Abstract Natural immunity is the first defense line of the host immune system, which plays a significant role in combating foreign pathogenic microorganisms. The IFN-β (interferon-beta) signaling pathway, being a typical example of innate immunity, plays a vital function. This study aimed to elucidate the function of pseudorabies virus (PRV) UL38 protein (unique long region 38) in suppressing the activation of the IFN-β signaling pathway. The findings from our study indicate that the PRV UL38 protein effectively hampers the activation of IFN-β by poly (dA: dT) (poly(deoxyadenylic-deoxythymidylic)) and 2’3’-cGAMP (2’-3’-cyclic GMP-AMP). Furthermore, UL38 exhibits spatial co-localization with STING (stimulator of interferon genes) and effectively hinders STING dimerization. Subsequently, STING was downgraded to suppress the production of IFN-β and ISGs (interferon stimulated genes). Immunoprecipitation analysis revealed that the interaction between UL38 and STING, which subsequently initiated the degradation of STING via selective autophagy mediated by TOLLIP (toll interacting protein). To summarize, this research elucidates the function of UL38 in counteracting the cGAS (cGAMP synthase)-STING-induced IFN-β pathway. The PRV UL38 protein may attenuate the activation of IFN-β as a means of regulating the virus’s persistence in the host.

ABSTRACT Mammalian orthoreovirus (MRV) outer capsid protein σ3 is a multifunctional protein containing a double-stranded RNA-binding domain, which facilitates viral entry and assembly. We reasoned that σ3 has an innate immune evasion function. Here, we show that σ3 protein localizes in the mitochondria and interacts with mitochondrial antiviral signaling protein (MAVS) to activate the intrinsic mitochondria-mediated apoptotic pathway. Consequently, σ3 protein promotes the degradation of MAVS through the intrinsic caspase-9/caspase-3 apoptotic pathway. Moreover, σ3 protein can also inhibit the expression of the components of the RNA-sensing retinoic acid-inducible gene (RIG)-like receptor (RLR) signaling pathway to block antiviral type I interferon responses. Mechanistically, σ3 inhibits RIG-I and melanoma differentiation-associated gene 5 expression is independent of its inhibitory effect on MAVS. Overall, we demonstrate that the MRV σ3 protein plays a vital role in negatively regulating the RLR signaling pathway to inhibit antiviral responses. This enables MRV to evade host defenses to facilitate its own replication providing a target for the development of effective antiviral drugs against MRV.IMPORTANCEMammalian orthoreovirus (MRV) is an important zoonotic pathogen, but the regulatory role of its viral proteins in retinoic acid-inducible gene-like receptor (RLR)-mediated antiviral responses is still poorly understood. Herein, we show that MRV σ3 protein co-localizes with mitochondrial antiviral signaling protein (MAVS) in the mitochondria and promotes the mitochondria-mediated intrinsic apoptotic pathway to cleave and consequently degrade MAVS. Furthermore, tryptophan at position 133 of σ3 protein plays a key role in the degradation of MAVS. Importantly, we show that MRV outer capsid protein σ3 is a key factor in antagonizing RLR-mediated antiviral responses, providing evidence to better unravel the infection and transmission mechanisms of MRV.

Abstract Breast cancer (BC) is a prominent cause of cancer incidence and mortality around the world. Disulfidptosis, a type of cell death, can induce tumor cell death. The purpose of this study was to analyze the potential impact of disulfidptosis-related genes (DRGs) on the prognosis and immune infiltration features of BC. Based on DRGs, we conducted an unsupervised clustering analysis on gene expression data of BC in TCGA-BRCA dataset and identified two BC subtypes, cluster1 and cluster2, with cluster1 showing a higher likelihood of favorable survival. Through immune analysis, we found that cluster1 had lower proportions of infiltration in immune-related cells, including aDCs, DCs, NK_cells, Th2_cells, and Treg. Based on the immunophenoscore (IPS) results, we inferred that cluster1 might benefit more from immune checkpoint inhibitors targeting CTLA-4 and PD1. Targeted small molecule prediction results showed that patients with cluster2 BC might respond better to antagonistic small molecule compounds, including clofazimine, lenalidomide, and epigallocatechin. Differentially expressed genes between the two subtypes were found to be enriched in signaling pathways related to steroid hormone biosynthesis, ovarian steroidogenesis, and neutrophil extracellular trap formation, according to enrichment analyses. In conclusion, this study identified BC subtypes based on DRGs so as to help predict patient prognosis and provide valuable tools for guiding clinical management and precise treatment of BC patients.

Abstract Osteosarcoma (OS) is a highly aggressive form of bone cancer that predominantly affects adolescents and young adults. In this study, we have undertaken an investigation into the potential anti-OS cell activity of IMT1 (inhibitor of mitochondrial transcription 1), a first-in-class inhibitor of RNA polymerase mitochondrial (POLRMT). IMT1 exhibited a profound inhibitory effect on cell survival, proliferation, cell cycle progression, and migration in primary and immortalized OS cells. Furthermore, this POLRMT inhibitor elicited apoptosis in the OS cells, without, however, inducing cytotoxicity in human osteoblasts or osteoblastic cells. IMT1 disrupted mitochondrial functions in OS cells, resulting in mitochondrial depolarization, oxidative injury, lipid peroxidation, and ATP reduction in OS cells. Silencing POLRMT using targeted shRNA closely mimicked the actions of IMT1 and exerted potent anti-OS cell activity. Importantly, IMT1’s effectiveness was diminished in POLRMT-silenced OS cells. Subsequent investigations revealed that IMT1 suppressed the activation of the Akt-mammalian target of rapamycin (mTOR) cascade in OS cells. IMT1 treatment or POLRMT silencing in primary OS cells led to a significant reduction in Akt1-S6K-S6 phosphorylation. Conversely, it was enhanced upon POLRMT overexpression. The restoration of Akt-mTOR activation through the introduction of a constitutively active S473D mutant Akt1 (caAkt1) mitigated IMT1-induced cytotoxicity in OS cells. In vivo, oral administration of IMT1 robustly curtailed the growth of OS xenografts in nude mice. Furthermore, IMT1 suppressed POLRMT activity, impaired mitochondrial function, repressed Akt-mTOR activation, and induced apoptosis within xenograft tissues. Collectively, these findings underscore the potent growth-inhibitory effects attributed to IMT1 via targeted POLRMT inhibition. The utilization of this POLRMT inhibitor carries substantial therapeutic promise in the context of OS treatment.

Rare earth and transition metal ion-doped CaZnOS has garnered significant attention for its exceptional mechanoluminescence (ML) performance under mild mechanical stimuli and its capability for multicolor emissions. Since powdered phosphors are not directly usable, they require encapsulation within with polymers to create stable structures. This study investigates Mn2+-doped CaZnOS (CaZnOS:Mn2+) as the ML phosphor, optimizing its performance by varying the Mn2+ content, resulting in bright orange-red emissions from the d-d transitions of the Mn2+ activator. A quantum efficiency of 59.08% was achieved through the self-sensitization of the matrix lattice and energy transfer to the Mn2+ luminescent centers. The enhancement in ML due to Mn2+ doping is attributed to the reduced trap depth and increased trap concentration. Encapsulation with four polymers—PDMS, PU, SIL, and RTV-2—was explored to further optimize ML performance. Among these, PDMS provides the best ML output and sensitivity, owing to its slightly cross-linked structure and good triboelectric properties. The optimized CaZnOS:0.03Mn2+/PDMS composite, featuring excellent flexibility and recoverability, shows great potential for applications in anti-counterfeiting encryption, stress sensors, and wearable devices.

In order to study the microscopic reaction mechanism and kinetic model of Al/PTFE, a reactive force field (ReaxFF) was used to simulate the interface model of the Al/PTFE system with different oxide layer thicknesses (0 Å, 5 Å, 10 Å), and the thermochemical behavior of Al/PTFE at different heating rates was analyzed by simultaneous thermal analysis (TG-DSC). The results show that the thickness of the oxide layer has a significant effect on the reaction process of Al/PTFE. In the system with an oxide layer thickness of 5 Å, the compactness of the oxide layer changes due to thermal rearrangement, resulting in the diffusion of reactants (fluorine-containing substances) through the oxide layer into the Al core. The reaction mainly occurs between the oxide layer and the Al core. For the 10 Å oxide layer, the reaction only exists outside the interface of the oxide layer. With the movement of the oxygen ions in the oxide layer and the Al atoms in the Al core, the oxide layer moves to the Al core, which makes the reaction continue. By analyzing the reaction process of Al/PTFE, the mechanism function of Al/PTFE was obtained by combining the shrinkage volume model (R3 model) and the three-dimensional diffusion (D3 model). In addition, the activation energy of Al/PTFE was 258.8 kJ/mol and the pre-exponential factor was 2.495 × 1015 min−1. The research results have important theoretical significance and reference value for the in-depth understanding of the microscopic chemical reaction mechanism and the quantitative study of macroscopic energy release of Al/PTFE reactive materials.

Lift-off velocity, the wind speed at which a diaspore starts moving during secondary wind dispersal, representing diaspore mobile capacity, may greatly depend on vegetation where dispersal takes place. However, how vegetation structure influences lift-off velocity and which is the most important vegetation attribute remain unknown. The vegetation with various attributes (four coverages-0, 10 %, 20 % and 30 %, three life-form compositions-herb, shrub and herb + shrub, two vertical patterns-one and two layers, and two horizontal patterns-uniform and aggregated) were set up for the wind tunnel experiment. Diaspores of 30 species varied in wing loading, terminal velocity, mass, projected area, length, width, height, and shape index were selected to measure their lift-off velocity within various vegetation structures. Lift-off velocity significantly increased with the increasing vegetation coverage, in the case of any life-form composition, vertical pattern and horizontal pattern of vegetation. For the vertical pattern of vegetation, the lift-off velocity in one-layer vegetation was significantly higher than that in two-layer vegetation, but only for 10 % vegetation coverage. Both the life-form composition and the horizontal pattern of vegetation had no influences on lift-off velocity. The correlation between lift-off velocity and diaspore attributes (length, width, height, projected area, mass and wing loading) increased with vegetation coverage and the correlation in two-layer vegetation was larger than that in one-layer vegetation. Our results indicate that vegetation coverage is the key vegetation attribute determining lift-off velocity during secondary wind dispersal. Lift-off velocity is also influenced by diaspore traits, but their relationships tend to be regulated by vegetation attributes, especially the coverage and vertical pattern of vegetation. Our study demonstrates the important role of vegetation coverage in determining the second dispersal capacity, which is helpful to understand the dispersal strategies of plants in response to vegetation structure and succession.

In the work, high nitrogen γ-Fe based alloy was fabricated by N2 pressure sintering in which the diffused N content could be up to 0.521wt%. Microstructure and mechanical properties were systematically studied. The increased activity of N atom with the added N2 pressure accelerated the generation of MnSiN2, which had an orthorhombic structure of an orientation relation of (11¯1)γ-Fe//(121¯)MnSiN2 with γ-Fe. HRTEM map displayed the incoherent interface of duplex phase on account of a large difference of the crystal plane distance between (11¯1)γ-Fe (d11¯1 = 0.2082 nm) and (121¯)MnSiN2 (d121¯ = 0.2428 nm). Besides, due to the solute N atoms in γ-Fe, an ordered structure with a superlattice structure of selective electron diffraction spots was formed. The elevated nanohardness for austenite grains implied the solution strengthening mechanism of N. The EBSD diagrams showed that the grain sizes of austenite are 9.81, 7.27 and 6.22um respectively, indicating that the formation and precipitation of MnSiN2 hindered the grain growth and led to the finer grain structure with the increased pressure of 0.05Mpa, 0.5Mpa and 1Mpa. The combination of solution strengthening and fine crystal reinforcing was the main reinforcement mechanism which enhanced the hardness austenite grains and wear resistance.

Soil carbon content and stability are primarily influenced by the stabilization of particulate organic matter (POM) and mineral-associated organic matter (MAOM). Despite extensive research on the stabilization processes of POM and MAOM carbon components under various land-use types, the investigation into stabilization processes of soil carbon remains limited in saline–alkali soils. Therefore, we collected soil samples from different positions of saline–alkali drainage ditches at four reclamation times (the first, seventh, fifteenth, and thirtieth year) to determine their carbon content and physicochemical properties. Moreover, POM and MAOM fractions were separated from soil samples, and Fourier transform infrared spectra (FTIR) were used to investigate changes in their chemical composition. The results showed that with increasing reclamation time, the soil total carbon and soil organic carbon (SOC) contents significantly increased from 14 to 15 and 2.9 to 5.5 g kg−1, respectively. In contrast, soil inorganic carbon content significantly decreased from 11 to 9.6 g kg−1. Notably, the changes in soil carbon components following the increasing reclamation time were primarily observed in the furrow sole at a depth of 20–40 cm. While the SOC content of the POM fraction (SOCPOM) decreased significantly, the SOC content of the MAOM fraction (SOCMAOM) increased significantly. These alterations were largely dominated by drainage processes after reclamation instead of a possible conversion from SOCPOM to SOCMAOM. FTIR results revealed that MAOM was greatly influenced by the reclamation time more than POM was, but the change in both POM and MAOM contributed to an increase in soil carbon stability. Our findings will deepen the comprehension of soil carbon stabilization processes in saline–alkali drainage ditches after reclamation and offer a research framework to investigate the stability processes of soil carbon components via alterations in POM and MAOM fractions.

Diaspore dispersal is crucial in shaping plant population dynamics, biodiversity, and ecosystem functions. The effect of the vegetation structure on the secondary wind dispersal of diaspores with different appendage types is not well understood. Using a wind tunnel and a high-definition video camera, we accurately measured the lift-off and dispersal velocities of diaspores from sixteen plant species across six wind velocities (2, 4, 6, 8, 10, and 12 m s−1) under six simulated vegetation structures. Vegetation structure and appendage type were pivotal factors, explaining 41.1% and 42.3% of the variance in lift-off velocity and accounting for 12.0% and 25.3% of the variability in diaspore dispersal velocity, respectively. Vegetation coverage was the main factor influencing near-surface wind velocity, and the lift-off and dispersal velocities of diaspores changed significantly when vegetation coverage exceeded 40%. Diaspores with one wing, having high lift-off velocities and low dispersal velocities, adopt the anti-long-distance wind dispersal strategy, whereas diaspores with pappus, having low lift-off velocities and high dispersal velocities, adopt the long-distance wind dispersal strategy. In contrast, diaspores with thorn, discoid, balloon, and four wings adopt the non-long-distance wind dispersal strategy, suitable for environments such as low-coverage deserts and desert steppes but not high-coverage typical steppes. This study could help comprehend the effect of the vegetation structure on the dispersal process of diaspores, which facilitate habitat restoration and biodiversity conservation of grassland and forest ecosystems.

Recently, the growth of blockchain technology and the economic benefits of cryptocurrencies have led to a proliferation of malicious cryptomining activities on the internet, resulting in significant losses for companies and institutions. Therefore, accurately detecting and identifying these behaviors has become essential. To address low accuracy in detecting and identifying cryptomining behaviors in encrypted traffic, a technique for identifying cryptomining behavior traffic is proposed. This technique is based on the time series characteristics of network traffic and introduces the feature of long-range dependence, and the recognition effect is not easily affected by the encryption algorithm. First, 48-dimensional features are extracted from the network traffic using statistical methods and the rescaled range method, of which 47 dimensions are statistical features and 1 dimension is a long-range dependence feature. Second, because there is much less cryptomining traffic information than normal network traffic information in the dataset, the dataset is processed using oversampling to make the two types of traffic data balanced. Finally, a random forest model is used to identify the type of traffic based on its features. Experiments demonstrate that this approach achieves good detection performance and provides an effective solution for identifying encrypted network traffic with malicious cryptomining behavior. The long-range dependence features introduced therein together with the statistical features describe a more comprehensive flow characteristics, and the preprocessing of the dataset improves the performance of the identification model.

Rice is one of the most important food crops in the world. Rice disease is a major threat to its yield loss. Bacterial blight caused by the gram-negative bacterium Xanthomonas oryzae pv. oryzae (Xoo), is one of the three major diseases that seriously affect rice yield. The traditional chemical control methods have many drawbacks. The cultivation and planting of rice varieties resistant to bacterial blight is recognized worldwide as the most safe and effective means to control the disease. Identification of resistant germplasm resources and discovery of resistant genes are the premise of resistant varieties breeding. Under the pressure of both natural and artificial selections, new pathogenic varieties have emerged one after another due to the constant variation of bacterial blight pathogens, and the main varieties have gradually lost their disease resistance. Therefore, it is necessary to explore and identify more durable disease resistance genes. In the study, 16 bacterial blight resistance genes that have been cloned are briefly classified, and then three genes Xa23, Xa7 and xa5 that are broad-spectrum resistant to bacterial blight with good application are described in detail from the aspects of gene discovery, location, cloning, mechanism of action, and etc. The conventional breeding, molecular marker-assisted selection breeding, transgenic breeding and gene marker breeding for bacterial blight resistance are introduced. In addition, the prospect of molecular breeding for broad-spectrum resistance to bacterial blight is put forward based on the characteristics of different bacterial blight genes and the problems in current breeding and utilization.

Histone H1.2 is a member of the linker histone family, which plays extensive and crucial roles not only in the regulation of chromatin dynamics, cell cycle, and cell apoptosis, but also in viral diseases and innate immunity response. Recently, it was discovered that H1.2 regulates interferon-β and inhibits influenza virus replication, whereas its role in other viral infections is poorly reported. Here, we first found the up-regulation of H1.2 during Encephalomyocarditis virus (EMCV) infection, implying that H1.2 was involved in EMCV infection. Overexpression of H1.2 inhibited EMCV proliferation, whereas knockdown of H1.2 showed a significant promotion of virus infection in HEK293T cells. Moreover, we demonstrated that overexpression of H1.2 remarkably enhanced the production of EMCV-induced type I interferon, which may be the crucial factor for H1.2 proliferation–inhibitory effects. We further found that H1.2 up-regulated the expression of the proteins of the MDA5 signaling pathway and interacted with MDA5 and IRF3 in EMCV infection. Further, we demonstrated that H1.2 facilitated EMCV-induced phosphorylation and nuclear translocation of IRF3. Briefly, our research uncovers the mechanism of H1.2 negatively regulating EMCV replication and provides new insight into antiviral targets for EMCV.

Extensive drainage ditches are constructed to reduce soil salinity in reclaimed saline–alkali farmland, consequently forming plant growth hotspots and impacting soil carbon stocks therein. However, the investigation into changes in soil carbon stocks remains limited in these ditches. To address this, soil samples were collected from drainage ditches, which originated from the reclamation of saline–alkali farmland, at different reclamation years (the first, seventh, and fifteenth year). Moreover, fractions were separated from soil samples; a particle size separation method (particulate organic matter, POM; mineral–associated organic matter, MAOM) and a spatio–temporal substitution method were conducted to analyze the variations in soil carbon components and the underlying mechanisms. The results indicate that there were no significant variations in the contents and stocks of soil organic carbon (SOC) and soil inorganic carbon (SIC) following the increase in reclamation time. However, in the POM fraction, the SOC content (SOCPOM) and stock significantly decreased from 2.24 to 1.12 g kg−1 and from 19.02 to 12.71 Mg ha−1, respectively. Conversely, in the MAOM fraction, the SOC content (SOCMAOM) and stock significantly increased from 0.65 to 1.70 g kg−1 and from 5.30 to 12.27 Mg ha−1, respectively. The different changes in SOCPOM and SOCMAOM, as well as the result of the structural equation model, showed a possible transformation process from SOCPOM to SOCMAOM in the soil carbon pool under the driving force of reclamation time. The results in terms of the changes in soil carbon components demonstrate the stability rather than the stock of the soil carbon pool increase in coastal saline–alkali ditches following the excavation formation time. Although more long time series and direct evidence are needed, our findings further provide a case study for new knowledge about changes in the soil carbon pool within saline–alkali ditches and reveal the potential processes involved in the transformation of soil carbon components.

Covalent organic frameworks (COFs) have gained considerable attention due to their highly conjugated π-skeletons, rendering them promising candidates for the design of light-emitting materials. In this study, we present two vinylene-linked COFs, namely, VL-COF-1 and VL-COF-2, which were synthesized through the Knoevenagel condensation of 2,4,6-trimethyl-1,3,5-triazine with terephthalaldehyde or 4,4′-biphenyldicarboxaldehyde. Both VL-COF-1 and VL-COF-2 exhibited excellent chemical and thermal stability. The presence of vinylene linkages between the constituent building blocks in these COFs resulted in broad excitation and emission properties. Remarkably, the designed VL-COFs demonstrated bright emission, fast fluorescence decay, and high stability, making them highly attractive for optoelectronic applications. To assess the potential of these VL-COFs in practical devices, we fabricated white-light-emitting diodes (WLEDs) coated with VL-COF-1 and VL-COF-2. Notably, the WLEDs coated with VL-COF-1 achieved high-quality white light emission, closely approximating standard white light. The promising performance of VL-COF-coated WLEDs suggests the feasibility of utilizing COF materials for stable and efficient lighting applications.

Objectives: This study aims to explore the associations of personality traits and extra-family social relationship with depressive symptoms among Chinese adults.Methods: A nationally representative sample of 29,810 adults aged 16 and above were selected from 2018 CFPS. Personality and depressive symptoms were measured using CBF-PI-15 and the CES-D8 scale. Extra-family social relationship was assessed through the self-rated evaluation. The multiple regression analysis and the PROCESS macro were used for the mediation analysis.Results: Extraversion (OR = 0.807, 95% CI = 0.773, 0.842), agreeableness (OR = 0.795, 95% CI = 0.756, 0.835) and extra-family social relationship (OR = 0.927, 95% CI = 0.913, 0.941) had negative associations with depressive symptoms. Extra-family social relationship could mediate between extraversion and depressive symptoms (Indirect effect = −0.049,95% CI = −0.060, −0.039) as well as agreeableness (Indirect effect = −0.056, 95% CI = −0.068, −0.046) and depressive symptoms. Comparing to females, the indirect effect accounts for a higher proportion of total effect in males.Conclusion: Extra-family social relationship might mediate the association between extraversion and depressive symptoms as well as agreeableness and depressive symptoms.

Pseudorabies virus (PRV) is an agent of Aujeszky's disease, and causes great economic losses to pig farming. Re-outburst of pseudorabies implies that new control measures are urgently needed. We show here that DDX56 possesses the ability to inhibit PRV replication in vitro, which may be an important factor for PRV infection. Overexpression of DDX56 inhibited PRV genomic DNA transcription and lower titers of PRV infection in PK15 cells, whereas down-regulation of the DDX56 expression had a promotion role on virus replication. Further study demonstrated that DDX56 exerted its proliferation-inhibitory effects of PRV through up-regulating cGAS-STING-induced IFN-β expression. Moreover, we found that DDX56 could promote cGAS expression and direct interaction also existed between DDX56 and cGAS. Based on this, DDX56-regulated IFN-β pathway may be targeted at cGAS. To verify this, down-regulated cGAS expression in DDX56 over-expression cells was performed. Results indicated that knockdown of cGAS expression could abrogate the inhibition role of DDX56 on PRV proliferation and weaken the effect of DDX56 on IFN-β expression. In addition, DDX56 played a promotion role in IRF3 phosphorylation and nucleus translocation. Altogether, our results highlight DDX56's antiviral role in PRV infection, and our findings contribute to a better understanding of host factors controlling PRV replication.

Abstract Background Most Chinese migrants have been faced with obstacles to getting access to local public health services. Young migrants among internal migrants make a major contribution to the economy. However, the establishment of their health records has been ignored. This research was aimed at exploring the status and associated factors of the establishment of health records among young Chinese migrants. Methods Data were obtained from the 2017 China Migrants Dynamic Survey (CMDS). Chi-square test and binary logistic regression were performed to investigate the associated factors of the establishment of health records among young Chinese migrants. Results Approximately 30.2% of young migrants had their health records established in inflow communities. Urban residence, medical insurance (insured), and lower average monthly household income were attributed to the establishment of more health records. Young migrants who were in northeast China and across provinces and immigrated for working or engaging in trade were less likely to have health records established. Young migrants who participated in social activities and public affairs activities and took type of people in touch as natives in the inflow area showed a higher possibility of establishing health records. Meanwhile, receiving health education and hearing about national basic public health services (BPHSs) were beneficial for establishing the health records of more young migrants. Conclusion This study showed that the health records of young migrants had a relatively low establishment rate. Sociodemographic and migration characteristics, health status, public health services, and social integration factors were all related to the health record establishment of young Chinese migrants.

Interferon (IFN) helps cells fight viral infections by further inducing the expression of many downstream IFN-stimulated genes (ISGs). Human interferon-inducible transmembrane proteins (IFITM) are one of these ISGs. The antiviral function of human IFITM1, IFITM2, and IFITM3 are well known. In this study, we report that IFITM can significantly inhibit EMCV infectivity in HEK293 cells. Overexpression of IFITM proteins could promote IFN-β production. Meanwhile, IFITMs facilitated type I IFN signaling pathway adaptor MDA5 expression. We detected the binding of IFITM2 to MDA5 in a co-immunoprecipitation assay. It was also found that the ability of IFITM2 to activate IFN-β was significantly inhibited after interfering with MDA5 expression, suggesting that MDA5 may play an important role in the activation of the IFN-β signaling pathway by IFITM2. Moreover, the N-terminal domain plays an active role in the antiviral activity and the activation of IFN-β by IFITM2. These findings suggest that IFITM2 plays a vital role in antiviral signaling transduction. In addition, a positive feed-forward loop between IFITM2 and type I IFN establishes a key role for IFITM2 in enforcing innate immune responses.

Sarcophaga tsinanensis (Fan, 1964) (Diptera: Sarcophagidae), a species from subgenus Heteronychia. In this study, we report the complete mitochondrial genome (mitogenome) of S. tsinanensis. The length of this mitogenome was 14,972 bp in total, containing 13 protein-coding genes, 2 ribosomal RNAs, 22 transfer RNAs, and a non-coding control region. The arrangement of genes was the same as that of ancestral metazoan. Sarcophaga tsinanensis has a high nucleotide bias in A/T accounting for 76.80% of total nucleotides (A 39.9%, G 9.2%, C 13.9%, and T 36.9%). The result of phylogenetic analysis revealed that S. tsinanensis cluster together with species from the same subgenus Heteronychia, showing a clear monophyletic relationship. Sarcophaga tsinanensis is closely related to its sister species Sarcophaga depressifrons, Sarcophaga plotnikovi, and Sarcophaga shnitnikovi. This study provides the mitochondrial data of S. tsinanensis for further research on evolutionary relationships and species identification of flesh flies.

Lucilia shenyangensis Fan, 1965 (Diptera: Calliphoridae) is of potential importance in epidemiology, veterinary medicine, and forensic entomology due to their necrophilous habit and behaviors associated with mammals. In this study, we report the complete mitochondrial genome (mitogenome) of L. shenyangensis. The mitogenome is 14,989 bp in length, comprising 13 protein-coding genes (PCGs), two ribosomal RNAs (rRNAs), 22 transfer RNAs (tRNAs), and a non-coding control region. The arrangement of genes is identical to that of the ancestral metazoan. Nucleotide composition revealed a high A/T bias, accounting for 76.50% total mitogenome nucleotides (A 39.2%, G 9.6%, C 14.0%, T 37.3%). Phylogenetic analysis indicated that L. shenyangensis was clear separated from other blow flies and emerged as the sister lineage to the rest species from genus Lucilia (L. illustris, L. sericata, L. coeruleiviridis, and L. porphyrina). The mitogenome data of L. shenyangensis could facilitate further evolutionary genetic researches on blow flies.

Wear and corrosion resistant austenitic cast irons were prepared by powder metallurgy (P/M). In this system, equivalent Mn were doped to replace nickel with a constant Nieq (the Ni equivalent number) to reduce the cost. Conventional high nickel cast iron was employed as a reference in this study. The hardness, dry sliding wear behavior and corrosion behavior of conventional and P/M cast iron were investigated under the same conditions. The result demonstrates that a little difference in hardness is found between Mn0-C (Mn-0wt% prepared by cast) and Mn0-PM (Mn-0wt% prepared by P/M). However, the wear resistance of Mn0-PM is better than Mn0-C due to the evenly distributed lubricating graphite. The wear resistance of P/M cast iron increases with the increase of Mn content which is attributed to the increased hardness. The corrosion behaviors for all the materials tested by using potentiodynamic polarization curves and electrochemical impedance spectroscopy (EIS) are almost the same, largely due to the formation of austenite matrix. Thereinto, Mn5-PM holds the best corrosion behavior with the lowest corrosion current density and highest polarization resistance. Because the potential difference between matrix and carbide is lower than that between matrix and graphite. The decreased graphite with the increased carbide induces a strong corrosion resistance in Mn5-PM.

Abstract The subspace technique has been widely used to solve unconstrained/constrained optimization problems and there exist many results that have been obtained. In this paper, a subspace algorithm combining with limited memory BFGS update is proposed for large-scale nonsmooth optimization problems with box-constrained conditions. This algorithm can ensure that all iteration points are feasible and the sequence of objective functions is decreasing. Moreover, rapid changes in the active set are allowed. The global convergence is established under some suitable conditions. Numerical results show that this method is very effective for large-scale nonsmooth box-constrained optimization, where the largest dimension of the test problems is 11,000 variables.

The aim of this study was to investigate the extraction of antibacterial substances from Galla chinensis (ASGC) that act against the aquatic pathogen Vibrio harveyi, to identify the chemical composition and explore the mechanism of action. The optimal extraction process was as follows: anhydrous methanol, liquid/material ratio (mL/g) 26:1, extraction temperature 98 °C and extraction time 157 min. The diameter of the inhibition zones (21.31 ± 0.51 mm) under the optimal conditions agreed with the predicted value by experimental rechecking. The yield of ASGC was 39.39% ± 0.69%, and two compounds including gallic acid (GA) and penta-O-galloyl-β-d-glucose (5GG) were identified in ASGC. The minimal inhibitory concentration (MIC) values of ASGC and GA were 0.50 and 0.25 mg/mL, respectively. Meanwhile, 5GG and other chemical constituents did not exert antibacterial activities against V. harveyi (MICs > 1 mg/mL). Further investigation of the mechanisms of action of ASGC and GA against V. harveyi revealed their strong destructive effects on the growth of bacterial cells, the integrity and permeability of the cell membrane, and morphological alterations. The report provides a favorable prospect of GA as a potential control agent against V. harveyi in aquaculture.

Background The impact of perceived life stress on subjective well-being has been well-established; while few studies have explored the mediating and moderating mechanisms of the association between perceived life stress and subjective well-being among perimenopausal women. This study is aimed at exploring the mediating effect of depressive symptoms and the role of interests/hobbies as a moderator in the association between perceived life stress and subjective well-being among perimenopausal women. Methods The participants were 1,104 perimenopausal women at the age of 40 to 60, who were asked to complete a paper-based questionnaire. A single item was used to measure self-perceived life stress and interests/hobbies. The Zung Self-rating Depression Scale (SDS) and Subjective Well-being Scale for Chinese Citizens (SWBS-CC) were applied to assess both depressive symptoms and subjective well-being. Multiple linear regression analysis and the PROCESS macro were adopted to analyse not only the mediating effect of depressive symptoms but also the moderating role of interests/hobbies. Heat shock proteins (HSPs) are a protein family that respond to physiological stress, such as heat, starvation, and infection. As cellular protein chaperones, they play an important role in protein folding, assembly, and degradation. Though it is well known that HSP27 is involved in a range of viral infections, its role during an encephalomyocarditis virus (EMCV) infection is not known. Here, we report that EMCV degrades HSP27 and that EMCV proteins 2Cpro and 3Apro are primarily responsible for its degradation. Consequently, loss of cellular HSP27 augmented EMCV proliferation, an effect that could be reversed upon HSP27 overexpression. Importantly, we found that HSP27 positively regulated EMCV-triggered type I interferon (IFN) production. Moreover, overexpression of 2Cpro and 3Apro significantly blocked type I IFN production. We also found for the first time that HSP27, as a molecular chaperone, can specifically interact with MDA5 and stabilize the expression of MDA5. Collectively, this study shows that HSP27 dampens EMCV infectivity by positively regulating EMCV-triggered retinoic acid-inducible gene (RIG)-I-like receptor (RLR)/melanoma differentiation-associated gene 5 (MDA5) signal pathway, while EMCV proteins 2Cpro and 3Apro interact with HSP27 and degrade HSP27 protein expression to allow EMCV proliferation. Our findings provide further mechanistic evidence for EMCV partaking in immune escape mechanisms, and that 2Cpro and 3Apro could serve as potential antiviral targets.

To determine the mechanism of penetration of multi-layer aluminum targets (MLAT) by a reactive materials projectile (RMP), AUTODYN-3D numerical simulations and experimental tests were carried out. The Powder Burn equation of the state ignition model was introduced for the reactive core activation under different projectile–target interaction conditions, which effectively simulated the deflagration reaction damage effects behavior of the RMP and the damage evolution behavior of the MLAT. The activation rate of the reactive core increased significantly when the thickness of the steel target was 8–15 mm; a significant combined destructive effect of kinetic and chemical energy was produced on the MLAT. The initial velocity was proportional to the penetration and destruction effect of the front-layer aluminum target. For the rear-layer aluminum target, the detonation damage showed a tendency to increase and then decrease. If the head metal block was too thick, the penetration ability would be improved at the same time, and the deflagration reaction damage effects ability of the steel target would be significantly reduced. In order to achieve good battlefield damage efficacy, all of the influencing factors should be comprehensively considered.

Pseudorabies virus is a typical swine alphaherpesvirus, which can cause obvious neurological disorders and reproductive failure in pigs. It is capable of evading host antiviral immune response. However, the mechanism by which many PRV proteins assist the virus to evade innate immunity is not fully understood. This study identified PRV US3 protein as a crucial antagonistic viral factor that represses interferon beta (IFN-β) expression. A in-depth study showed that US3 protein restricted type I IFN production by targeting interferon regulatory factor 3 (IRF3), a key molecule required for type I IFN induction. Additionally, US3 protein interacted with IRF3, degraded its protein expression to block the phosphorylation of IRF3. These findings suggested a novel strategy utilized by PRV to inhibit IFN-β production and escape the host innate immunity.

The Mo-12Si-8.5B alloy was surface-remelted by laser and electron beam, and the microstructure of its melt pool and substrate regions were analyzed by scanning electron microscopy (SEM), X-ray diffraction (XRD), and energy spectrometry (EDS) techniques. It was found that the composition of the surface phases in the Mo-12Si-8.5B alloy did not change by the high-energy beam surface remelting process, but the microstructure of the molten pool region was significantly different from that of the substrate region, and its phase distribution was more uniform. Dendrites appeared on the surface of the material under the action of both processes, and the Si- and B-rich phases were mainly gathered in the interdendritic region. In the melt pool of the laser-remelted specimens, the α-Mo phase was continuously distributed with an average dendrite length of 70 µm, while the α-Mo phase distribution in the melt pool of the electron beam remelted specimens were relatively concentrated, with a larger dendrite size and an average dendrite length of 120 µm. The dendrite size in the melt pool of the laser remelted material was smaller, and the distribution of the elements was relatively uniform. Using a laser beam as the heat source was more favorable for the next step of the additive manufacturing of the core parts of hypersonic vehicles.

Pseudorabies (PR) is a domestic and wild animal infectious disease caused by the pseudorabies virus (PRV) and is one of the major infectious diseases that endanger the global swine industry. Studies have reported that PRV may achieve cross-species transmission from pigs to humans in recent years. Therefore, in-depth exploration of the relationship between PRV and host proteins is of great significance for elucidating the pathogenic mechanism of PRV and anti-PRV infection. Here, we report that heat shock protein 27 (HSP27) ubiquitinates and degrades cyclic GMP-AMP synthase (cGAS) and attenuates cGAS-mediated antiviral responses, thereby promoting PRV infection. Overexpression of HSP27 promoted PRV proliferation in vitro, while knockdown of HSP27 inhibited PRV infection. Importantly, we found that HSP27 inhibited PRV infection or poly(dA:dT)-activated IFN-β expression. Further studies found that HSP27 may inhibit cGAS-STING-mediated IFN-β expression through targeting cGAS. In addition, we found that HSP27 can suppress the expression of endogenous cGAS in different cells at both gene transcription and protein expression levels, and that HSP27 interacts with and ubiquitinates cGAS. In conclusion, we reveal for the first time that HSP27 is a novel negative regulator of the cGAS-STING signaling pathway induced by PRV infection or poly(dA:dT) activation and demonstrate that HSP27 plays a crucial role in PRV infection.

Abstract Background Parkinson's disease is a progressive degenerative nervous system disease. Recent studies have shown that secondary changes in the GABA system play directly affect the pathogenesis of PD. There is still much debate about GABA concentrations because currently, GABA concentrations in the brain tissue are obtained indirectly by measuring its concentration in the blood and cerebrospinal fluid. These results are unreliable. Magnetic resonance spectroscopy (MRS) is the only noninvasive method for evaluating the concentration of metabolites in living brain tissue and has been widely applied in research and clinical practice. In addition, combining MEGA-PRESS technology with LCModel software for quantitative GABA measurements is largely recognized. At present, the PD monkeys model in primates has been increasingly proficient. Primates are more similar to humans in terms of brain structure and function than other animals. However, 3.0 T MRS studies involving the PD monkey model to measure metabolites in living subjects with PD are still rare. The study was performed at 3.0 T MRI with control monkeys and PD monkeys that were injected methyl-phenyl-tetrahydropyridine (MPTP) in one side of common carotid artery before and 3 months after successful model establishment to measure GABA concentrations in the bilateral striatum. Behavioral observations were performed for all animals, and the behavioral score was recorded. After 3 months, the GABA concentration in the bilateral striatum was measured in both groups by high-performance liquid chromatography (HPLC). The data obtained from magnetic resonance spectroscopy (MRS) were compared with the actual measured GABA concentrations in tissues isolated from the corresponding regions, and their correlations with the behavior score were analyzed. The research objectives are to investigate the changes of γ-aminobutyric acid (GABA) concentration in the bilateral striatum of monkeys with Parkinson's disease (PD) and the value of quantitatively measuring its concentration by noninvasive 3.0 T spectroscopy. Results (1) The MRS results showed that the GABA concentration in the injured side of the striatum of the PD monkeys was higher than in the contralateral side, but the difference was not statistically significant (P = 0.154). To recognize the tender shoots for high-quality tea and to determine the picking points accurately and quickly, this paper proposes a method of recognizing the picking points of the tender tea shoots with the improved YOLO-v3 deep convolutional neural network algorithm. This method realizes the end-to-end target detection and the recognition of different postures of high-quality tea shoots, considering both efficiency and accuracy. At first, in order to predict the category and position of tender tea shoots, an image pyramid structure is used to obtain the characteristic map of tea shoots at different scales. The residual network block structure is added to the downsampling part, and the fully connected part is replaced by a 1 × 1 convolution operation at the end, ensuring accurate identification of the result and simplifying the network structure. The K-means method is used to cluster the dimension of the target box. Finally, the image data set of picking points for high-quality tea shoots is built. The accuracy of the trained model under the verification set is over 90%, which is much higher than the detection accuracy of the research methods.

The electronic components inside a main battle tank (MBT) are the key components for the tank to exert its combat effectiveness. However, breakdown of the inner electronic components can easily occur inside the MBT due to the strong transient shock and large vibration during artillery fire. As a typical key electronic component inside an MBT, the fault mechanism and fault patterns of the CPU board of the fire control computer (FCC) are discussed through numerical simulation and experimental research. An explicit nonlinear dynamic analysis is performed to study the vibration features and fault mechanism under instantaneous shock load. By using finite element modal analysis, the first six nature frequencies of the CPU board are calculated. Meanwhile, curves of stress–frequency and strain–frequency of the CPU board under different harmonic loads are obtained, which are applied to further identify the peak response of the structure. Validation of the finite element model and simulation results are performed by comparing those obtained from the modal with experiments. Based on the dynamic simulation and experimental analysis, fault patterns of CPU board are discussed, and some optimization suggestions were proposed. The results shown in this work can provide a potential technical basis and reference for the optimization design of the electronic components that are commonly used in the modern weapon equipment and wartime support.

In this paper, we present the design of an integrated temperature and strain dual-parameter sensor based on surface acoustic waves (SAWs). First, the COMSOL Multiphysics simulation software is used to determine separate frequencies for multiple sensors to avoid interference from their frequency offsets caused by external physical quantity changes. The sensor consists of two parts, a temperature-sensitive unit and strain-sensitive unit, with frequencies of 94.97 MHz and 90.05 MHz, respectively. We use standard photolithography and ion beam etching technology to fabricate the SAW temperature–strain dual-parameter sensor. The sensing performance is tested in the ranges 0–250 °C and 0–700 μԑ. The temperature sensor monitors the ambient temperature in real time, and the strain sensor detects both strain and temperature. By testing the response of the strain sensor at different temperatures, the strain and temperature are decoupled through the polynomial fitting of the intercept and slope. The relationship between the strain and the frequency of the strain-sensitive unit is linear, the linear correlation is 0.98842, and the sensitivity is 100 Hz/μԑ at room temperature in the range of 0–700 μԑ. The relationship between the temperature and the frequency of the temperature-sensitive unit is linear, the linearity of the fitting curve is 0.99716, and the sensitivity is 7.62 kHz/°C in the range of 25–250 °C. This sensor has potential for use in closed environments such as natural gas or oil pipelines.

Oscillatory pressure sintering is a novel crystal refinement technology. The doping of different concentrations of ZrB2 under oscillatory sintering technology (9 Hz) is discussed here, focusing on its macroscopic mechanics and oxidation resistance. In particular, doping 2.5 wt% ZrB2 can effectively increase the hardness of the alloy, slightly increase the fracture toughness of the alloy and have an outstanding effect on the oxidation resistance of the alloy at 1300 °C, achieving the effect of reducing mass loss by 80.3%.

Although the ignition-and-growth model can simulate the ignition and detonation behavior of traditional energy materials well, it seems insufficient to simulate the impact-induced deflagration behavior of reactive materials (RMs) using current finite element codes due to their more complicated ignition threshold and lower reaction rates. Therefore, a simulation method for the impact-induced deflagration behavior of a reactive materials projectile (RMP) is developed by introducing tunable ignition threshold conditions for RMs, and a user-defined subroutine is formed by the secondary development on the equation of state (EOS). High-velocity impact experiments were performed to prove the validity of simulations. The results show that the user-defined subroutine for RMs is competent in simulating the ignition and deflagration behavior under impact conditions, because the reaction ratio, morphology and temperature distribution of RMP fragments are all well consistent with experiments, theory, and current reports from other researchers. In this way, the quantitative study on the deflagration reaction of RMs can be implemented and relevant mechanisms are revealed more clearly.

Objectives The study aimed to explore the association of coping styles and social support with psychological distress among patients with end-stage renal disease (ESRD) as well as gender differences in these associations. Methods A cross-sectional study of 769 ESRD patients was conducted at 25 hospitals in Qiqihar City, China. All information was collected using structured questionnaires that were self-administered by the patients. Patients’ psychological health status, coping styles and perceived social support were measured using the 12-item General Health Questionnaire, the Medical Coping Modes Questionnaire and the Multidimensional Scale of Perceived Social Support. Student’s t test, analysis of variance (ANOVA) and binary logistic regression analysis were used for data analysis. Results A total of 72.3% of participants reported psychological distress, and the prevalence of distress was higher in females (77.0%) than in males (68.8%). Encephalomyocarditis virus (EMCV) causes encephalitis, myocarditis, neuropathy, reproductive disorders, and diabetes in animals. EMCV is known to induce cell autophagy; however, the molecular mechanisms underlying this remain unclear. Here, we show that the type III-transmembrane protein, transmembrane protein 39A (TMEM39A), plays a critical role in EMCV replication. We showed that EMCV GS01 strain infection upregulated TMEM39A expression. Importantly, EMCV induced autophagy in a range of host cells. The autophagy chemical inhibitor, 3-MA, inhibited EMCV replication and reduced TMEM39A expression. This is the first study demonstrating TMEM39A promoting the replication of EMCV via autophagy. Overall, we show that TMEM39A plays a positive regulatory role in EMCV proliferation and that TMEM39A expression is dependent on the autophagy pathway.

Vernonia amygdalina Delile is widely used in folkloric medicine for the treatment of various diseases. In this study, the complete chloroplast genome of V. amygdalina Delile was reported, which was assembled and annotated base on genome high-throughput sequencing data. This work provided the clues for the taxonomy of the herb and the potential to utilize the chloroplast genome sequence as a new study target. The length of V. amygdalina Delile chloroplast genome was 153,133bp, with two single-copy regions, each has the length of 84,245bp and 13,152bp respectively. This region were separated by two inverted repeat regions with 27,868bp in length. It was predicted to consist of 131 genes in total, including 86 protein-coding genes, 37 tRNA genes, 8 rRNA genes with GC content at 37.68%. Phylogenetic analysis by RAxML (Random Axelerated Maximum Likelikhood) showed V. amygdalina Delile is closest to Sonchus webbii in Compositae.

Molybdenum silicon boron alloy is regarded as the next generation of superalloy that is expected to replace nickel-based superalloys. However, the high-temperature oxidation resistance of Mo-Si-B alloy has always been an issue worth studying. In this study, Mo-9Si-8B alloy was prepared via a plasma oscillatory pressure sintering process and pre-oxidized at 1300 °C while maintaining a certain balance of mechanical and oxidation properties. The influence of the oxide protective layer on its performance at high temperature of 1150 °C was explored, the micro-mechanism of its performance and its failure mode of the hinge-locking mechanism was illustrated, and finally, its oxidation kinetics was inferred. In conclusion, pre-oxidized Mo-9Si-8B (at.%) alloy did play a role in delaying the oxidation process during the initial period of cyclic oxidation. However, with the increase of cyclic oxidation time, the improvement of high-temperature oxidation resistance was limited.

Abstract In this paper, a modified BFGS algorithm is proposed for unconstrained optimization. The proposed algorithm has the following properties: (i) a nonmonotone line search technique is used to obtain the step size α k $\alpha_{k}$ to improve the effectiveness of the algorithm; (ii) the algorithm possesses not only global convergence but also superlinear convergence for generally convex functions; (iii) the algorithm produces better numerical results than those of the normal BFGS method.

The present case study investigated university students’ academic and linguistic gains during a semester-long exchange program abroad. Thirty three third-year English majors from a Chinese university answered a battery of questionnaires and 13 of them participated in semi-structured interviews both prior to and after the program. Analyses of the data showed that the participants gained greatly from the exchange program both academically and linguistically. Based on the findings, some implications about exchange programs are discussed.

It is generally acknowledged that the conjugate gradient (CG) method achieves global convergence--with at most a linear convergence rate--because CG formulas are generated by linear approximations of the objective functions. The quadratically convergent results are very limited. We introduce a new PRP method in which the restart strategy is also used. Moreover, the method we developed includes not only n-step quadratic convergence but also both the function value information and gradient value information. In this paper, we will show that the new PRP method (with either the Armijo line search or the Wolfe line search) is both linearly and quadratically convergent. The numerical experiments demonstrate that the new PRP algorithm is competitive with the normal CG method.

A limited memory BFGS (L-BFGS) algorithm is presented for solving large-scale symmetric nonlinear equations, where a line search technique without derivative information is used. The global convergence of the proposed algorithm is established under some suitable conditions. Numerical results show that the given method is competitive to those of the normal BFGS methods.

A series of C2-symmetric chiral benzimidazolium salts, the precursor of N-heterocyclic carbene ligands, were designed and synthesized from 1,2-dibromobenzene. In situ prepared corresponding carbenes were tested in the asymmetric palladium-catalyzed intramolecular α-arylation of amides, affording chiral diarylmethanols with high yields and moderate enantioselectivities.
