Abstract Quaternary climate changes are driven in part by variations in the distribution and strength of insolation due to orbital parameters. Continental climate variability is well documented for the most recent glacial-interglacial cycles, yet few records extend further back in time. Such records are critically needed to comprehensively assess the entire spectrum of natural climate variability against the backdrop of anthropogenic warming. Here, we apply uranium isotope geochronology to calcite deposits to date groundwater-table changes in Devils Hole cave, Nevada. The deposits record multi-meter groundwater-table fluctuations over the last 750,000 years, reflecting the long-term evolution of hydroclimate in this presently arid region. During periods between glacial or interglacial extremes, the water table responded sensitively to variations in 65°N summer insolation, likely caused by the increasing extent of North American ice sheets during cold period, which steered moisture-laden trajectories towards the southwestern USA. These orbitally-driven hydroclimatic changes are superimposed on a tectonically-driven long-term decline in the regional groundwater table observed prior to 438,000 ± 14,000 years ago.

Abstract The objectives of this study were to explore the changes in soil stoichiometry and enzyme activities at different distances from an opencast coal mine in the Hulun Buir Grassland of China. Four transects were established on north and east sides of the opencast coal mining area, and samples were collected at 50 m, 550 m, and 1550 m from the pit on each transect. Control samples were collected from a grassland station 8 km from the opencast coal mining area that was not disturbed by mining. Four replicate soil samples were collected at each point on the four transects. Soil physicochemical properties and enzyme activities were determined, and correlations between soil properties and stoichiometric ratios and enzyme activities were explored using redundancy analysis. The increase in distance from mining did not significantly affect soil properties, although soil urease activity was significantly lower than that of the control area. Soil properties 1550 m from the mine pit were similar to those at the grassland control. In addition, soil total nitrogen had the greatest effect on soil stoichiometry, and soil total potassium had the greatest effect on soil enzyme activities. Coal dust from opencast mining might be the main factor affecting soil stoichiometry and enzyme activities. The results of this study provide direction for the next step in studying the influence of mining areas on soil properties and processes.

With the gradual expansion of mining scale in open-pit coal mines, slope safety problems are increasingly diversified and complicated. In order to reduce the potential loss caused by slope sliding and reduce the major threat to the safety of life and property of residents in the mining area, this study selected two mining areas in Xinjiang as cases and focused on the relationship between phase noise and deformation. The study predicts the specific time point of slope sliding by analyzing the dynamic history correlation tangent angle between the two. Firstly, the time series data of the micro-variation monitoring radar are used to obtain the small deformation of the study area by differential InSAR (D-InSAR), and the phase noise is extracted from the radar echo in the sequence data. Then, the volume of the deformation body is calculated by analyzing the small deformation at each time point, and the standard deviation of the phase noise is calculated accordingly. Finally, the sliding time of the deformation body is predicted by combining the tangent angle of the ratio of the volume of the deformation body to the standard deviation of the phase noise. The results show that the maximum deformation rates of the deformation bodies in the studied mining areas reach 10.1 mm/h and 6.65 mm/h, respectively, and the maximum deformation volumes are 2,619,521.74 mm3 and 2,503,794.206 mm3, respectively. The predicted landslide time is earlier than the actual landslide time, which verifies the effectiveness of the proposed method. This prediction method can effectively identify the upcoming sliding events and the characteristics of the slope, provide more accurate and reliable prediction results for the slope monitoring staff, and significantly improve the efficiency of slope monitoring and early warning.

Abstract Qunf Cave oxygen isotope (δ18Oc) record from southern Oman is one of the most significant of few Holocene Indian summer monsoon cave records. However, the interpretation of the Qunf δ18Oc remains in dispute. Here we provide a multi-proxy record from Qunf Cave and climate model simulations to reconstruct the Holocene local and regional hydroclimate changes. The results indicate that besides the Indian summer monsoon, the North African summer monsoon also contributes water vapor to southern Oman during the early to middle Holocene. In principle, Qunf δ18Oc values reflect integrated oxygen-isotope fractionations over a broad moisture transport swath from moisture sources to the cave site, rather than local precipitation amount alone, and thus the Qunf δ18Oc record characterizes primary changes in the Afro-Asian monsoon regime across the Holocene. In contrast, local climate proxies appear to suggest an overall slightly increased or unchanged wetness over the Holocene at the cave site.

Abstract We present a speleothem record from western Cuba, spanning the period 98.7–84.9 ka BP. Our record shows two distinctive periods of high δ18O corresponding to dry and/or cold periods during 85–87.6 and 90.2–93.1 ka BP, synchronous with Heinrich events 8 and 9 (H8 and H9). Hence, we provide the first proxy evidence of the local Caribbean climate response to H8 and H9. Interestingly, H8 is more pronounced compared to H9, which may be a local response to lower temperatures in the North Atlantic resulting in a weak AMOC and reduced deep water formation, therefore a stronger south shift of the ITCZ. Our data complement existing speleothem records from western Cuba which, collectively, provide a nearly continuous paleoclimate time-series spanning the last 100 ka BP, indicating a consistent response to millennial-scale events as dry and/or cooler conditions. The comparison with regional paleoclimate records reveals an anti-phased relationship with South America, caused by the southern movements of the ITCZ during millennial-scale events which lead to dry conditions in the Caribbean and a stronger South American Monsoon System.

Abstract Infrared imaging technology is widely used in the fault diagnosis of power equipment. The power equipment images obtained by infrared thermal cameras generally have the characteristics of low resolution, low contrast, similar geometric features, and complex image backgrounds. The traditional infrared image detection methods have problems of low detection accuracy, low real‐time performance, and missed and fault detection, all of which will lead to great difficulties in the automatic identification process of infrared images of power equipment. This paper adopted an algorithm based on You Only Look Once (YOLO) v4 to realize the detection of electrical equipment under infrared images. First of all, the authors utilized the Fast Super‐Resolution Convolutional Neural Network (FSRCNN) algorithm to perform super‐resolution reconstruction on the infrared images of power equipment to achieve image enhancement. Moreover, the lightweight network Mobilenetv3 was selected as the backbone network of YOLOv4 to improve the running speed by reducing network parameters. Finally, the performance of the YOLOv4 algorithm was compared to verify the effectiveness of the algorithm proposed in this paper. The experimental results showed that the proposed method had strong generalization ability and reliable detection of infrared targets of various types of power equipment, with the mean average precision (mAP) reaching 91.3%.

Abstract Object Early‐life neglect has irreversible emotional effects on the central nervous system. In this work, we aimed to elucidate distinct functional neural changes in medial prefrontal cortex (mPFC) of model rats. Methods Maternal separation with early weaning was used as a rat model of early‐life neglect. The excitation of glutamatergic and GABAergic neurons in rat mPFC was recorded and analyzed by whole‐cell patch clamp. Results Glutamatergic and GABAergic neurons of mPFC were distinguished by typical electrophysiological properties. The excitation of mPFC glutamatergic neurons was significantly increased in male groups, while the excitation of mPFC GABAergic neurons was significant in both female and male groups, but mainly in terms of rest membrane potential and amplitude, respectively. Conclusions Glutamatergic and GABAergic neurons in medial prefrontal cortex showed different excitability changes in a rat model of early‐life neglect, which can contribute to distinct mechanisms for emotional and cognitive manifestations.

IntroductionDespite the widespread use of the unilateral striatal 6-hydroxydopamine (6-OHDA) lesion model in mice in recent years, the stability of behavioral deficits in the 6-OHDA striatal mouse model over time is not yet clear, raising concerns about using this model to evaluate a compound’s long-term therapeutic effects.Materials and methodsIn the current study, mice were tested at regular intervals in the cylinder test and gait analysis beginning 3 days after 6-OHDA injection of 4 and 8 μg and lasting until 56 days post-lesion. Apomorphine-induced rotational test and rotarod test were also performed on Day 23 and 43 post-lesion, respectively. Immunohistochemistry for dopaminergic neurons stained by tyrosine hydroxylase (TH) was also performed.ResultsOur results showed that both the 4 and 8 μg 6-OHDA lesion groups exhibited forelimb use asymmetry with a preference for the ipsilateral (injection) side on Day 3 and until Day 21 post-lesion, but did not show forelimb asymmetry on Day 28 to 56 post-lesion. The 8 μg 6-OHDA lesion group still exhibited forelimb asymmetry on Day 28 and 42 post-lesion, but not on Day 56. The gait analysis showed that the contralateral front and hind step cycles increased from Day 3 to 42 post-lesion and recovered on Day 56 post-lesion. In addition, our results displayed a dose-dependent reduction in TH+ cells and TH+ fibers, as well as dose-dependent apomorphine-induced rotations. In the rotarod test, the 8 μg 6-OHDA lesion group, but not the 4 μg group, decreased the latency to fall on the rotarod on Day 43 post-lesion.ConclusionIn summary, unilateral striatal 6-OHDA injections of 4 and 8 μg induced spontaneous motor impairment in mice, which partially recovered starting on Day 28 post-lesion. Forced motor deficits were observed in the 8 g 6-OHDA lesion group, which remained stable on Day 43 post-lesion. In addition, the rotarod test and apomorphine-induced rotational test can distinguish between lesions of different extents and are useful tools for the assessment of functional recovery in studies screening novel potential therapies.

Abstract Background Alzheimer's disease (AD) is an incurable and irreversible neurodegenerative disease, without a clear pathogenesis. Therefore, identification of candidates before amyloid‐β plaque (Aβ) deposition proceeds is of major significance for earlier intervention in AD. Methods To explore the potential noninvasive earlier biomarkers of AD in a 5XFAD mouse model, microRNAs (miRNAs) from urinary exosomes in 1‐month‐old pre‐Aβ accumulation 5XFAD mice models and their littermate controls were profiled by microarray analysis. The differentially expressed miRNAs were further analyzed via droplet digital PCR (ddPCR). Results Microarray analysis demonstrated that 48 differentially expressed miRNAs (18 upregulated and 30 downregulated), of which six miRNAs – miR‐196b‐5p, miR‐339‐3p, miR‐34a‐5p, miR‐376b‐3p, miR‐677‐5p, and miR‐721 – were predicted to display gene targets and important signaling pathways closely associated with AD pathogenesis and verified by ddPCR. Conclusions Urinary exosomal miRNAs showing differences in expression prior to Aβ‐plaque deposition were identified. These exosomal miRNAs represent potential noninvasive biomarkers that may be used to prevent AD in clinical applications.

Abstract As an important equipment of power system, the insulator's normal operation is the basis to ensure the safe operation of the power system. The insulator positioning and identification technology based on machine vision can quickly and accurately complete the inspection of insulators on site and effectively save the cost of operation and maintenance. This paper proposes an insulator inspection method based on region‐convolutional neural networks (RCNNs). First, the dataset of the insulator image is preprocessed by means of data expansion. Then, the feature extraction of the insulator image is realised by using the zeiler fergus (ZF) network. The k‐means clustering method is used to optimise the selection of anchor points. Meanwhile, the non‐maximum suppression post‐processing algorithm is improved, and a non‐linear penalty factor is introduced to adapt to multi‐scale and overlapping occlusion insulator inspection. Experimental results show that the improved faster RCNNs insulator inspection method can accurately obtain the coordinate frame and the corresponding probability value of the insulator object and improve the average precision by 10.43%, achieving the accurate inspection of the insulator object.

Abstract Background In isolated premature thelarche (IPT) girls, bone age (BA) is considered consistent with chronological age. However, some IPT girls confirmed by gonadotropin-releasing hormone (GnRH) stimulation test could show another trend. We analysed BA and possible potentiating factors in a selected group of girls aged 4–8 years with IPT. Methods IPT girls confirmed by GnRH stimulation test aged 4–8 years hospitalized from January 2015 to April 2018 at Shenzhen Children’s Hospital were included in this retrospective study. They were divided into two groups with advanced BA of 2 years as the cut-off. Body mass index (BMI) and hormone levels were the main outcome measures, and regression analysis was used to identify independent risk factors. IPT girls were divided into subgroups according to the levels of BMI standard deviation score (SDS), insulin-like growth factor-1 (IGF-1) SDS and dehydroepiandrosterone sulfate (DHEAS) SDS for comparisons of advanced BA. Results Overall, 423 subjects were included and classified into the advanced BA group (48.7%, n = 206) and control group (51.3%, n = 217). Abstract The vehicle group chain conveyor is the new kind of continuous material conveying equipment in bulk materials field. For its essential structure， which is the continuous track group car carrying the conveyor belt and relying on the rail runs. Curve passing performance at curve working conditions is studied. A nonlinear dynamics model of the conveyor track group passing the curve is established and analyzed. The multi-body dynamics simulation software SIMPACK is used to build the track group car model. Based on the different working conditions and the structure of the car， the variation of the curve passing performance of the track car is analyzed. The simulation results show that dynamics performance index of the track car in vehicle group chain conveyor under various working conditions decreases with the increase of the vehicle speed and spacing， and increases with the increase of the curve radius and transition curve length. Track car with smaller wheelbase and articulated body can adapt to curved track. The curve of the track car has good performance and all indicators are within the safe limit range.

In the paper, we proposed a deep learning-based industrial equipment detection algorithm ROMS R-CNN (Rotation Occlusion Multi-Scale Region-CNN). It can solve the problem of inaccurate detection of industrial equipment under complex working conditions such as multi-scale ratio, rotation tilt, occlusion and overlap. The method proposed in this paper first is to construct the MobileNetV2 as the feature pyramid network, and then to combine high semantic information with high resolution information solved industrial equipment detection of different scales. Secondly, a specific rotation anchor scheme is proposed, and the data set is clustered through the k-means algorithm to obtain a specific aspect ratio. Combined with the rotation angle, a rotation anchor of any direction and size is generated to solve the problem of easy tilting of industrial equipment. Finally, a Non-Maximum Suppression algorithm with penalty factors is introduced to solve the overlapping in industrial equipment detection. The experimental results in common industrial equipment detection show that this method is better than other algorithms, significantly improves the missed detection and false detection, and the mAP reaches 0.939.

Coal mining results in reduced soil quality and makes environments less stable. Soil fungi are suitable indicators of soil quality for monitoring purposes. Here, the objective was therefore to investigate the effects of grazing and mining on the composition of the soil fungal community at the periphery of an opencast coal-mine dump in the Shengli mining area, Xilingol League, Inner Mongolia. A total of 2,110 fungal operational taxonomic units were identified and subdivided into 81 orders and nine categories, based on trophic modes. The sensitive factor to mining was soil pH, and that to grazing were soil nitrate-nitrogen and alkaline phosphatase activity. According to the Pearson correlation and Mantel test, we propose interactions between grazing and coal-mining exist a co-effect and could regulate edaphic variables to alter the behavior of soil fungal community. Moreover, compared with coal-mining, grazing has a greater impact on it. The results provide a basis to further clarify soil fungal ecological functions, and may also contribute to the practice of soil remediation and environmental management in coal-mining areas.

In Parkinson's disease, synucleinopathy is hypothesized to spread from the enteric nervous system, via the vagus nerve, to the central nervous system. Recent evidences collected in non-human primates challenge however the hypothesis of a transmission of α-synuclein (α-syn) pathology through the vagus nerve. Would the hypothesis whereby the bloodstream acts as a route for long-distance transmission of pathological α-syn hold true, an inter-individual transmission of synucleinopathy could occur via blood contact. Here, we used a parabiosis approach to join the circulatory systems of wild type and GFP transgenic C57BL/6 J mice, for which one of the partners parabiont received a stereotaxic intranigral injection of patient-derived α-syn aggregates. While the Lewy Body-receiving mice exhibited a loss of dopamine neurons and an increase in nigral S129 phosphorylated α-syn immunoreactivity, their parabiotic bloodstream-sharing partners did not show any trend for a lesion or change in S129 phosphorylated-α-syn levels. Altogether, our study suggests that, in the patient-derived α-synuclein aggregates-injected mouse model and within the selected time frame, the disease is not “transmitted” through the bloodstream.

Woodchuck Hepatitis Virus Post-transcriptional Regulatory Element (WPRE) is thought to enhance transgene expression of target genes delivered by adeno-associated viral (AAV) vectors. This study assessed the protein expression of α-synuclein, phosphorylated α-synuclein at Serine 129, extent of nigrostriatal degeneration as well as subsequent behavioral deficits induced by unilateral intranigral stereotactic injection in male adult C57BL/6J mice of an AAV2/9 expressing A53T human α-synuclein under the control of the synapsin promoter in presence or absence of the WPRE. The presence of WPRE enabled to achieve greater nigrostriatal degeneration and synucleinopathy which was concomitant with worsened forelimb use asymmetry. This work refines a mouse Parkinson's disease model in which anatomo-pathology is related to behavioral deficits.

Early-life neglect in critical developmental periods has been associated with emotional and cognitive consequences. Maternal separation (MS) has been commonly used as a rodent model to identify the developmental effects of child neglect. However, reports have shown considerable variability in behavioral results from MS studies in both mice and rats. Difficulties in developing reliable child neglect models have impeded advances in identifying the effects of early-life stress. Accumulating evidence shows that neuronal intrinsic excitability plays an important role in information processing and storage in the brain. The prefrontal cortex (PFC) integrates information from many cortical and subcortical structures. No studies to date have examined the impact of early-life stress on glutamatergic neuronal excitability in the PFC. This study aimed to develop a reliable child neglect rat model and observe glutamatergic neuronal excitability in the PFC. An MS with early weaning (MSEW) rat model was developed. Rats were separated from the dam for 4 h per day on postnatal days (PNDs) 2–5 and for 8 h per day on PNDs 6–16 and then weaned on PND 17. A battery of behavioral tests was used to assess anxiety-like behavior, coping behavior, working memory, spatial reference memory, and fear memory. The action potentials (APs) of glutamatergic neuronal membranes were recorded. MSEW resulted in anxiety-like behavior, a passive coping strategy and increased fear memory in male rats and decreased locomotor activity in both sexes. MSEW slightly impaired working memory during non-stressful situations in female rats but did not change spatial reference memory or associative learning under stressful circumstances in either sex. MSEW reduced the number of glutamatergic neuron APs in male rats. Our findings showed that MS with early weaning induced anxiety-like behavior in male rats. The reduced glutamatergic neuronal excitability may be associated with the emotional alteration induced by MSEW in male rats. In addition, MSEW induced adaptive modification, which depended on a non-stressful context.

BackgroundAlzheimer’s disease (AD) is an incurable neurodegenerative disease characterized by irreversible progressive cognitive deficits. Identification of candidate biomarkers, before amyloid-β-plaque deposition occurs, is therefore of great importance for early intervention of AD.ObjectiveTo investigate the potential non-invasive early biomarkers of AD in 5XFAD mouse model, we investigate the proteome of urinary exosomes present in 1-month-old (before amyloid-β accumulation) 5XFAD mouse models and their littermate controls. Another two groups of 2 and 6 months-old urinary samples were collected for monitoring the dynamic change of target proteins during AD progression.MethodsProteomic, bioinformatics analysis, multiple reaction monitoring (MRM), western blotting (WB) or ELISA were performed for analyzing these urinary exosomes.ResultsA total of 316 proteins including 44 brain cell markers were identified using liquid chromatography tandem mass spectrometry. Importantly, 18 proteins were unique to the 5XFAD group. Eighty-eight proteins including 11 brain cell markers were differentially expressed. Twenty-two proteins were selected to be verified by WB. Furthermore, based on an independent set of 12 urinary exosomes samples, five in these proteins were further confirmed significant difference. Notably, Annexin 2 and Clusterin displayed significant decreased in AD model during the course detected by ELISA. AOAH, Clusterin, and Ly86 are also brain cell markers that were first reported differential expression in urinary exosomes of AD model.ConclusionOur data demonstrated that some urinary exosome proteins, especially Annexin 2 and Clusterin, as nanometer-sized particles, enable detection of differences before amyloid-β-plaque deposition in 5XFAD mouse model, which may present an ideal non-invasive source of biomarkers for prevention of AD.

Abstract Background Bupleurum yinchowense Shan et Y. Li is widely used to treat depressive and anxiety disorders for hundreds of years in China. Total saikosaponins (TSS) is the major ingredient of Bupleurum yinchowense. A-amino-3-hydroxy-5-methyl-4-isoxazolepropionic acid (AMPA) receptor and subsequent mammalian target of rapamycin (mTOR) signaling is responsible for synaptic maturation and may contribution to the synaptic alteration underlying depression. The aim of the study was to investigate the antidepressant-like and anxiolytic effect of TSS in chronic corticosterone-treated mice. The effect of TSS on synaptic proteins expression and AMPA receptor-mTOR signaling pathway alteration was also evaluated. Methods Dose-response effect of TSS (12.5, 25, 50 mg/kg) was investigated in forced swim test (FST) in ICR male mice. In the chronic corticosterine-treated model, TSS was given intragastrically once a day for 2 weeks and continued through the behavior testing period. Behavior tests and AMPA receptor related signaling pathway were investigated. Results TSS (25 and 50 mg/kg) decreased the immobility time in the FST when compared with the control group. TSS (25 mg/kg) showed antidepressant-like and anxiolytic effects in the chronic corticosterone treatment model in mice. TSS increased hippocampal synaptic proteins (synapsin-1 and postsynaptic density protein 95) expression. Immunohistochemistry analysis showed that TSS significantly increased the synapsin-1 expression in CA3 of hippocampus. TSS also increased hippocampal phosphorylation expression of GluR1 Ser 845 (AMPA receptor subunit) and its downstream regulators extracellular signaling-regulated kinase (ERK), protein kinase B (Akt) and mTOR. Conclusion TSS produces antidepressant-like and anxiolytic effects and increases synaptic proteins expression which may be mediated by induction of AMPA receptor and subsequent mTOR signaling pathway.

Abstract The precipitation variability associated with the East Asian summer monsoon (EASM) has profound societal implications. Here, we use precisely dated and seasonally-resolved stalagmite oxygen isotope (δ18O) records from Shihua Cave, North China to reconstruct the EASM variability over the last 145 years. Our record shows a remarkable weakening of the EASM strength since the 1880s, which may be causally linked to the warming of the tropical Pacific and Indian Oceans. The δ18O record also exhibits a significant ~30-year periodicity, consistent with the instrumental, historical and proxy-based rainfall records from North China, plausibly driven by the Pacific Decadal Oscillation (PDO). Together, these observations imply that ~30-year periodicity is a persistent feature of the EASM, which remains significant with or without anthropogenic forcing. If indeed, the EASM rainfall in North China might decline significantly in the near future, which may affect millions of people in this region.

Infantile hemangioma (IH) is one of the most common soft-tissue neoplasms of infancy. Although clinical diagnosis for IH is well-established, the haematological parameters associated with IH are not well explored. In this short study, we observed significantly higher eosinophil (EO) numbers in IH patient blood compared to healthy controls. This contributed to the observed higher EO % in the peripheral blood of IH patients and was irrespective of age. This new haematological finding could carry a potential diagnostic/prognostic relevance for IH.

Mitophagy plays a key role in cleaning damaged and depolarized mitochondria to maintain cellular homeostasis and viability. Although it was originally found in neurodegenerative diseases, mitophagy is reported to play an important role in acute kidney injury. PINK1 and Parkin are key molecules in mitophagy pathway. Here, we used PINK1 knockout rats to examine the role of PINK1/Parkin-mediated mitophagy in cisplatin nephrotoxicity. After cisplatin treatment, PINK1 knockout rats showed lower plasma creatinine and less tubular damage when compared with wild-type rats. Meanwhile, mitophagy indicated by autophagosome formation and LC3B-II accumulation was also attenuated in PINK1 knockout rats. Renal expression of PINK1 and Parkin were down-regulated while BNIP3L was up-regulated by cisplatin treatment, indicating a major role of BNIP3/BNIP3L pathway in cisplatin-induced mitophagy. Transmission electron microscopy showed that PINK1 deficiency inhibited cisplatin-induced mitochondrial fragmentation indicating an involvement of mitochondrial fusion and fission. Renal expression of mitochondrial dynamics related proteins including Fis1, Drp1, Mfn1, Mfn2, and Opa1 were checked by real-time PCR and western blots. The results showed PINK1 deficiency distinctly prevented cisplatin-induced up-regulation of DRP1. Finally, PINK1 deficiency alleviated cisplatin-induced tubular apoptosis indicated by TUNEL assay as well as the expression of caspase3 and cleaved caspase3. Together, these results suggested PINK1 deficiency ameliorated cisplatin-induced acute kidney injury in rats, possibly via inhibiting DRP1-mediated mitochondrial fission and excessive mitophagy.

Solar thermochemical energy storage based on calcium looping (CaL) process is a promising technology for next-generation concentrated solar power (CSP) systems. However, conventional calcium carbonate (CaCO3) pellets suffer from slow reaction kinetics, poor stability, and low solar absorptance. Here, we successfully realized high power density and highly stable solar thermochemical energy storage/release by synergistically accelerating energy storage/release via binary sulfate and promoting cycle stability, mechanical strength, and solar absorptance via Al–Mn–Fe oxides. The energy storage density of proposed CaCO3 pellets is still as high as 1455 kJ kg−1 with only a slight decay rate of 4.91% over 100 cycles, which is higher than that of state-of-the-art pellets in the literature, in stark contrast to 69.9% of pure CaCO3 pellets over 35 cycles. Compared with pure CaCO3, the energy storage power density or decomposition rate is improved by 120% due to lower activation energy and promotion of Ca2+ diffusion by binary sulfate. The energy release or carbonation rate rises by 10% because of high O2− transport ability of molten binary sulfate. Benefiting from fast energy storage/release rate and high solar absorptance, thermochemical energy storage efficiency is enhanced by more than 50% under direct solar irradiation. This work paves the way for application of direct solar thermochemical energy storage techniques via achieving fast energy storage/release rate, high energy density, good cyclic stability, and high solar absorptance simultaneously.

Prostate stem cell antigen (PSCA) is expressed in all stages of prostate cancer, including in advanced androgen-independent tumors and bone metastasis. PSCA may associate with prostate carcinogenesis and lineage plasticity in prostate cancer. PSCA is also a promising theranostic marker for a variety of other solid tumors, including pancreatic adenocarcinoma and renal cell carcinoma. Here, we identified a novel fully human PSCA antibody using phage display methodology. The structure-based affinity maturation yielded a high-affinity binder, F12, which is highly specific and does not bind to 6,000 human membrane proteins based on a membrane proteome array assay. F12 targets PSCA amino acids 63–69 as tested by the peptide scanning microarray, and it cross-reacts with the murine PSCA. IgG1 F12 efficiently internalizes into PSCA-expressing tumor cells. The antimitotic reagent monomethyl auristatin E (MMAE)-conjugated IgG1 F12 (ADC, F12-MMAE) exhibits dose-dependent efficacy and specificity in a human prostate cancer PC-3-PSCA xenograft NSG mouse model. This is a first reported ADC based on a fully human PSCA antibody and MMAE that is characterized in a xenograft murine model, which warrants further optimizations and investigations in additional preclinical tumor models, including prostate and other solid tumors.

The investigation of cryogenic cave minerals (CCMs) has developed in recent decades to be a particularly valuable proxy for palaeo-permafrost reconstruction. Due to difficulties, however, in obtaining reliable chronologies with the so-called “fine” form of these minerals, such studies have thus far utilised the “coarse” form. In this study, we successfully investigate the northernmost-known deposit of fine-grained cryogenic cave minerals (CCMs), which are situated in Cove Cave (Greenlandic translation: Eqik Qaarusussuaq), a low-elevation permafrost cave in northeast Greenland (80° N). The Cove Cave CCMs display a complex mineralogy that consists of fine-grained cryogenic cave carbonates (CCCfine) as well as sulphate minerals (gypsum, eugsterite, mirabilite, and löweite). In comparison to CCCfine from the mid-latitudes, positive δ13C values (7.0 to 11.4 ‰) recorded in Cove Cave are similar. In contrast, Cove Cave CCCfine δ18O values are ca. 8 to 16 ‰ lower. Furthermore, despite previous CCCfine dating efforts being unsuccessful, here we demonstrate that precise dating is possible with both isochron-based 230Th/U dating and 14C dating if the dead carbon fraction is reliably known. The dating result (65 ± 17 a BP; 1885 ± 17 CE) shows that the CCMs formed during the late Little Ice Age, a time interval characterised by cold temperatures and abundant permafrost in northeast Greenland, making water infiltration into Cove Cave dependent on water amount and latent heat. We relate the CCM formation to a combination of black carbon deposition and anomalously high temperatures, which occurred over a few days, in the summer of 1889 CE. Such extreme conditions led to widespread melting over large areas of the Greenland ice sheet. We propose that the anomalous (weather) conditions of 1889 CE also affected northeast Greenland, where the enhanced melting of a local ice cap resulted in water entering the cave and rapidly freezing. While CCCfine and gypsum likely precipitated concurrently with freezing, the origin of the other sulphate minerals might not be purely cryogenic but could be linked to subsequent sublimation of this ice accumulation in the very dry cave environment.
