14. Efficacy and safety of diacerein monotherapy in adults with obesity: A randomized, double‐blind, placebo‐controlled trial.

Abstract

Aim Methods Results Conclusions To assess the efficacy and safety of diacerein monotherapy in adults with obesity.Forty‐two adults with obesity participated in the study and were randomly assigned to receive diacerein or placebo in addition to lifestyle modification for 14 weeks, in a double‐blinded fashion. Differences in changes in body weight, body composition, metabolic variables, fatty liver‐related indicators, cardiovascular system variables, lifestyle score and metabolic factors were compared.Post‐treatment weight loss percentage from baseline was −6.56% (−8.71%, −4.41%) in the diacerein group and −0.59% (−2.74%, 1.56%) in the placebo group. Compared with the placebo group, the diacerein group showed significant improvements in body composition, metabolic variables and indicators related to fatty liver. In addition, after 14 weeks of treatment, diacerein led to a significant reduction in serum visfatin concentration versus the placebo group. The reductions in total body fat mass and visceral fat area mediated the weight loss induced by diacerein. No significant differences were found between the groups in the number of adverse events and safety variables.For adults with obesity, diacerein led to a clinically meaningful weight loss and provided multiple metabolic benefits with acceptable safety. Abstract

This study combined population genetics and parentage analysis to obtain foundational data for the conservation of Magnolia kwangsiensis. M. kwangsiensis is a Class I tree species that occurs in two disjunct regions in a biodiversity hotspot in southwest China. We assessed the genetic diversity and structure of this species across its distribution range to support its conservation management. Genetic diversity and population structure of 529 individuals sampled from 14 populations were investigated using seven nuclear simple sequence repeat (nSSR) markers and three chloroplast DNA (cpDNA) fragments. Parentage analysis was used to evaluate the pollen and seed dispersal distances. The nSSR marker analysis revealed a high genetic diversity in M. kwangsiensis, with an average observed (Ho) and expected heterozygosities (He) of 0.726 and 0.687, respectively. The mean and maximum pollen and seed dispersal distances were 66.4 and 95.7 m and 535.4 and 553.8 m, respectively. Our data revealed two distinct genetic groups, consistent with the disjunct geographical distribution of the M. kwangsiensis populations. Both pollen and seed dispersal movements help maintain genetic connectivity among M. kwangsiensis populations, contributing to high levels of genetic diversity. Abstract

Maize is a crucial staple crop that ensures global food security by supplying essential nutrients. However, heavy metal (HM) contamination inhibits maize growth, reduces output, and affects food security. Some endophytic fungi (EFs) in maize seeds have the potential to enhance growth and increase dry biomass, offering a solution to mitigate the negative effect of HM contamination. Using these functional EFs could help maintain crop production and ensure food safety in HM-contaminated areas. In the present study, the diversity of EFs in corn grains from various HM-contaminated areas in China was studied through culture-dependent and culture-independent methods. We tested the plant growth-promoting (PGP) traits of several dominant culturable isolates and evaluated the growth-promoting effects of these twenty-one isolates through pot experiments. Both studies showed that HM contamination increased the diversity and richness of corn grain EFs and affected the most dominant endophytes. Nigrospora and Fusarium were the most prevalent culturable endophytes in HM-contaminated areas. Conversely, Cladosporium spp. were the most isolated endophytes in non-contaminated areas. Different from this, Saccharomycopsis and Fusarium were the dominant EFs in HM-contaminated sites, while Neofusicoccum and Sarocladium were dominant in non-contaminated sites, according to a culture-independent analysis. PGP trait tests indicated that 70% of the tested isolates (forty-two) exhibited phosphorus solubilization, IAA production, or siderophore production activity. Specifically, 90% of the tested isolates from HM-contaminated sites showed better PGP results than 45% of the isolates from non-contaminated sites. The benefit of the twenty-one isolates on host plant growth was further studied through pot experiments, which showed that all the isolates could improve host plant growth. Among them, strains derived from HM-contaminated sites, including AK18 (Nigrospora), AK32 (Beauveria), SD93 (Gibberellia), and SD64 (Fusarium), had notable effects on enhancing the dry biomass of shoots and roots of maize under Cd stress. We speculate that the higher ratio of PGP EFs in corn grains from HM-contaminated areas may explain their competitiveness in such extreme environments. Fusarium and Cladosporium isolates show high PGP properties, but they can also be phytopathogenic. 28. Dominant Dissolved Oxygen-Independent Pathway to Form Hydroxyl Radicals and the Generation of Reactive Chlorine and Nitrogen Species in Breakpoint Chlorination

Abstract

Due to the complexities of the interactions between ammonia, chlor(am)ine, and intermediate species such as ONOOH, the radical formation in breakpoint chlorination and the consequential removal of micropollutants remain largely unexplored. In this study, the dominant generation pathway of HO center dot, as a primary radical in breakpoint chlorination, was examined, and the generations of HO center dot, reactive chlorine species (RCS), and reactive nitrogen species (RNS) were quantitatively evaluated. A dissolved oxygen (DO)-independent pathway was verified by 18O labeling and contributed over 90% to HO center dot generation. The commonly believed pathway, the decomposition of ONOOH involving DO, contributed only 7% to HO center dot formation in breakpoint chlorination. The chlorine to nitrogen (Cl/N) ratio and pH greatly affected the generations and speciations of the reactive species. An optimum Cl/N mass ratio for HO center dot, Cl2 center dot-, and RNS generations occurred at the breakpoint (i.e., Cl/N mass ratio = 9), whereas excessive free chlorine shifted the radical speciation toward ClO center dot at Cl/N mass ratios above the breakpoint. Basic conditions inhibited the generations of HO center dot and RNS but significantly promoted that of ClO center dot. These findings improved the fundamental understanding of the radical chemistry of breakpoint chlorination, which can be extended to estimate the degradations of micropollutants of known rate constants toward the reactive species with influences from the Cl/N ratio and pH in real-world applications.

29. Role of UV-based advanced oxidation processes on NOM alteration and DBP formation in drinking water treatment: A state-of-the-art review

Abstract

Oxidative treatment of drinking water has been practiced for more than a century. UV-based advanced oxidation processes (UV-AOPs) have emerged as promising oxidative treatment technologies to eliminate recalcitrant chemicals and biological contaminants in drinking water. UV-AOPs inevitably alter the properties of natural organic matter (NOM) and affect the disinfection byproduct (DBP) formation in the post-disinfection. This paper provides a state-of-the-art review on the effects of UV-AOPs on the changes of NOM properties and the consequent impacts on DBP formation in the post-chlorination process. A tutorial review to the connotations of NOM properties (e.g., bulk properties, fractional constituents, and molecular structures) and the associated state-of-the-art analytical methods are firstly presented. The impacts of different radical-based AOPs on the changes of NOM properties together with the underlying NOM-radical reaction mechanisms are discussed. The impacts of alteration of NOM properties on DBP formation in the post-chlorination process are then reviewed. The current knowledge gaps and future research needs are finally presented, with emphases on the needs to strengthen the comparability of research data in literature, the accuracy in quantifying the reactive moieties of NOM, and the awareness of unknown DBPs in oxidative water treatment processes. The review and discussion improve the fundamental understanding of NOM-radical and NOM-chlorine chemistry. They also provide useful implications on the engineering design and operation of next-generation drinking water treatment plants. © 2022 Elsevier Ltd

30. Chlorine Reuse in UV/Electrochemical Oxidation to Advance Sewage Cotreatment of Landfill Leachate

Abstract

Landfill leachate cotreatment with municipal sewage remains a widespread practice in waste management. However, the quality of the resulting effluent is often compromised by dissolved organic matter (DOM) from leachate, especially when it exhibits high ultraviolet absorbance at 254 nm (UV254). While degrading the DOM through oxidation is a promising approach, substantial chemical dosing is required for landfill leachate. An emerging alternative is electrochemical oxidation with ultraviolet radiation (UV/EO), which generates chlorine and its derived radicals in situ. Yet, prolonged UV/EO introduces concerns of residual chlorine accumulation and consequent toxicity. In response, we adapted UV/EO to reuse the residual chlorine, wherein partially replacing the treated leachate with untreated volumes reduced the UV254 of the subsequent batch. The energy efficiency of the chlorine-reuse process was optimized by using the response surface methodology, adjusting the applied current density and Reynolds number. We incorporated a UV dechlorination unit in sequence, eliminating formulas recalcitrant to UV/EO and resulting in an overall 81% reduction in UV254. Particularly in areas with established cotreatment infrastructures, this approach advances landfill leachate management, having demonstrated minimal risks of acute toxicity and genotoxicity upon discharge with municipal sewage at 3% v/v. © 2023 American Chemical Society.

Abstract

Enterococcus faecalis is a common pathogen causing refractory periapical periodontitis and secondary intraradicular infections. In this study, E. faecalis YN771 isolated from a re-treated root canal at a stomatology department was used as the host bacterium and was co-cultured with wastewater from the same department and patient samples to isolate a phage that lyses E. faecalis. We studied the biological and genomic characteristics of this phage. Transmission electron microscopy showed that this phage’s head is icosahedral in structure, with a head diameter of around 98.4 nm, and a contractile tail of around 228.5 nm in length and a diameter of 17.3 nm. The phage was identified as a member of the Myoviridae family and named PEf771. It is sensitive to proteinase K but resistant to chloroform and Triton X-100. Its lytic cycle is 45 min, burst size is 78, optimal multiplicity of infection is 0.1, lysis spectrum is narrow, and host strain specificity is strong. Its optimal growth temperature is 37 °C, most suitable pH is 6.0, and is sensitive to ultraviolet radiation. Whole-genome sequencing of PEf771 indicated it has a genome size of 151 052 bp, with a GC content of 36.97%, and encodes 197 proteins plus 26 tRNAs. PEf771 is most closely related to E. faecalis phage EFDG1. Phage PEf771 has strong host specificity and lytic ability, so it is important to further characterize this phage and its interaction with E. faecalis.

Abstract

Phages are considered the most abundant and diverse biological entities on Earth. Although many phages have been isolated, most of this diversity remains completely unexplored. Recent advances in genome sequencing, however, have provided unprecedented glimpses into the virosphere. The lytic cold-active Siphoviridae bacteriophage VW6S, infecting the host Pseudomonas fluorescens W-6, was isolated from the Napahai plateau wetland in China. The newly sequenced phage VW6S contained double-stranded DNA with a genome size of 37,917 bp, an overall GC content of 56.90% and 52 open reading frames (ORFs). The genome was organized into several modules containing genes for packaging, structural proteins, replication/transcription, and cell lysis. The sequence contained 16 transcription terminators, no tRNAs. The lysis cassette of VW6S consisted of 4 proteins (holin protein, lysin protein and 2 putative spanins). This is the first report on cold-active SiphoviridaePseudomonas fluorescens bacteriophage genome sequencing.

Abstract

Carbon dots (CDs) have broad prospective applications in various fields, and expanding the applications of fluorescent CDs, especially for CDs derived from bacteria, is a major research goal. In this study, novel CDs derived from Escherichia coli BW25113 (WT) were successfully synthesized via a one-step hydrothermal method. Unlike previously developed CDs-E. coli, CDs-WT can be used for microbial imaging of both live and dead cells. We demonstrated the biocompatibility, excellent penetrability, and nontoxic characteristics of CDs-WT for use as fluorescent probes for bioimaging both in vitro and in vivo. Importantly, we provide the first demonstration of CDs-WT distribution in various organs of mice, including the ability to cross the blood-brain barrier and the potential for rapid excretion through the intestines. Additionally, CDs-WT can be instantly utilized as a fluorescent probe for the highly selective and rapid detection of p-nitrophenol (p-NP) by the inner filter effect, with a limit of detection for p-NP of 11 nM, the lowest value reported to date. Hence, our results demonstrate the feasibility of p-NP detection and extend the bio-imaging applications of CDs prepared from bacteria.
