Sox2 is an embryonal stem cell Ag essential for early embryonic development, tissue homeostasis and immune regulation. In the current study, one complete Sox2 cDNA sequence was cloned from freshwater bivalve Anodonta woodiana and named AwSox2. Histological changes of testis derived from Bisphenol A (BPA) treatment were analyzed by hematoxylin and eosin staining. Expressions of AwSox2 derived from BPA, LPS and polyinosinic:polycytidylic (Poly I:C) challenge were measured by quantitative real-time PCR. The full-length cDNA of AwSox2 contained an open reading frame of 927 nucleotides bearing the typical structural features of Sox2 family. Obvious degeneration, irregular arrangement of spermatids, and clotted dead and intertwined spermatids were observed in BPA-treated groups. Administration of BPA could result in a dose-dependent up-regulation of AwSox2 expression in the male gonadal tissue of A. woodiana. In addition, expression of AwSox2 was significantly induced by LPS and Poly I:C treatment in the hepatopancreas, gill and hemocytes, compared with that of control group. These results indicated that up-regulations of AwSOx2 are closely related to apoptosis of spermatogonial stem cells derived from BPA treatment as well as enhancement of immune defense against LPS and Poly I:C challenge in A. woodiana. Aim Of Study: This study is to investigate the effects of a novel peroxisome proliferator-activated receptor (PPAR) α/γ dual agonist TZD18 on cell growth, apoptosis, caspase activity, mitochondrial membrane potential, cytochrome c release, and apoptotic-related protein expression in MKN-45 cells.Materials and Methods: 3-(4, 5-dimethylthiazolyl)-2,5-diphenyltetrazolium bromide assay against various human cancer cell lines was performed to investigate the whether TZD18 could in reduce the proliferation rates of cancer cells. The percentages of apoptotic cells and mitochondrial membrane potential level were determined by flow cytometry. The subcellular localization of cytochrome c was examined by immunofluorescence microscopy. Western blotting assay was performed to reveal the expression of apoptosis-related proteins.Results: The results showed that the administration of TZD18 could inhibit the growth of MKN-45 cells in a dose- and time-dependent manner. In addition, the apoptotic ratio increased sharply along with a significant increase of caspase activities, mitochondrial membrane potential, and cytochrome c release following TZD18 exposure. The expression of Bax and p27kip1 increased significantly, whereas the expression level of Bcl-2 protein was downregulated.Conclusion: These results indicated that the administration of PPAR α/γ agonist TZD18 may inhibit cell growth by inducing the apoptotic process in MKN-45 cells. Aims: To explore effect of Ca2+ and Cd2+ on the calmodulin (CaM), one complete cDNA sequence (AwCaM1) was cloned and characterized from the freshwater mussel Anodonta woodiana and its expressions were analyzed. Materials and methods: The AwCaM1 was cloned from the A. woodiana using the rapid amplification of cDNA ends methods and its expression was determined by real-time PCR. Results: In the hepatopancreas, AwCaM1 expression was up-regulated with a time and dose dependent pattern in the Ca2+ treated groups (0.01, 0.02, 0.04 and 0.08 mg/L) during experiment observed, and increased more than 56.15% (p<0.05) compared with that of control group. AwCaM1 mRNA level increased more 65.04% (p<0.05) in the Cd2+ treated groups (8 and 16 mg/L). In the gill, AwCaM1 expression increased more than 79.41% (p<0.05) compared with that of control group in all the Ca2+ treated groups, and more than 88.23% (p<0.05) in all the Cd2+ treated groups. Conclusion: These results indicated that up-regulations of AwCaM1 expression in bivalve A. woodiana are associated with Ca2+ absorb and environmental adaption derived from Ca2+ and Cd2+ treatment. Objective: Insects dwelled in soil play a key role in monitoring of metal contaminations. In order to explore the toxicity of lead (Pb) in soil, juvenile of oriental beetle Anomala corpulenta, were firstly applied to analyze effect of Pb. Methods: In the current study, toxicity of different concentrations Pb on A. corpulenta in the laboratory was performed by measuring survival, growth and avoidance of animals. Meanwhile, activities of acetylcholinesterase (ChE), superoxide dismutase (SOD), catalase (CAT) and glutathione peroxidase (GSH-Px) in A. corpulenta were examined to quantify the extent of alterations caused by Pb treatments. Result: Events of mortality were occurred in the Pb treated groups (1200 and 1600 mg Pb/kg soil). Administration of Pb treatments could result in a significant decrease of mean end weights in contrasted with that of control group. Treatment of Pb at 800 mg Pb/kg soil caused an obviously avoidance behavior. Expressions of ChE, SOD, CAT and GSH-Px of Pb treated groups were significant decreased compared with that of control group in the experiment observed. Conclusion: Pb contamination of soil may cause multiple effects on juvenile A. corpulenta including occurrence of mortality, delay of growth and avoidance of contaminated environment. Objective: To investigate the role of Eclipta prostrata (E. prostrata) extract in improving spatial learning and memory deficits in D-galactose-induced aging in rats., Methods: Rats were divided into five groups, with 10 animals in each group. Aging rats were produced by treatment with 100 mg·kg-1·d-1 of D-galactose for 6 weeks. Rats in the E. prostrata treatment groups received an aqueous extract of E. prostrata orally at a concentration of 50, 100, or 200 mg·kg-1·d-1 for 3 weeks. Animals in both the normal and model groups were treated with similar volumes of saline. Spatial memory performance was measured using the Morris water maze. The mRNA levels and enzyme activities of superoxide dismutase (SOD), catalase (CAT), glutathione peroxidase (GPx) and glutathione reductase (GR) were analyzed using real-time quantitative PCR and spectrophotometry, respectively. The levels of induced nitric oxide synthase (iNOS), nitric oxide (NO), dopamine (DA), norepinephrine (NE), and serotonin (5-HT) were determined using enzyme-linked immunosorbent assay and spectrophotometry., Results: Compared with the normal group, rats in the D-galactose-treated model group exhibited significant memory loss. There was severe damage to the hippocampal CA1 area, and expression levels of SOD, CAT, GPx, and GR were significantly decreased in the model group compared with the normal group. In the model group, levels of iNOS and NO were significantly increased compared with the normal group. However, treatment with E. prostrata extract reversed the conditions caused by D-galactose-induced aging, especially in the groups with higher treatment concentrations. Compared with the normal group, the levels of DA, NE, and 5-HT were significantly lower in the D-galactose-treated model group. In the E. prostrata extract-treated groups, however, there was a dose-dependent upregulation of DA, NE, and 5-HT expression., Conclusion: Our results suggest that administration of E. prostrata extract can result in an improvement in the learning and memory impairments that are induced by D-galactose treatment in rats. This improvement may be the result of enhanced antioxidative ability, decreased iNOS and NO levels, and the induction of DA, NE, and 5-HT expression in the brain.
