Atherosclerosis is a chronic inflammatory disease. The triggering receptor expressed on myeloid cells-1 (TREM-1) plays a crucial role in inflammatory diseases; recently, it was identified as a major upstream proatherogenic receptor, but its mechanism is still unclear. In this study, we explore the role of TREM-1 on dendritic cells maturation and inflammatory responses induced by ox-LDL and its possible mechanism. Human dendritic cells were differentiated from blood monocytes and treated with ox-LDL. Naive autologous T cells were cocultured with pretreated DCs or treated directly. The expression of TREM-1 and inflammatory factors were evaluated by real-time PCR, western blot, and ELISA methods. And the expression of immune factors to evaluate the DCs maturation and T-cell activation were determined by the FACS. Our study showed that ox-LDL induced TREM-1 expression, DC maturation, and T-cell activation. T cells exposed to ox-LDL-treated DCs produced interferon-γ and interleukin-17 (IL-17). Blocking TREM-1 suppressed the DC maturation, showing lower expression of CD1a, CD40, CD86, CD83, and HLA-DR, and limited their production of tumor necrosis factor-alpha (TNF-α), IL-1β, IL-6, and monocyte chemoattractant protein-1 (MCP-1), meanwhile increased transforming growth factor-β(TGF-β) and IL-10 production. Ox-LDL induced miR-155, miR-27, Let-7c, and miR-185 expression; however, TREM-1 inhibiting decreased miRNA-155 expression. Furthermore, silencing miRNA-155 restores SOCS1 repression induced by ox-LDL. Experiments with T cells derived from carotid atherosclerotic plaques or healthy individuals showed similar results. Our results uncover a new link between ox-LDL and TREM-1 and may provide insight into this interaction in the context of atherosclerosis.

Cuticular proteins (CPs) play important roles in insect growth and development. However, it is unknown whether CPs are related to heat tolerance. Cnaphalocrocis medinalis, a serious pest of rice, occurs in summer and exhibits strong adaptability to high temperature, but the underlying mechanism is unclear. Here, the role of CP genes in heat acclimation was studied. Heat tolerance of the heat-acclimated larvae was significantly stronger than the unacclimated larvae. The cuticular protein content in the heat-acclimated larvae was higher than that of the unacclimated larvae. 191 presumed CP genes of C. medinalis (CmCPs) were identified. Expression patterns of 14 CmCPs were different between the heat acclimated (S39) and unacclimated (S27) larvae under heat stress. CmCPs were specifically expressed in epidermis and the head except CmCPR20 mainly expressed in Malpighian tubules. CmCPR20 was upregulated in S39 while downregulated in S27, but CmTweedle1 and CmCPG1 were upregulated in S27 and downregulated in S39. RNAi CmTweedle1 or CmCPG1 remarkably decreased heat tolerance and cuticular protein content of the heat-acclimated larvae but not the unacclimated larvae. RNAi CmCPR20 decreased heat tolerance and cuticular protein content of the unacclimated larvae but not the heat-acclimated larvae. CmTweedle1 and CmCPG1 genes involve heat acclimation of C. medinalis.

Wing dimorphism is a fascinating feature of the ability of insects to adapt to environments. The brown planthopper (BPH) Nilaparvata lugens, a serious pest of rice, can switch between the long- and short-winged morphs. It has been known that environmental factors can affect the wing morph of BPH. However, it is still unclear whether the effect of environment is dependent on BPH genetic backgrounds or not. In the present study, we established the pure-bred lineages of short- and long-winged BPHs via multigenerational selection, and we found that survival and fecundity were similar between these 2 lineages. Wing morphs of the pure-bred lineages were almost fully dependent on genetics, but independent of the environmental factors, nymphal density and rice plant stage, 2 key factors affecting BPH wing morphs. In the unselected BPH population, short- and long-winged morphs were produced depending on those 2 environmental factors, indicating the contribution of environment to wing morph. In the wing-selected lineages, 4 developmental regulated genes of wing, NlInR1, NlInR2, NlAkt, and NlFoxo were expressed stably in the short-winged adults, but almost silenced in the long-winged adults. However, all these genes were expressed normally with a similar level in both the short- and long-winged adults in an unselected population except NlFoxo. The pure-bred lineages of long- and short-winged morphs exhibited different expression patterns of wing development-regulated genes, suggesting the genetic determination of wing morphs. Effects of environmental factors on wing morphs occurred only in the genetic mix population.

Objective: To observe the efficacy and imaging of surgical treatment of thoracolumbar fractures via the paravertebral muscle space approach. Methods: A retrospective analysis was conducted on patients with thoracolumbar fractures receiving surgery in Baoding First Central Hospital from January 2019 to December 2020. According to different surgical approaches, they were divided into paravertebral approach group, posterior median approach group and minimally invasive percutaneous approach group. They received surgery via the paravertebral muscle space approach, posterior median approach and minimally invasive percutaneous approach, respectively. Results: Statistically significant differences were found in surgical duration, intraoperative bleeding volume, intraoperative fluoroscopy frequency, postoperative drainage volume and hospital stay among the three groups. One year after surgery, the VAS, ADL and JOA scores of the paravertebral approach group and the minimally invasive percutaneous approach group had statistically significant differences from the posterior median approach group (P < 0.05). Conclusion: For the surgical treatment of thoracolumbar fractures, the clinical efficacy of the paravertebral muscle space approach is superior to that of the traditional posterior median approach, and the clinical efficacy of the minimally invasive percutaneous approach is similar to that of the posterior median approach. All the three approaches can effectively improve the postoperative function and pain symptoms of patients without increasing the incidence of complications. Compared with the posterior median approach, the surgery via the paravertebral muscle space and minimally invasive percutaneous approaches presents shorter surgical duration, less bleeding and shorter hospital stay, which is more conducive to postoperative recovery of patients. doi: https://doi.org/10.12669/pjms.39.3.3295 How to cite this: Li H, Liu X, Ma W, Wang S. Transmission rate and role in hosts contribute to the prevalence of an endosymbiont. However, factors affecting transmission and role of facultative endosymbionts are still not well understood. Here, we illustrated that host plants and environmental temperatures affected the transmission, relative abundance and role of Arsenophonus in the cotton aphid Aphis gossypii. The transmission rate of this endosymbiont from mother aphids to offspring was relatively lower. High temperatures impeded the transmission, and infection rates declined as aphids were exposed to 30°C. Contents of amino acids and secondary metabolites were remarkably different among host plants. Aphids feeding on zucchini leaves containing a higher titre of amino acids and lower secondary metabolites harboured a relatively lower abundance of Arsenophonus. Concentrations of an amino acid and a plant secondary metabolite, cucurbitacin B, in aphid diet were not associated with Arsenophonus abundance. However, gossypol, another plant secondary metabolite, was strongly related with the abundance. Arsenophonus imparted a fitness benefit to aphids, and the benefit was dependent on host plants and gossypol concentration. In sum, plant secondary metabolite and environmental temperature affect transmission, relative abundance and role of Arsenophonus, which determine the endosymbiont prevalence in aphid populations.

Background: Modern medical researches show that the health of a population is closely related to their behavior, environmental factors, social and economic conditions, aging and genetic susceptibility. Therefore, it is very important to study the relationship between these factors and health of a population. Objective: This study reviews the latest research progress in population health management and life/disease risk assessment, analyzes the existing problems in relevant researches, and proposes some improvement plans to solve the problems. Methods: It also introduces the general methods of human health management and life/disease risk assessment, analyzes the defects in these methods in practical application, and then draws conclusions regarding industrial system equipment health management and residual life prediction, and puts forward some new ideas to solve the problems. Results: In the study of health assessment, although self-assessment of health is a common method of determining health variables, its subjectivity and inaccuracy are problems that cannot be ignored. In life/disease risk assessment studies, medical effect and efficacy are mainly evaluated, and little consideration is given to other factors, such as behaviour, environment, and socio-economic status. In the construction of statistical graphs, great individual differences, internal symbiotic variables and external impact variables are seldom considered. Conclusion: This study helps to understand the basic ideas and methods of health management and life/disease risk assessment, and proposes some future research programs based on the author's knowledge.

Tissue damage in diabetes is at least partly due to elevated reactive oxygen species production by the mitochondrial respiratory chain during hyperglycemia. Sustained hyperglycemia results in mitochondrial dysfunction and the abnormal expression of mitochondrial genes, such as NADH: Ubiquinone oxidoreductase subunit A13 (NDUFA13). Metformin, an AMP‑activated protein kinase (AMPK) activator, protects cardiomyocytes from oxidative stress by improving mitochondrial function; however, the exact underlying mechanisms are not completely understood. The aim of the present study was to investigated the molecular changes and related regulatory mechanisms in the response of H9C2 cardiomyocytes to metformin under high glucose conditions. H9C2 cells were subjected to CCK‑8 assay to assess cell viability. Reactive oxygen species generation was measured with DCFH‑DA assay. Western blotting was used to analyze the expression levels of NDUFA13, AMPK, p‑AMPK and GAPDH. Reverse transcription‑quantitative PCR was used to evaluate the expression levels of mitochondrial genes and transcription factors. It was observed that metformin protected H9C2 cardiomyocytes by suppressing high glucose (HG)‑induced elevated oxidative stress. In addition, metformin stimulated mitochondrial biogenesis, as indicated by increased expression levels of mitochondrial genes (NDUFA1, NDUFA2, NDUFA13 and manganese superoxide dismutase) and mitochondrial biogenesis‑related transcription factors [peroxisome proliferator‑activated receptor‑gamma coactivator‑1α, nuclear respiratory factor (NRF)‑1, and NRF‑2] in the metformin + HG group compared with the HG group. Moreover, metformin promoted mitochondrial NDUFA13 protein expression via the AMPK signaling pathway, which was abolished by pretreatment with the AMPK inhibitor, Compound C. The results suggested that metformin protected cardiomyocytes against HG‑induced oxidative stress via a mechanism involving AMPK, NDUFA13 and mitochondrial biogenesis.

Flexibility and variations in morphological and physiological traits are the key for insect herbivores to adapt to environments. The cotton–melon aphid Aphis gossypii Glover is a polyphagous insect which populations exhibit host specialization. Feeding experience on specific host plants converted host specialization in this aphid. However, adaptive changes of this aphids during the host conversion are still unclear. Here, we examined variations in body length, leg segments, and host preference of the cotton-specialized and cucurbit-specialized aphids, A. gossypii, induced by novel host plants. We found that on the original host plants, body, femur, tibia, and tarsus of the cucurbit-specialized aphids were significantly longer than these of the cotton-specialized aphids. However, feeding on zucchini or cowpea resulted in the short body and leg segments in the cucurbit-specialized aphids, whereas led to the long body and leg segments or no changes in the cotton-specialized aphids, and consequently, the tarsus lengths became equal between the cotton- and cucurbit-specialized aphids. Interestingly, the change trends of the relative lengths of leg segments to body length with the increase of reared generation on zucchini were almost different between the cucurbit-specialized and cotton-specialized aphids, especially the relative length of tarsus, but on cowpea, the trends were similar. Meanwhile, feeding on zucchini altered host preference of the cotton-specialized aphids, but not of the cucurbit-specialized aphids. Novel host plants induce adaptive changes in leg segments and host preference of aphids, suggesting the conversion of host specialization.

The rice leaf folder (RLF) Cnaphalocrocis medinalis, a serious pest of rice, is vulnerable to high temperature. However, population outbreak of RLF became more frequent during first twenty years of the 21st century, suggesting that this pest might have adapt to the frequent heat stress under global warming. RLF adults are more liable to heat stress than larvae. Here, we hypothesized that besides migration, adults might have other ways to cope with heat stress. Therefore, distribution patterns of female and male adults was observed under gradually increasing temperatures, and the effect of heat exposure on mating frequency, longevity, ovary development, fecundity of adults, and hatchability of eggs were compared when female and male adults were separate (♀♂-separate mode) and coexistent (♀♂-coexistent mode) during heat exposure. The result showed that the separate pattern of female and male moths gradually appeared as temperature increased from 27 to 39 °C. The mating frequency of females was significantly lower in the ♀♂-coexistent mode than that in the ♀♂-separate mode. The ♀♂-coexistent mode led to the shorter longevity, slower ovary development, lower fecundity of adults, and lower hatchability of eggs, compared to the ♀♂-separate mode, indicating that coexistence of female and male adults was adverse to reproductive performance of RLF under heat stress. Female and male adults might adopt a separate pattern to deal with a short-term high temperature.

Sex ratio bias is common in migratory animals and can affect population structure and reproductive strategies, thereby altering population development. However, little is known about the underlying mechanisms that lead to sex ratio bias in migratory insect populations. In this study, we used Cnaphalocrocis medinalis, a typical migratory pest of rice, to explore this phenomenon. A total of 1,170 moths were collected from searchlight traps during immigration periods in 2015–2018. Females were much more abundant than males each year (total females: total males = 722:448). Sex-based differences in emergence time, take-off behaviour, flight capability and energy reserves were evaluated in a laboratory population. Females emerged 0.78 days earlier than males. In addition, the emigratory propensity and flight capability of female moths were greater than those of male moths, and female moths had more energy reserves than did male moths. These results indicate that female moths migrate earlier and can fly farther than male moths, resulting more female moths in the studied immigratory population.

The relational measurement based on Markov-switching GRG copula constructed by this paper is harnessed to analyze the dependence structure between WTI (BRENT) crude oil futures price and 12 kinds of Chinese agricultural commodity futures prices. The empirical results show that there exist two structural states of Markov switching between the futures prices of different agricultural commodities and crude oil futures price. The two states have different duration periods, and the degree of correlation with crude oil futures prices varies under different agricultural commodity futures prices. Among all the 12 kinds of agricultural commodity futures, 11 kinds of agricultural commodity futures prices mainly present positive correlations with crude oil futures prices, although the positive correlation differs between non-extreme and extreme conditions. The remaining agricultural commodity futures price is not related to crude oil futures prices.

Endosymbionts play important roles in protecting hosts from environmental stress, such as natural enemies, heat, and toxins. Many insects are infected with the facultative nonessential endosymbionts Wolbachia and Rickettsia, which are the crux in this review, although other relevant symbiont genera will also be treated. Insecticide resistance of hosts can be related to infections with Wolbachia and Rickettsia. These endosymbionts commonly increase host susceptibility to chemical insecticides, but cases of increased resistance also exist. The symbiont-mediated insecticide resistance/susceptibility varies with species of insect, species of symbiont, and chemical compound. Changes in insecticide resistance levels of insects can be associated with fluctuations in population density of endosymbionts. Effects of endosymbionts on host fitness, metabolism, immune system, and gene expression may determine how endosymbionts influence insecticide resistance. A clearer understanding of these interactions can improve our knowledge about drivers of decreasing insecticide resistance.

The aim of the present study was to evaluate the biomechanical mechanism of injuries of the thoracolumbar junction by the methods of a backward fall simulation experiment and finite element (FE) analysis (FEA). In the backward fall simulation experiment, one volunteer was selected to obtain the contact force data of the sacrococcygeal region during a fall. Utilizing the fall data, the FEA simulation of the backward fall process was given to the trunk FE model to obtain the stress status of local bone structures of the thoracolumbar junction during the fall process. In the fall simulation test, the sacrococcygeal region of the volunteer landed first; the total impact time was 1.14±0.58 sec, and the impact force was up to 4,056±263 N. The stress of thoracic (T)11 was as high as 42 MPa, that of the posterior margin and the junction of T11 was as high as 70.67 MPa, and that of the inferior articular process and the superior articular process was as high as 128 MPa. The average stress of T12 and the anterior margin of lumbar 1 was 25 MPa, and that of the endplate was as high as 21.7 MPa, which was mostly distributed in the back of the endplate and the surrounding cortex. According to the data obtained from the fall experiment as the loading condition of the FE model, the backward fall process can be simulated to improve the accuracy of FEA results. In the process of backward fall, the front edge of the vertebral body and the root of vertebral arch in the thoracolumbar junction are stress concentration areas, which have a greater risk of injury.

Background Gastric cancer (GC) remains an important cancer worldwide. Further understanding of the molecular mechanisms of gastric carcinogenesis will enhance the diagnosis and treatment of GC. Methods The expression of DLEU2 and ETS2 was analyzed in several GC cell lines using GEPIA online analyze, qRT-PCR and immunohistochemistry. The biological behavior of GC cells was detected by CCK8, clone formation, transwell, wound healing, western blot, and flow cytometry assay. More in-depth mechanisms were studied. Results DLEU2 was significantly up-regulated in GC tissues and cell lines. The expression of DLEU2 was significantly associated with pathological grading and TNM stage of GC patients. Furthermore, knockdown of DLEU2 inhibited the proliferation, migration, and invasion of AGS and MKN-45 cells, while overexpression of DLEU2 promoted the proliferation, migration, and invasion of HGC-27 cells. MiR-30a-5p could directly bind to the 3’ UTR region of ETS2. Moreover, DLEU2 bound to miR-30a-5p through the same binding site, which facilitated the expression of ETS2. Knockdown of DLEU2 reduced the protein level of intracellular ETS2 and inhibited AKT phosphorylation, while overexpression of DLEU2 induced the expression of ETS2 and the phosphorylation of AKT. ETS2 was highly expressed in GC tissues. The expression of ETS2 was significantly associated with age, pathological grading, and TNM stage. ETS2 overexpression promoted cell proliferation and migration of AGS and MKN-45 cells. Furthermore, ETS2 overexpression rescued cell proliferation and migration inhibition induced by DLEU2 down-regulation and miR-30a-5p up-regulation in AGS and MKN-45 cells. Conclusions DLEU2 is a potential molecular target for GC treatment.

Gel-mediated crystallization is a common system to produce self-organized materials, which is fundamental to the development of bottom-up approaches to functional complex materials. Mineralization in hydrogel matrices nevertheless remains empirical in the generation of crystallization products with tailored heterogeneous structures. We demonstrate that the employment of the hydrogels with proper cationic diffusivity can trigger the consecutive growth of oriented, granular-rhombohedral heterogeneous structures. The controllable morphogenesis leads to continuous calcitic CaCO3 films comprising spatial heterogeneity, where epitaxial match assumedly favors the successive deposition of both granular and rhombohedral layers. The scenario of consecutive growth is disclosed, where the thickness of the granular layers can become a valuable indicator to reflect the retardancy degree of crystallization. The evaluation of the physicochemical properties of the hydrogels finally establishes a direct correlation between the cationic diffusivity of the hydrogels and the appearance of the heterogeneous structures. The current work therefore sheds light on the implementation of rational morphogenetic approaches to crystalline materials with tailored complex architectures.

This systematic review aimed to assess the efficacy of occlusal splints in the treatment of temporomandibular disorders (TMDs).This systematic review was conducted according to Preferred Reporting Items for Systematic Reviews and Meta-Analysis (PRISMA) guidelines. Four databases (MedlineEleven studies were included. The sample size ranged from 12 to 96 subjects. The male to female ratio was 0 to 25%. The mean length of follow-up was 4 months. Occlusal splint had a positive effect on mandibular movements in all included studies. Seven studies showed a positive effect of occlusal splint on chronic pain reduction and pain intensity, while two others showed improvement of temporomandibular joint clicking sounds and locking of the jaws. Moreover, improvements in mouth opening, depression, and anxiety symptoms, were reported in four studies.An occlusal splint can be considered as a non-invasive treatment approach for patients with TMD, especially those with signs and symptoms of restriction of mandibular movement and pain. Moreover, the present findings highlighted an urgent need of a standardised consensus regarding the prognostic evaluation of TMD.

Maternal stress during pregnancy exposes fetuses to hyper glucocorticoids (GC), which increases the risk of metabolic dysfunctions in offspring. Despite of being a key tissue for maintaining metabolic health, the impacts of maternal excessive GC on fetal brown adipose tissue (BAT) development and its long-term thermogenesis and energy expenditure remain unexamined. To test, pregnant mice were administrated with dexamethasone (DEX), a synthetic GC, in the last trimester of gestation, when BAT development is the most active. DEX offspring had glucose, insulin resistance and adiposity, also displayed cold sensitivity following cold exposure. In BAT of DEX offspring, Ppargc1a expression was suppressed, together with reduced mitochondrial density, and the brown progenitor cells sorted from offspring BAT demonstrated attenuated brown adipogenic capacity. Increased DNA methylation in Ppargc1a promoter had a fetal origin because elevated DNA methylation was also detected in neonatal BAT and brown progenitors. Mechanistically, fetal GC exposure increased GC receptor (GR)/ DNMT3b complex in binding to the Ppargc1a promoter, potentially driving its de novo DNA methylation and transcriptional silencing, which impaired fetal BAT development. In summary, maternal GC exposure during pregnancy increases DNA methylation in the Ppargc1a promoter, which epigenetically impairs BAT thermogenesis and energy expenditure, predisposing offspring to metabolic dysfunctions.

Behavioral thermoregulation is a key strategy for insects to cope with heat stress. The rice leaf folder Cnaphalocrocis medinalis Guenée (Lepidoptera: Pyralidae) larvae usually fold one leaf to construct a leaf shelter. The larvae are vulnerable to heat stress, and the temperature in summer is often beyond the optimal range of them. Shelters confer protection against environmental stress but unclear whether larvae will alter shelter-building behavior when encountering heat stress. We observed the shelter-building behavior of larvae during and after heat shock, and then examined the shape and structure of shelters. Larvae spent more time in selecting a site and building a shelter during and after heat shock than at the optimal temperature. More than 70% of larvae folded two or three leaves to build a shelter during and after heat shock, but more than 60% of larvae only folded one leaf at the optimal temperature. Larvae built more single-leaf longitudinal shelters at the optimal temperature, but they built more multileaf overlapping shelters during and after heat stress. Larvae constructed a short leaf shelter using a small amount of silk binds when they were exposed to 40°C for 4 h. The rice leaf folder larvae can alter their shelter-building behavior and shelter structure in response to heat stress.
