We found that β-HgS nanocrystals could catalyze the reactions of oxidase substrates such as 3,3′,5,5′-tetramethylbenzidine (TMB) and O-phenylenediamine (OPD) to form coloured and fluorescent products, respectively, but α-HgS nanocrystals could not catalyze these reactions. Further investigation revealed that the predominantly exposed crystal planes may play an important role in determining the catalytic activity. On the basis of these findings, taken together with the reaction of forming a Schiff base between the –NH2 group of OPD and the –CHO group of formaldehyde, a novel, simple, and highly selective photoluminescence (PL) sensing platform for detecting formaldehyde could thereby be successfully developed.

The aim of the present study was to evaluate the potential use of 7 plasma miRNAs for liver fibrosis staging in patients with chronic hepatitis B virus (HBV) infection. Relative levels of miRNAs were measured using quantitative polymerase chain reaction and used to develop a diagnostic panel. A receiver operating characteristic (ROC) curve was drawn to evaluate the performance of individual miRNAs and the whole panel. Ascorbic acid and glycol were employed as the carbon resource and co-solvent, respectively, to prepare extraordinary green photoluminescent carbon dots (CDs) by means of a hydrothermal method. Furthermore, the resulting CDs could be successfully employed to establish highly sensitive and selective sensing systems for Al3+ ions with a photoluminescence (PL) enhancement response and for F− anions with a PL “ON–OFF” model.

A sensitive and selective chemosensing system for bromate anions (BrO3− anions) is highly demand due to the serious toxicity of BrO3− anions. In this report, polymeric ethylene imine (PEI) functionalized photoluminescent carbon dots (CDs@PEI) were prepared by the thermal pyrolysis of citric acid and PEI. The as-prepared CDs@PEI were successfully employed to establish a photoluminescence (PL)-based sensing system for BrO3− anions with high sensitivity and selectivity. The experimental conditions were systemically optimized, and a linear relationship between the PL quenching rate of the CDs@PEI and BrO3− anion concentration in the range from 0.04 to 0.35 μM could be obtained, with a very low limit of detection of ca. 0.17 nM. The proposed method was further employed to determine BrO3− anions in drinking water, bottled water and lake water samples with good recoveries. This established method was also successfully employed to detect BrO3− anion content in practical pastry samples.

Background: Accurate diagnosis of chronic Hepatitis B virus (HBV) infection related diseases is crucial to guide the therapy and to understand the mechanisms of disease progression. Plasma microRNAs, as stable biomarkers, have drawn significant attentions for distinguishing HBVrelated diseases. Methods: In this study, a new HBV-related disease identification method based on a two-layer logistic regression model was presented. A total of nine effective plasma microRNA biomarkers were selected through sample collection, data processing, model selection, feature selection and model optimization to distinguish HBV-related chronic hepatitis and cirrhosis samples as well as healthy controls. The first layer utilized three microRNAs to distinguish HBV-related disease samples from healthy controls. Then the second layer divided the HBV-related disease samples into cirrhosis and chronic hepatitis samples by using eight microRNAs. Result: Test on two independent cohorts showed high accuracy and robustness of our model. Functional analysis of the selected microRNAs and their target genes confirmed that they were significantly associated with HBV-related diseases and related functional pathways. Conclusion: Compared with previous models, the two-layer model was more consistent with the underlying pathological progress of HBV related diseases from health to chronic hepatitis and further to liver cirrhosis. It could also take the results of other diagnostic tests into account, which could be useful in both physical examination and disease diagnosis.

In this paper, a novel DNA-length-dependent fluorescent sensor was constructed based on the fluorescence resonance energy transfer. In the self-assembled multilayers (Quartz/GO/PDDA/Tx-DNA/PDDA/ZnO@CdS), ZnO@CdS and graphene oxide(GO) were employed as an energy donor and an energy acceptor, respectively. Single-stranded Tx-DNA (x represents different chain length of DNA) and poly(diallydimethylammonium) chloride (PDDA) were used as a linker. In the presence of complementary Px-DNA, the formation of double-stranded DNA leads to a change in chain length and achieves the purpose of changing the distance between ZnO@CdS and GO. Thereby, it enhances the efficiency of energy transfer between ZnO@CdS and GO resulting in the quench of fluorescence of ZnO@CdS, and thus different length DNA sequence was detected.

An important unresolved clinical issue is to distinguish hepatitis B virus (HBV) infection caused chronic hepatitis and their corresponding liver cirrhosis (LC). Recent research suggests that circulating microRNAs are useful biomarkers for a wide array of diseases. We analyzed microRNA profiles in the plasmas of a total of 495 chronic hepatitis B (CHB) patients, LC patients and healthy donors and identified 10 miRNAs that were differentially expressed between CHB and LC patients. Our logistic models show that three panels of miRNAs have promising diagnostic performances in discriminating CHB from LC. Blinded tests were subsequently conducted to evaluate the diagnostic performances in clinical practice and a sensitivity of 85% and specificity of 70% have been achieved in separating CHB from LC pateints. The expression levels of some circulating miRNAs were significantly correlated with HBV DNA load and liver function, such as prothrombin activity (PTA) and levels of alanin aminotransferase (ALT), albumin (ALB) and cholinesterase (CHE). Our results provide important information for developing novel diagnostic tools for distinguishing chronic HBV hepatitis and their corresponding cirrhosis.

A novel dual-photoluminescence probe for Cu(2+) has been developed, in which the graphene quantum dots with blue emission and CdTe QDs with yellow emission act as internal standard and probe, respectively. The photoluminescence probe exhibited selective sensing for Cu(2+) with a limit of detection (3SD/k) of 5.3 × 10(-8) M and showed its potential application in visual imaging. The results indicated that the constructed probe can be employed for sensing Cu(2+) by the naked eye, and also for monitoring intracellular Cu(2+).

Studies of congestive heart failure (CHF) in man and in experimental CHF have demonstrated elevated circulating levels of endothelin (ET). In order to examine whether there are concomitant ET receptor alterations, the vasomotor effects of endothelin-1 (ET-1) and sarafotoxin 6c (S6c) were examined in endothelium-intact and -denuded isolated mesenteric arteries from rats with CHF. CHF was induced by ligation of the left anterior descending coronary artery. Vasomotor responses were studied using small mesenteric arteries (approx. 250 microm in diameter, determined after normalisation). The antagonists IRL2500 and FR139317 were used in order to characterise the ET-1-induced response. In mesenteric arteries with intact endothelium, ET-1-induced contractions were more potent in CHF as compared to sham (pEC(50) 9.6+/-0.2 and 9.1+/-0.1, respectively, P0.01). In endothelium-denuded arteries, there was no difference in potency of ET-1 between CHF and sham arteries, or in maximum contraction. In the presence of IRL2500, a selective ET(B)-receptor antagonist, ET-1 was more potent in endothelium-denuded arteries of CHF rats, while this difference was not seen in sham arteries. S6c had no consistent contractile or dilatory effect in CHF and sham rats. The results indicate that the enhanced contractile effects of ET-1 noted in CHF might be due to an attenuated endothelial function and that inhibition of smooth muscle cell ET(B) receptors increase the effects of contractile ET(A) receptors in CHF rats.

Aim: The aim of the present study was to examine the role of Ca2+ -mediated contractile responses in isolated mesenteric resistance arteries from rats with congestive heart failure (CHF). Methods: Heart failure was induced by ligation of the left coronary artery. Rats exposed to the same surgical procedure except ligation served as controls (Sham). The following experiments were conducted: (1) passive increase in radial stretch (the length–tension relationship) in Ca2+ -free and in depolarizing high K+- solution; (2) the contractile responses to external application of Ca2+ and high K+ -solutions in the presence of nifedipine and phentolamine; and (3) a histological evaluation of CHF and Sham vessels. Results: The length–tension induced response in Ca2+ -free buffer solution was significantly lower in arteries from CHF rats, starting at a very low tension (0.9 ± 0.2 mN/mm for heart failure and 1.7 ± 0.2 mN/mm for Sham). This difference, but at a higher degree of stretch, was also present in K+ -activated vessels. The external application of Ca2+ in K+ -depolarized vascular segments in the presence of phentolamine (1 μM) induced an enhanced contractile response in arteries from CHF rats compared with Sham (4.8 ± 0.3 mN/mm and 3.6 ± 0.6 mN/mm, respectively, P = 0.059). In the absence of phentolamine the reverse response was found (4.0 ± 0.4 mN/mm and 5.7 ± 0.3 mN/mm for CHF vs. Sham respectively, P = 0.035). Application of increasing concentrations of K+ -solution induced a stronger contractile response in Sham compared with CHF arteries (Sham 4.9 ± 0.4 and heart failure 4.0 ± 0.3, P = 0.04). Microscopic examination of vessels yielded no difference in gross morphology, media thickness or wall to lumen ratio between CHF and Sham arteries. Conclusion: The results indicate an attenuation of α-adrenoceptors and a difference of Ca2+ -mediated vascular contractility in resistance arteries of congestive heart failure rats.

We investigated whether the effect of long-term intermittent hypoxia (LTIH) on cardiovascular function may be modified by preexisting genetic traits. To induce LTIH experimentally, cycles of 90-s hypoxia (nadir 6%) followed by 90-s normoxia were applied to six Wistar-Kyoto and six spontaneously hypertensive rats during 8 h daily. Comparison with the same number of control animals after 70 days revealed no alteration of intra-arterial blood pressure or heart rate. Blood pressure responsiveness to a brief hypoxic stimulus was enhanced in the LTIH animals, regardless of strain, whereas the hypoxia-induced increase in heart rate was abolished. In the spontaneously hypertensive but not the Wistar-Kyoto rats, LTIH increased left ventricular weight-to-body weight ratio and content of atrial natriuretic peptide mRNA. Expression of B-type natriuretic peptide was unchanged (Northern blot). Slightly increased right ventricular weight-to-body weight ratios in the LTIH animals were associated with higher right ventricular atrial natriuretic peptide and B-type natriuretic peptide mRNA amounts. Consequently, the effects of LTIH on different components of cardiovascular function appear incompletely related to each other and differentially influenced by constitutional traits.

Heart failure is associated with increased sympathetic nerve activity. We hypothesized that chronic sympathetic stimulation in heart failure resulted in decreased vascular sympathetic responsiveness. A pithed rat model was employed to evaluate peripheral vascular alpha-adrenoceptor and neuropeptide Y (NPY) receptor responsiveness. Heart failure was induced in Sprague-Dawley rats by coronary artery ligation. Sham operated rats (Sham) served as controls. Two months after this surgical procedure, both heart failure (n = 30) and Sham (n = 30) rats underwent standard pithing procedure. Pressor responses to preganglionic sympathetic nerve stimulation (PNS) and activation of postjunctional alpha1- and alpha2-adrenoceptors as well as Y1 receptors were studied. In response to PNS, cardiac index was similar between heart failure and sham rats (P = n.s.). Mean arterial pressure (MAP) increased in a frequency-dependent fashion after PNS in heart failure rats as well as in control rats. All the agonists used, i.e. the alpha1-adrenoceptor agonist phenylephrine, the alpha2-adrenoceptor agonists clonidine and BHT933 as well as NPY, induced dose-dependent increases in MAP in heart failure and in sham rats. However, in rats with heart failure, the response to all the agonists studied was significantly decreased and the dose response curves were shifted to the right (P < 0.01). We conclude that in vivo vascular response to postjunctional alpha1- and alpha2-adrenoceptors as well as Y1 receptors are decreased in rats with heart failure.

Objective: Congestive heart failure (CHF) is accompanied by enhanced peripheral sympathetic nerve activity, increased vascular resistance and impaired peripheral blood flow. Besides noradrenaline and neuropeptide Y, the sympathetic nervous system also releases ATP, which has contractile effects mediated by different subtypes of P2-receptors on the vascular smooth muscle cells. The present study was designed to examine postsynaptic changes of the contractile responses to ATP and other extracellular nucleotides in CHF. Methods: CHF was induced by left coronary artery ligation resulting in a reproducible myocardial infarction in Sprague–Dawley rats. Contractile responses were examined in cylindrical segments of aorta and the mesenteric artery after endothelium removal. To determine if an altered response was regulated on the transcriptional level, competitive reverse transcription polymerase chain reaction (RT-PCR) was used to estimate the amount of P2X1-receptor mRNA. Results: ATP, which is both a P2X1- and a P2Y-receptor agonist, induced a weaker contraction in the mesenteric artery from CHF as compared to sham operated rats. A decrease in both potency and maximum contraction was shown for the selective P2X1-receptor agonist, αβ-MeATP, in the mesenteric artery (pEC50=6.04 vs. 5.76, C max=57% vs. 33%, sham vs. CHF operated rats), but not in the aorta. Competitive RT-PCR also revealed decreased P2X1-receptor mRNA levels in CHF operated rats in the mesenteric artery (9106·103 vs. 714·103 molecules/μg, sham vs. CHF operated rats), while it remained unaltered in the aorta. To study the P2Y-receptor induced contractile effects, the P2X1-receptors were first desensitised with αβ-MeATP (10−5 M for 8 min). After P2X1-receptors desensitisation, UTP and UDP induced strong contractions in both the mesenteric artery and in the aorta, while ATP and ADP were much less effective. These contractions were not altered by CHF, indicating that vascular contraction mediated by P2Y-receptors are unaffected by CHF. Conclusion: CHF induces downregulation of P2X1-receptor stimulated contraction in the mesenteric artery depending on decreased mRNA synthesis for the receptor, while the P2Y-receptor activity remains unchanged. Downregulation of P2X1-receptors appears to be specific for peripheral resistance arteries. This may represent a compensatory response to enhanced peripheral sympathetic nerve activity and increased vascular resistance in CHF.

It was examined to what extent congestive heart failure (CHF) in rats, induced by ligation of the left coronary artery, affects the vascular responses to the vasodilatory substances acetylcholine (ACh), calcitonin gene-related peptide (CGRP), and substance P (SP). After induction of CHF status, the basilar, mesenteric and renal arteries and the iliac vein were studied in vitro. Dilatory responses were determined in relation to pre-contraction by the thromboxane mimetic U46619. Sham-operated animals (Sham) served as controls. U46619 induced stronger contraction in CHF basilar and renal arteries compared with the corresponding segments in Sham. ACh induced concentration-dependent dilations in all vessels examined with no difference of maximum relaxation or potency between CHF and Sham. SP induced weak dilations in all arteries examined while the response was markedly attenuated in CHF iliac veins compared with Sham (Emax% 12.2 +/- 3.4 vs. 32.3 +/- 4.8, P = 0.01). The CGRP induced dilation in the CHF basilar artery was weaker (Emax% 18.6 +/- 6.5 vs. 66.9 +/- 5.0, P < 0.001) and less potent (pEC50: 8.2 +/- 0.2 vs. 9.0 +/- 0.2, P = 0.01) compared with Sham. Further, CGRP was less potent in the renal artery of CHF rats compared with Sham (pEC50: 8.1 +/- 0.2 vs. 9.5 +/- 0.3, P < 0.01). In the CHF iliac vein, CGRP was more potent compared with Sham (pEC50: 9.7 +/- 0.4 vs. 8.3 +/- 0.4, P < 0.05). It can be concluded CHF is accompanied by alterations in the vascular response to the dilatory substances studied. The changes differ between vascular beds and between the different substances.

In order to assess the effects of dihydropyridine calcium antagonist on sympathetic nerve activity (SNA) in experimental chronic heart failure (CHF), felodipine was given to rats with CHF induced by coronary artery ligation. Anesthetized CHF (n = 7) and sham-operated (n = 9) rats were injected with a bolus dose of felodipine (20 µg/kg) and then infused with felodipine (30 µg/kg/h) for 3 hours. Control CHF rats (n = 8) were given vehicle in the same way. After felodipine treatment, mean blood pressure (MBP) rapidly decreased to 75–85 mmHg, and there was a reflex tachycardia and reflex activation of renal SNA. The heart rate (HR) had returned to baseline level after 3 hours of continuous felodipine infusion, and the SNA returned to baseline level after 2 hours of infusion. At the end of the experiment, renal SNA was 65.4 ± 11.5% of the baseline level in CHF rats receiving felodipine (P 0.05), but there was no statistical difference between the two groups. Arterial baroreceptor sensitivity (assessed by phenylephrine infusion), which was impaired in CHF rats (−2.7 ± 0.2 SNA%/mmHg in all CHF rats together vs. −3.6 ± 0.4 in sham-operated rats, P < 0.5) did not differ significantly from that in sham-operated rats during felodipine infusion (−3.2 ± 0.4 in felodipine-treated CHF rats vs. −3.7 ± 0.6 in sham-operated rats) but deteriorated without felodipine treatment (−2.1 ± 0.3 in CHF rats receiving vehicle, P < 0.05). The biphasic renal SNA response during felodipine infusion suggests that felodipine does not cause persistent sympathetic activation and relatively improves baroreceptor sensitivity in CHF rats.

Objective To study the effects of the selective neuropeptide Y (NPY) Y1 receptor antagonist BIBP 3226 in spontaneously hypertensive rats (SHR) in order to elucidate whether NPY function may be altered in the SHR and whether the NPY Y1 receptor plays a specific role in the maintenance of high blood pressure in this genetic form of hypertension. Methods Pithed and conscious SHR were studied after intravenous administration of 0.125-1 mg/kg BIBP 3226. The cardiovascular effects were evaluated under baseline conditions, under acute stress and after exogenous administration of 20 microg/kg NPY. The potentiating effects of NPY on pressor responses to phenylephrine and tyramine were studied in the SHR. Results Intravenous administration of 0.125-1 mg/kg BIBP 3226 dose-dependently inhibited the pressor response to exogenous NPY in pithed SHR. At higher doses BIBP 3226 had an effect duration of 20-40 min. In the pithed SHR, a 0.5 mg/kg bolus injection of BIBP 3226 shifted the pressor response curve for exogenous NPY significantly to the right It also inhibited significantly the potentiating effects of NPY on pressor responses to phenylephrine and tyramine. In conscious SHR, 0.125-1 mg/kg BIBP 3226 did not reduce the basal blood pressure. In combination with a hypotensive dose of prazosin, administration of 0.5 mg/kg BIBP 3226 had no added effects lowering the basal blood pressure. A stressful stimulus, namely an air jet, caused a brief increase in blood pressure and heart rate in the conscious SHR. In this model, 0.5 mg/kg BIBP inhibited the heart rate response slightly but had no effect on the blood pressure response. Conclusions Our results demonstrate that, although the selective NPY Y1 receptor antagonist BIBP 3226 may shift the pressor response to exogenous NPY potently, it does not influence basal blood pressure significantly in the SHR.

Using microcalorimetry and measuring oxygen consumption rate, we have studied metabolic activity in myocardial tissue from rats suffering from congestive heart failure (CHF) after ligation of the left coronary artery followed by myocardial infarction, comprising 30–40% of the endocardial circumference, known to be large enough to induce heart failure and neuroendocrine counter-regulation. The heat production in the preserved part of the left ventricle, harbouring the myocardial infarction, was 0.94 ± 0.05 mW g −1 ( n = 10) and not different from the controls i.e 0.94 ± 0.05 mW g −1 , ( n = 23). The oxygen consumption rate was 1.36 ± 0.20 nmol O 2 s −1 g −1 in CHF myocardium and 1.55 ± 0.24 nmol O 2 s −1 g −1 in the control rats, ( n = 20), n.s. The calculated contribution from aerobic metabolism to total heat production was 71 ± 9% in the CHF rats and 80 ± 6% in the control group, n.s. In tissue samples taken from the region of the border of and into the macroscopically damaged area of the myocardial infarction ( n = 5), the value for heat production was 1.15 ± 0.13 mW g −1 and oxygen uptake 1.79 ± 0.23 nmol O 2 s −1 g −1 . These results show that the contribution from aerobic metabolism to total energy expenditure in preserved left ventricular tissue is not higher than normal and suggest that the effects of left coronary occlusion are not limited to the generation of a myocardial infarction, but may also have a substantial impact on the ability of myocardial cells to increase metabolic activity in macro- and microscopically preserved regions of the left ventricle. In addition, the data demonstrate persistent metabolic activity within the region of ischemically damaged myocardium not different from that found in undamaged myocardium.

Metabolically stabilized (phosphorothioate) human and rat NPY Y1 receptor oligodeoxynucleotides (ODNs) complimentary to the rat or human Y1 mRNA were synthesized; [sense (rY1-SODN, 5'-AATTCAACTCTGTTCTCC-3'), antisense (hY1-ASODN, 5'-CCTGGGAAAATAATGTTG-3' and rY1-ASODN, 5'-GGAGAACAGAGTTGAATT-3') and mismatches (hY1-MMODN, 5'-CCTGAGATAA-TAAGGTTG-3' and rY1-MM 5'-GTAGATCAGAGATGAAGT-3')] and used to modulate cardiovascular function in vitro in human vessels as well as in vivo in the rat. 2. The objectives of the experiments were to assess the influence of the NPY Y1 receptor on vasomotor function human resistance arteries in vitro and to investigate the contribution of the NPY receptor system to cardiovascular haemodynamics in vivo. 3. Human subcutaneous resistance arteries removed from patients who underwent surgery for nonvascular diseases were incubated in vitro with the stabilized phosphorothioated hY1-receptor ASODN or MMODN (10(-7) TO 10(-5) M). 4. In human resistance vessels preincubated with hY1-AS (10(-7) to 10(-5) M), the contractile response to NPY was significantly reduced in a dose-dependent fashion. No effects were observed in the hY1-MMODN-incubated vessels at lower concentrations (10(-7) M to 10(-6) M). 5. The haemodynamic effects of the phosphorothioated rY1-ASODN, SODN or MMODN were investigated in conscious rats during 48 h of continuous infusions. The continuous infusion with rY1-ASODN did not change MAP while the rY1-SODN unexpectedly induced an early (10-20) increase in ambulatory MAP and the rY1-MMODN a late (24-44 h) increase. 6. Contractile responses to NPY (2, 4, 8, 16 and 32 micrograms kg-1) were significantly reduced in the rats treated with long-term infusion of rY1-ASODN (2.1 mg kg-1 h-1, i.v. infusion for 48 h) compared with animals treated with rY1-SODN and MMODN, as well as animals treated with saline and glucose. Notably, the group infused with the rY1-SODN showed an exaggerated response to tested doses of NPY. 7. We conclude that the incubation of human subcutaneous arteries with a metabolically stabilized 18 base pair hY1-ASODN and long-term infusion with a corresponding rY1-ASODN attenuate NPY-induced vasoconstriction.

Objective: Vascular alpha-2 adrenoceptor function of rats with congestive heart failure (CHF) was characterized in both in vivo and in vitro experiments. Methods: CHF was induced in Sprague-Dawley rats by coronary artery ligation. Sham-operated rats served as normal controls. Postjunctional alpha-2 adrenergic responsiveness was assessed in vivo using the pithed rat model and in vitro in organ bath. Vascular alpha-2 adrenoceptor density was studied by receptor binding assay. Results: Four to 6 weeks after this surgical procedure, plasma catecholamines were markedly increased in CHF rats. In vivo vascular responses to alpha-2 adrenoceptor agonists BHT933 and clonidine were significantly decreased in CHF rats ( P < 0.001). Clonidine elicited dose-dependent responses in endothelium intact mesenteric arteries in both CHF and sham-operated rats. The dose-response curve in CHF was shifted to the right with a pD2 value of 5.5 ± 0.2 compared with control rats 6.2 ± 0.2 ( P < 0.05). The response to clonidine was selectively blocked by an alpha-2 adrenergic antagonist rauwolscine in both groups. Endothelium denuded arteries showed an enhanced response to clondine in both CHF and control rats. However, the response to clondine is still decreased in CHF compared to sham-operated rats ( P < 0.05). Alpha-2 adrenoreceptor density, as determined by [3H]yohimbine binding in membrane preparations from mesenteric arteries was decreased in CHF compared to sham-operated rats (Bmax 43 ± 6 vs. 104 ± 20 fmol/mg protein, P < 0.05). Conclusions: Vascular alpha-2 adrenoceptor function is decreased in rats with CHF.

In addition to its direct vasoconstrictive effect, neuropeptide Y (NPY) potentiates noradrenaline-(NA) induced contraction and inhibits acetylcholine-(ACh) induced relaxation: The aim of the present study was to elucidate the NPY receptor subtypes responsible for mediating these three responses. NPY, peptide YY (PYY) and pro34NPY (a NPY Y1 receptor agonist) induced equipotent and equally strong concentration-dependent contractions of guinea pig basilar arteries. NPY13-36 (a NPY Y2 receptor agonist), however, caused only weak contraction with significantly lower potency. The NPY-induced contraction was significantly inhibited by the selective NPY Y1 receptor antagonist BIBP3226 (1 microM). NPY, PYY and pro34NPY but not NPY13-36 significantly potentiated the NA-induced contraction in guinea pig mesenteric arteries. The potentiation was significantly inhibited by BIBP3226 (1 microM). In precontracted guinea pig basilar arteries, ACh induced a concentration-dependent relaxation which was significantly inhibited by NPY, PYY and NPY13-36 but not by pro34NPY. BIBP3226 had no significant effect on the NPY-induced inhibition of the relaxation. These results suggests that the NPY Y1 receptors mediate the direct contraction and the potentiation of the NA-induced contraction but not the inhibition of the ACh-induced relaxation. This effect seems to be mediated by another NPY receptor subtype, presumably by the Y2 receptor, as judged from the agonist potency order.

Neuropeptide Y is a sympathetic co-neurotransmitter released with noradrenaline upon sympathetic nerve stimulation. This study describes the ability of a synthetic inositol phosphate, α-trinositol (d-myo-inositol 1,2,6−triphosphate; PP 56) to antagonize vasoconstrictor responses to neuropeptide Y in-vitro as well as in-vivo. In human and guinea-pig isolated arteries α-trinositol potently (10 nM to 1 μM extracellular concentration) suppressed the constriction evoked by neuropeptide Y alone, the potentiation by neuropeptide Y of noradrenaline-evoked constriction, and the neuropeptide Y-induced inhibition of relaxation. Moreover, in the pithed (areflexive) rat, a non-adrenergic portion of the pressor response to preganglionic sympathetic nerve stimulation was sensitive to α-trinositol. As studied in the recently cloned human (vascular-type) Y1 receptor, the action of α-trinositol does not occur through antagonism at the neuropeptide Y recognition site nor does it induce allosteric changes of this receptor. However, we found α-trinositol to inhibit the rise in intracellular Ca2+ as well as inositol triphosphate concentrations induced by neuropeptide Y. It is, therefore, proposed that α-trinositol represents a non-receptor, but yet selective antagonist of neuropeptide Y in vasculature, opening up the possibility to investigate involvement of neuropeptide Y in sympathetic blood pressure control and in cardiovascular disorders.

Previous studies have demonstrated that alpha-trinositol (D-myo-inositol-1.2. 6-trisphosphate; PP56) may act as a functional neuropeptide Y (NPY) inhibitor. Because NPY is known to be a potent vasoconstrictor, the effects of alpha-trinositol on renal function, vascular responses and the potentiating effects of NPY were investigated in rats with congestive heart failure (CHF) induced by ligation of the left coronary artery. Incremental doses of alpha-trinositol were given to conscious rats (bolus 2, 4 or 10 mg/kg i.v. followed by a 15-minute infusion 20, 40 and 100 mg/kg/h, respectively). Urinary volume, sodium and potassium excretions were significantly increased in both CHF and sham-operated control animals after alpha-trinositol administration compared with saline. Diuresis and natriuresis were observed also during co-administration of alpha-trinositol with NPY but not with norepinephrine (NE). In the pithed CHF rats, threshold doses of NPY potentiated the presser effects of endothelin-1 (ET-1) and angiotensin II (AII), but not preganglionic nerve stimulation or phenylephrine administration. Alpha-trinositol antagonized both the pressor response to NPY and the potentiation by NPY of pressor responses to effects of ET-1 and AIL Our data show that alpha-trinositol exhibis diuretic and natriuretic effects as well as vascular antagonistic effects on NPY in normal and CHF rats. These effects of alpha-trinositol may be due to an interaction with NPY mediated antidiuresis and antinatriuresis.

The modulatory effects of alpha-trinositol (D-myo-inositol-1.2.6- trisphosphate; PP 56) on the systemic arterial blood pressor responses induced by neuropeptide Y, preganglionic nerve stimulation, phenylephrine and vasopressin were studied in pithed rats. Intravenous administration (within 2 min.) of alpha-trinositol reduced the neuropeptide Y-induced increase in mean arterial pressure within a defined dose range without altering the heart rate. The influence of alpha-trinositol on the neuropeptide Y-induced pressor response in the presence of non-selective as well as H1- and H2-selective histamine antagonists (diphenhydramine, mepyramine and cimetidine respectively) were investigated. The maximal increase in mean arterial pressure induced by neuropeptide Y as well as the duration of the pressor response was enhanced after nonselective (diphenhydramine) or H1-selective (mepyramine) histamine blockade. The enhancement of the neuropeptide Y-induced pressor response by the H1 specific antagonist mepyramine was significantly more pronounced compared to the H2-selective agent. The exaggerated increase in mean arterial pressure in response to neuropeptide Y after histamine blockade was inhibited by alpha-trinositol to a similar extent as without such pretreatment. We conclude that neuropeptide Y interacts with histamine in the pithed rat and that this action may partially offset the pressor actions of the peptide. The neuropeptide Y-induced pressor responses may be inhibited by alpha-trinositol within a defined dose range indicating that this non-peptide agent may act as a functional inhibitor to neuropeptide Y in vascular tissue.

Since the discovery of neuropeptide Y which is co-stored and co-operate with noradrenaline (NA) in sympathetic nerve fibers, several scientific groups have searched for structures with neuropeptide Y antagonistic properties. Research has mainly focused on various peptide fragments which originate from or are related to the neuropeptide Y sequence. Some non-peptide antagonists have been proposed but they are mostly of low potency and non-selective. Our recent observations that alpha-trinositol (D-myo-inositol 1.2.6-trisphosphate) is an inhibitor of neuropeptide Y effects will hopefully lead to the development of useful non-peptide neuropeptide Y inhibitors. As a novel approach the highly selective approach of down-regulating neuropeptide Y receptors with antisense oligodeoxynucleotides is also discussed. Neuropeptide Y antagonistic agents would help us to understand the physiological role of neuropeptide Y and may serve as useful medication in circulation disorders.

In the present paper, the authors studied the effects of anionic polyelectrolyte sodium polystyrene sulfonate (PSS) on the cationic dye thioflavin T (ThT) fluorescence spectra in aqueous solution. The influencing factors of the formed thioflavin T excimer in the existence of PSS were investigated. Experimental results show that PSS could increases the local concentration of ThT by electrostatic attraction, therefore enhances excimer formation of thioflavin T. The fluorescence intensity of ThT monomer and excimer (I(M) and I(E)) changes regularly along with the change in temperature; ln(I(E)/I(M)) and 1/T have good linear relationship. Hereby we have developed a new type of fluorescence ratiometric sensor for temperature which could be recognized by naked eye, and it could effectively eliminate environmental effects such as instability of exciting light intensity and photobleaching, therefore improve the accuracy of identification.

Objective: The aim was to study the Gi protein mediated muscarinic signalling system in the myocardium of rats with chronic ischaemic heart failure. Methods: Chronic ischaemic heart failure was induced by myocardial ischaemia (four weeks after coronary artery ligation) in rats. The densities and agonist affinities of muscarinic receptors, and the functional activity and concentration of Gi proteins were studied. Results: In failing hearts, the activity of adenylyl cyclase stimulated by guanyliminodiphosphate (Gpp(NH)p) was decreased by 46%. Stimulated activities of adenylyl cyclase by both sodium fluoride and forskolin, however, remained unchanged. Carbachol depressed forskolin stimulated adenylyl cyclase more in membranes from failing hearts than those from normal hearts. The functional level of Gs protein as measured by a reconstitution assay in sarcolemmal membrane did not differ between the two groups. Furthermore, muscarinic receptors exhibited superhigh and low affinities for agonist in failing hearts whereas those in control hearts displayed only high and low affinities. No significant difference in the peptide equivalent amount of membrane bound Gi protein was found in either group. Conclusions: The experimental chronic failing heart due to myocardial ischaemia showed a depressed myocardial adenylyl cyclase signalling system. This may be due to the hypersensitivity of the Gi protein mediated muscarinic receptor-adenylyl cyclase system as shown by the increased inhibition of Gpp(NH)p mediated adenylyl cyclase, more potent inhibition of stimulated adenylyl cyclase by carbachol, and the superhigh afinity of the muscarinic receptors for carbachol. Mesenteric artery and cardiac ventricular endothelin receptors and endothelin-1-induced pressor responses were studied in normal rats and rats with chronic congestive heart failure induced by myocardial ischemia (4 weeks after coronary artery ligation). In mesenteric arteries of rats with chronic ischemic heart failure, endothelin receptor density was significantly decreased by 59%, whereas the dissociation constant was increased 2.8-fold, as compared with controls. There were, however, no changes in endothelin-receptor density or the dissociation constant in cardiac ventricular membrane preparations from rats with congestive heart failure as compared with controls. In pithed rats with congestive heart failure there was a reduced pressor response to a bolus injection of endothelin-1 (800 pmole/kg body weight), while the vasodilatory response was unaltered as compared with sham-operated controls. These results demonstrate that there is a decreased vascular endothelin-receptor function due to a down-regulated endothelin receptor. The in vivo data indicate that this is due to impaired endothelin A but not endothelin B receptor function. Thus, there is an impaired arterial but not cardiac ventricular endothelin receptor-mediated signalling system in the rat with chronic ischemic heart failure.

The effects of the novel ETA receptor antagonist, FR 139317, on ET-1 induced blood pressure changes were studied in the pithed Sprague-Dawley rat. FR139317 in a dose of 0.025 mg/kg b.w. had no effect while 0.05-1 mg/kg b.w. dose dependently inhibited the pressor response to an i.v. bolus injection of ET6-1 (800 pmoles/kg). While FR139317 potently inhibited the magnitide and duration of the ET-1 induced pressor response, the ETA antagonist did not significantly influence the shortlasting initial depressor response. We conclude that FR139317 in the pithed rat potently inhibits ET-1 mediated pressor responses and that this agent may become a significant tool to elucidate the putative physiological and pathophysiological role of ETA receptor mediated responses.

From the digital images of the red complex which resulted in the interaction of nitrite with N-(1-naphthyl) ethylenediamine dihydrochloride and P-Aminobenzene sulfonic acid, it could be seen that the solution colors obviously increased with increasing the concentration of nitrite ion. The JPEG format of the digital images was transformed into gray-scale format by origin 7.0 software, and the gray values were measured with scion image software. It could be seen that the gray values of the digital image obviously increased with increasing the concentration of nitrite ion, too. Thus a novel digital imaging colorimetric (DIC) method to determine nitrogen oxides (NO(x)) contents in air was developed. Based on the red, green and blue (RGB) tricolor theory, the principle of the digital imaging colorimetric method and the influential factors on digital imaging were discussed. The present method was successfully applied to the determination of the daily changes curve of nitrogen oxides in the atmosphere and NO2- in synthetic samples with the recovery of 97.3%-104.0%, and the relative standard deviation (RSD) was less than 5.0%. The results of the determination were consistent with those obtained by spectrophotometric method.

The fluorescence spectra of the succinic acid (7-hydroxy-coumarin) ester (SCE) of different concentration in water and ethanol solvents were studied. It was found that the peak wavelength changes not only with solvent, but also with the concentration. These phenomena of spectra have been discussed on the basis of solvent properties and dimerisation of SCE in hydrogen bonding solvents. The author thought that there are dimerisation phenomena in the SCE solutions at high concentration. The formation of dimer was caused by the aid of hydrogen bonding between SCE and solvent or between SCE themselves. Its acidic dissociation constant at excited state was determined based on the changes of the fluorescence spectra in water. And a simple and sensitive fluorescence quenching method for the determination of trace iron was developed. The method is based on the reaction of SCE with iron in hydrochloric acid medium. The fluorescence intensity was measured with excitation and emission wavelengths of 347 and 457 nm, respectively. The linear range for the determination of iron was 0.66-6.59 microg x L(-1). The detection limit was 51 ng x L(-1). The author contrasted SCE with 7-hydroxy-coumarin to determine Fe3+, and found the sensitivity of SCE is 10(4) higher than that of 7-hydroxy-coumarin. To our thinking there is a carboylate group in SCE, whose coordinate force with iron is stronger than hydroxide radical in 7-hydroxy-coumarin.

Fluorescent dyes butyl rhodamine B were assembled via a dl-cystenine intermediate onto quartz wafers whose surface had first adsorbed gold nanoparticles. Hence self-assembled sandwich bilayers with nanocomposite structure were constructed which can be used as a biosensor for bovine serum albumin. The biosensor-based self-assembled monolayers (SAMs) are regenerable and have high sensitivity, five orders of magnitude higher than that of bulk solution phase sensing. Quartz substrate was modified with self-assembled monolayer of gamma-aminopropyl-triethylsilane by aqueous phase silanization, giving an homogenous surface terminated with amino groups. Colloidal gold nanoparticles were further assembled on this functionalized monolayer substrate, forming a gold nanoparticles/APES/quartz nanocomposite structure. Flourescent reagent morin was assembled onto the self-assembled multilayer films of gold nanoparticles via DL-cystenine (Cys) intermediate. The morin-modified self-assembled multilayer films of gold nanoparticles could detect triphenyltin fluorimetrically with high sensitivity, and the detection limit was as low as 1.297 x 10(-8) mol x L(-1). The effects of various factors such as concentration of solution, pH, and assembling time were investigated. The response mechanism behind caused the enhancement of relative fluorescence intensity was also discussed.

In this paper, an internal conversion (IC) fluorescence probe N-(o-boronic acid)benzyl-1-naphthylamine (BBNA) was prepared from 1-naphthylamine and 2-formylbenzeneboronic acid. The fluorescence parameters of BBNA were investigated in a variety of solvents. When BBNA interacted with D-fructose in phosphate buffer solution of 30% MeOH, pH 8.21 (v/v), the fluorescence intensity increased and emission maximum red-shifted slightly with increasing D-fructose concentration. In the presence of D-fructose, the fluorescence quantum yield of BBNA increased with increasing solvent polarity, suggesting that internal conversion (IC) occurred with BBNA. The binding force of BBNA with d-fructose was the strongest, and the stability constant (K) of D-fructose was 99.9 mol/L. Therefore, a selective recognition system based on IC was constructed for D-fructose.

OBJECTIVE: Congestive heart failure (CHF) is accompanied by impaired peripheral blood flow and endothelial dysfunction with decreased release of nitric oxide (NO). Strong evidence supports the existence of another vasodilatory substance, endothelium derived hyperpolarising factor (EDHF), which has not previously been studied in CHF. METHOD: CHF was induced by left coronary artery ligation resulting in a reproducible myocardial infarction in Sprague Dawley rats. Vasodilatory responses to acetylcholine and extracellular nucleotides (ATP, ADP beta S, ADP and UTP) were examined in cylindrical segments of the mesenteric artery, precontracted with noradrenaline. The combined NO- and EDHF-dilatation (after inhibition of cyclo-oxygenase pathways) was called "total dilatation", as indomethacin had only minor effects in this system. NO-dilatation was studied in segments pretreated with indomethacin and the potassium channel inhibitors charybdotoxin (10(-7.5) M) and apamin (10(-6) M), while EDHF-dilatations were studied in the presence of indomethacin (10(-5) M) and L-NOARG (10(-3.5) M). RESULTS: EDHF-dilatations in CHF were strongly up-regulated for ACh (36% vs. 73%; sham vs. CHF operated rats), ADP beta S (10% vs. 42%), ADP (0% vs. 21%) and UTP (3% vs. 35%). These dilatations were abolished by a combination of charybdotoxin and apamin, confirming that they were mediated by EDHF. The NO-dilatations on the other hand were down-regulated in CHF as compared to sham operated rats for ACh (93% vs. 76%; sham vs. CHF operated rats), ADP beta S (61% vs. 37%). ADP (60% vs. 30%), ATP (49% vs. 34%) and UTP (65% vs. 47%), while a minor decrease was seen in the total dilatation for ACh (87% vs. 75%; sham vs. CHF operated rats), ADP beta S (47% vs. 42%), ADP (59% vs. 39%), ATP (52% vs. 39%) and UTP (59% vs. 44%). CONCLUSION: In this model of non-atherosclerotic CHF there was a minor decrease in the total dilatation and a marked down-regulation of the NO-mediated dilatation, while the EDHF-dilatation was up-regulated. Increased EDHF-activity in CHF may represent a compensatory response to decreased NO-activity to preserve endothelial function and tissue perfusion. (Less)

Neuropeptide Y (NPY) has been shown to potentiate the effects of various vasoactive agents in both in vitro and in vivo experiments. The present study was designed to investigate the potentiation effects of NPY on various vasoconstrictive agents and the influence of NPY on the dilatation effects of endothelin-1 in rats with congestive heart failure (CHF). CHF was induced by left coronary artery ligation. Sham-operated rats subjected to thoracotomy served as normal controls. Experiments in conscious rats were performed 4-6 weeks after coronary artery ligation or sham operation. The pressor responses of intravenous phenylephrine (12.5 nmol/kg), endothelin-1 (400 pmol/kg) and angiotensin II (10 ng) but not tyramine (40 micrograms) were significantly decreased in CHF rats compared with sham-operated rats (p < 0.01). In sham-operated rats, subthreshold dose of NPY (0.1 microgram/kg/min) potentiated the pressor responses of all the agonists (p < 0.05). In CHF rats, significant enhancement of mean arterial pressure (MAP) by subthreshold dose of NPY was observed with angiotensin II (p < 0.05). The MAP was enhanced by 45.4% in CHF and 40.6% in sham-operated rats respectively. NPY did not enhance the pressor responses induced by phenylephrine, endothelin-1 or tyramine in CHF rats. The initial decrease of MAP after bolus injection of endothelin-1 was observed in both CHF and sham-operated control rats, and magnitude of this response was similar in both groups. Subthreshold dose of NPY significantly reduced the vasodilatation effect of endothelin-1 in CHF (p < 0.05) but not in normal control rats. We conclude that NPY potentiates pressor effects of angiotensin II and reduces vasodilatation effects of endothelin-1 in rats with CHF. These effects of NPY may contribute to the increased vascular tone in CHF.

Binding experiments show that ZD 7155 is a potent angiotensin II type 1 receptor antagonist. In this study this novel substance was studied in normotensive and hypertensive rats. The relative potency and duration of the antihypertensive effects of ZD 7155 were compared with those of the reference substance, losartan. The inhibitory effects of both compounds on angiotensin II-induced pressor actions were studied in the conscious normotensive Sprague-Dawley (SD) rat and in the conscious, spontaneously hypertensive rat (SHR). Arterial blood pressure and heart rate (HR) were obtained by direct intraarterial recording. Angiotensin II infusion was administered intravenously in the dose range 53.3 ng—12.8 μg kg−1 min−1 to the conscious rats. ZD 7155 was administered in a bolus dose of 1.082 μmol kg−1 (0.51 mg kg−1) and losartan in bolus doses of 2.165 and 6.495 μmol kg−1 (1.0 and 3.0 mg kg−1). In conscious SD rats, ZD 7155 and losartan behaved as competitive antagonists and the pressor response curve to angiotensin II was shifted to the right. Experiments in conscious SD rats also showed that ZD 7155 was approximately ten times as potent as losartan in suppressing the angiotensin II-induced pressor response (240 ng kg−1; 10 min infusion). In addition, experiments with conscious rats demonstrated that ZD 7155 could suppress the angiotensin II-induced pressor response for approximately 24 h when ZD 7155 was administered intravenously in a 1.082 μmol kg−1 bolus dose and angiotensin II was given at 240 ng kg−1 (in a 10−min infusion). Experiments in conscious SHRs using ZD 7155 (1.082 μmol kg−1) and losartan (6.495 μmol kg−1) as intravenous boluses indicated that both ZD 7155 and the reference compound losartan exhibited a significant antihypertensive effect. These results demonstrate that ZD 7155 is a potent angiotensin II-type 1 antagonist which is approximately ten times as potent as losartan in suppressing the angiotensin II-induced pressor response. Furthermore, ZD 7155 may suppress the angiotensin II-induced pressor response for 24 h and in the SHR ZD 7155 induces a pronounced and persistent antihypertensive effect.

A novel dual-fluorescence quantum dots (QD) nanocomposite with tuning emission wavelength and fluorescence intensity was synthesized, in which CdS and CdTe were the internal standard and probe, respectively. This nanocomposite exhibited good photobleaching and pH stability, and exhibited selective sensing for Hg(2+) with a detection limit (3SD/k) of 5.6 nM. Based on the blue background emitted by the internal standard CdS, a novel visual fluorescence detection method has been established, and can be used for the qualitative and semi-quantitative colorimetric analysis of Hg(2+).

The objective of this study was to compare the cardiovascular and renal effects of the two gamma-melanocyte-stimulating hormone (MSH) peptide sequences in the pro-opiomelanocortin prohormone structure in conscious, anesthetized, and pithed spontaneous hypertensive rats (SHR) and normotensive Wistar Kyoto (WKY) controls. In the conscious but not in the pithed rats, intravenous injection of gamma 2- and gamma 1-MSH induced a rapid and dose-dependent increase in mean arterial pressure (MAP), and gamma 2-MSH was more potent than gamma 1-MSH. The pressor response was more pronounced and more sustained in the SHR compared with the WKY. There were dose-dependent and significant increases in heart rate (HR) after gamma 2- and gamma 1-MSH in the SHR. At intravenous infusions of low doses of gamma 2-MSH, which did not significantly influence MAP or HR, urinary sodium excretion was significantly increased in both SHR and WKY. In conscious, but not in anesthetized rats, intracerebroventricular administration of the gamma 2-MSH peptide induced sustained increases in MAP in both SHR and WKY. After intrathecal administration, there were transient pressor effects of gamma 2-MSH. We conclude that the pro-opiomelanocortin-derived gamma 2- and gamma 1-MSH peptide sequences possess potent rapid pressor actions. The pressor effects, which require an intact sympathetic nervous system, are more pronounced in the SHR strain. Moreover, gamma 2-MSH induces natriuresis when administered in nonpressor doses in WKY and SHR.

The antagonistic effects of a new inositol phosphate derivative, D-myoinositol-1,2,6-trisphosphate (PP56), on pressor responses to preganglionic sympathetic nerve stimulation and exogenously administered phenylephrine or neuropeptide Y (NPY) were investigated in vivo in the pithed rat. In this model an intravenous (i.v.) bolus administration of PP56 (1-50 mg/kg) dose dependently inhibited the increase in mean arterial blood pressure (MAP) induced by a continuous infusion of NPY (2 micrograms/kg per min for 10 min). PP56 in a dose of 5 mg/kg i.v. bolus reduced the entire NPY dose-response curve (0.4-8 microgram/kg per min 10 min infusion) without any shift to the right indicating a non-competitive interaction. Furthermore, PP56 (10-50 mg/kg i.v.) was found to inhibit the pressor response to preganglionic sympathetic nerve stimulation and i.v. bolus injection of the alpha 1-adrenoceptor agonist, phenylephrine. The dose-response curves for increasing doses of phenylephrine and incremental preganglionic sympathetic nerve stimulation were not significantly altered by a lower dose of PP56 (5 mg/kg i.v. bolus). We conclude that PP56, representing a new class of synthetic drugs, can antagonize the actions of exogenous and endogenous NPY in vivo, an action which is specific for NPY within a limited dose range.

We wanted to study the expression of P2-receptors at the mRNA-level in the heart and if it is affected by congestive heart failure (CHF). To quantify the P2 receptor mRNA-expression we used a competitive RT-PCR protocol which is based on an internal RNA standard. The P2 receptor mRNA-expression was quantified in hearts from CHF rats and compared to sham-operated rats. Furthermore, the presence of receptor mRNA was studied in the myocardium from patients with heart failure. In the sham operated rats the G-protein coupled P2Y-receptors were expressed at a higher level than the ligand gated ion-channel receptor (P2X1). Among the P2Y-receptors the P2Y6-receptor was most abundantly expressed (P2Y6 > P2Y1 > P2Y2 = P2Y4 > P2X1). A prominent change was seen for the P2X1- and P2Y2-receptor mRNA levels which were increased 2.7-fold and 4.7-fold respectively in the myocardium from the left ventricle of CHF-rats. In contrast, the P2Y1-, P2Y4- and P2Y6-receptor mRNA levels were not significantly altered in CHF rats. In human myocard the P2X1-, P2Y1-, P2Y2-, P2Y6- and P2Y11-receptors were detected by RT-PCR in both right and left atria and ventricles, while the P2Y4-receptor band was weak or absent. In conclusion, most of the studied P2-receptors were expressed in both rat and human hearts. Furthermore, the P2X1- and P2Y2-receptor mRNA were upregulated in CHF, suggesting a pathophysiological role for these receptors in the development of heart failure.

Neurohormonal changes in congestive heart failure (CHF) include an enhanced peripheral sympathetic nerve activity which results in increased release of noradrenaline, neuropeptide Y and ATP. To examine if such changes in CHF would modulate peripheral pre- and postsynaptic receptors of ATP and its degradation product adenosine, experiments were performed in a rat model of ischaemic CHF. In this model, ischaemia was induced in rats by ligation of the left coronary artery. Our results demonstrate that there is a selective downregulation of P2X receptor-mediated pressor effects, while the hypotensive effects mediated by the endothelial P2Y receptors are unaffected in CHF. Moreover, the adenosine-mediated inhibitory effects on heart rate and blood pressure were also attenuated in the CHF rats. The most important changes in adenosine and P2-receptor function induced by ischaemic CHF were the reduced pressor effect mediated by the P2X receptor and the increased heart rate due to an attenuated inhibitory effect of adenosine.
