Abstract Gas Insulated Switchgear (GIS) was a crucial electrical equipment that provides the safety and security of power systems, and finding the eco-friendly alternatives of sulphur hexafluoride (SF6) as insulating medium in GIS has been an urgent demand in past decades. However, few studies reported the specific dielectric strength under various electrodes and partial discharge (PD) decomposition products detection of applying the eco-friendly gas in GIS. In this paper, the insulation properties among C4F7N/CO2 mixtures, dry air and CO2 were explored under the reduced-scale gas-insulated experimental device. The relationship between AC breakdown voltage and gas pressure was gained under the different electrodes and gap distances, and 0.6 MPa 7% C4F7N/93% CO2, 0.9 MPa dry air, 0.9 MPa CO2 all possess the capacity to be the insulating medium in eco-friendly gas insulated switchgear. Furthermore, the partial discharge experimental was also carried out to realize the decomposition products detection of dry air, which designs three common defect types including metal protrusion defects, air gap defects, and metal contamination defects. The decomposition products detection result shows that the contains of CO2, CO, and NO2 linearly increase with the increasing applied voltages and times, and the partial discharge defects are distinguished according to the ratios of c(CO2 + CO)/c(NO2) and c(CO2)/c(CO). The results can provide the basis for the further development of eco-friendly gas insulated switchgear.

Abstract Existing natural language processing (NLP) methods to convert free-text clinical notes into structured data often require problem-specific annotations and model training. This study aims to evaluate ChatGPT’s capacity to extract information from free-text medical notes efficiently and comprehensively. We developed a large language model (LLM)-based workflow, utilizing systems engineering methodology and spiral “prompt engineering” process, leveraging OpenAI’s API for batch querying ChatGPT. We evaluated the effectiveness of this method using a dataset of more than 1000 lung cancer pathology reports and a dataset of 191 pediatric osteosarcoma pathology reports, comparing the ChatGPT-3.5 (gpt-3.5-turbo-16k) outputs with expert-curated structured data. ChatGPT-3.5 demonstrated the ability to extract pathological classifications with an overall accuracy of 89%, in lung cancer dataset, outperforming the performance of two traditional NLP methods. The performance is influenced by the design of the instructive prompt. Our case analysis shows that most misclassifications were due to the lack of highly specialized pathology terminology, and erroneous interpretation of TNM staging rules. Reproducibility shows the relatively stable performance of ChatGPT-3.5 over time. In pediatric osteosarcoma dataset, ChatGPT-3.5 accurately classified both grades and margin status with accuracy of 98.6% and 100% respectively. Our study shows the feasibility of using ChatGPT to process large volumes of clinical notes for structured information extraction without requiring extensive task-specific human annotation and model training. The results underscore the potential role of LLMs in transforming unstructured healthcare data into structured formats, thereby supporting research and aiding clinical decision-making.

Accurately estimating the volume of woody vegetation is critical for assessing fuel characteristics and associated wildfire risks in shrublands. However, few studies have investigated the branch volume of multi-stemmed shrubs, a dominant life form in wildfire-prone drylands. This study predicts branch volume using the inflection point of branch diameter. This inflection point, identified using the “Segmented” package in R, marks the transition from a gradual decrease to a significant reduction in diameter along the stem. The volume of branch segment above this point is calculated as a cone, and below it, a cylinder. We validated this method on various species such as Caragana korshinskii, Salix psammophila, and Vitex negundo. Good estimations were achieved with an average 19.2% bias relative to reference branch volumes, outperforming conventional methods that subjectively treated the whole branch as either a cylinder (96.9% bias) or a cone (−34.4% bias). We tallied branches by basal diameter and provided inventories for easily locating the inflection point, as well as using two-way branch volume tables for rapid volume predictions in shrubland. In general, we developed an effective method for estimating branch volumes of multi-stemmed shrubs, enabling its application to larger-scale shrubland volumetric prediction. This advancement supports wildfire hazard assessment and informs decision-making in fuel treatments.

Polyimide (PI) has been widely used in cable insulation, thermal insulation, wind power protection, and other fields due to its high chemical stability and excellent electrical insulation and mechanical properties. In this research, a modified PI composite film (MoS2@PDA/PI) was obtained by using polydopamine (PDA)-coated molybdenum disulfide (MoS2) as a filler. The low interlayer friction characteristics and high elastic modulus of MoS2 provide a theoretical basis for enhancing the flexible mechanical properties of the PI matrix. The formation of a cross-linking structure between a large number of active sites on the surface of the PDA and the PI molecular chain can effectively enhance the breakdown field strength of the film. Consequently, the tensile strength of the final sample MoS2@PDA/PI film increased by 44.7% in comparison with pure PI film, and the breakdown voltage strength reached 1.23 times that of the original film. It can be seen that the strategy of utilizing two-dimensional (2D) MoS2@PDA nanosheets filled with PI provides a new modification idea to enhance the mechanical and electrical insulation properties of PI films.

Oral submucous fibrosis is a chronic, inflammatory and potentially malignant oral disease. Local delivery of triamcinolone to lesion site is a commonly used therapy. The existing methods for local drug delivery include topical administration and submucosal injection. However, in the wet and dynamic oral microenvironment, these methods have drawbacks such as limited drug delivery efficiency and injection pain. Therefore, it is urgently needed to develop an alternative local drug delivery system with high efficiency and painlessness. Inspired by the structure of band-aid, this study proposed a novel double-layered mucoadhesive microneedle patch for transmucosal drug delivery. The patch consisted of a mucoadhesive silk fibroin/tannic acid top-layer and a silk fibroin microneedle under-layer. When applying the annealing condition for the medium content of β-sheets of silk fibroin, the microneedles in under-layer displayed both superior morphology and mechanical property. The mechanical strength of per needle (0.071N) was sufficient to penetrate the oral mucosa. Sequentially, the gelation efficiency of silk fibroin and tannic acid in top-layer was maximized as the weight ratio of tannic acid to silk fibroin reached 5:1. Moreover, in vitro results demonstrated the double-layered patch possessed undetectable cytotoxicity. The sustained release of triamcinolone was observed from the double-layered patch for at least 7 days. Furthermore, compared with other commercial buccal patches, the double-layered patch exhibited an enhanced wet adhesion strength of 37.74 kPa. In addition, ex vivo mucosal tissue penetration experiment confirmed that the double-layered patch could reach the lamina propria, ensuring effective drug delivery to the lesion site of oral submucous fibrosis. These results illustrate the promising potential of the drug-loaded mucoadhesive microneedle patch for the treatment of oral submucous fibrosis.

In this study, the coupled use of a double dielectric barrier discharge (DDBD) and CoOOH catalyst was investigated for the degradation of methylene blue (MB). The results indicated that the addition of CoOOH significantly promoted MB degradation performance compared to the DDBD system alone. In addition, both the removal rate and energy efficiency increased with an increase in CoOOH dosage and discharge voltage. After 30 min of discharge treatment in the coupled system (with CoOOH of 150 mg), the removal rate reached 97.10% when the discharge voltage was 12 kV, which was 1.92 times that in the single DDBD system. And when the discharge time was 10 min, the energy efficiency could reach 0.10 g (k·Wh)−1, which was 3.19 times better than the one in the single DDBD system. Furthermore, the addition of CoOOH could also significantly enhance the TOC and COD removal rates of MB. In the DDBD-coupled-with-CoOOH system, TOC and COD were 1.97 times and 1.99 times those of the single DDBD system after 20 min of discharge treatment with a discharge voltage of 12 kV and 100 mg of CoOOH. The main active substances detected in the coupled system indicated the conversion of the active species H2O2 and O3 into a more oxidizing ·OH was enhanced through the addition of a CoOOH catalyst, resulting in the more effective decomposition of MB and intermediate molecules.

The therapeutic precision and clinical applicability of drug-eluting coatings can be substantially improved by facilitating tunable drug delivery. However, the design of coatings which allows for precise control over drug release kinetics is still a major challenge. Here, a double-layered silk fibroin (SF) coating system was constructed by sequential electrophoretic deposition. A mixture of dissolved Bombyx mori SF (bmSF) molecules and pre-made bmSF nanospheres at different ratios was deposited as under-layer. Subsequently, this underlayer was covered by a top-layer comprising Antheraea pernyi SF (apSF) molecules (rich in arginylglycylaspartic acid, RGD) to improve the cellular response of the resulting double-layered coatings. Additionally, model drug doxycycline was either pre-mixed with dissolved bmSF molecules or pre-loaded into pre-made bmSF nanospheres at the same amount before their mixing and deposition. The thickness and nanosphere content of the under-layer architecture were proportional to the deposition time and nanosphere concentration in precursor mixtures, respectively. The surface topography, wettability, degradation rate and adhesion strength were comparable within the double-layered coating system. As expected, RGD-rich apSF top-layer improved cell adhesion, spreading and proliferation compared with bmSF top-layer. Furthermore, the amount and duration of drug release increased linearly with increasing nanosphere concentration at fixed deposition time, whereas drug release amount increased linearly with increasing deposition time. These results indicate that the dosage and kinetics of loaded drugs can be quantitatively tailored by altering nanosphere concentration and deposition time as main processing parameters. Overall, this study illustrates the strong potential of pre-defining coating architecture to facilitate control over drug delivery.

Cinnamon oil is a blend of secondary metabolites and is widely used as spice. Endophytic bacteria are always related to the secondary metabolites production. However, the potential of endophytic bacteria communities for cinnamon oil production during cinnamon shade-drying process is still not clear. In this study, we investigated the composition and metabolic function of endophytic bacterial community during 80-day shade-drying process. The temporal dynamics of essential oil content and its dominant constituents were analyzed. The succession of endophytic bacterial community from d0 to d80 was identified. The influence of endophytic bacterial community evolution on cinnamon oil is significant positive. Predictive functional analysis indicated that shade-drying process was rich in Saccharopolyspora that produce enzymes for the conversion of phenylalanine to cinnamaldehyde. These findings enhance our understanding of the functional bacterial genera and functional genes involved in the production of cinnamon oil during cinnamon shade-drying process.

BackgroundObesity plays an important role in type 2 diabetes mellitus (T2DM) and myocardial infarction (MI). Ferroptosis and ferritinophagy are related to metabolic pathways, such as fatty acid metabolism and mitochondrial respiration. We aimed to investigate the ferroptosis- and autophagy-related differentially expressed genes (DEGs) that might be potential targets for MI progression.MethodsGSE116250 was analyzed to obtain DEGs. A Venn diagram was used to obtain the overlapping ferroptosis- and autophagy-related DEGs. The enrichment pathway analysis was performed and the hub genes were obtained. Pivotal miRNAs, transcription factors, and drugs with the hub genes interactions were also predicted. The MI mice model was constructed, and qPCR analysis and single-cell sequencing were used to validate the hub genes.ResultsUtilizing the limma package and the Venn diagram, 26 ferroptosis-related and 29 autophagy-related DEGs were obtained. The list of ferroptosis-related DEGs was analyzed, which were involved in the cellular response to a toxic substance, cellular oxidant detoxification, and the IL-17 signaling pathway. The list of autophagy-related DEGs was involved in the regulation of autophagy, the regulation of JAK-STAT signaling pathway, and the regulation of MAPK cascade. In the protein-protein interaction network, the hub DEGs, such as IL-6, PTGS2, JUN, NQO1, NOS3, LEPR, NAMPT, CDKN2A, CDKN1A, and Snai1, were obtained. After validation using qPCR analysis in the MI mice model and single-cell sequencing, the 10 hub genes can be the potential targets for MI deterioration.ConclusionThe screened hub genes, IL-6, PTGS2, JUN, NQO1, NOS3, LEPR, NAMPT, CDKN2A, CDKN1A, and Snai1, may be therapeutic targets for patients with MI and may prevent adverse cardiovascular events.

BackgroundThe macrophages are involved in all stages of cardiovascular diseases, demonstrating the correlation between inflammation, atherosclerosis, and myocardial infarction (MI). Here, we aim to investigate macrophages-related genes in the deterioration of atherosclerosis.MethodsGSE41571 was downloaded and the abundance of immune cells was estimated by utilizing the xCell. By utilizing the limma test and correlation analysis, differentially expressed macrophages-related genes (DEMRGs) were documented. The functional pathways and the protein–protein interaction (PPI) network were analyzed and the hub DEMRGs were obtained. The hub DEMRGs and their interactions were analyzed using NetworkAnalyst 3.0 and for validation, the expressions of hub DEMRGs were analyzed using the GSE135055 and GSE116250 datasets as well as atherosclerosis and MI mice model.ResultsA total of 509 differentially expressed genes (DEGs) were correlated with the abundance of macrophages and were identified as DEMRGs (Pearson correlation coefficients (PCC) > 0.6), which were mainly enriched in extracellular structure organization, lysosomal membrane, MHC protein complex binding, and so on. After screening out, 28 hub DEMRGs were obtained with degrees ≥20, including GNAI1 (degree = 113), MRPS2 (degree = 56), HCK (degree = 45), SOCS3 (degree = 40), NET1 (degree = 28), and so on. After validating using Gene Expression Omnibus (GEO) datasets and the atherosclerosis and MI mice model, eight proteins were validated using ApoE-/- and C57 mice. The expression levels of proteins, including SYNJ2, NET1, FZD7, LCP2, HCK, GNB2, and PPP4C were positively correlated to left ventricular ejection fraction (LVEF), while that of EIF4EBP1 was negatively correlated to LVEF.ConclusionThe screened hub DEMRGs, SYNJ2, NET1, FZD7, LCP2, HCK, GNB2, EIF4EBP1, and PPP4C, may be therapeutic targets for treatment and prediction in the patients with plaque progression and MI recurrent events. The kit of the eight hub DEMRGs may test plaque progression and MI recurrent events and help in the diagnosis and treatment of MI-induced heart failure (HF), thus decreasing mortality and morbidity.

Abstract Silk fibroin (SF) can be used to construct various stiff material interfaces to support bone formation. An essential preparatory step is to partially transform SF molecules from random coils to β-sheets to render the material water insoluble. However, the influence of the SF conformation on osteogenic cell behavior at the material interface remains unknown. Herein, three stiff SF substrates were prepared by varying the β-sheet content (high, medium, and low). The substrates had a comparable chemical composition, surface topography, and wettability. When adsorbed fibronectin was used as a model cellular adhesive protein, the stability of the adsorbed protein-material interface, in terms of the surface stability of the SF substrates and the accompanying fibronectin detachment resistance, increased with the increasing β-sheet content of the SF substrates. Furthermore, (i) larger areas of cytoskeleton-associated focal adhesions, (ii) higher orders of cytoskeletal organization and (iii) more elongated cell spreading were observed for bone marrow-derived mesenchymal stromal cells (BMSCs) cultured on SF substrates with high vs. low β-sheet contents, along with enhanced nuclear translocation and activation of YAP/TAZ and RUNX2. Consequently, osteogenic differentiation of BMSCs was stimulated on high β-sheet substrates. These results indicated that the β-sheet content influences osteogenic differentiation of BMSCs on SF materials in vitro by modulating the stability of the adsorbed protein-material interface, which proceeds via protein-focal adhesion-cytoskeleton links and subsequent intracellular mechanotransduction. Our findings emphasize the role of the stability of the adsorbed protein-material interface in cellular mechanotransduction and the perception of stiff SF substrates with different β-sheet contents, which should not be overlooked when engineering stiff biomaterials.

Fatigue failure is invariably the most crucial failure mode for metallic structural components. Most microstructural strategies for enhancing fatigue resistance are effective in suppressing either crack initiation or propagation, but often do not work for both synergistically. Here, we demonstrate that this challenge can be overcome by architecting a gradient structure featuring a surface layer of nacre-like nanolaminates followed by multi-variant twinned structure in pure titanium. The polarized accommodation of highly regulated grain boundaries in the nanolaminated layer to cyclic loading enhances the structural stability against lamellar thickening and microstructure softening, thereby delaying surface roughening and thus crack nucleation. The decohesion of the nanolaminated grains along horizonal high-angle grain boundaries gives rise to an extraordinarily high frequency (≈1.7 × 103 times per mm) of fatigue crack deflection, effectively reducing fatigue crack propagation rate (by 2 orders of magnitude lower than the homogeneous coarse-grained counterpart). These intriguing features of the surface nanolaminates, along with the various toughening mechanisms activated in the subsurface twinned structure, result in a fatigue resistance that significantly exceeds those of the homogeneous and gradient structures with equiaxed grains. Our work on architecting the surface nanolaminates in gradient structure provides a scalable and sustainable strategy for designing more fatigue-resistant alloys.

When delivered across a cell, certain pulsed electric fields can cause an increase in the cell membrane permeability through a biophysical process known as electroporation. The current signals during the electric pulses could be used as a method for noninvasive electroporation measurement because of the sharp change in the conductivity of cells due to electroporation. To add to the existing knowledge on electroporation current signals under different pulse parameters, we undertook a study in which the electric current across the cells was recorded during electroporation pulses. The experimental current response to a pulsed electric field consisted of three stages: a) a rapid initial increase followed by b) an exponential decrease and then c) a monotonic increase. The rise time of the current signals was not affected by the intensity of the electric field or the number of pulses. However, the time at which the current increased again, deemed the electroporation onset time, shortened as the electric field became more intense and as the number of pulses increased. The transient conductivity change rate, defined to describe the electroporation degree during the pulse, increased under a higher electric field strength. However, the transient conductivity change rate first decreased and then gradually increased with additional pulses. This work may provide insight into the change in current during real-time electroporation detection.

In order to improve the anti-disturbance performance of permanent magnet synchronous motor (PMSM) servo system, a sliding-mode control strategy using a new reaching law (NRL) is proposed. The NRL incorporates power term and switching gain term of the system state variables into the conventional exponential reaching law (CERL), which can effectively suppress the sliding-mode chattering and increase the convergence rate of system state reaching sliding-mode surface. Based on this new reaching law, a sliding-mode speed controller (SMSC) of PMSM is designed. At the same time, to solve the chattering problem caused by the large sliding-mode switching gain, an anti-disturbance sliding-mode speed controller method with an extended sliding-mode disturbance observer (ESMDO), called SMSC+ESMDO method, is developed. The sliding-mode disturbance observer is designed to accurately estimate the motor speed and external load disturbances, and the disturbance estimator is used as a feed-forward to compensate the sliding-mode speed controller (SMSC) to improve the system robustness and reduce the system chattering. Simulation and experimental results show that the proposed compound sliding-mode control strategy can effectively improve the dynamic performance and robustness of the system compared with the PI controller.

The output power of PV array changes with the variation of environmental factors, such as temperature and solar irradiation. Therefore, a maximum power point (MPP) tracking (MPPT) algorithm is essential for the photovoltaic generation system. However, the P-U curve changes dynamically with the variation of the environmental factors; here, the misjudgment may occur if a simple perturb-and-observe (P&O) MPPT algorithm is used. In order to solve this problem, this paper takes MPPT as the main research object, and an improved MPPT algorithm for PV generation applications based on P-U curve reconstitution is proposed. Firstly, the mathematical model of PV array is presented, and then the output dynamic characteristics are analyzed. Based on this, a P-U curve reconstitution strategy is introduced, and the improved MPPT algorithm is proposed. At last, simulation and comparative analysis are conducted. Results show that, with the proposed algorithm, MPP is tracked accurately, and the misjudgment problem is solved effectively.

Aquatic environmental deterioration is becoming a serious problem due to rapid urbanization and economic development, particularly in developing countries. As two important components of the aquatic environment, water quality and sediment pollution are widely considered to be concerns; however, they are considered separately in most cases. The relationship between water quality and sediment pollution with heavy metals has been little addressed. In this study, the Haihe River Basin (HRB), one of the most polluted areas in China, was used as a case study, and the eutrophication index (EI) and the potential ecological risk index (RI) were employed to evaluate water quality and sediment pollution of heavy metals, respectively. The results showed that generally in the HRB, the water quality was poor, while the risk of heavy metal pollution was relatively low. Surface water quality was mainly influenced by sewage discharges from human daily life, and heavy metal pollution was affected by industry structure, in that the areas with resource/energy consumption industries and high-pollution industries often have high risks of heavy metal pollution Synergic pollution from water eutrophication and sediment pollution with heavy metals was found, especially in the central areas of the HRB, and it was largely dependent on the type of human activities. In the places with intensive human activities, such as secondary industry, eutrophication occurred simultaneously with heavy metal pollution, other than in less human-affected areas. These findings are useful for planning aquatic environment protections and river ecosystem management.
