virus (BVDV) infection causes significant economic losses to the cattle industry worldwide and still represents a huge pressure on agricultural production. Thus, the development of novel anti-BVDV strategies are urgently needed. The nonstructural protein 5 (NS5B) of BVDV is essential for viral replication. Further, the camel single-domain antibody (nanobody) represents a promising antiviral approach with the advantages of small size, stable structure, high specificity and solubility, and the recognition of specific epitopes. However, no NS5B-specific nanobodies against BVDV have been reported. In this study, NS5B-specific nanobodies were isolated from a phage display library of variable domains of Camellidae heavy chain-only antibodies (VHHs). Further, an MDBK cell line stably expressing Nb1 was established to explore antiviral activity. Results showed that Nb1 could markedly suppress BVDV replication and interact with the BVDV NS5B protein.
