Abstract Color is one of the most important indicators to characteristic the quality of tobacco, which is strongly related to the variations of chemical components. In order to clarify the relationship between the changes of tobacco color and chemical components, here we established several prediction models of chemical components with the color values of tobacco based on machine learning algorithms. The results of correlation analysis showed that tobacco moisture content was highly significantly correlated with the parameters such as a*, H* and H°, the reducing sugar and total sugar content of tobacco was significantly correlated with the color values, and the starch content was highly significantly correlated with the color values except for b* and C*. The random forest models performed best in predicting tobacco moisture, reducing sugar, total sugar and starch constructed with the R 2 of the model validation set was higher than 0.90, and the RPD value was greater than 2.0. The consistent between the predictions and measurements verified the availability and feasibility using color values to predict some chemical components of the tobacco leaves with high accuracy, and which has distinct advantages and potential application to realize the real-time monitoring of some chemical components in the tobacco curing process.

ABSTRACT To explore the factors influencing the occurrence of bacterial wilt, the differences in the physicochemical properties, microbial community composition and function between rhizosphere soil of tobacco plants with bacterial wilt and healthy plants in the tobacco planting area of Fuzhou City, Jiangxi Province were analyzed and compared. The results showed that the rhizosphere soil of diseased tobacco exhibited significantly reduced levels of exchangeable potassium, water-soluble potassium, nitrate nitrogen, total nitrogen and pH, in comparison to the rhizosphere soil of healthy plants. Conversely, the available phosphorus content of the rhizosphere soil of diseased tobacco was significantly increased. The amount of Ralstonia solanacearum in soil was negatively correlated with pH, nitrate nitrogen and total nitrogen, and positively correlated with exchangeable potassium and water-soluble potassium. A total of 43 genera were significantly different between the two groups of rhizosphere soil, of which 24 genera were enriched in the rhizosphere of healthy plants, including Ideonella, Rhizophagus, Rhizobacter, Altererythrobacter and Ignavibacterium associated with plant disease resistance, Thermodesulfovibrio, Syntrophorhabdus, Syntrophus, Chlorobium, Hydrogenophaga and Limnohabitans associated with soil sulfur metabolism, as well as Ignavibacterium, Ideonella, Derxia and Azohydromonas associated with soil nitrogen cycling. Kyoto Encyclopedia of Genes and Genomes functional analysis of the unigenes obtained by metagenomic sequencing also showed that the differential unigenes were significantly enriched in the sulfur metabolism pathway. In addition, the rhizosphere soil of diseased tobacco plants exhibited a higher abundance of antibiotic-producing actinomycetes and an increased load of antibiotic resistance genes compared to that of healthy plants. In general, lower pH value, less content of nitrate nitrogen and total nitrogen, and more content of exchangeable potassium and water-soluble potassium could contribute to onset of bacterial wilt. Twenty-four genera, including Ideonella and Rhizophagus, may construct a healthy microecological network in the rhizosphere of tobacco plants. All these factors may interact with each other to control the development of bacterial wilt. This complicated interaction network needs to be explored further.IMPORTANCEPrevious studies have mainly focused on the differences in microbial species composition between healthy and diseased soils, but the differences in microbial community functions between two types of soil have not been well characterized. In this study, soil samples in diseased and healthy plant rhizospheres were collected for physicochemical property testing and metagenomic sequencing. We focused on analyzing the differences in physicochemical properties and microbial community functions between these soils, as well as the correlation between these factors and pathogen content. The results of this study provide a theoretical basis for further understanding the occurrence of tobacco bacterial wilt in the field.

In order to explore a resource-saving and environmentally friendly approach to reduce nitrogen fertilizer and increase soil carbon pool in crop production. There are 5 treatments in the experiment namely CK: conventional fertilization and irrigation methods (pure nitrogen 30 kg/hm2); T1: drip irrigation fertilization method (pure nitrogen 15 kg/hm2); T2: drip irrigation fertilization method (pure nitrogen 15 kg /hm2), biochar-based fertilizer 600 kg/hm2; T3: drip irrigation method (pure nitrogen 15 kg/hm2), biochar-based fertilizer 1200 kg/hm2; T4: drip irrigation method (pure nitrogen 15 kg/hm2), biochar-based fertilizer is 1800 kg/hm2. The results showed that the microbial carbon content of the biochar-based fertilizer treatment under the condition of drip irrigation to reduce nitrogen was increased by 39.9%-68.4% as compared with the control. All treatments of the biochar-based fertilizer can significantly increase the soil easily oxidizable carbon in the early and mid-term growth. The content of solid carbon and soluble organic carbon increased by 31.3%-148.31% and 12.2%-63.92%, respectively. The application of biochar-based fertilizers significantly increased the net photosynthetic rate and stomatal conductance in the early and mid-transplanting period, as well as the transpiration rate and intercellular CO2 concentration throughout the growth period, and the SPAD, NDVI value, and dry matter accumulation. Under the condition of reducing nitrogen by 50%, drip irrigation combined with biochar-based fertilizer can significantly increase the soil microbial biomass carbon content, oxidizable carbon and soluble carbon content, promote the photosynthetic effect and dry matter accumulation of tobacco plants, and it is beneficial to promote crop planting.

Abstract Root-knot nematode disease is a catastrophic soil-borne disease in tobacco production. The regulation of natural microbial communities is considered a good disease management approach to suppress the incidence of soilborne diseases. In this study, the effects of tobacco (Nicotiana tabacum L.)-marigold (Tagetes erecta L.) rotation on the diversity and structure of soil microbial communities in continuously cropped tobacco fields were analyzed to manage this devastating pathogen. The results showed that the soil bacterial OTUs increased after marigold rotation and that the bacterial Shannon, ACE, Chao1 index, and fungal Shannon index were higher in the tobacco-marigold rotation fields than in the continuously cropped tobacco fields by 3.98%, 10.37%, 5.46%, and 3.43%, respectively. After marigold rotation, the relative abundances of Actinobacteria, Acidobacteria, and Ascomycota increased by 28.62%, 107.50%, and 57.44%, respectively, and the proportion of beneficial bacterial genera such as Nocardioides, Gemmatimonas, and Bradyrhizobium increased. In addition, our results also showed that rotation of marigold could effectively reduce the incidence of root-knot nematodes in the next crop of tobacco. These results indicate that marigold rotation had a positive effect on the soil microecological environment of continuously cropped tobacco fields, reducing the obstacles to continuous cropping of tobacco.

Bacterial wilt, which is a major soil-borne disease with widespread occurrence, poses a severe danger in the field of tobacco production. However, there is very limited knowledge on bacterial wilt-induced microecological changes in the tobacco root system and on the interaction between Ralstonia solanacearum and fungal communities in the rhizosphere soil. Thus, in this study, changes in fungal communities in the rhizosphere soil of tobaccos with bacterial wilt were studied by 18S rRNA gene sequencing. The community composition of fungi in bacterial wilt-infected soil and healthy soil in two tobacco areas (Gengma and Boshang, Lincang City, Yunnan Province, China) was studied through the paired comparison method in July 2019. The results showed that there were significant differences in fungal community composition between the rhizosphere soil of diseased plants and healthy plants. The changes in the composition and diversity of fungal communities in the rhizosphere soil of tobaccos are vital characteristics of tobaccos with bacterial wilt, and the imbalance in the rhizosphere microecosystem of tobacco plants may further aggravate the disease.

Abstract We tested the hypothesis that soil texture and nitrogen (N) fertilisation are the primary factors regulating the N cycle and soil bacterial community structure. The response of soil bacterial communities to N fertilisation in different textured soils might help in identifying the specific underlying mechanism and hence management of N fertiliser application in fields. We examined how N fertiliser accumulates in flue-cured tobacco and influences soil bacterial community structure in different textured soils. We conducted plot and micro-plot experimental measurements of N content in soil and tobacco samples using the KNO3 15N isotope technique. Soil bacterial community structure was determined using high-throughput sequencing of 16S rRNA. Nitrogen absorption and utilisation by tobacco plants were highest in sandy loam soils, followed by loam soil and clay loam. The ability of clay loam to supply N was weak during the plant growth period. Absence of fertilisation could reduce bacterial abundance in soils to various degrees. Bacterial diversity was higher in sandy loam soil than in loam soil and clay loam. Soil texture and N fertilisation significantly affected soil bacterial community structure and diversity. Proteobacteria, Acidobacteria, Firmicutes, Bacteroidetes, Actinobacteria, and Chloroflexi were the dominant bacterial phyla, while Bacillus, Nitrobacter, Nitrosospira, Nitrospira, and Rhizobium were the primary N transformation bacteria at the genus level in all treatments. However, relative abundances differed with N fertiliser application, which could lead to differential N availability and N use efficiency of tobacco among soil types. We conclude that both soil texture and N fertilisation influence N accumulation and distribution in flue-cured tobacco and thus regulate soil bacterial communities. N fertiliser application in sandy loam soil should be strictly controlled for its higher N use efficiency, soil bacterial abundance, and diversity.

Bacterial communities in the rhizosphere play an important role in sustaining plant growth and the health of diverse soils. Recent studies have demonstrated that microbial keystone taxa in the rhizosphere microbial community are extremely critical for the suppression of diseases. However, the mechanisms involved in disease suppression by keystone species remain unclear. The present study assessed the effects of three Pseudomonas strains, which were identified as keystone species in our previous study, on the growth performance and root-associated bacterial community of tobacco plants. A high relative abundance of Ralstonia was found in the non-inoculated group, while a large Azospira population was observed in all groups inoculated with the three Pseudomonas strains. Correspondingly, the activities of the defense-related enzymes and the expression levels of the defense signaling marker genes of the plant were increased after inoculation with the Pseudomonas strains. Unhindered transportation of substances in the brain extracellular space (ECS) is essential for maintaining brain function. Regulation of transportation is a novel strategy for treating ECS blockage-related brain diseases, but few techniques have been developed to date. In this study, we established a novel approach for accelerating the drainage of brain interstitial fluid (ISF) in the ECS using minimally invasive surgery, in which a branch of the external carotid artery is separated and implanted epidurally (i.e., epidural arterial implantation [EAI]) to promote a pulsation effect on cerebrospinal fluid (CSF) in the frontoparietal region. Tracer-based magnetic resonance imaging was used to evaluate the changes in ISF drainage in rats 7 and 15 days post-EAI. The drainage of the traced ISF from the caudate nucleus to ipsilateral cortex was significantly accelerated by EAI. Significant increases in the volume fraction of the ECS and molecular diffusion rate were demonstrated using the DECS-mapping technique, which may account for the mechanisms underlying the changes in brain ISF. The drainage of brain interstitial fluid (ISF) has been observed to slow down following neuronal excitation, although the mechanism underlying this phenomenon is yet to be elucidated. In searching for the changes in the brain extracellular space (ECS) induced by electrical pain stimuli in the rat thalamus, significantly decreased effective diffusion coefficient (DECS) and volume fraction (a) of the brain ECS were shown, accompanied by the slowdown of ISF drainage. The morphological basis for structural changes in the brain ECS was local spatial deformation of astrocyte foot processes following neuronal excitation. We further studied aquaporin-4 gene (APQ4) knockout rats in which the changes of the brain ECS structure were reversed and found that the slowed DECS and ISF drainage persisted, confirming that the down-regulation of ISF drainage following neuronal excitation was mainly attributable to the release of neurotransmitters rather than to structural changes of the brain ECS. Meanwhile, the dynamic changes in the DECS were synchronized with the release and elimination processes of neurotransmitters following neuronal excitation. In conclusion, the downregulation of ISF drainage following neuronal excitation was found to be caused by the restricted diffusion in the brain ECS, and DECS mapping may be used to track the neuronal activity in the deep brain. Biomass moulding fuel is an environmentally friendly and renewable energy source. We studied an independently-developed automatic flue-cured tobacco furnace, employing a biomass fuel source and studied the effect of biomass moulding fuel onbacco flue curing. We found that 1 kg of dried tobacco required 1.2 kg of biomass moulding fuel (heating value = 3550 kcal per kg). The energy input cost was 1.43 RMB** (fuel and electricity) and the labor cost was reduced by 75%, a reduction of 18.6% compared to the cost of burning coal. With respect to the environmental impact, the average emission concentration of smoke in the exhaust gas from the furnace was 16.2 mg/m³, SO2 was 13.6 mg/m³, and NOx was 2.3 mg/m³. Ringelmann blackness was less than 1. Compared to burning coal, all emissions were very low, demonstrating that the biomass moulding fuel furnace saved energy and reduced emissions compared to coal.
